哺乳動物細胞移植治療を必要とする疾患(例えば、外傷性疾患、組織変性疾患、がん)の治療における使用のための、哺乳動物細胞を含む本件哺乳動物細胞保存用液;や、
アカルボース類及び/又はスタキオース類を含む液(好ましくは、等張液)に、哺乳動物細胞を加えるか、或いは、哺乳動物細胞を含む液(好ましくは、等張液)に、アカルボース類及び/又はスタキオース類を加えることにより、本件哺乳動物細胞保存用液を調製する工程を含む、哺乳動物細胞を含む本件哺乳動物細胞保存用液の調製方法;や、
哺乳動物細胞を含む本件哺乳動物細胞保存用液
を挙げることができる。なお、上記移植方法の、哺乳動物細胞を保存する工程は、通常、哺乳動物細胞を含む本件哺乳動物細胞保存用液を、当該保存用液が液体の状態で存在する温度条件下で保存するものであり、当該保存用液が固体の状態で保存する工程(例えば、凍結保存する工程、凍結乾燥保存する工程等の哺乳動物細胞を休眠状態で保存する工程)を含まない。
また、本発明には、アカルボース若しくはその塩、及び/又はスタキオース若しくはその塩を含む、本件哺乳動物細胞保存用液を調製するための粉末製剤が含まれる。当該粉末製剤には上記の任意成分を含んでいてもよい。
本発明者らは、これまでに、哺乳動物細胞を含む液中に、トレハロースを添加すると、溶液の状態で保存したときの細胞生存率低下を抑制できることを報告している(特開2012-115253号公報、国際公開第2014/208053号パンフレット)。そこで、本実施例においては、トレハロース以外の糖類から、細胞生存率低下の抑制効果を示すものを探索した。
[被験用等張液の調製]
93種類の糖類を選択し、分子量が明確な糖類については、87.5mMの濃度となるように、それぞれ乳酸リンゲル液であるラクテック(登録商標)注(大塚製薬工場社製)に添加・融解して調製し、また、分子量が不明な糖類については、4~5%の濃度となるように、それぞれラクテック注に添加・融解し、93種類の糖類を含む等張液を調製した。また、トレハロース(株式会社林原社製)をラクテック注に添加・融解して調製し、88mMトレハロースを含む等張液を調製した。
哺乳動物細胞は、骨髄由来ヒト間葉系幹細胞(Lonza Walkersville社製)(以下、「hMSC」という)を用いた。
hMSCを含むPBSは、以下の手順〔1〕~〔8〕に従って調製した。
〔1〕4×105個のhMSCを、75cm2フラスコに加え、培地存在下で37℃、5%CO2インキュベーターにて培養を行った。顕微鏡下で細胞の状態を観察し、約80%程度コンフルエントになるまで培養した。
〔2〕培地を除き、8mLのPBS(-)でhMSCをリンスした。
〔3〕PBS(-)を除き、3.75mLのトリプシン-EDTA(Lonza社製)を加え、室温で5分間静置した。
〔4〕hMSCが90%程度剥離するまで顕微鏡下で観察しながら、ゆっくりと揺らした。
〔5〕3.75mLの培地を加え、トリプシン反応を停止させ、ピペッティングによりhMSCを回収し、50mL遠心チューブに移した。
〔6〕600×g、5分間、22℃で遠心分離を行った。
〔7〕上清(培地)を除き、1.5mLのPBS(-)を加え、沈殿(hMSC)を懸濁した。
〔8〕細胞数を計測し、5×105cells/mLとなるようにPBS(-)で調整し、hMSCを含むPBSを調製した。
各種被験用等張液中でのhMSCの保存は、以下の手順〔1〕~〔2〕に従って行った。
〔1〕調製したhMSCを含むPBSを、各チューブに0.5mLずつ分注し、遠心分離(600×g、5分間)を行った。
〔2〕上清(PBS)を除き、沈殿(hMSC)を、0.5mLの各種被験用等張液で懸濁し、4℃で3日間保存した。
トリパンブルー染色法によるhMSCの細胞生存率の解析は、以下の手順〔1〕~〔2〕に従って行った。
〔1〕4℃で3日間保存後のhMSCを含む各種被験用等張液のそれぞれから、20μL(1×104cells相当)を採取し、チューブに移した後、トリパンブルー染色液(Gibco社製)20μLを混合した。なお、比較対照として、各種被験用等張液に懸濁する前(4℃で保存前)のhMSCを含むPBSのそれぞれから、20μL(1×104cells相当)を採取し、チューブに移した後、トリパンブルー染色液20μLを混合した。
