Abstract
본 발명은 서열번호 2의 아미노산 서열로 이루어진 피부 세포 증식 및 항산화 효과가 증가한 보툴리눔 톡신-인간상피세포성장인자 융합단백질, 대장균 코돈 최적화된 서열번호 1의 염기서열로 이루어진 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 코딩하는 유전자, 상기 유전자를 포함하는 재조합 벡터, 상기 재조합 벡터로 형질전환된 숙주세포, 상기 재조합 벡터로 숙주세포를 형질전환시켜 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 생산하는 방법 및 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 유효성분으로 포함하는 피부 주름 개선 및 피부 탄력 유지용 화장료 조성물에 관한 것으로, 본 발명의 화장료 조성물은 피부 주름 개선 및 피부 재생 효과를 증진시킬 수 있는 효과가 있으므로, 추후 기능성 화장품 분야 또는 미용성형 분야에 유용하게 사용될 수 있을 것이다.
Description
본 발명은 피부 세포 증식 및 항산화 효과가 증가한 보툴리눔 톡신-인간상피세포성장인자 융합단백질 및 이를 유효성분으로 함유하는 피부 재생 및 주름 개선용 화장료 조성물에 관한 것이다.
현대 화장품 산업의 핵심은 화장품 원료의 고갈에 따른 신소재 원료 개발과 적용에 있다. 현재 화장품 산업 전반에 걸쳐 신소재 개발을 위한 기술과 고기능성 화장품의 개발 등이 지속적으로 이루어지고 있는 실정이다. 특히 인간 상피세포성장인자(Human Epidermal Growth Factor, hEGF)는 주름 개선, 미백 등 피부 재생 효과가 뛰어나 소비자의 주목을 받고 있는 소재 중 하나이다.
사람의 피부는 성년(~25세) 이후 신진대사나 세포의 재생 능력이 저하됨에 따라 색소 침착 및 주름 형성 등이 유발되며 피부 노화 현상이 진행된다. 피부 재생 효과가 우수한 인간 상피세포성장인자는 피부 재생을 위한 의약품 치료제로서 사용되어 왔으나, 피부의 저하된 재생능력을 돕고 피부 세포의 새로운 세포 성장을 촉진할 수 있는 기능을 가진 노화 방지용 기능성 화장품의 원료로 사용하기 위한 연구가 집중되어 왔다.
인간 상피세포성장인자(Human epidermal growth factor, hEGF)는 세포 표면에 존재하는 상피세포성장인자 수용체와 결합하여 상피세포성장인자 수용체의 이합체화 반응(dimerization)을 유도한다. 이합체의 상피세포성장인자 수용체는 수용체 내에 존재하는 타이로신 키나아제를 활성화시켜 세포 내 신호전달 시스템을 유도하게 되고, 이러한 과정을 통해 세포 내에서는 해당 작용(glycolysis)과 단백질 합성이 촉진되어 최종적으로 세포의 성장이 진행된다.
피부 재생의 중요한 역할을 하는 상피세포성장인자는 노화가 진행될수록 감소되는데, 상피세포성장인자의 감소는 피부 세포 증식 및 이동의 감소를 유발하여 피부의 노화, 주름 증가, 탄력 감소 등의 현상을 나타낸다.
클로스트리디움 보툴리눔(Clostridium botulinum)으로부터 처음 발견된 보툴리눔 톡신(Botulinum toxin)은 근육 신경을 마비시키는 효과를 통해 의료용으로 처음 사용되었다. 특히 극소량의 보툴리눔 톡신을 한정된 부위에 선택적으로 사용시 근육 및 신경질환 등의 증상을 치료할 수 있어 미국의 제약회사 엘러간(Allergan Inc.)사에서 저농도의 보툴리눔 톡신 A를 보톡스(Botox)라는 상품명으로 판매하고 있으며, 현재 국내에는 보톡스, 보툴렉스(국내산), 메디톡신(국내산), 디스포트(유럽산), BTXA(중국산) 등 모두 4종이 판매되고 있다. 1990년대에 보톡스가 주름제거의 효과가 있는 것이 알려지면서 현재는 미용 성형 분야에서 주름제거 목적으로 전 세계적으로 널리 사용되고 있다.
