Abstract
本発明の課題は、比較的簡便かつ安価に製造することができる低分子化合物を有効成分とする、哺乳類精子の受精機能増強剤や、比較的簡便かつ安価に製造することができる低分子化合物を用いて、哺乳類精子の受精機能を増強する方法や、哺乳類受精卵を作製する方法を提供することにある。 以下の一般式(I0)、一般式(II)、及び一般式(III)で表される化合物、並びに、R3がOHのときそれらの生理学的に許容される塩からなる群から選択される1種又は2種以上の化合物を含むものを、哺乳類精子の受精機能増強剤とする。
Description
本発明は、哺乳類精子の受精機能増強剤、哺乳類精子の受精機能を増強する方法、及び哺乳類受精卵を作製する方法に関する。
日本においては、晩婚化や晩産化の影響により、4組に1組の夫婦が不妊の問題を抱えているといわれている。このため、人工授精(IUI)による一般的な不妊治療の他、体外受精(IVF)、顕微授精(ICSI)等の高度不妊治療(ART)の実施が増加傾向にある。
WHO(世界保健機関)によると、不妊原因のうち、「男性のみ」に原因がある場合が24%であり、男女双方に原因がある場合を加えると、約50%のケースで男性が関係している。しかしながら、日本の不妊治療は女性側に偏っており、男性側の対策はほとんどなされていないのが現状である。
男性不妊の症例において運動能の低下した精子が確認され、その精子には突然変異を起こしたミトコンドリアゲノムが存在することが報告されている。また、ミトコンドリアゲノムに突然変異を導入したマウスを用いて、ミトコンドリアの機能低下が男性不妊に関連していることも報告されている(非特許文献1)。
一方、本発明者らは、後述する本件化合物群が、エリスロポエチン発現増強効果及びミトコンドリア病の治療効果(特許文献1)や、臓器線維化抑制効果(特許文献2)を有することを報告している。しかしながら、本件化合物群が、哺乳類精子の受精機能の増強効果を有することについては、これまで知られていなかった。

本発明の課題は、比較的簡便かつ安価に製造することができる低分子化合物を有効成分とする、哺乳類精子の受精機能増強剤や、比較的簡便かつ安価に製造することができる低分子化合物を用いて、哺乳類精子の受精機能を増強する方法や、哺乳類受精卵を作製する方法を提供することにある。
本発明者らは、上記課題を解決すべく鋭意研究を続けている。その過程において、後述する本件化合物群が、受精機能が低下した哺乳類精子に対しても、効果的に受精機能を増強する作用を有することを見いだし、本発明を完成するに至った。
すなわち、本発明は以下のとおりである。
〔1〕以下の一般式(I0)、一般式(II)、及び一般式(III)で表される化合物、並びに、R3がOHのときそれらの生理学的に許容される塩からなる群から選択される1種又は2種以上の化合物を含む、哺乳類精子の受精機能増強剤。
[式中、R1は、ベンゼン環が非置換のベンゾイルメチル基であるか、あるいはベンゼン環が炭素数1~7のアルキル基、炭素数1~7のアルコキシル基、フッ素及び/又は塩素で置換されたベンゾイルメチル基;非置換若しくはフッ素で置換された鎖状又は分枝状の炭素数4~6のアルキル基;又は、フェニル基若しくはシクロペンチル基で置換されたメチレン又はエチレン;を表し、前記フェニル基はさらに1以上のフェニル基で置換されていてもよく、Z1、Z2、Z3、Z4は、同一でも異なっていてもよく、水素原子、ハロゲン原子、C1~C6のアルキル基、C2~C6のアルケニル基、C2~C6のアルキニル基、OR8で表される有機オキシ基を表し、R8は、C1~C7のアルキル基、C2~C6のアルケニル基、C2~C6のアルキニル基を表し、Z5は、水素原子又はC1~C6のアルキル基を表し、R3はOH、OR4、NHR4及びNR4R5のいずれか一つから選ばれる基であり、R4及びR5は同一又は異なって、置換若しくは非置換の炭素数1~4のアルキル基である。]

