Abstract
본 발명은 GDF15를 유효성분으로 함유하는 면역질환의 예방 및 치료용 조성물에 관한 것으로, 본 발명의 GDF15는 TH17 세포 저하시 발현이 증가하며, 간이식 환자의 혈액에서 STAT3 처리에 따라 발현이 증가하고, GDF15 처리에 의해 Treg 세포 유도가 증가되며, 간세포의 섬유화가 억제되는 효과가 있으므로, 면역반응의 조절 이상으로 유발되는 자가면역질환, 염증성질환 및 이식거부질환과 같은 면역질환을 예방 및 치료할 수 있는 약학적 조성물 또는 면역 억제제로 유용하게 사용될 수 있다.
Description
본 발명은 GDF15를 유효성분으로 함유하는 면역질환의 예방 및 치료용 조성물에 관한 것이다.
면역체계는 크게 선천성 면역(innate immunity)과 후천성 면역(acquired immunity)으로 나눌 수 있다. 후천성 면역은 척추동물에만 존재하는 면역체계로 여러 세포들이 네트워크처럼 얽혀 있으며, 항원제시세포, B세포, T세포 등이 관여한다. 효과기 세포(Effector cell)로는 B세포, T세포가 있는데, B세포는 항체를 분비하는 체액성면역(humoral immunity)에 관여하고, T세포는 주로 세포성면역(cellular immunity)을 담당하여 세포독성 T세포(cytotoxic T cell)와 조력 T세포(helper T cell)로 분류된다. 면역계는 정상상태에서는 자가항원에 대한 특이적 면역반응을 제어하고 있으며, 외부항원에 대한 면역반응도 억제하고 있는 경우가 있는데, 예컨대 임산부의 태아에 대한 반응 및 만성감염상태에 있는 미생물에 대한 면역반응을 들 수 있다. 이러한 현상들은 항원 특이적 면역관용이 유도될 수 있는 기전으로 클론 제거(clonal deletion), 클론 무반응(anergy) 및 면역 조절 T 세포(Regulatory T cell: Treg)에 의한 능동적 통제에 의해 유도되는 것으로 알려져 있다. 이식항원에 대한 면역관용이 우연히 획득된 일부 환자나 실험적으로 면역관용을 유도한 동물모델을 조사해 보면 위의 세 가지 기전 모두 이식면역관용에 관여한다는 사실이 확인되고 있고, 특히 최근에는 면역조절 T림프구가 이식면역반응 뿐만 아니라 자가면역, 종양면역, 감염면역반응 등 생체의 거의 모든 면역반응을 통제하는데 관여하는 중요한 세포로 주목받고 있다.
각종 병원체에 대한 생체 방어 시스템으로 면역계에서 중심적 역할을 담당하는 세포군의 하나로 T 세포가 있다. T 세포는 인체의 흉선에서 생성되며 일련의 분화 과정을 거치면서 고유의 특성을 지닌 T 세포로 분화하게 되는데, 분화를 완료한 T 세포는 그 기능에 따라 크게 1형 보조 세포(Th1)와 2형 보조 세포(Th2)로 구분된다. 이 중에서 Th1 세포의 주된 기능은 세포 매개성 면역에 관여하고, Th2 세포는 체액성 면역에 관여하며, 면역계에서 이러한 두 세포 집단은 서로 과 활성화되지 않도록 서로 견제를 통해 면역계의 균형을 유지하고 있다. 따라서 면역질환의 대부분은 이러한 두 가지 면역 세포간의 불균형에 기인하는 것으로 볼 수 있는데, 예를 들어 Th1 세포의 활성이 비정상적으로 증가하는 경우 자가면역질환이 발생할 수 있고, Th2 세포의 활성이 비정상적으로 증가하는 경우 과민반응에 의한 면역질환이 발생하는 것으로 알려져 있다.
세포독성 T세포는 감염된 세포를 직접 죽임으로써 숙주를 보호하고, 여러 종류의 세포들을 움직이게 하고 조절하여 숙주 방어에 참여하고, 조력 T세포는 다른 임파구, 선천성 면역에 관여하는 세포 및 비면역세포인 상피세포에도 작용하여 숙주를 방어한다. 후천성 면역의 중요한 개념 중의 하나가 다양성이다. 각각의 T세포는 항원에 특이한 항원수용체 (T cell receptor, TCR)를 가지고 있어 다양한 병원균에 특이적으로 반응한다. 조력 T세포가 분화하기 위해서는 항원제공세포인 수지상 세포(dendritic cell)가 병원균을 찾아 임파선으로 이동하여 병원균을 만나야 한다. 수지상 세포가 항원을 제시하면 분화되지 않은 조력 T세포(naive T cell)는 수차례 분할하여 병원체에 대응할 정도의 충분한 수가 된다. 병원균 자체도 다양한 생활 방식과 면역 회피 기전을 가지고 있기 때문에 수지상 세포는 여러 기전으로 T세포에게 병원균의 양상과 위치에 관하여 정보를 제공하여, 병원체가 세포 내에 있는지 세포 외에 있는지 또는 박테리아인지 바이러스인지 아니면 진핵생물인지, 어떤 조직에 침범하고 있는지 등에 관해 알 수 있게 한다. 따라서 병원체에 특이한 T세포는 제거하고자 하는 병원균의 생활 주기에 맞추어 유전자의 발현을 하며 분비되는 사이토카인의 종류에 따라 어떤 조력 T세포를 더 많이 생성할지 또 어떤 세포들을 동원(recruitment)할지 결정한다. 조력 T세포의 분화에 가장 중요한 신호(signal)는 사이토카인이다. T 세포 분화에서 사이토카인은 Th1 또는 Th2 세포로의 분화에 필수적인 전사인자뿐만 아니라 성장인자(growth factor)를 유도 또는 억제하며, 분화된 조력 T세포 자체에서도 분비되어 다른 면역세포의 성장 및 분화를 일으키는 매개체 역할도 한다. Th1 세포에 의해 분비되는 대표적인 사이토카인이 인터페론-γ(interferon-γ)이고, Th2 세포에 의해 분비되는 대표적인 사이토카인은 IL-4, IL-5, IL-13이다. IL-17은 최근에 발견된 조력 T세포의 일종인 Th17세포의 대표적 사이토카인이다. 각각의 조력 T세포에 의해 분비되는 사이토카인은 해당 조력세포의 분화를 촉진하며 또 다른 조력 T세포의 분화를 억제한다.
한편, Th1 세포의 분화에 대한 최근 연구 결과에 따르면, 최근 그 존재가 밝혀진 Th1 세포의 활성을 조절할 수 있는 새로운 그룹인 면역조절 T 세포(Treg), 즉, 면역조절 T 림프구(Treg)는 크게 자연성(natural) Treg 와 적응성(adaptive) Treg 세포로 나눌 수 있으며, 자연성 Treg인 CD4+ CD25+ T 세포는 이 세포가 흉선에서 새로이 만들어질 때부터 면역억제기능을 부여받게 되며, 정상개체의 말초 CD4+ T 림프구 중 5 ~ 10%의 빈도로 존재한다. 아직까지 이 세포의 면역억제 기전은 정확히 파악되지 못하고 있지만, Foxp3라는 유전자의 발현 제어인자가 이 세포의 분화와 활성에 중요한 역할을 수행한다는 사실이 최근에 밝혀졌다. 또한, 말초 자연성 T 세포는 특정 환경하에서 자가 또는 외부항원의 자극을 받으면 면역억제효과를 나타내는 세포로 분화될 수 있는데, 이를 적응성(adaptive) 또는 유도성(inducible) Treg로 부르며, IL-10을 분비하는 Th2, TGF-β를 분비하는 Th3 및 CD8 Ts등이 여기에 해당한다. Treg 세포는 비정상적으로 활성화된 면역세포의 기능을 억제하여 염증 반응을 제어하는 특성이 있어, Treg 세포의 활성을 증가시키는 작용을 통해 면역질환을 치료하고자 하는 연구들이 많이 진행되고 있다. 또한, Treg 세포 이외에 T 세포의 분화 과정에서 만들어지는 또 다른 그룹으로 Th17 세포가 있는데, Th17 세포는 미분화 T세포의 분화 과정에서 Treg 세포의 분화와 유사한 과정을 거치며 형성되는 것으로 알려져 있다. 즉, Treg 세포와 Th17 세포의 분화는 공통적으로 TGF-β의 존재 하에서 이루어지지만, Treg 세포의 경우 IL-6을 필요로 하지 않는 반면, Th17 세포의 경우에는 TGF-β와 함께 IL-6가 존재하는 상황에서 분화를 한다. 또한, 분화된 Th17 세포는 IL-17을 분비하는 것을 특징으로 한다. Th17 세포는 Treg 세포와는 달리 면역질환에서 보이는 염증반응의 최전방에서 관여하여 염증 반응의 신호를 최대화시켜 질병의 진행을 가속화시키는 것이 밝혀지고 있다. 그러므로 자가면역질환 중 Treg 세포에 의해 제어되지 않는 자가면역질환의 경우, Th17 세포 활성의 억제를 표적으로 한 자가면역질환의 치료제 개발이 크게 부각되고 있다.
한편, 이식면역 거부반응의 주요 매개체는 T 세포로서, 이식편(graft)에 발현되어져 있는 주조직적합성분자(major histocompatibility complex, MHC)를 T 세포 수용체(T cell receptor)가 인지함으로써 면역반응이 유도되어 이식 거부반응이 발생하게 된다. 이와 같은 면역거부반응과 면역억제제의 부작용에 의한 사망률이 높아 효과적이고 안전한 새로운 면역억제제의 개발이 요구되고 있다. 현재 사용되고 있는 면역질환치료제로는 T 세포에서의 신호변환 경로를 차단하는 면역 억제제가 가장 많이 사용되고 있는데, 이러한 면역억제제들은 독성, 감염, 임파종, 당뇨병, 진전(tremor), 두통, 설사, 고혈압, 오심, 신기능 장애 등의 부작용이 발생하는 문제점이 있다. 또한, T 세포의 활성화를 억제하는 방법을 통해 면역질환을 치료하는 방법 이외에도 면역 세포로부터 분비되는 사이토카인의 양을 조절하는 치료법 및 면역 세포로부터 분비되는 사이토카인을 표적으로 하는 항체를 이용한 치료법이 개발 중에 있다. 그러나 이러한 방법은 실제적으로 임상실험을 거쳐 환자들에게 적용하기까지 많은 시간이 소요되어야 하고, 항체를 이용하는 방법은 항체 제작 과정에서 너무 많은 비용이 든다는 문제점이 있다.