〔2〕顕微鏡下にてワンセルカウンター(バイオメディカルサイエンス社製)を用いて全細胞数とトリパンブルー陽性細胞(死細胞)数の測定を行い、全細胞数に対するトリパンブルー陰性細胞の割合、すなわち、細胞生存率を算出した。
93種類の糖類のうち、トレハロースよりも細胞生存率低下の抑制効果が高いものとして、アカルボースとスタキオースが得られた。
一方、D-ラクトビオン酸、(-)-2,3-O-イソプロピリデン-D-スレイトール、D-ガラクツロン酸、及びD-マンノサミンについては、乳酸リンゲル液中でhMSCを保存した場合よりも細胞生存率が低下した。
アカルボースとスタキオースは四糖類で共通する。そこで、アカルボースとスタキオース以外の四糖類についても、細胞生存率低下抑制効果があるか否かを確認するために、2種類の四糖(ニストース及びマルトテトラオース)を用いて検討した。
[被験用等張液の調製]
アカルボース(和光純薬工業社製)をラクテック注に添加・融解し、175mMアカルボースを含む等張液を調製した。
また、スタキオースn水和物(和光純薬工業社製)をラクテック注に添加・融解し、175mMスタキオースを含む等張液を調製した。また、ニストース三水和物(和光純薬工業社製)をラクテック注に添加・融解し、175mMニストースを含む等張液を調製した。また、マルトテトラオース(和光純薬工業社製)をラクテック注に添加・融解し、175mMマルトテトラオースを含む等張液を調製した。なお、比較対照として、ラクテック注を用いた。
哺乳動物細胞は、実施例1で使用したhMSCを用いた。
hMSCを含むPBSは、実施例1に記載の方法に従って調製した。
hMSCを、実施例1に記載の方法に従い、各種被験用等張液中に4℃で2日間保存した。
トリパンブルー染色法によるhMSCの細胞生存率の解析は、実施例1に記載の方法に従って行った。
hMSCを、アカルボース又はスタキオースを含むラクテック注中に保存すると、ラクテック注中に保存した場合と比べ、細胞生存率は上昇することが確認されたのに対して、hMSCを、ニストース又はマルトテトラオースを含むラクテック注中に保存しても、かかる細胞生存率の上昇は認められなかった(表1及び図1参照)。
この結果は、アカルボース及びスタキオースによる細胞生存率低下抑制効果は、四糖類に共通するものではなく、アカルボース及びスタキオースに特異的なものであることを示している。
アカルボースによる細胞生存率低下抑制効果と、トレハロースやグルコースによる細胞生存率低下抑制効果とを比較検討した。
[被験用等張液の調製]
アカルボース(和光純薬工業社製)をラクテック注に添加・融解し、88mMアカルボースを含む等張液、及び175mMアカルボースを含む等張液を調製した。また、トレハロース(株式会社林原社製)をラクテック注に添加・融解して調製し、88mMトレハロースを含む等張液、及び175mMトレハロースを含む等張液を調製した。また、グルコース(和光純薬工業社製)をラクテック注に添加・融解し、88mMグルコースを含む等張液、及び175mMグルコースを含む等張液を調製した。なお、比較対照として、生理食塩液である大塚生食注(大塚製薬工場社製)、ラクテック注、及びヒト間葉系幹細胞専用培地キット(MSCGM BulletKit、以下「培地」という)(Lonza Walkersville社製、PT-3001)を用いた。
哺乳動物細胞は、実施例1で使用したhMSCを用いた。
hMSCを含むPBSは、実施例1に記載の方法に従って調製した。
hMSCを、実施例1に記載の方法に従い、各種被験用等張液中に4℃で3日間保存した。
XTT(sodium 3'-[1(-phenylaminocarbonyl)-3,4-tetrazolium]-bis(4-methoxy-6-nitro)benzene sulfonic acid hydrate)法による細胞生存率の解析は、以下の手順〔1〕~〔3〕に従って行った。
〔1〕4℃で3日間保存後のhMSCを含む各種被験用等張液のそれぞれから、100μL(5×104cells相当)を採取し、チューブに移した後、600×g、5分間、22℃で遠心分離を行った。
〔2〕上清(各種被験用等張液)を除き、100μLの培地を加え、沈殿(hMSC)を懸濁した後、96ウェルプレートに移した。