また本発明の実施の他の形態として、哺乳類精子の受精機能増強剤として使用するための、本件化合物群から選択される1種又は2種以上の化合物や、哺乳類精子の受精機能増強における使用のための、本件化合物群から選択される1種又は2種以上の化合物や、哺乳類精子の受精機能増強剤を製造するための、本件化合物群から選択される1種又は2種以上の化合物の使用を挙げることができる。
また本発明の実施の他の形態として、採取された少なくとも1つの哺乳類精子を、本件化合物群から選択される1種又は2種以上の化合物を含む生理的水溶液中に移す工程(a)であって、前記哺乳類精子が非ヒト哺乳類精子である、工程(a)と、前記工程(a)で得られた非ヒト哺乳類精子を、排卵期の雌の非ヒト哺乳類子宮内に注入する(この過程を人工授精[IUI]という)工程(p-1)とを含む、前記雌の非ヒト哺乳類の受精効率及び/又は妊娠効率を上昇させる方法を挙げることができる。
また本発明の実施の他の形態として、採取された少なくとも1つの哺乳類精子を、本件化合物群から選択される1種又は2種以上の化合物を含む生理的水溶液中に移す工程(a)であって、前記哺乳類精子がヒト精子である、工程(a)と、前記工程(a)で得られたヒト精子を、排卵期の女性の子宮内に注入する工程(p-2)とを含む、前記女性の受精効率及び/又は妊娠効率を上昇させる方法を挙げることができる。
本発明によると、哺乳類精子の受精機能を効果的に増強することができ、男性側を原因とする不妊(男性不妊)治療において、低下した精子の受精機能を増強できるため、受精率や妊娠率の向上が期待される。さらに、本件化合物群は、比較的簡便かつ高収率で製造できる低分子化合物を、哺乳類精子の受精機能増強の有効成分とするため、比較的簡便かつ安価に製造することができる点でも優れている。
本発明の哺乳類精子の受精機能増強剤は、「哺乳類精子の受精機能を増強するため」という用途が限定された、本件化合物群から選択される1種又は2種以上の化合物を有効成分として含有する剤(以下、「本件増強剤」ということがある)である。
本発明の哺乳類精子の受精機能の増強方法としては、採取された少なくとも1つの哺乳類精子を、本件化合物群から選択される1種又は2種以上の化合物を含む生理的水溶液(以下、「本件化合物群含有生理的水溶液」ということがある)中に移す工程(a)を含む方法(以下、「本件増強方法」ということがある)であれば特に制限されず、工程(a)で得られた哺乳類精子を、受精又は媒精前に培養する工程(以下、「受精前/媒精前培養工程」ということがある)をさらに含む方法が好ましい。本件増強方法により得られた哺乳類精子は、インビトロでの媒精方法(例えば、体外受精による媒精)や、インビボでの媒精方法(例えば、人工授精)に用いることができる。このため、本件増強方法としては、工程(a)の後や、受精前/媒精前培養工程の後に得られた哺乳類精子を、顕微鏡下でガラスピペット(顕微授精用インジェクションピペット)等を用いて、採取された少なくとも1つの哺乳類卵子内に注入する工程(以下、「顕微授精工程」ということがある)をさらに含むものであってもよい。
本発明の哺乳類受精卵の作製方法としては、本件増強方法により得られた哺乳類精子を、採取された少なくとも1つの哺乳類卵子を含む培養液(以下、「哺乳類卵子含有培養液」ということがある)中で培養(体外受精[IVF]による媒精)する工程(b-1)を含む、インビトロでの方法(以下、「本件作製方法1」ということがある)や、採取された少なくとも1つの哺乳類精子と、採取された少なくとも1つの哺乳類卵子とを、本件化合物群から選択される1種又は2種以上の化合物を含む培養液(以下、「本件化合物群含有培養液」ということがある)中で培養(媒精)する工程(b-2)を含む、インビトロでの方法(以下、「本件作製方法2」ということがある)であれば特に制限されず、その後受精の確認のために、哺乳類受精卵をインビトロで培養する工程も含んでもよい。本件作製方法1及び本件作製方法2は、哺乳類精子と、哺乳類卵子とが培養液中で自然に受精させる体外受精(いわゆる体外受精による媒精)により哺乳類受精卵を作製する方法であり、哺乳類精子を、顕微鏡下でガラスピペット(顕微授精用インジェクションピペット)等を用いて、採取された少なくとも1つの哺乳類卵子内に注入する工程(顕微授精の工程)を含まない。
本件増強方法、本件作製方法1、及び本件作製方法2は、受精卵を女性の子宮に着床させる工程(胚移植[ET])等のいわゆる医師による医療行為を含まない。
本明細書において、「哺乳類精子の受精機能が増強する」とは、哺乳類精子のミトコンドリア機能が上昇し、運動性(より具体的には前進運動性)が増強すること等の要因により、哺乳類卵子との受精率が上昇することを意味する。哺乳類精子の受精機能が増強する作用には、正常なレベルの受精機能がさらに増強する作用の他、低下した受精機能、例えば、健常者精子の前進運動率の下限値32%(文献「WHO精液検査ラボマニュアル第5版(世界保健機構刊行、高度生殖医療技術研究所訳、2010年)」参照)と比べ、低下した精子の前進運動率を増強し、受精機能を改善させる作用が含まれる。つまり、本件増強方法により、哺乳類精子の受精機能が増強した結果、体外受精(IVF)の適応とされた不妊患者が、人工授精(IUI)により妊娠できる可能性がある。また、近年AYA(Adolescent and Young Adult)世代のがんサバイバーに対する妊孕性温存治療(がん生殖医療)が注目されているが、凍結融解操作には精子機能の低下が避けられない。上記「哺乳類精子の受精機能が増強する作用」には、こうしたがんサバイバーの凍結融解精子を利用した生殖補助医療における受精機能増強作用も含まれる。
上記哺乳類としては、ヒトや、マウス、ラット、ハムスター、モルモット等のげっ歯類、ウサギ等のウサギ目、ブタ、ウシ、ヤギ、ウマ、ヒツジ等の有蹄目、イヌ、ネコ等のネコ目、サル、アカゲザル、カニクイザル、マーモセット、オランウータン、チンパンジー等の非ヒト霊長類などの非ヒト哺乳類を例示することができ、中でも、マウス、ブタ、ヒトを好適に例示することができる。
本件増強剤は、液体タイプと非液体タイプとに大別される。液体タイプ増強剤においては、本件化合物群を、溶媒(例えば、精子調整液等の生理的水溶液、培養液)に融解した液体を挙げることができる。また、非液体タイプ増強剤においては、ふつう上記溶媒に添加される紛体等の本件化合物群含有物として構成され、かかる非液体タイプ増強剤をそれぞれ生理的水溶液及び培養液に添加すると、液体タイプ増強剤である本件化合物群含有生理的水溶液及び本件化合物群含有培養液をそれぞれ調製することができる。
上記生理的水溶液としては、体液や細胞液の浸透圧とほぼ同じになるようにナトリウムやカリウムなどによって塩や糖濃度等調整した等張水溶液であれば特に制限されず、例えば、家畜等における精子調整液(精子洗浄濃縮用の密度勾配遠心用液)、例えば、Percoll液等;ヒトの生殖補助医療における精子調整液、例えば、SepaSperm(株式会社 北里コーポレーション、静岡、日本)、PureCeption(SAGE[登録商標]CooperSurgical,Inc.、コネチカット、米国)、Isolate(Irvine Scientific、カリフォルニア、米国)、SupraSperm(ORIGIO[登録商標]CooperSurgical,Inc.、コネチカット、米国)等;生理食塩水;リン酸緩衝化生理食塩水[PBS];トリス緩衝化生理食塩水[TBS];HEPES緩衝化生理食塩水;リンゲル液(乳酸リンゲル液、酢酸リンゲル液、重炭酸リンゲル液等);5%グルコース水溶液;培養液;などを挙げることができ、本件増強方法において、哺乳類精子を受精又は媒精前に培養する場合、培養液が好ましく、本件増強方法において、哺乳類精子を受精又は媒精前に培養しない場合、精子調整液が好ましく、ヒトの生殖補助医療における精子調整液がより好ましい。本明細書において「等張」とは、浸透圧が250~380mOsm/Lの範囲内であることを意味する。
上記培養液としては、哺乳類精子の生存・維持に適したものであればよく、例えば、TYH培養液、HTF培養液、KSOM培養液、Dulbeccos’s PBS培養液、M2培養液、PB1培養液、Hanks培養液、Hepes-TALP培養液、Hoppe&Pitts培養液、m-KRB培養液、HIS培養液、BO培養液、mTALP培養液、mT培養液、MCM培養液、CCM培養液、K-MCM培養液、BWW培養液、Whitten培養液、BMOC培養液、T6培養液、HT6培養液、Bavister-TALP培養液、SOF培養液、Menezo-B2培養液、Ham’s培養液、Medium199培養液、MEM培養液、mWM培養液等、ヒト胚専用の培養液、例えば、10%の血漿タンパク質分画(血漿タンパク質分画[PPF;plasma protein fraction]、ヒトアルブミン[HAS;human albumin]、SSS;serum substitute supplement[Irvine Scientific社製]等)を含むクリベージメディウム(SAGE[登録商標]Cleavage Medium CooperSurgical,Inc.、コネチカット、米国)、スパームウォッシングメディウム(Irvine Scientific、カリフォルニア、米国)、ファティリゼイション(HTF)メディウム(SAGE In-Vitro Fertilization、コネチカット、米国)、Ferticult(登録商標)Sperm Washing Flushing Medium(FertiPro N.V.、ベールネム、ベルギー)、Insemination Medium、NI fertilization Medium(NAKA ivf medium, ナカメディカル、日本)等を挙げることができる。
本件増強剤としては、本件化合物群以外の、哺乳類精子の受精機能増強成分を含むものであってもよいが、本件化合物群単独でも優れた哺乳類精子の受精機能増強効果を発揮するため、本件化合物群以外の、哺乳類精子の受精機能増強成分(例えば、タンパク質、DNA、RNA、植物由来の抽出物)を含まないものが好ましい。
上記工程(a)において、哺乳類精子は、哺乳類精子含有液(例えば、精液、精子浮遊液)であっても、液体中に含まれない哺乳類精子それ自体であってもよく、これら哺乳類精子を、例えば、哺乳類精子の吸引が可能な内径を有するキャピラリー(ガラス製や合成樹脂製等の毛細管)又はポリプロピレン製マイクロピペットチップ内に吸引し、顕微鏡観察用容器上に載置又は接触させた滴状の本件化合物群含有生理的水溶液中に移す(混合又は注入する)ことができる。工程(a)後の哺乳類精子を、受精又は媒精前に培養する場合であって、生理的水溶液が培養液であるときは、通常、そのまま滴状の本件化合物群含有生理的水溶液を培養する。また、工程(a)後の哺乳類精子を、受精又は媒精前に培養若しくは洗浄濃縮する場合であって、生理的水溶液が培養液でないときは、哺乳類精子を、上述の操作によりキャピラリー又はポリプロピレン製マイクロピペットチップ内に吸引し、顕微鏡観察用容器上に載置又は接触させた滴状の本件化合物群含有培養液中か、あるいは、ガラス、高密度ポリプロピレン(PP)遠心分離用コニカルチューブ等に静置した本件化合物群含有培養液中に移(混合又は注入)し、当該滴状の本件化合物群含有培養液中で培養又は洗浄濃縮することが好ましい。本件増強方法により得られた、哺乳類精子を含む本件化合物群含有生理的水溶液(本件化合物群含有培養液)は、哺乳類精子浮遊液として、そのまま、その後の工程(b-1)における培養(体外受精による媒精)や、顕微授精工程に用いることができる。上記顕微授精工程において、採取された哺乳類卵子が、その周りに顆粒膜細胞(卵丘細胞)がついたもの(卵丘細胞-卵子複合体[COC;cumulus oocyte complex])である場合、顆粒膜細胞を除去するため、ヒアルロニターゼで処理した哺乳類卵子が好ましい。上記顕微授精工程において、工程(a)又は受精前/媒精前培養工程後、哺乳類精子を含む滴状の本件化合物群含有生理的水溶液(本件化合物群含有培養液)を、例えば、PVP(polyvinylpyrrolidone)と混合した後、上述の操作によりガラスピペット(顕微授精用インジェクションピペット)内に吸引し、顕微操作用容器上に載置又は接触させた滴状の哺乳類卵子含有生理的水溶液中にて、哺乳類精子を哺乳類卵子へ顕微注入する。
