Abstract
Description
医療容器等の用途に使用されるプロピレン系樹脂組成物は、最終段階で各種滅菌処理に供されることから、これらの処理に対する耐性を有することが要求される。一般的に行う滅菌処理としては、高圧蒸気滅菌、エチレンオキサイドガス滅菌、放射線滅菌がある。放射線滅菌にはガンマ線照射による滅菌と、電子線、特に紫外線照射による滅菌とがある。
[1] エチレン含量が1~5質量%、JIS K7210(230℃、2.16kg荷重)に準拠したメルトフローレイト(以下、MFRと略称することがある。)が10~100g/10分であるプロピレン-エチレン共重合体(a)の90質量%超97質量%以下と、エチレン含量が15~22質量%、MFRが1~50g/10分であるプロピレン-エチレン共重合体(b)3質量%以上10質量%未満とからなるプロピレン-エチレン系樹脂組成物(A)(ただし、(a)及び(b)の合計は100質量%である)88~95質量部、及び密度0.880~0.920g/cm3のエチレン-α-オレフィンランダム共重合体であるエラストマー(B)5~12質量部(ただし、(A)及び(B)の合計は100質量部である)、および耐候安定剤を0.01~0.20質量部を含む医療用プロピレン-エチレン系樹脂組成物を用いて得られる射出成形品であって、該射出成形品はγ線又は電子線滅菌されたものである医療用途向け射出成形品。
[3] 造核剤を含有する[1]又は[2]に記載の医療用途向け射出成形品。
本発明で使用するプロピレン-エチレン共重合体(a)は以下の特性1a及び2aを満足する。
(特性1a:MFR)
本発明に用いるプロピレン-エチレン共重合体(a)のMFRは10~100g/10分の範囲であることが必要であり、好ましくは20~60g/10分、より好ましくは20~40g/10分である。この範囲の下限値以上であると流動性の向上により成形加工性が良好となり、特に成形品の肉厚が2.5mm厚以下のものを成形した場合でも成形配向がかかり難くなり、衝撃を受けた場合、成形配向方向に亀裂が生じるのを防ぐことが出来、上限値以下のものは樹脂組成物の生産性が良好となり経済上好ましいと共に、成形品の放射線滅菌後の耐衝撃性に優れる。
MFR値の制御の方法は周知であり、重合条件である温度や圧力を調節したり、水素等の連鎖移動剤を重合時に添加する水素添加量の制御により、容易に調整を行うことができる。
なお、本発明において、プロピレン系樹脂のMFRは、JIS K7210:1999「プラスチック-熱可塑性プラスチックのメルトマスフローレイト(MFR)及びメルトボリュームフローレイト(MVR)の試験方法」のA法、条件M(230℃、2.16kg荷重)に準拠して測定され、単位はg/10分である。また、分子量調整剤を用いてMFRをCR(コントロールドレオロジー)してMFRを調整する方法が一般に知られているが、本発明においてはCRせずに重合条件のみでMFRを調整することが成形時のエラストマー由来の樹脂焼け防止の観点から好ましい。
本発明に用いるプロピレン-エチレン共重合体(a)のエチレン含量は1~5質量%の範囲であることが必要であり、好ましくは1.5~3.0質量%である。この範囲の下限値以上であると成形品の透明性が良好となると共に、放射線滅菌後の耐衝撃性に優れる。
また上限値以下であると結晶化温度の上昇により成形時の固化が速くなり成形加工性が良好となる。
エチレン含量は、重合時におけるプロピレンとエチレンのモノマー組成の制御によって調整することができる。
本発明で使用するプロピレン-エチレン共重合体(b)は以下の特性1b及び2bを満足する。
(特性1b:MFR)
