Abstract

본 발명은 표면자유에너지가 높은 박막을 형성하도록 하는 제1단량체와 표면자유에너지가 낮은 박막을 형성하도록 하는 제2단량체가 형성한 공중합체를 포함하는 배양 플레이트와 이의 제조방법, 및 상기 배양 플레이트를 이용하여 세포 시트 형태의 세포 집합체를 제조 및 전사하는 방법을 제공한다. 상기와 같은 본 발명에 따르면, 종래기술과 비교하여 쉽고 간단한 공정으로 세포 시트 형태의 세포 집합체를 생산할 수 있는 효과가 있다.

Description

세포시트 제작방법 및 응용을 위한 고분자 박막 배양플레이트 제작방법 및 용도
본 특허출원은 2016년 7월 5일에 대한민국 특허청에 제출된 대한민국 특허출원 제10-2016-0084856호에 대하여 우선권을 주장하며, 상기 특허출원의 개시 사항은 본 명세서에 참조로서 삽입된다.
본 발명은 배양 플레이트에 관한 것으로서, 더욱 상세하게는 표면자유에너지 조절이 가능한 고분자 박막코팅을 포함하는 배양 플레이트와 이의 제조방법 및 상기 배양 플레이트를 이용하여 세포 시트 형태의 세포 집합체를 제조하는 방법에 관한 것이다.
재생의학에서 조직공학적 치료는 생분해성 고분자를 기반으로 세포를 배양하여 조직을 재건하고, 손상되거나 기능 부전에 빠진 장기에 이식하여 정상적인 기능을 하게 하는 치료법으로 발전하고 있다. 그러나 생분해성 고분자 지지체가 이식되었을 때, 면역 반응 및 생분해성 고분자의 분해에 의한 염증반응 등의 문제점을 피할 수는 없다 (Ronneberger B et al.,J Biomed Mater Res, 30 (1) (1996), 31-40). 다른 방법으로는 세포를 생분해성 고분자 용액에 섞어 인체에 주입하는 방법이 있으나, 이 과정 중 세포의 ECM (Extra Cellular Matrix : 세포외기질)이 깨져서 타겟 조직에 이식했을 때 세포재생 효율이 많이 저하되게 된다 (Canavan H et al.,J Biomed Mater Res A. 2005 Oct 1;75(1):1-13).
지지체 없이 세포를 이식하기 위해 세포 시트가 개발되었으며 (Yang J et al. Biomaterials. 2005 Nov;26(33):6415-22), 현재 가장 많이 사용되는 방법은 일본의 Teruo Okano에 의해 개발되어진 기술로서, 폴리스티렌(polystyrene) 배양접시 표면에 전자 빔 조사를 통해 온도 감응형 고분자 poly(N-isopropylacrylamide) (PIPAAm)를 20 nm 이하의 두께로 공유결합시키는 것이다 (Tang J et al, Polymers, 2012, 4, 1478-1498,Yang J et al Biomaterials 2007;28(34):5033-5043, Patent: US2008-0131476-A1). 온도 감응형 배양접시는 표면의 하한임계온도(lower critical solution temperature, LCST) 이상에서는 세포가 부착하여 세포시트를 형성하고, 하한임계온도 이하에서는 고분자가 팽윤하여 세포를 시트 형태로 회수할 수 있다 (Haraguchi, Y. et al. Nat. Protoc. 7, 850-858 (2012). 하지만, 이 방식은 세포의 온도를 20℃ 이하로 낮추어야 하며, 표면의 화학적 기능기를 바꾸는데 많은 제약이 있고, 제작방법이 까다로울 뿐만 아니라 많은 시간이 소요되는 등의 한계점으로 범용성 및 상용화가 어려운 문제가 있다 (Akiyama, Y. et al Langmuir 20, 5506-5511 (2004).
온도감응형 배양접시 이외에 전기 자극, 초음파 자극, pH 감응형을 이용한 연구가 진행되었으나, 전기 자극 및 초음파 효과를 발생시키기 위해서는 주로 금속물질 (Au, Ag, 또는 CNT)을 사용해야 하므로, 세포배양용 기판을 제작하는데 있어서 코팅 방법에 어려움이 발생하고, 또한 고가의 장비가 필요하므로 사용의 한계점이 발생한다 (Guillaume-Gentil O et al, Adv. 또한 세포 시트를 제작하였더라도, 이를 실제로 임상적으로 적용하기 위해서는 시트들을 적층하는 과정이 반복적으로 필요하고, 이를 효율적으로 적용하고자 하는 부위에 전사 가능해야 한다. 그러나, 현재까지 알려진 방법들은 여전히 적층과 전사(transfer) 시 재현성이 부족하며, 과정이 복잡하고 소모시간이 길다는 단점이 있다. 특히 기존의 대부분의 세포시트 전사 방법들은 여러장의 세포시트를 한 번에 전사하기보다는 단층의 세포시트를 개별적으로 전사하여 적층하는 방법을 선택하였다. 따라서 보다 쉽고 빠르게 세포시트를 적층하는 동시에 다른 기판 및 질병 모델 등으로 전사할 수 있는 새로운 세포시트의 전사 방법 개발에 대한 필요성이 제기되고 있는 실정이다.

