코로나 바이러스의 감염증의 개선 또는 치료용 약학 조성물에 대한 기술로는 한국등록특허 제1913789호에 코로나 바이러스 감염으로 인한 질환 치료용 화합물이 개시되어 있고, 미국공개특허 제2006-0257852호에 중증급성호흡기증후군 코로나바이러스에 대한 기술이 개시되어 있다.
그러나, 아직까지 유효한 치료 효과를 나타내는 치료제 기술은 개발되지 않았다.
한편, 마크로락틴(Macrolactin)은 1989년 Gustafson에 의하여 미분류된 해양미생물에서 세계최초로 분리되어 발표되었다. Macrolactin A~F까지 총 6종류의 Macrolactin류를 분리규명하였고, 추가로 Macrolactinic acid와 isomacrolactinic acid를 분리하여 보고하는 등 Macrolactin 류로 20여종을 보고하였다.
Decursinol은 천연약제에 존재하는 물질이며 특히 한국산 토종당귀인 참당귀에 함유되어 있으며 피라노쿠마린 계열의 데커신, 데커시놀안젤레이트, 데커신놀이 건조중량의 약 10%를 차지하고 있으며, 이외의 함유 성분으로는 노다케닌 (nodakenin), 엄벨리페론(umbelliferon), β-시토스테롤(β-sitosterol), α-피넨(α-pinene), 리모넨(limonene) 등이 알려져 있다. 그러나, 참당귀추출물을 약학적 제제로 사용하는데에 제약이 따른다. 데커신, 데커시놀 안젤레이트를 주성분으로 포함하는 참당귀추출물은 정제하기가 까다로워서 50%이상의 정제를 하기는 대단히 어렵다. 순도가 낮을 경우에는 고혈압을 유발하는 심각한 부작용을 동반한다. 데커시놀(decursinol)은 데커신, 데커시놀 안젤레이트와 함께 당귀의 생리활성 성분 중의 하나로, 해열 진통효과, 패혈증쇼크 방지, 위점막보호작용, 아토피피부염 예방, 암세포 억제, 신경독성에 대한 보호 등의 효능이 있는 것으로 밝혀졌다. 데커시놀은 당귀 뿌리 내에서 극히 소량 존재하나 참당귀의 주성분인 데커신(decursin)이나 데커시놀 안젤레이트(decursinol angelate)를 염기로 가수분해하여 대량으로 높은 순도로 얻을 수 있다.
본 발명의 하나의 목적은 마크로락틴 유도체, 또는 이의 약학적으로 허용 가능한 염을 포함하는 제2형 중증급성호흡기증후군 코로나바이러스 감염 질환 예방 또는 치료용 약학적 조성물을 제공하는 것이다.
본 발명의 다른 하나의 목적은 마크로락틴 유도체, 또는 이의 약학적으로 허용가능한 염을 포함하는 제2형 중증급성호흡기증후군 코로나바이러스 감염 질환 예방 또는 개선용 식품 조성물을 제공하는 것이다.

이하, 본 발명을 상세히 설명한다.
본 명세서에서 어떤 부분이 어떤 구성요소를 "포함" 한다고 할 때, 이는 특별히 반대되는 기재가 없는 한 다른 구성요소를 제외하는 것이 아니라 다른 구성 요소를 더 포함할 수 있는 것을 의미한다.
본 발명에서 항 제2형 중증급성호흡기증후군 코로나바이러스는 제2형 중증급성호흡기증후군 코로나바이러스 감염 질환, 즉, 제2형 중증급성호흡기증후군 코로나바이러스 감염 또는 이로 인해 유발된 질환의 예방 치료 또는 개선을 의미할 수 있다. 또한, 항 제2형 중증급성호흡기증후군 코로나바이러스는 제2형 중증급성호흡기증후군 코로나바이러스의 대상체 내 증식의 억제, 사멸 등을 포함하여 바이러스의 활성을 감소시키는 모든 행위를 포함하는 것으로 해석될 수 있다.
본 명세서에서 상기 "예방"이란 상기 약학적 조성물의 투여로 제2형 중증급성호흡기증후군 코로나바이러스의 감염 또는 제2형 중증급성호흡기증후군 코로나바이러스 감염으로 인해 유발되는 질환을 억제 또는 지연시키는 모든 행위를 의미한다. 또한, 상기 "치료"란 상기 약학적 조성물의 투여로 제2형 중증급성호흡기증후군 코로나바이러스의 감염 또는 제2형 중증급성호흡기증후군 코로나바이러스 감염으로 인해 유발되는 질환의 증세가 호전되거나 이롭게 변경하는 모든 행위를 의미한다.
