Abstract
본 발명은 페록시레독신 1 단백질의 활성 억제제를 유효성분으로 함유하는 골질환의 예방 또는 치료용 약학적 조성물에 관한 것으로, 구체적으로 본 발명의 페록시레독신 1 단백질은 전구 파골세포의 분화를 유도하고 페록시레독신 1 단백질에 대한 항체가 리포폴리사카라이드에 의해 유도된 파골세포의 증가 및 골손실 동물모델에서의 골손실을 억제함으로써, 상기 페록시레독신 1 단백질의 활성 억제제는 골질환의 예방 또는 치료에 유용하게 활용될 수 있다.
Description
본 발명은 페록시레독신 1 단백질의 활성 억제제를 유효성분으로 함유하는 골질환의 예방 또는 치료용 약학적 조성물에 관한 것이다.
대표적인 대사성 골질환인 골다공증(osteoporosis)은 골 조직의 석회가 감소되어 뼈의 치밀질이 엷어지고 그로 인해 골수강(骨髓腔)이 넓어지게 되는 질환으로, 증세가 진전됨에 따라 뼈가 약해지기 때문에 작은 충격에도 골절되기가 쉽다.
골다공증은 골량의 감소와 미세구조의 이상과 같은 특징을 갖는다. 이는 낡은 뼈의 흡수와 새로운 뼈의 형성 사이에 균형이 무너져 새로운 뼈의 대체(bone remodeling)가 원활히 이루어지지 않아 뼈가 엉성해지고, 부러지거나 부서질 위험성을 증가시킨다. 이러한 골량은 유전적 요인, 영양 섭취, 호르몬의 변화, 운동 및 생활 습관의 차이 등 여러 가지 요인들에 의해 영향을 받는다. 골량감소의 주요 원인으로는 노령, 운동 부족, 저체중, 흡연, 저칼슘 식이, 폐경, 난소 절제 또는 염증 발생 등이 있다.
개인차는 있으나, 일반적으로 골량은 14 내지 18세에 가장 높고 노후에는 1년에 약 1%씩 감소한다. 특히, 여성의 경우 30세 이후부터 골 감소가 지속적으로 진행되며, 폐경기에 이르면 호르몬 변화에 의해 골 감소가 급격히 진행된다. 즉, 폐경기는 체내 에스트로겐 농도의 감소에 의해 파골세포의 활성을 증가시키므로 결국 골량이 감소하게 된다.
이와 같이, 골다공증은 정도에 차이는 있으나 노년층, 특히 폐경기 이후의 여성에게 있어서는 피할 수 없는 증상으로, 선진국에서는 인구가 노령화됨에 따라 골다공증 및 그 치료제에 대한 관심이 점차 증가하고 있다. 또한, 전 세계적으로 골질환 치료와 관련되어 약 1300억 달러의 시장이 형성되어 있으며, 앞으로 더 증가할 것으로 예상되기 때문에 세계적인 연구기관과 제약회사에서는 골질환 치료제 개발에 많은 투자를 하고 있다.
국내에서도 근래에 평균수명이 80세에 육박하면서 골다공증 유병률이 급격하게 증가하고 있다. 최근 지역 주민을 대상으로 실시된 연구에 의하면 전국 인구로 표준화 하였을 경우 남성의 4.5%, 여성의 19.8%가 골다공증 환자임이 보고되었다. 이는 골다공증이 당뇨병이나 심혈관계 질환보다 더 흔한 질환이며, 골절로 인해 받는 환자들의 고통이나 치료를 위해 들어가는 비용을 추정할 때 골다공증은 매우 중요한 보건 문제임을 시사한다.
현재까지 개발된 골다공증 치료제 중 가장 많이 사용되는 에스트로겐은 그 실제적인 효능이 아직 검증되지 않았고 생애 동안 계속 복용해야 하며, 장기간 투여하는 경우 유방암이나 자궁암이 증가하는 부작용이 있다. 또한, 알렌드로네이트(alrendronate)도 그 효능이 명확하지 않고 소화관에서의 흡수가 더디며 위장과 식도점막에 염증을 유발하는 문제가 있다. 한편, 칼슘제제는 부작용이 적으면서도 효과가 우수하나 치료제라기보다는 예방제에 해당한다. 그 외에도 칼시토닌과 같은 비타민 D 제제는 아직 효능 및 부작용에 대한 연구가 충분하지 않다. 이에, 부작용이 적고 효과가 우수한 새로운 골질환 치료제가 요구되고 있는 실정이다.
파골세포는 척추동물의 뼈가 성장하는 과정에서 불필요하게 된 뼈조직을 파괴 또는 흡수하는 대형의 다핵세포로써, 파골세포 전구세포(osteoclast precursor)로부터 분화된다. 파골세포 전구세포들은 M-CSF 및 RANKL 존재 하에서 전구 파골세포(preosteoclast, pOC)를 거쳐 단핵 파골세포로 분화되며, 융합을 통해 다핵 파골세포(multinucleated osteoclast)를 형성한다. 파골세포는 αvβ3 인테그린(integrin) 등을 통해 골에 결합하여 산성 환경을 조성하는 한편, 각종 콜라게네이즈(collagenase) 및 프로테아제(protease)를 분비하여 골 흡수(bone resorption)를 일으키는데, 이러한 파골세포의 억제는 골질환 치료의 효과적인 방법이 될 수 있다.