따라서 부작용이 없고 저렴하면서도 치료 효과가 우수한 새로운 면역질환 치료제의 개발이 시급하다.
본 발명의 발명자들은 TH17 세포 저하시 GDF15 발현이 증가하며, 간이식 환자의 혈액에서 STAT3 억제제 처리에 따라 GDF15 발현이 증가하고, GDF15 처리에 의해 Treg 세포 유도가 증가되며, 간세포의 섬유화가 억제되는 것을 발견하여 본 발명을 완성하였다.
이에, 본 발명의 목적은 GDF15 단백질 또는 이를 코딩하는 핵산서열의 면역질환의 예방 또는 치료, 이식 관련 질환 또는 면역질환의 모니터링과 같은 다양한 용도를 제공하는 것이다.
본 발명의 또 다른 목적은 GDF15 단백질 또는 이를 코딩하는 핵산서열의 약학적으로 유효한 양을 개체에 투여하는 단계;를 포함하는 면역질환의 치료 방법을 제공하는 것이다.
상기 과제를 해결하기 위하여, 본 발명은 GDF15 단백질 또는 이를 코딩하는 핵산서열을 유효성분으로 포함하는 면역질환의 예방 또는 치료용 약학 조성물을 제공한다.
또한, 본 발명은 GDF15의 발현량을 확인하는 단계를 포함하는, 미트콘드리아 기능손상을 진단하는 방법을 제공한다.
또한, 본 발명은 GDF15의 발현량을 확인하는 단계를 포함하는, 혈액내 Th17 세포의 양을 모니터링하는 방법을 제공한다.
또한, 본 발명은 GDF15의 발현량을 확인하는 단계를 포함하는, 간이식 후 간이식 예후를 예측하는 방법을 제공한다.
또한, 본 발명은 GDF15의 발현양을 확인하는 단계를 포함하는 1)류마티스 관절염 또는 2)이식 후, 이식 관련 질환 치료의 예후를 모니터링하는 방법을 제공한다.
또한, 본 발명은 분리된 시료에 후보물질을 처리하는 단계; 및 상기 시료에서 GDF15의 발현량을 확인하는 단계를 포함하는, IL-17의 발현을 감소시켜 Th17 세포의 양을 감소시키며, IL-10의 발현을 증가시켜 Treg 세포의 양을 증가시키는 화합물을 스크리닝하는 방법을 제공한다.
또한, 본 발명은 GDF15의 발현량을 측정하는 제제를 포함하는 1)류마티스 관절염 또는 2)이식 후, 이식관련 질환의 예후를 모니터링하기 위한 조성물 및 이 조성물을 포함하는 1)류마티스 관절염 또는 2)이식 후, 이식관련 질환의 예후를 모니터링하기 위한 키트를 제공한다.
또한, 본 발명은 GDF15 단백질 또는 이를 코딩하는 핵산서열의 약학적으로 유효한 양을 개체에 투여하는 단계;를 포함하는 면역질환의 치료 방법을 제공한다.
본 발명에 따른 GDF15는 TH17 세포 저하 시 발현이 증가하며, 간이식 환자의 혈액에서 STAT3 억제제 처리에 따라 발현이 증가하고, GDF15 처리에 의해 Treg 세포 유도가 증가되며, 간세포의 섬유화가 억제되는 효과가 있으므로, 면역반응의 조절 이상으로 유발되는 자가면역질환, 염증성질환 및 이식거부질환과 같은 면역질환을 예방 및 치료할 수 있는 약학적 조성물 또는 면역 억제제로 유용하게 사용될 수 있다.
또한, TH17 세포 및 IL17과 GDF15의 발현과의 상관관계를 확인하여 이를 이용한 면역질환의 진단 방법 및 이식 후 이식관련 질환의 치료 예후를 예측할 수 있는 방법을 확립할 수 있다.

도 20은 본 발명의 류마티스 관절염이 유도된 마우스에서, 에크멘시아 뮤신필리아 균주의 투여에 따른 GDF15의 발현을 면역조직화학 염색으로 확인한 도이다(A: 면역조직화학 염색 결과, B: GDF 양성 면적 정량화).

이하, 첨부된 도면을 참조하여 본 발명의 구현예로 본 발명을 상세히 설명하기로 한다. 다만, 하기 구현예는 본 발명에 대한 예시로 제시되는 것으로, 당업자에게 주지 저명한 기술 또는 구성에 대한 구체적인 설명이 본 발명의 요지를 불필요하게 흐릴 수 있다고 판단되는 경우에는 그 상세한 설명을 생략할 수 있고, 이에 의해 본 발명이 제한되지는 않는다. 본 발명은 후술하는 특허청구범위의 기재 및 그로부터 해석되는 균등 범주 내에서 다양한 변형 및 응용이 가능하다.
또한, 본 명세서에서 사용되는 용어(terminology)들은 본 발명의 바람직한 실시예를 적절히 표현하기 위해 사용된 용어들로서, 이는 사용자, 운용자의 의도 또는 본 발명이 속하는 분야의 관례 등에 따라 달라질 수 있다. 따라서, 본 용어들에 대한 정의는 본 명세서 전반에 걸친 내용을 토대로 내려져야 할 것이다. 명세서 전체에서, 어떤 부분이 어떤 구성요소를 "포함"한다고 할 때, 이는 특별히 반대되는 기재가 없는 한 다른 구성요소를 제외하는 것이 아니라 다른 구성 요소를 더 포함할 수 있는 것을 의미한다.
본 발명에서 사용되는 모든 기술용어는, 달리 정의되지 않는 이상, 본 발명의 관련 분야에서 통상의 당업자가 일반적으로 이해하는 바와 같은 의미로 사용된다. 또한 본 명세서에는 바람직한 방법이나 시료가 기재되나, 이와 유사하거나 동등한 것들도 본 발명의 범주에 포함된다. 본 명세서에 참고문헌으로 기재되는 모든 간행물의 내용은 본 발명에 도입된다.
본 발명에서는 정상환자의 혈액에서 GDF15와 Th17 세포의 발현 수와 역관계를 갖는다는 점을 명확히 확인하였고(도 1), GDF15와 미토콘드리아와 기능이상과 양의 상관관계를 확인하였다.
또한, 간이식 환자의 혈액에서 STAT3 억제제가 처리되는 경우, Th17 세포가 감소하고, Treg가 유도되며, GDF15의 발현양이 증가됨을 확인하여, GDF15의 발현양을 확인함으로써, 간이식 거부질환의 후보 치료제의 치료효과 예측시, Th17세포와 Treg의 항상상 유지여부를 평가할 수 있는 마커로 이용될 수 있음을 확인하였다(도 2).
또한, 간이식 환자에서 IL-17 발현과 GDF15의 발현과 상관관계를 확인하여 음의 상관관계가 있음을 확인하여, GDF15를 간이식 환자의 상태를 모니터링할 수 있는 마커로 이용가능할 수 있음을 확인하였다(도 8).
또한, 기존 Th17과 Treg 세포의 항상성을 유지시킬 수 있다고 알려진 SD282 화합물을 간이식 환자의 면역세포에 처리 후, GDF15의 발현양을 확인한 경우, SD282 화합물이 GDF15의 발현양을 증가시킴을 확인하였다(도 9). 또한, 이식거부질환(GVHD) 동물 모델에 SD282 화합물을 처리하는 경우, 이식거부질환의 치료효과와 GDF15의 발현량과의 상관관계를 확인하였다(도 10). 또한, 간이식 동물 모델에서 SD282의 치료효과의 IL-10 발현량, IL-17의 발현량 및 GDF15 발현량과의 관계를 확인하였고, GDF15 발현량이 현저히 증가함을 확인하였고, IL-10의 발현이 증가함을 확인하였으나, IL-17의 발현이 감소됨을 확인하였다(도 11, 12). 상기 실험 결과를 근거로, GDF15의 발현양을 확인하여, 이식거부질환 치료제 또는 Th17를 감소시키고, Treg 세포를 증가시키는 후보물질을 스크리닝할 수 있는 방법, 면역환자에 있어서 면역상태를 모니터링할 수 있는 키트로 이용가능할 수 있음을 확인하였다(도 9 내지 12).
또한, 류마티스 관절염에서 SD282 화합물을 처리하는 경우, 관절손상 및 염증세포 침윤이 현저하게 감소됨과 더불어, GDF15 발현 세포가 증가됨을 확인하였다. 이를 근거로 상기 GDF15의 경우, 류마티스 관절염 환자의 치료효과를 모니터링할 수 있는 키트로 이용가능할 수 있음을 확인하였다(도 14).
또한, 정상인의 혈액에서 GDF15의 처리시 IL-10의 증가를 확인하였고, 간이식 환자 혈액에서도 IL-10 및 Treg 유도를 확인하였다(도 3, 4).
또한, GDF15가 TGF-β 및 α-SMA의 mRNA 수준이 감소하는 것으로 나타나, GDF15가 섬유증을 억제하는 효과가 있음을 확인하였다(도 5)
또한, 간 이식질환 환자의 면역세포에서 GDF15를 처리하는 경우, IL-10 분비 및 Treg 유도효과를 확인하여, GDF15가 이식거부질환등의 질환에 이용가능할 수 있음을 확인하였다(도 6).
또한, 쇼그렌 전임상 모델에서, GDF15를 유전자 치료제 형태로 투여하는 경우, 동물 모델의 체중이 증가하고, 질병의 스코어가 감소됨을 확인하여, GDF15를 쇼그렌 증후군의 예방 또는 치료에 이용할 수 있음을 확인하였다(도 7).
