Abstract
オキシブチニン又はその薬学的に許容される塩、水及び低級アルコールを含有する、多汗症治療用の外用液剤であって、外用液剤中のオキシブチニン又はその薬学的に許容される塩の含有量が15質量%~25質量%であり、1日1回投与するように用いられ、1回のオキシブチニン又はその薬学的に許容される塩の投与量が60mg~150mgである外用液剤を開示する。
Description
本発明は、多汗症治療用の外用液剤に関する。より詳細には、オキシブチニン又はその薬学的に許容される塩を含有する、多汗症治療用の外用液剤に関する。
多汗症の治療方法として、オキシブチニンなどの抗コリン作用薬を含む外用組成物を投与する方法が提案されており、薬物の経口投与による全身性副作用を回避することが期待されている(特許文献1及び2)。また、多汗症治療用の外用剤としては、水、抗コリン作用薬及び所定の塩を含有する外用剤(特許文献3)、水、抗コリン作用薬及びジカルボン酸エステルを含有する外用剤(特許文献4)などが知られている。
多汗症治療用のオキシブチニン外用液剤は上市された製品が未だなく、そのため、外用液剤における望ましい用法用量も判明していない。
上記課題を解決するため、本発明者らは鋭意検討を行ったところ、十分な発汗抑制効果を奏する外用液剤を見出し、本発明を完成するに至った。
本発明の一側面に係る多汗症治療用の外用液剤は、オキシブチニン又はその薬学的に許容される塩、水及び低級アルコールを含有し、外用液剤中のオキシブチニン又はその薬学的に許容される塩の含有量は15質量%~25質量%である。外用液剤は1日1回投与するように用いられ、1回のオキシブチニン又はその薬学的に許容される塩の投与量が60mg~150mgである。
本発明の別の一側面に係る多汗症治療用の外用液剤は、オキシブチニン又はその薬学的に許容される塩、水及び低級アルコールを含有し、外用液剤中のオキシブチニン又はその薬学的に許容される塩の含有量は15質量%~25質量%である。外用液剤は1日1回投与するように用いられ、1回のオキシブチニン又はその薬学的に許容される塩の投与量が90mg~100mgである。外用液剤は、以下の(a)~(i)の少なくとも一つの薬物動態パラメータを示す:

本発明の多汗症治療用の外用液剤は、十分な発汗抑制効果を奏する。
(第1実施形態)
第1実施形態に係る外用液剤は、オキシブチニン又はその薬学的に許容される塩、水及び低級アルコールを含有する。
第1実施形態に係る外用液剤は、オキシブチニン又はその薬学的に許容される塩、水及び低級アルコールを含有する。
オキシブチニン又はその薬学的に許容される塩は、抗コリン作用を有する薬物であり、多汗症患者に投与することで発汗を抑制する作用を発揮する。オキシブチニンの薬学的に許容される塩として、オキシブチニン塩酸塩などが挙げられる。外用液剤中のオキシブチニン又はその薬学的に許容される塩の含有量は15質量%~25質量%(外用液剤の全質量を基準として。以下同じ。)であり、18質量%~22質量%であることが好ましい。
外用液剤中の水は、溶媒であり、オキシブチニン又はその薬学的に許容される塩及びその他の成分を溶解又は分散させるための媒体となる。外用液剤中の水の含有量は10質量%~40質量%とすることができ、20質量%~30質量%であることが好ましく、24質量%~28質量%であることがより好ましい。
外用液剤中の低級アルコールは、オキシブチニン又はその薬学的に許容される塩の溶解性及び分散性を高め、オキシブチニン又はその薬学的に許容される塩の皮膚への分配性を高める。低級アルコールとして、メタノール、エタノール及びイソプロパノールなどが挙げられ、好ましくはエタノールである。外用液剤中の低級アルコールの含有量は30質量%~50質量%とすることができ、36質量%~44質量%であることが好ましい。
