Description
本発明は、液体クロマトグラフの分析流路に対して試料を自動的に注入するオートサンプラとそのオートサンプラを備える液体クロマトグラフに関するものである。
液体クロマトグラフィー用の分析流路中に試料を注入する装置として、全量注入方式のオートサンプラが知られている。全量注入方式のオートサンプラは、先端にニードルを有しニードルを介して吸入した試料を一時的に保持するためのサンプリング流路を備え、サンプリング流路を分析流路に組み込んだ状態(インジェクティングモード)と、サンプリング流路を分析流路に組み込まない状態(ローディングモード)とに切り替えられるように構成されている(特許文献1参照)。
液体クロマトグラフでは、分析の系内において塩の析出などに起因した流路の詰まりが発生する場合がある。分析系内で詰まりが発生するとシステム圧力が上昇するため、システム圧力を監視していれば分析系内で詰まりが発生したことを検知することは可能である。しかし、分析系内での詰まりを検知することができても、その詰まりがどのような場所で発生しているのかを特定することはできない。
分析系の中でもオートサンプラのニードル内で詰まりが発生しやすい。分析系内で詰まりが発生したときにそれがオートサンプラのニードル内で起こっていることを容易に特定することができれば、ニードルを洗浄又は交換するだけで問題を解決することができる。
そこで、本発明は、液体クロマトグラフの分析系内における詰まりの発生個所を特定しやすくすることを目的とするものである。
本発明に係るオートサンプラは、試料を一時的に保持するためのサンプリング流路を有し、液体クロマトグラフの分析流路に前記サンプリング流路を組み込むインジェクティングモードと前記分析流路に前記サンプリング流路を組み込まないローディングモードに選択的に切り替えられ、前記サンプリング流路に試料を保持させた状態で前記インジェクティングモードとなることにより、前記分離カラムの上流において前記分析流路中に試料を注入するオートサンプラであって、前記分析流路中で移動相を送液する送液ポンプの送液圧力を取り込み、前記インジェクティングモードと前記ローディングモードとの間で切り替えられたときの前記送液圧力の変動値を求め、求めた前記変動値に基づいて前記インジェクティングモードで前記分析流路に組み込まれる系内における詰まりの有無を判定するように構成された詰まり判定部と、を備えている。
ところで、オートサンプラのモードが切り替えられたときには移動相の圧縮・膨張に起因した送液圧力の変動が生じるが、その変動は送液圧力の大きさに応じて大きくなるため、送液圧力の変動値の絶対値を詰まりの有無の判定に用いると、送液圧力が大きいときに誤判定してしまう虞がある。
そこで、本発明のオートサンプラにおいては、前記詰まり判定部は、前記インジェクティングモードと前記ローディングモードとの間で切り替えられたときの前記送液圧力の変動値を前記送液圧力で除算した値、すなわち送液圧力に対する変動の割合を予め設定されたしきい値と比較することにより、前記インジェクティングモードで前記分析流路に組み込まれる系内における詰まりの有無を判定するように構成されていることが好ましい。そうすれば、オートサンプラのモード切替え時の圧力変動を流路の詰まりとして誤判定されることが防止できる。
上記の場合、前記しきい値は、前記ローディングモードから前記インジェクティングモードに切り替わったときに生じる圧力変動の送液圧力に対する割合よりも大きな値に設定することができる。
本発明に係る液体クロマトグラフは、分析流路と、前記分析流路に試料を注入する請求上述のオートサンプラと、前記オートサンプラにより前記分析流路中に注入された試料を成分ごとに分離するための分離カラムと、前記分析流路上における前記分離カラムよりも下流に設けられ、前記分離カラムで分離された試料成分を検出するための検出器と、を備えている。
本発明に係るオートサンプラでは、分析流路中で移動相を送液する送液ポンプの送液圧力を取り込み、インジェクティングモードとローディングモードとの間で切り替えられたときの送液圧力の変動値を求め、求めた変動値に基づいてインジェクティングモードで分析流路に組み込まれる系内における詰まりの有無を判定するように構成された詰まり判定部を備えているので、インジェクティングモードで分析流路に組み込まれる系内での詰まりの有無を容易に認識することができる。これにより、液体クロマトグラフの分析系内における詰まりの発生個所の特定が容易になる。
本発明に係る液体クロマトグラフは、上記オートサンプラを備えているので、分析系内における詰まりの発生個所の特定が容易になる。
オートサンプラとそのオートサンプラを備える液体クロマトグラフの一実施例について、図1を用いて説明する。
