면역억제제란 항원의 작용에 대하여 숙주가 항체를 만드는 능력(체액성 면역반응) 또는 세포성 면역반응을 일으키는 능력을 저하시키거나 차단하기 위해 사용되는 다양한 물질들을 말한다. 이러한 면역억제제는 장기이식분야 뿐만 아니라 루푸스, 류마티스 관절염 등과 같은 자가면역질환과, 아토피, 알러지 등의 피부과민 반응에도 유용하게 사용될 수 있다. 우수한 면역억제제는 면역반응의 불균형을 조절할 수 있어야 하고, 인체에 대한 안전성이 확보되어야 하며, 장기간 치료시에 질환의 재발 발생 빈도가 낮아야 한다.
현재 사용되고 있는 면역억제제로는 사이클로스포린 A와 FK506 등이 있는데, 이들은 복잡한 화학구조를 가진 천연물 유래의 화합물로서 원료 수급의 측면에서 고비용이 드는 문제점이 있어 비경제적이고, 장기투여로 인해 각종 부작용이 야기될 수 있다는 위험성을 내포하고 있다. 따라서 낮은 독성 및 면역관용 유도와 함께 경제적인 생산이 가능한 새로운 면역억제제의 개발이 절실히 요구되고 있는 실정이다.
한편, 각종 병원체에 대한 생체 방어 시스템으로 면역계에서 중심적 역할을 담당하는 세포군의 하나로 T 세포가 있다. T 세포는 인체의 흉선에서 생성되며 일련의 분화 과정을 거치면서 고유의 특성을 지닌 T 세포로 분화하게 되는데, 분화를 완료한 T 세포는 그 기능에 따라 크게 1형 보조 세포(Th1)와 2형 보조 세포(Th2)로 구분된다. 이 중에서 Th1 세포의 주된 기능은 세포 매개성 면역에 관여하고, Th2 세포는 체액성 면역에 관여하며, 면역계에서 이러한 두 세포 집단은 서로 과 활성화되지 않도록 서로 견제를 통해 면역계의 균형을 유지하고 있다.
따라서 면역질환의 대부분은 이러한 두 가지 면역 세포간의 불균형에 기인하는 것으로 볼 수 있는데, 예를 들어 Th1 세포의 활성이 비정상적으로 증가하는 경우 자가면역질환이 발생할 수 있고, Th2 세포의 활성이 비정상적으로 증가하는 경우 과민반응에 의한 면역질환이 발생하는 것으로 알려져 있다.
한편, Th1 세포의 분화에 대한 최근 연구 결과에 따르면, Th1 세포의 활성을 조절할 수 있는 새로운 그룹인 면역조절 T 세포(Treg)의 존재가 알려지면서 이를 이용한 면역질환의 치료에 대한 연구가 대두되고 있는데, Treg 세포는 비정상적으로 활성화된 면역세포의 기능을 억제하여 염증 반응을 제어하는 특성이 있어, Treg 세포의 활성을 증가시키는 작용을 통해 면역질환을 치료하고자 하는 연구들이 많이 진행되고 있다.
또한, Treg 세포 이 외에 T 세포의 분화 과정에서 만들어지는 또 다른 그룹으로 Th17 세포가 있는데, Th17 세포는 미분화 T세포의 분화 과정에서 Treg 세포의 분화와 유사한 과정을 거치며 형성되는 것으로 알려져 있다. 즉, Treg 세포와 Th17 세포의 분화는 공통적으로 TGF-β의 존재 하에서 이루어지지만, Treg 세포의 경우 IL-6을 필요로 하지 않는 반면, Th17 세포의 경우에는 TGF-β와 함께 IL-6가 존재하는 상황에서 분화를 한다. 또한, 분화된 Th17 세포는 IL-17을 분비하는 것을 특징으로 한다.
Th17 세포는 Treg 세포와는 달리 면역질환에서 보이는 염증반응의 최전방에서 관여하여 염증 반응의 신호를 최대화시켜 질병의 진행을 가속화시키는 것이 밝혀지고 있다. 그러므로 자가면역질환 중 Treg 세포에 의해 제어되지 않는 자가면역질환의 경우, Th17 세포 활성의 억제를 표적으로 한 자가면역질환의 치료제 개발이 크게 부각되고 있다.
현재 사용되고 있는 면역질환치료제로는 T 세포에서의 신호변환 경로를 차단하는 면역 억제제가 가장 많이 사용되고 있는데, 이러한 면역억제제들은 독성, 감염, 임파종, 당뇨병, 진전(tremor), 두통, 설사, 고혈압, 오심, 신기능 장애 등의 부작용이 발생하는 문제점이 있다.
또한, T 세포의 활성화를 억제하는 방법을 통해 면역질환을 치료하는 방법 이외에도 면역 세포로부터 분비되는 사이토카인의 양을 조절하는 치료법 및 면역 세포로부터 분비되는 사이토카인을 표적으로 하는 항체를 이용한 치료법이 개발 중에 있다. 그러나 이러한 방법은 실제적으로 임상실험을 거쳐 환자들에게 적용하기까지 많은 시간이 소요되어야 하고, 항체를 이용하는 방법은 항체 제작 과정에서 너무 많은 비용이 든다는 문제점이 있다.
따라서 부작용이 없고 저렴하면서도 치료 효과가 우수한 새로운 면역질환 치료제 및 치료방법의 개발이 시급하다.
본 발명의 목적은 RIPK(receptor interacting serine/threonine protein kinase) 억제제를 유효성분으로 포함하는 면역질환의 예방 또는 치료용 약학적 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 RIPK1(receptor interacting serine/threonine protein kinase 1) 억제제를 유효성분으로 포함하는 골 손실 억제용 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 RIPK3(receptor interacting serine/threonine protein kinase 3) 억제제를 유효성분으로 포함하는 면역거부 질환의 예방 또는 치료용 약학적 조성물을 제공하는 것이다.
상기 목적을 달성하기 위하여, 본 발명은 RIPK(receptor interacting serine/threonine protein kinase) 억제제를 유효성분으로 포함하는 면역질환의 예방 또는 치료용 약학적 조성물을 제공한다.

본 발명은 RIPK 억제제를 면역질환의 새로운 치료제로서 사용할 수 있음을 규명한 것으로 면역질환이 유발된 동물모델에 RIPK 억제제를 처리하게 되면 질환의 증상이 완화 및 개선되는 것으로 나타났고, 염증성 사이토카인 및 병인성 사이토카인의 생성이 억제되고 반면 항염증성 사이토카인의 생성은 증가되는 것으로 나타났다. 따라서 본 발명은 면역질환 치료를 위한 새로운 타겟으로 RIPK을 발견하고 RIPK 억제제를 새로운 치료제로 활용할 수 있는 효과가 있다.
도 1은 정상 마우스(Normal) 및 콜라겐 유도 류마티스 관절염 동물모델(CIA, collagen-induced arthritis)의 관절 조직에서 RIPK1 발현량을 면역조직화학염색법에 의해 분석한 결과를 나타낸 것이다.

실시예
1. 정상조직과 관절염 조직에서의

1) 발현량 분석
본 발명자들은 정상상태와 질환유발 상태 시 RIPK1의 발현변화를 분석하기 위해, 정상 마우스와 관절염이 유도된 마우스의 관절 조직에서 RIPK1의 발현량을 면역조직화학염색법을 수행하여 분석하였다.
먼저, 관절염 유도 마우스는 DBA/1J 마우스에 type II collagen(CII)과 CFA(complete Freund's adjuvant)를 1:1로 혼합하여 마우스당 100μg의 CII를 tail base에 50μl 용량으로 주사하였다. 2주 후, CII와 CFA 1:1로 혼합액을 100μg/50μl로 2차로 주입하는 방법으로 관절염이 유발된 마우스를 제조하여 사용하였고, 상기 정상마우스는 CII를 처리하지 않은 DBA/1J 마우스를 사용하였다.
정상 마우스와 관절염 유도 마우스로부터 수득된 관절 조직을 10% 포르말린으로 고정시키고 뼈에서 석회질을 제거한 후 파라핀에 포매한 후 조직을 7 μm 두께의 절편을 만들어서 슬라이드에 부착하였다. 면역조직 화학염색을 진행하기 전에 자일렌을 이용하여 탈 파라핀 과정을 통과 후, 에탄올을 고농도에서 저농도까지 함수 시켰다. 이때 상기 RIPK1의 발현량을 감지하기 위해서 RIPK1에 대한 항체(Santa Cruz, SC-7881)를 이용하여 면역염색을 수행하였고, 이후 광학현미경으로 분석을 통해 단백질의 발현량을 측정하였다.
그 결과, 정상 마우스의 관절조직에 비해 류마티스 관절염이 유발된 관절조직에서 RIPK1의 발현이 현저하게 증가되었음을 확인하였다(도 1).