外用液剤は、さらに、乳酸塩、酒石酸塩及びリン酸塩からなる群から選択される1以上の塩を含有することができる。かかる塩は、皮膚の付属器官(汗腺など)に対するオキシブチニン又はその薬学的に許容される塩の蓄積性が高まり、発汗抑制作用を高めることができる。上記塩は無水物でも水和物でもよい。乳酸はL体及びD体のいずれでもよく、これらの任意の混合物であってもよい。酒石酸はL体、D体及びメソ体のいずれでもよく、これらの任意の混合物であってもよい。上記塩は、例えば、ナトリウム、カリウム及びリチウムなどの一価の金属との塩、カルシウム及びマグネシウムなどの二価の金属との塩、アルミニウムなどの三価の金属との塩並びにアンモニア、エチレンジアミン、トリエチルアミン、ジエタノールアミン、トリエタノールアミン及びメグルミンなどのアミン化合物との塩などが挙げられる。皮膚の付属器官に対するオキシブチニン又はその薬学的に許容される塩の蓄積性を向上させる観点から、上記塩は、乳酸塩であることが好ましく、乳酸ナトリウムであることがより好ましい。外用液剤中の上記塩の含有量は、5質量%~10質量%とすることができ、5.4質量%~6.6質量%であることが好ましい。
外用液剤は、さらに、ジカルボン酸エステルを含有することができる。ジカルボン酸エステルは、外用液剤の粘性を低下させ、オキシブチニン又はその薬学的に許容される塩に由来する「べたつき」を抑制する働きをする。ジカルボン酸エステルとして、アジピン酸ジイソプロピル、セバシン酸ジエチル、セバシン酸ジイソプロピル、コハク酸ジメチル、アジピン酸ジブチル、アジピン酸ジイソブチル、アジピン酸ジオクチル、セバシン酸ジオクチル、フタル酸ジエチル、及びフタル酸ジブチルが挙げられ、好ましくはアジピン酸ジイソプロピルである。外用液剤中のジカルボン酸エステルの含有量は5質量%~10質量%とすることができ、7.2質量%~8.8質量%であることが好ましい。
外用液剤は、上記成分の他に、界面活性剤、保存安定剤、油脂、溶解剤、充填剤、保湿剤、pH調節剤、浸透圧調節剤、増粘剤、清涼剤、収斂薬及び血管収縮薬などを含んでいてもよい。
界面活性剤は、オキシブチニン又はその薬学的に許容される塩を水などの媒体に乳化させるために有用である。界面活性剤の具体例としては、非イオン性界面活性剤(ポリソルベート20、ポリソルベート80、ポリソルベート60、ポリオキシエチレン硬化ヒマシ油20、ポリオキシエチレン硬化ヒマシ油40及びポリオキシエチレン硬化ヒマシ油60など)、イオン性界面活性剤及び両性界面活性剤が挙げられる。界面活性剤の含有量は、外用液剤の全質量を基準として、例えば、0質量%~10質量%であってよい。
保存安定剤の具体例としては、パラベン、イソプロピルメチルフェノール、フェノキシエタノール及びチモールが挙げられる。
油脂及び溶解剤の具体例としては、脂肪酸及び脂肪族アルコールが挙げられる。
充填剤の具体例としては、無機粉体(タルク、モンモリロナイト、スメクタイト及びカオリンなど)及び有機粉体が挙げられる。
保湿剤の具体例としては、多価アルコール、糖類、尿素、ワセリン、及びパラフィンが挙げられる。
外用液剤のpHは4.5~7.5の範囲内とすることができる。pHの測定は、第17改正日本薬局方の一般試験法の「2.54 pH測定法」に従い、複合ガラス電極を用いて行う。
外用液剤は、ローション剤及びリニメント剤などの形態であってもよく、また、適切な容器(例えば、液剤を噴霧するためのスプレー容器、液剤を塗布するための容器及びエアゾール剤容器など)に収容された塗布剤及びスプレー剤などの形態であってもよい。好ましい剤形はローション剤である。
外用液剤は、上記各成分を混合し混和することで製造することができる。
第1実施形態に係る外用液剤は、ヒト成人に対し、1日1回投与する。投与部位は、発汗量の多い部位であることが好ましく、例えば掌である。投与するタイミングは限られないが、例えば、就寝前とすることができる。投与1回あたりの外用液剤に含まれる、オキシブチニン又はその薬学的に許容される塩の量は60mg~150mgであり、好ましくは70mg~120mgであり、さらに好ましくは80mg~110mgであり、特に好ましくは90mg~100mgである。投与1回あたりの外用液剤の体積は、400μL~600μLとすることができ、好ましくは450μL~550μLである。外用液剤の密度は、0.90~1.0g/cm3とすることができる。