본 발명의 “GDF15”는 성장 분화 인자 15(Growth differentiation factor 15)로서, 세포 스트레스 및 질병에 관여하며, TGFβ 슈퍼패밀리의 구성원으로서 공지되어 있는 인자이다. GDF15는 1997년 subtraction cloning을 통해 처음 발견되었으며, 전염증성 사이토카인에 반응하여 분비가 증가하여, 후기 대식세포의 추가 활성화를 제한하는 것이 확인되면서, 대식세포 억제 사이토카인(macrophage inhibitory cytokine-1, MIC-1)으로 명명되기도 하였다.
한편, 이러한 GDF15는 심혈관질환과 관련된 예측 바이오마커로 알려져 있고, 심장이나 근육 손상 시에 혈청 내에서 증가하는 것으로 알려져 있다. 이러한 발현의 증가는 조직이나 세포의 재생에 관여하는 것으로 파악되고 있으며, 심혈관 쥐 모델에서 GDF15는 심장의 비대와 혀혈성 재관류 손상을 보호하는 기능이 있고, 대식세포의 항염증 기능을 가지는 M2의 분화를 촉진함이 알려져있다. 또한 일부 암 질환에서 암세포의 사멸을 유도함이 알려져 있으며, 특히 미토콘드리아 기능 이상을 가진 동물 모델에서 GDF15는 미토콘드리아의 기능을 회복하고 세포 대사를 조절 할 수 있음이 알려져 있다. 따라서 GDF15의 발현은 증가시키는 것은, 세포의 항염기능을 향상 시키고, 세포의 대사 조절 (미토콘드리아 기능회복)을 할 수 있음을 의미한다.
본 발명의 용어 "SD282 화합물"은 하기 화학식 1로 표현되는 화합물(N-Ethyl-4-fluorophenyl biguanide)일 수 있으며, 하기 화학식은 한국 특허출원 10-2014-0155182에서 개시된 화합물이다.

본 발명의 용어 "SD217 화합물"은 하기 화학식 2로 표현되는 화합물(1-(2,4-difluorophenyl)biguanide)일 수 있으며, 하기 화학식은 한국 특허출원 10-2014-0110919에서 개시된 화합물이다.

따라서, 일 측면에서, 본 발명은 GDF15 (growth/differentiation factor 15) 단백질 또는 이를 코딩하는 핵산서열을 유효성분으로 포함하는 면역질환의 예방 또는 치료용 약학 조성물에 관한 것이다.
일 구현예에서, 면역질환은 IL-10 감소되고 및 IL-17의 발현이 증가되어, Treg의 수가 감소되었고, Th17 세포의 수가 증가된 면역질환으로, 예를 들어, 자가면역질환, 염증성질환 및 세포, 조직 또는 기관의 이식거부(transplantation rejection)질환으로 이루어진 군 중에서 선택된 어느 하나 이상일 수 있다.
일 구현예에서, 상기 조직 또는 기관은 간 조직 또는 간일 수 있다.
일 구현예에서, 상기 면역질환은 이식 거부질환, 이식질환, 섬유증 등을 포함할 수 있다. 또한, 상기 면역질환은 예를 들어, 간섬유증, 간이식 후 섬유증, 간이식 질환, 이식거부질환, 쇼그렌 증후군, 이식편대숙주질환(GVHD) 등을 포함할 수 있다.
일 구현예에서, 상기 이식은 간 세포 또는 조직의 이식일 수 있다.
성공적인 장기 이식을 위해서는 이식할 세포 및 장기에 대한 수혜자의 면역 거부반응을 극복해야 한다. 이식면역거부반응의 주요 매개체는 T 세포로서, 이식편(graft)에 발현되어져 있는 주조직적합성분자(major histocompatibility complex, MHC)를 T 세포 수용체(T cell receptor)가 인지함으로써 면역반응이 유도되어 이식거부반응이 발생되게 된다.
일 구현예에서, 상기 GDF15 단백질을 코딩하는 핵산서열은 서열번호 1 내지 4 중 어느 하나로 표시되는 염기서열을 포함할 수 있다.
일 구현예에서, 상기 GDF15 단백질은 서열번호 5 내지 8 중 어느 하나의 서열을 포함할 수 있다.
일 구현예에서, 상기 GDF15 단백질을 코딩하는 핵산서열은 미분화 T 세포의 Th17 세포로의 분화를 감소 또는 억제할 수 있다.
일 구현예에서, 상기 GDF15 단백질을 코딩하는 핵산서열은 IL-10의 분비를 증가시키고 조절 T 세포(Regulatory T cell: Treg)를 유도할 수 있으며, 구체적으로 Treg의 활성 또는 증폭을 촉진 또는 증가시킬 수 있다.
Foxp3는 흉선(thymus)으로부터 유래하는 면역조절 T 세포(Regulatory T cell)에 주로 존재하고, CD4+ CD25+ 표지 항원을 가진 세포에 존재하는 전사 조절 인자 (transcriptional factor)로서, 그 기능은 Foxp3를 발현하는 T 세포에 대한 항원 인지시 항원에 대해 저반응성을 가짐과 동시에 흉선으로부터 분화되어 나온 Foxp3를 발현하지 않는 CD4+ CD25- T 세포 중 잠재적으로 자가 면역증을 유발할 수 있는 T 세포들에 대하여 IL-2의 생성과 세포분열 현상을 억제하는 억제자 T 세포 (suppressor T cell)로서의 역할을 가지고 있다. 또한, Foxp3는 Foxp3를 발현하는 조절 T 세포 및 이와 세포-세포간 접촉(cell-cell contact)을 통해 CD25- T 세포에 대하여서는 IL-2 뿐만이 아니라 전사인자인 NFAT의 영향을 받는 IL-4, IFN-감마 등의 전사 조절을 억제하는 기능을 하는 것으로 밝혀져 있다. 따라서 상기와 같은 작용을 하는 Foxp3를 발현하고 있는 T 세포의 경우, 면역반응을 억제 또는 조절하는 작용을 통해 면역질환을 치료하는데 응용되고 있고, 또한, 인간에 존재하는 Foxp3를 발현하는 CD4 T 세포의 자가 항원 특이적인 T 세포(self-antigen specific T cell clone)를 고농도의 IL-2 사이토카인 처리 및 항-CD3, 항-CD28 항체와의 조합처리를 통해서 그 수를 증가시켜 세포 치료방법으로 응용하고자 하는 시도들이 있어왔다. 따라서 Foxp3의 발현 확인은 Treg 세포의 활성 또는 증폭을 측정할 수 있는 지표가 된다.
본 발명에서 상기 "활성(activation)"이란 생체 내에서 조절 T 세포(Regulatory T cell: Treg), 즉, 자연성(natural) Treg 와 적응성(adaptive) Treg 세포를 모두 포함하는 Treg 세포가 가지는 모든 기작이 촉진 또는 증진되는 것을 말하며, 생체 내의 면역반응이 정상상태를 유지하도록 면역조절작용, 예컨대 면역억제반응이 촉진 또는 증진되는 것을 말한다.
또한, 상기 "증폭(expansion)"이란 미분화 T 세포가 조절 T 세포로 분화 및 증식되는 것을 말하는 것으로서, '분화(differentiation)'는 세포가 분열 증식하여 성장하는 동안에 서로 구조나 기능이 특수화하는 현상, 즉 생물의 세포, 조직 등이 각각에게 주어진 일을 수행하기 위하여 형태나 기능이 변해가는 것을 말하며, '증식(proliferation)' 은 세포가 분열되어 동질의 것이 불어나는 것으로서 보통 다세포생물의 체내에서 세포수가 증가되어 가는 것을 말한다.
일 구현예에서, GDF15 단백질을 코딩하는 핵산서열은 TGF-β 및 α-SMA의 mRNA 발현수준을 감소시켜 섬유화, 예를 들어, 장기의 섬유화를 억제할 수 있다. 상기 장기는 예를 들어 간일 수 있다.
일 구현예에서, GDF15 또는 이의 단편이 조성물에 0.2 내지 200 ng/ml로 포함될 수 있다.
본 발명에서 상기 "면역질환"은 포유류 면역계의 구성성분들이 포유류의 병리상태를 야기하거나, 매개하거나 또는 기타 공헌하는 질환을 의미한다. 또한, 면역 반응의 자극 또는 중단이 그 질병의 진행에 보상적인 효과를 갖는 질환을 모두 포함할 수 있는데, 본 발명에서는 과민성 면역반응으로 인해 야기되는 질환들을 포함할 수 있다. 이러한 면역질환의 예로는 이에 제한되지는 않으나, 자가면역질환; 염증성질환; 및 세포, 조직 또는 기관의 이식거부(transplantation rejection)질환 등을 모두 포함할 수 있다.
또한, 모든 정상 개체에 있어서 가장 중요한 특성 중의 하나는 자기(self)를 구성하고 있는 항원물질에 대해서는 해롭게 반응하지 않는 반면, 비자기(non-self) 항원들에 대해서는 이를 인식하고 반응하여 제거할 수 있는 능력을 가지고 있다. 이처럼 자기항원에 대한 생체의 무반응을 면역학적 무반응성(immunologic unresponsiveness) 또는 관용(tolerance)이라고 한다. 그러나 이러한 자기관용을 유도하거나 계속 유지하는데 있어서 문제가 생기게 되면 자기항원에 대하여 면역반응이 일어나게 되고, 이로 인하여 자신의 조직을 공격하는 현상이 발생하는데 이러한 과정에 의해 발생되는 질환을 "자가면역질환"이라고 한다.
또한, "염증성 질환"이란 염증유발인자 또는 방사선조사 등 유해한 자극으로 인해 인체 면역체계를 과도하게 항진시켜 대식세포와 같은 면역세포에서 분비되는 TNF-α(tumor necrosis factor-α), IL-1(interleukin-1), IL-6, 프로스타글란딘(prostagladin), 루코트리엔(luecotriene) 또는 산화질소(nitric oxide, NO)와 같은 염증 유발물질(염증성 사이토카인)에 의해 유발되는 질환을 말한다.
본 명세서에서 사용되는 용어 "치료"는 이롭거나 바람직한 임상적 결과를 수득하기 위한 접근을 의미한다. 본 발명의 목적을 위해서, 이롭거나 바람직한 임상적 결과는 비제한적으로, 증상의 완화, 질병 범위의 감소, 질병 상태의 안정화 (즉, 악화되지 않음), 질병 진행의 지연 또는 속도의 감소, 질병 상태의 개선 또는 일시적 완화 및 경감 (부분적이거나 전체적으로), 검출가능하거나 또는 검출되지 않거나의 여부를 포함한다.
