Abstract
본 발명은 FCHo1의 활성을 촉진 또는 억제하여 세포 분열을 조절하기 위한 조성물 및 이를 이용한 방법에 관한 것이다. 본 발명의 FCHo1 활성 조절제는 세포 분열에서 중요한 인자로 기능하는 FCHo1을 타깃으로 하여 이의 활성을 촉진 또는 억제시킴으로써, 세포 분열을 촉진하거나 세포 분열을 억제할 수 있으므로 세포 분열과 관련된 질병의 치료에 효과적으로 활용될 수 있다.
Description
본 발명은 FCHo1의 활성을 촉진 또는 억제하여 세포 분열을 조절하기 위한 조성물 및 이를 이용한 방법에 관한 것이다.
세포분열은 생명체의 성장과 자연적인 손실을 보충하기 위해 수행되는 필수적인 과정으로, 생명체 유지에 있어서 매우 중요한 과정이다. 따라서 세포 분열을 매우 정교하고 복잡한 과정을 통해 수행되게 되며 이와 같은 세포 분열에 이상이 발생하면 암과 같은 질병이 발생될 수 있다.
인류가 정복해야 할 중요한 질병중의 하나인 암은 세포분열상의 결함으로 인하여 휴지기 상태에 있어야 할 세포가 이상적으로 증식하여 그 주변의 세포를 파괴하는 특성을 갖는다. 이러한 암은 세포의 성장 및 분화를 조절하는 신호전달체계 (signal transduction pathway) 에 관여하는 여러 인자들의 유전적 변이로 발생한다고 알려져 있다.
진핵세포는 외부에서 세포성장에 대한 신호를 받으면 신호전달체계를 통하여 세포주기에 따라 증식하게 된다. 진핵세포는 DNA 합성 (synthesis phase; S기 라고함) 과 유사분열(mitosis phase; M기 라고 함)의 과정을 주기적으로 거치면서 증식하는데, S기와 M기 사이에는 G1기와 G2기가 있으므로 G1-- S--G2--M기의 주기를 차례로 돌면서 분열한다. 세포분열이 완결된 후에 세포는 다시 G1기로 들어가는데 이 G1 기는 세포내 대사가 가장 활발한 시기로 대부분 의 세포는 많은 시간을 G1 기의 상태로 존재한다. G1 기에서 세포는 디플로이드 (diploid) 즉 2N 상태이고, S기에서 DNA 복제가 일어나 2N이 4N (tetraploid)으로 되며, G 2 기를 거쳐 짧은 M기 동안 4N이 2N으로 분열하고 다시 G1 기로 돌아가게 된다. 정상세포는 G1 기에서 외부로부터 성장 자극 신호가 있으면 S기로 진입하여 G2 기를 거쳐 세포분열을 일으켜 증식하고, 외부신호가 없으면 세포주기의 진행이 중단되어 G1 기에서 멈추어진 G0 기 (휴지기)로 존재한다.
유사분열은 활발히 분열하는 세포에서 다수 관찰되며, 염색질이 응축하여 염색체가 형성되는 동시에 미소관을 골격으로 하는 방추사 또는 분열장치가 생기는 것, 염색체가 방추체 상에서 양극으로 2분하여 핵분열이 완료되는 것을 특징으로 한다. 유사분열은 전기, 중기, 후기로 나뉘며 방추사가 연결된 염색체들이 세포의 적도면에 배열되는 것이 중기, 쌍으로 된 염색체가 세로로 갈라져 둘로 분리되며 방추사에 의해 양극으로 이동하는 것이 후기, 방추사가 소실되고 성상체가 중심체로 되며 핵막과 인이 나타나 두개의 새로운 딸핵을 형성하는 것이 말기에 해당한다.
반면, 암세포는 외부신호와 상관없이 휴지기에 있어야 할 세포들이 세포주기를 따라 DNA합성과 세포분열을 계속하면서 증식한다. 이와 같이 휴지기와 세포분열주기를 결정하기 위해서는 각 기의 세포주기인자들이 세포 내에 존재하여 정확하게 세포주기가 수행되도록 하여야 하며 외부신호를 받아들이는 수용체가 존재하여야 한다. 따라서 휴지기와 세포분열주기의 경계를 결정하는 수용체 및 세포주기인자들의 기능이 왜곡되면 세포가 비정상적으로 성장하게 되어 암세포로 형질 전환되게 된다.
따라서 특히 암세포의 치료에 있어 암세포의 세포분열을 조절하는 것은 매우 흥미로운 주제이나, 아직까지 중앙체 형성의 조절을 통한 암세포 분열의 조절에 관해서는 널리 알려진 바 없다.
본 발명자들은 세포 분열을 조절하고, 더 나아가 암세포 분열의 조절을 통해 새로운 치료제를 개발하기 위한 연구를 수행하던 중, FCHo1 단백질이 세포 분열 중 중앙체(midbody) 형성에 관여하는 중요한 인자이며, 이를 타깃으로 하는 경우 세포 분열을 효과적으로 조절할 수 있음을 발견하고 본 발명을 완성하였다.
따라서 본 발명은 FCHo1 활성 조절제를 포함하는 세포 분열 조절용 조성물 및 이를 이용한 세포 분열능 조절 방법에 관한 것이다.

본 발명은 FCHo1 (FCH domain only 1) 활성 조절제를 포함하는, 세포 분열 조절용 조성물에 관한 것이다.
본 발명의 FCHo1은 세포 분열 동안 위치 특이적인 절단을 통해 중앙체 형성에 관여하는 단백질로, 세포 분열, 특히 유사분열(mitosis) 단계에 있어서 필수적인 역할을 수행함으로, 이를 타깃으로 한 FCHo1 활성 조절제를 통해 세포 분열을 조절하여 세포 분열과 관련된 다양한 질환, 특히 암을 효과적으로 치료할 수 있다.
상기 FCHo1은 인간 FCHo1의 경우 Gene ID: 23149 로 등재되어 있으며, F-BAR(FER/Cip4 homology Bin-Amphiphysin-Rvs) 도메인, 프롤린 풍부 도메인 (proline-rich domain; PRD) 및 MHD(mu-homology domain)로 구성되는 단백질로, 세포 분열 시 세포질 분열의 마지막에 ICB(intracellular cellular bridge)의 중앙에 위치하는 중앙체의 형성에 중요한 역할을 하는 단백질이다.
중앙체(midbody)란, 세포 분열의 마지막 단계에서 두개의 딸 세포를 연결하고 있는 무릎관절 유사 구조를 갖는 것으로 본 발명에 따르면 FCHo1 에 의해서 조절되는 것을 특징으로 한다.
상기 FCHo1은 위치 특이적 절단을 통해 중앙체의 형성을 조절하며, 이와 같은 절단 위치는 PRD 및 MHD 도메인 사이의 위치 및 F-BAR 및 PRD 사이의 위치에서 나타날 수 있으며, 해당 위치는 MMP-9에 의해서 절단되는 것을 특징으로 할 수 있다.
FCHo1의 위치 특이적 절단은 세포 분열의 단계의 진행에 따라 일어나게 되며, 이의 절단된 C-말단 절편은 중앙체에 남아있게 되나, N 말단 절편은 세포 분열의 말기에는 ICB(intracellular cellular bridge)에 위치하는 특성을 나타내어 절단 후 각각의 절편들이 이동하는 양상을 나타낸다.
따라서 본 발명은 상기 FCHo1활성 조절제는 중앙체 형성 조절용인, 세포 분열 조절용 조성물에 관한 것이다.
본 발명에 있어서, FCHo1의 활성 조절제란, FCHo1의 활성을 촉진하거나 억제시킬 수 있는 물질을 말하며, 여기서 FCHo1의 활성이란 'FCHo1이 중앙체 형성, 나아가 세포 분열이 정상적으로 일어날 수 있도록 하는 활성'을 의미한다. 따라서 FCHo1의 활성 촉진제란, FCHo1 가 세포 분열 단계에서 하는 역할을 촉진함으로써, 세포 분열을 촉진할 수 있는 물질을 말한다. 예컨대 FCHo1의 활성 촉진제는 FCHo1의 위치 특이적 절단을 촉진하는 물질 (MMP-9), FCHo1의 적절한 인산화를 촉진하는 물질 (Akt1), FCHo1과 14-3-3ζ과의 결합을 촉진하는 물질로 구성될 수 있으며, FCHo1 특이적 전사 또는 발현을 촉진시킬 수 있는 FCHo1 특이적 프로모터와 같은 당 분야에 알려진 유전자 전사, 발현 조절 물질을 제한없이 포함할 수 있다. 또한 FCHo1 활성 억제제는 FCHo1가 세포 분열 단계에서 하는 역할을 억제 또는 파괴함으로써 세포 분열을 정상적으로 일어날 수 없도록 하는 물질을 말한다. 예컨대 FCHo1의 활성 억제제는 FCHo1의 위치 특이적 절단을 억제하는 물질, FCHo1의 인산화 억제제, 14-3-3ζ 과 FCHo1사이의 결합 억제제일 수 있으며, FCHo1의 변이체 또는 FCHo1의 결실 서열일 수 있다.
특히 FCHo1의 변이체 또는 결실 서열은 FCHo1의 서열 중 MMP-9(matrix metallopeptidase 9)에 의해 절단되는 서열에 변이 또는 결실이 일어난 것으로, 정상적인 FCHo1의 위치 특이적 절단을 불가능하게 함으로써, 세포 분열을 억제할 수 있도록 하는 서열일 수 있고, FCHo1의 서열 중 Akt1에 의한 인산화 부위에 변이 또는 결실이 일어나, Akt1이 FCHo1에 정상적으로 결합하지 못하게 함으로써 FCHo1의 정상적인 절단을 차단하는 서열일 수 있고, FCHo1의 서열 중 14-3-3ζ 과의 결합 부위에 변이 또는 결실이 일어나 결합을 방해함으로써, 중앙체의 형성을 방해하는 서열일 수 있다.
예시적인 FCHo1의 억제제는 이의 단백질 발현, 이의 유전자 발현을 억제하는 유전 공학적 기법에 의한 억제제를 다양하게 포함할 수 있으며, 예컨대 FCHo1유전자의 mRNA에 상보적으로 결합하는 안티센스 뉴클레오티드, 짧은 헤어핀 RNA(small hairpin RNA, shRNA), 작은 간섭 RNA(small interfering RNA, siRNA) 및 리보자임(ribozymee)으로 이루어진 군으로부터 선택된 1종 이상일 수 있으며, FCHo1 단백질에 특이적으로 결합하는 화합물, 펩티드, 펩티드모방체, 기질 유사체, 앱타머, 및 항체로 이루어진 군으로부터 선택된 1종 이상일 수 있다.
따라서 본 발명은 FCHo1의 활성 억제제는 FCHo1의 위치 특이적 절단을 억제하는 것인, 세포 분열 조절용 조성물에 관한 것이다. 또한 본 발명은 상기 FCHo1 활성 억제제는 FCHo1 인산화 억제제인, 세포 분열 조절용 조성물에 관한 것이다.