第1実施形態に係る外用液剤を多汗症患者に1日1回、8週間、ヒト成人に反復投与した後のオキシブチニンの平均血漿中濃度は15ng/mL~25ng/mLであり、かつ、活性代謝物であるN-デスメチルオキシブチニンの平均血漿中濃度は4.5ng/mL~8ng/mLである。また、その場合のオキシブチニンの平均血漿中濃度/N-デスメチルオキシブチニンの平均血漿中濃度の比は、1.875~5.556である。かかる濃度範囲及び/又は比となることで、十分な発汗抑制作用を発揮する。
(第2実施形態)
第2実施形態に係る外用液剤は、オキシブチニン又はその薬学的に許容される塩、水及び低級アルコールを含有する。外用液剤に含まれる、オキシブチニン又はその薬学的に許容される塩、水、低級アルコール及びその他の成分、外用液剤のpH、外用液剤の剤形、並びに外用液剤の製造方法は、上述の第1実施形態に係る外用液剤に関する説明と同様である。
第2実施形態に係る外用液剤は、オキシブチニン又はその薬学的に許容される塩、水及び低級アルコールを含有する。外用液剤に含まれる、オキシブチニン又はその薬学的に許容される塩、水、低級アルコール及びその他の成分、外用液剤のpH、外用液剤の剤形、並びに外用液剤の製造方法は、上述の第1実施形態に係る外用液剤に関する説明と同様である。
第2実施形態に係る外用液剤は、ヒト成人に対し、1日1回投与する。投与部位は、発汗量の多い部位であることが好ましく、例えば掌である。投与するタイミングは限られないが、例えば、就寝前とすることができる。投与1回あたりの外用液剤に含まれる、オキシブチニン又はその薬学的に許容される塩の量は90mg~100mgである。投与1回あたりの外用液剤の体積は、400μL~600μLとすることができ、好ましくは450μL~550μLである。外用液剤の密度は、0.90~1.0g/cm3とすることができる。
第2実施形態に係る外用液剤をヒト成人に単回投与した場合、オキシブチニンの最大血漿中濃度は0.8ng/mL~40ng/mLであり、上記最大血漿中濃度に達するまでに要する時間は10時間~24時間であり、オキシブチニンの投与時から濃度測定最終時点までの血漿中薬物濃度-時間曲線下面積は9ng・h/mL~291ng・h/mLである。また、この場合、投与時から24時間後のオキシブチニンの血漿中濃度(C24)は0.7ng/mL~12ng/mLであることが好ましい。かかる薬物動態パラメータを示す外用液剤は、十分な発汗抑制作用を発揮する。
第2実施形態に係る外用液剤をヒト成人に単回投与した場合、オキシブチニンの活性代謝物であるN-デスエチルオキシブチニンの最大血漿中濃度とオキシブチニンの最大血漿中濃度との比(N-デスエチルオキシブチニンの最大血漿中濃度/オキシブチニンの最大血漿中濃度)は0.04~0.55であり、N-デスエチルオキシブチニンの投与時から濃度測定最終時点までの血漿中薬物濃度-時間曲線下面積とオキシブチニンの投与時から濃度測定最終時点までの血漿中薬物濃度-時間曲線下面積との比(N-デスエチルオキシブチニンの投与時から濃度測定最終時点までの血漿中薬物濃度-時間曲線下面積/オキシブチニンの投与時から濃度測定最終時点までの血漿中薬物濃度-時間曲線下面積)は0.06~0.31である。また、この場合、投与時から24時間後のN-デスエチルオキシブチニンの血漿中濃度(C24)とオキシブチニンの血漿中濃度(C24)との比(N-デスエチルオキシブチニンのC24/オキシブチニンのC24)は0.1~0.6であることが好ましい。かかる薬物動態パラメータを示す外用液剤は、十分な発汗抑制作用を発揮する。
