Abstract
본 발명은 도코사헥사에노산의 효과를 향상시켜 주는 12-LOX, 또는 12-LOX의 발현 촉진제를 유효성분으로 포함하는 조성물 등에 관한 것으로, 본 발명에 따른 12-LOX, 또는 12-LOX의 발현 촉진제는 도코사헥사에노산의 간 질환 개선, 예방, 치료 효과 등을 현저히 증가시킬 수 있다.
Description
본 발명은 12-LOX, 또는 12-LOX의 발현 촉진제를 유효성분으로 포함하는 간 질환 예방 또는 치료용 조성물에 관한 것이다.
간은 체외에서 들어오는 물질 및 체내의 물질 대사에서 중추적인 역할을 담당하는 생체조직으로서, 최근에는 고열량 및 고지방 음식의 섭취가 현저히 증가되면서 간 질환의 발생이 증가되고 있는 추세이다. 이러한 간 질환에는 지방간증(hepatosteatosis), 간 섬유화(fibrosis), 간경화(cirrhosis), 비알콜성 지방간 질환(nonalcoholic fatty liver disease; NAFLD), 비알콜성 지방간염(nonalcoholic steatohepatitis; NASH) 등이 있으며, 이 중 비알콜성 지방간 질환 및 비알콜성 지방간염은 단순 지방간증과 비교하여 훨씬 중증의 간질환을 의미하나 종종 혼용되어 사용되고 있다. 지방간증은 염증 반응 및 세포 손상을 동반하지 않을 때는 양성(benign) 장애일 가능성이 매우 높으나, 비알콜성 지방간염은 지방간증에 더해, 간 세포 손상 및 염증 반응이 존재하며, 경화증, 간 기능부진 등과 더불어 간암의 발생 위험을 급격히 증가시키기 때문에, 비알콜성 지방간염의 치료제에 대한 관심이 증가되고 있는 추세이다.
최근에는 도코사헥사에노산(docosahexaenoic acid; DHA), 알파 리놀렌산(alpha-linolenic acid; ALA), 에이코사펜타에노산(eicosapentaenoic acid; EPA), 스테아리돈산(stearidonic acid; SDA), 에이코사테트라에노산(eicosatetraenoic acid; ETA) 등과 같은 오메가-3 지방산이 지질의 축척을 감소시켜, 지방간증을 감소시킨다는 연구 결과들이 발표되고 있으나, 임상시험에서 그 효과가 미비하여 치료제로 개발되고 있지 못하고 있는 실정이다. 따라서, 오메가-3 지방산의 치료 효능을 증가시켜줄 수 있는 물질이 있다면, 다양한 간 질환의 예방 및 치료에 효과적으로 사용될 수 있을 것으로 기대된다(대한민국 공개특허 10-2017-0140376).
본 발명은 상기와 같은 종래 기술상의 문제점을 해결하기 위해 안출된 것으로, DHA의 효과를 향상시켜 주는 12-LOX, 또는 12-LOX의 발현 촉진제를 유효성분으로 포함하는 간 질환 개선, 예방 또는 치료용 조성물 등을 제공하는 것을 그 목적으로 한다.
그러나 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 본 발명이 속하는 기술 분야의 통상의 지식을 가진 자에게 명확하게 이해될 수 있을 것이다.
본 발명은 도코사헥사에노산(docosahexaenoic acid; DHA) 및 12-LOX(platelet type 12-Lipoxygenase)를 유효성분으로 포함하는 간 질환의 예방 또는 치료용 약학적 조성물을 제공한다.
또한, 본 발명은 도코사헥사에노산(docosahexaenoic acid; DHA), 및 12-LOX(platelet type 12-Lipoxygenase)의 발현 촉진제를 유효성분으로 포함하는 간 질환의 예방 또는 치료용 약학적 조성물을 제공한다.
또한, 본 발명은 도코사헥사에노산(docosahexaenoic acid; DHA), 및 12-LOX(platelet type 12-Lipoxygenase)를 유효성분으로 포함하는 간 질환 개선 또는 예방용 식품 조성물을 제공한다.
또한, 본 발명은 도코사헥사에노산(docosahexaenoic acid; DHA), 및 12-LOX(platelet type 12-Lipoxygenase)의 발현 촉진제를 유효성분으로 포함하는 간 질환 개선 또는 예방용 식품 조성물을 제공한다.
본 발명의 일 구체예에 있어서, 상기 간 질환은 바람직하게는 고열량 및/또는 고지방 음식의 과다 섭취로 인한 간 질환이며, 더욱 바람직하게는 지방간증, 간 섬유화, 간경화, 비알콜성 지방간 질환, 비알콜성 지방간염 등일 수 있으나, 도코사헥사에노산에 의한 개선, 예방, 또는 치료 효과를 가지는 질환이라면 이에 제한되지 않는다.