한편, 페록시레독신 1 단백질(peroxiredoxin 1, PRX1)은 자연계에 널리 존재하는 항산화 효소인 페록시레독신 단백질의 한 종류이다. PRX1은 사이토카인에 의해 생기는 과산화물(peroxide)의 농도를 조절함으로써 포유동물의 세포 내에서의 신호전달과정을 매개한다. 이는 세포질에 존재하는 형태와 세포 밖으로 분비되는 수용성 형태의 두 가지 종류로 존재한다. 골 항상성과 관련하여, 에스트로겐 결핍에 의해 산화적 스트레스(oxidative stress)가 발생하면 조골세포의 세포질에 존재하는 PRX1의 발현이 증가됨으로써 산화적 손상(oxidative damage)을 낮춤(Du J et al., Sci Rep. 2016; 27; 6:35995)이 알려진 바 있으나, 분비되는 수용성 형태의 PRX1의 기전은 알려진 바가 없다.
이에 본 발명자들은, 파골세포의 활성을 조절하여 골질환을 치료하는 물질을 탐색하던 중, 분비된 페록시레독신 1 단백질이 파골세포의 분화를 촉진함을 발견하였고, 이의 활성 억제제가 파골세포의 분화 및 골손실을 억제시키는 것을 확인함으로써 본 발명을 완성하였다.
본 발명의 목적은 페록시레독신 1 단백질의 활성 억제제를 유효성분으로 함유하는 골질환의 예방, 개선 또는 치료용 조성물을 제공하는 것이다.

본 발명은 페록시레독신 1 단백질의 활성 억제제를 유효성분으로 함유하는 골질환의 예방 또는 치료용 약학적 조성물을 제공한다.

상기 폴리펩티드는 단백질의 기능에 영향을 미치지 않는 범위 내에서, 아미노산 잔기의 결실, 삽입, 치환 또는 이들의 조합에 의해서 상이한 서열을 가지는 아미노산의 변이체 또는 단편일 수 있다. 분자의 활성을 전체적으로 변경시키지 않는 단백질 또는 펩티드에서의 아미노산 교환은 당해 분야에 공지되어 있다. 경우에 따라서는 인산화(phosphorylation), 황화(sulfation), 아크릴화(acrylation), 당화(glycosylation), 메틸화(methylation) 또는 파네실화(farnesylation) 등으로 수식(modification)될 수 있다. 따라서, 본 발명은 상기 서열번호 1 또는 서열번호 2로 기재되는 아미노산 서열을 갖는 폴리펩티드와 실질적으로 동일한 아미노산 서열을 갖는 폴리펩티드, 및 이의 변이체 또는 단편을 포함할 수 있다. 상기 실질적으로 동일한 폴리펩티드는 본 발명의 폴리펩티드와 80% 이상, 구체적으로 90% 이상, 더욱 구체적으로 95% 이상으로 상동성을 가질 수 있다.
상기 활성 억제제는 페록시레독신 1 단백질에 상보적으로 결합하는 화합물, 펩티드, 펩티드 미메틱스 및 항체로 구성된 군에서 선택되는 어느 하나 이상일 수 있다.
상기 펩티드 및 펩티드 미메틱스(Peptide Mimetics)는 페록시레독신 1 단백질이 다른 단백질과 결합하는 것을 억제함으로써 페록시레독신 1 단백질의 활성을 억제하는 것일 수 있다. 펩티드는 단백질의 기능에 영향을 미치지 않는 범위 내에서, 아미노산 잔기의 결실, 삽입, 치환 또는 이들의 조합에 의해서 상이한 서열을 가지는 아미노산의 변이체 또는 단편일 수 있다. 비가수분해성 펩티드 미메틱스는 주요 잔기로서 β-턴 디펩티드 코어, 케토-메틸렌 슈도펩티드류, 아제핀, 벤조디아제핀, β-아미노알콜 또는 치환 감마락탐환을 사용하여 제조할 수 있다.
상기 항체는 단일클론항체, 다클론항체 또는 재조합항체일 수 있다. 특정 단백질에 대한 항체는 단백질의 서열이 공지되어 있다면 당업계에 잘 알려진 기술을 이용하여 용이하게 제조할 수 있다. 상기 단일클론 항체는 당업계에 공지된 하이브리도마 방법, 또는 파지 항체 라이브러리 기술을 이용하여 제조될 수 있다. 일반적으로, 단일클론항체를 분비하는 하이브리도마 세포는 항원 단백질을 주사한 마우스와 같이 면역학적으로 적합한 숙주 동물로부터 분리된 면역 세포와 암 세포주를 융합하여 만들 수 있다. 이런 두 집단의 세포 융합은 폴리에틸렌글리콜과 같이 본 발명이 속하는 기술 분야에 공지되어 있는 방법을 이용하여 융합시키고 항체 생산 세포를 표준적인 배양 방법에 의해 증식시킨다. 한계 희석법을 이용하여 서브 클로닝을 실시하고, 균일한 세포 집단을 수득한 뒤 항원에 특이적인 항체를 생산할 수 있는 하이브리도마 세포를 시험관 또는 생체 내에서 대량으로 배양하여 제조할 수 있다. 상기 방법으로 제조된 항체는 겔 전기영동, 투석, 염 침전, 이온교환 크로마토그래피, 친화성 크로마토그래피 등의 방법을 이용하여 분리 및 정제될 수 있다.
