持久力を必要とする運動選手のトレーニングにおいて、栄養素の摂取により行動体力を向上させる方法として、グリコーゲンローディングが知られている。筋肉中のグリコーゲン量の増加は運動能力を向上させることが知られており、グリコーゲンローディングとは、あらかじめ低糖質食を摂取して激しい運動を行い、筋肉中のグリコーゲン量を大幅に減少させた後、高糖質食に切り替えることにより、筋肉中のグリコーゲンをより効果的に増加させるというものである。この方法は、栄養素摂取による行動体力向上方法であり、望ましいと考えられるが、長期間の実施が必要であり、栄養価の計算された食事の準備や、グリコーゲンを枯渇させる運動プログラムの設定など、複雑なプロセスが必要となる。
栄養素摂取による運動能力の向上の例として、特許文献1、2にアミノ酸混合物による持久力の向上が報告されている。アミノ酸は食物に普通に含まれている栄養素であり、アミノ酸混合物の摂取は手段としては望ましいが、前記特許文献では、複数のアミノ酸からなる組成物による効果は示されているが、シスチンの効果については何ら示唆されていない。
非特許文献1、2には、長距離ランナーにシスチンおよびテアニンを摂取させることによって、強度な運動負荷後の免疫機能低下を抑制し、また骨格筋崩壊による筋障害を緩和したことが報告されている。
しかし、非特許文献1、2には、シスチンおよびテアニンの運動能力や行動体力に対する効果に関しての言及はない。

本発明は、上記のような事情に鑑みてなされたものであり、その解決しようとする課題は、身体機能に関わる行動体力の向上促進に関して、従来技術に比べて優れた改善効果を奏し得る、栄養組成物を提供することにある。
本発明者らは、上記課題を解決すべく鋭意検討した結果、シスチンが運動(トレーニング)による筋肉中のグリコーゲン量の増加効果や、行動体力向上効果を一層高めることを見出し、かかる知見に基づいてさらに研究を進めることにより、本発明を完成するに至った。
即ち、本発明は以下のとおりである。
[1]シスチンを有効成分として含有する、行動体力向上剤。
[2]行動を持続する能力の向上剤である、[1]に記載の向上剤。
[3]シスチンの含有量が、1回あたりの摂取量または投与量として2.5mg~15gである、[1]または[2]に記載の向上剤。

本発明の行動体力向上剤は、行動体力、特に筋持久力、全身持久力といった行動を持続する能力を向上させる。また、本発明の行動体力向上剤は、筋肉中のグリコーゲン量を増加させる。
従って、本発明の行動体力向上剤は、筋肉中グリコーゲン量の増加や、行動を持続する能力(以下、本明細書中において「持久力」ともいう)の向上のために効果的に用いられ、特に、運動(トレーニング)による筋肉中グリコーゲン量増加効果や、筋持久力、全身持久力の向上効果をより高めることができる。
また、本発明の行動体力向上剤は、シスチン等の食経験が豊富なアミノ酸を有効成分とすることから、安全性が高く副作用がほとんどないため、極めて有利である。
従って、本発明の行動体力向上剤は、筋肉中グリコーゲン量の増加や、行動を持続する能力(以下、本明細書中において「持久力」ともいう)の向上のために効果的に用いられ、特に、運動(トレーニング)による筋肉中グリコーゲン量増加効果や、筋持久力、全身持久力の向上効果をより高めることができる。
また、本発明の行動体力向上剤は、シスチン等の食経験が豊富なアミノ酸を有効成分とすることから、安全性が高く副作用がほとんどないため、極めて有利である。
本発明の行動体力向上剤(以下、本明細書において、「本発明の剤」ともいう)は、シスチンを有効成分として含有する。
本明細書において、「行動体力」とは、「体力」を大きく二つに分けた場合に、行動の基礎となる身体的な能力をいう。
「体力」は、行動体力、すなわち、実際に身体を動かし行動する身体的な能力と、病気やストレスに対する免疫力や抵抗力、環境に適応する能力を指す防衛体力に分けられる。そして、「行動体力」は、筋力、瞬発力等の行動を起こす能力、筋持久力や全身持久力等の行動を持続する能力、敏捷性、平衡性、巧緻性、柔軟性といった行動を調節する能力に分けられる。
本発明の行動体力向上剤は、主として行動を持続する能力、すなわち筋持久力や全身持久力を向上させるものである。
ここで、「筋持久力」とは、ある筋肉が繰り返し収縮し続ける能力、すなわち、体の筋肉の一部を使った運動を長く続ける能力をいい、「全身持久力」とは、体全体の筋肉を使った運動を長く続けるために必要な能力をいう。
また、本発明の行動体力向上剤は、筋肉中のグリコーゲン量を増加させるものである。従って、本発明の行動体力向上剤は、グリコーゲンローディングを促進して、運動能力の限界を上昇させることができる。
さらに、本発明の行動体力向上剤は、運動(トレーニング)による筋肉中のグリコーゲン量増加効果や、筋持久力の向上、全身持久力の向上といった効果をより高めるものである。
「体力」は、行動体力、すなわち、実際に身体を動かし行動する身体的な能力と、病気やストレスに対する免疫力や抵抗力、環境に適応する能力を指す防衛体力に分けられる。そして、「行動体力」は、筋力、瞬発力等の行動を起こす能力、筋持久力や全身持久力等の行動を持続する能力、敏捷性、平衡性、巧緻性、柔軟性といった行動を調節する能力に分けられる。

さらに、本発明の行動体力向上剤は、運動(トレーニング)による筋肉中のグリコーゲン量増加効果や、筋持久力の向上、全身持久力の向上といった効果をより高めるものである。
本発明の剤に有効成分として含まれるシスチン、すなわち3,3’-ジチオビス(2-アミノプロパン酸)は、L-体、D-体、DL-体のいずれも使用可能であるが、好ましくは、L-体およびDL-体であり、さらに好ましくは、L-体である。
なお、構成アミノ酸としてシスチンを含有するタンパク質やペプチドも、シスチン供給源として使用することができる。消化を要しないため、遊離のシスチンを用いることが好ましい。
また、シスチンは、遊離体のみならず、塩の形態でも使用することができる。本明細書における「シスチン」という用語は、塩をも包含する概念である。塩の形態としては、酸付加塩や塩基との塩等を挙げることができ、薬理学上許容される塩を選択することが好ましい。
なお、構成アミノ酸としてシスチンを含有するタンパク質やペプチドも、シスチン供給源として使用することができる。消化を要しないため、遊離のシスチンを用いることが好ましい。
また、シスチンは、遊離体のみならず、塩の形態でも使用することができる。本明細書における「シスチン」という用語は、塩をも包含する概念である。塩の形態としては、酸付加塩や塩基との塩等を挙げることができ、薬理学上許容される塩を選択することが好ましい。
具体的なシスチンの塩としては、無機塩基、有機塩基、無機酸、有機酸との塩およびアミノ酸との塩等が挙げられる。
無機塩基との塩としては、例えば、リチウム、ナトリウム、カリウム等のアルカリ金属との塩、マグネシウム、カルシウム等のアルカリ土類金属との塩、アンモニウム塩等が挙げられる。
有機塩基との塩としては、例えばモノエタノールアミン、ジエタノールアミン、トリエタノールアミン等のアルカノールアミンとの塩、モルホリン、ピペリジン等の複素環式アミンとの塩等が挙げられる。
無機酸との塩としては、例えば、ハロゲン化水素酸(塩酸、臭化水素酸、ヨウ化水素酸等)、硫酸、硝酸、リン酸等との塩が挙げられる。
有機酸との塩としては、例えば、ギ酸、酢酸、プロパン酸等のモノカルボン酸との塩;シュウ酸、マロン酸、リンゴ酸、コハク酸等の飽和ジカルボン酸との塩;マレイン酸、フマル酸等の不飽和ジカルボン酸との塩;クエン酸等のトリカルボン酸との塩;α-ケトグルタル酸等のケト酸との塩が挙げられる。
アミノ酸との塩としては、グリシン、アラニン等の脂肪族アミノ酸との塩;フェニルアラニン等の芳香族アミノ酸との塩;リシン等の塩基性アミノ酸との塩;アスパラギン酸、グルタミン酸等の酸性アミノ酸との塩;ピログルタミン酸等のラクタムを形成したアミノ酸との塩等が挙げられる。
無機塩基との塩としては、例えば、リチウム、ナトリウム、カリウム等のアルカリ金属との塩、マグネシウム、カルシウム等のアルカリ土類金属との塩、アンモニウム塩等が挙げられる。
有機塩基との塩としては、例えばモノエタノールアミン、ジエタノールアミン、トリエタノールアミン等のアルカノールアミンとの塩、モルホリン、ピペリジン等の複素環式アミンとの塩等が挙げられる。

