フラボノイド配糖体の分解方法及びフラボノイドの製造方法
 本発明は、フラボノイド配糖体の分解方法及びフラボノイドの製造方法に関する。
 フラボノイドは、天然に存在する有機化合物群であり、柑橘類及び豆類をはじめとして、様々な植物の花、葉、根、茎、果実、種子等に含まれている。フラボノイドは、種類によって特徴及び作用が異なるが、その多くが強い抗酸化作用を有している。例えば、柑橘類に含まれるフラボノイドであるポリメトキシフラボンは、抗酸化作用、発ガン抑制作用、抗菌作用、抗ウイルス作用、抗アレルギー作用、メラニン生成抑制作用、血糖値抑制作用等を有することが知られており、医薬品、健康食品、化粧品等の様々な用途への応用が期待されている。
 柑橘類からフラボノイドを製造する方法としては、例えば、柑橘類の果皮等からエタノール水溶液でフラボノイドを抽出し、抽出されたフラボノイドを溶液中から回収する方法が知られている(例えば、特許文献1参照)。

 しかしながら、従来のフラボノイドの製造方法では、フラボノイドの収率が低いという問題がある。そのため、フラボノイドの収率を向上できる製造方法の開発が求められている。
 例えば柑橘類の果皮には、フラボノイドの他に、それよりも多量のフラボノイド配糖体が含まれているが、これをフラボノイドとして回収できれば、フラボノイドの収率を向上させることが可能である。フラボノイド配糖体をフラボノイドに分解する方法としては、フラボノイド配糖体を塩酸等の酸と反応させる方法が挙げられる。しかしながら、この方法では、使用した酸が残存して製品中に混入するおそれがあること、酸とフラボノイドとの副反応生成物が生じる恐れがあるという問題がある。酸及び副生成物等の不純物を除去する方法としては、分解生成物中のフラボノイドを液体クロマトグラフィーにより分離・精製する方法が挙げられるが、高コストであり且つ生産効率が悪いという問題がある。そのため、酸を用いない新たなフラボノイド配糖体の分解方法が求められている。
 本発明は、上記従来技術の有する課題に鑑みてなされたものであり、酸を用いなくてもフラボノイド配糖体を効率的にフラボノイドに分解できるフラボノイド配糖体の分解方法、及び、フラボノイドの収率を向上できるフラボノイドの製造方法を提供することを目的とする。
 上記目的を達成するために、本発明は、フラボノイド配糖体を含む原料を水熱処理することで、上記フラボノイド配糖体をフラボノイドに分解する、フラボノイド配糖体の分解方法を提供する。
 上記方法によれば、酸を用いることなく、水熱処理によりフラボノイド配糖体を効率的にフラボノイドに分解することができる。また、この方法を用いることで、フラボノイドを低コストで効率的に製造することが可能となる。
 上記方法において、上記フラボノイド配糖体はスダチチン配糖体及び/又はデメトキシスダチチン配糖体を含んでいてもよい。上記方法によれば、スダチチン配糖体及びデメトキシスダチチン配糖体を特に効率的に分解することができる。
 上記方法において、水熱処理温度は110~300℃の範囲内であってもよい。上記範囲内の温度であると、フラボノイド配糖体の分解をより促進することができる。
 本発明はまた、上記本発明の方法によりフラボノイド配糖体を分解する分解工程と、上記分解工程で得られた分解生成物からフラボノイドを抽出する抽出工程と、を含む、フラボノイドの製造方法を提供する。かかる製造方法によれば、フラボノイドを高い収率で、低コスト且つ効率的に製造することができる。
 本発明によれば、酸を用いなくてもフラボノイド配糖体を効率的にフラボノイドに分解できるフラボノイド配糖体の分解方法、及び、フラボノイドの収率を向上できるフラボノイドの製造方法を提供することができる。
 以下、本発明をその好適な実施形態に即して詳細に説明する。但し、本発明は以下の実施形態に限定されるものではない。