상기 다클론 항체는 당업계에 알려진 방법에 따라 면역원인 바이오마커 단백질 또는 그 단편을 외부 숙주에 주사함으로써 제조될 수 있다. 외부 숙주는 마우스, 래트, 양, 토끼와 같은 포유동물일 수 있다. 상기 면역원이 근내, 복강내 또는 피하 주사 방법으로 주사되는 경우, 항원성을 증가시키기 위한 보조제(adjuvant)와 함께 투여될 수 있다. 이후, 외부 숙주로부터 정기적으로 혈액을 채취하여 향상된 역가 및 항원에 대한 특이성을 보이는 혈청을 수득하고 이로부터 항체를 분리 및 정제하여 제조될 수 있다.
본 명세서의 항체는 또한, 중쇄 또는 경쇄를 포함할 수 있다. 경쇄 및 중쇄 각각은 N-말단에 가변 영역을 가질 수 있고, 그리고 각각의 가변 영역은 번갈아 4개의 프레임워크 구역(FR) 및 세 개의 상보성 결정 영역(CDR)을 포함할 수 있다. 통상적으로, 가변 영역의 잔기는 Kabat 등에 의해 고안된 시스템에 따라 번호가 부여된다. 상기 시스템은 미국의 보건 사회 복지부(US Department of Health and Human Services) NIH의 면역학적 관심 단백질의 서열(Sequences of Proteins of Immunological Interest, 1987년, Kabat) 등에 기재되어 있다.
또한, 본 발명의 항체는 Fab, F(ab'), F(ab')2 및 Fv 등의 항체 변이체를 구성으로 포함할 수 있다. 이중 Fab는 경쇄 및 중쇄의 가변영역과 경쇄의 불변영역 및 중쇄의 첫 번째 불변영역(CH1)을 가지는 구조로 1개의 항원 결합 부위를 가진다. Fab'는 중쇄 CH1 도메인의 C-말단에 하나 이상의 시스테인 잔기를 포함하는 힌지 영역(hinge region)을 가진다는 점에서 Fab와 차이가 있다. F(ab')2 항체는 Fab'의 힌지 영역의 시스테인 잔기가 디설파이드 결합을 이루면서 생성된다. Fv는 중쇄 가변영역 및 경쇄 가변영역만을 가지고 있는 최소의 항체조각으로, 이를 생성하는 재조합 기술은 PCT 국제 공개특허출원 WO 88/10649, WO 88/106630, WO 88/07085, WO 88/07086 및 WO 88/09344에 개시되어 있다. 이중쇄 Fv(two-chain Fv)는 비공유 결합으로 중쇄 가변영역과 경쇄 가변영역이 연결되어 있고 단쇄 Fv(single-chain Fv)는 일반적으로 펩타이드 링커를 통하여 중쇄의 가변영역과 단쇄의 가변영역이 공유결합으로 연결되거나 또는 C-말단에서 바로 연결되어 있어서 이중쇄 Fv와 같이 다이머와 같은 구조를 이룰 수 있다. 이러한 항체는 단백질 가수분해 효소를 이용해서 얻을 수 있고(예를 들어, 전체 항체를 파파인으로 제한 절단하면 Fab를 얻을 수 있고 펩신으로 절단하면 F(ab')2를 얻을 수 있음), 바람직하게는 유전자 재조합 기술을 통하여 제작할 수 있다.
또한, 상기 골질환은 골절, 골다공증(osteoprosis), 류마티스성 관절염(rheumatoid arthritis), 파제트병(Paget disease) 또는 전이성 골암(metastatic bone cancer)일 수 있다.
본 발명의 구체적인 실시예에서, 본 발명자들은 전구 파골세포에 RANKL를 처리하면 분비되는 PRX1의 양이 증가하고, PRX1을 첨가하면 이의 농도의존적으로 파골세포의 분화가 촉진됨을 확인하였다(도 1 및 2 참조). 또한, 전구 파골세포에 PRX1을 처리하였을 때 NFATc1의 발현과, IκB의 분해 및 p38의 인산화 정도가 증가함으로써(도 3 내지 5 참조), PRX1에 의한 파골세포 분화 관련 신호전달 경로를 확인하였다.
또한, 동물모델에 PRX1을 투여하면, 파골세포 수가 증가하며 골 손실이 유발되고(도 9 참조), PRX1에 대한 항체에 의해 RANKL에 의해 유도되는 파골세포의 분화가 농도 의존적으로 억제되었으며, 동물 모델에서의 염증 유도성 파골세포 분화 및 이로 인한 골 손실이 감소됨을 육안으로 확인하였다(도 10 및 11 참조).
나아가, PRX1 항체는 꼬리 매달기를 통해 골손실이 유발된 동물모델에서 골손실을 억제하고(도 13 참조), 골손실에 의해 감소된 골 미네랄 밀도, 조직 부피에 대한 골 부피, 섬유주 두께 및 섬유주 갯수와 같은 지표를 증가시키는 한편, 골손실에 의해 증가된 섬유주 분리 및 구조 모델 색인과 같은 지표를 감소시켰다(도 14a 내지 14c 참조).
따라서, 페록시레독신 1 단백질의 활성을 억제하는 물질은 골질환의 예방 또는 치료에 유용하게 사용될 수 있다.