〔3〕細胞増殖キットII(Cat.No.1465015、Roche社製)のXTT混合試薬(XTT試薬と、電子カップリング液とを50:1の比率で混合したもの)を、各ウェルに50μLずつ加えた。
〔4〕24分後にプレートリーダー(Viento XS [KC junior]、大日本製薬社製)を用いて、生細胞により代謝され、産生されるフォルマザン色素の吸光度である480nmの吸光度(A480)と、対照波長の吸光度である650nmの吸光度(A650)とを測定し、A650で補正したA480の吸光度(A480-A650)を算出した。
hMSCを、88mM又は175mMのアカルボースを含むラクテック注中に保存すると、ラクテック注中に保存した場合と比べ、生細胞の指標である「A480-A650」値が高かった(表2及び図2参照)。また、hMSCを、88mM又は175mMのトレハロースやグルコースを含むラクテック注中に保存すると、ラクテック注中に保存した場合と比べ、「A480-A650」値は高かったものの、同じ濃度のアカルボースを含むラクテック注中にhMSCを保存した場合の方が、有意に高かった(表2及び図2参照)。
この結果は、アカルボースを等張液中に添加し、哺乳動物細胞を保存すると、アカルボース非添加の等張液や、トレハロース又はグルコースを添加した等張液中に哺乳動物細胞を保存した場合と比べ、当該液中の哺乳動物細胞の細胞死が効果的に抑制され、生細胞の割合を高めることができることを示している。
44~350mMアカルボースを含む等張液を用いて、アカルボースによる細胞生存率低下抑制効果の至適濃度を検討した。
[被験用等張液の調製]
アカルボース(和光純薬工業社製)をラクテック注に添加・融解し、44mMアカルボースを含む等張液、88mMアカルボースを含む等張液、175mMアカルボースを含む等張液、及び350mMアカルボースを含む等張液を調製した。なお、比較対照として、ラクテック注を用いた。
哺乳動物細胞は、実施例1で使用したhMSCを用いた。
hMSCを含むPBSは、実施例1に記載の方法に従って調製した。
hMSCを、実施例1に記載の方法に従い、各種被験用等張液中に4℃で3日間保存した。
トリパンブルー染色法によるhMSCの細胞生存率の解析は、実施例1に記載の方法に従って行った。
hMSCを、44~175mMのアカルボースを含むラクテック注中に保存すると、細胞生存率は、アカルボースの濃度依存的に上昇したのに対して、hMSCを、175~350mMのアカルボースを含むラクテック注中に保存すると、細胞生存率はほとんど変わらなかった(表3及び図3参照)。
この結果は、少なくとも44mM前後(好ましくは、少なくとも175mM前後)のアカルボースを等張液中に添加し、哺乳動物細胞を保存すると、当該液中の哺乳動物細胞の細胞死を効果的に抑制できることを示している。
アカルボースによる細胞生存率低下抑制効果が、哺乳動物細胞を長期間保存したときも認められるかどうかを確かめるために、hMSCを、アカルボースを含む等張液中に4℃で28日間保存し、細胞生存率を解析した。
[被験用等張液の調製]
アカルボース(和光純薬工業社製)をラクテック注に添加・融解し、44mMアカルボースを含む等張液、88mMアカルボースを含む等張液、及び175mMアカルボースを含む等張液を調製した。また、トレハロース(株式会社林原社製)をラクテック注に添加・融解して調製し、88mMトレハロースを含む等張液を調製した。なお、比較対照として、ラクテック注を用いた。
哺乳動物細胞は、実施例1で使用したhMSCを用いた。
hMSCを含むPBSは、実施例1に記載の方法に従って調製した。
hMSCを、実施例1に記載の方法に従い、各種被験用等張液中に4℃で28日間保存した。
トリパンブルー染色法によるhMSCの細胞生存率の解析は、実施例1に記載の方法に従って行った。
