Description
(1) ナルフラフィン又はその薬理学的に許容される酸付加塩と、マルトース、マルチトール、デキストリン及びプルランからなる群から選択される結合剤成分と、担体と、を含有し、上記ナルフラフィン又はその薬理学的に許容される酸付加塩は、0.1~10μg含有され、上記結合剤成分の重量は、上記ナルフラフィン又はその薬理学的に許容される酸付加塩の重量に対して100,000~2,000,000重量%であり、かつ、得られる医薬組成物の全重量に対して5~20重量%である、錠剤化された医薬組成物。
(2) ナルフラフィン又はその薬理学的に許容される酸付加塩と、20℃における2%水溶液の粘度が5.9mPa・sより大きいヒドロキシプロピルセルロースを含む結合剤成分と、担体と、を含有し、上記ナルフラフィン又はその薬理学的に許容される酸付加塩は、0.1~10μg含有され、上記結合剤成分の重量は、上記ナルフラフィン又はその薬理学的に許容される酸付加塩の重量に対して20,000~500,000重量%であり、かつ、得られる医薬組成物の全重量に対して1~5重量%である、錠剤化された医薬組成物。
(3) 上記結合剤成分の重量は、上記ナルフラフィン又はその薬理学的に許容される酸付加塩の重量に対して100,000~400,000重量%である、(1)記載の医薬組成物。
(4) 上記結合剤成分の重量は、上記ナルフラフィン又はその薬理学的に許容される酸付加塩の重量に対して20,000~100,000重量%である、(2)記載の医薬組成物。
(5) 上記ナルフラフィン又はその薬理学的に許容される酸付加塩は、1~5μg含有されている、(1)~(4)のいずれか記載の医薬組成物。
(6) 上記担体は、マンニトール、ブドウ糖、無水結晶果糖、乳糖及びマルチトールからなる群から選択される、(1)~(5)のいずれか記載の医薬組成物。
(7) (1)~(6)のいずれか記載の医薬組成物からなる、錠剤。
<HPLC条件>
試料調製 :医薬組成物に測定時のナルフラフィン又はその薬理学的に許容される酸付加塩の濃度が1μg/mLになるように水を添加し、適宜撹拌して抽出する。この抽出液を遠心分離し、上澄みを測定試料とする。
標準液調製:ナルフラフィン又はその薬理学的に許容される酸付加塩の濃度が1μg/mLになるように水を添加し溶解し、測定試料とする。

図1に示されるように、高速クロマトグラフィーを用いた分析法で参考例2及び比較例1の錠剤を分析し、それらのクロマトグラムを比較すると、参考例2は20~75分以内に、対有効成分に換算して不純物の成分量が1%以上のピークを認めなかったことから、高純度に錠剤化された医薬組成物であった。
有効成分であるナルフラフィン又はその薬理学的に許容される酸付加塩を100mgあたり1μg相当含有すると想定した錠剤として、マンニトール(ペアリトール(登録商標)50C、ロケットジャパン社製)945mgとステアリン酸マグネシウム(太平化学産業株式会社製)5mgを袋内にて混合し、混合試料とした。次にソルビトール(ロケットジャパン社製)50mgを200μLの水に溶解させて結合剤成分の溶液とし、これを混合試料に添加しつつ乳鉢造粒し、造粒物を調製した。この造粒物100mgを油圧プレス機にて100kgfで加圧して6mmφ錠剤(1錠)を調製した(有効成分の想定含量1μg/錠剤)。この錠剤の硬度を硬度計(岡田精工株式会社製、PC-30)にて測定した。また、この造粒物0.5g(有効成分の想定含量5μg)から水5mLで抽出した抽出物をHPLCにて分析し、得られたピークの面積値を1μg/mLナルフラフィン塩酸塩の面積値と比較し最大の不純物の成分量を算出した。