본 명세서 전체에 걸쳐 다수의 논문 및 특허문헌이 참조되고 그 인용이 표시되어 있다. 인용된 논문 및 특허문헌의 개시 내용은 그 전체로서 본 명세서에 참조로 삽입되어 본 발명이 속하는 기술 분야의 수준 및 본 발명의 내용이 보다 명확하게 설명된다.
본 발명자들은 상기 문제점을 해결하기 위하여 예의 노력한 결과, 세포와 배양표면의 접착력에 영향을 주는 표면자유에너지(Surface free energy)를 조절함으로써 다양한 형태로 세포시트의 형성 및 분리가 가능할 뿐만 아니라, 세포외기질(extracellular matrix)이 포함되어진 상태로 회수되기 때문에 세포 시트의 적층이 가능하다는 것을 확인하였다. 또한 개시제를 이용한 화학기상증착법 (initiated chemical vapor deposition : iCVD)을 통한 배양 플레이트의 코팅은 기상증착 공정이므로 기판의 제약이 없고, 비교적 짧은 시간에 다양한 기능성 고분자를 코팅할 수 있어, 범용성 및 상용화에 있어 큰 장점을 가지고 있을 뿐만 아니라, 공중합체 코팅을 통해 표면자유에너지를 조절하여 세포 시트를 형성할 수 있는 것을 확인하고 본 발명을 완성하게 되었다.
따라서 본 발명의 목적은 배양플레이트를 제공하는데 있다.
본 발명의 다른 목적은 세포 시트 형태의 세포 집합체 제조방법을 제공하는데 있다.
본 발명의 또 다른 목적은 개시제를 이용한 화학기상증착법을 이용하여 배양 플레이트 표면 개질방법을 제공하는데 있다.
본 발명의 목적들은 이상에서 언급한 목적으로 제한되지 않으며, 언급되지 않은 본 발명의 다른 목적 및 장점들은 하기의 설명에 의해서 이해될 수 있으며, 본 발명의 실시예에 의해 보다 분명하게 알게 될 것이다. 또한, 본 발명의 목적 및 장점들은 청구범위에 나타낸 수단 및 조합에 의해 실현될 수 있음을 쉽게 알 수 있을 것이다.
본 발명의 일 양태에 따르면, 본 발명은 표면자유에너지가 낮은 박막을 형성하도록 하는 제1단량체와 표면자유에너지가 높은 박막을 형성하도록 하는 제2단량체가 형성한 공중합체를 포함하는 배양 플레이트를 제공한다.
본 명세서에서 사용된 표현, "표면자유에너지가 낮은 박막을 형성하도록 하는 제1단량체"는 표면자유에너지가 60 mJ/m2 이하인 단량체를 의미한다. "표면자유에너지가 높은 박막을 형성하도록 하는 제2단량체"는 표면자유에너지가 60 mJ/m2 이상인 단량체를 의미한다. 또한 "제1단량체와 제2단량체가 형성한 공중합체"는 상기 제1단량체와 제2단량체를 이용하여 형성된 표면자유에너지가 30 mJ/m2 - 90 mJ/m2 인 공중합체를 의미한다.
본 발명에 따르면, 배양플레이트의 표면자유에너지가 제1단량체 또는 제2단량체에 의해 형성된 동종중합체(homopolymer)를 통해 30 mJ/m2 - 90 mJ/m2로 구현되는 경우를 의미하기도 한다.
본 명세서에서 사용된 표현, "표면자유에너지가 낮은 박막을 형성하도록 하는 제1단량체와 표면자유에너지가 높은 박막을 형성하도록 하는 제2단량체가 형성한 공중합체를 포함하는 배양 플레이트"는 표면자유에너지가 낮은 박막을 형성하도록 하는 제1단량체와 표면자유에너지가 높은 박막을 형성하도록 하는 제2단량체가 형성한 공중합체를 포함하는 배양 플레이트의 일부분인 경우(예를 들어, 상기 공중합체로 표면이 코팅된 배양 플레이트)를 의미하는 것 뿐만 아니라, 표면자유에너지가 낮은 박막을 형성하도록 하는 제1단량체와 표면자유에너지가 높은 박막을 형성하도록 하는 제2단량체가 형성한 공중합체 자체를 배양 플레이트로 사용할 수 있음을 의미하기 위하여 사용된다.