 本明細書において、「~」を用いて示された数値範囲は、「~」の前後に記載される数値をそれぞれ最小値及び最大値として含む範囲を示す。本明細書に段階的に記載されている数値範囲において、ある段階の数値範囲の上限値又は下限値は、他の段階の数値範囲の上限値又は下限値と任意に組み合わせることができる。本明細書に記載されている数値範囲において、その数値範囲の上限値又は下限値は、実施例に示されている値に置き換えてもよい。「A又はB」とは、A及びBのどちらか一方を含んでいればよく、両方とも含んでいてもよい。本明細書に例示する材料は、特に断らない限り、1種を単独で又は2種以上を組み合わせて用いることができる。
(フラボノイド配糖体の分解方法)
 本実施形態に係るフラボノイド配糖体の分解方法は、フラボノイド配糖体を含む原料を水熱処理することで、フラボノイド配糖体をフラボノイドに分解する方法である。
 フラボノイド配糖体は、フラボノイドと糖とがグリコシド結合により結合した構造を有する親水性の化合物である。フラボノイド配糖体の元となるフラボノイドは、フェニルクロマン骨格を基本構造とする芳香族化合物であり、フラボン類、フラボノール類、フラバノン類、フラバノノール類、イソフラボン類、アントシアニン類、フラバノール類、カルコン類、オーロン類等が挙げられる。これらの中でも、フラボノイドは、フラボン類であるポリメトキシフラボンであってもよい。
 ポリメトキシフラボンとしては、スダチチン、デメトキシスダチチン、ノビレチン、タンゲレチン、ペンタメトキシフラボン、テトラメトキシフラボン、ヘプタメトキシフラボン等が挙げられる。これらの中でも、ポリメトキシフラボンは、スダチチン、又は、デメトキシスダチチンであってもよい。
 フラボノイド配糖体の元となる糖としては特に限定されず、上述したフラボノイドとグリコシド結合により結合して配糖体を形成することができる公知の糖が挙げられる。
 水熱処理に供する原料は、フラボノイド配糖体以外の他の成分を含んでいてもよい。他の成分としては、例えば、フラボノイド、水溶性食物繊維、難溶性食物繊維、糖類等が挙げられる。原料におけるフラボノイド配糖体の含有量は、原料の固形分全量を基準として、0.1質量%以上であることが好ましく、0.25~30質量%であることがより好ましく、0.5~5質量%であることが更に好ましい。原料がフラボノイドを更に含む場合、フラボノイド配糖体の含有量は、フラボノイドの含有量1質量部に対して、0.25質量部以上であることが好ましく、0.5~100質量部であることがより好ましく、5~50質量部であることが更に好ましい。
 原料として具体的には、植物及び海草の花、葉、根、茎、果実、種子等を用いることができる。特に果皮はポリメトキシフラボン、及びそれらの配糖体を多く含有するため、柑橘果実の搾汁残渣を好適に用いることができる。また、原料は、柑橘類から得られた乾燥粉末であってもよく、柑橘類の果皮から得られた乾燥粉末であってもよい。柑橘類としては、スダチ、温州みかん、ポンカン、シークワサー等が挙げられる。柑橘類は、スダチチン及びデメトキシスダチチン等のポリメトキシフラボン、及びそれらの配糖体を多く含有するスダチであってもよい。
 水熱処理は、原料を水と共に耐圧性の密閉容器内に封入し、密閉したまま100℃を超える温度で加熱することで行うことができる。上記原料及び水を含む反応液が密閉容器内で加熱されることで、密閉容器内が加熱及び加圧環境となり、水熱処理(水熱合成)が行われる。水熱処理は、反応液を撹拌しながら行ってもよい。耐圧性の密閉容器としては、水熱処理に使用可能な公知の容器を特に制限なく用いることができる。密閉容器における反応液の充填率は、高い分解効率を得る観点から、密閉容器の容積を基準として20体積%以上であることが好ましく、40~80体積%であることがより好ましい。
