Abstract
본 발명은 면역 마이크로버블 복합체 및 이의 이용에 관한 것으로, 본 발명에 따른 면역 마이크로버블 복합체(IMC)는 마이크로버블에 항체가 결합(conjugation)된 것으로, 상기 마이크로버블은 안정성이 뛰어나고, 항체의 결합력이 뛰어나며, 상기 면역 마이크로버블 복합체에 고강도 집중초음파(HIFU)를 처리할 경우 상기 HIFU 처리에 의해 항종양 효과가 유의적으로 증가하고 면역 증강 효과가 나타나는 것을 확인하였다. 이에, 본 발명에 따른 면역 마이크로버블 복합체는 결합된 항체의 전달 효율을 증가시키고 암의 진단과 동시에 치료에 이용될 수 있으며, 초음파를 통하여 조영 효과, 반감기 개선, 개선된 약물전달, 림프구 밀집효과, 면역항암치료 및 면역치료유도 등 면역치료 분야에서 여러 가지 기능을 나타낼 것으로 기대된다.
Description
본 발명은 마이크로버블; 및 상기 마이크로버블의 표면에 결합된 면역 체크포인트 저해(checkpoint inhibition)능 항체를 포함하는 면역 마이크로버블 복합체 및 이의 이용에 관한 것이다.
본 발명은 2019년 09월 02일에 출원된 한국특허출원 제10-2019-0108278호에 기초한 우선권을 주장하며, 해당 출원의 명세서 및 도면에 개시된 모든 내용은 본 출원에 원용된다.
암은 인류가 해결해야 할 난치병 중의 하나로, 전 세계적으로 이를 치유하기 위한 개발에 막대한 자본이 투자되고 있는 실정이며, 우리나라의 경우, 질병 사망원인 중 제 1위의 질병으로서 연간 약 10만 명 이상이 진단되고, 약 6만 명 이상이 사망하고 있다.
현재 임상학적으로 사용하고 있는 암 치료 방법은 화학 요법, 방사선, 표적 치료, 및 수술로서 병소를 절제하는 방법 등이 있으며, 현재까지 사용되고 있는 항암치료를 위한 화학요법제의 대표적인 예는 독소루비신(Doxorubicin) 또는 아드리아마이신(Adriamycin), 시스플라틴(Cisplatin), 탁솔(Taxol), 5-fluorouracil 등이 있으며 암치료를 위한 화학치료법으로 광범위하게 사용되고 있다. 그러나, 상기 방법들은 한계를 지니고 있으며, 완치하는 경우보다는 환자에게 심한 부작용 및 고통을 수반하고 있어, 부작용을 최소화할 수 있는 암 치료 기술의 개발이 중요한 실정이다.
이에, 암 면역치료 방법이 개발되었으며, 이는 암 특이적 면역세포의 활성화를 통해 암세포를 죽이는 방법으로, 기억면역세포의 생성으로 완치의 가능성이 높고 재발 억제까지 기대할 수 있으며 수술을 필요로 하지 않아 종래의 암 치료 방법에 비해 부작용을 최소화 할 수 있다. 현재 암 면역치료 시장은 약 4조 규모이며 이는 2024년도까지 10조 규모로 급증할 것으로 예상된다. FDA 승인 암 면역치료제는 현재까지 약 15건 정도로 면역 항암요법의 적용이 본격화 되고 있는 추세이다. 암 면역치료 방법 중 대표적인 방법으로는 백신 치료와 면역 체크포인트(checkpoint) 억제제 치료가 있으며, 이는 현재 좋은 임상 결과를 나타내는 면역치료 항암제로 알려져 있다. 그러나, 현재 사용되는 암 면역치료 방법은 암세포가 자체적으로 방해 항체를 만들어 면역항암제의 효과를 떨어뜨리는 현상이 발생하고, 이로 인해 투여한 면역항암제의 일정량이 손실되는 것과 같은 결과를 초래하여 치료효과를 떨어뜨리는 문제가 있으며, 면역항암제 치료 또한 약물의 체순환으로 인한 다양한 부작용과 약물 손실이 발생하고 있다.
한편, 치료용 조영 물질(theragnostic agent)은 병의 진단과 치료를 동시에 가능하게 하는 물질들을 일컫는다. 이들은 보통 작은 크기의 물질로 이뤄져 있으며, 대개 형광 염료, 방사성 분자 등이 리포좀, 고분자 및 나노 입자 등의 내부에 담지되고 약물이나 진단용 마커가 외부에 도입된 형태를 갖는다. 최근에는 생체 적합성(biocompatibility)이 우수한 지질구조체(lipid structure)로 이루어진 치료용 조영 물질의 합성 연구가 주를 이루고 있으며, 특히 지질구조체로 이뤄진 마이크로미터 크기의 공기방울 초음파 조영제인 마이크로버블(Microbubble ultrasound agent, MB)을 이용한 생체 영상 분석 연구가 활발히 진행되고 있으며, 마이크로버블, 나노 입자 약물 전달 시스템 및 이의 병용 요법을 이용한 치료 방법에 대해서도 많은 연구가 이루어지고 있다.
초음파에 노출된 마이크로버블은 공동현상(cavitation)이 일어나 주변 세포의 세포막이 일시적으로 기공을 형성하게 되고, 이로 인해 암세포 부근의 lymphocyte 증가현상을 나타내며, tumor ablation이 수지상세포를 활성화시켜 이를 통한 면역시스템 활성화 효과를 나타내므로, 초음파를 이용한 면역치료 방법은 간접적인 치료요법으로써 단독적으로 활용할 경우에는, 그 범위나 치료효과가 면역항암제를 이용한 치료법보다 약하지만, 초음파가 지니고 있는 고유의 물리적인 메커니즘을 기존의 면역치료 방법과 융합하여 사용할 경우 시너지 효과를 낼 수 있을 것으로 예상되었다.
이에, 본 발명자들은 면역치료에 초음파 기술을 융합하여 마이크로버블 복합체 및 초음파를 활용함으로써 새로운 면역치료 방법을 개발하였으며, 이는 암의 진단 및 치료 등과 같은 암 관련 연구, 개발 분야에 있어서 아직까지 잘 알려진 바가 없다.
