Description
本発明は、例えば溶出試験の溶出率やプロセス合成の反応率といった試料の経時的な変化を測定するための分析システムにおいて用いられるオートサンプラに関するものである。
近年、溶出試験の溶出率やプロセス合成の反応率を経時的に分析するために液体クロマトグラフを用いることが提案され、実施もなされている(特許文献1参照。)。その場合、分析対象の試料をオンラインで液体クロマトグラフに導入できるように、液体クロマトグラフのオートサンプラにフローバイアルが設置されることがある。
フローバイアルは、下端側に液を流入させるための入口部、上端側に液を流出させるための出口部を有し、上面の開口が弾力性を有するセプタムによって封止された容器である。フローバイアルの入口部に溶出試験機などの装置が接続され、分析対象試料がオンラインでフローバイアルに導入される。フローバイアル内に導入された試料はサンプリングニードルによって吸引され、液体クロマトグラフの分析流路に注入される。
液体クロマトグラフ用のオートサンプラは、試料や試薬の入ったバイアルを保持するラックが所定の位置に設置されるようになっており、所定の位置に設置されたラック上においてサンプリングニードルが移動し、所望のバイアルから液を吸引するように構成されている。さらに、ラックはペルチェ素子によって冷却される冷却ブロック上に配置され、その冷却ブロックからの冷却熱によって試料や試薬の蒸発や変質が抑制されるように構成されている。
上記のオートサンプラにフローバイアルを設置しようとした場合、サンプリングニードルの移動可能な範囲内にフローバイアルを配置する必要がある。そのため、フローバイアルを必然的に冷却ブロック上又はその近傍に配置される。
しかし、液体クロマトグラフを用いた分析では、フローバイアル内での試料液中の成分の析出による問題があることがわかった。すなわち、フローバイアル内で試料液中の成分の析出が生じると、その析出物をサンプリングニードルが吸引してサンプリングニードル内や液体クロマトグラフの流路内で詰まったり、試料液中の成分濃度が変動したりするということがわかった。
試料液中の成分の析出は、フローバイアルの温度が低いほど生じやすい。フローバイアルを冷却ブロック上又はその近傍に配置すると、冷却ブロックからの冷却熱によってフローバイアルの温度が低くなり、フローバイアル内で試料液中の成分の析出が生じやすくなるという問題がある。
そこで、本発明は、フローバイアル内での試料成分の析出を抑制することができるオートサンプラを提供することを目的とするものである。
本発明に係るオートサンプラの第1の形態は、冷却素子によって冷却される冷却ブロックと、前記冷却ブロック上に設けられ、分析に用いられる液を収容した複数の分析用液バイアルを前記冷却ブロックからの冷却熱によって冷却しながら同一平面内において保持する分析用液バイアルラックと、先端に液を吸引するための吸引口をもち、前記分析用液バイアルラックに保持された前記分析用液バイアルの分析用液を前記吸引口から吸引することができるように、先端が鉛直下方を向いた状態で水平面内方向と鉛直方向へ移動するように構成されたサンプリングニードルと、試料液を収容する空間を内部に有するとともに前記空間に通じる入口部及び前記空間の前記入口部よりも高い位置に通じる出口部を有する少なくとも1つのフローバイアルを、前記冷却ブロックとは熱的に分離しながら前記サンプリングニードルの移動範囲内で保持するフローバイアルラックと、を備えている。
ところで、フローバイアルを冷却ブロックとは熱的に分離するために、フローバイアルラックを冷却ブロックから離れた位置に配置することも考えられる。しかし、既存の液体クロマトグラフ用のオートサンプラでは、冷却ブロックから離れた位置にフローバイアルラックを配置できるようなスペースがなく、そのようなスペースを確保するためにはオートサンプラ全体の大きさを大きくする必要がある。また、フローバイアルラックを冷却ブロックから離して配置すると、サンプリングニードルの移動範囲をフローバイアルラックが配置されている位置にまで拡大する必要があり、既存のオートサンプラの規格変更を余儀なくされることになる。
そこで、本発明のオートサンプラの第1の形態においては、前記フローバイアルラックは、前記冷却ブロックと前記フローバイアルとの間に断熱材を介在させながら前記フローバイアルを前記冷却ブロック上で保持するものであることが好ましい。そうすれば、フローバイアルラックを冷却ブロックから離れた位置に配置しなくてよいので、オートサンプラの大型化や規格の変更の必要性を排除することができる。
