式(I)の化合物及びその塩は、その基本構造あるいは置換基の種類に基づく特徴を利用し、種々の公知の合成法を適用して製造することができる。その際、官能基の種類によっては、当該官能基を原料から中間体へ至る段階で適当な保護基(容易に当該官能基に転化可能な基)に置き換えておくことが製造技術上効果的な場合がある。このような保護基としては、例えば、ウッツ(P. G. M. Wuts)及びグリーン(T. W. Greene)著、「Greene’s Protective Groups in Organic Synthesis(第4版、2006年)」に記載の保護基等を挙げることができ、これらの反応条件に応じて適宜選択して用いればよい。このような方法では、当該保護基を導入して反応を行ったあと、必要に応じて保護基を除去することにより、所望の化合物を得ることができる。
前記各製法で得た式(I)の化合物を原料として、更に、エステル化、アミド化等の当業者が通常用いる化学修飾反応を行うことによって、別の式(I)の化合物を得ることができる。

この反応では、化合物(3)と化合物(5)とを等量若しくは一方を過剰量用い、これらの混合物を、塩基の存在下、反応に不活性な溶媒中、冷却下から室温下、好ましくは冷却下において、通常0.1時間~5日間攪拌する。ここで用いられる溶媒の例としては、特に限定はされないが、ベンゼン、トルエン、キシレン等の芳香族炭化水素類、ジエチルエーテル、テトラヒドロフラン、1,4-ジオキサン、ジメトキシエタン等のエーテル類、ヘキサン及びこれらの混合物が挙げられる。塩基の例には、リチウムジイソプロピルアミド、トリエチルアミン、ジイソプロピルエチルアミン、リチウムヘキサメチルジシラジド、カリウムヘキサメチルジシラジド、1,8-ジアザビシクロ[5.4.0]-7-ウンデセン、n-ブチルリチウム、カリウムtert-ブトキシド等の有機塩基、炭酸ナトリウム、炭酸カリウム、炭酸セシウム、水素化ナトリウム等の無機塩基が含まれる。

その他、当業者に公知の方法を用いて所望の原料化合物を得ることができる。例えば、以下の反応スキームに記載の方法を用いることができる。
ラット精巣上体の脂肪組織をホモジナイズし、100000 x gで30分間超遠心することでIRAPを含むミクロゾームを得た。取得したミクロゾーム(タンパク総量として0.55 μg/well)を溶媒(ジメチルスルホキシド(DMSO)、終濃度0.1%)又は被験化合物(公比3、最大濃度10 μM)と混和した後、最終濃度が25 μMになるようにAVPを添加し、37℃で1時間反応させた。トリフルオロ酢酸(TFA)水溶液(終濃度1%)を添加して酵素反応を停止させた後、残存するAVP量を質量分析法(MALDI-MS)によって定量した。得られた結果よりSigmoid-Emaxモデル非線形回帰分析で、溶媒を添加したコントロール群のAVP量の減少を50 %阻害する被験化合物濃度IC50値(nM)をIRAP活性阻害作用として算出した。
結果を表1に示す。実施例化合物は、P-LAPのラットホモログであるIRAPによるAVP分解を良好に阻害することが確認された。
リポフェクション法によりhP-LAP(J Biol Chem 1996; 271: 56-61)を一過性に強制発現させたHEK293細胞をホモジナイズし、100000 x gで30分間超遠心することでhP-LAPを含むミクロゾームを取得した。取得したミクロゾーム(タンパク総量として0.5~1.5 μg/well)を溶媒(DMSO、終濃度0.1%)又は被験化合物(公比3、最大濃度10 μM)と混和した後、最終濃度が25 μMになるようにAVPを添加し、37℃で1時間反応させた。TFA水溶液(終濃度1%)を添加して酵素反応を停止させた後、残存するAVP量を質量分析法(MALDI-MS)によって定量した。得られた結果よりSigmoid-Emaxモデル非線形回帰分析で、溶媒を添加したコントロール群のAVP量の減少を50%阻害する被験化合物濃度IC50値(nM)をhP-LAP活性阻害作用として算出した。結果を表1に示す。実施例化合物はhP-LAPによるAVP分解を良好に阻害することが確認された。
なお、後記表1及び表2において、Exは被験化合物の実施例番号を示す。