본 발명자들은 암 면역치료 방법에 초음파 기술을 융합하여 항체를 결합(conjugation)시킨 마이크로버블에 초음파를 처리하였을 때, 초음파를 처리하지 않은 경우에 비해 뛰어난 항종양 효과가 나타나는 것을 확인하였는바, 이에 기초하여 본 발명을 완성하였다.
이에, 본 발명의 목적은 마이크로버블; 및 상기 마이크로버블의 표면에 결합된 면역 체크포인트 저해(checkpoint inhibition)능 항체를 포함하는 면역 마이크로버블 복합체(immuno microbubble complex) 및 이의 이용을 제공하는 것이다.
그러나, 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 본 발명이 속하는 기술 분야의 통상의 지식을 가진 자에게 명확하게 이해될 수 있을 것이다.
상기와 같은 목적을 달성하기 위해 본 발명은 마이크로버블; 및 상기 마이크로버블의 표면에 결합된 면역 체크포인트 저해(checkpoint inhibition)능 항체를 포함하는 면역 마이크로버블 복합체(immuno microbubble complex)로서,
초음파의 소노포레시스(sonophoresis)로 면역반응이 부스팅(boosting)된 환경에서 종양세포의 면역 회피를 저해하여 T세포를 활성화 시키는 것을 특징으로 하는, 면역 마이크로버블 복합체를 제공한다.
본 발명의 일 구현예로서, 상기 마이크로버블과 항체의 결합은 아미드 결합, 싸이올 간의 결합 또는 바이오틴-아비딘 결합일 수 있다.
본 발명의 다른 구현예로서, 상기 마이크로버블은 DSPC(1,2-Distearoyl-sn-glycero-3-phosphorylcholine) 및 DSPE(1,2-distearoyl-sn-glycero-3-phosphoethanolamine)를 포함할 수 있다.
본 발명의 또 다른 구현예로서, 상기 DSPE는 DSPE-PEG2000-NHS(1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[succinyl(polyethylene glycol)-2000])일 수 있다.
또한, 본 발명은 (a) 인지질을 유기용매에 혼합한 다음 수화하는 단계;
(b) 상기 (a) 단계에서 수화된 리포좀 전구체를 분산 후 교반하여 마이크로버블을 제조하는 단계; 및
(c) 상기 마이크로버블에 면역 체크포인트 저해(checkpoint inhibition)능 항체를 결합(conjugation)시키는 단계를 포함하는, 면역 마이크로버블 복합체의 제조 방법을 제공한다.
본 발명의 일 구현예로서, 상기 인지질에는 DSPC(1,2-Distearoyl-sn-glycero-3-phosphorylcholine) 및 DSPE(1,2-distearoyl-sn-glycero-3-phosphoethanolamine)가 포함될 수 있다.
본 발명의 또 다른 구현예로서, 상기 DSPC 및 DSPE의 몰(mol) 비율은 6 내지 9:1 내지 4(mol)일 수 있다.
본 발명의 또 다른 구현예로서, 상기 면역 체크포인트 저해능 항체는 항 PD-L1(Programmed death ligand-1) 항체, 항 B7-1 항체, 및 항 B7-2 항체로 이루어진 군으로부터 선택되는 하나 이상일 수 있다.
또한, 본 발명은 상기 면역 마이크로버블 복합체를 포함하는, 약물 전달 담체를 제공한다.

본 발명은 마이크로버블; 및 상기 마이크로버블의 표면에 결합된 면역 체크포인트 저해(checkpoint inhibition)능 항체를 포함하는 면역 마이크로버블 복합체(immuno microbubble complex)로서,
초음파의 소노포레시스(sonophoresis)로 면역반응이 부스팅(boosting)된 환경에서 종양세포의 면역 회피를 저해하여 T세포를 활성화 시키는 것을 특징으로 하는, 면역 마이크로버블 복합체를 제공한다.
본 발명에 있어서, 상기 마이크로버블은 초음파의 소노포레시스로 면역 반응을 부스팅하고 상기 면역 체크포인트 저해능 항체는 면역 세포가 표적 질환 세포를 인식하게 할 수 있다.
본 발명에 있어서, 상기 마이크로버블은 DSPC(1,2-Distearoyl-sn-glycero-3-phosphorylcholine) 및 DSPE(1,2-distearoyl-sn-glycero-3-phosphoethanolamine)를 포함할 수 있다.
본 발명에 있어서, 상기 항체는 링커 또는 마이크로버블 표면의 활성화된 반응기에 의해 마이크로버블에 결합될 수 있다. 상기 반응기로는 싸이올기(thiol), 아민기(amine)이거나, 링커로는 상기 반응기를 포함하는 화합물일 수 있으며, 상기 마이크로버블과 항체의 결합은 아미드(amide) 결합, 싸이올(thiol) 간의 결합 또는 바이오틴(biotin)-아비딘(avidin) 결합일 수 있다.
본 발명에 있어서, "링커(linker)"는 항체 또는 약물 등을 마이크로버블에 결합시키는 물질을 의미하며, 상기 링커의 종류에는 제한이 없고, 예컨대 NHS(N-hydroxysuccinimide) 또는 maleimide일 수 있으며, 본 발명의 일 실시예에 따르면 상기 DSPE는 링커로 NHS가 연결된 DSPE-PEG2000-NHS(1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[succinyl(polyethylene glycol)-2000])일 수 있다.
본 발명에 있어서, 상기 마이크로버블(microbubble)의 입자 직경은 0.8 μm 내지 1.5 μm일 수 있고, 본 발명의 일 실시예에 따르면 상기 마이크로버블의 평균 직경은 1.19±0.245 μm일 수 있다.
