Abstract
本発明は、担体分子と、担体分子に結合している蛍光色素a及び蛍光色素bを含み、蛍光色素aと蛍光色素bは励起波長が異なり、蛍光色素aと蛍光色素bの間に蛍FRETが生じない蛍光プローブに関する。本発明は、また、標的細胞を前記の蛍光プローブで標識する工程と、蛍光プローブで標識された標的細胞に励起光を照射して、蛍光プローブからの蛍光を観察する工程を含む蛍光検出方法に関する。本発明は、また、細胞を前記の蛍光プローブによって蛍光標識する工程と、蛍光標識細胞をフローサイトメーター又は蛍光顕微鏡によって選別する工程と、選別した蛍光標識細胞を生体内に移植する工程と、生体内の蛍光標識細胞をインビボ蛍光イメージング装置で観察する工程を含む蛍光プローブの使用方法に関する。
Description
本発明は、二種類の蛍光色素を含む蛍光プローブ、蛍光検出方法及び蛍光プローブの使用方法に関する。
近年、癌や発生学の研究領域において、マウス等の動物を用いた生体内の蛍光イメージング解析が広く行われている。マウスに移植したiPS細胞やES細胞等の幹細胞、癌細胞及び肝硬変細胞等の疾患関連細胞等の細胞が移植後、どの位置で分化、生育、転移するかと言った情報の解析は、蛍光イメージング解析がメジャーな手法である。特にマウス等の動物の生存を維持したまま、移植細胞の経過を観察するには、現状、蛍光イメージング解析を用いる手法しかなく、特に重要な解析手法として位置づけられている。
生体内の移植細胞の蛍光イメージグ解析は、通常、移植する前に予めインビトロ(invitro)で細胞を蛍光プローブで標識し、該細胞をマウス等の生体内に移植し、インビボ(in vivo)蛍光イメージング装置で観察することで行われる。例えば、特許文献1には、アニオン性ポルフィリンとカチオン性オルガノアルコキシシラン及び非カチオン性シランを結合させたポルフィリン含有複合体を生体観察のための近赤外蛍光プローブとして用いることが記載されている。
一方、所定量の細胞を蛍光プローブで標識した場合、通常、全ての細胞が蛍光プローブで標識されることはない。そのため、蛍光プローブと結合していない細胞もマウス等の生体内に移植されてしまうことがある。生体観察用の近赤外蛍光プローブの場合、現状、蛍光顕微鏡やフローサイトメーター等のインビトロ蛍光イメージング装置で蛍光を検出することができず、インビトロで近赤外蛍光プローブと結合している蛍光標識細胞を確認することは困難である。それは、インビトロ蛍光イメージング装置とインビボ蛍光イメージング装置で求められる波長特性が異なるためである。インビトロ蛍光イメージング装置では励起波長350~650nm、蛍光波長400~800nmの波長領域で解析を行うことが一般的であり、インビボ蛍光イメージング装置では励起波長600~850nm、蛍光波長650~920nmの波長領域で解析を行うことが一般的である。
上述したとおり、インビトロ蛍光イメージング解析とインビボ蛍光イメージング解析に適した蛍光色素の波長特性は大きく異なるため、現在のところ、インビトロ及びインビボの両方の蛍光イメージング解析に使用できる蛍光プローブは存在しない。
本発明は、前記従来の問題を解決するため、インビトロ及びインビボの両方の蛍光イメージング解析に使用できる蛍光プローブ、蛍光検出方法及び蛍光プローブの使用方法を提供する。
本発明は、担体分子と、前記担体分子に結合している蛍光色素a及び蛍光色素bを含む蛍光プローブにおいて、蛍光色素aの励起波長と蛍光色素bの励起波長は異なり、蛍光色素aと蛍光色素bの間に蛍光共鳴エネルギー移動(FRET)が生じないことを特徴とする蛍光プローブに関する。
前記蛍光プローブは、細胞標識用蛍光プローブであり、蛍光色素aは、インビトロ蛍光イメージング用蛍光色素であり、蛍光色素bは、インビボ蛍光イメージング用蛍光色素であることが好ましい。前記蛍光プローブは、細胞表面に特異的に結合することで細胞を標識してもよく、細胞内部に取り込まれることで細胞を標識してもよい。
蛍光色素aは、励起波長が350~650nmの範囲であることが好ましく、蛍光色素bは、励起波長が600~850nmの範囲であることが好ましい。蛍光色素aがポルフィリンであることが好ましい。蛍光色素bがインドシアニングリーンであることが好ましい。
前記担体分子は、ポリシロキサンであることが好ましい。
前記蛍光プローブは、フローサイトメーターによる蛍光標識細胞の選別に用いてもよい。
本発明は、また、標的細胞を観察する蛍光検出方法であって、標的細胞を蛍光プローブで標識する工程と、蛍光プローブで標識された標的細胞に励起光を照射して、蛍光プローブからの蛍光を観察する工程を含み、前記蛍光プローブは、担体分子と、前記担体分子に結合している蛍光色素a及び蛍光色素bを含み、蛍光色素aの励起波長と蛍光色素bの励起波長は異なり、且つ蛍光色素aと蛍光色素bの間に蛍光共鳴エネルギー移動が生じないことを特徴とする蛍光検出方法に関する。
蛍光プローブで標識された標的細胞に蛍光色素aを励起する励起光を照射し、インビトロ蛍光イメージングによって蛍光プローブからの蛍光を観察してもよい。また、蛍光プローブで標識された標的細胞を生体(ただし、人体を除く)内に移植し、該生体に蛍光色素bを励起する励起光を照射し、インビボ蛍光イメージングによって蛍光プローブからの蛍光を観察してもよい。
本発明は、また、前記の蛍光プローブの使用方法であって、細胞を蛍光プローブによって蛍光標識する工程と、蛍光プローブによって蛍光標識された蛍光標識細胞をフローサイトメーター又は蛍光顕微鏡によって確認して選別する工程と、選別した蛍光標識細胞を生体(ただし、人体を除く)内に移植する工程と、生体内の蛍光標識細胞をインビボ蛍光イメージング装置で観察する工程を含み、前記フローサイトメーターによる蛍光標識細胞の選別は、蛍光色素a由来の蛍光を検出することで行い、前記生体内の蛍光標識細胞のインビボ蛍光イメージング装置による観察は、蛍光色素b由来の蛍光を検出することで行うことを特徴とする蛍光プローブの使用方法に関する。
本発明は、励起波長が異なり、且つ蛍光共鳴エネルギー移動が生じない蛍光色素a及び蛍光色素bを有する蛍光プローブを用いることにより、インビトロ及びインビボの両方の蛍光イメージング解析を行うことができる。特に、既存の蛍光顕微鏡、フローサイトメーター等のインビトロ蛍光イメージング装置にて、細胞のインビトロ蛍光イメージング解析を行うことができ、既存のインビボ蛍光イメージング装置にて、生体内のインビボ蛍光イメージング解析を行うことができる。また、フローサイトメーターにより蛍光プローブで標識された蛍光標識細胞を選別し、選別した蛍光標識細胞をマウス等の生体内に移植し、生存しているマウス等の生体内における蛍光標識細胞の局在を経時的に観察することができる。
本発明の発明者らは、インビトロ及びインビボの両方の蛍光イメージング解析に用いることができる蛍光プローブについて鋭意検討した結果、担体分子と、前記担体分子に結合している蛍光色素a及び蛍光色素bを含む蛍光プローブにおいて、蛍光色素a及び蛍光色素bとして、励起波長が互いに異なり、且つ、互いに蛍光共鳴エネルギー移動が生じない蛍光色素を用いることで、インビトロ及びインビボの両方の蛍光イメージング解析に用いることができる蛍光プローブが得られることを見出し、本発明に至った。従来、二種類の蛍光色素を含む蛍光プローブが存在しているものの、これらの蛍光プローブは、二種類の蛍光色素間で蛍光共鳴エネルギー移動が生じるものであって、インビトロ及びインビボの両方の蛍光イメージング解析に用いることはできない。
(蛍光プローブ)
本発明の蛍光プローブは、担体分子と、前記担体分子に結合している蛍光色素a及び蛍光色素bを含み、蛍光色素a及び蛍光色素bは、励起波長が異なり、且つ、蛍光色素aと蛍光色素bの間に蛍光共鳴エネルギー移動が生じない。蛍光色素aと蛍光色素bは、担体分子と共有結合することが好ましい。
本発明の蛍光プローブは、担体分子と、前記担体分子に結合している蛍光色素a及び蛍光色素bを含み、蛍光色素a及び蛍光色素bは、励起波長が異なり、且つ、蛍光色素aと蛍光色素bの間に蛍光共鳴エネルギー移動が生じない。蛍光色素aと蛍光色素bは、担体分子と共有結合することが好ましい。
蛍光色素aは、インビトロ蛍光イメージング用蛍光色素(可視光線蛍光イメージング用蛍光色素とも称される。)であることが好ましく、励起波長が350~650nmの範囲であり、蛍光波長が400~800nmの範囲であることがより好ましく、励起波長が380~580nmの範囲であり、蛍光波長が450~680nmの範囲であることがさらに好ましい。励起波長と蛍光波長が上述した範囲であると、汎用の蛍光顕微鏡やフローサイトメーター等のインビトロ蛍光イメージング装置にて蛍光プローブで標識された蛍光標識細胞を観察することができる。
蛍光色素aとしては、例えば、ポルフィリン、フルオレセイン、ローダミン等のインビトロ蛍光イメージング用蛍光色素を用いることができる。ポルフィリンは、特に限定されないが、インビトロ蛍光イメージング用蛍光色素として好適である観点から、カルボキシル基を有するポルフィリンを用いることができる。なお、“ポルフィリン”は、4つのピロール環がα位置で4つのメチン基と交互に結合した環状化合物とその誘導体の総称の意味で用いている。カルボキシル基を有するポルフィリンは、一般的に、励起波長が400~650nmの範囲であり、蛍光波長が600~740nmの範囲である。
カルボキシル基を有するポルフィリンとしては、例えば、下記一般式(I)で表される化合物を用いることができる。