본 발명에 있어서, 세포 분열이 조절되는 세포는 그 종류가 제한되지 않으나 특별히 유용한 목적을 위하여 세포의 이상 분열과 관련된 질환을 갖는 세포, 특히 바람직하게는 암세포일 수 있다.
또한 본 발명은 FCHo1 활성 조절제를 포함하는 세포 분열 조절제에 관한 것이다.
상기 세포 분열 조절제는 개체에서의 세포 분열을 조절함으로써, 세포 분열과 관련된 질병, 질환의 치료에 유용하게 사용될 수 있다.
상기 세포 분열과 관련된 질환은 세포 증식성 질환, 예컨대 비정상적인 세포 분열을 특징으로 하는 암, 비암성 이상 증식-세포분열에 의한 각종 난치성 질환, 또는 비정상적으로 낮은 수준의 세포 분열에 의한 각종 난치성 질환일 수 있다. 상기 세포증식성 질환은, 이에 한정하는 것은 아니지만, 종양, 악성종양, 혈관증식성질환(blood vessel proliferative disorders), 자가면역질환 및 섬유증질환(fibrotic disorders)을 포함한다.
상기 이상 증식은 일반적인 수준의 세포 증식 수준을 벗어나 부적절하게 높은 수준으로 세포 분열이 일어나는 것을 것을 의미할 수 있다.
또한 본 발명은 FCHo1 활성 조절제를 포함하는 배지 조성물에 관한 것이다.
본 발명에서 용어, '배지(media)'는 체외배양 조건에서 세포의 성장 및 생존을 지지할 수 있게 하는 배양액을 의미하고, 배양에 적절한 당 분야에서 사용되는 통상의 배지를 모두 포함한다. 또한, 세포의 종류에 따라 배지와 배양 조건을 선택할 수 있다. 배양에 사용되는 배지는 바람직하게는 세포 배양 최소 배지(cell culture minimum medium; CCMM)로, 일반적으로 탄소원, 질소원 및 미량원소 성분을 포함한다. 이런 세포 배양 최소 배지에는 예들 들어, DMEM(Dulbecco's Modified Eagle's Medium), MEM(Minimal essential Medium), BME(Basal Medium Eagle), RPMI1640, F-10, F-12, MEM(Minimal essential Medium), GMEM(Glasgow's Minimal essential Medium), Iscove's Modified Dulbecco's Medium 등이 있으나, 반드시 이들로 제한되는 것은 아니다.
또한 본 발명은 세포 내 FCHo1 (FCH domain only 1) 를 검출하는 단계; 를 포함하는 세포 분열능에 대한 정보를 제공하는 방법에 관한 것이다.
본 발명의 방법에 따르면, 세포 내 FCHo1을 검출하고, 이의 활성을 검출함으로써 중앙체의 형성이 정상적으로 이루어지는지 여부 및 이에 따른 세포 분열이 정상적으로 이루어지는지에 관한 세포 분열능에 대한 정보를 효과적으로 제공할 수 있다. 만약 FCHo1에서 비정상적인 변이가 일어나 FCHo1의 위치 특이적 절단이 정상적으로 이루어지지 않는다면, 세포 분열 단계에서 중앙체의 형성이 비정상적으로 이루어질 수 있으며, 이에 따라 세포 분열능이 저해될 수 있다.
따라서 상기 검출은 FCHo1의 절단, FCHo1의 인산화, FCHo1의 중앙체로의 이동 등으로 이루어진 군에서 선택된 1종 이상을 검출하는 것을 포함할 수 있다.
특히 FCHo1은 세포 분열 중 나타나는 중앙체(midbody) 형성에 필수적인 역할을 수행함으로, 상기 세포 분열능에 대한 정보는 중앙체 형성능에 관한 정보일 수 있다.
또한 본 발명은 FCHo1 (FCH domain only 1) 의 활성을 조절하는 단계; 를 포함하는 인간의 제외한 동물 세포의 분열능을 조절하는 방법에 관한 것이다.
본 발명의 FCHo1은 세포 분열 중 중앙체 형성에 필수적이므로, 이의 활성을 조절함으로써, 세포의 분열능을 효과적으로 조절할 수 있다.
상기 조절은 예컨대 FCHo1의 활성을 억제함으로써 중앙체 형성 억제를 유도하여 세포 분열이 정상적으로 일어나지 않도록 조절하는 것을 말하며, FCHo1의 활성 억제는 FCHo1의 활성 억제제를 통해 수행할 수 있다.
상기 동물 세포는 인간을 제외한 동물 세포 또는 인간을 포함하는 동물 세포일 수 있으며, 특히 세포 분열과 관련된 질병, 예컨대 암을 가진 개체로부터 유래된 세포 일 수 있다.
또한 본 발명은 생체 외(in vitro)에서 FCHo1 (FCH domain only 1) 의 활성을 조절하는 단계; 를 포함하는 세포의 분열능을 조절하는 방법에 관한 것이다.
본 발명에 따르면, 개체로부터 분리된 세포를 생체 외에서 FCHo1의 활성을 조절하여, 세포 분열능을 조절할 수 있으며, 이를 세포 분열과 관련된 질병 또는 기전 연구에 유용하게 사용할 수 있다.
또한 본 발명은 FCHo1 (FCH domain only 1) 활성 조절제를 포함하는, 암 예방 또는 치료용 약학적 조성물에 관한 것이다.
다양한 암 조직에서 FCHo1 의 전사 정도를 분석한 EST 결과 FCHo1이 암 조직 중 특히 B-림프아구(B-lymphoblast), 골수, 근육, 난소암 조직에서 높은 전사를 나타내며, 골, 안구, 폐, 췌장, 태반, 피부, 대장, 위, 정소에서도 높은 전사를 나타냄이 보고되어 있다. 따라서 유사분열에 기능하고, 분열이 빠른 세포에 더 많이 존재하는 본 발명의 FCHo1은 각종 암에서 질병 치료를 위한 타깃이 될 수 있으며, 이의 활성을 조절함으로써, 각종 이상 세포 증식에 의한 암을 예방 또는 치료할 수 있다.
상기 암은 세포분열, 유사분열(mitosis)에 이상이 생겨 유발되는 암을 모두 제한없이 포함할 수 있으나, 폐암, 위암, 대장암, 간암, 골암, 췌장암, 피부암, 두부 또는 경부암, 피부 또는 안구 내 흑색종, 자궁암, 난소암, 직장암, 항문부근암, 결장암, 유방암, 나팔관암종, 자궁내막암종, 자궁경부암종, 질암종, 음문암종, 호지킨병, 식도암, 소장암, 내분비선암, 갑상선암, 부갑상선암, 부신암, 연조직 육종, 요도암, 음경암, 전립선암, 만성 또는 급성 백혈병, 림프구 림프종, 방광암, 신장 또는 수뇨관 암, 신장세포 암종, 신장골반 암종, CNS 종양, 1차 CNS 림프종, 척수 종양, 뇌간신경교종 및 뇌하수체 선종과 같은 고형암으로 이루어진 군으로부터 선택된 1종, 바람직하게는 B-림프아구(B-lymphoblast), 골수암, 근육암, 난소암, 골암, 안암, 폐암, 췌장암, 태반암, 피부암, 대장암, 위암 및 정소암으로 이루어진 군에서 선택된 1종 이상일 수 있다.
상기 FCHo1 활성 조절제는 암 세포의 이상 세포 분열을 FCHo1 활성 조절을 통해 적절하게 조절할 수 있으며, 예컨대 FCHo1의 활성을 비정상적인 FCHo1 절단을 유도하거나, FCHo1 변이체 또는 결실 서열의 도입을 통해 억제함으로써, 암세포의 분열을 억제하도록 할 수 있다.
따라서 본 발명은 FCHo1 활성 조절제를 포함하는 암 치료제에 관한 것이다.
FCHo1의 변이체 또는 결실 서열은 FCHo1의 서열 중 MMP-9(matrix metallopeptidase 9)에 의해 절단되는 서열에 변이 또는 결실이 일어난 것으로, 정상적인 FCHo1의 위치 특이적 절단을 불가능하게 함으로써, 세포 분열을 억제할 수 있도록 하는 서열일 수 있고, FCHo1의 서열 중 Akt1에 의한 인산화 부위에 변이 또는 결실이 일어나, Akt1이 FCHo1에 정상적으로 결합하지 못하게 함으로써 FCHo1의 정상적인 절단을 차단하는 서열일 수 있고, FCHo1의 서열 중 FCHo1의 서열 중 14-3-3ζ 과의 결합 부위에 변이 또는 결실이 일어나 결합을 방해함으로써, 중앙체의 형성을 방해하는 서열일 수 있다. 보다 구체적으로 인간 FCHo1 서열을 기준으로 S570 위치를 변이 또는 결실시킨 서열, S155 부위를 변이 또는 결실시킨 서열, FCHo1의 R563-564 부위가 변이 또는 결실된 서열, FCHo1의 407-421aa가 변이 또는 결실된 서열일 수 있다.
상기 FCHo1의 변이체 또는 결실 서열의 도입은 당 분야에 공지된 방법으로 제한없이 도입될 수 있으며, 본 발명의 일 구현예와 같이 적절한 벡터, 예컨대 렌티바이러스 벡터에 FCHo1의 변이체 또는 결실 서열을 도입하여 암을 가진 개체에 도입할 수 있다.
본 발명의 약학적 조성물은 공지의 방법에 따라 다양한 비경구 또는 경구 투여용 형태로 제조될 수 있다. 비경구 투여용 제형의 대표적인 것으로는 에어로졸 제형, 주사용 제형이 있을 수 있다.
경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 상기 유효 성분에 적어도 하나 이상의 부형제, 예를 들면, 전분, 칼슘 카보네이트, 수크로오스 또는 락토오스, 젤라틴 등을 혼합하여 조제된다. 또한, 단순한 부형제 이외에도 마그네슘 스테아레이트, 탈크와 같은 윤활제들도 사용될 수 있다.
경구투여를 위한 액상제제에는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데, 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러가지 부형제, 예를 들면, 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다.
비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 비수성용제, 현탁제로는 프로필렌글리콜, 폴리에틸렌글리콜, 올리브오일과 같은 식물성 오일, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔, 마크로골, 트윈 61, 카카오지, 라우린지, 글리세로젤라틴 등이 사용될 수 있다.
특히 본 발명 약학적 조성물의 바람직한 제형은 에어로졸 전달을 통해 표적 부위에 전달되도록 제제화된 흡입 투여용 제형(에어로졸 제형) 일 수 있다.
흡입을 통한 약물 전달은 기도를 통과하여 폐의 점막을 통하여 직접 폐세포로 약물을 전달하는 비침습적(non-invasive) 방법 중 하나로, 특히 폐 질환의 광범위한 치료에 에어로졸 전달을 통한 핵산 전달이 유리하게 이용될 수 있다. 이는 폐의 해부학적 구조 및 위치가 즉각적이고 비 침습적인 접근을 가능케 하고, 다른 기관에는 영향을 미치지 않으면서 핵산 전달 시스템의 국소 적용을 받을 수 있기 때문이다. 따라서, 본 발명의 FCHo1의 변이체 또는 결실 서열을 적절한 전달체와 결합하여 에어로졸 방식으로 폐 등을 포함하는 병변 부위에 전달하면, 상기 질환에 대한 예방 또는 치료효과를 기대할 수 있다.
본 발명에 있어서, 약학적 조성물은, 본 발명이 목적으로 하는 주 효과를 손상시키지 않는 범위 내에서 바람직하게는 주 효과에 상승 효과를 줄 수 있는 다른 성분 등을 함유할 수 있다.
또한, 본 발명의 약학적 조성물은 투여를 위해서 상기 기재한 유효성분 외에 추가로 약학적으로 허용 가능한 담체, 부형제 및 희석제를 더 포함할 수 있다.
예를 들어 담체, 부형제 및 희석제로는 락토오스, 텍스트로오스, 수크로오스, 소르비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로오스, 메틸셀룰로오스, 미정질 셀룰로오스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트, 광물유로 이루어진 군에서 선택될 수 있다.
본 발명의 약학적 조성물의 유효 투여량은 환자의 나이, 성별 체중에 따라 달라질 수 있으나 0.0001 내지 50mg/kg으로 투여할 수 있으며, 바람직하게는 0.001 내지 20mg/kg으로 투여될 수 있다.
본 발명의 조성 물은 암의 예방 또는 치료를 위하여 단독으로, 또는 수술, 화학적 치료, 방사성 치료, 호르몬 치료, 약물 치료 및 생물학적 반응 조절 제를 사용하는 방법들과 병용하여 사용할 수 있다.
또한 본 발명은 FCHo1 (FCH domain only 1) 단백질 절편을 검출하는 단계;를 포함하는 개체의 암 진단에 대한 정도를 제공하는 방법을 제공한다.
상기 개체는 인간을 포함하는 포유류인 것이 바람직하며, 상기 암 진단에 대한 정보를 제공하는 방법은, FCHo1의 단백질 절편을 검출하여 이를 상호 비교함으로써, FCHo1 단백질 절편이 정상 개체와 비교하여 감소되어 있는 경우, 이를 암을 가진 개체로 진단하는 것을 특징으로 한다.