상기 약학적 조성물은 약학적 조성물 전체 중량에 대하여 유효성분인 본 발명에 따른 페록시레독신 1 단백질 활성 억제제를 10 내지 95 중량%로 포함할 수 있다. 또한, 본 발명의 약학적 조성물은 상기 유효성분 외에 추가로 동일 또는 유사한 기능을 나타내는 유효성분을 1종 이상 추가로 함유할 수 있다.
본 발명의 조성물은 또한 생물학적 제제에 통상적으로 사용되는 담체, 희석제, 부형제 또는 둘 이상의 이들의 조합을 포함할 수 있다. 약제학적으로 허용 가능한 담체는 조성물을 생체 내에 전달하는데 적합한 것이면 특별히 제한되지 않으며, 예로서, Merck Index, 13th ed., Merck & Co. Inc. 에 기재된 화합물, 식염수, 멸균수, 링거액, 완충 식염수, 덱스트로스 용액, 말토 덱스트린 용액, 글리세롤, 에탄올 또는 이들 성분 중 1 성분 이상을 혼합한 것일 수 있다. 이때, 필요에 따라 항산화제, 완충액, 정균제 등 다른 통상의 첨가제를 첨가할 수 있다.
상기 조성물을 제제화할 경우, 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 제조된다.
본 발명의 조성물은 경구제제 또는 비경구제제로 제형화 될 수 있다. 경구 투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제, 트로키제 등이 포함되며, 이러한 고형 제제는 하나 이상의 조성물에 적어도 하나 이상의 부형제, 예를 들면, 전분, 탄산칼슘, 수크로스, 락토오스 및 젤라틴 등을 섞어 조제될 수 있다. 또한, 마그네슘 스티레이트, 탈크 같은 윤활제들도 첨가될 수 있다. 한편, 액상 제제로는 현탁제, 내용액제, 유제 또는 시럽제 등이 해당되는데, 여기에는 습윤제, 감미제, 방향제, 보존제 등과 같은 부형제가 포함될 수 있다.
비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁용제, 유제 등의 주사제가 포함될 수 있다.
비수성용제, 현탁용제로는 프로필렌글리콜, 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다.
또한, 본 발명은 페록시레독신 1 단백질의 활성 억제제를 유효성분으로 함유하는 골질환의 개선용 건강기능식품을 제공한다.
상기 페록시레독신 1 단백질 활성 억제제는 상술한 바와 같은 특징을 가질 수 있다. 일례로, 상기 페록시레독신 1 단백질은 서열번호 1 또는 서열번호 2로 기재되는 아미노산 서열로 구성되는 폴리펩티드일 수 있다. 또한, 상기 활성 억제제는 페록시레독신 1 단백질에 상보적으로 결합하는 화합물, 펩티드, 펩티드 미메틱스 및 항체로 구성된 군에서 선택되는 어느 하나 이상일 수 있다.
상기 골질환은 골절, 골다공증(osteoprosis), 류마티스성 관절염(rheumatoid arthritis), 파제트병(Paget disease) 또는 전이성 골암(metastatic bone cancer)일 수 있다.
본 발명의 페록시레독신 1 단백질 활성 억제제는 식품에 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용될 수 있다. 이때, 첨가되는 유효성분의 함량은 목적에 따라 결정될 수 있다. 일반적으로, 건강기능식품 중의 함량은 전체 식품 중량의 0.01 내지 90 중량부일 수 있다.
또한, 상기 건강기능식품의 형태 및 종류에는 특별한 제한은 없다. 상기 물질을 첨가할 수 있는 건강기능식품의 형태는 정제, 캅셀, 분말, 과립, 액상 및 환 등일 수 있다.
본 발명의 건강기능식품은 통상의 건강기능식품과 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상술한 천연 탄수화물은 포도당, 과당과 같은 모노사카라이드, 말토스, 슈크로스와 같은 디사카라이드, 덱스트린, 사이클로덱스트린과 같은 폴리사카라이드, 자일리톨, 소르비톨, 에리트리톨 등의 당알코올이다. 감미제로서는 타우마틴, 스테비아 추출물과 같은 천연 감미제나, 사카린, 아스파르탐과 같은 합성 감미제 등을 사용할 수 있다.
상기 외에 본 발명의 건강기능식품은 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올 등을 함유할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용 될 수 있다. 이러한 첨가제의 비율은 본 발명의 조성물 100 중량부당 0.01 내지 0.1 중량부의 범위에서 선택될 수 있다.
아울러, 본 발명은 1) 골세포 또는 골조직에 피검물질을 처리하는 단계; 2) 상기 단계 1)의 골세포 또는 골조직으로부터 페록시레독신 1 단백질의 분비량을 측정하는 단계; 및 3) 상기 단계 2)의 페록시레독신 1 단백질의 분비량이 무처리 대조군에 비해 감소된 피검물질을 선별하는 단계를 포함하는 골질환의 예방 또는 치료용 물질 스크리닝 방법을 제공한다.

단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예에 의해서 한정되는 것은 아니다.