本発明に用いるプロピレン-エチレン共重合体(b)のMFRは1~50g/10分の範囲であることが必要であり、好ましくは1~30g/10分、より好ましくは1~20g/10分、最も好ましくは1~10g/10分である。この範囲の下限値以上であるとプロピレン-エチレン共重合体(a)への分散性が向上し、成形品にフィッシュアイが発生することを抑制することが可能となる。また上限値以下であると低結晶成分が表面にブリードしにくくなることにより薬剤吸着性が良好となると共に、放射線滅菌後の耐衝撃性が良好となる。また、分子量調整剤を用いてMFRをCR(コントロールドレオロジー)してMFRを調整する方法が一般に知られているが、本発明においてはCRせずに重合条件のみでMFRを調整することが成形時のエラストマー由来の樹脂焼け防止の観点から好ましい。
本発明に用いるプロピレン-エチレン共重合体(b)のエチレン含量は15~22質量%の範囲であることが必要であり、好ましくは15~21質量%であり、より好ましくは15~19質量%であり、さらに好ましくは17~19質量%である。この範囲の下限値以上であると成形品の放射線滅菌後の耐衝撃性が向上する。また上限値以下であるとプロピレン-エチレン共重合体(a)との相溶性が向上することにより成形品の透明性が良好となると共に、プロピレン-エチレン共重合体(b)が成形品表面にブリードしにくくなることにより、べたつきや薬剤吸着性が良好となる。
本発明に用いるプロピレン-エチレン共重合体(a)とプロピレン-エチレン共重合体(b)の質量比はプロピレン-エチレン共重合体(a)が90質量%超97質量%以下、プロピレン-エチレン共重合体(b)が3質量%以上10質量%未満の範囲であることが必要である。好ましくはプロピレン-エチレン共重合体(a)が91質量%~95質量%、プロピレン-エチレン共重合体(b)が5質量%~9質量%である。プロピレン-エチレン共重合体(a)の質量比の下限値90質量%超であると、成形品の剛性や水中での光線透過率が向上し、上限値97質量%以下であると成形品の耐衝撃性に優れる。
また、各エチレン含量は13C-NMR法で測定して求めた値である。
本発明のプロピレン-エチレン系樹脂組成物には密度0.880~0.920g/cm3のエチレン-α-オレフィンランダム共重合体であるエラストマー(B)を含む。前記エラストマー(B)としては、メタロセン触媒を用いて重合され、JIS K7210(190℃、2.16kg荷重)に準拠したMFRが1~100g/10分であるエチレン-α-オレフィンランダム共重合体であることが好ましい。エラストマー(B)は1種単独で使用することもできるし、2種以上を組み合せて使用することもできる。
これらのα-オレフィンは、単独で又は組み合せて用いることができる。これらの中では、特にプロピレン、1-ブテン、1-ヘキセン、1-オクテンが好ましく用いられる。さらに、α-オレフィンを組み合わせて用いるときは、プロピレン、1-ブテンを組み合わせることが好ましい。以下にエチレン-α-オレフィンランダム共重合体、エチレン-プロピレン-ブテンランダム共重合体についての詳細を記す。
本発明に用いるエラストマー(B)の中でも、エチレン-α-オレフィンランダム共重合体を特定量含有させることにより材料のモルフォロジーを変化させ、透明性、低臭気性、剛性及び低異物出現性を保持したまま、さらに耐衝撃性を向上させることができる。
このようなエチレン-α-オレフィンランダム共重合体は、密度が0.880~0.920g/cm3、好ましくは0.880~0.915g/cm3であるエチレン-α-オレフィンランダム共重合体である。α-オレフィンの例としては、プロパン、1-ブテン、1-ペンテン、1-ヘキセン、4-メチル-1-ペンテン、3-メチル-1-ペンテン、1-ヘプテン、1-オクテン、1-デセン等を挙げることができる。
具体的なエチレン-α-オレフィンランダム共重合体の共重合体は、エチレン-プロピレンランダム共重合体、エチレン-1-ブテンランダム共重合体、エチレン-1-ペンテンランダム共重合体、エチレン-1-ヘキセンランダム共重合体、エチレン-4-メチル-1-ペンテンランダム共重合体、エチレン-3-メチル-1-ペンテンランダム共重合体、エチレン-1-ヘプテンランダム共重合体、エチレン-1-オクテンランダム共重合体、エチレン-1-デセンランダム共重合体等を挙げることができる。