본 발명의 다른 구체예에 있어서, 상기 12-LOX의 발현 촉진제는 바람직하게는 12-LOX의 발현량을 증가시킬 수 있는 물질이며, 더욱 바람직하게는 12-LOX를 코딩하는 핵산을 포함하는 재조합 벡터일 수 있으나, 12-LOX의 발현을 촉진시키는 물질이라면 이에 제한되지 않는다. 상기 12-LOX를 코딩하는 핵산은 바람직하게는 서열번호 2 또는 4의 아미노산 서열을 코딩하는 핵산이며, 더욱 바람직하게는 서열번호 1 또는 3의 염기서열을 포함하는 폴리뉴클레오티드이나, 서열번호 2 또는 4의 아미노산 서열로 표시되는 12-LOX 단백질의 기능성 동등물도 본 발명의 권리범위에 포함되며, 상기 기능적 동등물이란, 아미노산의 부가, 치환, 또는 결실의 결과, 상기 아미노산 서열과 적어도 60 % 이상, 바람직하게는 70 % 이상, 더욱 바람직하게는 80 % 이상, 가장 바람직하게는 90 % 이상의 서열 상동성을 가지는 것으로서, 상기 서열번호 2로 표시되는 아미노산 서열과 실질적으로 동질의 활성을 나타내는 폴리펩티드를 의미한다.
또한, 본 발명은 하기 단계를 포함하는 도코사헥사에노산(docosahexaenoic acid; DHA)과의 병용투여용 간 질환 치료제의 선별 방법을 제공한다: (a) 세포를 배양하는 단계; (b) 상기 배양된 세포에 후보 물질을 처리하는 단계; (c) 상기 후보 물질이 처리된 세포의 12-LOX(platelet type 12-Lipoxygenase) 발현량을 측정하는 단계; 및 (d) 상기 12-LOX의 발현량을 증가시킨 후보 물질을 선택하는 단계.
본 발명의 일 구체예에 있어서, 상기 후보 물질은 뉴클레오티드, DNA, RNA, 아미노산, 압타머, 단백질, 화합물, 천연물, 천연추출물, 벡터 등으로 제한이 없다.
또한, 본 발명은 도코사헥사에노산(docosahexaenoic acid; DHA) 및 12-LOX(platelet type 12-Lipoxygenase)를 유효성분으로 포함하는 조성물, 또는 도코사헥사에노산(docosahexaenoic acid; DHA), 및 12-LOX(platelet type 12 Lipoxygenase)의 발현 촉진제를 유효성분으로 포함하는 조성물을 개체에 투여하는 단계를 포함하는 간 질환의 치료 방법을 제공한다.
또한, 본 발명은 도코사헥사에노산(docosahexaenoic acid; DHA) 및 12-LOX(platelet type 12-Lipoxygenase)를 유효성분으로 포함하는 조성물, 또는 도코사헥사에노산(docosahexaenoic acid; DHA), 및 12-LOX(platelet type 12 Lipoxygenase)의 발현 촉진제를 유효성분으로 포함하는 조성물의 간 질환 치료 용도를 제공한다.
본 발명에 따른 12-LOX, 또는 12-LOX의 발현 촉진제는 도코사헥사에노산의 간 질환 개선, 예방, 치료 효과 등을 현저히 증가시킬 수 있기 때문에, 도코사헥사에노산과의 병용투여를 통하여 간 질환의 치료 효과를 현저히 증가시킬 수 있을 것으로 기대되며, 이를 통하여 실질적으로 간 질환의 치료뿐만 아니라, 예방, 개선 등의 목적을 위하여 폭넓게 사용될 수 있을 것으로 기대된다.

본 발명에서는 12-LOX 단백질이 도코사헥사에노산의 간 질환 치료 효과를 현저히 향상시킬 수 있다는 것을 확인하였으므로, 12-LOX 단백질 자체로, 또는 12-LOX의 발현량을 증가시키는 물질을 도코사헥사에노산과 병용투여함으로써, 다양한 간 질환의 효과적인 치료제로서 사용될 수 있다.
본 명세서에 있어서, “재조합 벡터(recombination vector)”란 12-LOX를 발현시킬 수 있도록 제작된 벡터를 총칭하며, 바람직하게는 리포좀, 플라스미드 벡터, 코즈미드 벡터, 박테리오파아지 벡터, 바이러스 벡터 등을 포함하며, 더욱 바람직하게는 바이러스 벡터이나, 12-LOX를 체내에서 발현시킬 수 있는 벡터라면 이에 제한되지 않는다. 상기 바이러스 벡터의 예로는 아데노바이러스(adenovirus), 아데노부속바이러스(adeno-associated virus), 레트로바이러스(retrovirus), 렌티바이러스(lentivirus), 단순포진바이러스(herpes simplex virus), 알파바이러스(alpha virus) 등이 있다.
