背景技术
鸡球虫病是一种在鸡上发病率和致死率都非常高的寄生虫病,是一种全球性流行的严重危害养鸡产业的重大疾病,每年给养殖户产生巨大的经济影响数以亿计,影响整个养禽行业的发展,造成重大威胁。目前造成鸡球虫病的种类众多,世界公认的有7种,其中毒害艾美耳球虫(Eimeria necatrix)和柔嫩艾美耳球虫(Eimeria tenella)的危害最为严重。
目前我国主要采用抗菌素来预防和治疗鸡球虫病的发生,包括聚醚类载体抗生素和化学合成抗球虫药,如盐霉素、马杜霉素、地克珠利、妥曲珠利、克球粉、尼卡巴嗪、氨丙啉等药物。但是,长期使用抗生素会导致耐药性、药物残留、毒副作用强,且治疗费用支出高等问题。加之,人们现在已经意识到食品安全问题,且农业农村部已出台相关方案,力争通过3年时间,实施养殖环节兽用抗菌药使用减量化行动试点工作,推广兽用抗菌药使用减量化模式,减少使用抗菌药类药物饲料添加剂,兽用抗菌药使用量实现“零增长”,兽药残留和动物细菌耐药问题得到有效控制。
因此,寻求新型的替代产品成为大势所趋。例如,开发中草药(如青蒿、野菊花、常山、柴胡等)、微生态制剂(如乳酸菌)、强毒或弱毒疫苗、植物提取物(如鸦胆子、金丝桃等)。而植物精油作为植物提取物中的一种,鲜有报道。
发明内容
本发明为了解决现有技术长期使用抗生素来预防和治疗禽类球虫病,导致的耐药性、药物残留、毒副作用强和治疗费用高等技术问题,提供了一种禽用植物精油组合物及其制备方法和应用。
本发明解决上述技术问题的技术方案如下:一种禽用植物精油组合物,由如下重量份的原料组成:
香芹酚0-10份、百里香酚0-10份和桉叶油1-10份。

本发明人经多年研究意外发现,将植物精油香芹酚、百里香酚和桉叶油按特定比例组成的植物精油组合物,对禽类球虫病有明显的治疗效果,且长期使用不会产生耐药性、药物残留和毒副作用,治疗费用低,能够作为抗菌素的替代产品在畜牧业生产中发挥重要作用。

上述两组禽用植物精油组合物,所用香芹酚、百里香酚和桉叶油对大多数革兰氏阳性菌和革兰氏阴性菌都有不同程度的抑制作用,抗菌能力很强,作用于细菌细胞的膜结构,改变其通透性,达到杀灭细菌的效果。香芹酚、百里香酚作用于机体肠道,在肠道发挥有益作用,提高生产性能和机体免疫力,缓解坏死性肠炎。而桉叶油具有广谱杀虫效果,是天然杀虫剂研究的方向之一。艾美耳球虫作为一种寄生虫,桉叶油在其方面有抑制作用。三种植物精油组合搭配可达到治疗和预防球虫病的发生,同时使机体肠道健康,最终提高禽类生产性能和机体免疫力。
本发明还提供一种如上所述的禽用植物精油组合物的制备方法,包括:
1)取百里香酚,加热到85℃以上熔融,再加入香芹酚和桉叶油,混合均匀,得混合精油;
2)向1)所得混合精油中加入吸附剂吸附至饱和,包衣或不包衣,即得禽用植物精油组合物。

测定效果实验
1、试验材料和试验设计
供试雏鸡,品种为海兰灰蛋鸡公雏,0日龄开始饲喂基础日粮,从12 日龄开始分组,随机分为八组,每组3个重复,每个重复30只鸡。其中,
第一组为阴性对照组,饲喂基础日粮+沸石粉1000克;
第二组饲喂基础日粮+百球清0.5ml/L;
第三组饲喂基础日粮+12.5%香芹酚和百里香酚(香芹酚5%+百里香酚7.5%/香芹酚8%+百里香酚4.5%)组合物2000克/吨,此精油组合物的制备方法同实施例1;
第四组饲喂基础日粮+5%桉叶油2000克/吨,此精油组合物的制备方法同实施例;
第五组饲喂基础日粮+实施例1组合物500克/吨;
第六组饲喂基础日粮+实施例1组合物1000克/吨;
第七组饲喂基础日粮+实施例1组合物2000克/吨;
第八组为阳性对照组,饲喂基础日粮+沸石粉1000克。
第二组-第八组在14日龄时感染柔嫩艾美耳球虫的孢子化乱囊7万,试验22日龄结束。
本试验由第三方北京农学院安健教授完成,安健教授一直从事鸡球虫病研究20余年,试验为盲试。
2、试验日粮及饲养管理
基础日粮配方为:玉米65wt%、豆粕31wt%、石粉1.6wt%、磷酸氢钙1.1wt%、预混料0.5wt%、氯化钠0.3wt%、赖氨酸0.22wt%、蛋氨酸0.18wt%和氯化胆碱0.1wt%。
上述基础日粮配方中营养成分为:代谢能2823千卡/千克饲料,粗蛋白18.5wt%,钙0.95wt%,磷0.37wt%,盐0.29wt%,赖氨酸1.1wt%,蛋氨酸+胱氨酸0.8wt%。
雏鸡自由采食,自由饮水,按照蛋鸡饲养管理进行。