エチレン-α-オレフィンランダム共重合体のガラス転移温度(Tg)は-130℃~-20℃のものが使用できるが、一般には、プロピレン系重合体よりかなり低いので、これをプロピレン系重合体へブレンドして、透明性を維持しつつ放射線照射後の耐衝撃性を改良するという試みが為されているが、期待以上の成果を達成できなかった。しかし、本発明のプロピレン-エチレン系樹脂組成物(A)とエラストマー(B)の併用は優れた効果が期待できる。
具体的には、エチレン-α-オレフィンランダム共重合体のMFR(190℃)/プロピレン-エチレン系樹脂組成物(A)のMFR比は、0.05~1.2が好ましく、0.1~1.0がさらに好ましい。この範囲は、本発明の医療用成形品としての透明性、異物の混入、臭気防止などの機能を適正に発現するためにおいても有意義であることが予測される。
このようなエチレン-α-オレフィンランダム共重合体は、市販品として、日本ポリエチレン(株)製のノバテックLLシリーズやハーモレックスシリーズ、カーネルシリーズ、三井化学(株)製のタフマーPシリーズやタフマーAシリーズ、(株)プライムポリマー製のエボリューシリーズ、住友化学(株)製のスミカセンE、EPシリーズ、エクセレンGMHシリーズなどが例示できる。
また、メタロセン触媒を用いて重合されたエチレン-α-オレフィンランダム共重合体としては、日本ポリエチレン(株)製のハーモレックスシリーズ、カーネルシリーズ、プライムポリマー製のエボリューシリーズ、住友化学(株)製のエクセレンFXシリーズ等が例示できる。
本発明の医療用プロピレン-エチレン系樹脂組成物が造核剤を含有することで、より透明性が良好な成形品を得ることができる。造核剤としては、特に限定はないが、ソルビトール系造核剤、リン系造核剤、カルボン酸金属塩系造核剤、ポリマー造核剤、無機化合物等を用いることができる。造核剤としては、ソルビトール系造核剤、リン系造核剤、ポリマー造核剤を用いることが好ましい。
ポリマー造核剤としては分岐状α-オレフィン重合体が好適に用いられる。分岐状α-オレフィン重合体の例として、3-メチル-1-ブテン、3-メチル-1-ペンテン、3-エチル-1-ペンテン、4-メチル-1-ペンテン、4-メチル-1-ヘキセン、4,4-ジメチル-1-ヘキセン、4,4-ジメチル-1-ペンテン、4-エチル-1-ヘキセン、3-エチル-1-ヘキセンの単独重合体、あるいはそれら相互の共重合体、さらにはそれらと他のα-オレフィンとの共重合体を挙げることができる。透明性、低温耐衝撃性、剛性の特性が良好であること、および経済性の観点から、特に、3-メチル-1-ブテンの重合体が好ましい。
無機化合物としては例えば、タルク、マイカ、炭酸カルシウムを用いることができる。
本発明に用いる造核剤としては、上記の通り、一部は市販品として容易に入手することができる。
またこれらの造核剤は一種単独でも、二種以上を用いてもよい。
本発明の医療用プロピレン-エチレン系樹脂組成物は、耐候安定剤を含有する。
耐候安定剤の具体例としては、ビス(2,6-ジ-t-ブチル-4-メチルフェニル)ペンタエリスリトール-ジ-フォスファイト、ジ-ステアリル-ペンタエリスリトール-ジ-フォスファイト、ビス(2,4-ジ-t-ブチルフェニル)ペンタエリスリトール-ジ-フォスファイト、トリス(2,4-ジ-t-ブチルフェニル)フォスファイト、テトラキス(2,4-ジ-t-ブチルフェニル)-4,4’-ビフェニレン-ジ-フォスフォナイト、テトラキス(2,4-ジ-t-ブチル-5-メチルフェニル)-4,4’-ビフェニレン-ジ-フォスフォナイト等のリン系酸化防止剤、n-ヘキサデシル-3,5-ジ-t-ブチル-4-ヒドロキシベンゾエート、2,4-ジ-t