トリスルフィド化合物又はセレノトリスルフィド化合物の製造方法を開示する。かかる製造方法は、ジスルフィド化合物を酸化剤により酸化させてスルホキシド化合物を得る工程、及び得られたスルホキシド化合物を硫黄源又はセレン源と反応させてトリスルフィド化合物又はセレノトリスルフィド化合物を得る工程を含む。
Description
本発明は、トリスルフィド化合物又はセレノトリスルフィド化合物の製造方法に関する。
3つの硫黄原子による共有結合構造(-S-S-S-)を含む化合物はトリスルフィド化合物と呼ばれる。トリスルフィド化合物の中には、ジメチルトリスルフィド及びジプロピルトリスルフィドなどのようにフレーバーとして使用されるもの、グルタチオントリスルフィドなどのように抗酸化成分として期待されているものがある。
トリスルフィド化合物の製造方法としては、特許文献1~2及び非特許文献1に記載された方法などが知られている。
トリスルフィドの中央の硫黄原子がセレン原子に置き換わった共有結合構造(-S-Se-S-)を含む化合物はセレノトリスルフィド化合物と呼ばれる。セレノトリスルフィド化合物の中には、グルタチオンセレノトリスルフィドなどのように抗がん活性を有する化合物が見つかっており、セレノトリスルフィド化合物が医薬品として注目を集めている。

本発明の課題は、安全かつ安価なトリスルフィド化合物又はセレノトリスルフィド化合物の製造方法を提供することにある。
本発明者らは、上記課題を解決するために鋭意努力したところ、ジスルフィド化合物又はチオール化合物の酸化反応によりスルホキシド化合物を経由させることで、トリスルフィド化合物又はセレノトリスルフィド化合物を得られることを見出し、本発明を完成するに至った。
すなわち、本発明は、以下の(1)~(14)に関する。
(1)ジスルフィド化合物を酸化剤により酸化させてスルホキシド化合物を得る工程、及び、得られたスルホキシド化合物を硫黄源又はセレン源と反応させてトリスルフィド化合物又はセレノトリスルフィド化合物を得る工程を含む、トリスルフィド化合物又はセレノトリスルフィド化合物の製造方法。
(2)スルホキシド化合物を得る工程、及び、トリスルフィド化合物又はセレノトリスルフィド化合物を得る工程をワンポットで行う、上記(1)に記載の方法。
(3)酸化剤が、ペルオキシ一硫酸カリウム、過酢酸、過酸化水素、過酸化水素及びメチルトリオキソレニウム又は過ヨウ素酸ナトリウムである、上記(1)又は(2)に記載の方法。
(4)硫黄源が、硫化ナトリウム、硫化カリウム、硫化水素ナトリウム、硫化水素カリウム又は硫化水素であり、セレン源が、セレン化ナトリウム、セレン化カリウム、セレン化水素ナトリウム、セレン化水素カリウム又はセレン化水素である、上記(1)~(3)のいずれかに記載の方法。
(5)ジスルフィド化合物がR1-S-S-R2で表される化合物であり、R1及びR2は、同一又は異なってもよく、それぞれ又は一緒になって、置換基群Aから選択される一以上の置換基で置換されていてもよいアルキル基を表し、置換基群Aは、ハロゲン原子、ヒドロキシ基、置換基群Bから選択される一以上の置換基で置換されていてもよいアミノ基及びオキソ基からなり、置換基群Bは、ヒドロキシ基、アミノ基及びオキソ基からなる群から選択される一以上の置換基で置換されていてもよいアルキル基並びにアセチル基からなる、上記(1)~(4)のいずれかに記載の方法。
(6)ジスルフィド化合物がR3-S-S-R4で表される化合物であり、R3及びR4は、同一又は異なってもよく、保護基で保護されていてもよいシステインからSH基を除いた基又は保護基で保護されていてもよいシステイン含有ペプチドからSH基を除いた基を表す、上記(1)~(4)のいずれかに記載の方法。
(7)チオール化合物を酸化剤により酸化させてスルホキシド化合物を得る工程、及び、得られたスルホキシド化合物を硫黄源又はセレン源と反応させてトリスルフィド化合物又はセレノトリスルフィド化合物を得る工程を含む、トリスルフィド化合物又はセレノトリスルフィド化合物の製造方法。
(8)スルホキシド化合物を得る工程、及び、トリスルフィド化合物又はセレノトリスルフィド化合物を得る工程をワンポットで行う、上記(7)に記載の方法。