前記一般式(I)中、R1b、R2b、R3b、及びR4bは、同一又は異なっていてもよく、カルボキシル基(COOH)、スルホ基(SO3H)又は水素原子(H)を示す(ただし、全てが水素原子又は全てがスルホ基である場合は除く。)。好ましくは、前記一般式(I)中、R1b、R2b、R3b、及びR4bが全てカルボキシル基、又はR1bはカルボキシル基でR2b、R3b、及びR4bは水素原子である。カルボキシル基を有するポルフィリンは、より好ましくは、前記一般式(I)で表され、R1b、R2b、R3b、及びR4bが、全てカルボキシル基である化合物である。R1b、R2b、R3b、及びR4bが全てカルボキシル基である化合物は、テトラキス(4-カルボキシフェニル)ポルフィリン(TCPP)と呼ばれる。
また、例えばビリルビン、ヘミン、プロトポルフィリン等も、カルボキシル基を有するポルフィリンとして用いることができる。
本発明に用いるカルボキシル基を有するポルフィリンは、公知の化合物であるか、又は公知の方法により容易に製造できる。例えば東京化成工業株式会社等から市販のものを入手することができる。
蛍光色素bは、蛍光色素aと励起波長が異なり、且つ、蛍光色素aとの間に蛍光共鳴エネルギー移動が生じない蛍光色素を用いる。蛍光色素bは、インビボ蛍光イメージング用蛍光色素である(近赤外線蛍光イメージング用蛍光色素とも称される。)ことが好ましく、励起波長が600~850nmの範囲であり、蛍光波長が650~920nmの範囲であることがより好ましく、励起波長が630~790nmの範囲であり、蛍光波長が680~910nmの範囲であることがさらに好ましい。励起波長と蛍光波長が上述した範囲であると、汎用のインビボ蛍光イメージング装置にてマウス等の生体内の蛍光プローブで標識された蛍光標識細胞を観察することができる。
蛍光色素bとしては、例えば、インドシアニン化合物、クマリン、ローダミン、キサンテン、ヘマトポルフィリン、及びフルオレスカミン等のインビボ蛍光イメージング用蛍光色素を用いることができる。インドシアニン化合物としては、生体内の安全性の観点から、インドシアニングリーンやその誘導体を用いることが好ましい。インドシアニングリーンは、一般的に、励起波長が740~790nmの範囲であり、蛍光波長が810~860nmの範囲である。インドシアニングリーンの誘導体は、インドシアニングリーンの主要な骨格及び機能を維持しつつその一部を他の官能基等で置換した化合物をいい、励起波長が750~790nmの範囲であり、蛍光波長が790~900nmの範囲である。蛍光色素bとして、励起波長が740~790nmの範囲であり、蛍光波長が790~900nmの範囲のインドシアニングリーンやその誘導体を用いることで、マウス等の生体におけるエサ等からの自家蛍光に影響されることなく、汎用のインビボ蛍光イメージング装置にてマウス等の生体内の蛍光プローブで標識された蛍光標識細胞を観察することができる。また、マウス等の生体内の臓器に局在する蛍光プローブで標識された蛍光標識細胞を観察することができる。
前記担体分子は、蛍光色素a及び蛍光色素bと結合し、蛍光色素a及び蛍光色素bの異なる励起波長を維持することができるものであればよい。例えば、ポリシラン、ポリゲルマン、ポリスタナン、ポリシロキサン、ポリシルセスキオキサン、ポリシラザン、ポリボラジレン、ポリホスファゼン等の無機ポリマーやポリピロール、ポリエチレングリコール、多糖等の有機ポリマー等のポリマーを用いることができる。蛍光色素a及び蛍光色素bとして、蛍光色素aの蛍光が蛍光色素bを励起しないように、適宜選択して用いることができる。蛍光色素a及び蛍光色素bのいずれか一方は、担体分子であるポリマーの主鎖構造中に組み込まれていることが好ましく、他方は同担体の表面に存在する官能基と結合することが好ましい。このような構造であると、FRETが生じにくい。蛍光色素a及び蛍光色素bの間にFRETを生じさせない観点から、前記担体分子は、ポリシロキサン(以下において、シロキサン結合を有するポリマーとも記す。)であることが好ましい。ポリシロキサンは、例えば、後述するように、シラン化合物が加水分解、重縮合することで形成される。
本発明の蛍光プローブは、細胞標識用蛍光プローブとして用いることが好ましい。細胞標識用蛍光プローブとして用いる場合、細胞表面に特異的に結合することで細胞を標識してもよい。この場合、本発明の蛍光プローブは、細胞の表面を特異的に認識する物質と結合する細胞表面結合物質を有することが好ましい。蛍光プローブが細胞表面結合物質を介して細胞表面に特異的に結合して細胞を標識することで、細胞標識用蛍光プローブとして用いることができる。細胞の表面を特異的に認識する物質とは、特定の細胞の表面に存在するタンパク質、脂質、糖鎖、及び/又は核酸等である。例えば、癌細胞の表面には、癌細胞に特異的な葉酸受容体、トランスフェリン受容体、抗原等が存在する。蛍光プローブに葉酸、トランスフェリン、抗体等を結合することにより、癌細胞を特異的に標識することができる。また、iPS細胞やES細胞等の幹細胞の表面には細胞表面マーカー(膜タンパク質)等が存在する。蛍光プローブに細胞表面マーカーと特異的に結合する分子等を結合することにより、iPS細胞やES細胞等の幹細胞を特異的に標識することができる。
本発明の蛍光プローブは、また、細胞の内部に取り込まれることで細胞を標識してもよい。このような細胞としては、例えば、マクロファージ、樹状細胞、免疫細胞、癌細胞、iPS細胞等が挙げられる。細胞内部に蛍光プローブを取り込んだマクロファージ、樹状細胞、免疫細胞、癌細胞、iPS細胞等を用い、免疫細胞療法において、マクロファージや樹状細胞の体内での動態を確認することができる。
本発明の蛍光プローブは、担体分子と蛍光色素aを結合、好ましくは共有結合させて担体分子と蛍光色素aの複合体を形成し、その後、担体分子と蛍光色素aの複合体からなるナノ粒子表面に蛍光色素bを結合、好ましくは共有結合させて、担体分子、蛍光色素a及び蛍光色素bの複合体を形成することで作製することができる。担体分子と蛍光色素aの複合体からなるナノ粒子に蛍光色素bを結合させる際に、同時に、細胞表面結合物質を担体分子と蛍光色素aの複合体からなるナノ粒子に結合させてもよい。或いは、本発明の蛍光プローブは、担体分子と蛍光色素bを結合、好ましくは共有結合させて担体分子と蛍光色素bの複合体を形成し、その後、担体分子と蛍光色素bの複合体からなるナノ粒子に蛍光色素aを結合、好ましくは共有結合させて、担体分子、蛍光色素a及び蛍光色素bの複合体を形成することで作製してもよい。担体分子と蛍光色素bの複合体からなるナノ粒子に蛍光色素aを結合させる際に、同時に、細胞表面結合物質を担体分子と蛍光色素bの複合体からなるナノ粒子に結合させてもよい。なお、担体分子と蛍光色素aの複合体、担体分子と蛍光色素bの複合体、担体分子、蛍光色素a及び蛍光色素bの複合体において、蛍光色素a及び蛍光色素bの励起波長及び蛍光波長は、担体分子と結合する前後でほとんど変化しないことが好ましい。
蛍光色素aと蛍光色素bとして、励起波長が異なりかつ一方の蛍光色素の蛍光が他方の蛍光色素を励起しない蛍光色素を選ぶこと、担体分子、蛍光色素a及び蛍光色素bの複合体中の蛍光色素aと蛍光色素bの配置等を調整すること等により、蛍光色素aと蛍光色素bの間でFRETが生じないようにする。担体分子、蛍光色素a及び蛍光色素bの複合体において、担体分子がポリシロキサンである場合、蛍光色素a及び蛍光色素bのいずれかはシロキサンネットワーク(ポリマーの主鎖)中に組み込まれ、他方は、シロキサンネットワーク中に組み込まれず、ポリシロキサンと一方の蛍光色素の複合体からなるナノ粒子表面の官能基(シロキサンネットワークを形成していない官能基)と結合することが好ましい。このような構造であると、蛍光色素a及び蛍光色素bの間でFRETが生じにくい。
担体分子がポリシロキサンであり、蛍光色素aがポルフィリンであり、蛍光色素bがインドシアニングリーンである場合、例えば下記のように蛍光プローブを作製することができる。
まず、アミノ基を有するシランとカルボキシル基を有するポルフィリンを反応させてポルフィリンを分子内に含むシラン(以下において、「ポルフィリン‐シラン」とも記す。)を得る。具体的には、溶媒にアミノ基を有するシランとカルボキシル基を有するポルフィリンを溶解させ、縮合剤を加えアミド化反応を起こす。溶媒としては、例えば、N,N-ジメチルホルムアミド(DMF)等を用いることができる。縮合剤としては、例えば、カルボジイミド等を使用することができる。カルボジイミドとしては、特に限定されないが、例えば、N,N´-ジプロピルカルボジイミド等が挙げられる。副生成物を減らすために、スクシンイミド等を加えてもよい。スクシンイミドとしては、特に限定されないが、例えば、N-ヒドロキシスクシンイミド等が挙げられる。反応温度は、特に限定されないが、合成コストの観点から、例えば、20~150℃であることが好ましく、20~80℃であることがより好ましい。反応時間は、特に限定されないが、例えば、1~72時間であることが好ましく、1~24時間であることがより好ましい。反応後、遠心分離して生成物を沈殿として回収することで、ポルフィリン‐シランを得ることができる。
上記反応において、アミノ基を含むシランとカルボキシル基を有するポルフィリンのモル比(アミノ基を含むシラン:カルボキシル基を有するポルフィリン)は、4:1~1:1であることが好ましく、より好ましくは4:1~2:1であり、さらに好ましくは4:1である。