상기 실시예 1의 결과를 통해, RIPK1의 발현이 면역질환 유발시 정상에 비해 증가되어 있는 것을 동물모델로 확인한 사실을 토대로, 실제 임상적으로도 동일한 결과를 보이는지 확인하기 위해, 정상인, 골관절염 환자 및 류마티스 관절염 환자로부터 수득한 활막조직을 대상으로 마이크로어레이(microarray) 분석을 통해 RIPK1의 발현량을 분석하였다.
그 결과, 정상인에 비해 골관절염 및 류마티스 관절염이 발병하면 RIPK1의 발현량이 실제적으로 증가되었음을 확인하였다(도 2).
실시예
3.
RIPK1
억제제에 의한 염증성 사이토카인 및 항염증성 사이토카인의 조절효과
RIPK1이 면역질환 유발과 관련성이 있음을 확인함에 따라, 본 발명자들은 RIPK1을 억제하게 되면 면역질환을 치료할 수 있는 효과가 있는지 알아보기 위해, RIPK1 억제제에 의한 염증성 사이토카인 및 항염증성 사이토카인의 조절 효과를 분석하였다.
T 세포 활성 조건인 anti-CD3로 자극한 C57BL/6 정상 마우스로부터 분리한 비장세포(splenocytes)에 RIPK1 억제제인 Necrostatin-1 (Selleckchem, S8037)를 각각 1μM 및 10μM의 농도로 처리한 후, 48시간 배양하였고, 이후 배양액을 수집하여 대표적인 병인 사이토카인인 IL-17과 염증성 사이토카인인 IFN-γ 및 항염증성 사이토카인인 IL-10의 발현량을 ELISA 분석을 통해 확인하였다. 구체적으로, ELISA 분석을 위해, 먼저 96 well 플레이트에 anti-IL-17, anti-IFN-γ, anti-IL-10을 각각 2μg/ml로 코팅한 후, 반응 후 차단용액(1% BSA/PBST)으로 비특이적 결합을 차단시켰고, 이후 IL-17, IFN-γ, IL-10 recombinant를 1/2씩 연속 희석하여 이를 표준시약(Standard)으로 사용하였는데, 여기에 세포배양 상층액을 첨가하고 실온에서 2시간 반응시켰다. 이후, 바이오티닐(biotinylated) anti-IL-17, anti-IFN-γ, anti-IL-10을 2시간 동안 실온에서 반응시키고 3회 세척 후 ExtraAvidin-Alkaline Phosphatase conjugate를 희석하여 첨가한 후, 다시 실온에서 2시간 반응시켰다. 대조군은 anti-CD3로만 세포를 자극하고 아무것도 처리하지 않은 군을 사용하였다.
그 결과, T 세포가 활성화된 조건에서 RIPK1 억제제를 처리한 군의 경우, RIPK1 억제제를 처리하지 않은 군에 비해, 병인성 사이토카인(IL-17)과 염증성 사이토카인(IFN-γ)의 발현은 감소된 것으로 나타난 반면, 항염 활성을 갖는 사이토카인(IL-10)은 증가한 것으로 나타났다(도 3).
따라서, RIPK1을 억제하는 화합물의 경우, 면역질환을 치료할 수 있는 치료제로서 사용할 수 있음을 확인하였다.

4. 대식세포 활성화 조건에서의
RIPK1
억제제에 의한 염증성 사이토카인 및 항염증성 사이토카인의 조절분석
본 발명자들은 대식세포 활성화 조건인 LPS(lipopolysaccharide)로 자극한 C57BL/6 정상 마우스로부터 분리한 비장세포(splenocytes)에 RIPK1 억제제인 Necrostatin-1 (Selleckchem, S8037)를 각각 1μM 및 10μM의 농도로 처리한 후, 48시간 배양하고, 배양액을 수집하여 대표적인 염증성 사이토카인인 TNF-α 및 항염증성 사이토카인인 IL-10의 발현량을 ELISA 분석을 통해 확인하였다. 구체적으로 ELISA 분석을 위해, 먼저 96 well 플레이트에 anti-TNF-α, anti-IL-10을 2μg/ml로 코팅한 후, 반응 후 차단용액 (1% BSA/PBST)으로 비특이적 결합을 차단시켰다. 이후 TNF-α, IL-10 recombinant를 1/2씩 연속 희석하여 이를 표준시약(Standard)로 사용하였고 세포배양 상층액을 넣고 실온에서 2시간 반응시켰다. 이후 바이오티닐(biotinylated) anti-TNF-α, anti-IL-10을 2시간 동안 실온에서 반응시키고 3회 세척 후, ExtraAvidin-Alkaline Phosphatase conjugate를 희석하여 첨가한 후, 실온에서 2시간 반응시켰다. 대조군은 LPS로만 세포를 자극하고 아무것도 처리하지 않은 군을 사용하였다.
그 결과, 대식세포를 활성화시키는 조건에서 RIPK1 억제제를 처리한 군은 처리하지 않은 군에 비해 LPS로 유도된 염증성 사이토카인인 TNF-α의 생성은 현저하게 억제하는 것으로 나타난 반면, 항염증성 사이토카인인 IL-10에는 영향을 미치지 않는 것으로 나타났다(도 4). 즉, RIPK1 억제제의 경우, 염증성 사이토카인의 생성은 억제하였으나, 항염증성 사이토카인의 생성은 억제하지 않는 것으로 나타났다.

관절염 동물모델을 대상으로
RIPK1
억제제에 의한 치료 효과 분석
RIPK1 억제제가 실제로 자가면역질환이 유발된 마우스 동물에게 투여할 경우, 효과적인 치료 효능이 있는지 확인하기 위해 다음과 같은 실험을 수행하였다.관절염이 유발된 마우스를 제조하기 위해 IL-10 넉아웃(knockout) 마우스에 Type II Collagen으로 CIA(collagen-induced arthritis)를 유도하여 관절염 발병 마우스 모델을 제조한 후, 처음 Immunization 후 10일째부터 1주일에 3회 복강 내 RIPK1 억제제인 Necrostatin-1 (Selleckchem,S8037)을 0.02mg/mice로 투여한 후 관절염 지수를 분석하였다.
관절염 평가 기준은 다음과 같다.
-평가 기준-
0점: 부종이나 종창이 없다.
1점: 발 또는 발목관절에 국한된 경한 부종과 발적
2점: 발목관절에서 족근골(metatarsal)에 걸친 경한 부종과 발적
3점: 발목관절에서 족근골에 걸친 중등도의 부종과 발적
4점: 발목에서 다리 전체에 걸쳐 부종과 발적이 있는 경우
5점 이상: 부종과 발적의 증상이 심각하게 계속 진행되는 경우
그 결과, 관절염이 유도된 마우스에 RIPK1 억제제를 투여한 경우, RIPK1 억제제를 투여하지 않은 군에 비해 관절염 지수(arthritis score)가 현저하게 억제되는 것으로 나타났고, 관절염 발병 정도(incidence)도 2.5배 이상 감소하는 것으로 나타났다(도 5).
따라서, 이러한 결과를 통해 본 발명자들은 실제적으로 RIPK1의 활성을 억제할 경우, 면역질환을 치료할 수 있음을 알 수 있었다.

RIPK1
억제제에 의한 항체 조절 효과분석
상기 실시예 5에서 실험에 사용한 마우스들을 대상으로 RIPK1 억제제를 4주간 투여한 마우스로부터 혈액을 수득하고 다시 혈청을 분리한 후, CII specific Total IgG, CII specific IgG1, CII specific IgG2a의 발현량을 ELISA에 의해 분석하였다.
그 결과, RIPK1 억제제를 투여한 마우스의 혈청에서는 CII specific Total IgG의 발현이 감소되었고, Th2 반응과 관련 있는 CII specific IgG1의 발현은 오히려 증가되었다. 또한, Th1 반응과 관련이 있는 CII specific IgG2a의 발현도 감소되었다(도 6).
따라서, RIPK1 억제제는 Th2와 같은 항염증성 항체는 증가시키고 Th1과 같은 염증성 항체의 생성을 억제시킬 수 있음을 알 수 있었다.