卵囊比数小于1.0(%)、1-25(%)、26-50(%)、51-75(%)、76-≥100(%)的,卵囊值分别为0、5、10、20和40。
当ACI≥180,判为敏感(有效),
ACI=160-179,判为部分抗药(部分敏感或部分有效),
ACI<160,判为抗药(无效)。
3.5抗球虫指数(ACI)
ACI=(存活率+相对增重率)-(病变值+卵囊值)
4、数据记录及分析
12日龄称取每组鸡只重量,试验22日龄结束后称取鸡只重量,计算每组平均值进行数据分析,判定各组植物精油组合物的敏感性。
5、结果与分析

从表1可以得出,以组1(阴性对照组)为基准,从相对增重率指标表明,组7的相对增重率达到99.05%,说明感染球虫后添加2000g/t实施例1组合物达到未感染球虫效果,组3的相对增重率最低,表明香芹酚和百里香酚组合物的效果明显低于三种精油组合物搭配的效果,组4桉叶油组的相对增重率虽比组3高,但效果仍未达到三种精油组合物;从存活率指标表明,各组均无鸡只死亡,可能与试验周期短有关;从病变分值指标表明,组4的病变最少。
综合各项指标,得出抗球虫指数ACI,表明组3和组5剂量的抗球虫作用无效,组4和组6剂量的抗球虫作用部分有效,组7剂量的抗球虫作用有效,达到与抗菌素组同样效果。因此说明,在日粮中添加2000g/t的实施例1组合物可以达到替代组2抗菌素效果,可以预防和治疗鸡的抗球虫疾病。同时,实施例1组合物效果明显优于香芹酚和百里香酚组合物搭配效果,也优于桉叶油组的效果,说明三种植物精油组合物明显优于单独使用效果。
以上所述仅为本发明的较佳实施例,并不用以限制本发明,凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。

Claims (10)

  1. 一种禽用植物精油组合物,其特征在于,由如下重量份的原料组成:
    香芹酚0-10份、百里香酚0-10份和桉叶油1-10份。
  2. 根据权利要求1所述的禽用植物精油组合物,其特征在于,由如下重量份的原料组成:
    香芹酚3份、百里香酚8份和桉叶油4份。
  3. 根据权利要求1所述的禽用植物精油组合物,其特征在于,由如下重量份的原料组成:
    香芹酚7份、百里香酚3份和桉叶油5份。
  4. 一种如权利要求1-3任一项所述的禽用植物精油组合物的制备方法,其特征在于,包括:
    1)取百里香酚,加热到85℃以上熔融,再加入香芹酚和桉叶油,混合均匀,得混合精油;
    2)向1)所得混合精油中加入吸附剂吸附至饱和,包衣或不包衣,即得禽用植物精油组合物。
  5. 根据权利要求4所述的制备方法,其特征在于,在2)中,所述吸附剂包括白炭黑、环糊精类、改性淀粉和多孔淀粉中的任意一种或几种的混合物。
  6. 根据权利要求4所述的制备方法,其特征在于,在2)中,所述包衣材料为硬脂酸及其盐类和肠溶性缓释包材中的任意一种。
  7. 根据权利要求6所述的制备方法,其特征在于,所述硬脂酸及其盐类包括C16-C18脂肪酸、单硬脂酸甘油酯、氢化脂肪酸和脂肪醇中的任意一种,所述肠溶性缓释包材包括聚丙烯树脂IV和壳聚糖中的任意一种。
  8. 根据权利要求4所述的制备方法,其特征在于,在2)中,还包括当不包衣时,用有机载体吸附剂或无机载体吸附剂吸附禽用植物精油组合物。
  9. 根据权利要求8所述的制备方法,其特征在于,所述无机载体吸附剂包括淀粉类、氧化镁、磷酸氢钙和硅酸盐类中的任意一种或几种的混合物。
  10. 一种预防和治疗禽类球虫病的药物,其特征在于,包括权利要求1-3任一项所述的禽用植物精油组合物。