2002년 미국 식품 의약국(U.S. Food and Drug Administration, FDA)은 보톡스를 미간 곡선 라인 치료제로 승인하였으며, 화장용 치료제로 현재까지 가장 각광받고 있다. 화장품 산업에서 주름 개선 치료제로 가장 인기가 있는 보툴리눔 톡신은 주사 요법을 사용하고 있으며, 그 효과가 3~6개월 내외라는 점과 매우 고가라는 최대 단점을 가지고 있다.
본 발명에서는 미용 성형 분야에서 주름제거 효과가 우수한 보툴리눔 톡신의 기능을 보다 효과적으로 지속하기 위해, 특히 피부재생효과를 부각시키기 위한 새로운 단백질 개발을 진행한 결과, 피부 재생 효과가 우수한 인간상피세포성장인자를 결합하여 새로운 이중 기능성 융합단백질이 주름 개선 및 피부재생 촉진을 극대화시킬 수 있음을 확인하였다.
한편, 한국공개특허 제2015-0144735호에는 '필러와 보틀리눔 독소를 포함하는 피부 주름, 노화 개선 또는 신경근육 관련 질환 치료용 조성물'이 개시되어 있고, 한국공개특허 제2015-0056022호에는 '상피세포 성장인자의 융합단백질을 포함하는 피부개선용 화장료 조성물'이 개시되어 있으나, 본 발명의 세포 증식 및 항산화 효과가 증가한 보툴리눔 톡신-인간상피세포성장인자 융합단백질 및 이를 유효성분으로 함유하는 피부 재생 및 주름 개선용 화장료 조성물에 대해서는 기재된 바가 없다.
본 발명은 상기와 같은 요구에 의해 도출된 것으로서, 본 발명자들은 보툴리눔 톡신 단백질에 인간상피세포성장인자를 융합하여 신규의 세포 증식 및 항산화 효과가 증가한 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 생산하였다. 상기 보툴리눔 톡신-인간상피세포성장인자 융합단백질은 피부 세포의 증식 및 항산화 효과가 인간상피세포성장인자보다도 매우 우수함을 확인함으로써, 본 발명을 완성하였다.
상기 과제를 해결하기 위해, 본 발명은 서열번호 2의 아미노산 서열로 이루어진 피부 세포 증식 및 항산화 효과가 증가한 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 제공한다.
또한, 본 발명은 상기 융합단백질을 코딩하는 유전자를 제공한다.
또한, 본 발명은 상기 유전자를 포함하는 재조합 벡터를 제공한다.
또한, 본 발명은 상기 재조합 벡터로 형질전환된 숙주세포를 제공한다.
또한, 본 발명은 상기 재조합 벡터로 숙주세포를 형질전환시켜 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 코딩하는 유전자를 과발현시키는 단계를 포함하는, 숙주세포에서 보툴리눔 톡신-인간상피세포성장인자 융합단백질의 생산 방법을 제공한다.
또한, 본 발명은 상기 방법에 의해 생산된 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 제공한다.
또한, 본 발명은 서열번호 2의 아미노산 서열로 이루어진 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 유효성분으로 포함하는 피부 재생 및 주름 개선용 화장료 조성물을 제공한다.
본 발명의 대장균 코돈 최적화된 보툴리눔 톡신-인간상피세포성장인자 융합단백질 코딩 유전자를 이용한 대장균에서의 생산 방법은 대장균 내에서 단백질이 응집체 형태로 발현되어 생산공정을 간소화하였으며, 단백질의 대량생산을 가능하게 하였고, 상기 방법에 의해 생산된 보툴리눔 톡신-인간상피세포성장인자 융합단백질은 피부 주름 개선 및 피부 재생 기능이 우수하여, 기능성 화장료 원료로 유용하게 사용될 수 있을 것이다.
도 1은 보툴리눔 톡신-인간상피세포성장인자 융합단백질 암호 유전자를 포함하는 재조합 플라스미드(pET22b::BTALEGF)의 제작 과정 및 대장균으로의 형질전환에 관한 모식도이다.