 水の量は、水熱処理を行うのに十分な量であればよく、特に限定されないが、水100質量部に対して原料の固形分が1質量部以上であってもよく、5~100質量部であってもよい。水の量が上記範囲内であると、フラボノイド配糖体の分解を効率的に行うことができる。
 水熱処理の反応条件は特に限定されないが、例えば、110~300℃で0.5~20時間とすることができる。反応温度は、120~190℃であることが好ましく、140~185℃であることがより好ましい。反応温度が110℃以上であると、水熱反応がより良好に発生しやすい傾向があり、300℃以下であると、原料及びフラボノイドの炭化が進行しにくく、収率がより向上する傾向がある。反応時間は、0.5~20時間であることが好ましく、1~10時間であることがより好ましい。反応時間が0.5時間以上であると、反応がより進みやすくなる傾向があり、20時間以下であると、反応の進行とコストとのバランスがとりやすくなる傾向がある。
 水熱処理時の容器内の圧力は、上記反応温度に対応する飽和水蒸気圧又はそれ以上であればよいが、装置の耐圧性の観点から、飽和水蒸気圧であることが好ましい。
 上記条件で水熱処理を行うことで、フラボノイド配糖体をフラボノイドに(より具体的にはフラボノイドと糖に)、効率的に分解することができる。
(フラボノイドの製造方法)
 本実施形態に係るフラボノイドの製造方法は、フラボノイド配糖体を分解する分解工程と、分解工程で得られた分解生成物からフラボノイドを抽出する抽出工程と、を含む。分解工程は、上述した本実施形態に係るフラボノイド配糖体の分解方法によりフラボノイド配糖体を分解する工程である。
 抽出工程では、分解工程で得られた分解生成物からフラボノイドを抽出する。分解生成物には、フラボノイドの他に、糖、分解させずに残ったフラボノイド配糖体、水溶性及び難溶性セルロース並びにその分解物等が含まれている。ここで、フラボノイドは疎水性であるのに対し、糖、フラボノイド配糖体、水溶性セルロース及びその分解物は親水性である。そのため、水熱処理後の水溶液に不溶な成分にはフラボノイドが高濃度で含まれており、水熱処理後に水溶液と不溶分とを分離することで、フラボノイドを濃縮することができる。また、さらに水不溶分を、フラボノイドを溶解する溶媒、例えばエタノール、酢酸エチル、ヘキサン、トルエン等、及び、それらの混合溶媒に溶解し、不溶物をろ過等により除去することで、フラボノイドをさらに抽出・精製することができる。その後、ろ液を乾燥させることにより、高濃度フラボノイドを得ることができる。
 上記方法により、フラボノイドを高い収率で効率的に製造することができる。本実施形態の製造方法で製造されるフラボノイドは、ポリメトキシフラボンであってもよく、スダチチン及び/又はデメトキシスダチチンであってもよい。本実施形態の製造方法は、ポリメトキシフラボン、特にスダチチン及びデメトキシスダチチンの製造に好適であり、その収率を大きく向上させることができる。
 以下、実施例及び比較例に基づいて本発明をより具体的に説明するが、本発明は以下の実施例に限定されるものではない。
(実施例1)
 スダチチン含有量1000質量ppm、デメトキシスダチチン含有量1100質量ppm、配糖体由来スダチチン含有量8000質量ppm、配糖体由来デメトキシスダチチン含有量2000質量ppmであるスダチ果皮エキス粉(池田薬草株式会社製)2gを超純水50gに溶解/分散させ、容量100mlのテフロン(登録商標)容器に封入し、更にそのテフロン(登録商標)容器をステンレス製耐圧容器に収め、耐圧容器を密閉した。密閉した耐圧容器内で、テフロン(登録商標)容器内の溶液をマグネティックスターラーを用いて回転数600rpmで撹拌しながら、溶液の温度が180℃となるようにヒーターで加熱した。180℃到達後、撹拌を続けながら180℃で60分間水熱処理を行った。その後、加熱及び撹拌を止めて常温(25℃)まで自然冷却した。なお、水熱処理中の最高到達温度は181℃であった。冷却後、テフロン(登録商標)容器内の溶液及び固形分をビーカーに取り出し、60℃加温下でダイアフラムポンプを用いて真空乾燥し、粉末状の配糖体分解サンプル1を1.85g得た。