-ブチルフェニル-3’,5’-ジ-t-ブチル-4’-ヒドロキシベンゾエート、ビス(2,2,6,6-テトラメチル-4-ピペリジル)セバケート、コハク酸ジメチル-2-(4-ヒドロキシ-2,2,6,6-テトラメチル-1-ピペリジル)エタノール縮合物、ポリ{[6-〔(1,1,3,3-テトラメチルブチル)アミノ〕-1,3,5-トリアジン-2,4ジイル]〔(2,2,6,6-テトラメチル-4-ピペリジル)イミノ〕ヘキサメチレン〔(2,2,6,6-テトラメチル-4-ピペリジル)イミノ〕}、N,N’-ビス(3-アミノプロピル)エチレンジアミン-2,4-ビス〔N-ブチル-N-(1,2,2,6,6-ペンタメチル-4-ピペリジル)アミノ〕-6-クロロ-1,3,5-トリアジン縮合物等のヒンダートアミン系安定剤、2,6-ジ-t-ブチル-p-クレゾール、テトラキス[メチレン(3,5-ジ-t-ブチル-4-ヒドロキシヒドロシンナメート)]メタン、1,3,5-トリメチル-2,4,6-トリス(3,5-ジ-t-ブチル-4-ハイドロキシベンジル)ベンゼン、トリス(3,5-ジ-t-ブチル-4-ハイドロキシベンジル)イソシアヌレート等のフェノール系酸化防止剤、ジ-ステアリル-β,β’-チオ-ジ-プロピオネート、ジ-ミリスチル-β,β’-チオ-ジ-プロピオネート、ジ-ラウリル-β,β’-チオ-ジ-プロピオネート等のチオ系酸化防止剤等が挙げられる。
なお、耐候安定剤は単独、又は複数用いても構わないが、本発明に係るプロピレン-エチレン系樹脂組成物は、放射線滅菌を行う医療用途向け射出成形品に使用されるため、滅菌後の変色の観点からリン系酸化防止剤又はヒンダートアミン系安定剤を配合することが好ましい。リン系酸化防止剤の中でも特にトリス(2,4-ジ-t-ブチルフェニル)フォスファイトが放射線滅菌時の樹脂劣化の抑制と変色のバランスに優れている為、好ましい。ヒンダートアミン系安定剤の中でも低溶出の観点から高分子量型ヒンダートアミン系安定剤が好ましく、特にコハク酸ジメチル-2-(4-ヒドロキシ-2,2,6,6-テトラメチル-1-ピペリジル)エタノール縮合物が放射線滅菌時の樹脂劣化の抑制と滅菌後の成形品の長期安定性、及び変色のバランスに優れており、かつ、低溶出性であり、塩基性もヒンダートアミン系安定剤の中では弱く中性に近い為、内容液への影響が少なく好ましい。放射線滅菌時の樹脂劣化抑制及び滅菌後の成形品の長期安定性保持の観点からリン系酸化防止剤とヒンダートアミン系酸化防止剤を併用することが最も好ましい。
さらに、本発明の医療用プロピレン-エチレン系樹脂組成物には性能を損なわない範疇でその他に、公知の銅害防止剤、紫外線吸収剤、帯電防止剤、親水化剤、スリップ剤、抗ブロッキング剤、防曇剤、着色剤、充填剤、石油樹脂、抗菌剤などを含有することができる。
本発明の医療用プロピレン-エチレン系樹脂組成物は、前述のプロピレン-エチレン共重合体(a)、プロピレン-エチレン共重合体(b)、エラストマー(B)、造核剤、中和剤、滑剤、酸化防止剤、及びその他添加剤等の各種配合成分の所定量を、例えばヘンシェルミキサー(商品名)、スーパーミキサー、リボンブレンダー、タンブラーミキサー、バンバリーミキサー、等の通常の混合装置を用いて混合することによって得ることができる。得られた混合物を、単軸押出機、二軸押出機、バンバリーミキサー、プラベンダー、ロールなどを用いて、溶融混練温度150~300℃、好ましくは180~250℃で溶融混練してペレタイズすることによって、ペレット状の組成物とすることもできる。
本発明の成形品は、上記の医療用プロピレン-エチレン系樹脂組成物を、公知の射出成形機により成形することで得られる医療用途向け射出成形品である。