ELISA는 고체 지지체에 부착된 항체와 항원의 복합체에서 항원을 인지하는 표지된 또 다른 항체를 이용하는 직접적 샌드위치 ELISA, 고체 지지체에 부착된 항체와 항원의 복합체에서 항원을 인지하는 또 다른 항체와 반응시킨 후 이 항체를 인지하는 표지된 2차 항체를 이용하는 간접적 샌드위치 ELISA 등의 다양한 ELISA 방법을 포함한다.
이하, 본 발명을 실시예 및 제제예에 의해 상세히 설명한다. 단, 하기 실시예 및 제제예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예 및 제제예에 의해 한정되는 것은 아니다.
세포 배양 및 형질감염
실험에 사용된 A549 세포는 10% FBS 및 페니실린/스트렙토마이신이 보충된 DMEM/고 글루코오스 배지에서 37℃, 5% CO2 조건으로 배양하였다. 세포들은 Neon® 형질감염 시스템을 이용하여 형질감염되었다.
플라스미드, siRNA 및 화합물
모든 플라스미드 구조들은 In-fusion® HD 클로닝 키트(Clontech® Laboratories) 에 의해서 클로닝하였다. 인간 FCHo1 cDNA 클론, pCMV6-N-3xDDK 벡터 및 pCMV6-C-3xDDK 벡터는 Origene에서 구입하여 사용하였다. 생성된 클론들은 다음과 같다: FCHo1-N-3xDDK, FCHo1-N-3xDDK 돌연변이 (Δ273-294, S295A), FCHo1-C-3xDDK, FCHo1-C-3xDDK 돌연변이 (Δ150-155, Δ480-561, Δ531-561, Δ562-569, Δ562-570, Δ571-579, Δ580-589, Δ563-564, Δ531-589, 407-421), FCHo1-C-3xDDK 이중 돌연변이 (Δ150-155 및 Δ562-570) 부분 FCHo1-C-3xDDK (562-889, 582-889, 1-268), 부분 FCHo1 (1-268)-C-3xDDK 돌연변이 (Δ150-155) 및 부분 FCHo1 (562-889)-C-3xDDK 돌연변이 (R562N, R563N, L564G, S566A, R567N, K568N, V569G, S570A, C571N). FCHo1 siRNA 저항성 클론: siRNA 저항성 FCHo1-C-3xDDK 및 siRNA 저항성 FCHo1-C-3xHA. 하기 RNA 올리고뉴클레오티드의 쌍은 표적 인간 FCHo1에 대하여 합성되었다: AGACCUACUCGAAGGCGAU (dtdt), UCAAGGACGUUCUCCGCUA (dtdt), ACGUGGUGCUGCUGCGAUA (dtdt), UCUCAGUGGAGUACGGCUA (dtdt). 노코다졸(Nocodazole)은 Sigma-Aldrich로부터 구입하였으며, MG132 은 MP Biomedicals에서 구입하여 사용하였다.

