Abstract
Description
アレクニチブは難溶性の塩基性薬剤であるため、その製剤化において、溶解補助剤を共存させた組成物とすること(特許文献1)や、式(I)で示される化合物又はその塩を含む顆粒を形成し、該顆粒を崩壊剤と共存させることにより、溶出性の良好なカプセル剤とすること(特許文献2)が報告されている。
ラウリル硫酸ナトリウムはアレクニチブ製剤をはじめとする種々の医薬品製剤の添加剤として用いられている(非特許文献1)。ラウリル硫酸ナトリウムを含有する製剤は、安定性に乏しく、また劣化したラウリル硫酸ナトリウムを含有する製剤は、製剤特性に変化が生ずることが知られており、ラウリル硫酸ナトリウムを含有する製剤を安定化するために中性塩及び塩基性物質からなる群より選ばれた少なくとも1種を配合することが知られている(特許文献3)。
しかしながら、医薬用製剤原料として用いられるラウリル硫酸ナトリウムのわずかな品質の違いが、製剤の劣化に影響すること、及びラウリル硫酸ナトリウムのわずかな品質の違いを検出する方法は、これまで知られていなかった。
すなわち、本発明は、以下のラウリル硫酸ナトリウムからなる医薬用製剤原料の選別方法に関する。
<1> ラウリル硫酸ナトリウムからなる医薬用製剤原料の選別方法であって、前記製剤原料を加速条件で前処理後、不純物の検出を行うことを特徴とする、前記選別方法。
<2> 前記加速条件が、65~75℃の温度条件であることを特徴とする、<1>に記載の選別方法。
<3> 前記加速条件が、79%RHを下回る湿度条件であることを特徴とする、<1>または<2>に記載の選別方法。
<4> 前記加速条件が、約10%RHの湿度条件であることを特徴とする、<1>~<3>のいずれか一項に記載の選別方法。
<5> 前記加速条件が、約70℃の温度条件であり、かつ、約10%RH以下の湿度条件であることを特徴とする、<1>~<4>のいずれか一項に記載の選別方法。
<6> 前記不純物が、硫酸イオン、ドデカノール又はラウリル硫酸ナトリウム・ドデカノール複合体である、<1>~<3>のいずれか一項記載の選別方法。

本発明の選別方法によれば、医薬用製剤原料ラウリル硫酸ナトリウムを所定の加速条件下で前処理することにより、安定性の優れた医薬品製剤を製造することができる医薬用製剤原料として選別することができる。あるいは、本発明の選別方法では、医薬用製剤原料の保存状態により、不純物の検出を省略することができる。
また、本発明の品質評価方法によれば、医薬用製剤原料であるラウリル硫酸ナトリウムを所定の加速条件下で前処理し、不純物量を検出することにより、当該医薬品製剤の品質を評価することができる。あるいは、本発明の品質評価方法によれば、ラウリル硫酸ナトリウムを含む医薬品製剤が、所定の加速条件下で前処理し、不純物量により選別された医薬用製剤原料であるラウリル硫酸ナトリウムを用いて製造されたか否かにより、医薬品製剤の不純物の測定を省略可能な品質であると評価することができる。
さらにまた、本発明の製造方法によれば、ラウリル硫酸ナトリウムの医薬用製剤原料を所定の加速条件下で前処理することにより不純物の低い製剤原料を選別し、当該選別された医薬用製剤原料を用いて医薬品製剤を製造することにより、ラウリル硫酸ナトリウムを含む安定性に優れた医薬品製剤を製造することができる。
また、本発明によれば、安定性に優れた医薬品製剤の製造に有用な医薬用製剤原料、あるいは、溶解性に影響を与えない、所定割合以下で不純物を含有するアレクチニブ含有組成物または医薬品製剤を提供することができる。