被験化合物は溶媒(10% N,N-ジメチルホルムアミド、10 %プロピレングリコール及び80 %蒸留水)に溶解し、ラットに経口投与した。なお、被験化合物がフリー体の場合は1モル等量の塩酸を溶媒に添加して溶解した。溶媒投与群には溶媒のみを投与した。その1時間後に水負荷として30 ml/kgの蒸留水をラットに強制投与した。その後1時間の尿量を測定し(但し、0.3 ml未満の尿量は0 mlと見做した)、水負荷量に対する排尿量の割合(尿排泄率)を算出した。下式により、化合物投与群の溶媒投与群に対する排尿阻害率(%)を算出した(各群4~5例)。

式(I)の化合物である、いくつかの実施例化合物について、3 mg/kg投与時の阻害率(%)を表2に示す。これらの実施例化合物は良好な抗利尿作用を有することが確認された。

また、本明細書に記載される粉末X線回折結果は、RINT-TTRIIを用い、管球:Cu、管電流:300 mA、管電圧:50 kV、サンプリング幅:0.020°、走査速度:4°/分、波長:1.54056Å、測定回折角範囲(2θ):2.5~40°の条件で測定したものである。なお、粉末X線回折パターンは、結晶の同一性認定においては、データの性質上、結晶格子間隔や全体的なパターンが重要であり、回折角及び回折強度は結晶成長の方向、粒子の大きさ、測定条件によって多少の誤差が生じうるものであるから、厳密に解されるべきではない。本明細書中、粉末X線回折パターンにおける回折角(2θ(°))は、当該測定法において通常許容される誤差範囲を考慮して解釈され、例えば、±0.2°の範囲が包含されると解釈される。
(3R,4R)-3-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-4-[(エチルスルファニル)メチル]-3-(メトキシメトキシ)-1-(メトキシメチル)アゼチジン-2-オン(65mg)、DOX(0.75ml)、6M塩酸(1.5ml)の混合物を60℃で1.5時間攪拌した。得られた反応混合物を氷冷した後、6M水酸化ナトリウム水溶液(1ml)とDOXを加え、減圧下で濃縮した。得られた残渣をODSカラムクロマトグラフィー(MeCN/0.1%蟻酸水溶液)にて精製し、(2R,3R)-3-アミノ-2-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-4-(エチルスルファニル)-2-ヒドロキシブタン酸(39mg)を固体として得た。
[(1R)-1-[(4R)-4-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル]-2-(メチルスルファニル)エチル]カルバミン酸 tert-ブチル(6g)とCH2Cl2(50ml)の混合物に、氷冷下でTFA(25ml)を加え、室温で1時間攪拌した。得られた反応混合物を、MeOH(85ml)と6M水酸化ナトリウム水溶液(77ml)の混合液に氷冷下で少しずつ加え、その後60℃で1.5時間攪拌した。得られた反応混合物に活性炭を加え、室温で30分間攪拌した後、不溶物をろ別した。得られたろ液を氷冷した後、6M塩酸を少しずつ加え、pHを約7に調整した後、同温で1時間攪拌した。生じた不溶物をろ取し、減圧下で乾燥した。得られた固体にEtOH:水(3:1)の混合液(45ml)を加え、80℃に加温し、不溶物が溶解するまで攪拌したのち、水(30ml)を加えた。得られた溶液を室温まで徐々に放冷しながら、終夜攪拌した。析出した固体をろ取して(2R,3R)-3-アミノ-2-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2-ヒドロキシ-4-(メチルスルファニル)ブタン酸(2.43g)を結晶として得た。得られた結晶は、粉末X線回折で2θ(°)6.5、8.6、12.3、14.1、14.7、17.4、17.9、18.5、19.1、19.6、20.7、22.7及び24.8にピークを有するものであった。