Immunofluorescence
)
A549 세포를 8 웰 챔버 커버글라스 또는 고정밀 커버글라스에 위치시키고 3% 파라포름알데하이드로 10분 동안 실온에서 고정시켰으며, 0.5% triton® X-100 를 5분 동안 침투시키고 30분 동안 차단하였다. 이 후 1차 또는 2차 항체와 함께 차단 용액 (0.1% 사포닌 및 3% BSA 를 갖는 PBS)에서 배양하였다. dSTORM(stochastic optical reconstruction microscopy) 이미징을 위하여, Alexa Fluor 647 접합 항-래빗 이차 항체 및 산소 소거 시스템(0.5mg/ml 글루코오스 옥시다아제, 40μg/ml 카탈라아제, 10% 글루코오스, pH 7.4)을 이용하였으며, 이미징 전에 최종 티올 농도 10-200mM를 달성하기 위하여 30mM 머캅토에틸아민(MES; 시그마) 을 첨가하였다. 모든 dSTORM 및 SIM 이미지는 Carl Zeiss ELYRA PS.1 (super-resolution microscopy)에서 얻었으며, 모든 CLSM 이미지는 Carl Zeiss LSM710 (Confocal Laser Scanning Microscope) 에서 얻었다.

세포들은 IP 용해 완충액에서 30분 동안 4℃에서 용해되었다. 17,000 x g에서 30분 동안 원심분리한 후, 상기 단백질의 농도를 측정하고, 용해물의 동일한 양을 면역 침강에 사용하였다. 면역침강은 항-flag M2 affinity gel (Sigma-Aldrich)을 이용하여 4℃에서 하룻밤동안 수행하였다. 침강물은 3회 세척용액으로 세척하고 침강된 단백질들은 SDS-PAGE를 이용하여 분리하였다. 특이항체 및 2차 항-마우스 IgG Veriblot 또는 홀스라다시 페록시다아제가 접합된 항-래빗 항체를 이용하여 웨스턴 블랏을 수행하였다. 결과물은 chemiluminescence detector (Atto Ez-Capture MG)를 이용하여 시각화하였다.

젤라틴 자이모그래피를 0.1% 젤라틴을 함유하는 10% SDS-PAGE 에 대한 면역침강(IP) 에서 수행하였다. IP 시료는 SDS-PAGE 겔에서 수행되었고 2.5% Triton X-100으로 세척하였다. 그 후 50mM TrisCl (pH 7.6), 0.2M NaCl, 5mM CaCl2, 및 0.2% Brij-35 를 함유하는 콜라게나아제 완충액을 이용하여 3일동안 배양하였다. 겔을 Coomassie Brilliant Blue 로 염색하고 탈염하였다.

PLA를 SIM 이미지 단백질-단백질 상호작용에 대하여 고-해상도 형광 현미경을 이용하여 수행하였다. 근접성 라이게이션(Proximity ligation)은 Duolink detection kit를 이용하여 제조사의 매뉴얼에 따라 수행하였다. 모든 PLA 이미지는 Carl Zeiss's ELYRA PS.1로 얻었다.

A549 세포를 2.5% 글루타알데하이드로 1시간동안 pH 7.2-7.4 0.1M 카코딜산(cacodylate) 완충액에서 고정시켰으며, 증류수로 세척하고 0.1M 카코딜산(cacodylate) 완충액에서 0.5% 사산화 오스뮴(osmium tetraoxide) 으로 고정시키고, 다시 증류수로 세척하였다. 그 후 50% 에탄올 내 0.1-0.5% 아세트산 우라닐로 하룻밤동안 En_bloc 염색을 수행한 후, 탈수화 하고 침윤시켰다. FEI's Helios 650 (FIB-FESEM)로 이미지를 얻었다.

A549 세포를 0.15M Hepes 내 4% 파라포름알데하이드 및 0.05% 글루타알데하이드의 혼합물 내에서 고정시키고, 0.15M Hepes 내 4% 파라포름알데하이드 내에서 후-고정시키고 세척하였다. 시료를 차단하고, FCHo1 항체 (584-804aa) 와 함께 배양하였으며, 세척하고 나노금(Nanoprobes, Yaphank, NY, USA)을 갖는 이차 항체와 함께 배양하고 다시 세척하였다. 1% 글루타알데하이드를 이용하여 다시 고정시킨 후 50mM 글리신으로 세척하였으며 이후 1% BSA 로 세척하였다. 다음 시료는 2% 사산화오스뮴(osmium tetraoxide) 및 3% 페로시안화칼륨(potassium ferrocyanide)의 1:1 혼합물로 고정하였고, 탈수시킨 후 침윤시켰다. 이미지들은 JEOL's JEM1010 (80 kV가속전압에서의 TEM)을 이용하여 얻었다.
프로테오믹스 분석을 위한 시료의 준비
RIPA 완충액을 이용하여 세포주들에서 단백질을 추출하였으며 단백질 농도를 bicinchoninic acid assay kit를 이용하여 결정하였다. 단백질 용해물을 FASP (filtered-aided sample preparation) 를 이용하여 소화시켰다. 200μg 의 단백질을 SDT 완충액((4 % SDS, 0.1 M DTT, 및 0.1 M Tris/HCl; pH 7.6)에서 변성시켰으며 그 후 45 분 동안 37℃에서 환원시켰다. 단백질 알킬화를 위하여 실온, 암 조건에서 30분 동안 50mM의 IAA(iodoacetamide) 를 첨가하였다. 시약 등을 제거하기 위하여 시료들을 UA 완충액 (8 M urea, 0. 1 M Tris-HCl; pH 8.5) 을 이용하여 원심분리하였으며, 그 후 UA 완충액을 TEAB 완충액 (100 mM; pH 8.5) 으로 교환하였다. 연속적 구배 트립신 (Promega, USA)을 첨가(100 mM TEAB 내 트립신: 단백질= 1 : 50 [w/w])하고 그 후 12시간동안 37℃에서 배양하였다. 분해된 시료들을 TMT 시약을 이용하여 제조사의 매뉴얼에 따라 표지하였다. 이 단계에서, 두개의 조건에 대한 3개의 시료들을 각각 채널을 이용하여 표지하였다; 대조군(control) 셋트 #1, #2, 및 #3을 126, 128 및 130 채널로, FCHo1 과다발현 셋트 #1, #2, 및 #3를 127, 129, 및 131 채널로 표지하였다. 동일한 양의 표지된 펩타이드를 모은 후 3100 OFFGEL fractionator (Agilent Technologies, USA) 을 이용하여 분획화하였다. 분획 이후, 각각의 분획물은 C18 스핀 컬럼을 이용하여 탈염화하였다.
LC-MS/MS 분석
펩티드 시료를 Q-exactive (Thermo Fisher Scientific, Germany)를 이용한 나노- LC-MS/MS에 의해 분석하였다. 상기 펩타이드 시료들을 나노-LC-MS/MS 에 의해 분석하였다. 상기 펩타이드들을 트랩 컬럼 (PepMap, 2 cm * 75 μm, 3 μm 입자크기, Thermo Scientific) 에 위치시키고, 용매 A (0.1 % 포름산 수용액) 및 용매 B (아세토니트릴 내 0.1 % 포름산) 를 이용하여 0.3μl/min에서 EASY-Spray column (PepMap, 50 cm * 75 μm, 2 μm 입자크기, Thermo Scientific)에 의해 분리하였으며, 그 후 분리 구배 프로그램을 하기와 같이 셋팅하였다: 160 분동안 linear 5-40% B, 2분 동안 linear 40-80% B, 10분 동안 등용매 80%, 2분동안 linear 80-5% B, 및 15분 동안 등용매 5% B. 컬럼의 온도는 60℃로 유지되도록 하였다. 전구체 이온 스캔을 3*106 의 AGC 타깃값, m/z 200에서의 70K 고분해능(mass resolution) 을 갖는 프로필 모드에서 얻었다. MS 스캔에서의 상위 10 전구체 이온들을 orbitrap analyzer로 선택하였다. NCE (normalized collision energy)를 갖는 HCD fragmentation에 의해서 이온 산물들이 생산되었다.
TMT
데이터 분석
인간 데이터베이스(uniprot/swissprot, release-Apr_2014, 89601 entries) 에 대하여 로우 데이터를 SEQUEST® search engine on Proteome Discoverer 1.4 (Thermo Scientific)을 이용하여 검색하였다. 검색은 하기와 같은 파라미터에서 수행되었다; 전구체 이온에 대해서 10ppm 으로 설정된 tolerance 및 프래그먼트 이온에 대해서 0.8Da; 최대 허용가능한 수의 잘못된 절단은 1이며; 리신의 TMT 변형 및 아미노산 말단의 자유 아민 그리고 시스테인의 카르바비도메틸레이션(carbamidomethylation)이 고정 변형으로 설정되었다; 메티오닌의 산화는 다양한 변형으로 설정되었다. 펩타이드 및 단백질 동정을 입증하기 위하여, Scaffold Q+ (version 4.3.3, Proteome Software Inc., USA)를 이용한 Peptide Prophet 및 Protein Prophet algorithms을 적용하였다. 펩타이드 가능성(peptide probability)의 역치는 >95% 이고, 단백질 가능성의 역치는 >99% 로 사용되었으며, 동정 단백질을 만약 그들이 2개 이상의 특이적 펩타이드에서 나타난다면, 그것은 정확한 것으로 간주하였다. TMT 리포터 이온의 피크 강도는 Scaffold Q+를 이용한 탠덤 질량 스펙트럼에서 추출하였다. 연속적으로, 다량의 단백질의 상대적인 비율 및 모든 p-값(sample p-value, ratio p-value)을 Isobar (R software package)를 이용한 두개의 군 (126/127, 128/129, 130/131) 사이에서 계산하였다. FCHo1 과다 발현 세포에서 시료 p-값 및 비율 p-값(ratio p-value) 이 모두 <0.05 인 단백질을 선택하였다. GO(Gene ontology) 분석을 DAVID 바이오인포매틱스 리소스를 이용하여 유의적인 단백질에서 수행하였다.