比較例6のソルビトールを乳糖(乳糖一水和物、DMV-Fonterra Excipients社製)に代えたこと以外は比較例6と同様の方法で実験した。具体的には、有効成分であるナルフラフィン又はその薬理学的に許容される酸付加塩を100mgあたり1μg相当含有すると想定した錠剤として、マンニトール(ペアリトール(登録商標)50C、ロケットジャパン社製)945mgとステアリン酸マグネシウム(太平化学産業株式会社製)5mgを袋内にて混合し、混合試料とした。次に乳糖50mgを200μLの水に溶解させて結合剤成分の溶液とし、これを混合試料に添加しつつ乳鉢造粒し、造粒物を調製した。この造粒物100mgを油圧プレス機にて100kgfで加圧して6mmφ錠剤(1錠)を調製した(有効成分の想定含量1μg/錠剤)。この錠剤の硬度を硬度計(岡田精工株式会社製、PC-30)にて測定した。また、この造粒物0.5g(有効成分の想定含量5μg)から水5mLで抽出した抽出物をHPLCにて分析し、得られたピークの面積値を1μg/mLナルフラフィン塩酸塩の面積値と比較し最大の不純物の成分量を算出した。
比較例6のソルビトールを無水結晶果糖(ダニスコジャパン株式会社製)に代えたこと以外は比較例6と同様の方法で実験した。具体的には、有効成分であるナルフラフィン又はその薬理学的に許容される酸付加塩を100mgあたり1μg相当含有すると想定した錠剤として、マンニトール(ペアリトール(登録商標)50C、ロケットジャパン社製)945mgとステアリン酸マグネシウム(太平化学産業株式会社製)5mgを袋内にて混合し、混合試料とした。次に無水結晶果糖50mgを200μLの水に溶解させて結合剤成分の溶液とし、これを混合試料に添加しつつ乳鉢造粒し、造粒物を調製した。この造粒物100mgを油圧プレス機にて100kgfで加圧して6mmφ錠剤(1錠)を調製した(有効成分の想定含量1μg/錠剤)。この錠剤の硬度を硬度計(岡田精工株式会社製、PC-30)にて測定した。また、この造粒物0.5g(有効成分の想定含量5μg)から水5mLで抽出した抽出物をHPLCにて分析し、得られたピークの面積値を1μg/mLナルフラフィン塩酸塩の面積値と比較し最大の不純物の成分量を算出した。

ナルフラフィン又はその薬理学的に許容される酸付加塩を想定添加量10μgとし、遠心管にマンニトール1gを入れ蒸留水10mLを添加して撹拌し、得られた上澄みを参考例11とした。参考例11をHPLCにて分析し、得られたピークの面積値を1μg/mLナルフラフィン塩酸塩の面積値と比較し最大の不純物の成分量を算出した。表5に示すとおり、参考例11の最大の不純物の成分量は0.00%であった。
参考例11のマンニトールをブドウ糖(三栄源エフ・エフ・アイ株式会社製)に代えたこと以外は同様の方法で参考例12を作成した。具体的には、ナルフラフィン又はその薬理学的に許容される酸付加塩を想定添加量10μgとし、遠心管にブドウ糖(三栄源エフ・エフ・アイ株式会社製)1gを入れ蒸留水10mLを添加して撹拌し、得られた上澄みを参考例12とした。また、参考例11と同様の方法で実験し、得られたピークの面積値を1μg/mLナルフラフィン塩酸塩の面積値と比較し最大の不純物の成分量を算出した。表5に示すとおり、参考例12の最大の不純物の成分量は0.00%であった。
参考例11のマンニトールを無水結晶果糖(ダニスコジャパン株式会社製)に代えたこと以外は同様の方法で参考例13を作成した。具体的には、ナルフラフィン又はその薬理学的に許容される酸付加塩を想定添加量10μgとし、遠心管に無水結晶果糖(ダニスコジャパン株式会社製)1gを入れ蒸留水10mLを添加して撹拌し、得られた上澄みを参考例13とした。また、参考例11と同様の方法で実験し、得られたピークの面積値を1μg/mLナルフラフィン塩酸塩の面積値と比較し最大の不純物の成分量を算出した。表5に示すとおり、参考例13の最大の不純物の成分量は0.00%であった。