また、本樹脂組成物を用いた成形品は、透明性の観点から、成形品の平均肉厚が3.0mm厚以下が好ましく、より好ましくは2.5mm厚以下であり、さらに好ましくは2.0mm厚以下であり、特に好ましくは1.5mmであり、より特に好ましくは1.2mm厚以下であり、最も好ましくは1.0mm以下である。上限厚以下であると十分な透明性を発現する。またここで言う成形品の平均肉厚とは、成形品の全表面積に占める割合で最も広範囲の部分の肉厚のことを指す。代表例として注射器(シリンジ又は筒状部をバレルという)では外筒(バレル)の円筒部分の肉厚のことを指す。
さらに、本樹脂組成物は、製品剛性と耐衝撃性のバランスが良好な射出成形品が得られる。なお、本樹脂組成物は、薬発第494号 透析型人工腎臓装置承認基準 IV血液回路の品質及び試験法に記載の重金属試験、鉛試験、カドミウム試験、溶出物試験を放射線滅菌後に満足するため人工透析用部材に好適であり、特にダイアライザーのハウジングやヘッダー、及びその関連部材に好適であり、JIS T3210:2011 滅菌済み注射筒に記載の6化学的要求事項を満足する為、注射器用部材に好適であり、特にディスポーザブル注射器に好適である。
(1)固体触媒成分の調製
無水塩化マグネシウム95.2g、デカン442mLおよび2-エチルヘキシルアルコール390.6gを130℃で2時間加熱反応を行って均一溶液とした後、この溶液中に無水フタル酸21.3gを添加し、さらに130℃にて1時間攪拌混合を行い、無水フタル酸を溶解させた。
このようにして得られた均一溶液を室温に冷却した後、-20℃に保持した四塩化チタン200mL中に、この均一溶液の75mLを1時間にわたって滴下装入した。装入終了後、この混合液の温度を4時間かけて110℃に昇温し、110℃に達したところでフタル酸ジイソブチル(DIBP)5.22gを添加し、これより2時間同温度にて攪拌保持した。
2時間の反応終了後、熱濾過にて固体部を採取し、この固体部を275mLの四塩化チタンに再懸濁させた後、再び110℃で2時間、加熱した。反応終了後、再び熱濾過にて固体部を採取し、110℃のデカンおよびヘキサンにて溶液中に遊離のチタン化合物が検出されなくなるまで充分洗浄した。
この吸収が観測されなくなるまで遊離チタンの洗浄除去および遊離チタンの検出を行った。
上記のように調製された固体状チタン触媒成分(A)は、デカンスラリーとして保存したが、この内の一部は触媒組成を調べる目的で乾燥した。このようにして得られた固体状チタン触媒成分(A)の組成は、チタン2.3質量%、塩素61質量%、マグネシウム19質量%、DIBP 12.5質量%であった。
内容積500mLの攪拌機付きの三つ口フラスコを窒素ガスで置換した後、脱水処理したヘプタンを400mL、トリエチルアルミニウム19.2mmol、ジシクロペンチルジメトキシシラン3.8mmol、上記固体状チタン触媒成分(A)4gを加えた。内温を20℃に保持し、攪拌しながらプロピレンを導入した。1時間後、攪拌を停止し結果的に固体状チタン触媒成分(A)1g当たり2gのプロピレンが重合した予備重合触媒成分(B)を得た。

PP成分(a)のMFRは45g/10分、エチレン含量は1.8質量%であった。
PP成分(a)の重合終了後(前記[工程1]の後)、内温を30℃まで降温し脱圧した。その後、水素0.94MPa-G装入し、続いてプロピレン/エチレン:(4.3L/min)/(1.1L/min)の混合ガスを導入した。内温60℃、全圧0.30MPa-G(導入ガス量により変動)で50分間プロピレン/エチレン共重合を行った。
所定時間経過したところで50mLのメタノールを添加し反応を停止し、降温、脱圧した。内容物を全量フィルター付きろ過槽へ移し60℃に昇温し固液分離した。更に、60℃のヘプタン6Lで固体部を2回洗浄した。このようにして得られたプロピレン/エチレン共重合体を真空乾燥した。
2段目で製造したPP成分(b)についてのインデックスを計算したところ、生産量は全体の重量に対し7質量%であり、MFRは7.0g/10分、エチレン含量は18.0質量%であった。