この実施例の液体クロマトグラフは、送液装置2、オートサンプラ3、分離カラム4、検出器6及びシステムコントローラ8を備えている。送液装置2、オートサンプラ3、分離カラム4及び検出器6は上流側から順に配管によって直列に接続されており、液体クロマトグラフィー分析のための分析流路を構成している。分離カラム4はカラムオーブン24内に収容されて一定温度に調節される。システムコントローラ8はこの液体クロマトグラフのシステム全体を管理するためのものである。
送液装置2は、2台の送液ポンプ9a、9bを有し、送液ポンプ9a、9bからそれぞれ互いに異なる溶媒をミキサ10へ送って混合し、混合された液を移動相として分析流路中で送液する。送液装置2には、送液圧力を検出するための圧力センサ11が設けられている。
オートサンプラ3は分析流路の流路構成を切り替えるための切替バルブ12を有する。この実施例では、切替バルブ12は(1)~(6)の6つのポートを有するマルチポートバルブであり、互いに隣り合うポート間の接続を切り替えるように構成されている。切替バルブ12のポート(1)にはサンプリング流路14の基端が接続され、ポート(2)にはシリンジポンプ17が接続されている。ポート(3)にはドレインへ通じるドレイン流路18が接続され、ポート(4)には注入ポート20が接続されている。ポート(5)には分離カラム4、検出器6を有する流路22が接続されており、ポート(6)には送液装置2からの流路が接続されている。切替バルブ12は、ポート(1)-(6)間、(2)-(3)間、(4)-(5)間を連通させた第1の状態(図示されている状態)と、ポート(1)-(2)間、(3)-(4)間、(5)-(6)間を連通させた第2の状態とのいずれかの状態に切り替えることができる。
サンプリング流路14の先端にニードル15が設けられており、ニードル15の基端側にサンプルループ16が設けられている。サンプリング流路14は、ニードル15の先端から吸入した試料をサンプルループ16内に一時的に保持するための流路である。ニードル15は図示しない移動機構によって移動させられる。
図示されているように、ニードル15の先端を注入ポート20に挿入接続した状態で切替バルブ12を第1の状態にすると、送液装置2-サンプリング流路14-分離カラム4が直列に接続された状態、すなわち、分析流路にサンプリング流路14が組み込まれた状態となる。この状態をインジェクティングモードと称する。
一方で、切替バルブ12を第2の状態に切り替えるとポート(5)-(6)間が連通するため、送液装置2と分離カラム4との間がサンプリング流路14を介することなく接続された状態、すなわち、分析流路にサンプリング流路14が組み込まれない状態となる。この状態をローディングモードと称する。
ローディングモードでは、切替バルブ12のポート(1)-(2)間が連通し、サンプリング流路14の基端がシリンジポンプ17と接続されるため、シリンジポンプ17により、図示されていない試料容器からニードル15の先端を介して試料を吸入することができる。ニードル15の先端から吸入された試料はサンプルループ16に滞留する。この状態で、オートサンプラ3をインジェクティングモードにすると、サンプリング流路14が分析流路に組み込まれ、サンプルループ16に保持された試料が送液装置2からの移動相によって分離カラム4へ搬送される。一般的に、試料が分離カラム4に導入されたタイミングで、オートサンプラ3はローディングモードに切り替えられる。分離カラム4に導入された試料は成分ごとに分離され、分離カラム4から溶出した試料成分が検出器6によって順に検出される。
オートサンプラ3は、切替バルブ12、ニードル15を移動させる移動機構(図示は省略)及びシリンジポンプ17の動作を制御するための制御部26を備えている。制御部26はマイクロコンピュータ等の演算素子やメモリ素子を搭載したコンピュータ回路によって実現されるものである。制御部26は詰まり判定部28を備えている。詰まり判定部28は、演算素子がプログラムを実行することによって得られる機能である。
オートサンプラ3の制御部26には、送液装置2の圧力センサ11によって検出される送液圧力がシステムコントローラ8を介して取り込まれるようになっている。詰まり判定部28は、オートサンプラ3がインジェクティングモードとローディングモードとの間で切り替わるときの送液圧力の変動値を求め、その変動値に基づいてインジェクティングモードで分析流路に組み込まれる系内における詰まりの有無、すなわちサンプリング流路14内や注入ポート内における詰まりの有無を判定するように構成されている。