上記工程(b-1)において、本件増強方法により得られた哺乳類精子は、哺乳類精子含有液(例えば、精子浮遊液)であっても、液体中に含まれない哺乳類精子それ自体であってもよく、これら哺乳類精子を、例えば、上述の操作によりキャピラリー又はポリプロピレン製マイクロピペットチップ内に吸引し、顕微鏡観察用容器上に載置又は接触させた滴状の哺乳類卵子含有培養液中、あるいは媒精用培養皿内の哺乳類卵子含有培養液中に、適切な濃度(ヒトの場合、精子濃度が、好ましくは10万~40万/mL)となるように移し(混合又は注入し)、当該本件化合物群含有培養液中で培養(媒精)する。また、上記工程(b-2)において、例えば、採取された哺乳類精子及び哺乳類卵子を、上述の操作により別々に又は共にキャピラリー又はポリプロピレン製マイクロピペットチップ内に吸引し、顕微鏡観察用容器上に載置又は接触させた滴状の本件化合物群含有培養液中に、あるいは媒精用培養皿内の哺乳類卵子含有培養液中に、適切な濃度(ヒトの場合、精子濃度が、好ましくは10万~40万/mL)となるように移し(混合又は注入し)、当該本件化合物群含有培養液中で培養(媒精)する。
上記滴状の本件化合物群含有生理的水溶液(本件化合物群含有培養液)や、上記滴状又は媒精用培養皿内の哺乳類卵子含有生理的水溶液(培養液)は、培養中の液の蒸発を防ぐために、油で被覆することが好ましい。
哺乳類精子及び哺乳類卵子の培養温度は、通常約30~40℃の範囲内であり、好ましくは約37℃である。培養時のCO2濃度は、通常約1~10%の範囲内であり、好ましくは約5%である。また、培養時の湿度は、通常約70~100%の範囲内であり、好ましくは約95~100%の範囲内である。また、培養時のO2濃度は、正常酸素濃度(例えば、18~22%O2)であっても、低酸素濃度(例えば、0~10%O2)であってもよい。
上記油としては、動物、植物、鉱物等から単離され、常温で液体であり、且つ水と相分離する疎水性の物質であればよく、具体的には、ミネラルオイル(鉱物油)、シリコーンオイル、サラダ油、ココナッツ油を挙げることができ、これらの中でもミネラルオイルを好適に例示することができる。
上記顕微鏡観察用容器としては、その一部又は全部が透明性に優れ、位相差顕微鏡による観察が可能な素材からなる容器であれば特に制限されず、また、上記顕微操作用容器としては、その一部又は全部が透明性に優れ、顕微授精が可能な素材からなる容器であれば特に制限されず、これら容器としては、具体的には、ポリメチルメタクリル酸メチル(PMMA)、ポリカーボネート、ポリプロピレン、ポリエチレン、ポリメチルペンテン、ポリスチレン、ポリテトラフルオロエチレン、ABS樹脂、ポリジメチルシロキサン、ポリエチレンテレフタレート、シクロオレフィンポリマー、フッ素樹脂、シリコン等の樹脂、それらの高分子化合物を含む共重合体又はその複合体を素材とする容器や、石英ガラス、パイレックス(登録商標)ガラス、ソーダガラス、ホウ酸ガラス、ケイ酸ガラス、ホウケイ酸ガラス等のガラス類又はその複合体を素材とする容器を挙げることができる。
上記顕微鏡観察用容器や顕微操作用容器の形状としては、例えば、フィルム(シート)形状、シャーレ(ディッシュ)形状、ウェルプレート形状、トレイ形状を挙げることができ、これらの中でもシャーレ(ディッシュ)形状を好適に例示することができる。
上記滴状の本件化合物群含有生理的水溶液(本件化合物群含有培養液)の体積や、上記滴状の哺乳類卵子含有生理的水溶液(培養液)の体積は、通常1.0μL以上であり、好ましくは10μL以上、より好ましくは30μL以上であり、また、数が極めて少ない精子の存在を顕微鏡下で確認しやすくするという観点から、通常500μL以下、好ましくは400μL以下、より好ましくは350μL以下、さらに好ましくは300μL以下である。したがって、上記滴状の本件化合物群含有生理的水溶液(本件化合物群含有培養液)の体積や、上記滴状の哺乳類卵子含有生理的水溶液(培養液)の体積としては、通常1.0~500μLの範囲内であり、好ましくは10~400μLの範囲内であり、より好ましくは100~350μLの範囲内であり、さらに好ましくは100~300μLの範囲内である。ウェル形状の培養又は媒精皿を用いた場合の本件化合物群含有生理的水溶液(本件化合物群含有培養液)の体積や、上記滴状の哺乳類卵子含有生理的水溶液(培養液)の体積としては、使用する培養又は媒精皿のメーカー推奨の体積が好ましい。
本明細書において、「少なくとも1つの哺乳類精子」とは、哺乳類精子の数が1以上であることを意味し、1つの哺乳類精子の他に、例えば、数が1~2、1~3、1~4、1~5、1~6、1~7、1~8、1~9、1~10、1~50、1~100等の範囲内の哺乳類精子も含まれる。
本明細書において、「少なくとも1つの哺乳類卵子」とは、哺乳類卵子の数が1以上であることを意味し、1つの哺乳類卵子の他に、例えば、数が1~2、1~3、1~4、1~5、1~6等の範囲内の哺乳類卵子も含まれる。
本件化合物群含有生理的水溶液(本件化合物群含有培養液)中の本件化合物群の濃度としては、哺乳類精子の受精機能増強効果が発揮される濃度であればよく、哺乳類の種類、哺乳類精子の濃度、哺乳類卵子の個数等によって異なるため、一概に特定することはできないが、例えば、0.01~100μMの範囲内であり、好ましくは0.05~50μM、より好ましくは0.1~10μMである。
本件増強方法において、哺乳類精子を培養する期間としては、哺乳類精子の受精機能増強効果が発揮される期間であればよく、哺乳類の種類、哺乳類精子の濃度、哺乳類卵子の個数等によって異なるため、一概に特定することはできないが、例えば、1分~12時間の範囲内であり、好ましくは5分~6時間、より好ましくは10分~3時間、さらに好ましくは30分~2時間である。
本件作製方法1の工程(b-1)において、本件増強方法により得られた哺乳類精子を、哺乳類卵子含有培養液中で培養(体外受精による媒精)する期間としては、受精が完了するのに十分な期間であればよく、哺乳類の種類、哺乳類精子の濃度、哺乳類卵子の個数等によって異なるため、一概に特定することはできないが、例えば、3~24時間の範囲内であり、好ましくは3~12時間又は12~24時間、通常媒精12~18時間後には受精していることを考慮すると(文献「Nagy ZP, et al. 1994 Human Reproduction 9, 1743-1748.」、「Zsolt P.Nagy et al. 1998 Human Reproduction 13, 1606-1612.」参照)、より好ましくは12~18時間である。
本件作製方法2の工程(b-2)において、哺乳類精子と哺乳類卵子とを、本件化合物群含有培養液中で培養(体外受精による媒精)する期間としては、哺乳類精子の受精機能増強効果が発揮され、かつ受精が完了するのに十分な期間であればよく、哺乳類の種類、哺乳類精子の濃度、哺乳類卵子の個数等によって異なるため、一概に特定することはできないが、例えば、3~24時間の範囲内であり、好ましくは3~12時間又は12~24時間、通常媒精12~18時間後には受精していることを考慮すると、より好ましくは12~18時間である。
本件化合物群に含まれる化合物の詳細な説明は以下に示す。
本発明の一態様において、上記一般式(I0)におけるR1は、ベンゼン環が非置換のベンゾイルメチル基であるか、あるいはベンゼン環が炭素数1~7のアルキル基、炭素数1~7のアルコキシル基、フッ素及び/又は塩素で置換されたベンゾイルメチル基である。かかる「ベンゼン環が炭素数1~7のアルキル基、炭素数1~7のアルコキシル基、フッ素及び/又は塩素で置換されたベンゾイルメチル基」とは、ベンゾイルメチル基のベンゼン環を構成する炭素原子に結合する1個若しくは2個以上の水素原子が、炭素数1~7のアルキル基、炭素数1~7のアルコキシル基、フッ素原子及び/又は塩素原子によって置き換わっていることを意味する。したがって、置換されたベンゼン環とは、ベンゼン環を構成する炭素原子に結合する水素原子のうち1~5個の水素原子が炭素数1~7のアルキル基、炭素数1~7のアルコキシル基、フッ素及び/又は塩素によって置き換えられたベンゼン環である。置換されたベンゼン環が2個以上の置換基を有する場合、当該置換基は同一であっても異なっていてもよい。例えば、1~5個の炭素数1~7のアルキル基で置換されたベンゼン環、1~5個の炭素数1~7のアルコキシル基で置換されたベンゼン環、1~5個のフッ素原子で置換されたベンゼン環、又は1~5個の塩素原子で置換されたベンゼン環を有するベンゾイルメチル基を挙げることができる。また、他の例として、炭素数1~4のアルキル基、炭素数1~4のアルコキシル基、フッ素原子及び塩素原子から選択された合計2~5個の置換基で置換されたベンゼン環を有するベンゾイルメチル基を挙げることができる。ここで、炭素数1~7のアルキル基としては、メチル基、エチル基、プロピル基、イソプロピル基、n-ブチル基、イソブチル基、sec-ブチル基、tert-ブチル基、n-ペンチル基、1-メチルブチル基、2-メチルブチル基、3-メチルブチル基、1-エチルプロピル基、1,1-ジメチルプロピル基、1,2-ジメチルプロピル基、n-ヘキシル基、1-メチルペンチル基、2-メチルペンチル基、3-メチルペンチル基、4-メチルペンチル基、1,1-ジメチルブチル基、1,2-ジメチルブチル基、1,3-ジメチルブチル基、2,2-ジメチルブチル基、2,3-ジメチルブチル基、3,3-ジメチルブチル基、1,1,2-トリメチルプロピル基、1-エチルブチル基、2-エチルブチル基、1-エチル-1-メチルプロピル基、1-エチル-2-メチルプロピル基、n-ヘキシル基、1-メチルペンチル基、2-メチルペンチル基、3-メチルペンチル基、4-メチルペンチル基、1,1-ジメチルブチル基、1,2-ジメチルブチル基、1,3-ジメチルブチル基、2,2-ジメチルブチル基、2,3-ジメチルブチル基、3,3-ジメチルブチル基、1,1,2-トリメチルプロピル基、1-エチルブチル基、2-エチルブチル基、1-エチル-1-メチルプロピル基、1-エチル-2-メチルプロピル基、n-ヘプチル基、1-メチルヘキシル基、2-メチルヘキシル基、3-メチルヘキシル基、4-メチルヘキシル基、5-メチルヘキシル基、1-エチルペンチル基、2-エチルペンチル基、3-エチルペンチル基、4,4-ジメチルペンチル基、1-プロピルブチル基等を挙げることができる。