제2형 중증급성호흡기증후군 코로나바이러스 감염에 따른 질환은 호흡기 질환일 수 있다. 상기 제2형 중증급성호흡기증후군 코로나바이러스 감염 질환은 예를 들어 바이러스 감염 후 2일 내지 14일의 잠복기를 거친 뒤 증상이 나타날 수 있다. 이러한 증상은 예를 들어, 고열, 기침, 숨가쁨, 폐렴, 설사와 같은 위-장관 증상, 기관 기능 부전(신부전, 신장 기능장애 등), 패혈성 쇼크, 심한 경우 사망을 포함하여, 상기 바이러스 감염에 의해 발생되는 증상이라면 어떠한 것이라도 포함된다.
본 발명에서, 약학적으로 허용가능한 염은 의약업계에서 통상적으로 사용되는 염을 의미하며, 예를 들어 칼슘, 포타슘, 소듐 및 마그네슘 등으로 제조된 무기이온염, 염산, 질산, 인산, 브롬산, 요오드산, 과염소산 및 황산 등으로 제조된 무기산염; 아세트산, 트라이플루오로아세트산, 시트르산, 말레인산, 숙신산, 옥살산, 벤조산, 타르타르산, 푸마르산, 만데르산, 프로피온산, 젖산, 글리콜산, 글루콘산, 갈락투론산, 글루탐산, 글루타르산, 글루쿠론산, 아스파르트산, 아스코르브산, 카본산, 바닐릭산, 하이드로 아이오딕산 등으로 제조된 유기산염; 메탄설폰산, 에탄설폰산, 벤젠설폰산, p-톨루엔설폰산 및 나프탈렌설폰산 등으로 제조된 설폰산염; 글리신, 아르기닌, 라이신 등으로 제조된 아미노산염; 및 트리메틸아민, 트라이에틸아민, 암모니아, 피리딘, 피콜린, 트로타민
등으로 제조된 아민염 등이 있으나, 열거된 이들 염에 의해 본 발명에서 의미하는 염의 종류가 한정되는 것은 아니다.
약학적으로 허용가능한 염은 의약업계에서 통상적으로 사용되는 염은 또한 식품학적으로 허용가능한 염으로도 이용될 수 있다.
본 발명의 약학적 조성물은 약효를 증가시키지는 않으나 약학적 조성물에 통상 사용되어 냄새, 맛, 시각 등을 향상시킬 수 있는 성분을 추가로 포함할 수 있다. 또한, 본 발명의 약학적 조성물은 약학적으로 허용 가능한 첨가제를 추가적으로 포함할 수 있다. 약학적으로 허용 가능한 첨가제는 예컨대, 전분, 젤라틴화 전분, 미결정셀룰로오스, 유당, 포비돈, 콜로이달실리콘디옥사이드, 인산수소칼슘, 락토스, 만니톨, 엿, 아라비아고무, 전호화전분, 옥수수전분, 분말셀룰로오스, 히드록시프로필셀룰로오스, 오파드라이, 전분글리콜산나트륨, 카르나우바 납, 합성규산알루미늄, 스테아린산, 스테아린산마그네슘, 스테아린산알루미늄, 스테아린산칼슘, 백당, 덱스트로스, 소르비톨 및 탈크 등이 있으나, 이를 한정하지 않는다. 추가로, 상기 약학적 조성물은 단독 사용하거나 기존에 사용된 항 제2형 중증급성호흡기증후군 코로나바이러스 활성을 가지는 물질을 포함할 수 있다.
본 발명의 상기 약학적 조성물은 약학적으로 허용 가능한 담체를 포함하고 경구 또는 비경구용의 인체 또는 수의용으로 제형화될 수 있다. 본 발명의 약학적 조성물을 제제화하는 경우 충진제, 증량제, 결합제, 습윤제, 붕해제 및 계면활성제 등의 희석제 또는 부형제를 사용할 수 있다. 경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제 및 캡슐제 등이 포함되며, 이러한 고형제제는 본 발명의 화합물을 포함하는 약학적 조성물에 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘카보네이트(Calcium carbonate), 수크로스(Sucrose) 또는 락토오스(Lactose) 및 젤라틴 등을 섞어 조제할 수 있다. 또한 단순한 부형제 이외에 마그네슘, 스티레이트, 탈크 같은 윤활제를 사용할 수 있다. 경구를 위한 액상제제로는 현탁제, 내용액제, 유제 및 시럽제 등이 해당되는데 흔히 사용되는 단순희석제인 물 및 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제 및 보존제 등이 포함될 수 있다. 비경구투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조제제 및 좌제가 포함된다. 비수성용제 및 현탁용제로는 프로필렌글리콜(Propylene glycol), 폴리에틸렌 글리콜 및 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다.
본 발명의 약학적 조성물은 목적하는 방법에 따라 경구 투여하거나 비경구 투여할 수 있으며, 비경구 투여시 피부 외용 또는 복강내주사, 직장내주사, 피하주사, 정맥주사, 근육 내 주사 또는 흉부 내 주사 주입방식을 선택하는 것이 바람직하다.