本発明においては、上記した遊離体および塩の形態のシスチンは、1種を単独で用いてもよく、2種以上を組み合わせて用いてもよい。
本発明の目的には、遊離体のシスチンおよび塩酸塩等が好ましく、遊離体のシスチンがより好ましく用いられる。

本発明において、遊離体および塩の形態のシスチンは、天然に存在する動植物等から抽出し精製したもの、或いは、化学合成法、発酵法、酵素法または遺伝子組換え法等によって得られるもののいずれを使用してもよいが、各社より提供されている市販の製品を利用してもよい。
本発明の剤におけるシスチンの含有量は、行動体力の向上効果の観点から、ヒトの1回あたりの摂取量または投与量として、好ましくは2.5mg以上であり、より好ましくは6mg以上であり、更に好ましくは10mg以上であり、特に好ましくは70mg以上である。また、摂取または投与のしやすさの観点から、本発明の剤におけるシスチンの含有量は、ヒトの1回あたりの摂取量または投与量として、好ましくは15g以下であり、より好ましくは12g以下であり、更に好ましくは10g以下である。
なお、本明細書において、シスチンをはじめ、本発明の剤におけるアミノ酸の含有量は、アミノ酸が塩の形態で含有される場合、遊離体に換算した含有量で表す。

さらに、行動体力向上効果の観点から、本発明の剤におけるシスチンの含有量は、本発明の剤における全アミノ酸含有量に対して、好ましくは1重量%以上であり、より好ましくは3重量%以上であり、特に好ましくは10重量%以上である。また、本発明の剤におけるシスチンの含有量は、本発明の剤における全アミノ酸含有量に対して、好ましくは90重量%以下であり、より好ましくは80重量%以下である。
また、本発明の剤に含有される全アミノ酸の中で、シスチンの含有量が最も多いか、または2番目に多いことが好ましい。本発明の剤において、他のアミノ酸に比べてシスチンが多く含有されることにより、有効成分として効率的に行動体力向上機能を発揮することが可能となる。
本発明の剤は、シスチンに加え、更に、他のアミノ酸として、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸を含有してもよい。シスチンに加え、更に、前記の少なくとも一つのアミノ酸を含有することにより、本発明の剤の行動体力向上効果が増強される。中でも、本発明の剤は、シスチンに加えて、他のアミノ酸として、ロイシン、イソロイシン、バリン、グルタミン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸を含有することが好ましい。
特に、本発明の一態様として、シスチンに加えて、他のアミノ酸として、ロイシン、イソロイシン及びバリンからなる群より選ばれる少なくとも一つのアミノ酸を含有することがより好ましく、ロイシン、イソロイシン及びバリンを含有することがさらに好ましい。
また、別の一態様として、シスチンに加えて、他のアミノ酸として、グルタミンを含有することがより好ましい。
さらに、他の一態様として、シスチンに加えて、他のアミノ酸として、アラニンおよびプロリンからなる群より選ばれる少なくとも一つを含有することがより好ましい。
特に、本発明の一態様として、シスチンに加えて、他のアミノ酸として、ロイシン、イソロイシン及びバリンからなる群より選ばれる少なくとも一つのアミノ酸を含有することがより好ましく、ロイシン、イソロイシン及びバリンを含有することがさらに好ましい。
また、別の一態様として、シスチンに加えて、他のアミノ酸として、グルタミンを含有することがより好ましい。
さらに、他の一態様として、シスチンに加えて、他のアミノ酸として、アラニンおよびプロリンからなる群より選ばれる少なくとも一つを含有することがより好ましい。
本発明において、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンとしては、それぞれ、L-体、D-体、DL-体のいずれも使用可能であるが、好ましくは、L-体およびDL-体であり、さらに好ましくは、L-体である。
また、これらのアミノ酸は遊離体のみならず、塩の形態でも使用することができる。本明細書における「グルタミン」、「セリン」、「ヒスチジン」、「アルギニン」、「バリン」、「ロイシン」、「イソロイシン」、「アラニン」および「プロリン」という用語は、それぞれ塩をも包含する概念である。
塩の形態としては、酸付加塩や塩基との塩等を挙げることができ、薬理学上許容される塩を選択することが好ましい。
また、これらのアミノ酸は遊離体のみならず、塩の形態でも使用することができる。本明細書における「グルタミン」、「セリン」、「ヒスチジン」、「アルギニン」、「バリン」、「ロイシン」、「イソロイシン」、「アラニン」および「プロリン」という用語は、それぞれ塩をも包含する概念である。
塩の形態としては、酸付加塩や塩基との塩等を挙げることができ、薬理学上許容される塩を選択することが好ましい。
グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンの塩としては、シスチンについて記載したのと同様の塩が挙げられる。
本発明の目的には、遊離体の上記アミノ酸、ならびにそれらの塩酸塩、酢酸塩、リンゴ酸塩、α-ケトグルタル酸塩、アスパラギン酸塩、グルタミン酸塩、ピログルタミン酸塩が好ましく、遊離体の上記アミノ酸およびそれらの塩酸塩がより好ましい。
本発明の目的には、遊離体の上記アミノ酸、ならびにそれらの塩酸塩、酢酸塩、リンゴ酸塩、α-ケトグルタル酸塩、アスパラギン酸塩、グルタミン酸塩、ピログルタミン酸塩が好ましく、遊離体の上記アミノ酸およびそれらの塩酸塩がより好ましい。
本発明において、遊離体および塩の形態のグルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンは、それぞれ天然に存在する動植物等から抽出し精製したもの、或いは、化学合成法、発酵法、酵素法または遺伝子組換え法によって得られるもののいずれを使用してもよいが、各社より提供されている市販の製品を利用してもよい。
本発明の剤が、シスチンに加え、他のアミノ酸として、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸を含有する場合、当該アミノ酸の含有量は、製剤安定性、行動体力向上機能および服用のしやすさ等の観点から、本発明の剤における全アミノ酸含有量に対して、好ましくは0.1重量%以上であり、より好ましくは1重量%以上であり、特に好ましくは3重量%以上である。また、当該アミノ酸の含有量は、本発明の剤における全アミノ酸含有量に対して、好ましくは99重量%以下であり、より好ましくは90重量%以下であり、特に好ましくは80重量%以下である。
ここで、当該他のアミノ酸の含有量とは、前記の群から選ばれる一つのアミノ酸を含有する場合はそのアミノ酸の含有量であり、前記の群から選ばれる2種以上のアミノ酸を含有する場合はそれらの含有量の合計を意味する。
ここで、当該他のアミノ酸の含有量とは、前記の群から選ばれる一つのアミノ酸を含有する場合はそのアミノ酸の含有量であり、前記の群から選ばれる2種以上のアミノ酸を含有する場合はそれらの含有量の合計を意味する。
本発明の剤が、シスチンに加え、他のアミノ酸として、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸を含有する場合、当該アミノ酸の含有量は、製剤安定性、行動体力向上機能および服用のしやすさ等の観点から、ヒトの1回あたりの摂取量または投与量として、通常5mg以上であり、好ましくは10mg以上であり、より好ましくは50mg以上であり、特に好ましくは100mg以上である。また、当該アミノ酸の含有量は、ヒトの1回あたりの摂取量または投与量として、好ましくは10g以下であり、より好ましくは5g以下である。
本発明の剤が、シスチンに加え、他のアミノ酸として、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸を含有する場合、当該アミノ酸の含有量とシスチンの含有量との合計は、行動体力向上効果の観点から、本発明の剤における全アミノ酸含有量に対して、好ましくは1.1重量%以上であり、より好ましくは2重量%以上であり、更に好ましくは4重量%以上であり、特に好ましくは11重量%以上である。また、当該アミノ酸の含有量とシスチンの含有量との合計は、本発明の剤における全アミノ酸含有量に対して、好ましくは95重量%以下であり、より好ましくは90重量%以下である。
本発明の剤が、シスチンに加え、他のアミノ酸として、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸を含有する場合、シスチンの含有量(a)と、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸の含有量(b)との重量比[(a):(b)]は、各アミノ酸の行動体力向上機能の観点から、好ましくは1:0.01~100であり、より好ましくは1:0.01~50であり、更に好ましくは1:0.1~50であり、より一層好ましくは1:0.1~30であり、最も好ましくは1:1~25である。