サンプリング流路14内や注入ポート20内に詰まりが発生している場合、オートサンプラ3がローディングモードからインジェクティングモードに切り替わると、送液装置2の送液圧力が上昇する。そこで、詰まり判定部28は、オートサンプラ3がローディングモードからインジェクティングモードに切り替わる前の送液圧力(P1)と切り替わった後の送液圧力(P2)の差分(P2-P1)を変動値ΔPとして求め、その変動値ΔPを送液圧力(P1)で除算した値を予め設定されたしきい値と比較し、変動値ΔPを送液圧力(P1)で除算した値がしきい値を超えていれば、サンプリング流路14内や注入ポート20内で詰まりが発生していると判定する。しきい値は、オートサンプラ3がローディングモードからインジェクティングモードに切り替わったときに生じる圧力変動の送液圧力に対する割合よりも大きな値に設定されており、制御部26若しくはシステムコントローラ8に保持されている。
詰まり判定部28は、インジェクティングモード時に組み込まれる系内に詰まりがあると判定したときに、何らかの警告を発するように構成されていてもよい。警告の方法として、例えばオートサンプラ3やシステムコントローラ8に設けられているディスプレイ、若しくはシステムコントローラ8に接続されたコンピュータのディスプレイに詰まりの発生を知らせる表示を行なうことのほか、警告音を発することも挙げられる。
なお、詰まり判定部28は必ずしもオートサンプラ3の制御部26に設けられている必要はない。図2に示されているように、システムコントローラ8の機能として詰まり判定部28を設けることもできる。さらには、システムコントローラ8に接続されたコンピュータに詰まり判定部28としての機能をもたせることもできる。そのような場合、詰まり判定部28を具備するシステムコントローラやコンピュータは、オートサンプラ3の一部を構成する。
上記のように、オートサンプラ3がローディングモードとインジェクティングモードとの間で切り替えられるときの送液圧力の変動値に基づいて系内の詰まりの判定を行なうことで、サンプリング流路14や注入ポート20を含む特定箇所での詰まりの発生を検知することができる。これにより、分析系内での詰まりの発生個所の特定が容易になる。

Claims (6)
- 試料を一時的に保持するためのサンプリング流路を有し、液体クロマトグラフの分析流路に前記サンプリング流路を組み込むインジェクティングモードと前記分析流路に前記サンプリング流路を組み込まないローディングモードに選択的に切り替えられ、前記サンプリング流路に試料を保持させた状態で前記インジェクティングモードとなることにより、前記分離カラムの上流において前記分析流路中に試料を注入するオートサンプラであって、
前記分析流路中で移動相を送液する送液ポンプの送液圧力を取得し、前記インジェクティングモードと前記ローディングモードとの間で切り替えられたときの前記送液圧力の変動値を求め、求めた前記変動値に基づいて前記インジェクティングモードで前記分析流路に組み込まれる系内における詰まりの有無を判定するように構成された詰まり判定部と、を備えたオートサンプラ。 - 前記詰まり判定部は、前記インジェクティングモードと前記ローディングモードとの間で切り替えられたときの前記送液圧力の変動値を前記送液圧力で除算した値を予め設定されたしきい値と比較することにより、前記インジェクティングモードで前記分析流路に組み込まれる系内における詰まりの有無を判定するように構成されている、請求項1に記載のオートサンプラ。
- 前記しきい値は、前記ローディングモードから前記インジェクティングモードに切り替わったときに生じる圧力変動の送液圧力に対する割合よりも大きな値に設定されている、請求項2に記載のオートサンプラ。
- 分析流路と、
前記分析流路中で移動相を送液する送液ポンプと、
前記分析流路に試料を注入する請求項1に記載のオートサンプラと、
前記オートサンプラにより前記分析流路中に注入された試料を成分ごとに分離するための分離カラムと、
前記分析流路上における前記分離カラムよりも下流に設けられ、前記分離カラムで分離された試料成分を検出するための検出器と、を備えた液体クロマトグラフ。 - 前記オートサンプラの詰まり判定部は、インジェクティングモードとローディングモードとの間で切り替えられたときの前記送液ポンプの送液圧力の変動値を前記送液圧力で除算した値を予め設定されたしきい値と比較することにより、前記インジェクティングモードで前記分析流路に組み込まれる系内における詰まりの有無を判定するように構成されている、請求項4に記載の液体クロマトグラフ。
- 前記しきい値は、前記ローディングモードから前記インジェクティングモードに切り替わったときに生じる圧力変動の送液圧力に対する割合よりも大きな値に設定されている、請求項5に記載の液体クロマトグラフ。