次に、上記で得られたポルフィリン‐シランと一種以上の官能基を有するシラン(以下において、「官能基シラン」とも記す。)との加水分解・縮合反応により、シロキサンとポルフィリンの複合体を得る。具体的には、溶媒にポルフィリン‐シランと官能基シランを溶解させ、さらにアルカリ溶液を加えて反応させる。溶媒としては、例えば、DMF等を用いることができる。アルカリ溶液としては、例えば、pHが8以上のアンモニア水、水酸化ナトリウムの水溶液等を用いることができる。反応温度は、特に限定されないが、合成コストの観点から、例えば、20~200℃であることが好ましく、60~80℃であることがより好ましい。反応時間は、特に限定されないが、例えば、1~72時間であることが好ましく、3~24時間であることがより好ましい。反応後、遠心分離することにより生成物を沈殿として回収することで、ポリシロキサンにポルフィリンが共有結合したポリシロキサンとポルフィリンの複合体を得ることができる。
上記反応において、ポルフィリン‐シランと官能基シランのモル比(ポルフィリン‐シラン:官能基シラン)は、1:2~1:100であることが好ましく、より好ましくは1:21~1:50であり、さらに好ましくは1:30~1:40である。
アミノ基を有するシランとしては、アミノ基を有するものであればよく、特に限定されない。例えば、下記一般式(II)で表される化合物を好適に用いることができる。

一般式(II)において、XはH2NCmH2m-、H2NCnH2n-HNCmH2m-、又はPh-NHCmH2m-〔ここでPhはフェニル基を示す。〕で示される基を示し、中でもH2NCmH2m-、又はH2NCnH2n-HNCmH2m-で示される基が好適である。m及びnは、同一又は異なって、1~6の整数を示す。mは1、2、3又は4が好ましく、1、2、又は3がより好ましい。nは1、2、3又は4が好ましく、1、2、又は3がより好ましい。また、上記のH2NCmH2m-、H2NCnH2n-HNCmH2m-、Ph-NHCmH2m-は、それぞれ、H2N(CH2)m-、H2N(CH2)n-HN(CH2)m-、Ph-NH(CH2)m-であることが好ましい。
また、R1a及びR2aは、同一又は異なって、炭素数1~6のアルキル基を示す。当該アルキル基は、直鎖状又は分岐鎖状であってよく、直鎖状が好ましい。また、炭素数1、2、3又は4のアルキル基が好ましい。炭素数1~6のアルキル基としては、具体的には、メチル、エチル、n-プロピル、イソプロピル、n-ブチル、イソブチル、tert-ブチル、sec-ブチル、n-ペンチル、1-エチルプロピル、イソペンチル、ネオペンチル、n-ヘキシル、1,2,2-トリメチルプロピル、3,3-ジメチルブチル、2-エチルブチル、イソヘキシル、3-メチルペンチル基等が例示される。中でも、メチル、エチル、n-プロピル、イソプロピル、n-ブチル、イソブチルが好ましい。
また、iは1又は2、jは0又は1、〔但し(4-i-j)≧2〕を示す。すなわち、(i、j)は(1、0)、(1、1)、又は(2、0)を示す。
一般式(II)で表される化合物としては、例えば、3-アミノプロピルトリエトキシシラン(APTES)、3-(2-アミノエチルアミノ)プロピルジメトキシメチルシラン、3-(2-アミノエチルアミノ)プロピルトリエトキシシラン、3-(2-アミノエチルアミノ)プロピルトリメトキシシラン、3-アミノプロピルジエトキシメチルシラン、3-アミノプロピルトリメトキシシラン、トリメトキシ[3-(フェニルアミノ)プロピル]シラン等が挙げられる。中でも、3-アミノプロピルトリエトキシシラン(APTES)及び/又は3-アミノプロピルトリメトキシシランが好ましい。
上述したアミノ基を有するシランは、例えば、東京化成工業株式会社製の市販品を用いることができる。また、アミノ基を有するシランは1種単独で又は2種以上を組み合わせて用いることができる。
官能基シランは、一種以上の官能基を有するものであれば特に限定されず、一種の官能基を有する単官能基シランであってもよく、二種以上の官能基を有する多官能基シランであってもよい。官能基を有するシランとして、例えば一般式(III):

上記において、α、β、δ、ε、ζ、η、θ及びιは、それぞれ独立して1~6の整数数を示し、好ましくは1、2、3又は4の整数を示し、γは0~8の整数を示し、好ましくは0、1、2、3、4、5、6又は7の整数を示す。また、炭素数1~18のアルキル基は、炭素数1~12が好ましく、炭素数1、2、3、4、5、6、7又は8がより好ましい。また、直鎖状であっても分岐鎖状であってもよく、直鎖状であることが好ましい。また、上記のClCαH2α-、HSCβH2β-、CF3CγF2γ-CδH2δ-、CH2=C(CH3)COOCεH2ε-、CH2=CHCOOCζH2ζ-、HN-CONH-CηH2η-は、それぞれ、Cl(CH2)α-、HS(CH2)β-、CF3(CF2)γ-(CH2)δ-、CH2=C(CH3)COO(CH2)ε-、CH2=CHCOO(CH2)ζ-、HN-CONH-(CH2)η-であることが好ましい。
また、R1cは、炭素数1~6のアルキル基、又は-(CH2)τ-OCH3を示し、R2cは、炭素数1~6のアルキル基、又はフェニル基を示す。R1c及びR2cは同一であっても異なってもよい。R1c及びR2cのいずれにおいても、炭素数1~6のアルキル基は、直鎖状又は分岐状であってよく、直鎖状が好ましい。また、炭素数1、2、3又は4のアルキル基が好ましい。炭素数1~6のアルキル基としては、具体的には、メチル、エチル、n-プロピル、イソプロピル、n-ブチル、イソブチル、tert-ブチル、sec-ブチル、n-ペンチル、1-エチルプロピル、イソペンチル、ネオペンチル、n-ヘキシル、1,2,2-トリメチルプロピル、3,3-ジメチルブチル、2-エチルブチル、イソヘキシル、3-メチルペンチル基等が例示される。中でも、メチル、エチル、n-プロピル、イソプロピル、n-ブチル、イソブチルが好ましい。また、τは1~4の整数(好ましくは1、2又は3)を示す。
また、pは1、2又は3、qは0、1又は2、但し、(4-p-q)≧1を示す。すなわち、(p、q)は(1、0)、(1、1)、(1、2)、(2、0)、(2、1)又は(3、0)を示す。
また、官能基シランとしては、例えば下記の一般式(IV)で表される化合物も好適に用いることができる。

また、多官能基性シランとして、N-[2-(N-ビニルベンジルアミノ)エチル]-3-アミノプロピルトリメトキシシラン塩酸塩を用いてもよい。

また、一般式(IV)で表される化合物としては、具体的には、アリルトリクロロシラン、トリクロロビニルシラン、3-クロロプロピルトリクロロシラン、トリクロロ(1H,1H,2H,2H-ヘプタデカフルオロデシル)シラン、トリクロロ(1H,1H,2H,2H-トリデカフルオロ-n-オクチル)シラン、ブチルトリクロロシラン、シクロヘキシルトリクロロシラン、デシルトリクロロシラン、ドデシルトリクロロシラン、エチルトリクロロシラン、n-オクチルトリクロロシラン、フェニルトリクロロシラン、トリクロロ-2-シアノエチルシラン、トリクロロヘキシルシラン、トリクロロ(メチル)シラン、トリクロロオクタデシルシラン、トリクロロ(プロピル)シラン、トリクロロテトラデシルシラン等が挙げられる。
上記官能基シランは、例えば東京化成工業株式会社製の市販品を用いることができる。また、官能基シランは1種単独で又は2種以上を組み合わせて用いることができる。
次に、ポリシロキサンとポルフィリンの複合体からなるナノ粒子と、インドシアニングリーンを反応させてポリシロキサン、ポルフィリン及びインドシアニングリーンの複合体を形成する。具体的には、ポリシロキサンとポルフィリンの複合体からなるナノ粒子の水溶液にインドシアニングリーンの水溶液を加えて、4~50℃の温度で、1~24時間撹拌することで反応させる。ポリシロキサンとポルフィリンの複合体からなるナノ粒子表面の官能基とインドシアニングリーンが共有結合してポリシロキサン、ポルフィリン及びインドシアニングリーンの複合体を形成する。インドシアニングリーンとして、ポリシロキサンとポルフィリンの複合体からなるナノ粒子表面の官能基と結合する官能基で修飾されたインドシアニングリーンを用いる。例えば、ポリシロキサンとポルフィリンの複合体からなるナノ粒子を形成した官能基シランが3-メルカプトプロピル(ジメトキシ)メチルシラン、(3-メルカプトプロピル)トリエトキシシラン及び(3-メルカプトプロピル)トリメトキシシラン等のチオール基を有する場合、マレイミド基が結合しているインドシアニングリーンを用いることができる。なお、ポリシロキサンとポルフィリンの複合体からなるナノ粒子とインドシアニングリーンを反応させて、ポリシロキサンとポルフィリンの複合体からなるナノ粒子にインドシアニングリーンを結合させると同時に、細胞表面結合物質、例えば、葉酸をポリシロキサンとポルフィリンの複合体からなるナノ粒子に結合させてもよい。葉酸としては、ポリエチレングリコール(PEG)の片末端にマレイミド基が、もう一方の片末端に葉酸が結合したものを用いることができる。
上記反応において、ポリシロキサンとポルフィリンの複合体からなるナノ粒子のインドシアニングリーンと共有結合する官能基と、インドシアニングリーンを修飾し、ポリシロキサンとポルフィリンの複合体からなるナノ粒子と共有結合する官能基のモル比(ポリシロキサンとポルフィリンの複合体からなるナノ粒子のインドシアニングリーンと共有結合する官能基:インドシアニングリーンを修飾した官能基)は、1:1~3:1であることが好ましく、1:1~2:1であることがより好ましく、1:1であることがさらに好ましい。