또한, "치료"는 치료를 받지 않았을 때 예상되는 생존율과 비교하여 생존율을 늘이는 것을 의미할 수도 있다. "치료"는 치료학적 치료 및 예방적 또는 예방조치 방법 모두를 가리킨다. 상기 치료들은 예방되는 장애뿐만 아니라 이미 발생한 장애에 있어서 요구되는 치료를 포함한다. 질병을 "완화(Palliating)"하는 것은 치료를 하지 않은 경우와 비교하여, 질병상태의 범위 및/또는 바람직하지 않은 임상적 징후가 감소되거나 및/또는 진행의 시간적 추이(time course)가 늦춰지거나 길어지는 것을 의미한다.
상기 치료란, 달리 언급되지 않는 한, 상기 용어가 적용되는 질환 또는 질병, 또는 상기 질환 또는 질병의 하나 이상의 증상을 역전시키거나, 완화시키거나, 그 진행을 억제하거나, 또는 예방하는 것을 의미하며, 본원에서 사용된 상기 치료란 용어는 치료하는이 상기와 같이 정의될 때 치료하는 행위를 말한다. 따라서 포유동물에 있어서 면역질환의 치료 또는 치료요법은 하기의 하나 이상을 포함할 수 있다:
(1) 면역질환의 성장을 저해함, 즉, 그 발달을 저지시킴,
(2) 면역질환의 확산을 예방함, 즉, 전이를 예방함,
(3) 면역질환을 경감시킴.
(4) 면역질환의 재발을 예방함, 및
(5) 면역질환의 증상을 완화함(palliating).
만약, 수혜동물이 조성물의 투여에 견딜 수 있거나, 조성물의 그 동물에의 투여가 적합한 경우라면, 조성물은 "약학적으로 또는 생리학적으로 허용가능함"을 나타낸다. 투여된 양이 생리학적으로 중요한 경우에는 상기 제제는 "치료학적으로 유효량"으로 투여되었다고 말할 수 있다. 상기 제제의 존재가 수혜 환자의 생리학적으로 검출가능한 변화를 초래한 경우라면 상기 제제는 생리학적으로 의미가 있다.
본 명세서에서 사용되는 "유효량"은, 이롭거나 바람직한 임상적 또는 생화학적 결과에 영향을 주는 적절한 양이다. 유효량은 한번 또는 그 이상 투여될 수 있다. 본 발명의 목적을 위하여, 촉진제의 유효량은 관련 질병 상태의 진행을 일시적으로 완화, 개선, 안정화, 되돌림, 속도를 늦춤 또는 지연시키는데 적절한 양이다.
일 측면에서, 본 발명은 GDF15 또는 이의 단편을 암호화한 폴리뉴클레오티드를 포함하는 벡터 또는 상기 벡터를 포함하는 세포를 유효 성분으로 포함하는 면역질환의 예방 및 치료용 조성물에 관한 것이다.
상기 벡터는 도 13에 도시된 벡터 맵과 같이 제작될 수 있으나, 이에 제한되지 않는다.
본 명세서에서 사용되는 용어 "벡터"는 숙주 세포에 삽입되어 숙주 세포 게놈과 재조합되고 이에 삽입되거나, 또는 에피좀으로서 자발적으로 복제하는 컴피턴트 뉴클레오티드 서열을 포함하는 임의의 핵산을 의미한다. 이러한 벡터로는 선형 핵산, 플라스미드, 파지미드, 코스미드, RNA 벡터, 바이러스 벡터 등이 있다.
본 명세서에서 사용되는 용어 "숙주 세포"는 하나 이상의 DNA 또는 벡터가 도입되는 진핵 또는 원핵 세포를 가리키며, 특정 대상 세포만이 아니라 그 자손 혹은 잠재적 자손까지도 가리키는 것으로 이해되어야 한다. 어떤 변형이 돌연변이 혹은 환경적 영향 때문에 후속 세대에 일어날 수 있기 때문에 사실 상기 자손은 부모 세포와 동일하지는 않지만, 본 명세서에서 사용된 바와 같이 상기 용어의 범주 내에서 여전히 포함된다.
본 발명에서, 용어 "예방"이란 본 발명에 따른 단백질 또는 이의 단편, 또는 이를 포함하는 조성물의 투여에 의해 면역질환의 발생, 확산 및 재발을 억제 또는 지연시키는 모든 행위를 의미한다.
본 발명의 조성물의 치료적으로 유효한 양은 여러 요소, 예를 들면 투여방법, 목적부위, 환자의 상태 등에 따라 달라질 수 있다. 따라서, 인체에 사용 시 투여량은 안전성 및 효율성을 함께 고려하여 적정량으로 결정되어야 한다. 동물실험을 통해 결정한 유효량으로부터 인간에 사용되는 양을 추정하는 것도 가능하다. 유효한 양의 결정시 고려할 이러한 사항은, 예를 들면 Hardman and Limbird, eds., Goodman and Gilman's The Pharmacological Basis of Therapeutics, 10th ed.(2001), Pergamon Press; 및 E.W. Martin ed., Remington's Pharmaceutical Sciences, 18th ed.(1990), Mack Publishing Co.에 기술되어있다.
본 발명의 약학 조성물은 약학적으로 유효한 양으로 투여한다. 본 발명에서 사용되는 용어, "약학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분하며 부작용을 일으키지 않을 정도의 양을 의미하며, 유효용량 수준은 환자의 건강상태, 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 방법, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 배합 또는 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고, 종래의 치료제와 순차적으로 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여, 부작용없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.
본 발명의 약학 조성물은 생물학적 제제에 통상적으로 사용되는 담체, 희석제, 부형제 또는 둘 이상의 이들의 조합을 포함할 수 있다. 본 발명에서 사용되는 용어, "약학적으로 허용가능한"이란 상기 조성물에 노출되는 세포나 인간에게 독성이 없는 특성을 나타내는 것을 의미한다. 상기 담체는 조성물을 생체 내 전달에 적합한 것이면 특별히 제한되지 않으며, 예를 들면, Merck Index, 13th ed., Merck & Co. Inc. 에 기재된 화합물, 식염수, 멸균수, 링거액, 완충 식염수, 덱스트로스 용액, 말토 덱스트린 용액, 글리세롤, 에탄올 및 이들 성분 중 1 성분 이상을 혼합하여 이용할 수 있으며, 필요에 따라 항산화제, 완충액, 정균제 등 다른 통상의 첨가제를 첨가할 수 있다. 또한, 희석제, 분산제, 계면활성제, 결합제 및 윤활제를 부가적으로 첨가하여 수용액, 현탁액, 유탁액 등과 같은 주이용 제형, 환약, 캡슐, 과립 또는 정제로 제제화할 수 있다. 더 나아가 당 분야의 적정한 방법으로 또는 Remington's Pharmaceutical Science(Mack Publishing Company, Easton PA, 18th, 1990)에 개시되어 있는 방법을 이용하여 각 질환에 따라 또는 성분에 따라 바람직하게 제제화할 수 있다.
일 구현예에서, 상기 약학 조성물은 경구형 제형, 외용제, 좌제, 멸균 주사용액 및 분무제를 포함하는 군으로부터 선택되는 하나 이상의 제형일 수 있다.
본 발명의 조성물은 또한 생물학적 제제에 통상적으로 사용되는 담체, 희석제, 부형제 또는 둘 이상의 이들의 조합을 포함할 수 있다. 약학적으로 허용 가능한 담체는 조성물을 생체 내 전달에 적합한 것이면 특별히 제한되지 않으며, 예를 들면, Merck Index, 13th ed., Merck & Co. Inc. 에 기재된 화합물, 식염수, 멸균수, 링거액, 완충 식염수, 덱스트로스 용액, 말토 덱스트린 용액, 글리세롤, 에탄올 및 이들 성분 중 1 성분 이상을 혼합하여 이용할 수 있으며, 필요에 따라 항산화제, 완충액, 정균제 등 다른 통상의 첨가제를 첨가할 수 있다. 또한, 희석제, 분산제, 계면활성제, 결합제 및 윤활제를 부가적으로 첨가하여 수용액, 현탁액, 유탁액 등과 같은 주이용 제형, 환약, 캡슐, 과립 또는 정제로 제제화할 수 있다. 더 나아가 당 분야의 적정한 방법으로 또는 Remington's Pharmaceutical Science(Mack Publishing Company, Easton PA, 18th, 1990)에 개시되어 있는 방법을 이용하여 각 질환에 따라 또는 성분에 따라 바람직하게 제제화할 수 있다.
본 발명의 조성물에 추가로 동일 또는 유사한 기능을 나타내는 유효성분을 1종 이상 함유할 수 있다. 본 발명의 조성물은, 조성물 총 중량에 대하여 상기 단백질을 0.0001 내지 10 중량 %로, 바람직하게는 0.001 내지 1 중량 %를 포함한다.
본 발명의 약학 조성물은 약제학적으로 허용 가능한 첨가제를 더 포함할 수 있으며, 이때 약제학적으로 허용 가능한 첨가제로는 전분, 젤라틴화 전분, 미결정셀룰로오스, 유당, 포비돈, 콜로이달실리콘디옥사이드, 인산수소칼슘, 락토스, 만니톨, 엿, 아라비아고무, 전호화전분, 옥수수전분, 분말셀룰로오스, 히드록시프로필셀룰로오스, 오파드라이, 전분글리콜산나트륨, 카르나우바 납, 합성규산알루미늄, 스테아린산, 스테아린산마그네슘, 스테아린산알루미늄, 스테아린산칼슘, 백당, 덱스트로스, 소르비톨 및 탈크 등이 사용될 수 있다. 본 발명에 따른 약제학적으로 허용 가능한 첨가제는 상기 조성물에 대해 0.1 중량부 내지 90 중량부 포함되는 것이 바람직하나, 이에 한정되는 것은 아니다.