(実施例2~4)
 水熱処理中の溶液の温度を160℃(実施例2)、140℃(実施例3)、120℃(実施例4)としたこと以外は実施例1と同様にして、粉末状の配糖体分解サンプル2(1.9g)、3(1.81g)、4(1.79g)を得た。
(実施例5~8)
 水熱処理の反応時間を600分(10時間)とし、処理中の温度を180℃(実施例5)、160℃(実施例6)、140℃(実施例7)、120℃(実施例8)としたこと以外は実施例1と同様にして、粉末状の配糖体分解サンプル5(1.83g)、6(1.82g)、7(1.75g)、8(1.92g)を得た。
(比較例1)
 実施例1で用いたものと同じスダチ果皮エキス粉(池田薬草株式会社製)2gを比較例1のサンプルとした。
(参考例1)
 実施例1で用いたものと同じスダチ果皮エキス粉(池田薬草株式会社製)2gを1N塩酸50gに溶解/分散させ、マグネティックスターラーを用いて回転数600rpmで撹拌しながら80℃で1時間加熱した後、加熱及び撹拌を止めて常温(25℃)まで自然冷却した。冷却後の反応液を1N水酸化ナトリウム水溶液で中和し、60℃加温下でダイアフラムポンプを用いて真空乾燥して、粉末状の配糖体塩酸分解サンプルを1.74g得た。
<評価方法>
 各実施例、比較例及び参考例で得られたサンプルのスダチチン濃度及びデメトキシスダチチン濃度は、以下の方法で測定した。まず、サンプル0.1gを希釈倍率500となるように、エタノールに溶解/分散させ、孔径0.1μmのPTFEフィルターでろ過して、エタノール溶液を得た。このエタノール溶液について、高速液体クロマトグラフィー(HPLC)により成分分析を行った。標準物質に市販のスダチチン標準精製試料及び市販のデメトキシスダチチン標準精製試料を用いてそれぞれ検量線を作成し、それを用いてサンプル中のスダチチン濃度及びデメトキシスダチチン濃度を概算した。HPLC装置には、日立ハイテク製「クロムマスター」を用いた。結果は表1にまとめて示した。

 表1に示すとおり、実施例1~8は全て、比較例1と比較してスダチチン濃度及びデメトキシスダチチン濃度が上昇しており、スダチチン配糖体及びデメトキシスダチチン配糖体の分解によってスダチチン及びデメトキシスダチチンが新たに生成し、スダチチン及びデメトキシスダチチンの収率を向上させることができることが分かった。また、実施例1~8では、参考例1のように塩酸を用いることなくスダチチン濃度及びデメトキシスダチチン濃度を向上させることができ、条件によっては塩酸を用いた場合と同程度にスダチチン濃度及びデメトキシスダチチン濃度を向上させることができることが分かった。

Claims (4)

  1.  フラボノイド配糖体を含む原料を水熱処理することで、前記フラボノイド配糖体をフラボノイドに分解する、フラボノイド配糖体の分解方法。
  2.  前記フラボノイド配糖体がスダチチン配糖体及び/又はデメトキシスダチチン配糖体を含む、請求項1に記載の分解方法。
  3.  水熱処理温度が110~300℃の範囲内である、請求項1又は2に記載の分解方法。
  4.  請求項1~3のいずれか一項に記載の方法によりフラボノイド配糖体を分解する分解工程と、
     前記分解工程で得られた分解生成物からフラボノイドを抽出する抽出工程と、
    を含む、フラボノイドの製造方法。