ポルフィリンとしてTCPPを用いた場合、ポリシロキサンとポルフィリンの複合体において、TCPPのカルボキシル基はアミド結合を形成して存在することが好ましく、TCPPはアミド結合によりシロキサンネットワーク(ポリシロキサンの主鎖)中に組み込まれていることがより好ましい。そして、インドシアニングリーンは、ポリシロキサンとポルフィリンの複合体からなるナノ粒子表面の官能基(ポリシロキサンとポルフィリンの複合体からなるナノ粒子を形成する官能基シランの官能基)と結合することが好ましい。このような構造になることで、ポルフィリンとインドシアニンの間にFRETが生じない。本発明において、ポリシロキサンとポルフィリンの複合体等の複合体や蛍光プローブの構造は、後述するように、フーリエ変換赤外分光法で解析することができる。
前記蛍光プローブは、特に限定されないが、インビトロ及びインビボの両方の蛍光イメージングに好適である観点から、蛍光色素aを1質量%以上含み、蛍光色素bを0.1質量%以上含むことが好ましく、より好ましくは蛍光色素aを5~90質量%含み、蛍光色素bを1~10質量%含む。また、蛍光色素aがポルフィリンであり、蛍光色素bがインドシアニングリーンであり、担体分子がポリシロキサンである場合、蛍光プローブにおいて、ポルフィリンの4つのピロールの環状構造部分(ポルフィン)の含有量が1質量%以上であることが好ましく、インドシアニングリーンの含有量が0.1質量%以上であることが好ましい。より好ましくは、ポルフィリンの4つのピロールの環状構造部分(ポルフィン)の含有量が5~90質量%であり、インドシアニングリーンの含有量が1~10質量%である。本発明において、後述するように、ポリシロキサンとポルフィリンの複合体等の複合体や蛍光プローブの熱重量・示差熱分析により、ポルフィリン等の蛍光色素やシリカポリマーなどの担体分子の含有量を測定することができる。
前記蛍光プローブは、特に限定されないが、細胞を標識しやすい観点から、平均粒子径が3~250nmであることが好ましく、10~80nmであることがより好ましい。本発明において、平均粒子径は、動的光散乱法で測定する。
(蛍光検出方法)
本発明の蛍光検出方法は、標的細胞を観察するのに用いる。具体的には、標的細胞を蛍光プローブで標識する工程と、蛍光プローブで標識された標的細胞に励起光を照射して、蛍光プローブからの蛍光を観察する工程を含む。蛍光プローブとしては、上述した本発明の蛍光プローブを用いる。
本発明の蛍光検出方法は、標的細胞を観察するのに用いる。具体的には、標的細胞を蛍光プローブで標識する工程と、蛍光プローブで標識された標的細胞に励起光を照射して、蛍光プローブからの蛍光を観察する工程を含む。蛍光プローブとしては、上述した本発明の蛍光プローブを用いる。
標的細胞の標識は、蛍光プローブを細胞表面に結合させること、あるいは、蛍光プローブを細胞内に取り込ませることで行うことができる。蛍光プローブを細胞表面に結合させる場合、蛍光プローブは細胞表面結合物質を有する。例えば、蛍光プローブが葉酸を有する場合、ヒトの子宮頸癌由来のHeLa S3細胞やヒトの大腸がん由来のHCT116細胞等の癌細胞の表面に結合してこれらの癌細胞を標識することができる。具体的には、細胞を蛍光プローブ入りの細胞培養培地で培養することで、細胞を蛍光プローブで標識することができる。標的細胞としては、iPS細胞やES細胞等の幹細胞、癌細胞及び肝硬変細胞等の疾患関連細胞等が挙げられる。
蛍光プローブで標識された標的細胞に蛍光色素aを励起する励起光を照射し、蛍光顕微鏡やフローサイトメーター等のインビトロ蛍光イメージング装置を用いたインビトロ蛍光イメージングによって蛍光プローブからの蛍光を観察することができる。例えば、波長範囲が350~650nmの励起光で蛍光色素aを励起し、400~800nm波長範囲の蛍光を観察する。好ましくは、波長範囲が380~580nmの励起光で蛍光色素aを励起し、450~680nm波長範囲の蛍光を観察する。
また、蛍光プローブで標識された標的細胞を生体(ただし、人体を除く)内に移植し、該生体内の蛍光プローブで標識された標的細胞に蛍光色素bを励起する励起光を照射し、インビボ蛍光イメージング装置を用いたインビボ蛍光イメージングによって蛍光プローブからの蛍光を観察することができる。例えば、波長範囲が600~850nmの励起光で蛍光色素bを励起し、650~920nm波長範囲の蛍光を観察する。好ましくは、波長範囲が630~790nmの励起光で蛍光色素bを励起し、680~910nm波長範囲の蛍光を観察する。本発明において、生体は、蛍光観察を行うことができる動物であれば特に限定は無い。好ましくは哺乳動物である。特に好ましくは、マウス、ラット等のげっ歯類の哺乳動物である。なお、マウス等の生体内への蛍光プローブで標識された標的細胞の移植は、後述するとおりに行うことができる。
本発明の蛍光プローブを用いることで、インビトロ及びインビボのいずれにおいても、蛍光プローブで標識された標的細胞を観察することができる。
(蛍光プローブの使用方法)
本発明の蛍光プローブの使用方法は、細胞を蛍光プローブによって蛍光標識する工程と、蛍光プローブによって標識された蛍光標識細胞をフローサイトメーター又は蛍光顕微鏡によって確認して選別する工程と、選別した蛍光標識細胞を生体(ただし、人体を除く)内に移植する工程と、生体内の蛍光標識細胞をインビボ蛍光イメージング装置で観察する工程を含む。
本発明の蛍光プローブの使用方法は、細胞を蛍光プローブによって蛍光標識する工程と、蛍光プローブによって標識された蛍光標識細胞をフローサイトメーター又は蛍光顕微鏡によって確認して選別する工程と、選別した蛍光標識細胞を生体(ただし、人体を除く)内に移植する工程と、生体内の蛍光標識細胞をインビボ蛍光イメージング装置で観察する工程を含む。
まず、細胞を蛍光プローブによって蛍光標識する。細胞の標識は、蛍光プローブを細胞表面に結合させること、あるいは、蛍光プローブを細胞内に取り込ませることで行うことができる。蛍光プローブを細胞表面に結合させる場合、蛍光プローブは細胞表面結合物質を有する。例えば、蛍光プローブが葉酸を有する場合、ヒトの子宮頸癌由来のHeLa S3細胞やヒトの大腸がん由来のHCT116細胞等の癌細胞の表面に結合してこれらの癌細胞を標識することができる。具体的には、細胞を蛍光プローブ入りの細胞培養培地で培養することで、細胞を蛍光プローブで標識することができる。細胞としては、iPS細胞やES細胞等の幹細胞、癌細胞及び肝硬変細胞等の疾患関連細胞等が挙げられる。
次に、蛍光プローブによって標識された蛍光標識細胞をフローサイトメーターによって選別する。フローサイトメーター又は蛍光顕微鏡による蛍光標識細胞の確認・選別は、蛍光色素a由来の蛍光を検出することで行う。具体的には、蛍光プローブによって蛍光標識操作を行った後の細胞をフローサイトメーターに供給し、蛍光色素aの励起する波長の励起光で蛍光色素aを励起し、励起された蛍光色素aからの蛍光を検出することで、蛍光プローブによって蛍光標識された蛍光標識細胞を選別することができる。フローサイトメーターによる観察は、例えば、励起波長350~640nm、蛍光波長630~780nmの条件下で行うことができる。或いは、蛍光プローブによって蛍光標識操作を行った後の細胞を蛍光顕微鏡に供給し、蛍光色素aの励起する波長の励起光で蛍光色素aを励起し、励起された蛍光色素aからの蛍光を検出することで、蛍光プローブによって蛍光標識された蛍光標識細胞を選別することができる。蛍光顕微鏡による観察は、例えば、励起波長350~640nm、蛍光波長630~780nmの条件下で行うことができる。従来は、蛍光プローブによって蛍光標識操作を行った後の細胞をそのままマウス等の生体内に移植していたが、本発明の蛍光プローブを用いることで、蛍光プローブによって蛍光標識された蛍光標識細胞のみをフローサイトメーターによって選別してマウス等の生体内に移植することができる。
次に、選別した蛍光標識細胞を生体(ただし、人体を除く)内に移植する。選別した蛍光標識細胞の生体内への移植は、例えば、皮下、静脈、腹腔等からの投与によって行うことができる。例えば、マウスの場合、蛍光標識細胞をリン酸緩衝生理食塩水に懸濁し、シリンジを用いて蛍光標識細胞の懸濁液をマウスの尾静脈に注射することで、蛍光標識細胞をマウス体内に移植する。
次に、生体内の蛍光標識細胞をインビボ蛍光イメージング装置で観察する。インビボ蛍光イメージング装置としては、特に限定されず、例えば、倉敷紡績社の蛍光インビボイメージング装置「KURAVIC KV-700」、パーキンエルマー社のIVIS Imaging System、MaestroTM等を用いることができる。生体内の蛍光標識細胞のインビボ蛍光イメージング装置による観察は、蛍光色素b由来の蛍光を検出することで行う。具体的には、蛍光色素bを励起する波長の励起光で蛍光色素bを励起し、励起された蛍光色素bからの蛍光を検出する。生体内の蛍光標識細胞のインビボ蛍光イメージング装置による観察は、例えば、励起波長650~760nm、蛍光波長760~850nmの条件下で行うことができる。生体内の蛍光標識細胞のインビボ蛍光イメージング装置による観察を経時的に行うことで、マウス等の生体内に移植したiPS細胞やES細胞等の幹細胞、癌細胞及び肝硬変細胞等の疾患関連細胞等の細胞が移植後、どの位置で分化、生育、転移するかと言った情報を取得することができる。