本発明において、「ラウリル硫酸ナトリウム」は、医薬品製剤の添加物として公知の物質であり、特に、難溶性医薬であるアレクチニブの溶解補助剤として当該医薬品製剤の医薬組成物を構成する物質である。ラウリル硫酸ナトリウムには、1水和物、1/2水和物、1/8水和物、および非溶媒和物が存在することが知られており(文献名:Journal of Crystal Growth 263 (2004) 480-490)、通常は室温で保存され、製造から1年以内に使用するのが好ましい。いずれの形態も本発明の医薬用製剤原料として使用することができる。
「医薬用製剤原料」とは、アレクチニブなどの有効成分を含む医薬品製剤の添加物として用いられる実質的に純粋な単一化合物(例えば、ラウリル硫酸ナトリウムなど)である。医薬用製剤原料は、当該医薬品製剤の工業的製造に用いるための、キログラムオーダー以上の量で所望の化合物を含むものをいう。実質的に純粋とは、ある程度の不純物が存在することを許容するが、所望の化合物を、一般的に流通可能な程度に含むことをいう。本発明において、医薬用製剤原料は、製造業者から市販のものを購入してもよく、自己であるいは他社に委託して製造したものでもよい。また、医薬用製剤原料は、購入または製造直後のものを用いても、あるいは購入/製造後所定の期間、例えば1年以上経過したものを用いてもよい。
なお、「ラウリル硫酸ナトリウムからなる医薬用製剤原料」は、実質的に純粋なラウリル硫酸ナトリウムを含む医薬用製剤原料を意味し、その不純物である、微量の硫酸イオン、ドデカノール又はラウリル硫酸ナトリウム・ドデカノール複合体が存在していてもよい。
イオンクロマトグラフィーは、主としてイオン種成分を測定する液体クロマトグラフィーの一種であり、溶離液を移動相として、イオン交換体などを固定相とした充填カラム内で試料溶液中のイオン種成分を分離し、定量することができる。ガスクロマトグラフィーは、固定相と物質の相互作用で分離を行い、検出器までの到達時間で定性、検出器からの信号量で定量することができる。粉末X線回折による測定は、例えば、日本薬局方(第十七改正)に記載されている「粉末X線回折測定法」などの常法に従って行うことができる。
定量NMRは、NMRスペクトル上に観測されるシグナルピークの面積が、試料に含まれる水素原子の数に比例し、物質の化学構造に依存しないという原理を用いて定量を行う方法である。したがって、分析対象成分以外の純度既知の物質を標準物質として使用することにより、分析対象成分の絶対定量が可能である。好ましくは、ガスクロマトグラフィー、または陰イオン交換カラムを用いるイオンクロマトグラフィーであり、イオンクロマトグラフィーの好ましい移動相、カラム、検出器は、以下のとおりである。

「加速条件」とは、通常の保存状態よりも、過酷な条件を意味し、具体的には温度や湿度などが、通常の保存状態よりも過酷な条件である。具体的には、医薬用製剤原料についての温度の加速条件は、50℃以上、好ましくは約60℃以上、より好ましくは、約60℃~80℃、さらに好ましくは約65℃~75℃、最も好ましくは約70℃の温度条件をいう。当該加速条件で用いられる湿度条件は、80%RH以下、好ましくは、79%RHを下回るか、より好ましくは約10%RHを下回る湿度条件である。
また、医薬用製剤原料が一定の条件下で保存されていれば、生じる不純物の割合を低く抑えることができるため、医薬用製剤原料が当該条件下で保存された当該製剤原料を含む医薬品製剤を選択すれば、不純物の測定を省略することができる。
医薬品製剤についての温度の加速条件は、50℃以上、好ましくは約50℃~70℃以上、より好ましくは、約55℃~65℃、最も好ましくは約60℃の温度条件をいう。
なお、「保存」とは、医薬用製剤原料が、製造されてから使用されるまでの間、温度や湿度が管理できる倉庫、保管庫、貨物内等で、一定期間取り置かれること(保管)を意味し、1日以上を要する輸送も含む。
