Abstract
本発明は、逆ナノディスクと、当該逆ナノディスクが親油性溶媒中に分散している親油性分散液を提供することを目的とする。本発明に係る逆ナノディスクは、非イオン界面活性剤、およびリポペプチドバイオサーファクタントを含有し、前記非イオン界面活性剤が、その親油性部分を外側に、その親水性部分を内側にして脂質二重層を形成し、前記リポペプチドバイオサーファクタントが前記脂質二重層の側面に結合していることを特徴とする。
Description
本発明は、逆ナノディスクと、当該逆ナノディスクが親油性溶媒中に分散している親油性分散液に関するものである。
化粧料や医薬部外品、医薬品などにおいて有効成分を皮膚に浸透させる技術として、ミセルやベシクル等の分子集合体が用いられることがある。一般的なミセルは、親水性溶媒中で親水性部分が外側を向き、親油性部分が内側を向いて界面活性剤が集合することにより形成される。ベシクルは二層以上の界面活性剤層で構成される球状粒子である。特にリン脂質や糖脂質で構成されたベシクルはリポソームと呼ばれている。一般にリポソームの大きさは0.1~数百μmと比較的大きく、これを親水性溶媒中に含む親水性分散液は白濁したものである。
近年、リポソームと同様に脂質二重層からなるナノ粒子またはナノディスクと呼ばれる微細な粒子が開発されている。一般的なナノディスクは、脂質二重層の側面に膜スキャフォールドタンパク質が結合して固定化された微粒子をいい、この様な構造の微粒子が親水性溶媒に分散している親水性分散物は、医薬品、医薬部外品、化粧品などの成分として利用されている。例えば本発明者らは、脂質二重層とリポペプチドバイオサーファクタントを含むナノディスクを開発している(特許文献1)。
一方、一般的なベシクルとは逆に、親油性溶媒中で最外面が脂質の親油性層からなり、内部に親水性層を有する構造の逆ベシクルも存在するが、その報告事例は少なく、その特性、用途、製法などについても十分研究が進んでいるとは言い難い。例えば特許文献2には、マンノシルアルジトールリピッド等の糖型バイオサーファクタント化合物と自己組織化2分子膜からなる逆ベシクルが開示されている。特許文献3には、シリコーン界面活性剤、水、および油剤を含有する逆ベシクル組成物が開示されている。特許文献4には、非イオン界面活性剤として特定の脂肪酸エステルを含む逆ベシクル(反転ベシクル)が開示されている。特許文献5には、特定のスフィンゴシン類、脂肪酸、炭化水素油、エステル油、および水を含む逆ベシクル組成物が開示されている。
上述したように、逆ベシクルは知られているが、一般的な逆ベシクルやベシクルの粒子径は比較的大きく、その分散液は白濁している。それに対してナノディスクの粒子径は比較的小さく、その分散液は肉眼では透明であるが、逆ベシクル自体の研究が未だ十分ではなく、逆ナノディスクは知られていない。しかし逆ナノディスクが実用化できれば、産業上の利用価値は非常に高いと考えられる。
そこで本発明は、逆ナノディスクと、当該逆ナノディスクが親油性溶媒中に分散している親油性分散液を提供することを目的とする。

本発明者らは、上記課題を解決するために鋭意研究を重ねた。その結果、脂質二重層を構成する脂質として非イオン界面活性剤を用い、且つ膜スキャホールド化合物としてリポペプチドバイオサーファクタントを用いることにより逆ナノディスクを製造できることを見出して、本発明を完成した。

本発明によれば、実用化できれば産業上の利用価値が非常に高いと考えられる逆ナノディスクが提供される。
本発明に係る逆ナノディスクは、非イオン界面活性剤とリポペプチドバイオサーファクタントを含有する。
非イオン界面活性剤とは、イオン性基を有さない親水性部分と親油性部分を有し、界面活性を示す化合物をいう。非イオン界面活性剤としては、例えば、ショ糖脂肪酸エステル、ソルビタン脂肪酸エステル、ポリオキシエチレンソルビタン脂肪酸エステル、ポリオキシエチレンアルキルエーテル、ポリオキシエチレンアルキルフェノールエーテル、脂肪酸アルカノールアミド等が挙げられる。