本発明の剤が、シスチンに加え、他のアミノ酸として、イソロイシン、ロイシンおよびバリンからなる群より選ばれる少なくとも一つのアミノ酸を含有する場合、シスチンの含有量(a)と、イソロイシン、ロイシンおよびバリンからなる群より選ばれる少なくとも一つのアミノ酸の含有量(b1)との重量比[(a):(b1)]は、1:0.01~100であることが好ましく、1:0.01~50であることがより好ましく、1:0.01~25であることが更に好ましく、1:0.1~10であることがより一層好ましい。
本発明の剤が、シスチンに加え、他のアミノ酸として、イソロイシン、ロイシンおよびバリンを含有する場合、それらの各含有量の重量比(イソロイシン:ロイシン:バリン)は、通常1:1.5~2.5:0.8~1.7であり、好ましくは1:1.9~2.2:1.1~1.3である。また、シスチンの含有量(a)と、イソロイシン、ロイシンおよびバリンの総含有量(b2)との重量比[(a):(b2)]は、1:0.01~100であることが好ましく、1:0.01~50であることがより好ましく、1:0.1~25であることが更に好ましく、1:0.5~20であることがより一層好ましい。
本発明の剤が、シスチンに加え、他のアミノ酸として、イソロイシン、ロイシンおよびバリンを含有する場合、それらの各含有量の重量比(イソロイシン:ロイシン:バリン)は、通常1:1.5~2.5:0.8~1.7であり、好ましくは1:1.9~2.2:1.1~1.3である。また、シスチンの含有量(a)と、イソロイシン、ロイシンおよびバリンの総含有量(b2)との重量比[(a):(b2)]は、1:0.01~100であることが好ましく、1:0.01~50であることがより好ましく、1:0.1~25であることが更に好ましく、1:0.5~20であることがより一層好ましい。
本発明の剤が、シスチンに加え、他のアミノ酸としてグルタミンを含有する場合、シスチンの含有量(a)と、グルタミンの含有量(b3)との重量比[(a):(b3)]は、1:0.01~100であることが好ましく、1:0.01~50であることがより好ましく、1:0.01~25であることが更に好ましく、1:0.1~10であることがより一層好ましい。
本発明の剤が、シスチンに加え、他のアミノ酸としてアラニンまたは、アラニンおよびプロリンを含有する場合、シスチンの含有量(a)と、アラニンまたは、アラニンおよびプロリンの含有量(b4)との重量比[(a):(b4)]は、1:0.01~100であることが好ましく、1:0.01~50であることがより好ましく、1:0.1~50であることが更に好ましく、1:1~50であることがより一層好ましく、1:5~25であることが特に好ましい。
また、本発明の剤が、シスチンに加え、他のアミノ酸としてプロリンを含有する場合、シスチンの含有量(a)と、プロリンの含有量(b5)との重量比[(a):(b5)]は、1:0.01~100であることが好ましく、1:0.01~50であることがより好ましく、1:0.01~25であることが更に好ましく、1:0.1~10であることがより一層好ましく、1:0.5~5であることが特に好ましい。
また、本発明の剤が、シスチンに加え、他のアミノ酸としてプロリンを含有する場合、シスチンの含有量(a)と、プロリンの含有量(b5)との重量比[(a):(b5)]は、1:0.01~100であることが好ましく、1:0.01~50であることがより好ましく、1:0.01~25であることが更に好ましく、1:0.1~10であることがより一層好ましく、1:0.5~5であることが特に好ましい。
本発明の剤は、更に、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリン以外のアミノ酸、たとえば、スレオニン、メチオニン、フェニルアラニン等の必須アミノ酸、アスパラギン酸、グリシン、チロシン等の非必須アミノ酸等をも含有し得るが、非タンパク質構成アミノ酸(non-proteinogenic amino acid)を実質的に含有しないことが好ましい。本発明の剤は、非タンパク質構成アミノ酸を実質的に含有しないことにより、本発明の剤の用量が限られる場合であっても、運動時に必要なタンパク質原料を十分に供給し、且つ、行動体力向上機能を発揮することができる。
非タンパク質構成アミノ酸としては、例えば、テアニン、オルニチン、シトルリン、タウリン、GABA(γ-アミノ酪酸)等が挙げられ、中でもテアニンは直接的にはタンパク質原料となり得ないことから、本発明の剤はテアニンを実質的に含有しないことが好ましい。
本発明において、非タンパク質構成アミノ酸(例えば、テアニン等)を「実質的に含有しない」とは、(1)非タンパク質構成アミノ酸を全く含有しない場合、または(2)非タンパク質構成アミノ酸を、本発明の効果に影響を与えない程度の量(具体的には、本発明の剤における全アミノ酸含有量に対して、通常28重量%以下、好ましくは20重量%以下、より好ましくは10重量%以下、更に好ましくは1重量%以下、特に好ましくは0.1重量%以下)で含有する場合のいずれかであることを意味する。
非タンパク質構成アミノ酸としては、例えば、テアニン、オルニチン、シトルリン、タウリン、GABA(γ-アミノ酪酸)等が挙げられ、中でもテアニンは直接的にはタンパク質原料となり得ないことから、本発明の剤はテアニンを実質的に含有しないことが好ましい。
本発明において、非タンパク質構成アミノ酸(例えば、テアニン等)を「実質的に含有しない」とは、(1)非タンパク質構成アミノ酸を全く含有しない場合、または(2)非タンパク質構成アミノ酸を、本発明の効果に影響を与えない程度の量(具体的には、本発明の剤における全アミノ酸含有量に対して、通常28重量%以下、好ましくは20重量%以下、より好ましくは10重量%以下、更に好ましくは1重量%以下、特に好ましくは0.1重量%以下)で含有する場合のいずれかであることを意味する。
本発明の剤は、他の栄養剤や抗疲労剤等と併用することもできる(以下、「併用薬」と称することもある)。併用薬としては、具体的には、グルコース、デキストラン等の糖質製剤、精製大豆油、精製卵黄レシチン等の脂肪乳剤、カゼイン、ホエイタンパク等のタンパク質製剤、カフェイン、ビタミン類、ミネラル類、ポリフェノール類等が挙げられる。
本発明の剤は、乳酸菌やビフィズス菌といった善玉菌と併用することもできる。善玉菌としては、特に制限されず、具体的には「VSL#3」(登録商標)(インカイン ファーマシューティカル(InKine Pharmaceutical) 社およびシグマ-タウ ファーマシューティカルズ(Sigma-Tau Pharmaceuticals)社)などが挙げられる。
本発明の剤は、錠剤、被覆錠剤、チュアブル錠、丸剤、(マイクロ)カプセル剤、顆粒剤、細粒剤、散剤、エリキシル剤、リモナーゼ剤、シロップ剤、懸濁剤、乳剤、経口ゼリー剤等の経口製剤、溶液状、懸濁液状、乳液状等の注射剤、用時溶解または懸濁して用いる固形状の注射剤、輸液剤、持続性注射剤等の注射用製剤等の剤形とすることができる。
上記剤形の本発明の剤は、製剤の分野で周知の製剤化手段、たとえば第十六改正日本薬局方製剤総則[2]製剤各条、第十七改正日本薬局方製剤総則[3]製剤各条等に記載された方法等により、調製することができる。その際、必要に応じて、薬理学的に許容し得る各種の製剤用添加剤を配合することができる。当該添加剤は、本発明の剤の剤形に応じて適宜選択することができるが、例えば、賦形剤、結合剤、崩壊剤、滑沢剤、被覆剤、基剤、溶剤、希釈剤、溶解補助剤、可溶化剤、乳化剤、分散剤、懸濁化剤、安定化剤、粘稠剤、無痛化剤、等張化剤、pH調整剤、抗酸化剤、防腐剤、保存剤、矯味剤、風味剤、甘味剤、香料、着色剤等が挙げられる。
具体的には、例えば賦形剤として、炭酸マグネシウム、二酸化チタン、糖類(例えば、ラクトース等)、糖アルコール(例えば、マンニトール等)、カゼイン等を挙げることができる。
結合剤としては、例えばゼラチン、澱粉、セルロースおよびその誘導体等を挙げることができる。
崩壊剤としては、例えばクロスポビドン、結晶セルロース等を挙げることができる。
滑沢剤としては、例えばタルク、ステアリン酸マグネシウム等を挙げることができる。
被覆剤としては、例えばメタクリル酸メチル・メタクリル酸ブチル・メタクリル酸ジメチルアミノエチル共重合体、アクリル酸エチル・メタクリル酸メチル・メタクリル酸塩化トリメチルアンモニウムエチル共重合体等を挙げることができる。
基剤としては、例えば動物および植物油、ポリエチレングリコール等を挙げることができる。
溶剤としては、例えば精製水、注射用水、一価または多価アルコール(例えば、グリセリン等)を挙げることができる。