RIPK1
억제제의 세포 독성 여부 확인
본 발명자들은 RIPK1 억제제가 세포 독성을 유발하는지 확인하기 위한 실험을 수행하였다. 구체적으로, 정상 마우스의 비장세포(splenocytes)에 RIPK1 억제제인 Necrostatin 1을 각각 0.1μM, 1μM, 10μM의 농도로 처리한 후, 3일 동안 배양하였고, MTT powder(Thiazolyl Blue Tetrazolium Bromide)를 PBS에 0.5mg/ml로 녹여 50μl씩 처리한 후 4시간을 배야하였다. 그 후, 1300rpm, 5분, 4°C에서 원심분리를 한 후 상층액 120μl을 제거하고 DMSO를 50μl 넣어준 후 540 nm에서 흡광도를 측정하였다.
그 결과, RIPK1 억제제를 처리한 군 모두에서 세포 독성이 유발되지 않음을 확인하였고, 0.1 μM, 1 μM, 10 μM 농도로 처리한 군 모두에서 세포 독성이 유발되지 않는 것으로 나타났다(도 7).

억제제에 의한
Th17
세포의 분화 억제 효과
RIPK1 억제제에 의한 Th17 세포의 분화를 억제하는 효과가 있는지를 확인하기 위한 실험을 수행하였다. 구체적으로, Th17 세포의 분화를 유도하기 위하여 정상 마우스의 비장세포(splenocytes)에 anti-CD3, anti-CD28, anti-IFN-γ, anti-IL-4, IL-6 및 TGF-β를 처리하고, RIPK1 억제제인 Necrostatin 1을 10μM 로 처리한 후 3일 동안 세포를 배양하였다. 그 후, 세포를 모은 후 anti-mouse CD4 PerCP-Cyanine5.5 와 anti-mouse IL-17 FITC 항체를 이용하여 염색하고, FACS(Fluorescence-activated cell sorting, BD FACS caliburTM)를 수행하였으며, FlowJo 프로그램을 이용하여 분석하였다.
그 결과, RIPK1 억제제를 처리하는 경우에는 대조군에 비해 Th17 세포로의 분화가 억제되었음을 확인하였다(도 8).

억제제에 의한 파골세포 분화 억제 효과
본 발명자들은 RIPK1 억제제에 의해 파골세포 분화가 억제되는 효과가 있는지 확인하기 위한 실험을 수행하였다. 구체적으로, IL-10 넉아웃(knockout) 마우스에 Type II Collagen으로 CIA를 유도하여 관절염 발병 마우스 모델로 유도한 후, 처음 Immunization 후 10일째부터 1주일에 3회 복강 내에 RIPK-1 억제제(Necrostatin 1)를 20mg/kg 및 10mg/kg으로 투여한 후, 실험종료 시점에 마우스의 골수세포에 50 ng/ml의 RANKL(Receptor activator of nuclear factor kappa-B ligand) 과 M-CSF(macrophage colony-stimulating factor)로 자극하고, 이후 RIPK1 억제제인 Necrostatin 1을 1μM, 10μM로 처리한 다음, TRAP(Tartrate-resistant acid phosphatase) 양성 세포를 조사하였다.
그 결과, RIPK-1 억제제의 파골세포 분화 조절능을 관찰하기 위한 Osteoclastogenesis 분화 실험에서 RIPK-1 억제제를 처리하는 경우에는 골파괴 기전과 관련 있는 TRAP 양성 세포가 농도 의존적으로 감소되었음을 확인하였다 (도 9).
따라서, RIPK1 억제제가 파골세포 분화를 억제할 수 있는 효과가 있고, 골질환의 치료에 효과가 있음을 알 수 있었다.

RIPK3
억제제에 의한 염증성 장질환 억제 효과
본 발명자들은 RIPK3 억제제를 염증성 장질환이 유발된 마우스 동물모델에 투여하여 치료효과를 확인하고자 하였다. 구체적으로 염증성 장질환 동물모델은 C57BL6 마우스의 음수에 3% DSS(dextran sulfate sodium)를 넣어주어 제조되었다. 체중은 DSS 약물의 투여를 시작하는 시점에서 측정되었고, 동시에 RIPK3 억제제인 GSK'872 (Calbiochem, 530389)은 경구투여되어 실험이 진행되었다.
그 결과, 염증성 장질환이 유발된 마우스에 RIPK3 억제제는 대조군(WT, wild type) 정도로 체중을 회복하지는 않았으나, 염증성 장질환 마우스보다는 약간 체중이 증가되었음을 확인하였다 (도 13).

억제제에 의한 동종반응(
Alloresponse
) 억제 효과
본 발명자들은 RIPK3 억제제가 조직이식에 대한 거부반응을 억제하는지 확인하고자 하였다. 구체적으로, in vitro에서 96 well의 각 웰 당 1×105개의 정상 수여자(Balb/c, responder)의 CD4+T cell와 1×105개의 방사선으로 조사시킨 수여자(동종동형) 또는 공여자(C57BL/6, stimulator, 동종이형) 유래의 T 세포를 제거한 비장세포를 넣고 혼합 배양시켜 동종반응을 유도하였다. 또한, RIPK3 억제제인 GSK'872 (Calbiochem, 530389)은 1μM 및 10μM을 함께 처리한 후 72시간 배양시켰다. 배양완료 16시간 전에 배양된 세포에서 T 세포 증식 반응 정도를 3H Thymidine을 처리하고 세포증식 정도를 b 카운터로 3H Thymidine의 양을 측정하여 확인하였다.
그 결과, RIPK3 억제제는 대조군에 비해 동종반응(Alloresponse)을 억제하는 효과가 있음을 확인하였다(도 14).
따라서, RIPK3 억제제가 이식 과정에서 발생할 수 있는 T 세포의 이식 후 거부반응 및 동종반응을 효과적으로 억제하여 이식에 의한 질환 치료를 성공적으로 유도할 수 있다는 사실을 확인하였다.
본 발명에서 RIPK1(receptor interacting serine/threonine protein kinase 1)은 pathogen recognition receptor (PRRS)와 DNA 손상 및 death receptor ligation에 의한 Cell-death 신호 (programmed necrosis) 전달에 관여한다.
본 발명에서 용어, "안티센스 올리고뉴클레오타이드"란 특정 mRNA의 서열에 상보적인 핵산 서열을 함유하고 있는 DNA 또는 RNA 또는 이들의 유도체를 의미하고, mRNA 내의 상보적인 서열에 결합하여 mRNA의 단백질로의 번역을 저해하는 작용을 한다. 본 발명의 안티센스 서열은 상기 유전자의 mRNA에 상보적이고 상기 mRNA에 결합할 수 있는 DNA 또는 RNA 서열을 의미하며, 상기 mRNA의 번역, 세포질내로의 전위(translocation), 성숙(maturation) 또는 다른 모든 전체적인 생물학적 기능에 대한 필수적인 활성을 저해할 수 있다.
또한, 상기 안티센스 핵산은 효능을 증진시키기 위하여 하나 이상의 염기, 당 또는 골격(backbone)의 위치에서 변형될 수 있다(De Mesmaeker et al., Curr