ショ糖脂肪酸エステルは、ショ糖と長鎖脂肪酸とのエステルであり、例えば、ショ糖ラウリン酸エステル、ショ糖ミリスチン酸エステル、ショ糖パルミチン酸エステル、ショ糖ステアリン酸エステル、およびショ糖オレイン酸エステル等のショ糖C12-20脂肪酸エステルが挙げられる。
ソルビタン脂肪酸エステルは、ソルビタンと長鎖脂肪酸とのエステルであり、例えば、ソルビタンモノオレエート、ソルビタンイソステアレート、ソルビタンモノラウレート、ソルビタンモノパルミテート、ソルビタンモノステアレート、ソルビタンセスキオレエート、ソルビタントリオレエート等のソルビタンC12-20脂肪酸エステルが挙げられる。
ポリオキシエチレンソルビタン脂肪酸エステルは、一般式R-COO-[ソルビタン]-((CH2CH2O)n-H)m(mは1以上、3以下の整数を示し、nは10以上、100以下の整数を示す。)で表される、ソルビタン脂肪酸エステルに酸化エチレンを付加したエステル・エーテル型の非イオン界面活性剤であり、例えば、POE(20)ソルビタンモノステアレートやPOE(20)ソルビタントリステアレートが挙げられる。
ポリオキシエチレンアルキルエーテルは、一般式R-O-(CH2CH2O)p-Hで表され、アルコールエトキシレートとも呼ばれ、アルコールを酸化エチレンでエトキシ化して得られるエーテル型の非イオン界面活性剤であり、例えば、エチレンオキサイド単位の繰返し数pが20以上、200以下のPOE-C12-20アルキルエーテルが挙げられる。
ポリオキシエチレンアルキルフェノールエーテルは、一般式R-(C6H4)-O-(CH2CH2O)q-H(qは5以上、60以下の整数を示す。)で表されるエーテル型の非イオン界面活性剤であり、例えば、ポリ(オキシエチレン)オクチルフェニルエーテルが挙げられる。
脂肪酸アルカノールアミドは、アンモニアの水素がR-CO-基と2つのエタノール基で置換された3級アミドであり、一般式R-CON(CH2CH2OH)2で表され、例えば、ラウリン酸ジエタノールアミド、ラウリン酸ミリスチン酸ジエタノールアミド、ステアリン酸ジエタノールアミド、オレイン酸ジエタノールアミド、C8-18脂肪酸メチルエタノールアミド、ヤシ油脂肪酸ジエタノールアミド、パーム核油脂肪酸ジエタノールアミド、モディファイドヤシ油脂肪酸ジエタノールアミド、ヤシ油脂肪酸モノエタノールアミド、ステアリン酸モノエタノールアミド、ラウリン酸モノイソプロパノールアミド、ヤシ油脂肪酸N-メチルエタノールアミドが挙げられる。
1種のみの非イオン界面活性剤を単独で用いてもよいし、2種以上の非イオン界面活性剤を併用してもよい。非イオン界面活性剤としては、ショ糖脂肪酸エステルが好ましい。
リポペプチドバイオサーファクタントは、疎水性基と、親水性部分を含むペプチド構造を有し、界面活性作用を示すものであり、微生物が生産するものをいう。リポペプチドバイオサーファクタントとしては、例えば、サーファクチン、アルスロファクチン、イチュリン、フェンジシン、セラウェッチン、ライケシン、ビスコシンを挙げることができる。
リポペプチドバイオサーファクタントとしては、下記式(I)で表されるサーファクチンまたはその塩(以下、「サーファクチン(I)」という)が好ましい。式(I)中、カルボキシメチル基およびカルボキシエチル基の根元は光学活性点を表す。
式(I)中、Xは、ロイシン、イソロイシンおよびバリンから選択されるいずれか1種のアミノ酸残基を表す。Xとしてのアミノ酸残基は、L体でもD体でもよいが、L体が好ましい。