(9)酸化剤が、ペルオキシ一硫酸カリウム、過酢酸、過酸化水素、過酸化水素及びメチルトリオキソレニウム又は過ヨウ素酸ナトリウムである、上記(7)又は(8)に記載の方法。
(10)硫黄源が、硫化ナトリウム、硫化カリウム、硫化水素ナトリウム、硫化水素カリウム又は硫化水素であり、セレン源が、セレン化ナトリウム、セレン化カリウム、セレン化水素ナトリウム、セレン化水素カリウム又はセレン化水素である、上記(7)~(9)のいずれかに記載の方法。
(11)チオール化合物がR1-SHで表される化合物であり、R1は、置換基群Aから選択される一以上の置換基で置換されていてもよいアルキル基を表し、置換基群Aは、ハロゲン原子、ヒドロキシ基、置換基群Bから選択される一以上の置換基で置換されていてもよいアミノ基及びオキソ基からなり、置換基群Bは、ヒドロキシ基、アミノ基及びオキソ基からなる群から選択される一以上の置換基で置換されていてもよいアルキル基並びにアセチル基からなる、上記(7)~(10)のいずれかに記載の方法。

(14)チオール化合物がR3-SH及びR4-SHで表される化合物であり、R3及びR4は、異なって、保護基で保護されていてもよいシステインからSH基を除いた基又は保護基で保護されていてもよいシステイン含有ペプチドからSH基を除いた基を表す、上記(7)~(10)のいずれかに記載の方法。

(4)硫黄源が、硫化ナトリウム、硫化カリウム、硫化水素ナトリウム、硫化水素カリウム又は硫化水素であり、セレン源が、セレン化ナトリウム、セレン化カリウム、セレン化水素ナトリウム、セレン化水素カリウム又はセレン化水素である、上記(1)~(3)のいずれかに記載の方法。

本発明に係るトリスルフィド化合物又はセレノトリスルフィド化合物の製造方法は安全かつ安価である。
本発明の一実施形態に係るトリスルフィド化合物又はセレノトリスルフィド化合物の製造方法は、ジスルフィド化合物を酸化剤により酸化させてスルホキシド化合物を得る工程(工程1)、及び、得られたスルホキシド化合物を硫黄源又はセレン源と反応させてトリスルフィド化合物又はセレノトリスルフィド化合物を得る工程(工程2)を含む。
上記製造方法は、スルホキシド化合物を単離することなく、工程1及び工程2をワンポットで反応を行ってもよい。
工程1に用いられる溶媒は、ジスルフィド化合物及び酸化剤を溶解し、酸化反応を阻害しないものであれば、特に限定されない。このような溶媒としては、例えば、水、硫酸水溶液、エタノール水溶液及びアセトニトリル水溶液が挙げられ、好ましくは、水である。工程1に用いられる溶媒の量は、ジスルフィド化合物1gに対して1mL~500mLとすることができ、好ましくは、10mL~20mLである。
工程1に用いられる酸化剤として、ペルオキシ一硫酸カリウム(Oxone(登録商標)などの商品名で販売される)、過酢酸、過酸化水素及び過ヨウ素酸ナトリウムが挙げられる。過酸化水素は触媒量のメチルトリオキソレニウムとともに使用してもよい。安全性及びコストの観点から、ペルオキシ一硫酸カリウムが好ましい酸化剤である。使用される酸化剤の量は、ジスルフィド化合物1当量に対して、0.8当量~2.0当量とすることができ、好ましくは、1.0当量~1.3当量である。
工程1の反応温度は、-20℃~30℃とすることができ、好ましくは、-5℃~5℃である。
工程1の反応時間は、5分間~24時間とすることができ、好ましくは、0.5時間~2時間である。
工程2に用いられる溶媒は、スルホキシド化合物及び硫黄源又はセレン源を溶解し、その後の反応を阻害しないものであれば、特に限定されない。このような溶媒としては、例えば、例えば、水、硫酸水溶液、エタノール水溶液及びアセトニトリル水溶液が挙げられ、好ましくは、水である。工程2に用いられる溶媒の量は、スルホキシド化合物1gに対して1mL~500mLとすることができ、好ましくは、10mL~20mLである。