안티센스 올리고뉴클레오타이드는 통상의 방법으로 시험관에서 합성되어 생체 내로 투여하거나 생체 내에서 안티센스 올리고뉴클레오타이드가 합성되도록 할 수 있다. 시험관에서 안티센스 올리고뉴클레오타이드를 합성하는 일예는 RNA 중합효소 I를 이용하는 것이다. 생체 내에서 안티센스 RNA가 합성되도록 하는 한 가지 예는 인식부위(MCS)의 기원이 반대 방향에 있는 벡터를 사용하여 안티센스 RNA가 전사되도록 하는 것이다. 이런 안티센스 RNA는 서열 내에 번역 중지 코돈이 존재하도록 하여 펩타이드 서열로 번역되지 않도록 하는 것이 바람직하다.
본 발명에서 "RNAi"는 RNA Interference를 지칭하는 말로, 우리말로 풀이하면 RNA 간섭이라는 뜻을 지니고 있다. RNA 간섭은 대부분의 생물체 사이에서 잘 보존되어 있는 특이적 유전자억제 현상이다. 바이러스 감염에 대한 방어나 트랜스포손을 억제하기 위하여 혹은 비정상적 mRNA를 제거하기 위하여 세포가 사용하는 유전자 감시기작의 일종으로 생각되고 있다. 특히, small RNA에 의한 유전자 억제현상을 넓은 의미에서 RNA간섭이라 하고, 좁은 의미의 RNA 간섭은 siRNA에 의한 mRNA 분해 현상을 뜻한다. 또한 RNA간섭은 siRNA를 이용한 유전자 억제 실험 기술을 뜻하기도 한다.
본 발명에서 "siRNA"라는 용어는 RNA 방해 또는 유전자 사일런싱을 매개할 수 있는 핵산 분자를 의미한다(국제공개 특허번호 00/44895, 01/36646, 99/32619, 01/29058, 99/07409 및 00/44914 참조). siRNA는 표적 유전자의 발현을 억제할 수 있기 때문에 효율적인 유전자 넉다운(knock-down) 방법으로서 또는 유전자치료 방법으로 제공된다.
본 발명의 siRNA 분자는, 센스 가닥(상기 마커 유전자의 mRNA 서열에 상응하는 서열)과 안티센스 가닥(상기 mRNA 서열에 상보적인 서열)이 서로 반대쪽에 위치하여 이중쇄를 이루는 구조를 가질 수 있으며, 또한, 본 발명의 siRNA 분자는 자기-상보성(self-complementary) 센스 및 안티센스 가닥을 가지는 단일쇄 구조를 가질 수 있다. 나아가 siRNA는 RNA끼리 짝을 이루는 이중사슬 RNA 부분이 완전히 쌍을 이루는 것에 한정되지 않고 미스매치(대응하는 염기가 상보적이지 않음), 벌지(일방의 사슬에 대응하는 염기가 없음) 등에 의하여 쌍을 이루지 않는 부분이 포함될 수 있다. 또한, siRNA 말단 구조는 상기 마커 유전자의 발현을 RNAi 효과에 의하여 억제할 수 있는 것이면 평활(blunt) 말단 혹은 점착(cohesive) 말단 모두 가능하고, 점착 말단 구조는 3'-말단 돌출 구조와 5'-말단 돌출 구조 모두 가능하다.
또한, 본 발명의 siRNA 분자는 자기-상보성(self-complementary) 센스 및 안티센스 가닥 사이에 짧은 뉴클레오티드 서열이 삽입된 형태를 가질 수 있으며, 이 경우 뉴클레오타이드 서열의 발현에 의해 형성된 siRNA 분자는 분자내 혼성화에 의하여 헤어핀 구조를 형성하게 되며, 전체적으로는 스템-앤드-루프 구조를 형성하게 된다. 이 스템-앤드-루프 구조는 인 비트로 또는 인 비보에서 프로세싱되어 RNAi를 매개할 수 있는 활성의 siRNA 분자를 생성한다.
siRNA를 제조하는 방법은 시험관에서 siRNA를 직접 합성한 뒤, 형질전환 과정을 거쳐 세포 안으로 도입시키는 방법과 siRNA가 세포 안에서 발현되도록 제조된 siRNA 발현 벡터 또는 PCR-유래의 siRNA 발현 카세트 등을 세포안으로 형질전환 또는 감염(infection)시키는 방법이 있다.
또한, 유전자 특이적인 siRNA를 포함하는 본 발명의 조성물은 siRNA의 세포내 유입을 촉진시키는 제제를 포함할 수 있다. siRNA의 세포내 유입을 촉진시키는 제제에는 일반적으로 핵산 유입을 촉진하는 제제를 사용할 수 있으며, 이러한 예로는, 리포좀을 이용하거나 콜레스테롤, 콜레이트 및 데옥시콜산을 비롯한 다수의 스테롤류 중 1종의 친유성 담체와 함께 배합할 수 있다. 또한, 폴리-L-라이신(poly-L-lysine), 스퍼민(spermine), 폴리실아잔(polysilazane), 폴리에틸레민(PEI:polyethylenimine), 폴리디하이드로이미다졸레늄(polydihydroimidazolenium), 폴리알리라민(polyallylamine), 키토산(chitosan) 등의 양이온성 고분자(cationic polymer)를 이용할 수도 있고, 숙실화된 PLL(succinylated PLL), 숙실화된 PEI(succinylated PEI), 폴리글루타믹산(polyglutamic acid), 폴리아스파틱산(polyaspartic acid), 폴리아크릴산(polyacrylic acid), 폴리메타아크릴산(polymethacylic acid), 덱스트란 설페이트(dextran sμlfate), 헤파린(heparin), 히아루릭산(hyaluronic acid) 등의 음이온성 고분자(anionic polymer)를 이용할 수 있다.
본 발명에서 항체는 기존의 치료제와 직접 또는 링커 등을 통하여 간접적으로 커플링(예를 들어, 공유결합)시킬 수 있다. 항체와 결합될 수 있는 치료제로는 이에 제한되지는 않으나, 131I, 90Y, 105Rh, 47Sc, 67Cu, 212Bi, 211At, 67Ga, 125I, 186Re, 188Re, 177Lu, 153Sm, 123I, 111In 등과 같은 방사성핵종(radionuclide); 메토트렉세이트(methotrexate), 아드리아마이신(adriamycin), 및 인터페론과 같은 림포카인(lympokine) 등의 생물학적 반응 변형체 또는 약물; 리신, 아브린, 디프테리아 등과 같은 독소; 이형기능성 항체(heterofunctional antibodies), 즉 다른 항체와 결합되어 그 복합체가 암세포와 효능 세포(예를 들면, T 세포와 같은 K 세포(killer cell) 모두에 결합하는 항체; 및 자연적인, 즉, 비-연관 또는 비-복합된 항체와 결합될 수 있다.
뼈(bone)는 인체의 연조직과 체중을 지탱해주고 내부기관을 둘러싸서 내부 장기를 외부의 충격으로부터 보호한다. 또한, 근육이나 장기를 구조적으로 지탱할 뿐만 아니라 체내의 칼슘이나 다른 필수 무기질 즉 인이나 마그네슘과 같은 물질을 저장하는 인체의 중요한 부분 중 하나이다. 따라서 성장이 끝난 성인의 뼈는 멈추지 않고 죽는 날까지 오래된 뼈는 제거하고 새로운 뼈로 대체하는 생성과 흡수과정을 매우 역동적, 지속적으로 반복 재생하면서 균형을 유지하게 된다. 이를 뼈의 재형성(bone remodeling)이라고 한다. 이러한 뼈의 재형성은 성장과 스트레스에 의해서 일어나는 뼈의 미세한 손상을 회복시키고 적절히 뼈의 기능을 유지하는데 필수적이다.
뼈의 재형성에는 크게 두 종류의 세포가 관여하는 것으로 알려져 있다. 두 세포 중 하나는 뼈를 생성하는 조골세포(osteoblast)이고, 다른 하나는 뼈를 파괴하는 파골세포(osteoclast)이다. 조골세포는 RANKL(receptor activator of NF-κB ligand: 이하 간략하게 'RANKL'이라 함)과 이것의 유도 수용체(decoy receptor)인 OPG(osteoprotegerin)를 생성한다. RANKL이 파골 전구세포(osteoclast progenitor cells) 표면에 있는 수용체인 RANK(receptor activator of NF-κB: 이하 간략하게 'RANK'라 함)에 결합하면 파골 전구세포가 파골세포로 분화되어 골 흡수(resorption)가 일어난다. 자세하게는 RANKL 자극에 의한 세포내 칼슘 신호 전달로 파골 전구세포에서 파골세포의 분화를 조절하는 핵심 전사인자로 알려져 있는 NFAT(nuclear factor of activated T cells: 이하 간단하게 'NFAT'라 함)의 발현이 유도되고, 그 후 세포외 신호조절인산화효소(extracellμlar signal-regμlated kinase: 이하 간단하게 'ERK'라 함), NF-κB, p38 MAP kinase 등의 활성화가 NFAT의 자가증폭(autoamplification)을 유발하여 활성이 지속되는 것이다.
또한, 파골세포의 생성을 위해서는 NF-κB 리간드의 수용체 활성제(RANKL)와 대식세포증식자극인자(macrophage colony stimμlating factor(M-CSF))와 같은 두 종류의 싸이토카인(cytokine)이 필수적인데, 이러한 싸이토카인이 파골세포의 분화를 유도한다(신정민 외, KOREAN J. FOODSCI. TECHNOL. 40(6), 674-679(2008)). 또한, 파골세포 표지 인자로 사용되고 있는 TRAP, 파골세포에 선택적으로 분포하여 골흡수를 일으키는 단백질인 카뎁신 K 및 파골세포의 이동에 중요한 역할을 하는 MMP-9와 같은 활성화된 파골세포에만 특이하게 발현된다.