本発明の他の態様において、上記一般式(I0)におけるR1は、非置換若しくはフッ素で置換された鎖状又は分枝状の炭素数4~6のアルキル基である。非置換若しくはフッ素で置換された鎖状又は分枝状の炭素数4~6のアルキル基としては、n-ブチル基、イソブチル基、sec-ブチル基、tert-ブチル基、n-ペンチル基、1-メチルブチル基、2-メチルブチル基、3-メチルブチル基、1-エチルプロピル基、1,1-ジメチルプロピル基、1,2-ジメチルプロピル基、2,2-ジメチルプロピル基、n-ヘキシル基、1-メチルペンチル基、2-メチルペンチル基、3-メチルペンチル基、4-メチルペンチル基、1,1-ジメチルブチル基、1,2-ジメチルブチル基、1,3-ジメチルブチル基、2,2-ジメチルブチル基、2,3-ジメチルブチル基、3,3-ジメチルブチル基、1,1,2-トリメチルプロピル基、1-エチルブチル基、2-エチルブチル基、1-エチル-1-メチルプロピル基、1-エチル-2-メチルプロピル基及びこれらのフッ素化体を挙げることができ、好ましくは1-エチルブチル基、2-エチルブチル基、1-メチルペンチル基、2-メチルペンチル基、3-メチルペンチル基、4-メチルペンチル基、5-メチルペンチル基、3,3,4,4,4-ペンタフルオロブチル基、4,4,5,5,5-ペンタフルオロペンチル基、5,5,6,6,6-ペンタフルオロヘキシル基であり、より好ましくは2-エチルブチル基、2-メチルペンチル基、3-メチルペンチル基、及び4,4,5,5,5-ペンタフルオロペンチル基であり、最も好ましくは4,4,5,5,5-ペンタフルオロペンチル基である。
本発明の他の態様において、上記一般式(I0)におけるR1は、フェニル基若しくはシクロペンチル基が置換したメチレン又はエチレンであり、前記フェニル基はさらに1又は2以上のフェニル基で置換されていてもよい。フェニル基若しくはシクロペンチル基が置換したメチレン又はエチレンとは、ベンジル基、2-フェネチル基、シクロペンチルメチル基又は2-シクロペンチルエチル基である。1又は2以上のフェニル基が置換したベンジル基又は2-フェネチル基としては、3-フェニルベンジル基、4-フェニルベンジル基、3,5-ジフェニルベンジル基、2-(1,1’-ビフェニル-3-イル)-エチル基、2-(1,1’-ビフェニル-4-イル)-エチル基、及び2-(3,5-ジフェニルフェニル)-エチル基を挙げることができる。上記一般式(I)におけるR1としては、2-フェネエチル基、シクロペンチルメチル基、2-シクロペンチルエチル基及び2-(1,1’-ビフェニル-3-イル)-エチル基を好適に例示することができる。