<실시예 1> 마우스 골수세포의 배양
먼저, 마우스의 골수세포를 얻기 위해, ICR 마우스(6 내지 9주령, 수컷)를 경추 탈골 후 70% 에탄올로 소독한 뒤, 경골 부분의 피부를 절개하여 부착 근육을 떼어냈다. 그리고 경골 원심부를 절단하고 슬개골을 탈골시켜 경골을 적출하였다. 뼈 양끝을 조금 잘라 한 쪽 끝에 25 G의 주사 바늘을 꽃고 α-MEM을 흘려주어 얻은 골수세포를 시험관에 모았다. 이를 원심 분리한 후, α-MEM에 현탁시켜 2배의 Gey’용액(Sigma)을 가한 뒤 적혈구를 제거하였다. 이를 다시 원심 분리하여 10%의 FBS가 함유된 α-MEM으로 재현탁하고 37℃, 5%의 CO2 조건에서 배양함으로써, 마우스의 골수세포를 준비하였다.
<실시예 2> 전구 파골세포의 배양
상기 <실시예 1>에 준비된 골수세포에 10 ng/ml의 대식세포증식자극인자(macrophage colony stimulating factor, M-CSF)를 첨가하여 하루 동안 배양하였다. 부유된 세포를 분리하고 30 ng/ml의 M-CSF를 첨가한 뒤 3일간 배양하여 전구 파골세포(Bone marrow Macrophages, BMM)를 생성하였다. 200 ng/ml의 RANKL(receptor activator of NF-κB ligand, 파골세포분화인자)를 첨가한 뒤 1일간 추가 배양하여 생성된 전구 파골세포(preosteoclast, pOC)를 수득하였다. 수득된 세포는 37℃, 5%의 CO2 조건에서 배양하여 사용하였다.
<실험예 1> 전구 파골세포에서 PRX1 분비여부 확인
상기 <실시예 2>에서 수득된 전구 파골세포에서 PRX1이 분비되는지 여부를 다음과 같은 방법으로 확인하였다.
구체적으로, 1×105 cells/well의 전구 파골세포에 200 ng/ml의 RANKL 및 30 ng/ml의 M-CSF를 첨가하여 48시간 동안 배양한 후, 배지 중의 분비 단백질을 다음과 같이 웨스턴 블롯팅 실험으로 분석하였다.
먼저, 회수한 배지의 단백질을 얻기 위해, 원심분리를 하여 상층액만을 회수하고 단백질의 농도를 BSA(bovine serum albumin)로 표준화하여 단백질 어세이 키트(Bio-Rad)를 사용하여 정량하였다. 20 ng의 단백질을 SDS-PAGE(poly acrylamide gel electrophoresis)로 변성 분리하고, 이를 80 V에서 약 1시간 45분 동안 PVDF 멤브레인으로 이전시켰다. PVDF 멤브레인을 5%의 탈지유(skim milk)가 함유된 PBST 용액(0.05% Tween-20 in PBS)으로 블로킹하고, 1차 항체로서 항-PRX1(1:1000, Ab Frontier) 및 항-β-actin(Sigma)항체와 각각 반응시켰다. 반응 후 PVDF 멤브레인을 PBST로 5회 세척하고 HRP(hoseradish peroxidase)가 결합된 2차 항체와 반응시킨 후 ECL Advance (Amersham. co.)로 발색시켜 분석하였다.
그 결과, 도 1에 나타난 바와 같이, 전구 파골세포에 RANKL를 첨가하면 PRX1의 양이 유의하게 증가하였다(도 1).
<실험예 2> 전구 파골세포에서 PRX1 분화유도 확인
다음으로, PRX1의 첨가가 파골세포의 분화에 미치는 영향을 다음과 같은 방법으로 확인하였다.
구체적으로, 1×105 cells/well의 전구 파골세포에 1 μg/ml의 PRX1(Ab Frontier)과 30 ng/ml의 M-CSF를 첨가하여 2일간 배양하였다. 배양된 세포를 10%의 포르말린으로 10분간 고정한 후 에탄올-아세톤 혼합용액(1:1)으로 1분간 재고정하여 TRAP(tartrate-resistant acid phosphatase)으로 염색하였다. 염색된 세포를 현미경으로 관찰하여 3개 이상의 핵을 가진 TRAP+ 세포를 분화된 다핵(multinucleated, MNC) 파골세포로 판정하여 계수하였다.
그 결과, 도 2에 나타난 바와 같이, 처리된 PRX1의 농도의존적으로 파골세포의 분화가 촉진되었다(도 2).
<실험예 3> PRX1에 의한 파골세포 분화 신호전달 경로 확인
<3-1> PRX1에 의한 NFATc1의 발현 확인
RANKL의 자극은 전구 파골세포에서 파골세포로의 분화를 조절하는 핵심 전사인자로 알려져 있는 NFATc1(nuclear factor of activated T cells c1)의 발현을 유도한다. NFATc1은 RANKL 자극에 의한 세포 내 칼슘 신호 전달에 의해 초기에 유도되며, 이후 NF-κB 및 MAPK(MAP kinase) 등의 활성화가 다시 NFATc1의 자가증폭(autoamplification)을 유발하여 활성을 지속시킨다.
상기 <실험예 1>에서 PRX1이 파골세포의 분화를 촉진함을 확인하였으므로, PRX1이 파골세포의 분화를 조절하는 핵심 전사인자인 NFATc1의 발현에 미치는 영향을 알아보고자, 다음과 같이 웨스턴 블롯팅 실험을 수행하였다.