本発明の剤の摂取量または投与量は、適用される対象(以下本明細書において、「適用対象」という)の状態または症状、性別、年齢、本発明の剤の剤形、投与方法等により適宜決定されるが、適用対象がヒト成人である場合、本発明の剤は、1日あたり、通常0.1mg/kg体重~2g/kg体重、好ましくは1mg/kg体重~1g/kg体重、より好ましくは5mg/kg体重~0.2g/kg体重のシスチンが摂取または投与されるように用いられ得る。
本発明の剤が、シスチンに加え、他のアミノ酸として、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸を含有する場合、ヒト成人に対し、本発明の剤は、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸が、以下の量で摂取または投与されるように用いられ得る。

上記の1日あたりの摂取または投与量は、1回で摂取させまたは投与してもよく、もしくは2回以上(例えば、2~5回)に分けて摂取させまたは投与することもできる。
本発明の剤は、適用対象がヒト成人である場合、1日あたりの全アミノ酸の摂取または投与量の合計が、通常0.1mg/kg体重~5g/kg体重となるように用いられ得る。特に適用対象が例えば運動選手等、筋持久力または全身持久力等の行動体力の向上が特に望まれる者である場合、本発明の剤は、本発明の剤の行動体力の向上機能と運動選手等が1日に摂取すべきタンパク質量との観点から、1日あたりの全アミノ酸の摂取または投与量の合計が、好ましくは10mg/kg体重~2g/kg体重、より好ましくは50mg/kg体重~1g/kg体重となるように用いられ得る。
本発明の剤の摂取または投与の時期は特に制限されず、例えば、運動開始前、運動中、運動終了後等のいずれの時期であってもよい。
本発明の剤の摂取または投与の回数は特に制限されないが、行動体力の向上を要する時に、少なくとも1回(1回または2回以上)である。本発明の剤は、適用の簡便性の観点から、運動開始前または運動中もしくは運動終了後に少なくとも1回摂取または投与されることが好ましい。また本発明の剤は、運動開始前に少なくとも1回摂取または投与された後、更に、運動中及び/または運動終了後に少なくとも1回摂取または投与されてもよい。
本発明の剤の摂取または投与の回数が2回以上である場合、本発明の剤の摂取または投与の期間(最初の摂取または投与から、最後の摂取または投与までの期間)は特に限定されないが、通常6時間~4週間であり、本発明の効果がより発揮される観点から、好ましくは1日間~2週間であり、より好ましくは3日間~1週間である。
一実施態様として、本発明の剤は、運動開始の1日前~2週間前(好ましくは3日前~1週間前)から、1日あたり1~5回(好ましくは1~3回)摂取または投与される。また、本発明の剤を運動終了後に摂取または投与する場合には、運動終了の直後~24時間後(好ましくは直後~6時間後)に、1~3回摂取または投与することができる。
本発明の剤の摂取または投与の回数は特に制限されないが、行動体力の向上を要する時に、少なくとも1回(1回または2回以上)である。本発明の剤は、適用の簡便性の観点から、運動開始前または運動中もしくは運動終了後に少なくとも1回摂取または投与されることが好ましい。また本発明の剤は、運動開始前に少なくとも1回摂取または投与された後、更に、運動中及び/または運動終了後に少なくとも1回摂取または投与されてもよい。
本発明の剤の摂取または投与の回数が2回以上である場合、本発明の剤の摂取または投与の期間(最初の摂取または投与から、最後の摂取または投与までの期間)は特に限定されないが、通常6時間~4週間であり、本発明の効果がより発揮される観点から、好ましくは1日間~2週間であり、より好ましくは3日間~1週間である。
一実施態様として、本発明の剤は、運動開始の1日前~2週間前(好ましくは3日前~1週間前)から、1日あたり1~5回(好ましくは1~3回)摂取または投与される。また、本発明の剤を運動終了後に摂取または投与する場合には、運動終了の直後~24時間後(好ましくは直後~6時間後)に、1~3回摂取または投与することができる。
本発明の剤が、シスチンと、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸とを含有する場合、これらはそれぞれ単独で、もしくは任意の組み合わせで二つ以上の製剤に含有されていてもよいし、または、全てが一つの製剤に含有されていてもよい。これらの有効成分が二つ以上の製剤に含有されている場合、各製剤が摂取または投与される時期は、同時であっても別々であってもよく、また各製剤が摂取または投与される経路は、同一であっても異なっていてもよい。本発明の剤は、全ての有効成分が一つの製剤に含有されている態様が、簡便に摂取または投与することができ、好ましい。シスチンおよびグルタミン等のアミノ酸の他に、上記した併用薬を用いる場合、その摂取または投与の方法(別の製剤とするか、同一の製剤とするか)および摂取または投与の時期は、併用薬の種類や効果によって適宜決定し得る。
本発明において、シスチンと、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニン、およびプロリンからなる群より選ばれる少なくとも一つのアミノ酸とが、二つ以上の製剤に含有されている場合、これらの各含有量(1回あたりの摂取量または投与量としての含有量、および本発明の剤における全アミノ酸含有量に対する含有量)や重量比は、各製剤における含有量を合計して表される。
本発明において、シスチンと、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニン、およびプロリンからなる群より選ばれる少なくとも一つのアミノ酸とが、二つ以上の製剤に含有されている場合、これらの各含有量(1回あたりの摂取量または投与量としての含有量、および本発明の剤における全アミノ酸含有量に対する含有量)や重量比は、各製剤における含有量を合計して表される。
本発明の剤は、単位包装形態とすることができる。本明細書において「単位包装形態」とは、特定量(例えば、1回あたりの摂取量等)を1単位とし、該1単位または2単位以上が一つの容器または包装体に収容された形態をいい、例えば、1回あたりの摂取量または投与量を1単位とする単位包装形態は、「1回あたりの摂取量または投与量単位の包装形態」と称する。単位包装形態に用いられる容器または包装体は、本発明の剤の形態等に応じて適宜選択し得るが、例えば、紙製の容器または袋体、プラスチック製の容器または袋体、パウチ、アルミ缶、スチール缶、ガラス瓶、ペットボトル、PTP(press through pack)包装シート等が挙げられる。
本発明の剤の適用対象としては、哺乳動物(例えば、ヒト、マウス、ラット、ハムスター、ウサギ、ネコ、イヌ、ウシ、ウマ、ロバ、ブタ、ヒツジ、サル等)や、鳥類(例えば、ニワトリ等)が挙げられる。なお、本発明の剤をヒト以外の適用対象動物(以下、単に「対象動物」ともいう)に適用する場合、本発明の剤の摂取または投与量は、対象動物の種類、性別、体重等に応じて適宜設定すればよい。
本発明の剤は、行動体力、特に筋持久力、全身持久力といった行動を持続する能力を向上させることができる。
特に、本発明の剤は、脂肪組織と横紋筋(心筋および骨格筋)に存在し、グルコースの取り込みに関与するグルコーストランスポーター4(GLUT4)等、グルコーストランスポーター遺伝子の発現を増強して、エネルギーとなるグルコースの取り込みを増強させることから、行動体力を効果的に向上させることができる。
また、本発明の剤は、筋肉中のグリコーゲン量を増加させることができる。従って、本発明の剤は、筋肉中のグリコーゲン量の増加剤として用いることができ、特に、体内にグリコーゲンを貯蔵させるグリコーゲンローディング促進剤としても好適に用いられ、運動能力の限界を上昇させることができる。
さらに、本発明の剤は、運動(トレーニング)による筋肉中のグリコーゲン量の増加、筋持久力、全身持久力の向上等の効果を、より高めることができる。
特に、本発明の剤は、脂肪組織と横紋筋(心筋および骨格筋)に存在し、グルコースの取り込みに関与するグルコーストランスポーター4(GLUT4)等、グルコーストランスポーター遺伝子の発現を増強して、エネルギーとなるグルコースの取り込みを増強させることから、行動体力を効果的に向上させることができる。
また、本発明の剤は、筋肉中のグリコーゲン量を増加させることができる。従って、本発明の剤は、筋肉中のグリコーゲン量の増加剤として用いることができ、特に、体内にグリコーゲンを貯蔵させるグリコーゲンローディング促進剤としても好適に用いられ、運動能力の限界を上昇させることができる。
さらに、本発明の剤は、運動(トレーニング)による筋肉中のグリコーゲン量の増加、筋持久力、全身持久力の向上等の効果を、より高めることができる。
また、本発明の剤は、そのまま、またはさらに上記した賦形剤、溶剤、希釈剤等の添加剤を加えて、行動体力の向上の求められる運動選手、高齢者、要介護者、運動療法を行っている患者等に投与される医薬とすることができる。
さらに、本発明の剤は、各種食品に添加して使用することができる。本発明の剤が添加される食品は特に制限されず、一般的に食事に供される形態の食品であれば如何なるものでもよい。例えば、本発明の剤を飲料に添加し、所望により適当な風味を加えて、ドリンク剤(例えば、清涼飲料等)とすることができる。より具体的には、本発明の剤は、例えば、ジュース、牛乳、菓子、ゼリー、ヨーグルト、飴等に添加することができる。