R1は、C9-18アルキル基を表す。ここで、「C9-18アルキル基」は、炭素数が9以上、18以下の直鎖状または分枝鎖状の一価飽和炭化水素基をいう。例えば、n-ノニル、6-メチルオクチル、7-メチルオクチル、n-デシル、8-メチルノニル、n-ウンデシル、9-メチルデシル、n-ドデシル、10-メチルウンデシル、n-トリデシル、11-メチルドデシル、n-テトラデシル、n-ペンタデシル、n-ヘキサデシル、n-ヘプタデシル、n-オクタデシルなどが挙げられる。
上記サーファクチン(I)は1種、または2種以上使用してもよい。例えば、R1のC9-18アルキル基が異なる複数のサーファクチン(I)を含むものであってもよい。
サーファクチン(I)は、公知方法に従って、微生物、例えばバチルス・ズブチリスに属する菌株を培養し、その培養液から分離することができ、精製品であっても未精製品であっても使用できる。例えば培養液のまま使用することもできる。また、化学合成法によって得られるものでも同様に使用できる。
サーファクチン(I)の塩も用いることができる。当該塩を構成するカウンターカチオンは特に制限されないが、例えばアルカリ金属イオンやアンモニウムイオンが挙げられる。
サーファクチン(I)の塩に使用できるアルカリ金属イオンは特に限定されないが、リチウムイオン、ナトリウムイオン、カリウムイオンなどを表す。また、2つのアルカリ金属イオンは、互いに同一であってもよいし、異なっていてもよい。
アンモニウムイオンの置換基としては、例えば、メチル、エチル、n-プロピル、イソプロピル、n-ブチル、t-ブチル等のアルキル基;ベンジル、メチルベンジル、フェニルエチル等のアラルキル基;フェニル、トルイル、キシリル等のアリール基等の有機基が挙げられる。アンモニウムイオンとしては、例えば、テトラメチルアンモニウムイオン、テトラエチルアンモニウムイオン、ピリジニウムイオン等が挙げられる。
なお、サーファクチン(I)の塩中、二つのカウンターカチオンは互いに同一であってもよいし、異なっていてもよいものとする。また、一方のカルボキシ基が-COOHまたは-COO-の状態になっていてもよいものとする。
一般的なナノディスクは、脂質二重層の側面部に両親媒性である膜スキャフォールドタンパク質の疎水性部分が結合して被覆し、且つ、脂質二重層において脂質の親水性部分が外側を向き、親油性部分が内側を向いている構造で安定化している。それに対して本発明に係る逆ナノディスクは、脂質二重層の側面疎水部にリポペプチドバイオサーファクタントが結合して被覆し、且つ、脂質二重層において脂質の親油性部分が外側を向き、親水性部分が内側を向いている構造で安定化していると考えられる。更に、環状ペプチドがイオンチャネルとして脂質二重層に刺さっていたり、嵩高い疎水性部分を有する脂質と嵩高い親水性部分を有する界面活性剤が二重層を形成している例も知られていることから、本発明に係る逆ナノディスクでも、一部のリポペプチドバイオサーファクタントが非イオン界面活性剤と共に二重層を形成している可能性もあり得る。
2以上の脂質二重層が互いに積層して多層構造を形成している場合がある。本発明に係る逆ナノディスクの大きさとしては、脂質二重層が単層の状態で、厚さが2nm以上、10nm以下、直径が5nm以上、20nm未満であってよい。当該厚さとしては3nm以上が好ましく、4nm以上がより好ましく、また、8nm以下が好ましく、6nm以下がより好ましい。当該直径としては、6nm以上が好ましく、7nm以上がより好ましく、また、15nm以下が好ましく、12nm以下がより好ましい。
本発明に係る逆ナノディスクは、非イオン界面活性剤およびリポペプチドバイオサーファクタント以外の成分を含んでいてもよい。例えば一価C4-20アルコールは、溶解性が十分でない一部の非イオン界面活性剤の溶解を助け、また、その両親媒性により非イオン界面活性剤を主成分とする脂質二重層の構成成分となり、当該脂質二重層の形成や安定化に寄与する可能性がある。