또한, 본 발명에 따른 약학적 조성물은 흡입에 의해 투여될 수 있다. 폐로 직접 약물을 전달하면서도 독성을 나타내지 않고 적은 용량으로도 더 긴 작용 지속 시간을 나타낼 수 있다. 흡입용 투여는 약물을 함유하는 호흡 가능한 입자 또는 액적을 포함하여 기도, 비강 등을 통해 흡입 가능한 약학 제제를 이용한 투여일 수 있다. 이러한 흡입용 투여에 있어서는 이에 제한되는 것은 아니나, 예를 들어 건조 분말 흡입기 장치 (DPI) 또는 압축 계량 용량 흡입기(pressurized metered dose inhaler, pMDI) 중 하나를 사용할 수 있다. 약물 입자는 예를 들어 전달 장치(건조 분말 흡입기) 내부에 함유된 취성의(frangible) 매트릭스 내로 가볍게(lightly) 압축된다. 작동 시 전달 장치는 매트릭스로부터 약물 입자의 일부를 벗겨내고(abrades), 기도에 약물 입자를 전달하는 들숨 호흡 내로 이들을 분산시킨다. 대안적으로, 약물 입자는 전달 장치(건조 분말 흡입기) 내 저장소 내부에 함유되는 자유 유동성(free flowing) 분말일 수 있다. 저장소는 장치 내부의 일체식 챔버, 또는 캡슐, 블리스터 또는 작동 전에 장치 내부로 삽입되는 유사한 성능의 저장소일 수 있다. 작동시 장치는 저장소로부터 약물 입자의 일부를 분산시키고 기도에 약물 입자를 전달하는 흡입 호흡 내로 이들을 분산시킨다.
본 발명의 상기 약학적 조성물은 개체에 투여하여 제2형 중증급성호흡기증후군 코로나바이러스 감염 또는 제2형 중증급성호흡기증후군 코로나바이러스로 인해 유발되는 질환을 예방 또는 치료할 수 있다. 본 발명에서 사용된 용어, "개체" 또는 “대상체”는 제2형 중증급성호흡기증후군 코로나바이러스 감염 또는 제2형 중증급성호흡기증후군 코로나바이러스로 인해 유발되는 질환을 가지고 있으며, 본 발명의 상기 약학적 조성물을 투여하여 증상이 호전될 수 있는 질환을 가진 인간을 포함한 말, 양, 돼지, 염소, 개, 고양이, 족재비, 호랑이
등의 포유동물을 의미하나, 바람직하게는 인간을 의미한다.
본 발명에서 사용된 용어, "투여"는 어떠한 적절한 방법으로 개체에 본 발명의 약학적 조성물을 도입하는 것을 의미한다. 투여 경로는 목적 조직에 도달할 수 있는 한 어떠한 일반적인 경로를 통하여 경구 또는 비경구 투여될 수 있다. 또한, 본 발명의 약학적 조성물이 표적 세포로 이동할 수 있도록 하는 임의의 장치에 의해 투여될 수 있다.
예를 들어, 본 발명에 따른 약학적 조성물에 있어서 마크로락틴 유도체 또는 이의 약학적으로 허용가능한 염을 0.0001 내지 500 mg/kg으로, 바람직하게는 0.001 내지 100 mg/kg의 용량으로 투여할 수 있으며, 상기 투여는 하루에 한번 또는 수회 나누어 투여할 수도 있다.
본 발명의 약학적 조성물은 약학적으로 유효한 양으로 투여한다. 본 발명에서 사용된 용어, 약학적으로 유효한 양은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효 용량 수준은 환자의 체중, 성별, 연령, 건강상태, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료 기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 약학적 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있다. 단일 또는 다중 투여될 수 있다. 상기 요소를 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 당업자에 의해 용이하게 결정될 수 있다.
본 발명의 상기 약학적 조성물은 상기 마크로락틴 유도체, 또는 이의 약학적으로 허용가능한 염 외에 동일 또는 유사한 약효를 나타내는 유효성분을 1종 이상 더 포함할 수 있다.
본 발명은 상기 마크로락틴 유도체 또는 이의 약학적으로 허용가능한 염을 포함하는 조성물의 치료학적으로 유효한 양의 투여를 포함하는 제2형 중증급성호흡기증후군 코로나바이러스 감염 또는 제2형 중증급성호흡기증후군 코로나바이러스 감염으로 인해 유발되는 질환의 예방 또는 치료하는 방법을 제공한다.
본 발명에 사용되는 "치료학적으로 유효한 양"이라는 용어는 제2형 중증급성호흡기증후군 코로나바이러스 감염 또는 제2형 중증급성호흡기증후군 코로나바이러스 감염으로 인해 유발되는 질환의 예방 또는 치료에 유효한 상기 마크로락틴 유도체, 또는 이의 약학적으로 허용가능한 염의 양을 나타낸다.