本発明はまた、本発明の剤を含有する、行動体力向上用の食品組成物(以下、「本発明の食品組成物」ともいう)を提供する。
本発明の食品組成物は、本発明の剤と、必要に応じて、製造用剤、増粘安定剤、ガムベース、乳化剤、保存料、酸化防止剤、光沢剤、pH調整剤、甘味料、苦味料、酸味料、着色料、香料等の食品添加物を含有し、あるいは、本発明の剤と、食品もしくは食品原材料とを含有して、例えばジュース類、清涼飲料水、茶類等の飲料;乳酸菌飲料、発酵乳、バター、チーズ、ヨーグルト、加工乳、脱脂乳等の乳製品;ハム、ソーセージ、ハンバーグ等の畜肉製品;蒲鉾、竹輪、さつま揚げ等の魚肉練り製品;だし巻き、卵豆腐等の卵製品;クッキー、ゼリー、チューイングガム、キャンディー、スナック菓子、冷菓等の菓子類;パン類;麺類;漬物類;燻製品;干物;佃煮;塩蔵品;スープ類;調味料等、種々の形態で提供することができ、瓶詰め食品、缶詰食品、レトルトパウチ食品であってもよい。また、粉末状、顆粒状、シート状、カプセル状、タブレット状、ゼリー状等の形態とすることもできる。
本発明の食品組成物は、行動体力の向上の求められる運動選手、高齢者、要介護者、運動療法を行っている患者等に好適に摂取され得る。
また、本発明の食品組成物は、運動選手ではないが日常的によく運動する者、高齢者ではないが行動体力の維持または向上を望む中・壮年者、介護は要しないが、負傷、疾患等のためにリハビリテーションを要する者等、行動体力の向上を希望する者に幅広く摂取させることができる。
本発明の食品組成物は、本発明の剤と、必要に応じて、製造用剤、増粘安定剤、ガムベース、乳化剤、保存料、酸化防止剤、光沢剤、pH調整剤、甘味料、苦味料、酸味料、着色料、香料等の食品添加物を含有し、あるいは、本発明の剤と、食品もしくは食品原材料とを含有して、例えばジュース類、清涼飲料水、茶類等の飲料;乳酸菌飲料、発酵乳、バター、チーズ、ヨーグルト、加工乳、脱脂乳等の乳製品;ハム、ソーセージ、ハンバーグ等の畜肉製品;蒲鉾、竹輪、さつま揚げ等の魚肉練り製品;だし巻き、卵豆腐等の卵製品;クッキー、ゼリー、チューイングガム、キャンディー、スナック菓子、冷菓等の菓子類;パン類;麺類;漬物類;燻製品;干物;佃煮;塩蔵品;スープ類;調味料等、種々の形態で提供することができ、瓶詰め食品、缶詰食品、レトルトパウチ食品であってもよい。また、粉末状、顆粒状、シート状、カプセル状、タブレット状、ゼリー状等の形態とすることもできる。
本発明の食品組成物は、行動体力の向上の求められる運動選手、高齢者、要介護者、運動療法を行っている患者等に好適に摂取され得る。
また、本発明の食品組成物は、運動選手ではないが日常的によく運動する者、高齢者ではないが行動体力の維持または向上を望む中・壮年者、介護は要しないが、負傷、疾患等のためにリハビリテーションを要する者等、行動体力の向上を希望する者に幅広く摂取させることができる。
従って、本発明の食品組成物は、行動体力の維持、向上用の特定保健用食品、栄養機能食品、機能性表示食品等の保健機能食品、病者用食品、高齢者用食品等の特別用途食品、健康補助食品等としても提供され得る。
さらに、本発明の剤を、濃厚流動食や、食品補助剤に添加して使用することも可能である。
「濃厚流動食」とは、1kcal/mL程度の濃度に調整され、長期間これのみを摂取する場合であっても、著しい栄養素の過不足が生じないように、各栄養素の質的構成が十分に考慮され、1日の栄養所要量をもとに設計された総合栄養食品(液体状食品)である。
本発明における「食品補助剤」とは、食品として摂取されるもの以外に栄養を補助する目的で摂取されるものをいい、栄養補助剤、サプリメントなどもこれに含まれる。本発明の剤を食品補助剤に添加する場合、所望により他の栄養成分や添加剤を加えて、例えば錠剤、カプセル、散剤、顆粒、懸濁剤、チュアブル剤、シロップ剤等の形態に調製することができる。
「濃厚流動食」とは、1kcal/mL程度の濃度に調整され、長期間これのみを摂取する場合であっても、著しい栄養素の過不足が生じないように、各栄養素の質的構成が十分に考慮され、1日の栄養所要量をもとに設計された総合栄養食品(液体状食品)である。
本発明における「食品補助剤」とは、食品として摂取されるもの以外に栄養を補助する目的で摂取されるものをいい、栄養補助剤、サプリメントなどもこれに含まれる。本発明の剤を食品補助剤に添加する場合、所望により他の栄養成分や添加剤を加えて、例えば錠剤、カプセル、散剤、顆粒、懸濁剤、チュアブル剤、シロップ剤等の形態に調製することができる。
上記した本発明の食品組成物は、本発明の剤に、必要に応じて食品添加物を加えて、あるいは、食品または食品原材料中に本発明の剤を添加し、一般的な食品の製造方法を用いることにより、加工、製造することができる。
本発明の食品組成物におけるシスチンの含有量、およびグルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸の含有量は、食品組成物の種類もしくは形態、当該食品組成物の摂取により期待される行動体力向上効果の程度等に応じて適宜決定されるが、食品の総重量に対して、通常シスチンが1重量%~95重量%程度、好ましくは3重量%~90重量%程度、より好ましくは10重量%~80重量%程度であり、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンから選ばれる少なくとも一つのアミノ酸の総量にして1重量%~95重量%程度、好ましくは3重量%~90重量%程度である。
また、本発明の食品組成物の摂取量は、本発明の剤について上述したシスチン等の摂取量を摂取できる量として設定され得る。
本発明の剤は、さらに、本発明の剤を行動体力の向上のために使用することができること、または使用すべきであることを記載した記載物を含む、商業的パッケージの態様でも提供され得る。
さらに本発明は、行動体力を向上させる必要のある対象動物の行動体力の向上方法(以下、本明細書において「本発明の方法」ともいう)をも提供する。
本発明の方法は、行動体力を向上させる必要のある対象動物に、当該対象動物の行動体力を向上させるのに有効な量のシスチンを摂取させること、または投与することを含む。
本発明の方法は、さらにシスチンに加え、対象動物の行動体力を向上させるのに有効な量のグルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸を摂取させること、または投与することを含む。
本発明の方法は、さらにシスチンに加え、対象動物の行動体力を向上させるのに有効な量のグルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸を摂取させること、または投与することを含む。
本発明の方法における対象動物としては、ヒトおよび、マウス、ラット、ハムスター、ウサギ、ネコ、イヌ、ウシ、ウマ、ロバ、ブタ、ヒツジ、サル等のヒト以外の哺乳動物、ならびにニワトリなどの鳥類が挙げられる。
本発明の方法の対象動物がヒトである場合、本発明の方法は、運動する者や、身体機能、運動能力等の低下傾向が見られる者を含め、筋持久力、全身持久力等行動体力の向上を望む者に対して幅広く適用することができるが、特に行動体力の向上が望まれる運動選手、あるいは筋持久力、全身持久力といった行動を持続する能力の低下の認められる高齢者や要介護者、糖尿病等の疾患を有し、運動療法を受けている患者等に好適に適用される。
本発明の方法の対象動物がヒトである場合、本発明の方法は、運動する者や、身体機能、運動能力等の低下傾向が見られる者を含め、筋持久力、全身持久力等行動体力の向上を望む者に対して幅広く適用することができるが、特に行動体力の向上が望まれる運動選手、あるいは筋持久力、全身持久力といった行動を持続する能力の低下の認められる高齢者や要介護者、糖尿病等の疾患を有し、運動療法を受けている患者等に好適に適用される。
本発明の方法におけるシスチン等の摂取量または投与量は、対象動物の種類、年齢、症状または状態等に応じて決定されるが、本発明の剤において、ヒトおよびヒト以外の対象動物について、上記した摂取量または投与量と同様の量を、上記した回数および期間にて摂取させまたは投与することができる。
なお、本発明の方法において、シスチンの1回あたりの摂取量または投与量(c)と、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸の1回あたりの摂取量または投与量(d)との重量比[(c):(d)]は、1:0.001~100であることが好ましく、1:0.01~100であることがより好ましい。
なお、本発明の方法において、シスチンの1回あたりの摂取量または投与量(c)と、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸の1回あたりの摂取量または投与量(d)との重量比[(c):(d)]は、1:0.001~100であることが好ましく、1:0.01~100であることがより好ましい。
さらに、本発明の方法におけるシスチン等の摂取または投与方法としては、経口投与、経腸経管投与、輸液による投与等が挙げられるが、医療機関にて医師の指導監督下に行う必要がなく、簡便に摂取させることができることから、経口投与が好ましい。
以下、実施例により本発明をさらに詳細に説明するが、本発明の実施範囲はこれらの例によってなんら限定されるものではない。
[試験例1]シスチンの持久力向上効果の検討
運動選手は激しいトレーニングを繰り返し、身体機能を向上させる。そこで、下記のような試験にて、持久力向上に対するシスチン摂取の効果を検討した。