구체적으로, 전구 파골세포를 1 μg/ml의 PRX1과 30 ng/ml의 M-CSF를 첨가하여 2일간 배양하였다. 배양된 세포를 회수하여 용해 버퍼(lysis buffer)를 첨가하고, 세포를 용해시켰다. 이를 원심분리하여 상층액을 회수하고, 이로부터 단백질의 농도를 BSA로 표준화하여 단백질 분석 키트(Bio-Rad)를 사용하여 정량하였다. 웨스턴 블롯팅 과정은 1차 항체로서 항-NFATc1(1:200, Santa cruz)을 사용한 것을 제외하고, 상기 <실험예 1>과 동일한 방법으로 수행하였다.
그 결과, 도 3에서 나타낸 바와 같이, 전구 파골세포에 PRX1을 처리하면 NFATc1의 발현이 증가하였다(도 3). 이로부터 PRX1이 NFATc1의 발현 증가를 통해 파골세포의 분화를 촉진함을 알 수 있었다.
<3-2> PRX1에 의한 NF-κB 및 p38 MAPK의 활성화 확인
다음으로, 좀 더 구체적인 신호전달 경로를 알아보기 위해, PRX1에 의한 NF-κB 및 p38 MAPK의 활성화 정도를 웨스턴 블롯팅 실험으로 확인하였다.
이때, NF-κB의 활성화는 NF-κB의 저해분자인 IκB의 분해 정도로, p38의 활성화는 p38에 대한 인산화 증가 정도로 확인하였다. 실험은 항체로서 항-IκB(1:1000, Cell signaling) 및 항 p-p38(1:1000, Cell signaling)을 사용한 것을 제외하고, 상기 <실험예 1>과 동일한 방법으로 수행하였다.
그 결과, 도 4 및 5에서 나타난 바와 같이, 전구 파골세포에 PRX1을 처리하면 IκB의 분해 및 p38의 인산화의 정도가 증가하였다(도 4 및 5). 이는 PRX1에 의해 NF-κB 및 p38 MAPK 각각의 신호전달 경로가 활성화됨을 의미한다. 따라서, 이로부터 PRX1은 NF-κB 및 p38 MAP kinase의 활성화를 통해 NFATc1의 발현을 증가시킴으로써 파골세포의 분화를 촉진함을 알 수 있었다.

실험예
4>
PRX1에
의한 성숙한 다핵 파골세포의 생존 조절 및 신호전달 경로 관여 확인
<4-1> PRX1에 의한 성숙한 다핵 파골세포의 생존 조절 확인
골 흡수에는 파골세포의 분화뿐만 아니라, 성숙한 다핵 파골세포의 생존기간 또한 중요한 영향을 미친다. 이와 관련하여 RANKL이 파골세포 분화를 촉진하는 기능과 함께 파골세포의 생존을 촉진함은 이미 잘 알려져 있다. 이에, PRX1이 성숙한 다핵 파골세포의 생존에 미치는 영향을 다음과 같이 확인하였다.
구체적으로, 1×105 cells/well의 전구 파골세포에 200 ng/ml의 RANKL 및 30 ng/ml의 M-CSF를 첨가하고, 이를 3일 동안 배양하여 성숙한 다핵 파골세포를 얻었다. 대조군에는 30 ng/ml의 M-CSF(veh), 실험군에는 200 ng/ml의 RANKL 및 30 ng/ml의 M-CSF(RANKL) 또는 1 μg/ml의 PRX1 및 30 ng/ml의 M-CSF(PRX1)를 처리 한 후 2일간 추가로 배양하였다. 배양된 세포를 10%의 포르말린으로 10분간 고정한 후 에탄올-아세톤 혼합용액(1:1)으로 1분간 재고정하여 TRAP(tartrate-resistant acid phosphatase)으로 염색하였다. 염색된 세포를 현미경으로 관찰하여 3개 이상의 핵을 가진 TRAP+ 세포를 분화된 다핵(multinucleated, MNC) 파골세포로 판정하여 계수하였다. 다핵 파골세포 생존율은 최초 다핵 파골세포 수 대비 살아남은 대조군 또는 실험군의 다핵 파골세포수의 퍼센트(%)로 표시하였다.
그 결과, 도 6에 나타난 바와 같이, 대조군의 성숙한 다핵 파골세포는 생존율이 낮았으나, RANKL을 첨가한 군은 성숙한 다핵 파골세포의 생존율이 높았다. 또한, PRX1을 추가 첨가한 실험군은 RANKL을 처리한 군보다 더 높은 성숙한 다핵 파골세포의 생존율을 보였다.
이로부터, PRX1가 성숙한 다핵 파골세포의 생존을 더욱 촉진함을 알 수 있다(도 6).
<4-2> PRX1의 신호전달 경로 관여 확인
성숙한 다핵 파골세포의 생존에 관여하는 신호전달 경로로서 NF-κB 및 AKT의 메커니즘 등이 알려져 있다. 상기 <실험예 4-1>에서 확인된 PRX1의 성숙한 다핵 파골세포 생존 촉진이 하위의 신호전달 경로를 조절하는지를 웨스턴 블롯팅 실험으로 확인하였다.
항체로서 항-IκB(1:1000, Cell signaling) 및 항-p-AKT(1:1000, Cell signaling)을 사용하는 것을 제외하고, 상기 <실험예 1>과 동일한 방법으로 웨스턴 블롯팅 실험을 수행하였다.
그 결과, 도 7 및 8에서 나타난 바와 같이, 성숙한 다핵 파골세포에 PRX1를 첨가하면 IκB의 분해 및 p-AKT의 인산화가 증가하였다(도 7 및 8). 이는 NF-κB와 AKT 신호전달 경로가 활성화됨을 의미하며, 이를 통해, 분비 PRX1이 NF-κB와 AKT의 활성화 과정을 거쳐 성숙한 다핵 파골세포의 생존을 촉진함을 알 수 있다.