上記一般式(I0)におけるR4及びR5は、同一又は異なって置換若しくは非置換の炭素数1~4のアルキル基である。置換若しくは非置換の炭素数1~4のアルキル基としては、メチル基、エチル基、プロピル基、イソプロピル基、n-ブチル基、イソブチル基、sec-ブチル基、tert-ブチル基、R4とR5が窒素と一緒になったピロリジンや、これらのメトキシ基、フェニル基、フッ素及び塩素により置換されたものを挙げることができ、好ましくは、メチル基、モノクロロメチル基、エチル基、2-メトキシエチル基、2,2,2-トリクロロエチル基、1-フェニルエチル基、2-フェニルエチル基、メトキシエチル基、イソプロピル基、ヘキサフルオロイソプロピル基、及びピロリジンであり、より好ましくは、メチル基及びエチル基である。
上記一般式(I0)の実施態様として下記の一般式(I)で表される化合物、及び、好ましくは、一般式(1)で表される化合物が例示される。
上記式(1)の化合物において、Z1、Z2、Z3、Z4は、同一でも異なっていてもよく、水素原子、ハロゲン原子、C1~C6のアルキル基、C2~C6のアルケニル基、C2~C6のアルキニル基、OR8で表される有機オキシ基を表し、R8は、C1~C7のアルキル基、C2~C6のアルケニル基、C2~C6のアルキニル基を表し、Z5は、水素原子又はC1~C6のアルキル基を表す。
式(1)におけるハロゲン原子としては、フッ素原子、塩素原子、臭素原子、ヨウ素原子等を挙げることができる。
式(1)におけるC1~C6のアルキル基とは、置換基を有していてもよい炭素数1~6の直鎖状または分岐状のアルキル基を意味し、具体的には、メチル基、エチル基、n-プロピル基、イソプロピル基、n-ブチル基、イソブチル基、sec-ブチル基、tert-ブチル基、n-ペンチル基、イソペンチル基、ネオペンチル基、n-へキシル基等を挙げることができる。
上記「置換基を有していてもよい」の置換基としては、ハロゲン原子、水酸基、カルボキシル基、炭素数1~6のアルキル基、炭素数2~6のアルケニル基、炭素数2~6のアルキニル基、C6~C10のアリール基を挙げることができる。上記炭素数1~6のアルキル基、炭素数2~6のアルケニル基、炭素数2~6のアルキニル基は、式(1)における炭素数1~6のアルキル基、炭素数2~6のアルケニル基、炭素数2~6のアルキニル基と同じである。また、上記C6~C10のアリール基としては、フェニル基、ナフチル基を挙げることができる。
式(1)におけるC2~C6のアルケニル基とは、置換基を有していてもよい炭素数2~6の直鎖状または分岐状のアルケニル基を意味し、具体的には、ビニル基、1-プロペニル基、2-プロペニル基、1-ブテニル基、2-ブテニル基、3-ブテニル基、1,3-ブテニル基、1-ペンテニル基、1-ヘキセニル基等を挙げることができる。
式(1)におけるC2~C6のアルキニル基とは、置換基を有していてもよい炭素数2~6の直鎖状または分岐状のアルキニル基を意味し、具体的には、エチニル基、1-プロピニル基、1-ブチニル基、1-ペンチニル基、1-ヘキシニル基等を挙げることができる。
上記Z1、Z2、Z3、Z4は、水素原子、ハロゲン原子、C1~C6のアルキル基、OR8で表される有機オキシ基が好ましく、水素原子、フッ素原子、塩素原子、メチル基、エチル基、n-プロピル基、イソプロピル基等のC1~C3のアルキル基、OR8で表される有機オキシ基がより好ましい。
上記Z5は、水素原子又はC1~C3のアルキル基が好ましく、水素原子又はメチル基がより好ましい。
上記R8は、C1~C6のアルキル基が好ましく、メチル基、エチル基、n-プロピル基、イソプロピル基等のC1~C3のアルキル基、ベンジル基がより好ましい。
前記式(1)で表される化合物の中でも、好ましくは、以下の式(2)、式(3)、式(4)、式(5)、式(6)で表される化合物又はその塩である。
上記式(2)、式(3)、式(4)、式(5)、式(6)中、Z1、Z2、Z3、Z4、Z5は、式(1)におけるZ1、Z2、Z3、Z4、Z5と同じ定義である。