hMSCをラクテック注中に28日間保存すると、細胞生存率は0%であったのに対して、hMSCを、88mMのアカルボースを含むラクテック注中に28日間保存すると、細胞生存率は15.9±5.90%に上昇した(表4及び図4参照)。また、トレハロースを含むラクテック注中にhMSCを28日間保存した場合も、細胞生存率は上昇したものの、アカルボースを含むラクテック注中にhMSCを28日間保存した場合の方がその割合は高かった(表4及び図4参照)。さらに、175mMのアカルボースを含むラクテック注中にhMSCを保存すると、88mMのアカルボースを含むラクテック注中にhMSCを保存した場合と比べ、細胞生存率はさらに上昇することが示された(表4及び図4参照)。
これらの結果は、アカルボースを等張液に添加し、哺乳動物細胞を長期間(4℃で28日間)保存した場合でも、トレハロースを当該等張液に添加し、哺乳動物細胞を長期間保存した場合と比べ、哺乳動物細胞の生存率低下を効果的に抑制できることを示している。
アカルボースによる細胞生存率低下抑制効果を、hMSC以外の哺乳動物細胞(ラット肝細胞)を用いて確認した。
[被験用等張液の調製]
アカルボース(和光純薬工業社製)をラクテック注に添加・融解し、175mMアカルボースを含む等張液を調製した。また、トレハロース(株式会社林原社製)をラクテック注に添加・融解して調製し、175mMトレハロースを含む等張液を調製した。また、グルコース(和光純薬工業社製)をラクテック注に添加・融解し、175mMグルコースを含む等張液を調製した。なお、比較対照として、大塚生食注、ラクテック注、及び培地を用いた。
哺乳動物細胞は、以下の表5に示すラット肝細胞を用いた。
ラット肝細胞を含む洗浄用培地は、以下の手順〔1〕~〔6〕に従って調製した。
〔1〕ラット肝細胞のストックチューブを、液体窒素タンクから取出し、37℃恒温槽で融解した。
〔2〕融解後、すぐに氷上に静置し、50mL遠心チューブに移した。
〔3〕25mLの氷冷した肝細胞洗浄用培地(Hepatocyte Wash Medium、以下「洗浄用培地」という)(GIBCO社製)を滴下した。
〔4〕50×g、3分間、4℃で遠心分離を行った。
〔5〕上清(洗浄用培地)を除き、6mLの洗浄用培地を加え、沈殿(ラット肝細胞)を2~3回ピペッティングにより懸濁した。
〔6〕細胞数を計測し、5×105cells/mLとなるように洗浄用培地で調整し、ラット肝細胞を含む洗浄用培地を調製した。
各種被験用等張液中でのラット肝細胞の保存は、以下の手順〔1〕~〔2〕に従って行った。
〔1〕調製したラット肝細胞を含む洗浄用培地を、各チューブに0.5mLずつ分注し、遠心分離(50×g、3分間、4℃)を行った。
〔2〕上清(洗浄用培地)を除き、沈殿(ラット肝細胞)を、0.5mLの各種被験用等張液で懸濁し、4℃で24時間保存した。
トリパンブルー染色法によるラット肝細胞の細胞生存率の解析は、実施例1に記載の方法において、hMSCをラット肝細胞に代えて行った。
ラット肝細胞を、アカルボースを含むラクテック注中に保存すると、ラクテック注中に保存した場合や、トレハロースやグルコースを含むラクテック注中に保存した場合と比べ、細胞生存率は有意に上昇した(表6及び図5参照)。かかる結果により、アカルボースによる細胞死抑制効果は、間葉系幹細胞(hMSC)に限らず、肝細胞でも認められることから、哺乳動物細胞全般に認められるものであると考えられる。
アカルボースは、αグルコシダーゼの活性部位に結合し、糖の加水分解を阻害する作用を有する。そこで、アカルボースによる細胞生存率低下抑制効果が、哺乳動物細胞中のαグルコシダーゼ活性が阻害された結果、生じたものであるか否かを確認するために、アカルボース以外のα-グルコシダーゼ阻害薬である2種類の物質(ミグリトール及びボグリボース)を用いて検討した。
[被験用等張液の調製]
アカルボース(和光純薬工業社製)をラクテック注に添加・融解し、175mMアカルボースを含む等張液を調製した。また、ミグリトール(東京化成工業社製)をラクテック注に添加・融解し、175mMミグリトールを含む等張液を調製した。また、ボグリボース(東京化成工業社製)をラクテック注に添加・融解し、175mMボグリボースを含む等張液を調製した。なお、比較対照として、ラクテック注を用いた。
哺乳動物細胞は、実施例1で使用したhMSCを用いた。
hMSCを含むPBSは、実施例1に記載の方法に従って調製した。
hMSCを、実施例1に記載の方法に従い、各種被験用等張液中に4℃で3日間保存した。