FCHo1
절단 위치의 확인
세포질 분열의 마지막에 ICB (intracellular cellular bridge) 의 중앙에 위치하는 중앙체 내부에는 FCHo1이 존재하며, FCHo1은 핵산 coat nucleator 로 조립되는 클라드린 표면에서 초기 워킹 단백질과 연관된 것으로 알려져있다. FCHo1은 도 1에 나타낸 바와 같이 F-BAR(FER/Cip4 homology Bin-Amphiphysin-Rvs) 도메인, 프롤린 풍부 도메인 (proline-rich domain; PRD) 및 MHD(mu-homology domain)로 구성된다. F-BAR 도메인은 초승달 형태의 역평행 다이머 구조를 형성하고 이 것은 PtdIns(4,5)P2-enriched membranes 에 결합한다. 도 1의 서열 하단의 밑줄은 다양한 FCHo1 항체에 의해 인식되는 위치에 해당되는 서열이며, 서열 내의 수직으로 표시된 부위는 잠재적인 FCHo1의 절단 부위를 나타낸다.
FCHo1의 부위 특이적 절단이 중앙체의 형성을 조절하는지 여부를 확인하기 위하여, 먼저 FCHo1의 절단 부위를 확인하였다. A549 세포 내에서 FCHo1 N-말단이 3xFLAG와 융합된 전장 길이의 야생형, Δ273-294, 또는 S295A 로 형질감염 세포로부터 항-FLAG 항체를 이용하여 면역블랏 분석을 수행하였으며, A549 세포 내에서 C-말단3xFLAG 태그 융합을 갖는 전장 야생형 또는 FCHo1 단백질의 부위 특이적 결실(Δ480-561, Δ531-561, Δ562-569, Δ562-570, 또는 Δ571-579) 의 형질감염 후 대표적인 항-FLAG 항체를 이용한 면역블랏 분석을 수행하였다. 면역블랏 분석결과를 도 2 및 도 3 에 나타내었다.
도 2에 나타낸 바와 같이, Δ273-294 에서 밴드가 사라진 것이 뚜렷하게 확인되었다. 또한 도 3에서는 Δ562-569, Δ562-570 에서 밴드가 사라지는 것을 확인하였다. 이에 따라, FCHo1 은 PRD 및 MHD 도메인 사이의 위치 및 F-BAR및 PRD 사이의 위치에서도 절단되는 것을 확인하였다. 즉 추정되는 절단 위치에 의해 나뉘는 FCHo1의 부위는 F-BAR 도메인 (F-BAR), PRD (프롤린-풍부 도메인(proline-rich domain)), mu 상동 도메인 (mu homology domain; MHD)이다.

중앙체에
특이적으로 위치하는
FCHo1
단백질 절편확인
절단된 FCHo1 단밸질 절편이 다른 서브 세포성 위치를 갖는지 여부를 확인하기 위하여 FCHo1 에 대한 다른 위치를 표적화하는 4개의 항체 (92-242aa, 285-364aa, 468-517aa, 또는 584-804aa) 를 사용하였고, CLSM(confocal laser scanning microscopy)을 이용하여 FCHo1의 현미경 이미지를 얻었다. Mitosis-synchronized A549세포를 4개의 다른 FCHo1 항체 및 Hoechst 33342 (DNA 염색)을 이용하여 염색하였다. 결과를 도 4에 나타내었다.
도 4에 나타낸 바와 같이, α584-804는 세포 분열의 마지막 단계에서 두개의 딸세포를 연결하는 무릎 관절-유사 구조와 같은 중앙체를 염색시킨 반면, 다른 FCHo1 항체(α92-242 및 α468-517)는 ICB를 염색시켰다. 즉 FCHo1은 위치 특이적 절단이 이루어지며 절단 부위 중 584-804aa 를 포함하는 절편들이 중앙체에 특이적으로 위치하는 절편임을 확인하였다.

3.
FCHo1을
절단하는 단백질 분해 효소의 확인
특이적 단백질 분해 효소가 FCHo1을 절단하는지 여부를 확인하기 위하여, 단백질 분해효소 억제제인 1 μM 의 MG132를 16시간 동안 처리하였다. MG132 분해효소 억제제의 존재 또는 부존재 하에서 A549 세포 추출물의 면역블랏을 수행하였으며, 그 결과를 도 5에 나타내었다.
도 5에 나타낸 바와 같이, MG132 처리군은 완벽하게 FCHo1 절단 형태가 사라지도록 하였으며, 이는 FCHo1의 절단이 특이적 단백질 분해효소에 의해 영향을 받는다는 것을 의미한다. 그 후 FCHo1 아미노산 서열을 PROSPER(https://prosper.erc.monash.edu.au)를 이용하여 분석하였다. 인상적이게도 분석 결과는 MMP-9 (matrix metallopeptidase 9) 가 Arg 563(R563) 위치에서 FCHo1을 절단할 수 있음을 예측하였다 (도 6). 이러한 결과에 입각하여 MMP-9이 FCHo1의 R563 위치에 결합하고 활성을 나타내는지 여부를 확인하기 위하여, 야생형(wt), 또는 563-564 또는 531-589aa 가 결실된 FCHo1 (Δ563-564 또는 Δ531-589) 에 대하여 유사분열 시 FLAG 항체를 이용한 면역침강을 수행하였다. 그후 MMP-9에 대하여 자이모그래피(zymography)를 수행하였으며 그 결과를 도 7에 나타내었다. 또한 중앙체 형성에서 FCHo1(584-804) 및 MMP-9의 PLA(proximity ligation assay) 를 수행하였으며 그 결과를 도 8에 나타내었다.
도 7에 나타낸 바와 같이, 자이모그래피(Zymography) 는 FCHo1 의 결실 돌연변이 Δ563-564 및 Δ531-589 에서 MMP-9의 활성이 야생형과 비교하여 억제되어 나타난다는 것을 입증한다. 또한 도 8에 나타낸 바와 같이 중앙체에서 FCHo1 및 MMP-9는 서로 분명하게 상호작용함을 확인하였다. 상기와 같은 결과는 FCHo1의 R563 위치가 MMP-9 단백질 분해효소에 의해 절단됨을 나타낸다.