本発明において、「所定の条件」は、保存期間中の温度や日数を意味し、具体的には、温度及び日数については、好ましくは40℃以下で47日間以下、より好ましくは30℃~40℃で47日間以下、最も好ましくは30℃~40℃で45日間以下である。
「所定の条件を超える」とは、温度についての条件の場合、保存中に40℃より高い温度で保存されることをいう。ただし、当該温度条件を超えた場合であっても、所定の範囲以内であれば、不純物の生成に対する影響は少ないため、不純物の測定を省略できる。具体的には、40℃を超えた場合は、40℃超60℃以下の時間が積算して48時間以内であれば、不純物の測定を省略できる。
また、日数についての条件の場合、「所定の条件を超える」とは、40℃以下の温度で47日間、好ましくは45日間を超えて保存されることをいう。ただし、保存温度が室温~30℃で1年間以下の期間で保存されたものについても、同様に不純物の測定を省略できる。
このように、不純物の測定を省略できる条件(追加条件)を超えて保管される場合にも、「所定の条件を超える」に該当する。
なお、本願明細書中において、「約」とは、±10%を許容する意味で用いる。
「前処理」とは、医薬品製剤の製造に用いる前に医薬用製剤原料に適用する、所定の処理をいう。所定の処理とは、具体的には、前記加速条件下で所定の期間医薬用製剤原料を静置することをいう。所定の期間とは、医薬用製剤原料については1日間以上をいい、好ましくは2日間以上、より好ましくは、3日間以上、さらに好ましくは4日間である。医薬品製剤については、5日間以上、好ましくは7日間以上、さらに好ましくは7日間以上14日間以内、最も好ましくは、7日間以内である。
「検出」とは、医薬品製剤または医薬用製剤原料中の各ロットもしくは包装単位に含まれる原料以外の物質(不純物)を定性的、または定量的に確認することをいう。例えば、医薬用製剤原料がラウリル硫酸ナトリウムであるとき、あるいはラウリル硫酸ナトリウムを含む医薬品製剤は、不純物である、硫酸イオン、ドデカノール又はラウリル硫酸ナトリウム・ドデカノール複合体の有無を、「選別」で用いられる方法と同様に、粉末X線回折、イオン交換クロマトグラフィー、ガスクロマトグラフィー又は定量NMRなどを用いることにより、定性的または定量的に確認することをいう。
「医薬組成物」とは、疾患の治療・予防などに用いられる、生理活性物質を有効成分として含み、さらに薬学的に許容可能な添加剤を含む、2以上の物質の混合物を意味する。本発明における生理活性物質としては、好ましくは、式(I)で示される化合物またはその塩である。
式(I)で示される化合物の「塩」は、薬学的に許容される塩が好ましく、「薬学的に許容される塩」としては、例えば、塩酸塩、臭化水素酸塩、ヨウ化水素酸塩、リン酸塩、ホスホン酸塩、硫酸塩、メタンスルホン酸塩、p-トルエンスルホン酸塩などのスルホン酸塩;酢酸塩、クエン酸塩、リンゴ酸塩、酒石酸塩、コハク酸塩、サリチル酸塩などのカルボン酸塩;ナトリウム塩、カリウム塩などのアルカリ金属塩;マグネシウム塩、カルシウム塩などのアルカリ土類金属塩;アンモニウム塩(NH4X:Xは1価の酸基)、アルキルアンモニウム塩、ジアルキルアンモニウム塩、トリアルキルアンモニウム塩、テトラアルキルアンモニウム塩などのアンモニウム塩などが含まれる。
好ましくは塩酸塩であり、最も好ましくは一塩酸塩である。
式(I)で示される化合物又はその塩は、公知の方法(例えば、特許文献2に記載の方法)により製造することができる。
式(I)で示される化合物の一塩酸塩は、非晶質であっても結晶であってもよく、結晶の場合は、粉末X線回折パターンにおいて8.4°、14.0°、16.7°、18.8°、23.3°付近の回折角(2θ)にピークを有する結晶(I型結晶)が好ましい。また、式(I)で示される化合物の一塩酸塩は、水和物であってもよい。式(I)で示される化合物の一塩酸塩の非晶質体は、WO2016/021707に記載の方法により、当該ピークを有する結晶は、WO2015/163447に記載の方法により製造することができる。