본 발명의 목적을 달성하기 위하여, 본 발명은 서열번호 2의 아미노산 서열로 이루어진 피부 세포 증식 및 항산화 효과가 증가한 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 제공한다.
본 발명에 따른 보툴리눔 톡신-인간상피세포성장인자 융합단백질의 범위는 서열번호 2의 아미노산 서열을 갖는 단백질 및 상기 단백질의 기능적 동등물을 포함한다. "기능적 동등물"이란 아미노산의 부가, 치환 또는 결실의 결과, 상기 서열번호 2의 아미노산 서열과 적어도 70% 이상, 바람직하게는 80% 이상, 더욱 바람직하게는 90% 이상, 더 더욱 바람직하게는 95% 이상의 서열 상동성을 갖는 것으로, 서열번호 2로 표시되는 단백질과 실질적으로 동질의 활성을 나타내는 단백질을 말한다. "실질적으로 동질의 활성"이란 피부 주름 개선 및 피부 재생 활성을 의미한다. 본 발명은 또한 보툴리눔 톡신-인간상피세포성장인자 융합단백질의 단편, 유도체 및 유사체 (analogues)를 포함한다. 본원에 사용된, 용어 "단편", "유도체" 및 "유사체"는 본 발명의 보툴리눔 톡신-인간상피세포성장인자 융합단백질과 실질적으로 같은 생물학적 기능 또는 활성을 보유하는 폴리펩타이드를 말한다.
본 발명의 보툴리눔 톡신-인간상피세포성장인자 융합단백질은 바람직하게는 서열번호 2의 아미노산 서열로 이루어지며, 상기 아미노산 서열 중 1번째 내지 448번째 아미노산 서열로 이루어진 보툴리눔 톡신 단백질과 449번째 내지 501번째 아미노산 서열로 이루어진 인간상피세포성장인자 단백질을 융합시켜 제작된 신규 단백질일 수 있다.
본 발명은 또한, 상기 피부 세포 증식 및 항산화 효과가 증가한 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 코딩하는 유전자를 제공한다. 상기 유전자는 대장균 코돈 최적화된 서열번호 1의 염기서열로 이루어질 수 있으나, 이에 제한되지 않는다.
본 발명에 따른 상기 피부 세포 증식 및 항산화 효과가 증가한 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 코딩하는 유전자는 서열번호 1의 염기서열을 포함할 수 있다. 또한, 상기 염기서열의 상동체가 본 발명의 범위 내에 포함된다. 구체적으로는, 상기 유전자는 서열번호 1의 염기서열로 이루어진 군으로부터 선택되는 염기서열과 각각 70% 이상, 더욱 바람직하게는 80% 이상, 더 더욱 바람직하게는 90% 이상, 가장 바람직하게는 95% 이상의 서열 상동성을 가지는 염기서열을 포함할 수 있다. 폴리뉴클레오티드에 대한 "서열 상동성의 %"는 두 개의 최적으로 배열된 서열과 비교 영역을 비교함으로써 확인되며, 비교 영역에서의 폴리뉴클레오티드 서열의 일부는 두 서열의 최적 배열에 대한 참고 서열(추가 또는 삭제를 포함하지 않음)에 비해 추가 또는 삭제(즉, 갭)를 포함할 수 있다.
"코돈 최적화"는 코딩된 단백질이 유기체에서 보다 효율적으로 발현되도록 특정 유기체에서 우선적으로 사용되는 것으로 단백질을 코딩하는 폴리뉴클레오티드의 코돈을 변화시키는 것을 의미한다. 대부분의 아미노산이 "동의어" 또는 "동의" 코돈이라고 하는, 몇몇 코돈에 의해 표시된다는 점에서, 유전자 코드가 축퇴적이지만, 특정 유기체에 의한 코돈 용법은 임의적이지 않고 특정 코돈 트리플렛에 편향적이다. 이러한 코돈 용법 편향성은 소정 유전자, 공통 기능 또는 조상 기원의 유전자, 고도로 발현되는 단백질 대 낮은 복제수 단백질, 및 유기체 게놈의 집합적 단백질 코딩 영역과 관련하여 더 높을 수 있다. 본 발명의 상기 서열번호 1의 염기서열은 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 코딩하는 유전자가 대장균 내에서 발현될 수 있도록 대장균의 코돈에 최적화된 서열이다.