第2実施形態に係る外用液剤をヒト成人に1日1回投与を繰り返して定常状態に達した場合、オキシブチニンの最大血漿中濃度(Cmax)は5ng/mL~46ng/mLであり、前記最大血漿中濃度に達するまでに要する時間(tmax)は1時間~22時間であり、オキシブチニンの投与間隔における血漿中薬物濃度-時間曲線下面積(AUC0-24)は、75ng・h/mL~610ng・h/mLであり、オキシブチニンの血漿中濃度の半減期(t1/2)は10時間~90時間である。また、この場合、投与時から24時間後のオキシブチニンの血漿中濃度(C24)は2ng/mL~44ng/mLであることが好ましく、オキシブチニンの最小血漿中濃度(Cmin)は2ng/mL~17ng/mLであることが好ましく、オキシブチニンの投与間隔における血漿中薬物濃度-時間曲線下面積(AUC0-24)から算出される静脈内投与以外の投与経路における見かけのクリアランス(CL/F)は140L/h~1150L/hであることが好ましい。かかる薬物動態パラメータを示す外用液剤は、十分な発汗抑制作用を発揮する。
第2実施形態に係る外用液剤をヒト成人に1日1回投与を繰り返して定常状態に達した場合、N-デスエチルオキシブチニンの最大血漿中濃度(Cmax)とオキシブチニンの最大血漿中濃度(Cmax)との比(N-デスエチルオキシブチニンのCmax/オキシブチニンのCmax)は、0.08~0.46であり、N-デスエチルオキシブチニンの投与間隔における血漿中薬物濃度-時間曲線下面積(AUC0-24)とオキシブチニンの投与間隔における血漿中薬物濃度-時間曲線下面積(AUC0-24)との比(N-デスエチルオキシブチニンの投与間隔におけるAUC0-24/オキシブチニンの投与間隔におけるAUC0-24)は、0.15~0.65である。また、この場合、投与時から24時間後のN-デスエチルオキシブチニンの血漿中濃度(C24)とオキシブチニンの血漿中濃度(C24)との比(N-デスエチルオキシブチニンのC24/オキシブチニンのC24)は、0.18~1.5であることが好ましい。かかる薬物動態パラメータを示す外用液剤は、十分な発汗抑制作用を発揮する。
また、第2実施形態に係る外用液剤をヒト成人に1日1回投与を繰り返した場合、定常状態でのオキシブチニンの最大血漿中濃度(Cmax)と初回投与時のオキシブチニンの最大血漿中濃度(Cmax)との比(定常状態でのCmax/初回投与時のCmax)は、0.5~13.5であることが好ましく、定常状態でのオキシブチニンの投与間隔における血漿中薬物濃度-時間曲線下面積(AUC0-24)と初回投与時のオキシブチニンの投与時から24時間後までの血漿中薬物濃度-時間曲線下面積(AUC0-24)との比(定常状態でのAUC0-24/初回投与時のAUC0-24)は、1~32であることが好ましく、定常状態でのN-デスエチルオキシブチニンの最大血漿中濃度(Cmax)と初回投与時のN-デスエチルオキシブチニンの最大血漿中濃度(Cmax)との比(定常状態でのCmax/初回投与時のCmax)は、1.4~17であることが好ましく、定常状態でのN-デスエチルオキシブチニンの投与間隔における血漿中薬物濃度-時間曲線下面積(AUC0-24)と初回投与時のN-デスエチルオキシブチニンの投与時から24時間後までの血漿中薬物濃度-時間曲線下面積(AUC0-24)との比(定常状態でのAUC0-24/初回投与時のAUC0-24)は、2~45であることが好ましい。かかる薬物動態パラメータを示す外用液剤は、十分な発汗抑制作用を発揮する。
本発明の別の好ましい実施形態は以下のとおりである。

1.外用液剤の製造
表1に示す成分を均一に混合することで外用液剤1~3(ローション剤)を製造し、容器に充填した。得られた外用液剤の密度は、外用液剤1が963mg/cm3、外用液剤2が945mg/cm3、外用液剤3が931mg/cm3であった。

2.外用液剤の投与
多汗症患者182人を3つの群にわけ、外用液剤1~3を500μL(それぞれ、96mg、24mg及び0mgのオキシブチニン塩酸塩を含む)を患者の両手掌部に1日1回就寝前に塗布した。外用液剤の投与は8週間繰り返した。