<실험예 5> 동물모델에서 PRX1의 파골세포 분화 효과 확인
시험관 내에서 확인한 PRX1의 파골세포 분화 촉진 효과가 동물모델에서도 동일하게 나타나는지를 다음과 같은 방법으로 확인하였다.
먼저, 11 내지 12주령의 수컷 ICR 마우스(Samtako)의 두개관(頭蓋冠)에 0.25 mg의 PRX1을 피하 주사하였고, 대조군에는 동량의 PBS를 주사하였다. 6일 뒤 마우스를 CO2 처리하여 희생시키고 두개관을 채취한 후 이를 4%의 파라포름알데히드에 넣어 고정시켰다. 고정된 두개관을 TRAP 염색하고 이를 육안으로 관찰하였다.
그 결과, 도 9에 나타난 바와 같이, PRX1을 주사한 마우스의 두개관에서TRAP+인 파골세포 수가 크게 증가하고 골 손실이 유발되었다(도 9). 이로부터, 동물모델에서도 PRX1이 파골세포의 분화를 효과적으로 촉진하는 것을 확인하였다.
<실험예 6> PRX1 항체의 파골세포 분화 억제 효과 확인
<6-1> 전구 파골세포에서의 파골세포 분화 억제 효과 확인
PRX1에 대한 항체에 의해, 전구 파골세포의 분화가 억제되는지를 다음과 같은 방법으로 확인하였다.
실험은 1×105 cells/well의 전구 파골세포에 200 ng/ml의 RANKL 및 30 ng/ml의 M-CSF를 첨가하고 여기에 1 μg/ml 또는 5 μg/ml의 PRX1의 항체(Ab Frontier)를 추가로 첨가한 것을 제외하고는 상기 <실험예 2>와 동일한 방법으로 수행하였다.
그 결과, 도 10에 나타난 바와 같이, RANKL의 첨가는 전구 파골세포의 분화를 촉진하였으나, PRX1에 대한 항체를 첨가함으로써 파골세포의 분화가 농도의존적으로 억제되었다(도 10).
<6-2> 동물모델에서의 파골세포 분화 억제 효과 확인
PRX1에 대한 항체의 파골세포 분화억제 효과를 동물모델을 사용하여 다음과 같은 방법으로 확인하였다. 임상학적으로 관절염 또는 치주염 등의 환자에서 염증 반응에 따른 파골세포 증가로 빈번한 골 손실이 관찰되는데, 이의 원인 물질로서 리포폴리사카라이드(lipopolysaccharide, LPS)가 잘 알려져 있다. 이에, PRX1에 대한 항체의 골 손실 억제효과 확인을 위해 리포폴리사카라이드를 사용하였다.
먼저, 11 내지 12주의 수컷 ICR 마우스의 두개관(頭蓋冠)에 0.1 mg의 리포폴리사카라이드를 매일 1회씩 피하 주사하여 골 손실을 유도하였다. 골손실이 유도된 마우스에 10 μg/ml의 PRX1에 대한 항체를 매일 1회 두개관에 피하 주사하였다. 이후의 과정은 상기 <실험예 5>와 동일한 조건 및 방법으로 두개관을 채취하고 TRAP염색하여 육안으로 관찰하였다.
그 결과, 도 11에 나타난 바와 같이, 대조군에서 파골세포 수가 뚜렷히 증가 하고 골손실이 발생하였으나, PRX1에 대한 항체를 투여하면 리포폴리사카라이드에 의해 유도된 파골세포 수의 증가가 유의하게 감소하였다(도 11). 이로부터, PRX1의 항체가 파골세포 분화를 효과적으로 억제할 수 있음을 확인하였다.
<실험예 7> PRX1 항체의 골손실 억제 효과 확인
<7-1> 골손실 동물모델에서 PRX1 농도 증가 확인
꼬리 매달기(tail suspension)를 통해 골손실이 유발된 동물모델에서 PRX1의 농도 변화를 다음과 같은 방법으로 확인하였다.
먼저, 8주령의 수컷 ICR 마우스를 뒷발이 지면에서 30도 정도 떠 있도록 쥐의 꼬리를 매달아 놓은 상태로 2주간 사육하였다. 이때, 사료 및 음용수에 자유롭게 접근이 가능하도록 하였다. 2주 후, 심장에서 혈액을 채취하고, PRX1에 대한 ELISA 키트(Cat#. MBS760435, Mybiosource, 미국)를 사용하여 혈청 내에 존재하는 PRX1의 농도를 측정하였다.
그 결과, 도 12에 나타난 바와 같이, 정상 마우스에 비해 골손실이 유발된 동물모델에서 혈청 내에 존재하는 PRX1의 농도가 유의적으로 증가하였다(도 12).
<7-2> PRX1 항체에 의한 골손실 억제 효과 확인
골손실이 유발된 동물모델을 이용하여 PRX1 항체의 골손실 억제 효과를 다음과 같은 방법으로 확인하였다.