4. 세포
분열 단계에 따른
FCHo1
및 이의 절편의 위치 확인
절단된 FCHo1 절편의 위치를 중앙체 형성 중간에 확인하기 위해서, 두개의 FCHo1 항체들 (α468-517 또는 α584-804)을 R563 위치에서 FCHo1의 절단된 N 말단(NT) 또는 C 말단(CT) 을 관찰하기 위하여 사용하였다. 중앙체 형성과 관련된 위치를 확인하기 위한 목적으로 유사분열의 후기 및 중앙체가 형성된 이후인 말기에서 FCHo1 절편의 위치를 확인하였다. SIM(Structured-illumination) 이미지를 세포 분열의 말기에 면역염색을 통해 얻었다. 또한 세포 분열의 말기 및 후기로 진행하며 각각의 절편이 위치 및 이동을 확인하기 위하여 siRNA 저항성 FCHo1 구조체인 이소성의 Δ563-564 단백질을 발현시키고 이소성의 FCHo1 에서 α468-517 또는 α584-804에 의한 FCHo1 절편의 위치를 확인하였다. 이와 같은 결과를 도 9에 나타내었다.
도 9에 나타낸 바와 같이, 유사분열의 후기 단계에서, α468-517 및 α584-804는 모두 중심 방추사를 염색시켰다, 반면 중앙체가 형성된 이후인 말기에서는 α584-804가 중앙체를 염색하는 반면, α468-517는 ICB를 염색시켰다. 흥미롭게도, α468-517 는 정상 FCHo1을 갖는 세포의 후기에서는 ICB를 염색시켰음에도 이소성 Δ563-564 발현의 말기에서는 중앙체 영역을 염색시켰다. 그러나, α584-804 의 경우, 정상 FCHo1과 동일하게 이소성의 FCHo1 Δ563-564에도 변화없이 중앙체에 남아있었다. 이와 같은 결과는 FCHo1이 말기 동안에는 중심방추사에 위치하지만 세포 분열이 진행되면서 FCHo1 의 R563가 MMP-9에 의하여 중앙체 형성 동안에 절단되며, 이에 따라 CT 절편은 중앙체에 남아있게 되는 반면, NT 절편은 ICB 에 위치한다는 점을 보여준다.

중앙체
특이적 위치 및 세포분열 진행에 따른 위치 확인
FCHo1이 위치하고 있는 중앙체의 나노 구조를 분석하기 위하여, FESEM (Field emission scanning electron microscopy) 및 TEM 분석을 수행하였고 그 결과를 도 10에 나타내었다. TEM에서 FCHo1 항체(α584-804) 가 면역-골드 라벨링을 통한 비히클 내 FCHo1 존재 검출을 위해 사용되었다. 또한 FCHo1 항체 (α584-804) 또는 칼모듈린 1을 이용한 SRM(Super-resolution microscopy)을 수행하여, FCHo1의 위치를 추가적으로 확인하였으며, 세포 분열 동안 대표적인 CLSM 이미지를 도 11에 나타내었다. A549세포는 FCHo1 항체(α584-804), 칼모듈린 항체, 및 Hoechst 33342 (DNA 염색)을 통해 염색되었다.
도 10에 나타낸 바와 같이, 전자 응축 구조인 중앙체는 어둡게 염색되었으며, 중앙체 및 ICB에 비히클이 풍부하게 존재한다는 것을 나타내었다(a, b). 또한 α584-804-골드는 중앙체의 경계에서 다수 염색되었으며(c, Post-embedding), 비히클의 세포막을 염색하였다(d, Pre-embedding). SRM(Super-resolution microscopy) 은 또한 FCHo1이 중앙체의 경계에 모인다는 것을 보여준다(e).
도 11에 나타낸 바와 같이, 항체 α584-804에 의해 검출되는 FCHo1은 세포 분열이 진행되는 동안 중앙체가 형성되는 동안 강하게 나타나고 세포질 분열이 일어나 절단이 되는 순간까지 중앙체에 지속적으로 존재한다는 것을 확인하였다.

기질 모티프 확인 및 절단 특성
도 12에 나타낸 바와 같이, FCHo1 내부에는 Ser155 (S155) 및 Ser570 (S570) 와 같은 2개의 Akt1 결합 모티프가 존재하며 이는 마우스 FCHo1에서 보존 부위인 것으로 나타났다. Akt1이 s570의 RxRxxS 모티프에 결합하는지 여부를 확인하기 위하여, 내생적 Akt1 의 공동-면역침강법을 항-3xFLAG-융합된 부분적 FCHo1 구조체(562-889 또는 582-889) 발현에 대한 FLAG 항체를 이용하여 수행하였으며, 추가적으로 FCHo1의 150-155 결실 돌연변이(F-BAR Δ150-155)에 대한 공동 면역침강법을 이용하여 Akt1 의 결합 부위를 확인하였다. 그 결과를 도 13에 나타내었다.
도 13에 나타낸 바와 같이, 562-889 는 Akt1에 결합하였으나, RxRxxS 가 결실된 582-889는 Akt1에 결합하지 않았고, 이는 FCHo1의 565-570aa 부위 (RSRKVS)가 Akt1 기질의 모티프에 해당됨을 나타낸다(a). 또한 F-BAR Δ150-155를 이용한 공동-면역침강 결과 Akt1이 F-BAR 도메인 (1-268aa) 에 결합한다는 것을 확인하였고, 이의 결실 F-BAR Δ150-155에서도 Akt1 결합이 약하게 관찰됨으로써, F-BAR 도메인의 Akt1 결합 모티프가 제거될지라도 Akt1 은 F-BAR Δ150-155와 어느정도는 상호작용을 하며, 이는 F-BAR 도메인이 호모다이머를 가지고 있음을 나타낸다(b). FCHo1 의 Akt1 인산화 부위 및 중앙체 및 ICB에서의 위치를 도 14 에 나타내었다.
FCHo1이 자가 결합하는지 여부를 확인하기 위하여, C-말단 HA-융합 또는 FLAG-융합 FCHo1 (FCHo1-HA 또는 FCHo1-FLAG), FCHo1 siRNA 를 공동 형질감염시키고, FCHo1-FLAG에 대한 항-FLAG 항체를 이용하여 FCHo1-HA의 co-IP를 수행하였으며, 결과를 도 15에 나타내었다.
도 15에 나타낸 바와 같이, 흥미롭게도 FCHo1-FLAG 는 전장 FCHo1-HA와 결합한 반면, FCHo1-FLAG는 FCHo1-1HA의 CT-절편(564-889aa)과 결합하지 않았다. 그러므로 이러한 결과는 F-BAR 도메인에 관련된 NT 절편이 FCHo1의 이량체 또는 다량체화에 중요하다는 것을 보여준다.
항-FLAG 항체를 이용한 Akt1 의 공동-면역 침강을 수행하였다. A549 세포가 전장-FCHo1, 절단된75kDa CT 절편, 절단된 38kDa CT 절편으로 형질감염되었으며, 분석 결과를 도 16에 나타내었다.
도 16에 나타낸 바와 같이, 38kDa-CT 절편은 간기 동안에 풍부하게 관찰되었으나, 세포 분열(mitosis) 동안에는 약하게 검출되고 새로운 절단된 75kDa CT 절편이 세포 분열 동안에 더욱 분명하게 나타나는 것을 확인하였다. 도면 내 붉은 별표는 75kDa의 C-말단 절편(F-BAR 절단)이 검출되지 않음을 나타낸다.
이러한 결과는 세포분열 동안에 R563의 절단이 간기 동안에 더 적게 발생한다는 것 및 75kDa CT-절편 밴드가 관찰된다는 것을 나타낸다. Glu269 (E269)에서 FCHo1이 절단되었다면, 3xFLAG (3kDa) 를 포함하는 CT 절편은 약 74kDa이다. 상기 74kDa는 F-BAR 도메인이 없는 269-889-3xFLAG 의 75kDa 밴드와 유사한 것으로 확인하였으며, 이는 F-BAR 도메인이 유사분열 동안에 절단됨을 나타낸다.