본 발명은 치료학적으로 유효한 양의 마크로락틴 유도체, 또는 이의 약학적으로 허용가능한 염을 이를 필요로 하는 대상체에게 투여하는 단계;를 포함하는 제2형 중증급성호흡기증후군 코로나바이러스 감염 질환의 치료 방법을 제공한다.
상기 마크로락틴 유도체, 또는 이의 약학적으로 허용가능한 염, 또는 이를 포함하는 조성물을 투여함으로써 징후의 발현 전에 질병 그 자체를 다룰 뿐만 아니라, 이의 징후를 저해하거나 피하는 것을 또한 포함한다. 질환의 관리에 있어서, 특정 활성 성분의 예방적 또는 치료학적 용량은 질병 또는 상태의 본성(nature)과 심각도, 그리고 활성 성분이 투여되는 경로에 따라 다양할 것이다. 용량 및 용량의 빈도는 개별 환자의 연령, 체중 및 반응에 따라 다양할 것이다. 적합한 용량 용법은 이러한 인자를 당연히 고려하는 이 분야의 통상의 지식을 가진 자에 의해 쉽게 선택될 수 있다.

본 발명은 제2형 중증급성호흡기증후군 코로나바이러스 감염 질환의 치료를 위한 마크로락틴 유도체, 또는 이의 약학적으로 허용가능한 염을 제공한다.
본 발명은 또한, 마크로락틴 유도체, 또는 이의 약학적으로 허용가능한 염을 포함하는 항 제2형 중증급성호흡기증후군 코로나바이러스 조성물을 제공한다.

본 발명의 이해를 돕기 위하여 실시예를 제시한다. 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
<실시예 1> 발효, 추출 및 분석 조건
Bacillus velezensis K-4를 배양하였고 7 liter 자동발효기를 이용하여 배양하였다. 자동발효기를 사용하기 전에는 보관된 종균을 멸균된 TSB(tryptic soy brith)에 무균적으로 접종하여 37 ℃에서 200 rpm 교반속도로 배양하였다. 600 nm에서 흡광도가 0.7 까지 도달할때까지 배양하였고, 이 용액 0.5 리터를 자동발효기의 7 리터 발효액에 분주하였다. 이용한 발효액은 MA medium -1을 이용하였다. MA medium-1의 1 리터 중의 구성요소는 다음과 같다.
MA medium -1 / 1 liter : 16 g Nutrient broth (Difco), 2.5 μM FeSO
4, 500 μM CaCl
2, 10 μM MnCl
2, 1 mM MgSO
4, 13 mM KCl and 10 g Skim milk.
배양온도는 30 ℃ 이며 교반속도는 1 VVM 이었다. 산도는 pH 6.8로 조절하였다.
1일에 2회씩 발효액을 무균적으로 소량 분취하여 실험에 사용하였다.
원심분리하여 상등액을 취하고 동일한 부피의 Ethyl-acetate (E.A.)을 넣었다. 잘 혼합하여 추출하고 그중에서 50% 내외의 E.A.를 수거하여 감압농축하였다. 농축물을 메탄올에 녹이고 LC/Mass를 이용하여 분석하였다. 분석기기는 Agilent 1100 high-pressure liquid chromatography 였으며, 검출기는 an online diode array detector (DAD) 였다. 분석조건은 양성 또는 음성 모드에서 electrospray ionization 장치를 가진 an Agilent mass selective detector (MSD)를 이용하였다. 사용된 컬럼은 A Zorbax SB C18 column (dimensions, 250 by 4.6 ㎜) 이었고, 유속은 1 ㎖/min로 하였다. 분석파장은 UV 262 nm 였고, 이동상은 0.1% formic acid를 함유하는 acetonitrile (ACN)과 정제수를 이용하였다. ACN의 농도는 20분동안 0%에서 100% 까지 일정하게 농도를 높여서 사용하였다. 필요에 따라서는 일정하게 40%농도를 유지하면서 분석하였다.
고속액체크래마토그래미(HPLC)분석을 위하여는 “Shimadzu Class vp"기종을 사용하여 분석하였다. 분석결과로 2.5일에 상당량의 주요성분이 검출되었으며 (Compound 1), 이후에는 생산량에 큰 증가는 없었다. 위 결과는 도 1에 나타내었다.
그리고 나서, 위의 결과를 그래프로 생산량을 조사하였는데 아래 그림과 같으며, 30℃에서 2.5일 동안 배양하는 것이 좋다는 결론을 얻었으며, 관련 내용은 도 2에 나타내었다.
<실시예 2> 주피크 분석
분리된 주 피크를 MPLC를 이용하여 또 다시 분리하였다. 아래 그림과 같이 3개의 주요 피크가 검출되었으며, 관련 내용을 도 3에 나타내었다.