본 발명에 있어서, 상기 DSPC(1,2-Distearoyl-sn-glycero-3-phosphorylcholine) 및 DSPE (1,2-distearoyl-sn-glycero-3-phosphoethanolamine)의 몰(mol) 비율은 6 내지 9:1 내지 4(mol), 6.5 내지 9:1 내지 3.5(mol), 또는 7 내지 9:1 내지 3(mol)일 수 있고, 바람직하게는 7:3(mol) 또는 9:1일 수 있으며, 가장 바람직하게는 9:1(mol)의 비율로 마이크로버블을 제조할 때, 제조된 마이크로버블의 안정성이 가장 높을 수 있으나, 이에 제한되지 않는다.
본 발명에서 사용되는 용어 "초음파(ultrasound)"는 일반적으로 사람의 귀가 들을 수 있는 음파의 주파수인 16 Hz~20 kHz의 주파수를 넘는 음파를 의미하며, 고강도 집중초음파는 연속적이고 고강도인 초음파 에너지를 초점에 제공하는 집속형 초음파를 도입하여 에너지와 진동수에 따라 순간 열 효과(65-100℃), 공동화(cavitation) 효과, 기계적 효과 및 음향화학적(sonochemical) 효과를 낼 수 있다. 초음파는 인체조직을 통과할 때 해롭지 않으나 초점을 형성하는 고강도의 초음파는 조직의 종류에 상관없이 응고괴사 및 열소작 효과를 일으킬 수 있을 만큼 충분한 에너지를 발생한다.
본 발명에 있어서, 상기 초음파는 고강도 집중초음파(high intensity focused ultrasound, HIFU)일 수 있으나, 이에 제한되는 것은 아니다. 본 발명에 있어서, 상기 HIFU에 사용되는 주파수는 0.1 Mhz 내지 10 Mhz, 0.1 Mhz 내지 9 Mhz, 0.1 Mhz 내지 8 Mhz, 0.1 Mhz 내지 7 Mhz, 0.1 Mhz 내지 6 Mhz, 0.1 Mhz 내지 5 Mhz, 0.1 Mhz 내지 4 Mhz, 0.1 Mhz 내지 3 Mhz , 또는 0.1 Mhz 내지 2 Mhz 일 수 있으며, 본 발명의 일 실시예에 따르면 1.1 Mhz일 수 있다. 본 발명에 있어서, 상기 HIFU는 1.1 Mhz, 50 W 내지 100 W, 1 내지 10 duty cycle, 및 40 PRF의 조건으로 10초 내지 20초 동안 처리할 수 있으나, 상기 조건에 제한되는 것은 아니다.
본 발명에서 사용되는 용어 "소노포레시스(sonophoresis)"는 액체 매개물을 통과하는 물질의 이송을 증진시키기 위해 음파, 대개 초음파의 파동 에너지를 사용하는 것을 가리키는 것으로, 약물의 흡수를 증가시키기 위해 초음파가 사용되는 약물 전달 방법을 말한다. 이 때 음파의 압축파는 액체 매개물에 "흐름" 및/또는 "케비테이션(cavitation)"을 유발한다. 본 발명에 따른 면역 체크포인트 저해능 항체에 초음파를 처리할 경우 소노포레시스에 의한 면역 반응 부스팅(boosting) 효과가 나타나, 상기 항체는 암세포 등의 표적 세포로 타겟팅이 더 잘 일어날 수 있다.
본 발명에서 사용되는 용어 "케비테이션(cavitation)"은 "공동화", "공동현상"등의 용어로도 사용될 수 있으며, 유체 속에서 압력이 낮은 곳이 생기면 물 속에 포함되어 있는 기체가 물에서 빠져나와 압력이 낮은 곳에 모이는데, 이로 인해 물이 없는 빈공간이 생긴 것을 의미하며, 본 발명에 있어서 상기 면역 마이크로버블 복합체는 초음파에 의해 케비테이션이 일어나, 주변 세포의 세포막이 일시적으로 기공을 형성하고, 이 기공을 통해 세포 내로 물질이 투과되며, 암세포 부근의 림프구(lymphocyte)를 밀집시킬 수 있다.
또한, 본 발명은 (a) 인지질을 유기용매에 혼합한 다음 수화하는 단계;
(b) 상기 (a) 단계에서 수화된 리포좀 전구체를 분산 후 교반하여 마이크로버블을 제조하는 단계; 및
(c) 상기 마이크로버블에 면역 체크포인트 저해(checkpoint inhibition)능 항체를 결합(conjugation)시키는 단계를 포함하는, 면역 마이크로버블 복합체의 제조 방법을 제공한다.
본 발명에 있어서, 상기 인지질에는 DSPC(1,2-Distearoyl-sn-glycero-3-phosphorylcholine) 및 DSPE(1,2-distearoyl-sn-glycero-3-phosphoethanolamine)가 포함될 수 있다.

본 발명에서 사용되는 용어 "항체"는 항원에 특이적으로 결합하여 인식하는 면역글로불린 유전자 또는 이의 단편에서 유래하는 프레임워크 영역을 포함하는 폴리펩티드를 의미한다. 인식된 면역글로불린 유전자는 카파, 람다, 알파, 감마, 델타, 엡실론, 및 뮤 불변 영역 유전자를 비롯하여, 무수한 면역글로불린 가변 영역 유전자를 포함한다. 경쇄는 카파 또는 람다로 분류되었다. 중쇄는 감마, 뮤, 알파, 델타, 또는 엡실론으로 분류되고, 결과적으로 각각 면역글로불린 부류 IgG, IgM, IgA, IgD 및 IgE를 한정한다. 전형적으로, 항체의 항원-결합 영역은 결합 특이성 및 친화성에서 가장 핵심적이게 된다. 본 발명에 있어서, 항체 또는 항체의 단편은 인간, 마우스, 래트, 햄스터, 낙타, 토끼 등을 포함한, 상이한 개체에서 유래될 수 있으나, 이에 제한되는 것은 아니다. 본 발명의 항체는 항체의 바람직한 기능(예를 들어, 당화, 발현, 항원 인식, 이펙터 기능, 항원 결합, 특이성 등)을 개선시키거나 또는 조정하기 위해 1 이상의 아미노산 위치에서 변형되거나 또는 돌연변이된 항체를 포함할 수도 있다.