본 발명은 또한, 상기 유전자를 포함하는 재조합 벡터 및 상기 재조합 벡터로 형질전환된 숙주세포를 제공한다.
용어 "재조합"은 세포가 이종의 핵산을 복제하거나, 상기 핵산을 발현하거나 또는 펩티드, 이종의 펩티드 또는 이종의 핵산에 의해 암호된 단백질을 발현하는 세포를 지칭하는 것이다. 재조합 세포는 상기 세포의 천연 형태에서는 발견되지 않는 유전자 또는 유전자 절편을, 센스 또는 안티센스 형태 중 하나로 발현할 수 있다. 또한 재조합 세포는 천연 상태의 세포에서 발견되는 유전자를 발현할 수 있으며, 그러나 상기 유전자는 변형된 것으로서 인위적인 수단에 의해 세포 내 재도입된 것이다.
본 발명에서, 상기 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 코딩하는 유전자는 재조합 발현 벡터 내로 삽입될 수 있다. 용어 "재조합 발현 벡터"는 세균 플라스미드, 파아지, 효모 플라스미드, 식물 세포 바이러스, 포유동물 세포 바이러스, 또는 다른 벡터를 의미한다. 대체로, 임의의 플라스미드 및 벡터는 숙주 내에서 복제 및 안정화할 수 있다면 사용될 수 있다. 상기 발현 벡터의 중요한 특성은 복제 원점, 프로모터, 마커 유전자 및 번역 조절 요소(translation control element)를 가지는 것이다.
보툴리눔 톡신-인간상피세포성장인자 융합단백질을 코딩하는 유전자 서열 및 적당한 전사/번역 조절 신호를 포함하는 발현 벡터는 당업자에 주지된 방법에 의해 구축될 수 있다. 상기 방법은 시험관 내 재조합 DNA 기술, DNA 합성 기술 및 생체 내 재조합 기술 등을 포함한다. 상기 DNA 서열은 mRNA 합성을 이끌기 위해 발현 벡터 내의 적당한 프로모터에 효과적으로 연결될 수 있다. 또한 발현 벡터는 번역 개시 부위로서 리보좀 결합 부위 및 전사 터미네이터를 포함할 수 있다.
본 발명의 일 구현 예에 따른 재조합 벡터는, pET22b 벡터에 합성한 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 코딩하는 유전자(서열번호 1)를 5' 말단(NdeI 제한효소 사이트)과 3' 말단 (XhoI 제한효소 사이트)에 인프레임(in frame)으로 융합하여 제조하였고, 상기 유전자를 lac 프로모터(lac promoter)와 lacI 리프레서(lacI repressor)에 의하여 효과적으로 발현시켜 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 제조하는 것을 특징으로 하는 벡터이다.
본 발명의 벡터를 원핵세포에 안정되면서 연속적으로 클로닝 및 발현시킬 수 있는 숙주세포는 당업계에 공지된 어떠한 숙주세포도 이용할 수 있으며, 예컨대, E.

본 발명의 벡터를 숙주세포 내로 운반하는 방법은, 숙주 세포가 원핵 세포인 경우, CaCl2 방법, 하나한 방법(Hanahan, D., 1983 J. Mol. Biol. 166, 557-580) 및 전기천공 방법 등에 의해 실시될 수 있다. 또한, 숙주세포가 진핵세포인 경우에는, 미세주입법, 칼슘포스페이트 침전법, 전기천공법, 리포좀-매개 형질감염법, DEAE-덱스트란 처리법, 및 유전자 밤바드먼트 등에 의해 벡터를 숙주세포 내로 주입할 수 있다.