工程2に用いられる硫黄源として、硫化ナトリウム、硫化カリウム、硫化水素ナトリウム、硫化水素カリウム及び硫化水素が挙げられる。使用される硫黄源の量は、スルホキシド化合物1当量に対して、0.5当量~4.0当量とすることができ、好ましくは、0.9当量~1.2当量である。工程2に用いられるセレン源として、セレン化ナトリウム、セレン化カリウム、セレン化水素ナトリウム、セレン化水素カリウム及びセレン化水素が挙げられる。使用されるセレン源の量は、スルホキシド化合物1当量に対して、0.5当量~4.0当量とすることができ、好ましくは、0.9当量~1.2当量である。
工程2の反応温度は、-20℃~30℃とすることができ、好ましくは、-5℃~25℃である。
工程2の反応時間は、10分間~2日間とすることができ、好ましくは、0.5時間~2時間である。
工程1及び工程2をワンポットで行う場合、反応溶媒としては、例えば、水、硫酸水溶液、エタノール水溶液及びアセトニトリル水溶液が挙げられ、好ましくは、水であり、溶媒の量は、ジスルフィド化合物1gに対して1mL~500mLとすることができ、好ましくは、10mL~20mLである。使用される酸化剤としては、ペルオキシ一硫酸カリウム、過酢酸、過酸化水素(触媒量のメチルトリオキソレニウムとともに使用してもよい)及び過ヨウ素酸ナトリウムが挙げられ、好ましくは、ペルオキシ一硫酸カリウムであり、使用される酸化剤の量は、ジスルフィド化合物1当量に対して、0.8当量~2.0当量とすることができ、好ましくは、1.0当量~1.3当量である。使用される酸化剤の量は、ジスルフィド化合物1当量に対して、0.8当量~2.0当量とすることができ、好ましくは、1.0当量~1.3当量である。使用される硫黄源としては、硫化ナトリウム、硫化カリウム、硫化水素ナトリウム、硫化水素カリウム及び硫化水素が挙げられ、使用される硫黄源の量は、ジスルフィド化合物1当量に対して、0.5当量~4.0当量とすることができ、好ましくは、0.9当量~1.2当量である。使用されるセレン源としては、セレン化ナトリウム、セレン化カリウム、セレン化水素ナトリウム、セレン化水素カリウム及びセレン化水素が挙げられ、使用されるセレン源の量は、ジスルフィド化合物1当量に対して、0.5当量~4.0当量とすることができ、好ましくは、0.9当量~1.2当量である。反応温度は、-20℃~30℃とすることができ、好ましくは、-5℃~25℃である。反応時間は、15分間~2日間とすることができ、好ましくは、1時間~4時間である。
工程1及び工程2の他に、必要に応じて、ヒドロキシ基、カルボニル基、アミノ基、カルボキシ基などの官能基を保護する工程及び保護された官能基を脱保護する工程を含んでいてもよい。これらの官能基の保護基、保護/脱保護反応は当業者にとって周知であり、"Greene's Protective Groups in Organic Synthesis"などを参照して、適切な保護基、保護/脱保護反応を選択することができる。
ジスルフィド化合物の一例として、R1-S-S-R2で表される化合物が挙げられる。R1及びR2は同一であってもよく(すなわち、対称ジスルフィド化合物)、R1及びR2は異なってもよい(すなわち、非対称ジスルフィド化合物)。R1及びR2は一緒になって以下に述べる基を形成してもよい(すなわち環状ジスルフィド化合物)。
R1及びR2は、置換基群Aから選択される一以上の置換基で置換されていてもよいアルキル基を表す。置換基群Aは、ハロゲン原子、ヒドロキシ基、置換基群Bから選択される一以上の置換基で置換されていてもよいアミノ基及びオキソ基からなる。置換基群Bは、ヒドロキシ基、アミノ基及びオキソ基からなる群から選択される一以上の置換基で置換されていてもよいアルキル基並びにアセチル基からなる。アルキル基の炭素数は1~6であってよく、例えば、メチル基、エチル基、プロパン-1-イル基、プロパン-2-イル基(イソプロピル基)、ブタン-1-イル基、ブタン-2-イル基、ペンタン-1-イル基、ペンタン-2-イル基、ペンタン-3-イル基、ヘキサン-1-イル基、ヘキサン-2-イル基および3-ヘキシル基が挙げられる。
ジスルフィド化合物が、R1-S-S-R2で表される化合物である場合、生成するスルホキシド化合物は、R1-S(=O)-S-R2又はR1-S-S(=O)-R2で表される化合物であり、生成するトリスルフィド化合物は、R1-S-S-S-R2で表される化合物であり、生成するセレノトリスルフィド化合物は、R1-S-Se-S-R2で表される化合物である。