본 발명에 있어서, 상기 항 PD-L1 항체는 PD-L1에 특이적으로 결합함으로써 PD-L1의 결합을 억제하여 종양에 대한 면역 반응 억제를 방지할 수 있으며, 이 때 상기 PD-L1의 아미노산 서열은 서열번호 1(NCBI GenBank: ADK70950.1)로 표시될 수 있고, 항 PD-L1 항체는 InVivoPlus anti-mouse PD-L1(B7-H1)(Catalog# BP0101, Clone 10F.9G2)을 구입하여 사용하였다.

본 발명에서 사용되는 용어 "조영제"는 생체 내에서 강력하고 특이적으로 암세포를 조영(contrast) 또는 영상화(imaging)하기 위해 체내 투여되는 물질을 말하는 것으로, 현재 의료, 진단 분야에서 조직 및 세포의 이미지 강화를 위해 광범위하게 사용되고 있다. 본 발명의 조영제라는 용어는 종래 알려진 CT, PET 또는 MRI 조영제의 범위로 한정되지 않으며 초음파 이미지 분석용 영상제, 형광 이미지 분석용 영상제 등을 포함하는 의미로 사용된다.
또한, 본 발명은 상기 면역 마이크로버블 복합체를 포함하는, 암 예방 또는 치료용 약학적 조성물을 제공한다.

본 발명에 따른 좌제에는 카카오지, 라놀린, 위텝솔, 폴리에틸렌글리콜, 글리세로젤라틴, 메칠셀룰로오스, 카르복시메칠셀룰로오스, 스테아린산과 올레인산의 혼합물, 수바날(Subanal), 면실유, 낙화생유, 야자유, 카카오버터+콜레스테롤, 레시틴, 라네트왁스, 모노스테아린산글리세롤, 트윈 또는 스판, 임하우젠(Imhausen), 모놀렌(모노스테아린산프로필렌글리콜), 글리세린, 아뎁스솔리두스(Adeps solidus), 부티룸 태고-G(Buytyrum Tego-G), 세베스파마 16(Cebes Pharma 16), 헥사라이드베이스 95, 코토마(Cotomar), 히드록코테 SP, S-70-XXA, S-70-XX75(S-70-XX95), 히드록코테(Hydrokote) 25, 히드록코테 711, 이드로포스탈(Idropostal), 마사에스트라리움(Massa estrarium, A, AS, B, C, D, E, I, T), 마사-MF, 마수폴, 마수폴-15, 네오수포스탈-엔, 파라마운드-B, 수포시로(OSI, OSIX, A, B, C, D, H, L), 좌제기제 IV 타입(AB, B, A, BC, BBG, E, BGF, C, D, 299), 수포스탈(N, Es), 웨코비(W, R, S, M ,Fs), 테제스터 트리글리세라이드 기제(TG-95, MA, 57)와 같은 기제가 사용될 수 있다.
경구 투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 상기 추출물에 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘카보네이트(calcium carbonate), 수크로스(sucrose) 또는 락토오스(lactose), 젤라틴 등을 섞어 조제된다. 또한 단순한 부형제 이외에 마그네슘 스티레이트 탈크 같은 윤활제들도 사용된다.
경구 투여를 위한 액상제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조제제, 좌제가 포함된다. 비수성용제, 현탁제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다.
본 발명에 따른 약학적 조성물은 약학적으로 유효한 양으로 투여한다. 본 발명에 있어서, "약학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효용량 수준은 환자 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다.
본 발명에 따른 약학적 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 본 발명이 속하는 기술분야에 통상의 기술자에 의해 용이하게 결정될 수 있다.
본 발명의 약학적 조성물은 개체에게 다양한 경로로 투여될 수 있다. 투여의 모든 방식은 예상될 수 있는데, 예를 들면, 경구 복용, 피하 주사, 복강 투여, 정맥 주사, 근육 주사, 척수 주위 공간(경막내) 주사, 설하 투여, 볼점막 투여, 직장 내 삽입, 질 내 삽입, 안구 투여, 귀 투여, 비강 투여, 흡입, 입 또는 코를 통한 분무, 피부 투여, 경피 투여 등에 따라 투여될 수 있다.
본 발명의 약학적 조성물은 치료할 질환, 투여 경로, 환자의 연령, 성별, 체중 및 질환의 중등도 등의 여러 관련 인자와 함께 활성성분인 약물의 종류에 따라 결정된다.
본 발명의 암 예방 또는 치료용 약학적 조성물에 있어서, 상기 조성물에 포함되는 면역 마이크로버블 복합체는 초음파 처리와 병용하여 사용될 수 있으며, 상기 조성물에는 면역 마이크로버블 복합체 외에 항암제, 영상 조영제, 항생제, 항염증제, 단백질, 싸이토카인, 펩타이드, 및 항체로 이루어진 군으로부터 선택되는 하나 이상이 더 포함될 수 있다.
또한, 본 발명은 생물학적 샘플에 면역 마이크로버블 복합체를 처리하는 단계; 및 초음파를 처리하는 단계를 포함하는, 암 진단을 위한 정보제공방법을 제공한다.
또한, 본 발명은 생물학적 샘플에 면역 마이크로버블 복합체를 처리하는 단계; 및 초음파를 처리하는 단계를 포함하는, 암 진단 방법을 제공한다.
본 발명에서 사용되는 용어 "진단"은 병리 상태의 존재 또는 특징을 확인하는 것을 의미한다. 본 발명의 목적상, 진단은 암의 발병 여부를 확인하는 것이다.
또한, 본 발명은 상기 면역 마이크로버블 복합체를 포함하는 조성물을 개체에 투여하는 단계; 및 초음파를 처리하는 단계를 포함하는 암 치료 방법을 제공한다.
또한, 본 발명은 상기 면역 마이크로버블 복합체를 포함하는 조성물의 암 치료 용도를 제공한다.
또한, 본 발명은 상기 면역 마이크로버블 복합체의, 암 치료에 이용되는 약제를 생산하기 위한 용도를 제공한다.
본 발명에 있어서, "투여"란 임의의 적절한 방법으로 개체에게 소정의 본 발명의 조성물을 제공하는 것을 의미한다.