上記一般式(I0)におけるR1が2,4-ジフルオロベンゾイルメチル基であり、Z1、Z2、Z3、Z4、Z5が水素であり、かつR3がOHのとき、一般式(I0)で表される化合物は、実施例で後述する化合物#5を表し、上記一般式(I0)におけるR1が4-フルオロベンゾイルメチル基であり、Z1、Z2、Z3、Z4、Z5が水素であり、かつR3がOHのとき、一般式(I0)で表される化合物は、実施例で後述する化合物#4を表し、上記一般式(I0)におけるR1が4,4,5,5,5-ペンタフルオロペンチル基であり、Z1、Z2、Z3、Z4、Z5が水素であり、かつR3がOHのとき、一般式(I0)で表させる化合物は、実施例で後述する化合物#21を表し、上記一般式(I0)におけるR1が2-シクロペンチルエチル基であり、Z1、Z2、Z3、Z4、Z5が水素であり、かつR3がOHのとき、一般式(I0)で表される化合物は、実施例で後述する化合物#24を表す。これら化合物の他、一般式(I0)で表される化合物の中で具体的なものとしては、実施例で後述する化合物#2、4、5、及び20や、実施例で後述する化合物#17~19や、実施例で後述する化合物#22及び23や、実施例で後述する化合物#25を挙げることができる。
上記一般式(II)におけるXは、炭素数4~6の直鎖のアルキレン基、即ちブチレン-(CH2)4-、ペンチレン-(CH2)5-、ヘキシレン-(CH2)6-、又は炭素数4のエーテル基であり、炭素数4のエーテル基としては、メチレン-O-プロピレン基、エチレン-O-エチレン基、プロピレン-O-メチレン基を挙げることができ、ブチレン、ヘキシレン及びエチレン-O-エチレン基が好ましい。
上記一般式(II)におけるR4及びR5は、同一又は異なって置換若しくは非置換の炭素数1~4のアルキル基である。置換若しくは非置換の炭素数1~4のアルキル基としては、メチル基、エチル基、プロピル基、イソプロピル基、n-ブチル基、イソブチル基、sec-ブチル基、tert-ブチル基、R4とR5が窒素と一緒になったピロリジンや、これらのメトキシ基、フェニル基、フッ素及び塩素により置換されたものを挙げることができ、好ましくは、メチル基、モノクロロメチル基、エチル基、2,2,2-トリクロロメチル基、1-フェニルエチル基、2-フェニルエチル基、メトキシエチル基、イソプロピル基、ヘキサフルオロイソプロピル基、及びピロリジンであり、より好ましくはメチル基及びエチル基である。
上記一般式(II)におけるXがブチレンであり、R6が水素であり、かつR3がOHのとき、一般式(II)で表させる化合物は、実施例で後述する化合物#15を表す。化合物#15の他、一般式(I)で表される化合物の中で具体的なものとしては、実施例で後述する化合物#13や、実施例で後述する化合物#14を挙げることができる。
上記一般式(III)におけるR7は、炭素数1~5のアルキル基又はベンジル基である。鎖状又は分枝状の炭素数1~5のアルキル基としては、メチル基、エチル基、プロピル基、イソプロピル基、n-ブチル基、イソブチル基、sec-ブチル基、tert-ブチル基、n-ペンチル基、1-メチルブチル基、2-メチルブチル基、3-メチルブチル基、1-エチルプロピル基、1,1-ジメチルプロピル基、1,2-ジメチルプロピル基、及び2,2-ジメチルプロピル基を挙げることができる。また、前記ベンジル基のベンゼン環は1又は2以上の炭素数1~3のアルキル基又は炭素数1~3のアルコキシ基で置換されていてもよい。炭素数1~3のアルキル基としては、メチル基、エチル基、n-プロピル基、及びイソプロピル基を挙げることができ、炭素数1~3のアルコキシ基としては、メトキシ基、エトキシ基、n-プロポキシ基、及びイソプロポキシ基を挙げることができる。上記一般式(III)におけるR7は、好ましくはメチル基、エチル基、プロピル基、n-ブチル基、n-ペンチル基、及び3,5-ジメトキシベンジル基であり、より好ましくは3,5-ジメトキシベンジル基である。
上記一般式(III)においてR4及びR5は、同一又は異なって置換若しくは非置換の炭素数1~4のアルキル基である。置換若しくは非置換の炭素数1~4のアルキル基としては、メチル基、エチル基、プロピル基、イソプロピル基、n-ブチル基、イソブチル基、sec-ブチル基、tert-ブチル基、R4とR5が窒素と一緒になったピロリジンや、これらのメトキシ基、フェニル基、フッ素及び塩素により置換されたものを挙げることができ、好ましくは、メチル基、モノクロロメチル基、エチル基、2,2,2-トリクロロメチル基、1-フェニルエチル基、2-フェニルエチル基、メトキシエチル基、イソプロピル基、ヘキサフルオロイソプロピル基、及びピロリジンであり、より好ましくはメチル基及びエチル基である。
上記一般式(III)におけるAがインドールであり、R7が3,5-ジメトキシベンジル基であり、かつR3がOHのとき、一般式(III)で表させる化合物は、実施例で後述する化合物#35を表す。化合物#35の他、一般式(I0)で表される化合物の中で具体的なものとしては、実施例で後述する化合物#36~38や、実施例で後述する化合物#33及び34を挙げることができる。
本件化合物群から選択される化合物が不斉炭素原子及び軸不斉に係わる不斉点をもつとき、かかる化合物は、考えられ得るすべての光学異性体を含み、それら光学異性体は任意の比で使用することができる。例えば、ある光学活性化合物は、エナンチオマーでもラセミでも任意の割合のエナンチオマー混合物でも使用することができ、不斉点が複数存在するときは、任意の割合のジアステレオマー混合物で使用してもよい。
本件化合物群における生理学的に許容される塩には、アルミニウム、カルシウム、リチウム、マグネシウム、カリウム、ナトリウム及び亜鉛から生成された金属塩や、N,N’-ジベンジルエチレンジアミン、クロロプロカイン、コリン、ジエタノールアミン、エチレンジアミン、N-メチルグルカミン、リジン、プロカイン等から生成された有機塩などが含まれる。
本件化合物群から選択される化合物の合成方法は、以下に例示することができるが、これらの方法に限られず、一般的に知られている合成法を用いることができる。また、以下に示す化合物は、シグマ-アルドリッチ社、東京化成工業、和光純薬、関東化学等から入手することができる。また、反応溶媒、反応温度に関して、特に記載のない場合は、通常その反応に利用される溶媒、温度で反応が行われる。反応は、通常、アルゴン又は窒素雰囲気下で行われる。保護基は、Green&Wuts, “PROTECTIVE GROUPS in ORGANIC SYNTHESIS” 3rded.John Wiley&Sons, Inc.を参照し、用いることできる。
上記一般式(I0)で示される化合物は、置換若しくは非置換のベンゼンと置換若しくは非置換のインドールを出発物質として合成することができる。まず、置換若しくは非置換のベンゼンと無水マレイン酸とを、フリーデル-クラフツ反応を用いて、4-アリール-4-オキソ-2-ブテン酸を合成する。このフリーデル-クラフツ反応は、ルイス酸、リン酸、ポリリン酸等を触媒として作用させることで行い、触媒として好適には塩化アルミニウムが用いられる。反応溶媒としては、塩素系の溶媒が好ましいが、出発物質の置換若しくは非置換のベンゼンを溶媒として用いることもできる。こうして得られた4-アリール-4-オキソ-2-ブテン酸と置換若しくは非置換のインドールとをマイケル反応させることにより、インドール酢酸のα位に置換若しくは非置換のベンゾイルオキシ基が置換した化合物を得て、一般式(I0)で示される化合物の基本骨格を構築することができる。このマイケル反応において、4-アリール-4-オキソ-2-ブテン酸のカルボキシル基は保護されていても保護されていなくてもよく、通常は保護する必要はないが、保護する場合、用いられる保護基としては、メチルエステル、tert-ブチルエステル、2,2,2-トリクロロエチルエステル及びtert-ブチルジメチルシリルエステル等を挙げることができる。一方、インドールの窒素原子も保護されていても保護されていなくてもよく、保護する場合はベンジル系の保護基が好ましく、アミド系の保護基は反応性を下げてしまうため好ましくない。また、マイケル反応は、反応系を加熱することで進行することもできるし、ルイス酸等の触媒を用いることもできる。一般式(I0)で示される化合物の骨格を得た後に、必要であれば保護基を除去することで、一般式(I0)で示される化合物が合成できる。この後、目的に応じて、カルボン酸部分を適宜、エステル化、アミド化又は生理学的に許容される塩とすることもできる。具体的には、次式に示すように、1,3-ジフルオロベンゼン、無水マレイン酸及びインドールから実施例で後述する化合物#5を合成することができる。
上記一般式(I0)で示される化合物の合成方法の他の態様としては、アルコールとインドール酢酸の保護体を出発原料として合成する方法を挙げることができる。アルコールの水酸基は、直接若しくは二段階の反応で、ヨウ素又は臭素へと変換することができる。直接変換する方法としては、これらに限られないが、アルコールに、トリフェニルホスフィン、イミダゾール及びヨウ素(I2)を作用させてヨウ素(I・)を置換させる方法、又はトリフェニルホスフィンと四臭化炭素を作用させて臭素を置換させる方法を挙げることができる。複数工程を経て合成する方法としては、アルコールをメタンスルホン酸、トリフルオロメタンスルホン酸、トルエンスルホン酸等のスルホン酸エステルに誘導した後に、アルカリ金属のヨウ化物塩又はアルカリ金属の臭化物塩を反応させる方法を挙げることができる。こうして得られたハロゲン体にインドール酢酸の保護体から生じたα位のエノラートを求核反応させることで、一般式(I0)で示される化合物の基本骨格を得ることができる。インドール酢酸の保護基としては、カルボキシル基の保護としてメチルエステル、tert-ブチルエステル、2,2,2-トリクロロエチルエステル及びtert-ブチルジメチルシリルエステル等へ誘導化する方法が挙げられる。一方、インドール酢酸のアミン部位は、炭酸アミドとして保護することが好ましく、保護基としては、メトキシカルボニル、エトキシカルボニル、tert-ブトキシカルボニル、ベンジルオキシカルボニル等を挙げることができる。こうして得られたインドール酢酸の保護体に塩基を作用させることでエノラートへと誘導し、生じたエノラートとハロゲン体とを求核反応させることで、一般式(I0)で示される化合物の基本骨格を得ることができる。この求核反応で用いることのできる塩基としては、炭酸リチウム、炭酸ナトリウム、炭酸カリウム、炭酸セシウム等のアルカリ金属の炭酸塩、メチルリチウム、n-ブチルリチウム、sec-ブチルリチウム、tert-ブチルリチウム等のアルキルリチウム、リチウムジイソプロピルアミド、リチウムヘキサメチルジシラザン、ナトリウムヘキサメチルジシラザン、カリウムヘキサメチルジシラザン等のアルカリ金属アミドなどを挙げることができる。用いる塩基によって、使用できる溶媒は異なるが、N,N-ジメチルホルムアミド(DMF)やテトラヒドロフラン(THF)等の非プロトン性極性溶媒が好ましい。また、ヘキサメチルリン酸トリアミド等の添加は反応を促進する効果がある。こうして得られた保護体から保護基を除去することで、目的の化合物を得ることができる。この後に、カルボン酸部分を適宜、エステル化、アミド化又はその生理学的に許容される塩とすることができる。具体的には、次式に示すように、4,4,5,5,5-ペンタフルオロペンタノールと1-メトキシカルボニル-3-インドール酢酸 メチルエステルを出発物質として合成例で後述する化合物#21が合成できる。
本発明における式(1)で表される化合物は、公知の有機化学反応を用いる有機合成手法によって得ることができる。例えば、以下に示すように(E)-4-(2,4-ジフルオロフェニル)-4-オキソ-2-ブテン酸と式(7)で表されるインドール誘導体とをマイケル反応させることにより、式(1)で表される化合物を得ることができる。
(上記式(7)におけるZ1、Z2、Z3、Z4、Z5は、式(1)におけるZ1、Z2、Z3、Z4、Z5と同じ定義である。)
上記(E)-4-(2,4-ジフルオロフェニル)-4-オキソ-2-ブテン酸は、以下に示すように1,3-ジフルオロベンゼンと無水マレイン酸とのフリーデル-クラフツ反応により合成することができる。かかるフリーデル-クラフツ反応は、ルイス酸、リン酸、ポリリン酸等を触媒として作用させることで行い、触媒として好適には塩化アルミニウムが好適に用いられる。
上記式(7)で表されるインドール誘導体は、市販品を用いることができる。市販のインドール誘導体としては、4-フルオロインドール、4-クロロインドール、4-ブロモインドール、6-フルオロインドール、6-クロロインドール、6-ブロモインドール、5-メチルインドール等を挙げることができる。
また、上記式(7)で表されるインドール誘導体は、公知の有機化学反応を用いる有機合成手法によって得ることもできる。例えば、R1、R2、R3、R4、R5がハロゲン原子である場合、市販のインドールに、N-ブロモスクシンイミド、N-クロロスクシンイミド、N-ヨードスクシンイミド等のハロゲン化剤を作用させることにより上記式(7)で表されるインドール誘導体を得ることができる。また、R1、R2、R3、R4がC1~C6のアルキル基、C2~C6のアルケニル基、C2~C6のアルキニル基、OR8で表される有機オキシ基である場合、上述のように市販のインドールをハロゲン化した後、アルキルリチウム等の有機リチウム試薬との反応、鈴木-宮浦カップリング反応等によって、上記式(7)で表されるインドール誘導体を得ることができる。さらに、R5がC1~C6のアルキル基である場合、ブロモメタン、ブロモエタン等のC1~C6のハロゲン化アルキルと市販のインドールを反応させることにより、上記式(7)で表されるインドール誘導体を得ることができる。
上記のすべての有機反応は、それぞれ溶媒中で行うことができるが、溶媒は反応温度や反応物等によって適宜選択される。また、上記有機反応の反応温度は、用いる溶媒の沸点等の条件によって適宜選択される。上記有機反応で溶媒を用いる場合、得られた反応溶液を必要に応じて濃縮した後、残渣をそのまま次の反応に使用してもよく、適宜な後処理を行った後に、式(1)で表される化合物として用いてもよい。後処理の具体的な方法としては、抽出処理及び/又は晶出、再結晶、クロマトグラフィー等の公知の精製を挙げることができる。
上述の一般式(I0)で示される化合物の合成方法は、一般式(II)で示される化合物を合成するために用いることもできる。すなわち、一般式(II)で示される化合物は、上述の一般式(I0)で示される化合物の合成方法において、出発原料として用いられるアルコールやインドール酢酸の保護体の代わりに、アミノ基がtert-ブトキシカルボニルで保護された直鎖のアミノアルコール又は鎖中に酸素を有する直鎖のアミノアルコールや、α位にメチル基が置換したインドール酢酸の保護体を出発原料として用い、同様の方法で合成することができる。直鎖のアミノアルコール及び鎖中に酸素を有する直鎖のアミノアルコールのtert-ブトキシカルボニルアミドへの変換は、定法により行うことができるが、通常炭酸ジtert-ブチルを用いる。α位にメチル基が置換したインドール酢酸の保護体は、上記の一般式(I0)で示される化合物の合成方法において、ハロゲン体をヨウ化メチルとしたときに得られる中間体であることは、当業者には容易に理解される。こうして調製した出発原料を用い、一般式(I0)で示される化合物の合成方法と同様の方法で、一般式(II)で示される化合物を合成することができる。具体的には、次式に示すように、4-アミノブタノールと1-メトキシカルボニル-3-インドール酢酸 メチルエステルを出発物質として実施例で後述する化合物#15が合成できる。
上記一般式(III)で示される化合物は、Aがインドール又はナフタレンであるときに共通して、5-ヒドロキシ-3-インドール酢酸エステル又はα-(7-ヒドロキシ-1-ナフタレニル)-酢酸エステルを出発原料として、合成することができる。5-ヒドロキシ-3-インドール酢酸エステル及びα-(7-ヒドロキシ-1-ナフタレニル)-酢酸エステルは、対応するカルボン酸をエステル化することによって得ることができるが、5-ヒドロキシ-3-インドール酢酸は活性プロトンを3つ、α-(7-ヒドロキシ-1-ナフタレニル)-酢酸は活性プロトンを2つ有し、反応の選択性が問題となる。このため、これら化合物のアルコール部分を保護し、エステル化を行った後に、保護基を除去し、出発原料を得ることもできる。また、E.Tsuda et. al.,“Alkoxy-auxins are selective inhibitors of auxin transport mediated by PIN, ABCB, and AUX1 transporters” Journal of Biological Chemistry, 286(3), 2354-2364; 2011.に記載の方法に従って、α-(7-ヒドロキシ-1-ナフタレニル)-酢酸 エチルエステルを合成することもできる。その他にも、5-ヒドロキシ-3-インドール酢酸エステルの合成法として、乾燥させたアルコール中で酸性条件下反応を行うことにより、良好な選択性で、溶媒として用いたアルコールとのエステルを合成できる。前記エステル化の反応条件としては、市販の塩酸/メタノールや、脱水したアルコールに乾燥した塩酸を吹き込む方法を挙げることができるが、予備乾燥したアルコールに酸クロライドを滴下し、系中で酸を発生させる方法が好ましい。この後に、カルボン酸部分を適宜、エステル化、アミド化又はその生理学的に許容される塩とすることができる。こうして準備した出発原料とヨウ化アルキル又は臭化アルキルとを反応させることで、一般式(III)で示される化合物の基本骨格を構築できる。これら5-ヒドロキシ-3-インドール酢酸エステル、又は7-ヒドロキシ-1-ナフタレニル酢酸エステルとヨウ化アルキル又は臭化アルキルとの反応に用いられる塩基としては、水素化ナトリウムや、炭酸リチウム、炭酸ナトリウム、炭酸カリウム、炭酸セシウムといったアルカリ金属の炭酸塩が挙げられる。反応溶媒としては、DMFやTHF等の非プロトン性極性溶媒が好ましい。こうして、一般式(III)で示される化合物の骨格を得た後に、必要であれば保護基を除去することで、一般式(III)で示される化合物が合成できる。この後、目的に応じて、カルボン酸部分を適宜、エステル化、アミド化又は生理学的に許容される塩とすることもできる。具体的には、次式に示すように、出発物質として1-ヨードブタンとα-(7-ヒドロキシ-1-ナフタレニル)-酢酸 エチルエステルを用いて実施例で後述する化合物#34を合成できる。
同様に、出発物質として3,5-ジメトキシ臭化ベンジルと7-ヒドロキシ-3-インドール酢酸を用いて実施例で後述する化合物#35を合成できる。
本件化合物群としては、本願明細書の実施例において、その効果が具体的に示されている化合物#5又は生理学的に許容される塩が好ましい。
以下、実施例により本発明をより具体的に説明するが、本発明の技術的範囲はこれらの例示に限定されるものではない。
1.本件化合物群の合成
以下に示す化合物の合成方法に用いる合成原料、反応試薬等は一般的な市販品である。また、反応溶媒、反応温度に関して特に記載のない場合は、通常その反応に利用される溶媒、温度で反応が行われる。また、反応は、アルゴン若しくは乾燥させた窒素雰囲気下で行われる。
以下に示す化合物の合成方法に用いる合成原料、反応試薬等は一般的な市販品である。また、反応溶媒、反応温度に関して特に記載のない場合は、通常その反応に利用される溶媒、温度で反応が行われる。また、反応は、アルゴン若しくは乾燥させた窒素雰囲気下で行われる。
[化合物#1の合成]
4-フェニル-2-(4-クロロ-1H-インドール-3-イル)-4-オキソ-ブタン(化合物#1)は、インドールの代わりに4-クロロインドールを用いて、後述する化合物#20の合成方法により合成した。
4-フェニル-2-(4-クロロ-1H-インドール-3-イル)-4-オキソ-ブタン(化合物#1)は、インドールの代わりに4-クロロインドールを用いて、後述する化合物#20の合成方法により合成した。
[化合物#2及び化合物#3の合成]
4-(4-クロロフェニル)-2-(1H-インドール-3-イル)-4-オキソ-ブタン酸(化合物#2)及び3-(1H-インドール-3-イル)-1-オキソ-1-フェニル-ブタン(化合物#3)は、Sayed,G. H. et al, “Synthesis and reactions of some β-aroyl-α-(indol-3-yl)propionic acids” Journal of the Chemical Society of Pakistan,7(4), 263-72; 1985の記載の方法に従って合成した。
4-(4-クロロフェニル)-2-(1H-インドール-3-イル)-4-オキソ-ブタン酸(化合物#2)及び3-(1H-インドール-3-イル)-1-オキソ-1-フェニル-ブタン(化合物#3)は、Sayed,G. H. et al, “Synthesis and reactions of some β-aroyl-α-(indol-3-yl)propionic acids” Journal of the Chemical Society of Pakistan,7(4), 263-72; 1985の記載の方法に従って合成した。