본 발명에 따른 조성물은 면역질환, 골 손실로 유발될 수 있는 골질환, 및 면역거부질환을 예방 및 치료할 수 있는 약학적 조성물로 사용될 수 있으며, 상기 약학적 조성물은 약학적으로 허용되는 담체를 추가로 포함할 수 있다. 상기에서 "약학적으로 허용되는"이란 생리학적으로 허용되고 인간에게 투여될 때, 통상적으로 위장 장애, 현기증 등과 같은 알레르기 반응 또는 이와 유사한 반응을 일으키지 않는 조성물을 말한다. 약학적으로 허용되는 담체로는 예를 들면, 락토스, 전분, 셀룰로스 유도체, 마그네슘 스테아레이트, 스테아르산 등과 같은 경구 투여용 담체 및 물, 적합한 오일, 식염수, 수성 글루코스 및 글리콜 등과 같은 비경구 투여용 담체 등이 있으며 안정화제 및 보존제를 추가로 포함할 수 있다. 적합한 안정화제로는 아황산수소나트륨, 아황산나트륨 또는 아스코르 브산과 같은 항산화제가 있다. 적합한 보존제로는 벤즈알코늄 클로라이드, 메틸- 또는 프로필-파라벤 및 클로로부탄올이 있다. 그 밖의 약학적으로 허용되는 담체로는 다음의 문헌에 기재되어 있는 것을 참고로 할 수 있다(Remington's Pharmaceutical Sciences, 19th ed., Mack Publishing Company, Easton, PA, 1995). 본 발명에 따른 약학적 조성물은 상술한 바와 같은 약학적으로 허용되는 담체와 함께 당업계에 공지된 방법에 따라 적합한 형태로 제형화 될 수 있다. 즉, 본 발명의 약학적 조성물은 공지의 방법에 따라 다양한 비경구 또는 경구 투여용 형태로 제조될 수 있으며, 비경구 투여용 제형의 대표적인 것으로는 주사용 제형으로 등장성 수용액 또는 현탁액이 바람직하다. 주사용 제형은 적합한 분산제 또는 습윤제 및 현탁화제를 사용하여 당업계에 공지된 기술에 따라 제조할 수 있다. 예를 들면, 각 성분을 식염수 또는 완충액에 용해시켜 주사용으로 제형화될 수 있다. 또한 경구 투여용 제형으로는, 이에 한정되지는 않으나, 분말, 과립, 정제, 환약 및 캡슐 등이 있다.
상기와 같은 방법으로 제형화된 약학적 조성물은 유효량으로 경구, 경피, 피하, 정맥 또는 근육을 포함한 여러 경로를 통해 투여될 수 있다. 상기에서 '유효량' 이란 환자에게 투여하였을 때, 예방 또는 치료 효과를 나타내는 양을 말한다. 본 발명에 따른 약학적 조성물의 투여량은 투여 경로, 투여 대상, 연령, 성별 체중, 개인차 및 질병 상태에 따라 적절히 선택할 수 있다. 바람직하게는, 본 발명의 약학적 조성물은 질환의 정도에 따라 유효성분의 함량을 달리할 수 있으나, 바람직하게는 1~10000㎍/체중kg/day, 더욱 바람직하게는 10~1000㎎/체중kg /day의 유효용량으로 하루에 수회 반복 투여될 수 있다. 또한 본 발명의 조성물은 면역질환을 예방, 개선 또는 치료하는 효과를 가지는 공지의 화합물과 병행하여 투여할 수도 있다.
또한, 본 발명은 RIPK1 유전자 또는 RIPK1 단백질을 포함하는 세포 또는 조직에 후보물질을 처리하는 단계; 및 RIPK1 유전자의 발현량 또는 RIPK1 단백질의 양 또는 RIPK1 단백질의 활성을 측정하는 단계를 포함하는, 면역질환 치료제 스크리닝 방법을 제공한다.
본 발명에 따른 상기 방법은 새로운 면역질환 질환의 치료제를 RIP-1을 이용하여 발굴할 수 있는 방법으로서, 상기 “후보물질”이란 RIPK1 유전자의 발현량, RIPK1 단백질의 양 또는 RIPK1 단백질의 활성에 영향을 미치는지 여부를 검사하기 위하여 스크리닝에서 이용되는 미지의 물질을 의미한다. 상기 시료는 화합물, 단백질, 펩타이드, 올리고뉴클레오티드 또는 천연물 추출물 등을 포함할 수 있으나, 이에 제한되지는 않는다.
상기 후보물질의 처리는 바람직하게 RIPK1 유전자 또는 RIPK1 단백질을 포함하는 세포 또는 조직에 후보물질을 처리한 후 배양하는 과정으로 수행할 수 있다.
또한, 상기에서 RIPK1 단백질의 세포 내 수준 증가의 의미는 RIPK1 유전자의 발현이 증가되거나 또는 RIPK1 단백질의 분해가 억제되어 RIPK1 단백질의 양이 증가되는 것을 말한다. 상기 RIPK1 유전자 발현은 RIPK1 유전자의 전사 및 단백질로의 번역 과정을 포함한다. 따라서 상기 후보물질이 세포 내에서 RIPK1 유전자의 발현 및 단백질의 수준을 감소기킬 경우, 상기 후보물질을 면역질환의 신규 치료제로 판단할 수 있다.
상기 유전자의 발현 수준 측정은 바람직하게는 mRNA의 수준을 측정하는 것이며, mRNA의 수준을 측정하는 방법으로는 역전사 중합효소연쇄반응(RT-PCR), 실시간 역전사 중합효소연쇄반응, RNase 보호 분석법, 노던 블럿 및 DNA 칩 등이 있으나, 이에 제한되지는 않는다.
또한, 상기 단백질의 양 또는 단백질의 활성 측정은 항체를 이용할 수 있는데, 이러한 경우, 생물학적 시료 내의 상기 마커 단백질과 이에 특이적인 항체는 결합물, 즉, 항원-항체 복합체를 형성하며, 항원-항체 복합체의 형성량은 검출 라벨(detection label)의 시그널의 크기를 통해서 정량적으로 측정할 수 있다. 이러한 검출 라벨은 효소, 형광물, 리간드, 발광물, 미소입자(microparticle), 레독스 분자 및 방사선 동위원소로 이루어진 그룹 중에서 선택할 수 있으며, 이에 제한되는 것은 아니다. 단백질 수준을 측정하기 위한 분석 방법으로는, 이에 제한되지는 않으나, 웨스턴 블럿, ELISA, 방사선면역분석, 방사선 면역 확산법, 오우크테로니 면역 확산법, 로케트 면역전기영동, 조직면역 염색, 면역침전 분석법, 보체 고정분석법, FACS, 단백질 칩 등이 있다.
이와 같이, 상기 RIPK1 유전자의 발현량 또는 RIPK1 단백질의 양 또는 RIPK1 단백질의 활성을 측정하는 방법은 기술된 바와 같이 당업계에 공지된 여러 가지 방법을 사용할 수 있는데, 바람직하게 역전사 중합효소연쇄반응(RT-PCR), 실시간 역전사 중합효소연쇄반응, 면역침전법(coimmunoprecipitation), 효소면역분석법(enzyme-linked immunosorbentassay), 방사능면역분석법(RIA), 면역조직화학, 웨스턴 블로팅(Western Blotting) 및 유세포 분석법(FACS)을 사용할 수 있다.
이하 본 발명을 실시예에 의하여 더욱 상세하게 설명한다. 이들 실시예는 단지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 범위가 이들 실시예에 국한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
실시예
1. 정상조직과 관절염 조직에서의
RIPK1
(Receptor-interacting serine/threonine-protein kinase 1) 발현량 분석
본 발명자들은 정상상태와 질환유발 상태 시 RIPK1의 발현변화를 분석하기 위해, 정상 마우스와 관절염이 유도된 마우스의 관절 조직에서 RIPK1의 발현량을 면역조직화학염색법을 수행하여 분석하였다.
먼저, 관절염 유도 마우스는 DBA/1J 마우스에 type II collagen(CII)과 CFA(complete Freund's adjuvant)를 1:1로 혼합하여 마우스당 100μg의 CII를 tail base에 50μl 용량으로 주사하였다. 2주 후, CII와 CFA 1:1로 혼합액을 100μg/50μl로 2차로 주입하는 방법으로 관절염이 유발된 마우스를 제조하여 사용하였고, 상기 정상마우스는 CII를 처리하지 않은 DBA/1J 마우스를 사용하였다.
정상 마우스와 관절염 유도 마우스로부터 수득된 관절 조직을 10% 포르말린으로 고정시키고 뼈에서 석회질을 제거한 후 파라핀에 포매한 후 조직을 7 μm 두께의 절편을 만들어서 슬라이드에 부착하였다. 면역조직 화학염색을 진행하기 전에 자일렌을 이용하여 탈 파라핀 과정을 통과 후, 에탄올을 고농도에서 저농도까지 함수 시켰다. 이때 상기 RIPK1의 발현량을 감지하기 위해서 RIPK1에 대한 항체(Santa Cruz, SC-7881)를 이용하여 면역염색을 수행하였고, 이후 광학현미경으로 분석을 통해 단백질의 발현량을 측정하였다.
그 결과, 정상 마우스의 관절조직에 비해 류마티스 관절염이 유발된 관절조직에서 RIPK1의 발현이 현저하게 증가되었음을 확인하였다(도 1).