式(I)で示される化合物又はその塩は、組成物全体に対し、フリー体に換算して20~70重量%、好ましくは35~60重量%、さらに好ましくは45~50重量%含有する。
本発明の製造方法により得られる製剤は、式(I)で示される化合物又はその塩を、単位製剤あたり、フリー体に換算して150mg~800mg、好ましくは150mg~400mg、特に好ましくは、150mg~300mg含有する。具体的には、150mgカプセル製剤、150mg、300mg、600mg錠剤である。
同一の製造業者から入手したラウリル硫酸ナトリウムの製造日の異なるロット(Lot1(製造日:2015.12.22)とLot2(製造日:2017.3.10))適量をガラスバイアルに入れ、密閉系にて条件1A:温度60℃、条件1B:温度70℃にて保管した。保管前、3日間、4日間保管後のサンプルについて粉末X線回折(XRPD)を下記に示す条件より測定した。

Lot1,Lot2の条件1Aと条件1B保管時の粉末X線回折パターンを図1に示す。Lot2では条件1A、条件1BともにXRPDパターンは4日間保管後まで変化がなかった。
一方、Lot1では、条件1Aでは4日間保管後に、条件1Bでは3日間以降に4.9°付近と7.4°付近の回折角(2θ)に新たなピークが観測されていた。
条件1A及び条件1BにてLot1およびLot2のXRPDにおいて新たなピークが初めて検出された時点を以下に示す。

したがって、温度の高い条件1Bがより早くラウリル硫酸ナトリウムLot1とLot2を評価可能な条件と判断し、実施例2の湿度の検討に進んだ。
実施例1で用いたラウリル硫酸ナトリウムの異なるロット(Lot1とLot2)約250mgをガラスバイアルに入れ、開放系にて条件2A:温度70℃、相対湿度約79%(NaCl飽和塩水溶液)、条件2B:温度70℃、相対湿度約10%(LiCl飽和塩水溶液)にて保管した。条件2Aでは保管前、1日間、2日間、3日間保管後*1、条件2Bでは保管前、1日間、2日間、3日間、6日間保管後のサンプルについて粉末X線回折を実施例1に示す条件より測定した。
*1:条件2Aの2日間、3日間保管後のLot1は液状になっていたため粉末X線回折の測定を実施しなかった。
条件2A及び条件2BにてLot1およびLot2のXRPDにおいて新たなピークが初めて検出された時点を示す。

以上の結果から、相対湿度の低い条件2Bがよりラウリル硫酸ナトリウムLot1とLot2を識別可能な条件と判断し、ラウリル硫酸ナトリウムサンプルの選別に用いた。
ラウリル硫酸ナトリウム(約85mg)をドデカノール(約2mL)に加え、30℃に保ちながら約12時間撹拌した。析出した固体を濾取し、白色の結晶を得た。上記の結晶(ラウリル硫酸ナトリウム・ドデカノール複合体)について粉末X線回折分析を実施例1の条件により測定し、測定結果を図3に示す。ラウリル硫酸ナトリウム・ドデカノール複合体において観測された4.9°付近と7.4°付近の回折角(2θ)のピークは、実施例1、実施例2でラウリル硫酸ナトリウムを保管した時に観測されていた未知物質のピークとよく一致した。
また、ラウリル硫酸ナトリウムとラウリル硫酸ナトリウム・ドデカノール複合体について走査型電子顕微鏡で下記条件にて観察した。その結果を図4に示す。ラウリル硫酸ナトリウムが薄板状の結晶であるのに対して、ラウリル硫酸ナトリウム・ドデカノール複合体は形状が異なっていた。

プロトン核磁気共鳴スペクトルのチャートを図5に示す。定量の結果、ラウリル硫酸ナトリウム・ドデカノール複合体中のラウリル硫酸ナトリウムとドデカノールのモル比は1.18:1.00であった。