3.発汗量の測定
投与前、投与開始4週間及び8週間時点において、患者の掌の発汗量を発汗量測定装置(換気カプセル型発汗計SKN-2000M;株式会社西澤電気計器製作所製)を用いて測定した。
(発汗量の変化量)=(外用液剤投与後の発汗量)-(外用液剤投与前の発汗量)として、各群の発汗量の変化量を算出した。発汗量の変化量の結果を図1に示す。統計解析(制限付き最尤法に基づく線形混合効果モデルによる反復測定データ解析法(Mixed-effects Model Repeated Measures;MMRM))の結果、外用液剤3投与群と比較して、外用液剤1投与群は、投与開始4週間(p=0.0015)及び8週間(p=0.0046)時点において、有意に発汗量が低下していた。

投与開始2週間、4週間、6週間及び8週間時点において、患者から採血し、液体クロマトグラフィータンデム質量分析システム(LC-MS/MS)を用いて、採血した各サンプルを分析し、オキシブチニン及びN-デスエチルオキシブチニンの平均血漿中濃度を測定した。使用したLC-MS/MS装置は、高速液体クロマトグラフProminence UFLCシステム(株式会社島津製作所)及び質量分析計QTRAP5500(株式会社エービー・サイエックス)、又は、高速液体クロマトグラフNexera UHPLCシステム(株式会社島津製作所)及び質量分析計Triple Quad 5500(株式会社エービー・サイエックス)であった。
投与開始2週間、4週間、6週間及び8週間時点において、患者から採血し、液体クロマトグラフィータンデム質量分析システム(LC-MS/MS)を用いて、採血した各サンプルを分析し、オキシブチニン及びN-デスエチルオキシブチニンの平均血漿中濃度を測定した。使用したLC-MS/MS装置は、高速液体クロマトグラフProminence UFLCシステム(株式会社島津製作所)及び質量分析計QTRAP5500(株式会社エービー・サイエックス)、又は、高速液体クロマトグラフNexera UHPLCシステム(株式会社島津製作所)及び質量分析計Triple Quad 5500(株式会社エービー・サイエックス)であった。
外用液剤1を投与した患者の平均血漿中濃度(ng/mL)は以下のとおりであった。
外用液剤2を投与した患者の平均血漿中濃度(ng/mL)は以下のとおりであった。
6.外用液剤の薬物動態試験
健康成人男性18人を対象に、外用液剤1を500μL両手掌部に1日1回8時間、14日間塗布した。外用液剤1の投与直前及び投与開始から432時間後まで採血し、液体クロマトグラフィータンデム質量分析システム(LC-MS/MS)を用いて、採血した各サンプルを分析し、オキシブチニン及びN-デスエチルオキシブチニンの血漿中濃度を測定した。オキシブチニンの血漿濃度推移の結果を図3に示す。使用したLC-MS/MS装置は、上記5の測定で用いた装置と同じである。
健康成人男性18人を対象に、外用液剤1を500μL両手掌部に1日1回8時間、14日間塗布した。外用液剤1の投与直前及び投与開始から432時間後まで採血し、液体クロマトグラフィータンデム質量分析システム(LC-MS/MS)を用いて、採血した各サンプルを分析し、オキシブチニン及びN-デスエチルオキシブチニンの血漿中濃度を測定した。オキシブチニンの血漿濃度推移の結果を図3に示す。使用したLC-MS/MS装置は、上記5の測定で用いた装置と同じである。
測定した血漿中濃度より、外用液剤1の以下の薬物動態パラメータを算出した。オキシブチニンについての結果を表4に、N-デスエチルオキシブチニンについての結果を表5に、それぞれ示す。
C24:各時点から24時間後の血漿中濃度
Cmax:各時点から24時間の最大血漿中濃度
tmax:上記最大血漿中濃度に達するまでに要する時間
AUC0-24:各時点から24時間後までの血漿中薬物濃度-時間曲線下面積
Cmin:各時点から24時間の最小血漿中濃度