従来は、細胞を蛍光プローブで標識を行った後、細胞が実際に蛍光プローブで標識されたか否かを確認せず、マウス等の生体内に移植していたが、本発明の蛍光プローブを用いることで、フローサイトメーターによって蛍光プローブで標識された蛍光標識細胞のみを選択し、マウス等の生体内に移植し、移植後の蛍光標識細胞の生体内における経過をインビボ蛍光イメージング装置で観察することができる。
以下、実施例を用いて本発明を具体的に説明する。なお、本発明は下記の実施例に限定されるものではない。
(試薬)
テトラキス(4-カルボキシフェニル)ポルフィリン(TCPP)、(3-メルカプトプロピル)トリメトキシシラン(MPTMS)、N,N-ジメチルホルムアミド(DMF)、マレイミド基が結合したインドシアニングリーン(分子量853、以下において、単に「ICG-Mal」とも記す。)、葉酸とマレイミド基が両末端に結合したポリエチレングリコール(PEG)(分子量3400、以下において、単に「FA-PEG-Mal」とも記す。)は、いずれも、東京化成工業株式会社から入手した。MPTMS、ICG-Mal及びFA-PEG-Malの構造式を下記に示した。
テトラキス(4-カルボキシフェニル)ポルフィリン(TCPP)、(3-メルカプトプロピル)トリメトキシシラン(MPTMS)、N,N-ジメチルホルムアミド(DMF)、マレイミド基が結合したインドシアニングリーン(分子量853、以下において、単に「ICG-Mal」とも記す。)、葉酸とマレイミド基が両末端に結合したポリエチレングリコール(PEG)(分子量3400、以下において、単に「FA-PEG-Mal」とも記す。)は、いずれも、東京化成工業株式会社から入手した。MPTMS、ICG-Mal及びFA-PEG-Malの構造式を下記に示した。
(実施例1)
(蛍光プローブの作製)
<担体分子と蛍光色素aの複合体の作製>
(1)TCPPをDMFに溶解させたTCPPのDMF溶液(3.8mM、32mL)にAPTES(462μmol)を加え、続いてN,N´-ジプロピルカルボジイミド(480μmol)とN-ヒドロキシスクシンイミド(480μmol)を加え、50℃で24時間撹拌した。
(2)上記で得られたポルフィリン‐シランのDMF溶液200μL(ポルフィリン‐シラン0.75μmol)を、MPTMS5μL(0.027mmol)と混合した。得られた混合液に、DMFを500μLとアンモニア水(濃度:28質量%、pH11)を300μL加えて、80℃で24時間反応を行った。
(3)24時間後、生成物を遠心分離(15000rpm、20分)で沈殿物として回収し、生成物を水とエタノールで数回洗浄し、最終的に1mLの水に分散させた。
(4)得られた物質は、ポリシロキサンを担体分子とし、ポルフィリンを蛍光色素aとするポリシロキサンにポルフィリンが共有結合したポリシロキサンとポルフィリンの複合体であり、動的光散乱法(Beckman Coulter製「DelsaMax PRO」)により測定したところ、平均粒子径が約40nmの蛍光ハイブリッドナノ粒子(以下において、単に「PPS HNPs」とも記す。)であった。
(蛍光プローブの作製)
<担体分子と蛍光色素aの複合体の作製>
(1)TCPPをDMFに溶解させたTCPPのDMF溶液(3.8mM、32mL)にAPTES(462μmol)を加え、続いてN,N´-ジプロピルカルボジイミド(480μmol)とN-ヒドロキシスクシンイミド(480μmol)を加え、50℃で24時間撹拌した。

(4)得られた物質は、ポリシロキサンを担体分子とし、ポルフィリンを蛍光色素aとするポリシロキサンにポルフィリンが共有結合したポリシロキサンとポルフィリンの複合体であり、動的光散乱法(Beckman Coulter製「DelsaMax PRO」)により測定したところ、平均粒子径が約40nmの蛍光ハイブリッドナノ粒子(以下において、単に「PPS HNPs」とも記す。)であった。
<担体分子と蛍光色素aの複合体に蛍光色素bを結合させる工程>
(1)上記で得られたPPS HNPsの水分散液1mLに、ICG-Malの水溶液を63μL(濃度:2mg/mL、ICG-Mal:1.48×10-4mmol)添加し、30℃で3時間撹拌して反応させた。
(2)反応後、生成物を遠心分離(15000rpm、20分)で沈殿物として回収し、水で数回洗浄し、最終的に1mLの水に分散させた。
(3)得られた物質は、ポリシロキサンを担体とし、ポルフィリンを蛍光色素aとし、インドシアニングリーンを蛍光色素bとするポリシロキサンにポルフィリン及びインドシアニングリーンが共有結合した複合体(蛍光プローブ)であり、動的光散乱法(Beckman Coulter製「DelsaMax PRO」)により測定したところ、平均粒子径が約50nmの蛍光ハイブリッドナノ粒子(以下において、単に「ICG-PPS HNPs」とも記す。)であった。
(1)上記で得られたPPS HNPsの水分散液1mLに、ICG-Malの水溶液を63μL(濃度:2mg/mL、ICG-Mal:1.48×10-4mmol)添加し、30℃で3時間撹拌して反応させた。

(実施例2)

(1)上記で得られたPPS HNPsの水分散液1mLに、FA-PEG-Malの水溶液を251μL(濃度:2mg/mL、FA-PEG-Mal:1.48×10-4mmol)と、ICG-Malの水溶液(濃度:2mg/mL、ICG-Mal:1.48×10-4mmol)を63μL添加し、30℃で3時間撹拌した。
(2)反応後、生成物を遠心分離(15000rpm、20分)で沈殿物として回収し、水で数回洗浄し、最終的に1mLの水に分散させた。
(3)得られた物質は、ポリシロキサンを担体分子とし、ポルフィリンを蛍光色素aとし、インドシアニングリーンを蛍光色素bとし、葉酸を細胞表面結合物質とするポリシロキサンにポルフィリン、インドシアニングリーン及び葉酸が共有結合した複合体(蛍光プローブ)であり、動的光散乱法(Beckman Coulter製「DelsaMax PRO」)により測定したところ、平均粒子径が約65nmの蛍光ハイブリッドナノ粒子(以下において、単に「FA-PEG/ICG-PPS HNPs」とも記す。)であった。
(1)上記で得られたPPS HNPsの水分散液1mLに、FA-PEG-Malの水溶液を251μL(濃度:2mg/mL、FA-PEG-Mal:1.48×10-4mmol)と、ICG-Malの水溶液(濃度:2mg/mL、ICG-Mal:1.48×10-4mmol)を63μL添加し、30℃で3時間撹拌した。
(2)反応後、生成物を遠心分離(15000rpm、20分)で沈殿物として回収し、水で数回洗浄し、最終的に1mLの水に分散させた。
(3)得られた物質は、ポリシロキサンを担体分子とし、ポルフィリンを蛍光色素aとし、インドシアニングリーンを蛍光色素bとし、葉酸を細胞表面結合物質とするポリシロキサンにポルフィリン、インドシアニングリーン及び葉酸が共有結合した複合体(蛍光プローブ)であり、動的光散乱法(Beckman Coulter製「DelsaMax PRO」)により測定したところ、平均粒子径が約65nmの蛍光ハイブリッドナノ粒子(以下において、単に「FA-PEG/ICG-PPS HNPs」とも記す。)であった。
(フーリエ変換赤外分光法による確認)
実施例1及び実施例2で得られたPPS HNPsと、原料として用いたTCPPをフーリエ変換赤外分光光度計(Nicolet社製「NEXUS470」)で解析し、PPS HNPs及びTCPPのFT-IRスペクトルを図1に示した。図1のFT-IRスペクトルから、どちらとも3430~3250cm-1にピロール環に由来するピークを示すことが分かった。PPS HNPsにおいて、TCPPの1800~1640cm-1のカルボン酸(カルボキシル基)のピークが消失し、1740~1620cm-1のアミド由来のピークが現れたことから、TCPPのカルボキシル基がアミド結合を形成してシロキサンと共有結合した(ポルフィリン‐シランとMPTMSの加水分解・縮合により形成された)ポリシロキサンとポルフィリンの複合体が確実に合成されていることが確認された。また、PPS HNPsの1260~1010cm-1(νSi-O-Si)、870~740cm-1(νSi-O-Si)、540~410cm-1(σSi-O-Si)にシロキサン結合に由来するピークが現れていた。すなわち、PPS HNPsでは、ポルフィリン‐シラン及びMPTMS(官能基シラン)にてシロキサンネットワークが形成され、ポルフィリンはアミド結合によりシロキサンネットワーク中に組み込まれていることが確認された。
実施例1及び実施例2で得られたPPS HNPsと、原料として用いたTCPPをフーリエ変換赤外分光光度計(Nicolet社製「NEXUS470」)で解析し、PPS HNPs及びTCPPのFT-IRスペクトルを図1に示した。図1のFT-IRスペクトルから、どちらとも3430~3250cm-1にピロール環に由来するピークを示すことが分かった。PPS HNPsにおいて、TCPPの1800~1640cm-1のカルボン酸(カルボキシル基)のピークが消失し、1740~1620cm-1のアミド由来のピークが現れたことから、TCPPのカルボキシル基がアミド結合を形成してシロキサンと共有結合した(ポルフィリン‐シランとMPTMSの加水分解・縮合により形成された)ポリシロキサンとポルフィリンの複合体が確実に合成されていることが確認された。また、PPS HNPsの1260~1010cm-1(νSi-O-Si)、870~740cm-1(νSi-O-Si)、540~410cm-1(σSi-O-Si)にシロキサン結合に由来するピークが現れていた。すなわち、PPS HNPsでは、ポルフィリン‐シラン及びMPTMS(官能基シラン)にてシロキサンネットワークが形成され、ポルフィリンはアミド結合によりシロキサンネットワーク中に組み込まれていることが確認された。