본 발명의 조성물은 목적하는 방법에 따라 비 경구 투여(예를 들어 정맥 내, 피하, 복강 내 또는 국소에 적용)하거나 경구 투여할 수 있으며, 투여량은 환자의 체중, 연령, 성별, 건강상태, 식이, 투여시간, 투여방법, 배설률 및 질환의 중증도 등에 따라 그 범위가 다양하다. 본 발명에 따른 조성물의 일일 투여량은 0.0001 ~ 10 ㎎/㎖이며, 바람직하게는 0.0001 ~ 5 ㎎/㎖이며, 하루 일 회 내지 수회에 나누어 투여하는 것이 더욱 바람직하다.
본 발명의 조성물의 경구 투여를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데, 통상적으로 사용되는 단순 희석제인 물, 액체 파라핀 이외에 다양한 부형제, 예컨대 습윤제, 감미제, 방향제, 보존제 등이 함께 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성 용제, 현탁제, 유제, 동결건조 제제, 좌제 등이 포함된다.
일 측면에서, 본 발명은 GDF15의 발현량을 확인하는 단계를 포함하는, 미트콘드리아 기능손상을 진단하는 방법 및 혈액내 Th17 세포의 양을 예측하는 방법에 관한 것이다.
일 구현예에서, 상기 GDF15의 발현량이 감소할 경우, 미토콘드리아의 기능이 손상되고, 혈액내 Th17 세포량이 증가한 것일 수 있다.
일 측면에서, 본 발명은 GDF15의 발현량을 확인하는 단계를 포함하는, 간이식 후 간이식 예후를 모니터링하는 방법에 관한 것이다.
일 구현예에서, 상기 GDF15의 발현량이 감소할 경우, 간이식 후 섬유증, 이식면역거부반응 등의 증상이 나타날 가능성이 높아 예후가 좋지 않은 것으로 판단할 수 있다.
일 측면에서, 본 발명은 GDF15의 발현양을 확인하는 단계를 포함하는 1)류마티스 관절염 또는 2)이식 후, 이식 관련 질환 치료의 예후를 모니터링하는 방법에 관한 것이다.
이식면역거부반응을 감소시키기 위해 면역억제제들이 사용되고 있는데 이러한 면역억제제들의 공통된 목적은 이식편에 대한 T 세포-매개 면역반응을 억제하는 것이다.
이러한 면역억제제의 투여 후 GDF15의 발현량이 증가할 경우 예후가 좋은 것으로 판단할 수 있으나, 이에 제한되지 않는다.
또한, 류마티스 관절염 또는 이식 관련 질환의 치료를 위한 치료제 또는 치료후보 물질의 투여 후 GDF15의 발현량이 증가할 경우 예후가 좋은 것으로 판단할 수 있으나, 이에 제한되지 않는다.
상기 치료제 또는 치료후보 물질은 하기 화학식 1로 표시되는 화합물 또는 STAT3 억제제일 수 있으나, 이에 제한되지 않는다.

본 발명에서 사용되는 용어 “배양액”은 미생물이 증식하기에 적합한 요소가 포함된 배지에, 목적 미생물을 종균하여, 일정시간 배양한 후 수득되는 것으로서, 미생물 및 배지를 포함하는 개념이다.
일 측면에서, 본 발명은 분리된 시료에 후보물질을 처리하는 단계; 상기 시료에서 GDF15의 발현량을 확인하는 단계를 포함하는, IL-17의 발현을 감소시켜 Th17 세포의 양을 감소시키며, IL-10의 발현을 증가시켜 Treg 세포의 양을 증가시키는 화합물을 스크리닝하는 방법에 관한 것이다.
일 측면에서, 본 발명은 GDF15의 발현양을 측정하는 제제를 포함하는 1)류마티스 관절염 또는 2)이식 후, 이식관련 질환의 예후를 모니터링 하기 위한 조성물에 관한 것이다.
상기 GDF15의 발현양을 측정하는 제제는 GDF15의 발현양을 mRNA 또는 단백질 수준에서 측정하는 제제라면 제한 없이 사용할 수 있으나, 이에 제한되지 않는다.
일 구현예에서, 상기 GDF15의 발현양을 mRNA 수준에서 측정하는 제제는, 마커의 핵산서열, 상기 핵산서열에 상보적인 핵산서열, 상기 핵산서열 및 상보적인 서열의 단편을 특이적으로 인식하는 프라미어 쌍, 프로브, 또는 프라이머 쌍 및 프로브를 포함할 수 있으며, 이의 측정은 중합효소연쇄반응, 실시간 RT-PCR (Real-time RT-PCR), 역전사 중합효소연쇄반응, 경쟁적 중합효소연쇄반응(Competitive RT-PCR), Nuclease 보호 분석(RNase, S1 nuclease assay), in situ 교잡법, 핵산 마이크로어레이, 노던블랏 또는 DNA 칩으로 이루어진 군으로부터 선택되는 방법으로 수행될 수 있다.
일 구현예에서, 상기 GDF15의 발현양을 단백질 수준에서 측정하는 제제는, 상기 마커의 단백질 전장 또는 그 단편을 특이적으로 인식하는 항체, 항체단편, 앱타머(aptamer), 아비머(avidity multimer) 또는 펩티도모방체(peptidomimetics)를 포함할 수 있으며, 이의 측정은 웨스턴블랏, ELISA(enzyme linked immunosorbent assay), 방사선면역분석(RIA: Radioimmunoassay), 방사면역확산법(radioimmunodiffusion), 면역 전기영동, 조직면역염색, 면역침전 분석법(Immunoprecipitation assay), 보체 고정 분석법(Complement Fixation Assay), FACS, 질량분석 또는 단백질 마이크로어레이로 이루어진 군으로부터 선택되는 방법으로 수행될 수 있다.
본 발명에서 사용된 용어 "검출" 또는 "측정"은 검출 또는 측정된 대상의 농도를 정량하는 것을 의미한다.
본 발명에서 용어, "프라이머"는 짧은 자유 3말단 수산화기 (free 3 hydroxyl group)를 가지는 핵산 서열로 상보적인 템플레이트(template)와 염기쌍 (base pair)를 형성할 수 있고 템플레이트 가닥 복사를 위한 시작 지점으로 기능을 하는 짧은 핵산 서열을 의미한다. 프라이머는 적절한 완충용액 및 온도에서 중합반응 (즉, DNA 폴리머레이트 또는 역전사효소)을 위한 시약 및 상이한 4가지 뉴클레오사이드 트리포스페이트의 존재하에서 DNA 합성이 개시할 수 있다.
본 발명에서 용어, "프로브"란 mRNA와 특이적 결합을 이룰 수 있는 짧게는 수 염기 내지 길게는 수백 염기에 해당하는 RNA 또는 DNA 등의 핵산 단편을 의미하며 라벨링 되어 있어서 특정 mRNA의 존재 유무를 확인할 수 있다. 프로브는 올리고 뉴클레오타이드(oligonucleotide) 프로브, 단쇄 DNA(single stranded DNA) 프로브, 이중쇄 DNA(double stranded DNA) 프로브, RNA 프로브 등의 형태로 제작될 수 있다. 본 발명에서는 상기 AFP, HMMR, NXPH4, PITX1, THBS4 및/또는 UBE2T 유전자와 상보적인 프로브를 이용하여 혼성화를 실시하여, 혼성화 여부를 통해 상기 유전자 발현 정도를 진단할 수 있다. 적당한 프로브의 선택 및 혼성화 조건은 통상의 기술분야에 공지된 것을 기초로 변형할 수 있으므로 본 발명에서는 이에 대해 특별히 한정하지 않는다.

N.Y.(1999)에서 확인할 수 있다. 예를 들어, 상기 엄격조건 중에서 고 엄격조건은 0.5 M NaHPO4, 7% SDS(sodium dodecyl sulfate),1mM EDTA에서 65℃ 조건으로 혼성화하고, 0.1 x SSC(standard saline citrate)/0.1% SDS에서 68℃ 조건으로 세척하는 것을 의미한다. 또는, 고 엄격조건은 6 x SSC/0.05% 소듐 파이로포스페이트에서 48℃ 조건으로 세척하는 것을 의미한다. 저 엄격조건은 예를 들어, 0.2 x SSC/0.1% SDS에서 42℃ 조건으로 세척하는 것을 의미한다.
본 발명에서 용어, "항체"란 당해 분야에서 공지된 용어로서 항원성 부위에 대해서 지시되는 특이적인 단백질 분자를 의미한다. 본 발명의 목적상, 항체는 본 발명의 마커인 AFP, HMMR, NXPH4, PITX1, THBS4 및/또는 UBE2T 유전자에서 발현되는 단백질에 대해 특이적으로 결합하는 항체를 의미하며, 상기 항체의 제조방법은 널리 공지된 방법을 사용하여 제조할 수 있다. 여기에는 상기 단백질에서 만들어질 수 있는 부분 펩티드도 포함된다. 본발명의 항체의 형태는 특별히 제한되지 않으며 폴리클로날 항체, 모노클로날 항체 또는 항원 결합성을 갖는 것이면 그것의 일부도 본 발명의 항체에 포함되고 모든 면역 글로불린 항체가 포함된다. 나아가, 본 발명의 항체에는 인간화 항체 등의 특수 항체도 포함된다.

본 발명에서 용어 "면역질환 진단 키트"는 본 발명의 면역질환 진단용 조성물이 포함된 키트를 의미한다. 따라서, 상기 표현 "면역질환 진단 키트"는 "면역질환 진단용 조성물"과 서로 교차 또는 혼용하여 사용이 가능하다. 본 명세서에서 용어 "진단"은 특정 질병 또는 질환에 대한 한 객체의 감수성(susceptibility)을 판정하는 것, 한 객체가 특정 질병 또는 질환을 현재 가지고 있는 지 여부를 판정하는 것, 특정 질병 또는 질환에 걸린 한 객체의 예후(prognosis)(예컨대, 면역질환 상태의 동정, 면역질환의 단계 결정 또는 치료에 대한 면역질환의 반응성 결정)를 판정하는 것, 또는 테라메트릭스(therametrics)(예컨대, 치료 효능에 대한 정보를 제공하기 위하여 객체의 상태를 모니터링 하는 것)을 포함한다.