以上の結果から、ラウリル硫酸ナトリウムが保管中に分解し、分解物の一つであるドデカノールと複合体を形成することが判明した。
ラウリル硫酸ナトリウム約250mgをガラスバイアルに入れ、開放系にて温度70℃、相対湿度約10%(LiCl飽和塩水溶液)で保管した。保管前、1日間、2日間、3日間、4日間、7日間、11日間保管後のサンプルについて粉末X線回折を実施例1に示す条件より測定した。
実施例1で用いたラウリル硫酸ナトリウムの異なるロット(Lot1,Lot2)の11日間保管後までのXRPDパターンを図6に示す。Lot2は11日間保管までにラウリル硫酸ナトリウム・ドデカノール複合体が有するピークが検出されなかったのに対して、Lot1は3日間保管後以降のサンプルに、ラウリル硫酸ナトリウム・ドデカノール複合体を示すピークが検出された。
また、4日間保管後のLot1とLot2についてラウリル硫酸ナトリウムの分解で生じる硫酸イオンを以下に示すイオンクロマトグラフィー法で定量した。
その結果、ラウリル硫酸ナトリウムLot2を用いて調製した試料溶液中の硫酸イオン濃度が1mg/L以下であるのに対して、ラウリル硫酸ナトリウムLot1を用いて調製した試料溶液中の硫酸イオン濃度が24.1mg/Lであり、4日間保管後のラウリル硫酸ナトリウムLot1は分解が進んでいることが判明した。
ラウリル硫酸ナトリウム約5mgを量りとり、水を加えて50mLとした。15分間超音波照射し、メンブランフィルター(DISMIC-25HP PTFE 0.45μm HYDROPHILIC、ADVANTEC社製)でろ過した。洗浄を施した固相抽出カートリッジ(Dionex OnGuard II RP、ThermoFisher Scientific製)にろ液を通し、溶出液を試料溶液とした。別途イオンクロマトグラフィー用硫酸イオン標準液(1g/L,和光純薬工業製)を用いて、硫酸イオン濃度が1mg/L、2mg/L、3mg/L、4mg/L、5mg/Lの溶液を調製し、標準溶液とした。イオンクロマトグラフィーはCompact IC Flex(Metrohm製)を用いて、以下の分析条件により試験を行った。
ガードカラム:Metrosep A Supp 4/5 S-Guard/4.0(Metrohm製)
サプレッサー:MSM,MCS(Metrohm製)
溶出液:5mM Na2CO3溶液
洗浄液:精製水
流速:0.8mL/min
サンプル温度:20℃
カラム温度:45℃
注入量:10ml
測定時間:24min
試料溶液中の硫酸イオン濃度は、標準溶液により作成した検量線に基づいて計算した。
実施例3よりラウリル硫酸ナトリウムはロットによって不純物が存在することが判明した。そこでラウリル硫酸ナトリウムLot1を用いて製剤LotAのアレセンサカプセル(150mg)製剤、および同一製造業者で製造日の異なるラウリル硫酸ナトリウムLot3(製造日:2015.10.02)を用いて製剤LotBのアレセンサカプセル(150mg)製剤を、特許5859712の実施例1に従って製造し、異なる保管条件にて不純物である複合体の存在を検討した。
製剤LotAとLotBをガラスバイアルに入れ、密閉系にて温度条件5A:60℃、条件5B:70℃、条件5C:80℃にて保管した。条件5Aでは保管後4日間、7日間、10日間、14日間、条件5Bでは保管後2日間、3日間、条件5Cでは保管後1日間のサンプルについて粉末X線回折を実施例1に示す条件より測定した。
LotAとLotBの条件5A、5B、5C保管時の粉末X線回折パターンを図7に示す。条件5Aにおいて、ラウリル硫酸ナトリウム・ドデカノール複合体が有するピークはLotBでは10日間保管後までのサンプルでは検出されなかったのに対して、LotAでは7日間保管後のサンプルで検出された。