본 명세서에서 제2단량체는 세포부착이 강한 높은 표면 자유에너지를 갖는 단량체이다. 예를 들면, 비닐계 아민 (2-비닐피리딘(2-vinylpyridine), 4-비닐피리딘(4-vinylpyridine), 1-vinylimidazole,1-vinylpyrrolidone, 2-비닐피리딘2-vinylpyridine,4-aminostyrene,9-vinylcabazole 등), 메타아크릴레이트계 아민 (2-(Dimethylamino)ethyl methacrylate, diethylaminoethylacrylate, dimethylaminoethylacrylate,diethylaminoethylacrylate 등), 산성의 작용기를 가진 단량체 (Maleic anhydride, Methacrylic acid 등), 아크릴아미드(acrylamide), 메타크릴아미드(methacrylamide), 염소계열 작용기를 가지는 단량체 (4-Vinylbenzyl chloride, 2-Chloroethyl acrylate 등), 시안계열 단량체 (Cyanoethyl acrylate, Vinyl benzyl cyanide 등), 비닐-N-메틸피리디늄클로라이드(vinyl-N-methylpyridinium chloride)로 구성된 군에서 선택되는 단량체일 수 있다.
본 발명에 있어서, 제1단량체와 제2단량체가 형성하는 공중합체 형성 시, 혼합비율의 제한은 없다. 제1단량체와 제2단량체의 혼합비율은 1% - 99% 까지 조절이 가능하다.
따라서 본 발명의 일구현예에 따르면, 제1단량체는 표면자유에너지가 낮은 박막을 형성하도록 하는 단량체이고, 제2단량체는 표면자유에너지가 높은 박막을 형성하도록 하는 단량체이다.
하나의 특정예에 따르면, 표면자유에너지가 낮은 박막을 형성하도록 하는 제1단량체는 divinylbenzene(이하 'DVB'라고 명명함)이고, 표면자유에너지가 높은 박막을 형성하도록 하는 제2단량체는 4-vinylpyridine(이하 '4VP'라고 명명함)이며, 상기 제1단량체와 제2단량체가 형성한 공중합체는 Poly(divinylbezene-co-4-vinylpyridine)(이하 'PD4V'라고 명명함)이다.
본 발명의 다른 구현예에 따르면 상기 표면자유에너지가 높은 박막을 형성하도록 하는 제1단량체와 표면자유에너지가 낮은 박막을 형성하도록 하는 제2단량체가 형성한 공중합체 상에서 세포를 배양함으로써 세포 시트 형태의 세포 집합체를 제조할 수 있다(도 1).
본 발명의 다른 양태에 따르면, 본 발명은 본 발명의 배양 플레이트에서 세포를 배양하는 단계를 포함하는 세포 시트 형태의 세포 집합체 제조방법을 제공한다.
본 발명의 또 다른 양태에 따르면, 본 발명은 개시제를 분해하여 유리 라디칼(free radical)을 형성하는 단계와, 유리 라디칼에 의해 제1단량체와 제2단량체를 중합반응시켜 공중합체를 형성하는 단계와, 배양 플레이트에 공중합체가 증착되어 박막을 형성하는 단계를 포함하는 화학기상증착법을 이용한 배양 플레이트 표면 개질방법을 제공한다.
본 명세서에서 화학기상증착법은 개시제를 이용한 화학기상증착법 (initiated chemical vapor deposition : iCVD)을 이용할 수 있고, 개시제는 TBPO(tert-butyl peroxide)를 사용할 수 있다. 한편, 개시제는 열 또는 전기에 의해 분해되어 유리 라디칼을 생성하고, 상기 유리 라디칼에 의해 제1단량체와 제2단량체를 활성화시킴으로써 상기 단량체를 연쇄 중합 반응시켜 공중합체를 형성하게 되고 상기 공중합체가 증착되어 박막을 형성함으로써 배양 플레이트 표면을 개질할 수 있다. 또한 배양플레이트의 고분자 박막의 두께는 특별히 제한되지 않으나, 예를 들면 5 ㎚ ~ 500 ㎛일 수 있다. 지지체층의 두께가 너무 얇거나 두꺼우면 박막 형성의 효율성과 세포배양을 위한 박막표면의 안정성에 영향을 미칠 수 있다.
한편, iCVD은 고분자 박박이 증착되는 기판 표면의 온도가 10℃에서 45℃ 사이로 낮게 유지되는 저온 및 저진공 공정이므로, 플라스틱 재질의 다양한 배양플레이트(35pi, 100pi dish, 6,12,24,96 well plate)에 손상 없이 다양한 고분자 코팅이 가능하다. 기존에 사용되는 딥코팅, 스핀 코팅과 같은 액상공정은 용매로 인한 기판 손상 문제, 불균형 코팅 등의 문제를 가지고 있어, iCVD를 통해 배양플레이트에 제작한다면 기존 코팅방법으로 불가능했던 코팅문제를 해결할 수 있다.