상기 실시예 1의 결과를 통해, RIPK1의 발현이 면역질환 유발시 정상에 비해 증가되어 있는 것을 동물모델로 확인한 사실을 토대로, 실제 임상적으로도 동일한 결과를 보이는지 확인하기 위해, 정상인, 골관절염 환자 및 류마티스 관절염 환자로부터 수득한 활막조직을 대상으로 마이크로어레이(microarray) 분석을 통해 RIPK1의 발현량을 분석하였다.
그 결과, 정상인에 비해 골관절염 및 류마티스 관절염이 발병하면 RIPK1의 발현량이 실제적으로 증가되었음을 확인하였다(도 2).
실시예
3.
RIPK1
억제제에 의한 염증성 사이토카인 및 항염증성 사이토카인의 조절효과
RIPK1이 면역질환 유발과 관련성이 있음을 확인함에 따라, 본 발명자들은 RIPK1을 억제하게 되면 면역질환을 치료할 수 있는 효과가 있는지 알아보기 위해, RIPK1 억제제에 의한 염증성 사이토카인 및 항염증성 사이토카인의 조절 효과를 분석하였다.
T 세포 활성 조건인 anti-CD3로 자극한 C57BL/6 정상 마우스로부터 분리한 비장세포(splenocytes)에 RIPK1 억제제인 Necrostatin-1 (Selleckchem, S8037)를 각각 1μM 및 10μM의 농도로 처리한 후, 48시간 배양하였고, 이후 배양액을 수집하여 대표적인 병인 사이토카인인 IL-17과 염증성 사이토카인인 IFN-γ 및 항염증성 사이토카인인 IL-10의 발현량을 ELISA 분석을 통해 확인하였다. 구체적으로, ELISA 분석을 위해, 먼저 96 well 플레이트에 anti-IL-17, anti-IFN-γ, anti-IL-10을 각각 2μg/ml로 코팅한 후, 반응 후 차단용액(1% BSA/PBST)으로 비특이적 결합을 차단시켰고, 이후 IL-17, IFN-γ, IL-10 recombinant를 1/2씩 연속 희석하여 이를 표준시약(Standard)으로 사용하였는데, 여기에 세포배양 상층액을 첨가하고 실온에서 2시간 반응시켰다. 이후, 바이오티닐(biotinylated) anti-IL-17, anti-IFN-γ, anti-IL-10을 2시간 동안 실온에서 반응시키고 3회 세척 후 ExtraAvidin-Alkaline Phosphatase conjugate를 희석하여 첨가한 후, 다시 실온에서 2시간 반응시켰다. 대조군은 anti-CD3로만 세포를 자극하고 아무것도 처리하지 않은 군을 사용하였다.
그 결과, T 세포가 활성화된 조건에서 RIPK1 억제제를 처리한 군의 경우, RIPK1 억제제를 처리하지 않은 군에 비해, 병인성 사이토카인(IL-17)과 염증성 사이토카인(IFN-γ)의 발현은 감소된 것으로 나타난 반면, 항염 활성을 갖는 사이토카인(IL-10)은 증가한 것으로 나타났다(도 3).
따라서, RIPK1을 억제하는 화합물의 경우, 면역질환을 치료할 수 있는 치료제로서 사용할 수 있음을 확인하였다.
실시예
4. 대식세포 활성화 조건에서의
RIPK1
억제제에 의한 염증성 사이토카인 및 항염증성 사이토카인의 조절분석
본 발명자들은 대식세포 활성화 조건인 LPS(lipopolysaccharide)로 자극한 C57BL/6 정상 마우스로부터 분리한 비장세포(splenocytes)에 RIPK1 억제제인 Necrostatin-1 (Selleckchem, S8037)를 각각 1μM 및 10μM의 농도로 처리한 후, 48시간 배양하고, 배양액을 수집하여 대표적인 염증성 사이토카인인 TNF-α 및 항염증성 사이토카인인 IL-10의 발현량을 ELISA 분석을 통해 확인하였다. 구체적으로 ELISA 분석을 위해, 먼저 96 well 플레이트에 anti-TNF-α, anti-IL-10을 2μg/ml로 코팅한 후, 반응 후 차단용액 (1% BSA/PBST)으로 비특이적 결합을 차단시켰다. 이후 TNF-α, IL-10 recombinant를 1/2씩 연속 희석하여 이를 표준시약(Standard)로 사용하였고 세포배양 상층액을 넣고 실온에서 2시간 반응시켰다. 이후 바이오티닐(biotinylated) anti-TNF-α, anti-IL-10을 2시간 동안 실온에서 반응시키고 3회 세척 후, ExtraAvidin-Alkaline Phosphatase conjugate를 희석하여 첨가한 후, 실온에서 2시간 반응시켰다. 대조군은 LPS로만 세포를 자극하고 아무것도 처리하지 않은 군을 사용하였다.
그 결과, 대식세포를 활성화시키는 조건에서 RIPK1 억제제를 처리한 군은 처리하지 않은 군에 비해 LPS로 유도된 염증성 사이토카인인 TNF-α의 생성은 현저하게 억제하는 것으로 나타난 반면, 항염증성 사이토카인인 IL-10에는 영향을 미치지 않는 것으로 나타났다(도 4). 즉, RIPK1 억제제의 경우, 염증성 사이토카인의 생성은 억제하였으나, 항염증성 사이토카인의 생성은 억제하지 않는 것으로 나타났다.