본 발명의 일 구현 예에 따른 화장료 조성물에서 상기 보툴리눔 톡신-인간상피세포성장인자 융합단백질의 함량은 화장료 조성물 총 중량 대비 0.000002 내지 0.002 중량%로 포함될 수 있으나, 이에 제한되지 않는다.
본 발명의 화장료 조성물에는 상기 유효성분 이외에 화장품 조성물에 통상적으로 이용되는 성분들을 포함하며, 예컨대 지방 물질, 유기 용매, 용해제, 농축제 및 겔화제, 연화제, 항산화제, 현탁화제, 안정화제, 발포제(foaming agent), 방향제, 계면활성제, 물, 이온형 또는 비이온형 유화제, 충전제, 금속이온 봉쇄제 및 킬레이트화제, 보존제, 비타민, 차단제, 습윤화제, 필수 오일, 염료, 안료, 친수성 또는 친유성 활성제, 지질 소낭과 같은 통상적인 보조제, 그리고 담체를 포함한다.
본 발명의 조성물은 당업계에서 통상적으로 제조되는 어떠한 제형으로도 제조될 수 있으며, 예를 들어, 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 오일, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션 및 스프레이 등으로 제형화될 수 있으나, 이에 한정되는 것은 아니다. 보다 상세하게는, 스킨, 스킨 소프트너, 스킨토너, 아스트린젠트, 로션, 밀크로션, 모이스쳐로션, 영양로션, 마사지크림, 영양크림, 아이 크림, 모이스쳐 크림, 핸드크림, 에센스, 영양에센스, 팩, 클렌징폼, 클렌징 워터, 클렌징 로션, 클렌징 크림, 바디로션, 바디클렌져, 비누 및 파우더의 화장품 제형으로 제조될 수 있다.
본 발명의 화장료 조성물의 제형이 페이스트, 크림 또는 겔인 경우에는 담체 성분으로서 동물성유, 식물성유, 왁스, 파라핀, 전분, 트라칸트, 셀룰로오스 유도체, 폴리에틸렌 글리콜, 실리콘, 벤토나이트, 실리카, 탈크 또는 산화아연 등이 이용될 수 있다.
본 발명의 화장료 조성물의 제형이 파우더 또는 스프레이인 경우에는 담체 성분으로서 락토스, 탈크, 실리카, 알루미늄 히드록시드, 칼슘 실리케이트 또는 폴리아미드 파우더가 이용될 수 있고, 특히 스프레이인 경우에는 추가적으로 클로로플루오로 히드로카본, 프로판/부탄 또는 디메틸 에테르와 같은 추진체를 포함할 수 있다.
본 발명의 화장료 조성물의 제형이 용액 또는 유탁액인 경우에는 담체 성분으로서 용매, 용해화제 또는 유탁화제가 이용되고, 예컨대 물, 에탄올, 이소프로판올, 에틸 카보네이트, 에틸 아세테이트, 벤질 알코올, 벤질 벤조에이트, 프로필렌글리콜, 1,3-부틸글리콜 오일, 글리세롤 지방족 에스테르, 폴리에틸렌 글리콜 또는 소르비탄의 지방산 에스테르가 있다.
본 발명의 화장료 조성물의 제형이 현탁액인 경우에는 담체 성분으로서 물, 에탄올 또는 프로필렌글리콜과 같은 액상의 희석제, 에톡실화 이소스테아릴 알코올, 폴리옥시에틸렌 소르비톨 에스테르 및 폴리옥시에틸렌 소르비탄 에스테르와 같은 현탁제, 미소결정성 셀룰로오스, 알루미늄 메타히드록시드, 벤토나이트, 아가 또는 트라칸트 등이 이용될 수 있다.
이하, 본 발명을 실시예에 의해 상세히 설명한다. 단 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예에 한정되는 것은 아니다.

보툴리눔 톡신(Botulinum toxin, 이하 BTAL) 단백질과 파트너 단백질로 사용되는 인간상피세포성장인자 단백질(Human epidermal growth factor, 이하 hEGF)을 코딩하는 유전자를 주형으로 사용하여 숙주 미생물 내에서 발현될 있도록 최적화된, 501개의 아미노산으로 이루어진 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 코딩하는 유전자(서열번호 1) 단편을 제작 및 합성하였다.