<실시예 3> MPLC에서 검출된 3개의 Peak의 HPLC 분석
MPLC에서 분리된 실시예 2의 첫 번째 피크 a], 두 번째 피크 b], 세 번째 피크 c]를 HPLC로 추가 분석하였다.
그 결과를 도 4에 나타내었다. a)는 혼합물인 상태의 검체이며, b)는 첫 번째 피크의 결과이고 C)는 두 번째 피크의 분석결과이며, D)는 세 번째 피크의 분석결과이다.
위 4개의 chromatogram에서 각각의 피크의 분자량을 체크하였고, 각각의 Peak의 특징은 아래 표 1과 같다. 분자량에 따른 추정 macrolactin은 “The predicted compound"에 표기하였다.

두 번째 Fraction에 존재하는 주 피크를 분석하였으며, 분리는 C-18 reversed-phase prep LC를 이용하였다.

각각의 피크를 분리하여 동일한 분석방법을 통하여 분석하여 구조를 얻었고, Macrolactin A, 7-O-malonyl macrolactin A, Macrolactin E, Macrolactin F, 7-O-succinyl Macrolactin A의 5개의 Macrolactin 류가 생성되는 것을 알 수 있었다.

멸균한 5% HP-20 resin (v/v %)을 포함하는 배지를 이용하였다. 이는 HP-20에 흡착하는 성질을 이용한 것이다.
예비실험으로 삼각 플라스크를 이용하여 배양하였으며, 30 ℃, 200 rpm에서 배양하였다.
2.5일 동안 배양 후에 여과를 통하여 HP-20 resin을 수거하였다. 에틸아세테이트로 세척한 후 이 용매를 휘산시키고, 잔류물에 메탄올을 넣어 녹여서 분석하였다.
그 결과를 도 5에 나타내었다. 이 분석의 결과로 주로 7-O-succinyl macrolactin A(SMA)로 분석되는 놀라운 결과를 보였다. 이는
Bacillus velezensis K-4 균주가 SMA를 주로 생성하고 Salt가 결합하면 분자량 526 물질이 발효액에서 분해되어 분자량 402의 macrolactin A (MA)로 잔류한다는 결론을 얻을 수 있었다.
<실시예 5> Macrolactin A의 급성독성시험
급성독성 Guideline(정부지침)에 따라서 수행하였다. 투여 후 14 일간 관찰하였고, 경구와 복강으로 투여하는 두 경로를 선택하였다. 각각 4마리를 하나의 군으로하여 실험하였다. 경구투여는 500, 1000, 2000 ㎎/㎏ mouse b.w. 으로 투여하였으며, 복강으로는 2000 ㎎/㎏ mouse b.w.으로 투여하였다. 사망이나 특별한 증상을 보이는 것은 없었다. 몸무게의 변화도 없었으며, 행동에 대한 특이한 점은 관찰되지 않았다.
실험 결과를 추가로 도 6에 나타내었다.
특이한 점은 14일 경과 후 수술에 의하여 개복하였을 때에 뭉치는 노란색 물질을 관찰할 수 있었으며 이것을 분석하였을 때에 MA로 분석되었다.
추가로 SMA에 대하여 동일한 방법으로 실험을 수행할 경우에 특이한 증상을 발견할 수 없었다. 따라서 경구투여나 복강투여에 의한 MA나SMA의 독성은 없는 것으로 보이며, MA의 경우에 복강투여할 경우에 분산이 될 수 있는 제제 개발이 필요하다고 사료되었다.
<실시예 6> 장부착력 실험
Bacillus polyfermenticus KJS2(BP2)를 이용한 장부착력실험을 수행하였다. 포유류의 장에는 Mannose가 많이 존재한다. 정장 생균제들 중에는 mannose수용체를 가지고 있음으로 장내에 mannose에 부착하여 장내에 머물려고 한다.
설치류를 대상으로 1 x 10
5 cfu/kg bw로 경구로 1회 투여하여 매일 분변을 받아 BP2의 검사를 하였다
. Endospore의 검사를 위하여는 80℃에서 2시간 처리하여 일반세균을 사멸시키고 endospore는 생존시켜 균수를 확인하였다. 또한 총균수는 BP2가 거의 모든 균수를 억제할수 있는 특징과 호기성에서 자라는 특징을 이용하여 배양하여 선택적으로 살수 있는 조건에서 배양하여 확인하였다.