トリパンブルー染色法によるhMSCの細胞生存率の解析は、実施例1に記載の方法に従って行った。
hMSCを、アカルボースを含むラクテック注中に保存した場合、ラクテック注中に保存した場合と比べ、細胞生存率が上昇することが確認されたのに対して、hMSCを、ミグリトール又はボグリボースを含むラクテック注中に保存した場合、ラクテック注中に保存した場合と比べ、細胞生存率はほとんど変わらなかった(表7及び図6参照)。
この結果は、アカルボースによる細胞生存率低下抑制効果は、哺乳動物細胞中のαグルコシダーゼ活性が阻害された効果、生じたものではないことを示している。
アカルボースが細胞凝集抑制効果を有するか否かを確認するために、アカルボースを含む等張液中にhMSCを保存し、細胞凝集レベルを解析した。
[被験用等張液の調製]
アカルボース(和光純薬工業社製)をラクテック注に添加・融解し、88mMアカルボースを含む等張液を調製した。なお、比較対照として、ラクテック注を用いた。
hMSCを含むPBSは、実施例1に記載の方法に従って調製した。
hMSCを、実施例1に記載の方法に従い、各種被験用等張液中に25℃で48時間保存した。
保存後のhMSCを含む各種被験用等張液を一部採取し、位相差顕微鏡下で3個以上の細胞が集まった細胞凝集塊の有無を観察した。
hMSCをラクテック注中に保存すると、細胞凝集塊の形成が多数認められた(図7の上段、黒丸で囲った箇所参照)。一方、hMSCを、アカルボースを含むラクテック注中に保存すると、細胞凝集塊の形成がほとんど認められなかった(図7の下段参照)。
この結果は、アカルボースを等張液に添加し、哺乳動物細胞を保存すると、当該液中の哺乳動物細胞の細胞凝集を効果的に抑制できることを示している。
アカルボースによる細胞生存率低下抑制効果が、hMSC以外の哺乳動物細胞を保存したときも認められることを確認するために、リンパ球細胞の1つであるCD8陽性T細胞を用いて検討した。
[被験用等張液の調製]
アカルボース(和光純薬工業社製)をラクテック注に添加・融解し、88mMアカルボースを含む等張液を調製した。また、比較対照として、トレハロース(株式会社林原社製)をラクテック注に添加・融解して調製し、88mMトレハロースを含む等張液を調製した。なお、比較対照として、ラクテック注を用いた。
哺乳動物細胞は、CD8陽性T細胞(VERITAS社製)を用いた。
CD8陽性T細胞の拡大培養は、以下の手順〔1〕~〔3〕に従って行った。
〔1〕凍結保存したCD8陽性T細胞を融解後、リンパ球培養培地(LGM3、Lonza社製)で洗浄し、当該培地中で約1時間インキュベート(37℃、5%CO2)した。
〔2〕CD8陽性T細胞を含む培地を分取し、遠心分離(300×g、10分間、室温)を行った。
〔3〕上清を除き、TLY CULTUREキット25(GCリンフォテック社製)に浮遊させ、CD8陽性T細胞を37℃、5%CO2条件下で7日間拡大培養した。
拡大培養したCD8陽性T細胞の保存及び細胞生存率の測定は、以下の手順〔1〕~〔3〕に従って行った。
〔1〕7日間拡大培養したCD8陽性T細胞(細胞濃度が約1.1×106細胞/5mL)を、88mMトレハロースを含むラクテック注で洗浄し、ステムフルチューブ(住友ベークライト社製)に分注後、遠心分離(300×g、10分間、室温)を行った。
〔2〕上清を除き、沈殿(CD8陽性T細胞)を、3種類の被験用等張液(88mMアカルボースを含むラクテック注、88mMトレハロースを含むラクテック注、及びラクテック注)で懸濁した後、5℃で24時間保存した。なお、保存前の細胞生存率を測定するために、懸濁直後に、各ステムフルチューブからCD8陽性T細胞懸濁液を一部(20μL)回収し、トリパンブルー染色液20μLを混合した後、顕微鏡下にてワンセルカウンター(バイオメディカルサイエンス社製)を用いて、1ヵ所の細胞計数部の四隅の細胞計数室のエリアの合計細胞数とトリパンブルー陽性細胞(死細胞)数を計測し、全細胞数に対するトリパンブルー陰性細胞の割合(細胞生存率)を算出した。
〔3〕24時間保存後、上記〔2〕に記載の方法に従って、細胞生存率を計測した。