参考例11のマンニトールを乳糖(乳糖一水和物、DMV-Fonterra Excipients製)に代えたこと以外は同様の方法で参考例14を作成した。具体的には、ナルフラフィン又はその薬理学的に許容される酸付加塩を想定添加量10μgとし、遠心管に乳糖(乳糖一水和物、DMV-Fonterra Excipients製)1gを入れ蒸留水10mLを添加して撹拌し、得られた上澄みを参考例14とした。また、参考例11と同様の方法で実験し、得られたピークの面積値を1μg/mLナルフラフィン塩酸塩の面積値と比較し最大の不純物の成分量を算出した。表5に示すとおり、参考例14の最大の不純物の成分量は0.35%であった。
参考例11のマンニトールをマルチトール(株式会社林原製)に代えたこと以外は同様の方法で参考例15を作成した。具体的には、ナルフラフィン又はその薬理学的に許容される酸付加塩を想定添加量10μgとし、遠心管にマルチトール(株式会社林原製)1gを入れ蒸留水10mLを添加して撹拌し、得られた上澄みを参考例15とした。また、参考例11と同様の方法で実験し、得られたピークの面積値を1μg/mLナルフラフィン塩酸塩の面積値と比較し最大の不純物の成分量を算出した。表5に示すとおり、参考例15の最大の不純物の成分量は0.32%であった。
参考例11のマンニトールを白糖(鈴粉末薬品株式会社製)に代えたこと以外は同様の方法で比較例10を作成した。具体的には、ナルフラフィン又はその薬理学的に許容される酸付加塩を想定添加量10μgとし、遠心管に白糖(鈴粉末薬品株式会社製)1gを入れ蒸留水10mLを添加して撹拌し、得られた上澄みを比較例10とした。また、参考例11と同様の方法で実験し、得られたピークの面積値を1μg/mLナルフラフィン塩酸塩の面積値と比較し最大の不純物の成分量を算出した。表5に示すとおり、比較例10の最大の不純物の成分量は1.28%であった。
図2に示されるように、高速クロマトグラフィーを用いた分析法で参考例13、14及び比較例10の錠剤を分析し、それらのクロマトグラムを比較すると、参考例13及び14は20~75分以内に、対有効成分に換算して不純物の成分量が1%以上のピークを認めなかったことから、高純度に錠剤化された医薬組成物であった。
マンニトール(ペアリトール200SD(登録商標)、ロケットジャパン社製)97.895部を秤りとって、乳鉢に投入した。この顆粒に対して、ナルフラフィン塩酸塩(東レ株式会社製)0.005部、チオ硫酸ナトリウム水和物(国産化学株式会社製)0.1部、20℃における2%水溶液の粘度が6~10mPa・sのヒドロキシプロピルセルロース(HPC-L、日本曹達株式会社製)1部を蒸留水に溶解した液を滴下しながら乳鉢で混合し、検体乾燥器にて45℃で2時間乾燥し造粒顆粒を得た。ここにステアリン酸マグネシウム(太平化学産業株式会社製)を1部添加して袋内で混合した。得られた造粒物100mgを油圧プレス機にて100kgfで加圧して6mmφ錠剤(1錠)を調製し、実施例1とした。実施例1の結合剤成分であるヒドロキシプロピルセルロースの重量は、錠剤化された医薬組成物に対して1重量%であり、ナルフラフィン塩酸塩に対しては20,000重量%となる。
実施例1のマンニトール(ペアリトール200SD(登録商標)、ロケットジャパン社製)の添加量を93.895部に変更し、20℃における2%水溶液の粘度が6~10mPa・sのヒドロキシプロピルセルロース(HPC-L、日本曹達株式会社製)の添加量を5部に変更した以外は実施例1と同様の方法で6mmφ錠剤(1錠)を調製した。具体的には、マンニトール(ペアリトール200SD(登録商標)、ロケットジャパン社製)93.895部を秤りとって、乳鉢に投入した。この顆粒に対して、ナルフラフィン塩酸塩(東レ株式会社製)0.005部、チオ硫酸ナトリウム水和物(国産化学株式会社製)0.1部、20℃における2%水溶液の粘度が6~10mPa・sのヒドロキシプロピルセルロース(HPC-L、日本曹達株式会社製)5部を蒸留水に溶解した液を滴下しながら乳鉢で混合し、検体乾燥器にて45℃で2時間乾燥し造粒顆粒を得た。ここにステアリン酸マグネシウム(太平化学産業株式会社製)を1部添加して袋内で混合した。得られた造粒物100mgを油圧プレス機にて100kgfで加圧して6mmφ錠剤(1錠)を調製し、実施例2とした。また、実施例2の最大の不純物の成分量及び錠剤の硬度について、同様の測定を行った。実施例2の結合剤成分であるヒドロキシプロピルセルロースの重量は、錠剤化された医薬組成物に対して5重量%であり、ナルフラフィン塩酸塩に対しては100,000重量%となる。