一価C4-20アルコールとしては、例えば、n-ブタノール、イソブタノール、s-ブタノール、t-ブタノール等のブタノール、ペンタノール、ヘキサノール、ヘプタノール、オクタノール、ノナノール、デカノール、ウンデカノール、ドデカノール、トリデカノール、テトラデカノール、ペンタデカノール、ヘキサデカノール、ヘプタデカノール、オクタデカノール、ノナデカノール、イコサノール等の一価C4-20アルコールを用いることができる。炭素数が4以上、20以下であれば、非イオン界面活性剤の脂質二重層の形成や安定化に寄与できると考えられる。アルコールとしては、一価C5-15アルコールが好ましく、一価C6-10アルコールがより好ましい。
本発明に係る逆ナノディスクは、水系溶媒を含んでいてもよい。本発明に係る逆ナノディスクの脂質二重層は、非イオン界面活性剤の親水性部分が集合した親水性層を内部に有し、水系溶媒はかかる親水性層中に存在することができ、一価C4-20アルコールと同様に、脂質二重層の形成や安定化に寄与すると考えられる。水系溶媒としては、例えば、水、および水混和性有機溶媒と水との混合溶媒が挙げられる。
水混和性有機溶媒とは、常温常圧で水と無制限に混和可能な有機溶媒をいい、例えば、メタノール、エタノール、n-プロパノール、イソプロパノール等の一価C1-4アルコールが挙げられ、エタノールおよび/またはイソプロパノールが好ましい。水系混合溶媒における水と水混和性有機溶媒との混合割合は適宜調整すればよいが、例えば、水系混合溶媒に対する水混和性有機溶媒の割合を50質量%以下とすることができる。当該割合としては、40質量%以下または30質量%以下が好ましく、20質量%以下または10質量%以下がより好ましく、5質量%以下、2質量%以下または1質量%以下がより更に好ましい。また、水系溶媒は緩衝液であってもよく、そのpHは特に制限されない。
本発明に係る逆ナノディスクの製造時には親油性溶媒を用いることから、脂質二重層には親油性溶媒が含まれる可能性がある。親油性溶媒は、水と混合した場合に二層を形成するものであれば特に制限されないが、例えば、脂肪族炭化水素溶媒、動植物油脂、水素添加動植物油脂、脂肪酸グリセリド、芳香族炭化水素溶媒などが挙げられる。
脂肪族炭化水素溶媒としては、例えば、ヘキサン、ヘプタン、オクタン、ノナン、デカン、ウンデカン、ドデカン等のC6-12脂肪族炭化水素溶媒の他、ミツロウ、キャンデリラロウ、米ぬかロウ等の混合脂肪族炭化水素溶媒が挙げられる。動植物油脂としては、パーム油、シア脂、サル脂、イリッペ脂、豚脂、牛脂、ナタネ油、米油、落花生油、オリーブ油、コーン油、大豆油、シソ油、綿実油、ヒマワリ油、月見草油、ゴマ油、サフラワー油、ヤシ油、カカオ脂、パーム核油、魚油、ワカメ油、コンブ油などが挙げられる。水素添加動植物油脂としては、パーム硬化油、パーム極度硬化油、ナタネ硬化油、ナタネ極度硬化油、大豆硬化油、豚脂硬化油、魚油硬化油などが挙げられる。脂肪酸グリセリドとしては、トリ-、ジ-、モノ-グリセリドのいずれでもよく、ステアリングリセリド、パルミチングリセリド、ラウリングリセリド等が挙げられる。芳香族炭化水素系溶媒としては、ベンゼン、トルエン、クロロベンゼン、ジクロロベンゼン等が挙げられる。親油性溶媒としては、脂肪族炭化水素溶媒が好ましく、C6-12脂肪族炭化水素溶媒またはC8-12脂肪族炭化水素溶媒がより好ましく、デカンがより更に好ましい。
本発明に係る逆ナノディスクの脂質二重層は、非イオン界面活性剤の親水性部分が集合した親水性層を内部に有し、非イオン界面活性剤の親油性部分が集合した親油性層をその外側に有する。よって、本発明に係る逆ナノディスクは、親水性化合物および/または親油性化合物を含み得る。
