Description
本実施形態に係る配糖体の分解方法は、配糖体を含む原料を溶媒存在下で加熱加圧処理することで、配糖体をアグリコンに分解する分解工程を有し、溶媒が、水と、水に不溶であり且つアグリコンが可溶な疎水性溶媒と含む混合溶媒である。
本実施形態に係るアグリコンの製造方法は、配糖体を分解する分解工程と、分解工程で得られた分解生成物から得られたアグリコンを分離する分離工程を含む。分解工程は、上述した本実施形態に係る配糖体の分解方法により配糖体を分解する工程である。
スダチチン含有量1000質量ppm、配糖体由来スダチチン含有量9000質量ppmであるスダチ果皮エキス粉(池田薬草株式会社製)2gを超純水47.5gとヘキサン2.5g(溶媒混合比95/5)に溶解/分散させた。この溶解/分散液を容量100mlのテフロン(登録商標)容器に封入し、更にそのテフロン(登録商標)容器をステンレス製耐圧容器に収め、耐圧容器を密閉した。密閉した耐圧容器内で、テフロン(登録商標)容器内の溶液をマグネティックスターラーを用いて回転数600rpmで撹拌しながら、溶液の温度が180℃となるようにヒーターで加熱した。180℃到達後、撹拌を続けながら180℃で60分間加熱処理を行った。その後、加熱及び撹拌を止めて常温(25℃)まで自然冷却した。なお、加熱処理中の最高到達温度は181℃であった。
スダチ果皮エキス粉2gを超純水37.5gとヘキサン12.5g(溶媒混合比75/25)に溶解/分散させたこと以外は、実施例1と同様にしてフラボノイド濃縮粉末2を0.022g得た。
スダチ果皮エキス粉2gを超純水25.0gとヘキサン25.0g(溶媒混合比50/50)に溶解/分散させたこと以外は、実施例1と同様にしてフラボノイド濃縮粉末3を0.053g得た。
スダチ果皮エキス粉2gを超純水12.5gとヘキサン37.5g(溶媒混合比25/75)に溶解/分散させたこと以外は、実施例1と同様にしてフラボノイド濃縮粉末4を0.052g得た。
スダチ果皮エキス粉2gを超純水2.5gとヘキサン47.5g(溶媒混合比5/95)に溶解/分散させたこと以外は、実施例1と同様にしてフラボノイド濃縮粉末5を0.05g得た。
玉ねぎ皮粉(淡路島産玉ねぎ粉末、自然健康社製)を超純水とエタノールとの混合溶液(混合比は、超純水:エタノール=50:50)に対して濃度が5重量%になるように分散させ、玉ねぎ皮粉の分散液を得た。得られた分散液を60℃で3時間加熱し、玉ねぎ皮粉に含まれるケルセチンアグリコン及びケルセチン配糖体を抽出した。抽出後、遠心分離機で10000rpm 10分で固液分離し、上澄み液を分取してオーブンで乾燥し、乾燥粉末を得た。得られた乾燥粉末をメノウ乳鉢で粉砕してケルセチンアグリコン7質量%及びケルセチン配糖体1質量%を含む玉ねぎ皮抽出粉末を得た。
スダチ果皮エキス粉2gを超純水25.0gとヘキサン25.0g(溶媒混合比50/50)に溶解/分散させたことに代えて、大豆由来グルコシルセラミド粉(和光純薬製)0.2gを超純水25.0gとヘキサン25.0g(溶媒混合比50/50)に溶解/分散させたこと以外は、実施例3と同様にして粉末状のセラミド濃縮粉末7を0.16g得た。
スダチ果皮エキス粉2gを超純水47.5gとヘキサン2.5g(溶媒混合比95/5)に溶解/分散させたことに代えて、スダチ果皮エキス粉2gを超純水50gに溶解/分散させたこと以外は、実施例1と同様にして作業をおこなったが、乾燥粉末は得られなかった。
スダチ果皮エキス粉2gを超純水47.5gとヘキサン2.5g(溶媒混合比95/5)に溶解/分散させたことに代えて、スダチ果皮エキス粉2gをヘキサン50gに溶解/分散させたこと以外は、実施例1と同様にして作業をおこない粉末状のフラボノイド濃縮粉末8を0.07g得た。
<アグリコン濃度の測定>
各実施例及び比較例で得られた濃縮粉末中のスダチチン、ケルセチン又はセラミドアグリコンの濃度は、以下の方法で測定した。まず、濃縮粉末0.1gを希釈倍率が500倍となるように、エタノールに溶解/分散させ、孔径0.1μmのPTFEフィルターでろ過して、エタノール溶液を得た。このエタノール溶液について、高速液体クロマトグラフィー(HPLC)により成分分析を行った。
Claims (8)
- 配糖体を含む原料を溶媒存在下で加熱加圧処理することで、前記配糖体をアグリコンに分解する分解工程を有し、
前記加熱加圧処理の反応時間が、0.5~20時間であり、
前記溶媒が、水と、水に不溶であり且つ前記アグリコンが可溶な疎水性溶媒と含む混合溶媒であり、
前記疎水性溶媒が、脂肪族炭化水素及び芳香族炭化水素のうち少なくとも一方であり、配糖体の分解方法。 - 前記疎水性溶媒が、ヘキサンである、請求項1に記載の分解方法。
- 前記溶媒及び前記原料の合計質量に占める前記配糖体の質量割合が、0.05質量%以上である請求項1又は2に記載の分解方法。
- 前記配糖体が、フラボノイド配糖体を含む、請求項1~3のいずれか一項に記載の分解方法。
- 前記配糖体が、スダチチン配糖体及び/又はデメトキシスダチチン配糖体を含む、請求項1~4のいずれか一項に記載の分解方法。
- 前記配糖体が、ケルセチン配糖体を含む、請求項1~5のいずれか一項に記載の分解方法。
- 前記加熱加圧処理は、温度110~300℃の条件で行われる、請求項1~6のいずれか一項に記載の分解方法。
- 請求項1~7のいずれか一項に記載の方法により配糖体を分解する分解工程と、
前記分解工程で得られた分解生成物から前記アグリコンを分離する分離工程と、
を含む、アグリコンの製造方法。