さらに、前記フローバイアルラックに保持された前記フローバイアルの温度を前記冷却ブロックの異なる温度に調節するためのフローバイアル温調機構をさらに備えていてもよい。そうすれば、フローバイアル内の試料液の温度を所望の温度に維持することができるので、フローバイアル内での試料成分の析出をより確実に防ぐことができる。また、フローバイアル内の試料液の温度を所望の温度に調節することにより、フローバイアル内で試料液の反応を促進させたり、試料液の反応を停止させたりすることも可能となり、実施可能な分析の幅を広げることができる。

本発明に係るオートサンプラの第2の形態は、上記第1の形態とは異なり、前記フローバイアルラックが前記冷却ブロックと熱的に分離されているか否かに関係なく、前記フローバイアルの前記入口部に接続された入口配管の温度を調節するための入口配管温調機構を備えているものである。
本発明に係るオートサンプラの第1の形態では、フローバイアルラックによってフローバイアルが冷却ブロックとは熱的に分離された状態でサンプリングニードルの移動範囲内に配置されるようになっているので、冷却ブロックからの冷却熱によってフローバイアルが冷却されることが防止され、フローバイアル内での試料成分の析出を抑制することができる。
本発明に係るオートサンプラの第2の形態では、フローバイアルの入口部に接続された入口配管の温度を調節するための入口配管温調機構を備えているので、フローバイアル内に導入される試料液の温度を所望の温度に調節することができ、それによって、フローバイアル内での試料成分の析出を抑制することも可能になる。
以下に、本発明に係るオートサンプラの実施形態について、図面を用いて説明する。
まず、本発明の対象となるオートサンプラが用いられる分析システムについて、図1を用いて説明する。
分析システムは、試料処理装置2、液体クロマトグラフ4、及び演算処理装置6を備えている。試料処理装置2としては、例えば、薬剤等の溶出試験を行なうための溶出試験機やフロー合成を行なうためのフロー合成装置などが挙げられる。演算処理装置6は、例えば、液体クロマトグラフ4の各モジュール8、10、12及び14を管理するシステムコントローラ(図示は省略。)と電気的に接続されたパーソナルコンピュータである。
液体クロマトグラフ4は、送液装置8、オートサンプラ10、カラムオーブン12、及び検出器14を備えている。
送液装置8は送液ポンプを用いて移動相を送液する装置である。送液装置8の出口は配管を介してオートサンプラ10に接続されている。
オートサンプラ10は、試料処理装置2から供給される試料を収容するフローバイアル38(図2を参照。)のほか、フローバイアル38から試料水を採取するためのサンプリングニードル20(図2を参照。)、サンプリングニードル20により採取された試料を送液装置8からの移動相が流れる流路中に導入するか否かを切り替えるためのマルチポートバルブ(図示は省略。)を備えている。
カラムオーブン12内には試料を成分ごとに分離するための分析カラム(図示は省略。)が収容されている。カラムオーブン12内の分析カラムは、配管を介してオートサンプラ10の出口に接続されており、オートサンプラ10により注入された試料が送液装置8からの移動相とともに分析カラムへ導入されるように構成されている。カラムオーブン12内の分析カラムの下流端は配管を介して検出器14に接続されている。
検出器14は、分析カラムで分離された試料成分を検出するためのものであり、例えば紫外線吸光度検出器である。検出器14で得られた検出器信号は演算処理装置6に取り込まれ、試料成分濃度の定量等に用いられる。
図2に示されているように、オートサンプラ10内には、分析に用いられる、例えば、標準試料や反応試薬などの分析用液の入った複数の分析用液バイアル24を保持する分析用液バイアルラック22と、フローバイアル38を保持するフローバイアルラック36と、が設置されている。図ではフローバイアル38が1つしか示されていないが、実際には複数のフローバイアル38が同図の紙面に対して垂直な方向に一列に並んで配列されている。フローバイアル38の個数に制限はない。

この実施例では、フローバイアルラック36も冷却ブロック32上に配置されている。しかし、これはフローバイアル38を冷却するためではなく、サンプリングニードル20の移動範囲内にフローバイアルラック36を配置する必要があるために分析用液バイアルラック22に近接した位置にフローバイアルラック36を配置した結果である。フローバイアル38の温度が低くなると、溶出試験機などの試料処理装置2から供給された試料液中の成分がフローバイアル38内で析出しやすくなる。
この実施例では、フローバイアルラック36の下端部は断熱層46となっており、フローバイアル38を冷却ブロック32から熱的に分離している。これにより、冷却ブロック32からの冷却熱によってフローバイアル38が冷却されることと防ぎ、フローバイアル38内での試料成分の析出を抑制している。断熱層46は、例えばPEEKからなる層と空気層とを備えたものである。