親水性化合物は、水分子に対して親和性を示す化合物であれば特に制限されないが、例えば、親水性化合物は、1gまたは1mLを水中に入れ、20±5℃で5分ごとに強く30秒間振り混ぜるとき、30分以内に溶解させるに要する水の量が30mL未満である化合物をいう。かかる水の量としては、10mL未満が好ましく、1mL未満がより好ましい。親水性化合物は水溶性を示すものが多く、脂質二重層の親水性層に含まれていたり、逆ナノディスクの親水性部分に吸着して安定化している可能性がある。また、金属酸化物など、親水性ではあるが水に対して不溶性である有効成分を逆ナノディスクの親水性部分に吸着させることにより安定化させることも可能になり得る。
親水性化合物としては、例えば、酸化チタン粒子や酸化亜鉛粒子などの紫外線散乱剤;ベンガラ、酸化チタン、酸化鉄などの無機顔料;アスコルビン酸やその誘導体などの水溶性ビタミン類;ローカストビーンガム、グァーガム誘導体、カラギーナン、ペクチン、キサンタンガム、ジェランガム、アルギン酸などの増粘剤を挙げることができる。
親油性化合物は、親油性基を有し、水に難溶または不溶で且つ油に可溶である化合物をいう。親油性化合物は、例えば、1gまたは1mLを水中に入れ、20±5℃で5分ごとに強く30秒間振り混ぜるとき、30分以内に溶解させるに要する水の量が30mL以上である化合物をいう。かかる水の量としては、100mL以上が好ましく、1000mL以上または10000mL以上がより好ましい。本発明で用いる親油性化合物は、化粧料や医薬組成物などに配合する有効成分であれば特に制限されないが、例えば、美白剤、抗老化剤、抗酸化剤、保湿剤、育毛剤、細胞賦活剤、ビタミン類、アミノ酸類などが挙げられる。より具体的には、例えば、トコフェロール、コエンザイムQ10、還元型コエンザイムQ10、およびそれらの誘導体などの脂溶性ビタミン類;スクワランなどのオイル;ヒドロコルチゾン、プレドニゾロン、デキサメタゾン、ベタメタゾンなどのステロイド系抗炎症薬;アセチルサリチル酸、イブプロフェン、インドメタシン、ロキソプロフェンなどの非ステロイド系抗炎症薬;抗生物質;抗真菌剤;メチルパラベンなどの防腐剤;メントールなどの香料;アリザニンなどの色素を挙げることができる。その他、アルブチン及びその誘導体;L-アスコルビン酸およびその誘導体;ハイドロキノン及びその誘導体;グルタチオン及びその誘導体;胎盤抽出物;パントテン酸およびその誘導体;トラネキサム酸およびその誘導体;コウジ酸およびその誘導体;システイン及びその誘導体;カミツレエキス、カンゾウ抽出物などの植物抽出物;ベータカロチン等のカロテノイド類;アスタキサンチン及びその誘導体;フラボノイド類;カテキン及びその誘導体;ビタミンA及びその誘導体;α-リポ酸およびその誘導体;グリチルリチン酸およびその誘導体;チオタウリン及びその誘導体;尿素およびその誘導体;グリセリン誘導体;サリチル酸およびその誘導体;ニコチン酸およびその誘導体;L-アミノ酸およびその誘導体;アデノシン及びその誘導体;イソフラボン等の植物性女性ホルモン様成分;グラブリジン及びその誘導体;マグノリグナン及びその誘導体;ミノキシジル及びその誘導体;フィナステリド及びその誘導体;フラバノン類;塩化カルプロニウム及びその誘導体;フェルラ酸およびその誘導体;フラーレン及びその誘導体;ラクトフェリン;D-アミノ酸およびその誘導体;オリゴ糖およびその誘導体;核酸およびその誘導体;D-グルコサミン及びその誘導体;コンドロイチン硫酸ナトリウム及びその誘導体;ジプロピレングリコール及びその誘導体;セラミド及びその誘導体;ソルビトール及びその誘導体;トレハロース及びその誘導体;ヒアルロン酸およびその誘導体;プロピレングリコール脂肪酸エステル及びその誘導体;マルチトール及びその誘導体;ラフィノース及びその誘導体;アルギン酸およびその誘導体;カフェイン及びその誘導体;カルボキシビニルポリマー及びその誘導体;キサンタンガム及びその誘導体;ヒドロキシエチルセルロース及びその誘導体;ポリビニルピロリドン及びその誘導体;ケラチン分解物およびその誘導体;シルクタンパク質分解物およびその誘導体;γ-アミノ酪酸およびその誘導体;アラントイン及びその誘導体;γ-オリザノール及びその誘導体;L-カルニチン及びその誘導体;β-1,3-グルカン及びその誘導体;ビオチン及びその誘導体などが挙げられる。