(3R,4R)-3-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-4-[(イソプロピルスルファニル)メチル]-3-(メトキシメトキシ)-1-(メトキシメチル)アゼチジン-2-オン(71mg)、MeOH(2ml)及びTHF(2ml)の混合物に6M水酸化ナトリウム水溶液(2ml)を加え、70℃で5時間攪拌した。得られた反応混合物に氷冷下、6M塩酸(2ml)を加え、減圧下で濃縮した。得られた残渣にMeCN(1ml)と1M塩酸(3ml)を加え、室温で3時間攪拌した。得られた反応混合物に6M塩酸(0.5ml)を追加し、室温で18時間攪拌した後、40℃で2時間攪拌した。反応混合物を室温まで放冷し、水を加えた。得られた混合物を減圧下で濃縮し、残渣をODSカラムクロマトグラフィー(MeCN/0.1%蟻酸水溶液)にて精製し、(2R,3R)-3-アミノ-2-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2-ヒドロキシ-4-(イソプロピルスルファニル)ブタン酸(21mg)を固体として得た。
[(1R)-1-[(4R)-4-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル]-2-(メチルスルファニル)エチル]カルバミン酸 tert-ブチル(1.1g)とトルエン(15ml)の混合物に、室温でTFA(5ml)を加え、同温で終夜攪拌した。得られた反応混合物を減圧下で濃縮し、残渣に飽和炭酸水素ナトリウム水溶液を加え、AcOEtで抽出した。得られた有機層を無水硫酸マグネシウムで乾燥した後、溶媒を減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(CHCl3/MeOH)で精製し(5R)-5-[(1R)-1-アミノ-2-(メチルスルファニル)エチル]-5-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2,2-ジメチル-1,3-ジオキソラン-4-オン(777mg)を油状物として得た。
得られた(5R)-5-[(1R)-1-アミノ-2-(メチルスルファニル)エチル]-5-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2,2-ジメチル-1,3-ジオキソラン-4-オン(65mg)に1M塩酸(1ml)を加え、その後減圧下で溶媒を留去して(5R)-5-[(1R)-1-アミノ-2-(メチルスルファニル)エチル]-5-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2,2-ジメチル-1,3-ジオキソラン-4-オン 二塩酸塩(70mg)を泡状固体として得た。
(5R)-5-[(1R)-1-アミノ-2-(メチルスルファニル)エチル]-5-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2,2-ジメチル-1,3-ジオキソラン-4-オン(95mg)、MeOH(5ml)及び炭酸カリウム(200mg)の混合物を室温で12時間攪拌した。得られた反応混合物にAcOEtを加え、不溶物をろ別した。得られた、ろ液を減圧下で濃縮し、残渣をシリカゲルカラムクロマトグラフィー(CHCl3/MeOH)で精製した。得られた粗精製物に1M塩酸(1ml)を加え、減圧下で溶媒を留去して(2R,3R)-3-アミノ-2-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2-ヒドロキシ-4-(メチルスルファニル)ブタン酸メチル 二塩酸塩(55mg)を泡状固体として得た。
(5R)-5-[(1R)-1-アミノ-2-(メチルスルファニル)エチル]-5-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2,2-ジメチル-1,3-ジオキソラン-4-オン(98mg)、MeOH(2ml)及び28%アンモニア水(2ml)の混合物を、マイクロ波照射下、120℃で30分間攪拌した。得られた反応混合物に水を加え、AcOEtとトルエンの1:1混合液で抽出した。得られた有機層を無水硫酸マグネシウムで乾燥した後、溶媒を減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(CHCl3/MeOH)で精製した。得られた粗精製物に1M塩酸(1ml)を加え、減圧下で溶媒を留去して(2R,3R)-3-アミノ-2-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2-ヒドロキシ-4-(メチルスルファニル)ブタンアミド 二塩酸塩(20mg)を泡状固体として得た。