본 발명에 있어서, "개체"란 질병의 치료를 필요로 하여 본 발명의 조성물이 투여될 수 있는 대상을 의미하고, 보다 구체적으로는, 인간 또는 비-인간인 영장류, 생쥐(mouse), 개, 고양이, 말, 및 소 등의 포유류를 의미한다.
본 발명의 일 실시예에서는 1,2-disteaoyl-sn-glycero-3-phosphocholine(DSPC) 및 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[succinyl(polyethylene glycol)-2000](DSPE-PEG2K-NHS)의 비율 조건을 달리하여 마이크로버블을 제조한 후 이의 물성을 비교하였으며, 9:1의 몰(mol) 비율로 제조하였을 때 마이크로버블의 안정성이 가장 높은 것을 확인하였다(실시예 2 참조).
종양 세포는 그들의 생리학적 방법을 이용하여 면역 반응을 회피하는데, 종양 세포의 표면에서, PD-L1(Programmed death ligand 1)은 정상적으로 상향 조절되고 T 세포의 표면에서 PD-1(Programmed death 1)은 PD-L1/PD-1 결합에 대한 면역 반응 및 공동 자극 분자 CD80의 관여 억제를 유도한다. CD80은 최대 면역 반응을 위해 중요한 항원 비특이적 공동 자극 신호를 제공하는 면역 글로불린 수퍼 패밀리의 구성원이다. 이러한 수용체와 리간드가 서로 결합할 때, Src homology 2 domain-containing protein tyrosine family phosphatases(SHPs)를 모집하고, 이는 T 세포가 MHC I의 인식의 유무에 관계없이 과립 및 퍼포린을 방출할 수 없게 되도록 하며, 조절 T 세포 자극, T 세포의 apoptosis 촉진, 및 effector T 세포 활성화를 저해한다.
PD-L1/PD-1 결합 억제는 면역 감소를 억제하여 항종양 면역 반응을 유지할 수 있는 방법 중 하나로, PD-L1/PD-1 결합을 표적으로 하는 면역 요법이 사용될 수 있다.
이에, 본 발명의 다른 실시예에서는 상기 제조한 마이크로버블에 항 PD-L1 항체를 결합하여 면역 마이크로버블 복합체(IMC)를 제조하였으며, 상기 항 PD-L1 항체의 결합 효율을 측정하여 항체가 100% 결합한 것을 확인하였다(실시예 3 참조).
본 발명의 또 다른 실시예에서는 상기 제조한 면역 마이크로버블 복합체(IMC)의 공초점 이미지를 관찰함으로써 마이크로버블의 표면에 결합된 항체를 확인하였다(실시예 4 참조).
본 발명의 또 다른 실시예에서는 상기 제조한 면역 마이크로버블 복합체(IMC)에 고강도 집중 초음파(HIFU)를 처리함에 따라 나타나는 항종양 효과 및 IFN-γ와 cytolytic T cell에 대한 활성을 확인한 결과, IMC에 HIFU를 처리할 경우 면역 증강 효과가 나타나는 것을 확인하였다(실시예 5 참조).
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
실시예 1. 재료 준비
1,2-disteaoyl-sn-glycero-3-phosphocholine(DSPC)은 Coatsome에서 얻었으며, 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[succinyl(polyethylene glycol)-2000](DSPE-PEG2000-NHS)은 Nanocs로부터 구입하였다. 또한, 래트(rat) IgG2b isotype대조군(7.62 mg/ml) 및 항-마우스 PD-L1(B7-H4)(6.76 mg/ml)은 BioXcell로부터 얻었다.
실시예 2. 마이크로버블 제조 및 DSPE-PEG2K 비율에 따른 마이크로버블 물성 확인
2-1. 마이크로버블 제조
1,2-disteaoyl-sn-glycero-3-phosphocholine(DSPC) 및 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[succinyl(polyethylene glycol)-2000](DSPE-PEG2K-NHS)를 이용하여 인지질 필름 수화 방법에 의해 마이크로버블(microbubble, 이하 MB)을 합성하였으며, 이 때 상기 DSPC와 DSPE-PEG2K-NHS의 비율에 따라 생성되는 마이크로버블의 물성을 비교하였다.
이를 위해, DSPC:DSPE-PEG2K-NHS를 9:1, 7:3, 6:4, 5:5, 또는 3:7의 각각의 몰(mol) 비율로 클로로포름(chloroform)에 용해시켰다. 그리고 나서, 클로로포름을 증발시켜 인지질 필름을 형성하였다. 1.0 mg/ml 농도의 상기 인지질 필름을 bath sonicator(초음파분쇄기)로 DSPC의 상 전이 온도에 걸쳐 0.01M PBS를 이용하여 수화시켰다. 이후 이를 병(vial)에 담고 여기에 sulfur hexafluoride gas(SF
6)를 채워넣었다. 이어, 수화된 리포좀 전구체를 마이크로버블로 변형시키기 위해 Vialmix
TM(Definity, United states)로 45초 동안 교반하여 마이크로버블을 제조하였다.
상기 방법으로 생성된 DSPC:DSPE-PEG2K-NHS 비율에 따른 마이크로버블의 크기 분포 및 zeta 전위는 Malvern Zetasizer Nano (Malvern Instrument Ltd., Worcesterchire, U.K.)를 사용하여 측정하였으며, 마이크로버블을 PBS(pH 7.4)에 100ug/ml로 용해시키고 25℃에서 5회 측정하였다.
그 결과, 상기 5가지 비율로 제조된 마이크로버블의 크기 및 개수를 각각 도 1에 나타내었으며, 상기 DSPC:DSPE-PEG2K-NHS의 mol 비율이 9:1일 경우 크기가 작고 많은 수의 마이크로버블이 생성된 것을 확인하였다.
2-2. 마이크로버블의 안정성 비교
상기 실시예 2-1에서 DSPC:DSPE-PEG2K-NHS의 비율에 따라 제조된 마이크로버블의 안정성을 시간별로 관찰하였다.