6週齢の雄性CD2F1マウス(日本チャールス・リバー株式会社)を3群に分け、それぞれ、普通食(AIN-93G組成)を摂取させ運動させない群(非運動群)、普通食を摂取させ運動させる群(運動群)、普通食のカゼインをシスチンに置き換えることにより2重量%のシスチン(遊離体)を含有させた食餌を与えて運動させる群(運動+2%シスチン群)とした。各群に8日間試験食(普通食または2重量%のシスチンを含有する普通食)を摂取させ、その後、運動群と運動+2%シスチン群については、3日間連続でトレッドミルにて疲労困憊するまで走行させ、そのときの走行時間を測定した(図1)。非運動群は、他の2群の走行中は絶食・絶水とした。3日間の走行が終了した翌日、各群のマウスの筋肉を採取し、筋肉中のグリコーゲン量を測定した。
なお、走行はアルコ(ARCO)社製の小型動物用トレッドミルを用いて行わせ、走行速度=18m/minで30分間走行させた後、30分毎に走行速度を3m/min増加させていき、走行速度が36m/minに達した場合は以後、36m/minを維持した。また、マウスがレーンの後方に下がり、タップしても走行レーンに戻って走行を再開できない場合に、マウスが疲労困憊したと判断した。
筋肉中のグリコーゲン量の測定は以下のように行った。マウスの腓腹筋50mgを300μLの30重量%水酸化カリウム水溶液に入れ、ヒートブロック上(100℃)で30分間消化し、グリコーゲンを抽出した後、飽和硫酸ナトリウム水溶液を用いて消化液を中性にした。その後95容量%のエタノールを加え、遠心分離後ペレットを形成させた。この操作を2回繰り返した後、得られた沈殿に0.6N塩酸を加えてヒートブロック上で2時間処理し、グルコースに加水分解した。この溶液中のグルコースを、「グルコースCII-テストワコー」(和光純薬工業株式会社製)を用いて検出、定量した。得られたグルコース量を0.93倍し筋肉中のグリコーゲン量を算出した。
疲労困憊するまでの走行時間の測定結果、および腓腹筋中のグリコーゲン量の測定結果については、分散分析を実施後、テューキー(Tukey)法により検定を実施した。
なお、走行はアルコ(ARCO)社製の小型動物用トレッドミルを用いて行わせ、走行速度=18m/minで30分間走行させた後、30分毎に走行速度を3m/min増加させていき、走行速度が36m/minに達した場合は以後、36m/minを維持した。また、マウスがレーンの後方に下がり、タップしても走行レーンに戻って走行を再開できない場合に、マウスが疲労困憊したと判断した。
筋肉中のグリコーゲン量の測定は以下のように行った。マウスの腓腹筋50mgを300μLの30重量%水酸化カリウム水溶液に入れ、ヒートブロック上(100℃)で30分間消化し、グリコーゲンを抽出した後、飽和硫酸ナトリウム水溶液を用いて消化液を中性にした。その後95容量%のエタノールを加え、遠心分離後ペレットを形成させた。この操作を2回繰り返した後、得られた沈殿に0.6N塩酸を加えてヒートブロック上で2時間処理し、グルコースに加水分解した。この溶液中のグルコースを、「グルコースCII-テストワコー」(和光純薬工業株式会社製)を用いて検出、定量した。得られたグルコース量を0.93倍し筋肉中のグリコーゲン量を算出した。
疲労困憊するまでの走行時間の測定結果、および腓腹筋中のグリコーゲン量の測定結果については、分散分析を実施後、テューキー(Tukey)法により検定を実施した。
運動1日目(day1)、2日目(day2)および3日目(day3)の、疲労困憊するまでの走行時間の測定結果を、図2に示した。両群(運動群(Ex)および運動+2%シスチン群(Ex+Cyt))ともに運動を連日行うことにより、疲労困憊するまでの走行時間は増加していったことから、運動により、持久力が向上することが示された。運動による走行時間の増加量は、運動群(Ex)よりも、運動+2%シスチン群(Ex+Cyt)の方が有意(P<0.05またはP<0.01)に増大していた。この結果から、シスチンは、運動による持久力向上効果を増強することがわかった。
3日間連続して運動させた翌日に、腓腹筋中のグリコーゲン量を測定した結果を図3に示した。非運動群(Sed)と比較して、運動を3日間連続して実施した運動群(Ex)の腓腹筋中のグリコーゲン量は、有意(P<0.05)に増加していた。さらに運動+2%シスチン群(Ex+Cyt)では、運動群(Ex)よりも腓腹筋中のグリコーゲン量が更に有意(P<0.05)に増加していた。
以上の結果から、シスチンは、運動のみ実施するよりも、筋肉中のグリコーゲン量を更に効率的に増加させることが判明した。