[(1R)-1-[(4R)-4-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル]-2-(メチルスルファニル)エチル]カルバミン酸 tert-ブチル(400mg)に濃塩酸(5ml)を加え、80℃で3日間攪拌した。得られた反応混合物をODSカラムクロマトグラフィー(MeCN/0.1%蟻酸水溶液)にて精製し、得られた2種の化合物のうち、高極性の化合物として(1)(2R,3R)-3-アミノ-2-ヒドロキシ-2-{[2-(3-ヒドロキシペンチル)フロ[3,2-c]ピリジン-4-イル]メチル}-4-(メチルスルファニル)ブタン酸(65mg)を泡状固体として得た。また、もう一方の低極性の化合物として得られた粗精製物に1M塩酸(2ml)を加え、その後減圧下で溶媒を留去して(2)(2R,3R)-3-アミノ-2-{[2-(3-クロロペンチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2-ヒドロキシ-4-(メチルスルファニル)ブタン酸 二塩酸塩(107mg)を泡状固体として得た。
[(1R)-1-{(4R)-4-[(2-ブチルチエノ[3,2-c]ピリジン-4-イル)メチル]-2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル}-2-(メチルスルファニル)エチル]カルバミン酸 tert-ブチル(83mg)、MeOH(0.83ml)及びDOX(0.83ml)の混合物に1M水酸化ナトリウム水溶液(0.83ml)を加え、50℃で5時間攪拌した。反応混合物を室温まで放冷し、減圧下で濃縮した。得られた残渣にDOX(0.83ml)を加え、氷冷下で塩化水素(4M DOX溶液、0.83ml)を加えた。得られた混合物を室温で1.5時間攪拌した後、減圧下で濃縮した。得られた残渣をODSカラムクロマトグラフィー(MeCN/0.1%蟻酸水溶液)にて精製し固体を得た。得られた固体をMeCN-MeOH(10:1)に懸濁し、不溶物をろ取した。得られた固体をMeCNで洗浄し、(2R,3R)-3-アミノ-2-[(2-ブチルチエノ[3,2-c]ピリジン-4-イル)メチル]-2-ヒドロキシ-4-(メチルスルファニル)ブタン酸(27mg)を固体として得た。
[(1R)-1-[(4R)-2,2-ジメチル-4-{[2-(3-メチルブチル)フロ[3,2-c]ピリジン-4-イル]メチル}-5-オキソ-1,3-ジオキソラン-4-イル]-2-(メチルスルファニル)エチル]カルバミン酸 tert-ブチル(140mg)とCH2Cl2(1.4ml)の混合物にTFA(0.635ml)を加え、室温で2時間攪拌した。得られた反応混合物を、MeOH(2.1ml)と6M水酸化ナトリウム水溶液(1.85ml)の混合液に氷冷下で少しずつ加え、その後60℃で3時間攪拌した。得られた反応混合物に6M塩酸(0.46ml)を氷冷下で加えた後、室温で10分間攪拌した。得られた溶液をODSカラムクロマトグラフィー(MeCN/0.1%蟻酸水溶液)にて精製した。得られた粗精製物に、過剰量の1M塩酸を加え、その後減圧下で溶媒を留去して(2R,3R)-3-アミノ-2-ヒドロキシ-2-{[2-(3-メチルブチル)フロ[3,2-c]ピリジン-4-イル]メチル}-4-(メチルスルファニル)ブタン酸 二塩酸塩(100mg)を固体として得た。
(3R,4R)-3-[(2,3-ジエチルフロ[3,2-c]ピリジン-4-イル)メチル]-4-[(エチルスルファニル)メチル]-3-(メトキシメトキシ)-1-(メトキシメチル)アゼチジン-2-オン(95mg)、DOX(0.95ml)及び6M塩酸(0.73ml)の混合物を、60℃で3時間攪拌した。得られた反応混合物に氷冷下で、1M水酸化ナトリウム水溶液を加えて中和した。得られた混合物をODSカラムクロマトグラフィー(MeCN/0.1%蟻酸水溶液)にて精製した。得られた粗精製物に、過剰量の1M塩酸を加え、その後減圧下で溶媒を留去して(2R,3R)-3-アミノ-2-[(2,3-ジエチルフロ[3,2-c]ピリジン-4-イル)メチル]-4-(エチルスルファニル)-2-ヒドロキシブタン酸 二塩酸塩(75mg)を固体として得た。