なお、この実施例では、オートサンプラ10内のスペースの制約上、フローバイアルラック36を冷却ブロック32上に配置しているが、必ずしも冷却ブロック32上である必要はない。サンプリングニードル20の移動範囲内で冷却ブロック32上ではない位置にフローバイアルラック36を配置することができるのであれば、そのほうが好ましい。
また、フローバイアル38の温度が低下することをより確実に防ぐために、図3に示されているように、フローバイアル温調機構としてのヒータ48と温度センサ50を用いてフローバイアル38の温度を積極的に制御するようにしてもよい。図3の例では、フローバイアルラック36を熱伝導率の良好な金属材料で構成し、そのフローバイアルラック36にヒータ48を埋設してフローバイアル38を加熱温調するように構成している。ヒータ48の出力を制御する制御部52はフローバイアルラック36に取り付けられた温度センサ50からの信号を読み取り、フローバイアルラック36の温度が所定の温度になるようにヒータ48の出力を制御するように構成されている。制御部52は、例えばオートサンプラ10に設けられたマイクロコンピュータがプログラムを実行することによって得られる機能である。
また、図4に示されているように、入口配管16にジャケットヒータなどを入口配管温調機構として取り付け、入口配管16を通じてフローバイアル38に供給される試料液の温度を積極的に制御するようにしてもよい。これにより、フローバイアル38内の試料液の温度を試料成分が析出しにくい一定温度(例えば35℃)以上に維持することも可能であるので、フローバイアル38内での試料成分の析出を抑制することができる。
図4では示されていないが、図3の例で示したフローバイアル温調機構48を入口配管温調機構54と組み合わせて実施してもよい。また、フローバイアルラック36の下端部に断熱層46を設けず、入口配管温調機構54のみによってフローバイアル38内での試料成分の析出を抑制するようにしてもよい。

Claims (5)
- 冷却素子によって冷却される冷却ブロックと、
前記冷却ブロック上に設けられ、分析に用いられる液を収容した複数の分析用液バイアルを前記冷却ブロックからの冷却熱によって冷却しながら同一平面内において保持する分析用液バイアルラックと、
先端に液を吸引するための吸引口をもち、前記分析用液バイアルラックに保持された前記分析用液バイアルの分析用液を前記吸引口から吸引することができるように、先端が鉛直下方を向いた状態で水平面内方向と鉛直方向へ移動するように構成されたサンプリングニードルと、
試料液を収容する空間を内部に有するとともに前記空間に通じる入口部及び前記空間の前記入口部よりも高い位置に通じる出口部を有する少なくとも1つのフローバイアルを、前記冷却ブロックとは熱的に分離しながら前記サンプリングニードルの移動範囲内で保持するフローバイアルラックと、を備えたオートサンプラ。 - 前記フローバイアルラックは、前記冷却ブロックと前記フローバイアルとの間に断熱材を介在させながら前記フローバイアルを前記冷却ブロック上で保持するものである、請求項1に記載のオートサンプラ。
- 前記フローバイアルラックに保持された前記フローバイアルの温度を前記冷却ブロックの異なる温度に調節するためのフローバイアル温調機構をさらに備えている、請求項1又は2に記載のオートサンプラ。
- 前記フローバイアルの前記入口部に接続された入口配管の温度を調節するための入口配管温調機構をさらに備えている、請求項1から3のいずれか一項に記載のオートサンプラ。
- 冷却素子によって冷却される冷却ブロックと、
前記冷却ブロック上に設けられ、分析に用いられる液を収容した複数の分析用液バイアルを前記冷却ブロックからの冷却熱によって冷却しながら同一平面内において保持する分析用液バイアルラックと、
先端に液を吸引するための吸引口をもち、前記分析用液バイアルラックに保持された前記分析用液バイアルの分析用液を前記吸引口から吸引することができるように、先端が鉛直下方を向いた状態で水平面内方向と鉛直方向へ移動するように構成されたサンプリングニードルと、
試料液を収容する空間を内部に有するとともに前記空間に通じる入口部及び前記空間の前記入口部よりも高い位置に通じる出口部を有する少なくとも1つのフローバイアルを、前記サンプリングニードルが前記フローバイアルから試料液を吸引することができるように前記サンプリングニードルの移動範囲内で保持するフローバイアルラックと、
前記フローバイアルの前記入口部に接続された入口配管の温度を調節するための入口配管温調機構と、を備えたオートサンプラ。