Cav:各時点から24時間の平均血漿中濃度
t1/2:14日目に塗布した外用液剤の除去(塗布後8時間時点)後(「最終除去後」ともいう)、薬物濃度が半分になるまでに要する時間(半減期)
CL/F:AUC0-24から算出した静脈内投与以外の投与経路における見かけのクリアランス
C24:各時点から24時間後の血漿中濃度
Cmax:各時点から24時間の最大血漿中濃度
tmax:上記最大血漿中濃度に達するまでに要する時間
AUC0-24:各時点から24時間後までの血漿中薬物濃度-時間曲線下面積
Cmin:各時点から24時間の最小血漿中濃度
Cav:各時点から24時間の平均血漿中濃度
t1/2:14日目に塗布した外用液剤の除去(塗布後8時間時点)後(「最終除去後」ともいう)、薬物濃度が半分になるまでに要する時間(半減期)
CL/F:AUC0-24から算出した静脈内投与以外の投与経路における見かけのクリアランス
上記薬学的パラメータ(C24、Cmax及びAUC0-24)の分子量補正したN-デスエチルオキシブチニン/オキシブチニン比を表6に示す。
血漿中オキシブチニン濃度及びN-デスエチルオキシブチニン濃度の累積係数(Robs:投与10回目及び投与14回目のCmax又はAUC0-24/投与1回目のCmax又はAUC0-24)を算出した。オキシブチニンについての結果を表7に、N-デスエチルオキシブチニンについての結果を表8に、それぞれ示す。
表7のとおり、血漿中オキシブチニン濃度の累積係数(平均値)は、Cmaxの投与10回目及び投与14回目では3.555及び3.628、AUC0-24の投与10回目及び投与14回目では5.528及び6.444であった。また、表6のとおり、血漿中N-デスエチルオキシブチニン濃度の累積係数(平均値)は、Cmaxの投与10回目及び投与14回目では3.434及び4.113、AUC0-24の投与10回目及び投与14回目では9.000及び10.752であった。
血漿中オキシブチニン濃度の累積係数は、いずれの薬物動態パラメータにおいても投与10回目と14回目では同程度であり、定常状態に達していると考えられた。また、血漿中N-デスエチルオキシブチニン濃度の累積係数についても同様に、いずれの薬物動態パラメータにおいても投与10回目と14回目では同程度であり、定常状態に達していると考えられた。
Claims (27)
- オキシブチニン又はその薬学的に許容される塩、水及び低級アルコールを含有する、多汗症治療用の外用液剤であって、外用液剤中のオキシブチニン又はその薬学的に許容される塩の含有量が15質量%~25質量%であり、1日1回投与するように用いられ、1回のオキシブチニン又はその薬学的に許容される塩の投与量が60mg~150mgである、外用液剤。
- さらに、乳酸塩、酒石酸塩及びリン酸塩からなる群から選択される1以上の塩を5質量%~10質量%含有する、請求項1に記載の外用液剤。
- 塩が乳酸ナトリウムである、請求項2に記載の外用液剤。
- さらに、ジカルボン酸エステルを5質量%~10質量%含有する、請求項1~3のいずれか一項に記載の外用液剤。
- ジカルボン酸エステルが、アジピン酸ジイソプロピルである、請求項4に記載の外用液剤。
- 低級アルコールがエタノールである、請求項1~5のいずれか一項に記載の外用液剤。
- 外用液剤中の低級アルコールの含有量が30質量%~50質量%である、請求項1~6のいずれか一項に記載の外用液剤。
- オキシブチニン又はその薬学的に許容される塩がオキシブチニン塩酸塩である、請求項1~7のいずれか一項に記載の外用液剤。
- ローションの剤形である、請求項1~8のいずれか一項に記載の外用液剤。
- 18質量%~22質量%のオキシブチニン塩酸塩、24質量%~28質量%の水、36質量%~44質量%のエタノール、5.4質量%~6.6質量%の乳酸ナトリウム、及び7.2質量%~8.8質量%のアジピン酸ジイソプロピルを含有する多汗症治療用のローション剤であって、1日1回投与するように用いられ、1回のオキシブチニン塩酸塩の投与量が90mg~100mgである、ローション剤。