본 명세서에 있어서, “12-LOX의 발현 촉진제”란 12-LOX에 직접 또는 간접적으로 작용하여 12-LOX의 발현을 개선, 유도, 자극, 증가시키는 물질을 의미하며, 상기 물질의 종류에는 제한이 없다. 상기 물질이 12-LOX의 발현을 촉진시키는 기작은 특별히 제한되지 않으며, 일례로 전사, 번역 등의 유전자 발현을 증대시키거나, 비활성형을 활성형으로 전환시키는 기작으로 작용할 수 있다.
본 명세서에 있어서, “발현량 측정”이란 mRNA 및/또는 단백질의 발현 수준을 측정하는 것으로서, mRNA의 발현 수준을 측정하는 것은 생물학적 시료에서 12-LOX mRNA 존재 여부 및 발현 정도를 확인하는 것으로서, mRNA의 양을 측정함으로써 확인할 수 있다. 이를 위한 분석 방법으로는, RT-PCR, 경쟁적 RT-PCR(competitive RT-PCR), 실시간 RT-PCR(Real-time RT-PCR), RNase 보호 분석법(RPA; RNase protection assay), 노던 블랏팅(northern blotting), DNA 마이크로어레이 칩 등이 있으나, 이들로 한정되는 것은 아니다. 또한, 단백질의 발현 수준을 측정하는 것은 생물학적 시료로부터 12-LOX 단백질의 존재 여부 및 발현 수준을 확인하는 것으로서, 상기 12-LOX에 대하여 특이적으로 결합하는 항체를 이용해 단백질의 양을 확인하거나, 단백질의 활성을 측정함으로써 알 수 있다. 이를 위한 분석방법으로는 웨스턴 블랏팅(western blotting), ELISA(enzyme linked immunosorbent assay), 방사선면역분석법(radioimmunoassay), 방사면역확산법(radioimmunodiffusion), 오우크레로니(Ouchterlony) 면역확산법, 로케트(Rocket) 면역전기동, 면역조직화학염색, 면역침전분석(immunoprecipitation assay), 보체고정분석(complete fixation assay), FACS, 단백질 칩(protein chip) 등이 있으나, 이들로 한정되는 것은 아니다.
본 명세서에 있어서, “예방(prevention)”이란 본 발명에 따른 조성물의 투여에 의해 간 질환을 억제시키거나 발병을 지연시키는 모든 행위를 의미한다.
본 명세서에 있어서, "치료(treatment)"란 본 발명에 따른 조성물의 투여에 의해 간 질환의 증세가 호전되거나 이롭게 변경되는 모든 행위를 의미한다.
본 명세서에 있어서 “개선(improvement)”이란, 치료되는 상태와 관련된 파라미터, 예를 들면, 증상의 정도를 적어도 감소시키는 행위를 의미한다. 이때 상기 본 발명의 조성물을 간 질환의 예방 또는 개선을 위하여 치료를 위한 약제와 동시에 또는 별개로서 사용될 수 있다.
본 명세서에 있어서, “개체(individual)”란 본 발명의 조성물이 투여될 수 있는 대상을 말하며, 그 대상에는 제한이 없다.
본 명세서에 있어서, “약학적 조성물(pharmaceutical composition)”이란 캡슐, 정제, 과립, 주사제, 연고제, 분말 또는 음료 형태임을 특징으로 할 수 있으며, 상기 약학적 조성물은 인간을 대상으로 하는 것을 특징으로 할 수 있다. 상기 약학적 조성물은 이들로 한정되는 것은 아니지만, 각각 통상의 방법에 따라 산제, 과립제, 캡슐, 정제, 수성 현탁액 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있다. 본 발명의 약학적 조성물은 약제적으로 허용가능한 담체를 포함할 수 있다. 약제학적으로 허용되는 담체는 경구투여시에는 결합제, 활탁제, 붕해제, 부형제, 가용화제, 분산제, 안정화제, 현탁화제, 색소, 향료 등을 사용할 수 있으며, 주사제의 경우에는 완충제, 보존제, 무통화제, 가용화제, 등장제, 안정화제 등을 혼합하여 사용할 수 있으며, 국소투여용의 경우에는 기제, 부형제, 윤활제, 보존제 등을 사용할 수 있다. 본 발명의 약학적 조성물의 제형은 상술한 바와 같은 약제학적으로 허용되는 담체와 혼합하여 다양하게 제조될 수 있다. 예를 들어, 경구투여시에는 정제, 트로키, 캡슐, 엘릭서(elixir), 서스펜션, 시럽, 웨이퍼 등의 형태로 제조할 수 있으며, 주사제의 경우에는 단위 투약 앰플 또는 다수회 투약 형태로 제조할 수 있다. 기타, 용액, 현탁액, 정제, 캡슐, 서방형 제제 등으로 제형할 수 있다.