実施例1及び実施例2で得られたPPS HNPsを29Si固体NMRで構造解析し、PPS HNPsの29Si固体NMRスペクトルを図2に示した。図2から、-58及び-69ppmにピークが現れることが確認された。-58ppmのピークは、下記の化学式で表されるT2構造を示し、-69ppmのピークは、下記の化学式で表されるT3構造を示していると推測される。この結果より、PPS HNPsには、一部T2構造のSiが存在するものの、ほとんどのMPTMSとポルフィリン‐シランが加水分解、重縮合を受けていることが確認された。ポリシロキサンにポルフィリンが共有結合して複合体を形成していることになる。
(熱重量・示差熱分析)
熱分析計(リガク製、TG8120)を用いてPPS HNPsに対して熱重量・示差熱分析(TG-DTA)を行った。図3にPPS HNPsのTG(熱重量)及びDTA(示唆熱)曲線を示した。解析の結果、100℃における重量減少は吸着水によるもの、300℃における重量減少はアミノプロピル部分及びメルカプトプロピル部分の燃焼によるもの、300~400℃における重量減少はポルフィリンの4つのピロールの環状構造部分(ポルフィン)の分解によるものであり、残りの部分がポリシロキサン部分と推測される。具体的には、PPS HNPs中には7重量%の吸着水、29重量%のアミノプロピル部分及びメルカプトプロピル部分、17重量%のポルフィリンの4つのピロールの環状構造部分(ポルフィン)、47重量%のポリシロキサン部分が含まれていると推測される。
熱分析計(リガク製、TG8120)を用いてPPS HNPsに対して熱重量・示差熱分析(TG-DTA)を行った。図3にPPS HNPsのTG(熱重量)及びDTA(示唆熱)曲線を示した。解析の結果、100℃における重量減少は吸着水によるもの、300℃における重量減少はアミノプロピル部分及びメルカプトプロピル部分の燃焼によるもの、300~400℃における重量減少はポルフィリンの4つのピロールの環状構造部分(ポルフィン)の分解によるものであり、残りの部分がポリシロキサン部分と推測される。具体的には、PPS HNPs中には7重量%の吸着水、29重量%のアミノプロピル部分及びメルカプトプロピル部分、17重量%のポルフィリンの4つのピロールの環状構造部分(ポルフィン)、47重量%のポリシロキサン部分が含まれていると推測される。
(PPS HNPsに結合したFA-PEG及びICGの定量)
PPS HNPsに結合したFA-PEG及びICGの量を求めるために、実施例2において、反応終了後の上澄み液(1mL)の吸収スペクトルを分光光度計(日本分光社製「V-570」)にて測定し、未反応のFA-PEG-Mal、ICG-Mal量を計算した。図4Aに、上澄み液の吸収スペクトルを示した。FA-PEG-Malは葉酸の水溶液中の吸光係数が不明であったため、葉酸のピーク波長377nmにおける検量線を作成し、そこから濃度を算出した。図4Bに葉酸の検量線を示した。図4Aにおいて377nmの吸光度は0.032であり、これより求められる上澄み中の葉酸の濃度は12nmol/mLである。使用したFA-PEG-Mal量は148nmolであるため、92%のFA-PEG-Malが反応したことが確認された。次にICG-Malについては、ICGの水溶液中の800nmにおけるモル吸光係数ε800=147000L/mol・cmを用いてランベルト・ベールの式A800=ε800cLから上澄み中の濃度を計算した結果、0.094nmol/mLであった。使用したICG-Mal量は148nmolであるため、99.9%のICG-Malが反応したと見積もることができる。以上の計算結果より92%のFA-PEG、ほぼ100%のICGがPPS HNPsに結合していると判断した。同様に、実施例1において、PPS HNPsに結合したICGの量を求めたところ、ほぼ100%のICGがPPS HNPsに結合していることが確認された。
PPS HNPsに結合したFA-PEG及びICGの量を求めるために、実施例2において、反応終了後の上澄み液(1mL)の吸収スペクトルを分光光度計(日本分光社製「V-570」)にて測定し、未反応のFA-PEG-Mal、ICG-Mal量を計算した。図4Aに、上澄み液の吸収スペクトルを示した。FA-PEG-Malは葉酸の水溶液中の吸光係数が不明であったため、葉酸のピーク波長377nmにおける検量線を作成し、そこから濃度を算出した。図4Bに葉酸の検量線を示した。図4Aにおいて377nmの吸光度は0.032であり、これより求められる上澄み中の葉酸の濃度は12nmol/mLである。使用したFA-PEG-Mal量は148nmolであるため、92%のFA-PEG-Malが反応したことが確認された。次にICG-Malについては、ICGの水溶液中の800nmにおけるモル吸光係数ε800=147000L/mol・cmを用いてランベルト・ベールの式A800=ε800cLから上澄み中の濃度を計算した結果、0.094nmol/mLであった。使用したICG-Mal量は148nmolであるため、99.9%のICG-Malが反応したと見積もることができる。以上の計算結果より92%のFA-PEG、ほぼ100%のICGがPPS HNPsに結合していると判断した。同様に、実施例1において、PPS HNPsに結合したICGの量を求めたところ、ほぼ100%のICGがPPS HNPsに結合していることが確認された。
(分光法による吸収スペクトルの確認)
実施例1及び実施例2で得られたPPS HNPsの水溶液及び実施例2で得られたFA-PEG/ICG-PPS HNPsの水溶液の吸収スペクトルを分光光度計(日本分光社製「V-570」)にて測定し、その結果を下記図5に示した。図5のPPS HNPs及びFA-PEG/ICG-PPS HNPsの吸収スペクトルから、PPS HNPsにICGが結合することにより、波長700~900nmあたりにICGに由来するピークが現れることが確認された。また、ICG-PPS HNPs及びFA-PEG/ICG-PPS HNPsにおいて、ポルフィリンに由来する380~650nmあたりのピークにはほとんど変化がないことが確認された。
実施例1及び実施例2で得られたPPS HNPsの水溶液及び実施例2で得られたFA-PEG/ICG-PPS HNPsの水溶液の吸収スペクトルを分光光度計(日本分光社製「V-570」)にて測定し、その結果を下記図5に示した。図5のPPS HNPs及びFA-PEG/ICG-PPS HNPsの吸収スペクトルから、PPS HNPsにICGが結合することにより、波長700~900nmあたりにICGに由来するピークが現れることが確認された。また、ICG-PPS HNPs及びFA-PEG/ICG-PPS HNPsにおいて、ポルフィリンに由来する380~650nmあたりのピークにはほとんど変化がないことが確認された。
(蛍光プローブの波長特性確認)
実施例2で得られたFA-PEG/ICG-PPS HNPsの水溶液を用い、FA-PEG/ICG-PPS HNPsの励起波長スペクトル及び蛍光波長スペクトルを、日本分光社製の分光蛍光光度計FP-6600にて解析した。その結果を図6に示した。図6Aはインビトロ蛍光イメージングに用いる短波長側の波長特性を示し、図6Bはインビボ蛍光イメージングに用いる長波長側の波長特性を示している。図6A及び図6Bのから、最大励起波長425nm、最大蛍光波長655nmの短波長側と、最大励起波長650nm、最大蛍光波長905nmの長波長側のどちらにおいてもストークスシフトが大きいことが分かった。ICG-PPS HNPs及びFA-PEG/ICG-PPS HNPsは、いずれも、担体分子(ポリシロキサン)に結合した異なる励起波長の2つの蛍光色素(ポルフィリンとICG)を有し、且つ2つの蛍光色素の間で蛍光共鳴エネルギー移動が生じない蛍光プローブであることが確認された。該蛍光プローブは、インビトロ及びインビボのどちらのイメージングにも用いることができる。さらに、ストークスシフトが大きいため、インビボイメージングにおいては生体の自家蛍光に妨げられることなく、非常に高い有用性が見込める。
実施例2で得られたFA-PEG/ICG-PPS HNPsの水溶液を用い、FA-PEG/ICG-PPS HNPsの励起波長スペクトル及び蛍光波長スペクトルを、日本分光社製の分光蛍光光度計FP-6600にて解析した。その結果を図6に示した。図6Aはインビトロ蛍光イメージングに用いる短波長側の波長特性を示し、図6Bはインビボ蛍光イメージングに用いる長波長側の波長特性を示している。図6A及び図6Bのから、最大励起波長425nm、最大蛍光波長655nmの短波長側と、最大励起波長650nm、最大蛍光波長905nmの長波長側のどちらにおいてもストークスシフトが大きいことが分かった。ICG-PPS HNPs及びFA-PEG/ICG-PPS HNPsは、いずれも、担体分子(ポリシロキサン)に結合した異なる励起波長の2つの蛍光色素(ポルフィリンとICG)を有し、且つ2つの蛍光色素の間で蛍光共鳴エネルギー移動が生じない蛍光プローブであることが確認された。該蛍光プローブは、インビトロ及びインビボのどちらのイメージングにも用いることができる。さらに、ストークスシフトが大きいため、インビボイメージングにおいては生体の自家蛍光に妨げられることなく、非常に高い有用性が見込める。