일 측면에서, 본 발명은 GDF15 또는 이의 단편의 미토콘드리아의 기능 활성화 용도에 관한 것이다. 미토콘드리아 활성이 조절되지 못하는 상황에서는 더 심한 염증이 생기며 이로 인하여 면역염증 질환의 발생에 미토콘드리아의 이상 반응이 매우 중요하다고 알려져 있다. 미토콘드리아의 활성이 저하될 경우에는 병인 세포의 제어 시스템이 불안하게 되고 이로 인하여 과도한 세포의 활성이나 증식을 유발하게 된다. 이와 관련하여, 본 발명의 GDF15의 발현은 미토콘드리아의 기능과 양의 상관관계에 있으므로, 이를 미토콘드리아 기능 활성화 또는 미토콘드리아 기능 확인을 위한 마커로서 사용할 수 있다.
일 측면에서, 본 발명은 GDF15 또는 이의 단편을 처리하는 단계를 포함하는 시험관(in vitro)내에서 미분화 T 세포의 Th17 세포로의 분화를 감소 또는 억제하는 방법에 관한 것이다.
일 측면에서, 본 발명은 GDF15 또는 이의 단편을 조절 T 세포(Regulatory T cell: Treg)에 처리하는 단계를 포함하는 조절 T 세포를 활성화시키는 방법에 관한 것이다.
일 구현예에서, 활성화된 조절 T 세포는 Foxp3의 발현이 증가될 수 있다.
또한, 본 발명은 GDF15 단백질 또는 이를 코딩하는 핵산서열의 약학적으로 유효한 양을 개체에 투여하는 단계;를 포함하는 면역질환의 치료 방법을 제공한다.
본 발명의 치료 방법은 상기 유효성분을 치료적 유효량으로 개체에 투여하는 것을 포함한다. 특정 개체에 대한 구체적인 치료적 유효량은 달성하고자 하는 반응의 종류와 정도, 경우에 따라 다른 제제가 사용되는지의 여부를 비롯한 구체적 조성물, 개체의 연령, 체중, 일반건강 상태, 성별 및 식이, 투여 시간, 투여 경로 및 조성물의 분비율, 치료기간, 구체적 조성물과 함께 사용되거나 동시 사용되는 약물을 비롯한 다양한 인자와 의약 분야에 잘 알려진 유사 인자에 따라 다르게 적용하는 것이 바람직하다. 일일 투여량은 본 발명의 약학조성물의 양을 기준으로 0.0001 내지 100 ㎎/㎏이고, 바람직하게는 0.01 내지 100 ㎎/㎏이며, 하루 1 ~ 6 회 투여될 수 있다. 다만, 각 유효성분의 복용량 또는 투여량이 각 유효성분의 함량을 지나치게 높게 포함하여 부작용을 초래하지 않을 정도이어야 함은 당업자에게 자명하다. 따라서 본 발명의 목적에 적합한 조성물의 유효량은 전술한 사항을 고려하여 결정하는 것이 바람직하다.
상기 개체는 임의의 포유동물에 적용가능하며, 상기 포유동물은 인간 및 영장류뿐만 아니라, 소, 돼지, 양, 말, 개 및 고양이 등의 가축을 포함한다.
본 발명의 약학적 조성물은, 쥐, 생쥐, 가축, 인간 등의 포유동물에 다양한 경로로 투여될 수 있다. 투여의 모든 방식은 예상될 수 있는데, 예를 들면, 경구, 직장 또는 정맥, 근육, 피하, 자궁내 경막 또는 뇌혈관내(intracerebroventricular) 주사에 의해 투여될 수 있다.
하기의 실시예를 통하여 본 발명을 보다 상세하게 설명한다. 그러나 하기 실시예는 본 발명의 내용을 구체화하기 위한 것일 뿐 이에 의해 본 발명이 한정되는 것은 아니다.
실시예 1. 정상 혈액에서의 TH17 세포 발현과 GDF15 발현량 연관성 확인
정상인으로부터 수득한 혈액에서 TH17 세포 발현량을 IL-17 양성 세포의 수로, 미토콘드리아 기능을 미토콘드리아 막전위 변화를 확인하는 JC-1으로 확인하고, 이들과 GDF15 발현량의 연관성을 확인하였다. 구체적으로, 정상인의 혈액에서 말초단핵구 세포 (PBMC)를 분리하여 FACS 분석을 수행하였다. PBMC 분리 후 Th17 세포 분석을 위해서 PMA, Ionomycin 및 Golgi stop 처리후 4시간 동안 반응하였다. 그 후 항-인간 CD4 PE-Cy7 처리 후 30분 동안 반응하고, Cytofix/Cytoperm을 투과화(eabilization)시켰다. 이 후 항-인간 IL-17 FITC로 염색하였다. 또한, JC-1 염색을 위해서는 키트 (JC-1 dye, Thermo Fisher)를 이용하여 수행하였다. 염색된 세포를 유세포 분석기(Flow cytometry)로 분석하여, CD4+ IL-17+ 세포, JC-1+ 세포를 세포의 수를 Flow jo program을 이용하여 분석하였다. 또한, 정상인의 혈청에서 GDF15 확인을 위해 ELISA를 실시하였다. 1차 Capture ab (항-마우스 GDF15)를 이용하여 상온에서 2시간 동안 96-웰 플레이트에 코팅한 후, 블로킹 버퍼(blocking buffer)를 이용하여 비특이적인 반응을 차단시켰다. 블로킹 후에 스탠다드(Standard)와 샘플을 플레이트에 넣고 두 시간 반응 후에 워시 버퍼(wash buffer)를 이용하여 세척하였다. 스탠다드 (재조합:항-인간 GDF15)와 혈청 원액을 넣어주고 2시간 반응 후 워시 버퍼를 이용하여 세척하였다. 세척 후 검출 ab (항-인간 GDF15)를 넣고 2시간 동안 반응시킨 후 세척하고 발색을 진행하였다.
그 결과, 정상인의 혈액에서 TH17 세포의 발현이 증가하면 미토콘드리아의 기능이 손상되고 GDF15 발현이 감소하였으며, TH17 세포의 발현이 감소하면 미토콘드리아의 기능이 회복되고 GDF15 발현이 증가하는 것으로 나타나 (도 1), TH17과 GDF15 발현이 반비례의 상관관계를 가지는 것을 알 수 있다.
실시예 2. 간이식 환자 혈액에서의 STAT3 억제제에 의한 GDF15 발현량과 TH17 세포 발현 및 Treg 연관성 확인
간이식 환자에서 STAT3 억제제 (메트포민) 처리에 의한 TH17 세포 발현, Treg 유도 (Foxp3 양성) 및 GDF15 발현량의 상관성을 확인하기 위해, 간이식 환자에서 수득한 혈액에서 TH17 세포 발현량을 CD4+IL-17+ 양성 세포의 수로, Treg 세포의 발현량을 CD4+CD25highFoxp3+ 양성 세포의 수로 확인하고, 이들과 GDF15 발현량의 연관성을 확인하였다. 구체적으로, 환자의 혈액에서 말초단핵구 세포를 분리하여 FACS 분석을 수행하였다. PBMC 분리 후 Th17 세포 분석을 위해서 PMA, Ionomycin 및 Golgi stop 처리후 4시간 동안 반응시켰다. 그 후 항-인간 CD4 PE-Cy7 처리후 30분 반응시키고, Cytofix/Cytoperm을 투과화시켰다. 이후 항-인간 IL-17 FITC를 염색하였다. Treg 세포 분석을 위해서 항-인간 CD4 PE-Cy7 및 항-인간 CD25 APC를 처리한 후 30분 동안 반응시키고, Cytofix/Cytoperm을 투과화시켰다. 이 후 항-인간 Foxp3 PE로 염색하였다.
그 결과, STAT3 억제제 복용 전에 비해 복용 후에 TH17 세포가 감소하였으며, Treg 유도가 증가하였고, GDF15의 발현이 증가하는 것으로 나타났다 (도 2). 이를 통해, GDF15의 발현이 STAT3 억제제에 의해 증가하고, TH17의 분화가 억제되며 Treg 유도가 증가하는 것을 알 수 있다.
실시예 3. 정상 혈액에서의 GDF15에 의한 IL-10 분비 및 Treg 유도 영향 확인
정상인으로부터 수득한 혈액에서 GDF15에 의한 IL-10의 분비량과의 연관성을 확인하였다. 구체적으로, 상기 인간의 말초혈액단핵구 세포에 anti-CD3 자극 단독 조건과 동시에 GDF 1ng/㎖를 처리한 후 3일간 배양 후에 sup(supernatant)을 수득하였다. 수득된 sup에 IL-10에 대해 ELISA를 진행하였다.
그 결과, GDF15에 의해 IL-10 분비가 증가하는 것으로 나타났다 (도 3).
실시예 4. 간이식 환자 혈액에서의 GDF15에 의한 IL-10 분비 및 Treg 유도 영향

간이식 환자로부터 수득한 혈액에서 T 세포를 활성화 시키기 위해 Anti CD3 0.5ug/ml (plate coating 2h) 항체와 GDF15 0.25, 10 또는 20 ng/ml을 처리한 뒤, 3일간 배양 후 세포에서 Treg의 발현을 조사하였다. Treg 세포 분석을 위해서 항-인간 CD4 PE-Cy7 및 항-인간 CD25 APC를 처리한 후 30분 동안 반응시키고, Cytofix/Cytoperm을 투과화시켰다. 이 후 항-인간 Foxp3 PE를 염색하였다. Flow jo program을 이용하여 상기 항체에 대해 양성인 세포들을 분석하였다.
그 결과, GDF15에 의해 간이식 환자의 혈액 세포에서 Treg 세포 유도가 증가한 것으로 나타났다 (도 4).
실시예 5. 간세포에서의 GDF15에 의한 섬유화 영향 확인
GDF15 처리에 의해 간세포에서 섬유화가 감소되는지 확인하기 위해, 간세포 (Hepg2)에 TGF-β와 GDF15를 처리하고 GDF15에 의한 TGF-β의 단백질 수준 및 α-SMA(α-smooth muscle actin)의 단백질 수준 (섬유화)을 웨스턴 블롯 분석을 통해 확인하였다.