보툴리눔 톡신의 카복실 말단(C-말단)에 인간상피세포성장인자 단백질이 결합된 보툴리눔 톡신-인간상피세포성장인자 융합단백질(서열번호 2)을 코딩하는 유전자를 합성하기 위해 정방향 프라이머1(5'-AAGGAGATATACATATGCCGTTCGTTAAC-3', 서열번호 3) 및 역방향 프라이머1(5'-GTCTGAGTTTTTGTTGTAAC-3', 서열번호 4)을 이용하여 대장균에 적합하도록 최적화된 보툴리눔 톡신을 코딩하는 1344개의 뉴클레오티드(서열번호 1의 1~1344번째 염기 서열)를 합성하였으며, 정방향 프라이머2(5'-GTTACAACAAAAACTCAGAC-3', 서열번호 5)와 역방향 프라이머2(5'-GTGCTCGAGGCGCAACTCCC-3', 서열번호 6)를 사용하여 대장균에 적합하도록 최적화된 인간상피세포성장인자 단백질을 코딩하는 159개의 뉴클레오티드(서열번호 1의 1345~1503번째 염기 서열)를 합성하였다. 상기의 방법으로 합성한 각각의 보툴리눔 톡신 단백질과 인간상피세포성장인자를 코딩하는 유전자를 주형으로 하여 정방향 프라이머1(서열번호 3)과 역방향 프라이머2(서열번호 6)를 사용하여 최종적으로 보툴리눔 톡신의 카복실 말단에 인간상피세포성장인자 단백질이 결합된 융합단백질을 코딩하는 1503개의 뉴클레오티드로 구성된 유전자를 중합효소 연쇄반응(PCR)을 통해 합성하였다.
상기 유전자 단편과 재조합 플라스미드를 동일한 제한효소(5' 말단 NdeI 및 3' 말단 XhoI)로 절단하고 삽입하여, 도 1에 나타낸 재조합 플라스미드(pET22b::BTALEGF)를 제작하였다. 제조된 재조합 플라스미드를 E.
coli TOP10에 형질전환시켜서, 숙주 미생물로부터 유전자 컨스트럭트를 대량 얻었다.
그 후, 제조된 재조합 플라스미드를 E.
coli Rosetta2(DE3) pLysS(Novagen, 독일)에 형질전환시켜서, 보툴리눔 톡신-인간상피세포성장인자 융합단백질 생산을 위한 재조합 미생물을 제작하였다.

발현 유도, 분리 및 정제
실시예 1에서 제조한 E.
coli Rosetta2(DE3) pLysS를 각각 1ℓ LB 배지(10% 트립토판, 10% 염화나트륨 및 5% 효모 추출물) 혹은 BSB 배지(1% 트립토판, 0.5% 효모 추출물, 1% 글루코스 및 0.1% HEPES(pH 7.0), ㈜넥스젠바이오텍)를 사용하여 회분 배양(batch culture) 시 OD600=0.6~0.8이 될 때까지 또는 20ℓ 발효기를 사용하여 연속 배양 시에는 OD600=15~20이 될 때까지 배양하였다. 이 후, 세포 배양 배지에 각각 최종 1~5mM IPTG 또는 2% 락토오즈를 가하여 재조합 대장균의 유전자 발현을 유도하였다. 유전자 발현 유도 후 3~4시간 더 배양하고 세포들을 원심분리방법으로 회수하였다. 이 세포들을 완충용액(Phosphate buffered saline, 8g 염화나트륨, 0.2g 염화칼륨, 1.44g 제2인산나트륨(Na2HPO4), 0.24g 인산이수소칼륨(KH2PO4)/ℓ, pH 7.4)에 완전히 현탁시킨 후, 초음파 파쇄기를 사용하여 세포를 파괴하고 세포 내 단백질을 포함하는 용액을 분리하였다.