본 발명에 따르면 상기 배양플레이트는 세포를 배양할 수 있는 임의의 공간을 제공하는 것으로 충분하기 때문에 이의 형태는 제한이 없다. 예를 들어, 배양 플레이트는 디쉬(배양 접시), 샬레나 플레이트(예컨대, 6웰, 24웰, 48웰, 96웰, 384웰, 9600웰 등의 마이크로타이터 플레이트, 마이크로 플레이트, 딥 웰 플레이트 등), 플라스크, 챔버 슬라이드, 튜브, 셀 팩토리, 롤러 보틀, 스피너 플라스크, 중공 섬유(hollow fibers), 마이크로 캐리어, 비즈 등의 형상을 가질 수 있다. 또한, 지지성을 갖는 물질이라면 상기 배양플레이트로 제한 없이 사용할 수 있으며, 예를 들어, 플라스틱(예컨대, 폴리스티렌, 폴리에틸렌, 폴리프로필렌 등), 금속, 실리콘 및 유리 등의 재질을 배양플레이트로 사용할 수 있다. 본 발명의 일실시예에 따른 배양 플레이트의 구조는 도 1에 도시되어있다.
본 발명의 일 구현예에 따르면, 제1단량체는 표면자유에너지가 높은 박막을 형성하도록 하는 단량체이고, 제2단량체는 표면자유에너지가 낮은 박막을 형성하도록 하는 단량체이다.
하나의 특정예에 따르면, 표면자유에너지가 낮은 박막을 형성하도록 하는 제1단량체는 DVB 이고, 표면자유에너지가 높은 박막을 형성하도록 하는 제2단량체는 4VP 이며, 상기 제1단량체와 제2단량체가 형성한 공중합체는 PD4V 이다.
다른 하나의 특정예에 따르면 도 2에 개시된 바와 같이 iCVD에 의해 공중합체 증착시 제1단량체인 DVB와 제2단량체인 4VP의 주입비율을 따라 세포 시트 (cell sheet) 형태 또는 세포 스페로이드(cell spheroid) 형태의 세포 집합체로 배양할 수 있고, DVB 주입비율이 높을수록 스페로이드 형태의 세포 집합체로 배양하기 적합한 배양 플레이트 표면개질이 가능하며, DVB 대비 4VP의 주입비율이 증가할수록 시트 형태의 세포 집합체로 배양하기 적합한 배양 플레이트 표면개질이 가능하다. 한편, 단량체의 주입비율을 조절하면 접촉각과 표면에너지가 다른 배양 플레이트 표면을 제작할 수 있고, 각 공중합체 코팅 표면의 에너지에 따라 세포와 배양 플레이트 표면과의 접착력이 달라져 각기 다른 형태로 세포가 자라게 되며, 온도변화 없이 배양 플레이트 표면으로부터 세포 시트 형태의 세포 집합체를 분리할 수 있다.
본 발명의 배양플레이트 상에서 다양한 세포를 배양하여 세포시트를 형성할 수 있다. 배양되는 세포는 본 발명에서 특별히 한정되지 않으며, 예를 들어, 심장, 근육, 간, 뼈, 골수, 흉선, 신장, 비장, 허파, 뇌, 정소, 난소, 소도(islet), 내장, 귀, 피부, 쓸개조직, 전립선, 방광, 배아(embryo), 면역계 및 조혈계 등으로부터 분리되거나 활성화할 수 있는 세포를 사용할 수 있다. 바람직하게는, 각종 줄기세포, 각막상피세포, 신경세포, 혈관내피세포, 연골세포, 섬유아세포, 골아세포, 근아세포, 신장세포, 간세포, 지방세포, 각질세포, 근육세포, 심장근육세포 또는 식도상피세포를 포함한다.
본 발명의 다른 양태에 따르면, 본 발명은 홀 구조체를 이용한 세포시트 형태의 세포 집합체의 적층 및 전사(transfer) 방법을 제공한다.
상기 세포 집합체의 적층 및 전사 방법은 다음 단계를 포함할 수 있다:
(a) 세포시트 형태의 세포 집합체를 홀 구조체(structure with holes) 표면에 1층 이상 부착하는 단계; 및
(b) 세포 집합체의 적용이 필요한 부위에 세포 집합체가 부착된 면이 대향하도록 홀 구조체를 배치한 후, 홀 구조체만을 박리하는 단계.
본 발명의 일 구현예에 따르면, 상기 홀 구조체는 세포시트 형태의 세포 집합체를 적용이 필요한 장소 또는 부위에 용이하게 적용할 수 있도록 세포 집합체를 올려 놓을 수 있는 멤브레인 형태의 구조물을 말한다. 상기 홀 구조체는 예컨대 니트로셀룰로오스 멤브레인, 나일론 멤브레인, PVDF(Polyvinylidene fluoride) 멤브레인, Polytetrafluoroethylene (PTFE) 멤브레인, 폴리카보네이트(polycarbonate) 멤브레인, MCE(mixed cellulose ester) 멤브레인, 폴리아마이드 멤브레인, 및 PES(Polyethersulfone) 멤브레인으로 이루어진 군으로부터 선택된 것으로서, 1개 이상의 구멍이 있는 것을 특징으로 하는 방법.
등일 수 있으나 이에 한정되는 것은 아니며, 당 업계에서 사용될 수 있는 다양한 종류의 멤브레인을 제한없이 사용할 수 있다.