したがって、条件5A60℃が製剤LotAとLotBの安定性を評価できる条件と判断し、製剤の品質評価に用いた。
ラウリル硫酸ナトリウムの不純物が製剤に与える影響を検討するため、実施例1で用いたラウリル硫酸ナトリウムLot1を用いて製剤LotCの、同一製造業者で製造日の異なるラウリル硫酸ナトリウムLot4(製造日:2016.06.28)を用いて製剤LotDのアレセンサカプセル(150mg)製剤を、実施例5と同様に製造し、それぞれの溶出率を検討した。
製剤LotCと製剤LotDをガラスバイアルに入れ、密閉系にて温度60℃で保管した。保管前及び7日間保管後のサンプルについて下記条件にて溶出試験を実施した結果、LotDの溶出プロファイルが保管前と同等であるのに対して、LotCの溶出率が保管前より顕著に低下した(図8)。
試験液にポリオキシエチレン(10)オクチルフェニルエーテル7%を含む日本薬局方溶出試験第1液900mLを用いて、日本薬局方溶出試験パドル法にて、37℃毎分100回転で試験を行った。
また、保管前と5日間、6日間、7日間、8日間、9日間、12日間、14日間保管後のサンプルについて粉末X線回折を実施例1に示す条件より測定した。その結果、LotDでは14日間保管後までラウリル硫酸ナトリウム・ドデカノール複合体が有するピークが検出されなかったのに対して、LotCでは7日間保管後に検出された(図9)。
さらに、7日間保管後のサンプルについてGC/MSによるドデカノールの定量を下記に示す方法で実施した。
各製剤3カプセル分のカプセル内容物を取り出して混合した。カプセル内容物50mgを量り取り、ヘキサンを加えて25mLとした。30分間超音波照射し、遠沈管に溶液に移し遠心分離を行い、上澄みを試料溶液とした。別途1-ドデカノール(和光純薬工業製)を用いてドデカノール濃度が1g/L、5g/L、10g/L、50g/L、100g/Lの溶液を調製し、標準溶液とした。GC/MSは7850A(Agilent製)を用いて、以下の分析条件により試験を行った。

以上のことから、ラウリル硫酸ナトリウムLot4を用いて製造した製剤LotDは60℃7日間保管においてラウリル硫酸ナトリウムの分解はなく溶出性の変化もないのに対して、ラウリル硫酸ナトリウムLot1を用いて製造した製剤LotCは60℃7日間保管によりラウリル硫酸ナトリウムの分解が確認され、製剤の溶出性も低下することが判明した。したがって、適切な品質のラウリル硫酸ナトリウムの選別を行い、製剤製造を行うことで、より高品質な製剤を製造可能であることが見出された。すなわち、Lot1のような、分解されやすいラウリル硫酸ナトリウムロットを除外し、Lot2のような、分解されにくいラウリル硫酸ナトリウムを選別するために、温度70℃、相対湿度約10%の加速条件下で4日間保管後、イオンクロマトグラフィーで検出される硫酸イオン濃度を測定する方法が、より安定性の優れた製剤の製造に用いられる医薬用製剤原料の選別に有効であることがわかった。そして、当該方法により測定される硫酸イオン濃度が、1mg/L以下のラウリル硫酸ナトリウムを、安定性の優れた医薬品製剤を製造することができる医薬用製剤原料として選別することができる。
ラウリル硫酸ナトリウムは通常室温にて保管するが、輸送中など室温より高温条件があり得る場合を想定して保管条件(温度)を設定した。ラウリル硫酸ナトリウムLot5(製造日:2016年10月13日)を用いて一次包装はポリ袋、二次包装はアルミニウム袋とし、表1に示す温度条件と期間で保管した。その後、約250mgをガラスバイアルに入れ、開放系にて温度70℃、相対湿度約10%(LiCl飽和塩水溶液)で4日間保管し、保管後のサンプルについてラウリル硫酸ナトリウムの分解で生じる硫酸イオンを実施例4に示すイオンクロマトグラフィー法で定量した。