중앙체에서의
위치 및 상호작용
면역형광법 및 PLA 분석법에 의해, F-BAR 도메인이 ICB 내 및 원형질막의 Akt1 에 결합하는지 여부 및 FCHo1 및 Akt1의 중앙체에서의 상호작용을 확인하였다. 중앙체에서 FCHo1 및 Akt1 의 SIM 이미지를 확인하였으며, 중앙체를 확대하여 관찰하였다. FCHo1 및 Akt1의 중앙체에서의 상호작용은 PLA를 통해 확인하였으며, 칼모듈린1을 ICB 마커로 사용하였다. 결과를 도 17에 나타내었다.
도 17에 나타낸 바와 같이, α-192-242는 ICB를 염색하는 반면, α-584-804는 중앙체에서 염색되는 것으로 나타났다. 즉 Akt1 과 F-BAR가 함께 위치하고 있으나, CT-절편은 중앙체에서 Akt1과 결합한다는 것이 확인되었다. 이러한 결과는 F-BAR 및 CT 절편이 다른 세포 위치, 즉 ICB 또는 중앙체에 위치하여 작용한다는 점을 나타내며 Akt1은 ICB 또는 중앙체에 위치하는 두 개의 F-Bar 또는 CT 절편의 각각의 Akt1 모티프에 각각 결합할 수 있음을 확인하였다. 그러므로, F-BAR 도메인(1-268)이 FCHo1 (1-889) 로부터 절단되고 F-BAR(1-268) 가 유사분열 동안에 ICB에 위치할 것으로 생각하였다. F-BAR 도메인은 유사분열 동안의 PM(Pleckstrin homology) 내의 Akt1과 관련이 있을 것으로 보이며, 기능은 세포내 분열 경로와 관련이 있을 것을 생각된다.

돌연변이에 의한
FCHo1의
절단 억제
FCHo1의 R563 절단 부위를 발견하였고, 이것은 FCHo1의 Akt1 결합 모티프(565-570aa)과 인접하게 위치하고 있다. S570 에서 FCHo1의 인산화를 파괴하기 위하여 FCHo1 S 570A 돌연변이 구조체를 제조하였다. 이와 같은 S570A 돌연변이 구조체에 대한 SIM 이미지는 FCHo1 항체 (α468-517)로 중앙체에서 염색하여 확인하였다. A549 세포는 야생형 FCHo1 또는 FCHo1 돌연변이(S570A)에 대하여 관찰하였다. 결과를 도 18에 나타내었다.
도 18에 나타낸 바와 같이, S570A 돌연변이는 FCHo1 항체의 염색을 ICB에서 중앙체로 변경시켰다. R563에서 절단된 FCHo1의 NT 절편을 검출하였다. 이러한 변화는 이전 실시예 3 에서 확인된 Δ563-564가 563R에서 FCHo1의 절단을 억제하는 것과 유사하다. 그러므로 FCHo1의 R563 부위에서의 절단은 Akt1 의존적인 것으로 생각된다.

의 상호 결합 및
중앙체
형성에 대한 영향 확인
14-3-3ζ는 MKLP1 kinesin-6 및 CYK4 RhoGAP로 구성된 헤테로테트라머이며, 이들은 각각 인간 MKLP1 및 RACGAP1의 상동분자로 알려져 있다. 중앙 방추사는 중앙체의 형성 및 유지를 위해 기능한다. 14-3-3ζ 는 중앙체 유래 RACGAP1/MKLP1 복합체의 분산 단백질이다.
FCHo1의 상호작용 파트너가 14-3-3ζ 인지 여부를 확인하기 위하여 항-FLAG항체를 이용하여 공동-면역침강법을 수행하였다. A549 세포는 전장 FCHo1, 절단된 FCHo1, 14-3-3ζ로 형질감염되었다. 형질감염된 세포들은 유사분열 또는 간기에서 확인하였으며, 결과를 도 19에 나타내었다.
도 19에 나타낸 바와 같이, FCHo1와 14-3-3ζ 의 결합이 확인되었으며, 이러한 결합은 간기보다 유사분열(mitosis) 단계에서 증가하였다. 또한 PLA 분석을 통해 중앙체에서 FCHo1에 14-3-3ζ이 결합한다는 것을 입증하였다.
FCHo1에 대한 14-3-3ζ의 추정적 결합 부위인 407-421aa를 검증하기 위하여, FCHo1 Δ407-421 구조를 제조하고 항-FLAG 항체를 이용한 14-3-3ζ 및 Akt1의 공동-면역 침강을 수행하였다. 형질감염 세포를 유사분열 시기에 관찰하였다. 또한 PLA 분석을 통해 중앙체에서 FCHo1(584-804)와 14-3-3ζ의 상호작용을 확인하였다. 결과를 도 20에 나타내었다.
도 20에 나타낸 바와 같이, Δ407-421는 FCHo1 에 대한 14-3-3ζ 의 결합을 억제하였다(a). 또한 PLA 실험결과는 중앙체에서 FCHo1(584-804)와 14-3-3ζ가 결합함을 나타냈다(b).
또한 도 21에 나타낸 바와 같이 Δ407-421 은 유사분열에서 중앙체의 형성을 강력하게 파괴하였다. 이는 14-3-3ζ이 FCHo1의 407-421aa와 결합하고 이 결합은 중앙체 형성에 필수적이라는 것을 나타낸다.
야생형 FCHo1 또는 FCHo1 Δ563-564 발현 A549 세포의 중앙체 이미지의 SIM 이미지를 추가적으로 확인하였다. 세포를 FCHo1 (α584-804), 14-3-3ζ, MKLP1, 및 RACGAP1 항체로 염색하였다. 중앙체 영역을 확대하여 관찰하고 그 결과를 도 22에 나타내었다.
도 22에 나타낸 바와 같이, 중앙체에서 α14-3-3 ζ 가 Δ563-564을 FCHo1보다 더 강하게 염색시킨다는 것을 확인하였다. 또한 Δ563-564 는 중앙체의 MKLP1/RACGAP1의 안정한 클러스터링을 격리시켰다. 이러한 결과는 R563 부위에서의 FCHo1 절단의 손상이 중앙체에서 14-3-3ζ의 축적을 유발하며 14-3-3ζ 의 축적이 MKLP1/RACGAP1 복합체의 비정상적인 클러스터링을 막도록 이끈다는 것을 나타낸다.

과발현에 의한
중앙체
연관 단백질 발현 변화
FCHo1의 과다발현에 의하여 뚜렷하게 상향 또는 하향으로 조절되는 단백질을 질량 분석을 통해 분석하였다. FCHo1을 과다발현시킨 A549 세포에서 세포 주기-연관 단백질 군의 변화를 프로테오믹스 분석을 통해 확인하였으며, 결과를 도 23에 나타내었다.
도 23에 나타낸 바와 같이, FCHo1의 과다발현은 세포주기 연관 단백질 군을 하향 발현되도록 하며, 반대로 미토콘드리아 전자전달-관련 단백질 군의 단백질을 상향 조절 시킨다는 것을 보여준다(a). 모든 중앙체 위치 단백질은 FCHo1의 과다발현에 의해 조절되었다(b). 이를 통해 FCHo1이 중앙체 연관 단백질을 조절한다는 것을 확인하였다.
동기화 방법(synchronization method)을 이용한 분석에서 세포들은 중앙 방추사 또는 중앙체 형태 분석을 위하여 MKLP1 항체로 염색되었다. 후기 (중앙 방추사) 및 말기(중앙체) 의 세포의 비율은 CLSM 이미지를 통해 정량화 하였다: FCHo1 siRNA (n=318), wt FCHo1 (n=702), FCHo1 Δ563-564 (n=756), FCHo1 Δ531-589 (n=1952). 결과를 도 24에 나타내었다.
도 24에 나타낸 바와 같이, Δ563-564 단백질은 야생형 FCHo1 단백질과 비교하여 중앙체의 형성을 억제하였다.
상기와 같은 결과를 토대로 FCHo1은 새로운 중앙체 조절자임을 새롭게 확인하였다. FCHo1은 단백질 분해 효소에 의해 위치 특이적으로 절단되며, 위치 특이적 절단의 단편들은 중앙체 또는 ICB로 다르게 이동한다. 키나아제로 알려진 Akt1은 FCHo1을 S155 및 S570에서 인산화시킨다. S570에서 인산화는 FCHo1을 절단하기 위해 중요하며, S570의 인산화는 R563 절단 의존적인 Akt1 에 의해 인산화된다. R563-564 결실 돌연변이는 중앙체 형성이 억제되고, 중심방추사(centralspindlin)의 구성요소로 알려진 RACGAP1 및 MKLP1을 불안정하게 형성한다. FCHo1의 절단은 중앙체에서 14-3-3ζ 의 축적을 막도록 유도한다. 이것은 MKLP1/RACGAP1 복합체의 안정성을 유지시키며 이를 통해 FCHo1은 중앙체 형성을 조절한다. 407-421aa 결실 돌연변이는 중앙체 형성을 폐치하였으며, 이는 14-3-3ζ와 FCHo1의 결합이 중앙체 형성에 필수적임을 의미한다. 중앙체 단백질 FCHo1은 중앙체 기능에 있어서 필수적이며, FCHo1의 위치 특이적 절단의 조절을 통해 중앙체 형성을 효과적으로 조절할 수 있다.