본 발명의 일 구현예에 따르면, 상기 홀 구조체는 1개 이상의 구멍이 있는 것이 바람직하고, 구멍의 개수와 모양에는 제한이 없으며, 구멍의 크기는 세포시트 형태의 세포집합체를 멤브레인의 구멍 위에 올려 놓아도 아래로 통과되지 않는다면 제한이 없다.
본 발명의 다른 일 구현예에 따르면, 상기 홀 구조체를 세포 집합체로부터 박리할 때 세포 집합체가 부착된 면의 반대 면에 인산완충용액, 또는 세포배양 배지를 점적하는 단계를 추가적으로 포함할 수 있다.
상술한 세포 집합체의 전사 방법에서 사용된 홀 구조체의 구조적 특징은 구조체 상에 구멍이 형성되어 있다는 것으로, 상기 구멍은 세포시트와 멤브레인 간의 과도한 부착력을 방지하면서 동시에 효과적인 전사가 가능하도록 한다.
첫째로, 구멍이 생기게 되면 멤브레인과 시트간의 접촉 면적이 상대적으로 줄어들어, 절대적인 부착력이 적어지고, 전사가 용이해진다.
두 번째로, 전사시 구멍을 통해 인산완충용액이나 세포 배양액을 소량 흘려주면 세포시트와 멤브레인이 서로 더 잘 떨어질 수 있다.
상기와 같은 본 발명에 따르면, 표면자유에너지가 높은 박막을 형성하도록 하는 제1단량체와 표면자유에너지가 낮은 박막을 형성하도록 하는 제2단량체가 형성한 공중합체를 포함하는 배양 플레이트와 이의 제조방법, 및 상기 배양 플레이트를 이용하여 세포 시트 형태의 세포 집합체를 제조하는 방법을 제공함으로써, 종래기술과 비교하여 쉽고 간단한 공정으로 세포 시트 형태의 세포 집합체를 생산 및 분리 회수할 수 있는 효과가 있다.

도 4 는 본 발명의 일실시예에서 제조된 제1단량체 고분자(pDVB), 제2단량체 고분자(p4VP), 공중합체인 pD4V가 코팅된 표면의 접촉각(contact angle)과 표면자유에너지(surface free energy)를 측정한 결과이다.
도 5 는 본 발명의 일실시예에서 제조된 제1단량체 고분자(pDVB), 제2단량체 고분자(p4VP), 공중합체(pD4V)가 코팅된 표면에서 배양된 세포(NIH3T3)의 현미경이미지결과이다.

본 발명의 상술한 목적, 특징 및 장점은 첨부된 도면을 참조하여 후술되어 있는 상세한 설명을 통하여 보다 명확해질 것이며, 그에 따라 본 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 본 발명의 기술적 사상을 용이하게 실시할 수 있을 것이다. 또한, 본 발명을 설명함에 있어서 본 발명과 관련된 공지기술에 대한 구체적인 설명이 본 발명의 요지를 불필요하게 흐릴 수 있다고 판단되는 경우에 그 상세한 설명을 생략하기로 한다. 이하, 첨부된 도면들을 함께 참조하여 본 발명에 따른 바람직한 실시예를 상세히 설명하기로 한다.
실시예 1. 배양플레이트의 제조
PD4V증착 시, 화학기상증착 반응기(iCVD, Daeki Hi-Tech Co., Ltd)를 사용하여, DVB (divinylbenzene)단량체 (Sigma-Aldrich), 4VP(4-vinylpyridine) 단량체 (Sigma-Aldrich)와 개시제 (tert-butyl peroxide, TBPO, Sigma-Aldrich)를 60:240:60의 비율로 iCVD 반응기 내에 흘러 주면서, 반응기 내의 필라멘트의 온도는 140℃, 반응기 내의 기판 온도는 23℃, 반응기 내 챔버의 압력은 300 mTorr로 유지하면서 1시간 30분 증착을 수행하여, 400㎚ 두께의 DVB-4VP 공중합체(nPD4V)가 증착된 배양플레이트를 얻었다.
실시예 2. 세포시트 배양플레이트 표면분석
중합체 박막을 증착한 후, 푸리에 변환 적외선 분광학(FT-IR, ALPHA FT-IR 흡광모드, Bruker Optics)을 이용하여 중합체의 분자 골격 및 분율을 측정하였다. 그 결과, 도 3에 나타난 바와 같이, 1596 와 1415 cm-1(좌측 2개 점선) 피크를 통해 4VP 분자의 존재를, 710 와 903 cm-1(우측 2개 점선) 피크를 통해 DVB 분자의 존재 및 중합체 합성을 확인하였다.