구체적으로, 실험예 7-1에서 제조된 골손실 유발 마우스에 100 ㎍/㎏의 용량으로 PRX1에 대한 항체를 정맥주사로 투여하였다. 투여는 하루에 1회씩 3일 간격으로 총 4회 하였다. 투여가 완료된 후, 마우스를 희생시켜 넙다리뼈를 수득하고, 이를 이용하여 마이크로 CT(micro CT) 분석을 수행한 2D의 결과 사진을 도 13에 나타내었다.
도 13에 나타난 바와 같이, 꼬리 매달기에 의해 골손실이 유발된 마우스에 PRX1 항체를 투여함으로써 골손실이 억제되었다(도 13).
<7-3> PRX1 항체에 의한 골관련 지표의 발현 변화 확인
골손실이 유발된 동물모델을 이용하여 PRX1 항체에 의한 다양한 골관련 지표의 발현 변화를 CTAn 분석 프로그램을 사용하여 확인하였다.
구체적으로, CTAn 분석 프로그램을 사용하여 2D 또는 3D의 마이크로 CT 데이터를 수집하고, 이로부터 형상 및 밀도를 측정하였다. 먼저, 2D 측정은 모델 1172(Model 1172, Skyscan, 벨기에) 마이크로 CT 스캐너 및 분석 소프트웨어를 사용하여 9 ㎛의 복셀크기로 수행하였다. 측정된 이미지는 35 ㎸의 에너지 220 ㎃의 강도 및 590 ㎳의 집적시간에서 수집되었다. 수집된 결괏값들의 임계 값은 뼈를 세분화하도록 설정하였고, 모든 샘플에는 동일한 임계값을 설정하였다. 한편, 3D의 미세구조 평가는 CTAn 소프트웨어인 릴리즈 2.5(release 2.5, Skyscan, 벨기에)를 이용하였고, 직육면체 모양의 관심용적(volume of interset, VOI)을 적용하였다. 이때, 관심용적은 각 검체 전체를 포함하는 가장 작은 크기로 설정하였다. 또한, 3차원 미세 골구조 관련지표는 CTAn 소프트웨어를 사용하여 측정하였다.
그 결과, 도 14에 나타난 바와 같이, 꼬리 매달기에 의해 감소한 골 미네랄 밀도(bone mineral density, BMD, g/㎤), 조직 부피에 대한 골 부피(bone volume per tissue volume, BV/TV, %), 섬유주 두께(trabecular thickness, Tb.Th, ㎛) 및 섬유주 갯수(trabecular number, Tb.N, ㎛)와 같은 지표들은 PRX1 항체에 의해 증가되었다(도 14a 및 14b). 한편, 꼬리 매달기에 의해 감소한 섬유주 분리(trabecular separation, Tb.Sp, ㎛) 및 구조 모델 색인(structure model index, SMI)과 같은 지표들은 PRX1 항체에 의해 감소되었다(도 14c). 따라서, 상기로부터 PRX1 항체의 투여가 골손실을 유의적으로 억제함을 알 수 있었다.
Claims (11)
- 페록시레독신 1 단백질의 활성 억제제를 유효성분으로 함유하는 골질환의 예방 또는 치료용 약학적 조성물.
- 제 1항에 있어서, 상기 페록시레독신 1 단백질이 서열번호 1 또는 서열번호 2로 기재되는 아미노산 서열인, 골질환의 예방 또는 치료용 약학적 조성물.
- 제 1항에 있어서, 상기 활성 억제제가 페록시레독신 1 단백질에 상보적으로 결합하는 화합물, 펩티드, 펩티드 미메틱스 및 항체로 구성된 군에서 선택되는 어느 하나 이상인, 골질환의 예방 또는 치료용 약학적 조성물.
- 제 1항에 있어서, 상기 골질환이 골절, 골다공증(osteoprosis), 류마티스성 관절염(rheumatoid arthritis), 파제트병(Paget disease) 또는 전이성 골암(metastatic bone cancer)인, 골질환의 예방 또는 치료용 약학적 조성물.
- 페록시레독신 1 단백질의 활성 억제제를 유효성분으로 함유하는 골질환의 개선용 건강기능식품.
- 제 5항에 있어서, 상기 페록시레독신 1 단백질이 서열번호 1 또는 서열번호 2로 기재되는 아미노산 서열인, 골질환의 개선용 건강기능식품.
- 제 5항에 있어서, 상기 골질환이 골절, 골다공증(osteoprosis), 류마티스성 관절염(rheumatoid arthritis), 파제트병(Paget disease) 또는 전이성 골암(metastatic bone cancer)인, 골질환의 개선용 건강기능식품.
- 1) 골세포 또는 골조직에 피검물질을 처리하는 단계;2) 상기 단계 1)의 골세포 또는 골조직으로부터 페록시레독신 1 단백질의 분비량을 측정하는 단계; 및3) 상기 단계 2)의 페록시레독신 1 단백질의 분비량이 무처리 대조군에 비해 감소된 피검물질을 선별하는 단계를 포함하는 골질환의 예방 또는 치료용 물질 스크리닝 방법.
- 페록시레독신 1 단백질의 활성 억제제를 이를 필요로 하는 대상에 투여하는 것을 포함하는 골질환의 예방, 개선 또는 치료방법.
- 골질환의 예방 또는 치료용 약학적 조성물의 제조에 사용하기 위한 페록시레독신 1 단백질의 활성 억제제의 용도.
- 골질환의 예방 또는 개선용 건강기능식품의 제조에 사용하기 위한 페록시레독신 1 단백질의 활성 억제제의 용도.