所定時間経過したところで50mLのメタノールを添加し反応を停止し、降温、脱圧した。内容物を全量フィルター付きろ過槽へ移し60℃に昇温し固液分離した。更に、60℃のヘプタン6Lで固体部を2回洗浄した。このようにして得られたプロピレン/エチレン共重合体を真空乾燥した。

所定時間経過したところで50mLのメタノールを添加し反応を停止し、降温、脱圧した。内容物を全量フィルター付きろ過槽へ移し60℃に昇温し固液分離した。更に、60℃のヘプタン6Lで固体部を2回洗浄した。このようにして得られたプロピレン/エチレン共重合体を真空乾燥した。
2段目で製造したPP成分(b)についてのインデックスを計算したところ、生産量は全体の重量に対し7質量%であり、MFRは8.0g/10分、エチレン含量は9.0質量%であった。
エラストマー(B)として、以下の(B-1)~(B-3)のエチレン-αオレフィン共重合体を用いた。
(B-1) メタロセン系エチレン-αオレフィン共重合体:密度(JIS K 7112に準拠して測定した。以下、密度と略称することがある。)903kg/m3、MFR(190℃)15g/10分(プライムポリマー製、商品名:SP00206)
(B-2) メタロセン系エチレン-αオレフィン共重合体:密度883kg/m3、MFR(190℃)20g/10分(三井化学製、商品名:A-2085S)
(B-3) メタロセン系エチレン-αオレフィン共重合体:密度913kg/m3、MFR(190℃)4.0g/10分(プライムポリマー製、商品名:SP1540)
<造粒・成形>
(1)造粒
表(実施例・比較例)に示す配合にて、プロピレン系重合体(A)と、エラストマー(B)、耐候安定剤としてコハク酸ジメチル-2-(4-ヒドロキシ-2,2,6,6-テトラメチル-1-ピペリジル)エタノール縮合物(「TINUVIN622」(商品名)、BASF社製)、造核剤としてノニトール 1,2,3-トリデオキシ-4,6:5,7-ビス-O-[(4-プロピルフェニル)メチレン](「ミラードNX8000J」(商品名)、ミリケン社製)または(C-1):リン酸-2,2’-メチレンビス(4,6-ジ-tert-ブチルフェニル)アルミニウム塩(「アデカスタブNA-21」(商品名)、ADEKA社製)を所定の量を配合し、さらに添加剤として、リン系酸化防止剤のトリス(2,4-ジ-t-ブチルフェニル)フォスファイト(「Irgafos168」(商品名)BASF社製)を0.13質量部、中和剤のステアリン酸カルシウム(日東化成社製)を0.10質量部、をヘンシェルミキサーにて攪拌混合した。
得られた混合物を東芝機械株式会社製の二軸押出機(TEM35BS)を用いて下記条件にて溶融混練してストランドを得た。
・型式:TEM35BS(35mm二軸押出機)
・スクリュー回転数:300rpm
・スクリーンメッシュ:#200
・樹脂温度:230℃
得られたストランドを水冷後ペレタイザーにて切断することにより、プロピレン系樹脂組成物のペレットを得た。
プロピレン系樹脂組成物のペレットを用いて、以下の方法で容器を成形した。
型締め力140トンの電動射出成形機(日精樹脂工業社製NEX140IV)を用いて、シリンダー温度230℃、金型温度25℃、射出1次圧力130MPa、射出速度50mm/sec、保圧圧力、150MPa、保圧時間5.0secの条件で、プロピレン系樹脂組成物のペレットを射出成形し、高さ80mm、直径16mm、側面肉厚1.0mmの10mLシリンジを射出成形した。
上記成形条件における連続成形において、100shot間離型不良、シリンジの変形、エラストマー樹脂の配向による容器流れ方向破損等のトラブルゼロで成形可能となる最少サイクルタイムを測定した。
(4)MFR
本発明のプロピレン-エチレン共重合体(a)及び(b)、プロピレン-エチレン系樹脂組成物(A)のメルトフローレイトMFRは、JISK-7210-1999(230℃、2.16kg荷重)、エラストマー(B)のMFRはJISK-7210-1999(190℃、2.16kg荷重)に準拠して測定した。