(3R,4R)-4-[(4S)-2,2-ジメチル-1,3-ジオキソラン-4-イル]-3-ヒドロキシ-1-(4-メトキシフェニル)アゼチジン-2-オン(21.94g)、1,2-ジクロロエタン(300ml)、クロロ(メトキシ)メタン(23.6ml)及びDIPEA(70ml)の混合物を110℃で12時間攪拌した。得られた反応混合物に、水を加え、CHCl3で抽出した。得られた有機層を無水硫酸マグネシウムで乾燥し、有機層を減圧下で濃縮した。生じた固体をジイソプロピルエーテル-MeOHの混合溶媒で洗浄し、(3R,4S)-4-[(4S)-2,2-ジメチル-1,3-ジオキソラン-4-イル]-3-(メトキシメトキシ)-1-(4-メトキシフェニル)アゼチジン-2-オン(12.30g)を固体として得た。更に、得られたろ液を濃縮し、残渣をシリカゲルカラムクロマトグラフィー(CHCl3/MeOH)にて精製し同化合物(12.75g)を固体として得た。
(3R,4S)-4-(2-シクロプロピルエチル)-3-(メトキシメトキシ)-1-(4-メトキシフェニル)アゼチジン-2-オン(1.24g)、MeCN(30ml)及び水(15ml)の混合物に、氷冷下、硝酸セリウム(IV)アンモニウム(6.3g)を加え、30分間攪拌した。得られた反応混合物に攪拌しながら、水及び飽和炭酸水素ナトリウム水溶液を加え、その後、2%亜硫酸水素ナトリウム水溶液を加えた。得られた反応混合物をセライトろ過し、ろ液をCHCl3で抽出した。得られた有機層を飽和塩化ナトリウム水溶液で洗浄し、無水硫酸マグネシウムで乾燥した後、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、(3R,4S)-4-(2-シクロプロピルエチル)-3-(メトキシメトキシ)アゼチジン-2-オン(601mg)を固体として得た。
(3R,4S)-4-[(4S)-2,2-ジメチル-1,3-ジオキソラン-4-イル]-3-(メトキシメトキシ)アゼチジン-2-オン(302mg)、クロロ(メトキシ)メタン(0.15ml)、テトラ-n-ブチルアンモニウムヨージド(500mg)及びTHF(9ml)の混合物に、氷冷下、カリウムヘキサメチルジシラジド(1.0M THF溶液、1.5ml)を加え1時間攪拌した後、室温で終夜攪拌した。得られた反応混合物に水を加え、AcOEtで抽出した。得られた有機層を無水硫酸マグネシウムで乾燥し、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、(3R,4S)-4-[(4S)-2,2-ジメチル-1,3-ジオキソラン-4-イル]-3-(メトキシメトキシ)-1-(メトキシメチル)アゼチジン-2-オン(247mg)を油状物として得た。
(3R,4S)-4-[(4S)-2,2-ジメチル-1,3-ジオキソラン-4-イル]-3-(メトキシメトキシ)-1-(4-メトキシフェニル)アゼチジン-2-オン(3.17g)、AcOH(50ml)及び水(13ml)の混合物を50℃で4時間攪拌した。得られた反応混合物を室温まで放冷した後、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(CHCl3/MeOH)にて精製し、(3R,4S)-4-[(1S)-1,2-ジヒドロキシエチル]-3-(メトキシメトキシ)-1-(4-メトキシフェニル)アゼチジン-2-オン(2.57g)を油状物として得た。
(3R,4S)-4-[(1S)-1,2-ジヒドロキシエチル]-3-(メトキシメトキシ)-1-(4-メトキシフェニル)アゼチジン-2-オン(2.09g)、CH2Cl2(40ml)及び飽和炭酸水素ナトリウム水溶液(1ml)の混合物に、過ヨウ素酸ナトリウム(2.3g)を加え、室温で1時間攪拌した。得られた反応混合物に無水硫酸マグネシウムを加え、30分間攪拌した。得られた反応混合物をセライトろ過し、ろ液を減圧下で濃縮し、(2R,3R)-3-(メトキシメトキシ)-1-(4-メトキシフェニル)-4-オキソアゼチジン-2-カルバルデヒド(1.80g)を固体として得た。