(蛍光顕微鏡による解析)
(1)細胞
マウスと同種の細胞であるマウスのマクロファージ由来のRAW264.7細胞、マウスと異種の細胞であるヒトの子宮頸癌由来のHeLa S3細胞とヒトの大腸がん由来のHCT116細胞を用いた。実施例において、細胞及び細胞培養用培地は、シグマーアルドリッチ社から入手した。細胞培養ディッシュは、サーモフィッシャーサイエンティフィック株式会社製のものを用いた。
(2)細胞の蛍光プローブ標識
(a)細胞を細胞培養ディッシュ(直径35mm)中の細胞培養用培地(10%FBS入りのDMEM培地)に播種(0.3x105cells/mL)し、一晩培養した。
(b)培養した細胞を蛍光プローブ(30μg/mL)入りの細胞培養用培地(10%FBS入りのDMEM培地)に交換し、24時間培養を行った。このとき、RAW264.7細胞は、蛍光プローブICG-PPS HNPs入りの細胞培養用培地に交換し、HeLa S3細胞とHCT116細胞は、蛍光プローブFA-PEG/ICG-PPS HNPs入りの細胞培養用培地に交換した。
(c)上清を捨て、培養細胞を2mLのリン酸緩衝生理食塩水(1×PBS、pH7.4、和光純薬「D-PBS(-)(045-29795)」)で洗浄した。
(d)上清を捨て、培養細胞に4%パラホルムアルデヒド(PFA)を2mL添加し、30分間室温(20±5℃)で静置した。
(e)上清を捨て、培養細胞に1×PBSを1mL加えた。この工程を3回繰り返した。
(3)蛍光顕微鏡による観察
ライフテクノロジーズ社の蛍光顕微鏡EVOS(登録商標)FL Cell Imaging Systemを用いて蛍光プローブで標識した蛍光標識細胞を観察した。ポルフィリンの波長特性に合わせ、励起波長は422nm、蛍光波長は655nmで観察を行った。その結果を図7に示した。
(1)細胞

図7Aには、RAW264.7細胞の明視野像(Bright field)、蛍光色素DAPI(4,6-ジアミジノ-2-フェニルインドール)を使用した細胞核の蛍光イメージ(DAPI)、ICG-PPS HNPs(蛍光プローブ)による蛍光イメージング、これら三つの画像を重ねあわせた像(Merge)が示されている。図7Aの明視野像から、細胞が存在していることが分かった。またDAPIの画像から、DAPIによって染色された細胞核が確認された。ICG-PPS HNPsの画像から、蛍光プローブによって細胞が標識されていることが分かった。Mergeの画像から、RAW264.7細胞において、蛍光プローブによる蛍光は、細胞質にて多く観察され、核は蛍光していないことがわかった。この結果からICG-PPS HNPs(蛍光プローブ)が細胞内に取り込まれて、細胞質で留まっていることが確認された。RAW264.7細胞は、マクロファージ細胞であり、貪食能により蛍光プローブを細胞内に取込み、細胞質に蛍光プローブが集まったと考えられる。
図7Bには、HeLa S3細胞の明視野像(Bright field)、蛍光色素DAPIを使用した細胞核の蛍光イメージ(DAPI)、FA-PEG/ICG-PPS HNPs蛍光プローブによる蛍光イメージング、これら三つの画像を重ねあわせた像(Merge)が示されている。図7Bの明視野像から、細胞が存在していることが分かった。またDAPIの画像から、DAPIによって染色された細胞核が確認された。FA-PEG/ICG-PPS HNPs蛍光プローブの画像から、蛍光プローブによって細胞が標識されていることが分かった。Mergeの画像から、HeLa S3細胞の蛍光プローブの蛍光は、細胞核も含めて細胞全体が蛍光していることがわかった。この結果から蛍光プローブが細胞表面に結合しており、細胞全体が光って観測されていることが明らかになった。がん細胞は細胞表面に葉酸を結合させる受容体を持っており、蛍光プローブ(FA-PEG/ICG-PPS HNPs)が葉酸を介してがん細胞であるHeLa S3細胞の表面に結合したと考えられる。
図7Cには、HCT116細胞の明視野像(Bright field)、蛍光色素DAPIを使用した細胞核の蛍光イメージ(DAPI)、FA-PEG/ICG-PPS HNPs蛍光プローブによる蛍光イメージング、これら三つの画像を重ねあわせた像(Merge)が示されている。図7Cの明視野像から、細胞が存在していることが分かった。またDAPIの画像から、DAPIによって染色された細胞核が確認された。FA-PEG/ICG-PPS HNPs蛍光プローブの画像から、蛍光プローブによって細胞が標識されていることが分かった。Mergeの画像から、HCT116細胞の蛍光プローブの蛍光は、細胞核も含めて細胞全体が蛍光していることがわかった。この結果から蛍光プローブが細胞表面に結合しており、細胞全体が光って観測されていることが明らかになった。がん細胞は細胞表面に葉酸を結合させる受容体を持っており、蛍光プローブ(FA-PEG/ICG-PPS HNPs)が葉酸を介してがん細胞であるHCT116細胞の表面に結合したと考えられる。
(フローサイトメーターによる解析)
(1)細胞
マウスと同種の細胞であるマウスのマクロファージ由来のRAW264.7細胞及びマウスと異種の細胞であるヒトの子宮頸癌由来のHeLa S3細胞とヒトの大腸がん由来のHCT116細胞を用いた。
(2)細胞の蛍光プローブ標識
(a)細胞を細胞培養ディッシュ(直径35mm)中の細胞培養培地(10%FBS入りのDMEM培地)に下記表1に示す播種量で播種し、一晩培養した。
(b)培養した細胞を下記表1に示す蛍光プローブ(50μg/mL)入りの細胞培養培地(10%FBS入りのDMEM培地)に交換し、さらに48時間培養を行った。
(c)細胞培養ディッシュから細胞を回収した。このとき、HeLa S3細胞及びHCT116細胞の回収には0.25%のトリプシン/EDTA溶液を使用した。
(d)回収液を1000回転にて5分間遠心分離した後、上清を捨て、細胞を400μLの細胞培養培地(10%FBS入りのDMEM培地)に懸濁した。
(e)懸濁液を1000回転、5分間遠心分離した後、上清を捨て、細胞に4%PFAを1mL添加し、30分間室温(20±5℃)で静置した。
(f)再懸濁液を1000回転、5分間遠心分離した後、上清を捨て、細胞に1xPBSを1mL加えた。この工程を3回繰り返した。
(1)細胞
マウスと同種の細胞であるマウスのマクロファージ由来のRAW264.7細胞及びマウスと異種の細胞であるヒトの子宮頸癌由来のHeLa S3細胞とヒトの大腸がん由来のHCT116細胞を用いた。

(b)培養した細胞を下記表1に示す蛍光プローブ(50μg/mL)入りの細胞培養培地(10%FBS入りのDMEM培地)に交換し、さらに48時間培養を行った。
(c)細胞培養ディッシュから細胞を回収した。このとき、HeLa S3細胞及びHCT116細胞の回収には0.25%のトリプシン/EDTA溶液を使用した。
(d)回収液を1000回転にて5分間遠心分離した後、上清を捨て、細胞を400μLの細胞培養培地(10%FBS入りのDMEM培地)に懸濁した。

(f)再懸濁液を1000回転、5分間遠心分離した後、上清を捨て、細胞に1xPBSを1mL加えた。この工程を3回繰り返した。
(3)フローサイトメーターによる観察
BD社のフローサイトメーターLSRFortessaX-20を用いて蛍光プルーブで標識を行った細胞を観察した。励起波長は405nm、蛍光波長は670nmで観察を行った。その結果を図8(RAW264.7細胞)、図9(HeLa S3細胞)及び図10(HCT116細胞)に示した。
BD社のフローサイトメーターLSRFortessaX-20を用いて蛍光プルーブで標識を行った細胞を観察した。励起波長は405nm、蛍光波長は670nmで観察を行った。その結果を図8(RAW264.7細胞)、図9(HeLa S3細胞)及び図10(HCT116細胞)に示した。
図8、図9及び図10から分かるように、全ての細胞サンプルで蛍光標識されている細胞と蛍光標識されていない細胞とを分離して観察することができた。光強度としては、100~10000倍ほどの差があった。細胞に対して蛍光プローブによる標識を行った後、蛍光標識された細胞と蛍光標識されていない細胞を明確に判別し、分離することができた。フローサイトメーターを用いることで、蛍光プローブで標識された蛍光標識細胞のみを選別し、マウス等の生体内へ移植することができ、蛍光インビボイメージング装置により、マウス等の生体内での蛍光標識細胞の局在をより正確に確認することができる。
(マウスへの蛍光標識細胞の移植)
(1)細胞
RAW264.7細胞及びHeLa S3細胞を用いた。
(2)細胞の蛍光プローブによる標識
(a)細胞を細胞培養ディッシュ(直径60mm)中の細胞培養用培地(10%FBS入りのDMEM培地)に播種(1×106cells/mL)し、一晩培養した。