상기 분리된 용액들을 시료로 하여 15% SDS-폴리아크릴아마이드 겔 전기영동으로 단백질 발현 여부를 확인하였다. 그 결과, IPTG 또는 락토오즈를 통해 발현 유도를 한 세포 조용해물에서 보툴리눔 톡신-인간상피세포성장인자 융합단백질의 발현을 확인할 수 있었다(도 2A).
보툴리눔 톡신-인간상피세포성장인자 융합단백질 내의 인간상피세포성장인자의 유무를 확인하기 위하여 EGF 검출 키트(넥스젠바이오텍, 한국)를 사용하였다. 그 결과 도 2A에서 확인된 보툴리눔 톡신-인간상피세포성장인자 융합단백질 내에 인간상피세포성장인자가 존재함을 확인할 수 있었다 (도 2B).
발현 확인된 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 분리, 정제하기 위해서 응집체(inclusion body)를 가용화 완충용액(5M 요소(urea), pH 11)으로 가용화한 후 초미세 여과방법(0.45㎛ 정밀여과막과 1K 초미세여과막)을 이용하여 재접힘(refolding) 과정을 수행하였으며, 저장용 완충용액(PBS)을 사용하여 최종적으로 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 분리하였다.

피부섬유아세포의
세포 증식 효과
실시예 2에서 분리, 정제된 보툴리눔 톡신-인간상피세포성장인자 융합단백질의 존재가 확인된 시료를 택하여 보툴리눔 톡신-인간상피세포성장인자 융합단백질의 활성을 측정하였다.
피부섬유아세포(Human Dermal Fibroblasts adult, HDFa cell)를 배양한 후 세포에 각각 0, 0.02ppm, 0.2ppm 농도의 융합단백질을 처리한 후 3일 동안 37℃에서 배양하였다. 이 후 크리스탈 바이올렛 염색법으로 피부섬유아세포의 증식 여부를 확인하였다.
그 결과 보툴리눔 톡신-인간상피세포성장인자 융합단백질의 농도(0.02~0.2 ppm)를 증가시킬수록 무처리 대조군(0ppm)과 비교하여 피부섬유아세포의 증식 효과가 우수함을 확인할 수 있었다(도 3). 또한, 보툴리눔 톡신 단백질을 융합하지 않은 인간 상피세포성장인자 단백질 단독 처리군에 비해서 융합단백질의 세포 증식 효과가 더 높게 관찰되었다. 상기 결과를 통해, 보툴리눔 톡신과 인간상피세포성장인자가 융합된 단백질의 상승효과를 확인하였다.

항산화 활성 측정
보툴리눔 톡신-인간상피세포성장인자 융합단백질의 항산화 효능을 측정하기 위해 자유라디칼 소거 효과 측정 방법 중 하나인 DPPH(1,1-diphenyl-2-picryl hydrazyl) 분석을 수행하였다. 상기 시약은 비교적 안정한 자유라디칼로 존재하여 자유라디칼 소거 작용을 확인하는데 일차적으로 사용되는 시험관적 방법 중 하나이다.
DPPH 분석 시 양성 대조군으로 L-아스코르브산(L-Ascorbic acid)을 사용하였다. 실험 방법은 L-아스코르브산, 보툴리눔 톡신-인간상피세포성장인자 융합단백질 및 인간 상피세포성장인자 단백질을 각각 1μM 농도로 준비하였으며, DPPH는 최종 농도가 200μM이 되도록 준비하였다. 상기 각 단백질과 DPPH를 1:1로 혼합한 후 37℃에서 30분 방치하였다. 이 후 ELISA 리더로 520nm 흡광도를 측정하였다. 자유라디칼 소거능(%)은 하기 식 1로 계산하였으며, 그 결과는 도 4에 나타내었다.

그 결과, 보툴리눔 톡신-인간상피세포성장인자 융합단백질은 L-아스코르브산 대조군보다 약 1.5배의 높은 자유라디칼 소거능을 보였으며, 보툴리눔 톡신을 융합하지 않은 인간 상피세포성장인자 단백질과 비교해도 약 2.5배의 높은 자유라디칼 소거능을 보여주었다(도 4). 따라서, 본 발명의 융합단백질은 보툴리눔 톡신에 의해 더욱 높은 항산화 효능을 갖게 된 것을 확인할 수 있었고, 이는 상기 융합단백질의 항산화 효능에 따른 피부 노화 방지 효과를 가질 수 있다는 것을 의미하였다.