CD8陽性T細胞を、アカルボースを含むラクテック注中に保存すると、ラクテック注中に保存した場合と比べ、細胞生存率は有意に上昇した(表8及び図8参照)。また、トレハロースを含むラクテック注中にCD8陽性T細胞を保存した場合も、細胞生存率は上昇したものの、アカルボースを含むラクテック注中にCD8陽性T細胞を保存した場合の方がその割合は有意に高かった(表8及び図8参照)。
この結果は、アカルボースによる生存率低下の抑制効果は、CD8陽性T細胞等のリンパ球細胞を保存した場合にも発揮されることを示すとともに、当該効果は、トレハロースよりも高いことを示している。
44~150mMスタキオースを含む等張液を用いて、スタキオースによる細胞生存率低下抑制効果の至適濃度を検討した。
[被験用等張液の調製]
スタキオースn水和物(和光純薬工業社製)をラクテック注に添加・融解し、44mMスタキオースを含む等張液、88mMスタキオースを含む等張液、及び175mMスタキオースを含む等張液を調製した。また、比較対照として、トレハロース(株式会社林原社製)をラクテック注に添加・融解して調製し、88mMトレハロースを含む等張液を調製した。なお、比較対照として、ラクテック注を用いた。
哺乳動物細胞は、実施例1で使用したhMSCを用いた。
hMSCを含むPBSは、実施例1に記載の方法に従って調製した。
hMSCを、実施例1に記載の方法に従い、各種被験用等張液中に5℃で4日間保存した。
トリパンブルー染色法によるhMSCの細胞生存率の解析は、実施例1に記載の方法に従って行った。
hMSCを、スタキオースを含むラクテック注中に保存すると、ラクテック注中に保存した場合と比べ、いずれの濃度のスタキオースにおいても細胞生存率は有意に上昇することや、細胞生存率は、スタキオースの濃度依存的に上昇することが示された(表9及び図9参照)。また、hMSCを、トレハロースを含むラクテック注中に保存した場合も、細胞生存率は上昇したものの、スタキオースを含むラクテック注中にhMSCを保存した場合の方がその割合は高かった(表9及び図9参照)。
この結果は、スタキオースによる細胞生存率低下の抑制効果は、等張液中のスタキオース濃度が、少なくとも44mM前後で発揮されることを示すとともに、当該効果は、トレハロースよりも高いことを示している。
等張液としてラクテック注以外の等張液についても検討を行った。
[被験用等張液の調製]
アカルボース(和光純薬工業社製)を、それぞれ4種類の等張液(ラクテック注、生理食塩液である大塚生食注[大塚製薬工場社製]、リンゲル液「オーツカ」[大塚製薬工場社製]、酢酸リンゲル液であるヴィーンF輸液[扶桑薬品工業社製])に添加・融解し、88mMアカルボースを含む上記4種類の等張液を調製した。また、比較対照として、上記4種類の等張液を用いた。
哺乳動物細胞は、実施例1で使用したhMSCを用いた。
hMSCを含むPBSは、実施例1に記載の方法に従って調製した。
hMSCを、実施例1に記載の方法に従い、各種被験用等張液中に5℃で3日間(図10B参照)又は9日間(図10C参照)保存した。
トリパンブルー染色法によるhMSCの細胞生存率の解析は、実施例1に記載の方法に従って行った。
hMSCを、アカルボースを含む4種類の等張液(ラクテック注、大塚生食注、リンゲル液「オーツカ」、ヴィーンF輸液)中に3日間保存すると、かかる4種類の等張液中に3日間保存した場合と比べ、細胞生存率が上昇し、特に、ラクテック注及びヴィーンF輸液を用いた場合、細胞生存率が有意に上昇することが示された(表10及び図10B参照)。また、hMSCを、アカルボースを含む上記4種類の等張液中に9日間保存すると、かかる4種類の等張液中に9日間保存した場合と比べ、細胞生存率が有意に上昇することが示された(表10及び図10C参照)。
この結果は、アカルボースによる細胞生存率低下抑制効果は、ラクテック注(すなわち、乳酸リンゲル液)に限らず、大塚生食注(すなわち、生理食塩液)、リンゲル液「オーツカ」(すなわち、リンゲル液)、ヴィーンF輸液(すなわち、酢酸リンゲル液)等の等張液全般で発揮されることを示している。

- 哺乳動物幹細胞が、哺乳動物間葉系幹細胞又は哺乳動物リンパ球細胞である、請求項20に記載の保存方法。
- アカルボース若しくはその塩、及び/又はスタキオース若しくはその塩を含む、請求項1~12のいずれか1項に記載の液を調製するための粉末製剤。