ジスルフィド化合物の別の一例として、R3-S-S-R4で表される化合物が挙げられる。R3及びR4は同一であってもよく(すなわち、対称ジスルフィド化合物)、R3及びR4は異なってもよい(すなわち、非対称ジスルフィド化合物)。R3及びR4は一緒になって以下に述べる基を形成してもよい(すなわち環状ジスルフィド化合物)。
R3及びR4は、保護基で保護されていてもよいシステインからSH基を除いた基又は保護基で保護されていてもよいシステイン含有ペプチドからSH基を除いた基を表す。ペプチドの長さは特に限定されないが、例えば、2アミノ酸~10アミノ酸からなるペプチドであってよく、2アミノ酸~5アミノ酸からなるペプチドであることが好ましい。保護基で保護されたシステインとしては、カルボキシ基及び/又はアミノ基が保護されたシステインが挙げられ、具体的には、N-アセチルシステインなどが挙げられる。保護基で保護されたペプチドとは、C末端カルボキシ基、N末端アミノ基及び/又は反応性官能基を有する側鎖(例えば、リジン残基のアミノ基、アスパラギン酸残基及びグルタミン酸残基のカルボキシ基など)が保護されたペプチドが挙げられる。
ジスルフィド化合物が、R3-S-S-R4で表される化合物である場合、生成するスルホキシド化合物は、R3-S(=O)-S-R4又はR3-S-S(=O)-R3で表される化合物であり、生成するトリスルフィド化合物は、R3-S-S-S-R4で表される化合物であり、生成するセレノトリスルフィド化合物は、R3-S-Se-S-R4で表される化合物である。
本発明の別の実施形態に係るトリスルフィド化合物又はセレノトリスルフィド化合物の製造方法は、チオール化合物を酸化剤により酸化させてスルホキシド化合物を得る工程(工程1’)、及び、得られたスルホキシド化合物を硫黄源又はセレン源と反応させてトリスルフィド化合物又はセレノトリスルフィド化合物を得る工程(工程2)を含む。
上記製造方法は、ジスルフィド化合物を単離することなく、ワンポットで反応を行ってもよい。
工程1’に用いられる溶媒は、チオール化合物及び酸化剤を溶解し、酸化反応を阻害しないものであれば、特に限定されない。このような溶媒としては、例えば、例えば、水、硫酸水溶液、エタノール水溶液及びアセトニトリル水溶液が挙げられ、好ましくは、水である。工程1’に用いられる溶媒の量は、チオール化合物1gに対して1mL~500mLとすることができ、好ましくは、10mL~20mLである。
工程1’に用いられる酸化剤及びその量は、工程1において説明したものと同様である。
工程1’の反応温度は-20℃~30℃とすることができ、好ましくは、-5℃~5℃である。
工程1’の反応時間は、10分間~24時間とすることができ、好ましくは、0.5時間~2時間である。
工程1’及び工程2をワンポットで行う場合、反応溶媒としては、例えば、水、硫酸水溶液、エタノール水溶液及びアセトニトリル水溶液が挙げられ、好ましくは、水であり、溶媒の量は、チオール化合物1gに対して1mL~500mLとすることができ、好ましくは、10mL~20mLである。使用される酸化剤としては、ペルオキシ一硫酸カリウム、過酢酸、過酸化水素(触媒量のメチルトリオキソレニウムとともに使用してもよい)及び過ヨウ素酸ナトリウムが挙げられ、好ましくは、ペルオキシ一硫酸カリウムであり、使用される酸化剤の量は、チオール化合物1当量に対して、0.8当量~2.0当量とすることができ、好ましくは、1.0当量~1.3当量である。使用される酸化剤の量は、チオール化合物1当量に対して、0.8当量~2.0当量とすることができ、好ましくは、1当量~1.3当量である。使用される硫黄源としては、硫化ナトリウム、硫化カリウム、硫化水素ナトリウム、硫化水素カリウム及び硫化水素が挙げられ、使用される硫黄源の量は、チオール化合物1当量に対して、0.5当量~4.0当量とすることができ、好ましくは、0.9当量~1.2当量である。使用されるセレン源としては、セレン化ナトリウム、セレン化カリウム、セレン化水素ナトリウム、セレン化水素カリウム及びセレン化水素が挙げられ、使用されるセレン源の量は、チオール化合物1当量に対して、0.5当量~4.0当量とすることができ、好ましくは、0.9当量~1.2当量である。反応温度は、-20℃~30℃とすることができ、好ましくは、-5℃~25℃である。反応時間は、15分間~2日間とすることができ、好ましくは、1時間~4時間である。