실험예 1. 피부 주름 개선, 피부 탄력 유지 효과 및 피부 자극 관능시험
상기 실시예 2에서 분리, 정제한 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 유효성분으로 하여 제조예 1, 2, 3 및 4와 비교예 1, 2, 3 및 4의 화장료 조성물을 제조하여 관능시험을 실시하였다(표 1, 3, 5 및 7 참조).
구체적으로는, 주름 개선 사항을 확인하기 위해 30세 이상 60세 미만의 여성과 남성 총 30명(30대 10명, 40대 10명, 5~60대 10명)을 대상으로 하여, 주름이 많이 분포하는 안면 왼쪽 눈가 부분에는 비교예(대조군)를, 안면 오른쪽 눈가 부분에는 제조예(시험군)를 1일 1회 2주간 지속적으로 사용하게 하였다. 평가는 눈 주위 부분의 주름 펴짐 정도를 평가하였다. 피부 자극 항목에 대해서도 동일한 방법으로 피부의 가려움, 따가움 및 홍반 현상 등에 관하여 관능시험을 실시하였다. 평가 기준은 매우 우수(5점), 우수(4점), 보통(3점), 나쁨(2점), 매우 나쁨(1점)의 오점법 기준에 의거하여 수행하였다. 단, 관능시험 결과에서 정수로 표시하기 힘든 경우에는 소수로 표시하였다. 그 결과는 하기 표 2, 4, 6 및 8에 나타내었다.
<제조예 1 및 비교예 1>
보툴리눔 톡신-인간상피세포성장인자 융합단백질을 유효성분으로 첨가하고, 하기 표 1에 기재된 성분과 함량으로 제조예 1의 스킨을 제조하였다. 또한, 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 유효성분으로 첨가하지 않고, 하기 표 1에 기재된 성분과 함량으로 비교예 1의 스킨을 제조하였다.

보툴리눔 톡신-인간상피세포성장인자 융합단백질을 유효성분으로 첨가하고, 하기 표 5에 기재된 성분과 함량으로 제조예 3의 로션을 제조하였다. 또한, 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 유효성분으로 첨가하지 않고, 하기 표 5에 기재된 성분과 함량으로 비교예 3의 로션을 제조하였다.

보툴리눔 톡신-인간상피세포성장인자 융합단백질을 유효성분으로 첨가하고, 하기 표 7에 기재된 성분과 함량으로 제조예 4의 크림을 제조하였다. 또한, 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 유효성분으로 첨가하지 않고, 하기 표 7에 기재된 성분과 함량으로 비교예 4의 크림을 제조하였다.

상기의 관능시험 결과들을 통해, 본 발명의 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 유효성분으로 포함하는 제조예 1, 2, 3 및 4가 해당 비교예에 비해 피부 주름 개선 효과가 있음을 확인할 수 있었다.

- 서열번호 2의 아미노산 서열로 이루어진 피부 세포 증식 및 항산화 효과가 증가한 보툴리눔 톡신-인간상피세포성장인자 융합단백질.
- 제1항의 피부 세포 증식 및 항산화 효과가 증가한 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 코딩하는 유전자.
- 제2항에 있어서, 상기 유전자는 대장균 코돈 최적화된 서열번호 1의 염기서열로 이루어진 것을 특징으로 하는 유전자.
- 제2항 또는 제3항의 유전자를 포함하는 재조합 벡터.
- 제4항의 재조합 벡터로 형질전환된 숙주세포.
- 제4항의 재조합 벡터로 숙주세포를 형질전환시켜 보툴리눔 톡신-인간상피세포성장인자 융합단백질을 코딩하는 유전자를 과발현시키는 단계를 포함하는, 숙주세포에서 보툴리눔 톡신-인간상피세포성장인자 융합단백질의 생산 방법.