조절을 통한 항암 효과 확인
11.1

결실 및
돌연변이형에
의한 종양 크기, 부피 감소 확인
FCHo1의 전장, 돌연변이 형태 및 결실 형태에 대한 렌티바이러스 벡터를 제작하고, 비침습적인 에어로졸 방식으로 폐암 모델 마우스에 전달한 후 이의 항암 효과를 확인하였다. 실시예 8에서 인간세포에서 FCHo1의 절단에 관여하는 부위가 FCHo1S570A 및 FCHo1del563
-564임 확인하였으므로, 마우스 FCHo1 에서 대응 위치를 확인하였으며, S570A 는 S554A, del563-564 는 del544-545에 해당됨을 확인하였다. 이를 토대로 렌티바이러스 벡터를 제작하였으며 9주령 K-ras 암컷 마우스에 4주간 총 8회를 흡입 노출을 하였을 때 항암 효과를 비교하였고, 결과를 도 25에 나타내었다.
도 25에 나타낸 바와 같이 총 종양의 수는 FCHo1S554A, FCHo1del544
-545에서 유의성 있게 감소하였다 (n=7). 또한 종양 부피를 관찰한 결과, FCHo1S554A은 FCHo1wt과 비교하여 유의성 있는 부피의 감소를 나타냈으며, FCHo1del544
-545 역시 부피가 감소되었음을 확인하였다. 이러한 결과를 토대로 FCHo1 의 돌연변이 및 결실형태의 경우 중앙체 형성의 억제 등을 통해 종양이 억제될 수 있음을 알 수 있다.

FCHo1
절단억제에 따른 병리분석
H&E 슬라이드를 제작하고, 아데노마(adenoma)와 과형성(hyperplasia)이 감소되는지 여부를 확인하였다. 결과를 표 1에 나타내었다. FCHo1 절단 억제를 위하여 shFCHo1을 이용하였으며 이의 서열은 도 26에 기초하여 서열번호 1 내지 4에 나타내었고, 이 중 서열번호 2의 shmFCHo1이 가장 FCHo1 억제 효과가 크므로 이를 이용하여 실험을 수행하였다.

표 1에 나타낸 바와 같이, FCHo1의 결실형 및 돌연변이 군에서는 대조군과 비교하여 유의성있는 수준의 아데노마 및 과형성의 억제가 관찰되었다. 이를 통해 FCHo1의 정상적인 절단 억제가 세포 분열에 이상을 유발하고 이를 통해 분화가 억제 되어서 항암 효과가 나타날 수 있음을 확인하였다.
11.3. 폐암 조직에서
FCHo1
절단 감소 확인
인간시료의 정상 조직 및 1,2,3기 폐암 조직에서 FCHo1 절단의 변화를 확인하였다. FCHo1의 발현양은 FCHo1의 MHD 도메인 부위를 일부 포함하는 584-804aa를 타깃으로 검출하는 항체를 이용하여 검출하였다. 결과를 도 27에 나타내었다.
도 27에 나타낸 바와 같이, 폐암 조직에서는 정상 폐 조직과 비교하여 FCHo1의 25kDa 이하의 절편이 눈에 띄게 감소되거나 사라지는 것을 확인하였다. 이 절편을 질량분석기로 분석하여 FCHo1의 642-665aa를 포함하고 있는 결과를 얻었다. 이는 Arg563이 절단되어 생긴 것이다. 이러한 결과는 폐암조직이 유사 분열 시기에 있기 때문임을 반영하며, FCHo1의 절편이 정상 개체와 비교하여 보이지 않거나 적은 것은 이용하여 FCHo1 의 절편을 암의 마커로 사용할 수 있음을 의미한다.

결합 사이트를 이용한
펩타이드
항암 치료제 개발
Akt1의 모티프를 가진 FCHo1 아미노산 서열 유래의 펩타이드들과 Akt1과의 결합 친화도를 확인하기 위해 펩타이드들에 바이오틴을 접합하여 합성하였다. 스트랩타비딘이 접합된 플레이트에 펩타이드들을 반응시켜 붙인뒤 세포의 lysate를 처리한 후 Akt1 항체로 펩타이드와 Akt1의 결합 친화도를 확인하였다. FCHo1 결합 사이트에서 유래된 각 펩타이드에 따른 in vitro 수준의 Akt1 결합 친화도를 확인한 결과 및 과정을 도 28에 나타내었다.
도 28에 나타낸 바와 같이, FCHo1의 562-571번 (RRLRSRKVSC)과 FCHo1의 560-571번 (PPRRLRSRKVSC) 펩타이드들에서 Akt1 결합친화도가 크게 상승했다. 이들은 Akt1 바인딩 모티프를 포함하고 있다. 펩타이드 치료제 개발의 목적은 Akt1, MMP9, 14-3-3에 대한 FCHo1 결합사이트 유래 짧은 펩타이드들로 하여금 FCHo1 결합파트너에 경쟁하는 안타고니스트 (Antagonist)를 발굴하는 것이다. 따라서 이들은 FCHo1의 절단을 억제하거나 인산화를 억제하거나 결합파트너의 결합을 억제할 것으로 예상되어 펩타이드 항암 치료제 개발에 사용될 수 있을 것으로 예상되었다.

펩타이드의
세포내
흡수 확인
바이오틴이 접합된 펩타이드들을 세포에 처리하고 세포를 고정시켜 스트랩타비딘-488 형광다이를 반응시킨후 CLSM으로 관찰하였으며 그 결과를 도 29에 나타내었다.
도 29에 나타낸 바와 같이 560-571번 (PPRRLRSRKVSC) 펩타이드는 농도 의존적으로 세포내 흡수가 증가하는 것을 확인하였다. 펩타이드 치료제의 한계점은 펩타이드의 극성 (chrage), 친수성과 같은 펩타이드의 성질에 의해 세포내 흡수가 잘 되지 않을 수 있어 캐리어를 쓰거나 Myristic acid를 펩타이드에 접합시켜야 한다는 것이다. 그러나 560-571 펩타이드의 경우 펩타이드 자체가 세포에 흡수가 잘 되기 때문에 펩타이드 치료제의 한계를 극복할 수 있는 장점이 있다. 이는 별도의 CPP(Cell Penetrating Peptide- 세포 투과 펩타이드) 와 캐리어의 접합을 요구하지 않고 펩타이드 자체가 세포막을 투과할 수 있다는 것을 보여주는 것이며, 효율성 높은 치료제로서 가치가 있음을 나타낸다.

의한 세포 생장률 변화 확인
세포생존률을 실시간으로 관찰하기 위해 Xcelligence를 사용하여 측정하였으며, 펩타이드들은 10μM로 처리되었다. 560-567, 562-571, 560-571 펩타이드에서 세포생장의 억제 효과를 확인하였으며 그 결과를 도 30에 나타내었다.
도 30에 나타낸 바와 같이, 560-567, 562-571, 560-571 펩타이드 모두 세포 생장의 억제 효과를 나타냈으며 특히 560-571 (PPRRLRSRKVSC) 펩타이드는 Akt1과의 결합친화력과 세포내 흡수력이 동시에 좋은 결과를 반영하듯 세포 생장 억제 효과가 우수한 것을 확인하였다. 이에 따라 FCHo1의 Akt1 결합모티프와 14-3-3 결합사이트는 펩타이드 항암제 및 케미칼의 개발을 위한 좋은 타겟사이트가 될 것으로 확인하였다.

- 제19항에 있어서 상기 FCHo1 활성 조절제는 암세포의 세포 분열을 억제하는 것인, 암 예방 또는 치료용 약학적 조성물.
- FCHo1 (FCH domain only 1) 활성 조절제를 포함하는, 암 치료제.
- FCHo1 (FCH domain only 1) 단백질 절편을 검출하는 단계;를 포함하는 개체의 암 진단에 대한 정도를 제공하는 방법.
- FCHo1 (FCH domain only 1) 단백질 절편 검출 제제를 포함하는 암 진단용 조성물.
- 제24항에 있어서, 상기 FCHo1 단백질 절편 검출 제제는 FCHo1 단백질 절편 특이적 항체인, 암 진단용 조성물.
- 제24항에 있어서, 상기 FCHo1 단백질 절편은 Arg 563 위치가 절단되어 형성된 것인, 암 진단용 조성물.