製剤の複数ロットを用いて、ラウリル硫酸ナトリウムの不純物であるドデカノールが製剤に検出される量と製剤の溶出性の関係性を調べた。製剤ロットLot E(製造日:2016.08.24), Lot F(製造日:2016.08.29), Lot G(製造日:2016.08.24), Lot H(製造日:2016.10.19), Lot I(製造日:2016.03.29), Lot J(製造日:2016.03.24), Lot K(製造日:2015.07.07)各製剤をガラスバイアルに入れ、密閉系にて温度60℃保管し、保管前及び7日間保管後のサンプルについて溶出試験、7日間保管後のサンプルについてGC/MSによるドデカノールの定量を実施した。溶出試験とGC/MSによるドデカノールの定量は実施例6に示す方法で実施した。
溶出試験の結果、Lot H, Lot I, Lot J, Lot Kの溶出プロファイルが保管前と同等であるのに対して、LotE, Lot F, Lot Gの溶出率が保管前より顕著に低下した(図10A~G)。また、保管後のサンプルのドデカノール定量の結果、Lot H, Lot I, Lot J, Lot K中のドデカノール/ラウリル硫酸ナトリウム(mol/mol%)が2%以下に対して、LotE, Lot F. Lot Gのドデカノール/ラウリル硫酸ナトリウム(mol/mol%)はすべて2%を超える値であった(表2)。
実施例6では製剤においてラウリル硫酸ナトリウムの分解速度が製剤ロットによって異なることが確認されたが、その分解の速度論的特性を確認した。製剤LotAとLotBを用いて、密閉系にて温度60℃保管し、LotAについては保管前、4日間、7日間、8日間、9日間、12日間、14日間保管後、LotBについては4日間、7日間、8日間、9日間、12日間保管後のサンプルについて、実施例6に示すGC/MSによるドデカノールの測定方法でドデカノールの定量を実施した。
その結果を図11に示す。製剤LotAとLotBはドデカノールが検出されるまでの期間が異なり、またドデカノール生成後は加速度的に量が増えることが確認された。
これらは製剤中のラウリル硫酸ナトリウムの分解から硫酸が発生し、その硫酸が触媒になって分解を促進し、その結果、ラウリル硫酸ナトリウムが加速度的に分解されることを反映していると考えられる。
Claims (35)
- ラウリル硫酸ナトリウムからなる医薬用製剤原料の選別方法であって、前記製剤原料を加速条件で前処理後、不純物の検出を行うことを特徴とする、前記選別方法。
- 前記加速条件が、65~75℃の温度条件であることを特徴とする、請求項1または2に記載の方法。
- 前記加速条件が、79%RHを下回る湿度条件であることを特徴とする、請求項1~3のいずれか一項に記載の方法。
- 前記加速条件が、約10%RHの湿度条件であることを特徴とする、請求項1~4のいずれか一項に記載の方法。
- 前記加速条件が、約70℃の温度条件であり、かつ、約10%RH以下の湿度条件であることを特徴とする、請求項1~5のいずれか一項に記載の方法。
- 前記不純物が、硫酸イオン、ドデカノール又はラウリル硫酸ナトリウム・ドデカノール複合体である、請求項1~6のいずれか一項記載の方法。
- 前記不純物を、粉末X線回折、イオン交換クロマトグラフィー、ガスクロマトグラフィー又は定量NMRを用いて検出する、請求項1~7のいずれか一項に記載の方法。
- 前記不純物が硫酸イオンであり、当該不純物を、イオン交換クロマトグラフィーを用いて検出する、請求項1~8のいずれか一項に記載の方法。
- 前記加速条件で、2日間以上前処置する、請求項1~9のいずれか一項に記載の方法。