그 결과를 도 7에 나타내었다. BP2는 단회 경구투여에서 3일동안 장내에 머물고 있었으며 4일째에는 거의 검출되지 않았다. 장부착능력이 있음을 알고 이것을 확인하고자 인공장점막인 Caco2를 이용하여 배양하여 부착여부를 확인하였다. Caco2를 배양하고 여기에 BP2를 넣어 방치시키고 4시간 후에 세포배양액을 관찰하였다. Caco2에 결합하는 Bp2를 확인할 수 있었다. 인체의 모든 점막은 외부바이러스, 박테리아의 침입경로이며, 코점막이나 눈의 점막 그리고 장내의 점막이 중요한 경로일 것이다. 따라서 장점막에 결합하는 정장생균제들은 각종 질병에 대한 방어 역할을 수행할 것이다. 장점막을 통해 침입한 바이러스나 박테리아는 인체의 모든 부위로 갈 수 있다. 생체에 침입하는 바이러스의 숫자를 줄이는 것은 생명보호에 꼭 필요한 요소일 것이다.
<실시예 7>
7-O-
Succinyl Macrolactin A의 Cell cytotoxicity assay (세포독성 평가) 및 CPE reduction Test(세포병변억제 효능 평가)
1종 SARS CoV-2 (NCCP-43326), VERO cell를 사용하여 시험을 수행하였고, CC
50, EC
50, SI에 대해 3반복으로 시험을 수행하였다.
Cell cytotoxicity assay (세포독성 평가)를 수행하였다. 세포배양액은 DMEM(5% FBS, 1% Antibiotic-Antimycotic)을 사용하였다.
시험시료에 대한 세포독성 평가는 MTT assay로 시험하였으며, 시험농도 별 시료 조제 시 사용한 용매(DMSO)는 최종농도 0.5%이하로 사용하였다.
96 wells plate에 well 당 5×10
4 cells/well로 분주 후 37℃, 5% CO
2 배양기에서 48시간 배양 후 세포단층을 얻은 후 PBS로 2회 세척하였다.
그리고 나서, 시험농도 별로 조제한 시료를 배양액을 포함하여 100 μL/well씩 분주 후 37℃, 5% CO
2 배양기에서 72시간 배양하였고, 세포상태 확인 후 MTT 용액을 10 μL/well씩 가한 후 37℃, 5% CO2 배양기에서 4시간 동안 정치하였다. 4시간 반응 후, MTT solution을 100 μL/well씩 가한 후 파이렛을 이용하여 formazan 결정체를 충분히 용해 후 plate reader기로 570 nm에서 흡광도를 측정하였다. 세포독성 비율은 정상세포와 시험시료를 처리한 군의 비율로 하기 식에 따라 산출하였다.
Cell viability(%) = Test OD/Control OD×100%
또한, 세포병변억제 효능 평가를 수행하였다.

2 배양기에서 1시간 정치하였다,
1시간 감염 후 바이러스를 제거 후 시험농도 별로 조제한 시료를 배양액을 포함하여 100 μL/well씩 분주 후 37℃, 5% CO
2 배양기에서 72시간 배양하였다.
그리고 나서, 세포상태 확인 후 MTT 용액을 10 μL/well씩 가한 후 37℃, 5% CO2 배양기에서 4시간 동안 정치하였고, 4시간 반응 후, MTT solution을 100 μL/well씩 가한 후 파이렛을 이용하여 formazan 결정체를 충분히 용해 후 plate reader기로 570 nm에서 흡광도를 측정하였다.
다음으로, 세포독성 비율은 정상세포와 시험시료를 처리한 군의 비율로 하기 식에 따라 산출하였다.
Virus inhibition rate(%) = (Test OD-Virus OD)/(Control OD-Virus OD)×100%
그 결과를 아래 표 2에 나타내었다.

Succinyl Macrolactin A(도 8)는 CC50>120 μM, EC50=13.6 μM, SI>8.8의 결과를 나타내어, 치료제로 효능을 나타내었다.
<실시예 8> Syrian Hamsters 동물모델을 이용하여 SARS-CoV-2 감염 후 약물 치료 실험
햄스터 암놈을 이용하여 그룹당 5마리를 배정하여 실험을 수행하였으며, 햅스터는 수령 약 1개월을 이용하였다. 이용한 햄스터는 ACE2 수용체를 발현시켜 SARS-CoV-2의 감염능력을 가지는 것을 예비실험을 통하여 확인 후 본 실험을 실시하였다. 바이러스의 접종은 Intranasal(비강)투여하였고 100 ul를 사용하였다.
SARS-CoV-2 바이러스의 역가 (Titer)는 2 X 10
5 PFU/ml을 사용하였다. 약물의 투여는 복강(intraperitoneal)으로 실시하였다. 투여용량은 500 ul로 하였으며, 아래 표 3과 같다.

Syrian Hamster 이용 코로나19 감염 동물모델 평가
실험을 실시하고 감염 후 4일째(PID4)에 부검을 통하여 폐조직을 채취하여 육안적 병변 정도를 평가하였으며, 그 결과를 도 9에 나타내었다.