한편, 제제화에 적합한 담체, 부형제 및 희석제의 예로는, 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말디톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로즈, 폴리비닐피롤리돈, 물, 메틸하이드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 또는 광물유 등이 사용될 수 있다. 또한, 충진제, 항응집제, 윤활제, 습윤제, 향료, 유화제, 방부제 등을 추가로 포함할 수 있다.
본 발명에 따른 약학적 조성물의 투여 경로는 이들로 한정되는 것은 아니지만 구강, 정맥내, 근육내, 동맥내, 골수내, 경막내, 심장내, 경피, 피하, 복강내, 비강내, 장관, 국소, 설하 또는 직장이 포함된다. 경구 또는 비경구 투하가 바람직하다. 본원에 사용된 용어 "비경구"는 피하, 피내, 정맥내, 근육내, 관절내, 활액낭내, 흉골내, 경막내, 병소내 및 두개골내 주사 또는 주입기술을 포함한다. 본 발명의 약학적 조성물은 또한 직장 투여를 위한 좌제의 형태로 투여될 수 있다.
본 발명의 약학적 조성물은 사용된 특정 화합물의 활성, 연령, 체중, 일반적인 건강, 성별, 정식, 투여시간, 투여경로, 배출율, 약물 배합 및 예방 또는 치료될 특정 질환의 중증을 포함한 여러 요인에 따라 다양하게 변할 수 있고, 상기 약학적 조성물의 투여량은 환자의 상태, 체중, 질병의 정도, 약물형태, 투여경로 및 기간에 따라 다르지만 당업자에 의해 적절하게 선택될 수 있고, 1일 0.0001 내지 50 mg/kg 또는 0.001 내지 50 mg/kg으로 투여할 수 있다. 투여는 하루에 한번 투여할 수도 있고, 수회 나누어 투여할 수도 있다. 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다. 본 발명에 따른 약학적 조성물은 환제, 당의정, 캡슐, 액제, 겔, 시럽, 슬러리, 현탁제로 제형화될 수 있다.
본 발명에 있어서 식품 조성물은, 각종 식품류, 예를 들어, 음료, 껌, 차, 비타민 복합제, 건강보조 식품류 등에 사용할 수 있으며, 환제, 분말, 과립, 침제, 정제, 캡슐 또는 음료의 형태로 사용할 수 있다. 상기 식품 조성물은 건강기능식품 조성물을 포함한다. 상기 건강기능식품(health functional food)이란 인체에 유용한 기능을 가지는 원료나 성분을 사용해 제조 및/또는 가공되는 식품으로써, 일반적인 식품과 비교하여 그 성분에 특징이 있어, 보다 적극적인 건강 유지, 또는 목적 또는 기대 효과를 갖는 식품을 말한다. 이때, 식품 또는 음료 중의 도코사헥사에노산과 상기 12-LOX, 또는 12-LOX의 발현 촉진제의 양은, 일반적으로 본 발명의 식품 조성물의 경우 전체 식품 중량의 0.01 내지 15 중량%로 가할 수 있으며, 건강 음료 조성물의 경우 100 mL를 기준으로 0.02 내지 10 g, 바람직하게는 0.3 내지 1 g의 비율로 가할 수 있다.
본 발명의 식품 조성물은 당업계에 통상적인 식품첨가제, 예를 들어 향미제, 풍미제, 착색제, 충진제, 안정화제 등을 포함할 수 있다. 본 발명에 따른 식품 조성물은 필수 성분으로서, 상기 도코사헥사에노산과 12-LOX, 또는 12-LOX의 발현 촉진제 외에 첨가되는 성분에는 특별한 제한은 없으며 통상의 식품과 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상기 천연 탄수화물의 예로는 모노사카라이드, 예를 들어, 포도당, 과당 등; 디사카라이드, 예를 들어 말토스, 슈크로스 등; 폴리사카라이드, 예를 들어 덱스트린, 시클로덱스트린; 등과 같은 통상적인 당 및 자일리톨, 소르비톨, 에리트리톨 등의 당알콜이다. 상술한 것 이외의 향미제로서 천연 향미제(타우마틴, 스테비아 추출물(예를 들어, 레바우디오시드 A, 리시르히진 등)) 및 합성 향미제(사카린, 아스파르탐 등)를 유리하게 사용할 수 있다. 상기 천연 탄수화물의 비율은 본 발명의 조성물 100 mL당 일반적으로 약 1 내지 20 g, 바람직하게는 약 5 내지 12 g이다.