중합체 박막을 증착한 후 접촉각 측정장비(Contact Angle Analyzer (Phoenix 150, SEO,Inc.)를 이용하여 5 ㎕의 증류수와 다이아이오도메탄(Diiodomethane, DIM)에 대하여 기판의 표면 접촉각을 측정하였다. 그 결과, 도 4에 나타난 바와 같이, 단량체의 혼합비율에 따라 형성된 중합체에 의해 표면이 개질되어 접촉각이 달라지는 것을 확인할 수 있었다. 이를 기반으로 Van Oss-Chaudhury-Good (OCG) 수식을 이용하여 기판의 표면자유에너지를 계산하였다. 그 결과 중합체의 분율에 따라 표면자유에너지 값이 달라짐을 확인하였다.
실시예 3.공중합체 분율에 따른 세포형태 관찰
DVB-4VP 공중합체(PD4V)가 코팅되어 있는 세포 배양 접시에 NIH3T3 세포를 배양한 후, 세포시트 형성 여부를 확인하였다. 세포가 충분히 자랐을 때, 4% 포름알데히드로 고정하고, DAPI와 팔로이딘(phalloidin)을 이용하여 핵과 액틴을 염색하여 형광현미경으로 관찰하였다. 도 5에 나타난 바와 같이, 모든 배양플레이에서 세포가 독성 없이 잘 자라는 것이 확인됐으며, DVB 배양표면에서는 세포스페로이드(spheroids)가 형성되고, 4VP 배양표면에서는 세포가 부착되어 자라는 것이 관찰되었다. 반면, 공중합체 배양표면(pD4V1, pD4V2)에서는 세포가 부착되어 자랐으나, DPBS(Dulbecco`s Phosphate Buffered Saline)을 이용하여 세척한 후에, 저절로 세포시트 형태로 떨어져 나오는 것이 관찰되었다.
실시예 4. 세포 시트 형성 및 분리
상기 실시예 1에 의해 제조된 pD4V가 증착된 35pi dish에서 NIH3T3 와 hMSC를 배양 후 세포시트 형성을 확인하였다. 세포배양은 NIH3T3 세포를 DMEM (Dulbecco's Modified Eagle Medium)/10% FBS/ 1% 항생제(penicillinestreptomycin, Gibco)로, hMSC 세포는 MEM α (Minimum Essential Medium α)/17% FBS/ 1% 항생제(penicillinestreptomycin, Gibco) 배지를 사용하고, 3-5일 배양하자 세포시트가 형성되었다. 이때 배양액을 제거하고, DPBS (Dulbecco's Phosphate buffer saline)로 세척해주면 형성된 세포 시트가 배양플레이트 표면으로부터 자발적으로 분리되어 나오는 것을 확인할 수 있었다(도 6 및 도 7).
실시예 5. 배양 플레이트에서 형성된 세포시트를 분리회수 세포시트의 적층 방법
세포시트를 제조하기 위해서는, PD4V 중합체가 코팅되어 있는 세포 배양 플레이트에 세포를 배양하여 세포 간의 접합이 충분히 일어날 수 있을 때까지 배양한 후, Dulbesco's phosphate buffered saline (DPBS) 용액을 이용하여 배양한 세포를 시트형태로 분리하면 된다. 배양접시 상에서 박리한 1장의 세포시트를 흡인하여 새로운 세포배양접시에 옮긴 후, 37℃ 포화수증기의 인큐베이터에 적당한 시간 (예를 들어 15∼30분간) 방치하였다. 그 동안에 세포시트는 배양접시 상에 접착하였다. 다음으로 박리한 직후의 두 번째 세포시트를 배양액과 함께 피펫으로 흡인하여, 배양접시 상에 고정된 첫 번째 세포시트 위에 적하하였다. 적하된 2장의 시트에 다시 새로운 배양액을 천천히 적하함으로써 두 번째의 시트를 첫 번째의 시트에 겹친 상태로 접합할 수 있었다. 동일한 수법을 반복하여 세포시트를 차례로 적층화 할 수 있었다.
실시예 6. 세포시트를 적층 후 전사시 홀 구조체를 이용하는 방법
본 발명에 따라 제작된 상기 세포시트를 적층하고, 이를 다른 세포 배양접시 또는 세포시트의 적용이 필요한 객체에 보다 쉽게 전사(transfer)하기 위하여 다음과 같은 방법을 사용하였다.
먼저 도 8에 나타낸 바와 같이, 배양접시 상에서 박리한 1장의 세포시트를 홀 구조체(예컨대, 1 이상의 구멍이 있는 니트로셀룰로오스 멤브레인)의 표면에 적하하여 부착시켰다. 이어서 박리한 다른 세포시트를 상기 니트로셀룰로오스 멤브레인에 먼저 부착된 세포시트의 표면에 추가적으로 적하하여 부착시킴으로써 2장의 세포시트를 적층하였다. 동일한 방법을 반복하면 원하는 수의 세포시트를 멤브레인에 적층할 수 있다.
상기 적층한 2장의 세포시트를 멤브레인과 함께 새로운 세포배양접시에 옮긴 후, 37℃ 포화수증기의 인큐베이터에 적당한 시간 (예를 들어 5∼30분간) 인큐베이션 하였다. 그 결과 적층된 세포시트는 배양접시 상에 접착되고 홀 구조체 멤브레인으로부터 떨어진다. 이와 같이 세포시트를 홀 구조체에 적층하여 원하는 곳에 전사(transfer)하는 것이 가능하다.