4-フェニル-2-(1H-5-エトキシインドール-3-イル)-4-オキソ-ブタン酸(化合物#7)は、インドールの代わりに5-エトキシインドールを用いて、化合物#20と同様の方法で合成した。
化合物#8、13~15、17~19、及び21~25はN-メトキシカルボニルインドール酢酸メチルを鍵中間体として合成した。

6-フルオロインドール(485mg,3.59mmol)を50mL丸底フラスコに入れた後、(E)-4-(2,4-ジフルオロフェニル)-4-オキソ-2-ブテン酸(508mg,2.39mmol)を加え、ベンゼン(20mL)で溶解し、80℃で7時間、加熱還流した。反応液に蒸留水(50mL)を加えたのち、酢酸エチル(50mL)で抽出した。有機層を食塩水(30mL)で洗浄してから、無水硫酸ナトリウムで脱水し、減圧濃縮した。濃縮して得られた生成物をシリカゲルクロマトグラフィー(ヘキサン:アセトン=2:1)で精製した、4-(2,4-ジフルオロフェニル)-2-(6-フルオロ-1H-インドール-3-イル)-4-オキソブタン酸(433mg,収率52%)を無色結晶で得た。

5-フルオロインドール(925mg,6.85mmol)を50mL丸底フラスコに入れた後、(E)-4-(2,4-ジフルオロフェニル)-4-オキソ-2-ブテン酸(969mg,4.57mmol)を加え、ベンゼン(20mL)で溶解し、80℃で11時間、加熱還流した。反応液に蒸留水(50mL)を加えたのち、酢酸エチル(50mL)で2回抽出した。有機層を食塩水(30mL)で洗浄してから、無水硫酸ナトリウムで脱水し、減圧濃縮した。濃縮して得られた生成物を桐山ロートでろ過し、クロロホルムと酢酸エチルから再結晶を行い、4-(2,4-ジフルオロフェニル)-2-(5-フルオロ-1H-インドール-3-イル)-4-オキソブタン酸(1122mg,収率71%)を無色結晶で得た。

7-メトキシインドール(1083mg,7.36mmol)を50mL丸底フラスコに入れた後、(E)-4-(2,4-ジフルオロフェニル)-4-オキソ-2-ブテン酸(1041mg,4.90mmol)を加え、ベンゼン(20mL)で溶解し、80℃で14時間、加熱還流した。反応液に蒸留水(50mL)を加えたのち、酢酸エチル(50mL)で抽出した。有機層を食塩水(30mL)で洗浄してから、無水硫酸ナトリウムで脱水し、減圧濃縮した。濃縮して得られた生成物を桐山ロートでろ過し、ベンゼンとアセトンから再結晶を行い、続いて、クロロホルムから再結晶を行って、4-(2,4-ジフルオロフェニル)-2-(7-メトキシ-1H-インドール-3-イル)-4-オキソブタン酸(1179mg,収率67%)を無色結晶で得た。