[試験例2]シスチンの摂取時期が持久力向上効果に及ぼす影響の検討
試験例1では、運動前に8日間シスチンを摂取した際にシスチンが持久力向上効果を有することを見出したが、運動後のシスチンの摂取によっても効果が見られるかについて、検討した。

6週齢の雄性CD2F1マウス(日本チャールス・リバー株式会社)に普通食(AIN-93G組成)を8日間摂取させた。その後、試験例1と同様の運動条件で疲労困憊するまで走行させ、そのときの走行時間を測定した。運動後に、前記マウスを、普通食を摂取させる群(運動群)、および、普通食のカゼインをシスチンに置き換えることにより2重量%のシスチン(遊離体)を含有させた食餌を摂取させる群(運動+2%シスチン群)の2群に分け、各群に試験食(普通食または2重量%のシスチンを含有する普通食)を摂取させた。その翌日に再度、試験例1と同様の運動条件で疲労困憊するまで走行させ、そのときの走行時間を測定し、走行時間の増加量を持久力向上効果として算出した。走行時間の増加量の算出結果については、対応のないt検定を実施した。
走行時間の増加量を、図4に示した。走行時間の増加量は、運動群(Ex)よりも、運動+2%シスチン群(Ex+Cyt)の方が増加する傾向が見られた(P<0.1)。この結果から、シスチンは、運動後に1日間摂取することによっても、持久力向上効果を増強することが示唆された。
[実施例1]行動体力向上剤
表1に示す組成に従い、各アミノ酸(リシンは塩酸塩を用い、その他のアミノ酸は遊離体を用いた)を混合した組成物を、実施例1の行動体力向上剤とした。また、特許文献2にて、持久力向上効果を有すると報告されたアミノ酸組成物を比較例として、その組成を表1に併せて示した。
表1に示す組成に従い、各アミノ酸(リシンは塩酸塩を用い、その他のアミノ酸は遊離体を用いた)を混合した組成物を、実施例1の行動体力向上剤とした。また、特許文献2にて、持久力向上効果を有すると報告されたアミノ酸組成物を比較例として、その組成を表1に併せて示した。
[試験例3]実施例1の行動体力向上剤の持久力向上効果の検討
特許文献2にて開示されているシスチンを含むアミノ酸組成物(比較例のアミノ酸組成物)について、持久力向上効果が報告されている。そこで、アミノ酸組成物中のシスチン含有量の違いにより、持久力向上効果が変化するかについて、下記試験で検討した。

6週齢の雄性CD2F1マウス(日本チャールス・リバー株式会社)を4群に分け、比較例のアミノ酸組成物(表1)を3重量%含有する普通食(AIN-93G組成)を与えて運動させない群(Sed-Con)と運動させる群(Ex-Con)、実施例1の行動体力向上剤(表1)を3重量%含有する普通食を与えて運動させない群(Sed-Cyt)と運動させる群(Ex-Cyt)をそれぞれ設定した。試験食(比較例のアミノ酸組成物を含有する普通食または実施例1の行動体力向上剤を含有する普通食)を7日間摂取させ、その後、運動させる群は2日間連続してトレッドミルにて疲労困憊するまで走行させ、そのときの走行時間を測定した。運動させない群は、運動させる群の走行中は絶食・絶水とした。2日間の走行が終了した翌日、各群のマウスの腓腹筋を採取し、試験例1と同様に筋肉中のグリコーゲン量を測定した。走行条件は試験例1の走行条件と同様とした。

運動1日目(1d)および2日目(2d)の、疲労困憊するまでの走行時間の測定結果を、図5に示した。また、運動1日目と2日目の走行時間の差(増加分)を、図6に示した。1dの走行時間については、運動させた両群間で差は見られなかったが、2dの走行時間については、比較例のアミノ酸組成物を含有する普通食を与えて運動させた群(Ex-Con)と比較して、実施例1の行動体力向上剤を含有する普通食を与えて運動させた群(Ex-Cyt)では、疲労困憊するまでの走行時間が有意(P<0.01)に増加していた。1日目と2日目の走行時間の差についても、Ex-Conと比較して、Ex-Cytでは有意(P<0.01)に高値を示した。
以上の結果から、特許文献2で開示されているシスチンを含有するアミノ酸組成物よりも、シスチンを高含有量で含有する本発明の行動体力向上剤は、運動により効率的に持久力を向上させることが明らかとなった。

各群の腓腹筋中のグリコーゲン量の測定結果を、図7に示した。比較例のアミノ酸組成物を含有する普通食を与えて運動させなかった群(Sed-Con)と比較して、実施例1の行動体力向上剤を含有する普通食を与えて運動させなかった群(Sed-Cyt)では、腓腹筋中のグリコーゲン量が有意(P<0.05)に高値を示しており、シスチンを高含有量で含有する本発明の行動体力向上剤が、筋肉中のグリコーゲン量を増加させることがわかった。
さらに、運動を2日間連続して実施させた場合、運動だけでも腓腹筋中のグリコーゲン量は増加することが、比較例のアミノ酸組成物を含有する普通食を与えて運動させた群(Ex-Con)の結果から示されたが、シスチンを高含有量で含有する本発明の行動体力向上剤を摂取させて運動させた群(Ex-Cyt)では、Ex-Conよりも更に腓腹筋中のグリコーゲン量が増加していた(P<0.01で有意)。
以上の結果から、シスチンを高含有量で含有する実施例1の行動体力向上剤は、これを摂取するだけでも、シスチン含有量の低い比較例のアミノ酸組成物に比べて筋肉中のグリコーゲン量を増加させるが、運動することにより、筋肉中のグリコーゲン量を増加させる効果をさらに高めることができることがわかった。
さらに、運動を2日間連続して実施させた場合、運動だけでも腓腹筋中のグリコーゲン量は増加することが、比較例のアミノ酸組成物を含有する普通食を与えて運動させた群(Ex-Con)の結果から示されたが、シスチンを高含有量で含有する本発明の行動体力向上剤を摂取させて運動させた群(Ex-Cyt)では、Ex-Conよりも更に腓腹筋中のグリコーゲン量が増加していた(P<0.01で有意)。
以上の結果から、シスチンを高含有量で含有する実施例1の行動体力向上剤は、これを摂取するだけでも、シスチン含有量の低い比較例のアミノ酸組成物に比べて筋肉中のグリコーゲン量を増加させるが、運動することにより、筋肉中のグリコーゲン量を増加させる効果をさらに高めることができることがわかった。
次いで、各群の腓腹筋における糖の取り込み遺伝子の発現について、以下の通り検討した。
各群のマウスより採取した腓腹筋からRNeasy Fibrous Tissue Mini Kit (キアゲン(QIAGEN)社)を用いてTotal RNAの抽出を行った。得られたTotal RNAはナノドロップシステム(株式会社エル・エム・エス)を用いて濃度を測定し、Prime Script RT reagent Kit(タカラバイオ(Takara Bio)株式会社)を用いて逆転写を行い、cDNAを調製した。得られたcDNAはリアルタイムPCRに供し、グルコーストランスポーター4(GLUT4)遺伝子の発現レベルを評価した(使用機器:Thermal Cycler Dice TP800(タカラバイオ(Takara Bio)株式会社)、試薬:SYBR Premix Ex Taq(登録商標) II KiT(タカラバイオ(Takara Bio)株式会社))。GLUT4遺伝子のプライマー配列としては、5’-AATGGAGACTGATGCGCTCT-3’(配列番号1)と5’-ACTCTTGCCACACAGGCTCT-3’(配列番号2)を用いた。なお、ハウスキーピング遺伝子としてGapdhを用いた。
GLUT4遺伝子の発現レベルの評価結果は、比較例のアミノ酸組成物含有食を与えて運動させなかった群(Sed-Con)群における発現レベルを1とした時の相対値で示し、統計的有意差の検定は、分散分析の実施後にテューキー(Tukey)法を用いて実施した。