測定モード:BCM(Bilevel Complete decoupling)
観測周波数:100.4MHz
観測範囲:17006.8Hz
パルス幅:C核45°(7.8μ秒)
パルス繰り返し時間:5秒
試料管:5mmφ試料管
回転数:12Hz
積算回数:20000回
測定温度:125℃
溶媒:1,2,4-トリクロロベンゼン:0.35ml/重ベンゼン:0.2ml
試料量:約40mg

射出成形法により試験片を成形し、成形後に室温23±5℃、相対湿度50±5%に調節された恒温室に72時間放置した後、JIS K 7161(ISO178)に準拠して求めた。
また、放射線滅菌後の引張弾性率は、10mLシリンジ成形後に室温23±5℃、相対湿度50±5%に調節された恒温室に72時間放置した後、空気雰囲気下、室温条件下で、γ線25kGy(平均線量)を照射した後、更に室温23℃±0.5℃、相対湿度50±5%の恒温室内で2週間状態調整した後に測定し求めた。
10mLシリンジを48~72時間23℃条件下で状態調整を行い、更に10℃の環境下で24時間以上状態調整を行った。
10℃環境下で、シリンジの胴部中心に対して、鉄棒(13.5mmΦ、120g)を垂直に落下させ、10回落下させたときに割れが発生しない最大の高さを測定した。
また、放射線滅菌後の落錘試験は、10mLシリンジ成形後に室温23±5℃、相対湿度50±5%に調節された恒温室に72時間放置した後、空気雰囲気下、室温条件下で、γ線25kGy(平均線量)を照射した後、更に室温23℃±0.5℃、相対湿度50±5%の恒温室内で2週間状態調整した後に測定し求めた。
第十七改正日本薬局方試験 7.02 プラスチック製医薬品容器試験法 1.4.透明性試験を参考に、下記方法にて試験を実施した。
容器の胴部の高さ40mm付近から、約0.9×4cmの大きさに切断したものを5個作り、それぞれ水を満たした紫外線吸収スペクトル測定用セルに浸し、水だけを満たしたセルを対象として、紫外可視吸光度測定法により波長450nmの透過率を測定し求めた。
また、放射線滅菌後の水中光線透過率は、10mLシリンジ成形後に室温23±5℃、相対湿度50±5%に調節された恒温室に72時間放置した後、空気雰囲気下、室温条件下で、γ線25kGy(平均線量)を照射した後、更に室温23℃±0.5℃、相対湿度50±5%の恒温室内で2週間状態調整した後に測定し求めた。
(a)100mm×120mm×1mmtプレスシートの作製
150mm×150mm×3mmtのアルミ板の間に100mm×120mm×1mmtのプレスシートが得られるスペーサーを置き、そのスペーサーの枠内に規定量のペレットを入れた。その後、230℃に加熱した加熱プレスを用いて、始めの7分間は圧力をかけずにペレットを加熱プレス機内で溶かし、その後、100kg/cm2の圧力を3分間かけた。その後、30℃の冷却プレスに速やかに移し、150kg/cm2の圧力を2分間かけてサンプルの冷却を行った。その後、プレスシートをアルミ板、及びスペーサーから剥がして取り出した。
(b)溶出物試験用の試験片の作製
(a)で作製したプレスシートを鋏で均等に4分割して60mm×50mm×2mmtのシートを4枚回収した。その後、蒸留水にてシート表面、及び、切断面の洗浄を行い、常温にて乾燥して溶出物試験用の試験片とした。
(c)試験片の放射線滅菌
試験片に空気雰囲気下、室温条件下で、γ線25kGy(平均線量)を照射した後、室温23℃±0.5℃、相対湿度50±5%の恒温室内で2週間状態調整した。
(d)試験液の調製
蒸留水で洗浄して室温にて乾燥させた500mlのホウケイ酸製のガラスビーカーに、蒸留水を250ml入れた。そこに、(C)で準備した溶出物試験用の試験片(60mm×50mm×2mmt)を4枚入れて水中に浸漬させた。その際、試験片表面に気泡が残らないようにした。そして、アルミ箔にてビーカーを密閉して、恒温槽中で37℃、8時間保った後、試験片を取り出して試験液とした。