그 결과, GDF15 처리에 의해 TGF-β 및 α-SMA의 mRNA 수준이 감소하는 것으로 나타나, GDF15가 섬유증을 억제하는 효과가 있음을 알 수 있다 (도 5).
실시예 6. 이식질환 환자 면역세포에서의 GDF15에 의한 IL-10 분비 및 Treg 유도 영향
확인
간 이식질환 환자로부터 수득한 면역세포에서 T 세포를 활성화시키기 위해 Anti CD3 0.5ug/ml (plate coating 2h) 항체와 GDF15 20 또는 100 ng/ml을 처리한 뒤, 3일간 배양 후 세포에서 Treg의 발현을 조사하였다. Treg 세포 분석을 위해서 항-인간 CD4 PE-Cy7 및 항-인간 CD25 APC를 처리한 후 30분 동안 반응시키고, Cytofix/Cytoperm을 투과화시켰다. 이 후 항-인간 Foxp3 PE를 염색하였다. Flow jo program을 이용하여 상기 항체에 대해 양성인 세포들을 분석하였다.
그 결과, GDF15에 의해 이식질환 환자의 면역세포에서 Treg 세포와 항염증 인자인 IL-10 유도가 증가한 것으로 나타났다 (도 6).
실시예 7. 쇼그렌 전임상 동물모델에서 GDF15에 의한 질환 치료 효과 확인
쇼그렌 전임상 동물모델(NOD/ShiLtJ)에서 GDF15 vector 주입 후 체중과 disease score를 확인하였다. 쇼그렌 전임상 모델 NOD/ShiLtJ에 GDF15 벡터를 처리한 후, 체중과 disease score를 확인하였다.
GDF15 벡터는 도 13과 같은 구성을 가지며, 하기와 같이 제조하였다. 우선, mGDF15 in pUC57-Amp 벡터는 서열번호 2에 표시되는 optimized Mus musculus GDF15 CDS에 5'말단 BglⅡ서열을 삽입하고 3'말단 stop codon을 제거하고 SalⅠ서열을 삽입하여 합성된 유전자를 확보하였다. mGDF15 in pFLAG-CMV5a 는 mGDF15 in pUC57-Amp을 BglⅡ와 SalⅠ으로 잘라서 Gel extraction을 통해 mGDF15 단편만 추출하였다. DNA 단편을 pFLAG-CMV5a 과발현 벡터에 ligation 하여 콜로니를 얻었다. 여러 콜로니를 랜덤선택하여 각각의 콜로니에서 plasmid DNA를 추출하고, BglⅡ와 SalⅠ을 처리하여 원하는 insert DNA (mGDF15)가 들어간 콜로니를 구분하여 얻었다. 선별된 클론을 시퀀싱하고 DNA 서열을 확인하였다. 선택된 클론을 HEK293세포에 형질도입한 뒤 3일 후 세포에서 단백질을 추출하고 이 단백질에서 Western Blotting을 진행하여 단백질 과발현이 확인된 clone을 선택하였다. 제조된 GDF15 벡터는 100 ug/100ul 씩 일주일에 한번 간격으로 muscle electroporation 방식으로 쇼그렌 전임상 동물모델에 주입하였다.
그 결과, GDF15 투여시 동물 모델의 체중이 증가하고, disease score가 감소함을 확인할 수 있었다(도 7).
실시예 8. 이식질환 환자의 혈청에서 IL-17과 GDF15의 상관관계 확인
IL-17의 발현과 GDF15 발현량의 상관성을 확인하기 위해, 간이식 환자에서 수득한 혈청이나 혈장에서 GDF15와 IL-17의 확인을 위해 ELISA를 실시하였다. 각각의 1차 Capture ab (항-휴먼 GDF15, IL-17)를 이용하여 상온에서 2시간 동안 각 96-웰 플레이트에 코팅한 후, 블로킹 버퍼(blocking buffer)를 이용하여 비특이적인 반응을 차단시켰다. 블로킹 후에 스탠다드(Standard)와 샘플을 플레이트에 넣고 두 시간 반응 후에 워시 버퍼(wash buffer)를 이용하여 세척하였다. 각각의 스탠다드 (재조합:GDF15, IL-17)와 혈청 원액을 넣어주고 2시간 반응 후 워시 버퍼를 이용하여 세척하였다. 세척 후 검출 각 ab (항-인간 GDF15, IL-17)를 넣고 2시간 동안 반응시킨 후 세척하고 발색을 진행하였다.
그 결과, 이식환자 혈청에서 IL17이 증가시, GDF15 발현이 저하되는 네가티브 코릴레이션을 확인하였다(도 8 참조).
실시예 9. 간이식 환자 면역세포에서 SD282 화합물 처리 후 GDF15 발현량 확인
메트포민 유도체인 SD282 화합물(대한민국 등록특허 10-1705446호에 공지됨)을 간이식 환자로부터 수득한 면역세포에 처리한 후 GDF15의 발현량을 확인하였다.
그 결과, 부형제 처리군에 비해 SD282 화합물 처리군(STAIM1)에서 GDF15 발현량이 현저히 증가함을 확인하였다(도 9 참조).
실시예 10. 이식거부질환 동물 모델에서 SD282 화합물 처리 후 GDF15 발현량 확인
이식거부질환(GVHD) 동물 모델로 유도하기 위해서 c57BL6 MICE의 비장세포와 골수세포를 각각 분리하여 각각의 세포를 5x106을 690cGY 로 조사Balb/c 마우스에 꼬리 정맥에 세포를 주입하였다. GVHD 유도한 당일에 SD282 화합물(STAIM1), FK506 또는 SD282 화합물과 FK506의 조합(STAIM2)을 경구 투여하였다, 약물의 투여는 실험종료 시점까지 투여하였다. 실험종료 시점에 얻은 소장 조직에서 IHC로 염색한 소장 세포에서 GDF15의 발현량을 확인하였다.
그 결과, SD282 화합물과 FK506의 병용 처리군(STAIM2)에서 GDF15 발현량이 현저히 증가함을 확인하였으며, 이는 이식거부질환의 치료 효과가 우수할수록 GDF15의 발현량도 증가함을 보이는 것이다(도 10 참조).
실시예 11. 간이식 환자 아바타 동물 모델에서 SD282 화합물 처리 후 치료 효과 및 GDF15 발현량 확인
간이식 동물 모델에 FK506 또는 SD282 화합물과 FK506의 조합(STAIM2)을 투여하였다. 그런 다음, 동물 모델의 간조직을 IHC 염색하여 GDF15 발현량과 IL-10 발현량을 확인하였다.
그 결과, SD282 화합물 처리군(STAIM1) 및 SD282 화합물과 FK506의 병용 처리군(STAIM2)에서 GDF15 발현량과 IL-10의 발현량이 현저히 증가함을 확인하였으며, 이는 GDF15 발현과 IL-10의 발현이 서로 밀접한 관련이 있음을 입증하는 것이다(도 11 참조).
실시예 12. 간이식 동물 모델에서 SD282 화합물 처리 후 IL-17 및 GDF15 발현량 확인
FK506 또는 SD282 화합물과 FK506의 조합(STAIM2)을 투여하였다. 그런 다음, 동물 모델의 간조직을 IHC 염색하여 GDF15 발현량과 IL-10 발현량을 확인하였다.
그 결과, SD282 화합물과 FK506의 병용 처리군(STAIM2)에서 GDF15 발현량이 증가함을 확인하였으며, IL-10의 발현량이 현저히 증가한 반면, IL-17의 발현량이 감소함을 확인하였다. 이는 GDF15 발현이 증가할 경우 IL-17의 발현이 감소됨을 보이는 것이다(도 12 참조).
실시예 13. 류마티스관절염 질환에서 SD282 화합물 처리 후 GDF15 발현량 확인
류마티스관절염 질환에서 SD282 화합물(HL237)에 의한 질환 제어 효력을 확인하였으며, SD282 화합물(HL237) 투여 후 6주에 희생된 류마티스관절염 마우스 관절 조직의 파괴 정도 (H&E, saflaninO)와 관절 조직 내에서의 GDF15 단백질 발현 세포를 분석하였다. 구체적으로는, 시노비움 조직 내의 전체 세포 내 GDF15 발현 세포 비율 분석하였다.
그 결과, 도 14에서 볼 수 있는 바와 같이 SD282 화합물(HL237)에 의해 관절 손상과 염증세포의 침윤이 현저하게 감소하였으며, 면역 바이오마커 GDF15 발현 세포가 현저하게 증가되었다. 따라서 GDF15는 염증 질환 치료 바이오마커로서 활용될 수 있음을 확인할 수 있었다.
실시예 14. 인간 말초단핵구 세포에서 SD282 화합물 처리 후 GDF15 발현량 확인
말초단핵구 세포에 T 세포 활성 조건인 antiCD3 0.5ug/ml로 자극한 조건에 SD282 처리 후 72시간 배양 후 그 상청액에서 GDF15의 발현을 ELISA 분석을 통해서 확인하였다.
그 결과, 도 15에서 볼 수 있는 바와 같이 SD282 화합물에 의해 GDF15 발현 세포가 현저하게 증가함을 확인할 수 있었다.
실시예 15. 류마티스 관절염 동물모델 제작
본 발명자들은, 대사질환에 치료 효과가 있는 것으로 알려진, 마이크로바이옴 균주인 에크멘시아 뮤신필리아(Akkermansia muciniphilia)의 GDF 발현 증가 및 류마티스 관절염에 대한 효과를 확인하기 위하여, 류마티스 동물모델을 제작하였다. 구체적으로, 정상 DBA/1 마우스에 꼬리 dermal에 type II collagen 100 ug을 주입 하였으며, 2주 후 type II collagen 100ug을 주입하여 관절염을 유발하였다. 류마티스 관절염 유발 후 3주차에 류마티스 관절염 징후를 보이는 군(관절염 지수 1점 이상)을 선별하였다. 류마티스 관절염 징후는, 관절염 지수로 평가하였으며, 류마티스 관절염 지수는 하기 표 1의 방법으로 평가하였다. 정상 대조군으로는, 야생형 마우스(WT)를 이용하였다.