대조군의 상태를 100 %로 보고 상대적인 병변 상태를 평가하였다. Vehicle control은 평균 1%를 보였고, 이에 비해 Remdesivir를 5 mg/kg/day로 1일 1회 투여한 군은 평균 44.3%를 보였다. 40mg/kg/day 군은 1일 2회 투여로 12.8%를 나타내었다.
위 확인할 수 있는 바와 같이, 본 발명에 따른 Succinyl Macrolactin A(SMA)의 처리는 우수한 치료 효과를 확인하였다.
<실시예 9> 데커시놀의 제조
실시예 9-1. 당귀 추출물의 제조
본 발명에서 사용된 당귀는 한국산 및 북한산 참당귀를 이용하였다. 참당귀를 40메시(mesh) 이하로 잘게 분쇄하여 수분함량이 5% 이하가 되도록 건조하였다. 분쇄 건조된 참당귀에 참당귀 무게의 2~4 배가 되도록 95%[v/v] 에탄올을 넣고 12시간 동안 반응하여 추출액을 확보하였고, 여과지를 이용해 고형물을 제거한 후, 증발 건조시켜 1차 당귀 농축물을 얻었다. 1차 농축물 1㎏당 에탄올 1ℓ를 넣어 현탁하고, -20℃에서 10시간 동안 방치한 후 생성된 침전물을 원심분리를 통해 제거하여 상층액을 확보하고, 확보한 상층액을 증발 건조하여 2차 당귀 농축물을 만들었다. 2차 당귀 농축물에 60%[v/v] 에탄올 50ℓ를 넣어 현탁한 후, 원심 분리하여 60%[v/v] 에탄올 층만을 분리하였다. 분리한 60%[v/v] 에탄올 용액을 증발 건조하여 농축하고, 이를 다시 90% 에탄올에 녹여 상등액을 취하여 최종의 참당귀 추출물을 얻었다.
실시예 9-2. 데커시놀의 제조
상기 실시예 9-1에서 제조한 참당귀 추출물 1000g을 95%[v/v] 에탄올로 현탁하고, 0.1N KOH 5000㎖을 처리하여 열분해 시킨 후, 6N HCl 575㎖ 넣어 중성화하였다. 중성화 과정에서 생성된 KCl 침전물을 원심 분리로 제거하고, 상층액은 소량의 에탄올이 남을 때까지 농축하여 4℃ 이하에서 방치하였다. 방치 중 에탄올에서 용해도가 낮은 데커시놀이 침전되면, 여과를 통해 침전물만을 따로 분리하고, 분리한 침전물을 0~4℃정도의 차가운 증류수를 이용하여 2회 세척하여 데커시놀 1차 정제물을 수득하였다.
또한 상기와 같은 조작을 반복하여 데커시놀 2차 정제물을 수득하였으며, 상기 조작을 (최소) 7회 반복하여 본 발명의 데커시놀 1,000 g을 확보하였다.
<실시예 10>
Bacillus polyfermenticus
KJS2와 Decursinol복합제를 투여한 후의 사람에서의 치료 효과 확인
Sars-Cov2 감염이 확정된 사람들 중에 자발적으로 복용을 원하는 환자 5명을 대상으로 Decursinol 160 mg 및 바실러스 폴리퍼멘티쿠스(
Bacillus Polyfermenticus) KJS-2 (기탁번호: KCCM10769P) 3 X 10
7 cfu를 1일 2회 경구투여하였다. 경구투여한 환자를 5일 후 검사하였고, PCR 음성, 항원음성, IgM 항체 양성, IgG한체 음성의 결과로 조기 완치판정 받았으며 이후 추가 검사에서도 정상으로 판별되었다.
또한, 부산의 한 학교 교사로 근무하던 중 감염되었고 감염 후 형제들 3명이 추가 감염되었다. 이들 4명 모두에게 Decursinol 160 mg 과 바실러스 폴리퍼멘티쿠스(
Bacillus Polyfermenticus) KJS-2 (기탁번호: KCCM10769P) 3 X 10
7 cfu를 1일 2회 경구투여하였다.
동일하게 5일 후에 PCR 검사에서 음성판정을 받았다.
더욱이 평소 Decursinol 45 mg 과
Bacillus polyfermenticus KJS 2 유산균 3 X 10
6CFU을 1일 1회 꾸준히 복용하였던 가족들은 모두 음성판정을 받았으며, 추가적인 PCR 검사에서도 모두 음성판정을 받았다.
상기 결과를 통해서 본 발명에 따른 병용 조합물이 우수한 치료 효과를 확인하였다.
본 명세서는 본 발명의 기술 분야에서 통상의 지식을 가진 자이면 충분히 인식하고 유추할 수 있는 내용은 그 상세한 기재를 생략하였으며, 본 명세서에 기재된 구체적인 예시들 이외에 본 발명의 기술적 사상이나 필수적 구성을 변경하지 않는 범위 내에서 보다 다양한 변형이 가능하다. 따라서 본 발명은 본 명세서에서 구체적으로 설명하고 예시한 것과 다른 방식으로도 실시될 수 있으며, 이는 본 발명의 기술 분야에 통상의 지식을 가진 자이면 이해할 수 있는 사항이다.