(2R,3R)-3-(メトキシメトキシ)-1-(メトキシメチル)-4-オキソアゼチジン-2-カルバルデヒド(5.08g)とTHF(50ml)の混合物に氷冷下、NaBH4(1.2g)を加え、30分間攪拌した。得られた反応混合物に水(5ml)を加えた後に無水硫酸マグネシウムを加え、室温で30分間攪拌した。得られた反応混合物をろ過した後に、ろ液を減圧下で濃縮した。得られた残渣をシリカゲルクロマトグラフィー(CHCl3/MeOH)にて精製し、(3R,4S)-4-(ヒドロキシメチル)-3-(メトキシメトキシ)-1-(メトキシメチル)アゼチジン-2-オン(4.43g)を油状物として得た。
(3R,4S)-4-(ヒドロキシメチル)-3-(メトキシメトキシ)-1-(メトキシメチル)アゼチジン-2-オン(100mg)、トリイソプロピルクロロシラン(0.21ml)、イミダゾール(140mg)及びDMF(2ml)の混合物を室温で終夜攪拌した。得られた反応混合物を水に加え、AcOEtで抽出した。得られた有機層を飽和塩化ナトリウム水溶液で洗浄し無水硫酸マグネシウムで乾燥した後に、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、(3R,4S)-3-(メトキシメトキシ)-1-(メトキシメチル)-4-{[(トリイソプロピルシリル)オキシ]メチル}アゼチジン-2-オン(137mg)を油状物として得た。
[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メタノール(850mg)とTHF(24ml)の混合物に、氷冷下でPBr3(0.25ml)のTHF(3ml)溶液を加えた後、室温で3時間攪拌した。得られた反応混合物をあらかじめ氷冷した飽和炭酸水素ナトリウム水溶液とCH2Cl2の混合液に注ぎ、得られた混合物を室温で5分間攪拌した。有機層を分離し、水層をCH2Cl2で抽出した。得られた有機層を合わせ、無水硫酸マグネシウムで乾燥した。得られた有機層をトルエンで希釈し、減圧下で約5mlになるまで濃縮した。得られた混合物を再度トルエンで希釈し、減圧下で約5mlになるまで濃縮した(混合物A)。窒素雰囲気下、(3R,4S)-3-(メトキシメトキシ)-1-(メトキシメチル)-4-{[(トリイソプロピルシリル)オキシ]メチル}アゼチジン-2-オン(1.2g)のTHF(20ml)溶液に-78℃でLDA(1.09Mヘキサン-THF溶液、4.5ml)をゆっくり加え、30分間攪拌した。この反応混合物に混合物Aを滴下し、その後、同温で1.5時間攪拌した。得られた反応混合物に飽和塩化アンモニウム水溶液を加えた後、室温へと昇温しAcOEtで2回抽出した。得られた有機層を無水硫酸マグネシウムで乾燥し、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(CHCl3/AcOEt)にて精製し(3R,4S)-3-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-3-(メトキシメトキシ)-1-(メトキシメチル)-4-{[(トリイソプロピルシリル)オキシ]メチル}アゼチジン-2-オン(1.6g)を油状物として得た。
アルゴン雰囲気下、5-[(シクロプロピルメチル)スルホニル]-1-フェニル-1H-テトラゾール(3.32g)とTHF(60ml)の混合物を-78℃に冷却し、リチウムヘキサメチルジシラジド(1.3M THF溶液、11ml)を加え、30分間攪拌した。得られた反応混合物に、(2R,3R)-3-(メトキシメトキシ)-1-(4-メトキシフェニル)-4-オキソアゼチジン-2-カルバルデヒド(3.00g)を加え、同温で30分間攪拌した。得られた反応混合物を室温まで昇温し、飽和塩化アンモニウム水溶液を加え、AcOEtで抽出した。得られた有機層を飽和塩化ナトリウム水溶液で洗浄し、有機層を無水硫酸マグネシウムで乾燥した。有機層を減圧下で濃縮し、得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、(3R)-4-(2-シクロプロピルビニル)-3-(メトキシメトキシ)-1-(4-メトキシフェニル)アゼチジン-2-オン(1.73g)を固体として得た。