各群の腓腹筋におけるGLUT4遺伝子の発現レベルを、図8に示した。比較例のアミノ酸組成物含有食を与えて運動させなかった群(Sed-Con)と比較して、実施例1の行動体力向上剤含有食を与えて運動させなかった群(Sed-Cyt)では、腓腹筋中のGLUT4の発現が、有意(P<0.05)に高値を示した。この結果から、シスチンを高含有量で含有する本発明の行動体力向上剤が、筋肉中のGLUT4遺伝子の発現量を増加させることが示唆された。
また、比較例のアミノ酸組成物含有食を与えて運動させた群(Ex-Con)においても、Sed-Conに比べてGLUT4遺伝子の発現レベルが高く、運動を連続して実施させた場合、運動のみによっても、腓腹筋中のGLUT4遺伝子の発現レベルが増加することが示された。シスチンを高含有量で含有する実施例1の行動体力向上剤含有食を与えて運動させた群(Ex-Cyt)では、Ex-Conよりも更に腓腹筋のGLUT4遺伝子の発現レベルは増加していた(P<0.05で有意)。
以上の結果から、シスチンを高含有量で含有する本発明の行動体力向上剤を摂取することにより、筋肉中にグルコースを取り込むトランスポーターを増加させて、行動体力を向上させ得ることが明らかになった。さらに、本発明の行動体力向上剤を摂取して運動することにより、行動体力の向上効果をさらに高めることができることがわかった。
また、比較例のアミノ酸組成物含有食を与えて運動させた群(Ex-Con)においても、Sed-Conに比べてGLUT4遺伝子の発現レベルが高く、運動を連続して実施させた場合、運動のみによっても、腓腹筋中のGLUT4遺伝子の発現レベルが増加することが示された。シスチンを高含有量で含有する実施例1の行動体力向上剤含有食を与えて運動させた群(Ex-Cyt)では、Ex-Conよりも更に腓腹筋のGLUT4遺伝子の発現レベルは増加していた(P<0.05で有意)。
以上の結果から、シスチンを高含有量で含有する本発明の行動体力向上剤を摂取することにより、筋肉中にグルコースを取り込むトランスポーターを増加させて、行動体力を向上させ得ることが明らかになった。さらに、本発明の行動体力向上剤を摂取して運動することにより、行動体力の向上効果をさらに高めることができることがわかった。
[実施例2]行動体力向上剤
表2に示す組成に従い、各アミノ酸(いずれも遊離体)を混合した組成物を、実施例2の行動体力向上剤とした。

[試験例4]実施例2の行動体力向上剤の持久力向上効果の検討

6週齢の雄性CD2F1マウス(日本チャールス・リバー株式会社)を2群に分け、普通食(AIN-93G組成)を与え運動させる群(Ex-Con)、実施例2の行動体力向上剤(表2)を3.2重量%含有する普通食を与えて運動させる群(Ex-CG)をそれぞれ設定した。試験食(普通食または実施例2の行動体力向上剤を含有する普通食)を7日間摂取させ、その後、2日間連続してトレッドミルにて疲労困憊するまで走行させ、そのときの走行時間を測定した。2日間の走行が終了した翌日、各群のマウスの腓腹筋を採取し、試験例1と同様に筋肉中のグリコーゲン量を測定した。走行条件は試験例1の走行条件と同様とした。
疲労困憊するまでの走行時間の測定結果については、試験例1の場合と同様に、分散分析を実施した後にテューキー(Tukey)法により検定を実施した。走行時間の増加量および腓腹筋中のグリコーゲン量の測定結果の検定には、対応のないt検定を用いた。

運動1日目(1d)および2日目(2d)の、疲労困憊するまでの走行時間の測定結果を、図9に示した。また、運動1日目と2日目の走行時間の差(増加分)を、図10に示した。1dの走行時間については、運動させた両群間で差は見られなかったが、2dの走行時間については、普通食を与えて運動させた群(Ex-Con)と比較して、実施例2の行動体力向上剤を含有する普通食を与えて運動させた群(Ex-CG)では、疲労困憊するまでの走行時間が有意(P<0.05)に増加していた。1日目と2日目の走行時間の差についても、Ex-Conと比較して、Ex-CGでは有意(P<0.05)に高値を示した。
また、各群の腓腹筋中のグリコーゲン量の測定結果を、図11に示した。実施例2の行動体力向上剤を含有する普通食を与えて運動させた群(Ex-CG)では、普通食を与えて運動させた群(Ex-Con)よりも腓腹筋中のグリコーゲン量が増加していた。
以上の結果から、シスチンとグルタミンを含有する本発明の実施例2の行動体力向上剤は、運動により効率的に持久力を向上させ、また、筋肉中のグリコーゲン量を増加させる効果を有することが明らかとなった。
以上、詳述したように、本発明により、行動体力、特に、筋持久力、全身持久力といった行動を持続する能力の向上に好適に用いられる行動体力向上剤を提供することができる。とりわけ、本発明の行動体力向上剤は、筋肉中のグリコーゲン量を増加させ、筋肉中のグリコーゲン量の増加剤として、特にグリコーゲンローディング促進剤として好適に用いられる。さらに、本発明の行動体力向上剤は、運動(トレーニング)による筋肉等におけるグルコースの取り込みの増加効果や、筋肉中のグリコーゲン量の増加効果、ならびに筋持久力、全身持久力を向上させる効果を高めることができる。
また、本発明の行動体力向上剤は、シスチン等の食経験が豊富なアミノ酸を有効成分とすることから、安全性が高く、副作用がほとんどないため、医薬品、食品等の分野において極めて有利である。

Claims (24)
- シスチンを有効成分として含有する、行動体力向上剤。
- 行動を持続する能力の向上剤である、請求項1に記載の向上剤。
- シスチンの含有量が、1回あたりの摂取量または投与量として2.5mg~15gである、請求項1または2に記載の向上剤。
- シスチンの含有量が、全アミノ酸含有量に対して1重量%以上である、請求項1~3のいずれか1項に記載の向上剤。
- 更に、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸を含有する、請求項1~4のいずれか1項に記載の向上剤。
- シスチンの含有量と、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸の含有量との合計が、全アミノ酸含有量に対して1.1重量%以上である、請求項5に記載の向上剤。
- シスチンの含有量(a)と、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸の含有量(b)との重量比[(a):(b)]が1:0.01~100である、請求項5または6に記載の向上剤。
- 経口的に摂取または投与される、請求項1~7のいずれか1項に記載の向上剤。
- 運動開始前に少なくとも1回摂取または投与される、請求項1~8のいずれか1項に記載の向上剤。
- 運動中または運動終了後に少なくとも1回摂取または投与される、請求項1~9のいずれか1項に記載の向上剤。
- 筋持久力の向上剤または全身持久力の向上剤である、請求項1~10のいずれか1項に記載の向上剤。
- 筋肉中のグリコーゲン量の増加剤である、請求項1~10のいずれか1項に記載の向上剤。
- グリコーゲンローディング促進剤である、請求項12に記載の向上剤。
- 請求項1~13のいずれか1項に記載の向上剤を含有する、医薬。
- 請求項1~13のいずれか1項に記載の向上剤を含有する、食品組成物。
- 行動体力の向上に有効な量のシスチンを、行動体力の向上を必要とする対象動物に摂取させることまたは投与することを含む、行動体力の向上方法。
- 行動を持続する能力を向上させる、請求項16に記載の方法。
- シスチンの1回あたりの摂取量または投与量が2.5mg~15gである、請求項16または17に記載の方法。
- 更に、行動体力の向上に有効な量のグルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸を摂取させること、または投与することを含む、請求項16~18のいずれか1項に記載の方法。
- グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸の1回あたりの摂取量または投与量が、10mg~10gである、請求項19に記載の方法。
- シスチンの1回あたりの摂取量または投与量(c)と、グルタミン、セリン、ヒスチジン、アルギニン、バリン、ロイシン、イソロイシン、アラニンおよびプロリンからなる群より選ばれる少なくとも一つのアミノ酸の1回あたりの摂取量または投与量(d)との重量比[(c):(d)]が1:0.01~100である、請求項19または20に記載の方法。
- 経口的に摂取させまたは投与する、請求項16~21のいずれか1項に記載の方法。
- 運動開始前に少なくとも1回摂取させまたは投与する、請求項16~22のいずれか1項に記載の方法。