マンニトール(ペアリトール200SD(登録商標)、ロケットジャパン社製)93.895部、マルチトール(粉末マビット、株式会社林原製)5部を秤りとって、乳鉢に投入した。この顆粒に対して、ナルフラフィン塩酸塩(東レ株式会社製)0.005部、チオ硫酸ナトリウム水和物(国産化学株式会社製)0.1部を蒸留水に溶解した液を滴下しながら乳鉢で混合し、検体乾燥器にて45℃で2時間乾燥し造粒顆粒を得た。ここにステアリン酸マグネシウム(太平化学産業株式会社製)を1部添加して袋内で混合した。得られた造粒物100mgを油圧プレス機にて100kgfで加圧して6mmφ錠剤を調製し、実施例3とした。実施例3の結合剤成分であるマルチトールの重量は、錠剤化された医薬組成物に対して5重量%であり、ナルフラフィン塩酸塩に対しては100,000重量%となる。
実施例3のマンニトール(ペアリトール200SD(登録商標)、ロケットジャパン社製)の添加量を78.895部に変更し、マルチトール(粉末マビット、株式会社林原製)の添加量を20部に変更した以外は実施例3と同様の方法で6mmφ錠剤(1錠)を調製した。具体的には、マンニトール(ペアリトール200SD(登録商標)、ロケットジャパン社製)78.895部、マルチトール(粉末マビット、株式会社林原製)20部を秤りとって、乳鉢に投入した。この顆粒に対して、ナルフラフィン塩酸塩(東レ株式会社製)0.005部、チオ硫酸ナトリウム水和物(国産化学株式会社製)0.1部を蒸留水に溶解した液を滴下しながら乳鉢で混合し、検体乾燥器にて45℃で2時間乾燥し造粒顆粒を得た。ここにステアリン酸マグネシウム(太平化学産業株式会社製)を1部添加して袋内で混合した。得られた造粒物100mgを油圧プレス機にて100kgfで加圧して6mmφ錠剤を調製し、実施例4とした。また、実施例4の最大の不純物の成分量及び錠剤の硬度について、同様の測定を行った。実施例4の結合剤成分であるマルチトールの重量は、錠剤化された医薬組成物に対して20重量%であり、ナルフラフィン塩酸塩に対しては400,000重量%となる。
実施例3のマルチトールをプルラン(日本薬局方プルラン、株式会社林原製)に変更した以外は実施例3と同様の方法で実験した。具体的には、マンニトール(ペアリトール200SD(登録商標)、ロケットジャパン社製)93.895部、プルラン(日本薬局方プルラン、株式会社林原製)5部を秤りとって、乳鉢に投入した。この顆粒に対して、ナルフラフィン塩酸塩(東レ株式会社製)0.005部、チオ硫酸ナトリウム水和物(国産化学株式会社製)0.1部を蒸留水に溶解した液を滴下しながら乳鉢で混合し、検体乾燥器にて45℃で2時間乾燥し造粒顆粒を得た。ここにステアリン酸マグネシウム(太平化学産業株式会社製)を1部添加して袋内で混合した。得られた造粒物100mgを油圧プレス機にて100kgfで加圧して6mmφ錠剤を調製し、実施例5とした。また、実施例5の最大の不純物の成分量及び錠剤の硬度について、同様の測定を行った。実施例5の結合剤成分であるプルランの重量は、錠剤化された医薬組成物に対して5重量%であり、ナルフラフィン塩酸塩に対しては100,000重量%となる。