상기 외에 본 발명의 식품 조성물은 여러 가지 영양제, 비타민, 광물(전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제 및 중진제(치즈, 초콜릿 등), 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 리세린, 알콜, 탄산 음료에 사용되는 탄산화제 등을 함유할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 이러한 첨가제의 비율은 그렇게 중요하진 않지만 본 발명의 조성물 100 중량부 당 0 내지 약 20 중량부의 범위에서 선택되는 것이 일반적이다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.

간염(hepatitis)과 12-LOX(platelet type 12-Lipoxygenase)와의 연관관계를 확인하기 위하여, US Biomax Inc.으로부터 구입한 인체 생검 슬라이드 시료(LV20812a)를 이용하여 간염 환자에서의 12-LOX의 발현량을 면역화학염색법(immunohistochemistry; IHC)을 통하여 확인하였다. 보다 자세하게는, 간염 환자 슬라이드와 대조군인 정상인의 슬라이드(Normal)를 자일렌(Xylene) 용액에 5분씩 3회 담가 세척하고, 100 % 에탄올(Ethanol)에 3 분씩 3 회, 95 % 에탄올에 2 분씩 2 회, 70 % 에탄올에 2 분씩 2 회, 증류수로 2 분씩 2 회 담가두어 수화(hydration)시켰다. 수화 과정을 마친 슬라이드에 1 : 100으로 희석된 12-LOX 토끼 다중클론 항체(Novus Biologicals)를 처리하고 4 ℃에서 16 시간 동안 반응시키고, 0.3 % 트리톤-X100이 첨가된 인산염완충용액(phosphate buffered saline; PBS)으로 세척하였다. 그리고 1 : 1,000으로 희석된 anti-rabbit IgG를 실온에서 30 분간 처리하여 반응시킨 후에, 다시 0.3 % 트리톤-X100이 첨가된 인산염완충용액을 이용하여 세척하였다. 그리고 1 % 헤마토실린 용액에 30 초 동안 담가두어 염색한 후에 증류수로 1 회 세척하고, 다시 0.25 % 염화수소(HCl)에서 1 초 반응시킨 후에 증류수로 세척하였다. 그리고 헤마토실린 매염제인 10 % 리튬카보네이트를 2~4 초간 반응시킨 후에 증류수로 세척하고, 95% 에탄올과 100% 에탄올을 이용하여 순차적으로 최종 탈수화 및 투명화 단계를 거친 후에 봉입(mounting) 하여 광학현미경으로 관찰하고 사진을 획득하였다. 그 결과는 도 1에 나타내었다.
도 1에 나타난 바와 같이, 간염 환자의 간 조직 시료에서 12-LOX의 발현이 현저히 감소된 것을 확인하였다.
또한, 간 조직 중 어떤 세포에서 12-LOX의 발현이 감소되는지 확인하기 위하여, 면역형광염색법을 실시하였다. 보다 자세하게는, CD68에 대한 항체인 Rat anti Mouse CD68(Santa Cruz)과 Invitrogen Green secondary antibody, 그리고 12-LOX에 대한 항체로 anti-12-LOX(Novus Biologicals)와 Invitrogen Red secondary antibody를 슬라이드에 처리하였다. 1차 항체는 4 ℃에서 18 시간 동안 처리하였고, 2차 항체는 실온에서 2 시간 동안 반응시켰다. 반응이 완료된 후에는 인산염완충용액으로 2회 세척하여 결합되지 않은 항체를 제거하고, DAPI(4',6'-Diamideine-2'-phenylindole) 용액을 1분간 처리하여 핵을 염색하였다. 염색이 완료된 세포는 마운팅 용액으로 마운팅한 후에 형광현미경을 사용하여 발현 여부를 분석하였다. 형광값은 ImageJ를 이용하여 정량화하였으며, 이후 모든 실험 측정값은 평균 ± 표준편차(mean ± standard deviation)로 표시하였고, 통계적 분석은 GraphPad Prism program ver 5.0(GraphPad Software Inc.)을 이용하여 Student's t-test로 대조군과 실험군 간의 차이를 분석하였으며,
p 값이 0.05 미만인 경우 통계적으로 유의한 것으로 판정하였다. 그 결과는 도 2에 나타내었다.
도 2에 나타난 바와 같이, 간염 환자의 간 대식세포에서 12-LOX가 감소되는 것을 확인하였다.