이상에서 설명한 본 발명은 본 발명이 속하는 기술분야에서 통상의 지식을 가진 자에게 있어 본 발명의 기술적 사상을 벗어나지 않는 범위 내에서 여러 가지 치환, 변형 및 변경이 가능하므로 전술한 실시예 및 첨부된 도면에 의해 한정되는 것은 아니다.

    상기 제1단량체 보다 표면자유에너지가 높은 박막을 형성하도록 하는 제2단량체가 형성한 공중합체를 포함하는 배양 플레이트.

    상기 제1단량체의 표면자유에너지는 60 mJ/㎡ 이하인 것을 특징으로 하는 배양플레이트.

    상기 제2단량체는 2-vinylpyridine, 4-vinylpyridine, vinylimidazole, vinylpyrrolidone, 4-aminostyrene, 9-vinylcabazole, 2-(Diethylamino)ethyl acrylate, diethylaminoethylacrylate, dimethylaminoethylacrylate, diethylaminoethylacrylate, 2-Chloroethyl acrylate, Cyanoethyl acrylate, 3-(dimethylamino)propyl acrylate, 2-(Dimethylamino)ethyl methacrylate, t-butylaminoethyl methacrylate, Dimethylaminomethyl styrene, Methacrylic acid, acrylamide, methacrylamide, N,N-Dimethylacrylamide, N-isopropylacrylamide, 4-Vinylbenzyl chloride, Vinyl benzyl cyanide, vinyl-N-methylpyridinium chloride, N-Vinylcaprolactam, Allylamine, N-(4-Vinylbenzyl)-N-dimethylamine, Acrylonitrile로 구성된 군에서 선택되는 것을 특징으로 하는 배양플레이트.
  7. 제 6 항에 있어서,
    상기 제2단량체는 4-vinylpyridine인 것을 특징으로 하는 배양플레이트.
  8. 제 1 항에 있어서,
    상기 공중합체의 표면자유에너지는 30 mJ/m2 - 90 mJ/m2인 것을 특징으로 하는 배양플레이트
  9. 제 1 항에 있어서,
    상기 배양 플레이트는 세포 시트 형태의 세포 집합체 제조용인 것을 특징으로 하는 배양플레이트.
  10. 제 1 항에 있어서,
    상기 배양플레이트의 소재는 유리, 금속, 금속 산화물, 섬유, 종이 및 플라스틱으로 구성된 군에서 선택되는 것을 특징으로 하는 배양플레이트.
  11. 제 10 항에 있어서,
    상기 플라스틱은 폴리에틸렌(polyethylene, PE), 폴리프로필렌(polypropylene, PP), 폴리스티렌(polystyrene, PS), 폴리에틸렌 테레프탈레이트(polyethylene terephthalate, PET), 폴리아미드(polyamides, PA), 폴리에스터(polyester, PES), 폴리염화비닐(polyvinyl chloride, PVC), 폴리우레탄(polyurethanes, PU), 폴리카보네이트(polycarbonate, PC), 폴리염화비닐리덴(polyvinylidene chloride, PVDC), 폴리테트라플루오르에틸(polytetrafluoroethylene, PTFE), 폴리에트르에테르케톤(polyetheretherrketone, PEEK) 및 폴리에테르이미드(polyetherimide, PEI)로 구성된 군에서 선택되는 것을 특징으로 하는 배양플레이트.
  12. 제1항 내지 제11항 중 어느 한 항의 배양플레이트에서 세포를 배양하는 단계를 포함하는 세포 시트 형태의 세포 집합체 제조방법.
  13. 개시제를 분해하여 유리 라디칼(free radical)을 형성하는 단계와,
    유리 라디칼에 의해 제1단량체와 제2단량체를 연쇄중합반응시켜 공중합체를 형성하는 단계와,
    배양플레이트에 공중합체가 증착되어 박막을 형성하는 단계를 포함하는 개시제를 이용한 화학적 기상 증착(initiated Chemical Vapor Deposition, iCVD) 공정을 이용한 배양플레이트 표면 개질방법.
  14. 제 13 항에 있어서,
    상기 제1단량체의 표면자유에너지는 60 mJ/㎡ 이하인 것을 특징으로 하는 배양플레이트 표면 개질방법.