구체적으로, 상기 실시예 2-1에서와 같이 5가지의 몰 비율로 마이크로버블을 제조한 다음, 상온에서 0시간, 3시간, 6시간, 24시간동안 마이크로버블이 사라지는 정도를 육안으로 확인하였다.
그 결과, 도 2에 나타낸 바와 같이, 시간이 지날수록 상기 DSPC:DSPE-PEG2K-NHS의 3:7, 5:5, 6:4 비율 순서대로 용액이 투명해져 마이크로버블이 사라지는 것을 관찰함으로써 7:3 및 9:1의 비율로 제조할 경우 안정적인 것을 확인하였다.
또한, 도 3에 나타낸 바와 같이 상기 5 가지 비율로 제조한 마이크로버블의 크기 분포를 확인한 결과, 3:7 및 5:5 비율의 경우, 1h에서의 peak 외에는 안정성이 낮아 zeta-size에서는 크기 분포가 나타나지 않았으며, 6:4의 경우 24h에서 불안정한 것으로 나타났다. 반면, 7:3 및 9:1 비율에서는 마이크로버블의 크기 분포가 안정적으로 나타났고, 특히 9:1 비율에서의 마이크로버블이 가장 안정성이 높은 것을 확인할 수 있었다.
따라서, 본 발명에서는 9:1의 DSPC:DSPE-PEG2K-NHS 비율로 마이크로버블을 제조한 후 이를 실험에 사용하였다. 이 때 상기 마이크로버블은 상기 실시예 2-1의 방법으로 제조하였으며, 인지질 필름의 농도는 0.5 mg/ml로 사용하였다.
마이크로버블의 제조에 사용한 DSPE-PEG-NHS의 구조 및 DSPC:DSPE-PEG2K-NHS = 9:1 비율로 제조한 마이크로버블의 크기 분포를 도 4에 나타내었으며, 마이크로버블은 크기가 1.19±0.245 μm이고 개수는 평균 1.99 x 10
9개로 나타난 것을 확인하였다.
2-3. 시간과 온도에 따른 마이크로버블의 안정성 확인
시간과 온도에 따른 마이크로버블(MB)의 안정성을 테스트하기 위해 4℃ 및 25℃의 온도에서 공기(air)를 충진한 후 제조한 MB(MB with air) 및 SF
6 가스를 충진한 후 제조한 MB(MB with SF
6)에서의 개수를 시간대별로 각각 광학 현미경으로 모니터링 하였다.
그 결과, 도 5에 나타낸 바와 같이, MB with SF
6(4℃)에서 가장 많은 수의 마이크로버블이 관찰되어 안정성이 높은 것을 확인하였다.
실시예 3. 면역 마이크로버블 복합체의 제조
3-1. 마이크로버블-항체 입자 조건
면역 마이크로버블 복합체(immuno microbubble complex, 이하 IMC)는 상기 실시예 2-1에서 제조한 마이크로버블의 NHS(N-hydroxysuccinimide)에 항 PD-L1 항체를 결합시켜 제조하였으며, 이 때 상기 항 PD-L1 항체가 NHS와 1:1로 반응한다고 가정하고 IMC를 제조하기 위한 항 PD-L1 항체 조건을 확인하였다.
이 경우 하기 표 1과 같이 NHS의 개수는 PD-L1 항체의 개수보다 약 100배가 많아 항 PD-L1 항체의 1회 투여량이 100~200 ug일 때, MB에 결합되는 항체는 충분할 것으로 예상하였다.
3-2. 면역 마이크로버블 복합체의 제조
상기 실시예 2-1에서 제조한 마이크로버블의 정제 후, 200 μg의 항 PD-L1 항체(이하, PD-L1 Ab)(BioXcell, United states)를 1.5 x 10
9개의 마이크로버블에 결합(conjugation)시켰다. 비결합 PD-L1 Ab를 제거하기 위해, 결합된 마이크로버블-항체의 정제를 5분 동안 300rpm에서 구배 원심분리로 수행하였으며, 동적광산란 (Malvern Zetasizer Nano series, England)으로 크기 및 표면 전하를 측정하였다.
3-3. 항 PD-L1 항체의 결합 효율 측정
IMC 합성 후, 여액에 남아있는 liposome+Ab(antibody) 때문에 결합력 측정에 대해 나타날 수 있는 오차를 우려하여 (1) lipid(micelle)+Ab 및 (2) Ab 농도를 먼저 측정하였다. 그 결과, 도 6에 나타낸 바와 같이 두 그룹간에 차이가 거의 없는 것을 확인하여, IMC 합성 후 여액에 존재하는 비결합 항체를 Bradford assay 방법으로 측정함으로써 항체 결합 효율을 확인하였다.
구체적으로, 여액에서 최대한 항체가 섞이지 않도록 충분히 기다려 준 다음, 주사기를 통해 여액을 분리하고 Bradford 용액을 섞어서 5분 동안 기다린 후, 항체를 측정하였다.
그 결과, STD curve에서 control 그룹(no Ab)과 큰 차이를 보이지 않았으며, 하기 표 2에서 나타낸 바와 같이, 여액에서의 Bradford assay 결과 항체 결합 효율이 99.9%인 것으로 나타나고, 여액과 control간의 큰 차이가 없는 것으로 볼 때, 200 ug 항체 기준으로 100% 결합 반응이 진행되었음을 알 수 있었다.
또한, Thiol을 사용하여 conjugation 시키는 방법을 이용하여 Bradford assay로 측정한 결과, 하기 표 3에 나타낸 바와 같이 항체가 0.581 nm 및 0.576 nm로 측정되었다.
실시예 4. 면역 마이크로버블 복합체(IMC)의 공초점 이미지 확인
4-1. 항체의 농도에 따른 공초점 이미지 확인
마이크로버블에서의 항체 결합을 확인하기 위해, IMC의 공초점 이미지를 확인하였다.