실시예 2: 간염 개선과 12-LOX와의 연관관계 확인
2.1. 12-LOX 활성 저해제와 DHA의 간염 개선 효과와의 연관관계 확인
12-LOX와 도코사헥사에노산의 간염 개선 효과와의 연관관계를 확인하기 위하여, 일차적으로 마우스를 이용하여 실험을 진행하였다. 보다 자세하게는, 비알콜성 지방간염을 유도하기 위하여, 6-7 주령의 C57BL/6 마우스 수컷을 실온 및 습도가 조절되는 환경에 1주일 동안 적응시키고, 고지질 식이(조지방(crude fat) 60 % 함유)를 12 주간 제공하였다. 그리고 간염 개선 효과를 가지고 있다고 알려져 있는 DHA(docosahexaenoic acid) 5 mg/kg 및/또는 12-LOX의 활성 저해제(inhibitor)인 바이칼레인(Baicalein) 5 mg/kg을 2 주간 1 일 1 회 복강 투여하였다. 그리고 마우스들을 마취하고 심장에서 채혈한 다음, 간을 적출하여 4 % 파라포름알데하이드 용액을 이용하여 고정하였다. 그리고 고정된 조직을 세척하고 탈석회(decalcification) 과정 및 탈수화 과정을 거친 후에 파라핀을 이용하여 파라핀 블록으로 제작하였다. 그리고 제작된 파라핀 블록을 3 ㎛의 두께로 잘라 절편을 제작한 후에 60 ℃ 가열블락(Heating block)에 45 분간 놓아두어 탈파라핀을 진행하였다. 그리고 자일렌(Xylene) 용액에 5 분씩 3 회 담가 세척하고, 100 % 에탄올(Ethanol)에 3 분씩 3 회, 95 % 에탄올에 2 분씩 2 회, 70 % 에탄올에 2 분씩 2 회, 증류수로 2 분씩 2 회 담가두어 수화(hydration)시켰다. 수화 과정을 마친 파라핀 절편을 1 % 헤마토실린 용액에 30 초 동안 담가두어 염색한 후에 증류수로 1 회 세척하고, 다시 0.25 % 염화수소(HCl)에서 1 초 반응시킨 후에 증류수로 세척하였다. 그리고 헤마토실린 매염제인 10 % 리튬카보네이트를 2~4 초간 반응시킨 후에 증류수로 세척하였다. 이 후 에오신을 2 초간 처리하여 세포질을 염색한 뒤에 95 % 에탄올과 100 % 에탄올을 이용하여 순차적으로 최종 탈수화 및 투명화 단계를 거친 후에 봉입(mounting) 하여 광학현미경으로 관찰하고 사진을 획득하여 간 조직 내의 지방 축척 상태를 관찰하였다. 그 결과는 도 3에 나타내었다.
도 3에 나타난 바와 같이, 고지질 식이와 DHA를 함께 투여한 실험군(DHA)에서는 간 조직 내에 지방이 축적되지 않았으며, 정상적인 간 형태를 보여주는 반면, 고지질 식이만을 제공한 대조군(Ctrl)과 고지질 식이와 DHA 및 바이칼레인을 병용투여한 실험군에서는 간 내에 지방이 많이 축적되었으며, 간 조직 또한 정상적인 형태가 아닌 것을 확인하였다.
또한, 간하 정맥에서 채취한 혈액을 이용하여 간내 중성지질(triglyceride; TG)의 농도는 EnzyChrom
TM Triglyceride Assay Kit(BioAssay System) 로 측정하고, 간독성 마커인 glutamic pyruvic transaminase(GPT)는 Fuji DRI-CHEM 3500s serum biochemistry analyzer (Fujifilm)를 이용하여 측정하였다. 그 결과는 도 4에 나타내었다.
도 4에 나타난 바와 같이, DHA를 투여한 실험군에서는 중성지질 및 GPT가 모두 유의성 있게 감소된 반면, DHA와 바이칼레인을 병용투여한 실험군에서는 모두 증가된 것을 확인하였다.
상기 결과들을 통하여, DHA에 의한 간염 개선 효과가 12-LOX 활성 저해제에 의하여 유의성 있게 억제된 것을 확인하였으며, 이를 통하여 12-LOX가 DHA에 의한 간염 개선 효과에 영향을 미친다는 것을 확인할 수 있었다.