- 前記加速条件で、4日間前処置する、請求項1~10のいずれか一項に記載の方法。
- 前記条件が、30℃~40℃で47日間以下である、請求項12に記載の選別方法。
- 前記条件が、30℃~40℃で47日間以下であり、40℃を超えた場合は、40℃超60℃以下の時間が積算して48時間以内である、請求項12に記載の選別方法。
- 前記条件が、輸送の際の保存条件である、請求項12~14のいずれか一項に記載の選別方法。
- ラウリル硫酸ナトリウムを含む医薬品製剤を、加速条件で前処理後、不純物を検出することを特徴とする、ラウリル硫酸ナトリウムを含む医薬品製剤の品質評価方法。
- 前記加速条件が、55~65℃の温度条件であることを特徴とする、請求項16または17に記載の品質評価方法。
- 前記加速条件が、約60℃の温度条件であることを特徴とする、請求項16~18のいずれか一項に記載の品質評価方法。
- 前記不純物が、硫酸イオン、ドデカノール又はラウリル硫酸ナトリウム・ドデカノール複合体である、請求項16~19のいずれか一項記載の品質評価方法。
- 前記不純物を、粉末X線回折、イオン交換クロマトグラフィー、ガスクロマトグラフィー又は定量NMRを用いて検出する、請求項16~20のいずれか一項に記載の品質評価方法。
- 前記不純物がドデカノールであり、当該不純物を、ガスクロマトグラフィーを用いて検出する、請求項16~21のいずれか一項に記載の品質評価方法。
- 前記不純物が硫酸イオンであり、当該不純物を、イオン交換クロマトグラフィーを用いて検出する、請求項16~21のいずれか一項に記載の品質評価方法。
- 前記前処理が、前記加速条件で、2日間以上行われる、請求項16~23のいずれか一項に記載の品質評価方法。
- 前記前処理が、前記加速条件で、7日間行われる、請求項16~24のいずれか一項に記載の品質評価方法。
- 前記不純物が、ドデカノール/ラウリル硫酸ナトリウム(mol/mol%)または硫酸イオン/ラウリル硫酸ナトリウム(mol/mol%)として、前記製剤中に2%以下で存在する医薬品製剤を選別する、請求項16~25のいずれか一項に記載の品質評価方法。
- ラウリル硫酸ナトリウムを含む医薬品製剤の製造方法であって、
a 加速条件で医薬用製剤原料であるラウリル硫酸ナトリウムを前処理するステップ、
b 前記ステップaの前処理後に、不純物の検出を行い、不純物である硫酸イオンの含有量が1mg/L以下である医薬用製剤原料を選別するステップ、及び
c 前記ステップbで選別された医薬用製剤原料を用いて医薬品製剤を製造するステップ、
を含む、前記製造方法。 - 前記組成物中のドデカノール/ラウリル硫酸ナトリウム(mol/mol%)または硫酸イオン/ラウリル硫酸ナトリウム(mol/mol%)が、粉末X線回折、イオン交換クロマトグラフィー、ガスクロマトグラフィー又は定量NMRを用いて測定して2%以下である、請求項30に記載の組成物。
- 前記製剤中のドデカノール/ラウリル硫酸ナトリウム(mol/mol%)または硫酸イオン/ラウリル硫酸ナトリウム(mol/mol%)が、粉末X線回折、イオン交換クロマトグラフィー、ガスクロマトグラフィー又は定量NMRを用いて測定して2%以下である、請求項32に記載の製剤。
- ラウリル硫酸ナトリウムを含有する医薬品製剤であって、前記製剤を加速条件で前処理後に測定した前記製剤中のドデカノール/ラウリル硫酸ナトリウムまたは硫酸イオン/ラウリル硫酸ナトリウムを、前記医薬品製剤の溶出性に影響を与えない割合で含む、製剤。
- ラウリル硫酸ナトリウムからなる医薬用製剤原料であって、前記製剤原料を加速条件で前処理後に測定した前記原料中の硫酸イオンが、イオン交換クロマトグラフィーを用いて測定して1mg/L以下である、前記製剤原料。