工程1’及び工程2の他に、必要に応じて、ヒドロキシ基、カルボニル基、アミノ基、カルボキシ基などの官能基を保護する工程及び保護された官能基を脱保護する工程を含んでいてもよい。これらの官能基の保護基、保護/脱保護反応は当業者にとって周知であり、"Greene's Protective Groups in Organic Synthesis"などを参照して、適切な保護基、保護/脱保護反応を選択することができる。
チオール化合物の一例として、R1-SHで表される化合物が挙げられる。2種類のチオール化合物R1-SH及びR2-SHを用いて反応を行ってもよい。R1及びR2の定義は上述のとおりである。
チオール化合物が、R1-SHで表される化合物である場合、生成するスルホキシド化合物は、R1-S(=O)-S-R1で表される化合物であり、生成するトリスルフィド化合物は、R1-S-S-S-R1で表される化合物であり、生成するセレノトリスルフィド化合物は、R1-S-Se-S-R1で表される化合物である。チオール化合物が、R1-SHで表される化合物及びR2-SHで表される化合物である場合、生成するスルホキシド化合物は、R1-S(=O)-S-R1で表される化合物、R2-S(=O)-S-R2で表される化合物、R1-S(=O)-S-R2で表される化合物及びR1-S-S(=O)-R2で表される化合物のいずれか又はこれらの混合物である。混合物である場合、必要に応じて、所望のスルホキシド化合物を分離することができる。生成するトリスルフィド化合物は、R1-S-S-S-R1で表される化合物、R2-S-S-S-R2で表される化合物及びR1-S-S-S-R2で表される化合物のいずれか又はこれらの混合物である。生成するセレノトリスルフィド化合物は、R1-S-Se-S-R1で表される化合物、R2-S-Se-S-R2で表される化合物及びR1-S-Se-S-R2で表される化合物のいずれか又はこれらの混合物である。混合物である場合、必要に応じて、所望のトリスルフィド化合物又はセレノトリスルフィド化合物を分離することができる。
チオール化合物の別の一例として、R3-SHで表される化合物が挙げられる。2種類のチオール化合物R3-SH及びR4-SHを用いて反応を行ってもよい。R3及びR4の定義は上述のとおりである。
チオール化合物が、R3-SHで表される化合物である場合、生成するスルホキシド化合物は、R3-S(=O)-S-R3で表される化合物であり、生成するトリスルフィド化合物は、R3-S-S-S-R3で表される化合物であり、生成するセレノトリスルフィド化合物は、R3-S-Se-S-R3で表される化合物である。チオール化合物が、R3-SHで表される化合物及びR4-SHで表される化合物である場合、生成するスルホキシド化合物は、R3-S(=O)-S-R3で表される化合物、R4-S(=O)-S-R4で表される化合物、R3-S(=O)-S-R4で表される化合物及びR3-S-S(=O)-R4で表される化合物のいずれか又はこれらの混合物である。混合物である場合、必要に応じて、所望のスルホキシド化合物を分離することができる。生成するトリスルフィド化合物は、R3-S-S-S-R3で表される化合物、R4-S-S-S-R4で表される化合物及びR3-S-S-S-R4で表される化合物のいずれか又はこれらの混合物である。生成するセレノトリスルフィド化合物は、R3-S-Se-S-R3で表される化合物、R4-S-Se-S-R4で表される化合物及びR3-S-Se-S-R4で表される化合物のいずれか又はこれらの混合物である。混合物である場合、必要に応じて、所望のトリスルフィド化合物又はセレノトリスルフィド化合物を分離することができる。