(3R)-4-(2-シクロプロピルビニル)-3-(メトキシメトキシ)-1-(4-メトキシフェニル)アゼチジン-2-オン(831mg)のトルエン(25ml)溶液に、PtO2(61mg)を加え、水素雰囲気下0℃で6時間攪拌した。得られた反応混合物から不溶物をろ別した後、ろ液を減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、(3R,4S)-4-(2-シクロプロピルエチル)-3-(メトキシメトキシ)-1-(4-メトキシフェニル)アゼチジン-2-オン(574mg)を油状物として得た。
(3R)-4-(2-シクロブチルビニル)-3-(メトキシメトキシ)-1-(4-メトキシフェニル)アゼチジン-2-オン(1.06g)とCH2Cl2(24ml)の混合物に、(1,5-シクロオクタジエン)(ピリジン)(トリシクロヘキシルホスフィン)イリジウム(I) ヘキサフルオロリン酸塩(270mg)を加え、水素雰囲気下、室温で終夜攪拌した。得られた反応混合物を減圧下で濃縮し、得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、(3R,4S)-4-(2-シクロブチルエチル)-3-(メトキシメトキシ)-1-(4-メトキシフェニル)アゼチジン-2-オン(960mg)を油状物として得た。
亜硫酸水素ナトリウム(19g)の水(130ml)溶液に、氷冷下で[(2R)-1-(メチルスルファニル)-3-オキソプロパン-2-イル]カルバミン酸 tert-ブチル(20g)、水(13ml)及びMeCN(54ml)の混合物を滴下し、室温で13時間攪拌した。得られた反応混合物にメチル-tert-ブチルエーテルを加えて攪拌した後、水層と有機層を分離した。得られた有機層を水で抽出し、先に得られた水層と混合した後、AcOEt(100ml)とシアン化カリウム(7.7g)を加え、室温で18時間攪拌した。得られた反応混合物から有機層を分離し、水層をAcOEtで2回抽出した。得られた有機層を混合した後、炭酸水素ナトリウム水溶液および飽和塩化ナトリウム水溶液で洗浄した後、無水硫酸ナトリウムで乾燥した。減圧下で溶媒を留去して[(2R)-1-シアノ-1-ヒドロキシ-3-(メチルスルファニル)プロパン-2-イル]カルバミン酸 tert-ブチル(17.2g)を油状物として得た。
氷冷下、[(2R)-1-シアノ-1-ヒドロキシ-3-(メチルスルファニル)プロパン-2-イル]カルバミン酸 tert-ブチル(17.2g)に濃塩酸(70ml)をゆっくり加えたのち、90℃で2時間攪拌した。得られた反応混合物を減圧下で濃縮し、残渣にトルエンを加え減圧下で溶媒を留去する操作を5回繰り返し、(3R)-3-アミノ-2-ヒドロキシ-4-(メチルスルファニル)ブタン酸 塩酸塩を含む残渣(18g)を油状物として得た。
(3R)-3-アミノ-2-ヒドロキシ-4-(メチルスルファニル)ブタン酸 塩酸塩(18g)、THF(90ml)、水(90ml)、4-ジメチルアミノピリジン(3.6g)及びEt3N(37ml)の混合物に2-(tert-ブトキシカルボニルオキシイミノ)-2-フェニルアセトニトリル(42g)を室温で加え、室温で3日間攪拌した。得られた反応混合物を減圧下で約半量まで濃縮し、残渣にメチル-tert-ブチルエーテルと飽和炭酸水素ナトリウム水溶液を加え、水層と有機層を分離した。得られた水層をメチル-tert-ブチルエーテルで3回洗浄し、1M塩酸を加えてpHを約2に調節した後、AcOEtで4回抽出した。得られた有機層を合わせ、飽和塩化ナトリウム水溶液で洗浄し、無水硫酸ナトリウムで乾燥した後、減圧下で溶媒を留去して、(3R)-3-[(tert-ブトキシカルボニル)アミノ]-2-ヒドロキシ-4-(メチルスルファニル)ブタン酸(13.4g)を油状物として得た。