実施例4のマルチトールをプルラン(日本薬局方プルラン、株式会社林原製)に変更した以外は実施例4と同様の方法で実験した。具体的には、マンニトール(ペアリトール200SD(登録商標)、ロケットジャパン社製)78.895部、プルラン(日本薬局方プルラン、株式会社林原製)20部を秤りとって、乳鉢に投入した。この顆粒に対して、ナルフラフィン塩酸塩(東レ株式会社製)0.005部、チオ硫酸ナトリウム水和物(国産化学株式会社製)0.1部を蒸留水に溶解した液を滴下しながら乳鉢で混合し、検体乾燥器にて45℃で2時間乾燥し造粒顆粒を得た。ここにステアリン酸マグネシウム(太平化学産業株式会社製)を1部添加して袋内で混合した。得られた造粒物100mgを油圧プレス機にて100kgfで加圧して6mmφ錠剤を調製し、実施例6とした。また、実施例6の最大の不純物の成分量及び錠剤の硬度について、同様の測定を行った。実施例6の結合剤成分であるプルランの重量は、錠剤化された医薬組成物に対して20重量%であり、ナルフラフィン塩酸塩に対しては400,000重量%となる。
乳糖(Pharmatose200M(登録商標)、DMV-Fonterra Excipients製)87.1464部、低置換度ヒドロキシプロピルセルロース(L-HPC)(LH-11、信越化学株式会社製)8.75部を秤りとって、乳鉢に投入した。この顆粒に対して、ナルフラフィン塩酸塩(東レ株式会社製)0.0036部、チオ硫酸ナトリウム水和物(国産化学株式会社製)0.1部、ヒドロキシプロピルメチルセルロース(HPMC2910、信越化学株式会社製)3部を蒸留水に溶解した液を滴下しながら乳鉢で混合し、検体乾燥器にて45℃で2時間乾燥し造粒顆粒を得た。ここにステアリン酸マグネシウム(太平化学産業株式会社製)を1部添加して袋内で混合した。得られた造粒物100mgを油圧プレス機にて100kgfで加圧して6mmφ錠剤(1錠)を調製した。比較例11の結合剤成分であるヒドロキシプロピルセルロースの重量は、錠剤化された医薬組成物に対して3重量%であり、ナルフラフィン塩酸塩に対しては83,333重量%となる。
Claims (7)
- ナルフラフィン又はその薬理学的に許容される酸付加塩と、マルトース、マルチトール、デキストリン及びプルランからなる群から選択される結合剤成分と、担体と、を含有し、
前記ナルフラフィン又はその薬理学的に許容される酸付加塩は、0.1~10μg含有され、
前記結合剤成分の重量は、前記ナルフラフィン又はその薬理学的に許容される酸付加塩の重量に対して100,000~2,000,000重量%であり、かつ、得られる医薬組成物の全重量に対して5~20重量%である、錠剤化された医薬組成物。 - ナルフラフィン又はその薬理学的に許容される酸付加塩と、20℃における2%水溶液の粘度が5.9mPa・sより大きいヒドロキシプロピルセルロースを含む結合剤成分と、担体と、を含有し、
前記ナルフラフィン又はその薬理学的に許容される酸付加塩は、0.1~10μg含有され、
前記結合剤成分の重量は、前記ナルフラフィン又はその薬理学的に許容される酸付加塩の重量に対して20,000~500,000重量%であり、かつ、得られる医薬組成物の全重量に対して1~5重量%である、錠剤化された医薬組成物。 - 前記結合剤成分の重量は、前記ナルフラフィン又はその薬理学的に許容される酸付加塩の重量に対して100,000~400,000重量%である、請求項1記載の医薬組成物。
- 前記結合剤成分の重量は、前記ナルフラフィン又はその薬理学的に許容される酸付加塩の重量に対して20,000~100,000重量%である、請求項2記載の医薬組成物。
- 前記ナルフラフィン又はその薬理学的に許容される酸付加塩は、1~5μg含有されている、請求項1~4のいずれか一項記載の医薬組成物。
- 前記担体は、マンニトール、ブドウ糖、無水結晶果糖、乳糖及びマルチトールからなる群から選択される、請求項1~5のいずれか一項記載の医薬組成物。
- 請求項1~6のいずれか一項記載の医薬組成物からなる、錠剤。