  15. 제 13 항에 있어서,
    상기 제1단량체는 divinylbenzene, Vinyl Benzoate, styrene, Benzyl Methacrylate, Cyclohexyl Methacrylate, butyl methacrylate, Isopropyl methacrylate, Acryl Amide, Allyl methacrylate, 2-Isocyanatoethyl Methacrylate, Ethylene glycol dimethacrylate, Di(ethylene glycol) methyl ester methacrylate, 2-Hydroxyethyle methacrylate, 1,2,4-trivinylcyclohexane, furfuryl methacrylate, Tetrahydrofurfuryl methacrylate, Hexyl Methacrylate, hydroxyethyl methacrylate, Glycidyl methacrylate, Propargyl Methacrylate, 1,4-Butanediol divinyl ether, Isobornyl acrylate, Ethylene glycol diacrylate, Propargyl acrylate, 2,4,6,8-tetramethyl-2,4,6,8-tetravinylcyclotetrasiloxane, hexavinyldisiloxane, Hexavinyldisiloxane, 1,3,5-trivinyl-1,3,5-trimethylcyclotrisiloxane, 2,4,6-Trimethyl-2,4,6-trivinylcyclotrisilazane, Dimethylphenylvinylsilane, heptadecafluorodecyl methacrylate,perfluorodecyl acrylate, Heptafluorobutyl methacrylate, 1,1,1,3,3,3-Hexafluoroisopropyl methacrylate, 2,2,3,3,4,4-hexafluoro-1,5-pentyl dicrylate, 2-perfluorohexylethyl methacrylate, 2,2,2-Trifluoroethyl methacrylate, Pentafluorophenyl methacrylate, 1H,1H,7H-Dodecafluoroheptyl acrylate, 1H,1H,2H,2H-heptadecaflurodecyl acrylate, di(ethyleneglycol)di(vinyl) ether, 1,9-Decadiene, Methacrylic Anhydride, 1,2,4-Trivinylcyclohexane, 2-(Methacryloyloxyl)ethyl acetoacetate, Allyl Acetoacetate, Maleic Anhydride로 구성된 군에서 선택되는 것을 특징으로 하는 배양플레이트 표면 개질방법.
  16. 제 15 항에 있어서,
    상기 제1단량체는 divinylbenzene인 것을 특징으로 하는 배양플레이트 표면 개질방법.
  17. 제 13 항에 있어서,
    상기 제2단량체의 표면자유에너지는 60 mJ/㎡ 이상인 것을 특징으로 하는 배양플레이트 표면 개질방법.
  18. 제 13 항에 있어서,
    상기 제2단량체는 2-vinylpyridine, 4-vinylpyridine, vinylimidazole, vinylpyrrolidone, 4-aminostyrene, 9-vinylcabazole, 2-(Diethylamino)ethyl acrylate,diethylaminoethylacrylate, dimethylaminoethylacrylate, diethylaminoethylacrylate, 2-Chloroethyl acrylate, Cyanoethyl acrylate,3-(dimethylamino)propyl acrylate, 2-(Dimethylamino)ethyl methacrylate,t-butylaminoethyl methacrylate, Dimethylaminomethyl styrene, Methacrylic acid, acrylamide, methacrylamide, N,N-Dimethylacrylamide, N-isopropylacrylamide, 4-Vinylbenzyl chloride, Vinyl benzyl cyanide, vinyl-N-methylpyridinium chloride,N-Vinylcaprolactam, Allylamine,N-(4-Vinylbenzyl)-N-dimethylamine, Acrylonitrile로 구성된 군에서 선택되는 것을 특징으로 하는 배양플레이트 표면 개질방법.
  19. 제 18 항에 있어서,
    상기 제2단량체는 4-vinylpyridine인 것을 특징으로 하는 배양플레이트 표면 개질방법.
  20. 제 13 항에 있어서,
    상기 공중합체의 표면자유에너지는 30 mJ/m2 - 90 mJ/m2인 것을 특징으로 하는 배양플레이트 표면 개질방법.
  21. 제 13 항에 있어서,
    상기 배양플레이트는 세포 시트 형태의 세포 집합체 제조용인 것을 특징으로 하는 배양플레이트 표면 개질방법.
  22. (a) 세포시트 형태의 세포 집합체를 홀 구조체(structure with holes) 표면에 1층 이상 부착하는 단계; 및
    (b) 세포 집합체의 적용이 필요한 부위에 세포 집합체가 부착된 면이 대향하도록 홀 구조체를 배치한 후, 홀 구조체만을 박리하는 단계를 포함하는 세포 집합체의 전사(transfer) 방법.
  23. 제 22 항에 있어서, 상기 홀 구조체는 니트로셀룰로오스 멤브레인, 나일론 멤브레인, PVDF(Polyvinylidene fluoride) 멤브레인, Polytetrafluoroethylene (PTFE) 멤브레인, 폴리카보네이트(polycarbonate) 멤브레인, MCE(mixed cellulose ester) 멤브레인, 폴리아마이드 멤브레인, 및 PES(Polyethersulfone) 멤브레인으로 이루어진 군으로부터 선택된 것으로서, 1개 이상의 구멍이 있는 것을 특징으로 하는 방법.
  24. 제 22 항에 있어서, 상기 (b)의 홀 구조체의 박리시 세포 집합체가 부착된 면의 반대 면에 인산완충용액, 또는 세포배양 배지를 점적하는 것을 특징으로 하는 방법.