2.2. 12-LOX 발현용 바이러스 시스템 제작
12-LOX의 과발현이 DHA에 의한 간염 개선 효과를 증진시키는지 확인하기 위하여, HEK293T 세포주에 AAV Helper-Free system(Cell biolabs)을 이용하여 제조사에서 제공되는 프로토콜에 따라 pAAV_RC, pAAV_Helper, 및 pAAV_12-LOX를 형질주입하였다. 12-LOX 서열은 서열번호 1의 염기서열을 이용하였고, 대조군으로는 GFP(green fluorescent protein)를 이용하였다. 형질주입된 벡터는 세포 내에서 rAAV_12-LOX가 자가조립되기 때문에, 이후 용해된 세포로부터 AAV_12-LOX 바이러스를 추출한 뒤 Heparin conjugated agarose beads를 이용하여 AAV_12-LOX를 정제하였다. 정제된 AAV_12-LOX는 표적 세포로 HEK293 세포주에 형질감염시키고, 표적 세포에서의 12-LOX 발현 여부를 웨스턴 블롯팅을 이용하여 확인하였다. 보다 자세하게는, 배양된 표적 세포를 차가운 인산염완충용액을 이용하여 세척하고, RIPA 버퍼를 이용하여 세포를 용해시킨 후에, 용해물을 10 % SDS-PAGE를 이용하여 크기별로 분리하고, 분리된 단백질을 PVDF 멤브레인으로 이동시켰다. 그리고 멤브레인은 블로킹 용액(5 % skim milk)에 침지시켜 1 시간 동안 반응시킨 후에, anti-12-LOX(Novus Biologicals) 또는 anti-actin(Santa Cruz)을 첨가하고 4 ℃에서 16 시간 동안 반응시켰다. 그리고 secondary anti-rabbit IgG(Invitrogen)를 처리하고 1 시간 동안 실온에서 반응시킨 후에, ECL 용액을 이용하여 관찰하였다. 실험의 전반적인 개략도 및 웨스턴 블롯팅 결과는 도 5에 나타내었다.
도 5에 나타난 바와 같이, AAV-12-LOX를 표적 세포에 형질입하면, 표적 세포 내의 염색체 내에 CMV_12-LOX 유전자를 삽입하여 표적 세포 내에서 12-LOX를 발현시킨다는 것을 확인하였다.
2.3. 12-LOX 과발현과 도코사헥사에노산의 간염 개선 효과와의 연관관계 확인
12-LOX와 간염 개선과의 연관관계를 확인하기 위하여, 실시예 2.1과 유사한 방법으로 C57BL/6 마우스 수컷에서 고지질 식이를 16 주간 제공하며, 비알콜성 지방간염을 유도하였다. 그리고 비알콜성 지방간염이 유도된 마우스에 실시예 2.2와 동일한 방법으로 제작된 AAV_12-LOX를 투여하여 12-LOX의 과발현을 유도하고, DHA 1 mg/kg을 3 주간 1 일 1 회 복강 투여하였다. 그리고 마우스들을 마취하고 심장에서 채혈한 다음, 간을 적출하여 사진을 찍고, 간의 무게를 측정하였다. 그 결과는 도 6에 나타내었다. 그리고 적출된 간은 4 % 파라포름알데하이드 용액을 이용하여 고정하고 세척한 뒤, 탈석회(decalcification) 과정 및 탈수화 과정을 거친 후에 파라핀을 이용하여 파라핀 블록으로 제작하였다. 그리고 제작된 파라핀 블록을 3 ㎛의 두께로 잘라 절편을 제작한 후에 간 조직 내의 콜라겐 침착 정도를 확인하기 위하여 시리우스 레드(Sirius red)를 이용하여 염색을 실시하였다. 그 결과는 도 7에 나타내었다. 또한, 획득된 간 조직을 이용하여 간경화 마커인 α-SMA 및 TGFβ의 발현을 웨스턴 블롯팅을 이용하여 확인하였다. 웨스턴 블롯팅은 실시예 2.2와 동일한 방법으로 진행하였으며, 항체로는 anti-α-SMA(Abcam), anti-TGF beta 1(Santa Cruz), anti-12-LOX(Novus Biologicals) 및 anti-actin(Santa Cruz)을 이용하였다. 그 결과는 도 8에 나타내었다. 또한, 실시예 2.1과 동일한 방법으로 심장에서 채취한 혈액을 이용하여 간내 중성지질 및 GPT의 농도를 측정하였다. 그 결과는 도 9에 나타내었다.
도 6에 나타난 바와 같이, 고지질 식이가 제공된 마우스에 DHA를 투여한 대조군(AAV-GFP)에서는 간의 무게가 약간 감소된 반면, DHA의 투여와 함께 12-LOX를 과발현시킨 실험군(AAV_12-LOX)에서는 간의 무게가 현저히 감소되었으며, 간의 형태도 정상에 가까운 것을 확인하였다.
도 7에 나타난 바와 같이, 고지질 식이를 제공한 마우스에서는 콜라겐의 축척이 증가되었으며, DHA를 투여한 실험 군에서는 콜라겐의 축척이 약하게 감소된 것을 확인하였다. 그러나 DHA의 투여와 함께 12-LOX를 과발현시킨 실험군에서는 콜라겐의 축척이 현저히 감소된 것을 확인하였다.
도 8에 나타난 바와 같이, DHA의 투여와 함께 12-LOX를 과발현시킨 실험군에서는 간경화 마커인 α-SMA 및 TGFβ의 발현이 현저히 감소된 것을 확인하였다.
