Abstract
Description
[1] エクオールを有効成分として含む腸内細菌に対する抗菌剤。
[2] 腸内細菌がクロストリディオイデス・ディフィシル(Clostridioides difficile)、クロストリディオイデス・パーフリンゲンス(C. perfringens)及びバクテロイデス・フラジリス(B. fragilis)からなる群から選択される、[1]の抗菌剤。
[3] クロストリディオイデス・ディフィシルの成長を阻害し、かつクロストリディオイデス・ディフィシルの芽胞形成を阻害する[1]又は[2]の抗菌剤。
[4] [1]~[3]のいずれかの抗菌剤を含むクロストリディオイデス・ディフィシル感染症の予防又は治療剤。
[5] エクオールを有効成分として含む整腸剤。
[6] 整腸剤が、腸内細菌における下痢及び/又は腹痛の症状を緩和する、[5]の整腸剤。
[7] エクオールを有効成分として含む大腸癌予防及び/又は治療剤。
クロストリディオイデス・ディフィシル(NAP1/027株)を、100μg/mL、75μg/mL、50μg/mL及び25μg/mLの濃度のエクオールを含む培地に添加し(0時間)、続けて培養し、培養開始後24時間までの成長への影響を確認した。培養条件は、すべて嫌気下、35℃で実施した。
エクオールは腸内細菌によって、ダイジン(daizine)からダイゼイン(daidzein),ジヒドロダイゼイン(dihydro daidzein)を経て生成される最終産物である。実施例1と同様の方法で、エクオール(100μg/mL、50μg/mL)、ダイゼイン(50μg/mL)及びジヒドロダイゼイン(100μg/mL、50μg/mL)のクロストリディオイデス・ディフィシルの成長への阻害を測定した。
027株以外のクロストリディオイデス・ディフィシルの臨床分離株(TUM12745株、TUM12806株及びTUM12855株)もエクオール(100μg/mL)の成長阻害を受けるかどうかを、実施例1と同様の方法で検証した。
クロストリディオイデス・ディフィシル以外の菌にもエクオール(100μg/mL)は阻害するかどうかを、実施例1と同様の方法で検証した。
エクオールのクロストリディオイデス・ディフィシル(NAP1/027株)の形態への影響を実施例1と同様の方法で、エクオール(100μg/mL)を作用させ、クロストリディオイデス・ディフィシルをグラム染色により染色することにより検証した。
クロストリディオイデス・ディフィシルの芽胞形成へのエクオール(100μg/mL)の影響を検証するため、飽和状態までクロストリディオイデス・ディフィシルが成長した培地にエクオールを加えて、形成される芽胞の数を数えた。