류마티스
관절염 동물모델을 대상으로
RIPK1
억제제에 의한 치료 효과 분석
RIPK1 억제제가 실제로 자가면역질환이 유발된 마우스 동물에게 투여할 경우, 효과적인 치료 효능이 있는지 확인하기 위해 다음과 같은 실험을 수행하였다.관절염이 유발된 마우스를 제조하기 위해 IL-10 넉아웃(knockout) 마우스에 Type II Collagen으로 CIA(collagen-induced arthritis)를 유도하여 관절염 발병 마우스 모델을 제조한 후, 처음 Immunization 후 10일째부터 1주일에 3회 복강 내 RIPK1 억제제인 Necrostatin-1 (Selleckchem,S8037)을 0.02mg/mice로 투여한 후 관절염 지수를 분석하였다.
관절염 평가 기준은 다음과 같다.
-평가 기준-
0점: 부종이나 종창이 없다.
1점: 발 또는 발목관절에 국한된 경한 부종과 발적
2점: 발목관절에서 족근골(metatarsal)에 걸친 경한 부종과 발적
3점: 발목관절에서 족근골에 걸친 중등도의 부종과 발적
4점: 발목에서 다리 전체에 걸쳐 부종과 발적이 있는 경우
5점 이상: 부종과 발적의 증상이 심각하게 계속 진행되는 경우
그 결과, 관절염이 유도된 마우스에 RIPK1 억제제를 투여한 경우, RIPK1 억제제를 투여하지 않은 군에 비해 관절염 지수(arthritis score)가 현저하게 억제되는 것으로 나타났고, 관절염 발병 정도(incidence)도 2.5배 이상 감소하는 것으로 나타났다(도 5).
따라서, 이러한 결과를 통해 본 발명자들은 실제적으로 RIPK1의 활성을 억제할 경우, 면역질환을 치료할 수 있음을 알 수 있었다.
실시예 6. RIPK1 억제제에 의한 항체 조절 효과분석
상기 실시예 5에서 실험에 사용한 마우스들을 대상으로 RIPK1 억제제를 4주간 투여한 마우스로부터 혈액을 수득하고 다시 혈청을 분리한 후, CII specific Total IgG, CII specific IgG1, CII specific IgG2a의 발현량을 ELISA에 의해 분석하였다.
그 결과, RIPK1 억제제를 투여한 마우스의 혈청에서는 CII specific Total IgG의 발현이 감소되었고, Th2 반응과 관련 있는 CII specific IgG1의 발현은 오히려 증가되었다. 또한, Th1 반응과 관련이 있는 CII specific IgG2a의 발현도 감소되었다(도 6).
따라서, RIPK1 억제제는 Th2와 같은 항염증성 항체는 증가시키고 Th1과 같은 염증성 항체의 생성을 억제시킬 수 있음을 알 수 있었다.
실시예 7. RIPK1 억제제의 세포 독성 여부 확인
본 발명자들은 RIPK1 억제제가 세포 독성을 유발하는지 확인하기 위한 실험을 수행하였다. 구체적으로, 정상 마우스의 비장세포(splenocytes)에 RIPK1 억제제인 Necrostatin 1을 각각 0.1μM, 1μM, 10μM의 농도로 처리한 후, 3일 동안 배양하였고, MTT powder(Thiazolyl Blue Tetrazolium Bromide)를 PBS에 0.5mg/ml로 녹여 50μl씩 처리한 후 4시간을 배야하였다. 그 후, 1300rpm, 5분, 4°C에서 원심분리를 한 후 상층액 120μl을 제거하고 DMSO를 50μl 넣어준 후 540 nm에서 흡광도를 측정하였다.
그 결과, RIPK1 억제제를 처리한 군 모두에서 세포 독성이 유발되지 않음을 확인하였고, 0.1 μM, 1 μM, 10 μM 농도로 처리한 군 모두에서 세포 독성이 유발되지 않는 것으로 나타났다(도 7).
실시예 8. RIPK1 억제제에 의한 Th17 세포의 분화 억제 효과
RIPK1 억제제에 의한 Th17 세포의 분화를 억제하는 효과가 있는지를 확인하기 위한 실험을 수행하였다. 구체적으로, Th17 세포의 분화를 유도하기 위하여 정상 마우스의 비장세포(splenocytes)에 anti-CD3, anti-CD28, anti-IFN-γ, anti-IL-4, IL-6 및 TGF-β를 처리하고, RIPK1 억제제인 Necrostatin 1을 10μM 로 처리한 후 3일 동안 세포를 배양하였다. 그 후, 세포를 모은 후 anti-mouse CD4 PerCP-Cyanine5.5 와 anti-mouse IL-17 FITC 항체를 이용하여 염색하고, FACS(Fluorescence-activated cell sorting, BD FACS caliburTM)를 수행하였으며, FlowJo 프로그램을 이용하여 분석하였다.
그 결과, RIPK1 억제제를 처리하는 경우에는 대조군에 비해 Th17 세포로의 분화가 억제되었음을 확인하였다(도 8).
실시예 9.
RIPK1 억제제에 의한 파골세포 분화 억제 효과
본 발명자들은 RIPK1 억제제에 의해 파골세포 분화가 억제되는 효과가 있는지 확인하기 위한 실험을 수행하였다. 구체적으로, IL-10 넉아웃(knockout) 마우스에 Type II Collagen으로 CIA를 유도하여 관절염 발병 마우스 모델로 유도한 후, 처음 Immunization 후 10일째부터 1주일에 3회 복강 내에 RIPK-1 억제제(Necrostatin 1)를 20mg/kg 및 10mg/kg으로 투여한 후, 실험종료 시점에 마우스의 골수세포에 50 ng/ml의 RANKL(Receptor activator of nuclear factor kappa-B ligand) 과 M-CSF(macrophage colony-stimulating factor)로 자극하고, 이후 RIPK1 억제제인 Necrostatin 1을 1μM, 10μM로 처리한 다음, TRAP(Tartrate-resistant acid phosphatase) 양성 세포를 조사하였다.
그 결과, RIPK-1 억제제의 파골세포 분화 조절능을 관찰하기 위한 Osteoclastogenesis 분화 실험에서 RIPK-1 억제제를 처리하는 경우에는 골파괴 기전과 관련 있는 TRAP 양성 세포가 농도 의존적으로 감소되었음을 확인하였다 (도 9).
따라서, RIPK1 억제제가 파골세포 분화를 억제할 수 있는 효과가 있고, 골질환의 치료에 효과가 있음을 알 수 있었다.
실시예
9.
RIPK1
억제제에 의한 병인 사이토카인 및 염증성 사이토카인의 조절분석
본 발명자들은 T 세포 활성 조건인 anti-CD3로 자극 또는 대식세포 활성화 조건인 LPS(lipopolysaccharide)로 자극한 C57BL/6 정상 마우스로부터 분리한 비장세포(splenocytes)에 RIPK1 억제제인 Necrostatin-1s (Calbiochem, 504297)를 주어진 농도로 처리한 후, 48시간 배양하고, 배양액을 수집하여 병인 사이토카인인 IL-17 및 염증성 사이토카인인 TNF-α의 발현량을 ELISA 분석을 통해 확인하였다. 구체적으로, ELISA 분석을 위해, 먼저 96 well 플레이트에 anti-IL-17, anti-TNF-α를 2μg/ml로 코팅한 후, 반응 후 차단용액 (1% BSA/PBST)으로 비특이적 결합을 차단시켰다. 이후 IL-17, TNF-α recombinant를 1/2씩 연속 희석하여 이를 표준시약(Standard)로 사용하였고 세포배양 상층액을 넣고 실온에서 2시간 반응시켰다. 이후 바이오티닐(biotinylated) anti-IL-17, anti-TNF-α을 2시간 동안 실온에서 반응시키고 3회 세척 후, ExtraAvidin-Alkaline Phosphatase conjugate를 희석하여 첨가한 후, 실온에서 2시간 반응시켰다. 대조군은 anti-CD3 또는 LPS로만 세포를 자극하고 아무것도 처리하지 않은 군을 사용하였다.
그 결과, RIPK1 억제제(Necrostain 1s)를 처리한 군은 처리하지 않은 군에 비해 병인 사이토카인인 IL-17 및 염증성 사이토카인인 TNF-α의 발현량이 억제되는 효과가 있음을 확인하였다 (도 10).
실시예