本発明に係る逆ナノディスクは、例えば、非イオン界面活性剤の水系溶媒溶液と親油性溶媒の混合物を撹拌することにより逆ベシクル分散液を得る工程、および、前記逆ベシクル分散液にポリペプチドバイオサーファクタントを添加することにより逆ナノディスクを得る工程を含む方法により製造することができる。なお、本発明に係る逆ナノディスクは、溶媒として用い得る成分を含む各成分を混合し、穏やかな撹拌のみで或いは撹拌することなく製造できる可能性もある。
水系溶媒の使用量は、常温常圧で非イオン界面活性剤を溶解できる範囲で適宜調整すればよいが、例えば、非イオン界面活性剤1質量部に対する水系溶媒の質量比を1以上、4以下とすることができる。当該比が1以上であれば、非イオン界面活性剤をより確実に溶解することができ、当該比が4以下であれば、逆ベシクルの形成を阻害しないと考えられる。上記比としては、1.5以上が好ましく、また、3以下が好ましく、2.5以下がより好ましい。
非イオン界面活性剤と水系溶媒との混合物には、非イオン界面活性剤の溶解のためや、逆ベシクルの形成を促進するために、一価C4-20アルコールを添加してもよい。一価C4-20アルコールの添加量は、上記効果が発揮される範囲で適宜調整すればよいが、例えば、非イオン界面活性剤1質量部に対する一価C4-20アルコールの質量比を0.5以上、2以下とすることができる。当該比が当該範囲内であれば、上記効果がより確実に廃棄される。上記比としては、0.8以上が好ましく、また、1.5以下が好ましく、1.2以下がより好ましい。
非イオン界面活性剤と水系溶媒、更に好適には一価C4-20アルコールを含む混合物を混合し、非イオン界面活性剤を溶解させる。次に、親油性溶媒を加え、逆ベシクルが形成されるよう激しく撹拌する。親油性溶媒の使用量は、逆ベシクルが形成される範囲で適宜調整すればよいが、例えば、非イオン界面活性剤1質量部に対する親油性溶媒の質量比を2以上、10以下とすることができる。当該比が2以上であれば、逆ベシクルがより確実に形成され、当該比が10以下であれば、次工程における逆ナノディスクの形成が阻害されないと考えられる。上記比としては、3以上または4以上が好ましく、5以上がより好ましく、また、8以下が好ましく、7以下がより好ましい。
非イオン界面活性剤溶液と親油性溶媒の混合物は、逆ベシクルが形成される程度に激しく撹拌することが好ましい。撹拌強度は、混合物の量などに応じて逆ベシクルが形成される範囲で適宜調整すればよいが、例えば、200rpm以上、10000rpm以下の回転数で、30秒間以上、30分間以下撹拌すればよい。撹拌手段としては、小規模であればボルテックスミキサー等を用いることができ、大規模であれば、撹拌翼などを備えたホモジナイザーやディゾルバー等を用いることができる。
次に、上記で得られた逆ベシクル分散液にポリペプチドバイオサーファクタントを添加することにより逆ナノディスクを得る。一般的なベシクルは脂質二重層で構成される20~400μm程度の球状粒子であり、その分散液は白濁している。本発明においても上記逆ベシクル分散液が白濁している場合であっても、ポリペプチドバイオサーファクタントの添加によってより微細な逆ナノディスクが形成されると、少なくとも肉眼では透明に見える傾向がある。
具体的には、逆ベシクル分散液にポリペプチドバイオサーファクタントを少量加えて撹拌するか、或いは撹拌した逆ベシクル分散液にポリペプチドバイオサーファクタントを連続的または断続的に加えていってもよい。