(b)培養した細胞を蛍光プローブ(30μg/mL)入りの細胞培養用培地(10%FBS入りのDMEM培地)に交換し、24時間培養を行った。このとき、RAW264.7細胞は、蛍光プローブICG-PPS HNPs入りの細胞培養用培地に交換し、HeLa S3細胞は、蛍光プローブFA-PEG/ICG-PPS HNPs入りの細胞培養用培地に交換した。
(c)細胞培養ディッシュから細胞を回収した。このとき、HeLa S3細胞の回収にはトリプシン/EDTA溶液を使用した。
(d)回収液を1000回転にて5分間遠心分離した後、上清を捨て、細胞を1000μLの無血清培地(DMEM培地)に懸濁した。
(e)懸濁液を1000回転にて5分間遠心分離した後、上清を捨て、1000μLの血清培地(10%FBS入りのDMEM培地)を加え、細胞を懸濁した。
(f)1x106細胞を100μLの1xPBSに懸濁し、シリンジを用いてマウス(KSN/Slc、雄、6週零、日本エスエルシー株式会社)の尾静脈に注射した。細胞数については、血球計算盤を用いて測定した。
(1)細胞
RAW264.7細胞及びHeLa S3細胞を用いた。

(a)細胞を細胞培養ディッシュ(直径60mm)中の細胞培養用培地(10%FBS入りのDMEM培地)に播種(1×106cells/mL)し、一晩培養した。
(b)培養した細胞を蛍光プローブ(30μg/mL)入りの細胞培養用培地(10%FBS入りのDMEM培地)に交換し、24時間培養を行った。このとき、RAW264.7細胞は、蛍光プローブICG-PPS HNPs入りの細胞培養用培地に交換し、HeLa S3細胞は、蛍光プローブFA-PEG/ICG-PPS HNPs入りの細胞培養用培地に交換した。
(c)細胞培養ディッシュから細胞を回収した。このとき、HeLa S3細胞の回収にはトリプシン/EDTA溶液を使用した。
(d)回収液を1000回転にて5分間遠心分離した後、上清を捨て、細胞を1000μLの無血清培地(DMEM培地)に懸濁した。
(e)懸濁液を1000回転にて5分間遠心分離した後、上清を捨て、1000μLの血清培地(10%FBS入りのDMEM培地)を加え、細胞を懸濁した。
(f)1x106細胞を100μLの1xPBSに懸濁し、シリンジを用いてマウス(KSN/Slc、雄、6週零、日本エスエルシー株式会社)の尾静脈に注射した。細胞数については、血球計算盤を用いて測定した。
(3)マウス体内の蛍光プローブで標識されて蛍光標識細胞の観察
倉敷紡績社の蛍光インビボイメージング装置「KURAVIC KV-700」を用いてマウス体内の蛍光標識細胞を観察した。観察は注射直前を0時間として、24時間まで間隔をあけて撮影を行った。ICGの波長特性に合わせ、励起波長は747nm、蛍光波長は850nmで観察を行った。その結果を図11(RAW264.7細胞)及び図12(HeLa S3細胞)に示した。
倉敷紡績社の蛍光インビボイメージング装置「KURAVIC KV-700」を用いてマウス体内の蛍光標識細胞を観察した。観察は注射直前を0時間として、24時間まで間隔をあけて撮影を行った。ICGの波長特性に合わせ、励起波長は747nm、蛍光波長は850nmで観察を行った。その結果を図11(RAW264.7細胞)及び図12(HeLa S3細胞)に示した。
図11から、蛍光プローブ(ICG-PPS HNPs)で標識されたRAW264.7細胞のマウス体内における24時間までの動きが確認できた。すなわち、RAW264.7細胞がマウス体内でどのような動きをしたか確認できた。特に、インビボ観察に適した蛍光波長の選択によって、自家蛍光を捉えることなく、明確にICGの蛍光のみを捉えることができた。24時間後の状態としては、RAW264.7細胞が脾臓や肝臓が多いものの、内臓全体に分布していることが分かった。
図12から、蛍光プローブ(FA-PEG/ICG-PPS HNPs)で標識されたHeLa S3細胞のマウス体内における24時間までの動きが確認できた。すなわち、HeLa S3細胞がマウス体内でどのような動きをしたか確認できた。特に、インビボ観察に適した蛍光波長の選択によって、自家蛍光を捉えることなく、明確にICGの蛍光のみを捉えることができた。24時間後の状態としては、細胞が肝臓に特に集積していることが分かった。
(4)マウス体内の蛍光プローブで標識された細胞の集積の確認
24時間経過後のマウスを解剖し、臓器を取り出し、蛍光インビボイメージング装置「KURAVIC KV-700」により、24時間後の蛍光プローブで標識された細胞のマウス体内での集積の状態を観察した。その結果を図13(RAW264.7細胞)、図14(RAW264.7細胞)、図15(HeLa S3細胞)及び図16(HeLa S3細胞)を示した。
24時間経過後のマウスを解剖し、臓器を取り出し、蛍光インビボイメージング装置「KURAVIC KV-700」により、24時間後の蛍光プローブで標識された細胞のマウス体内での集積の状態を観察した。その結果を図13(RAW264.7細胞)、図14(RAW264.7細胞)、図15(HeLa S3細胞)及び図16(HeLa S3細胞)を示した。
図13及び図14から分かるように、24時間後に、蛍光プローブ(ICG-PPS HNPs)で標識されたRAW264.7細胞は脾臓や肝臓に多く集積されているものの、内臓全体に分布していた。また、図15及び図16から分かるように、24時間後に、蛍光プローブ(FA-PEG/ICG-PPS HNPs)で標識されたHeLa細胞は、肝臓に特に多く集積した。
以上から分かるように、蛍光プローブとしてICG-PPS HNPs及びFA-PEG/ICG-PPS HNPs等の担体分子に結合した異なる励起波長の2つの蛍光色素を有し、且つ2つの蛍光色素の間で蛍光色素aと蛍光色素bの間に蛍光共鳴エネルギー移動が生じない蛍光プローブを用いることにより、蛍光プローブで標識された細胞を蛍光顕微鏡やフローサイトメーター等の蛍光インビトロイメージング装置で確認できるとともに、蛍光プローブで標識された細胞を移植した生体内における蛍光標識細胞の時間に伴う局在等を蛍光インビボイメージング装置で確認できる。
Claims (21)
- 担体分子と、前記担体分子に結合している蛍光色素a及び蛍光色素bを含む蛍光プローブにおいて、蛍光色素aの励起波長と蛍光色素bの励起波長は異なり、蛍光色素aと蛍光色素bの間に蛍光共鳴エネルギー移動が生じないことを特徴とする蛍光プローブ。
- 前記蛍光プローブは、細胞標識用蛍光プローブであり、蛍光色素aは、インビトロ蛍光イメージング用蛍光色素であり、蛍光色素bは、インビボ蛍光イメージング用蛍光色素である請求項1に記載の蛍光プローブ。
- 細胞表面に特異的に結合することで細胞を標識する請求項1又は2に記載の蛍光プローブ。
- 細胞内部に取り込まれることで細胞を標識する請求項1又は2に記載の蛍光プローブ。
- 蛍光色素aは、励起波長が350~650nmの範囲である請求項1~4のいずれか1項に記載の蛍光プローブ。
- 蛍光色素bは、励起波長が600~850nmの範囲である請求項1~5のいずれか1項に記載の蛍光プローブ。
- 蛍光色素aがポルフィリンである請求項1~6のいずれか1項に記載の蛍光プローブ。
- 蛍光色素bがインドシアニングリーンである請求項1~7のいずれか1項に記載の蛍光プローブ。
- 前記担体分子は、ポリシロキサンである請求項1~8のいずれか1項に記載の蛍光プローブ。
- フローサイトメーターによる蛍光標識細胞の選別に用いる請求項2~9のいずれか1項に記載の蛍光プローブ。
- 標的細胞を検出する蛍光検出方法であって、
標的細胞を蛍光プローブで標識する工程と、
蛍光プローブで標識された標的細胞に励起光を照射して、蛍光プローブからの蛍光を観察する工程を含み、
前記蛍光プローブは、担体分子と、前記担体分子に結合している蛍光色素a及び蛍光色素bを含み、蛍光色素aの励起波長と蛍光色素bの励起波長は異なり、且つ蛍光色素aと蛍光色素bの間に蛍光共鳴エネルギー移動が生じないことを特徴とする蛍光検出方法。 - 蛍光色素aは、インビトロ蛍光イメージング用蛍光色素であり、蛍光色素bは、インビボ蛍光イメージング用蛍光色素である請求項11に記載の蛍光検出方法。
- 蛍光プローブで標識された標的細胞に蛍光色素aを励起する励起光を照射し、インビトロ蛍光イメージングによって蛍光プローブからの蛍光を観察する請求項11又は12に記載の蛍光検出方法。
- 蛍光プローブで標識された標的細胞を生体(ただし、人体を除く)内に移植し、該生体に蛍光色素bを励起する励起光を照射し、インビボ蛍光イメージングによって蛍光プローブからの蛍光を観察する請求項11又は12に記載の蛍光検出方法。
- 前記蛍光プローブは、細胞表面に特異的に結合することで細胞を標識する請求項11~14のいずれか1項に記載の蛍光検出方法。
- 前記蛍光プローブは、細胞内部に取り込まれることで細胞を標識する請求項11~14のいずれか1項に記載の蛍光検出方法。
- 蛍光色素aは、励起波長が350~650nmの範囲である請求項11~16のいずれか1項に記載の蛍光検出方法。
- 蛍光色素bは、励起波長が600~850nmの範囲である請求項11~17のいずれか1項に記載の蛍光検出方法。

選別した蛍光標識細胞を生体(ただし、人体を除く)内に移植する工程と、
生体内の蛍光標識細胞をインビボ蛍光イメージング装置で観察する工程を含み、
前記フローサイトメーター又は蛍光顕微鏡による蛍光標識細胞の選別は、蛍光色素a由来の蛍光を検出することで行い、前記生体内の蛍光標識細胞のインビボ蛍光イメージング装置による観察は、蛍光色素b由来の蛍光を検出することで行うことを特徴とする蛍光プローブの使用方法。