상기 선별된 군의 마우스를 이용하여, 본 발명의 에크멘시아 뮤신필리아의 GDF 증가 및 류마티스 관절염 개선효과를 확인하고자 하였다.
실시예 16. 관절염 지수 및 유병율 감소 확인
본 발명의 마이크로바이옴 균주인 에크멘시아 뮤신필리아가 류마티스 관절염에 효과가 있는지 확인하기 위하여, 상기 실시예 1에서 제작된 동물모델에, 에크멘시아 뮤신필리아(1x1010/mice)를 매일 경구투여(Akkermansia 군, A. Muciniphilia)하여, 상기 표 1의 기준으로 관절염 지수 및 유발률을 관찰하였다. 유발률 평가의 경우 하나의 발에서 부종이 발생될 시 25%로 네 다리의 합을 100%로 계산하여 평가하였다. 대조군으로는 상기 실시예 1의 동물모델에 생리식염수를 투여한 Vehicle 군을 이용하였다.
그 결과, 도 16에 나타낸 바와 같이, Vehicle 군과 비교하여, 에크멘시아 뮤신필리아를 투여한 군에서는, 유의적으로 관절염 지수가 감소하는 것을 확인하였으며, 유발률 또한 Vehicle 군과 비교하여 감소한 것을 확인하였다.
실시예 17. 연골 보호 및 연골 손상 억제 확인
본 발명의 마이크로바이옴인 에크멘시아 뮤신필리아가 관절 파괴를 억제하는지 확인하고자 하였다. 구체적으로 상기 실시예 2의 마우스를 인도적으로 희생하여, 관절 synovium 조직을 수득하였으며, 수득된 조직을 헤마토실린 및 에오신(Hematoxylin and eosin, H&E) 염색과, 사프라닌 O(Safranin O) 염색으로 조직의 손상 정도를 관찰하였다.
그 결과, 야생형 대조군과 비교하여, Vehicle 군에서는 염증 지수, 관절 손상 및 연골 손상이 증가한 것을 확인하였으며, 본 발명의 에크멘시아 뮤신필리아를 투여한 군에서는 Vehicle 군과 비교하여, 염증 지수, 관절 손상 및 연골 손상이 유의적으로 감소한 것을 확인하였다(도 17).
실시예 18. IL-17 발현 억제 확인
IL-17은 염증을 유발하는 인터루킨으로서, IL-17 양성 세포의 분화가 촉진되면 발현되어, 염증을 유발하게 된다. 따라서, 본 발명의 마이크로바이옴인 아커멘시아 뮤시니필 리가, 류마티스 관절염에서 IL-17의 발현을 억제하기 위하여, 상기 실시예 2의 마우스의 관절 조직을 면역조직화학염색으로 염색하여 IL-17의 발현을 확인하였다.
그 결과, Vehicle 군과 비교하여, 에크멘시아 뮤신필리아를 투여한 군에서는, IL-17이 억제된 것을 확인하였으며, IL-17 양성으로 염색된 면적(IL-17 Positive area)이 유의적으로 감소한 것을 확인하였다(도 18).
실시예 19. 조절 T 세포(Regulatory T cell, Treg) 조절 확인
본원 발명의 상기 실시예 2의 마우스로부터 적출한 비장을, 각각 유세포 분석을 시행하였다. 구체적으로, 적출된 비장에서, Treg 세포 분석을 위해서 항-인간 CD4 PE-Cy7 및 항-인간 CD25 APC를 처리한 후 30분 동안 반응시키고, Cytofix/Cytoperm을 투과화시켰다. 이 후 항-인간 Foxp3 PE로 염색하였다.
그 결과, 도 19에 나타낸 바와 같이, Vehicle 군과 비교하여, 본 발명의 에크멘시아 뮤신필리아를 투여한 군에서는, Treg가 증가한 것을 확인하였다.
실시예 20. GDF15 발현 증가 확인
본 발명의 에크멘시아 뮤신필리아가 GDF의 발현을 증가시키는지 확인하기 위하여, 상기 실시예 2의 마우스로부터 적출한 비장을, 면역화학조직 염색을 이용하여, GDF의 발현을 확인하고자 하였다.
그 결과, 도 20에 나타낸 바와 같이, Vehicle 군과 비교하여, 본원 발명의 에크멘시아 뮤신필리아를 투여한 군의 비장 절편에서는 GDF15의 발현이 유의적으로 증가하여, GDF15 양성 면적(GDF15 Positive area)가 증가한 것을 확인하였다.
실시예 21. 루프스 동물모델 및 SD217 처리
본원 발명의 GDF15 발현을 유도하는 SD217이 루푸스에 대한 효과를 확인하고자, 루푸스 동물모델을 이용하였다. 구체적으로, 10주령의 루푸스 자연발생 마우스인 MRL/lpr를 이용하였으며, MRL/lpr 마우스에 SD217을 50 mg/kg의 농도로 매일 경구 투여 하였다. 약물 투여 시작 후 2주 간격으로 체중을 측정하였다. 약물투여 16주차에 마우스를 인도적으로 희생하였다. 대조군으로는, 생리식염수를 투여한 Vehicle군을 이용하였다.
그 결과, 도 21에 나타낸 바와 같이, 루푸스 자연발생 마우스인 MRL/lpr 마우스는 루푸스 병증이 증가될수록, 체중 감소가 일어나는 반면, GDF15 발현을 유도하는 SD217을 투여한 군에서는 체중이 유지 및 증가하는 것을 확인하여, 본 발명의 SD217이 루푸스의 병증 진행을 억제하는 것을 확인하였다.
실시예 22. SD217 처리에 따른 면역세포 조절 확인
본원 발명의 신규한 화합물인 GDF15 발현을 유도하는 SD217이, 루푸스 동물모델에서, 면역 세포를 조절하는지 확인하고자 하였다. 구체적으로 상기 실시예 1의 희생된 마우스의 전혈을 채취하고, 혈액세포에서 CD4-(percp cy5.5), CD8-(PE) T 세포(double negative T cell, DNT)의 발현을 유세포분석으로 분석하였다.
그 결과, 도 22에 나타낸 바와 같이, GDF15 발현을 유도하는 SD217을 투여한 군에서는 Vehicle군과 비교하여, DNT의 발현이 유의적으로 감소한 것을 확인하였다.
Claims (21)
- GDF15 단백질 또는 이를 코딩하는 핵산서열을 유효성분으로 포함하는 면역질환의 예방 또는 치료용 약학 조성물.
- 제 1 항에 있어서,상기 면역질환은 IL-10 감소되고 및 IL-17의 발현이 증가되어, Treg의 수가 감소되었고, Th17 세포의 수가 증가된 면역질환인 것인 면역질환의 예방 또는 치료용 약학 조성물.
- 제 1 항에 있어서,상기 면역질환은 이식 거부질환, 이식질환 및 섬유증으로 이루어진 군으로부터 선택된 것인 면역질환의 예방 또는 치료용 약학 조성물.
- 제 1 항에 있어서,상기 면역질환은 간섬유증, 간이식 후 섬유증, 간이식 질환, 이식거부질환, 쇼그렌 증후군 및 이식편대숙주질환(GVHD)으로 이루어진 군으로부터 선택된 것인 면역질환의 예방 또는 치료용 약학 조성물.
- 제 1 항에 있어서,상기 GDF15 단백질은 서열번호 5 내지 8 중 어느 하나로 표시되는 서열을 포함하는 것인 면역질환의 예방 또는 치료용 약학 조성물.
- 제 1 항에 있어서,상기 GDF15 단백질을 코딩하는 핵산서열은 서열번호 1 내지 4 중 어느 하나로 표시되는 염기서열을 포함하는 것인 면역질환의 예방 또는 치료용 약학 조성물.
- 제 1 항에 있어서,상기 GDF15 단백질 또는 이를 코딩하는 핵산서열은 미분화 T 세포의 Th17 세포로의 분화를 감소 또는 억제하는 것인 면역질환의 예방 또는 치료용 약학 조성물.
- 제 1 항에 있어서,상기 GDF15 단백질 또는 이를 코딩하는 핵산서열은 IL-10의 분비를 증가시키고 Treg 세포를 유도하는 것인 면역질환의 예방 또는 치료용 약학 조성물.
- 제 1 항에 있어서,상기 GDF15 단백질 또는 이를 코딩하는 핵산서열은 TGF-β 및 α-SMA의 mRNA 발현수준을 감소시켜 섬유화를 억제하는 것인 면역질환의 예방 또는 치료용 약학 조성물.
- GDF15의 발현량을 확인하는 단계를 포함하는, 미트콘드리아 기능손상을 진단하는 방법.
- GDF15의 발현량을 확인하는 단계를 포함하는, 혈액내 Th17 세포의 양을 예측하는 방법.
- GDF15의 발현량을 확인하는 단계를 포함하는, 간이식 후 간이식 예후를 모니터링하는 방법.

- 제 13 항에 있어서,상기 방법은 치료제 또는 치료후보 물질의 투여 후 GDF15의 발현량이 증가할 경우 예후가 좋은 것으로 판단하는 것인 1)류마티스 관절염 또는 2)이식 후, 이식관련 질환 치료의 예후를 모니터링하는 방법.
- 제 14 항에 있어서,상기 치료제 또는 치료후보 물질은 에크멘시아 뮤신필리아(Akkermansia muciniphilia) 균주 또는 이의 배양액인, 방법.
- 분리된 시료에 후보물질을 처리하는 단계;상기 시료에서 GDF15의 발현량을 확인하는 단계를 포함하는, IL-17의 발현을 감소시켜 Th17 세포의 양을 감소시키며, IL-10의 발현을 증가시켜 Treg 세포의 양을 증가시키는 화합물을 스크리닝하는 방법.
- GDF15의 발현양을 측정하는 제제를 포함하는 1)류마티스 관절염 또는 2)이식 후, 이식관련 질환의 예후를 모니터링 하기 위한 조성물.
- 제 19항에 따른 조성물을 포함하는 1)류마티스 관절염 또는 2)이식 후, 이식관련 질환의 예후를 모니터링 하기 위한 키트.
- GDF15 단백질 또는 이를 코딩하는 핵산서열의 약학적으로 유효한 양을 개체에 투여하는 단계;를 포함하는 면역질환의 치료 방법.