(e)pH試験、溶出金属の試験
JIS T3210:2011に記載の方法に準拠して試験を実施した。なお、空試験液は蒸留水を用い、溶出金属は原子吸光光度法によって分析した。
各試験結果の基準は、以下の通りである。適否について評価した。
(i)ΔpH:試験液と空試験液のpHの差は1以下
(ii)溶出金属:鉛,亜鉛,鉄の合計は5mg/L以下で、かつ、試験液のカドミウム測定値を空試験液のカドミウム測定値で補正したとき,試験液のカドミウム含量は 0.1 mg/L 以下。
現在、「薬発第494号 透析型人工腎臓装置承認基準」は「通知の廃止」となっているが、本試験が本用途での化学的安全性確認の目安となっている為、試験を実施した。
重金属試験、鉛試験、カドミウム試験(まとめて灰化試験という)はペレットを用いて、承認基準の操作方法に準拠して試験を行った。なお、試験に用いたペレットは本試験を行う2週間前にγ線25kGy(平均値)の照射滅菌を行い、室温23℃±0.5℃、相対湿度50±5%の恒温室内で2週間状態調整したものである。
また、溶出物試験は、本承認基準内のV透析器の品質及び試験法 5.支持体及び血液接続管に記載の溶出物試験を参考にして、ペレット15gに対して水150mlを加えた後、70℃で1時間の抽出試験を行い、各試験については承認基準の操作方法に準拠して試験を行った。なお、試験に用いたペレットは本試験を行う2週間前にγ線25kGy(平均値)の照射滅菌を行ない、室温23℃±0.5℃、相対湿度50±5%の恒温室内で2週間状態調整したものである。
各試験結果の基準は、以下の通りである。適否について評価した。
4.重金属試験:10μg/g以下
5.鉛試験:1μg/g以下
6.カドミウム試験:1μg/g以下
8.溶出物試験
(i)外観:無色透明、異物なし
(ii)あわだち:3分以内に消失
(iii)ΔpH:ブランクとの差が1.5以下
(iv)亜鉛:標準溶液(0.5μg/g)以下
(v)過マンガン酸カリウム(KMnO4)還元性物質:標準溶液との過マンガン酸カリウム消費量の差1.0ml以下
(vi)蒸発残留物:1.0mg以下
(vii)紫外吸収スペクトル(UV)220~350nm:0.1以下
*)現在、「薬発第494号 透析型人工腎臓装置承認基準」は「通知の廃止」となっているが、本試験が本用途での化学的安全性確認の目安となっている為、試験を実施する。
Claims (3)
- エチレン含量が1~5質量%、JIS K7210(230℃、2.16kg荷重)に準拠したメルトフローレイト(以下、MFRと略称することがある。)が10~100g/10分であるプロピレン-エチレン共重合体(a)の90質量%超97質量%以下と、エチレン含量が15~22質量%、MFRが1~50g/10分であるプロピレン-エチレン共重合体(b)3質量%以上10質量%未満とからなるプロピレン-エチレン系樹脂組成物(A)(ただし、(a)及び(b)の合計は100質量%である)88~95質量部、及び密度0.880~0.920g/cm3のエチレン-α-オレフィンランダム共重合体であるエラストマー(B)5~12質量部(ただし、(A)及び(B)の合計は100質量部である)、および耐候安定剤を0.01~0.20質量部を含む医療用プロピレン-エチレン系樹脂組成物を用いて得られる射出成形品であって、
該射出成形品はγ線又は電子線滅菌されたものである医療用途向け射出成形品。 - 前記エラストマー(B)は、メタロセン触媒を用いて重合され、JIS K7210(190℃、2.16kg荷重)に準拠したMFRが1~100g/10分であるエチレン-α-オレフィンランダム共重合体である請求項1に記載の医療用途向け射出成形品。
- 造核剤を含有する請求項1又は2に記載の医療用途向け射出成形品。