5-メトキシインドール(1166mg,7.93mmol)を50mL丸底フラスコに入れた後、(E)-4-(2,4-ジフルオロフェニル)-4-オキソ-2-ブテン酸を加え、ベンゼン(20mL)で溶解し、80℃で10時間、加熱還流した。反応液に蒸留水(50mL)を加えたのち、酢酸エチル(50mL)で2回抽出した。有機層を食塩水(30mL)で2回洗浄してから、無水硫酸ナトリウムで脱水し、減圧濃縮した。濃縮して得られた生成物を桐山ロートでろ過し、ベンゼンとアセトンから再結晶を行い、続いて、クロロホルムから再結晶を行って、4-(2,4-ジフルオロフェニル)-2-(5-メトキシ-1H-インドール-3-イル)-4-オキソブタン酸(1478mg,収率75%)を無色結晶で得た。

6-ベンジルオキシインドール(1255mg,5.62mmol)を50mL丸底フラスコに入れた後、(E)-4-(2,4-ジフルオロフェニル)-4-オキソ-2-ブテン酸(795mg,3.74mmol)を加え、ベンゼン(20mL)で溶解し、80℃で9時間、加熱還流した。反応液に蒸留水(50mL)を加えたのち、酢酸エチル(50mL)で抽出した。有機層を食塩水(30mL)で洗浄してから、無水硫酸ナトリウムで脱水し、減圧濃縮した。濃縮して得られた生成物を桐山ロートでろ過し、クロロホルムから再結晶を行って、2-(6-ベンジルオキシ-1H-インドール-3-イル)-4-(2,4-ジフルオロフェニル)-4-オキソブタン酸(531mg,収率33%)を淡黄色結晶で得た。

[参考例1]
(E)-4-(2,4-ジフルオロフェニル)-4-オキソ-2-ブテン酸
(E)-4-(2,4-ジフルオロフェニル)-4-オキソ-2-ブテン酸
本件化合物#1~10の合成で用いた(E)-4-(2,4-ジフルオロフェニル)-4-オキソ-2-ブテン酸は、以下のとおり合成した。すなわち、1,3-ジフルオロベンゼン(1300mg,11.39mmol)を100mL丸底フラスコに入れた後、無水マレイン酸(894mg,9.12mmol)を加え、ジクロロメタン(40mL)で溶解し、スターラーで撹拌した。撹拌しながら、無水塩化アルミニウム(2279mg,17.09mmol)を少しずつ投入して室温で6時間撹拌した。反応液を丸底フラスコの中の反応物を氷水(100mL)に入れて反応を停止させた後、水層を酢酸エチル(150mL)で抽出し、有機層を食塩水(100mL)で2回抽出操作をした後、有機層を無水硫酸ナトリウムで脱水した後、減圧濃縮して固形物を得た。この固形物を温浴中でベンゼンと少量のアセトンで再結晶させて精製したところ、淡黄色結晶の(E)-4-(2,4-ジフルオロフェニル)-4-オキソ-2-ブテン酸を収率48%で得た。

2.本件化合物群が哺乳類精子の受精機能増強効果を有することの確認
実施例1で合成した本件化合物群(化合物#5)を用い、以下に示す方法に従って、哺乳類精子の受精機能増強効果を確認した。なお、精子の前培養及び媒精は、5%CO2/20%O2、37℃条件下で行った。

[方法]
〔1〕メスのC57/B6マウスに、妊馬血清性腺刺激ホルモン(PMSG;pregnant mare serum gonadotropin)を腹腔内投与し、PMSG投与48時間後にヒト絨毛性性腺刺激ホルモン(hCG;Human chorionic gonadotropin)することにより過排卵を誘起した。hCG投与5~17時間後にマウスを安楽死させ、卵管を回収した。
〔2〕オスのC57/B6マウスを安楽死させた後、精巣上体の尾部を摘出し、回収した。
〔3〕100μLの4種類の培養液(0.0001%のDMSOを含む媒精用培養液[TYH培養液、LSIメディエンス社製]、及び1μMの化合物#5を含むTYH培養液、並びに、10μMのFCCP(Carbonyl cyanide-p-trifluoromethoxyphenylhydrazone)を含むTYH培養液、及び10μMのFCCPと1μMの化合物#5とを含むTYH培養液)からなる滴(ドロップ)をそれぞれ調製し、その上をミネラルオイル(シグマ-アルドリッチ社製)で覆った。上記回収した精巣上体の尾部を、上記ミネラルオイル中に移し、ピンセットで把持した状態で1mLの26G針付きツベルクリン用シリンジ(ニプロ社製)にて精巣上体管に傷をつけ、得られた精子塊を、ミネラルオイル中の上記4種類の培養液中にそれぞれ移し、媒精前の培養(前培養)を1時間行った。
〔4〕上記4種類の培養液中で前培養した精子3μLを、マイクロピペッター(エッペンドルフ社製、ドイツ)を用いて、卵丘細胞-卵子複合体(COC;cumulus oocyte complex)を含むTYH培養液のドロップ200μL中にそれぞれ移し、その上をミネラルオイル(シグマ-アルドリッチ社製)で覆い、媒精を行った。
〔5〕媒精24~28時間後に2細胞期卵の有無を観察し、2細胞期卵が観察された場合、受精したと評価し、受精率を測定した。

[結果]
化合物#5の溶媒であるDMSOを含むTYH培養液、すなわち、化合物#5不含の培養液中で前培養した精子を用いて媒精を行うと、受精率40%であったのに対して、化合物#5含有培養液中で前培養した精子を用いて媒精を行うと、受精率は100%であった(表1参照)。
この結果は、本件化合物群(化合物#5)は、精子の受精機能を賦活化(増強)する作用を有することを示している。
化合物#5の溶媒であるDMSOを含むTYH培養液、すなわち、化合物#5不含の培養液中で前培養した精子を用いて媒精を行うと、受精率40%であったのに対して、化合物#5含有培養液中で前培養した精子を用いて媒精を行うと、受精率は100%であった(表1参照)。
この結果は、本件化合物群(化合物#5)は、精子の受精機能を賦活化(増強)する作用を有することを示している。
精子は、ミトコンドリアにより産生されるATPを使用して鞭毛による前進運動を行う。かかるミトコンドリア機能を阻害し、受精機能が低下した精子に対する本件化合物群(化合物#5)の効果を検討した。
脱共役剤の1つであるFCCPを含む培養液中で前培養した精子を用いて媒精を行うと、受精率は30%であり、FCCP不含の培養液中で前培養した精子を用いて媒精した場合の受精率(47.4%)と比べ、低下することが確認された(表2参照)。一方、化合物#5をFCCP含有培養液中に添加し、かかる培養液中で精子を前培養すると、受精率は50%まで回復することが示された(表2参照)。
この結果は、本件化合物群(化合物#5)は、低下した精子の受精機能(例えば、ATPを利用した前進運動)を改善する作用を有することを示している。

Claims (7)
- 以下の一般式(I0)、一般式(II)、及び一般式(III)で表される化合物、並びに、R3がOHのときそれらの生理学的に許容される塩からなる群から選択される1種又は2種以上の化合物を含む、哺乳類精子の受精機能増強剤。
- 採取された少なくとも1つの哺乳類精子を、以下の一般式(I0)、一般式(II)、及び一般式(III)で表される化合物、並びに、R3がOHのときそれらの生理学的に許容される塩からなる群から選択される1種又は2種以上の化合物を含む生理的水溶液中に移す工程(a)を含む、哺乳類精子の受精機能の増強方法。
- 請求項3又は4に記載の増強方法により得られた哺乳類精子を、採取された少なくとも1つの哺乳類卵子を含む培養液中で培養する工程(b-1)を含む、哺乳類受精卵の作製方法。
- 採取された少なくとも1つの哺乳類精子と、採取された少なくとも1つの哺乳類卵子とを、以下の一般式(I0)、一般式(II)、及び一般式(III)で表される化合物、並び
に、R3がOHのときそれらの生理学的に許容される塩からなる群から選択される1種又は2種以上の化合物を含む培養液中で培養する工程(b-2)を含む、哺乳類受精卵の作製方法。