- オキシブチニン又はその薬学的に許容される塩、水及び低級アルコールを含有する、多汗症治療用の外用液剤であって、外用液剤中のオキシブチニン又はその薬学的に許容される塩の含有量が15質量%~25質量%であり、1日1回投与するように用いられ、1回のオキシブチニン又はその薬学的に許容される塩の投与量が90mg~100mgであり、
以下の(a)~(i)の少なくとも一つの薬物動態パラメータ:
(a)外用液剤の単回投与時のオキシブチニンの最大血漿中濃度が0.8ng/mL~40ng/mLであり、前記最大血漿中濃度に達するまでに要する時間が10時間~24時間である、
(b)外用液剤の単回投与時のオキシブチニンの投与時から濃度測定最終時点までの血漿中薬物濃度-時間曲線下面積が9ng・h/mL~291ng・h/mLである、
(c)1日1回の外用液剤の投与を繰り返して定常状態に達した場合のオキシブチニンの最大血漿中濃度が5ng/mL~46ng/mLであり、前記最大血漿中濃度に達するまでに要する時間が1時間~22時間である、
(d)1日1回の外用液剤の投与を繰り返して定常状態に達した場合のオキシブチニンの投与間隔における血漿中薬物濃度-時間曲線下面積が、75ng・h/mL~610ng・h/mLである、
(e)1日1回の外用液剤の投与を繰り返して定常状態に達した場合のオキシブチニンの血漿中濃度の半減期が10時間~90時間である、
(f)外用液剤の単回投与時のN-デスエチルオキシブチニンの最大血漿中濃度とオキシブチニンの最大血漿中濃度との比(N-デスエチルオキシブチニンの最大血漿中濃度/オキシブチニンの最大血漿中濃度)が0.04~0.55である、
(g)外用液剤の単回投与時のN-デスエチルオキシブチニンの投与時から濃度測定最終時点までの血漿中薬物濃度-時間曲線下面積とオキシブチニンの投与時から濃度測定最終時点までの血漿中薬物濃度-時間曲線下面積との比(N-デスエチルオキシブチニンの投与時から濃度測定最終時点までの血漿中薬物濃度-時間曲線下面積/オキシブチニンの投与時から濃度測定最終時点までの血漿中薬物濃度-時間曲線下面積)が0.06~0.31である、
(h)1日1回の外用液剤の投与を繰り返して定常状態に達した場合のN-デスエチルオキシブチニンの最大血漿中濃度とオキシブチニンの最大血漿中濃度との比(N-デスエチルオキシブチニンの最大血漿中濃度/オキシブチニンの最大血漿中濃度)が、0.08~0.46である、
(i)1日1回の外用液剤の投与を繰り返して定常状態に達した場合のN-デスエチルオキシブチニンの投与間隔における血漿中薬物濃度-時間曲線下面積とオキシブチニンの投与間隔における血漿中薬物濃度-時間曲線下面積との比(N-デスエチルオキシブチニンの投与間隔における血漿中薬物濃度-時間曲線下面積/オキシブチニンの投与間隔における血漿中薬物濃度-時間曲線下面積)が、0.15~0.65である、
を示す、外用液剤。 - (a)の薬物動態パラメータを示す、請求項11に記載の外用液剤。
- (b)の薬物動態パラメータを示す、請求項11に記載の外用液剤。
- (c)の薬物動態パラメータを示す、請求項11に記載の外用液剤。

- さらに、乳酸塩、酒石酸塩及びリン酸塩からなる群から選択される1以上の塩を5質量%~10質量%含有する、請求項11~21のいずれか一項に記載の外用液剤。
- 塩が乳酸ナトリウムである、請求項22に記載の外用液剤。
- さらに、ジカルボン酸エステルを5質量%~10質量%含有する、請求項11~23のいずれか一項に記載の外用液剤。
- 外用液剤中の低級アルコールの含有量が30質量%~50質量%である、請求項11~24のいずれか一項に記載の外用液剤。
- オキシブチニン又はその薬学的に許容される塩がオキシブチニン塩酸塩である、請求項11~25のいずれか一項に記載の外用液剤。
- ローションの剤形である、請求項11~26のいずれか一項に記載の外用液剤。