リポペプチドバイオサーファクタントの使用量は、逆ナノディスクが形成される範囲で適宜調整すればよい。例えば、非イオン界面活性剤1質量部に対するリポペプチドバイオサーファクタントの質量比を0.001以上、0.5以下とすることができる。当該比が0.001以上であれば、逆ナノディスクがより確実に形成され、当該比が0.5以下であれば、逆ナノディスクの形成に関与しないリポペプチドバイオサーファクタントの量がそれほど過剰にならず、経済的である。上記比としては、0.005以上が好ましく、0.01以上がより好ましく、また、0.5以下または0.1以下が好ましく、0.05以下がより好ましく、0.02以下がより更に好ましい。
非イオン界面活性剤、水系溶媒、一価C4-20アルコール、親油性溶媒、およびリポペプチドバイオサーファクタント以外の成分は、適時添加すればよい。例えば親水性化合物は、水系溶媒に適時添加すればよい。親油性化合物は、親油性溶媒と共存させることが好ましい。親油性化合物および親水性化合物の量は、それぞれ非イオン界面活性剤1質量部に対して0.001質量部以上、1質量部以下が好ましく、0.005質量部以上、0.5質量部以下がより好ましく、0.01質量部以上、0.2質量部以下がより更に好ましい。
逆ナノディスクは、凍結乾燥した後、再び親油性溶媒に分散させてもよい。再溶解液の逆ナノディスク濃度は、任意に調整することができる。
本発明に係る逆ナノディスクは、特に親水性化合物や親油性化合物をその内部に取り込み、親油性溶媒中に良好に分散させることができる上に、非常に微細であることから、その分散液は目視上十分に透明に見え、また、皮膚組織などへの浸透性も高いと考えられる。なお、特に化粧品業界においては、一般的に、分散質が目視で確認できなくなり、分散液が目視上透明になった状態を「可溶化」という場合がある。よって本発明に係る逆ナノディスクは、例えばO/Wエマルション製剤である化粧料などのパーソナルケア組成物や医薬組成物の成分として、非常に有効であるといえる。

以下、実施例を挙げて本発明をより具体的に説明するが、本発明はもとより下記実施例によって制限を受けるものではなく、前・後記の趣旨に適合し得る範囲で適当に変更を加えて実施することも勿論可能であり、それらはいずれも本発明の技術的範囲に包含される。
実施例1
(1)逆ベシクル分散液の調製
水と1-ヘキサノールを水:1-ヘキサノール=20:10の質量比で試験管に量り取り、1-ヘキサノールと同量の非イオン界面活性剤(「DKエステルSS」第一工業製薬社製,ショ糖脂肪酸エステル)を加えて溶液を得た。更に、非イオン界面活性剤に対して6質量倍のn-デカンを加え、ボルテックスミキサーを使って1分間撹拌することで、白濁液を得た。当該白濁液中の各成分の質量比は、n-デカン:水:非イオン界面活性剤:1-ヘキサノール=60:20:10:10である。
得られた白濁液を位相差顕微鏡(「BX41」OLYMPUS社製)で観察したところ、図1(a)に示す通り、ドーナツ状の分子集合体が観察された。
また、同白濁液を直交ニコル下で観察したところ、図1(b)に示す通りマルターゼクロスが確認されたことから、逆ベシクルが形成されていることが明らかとなった。
(1)逆ベシクル分散液の調製
水と1-ヘキサノールを水:1-ヘキサノール=20:10の質量比で試験管に量り取り、1-ヘキサノールと同量の非イオン界面活性剤(「DKエステルSS」第一工業製薬社製,ショ糖脂肪酸エステル)を加えて溶液を得た。更に、非イオン界面活性剤に対して6質量倍のn-デカンを加え、ボルテックスミキサーを使って1分間撹拌することで、白濁液を得た。当該白濁液中の各成分の質量比は、n-デカン:水:非イオン界面活性剤:1-ヘキサノール=60:20:10:10である。
得られた白濁液を位相差顕微鏡(「BX41」OLYMPUS社製)で観察したところ、図1(a)に示す通り、ドーナツ状の分子集合体が観察された。
