さらに錠剤は、必要に応じ、通常の剤皮を施した錠剤、たとえば、糖衣錠、ゼラチン被包錠、胃溶性被覆錠、腸溶性被覆錠および水溶性フィルムコーティング錠とすることができる。
カプセル剤は、上記で例示した各種の医薬品と混合し、硬質ゼラチンカプセルおよび軟質カプセルなどに充填して調製される。
また、溶剤、増量剤、等張化剤、溶解補助剤、乳化剤、懸濁化剤、増粘剤などの上記した各種の液体製剤化用添加物を用い、常法に従い調製して、水性または油性の懸濁液、溶液、シロップおよびエリキシル剤とすることもできる。
上記製剤の有効成分の投与量は、用法、患者の年齢、性別、疾患の形態、その他の条件などに応じて適宜選択されるが、通常成人に対して1日0.1~1000mgを1回から数回に分割して投与すればよく、好ましくは、1日40~500mgを1回から数回に分割して投与すればよい。
被験物質として、1-(3-(2-(1-ベンゾチオフェン-5-イル)エトキシ)プロピル)アゼチジン-3-オールのマレイン酸塩(以下、化合物Aとする)を用いた。
21種類の受容体に対する化合物Aの結合能を試験した。各受容体に対する試験は、それぞれα1 (non-selective): Greengrass and Bremner (1979)、α2 (non-selective): Uhlen and Wikberg (1991)、β1 (h): Smith and Teitler (1999)、β2 : Abrahamsson et al. (1988)、AT1 (h): Bergsma et al. (1992)、AT2 (h): Tsuzuki et al. (1994)、BZD (central): Speth et al. (1979)、D1: Trampus et al. (1991)、D2: Terai et al. (1989)、GABAA: Snodgrass (1978)、GABAB :Bowery et al. (1983)、NMDA: Sills et al. (1991)、H1 (h): Smit et al. (1996)、H2: Ruat et al. (1990)、M1: Watson et al. (1982)、M2 (h): Dorje et al. (1991)、N (neuronal) (α-BGTX-insensitive): Pabreza et al. (1991)、5-HT (non-selective) : Peroutka and Snyder (1979)、σ (non-selective): Shirayama et al. (1993)、TH : Inoue et al. (1983)、TRH: Sharif and Burt (1983)に準拠した以下の方法により行った。
試験に用いた各受容体および参照化合物を表1に示す。
各受容体に対する参照化合物および放射性リガンド(カッコ内は、最終濃度を示す。)、ならびにインキュベーション条件を表2に示す。
式:100-([measured specific binding]/[control specific binding]×100)
式中、measured specific bindingは、化合物Aと参照化合物存在下における放射性リガンドとの結合放射能量;control specific bindingは、参照化合物存在下における放射性リガンドとの結合放射能量を示す。
化合物Aは、シグマ受容体に選択的に高い親和性を有することが明らかとなった。
化合物Aのシグマ1受容体に対する阻害定数(Ki値)を同定した。試験は、Ganapathy, M. E. (1999)に準拠した以下の方法により行った。
Jurkat細胞に発現しているシグマ1受容体において、15nMの[3H](+)-pentazocineに対する化合物Aの各濃度(0.1、0.3、1、3、10、30、100、300、1000nM)の阻害率を測定した。反応条件は、120分間、37℃で実施した。阻害率は、試験例1に記載の方法と同様に算出した。Ki値は、非線形回帰分析を基にIC50を算出し、チェン=プルソフ式に従って同定した。
その結果、化合物AのKi値は、16nMであった。
化合物Aのシグマ2受容体に対する阻害定数(Ki値)を同定した。試験は、Ganapathy, M. E. (1999)に準拠した以下の方法により行った。
Jurkat細胞に発現しているシグマ2受容体において、1μM (+)-pentazocine存在下での25nMの[3H]DTGに対する化合物Aの各濃度(0.1、0.3、1、3、10、30、100、300、1000nM)の阻害率を測定した。反応条件は、60分間、室温で実施した。阻害率は、試験例1に記載の方法と同様に算出した。Ki値は、非線形回帰分析を基にIC50を算出し、チェン=プルソフ式に従って同定した。
その結果、化合物AのKi値は、160nMであった。
本発明の一般式[1]で表されるアルキルエーテル誘導体またはその塩を使用する方法は、シグマ受容体を活性化または阻害する方法として有用である。

- R1およびR2が、同一または異なって、水素原子、ハロゲン原子またはC1-6アルコキシ基である、請求項2に記載のシグマ1受容体結合剤。
- mが2、かつnが2または3である、請求項2または3に記載のシグマ1受容体結合剤。
- R3が、保護されていてもよいヒドロキシル基である、請求項2~4のいずれか一項に記載のシグマ1受容体結合剤。
- アルキルエーテル誘導体が、1-(3-(2-(1-ベンゾチオフェン-5-イル)エトキシ)プロピル)アゼチジン-3-オールである、請求項2に記載のシグマ1受容体結合剤。
- 1-(3-(2-(1-ベンゾチオフェン-5-イル)エトキシ)プロピル)アゼチジン-3-オールまたはその塩を含有することを特徴とする、シグマ2受容体結合剤。