구체적으로, 10 uL(0.01 mg/ml)의 MB에 1차 항체(mouse PD-L1 antibody) 20 ug~160 ug를 첨가하여 conjugation 시킨 후 제조된 IMC에 FITC-태그된 2차 항체(mouse PD-L1 antibody의 Fc 부분에 결합하는 항체)를 1차 항체의 2배로 4℃에서 60분 동안 처리하였다. 상기 2차 항체가 결합된 IMC 용액을 3,000 rpm에서 5분 동안 원심분리하여 정제하였으며, 이를 슬라이드 유리에 떨어뜨렸다. 그런 다음, 공초점 레이저 현미경(Confocal Laser Scanning Microscope, CLSM)으로 IMC를 관찰하였다.
그 결과, 도 7에 나타낸 바와 같이, 항체의 농도가 높을수록 형광이 뚜렷하게 나타나, 항 PD-L1 항체가 MB에 대해 결합력이 높은 것을 관찰하였다.
4-2. GFP 및 DIC 이미지 비교
1 mg의 MB에 1차 항체(항 PD-L1 항체) 1 mg을 첨가하여 conjugation 시킨 후 제조된 IMC에 GFP가 태그된 2차 항체를 1 mg 처리하고 상기 실시예 4-1의 방법으로 공초점 이미지를 관찰하였으며, DIC(differential interference contrast) 현미경으로 관찰한 이미지 및 상기 GFP와 DIC를 합친 이미지를 확인하여 도 8에 나타내었다.
도 8에 나타낸 바와 같이, 마이크로버블의 표면에 형광이 나타난 것을 관찰하여 본 발명에 따른 IMC는 마이크로버블의 표면에 항체가 결합한 것을 확인하였다.
실시예 5. 면역 마이크로버블 복합체(IMC) 및 고강도 집중초음파(HIFU) 처리에 따른 효과
5-1. B16F10 cell에서 PD-L1 발현 확인
도 9에 나타낸 바와 같이 실험을 설계하고 B16F10 cell에서의 PD-L1 발현을 확인하였다.
구체적으로, B16F10 cell을 well에 풀고 발현된 PD-L1을 검출하기 위해 50 ug의 항 PD-L1 항체를 넣었다. 그 후, PBS나 media로 washing 을 두 번 진행한 뒤, 공초점(confocal) 현미경을 통한 확인을 위해 형광이 달린 2차 PD-L1 항체를 50~100 ug 넣어준 후, 2시간정도 기다리고, 그 후에 또 다시 2~3번 washing 작업을 거친 후 confocal 촬영을 하였다.
그 결과, 도 10에 나타낸 바와 같이, PD-L1 발현량을 확인한 결과 형광 anti-PD-L1 를 통해, PD-L1 에 대해 붉은색으로 염색된 것을 관찰하여 세포 표면에 PD-L1 이 높게 발현된 것을 확인하였다.
5-2. 면역 마이크로버블 복합체(IMC) 및 고강도 집중초음파(HIFU) 처리에 따른 항종양 활성
상기 실시예 3-2에서 제조한 IMC 및 IMC에 고강도 집중초음파(high-intensity focused ultrasound, 이하 HIFU)를 처리했을 때의 항종양 활성을 확인하기 위해 4주령의 BALB/c 암컷 마우스의 우측 옆구리 부위에 CT26wt 세포 현탁액(마우스당 1 x 10
6 세포)으로 피하 접종하여 확립된 CT26wt 종양 보유 BALB/c 마우스 모델을 사용하였다. 종양의 부피가 50 mm
3에 도달한 후, 마우스를 치료를 위해 무작위로 분류하였다. 실험 그룹은 다음과 같이 정의되었다:
G1: Isotype Ab only (0.3 mg)
G2: PD-L1 Ab only (0.3 mg)
G3: PBS
G4: HIFU only
G5: Isotype Ab + MB (0.3 mg)
G6: Isotype Ab + MB + HIFU (0.3 mg)
G7: PD-L1 + MB (0.3 mg)
G8: IMC + HIFU (0.3 mg)
Isotype Ab로는 PD-L1 Ab의 기능이 없는 동종의 IgG2 Ab를 control로 사용하였다.
종양 크기는 디지털 캘리퍼(caliper)로 측정하였으며, 실험의 마지막에 조직학적 분석을 위해 종양 조직을 절제하였다.
그 결과, 도 11에 나타낸 바와 같이, G2에 비해 G1의 종양 크기가 약간 작게 나타났으나 큰 차이는 없었고(도 11의 상단 도면), G3에 비해 G4의 종양 크기가 약간 더 작게 나타났으며(도 11의 가운데 도면), Isotype Ab + MB 그룹(G5) 및 PD-L1 Ab + MB 그룹(G7) 모두 HIFU를 처리했을 때 종양의 크기가 현저하게 감소하였고, 특히 PD-L1 Ab + MB(IMC)에 HIFU를 처리한 G8 그룹에서 가장 종양 크기가 감소한 것을 확인하였다(도 11의 하단 도면).
따라서, 상기 결과로부터 본 발명에서 제조한 면역 마이크로버블 복합체(IMC)에 고강도 집중초음파(HIFU)를 처리할 경우 뛰어난 항종양 효과가 나타나는 것을 알 수 있었다.
5-3. IFN-γ 및 cytolytic T cell에 대한 활성
IFN-γ assay는 플라즈마 또는 복수를 마우스에서 빼낸 다음 quantikine mouse IFN-γ assay kit(R&D systems, Minneapolis, MN) 를 적용시켜 ELISA 로 측정하였다.
또한, cytolytic T cell 은 암세포를 PBS 로 resuspend 한후, 10 uM 의 CFSE(Carboxyfluorescein succinimidyl ester)를 37℃에서 10분간 반응시킨 뒤, 동량의 FCS 를 상온에서 2분간 적용하여 반응을 stop 시켰다. 그 후 AIM-V(+5% human A/B serum)으로 2번 washing 한뒤, AIM-V+IL2, IL-7 과 IL-15(+5% human A/B serum) 으로 resuspend 하였으며, DC(dendritic cell) 와 CTL(cytotoxic T cell)을 1:10 비율로 첨가하고 37℃에서 6시간동안 incubate 진행하였다. 그런 다음 1 ug/ml propidium iodide를 넣고 측정 전에 10 ul의 calibrate beads를 넣었다.