また、同白濁液を直交ニコル下で観察したところ、図1(b)に示す通りマルターゼクロスが確認されたことから、逆ベシクルが形成されていることが明らかとなった。
(2)逆ナノディスク分散液の調製
上記(1)で得られた逆ベシクル分散液(5mL)にサーファクチンナトリウム(SFNa)を徐々に添加していったところ、10.8mg添加したところで図2に示す通り透明になった。この際の非イオン界面活性剤の濃度は140mM、サーファクチンナトリウムの濃度は2mMであった。
動的光散乱装置(「DLS-7000」大塚電子社製)を用い、得られた透明溶液に含まれている粒子の粒度分布を測定した。その際、光源にはλ=488nmのArレーザーを用い、散乱角度は90度に設定した。その結果、図3に示すように、流体力学的直径が13.7±2.1nmの微細なナノ粒子が計測された。ベシクル構造体の最小粒子径はおよそ20nmとされていることから、ベシクルではなく、逆構造のナノディスクが形成したものと考えられる。
また、同溶液をネガティブ染色し、透過型電子顕微鏡(「H-7650」日立ハイテクノロジーズ社製)で観察した。ネガティブ染色には、1%リンタングステン酸ナトリウム水溶液を用い、加速電圧は100kVに設定し、200メッシュでカーボン補強処理したグリッドを用いた。リンタングステン酸ナトリウム水溶液は、リンタングステン酸ナトリウムを50mg秤量し、超純水を5mL加え、遮光下で5分間撹拌した上で、pHを5M水酸化ナトリウム水溶液により6.8に調整して調製し、0.1μmまたは0.22μmPVDFシリンジフィルターにより濾過してから使用した。結果を図4に示す。
図4の透過型電子顕微鏡写真の通り、多数の均一なナノ粒子が観測され、画像解析の結果から、図3の動的光散乱測定とほぼ同程度の、13.1±1.7nmの粒子径分布が極めて狭いナノ粒子が得られていることが判明した。
以上の結果より、いったん形成された逆ベシクルを構成する逆脂質二重層の側面にリポペプチドバイオサーファクタントであるサーファクチンナトリウムが結合して微細な逆ナノディスクが形成されたと考えられる。かかる機構を、図5に模式的に示す。
上記(1)で得られた逆ベシクル分散液(5mL)にサーファクチンナトリウム(SFNa)を徐々に添加していったところ、10.8mg添加したところで図2に示す通り透明になった。この際の非イオン界面活性剤の濃度は140mM、サーファクチンナトリウムの濃度は2mMであった。

前記非イオン界面活性剤が、その親油性部分を外側に、その親水性部分を内側にして脂質二重層を形成し、

- 前記脂質二重層が親油性化合物を含む請求項1~3のいずれかに記載の逆ナノディスク。
- 体積平均粒子径が5nm以上、20nm未満である請求項1~4のいずれかに記載の逆ナノディスク。
- 前記リポペプチドバイオサーファクタントがサーファクチンまたはその塩である請求項1~5のいずれかに記載の逆ナノディスク。
- 親油性溶媒中に請求項1~6のいずれかに記載の逆ナノディスクが分散していることを特徴とする親油性分散液。
- 逆ナノディスクを製造するための方法であって、
非イオン界面活性剤の水系溶媒溶液と親油性溶媒の混合物を撹拌することにより逆ベシクル分散液を得る工程、および、
前記逆ベシクル分散液にポリペプチドバイオサーファクタントを添加することにより逆ナノディスクを得る工程を含むことを特徴とする方法。 - 前記非イオン界面活性剤1質量部に対して1質量部以上、4質量部以下の前記水系溶媒溶液を用いる請求項8に記載の方法。
- 前記水系溶媒溶液が、更に一価C4-20アルコールを含む請求項8または9に記載の方法。
- 前記リポペプチドバイオサーファクタントがサーファクチンまたはその塩である請求項8~10のいずれかに記載の方法。
- 前記非イオン界面活性剤1質量部に対して0.001質量部以上、0.5質量部以下の前記リポペプチドバイオサーファクタントを用いる請求項8~11のいずれかに記載の方法。