Claims (19)
- 마크로락틴 유도체, 또는 이의 약학적으로 허용가능한 염을 포함하는 제2형 중증급성호흡기증후군 코로나바이러스 감염 질환 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서, 마크로락틴 유도체는 마크로락틴 A, 마크로락틴 B, 마크로락틴 C, 마크로락틴 D, 마크로락틴 E, 마크로락틴 F, 마크로락틴 G, 마크로락틴 H, 마크로락틴 I, 마크로락틴 J, 마크로락틴 K 마크로락틴 L, 마크로락틴 M, 마크로락틴 N 마크로락틴 O, 마크로락틴 P, 마크로락틴 Q, 마크로락틴 R, 7-O-숙시닐 마크로락틴 F, 7-O-숙시닐 마크로락틴 A, 및 7-O-말로닐 마크로락틴 F로 이루어진 군으로부터 선택되는 어느 하나인, 제2형 중증급성호흡기증후군 코로나바이러스 감염 질환 예방 또는 치료용 약학적 조성물.
- 제2항에 있어서, 마크로락틴 유도체는 7-O-숙시닐 마크로락틴 A인, 제2형 중증급성호흡기증후군 코로나바이러스 감염 질환 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서,제2형 중증급성호흡기증후군 코로나바이러스 감염 질환은 호흡기 질환인 약학적 조성물.
- 제1항에 있어서, 제2형 중증급성호흡기증후군 코로나바이러스 감염 질환은 상기 코로나바이러스 감염 후 2일 내지 14일의 잠복기를 거친 뒤 증상이 발생하는 것인, 약학적 조성물.
- 제1항에 있어서, 상기 제2형 중증급성호흡기증후군 코로나바이러스 감염 질환은 고열, 기침, 숨가쁨, 폐렴, 설사, 신부전, 신장 기능 장애 및 패혈성 쇼크로 이루어진 군으로부터 선택되는 어느 하나인, 약학적 조성물.
- 마크로락틴 유도체, 또는 이의 식품학적으로 허용가능한 염을 포함하는 제2형 중증급성호흡기증후군 코로나바이러스 감염 질환 예방 또는 개선용 식품 조성물.
- 제7항에 있어서, 마크로락틴 유도체는 마크로락틴 A, 마크로락틴 B, 마크로락틴 C, 마크로락틴 D, 마크로락틴 E, 마크로락틴 F, 마크로락틴 G, 마크로락틴 H, 마크로락틴 I, 마크로락틴 J, 마크로락틴 K 마크로락틴 L, 마크로락틴 M, 마크로락틴 N 마크로락틴 O, 마크로락틴 P, 마크로락틴 Q, 마크로락틴 R, 7-O-숙시닐 마크로락틴 F, 7-O-숙시닐 마크로락틴 A, 및 7-O-말로닐 마크로락틴 F로 이루어진 군으로부터 선택되는 어느 하나인, 제2형 중증급성호흡기증후군 코로나바이러스 감염 질환 예방 또는 개선용 식품 조성물.

- 치료학적으로 유효한 양의 마크로락틴 유도체, 또는 이의 약학적으로 허용가능한 염을 이를 필요로 하는 대상체에게 투여하는 단계;를 포함하는 제2형 중증급성호흡기증후군 코로나바이러스 감염 질환의 치료 방법.
- 제2형 중증급성호흡기증후군 코로나바이러스 감염 또는 제2형 중증급성호흡기증후군 코로나바이러스 감염으로 인해 유발되는 질환의 치료용 약제의 제조를 위한 마크로락틴 유도체, 또는 이의 약학적으로 허용가능한 염의 용도.
- 제2형 중증급성호흡기증후군 코로나바이러스 감염 질환에 사용하기 위한, 마크로락틴 유도체, 또는 이의 약학적으로 허용가능한 염을 포함하는 조성물.
- 바실러스 벨레젠시스( Bacillus velezensis), 바실러스 폴리퍼멘티쿠스( Bacillus Polyfermenticus) 및 바실러스 아밀로리퀴페시언스 ( Bacillus amyloliquefacience)로 이루어진 군으로부터 선택되는 어느 하나 이상의 균주; 및 데커시놀(decursinol), 데커신(decursin), 데커시놀 안젤레이트(Decursinol angelate), 데커시놀 티글레이트(decursinol tiglate) 및 노다케닌(nodakenin)로 이루어진 군으로부터 선택되는 어느 하나 이상의 화합물을 포함하는 제2형 중증급성호흡기증후군 코로나바이러스 감염 질환 예방 또는 치료용 약학적 조성물.