酸化型グルタチオン(GSSG)、硫酸水溶液を反応容器に仕込み、この溶液を温度調整した。ここに、過酢酸(AcO2H)、過酸化水素(H2O2)、メチルトリオキソレニウム(MeReO3)、過ヨウ素酸ナトリウム(NaIO4)及びOxone(登録商標)を表1に記載の量を加え、反応させた。反応後の生成物(GS(=O)SG)のHPLC(高速液体クロマトグラフィー)純度を測定した結果および反応条件を表1に示す。
HPLC条件は以下のとおりである。

カラム:LiChrosorb RP-18(関東化学、4.0×250mm、5μm)
カラム温度:40℃付近の一定温度
移動相A:リン酸水溶液(pH3)
移動相B:メタノール
移動相の送液:移動相A及び移動相Bの混合比を次のように変えて濃度勾配制御する。
流量:0.6mL/min
検出器:紫外吸光光度計(測定波長:220nm)
カラム:LiChrosorb RP-18(関東化学、4.0×250mm、5μm)
カラム温度:40℃付近の一定温度
移動相A:リン酸水溶液(pH3)
移動相B:メタノール
移動相の送液:移動相A及び移動相Bの混合比を次のように変えて濃度勾配制御する。
酸化型グルタチオン(GSSG)10g(13.97mmol)、1mol/L硫酸水溶液20mLを反応容器に仕込み、氷浴にて冷却した。ここに、AcO2H(8.7%酢酸溶液、15.8g、18.1mmol)を滴下し、約2時間反応させた。続いて、室温まで昇温後、表2に示す硫黄源を添加し、約2時間反応させた。反応液に、エタノールを27mL添加後、飽和した炭酸ナトリウム水溶液を3mL添加した。スラリー液をろ過後、50%エタノール水溶液20mLで結晶を洗浄した。ろ取した結晶を室温で減圧乾燥し、グルタチオントリスルフィド(生成物)を得た。結果および反応条件を表3に示す。
HPLC条件は実施例1に記載した条件と同一である。
α-リポ酸2.0g(9.02mmol)、75%エタノール水溶液40mLを反応容器に仕込み、内温0℃まで冷却した。ここに、Oxone(登録商標)3.4g(10.20mmol)を添加し、約2時間反応させた。反応液中の無機塩をろ過後、エタノール7mLで洗浄した。ろ液に、硫化ナトリウム九水和物5.8g(24.1mmol)を添加し、約1時間反応させた。この反応液に3mol/L硫酸水溶液を7mL滴下後、続けて、水20mL、酢酸エチル(AcOEt)45mLを添加し、AcOEtで抽出した。水層をAcOEt20mLで2回抽出し、有機層を合わせて減圧濃縮した。濃縮物にエタノール3mLを加えて溶解した後、溶解液をODSカラム(YMC Dispo PackAT、移動相:アセトニトリル水溶液)により精製し、α-リポ酸トリスルフィド0.7g(2.39mmol、HPLC純度:100%)を得た。
HPLC条件は以下のとおりである。

カラム:LiChrosorb RP-18(関東化学、4.0×250mm、5μm)
カラム温度:40℃付近の一定温度
移動相A:リン酸水溶液(pH3)
移動相B:メタノール
移動相の送液:移動相A及び移動相Bの混合比を次のように変えて濃度勾配制御する。
L-シスチン1.0g(4.16mmol)、1mol/L硫酸水溶液10mLを反応容器に仕込み、氷浴にて冷却した。ここに、Oxone(登録商標)1.5g(4.35mmol)を添加し、約1時間反応させた。続いて、硫化ナトリウム九水和物1.1g(4.37mmol)を添加し、約3時間反応させた。反応液に、1mol/L炭酸水素ナトリウム水溶液を19mL滴下後、スラリー液をろ過し、水10mLで洗浄した。ろ取した湿晶を、室温で減圧乾燥し、システイントリスルフィド0.75g(2.75mmol、HPLC純度:89%)を得た。
HPLC条件は実施例1に記載した条件と同一である。
N,N’-ジアセチル-L-シスチン1.0g(3.08mmol)、水10mLを反応容器に仕込み、内温1℃まで冷却した。ここに、Oxone(登録商標)1.25g(3.72mmol)を添加し、約3時間反応させた。続いて、0.44mol/L硫化ナトリウム水溶液8.5mL(3.71mmol)を滴下し、約3時間反応させた。反応液に、アセトニトリルを33mL添加後、無機塩をろ過し、アセトニトリル5mLで洗浄した。このろ液をエバポレーターで減圧濃縮し、濃縮物はODSカラム(移動相:アセトニトリル水溶液)により精製し、N,N’-ジアセチル-L-システイントリスルフィド0.2g(0.56mmol)を得た。