도 9에 나타난 바와 같이, DHA의 투여와 함께 12-LOX를 과발현시킨 실험군에서는 DHA 단독 투여와 비교하여 중성지질 및 GPT의 농도가 유의성 있게 감소된 것을 확인하였다.
상기 결과들을 통하여, 본 발명의 12-LOX는 도코사헥사에노산의 간 질환 개선 효과, 예를 들어, 간섬유화, 간경화, 간염, 비알콜성 지방간염 등의 개선 효과를 현저히 향상시킬 수 있다는 것을 확인하였다. 기존에 도코사헥사에노산이 간에서 지질 축척을 억제하는 것이 확인되었으나, 인체를 대상으로 한 임상시험에서 그 효과가 미비하여 약물로 개발되지 못하였는데, 본 발명을 통해 12-LOX와 병용투여하는 경우 그 치료 효과를 현저히 증가시킬 수 있다는 것을 확인하였다. 이를 통하여, 도코사헥사에노산과 함께, 12-LOX 자체, 또는 12-LOX를 발현시킬 수 있는 치료용 벡터 시스템, 또는 12-LOX를 과발현하는 세포 치료제, 또는 12-LOX의 발현을 촉진시키는 물질을 병용투여함으로써 간 질환의 치료 효과를 현저히 증가시킬 수 있다는 것을 확인할 수 있었다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다.
본 발명에 따른 12-LOX, 또는 12-LOX의 발현 촉진제는 도코사헥사에노산의 간 질환 개선, 예방, 치료 효과 등을 현저히 증가시킬 수 있기 때문에, 도코사헥사에노산과의 병용투여를 통하여 간 질환의 치료 효과를 현저히 증가시켜 다양한 간 질환의 효과적인 치료제 또는 예방제제로서 사용될 수 있다.
Claims (14)
- 도코사헥사에노산(docosahexaenoic acid; DHA) 및 12-LOX(platelet type 12-Lipoxygenase)를 유효성분으로 포함하는, 간 질환의 예방 또는 치료용 약학적 조성물.
- 제 1 항에 있어서,상기 간 질환은 지방간증, 간 섬유화, 간경화, 비알콜성 지방간 질환, 및 비알콜성 지방간염으로 이루어진 군으로부터 선택되는 어느 하나인 것을 특징으로 하는, 약학적 조성물.
- 도코사헥사에노산(docosahexaenoic acid; DHA), 및 12-LOX(platelet type 12-Lipoxygenase)의 발현 촉진제를 유효성분으로 포함하는, 간 질환의 예방 또는 치료용 약학적 조성물.
- 제 3 항에 있어서,상기 12-LOX의 발현 촉진제는 12-LOX의 발현량을 증가시키는 것을 특징으로 하는, 약학적 조성물.
- 제 3 항에 있어서,상기 12-LOX의 발현 촉진제는 12-LOX를 코딩하는 핵산을 포함하는 재조합 벡터인 것을 특징으로 하는, 약학적 조성물.
- 제 3 항에 있어서,상기 간 질환은 지방간증, 간 섬유화, 간경화, 비알콜성 지방간 질환, 및 비알콜성 지방간염으로 이루어진 군으로부터 선택되는 어느 하나 이상인 것을 특징으로 하는, 약학적 조성물.
- 도코사헥사에노산(docosahexaenoic acid; DHA) 및 12-LOX(platelet type 12-Lipoxygenase)를 유효성분으로 포함하는, 간 질환의 개선 또는 예방용 식품 조성물.
- 도코사헥사에노산(docosahexaenoic acid; DHA), 및 12-LOX(platelet type 12-Lipoxygenase)의 발현 촉진제를 유효성분으로 포함하는, 간 질환의 개선 또는 예방용 식품 조성물.
- 제 7 항 또는 제 8 항에 있어서,상기 간 질환은 지방간증, 간 섬유화, 간경화, 비알콜성 지방간 질환, 및 비알콜성 지방간염으로 이루어진 군으로부터 선택되는 어느 하나 이상인 것을 특징으로 하는, 식품 조성물.
- 하기 단계를 포함하는 도코사헥사에노산(docosahexaenoic acid; DHA)과의 병용투여용 간 질환 치료제의 선별 방법:(a) 세포를 배양하는 단계;(b) 상기 배양된 세포에 후보 물질을 처리하는 단계;(c) 상기 후보 물질이 처리된 세포의 12-LOX(platelet type 12 Lipoxygenase) 발현량을 측정하는 단계; 및(d) 상기 12-LOX의 발현량을 증가시킨 후보 물질을 선택하는 단계.
- 도코사헥사에노산(docosahexaenoic acid; DHA) 및 12-LOX(platelet type 12-Lipoxygenase)를 유효성분으로 포함하는 조성물을 개체에 투여하는 단계를 포함하는 간 질환의 예방 또는 치료 방법.