그 결과, 도 12에 나타낸 바와 같이, 실시예 5-2의 결과와 마찬가지로 PD-L1 Ab + MB(IMC)에 HIFU를 처리한 G8 그룹에서 IFN-γ specific spot이 가장 높게 나타났으며, cytolytic T cell의 활성도 가장 높게 나타난 것을 확인하였다.
상기로부터 본 발명에 따른 면역 마이크로버블 복합체에 HIFU를 처리함으로써 면역 증강 효과가 나타나는 것을 확인할 수 있었다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야 한다.
본 발명에 따른 면역 마이크로버블 복합체는 결합된 항체의 전달 효율을 증가시키고 암의 진단과 동시에 치료에 이용될 수 있으며, 초음파를 통하여 조영 효과, 반감기 개선, 개선된 약물전달, 림프구 밀집효과, 면역항암치료 및 면역치료유도 등 면역치료 분야에서 이용될 것으로 기대된다.
Claims (21)
- 마이크로버블; 및상기 마이크로버블의 표면에 결합된 면역 체크포인트 저해(checkpoint inhibition)능 항체를 포함하는 면역 마이크로버블 복합체(immuno microbubble complex)로서,초음파의 소노포레시스(sonophoresis)로 면역반응이 부스팅(boosting)된 환경에서 종양세포의 면역 회피를 저해하여 T세포를 활성화 시키는 것을 특징으로 하는, 면역 마이크로버블 복합체.
- 제1항에 있어서,상기 마이크로버블과 항체의 결합은 아미드(amide) 결합, 싸이올(thiol) 간의 결합 또는 바이오틴(biotin)-아비딘(avidin) 결합인 것을 특징으로 하는, 면역 마이크로버블 복합체.
- 제1항에 있어서,상기 마이크로버블은 DSPC(1,2-Distearoyl-sn-glycero-3-phosphorylcholine) 및 DSPE(1,2-distearoyl-sn-glycero-3-phosphoethanolamine)를 포함하는, 면역 마이크로버블 복합체.
- 제3항에 있어서,상기 DSPE는 DSPE-PEG2000-NHS(1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[succinyl(polyethylene glycol)-2000])인 것을 특징으로 하는, 면역 마이크로버블 복합체.
- 제3항에 있어서,상기 DSPC 및 DSPE의 몰(mol) 비율은 6 내지 9:1 내지 4(mol)인 것을 특징으로 하는, 면역 마이크로버블 복합체.
- 제1항에 있어서,상기 초음파는 고강도 집중초음파(high intensity focused ultrasound, HIFU)인 것을 특징으로 하는, 면역 마이크로버블 복합체.
- 제1항에 있어서,상기 면역 체크포인트 저해능 항체는 항 PD-L1(Programmed death ligand-1) 항체, 항 B7-1 항체, 및 항 B7-2 항체로 이루어진 군으로부터 선택되는 하나 이상인 것을 특징으로 하는, 면역 마이크로버블 복합체.
- (a) 인지질을 유기용매에 혼합한 다음 수화하는 단계;(b) 상기 (a) 단계에서 수화된 리포좀 전구체를 분산 후 교반하여 마이크로버블을 제조하는 단계; 및(c) 상기 마이크로버블에 면역 체크포인트 저해(checkpoint inhibition)능 항체를 결합(conjugation)시키는 단계를 포함하는, 면역 마이크로버블 복합체의 제조 방법.
- 제8항에 있어서,상기 인지질에는 DSPC(1,2-Distearoyl-sn-glycero-3-phosphorylcholine) 및 DSPE(1,2-distearoyl-sn-glycero-3-phosphoethanolamine)가 포함되는 것을 특징으로 하는, 면역 마이크로버블 복합체의 제조 방법.
- 제9항에 있어서,상기 DSPC 및 DSPE의 몰(mol) 비율은 6 내지 9:1 내지 4(mol)인 것을 특징으로 하는, 면역 마이크로버블 복합체의 제조 방법.
- 제1항 내지 제7항 중 어느 한 항의 면역 마이크로버블 복합체를 포함하는, 약물 전달 담체.
- 제1항 내지 제7항 중 어느 한 항의 면역 마이크로버블 복합체를 포함하는, 암세포 특이적 초음파, 자기공명영상 또는 형광 분석용 조영제 조성물.
- 제1항 내지 제7항 중 어느 한 항의 면역 마이크로버블 복합체를 포함하는, 암 예방 또는 치료용 약학적 조성물.
- 제13항에 있어서,상기 면역 마이크로버블 복합체는 초음파 처리와 병용하는 것을 특징으로 하는, 암 예방 또는 치료용 약학적 조성물.
- 제14항에 있어서,상기 초음파는 고강도 집중초음파(high intensity focused ultrasound, HIFU)인 것을 특징으로 하는, 암 예방 또는 치료용 약학적 조성물.
- 제13항에 있어서,상기 면역 마이크로버블 복합체는 초음파에 의해 케비테이션(cavitation)되어 암세포 부근의 림프구(lymphocyte)를 밀집시키는 것을 특징으로 하는, 암 예방 또는 치료용 약학적 조성물.
- 생물학적 샘플에 제1항 내지 제7항 중 어느 한 항의 면역 마이크로버블 복합체를 처리하는 단계; 및초음파를 처리하는 단계를 포함하는, 암 진단을 위한 정보제공방법.
- 제17항에 있어서,상기 초음파는 고강도 집중초음파(high intensity focused ultrasound, HIFU)인 것을 특징으로 하는, 암 진단을 위한 정보제공방법.
- 제1항 내지 제7항 중 어느 한 항의 면역 마이크로버블 복합체를 포함하는 조성물을 개체에 투여하는 단계; 및 초음파를 처리하는 단계를 포함하는 암 예방 또는 치료 방법.
- 제1항 내지 제7항 중 어느 한 항의 면역 마이크로버블 복합체를 포함하는 조성물의 암 예방 또는 치료 용도.
- 제1항 내지 제7항 중 어느 한 항의 면역 마이크로버블 복합체의, 암 치료에 이용되는 약제를 생산하기 위한 용도.