RIPK1
억제제에 의한 염증성 사이토카인 및 항염증성 사이토카인의 조절분석

실시예 11. RIPK3 억제제에 의한 병인 사이토카인의 조절분석
본 발명자들은 T 세포 활성 조건인 anti-CD3로 자극한 C57BL/6 정상 마우스로부터 분리한 비장세포(splenocytes)에 RIPK3 억제제인 GSK'872 (Calbiochem, 530389)를 주어진 농도로 처리한 후, 48시간 배양하고, 배양액을 수집하여 병인 사이토카인인 IL-17의 발현량을 ELISA 분석을 통해 확인하였다. 구체적으로, ELISA 분석을 위해, 먼저 96 well 플레이트에 anti-IL-17를 2μg/ml로 코팅한 후, 반응 후 차단용액 (1% BSA/PBST)으로 비특이적 결합을 차단시켰다. 이후 IL-17 recombinant를 1/2씩 연속 희석하여 이를 표준시약(Standard)로 사용하였고 세포배양 상층액을 넣고 실온에서 2시간 반응시켰다. 이후 바이오티닐(biotinylated) anti-IL-17을 2시간 동안 실온에서 반응시키고 3회 세척 후, ExtraAvidin-Alkaline Phosphatase conjugate를 희석하여 첨가한 후, 실온에서 2시간 반응시켰다. 대조군은 anti-CD3 로만 세포를 자극하고 아무것도 처리하지 않은 군을 사용하였다.
그 결과, RIPK3 억제제를 처리한 군은 처리하지 않은 군에 비해 병인 사이토카인인 IL-17의 발현량이 현저하게 억제되는 효과가 있음을 확인하였다 (도 12).
실시예 12. RIPK3 억제제에 의한 염증성 장질환 억제 효과
본 발명자들은 RIPK3 억제제를 염증성 장질환이 유발된 마우스 동물모델에 투여하여 치료효과를 확인하고자 하였다. 구체적으로 염증성 장질환 동물모델은 C57BL6 마우스의 음수에 3% DSS(dextran sulfate sodium)를 넣어주어 제조되었다. 체중은 DSS 약물의 투여를 시작하는 시점에서 측정되었고, 동시에 RIPK3 억제제인 GSK'872 (Calbiochem, 530389)은 경구투여되어 실험이 진행되었다.
그 결과, 염증성 장질환이 유발된 마우스에 RIPK3 억제제는 대조군(WT, wild type) 정도로 체중을 회복하지는 않았으나, 염증성 장질환 마우스보다는 약간 체중이 증가되었음을 확인하였다 (도 13).
실시예 13. RIPK3 억제제에 의한 동종반응(Alloresponse) 억제 효과
본 발명자들은 RIPK3 억제제가 조직이식에 대한 거부반응을 억제하는지 확인하고자 하였다. 구체적으로, in vitro에서 96 well의 각 웰 당 1×105개의 정상 수여자(Balb/c, responder)의 CD4+T cell와 1×105개의 방사선으로 조사시킨 수여자(동종동형) 또는 공여자(C57BL/6, stimulator, 동종이형) 유래의 T 세포를 제거한 비장세포를 넣고 혼합 배양시켜 동종반응을 유도하였다. 또한, RIPK3 억제제인 GSK'872 (Calbiochem, 530389)은 1μM 및 10μM을 함께 처리한 후 72시간 배양시켰다. 배양완료 16시간 전에 배양된 세포에서 T 세포 증식 반응 정도를 3H Thymidine을 처리하고 세포증식 정도를 b 카운터로 3H Thymidine의 양을 측정하여 확인하였다.
그 결과, RIPK3 억제제는 대조군에 비해 동종반응(Alloresponse)을 억제하는 효과가 있음을 확인하였다(도 14).
따라서, RIPK3 억제제가 이식 과정에서 발생할 수 있는 T 세포의 이식 후 거부반응 및 동종반응을 효과적으로 억제하여 이식에 의한 질환 치료를 성공적으로 유도할 수 있다는 사실을 확인하였다.

- RIPK(receptor interacting serine/threonine protein kinase) 억제제를 유효성분으로 포함하는 면역질환의 예방 또는 치료용 약학적 조성물.
- 제 1 항에 있어서,상기 RIPK 억제제는 RIPK1 억제제 또는 RIPK3 억제제인 것을 특징으로 하는 조성물.
- 제 2 항에 있어서,상기 RIPK1(receptor interacting serine/threonine protein kinase 1) 억제제는 RIPK1 유전자에 특이적으로 결합하여 RIPK1의 발현을 억제하는 물질이거나, RIPK1의 단백질에 특이적으로 결합하여 RIPK1의 활성을 억제하는 물질인 것을 특징으로 하는 조성물.
- 제 3 항에 있어서,상기 RIPK1은 서열번호 1의 핵산서열로 이루어진 것을 특징으로 하는 조성물.
- 제 3 항에 있어서,상기 RIPK1의 단백질은 서열번호 2의 아미노산 서열로 이루어진 것을 특징으로 하는 조성물.
- 제 3 항에 있어서,상기 RIPK1의 발현을 억제하는 물질은 RIPK1의 유전자에 대한 안티센스 올리고뉴클레오티드, siRNA 또는 shRNA인 것을 특징으로 하는 조성물.
- 제 3 항에 있어서,상기 RIPK1의 단백질에 특이적으로 결합하여 RIPK1의 활성을 억제하는 물질은 RIPK1에 대한 항체 또는 화합물인 것을 특징으로 하는 조성물.
- 제 7 항에 있어서,상기 화합물은 Necrostatin 1 또는 Necrostatin 1s 인 것을 특징으로 하는 조성물.
- 제 2 항에 있어서,상기 RIPK3(receptor interacting serine/threonine protein kinase 3) 억제제는 RIPK3 유전자에 특이적으로 결합하여 RIPK3의 발현을 억제하는 물질이거나, RIPK3의 단백질에 특이적으로 결합하여 RIPK3의 활성을 억제하는 물질인 것을 특징으로 하는 조성물.
- 제 9 항에 있어서,상기 RIPK3은 서열번호 3의 핵산서열로 이루어진 것을 특징으로 하는 조성물.
- 제 9 항에 있어서,상기 RIPK3의 단백질은 서열번호 4의 아미노산 서열로 이루어진 것을 특징으로 하는 조성물.
- 제 9 항에 있어서,상기 RIPK3의 발현을 억제하는 물질은 RIPK3의 유전자에 대한 안티센스 올리고뉴클레오티드, siRNA 또는 shRNA인 것을 특징으로 하는 조성물.

- 제 1 항에 있어서,상기 면역질환은 류마티스 관절염 (Rheumatoid Arthritis), 골관절염, 천식 (Asthma), 피부염 (Dermititis), 건선 (Psoriasis), 낭섬유증 (Cystic Fibrosis), 고형장기 이식 후기 및 만성 거부증 (Post transplantation late and chronic solid organ rejection), 다발성 경화증 (Mμltiple Sclerosis), 전신성 홍반성 루푸스(systemic lupus erythematosus), 쇼그렌 증후군(Sjogren syndrome), 하시모토 갑상선(Hashimoto thyroiditis), 다발성근염(polymyositis), 경피증(scleroderma), 아디슨병(Addison disease), 백반증(vitiligo), 악성빈혈(pernicious anemia), 사구체신염(glomerμlonephritis) 및 폐섬유증(pμlmonary fibrosis), 염증성 장질환 (Inflammatory Bowel Dieseses), 자가면역성 당뇨 (Autoimmune Diabetes), 당뇨 망막증 (Diabetic retinopathy), 비염 (Rhinitis), 허혈-재관류 손상 (Ischemia-reperfusion injury), 혈관성형술후 재협착 (Post-angioplasty restenosis), 만성 폐색성 심장 질환 (Chronic obstructive pμlmonary diseases; COPD), 그레이브병 (Graves disease), 위장관 알러지 (Gastrointestinal allergies), 결막염 (Conjunctivitis), 죽상경화증 (Atherosclerosis), 관상동맥질환 (Coronary artery disease), 협심증 (Angina), 암 전이, 소동맥 질환, 이식편대숙주질환(graft-versus-host disease) 및 미토콘드리아 관련 증후군으로 이루어진 군으로부터 선택되는 것을 특징으로 하는 조성물.
- RIPK1(receptor interacting serine/threonine protein kinase 1) 억제제를 유효성분으로 포함하는 골 손실 억제용 약학적 조성물.
- 제 16 항에 있어서,상기 RIPK1 억제제는 Necrostatin 1 또는 Necrostatin 1s 인 것을 특징으로 하는 조성물.
- 제 16 항에 있어서,상기 RIPK1 억제제는 파골세포의 형성 및 분화를 억제하는 효능를 갖는 것을 특징으로 하는 조성물.
- 제 16 항에 있어서,상기 골 손실 억제를 통해 치료할 수 있는 질환은 골다공증, 파제트 골질환, 구루병, 골연화증 및 퇴행성 골질환으로 이루어진 군으로부터 선택되는 어느 하나인 것을 특징으로 하는 조성물.
- RIPK3(receptor interacting serine/threonine protein kinase 3) 억제제를 유효성분으로 포함하는 면역거부 질환의 예방 또는 치료용 약학적 조성물.
- 제 20 항에 있어서,상기 RIPK3 억제제는 GSK'872 인 것을 특징으로 하는 조성물.