HPLC条件は以下のとおりである。
検出器:紫外吸光光度計(測定波長:220nm)
カラム:LiChrosorb RP-18(関東化学、4.0×250mm、5μm)
カラム温度:40℃付近の一定温度
移動相:40%(v/v)アセトニトリル水溶液
流量:0.5mL/min
検出器:紫外吸光光度計(測定波長:220nm)
カラム:LiChrosorb RP-18(関東化学、4.0×250mm、5μm)
カラム温度:40℃付近の一定温度
移動相:40%(v/v)アセトニトリル水溶液
流量:0.5mL/min
N-アセチル-L-システイン1.0g(6.13mmol)、20%アセトニトリル水溶液40mLを反応容器に仕込み、内温5℃まで冷却した。ここに、Oxone(登録商標)3.4g(10.14mmol)を添加し、約2.5時間反応させた。続いて、硫化ナトリウム九水和物1.5g(6.12mmol)を添加し、約1時間反応させた。アセトニトリル33mL加えた後、無機塩をろ過し、アセトニトリル3mLで洗浄した。このろ液をエバポレーターで減圧濃縮し、濃縮物はODSカラム(移動相:アセトニトリル水溶液)により精製し、N,N’-ジアセチル-L-システイントリスルフィド0.1g(0.28mmol)を得た。
HPLC条件は実施例5に記載した条件と同一である。
実施例7
酸化型グルタチオン(GSSG)500.3mg(0.69mmol)、0.2mol/L硫酸水溶液8.5mLを反応容器に仕込み、氷浴にて冷却した。ここに、Oxone(登録商標)278.0mg(0.83mmol)を添加し、約2.5時間反応させた。続いて、1.0mol/L炭酸水素ナトリウム水溶液6mLを滴下後、0.2mol/Lセレン化ナトリウム水溶液3.8mL(0.76mmol)を滴下し、約1時間反応させた。この反応液をLC/MS(HR-ESI-TOF-MS)で分析し、GSSeSG(グルタチオンセレノトリスルフィド)の生成を確認した。

実施例8
α-リポ酸246.8mg(1.20mmol)、75%エタノール水溶液4.9mLを反応容器に仕込み、氷浴にて冷却した。ここに、Oxone(登録商標)419.1mg(1.26mmol)を添加し、約1時間反応させた。反応液中の無機塩をろ過後、エタノール1mLで洗浄した。ろ液に、1.0mol/L炭酸水素ナトリウム水溶液2.6mLを滴下後、セレン化ナトリウム117mg(0.93mmol)を添加し、約3時間反応させた。この反応液をLC/MS(HR-ESI-TOF-MS)で分析し、α-リポ酸SSeS(α-リポ酸セレノトリスルフィド)の生成を確認した。

Claims (28)
- ジスルフィド化合物を酸化剤により酸化させてスルホキシド化合物を得る工程、及び
得られたスルホキシド化合物を硫黄源と反応させてトリスルフィド化合物を得る工程を含む、
トリスルフィド化合物の製造方法。 - スルホキシド化合物を得る工程及びトリスルフィド化合物を得る工程をワンポットで行う、請求項1に記載の方法。
- 酸化剤が、ペルオキシ一硫酸カリウム、過酢酸、過酸化水素、過酸化水素及びメチルトリオキソレニウム又は過ヨウ素酸ナトリウムである、請求項1又は2に記載の方法。
- 硫黄源が、硫化ナトリウム、硫化カリウム、硫化水素ナトリウム、硫化水素カリウム又は硫化水素である、請求項1~3のいずれか一項に記載の方法。
- ジスルフィド化合物がR1-S-S-R2で表される化合物であり、
R1及びR2は、同一又は異なってもよく、それぞれ又は一緒になって、置換基群Aから選択される一以上の置換基で置換されていてもよいアルキル基を表し、
置換基群Aは、ハロゲン原子、ヒドロキシ基、置換基群Bから選択される一以上の置換基で置換されていてもよいアミノ基及びオキソ基からなり、
置換基群Bは、ヒドロキシ基、アミノ基及びオキソ基からなる群から選択される一以上の置換基で置換されていてもよいアルキル基並びにアセチル基からなる、
請求項1~4のいずれか一項に記載の方法。 - ジスルフィド化合物がR3-S-S-R4で表される化合物であり、

R1は、置換基群Aから選択される一以上の置換基で置換されていてもよいアルキル基を表し、