(3R)-3-[(tert-ブトキシカルボニル)アミノ]-2-ヒドロキシ-4-(メチルスルファニル)ブタン酸(16.2g)、1,2-ジクロロエタン(186ml)、2,2-ジメトキシプロパン(82.2ml)及びp-トルエンスルホン酸ピリジニウム(770mg)の混合物を80℃で15時間攪拌した。得られた反応混合物を室温まで放冷後、2.5%炭酸水素ナトリウム水溶液(50ml)を加えて攪拌したのち、有機層を分離した。水層をCHCl3で抽出し、得られた有機層を混合した後、飽和塩化ナトリウム水溶液で洗浄した。得られた有機層を無水硫酸ナトリウムで乾燥後、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、[(1R)-1-(2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル)-2-(メチルスルファニル)エチル]カルバミン酸 tert-ブチル(12.2g)を油状物として得た。
窒素雰囲気下、N-(tert-ブトキシカルボニル)-L-セリン(20g)とDMF(480ml)の混合物に、NaH(60%鉱油分散体、8.6g)を氷冷下、内温を5℃以下に保ちながら5回に分けて加え、その後氷冷下で1時間攪拌した。得られた反応混合物に、(2-ヨードエチル)シクロプロパン(24g)を加え、室温で14時間攪拌した。得られた反応混合物を氷冷した後、水と1M塩酸を加えpHを2~3に調節した。得られた反応混合物をAcOEtで3回抽出したのち、有機層を飽和塩化ナトリウム水溶液で洗浄した。得られた有機層を無水硫酸ナトリウムで乾燥し、減圧下で濃縮した。得られた残渣に、MeOH(140ml)、CH2Cl2(420ml)を加えたのち氷冷し、(ジアゾメチル)(トリメチル)シラン(2Mヘキサン溶液、62ml)を、内温を6℃以下に保ちながら滴下し、その後氷冷下で10分間、室温で1時間攪拌した。得られた反応混合物にAcOHを加えた後、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、メチル N-(tert-ブトキシカルボニル)-O-(2-シクロプロピルエチル)-L-セリナート(6.51g)を油状物として得た。
窒素雰囲気下、[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メタノール(1.33g)のTHF溶液(42ml)にPBr3(0.58ml)を室温で滴下した。反応混合物を室温で1時間攪拌した後、氷冷した飽和炭酸水素ナトリウム水溶液とCH2Cl2の混合物に加えた。得られた混合物を30分間攪拌後、有機層を分離し、水層をCH2Cl2で抽出した。得られた有機層を合わせて飽和塩化ナトリウム水溶液で洗浄後、無水硫酸マグネシウムで乾燥し、トルエンで希釈し、減圧下で約2mlになるまで濃縮した。(混合物A)。
窒素雰囲気下、[(1R)-1-(2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル)-2-(メチルスルファニル)エチル]カルバミン酸 tert-ブチル(1.7g)のTHF溶液(34ml)をドライアイス-アセトン浴で冷却し、LDA(1.09Mヘキサン-THF溶液、12ml)を滴下した。得られた反応混合物をドライアイス-アセトン浴で冷却したまま30分間攪拌後、混合物Aを滴下し、さらに2時間攪拌した。得られた混合物に飽和塩化アンモニウム水溶液を加え、室温まで昇温した。得られた混合物をCHCl3で抽出し、無水硫酸ナトリウムで乾燥後、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、[(1R)-1-[(4R)-4-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル]-2-(メチルスルファニル)エチル]カルバミン酸 tert-ブチル(923mg)を油状物として得た。
窒素雰囲気下、メチル N-(tert-ブトキシカルボニル)-O-(2-シクロプロピルエチル)-L-セリナート(6.5g)、ジブロモメタン(8.0g)及びTHF(22ml)の混合物に、-20℃で2,2,6,6-テトラメチルピペリジニルマグネシウムクロリド リチウムクロリド錯体(1M THF-トルエン溶液、91ml)を内温-11℃以下を保ちながら2時間かけて滴下し、その後、-15℃で2時間攪拌した。反応混合物をあらかじめ氷冷した5%クエン酸水溶液とAcOEtの混合物に注ぎ、その後10分間攪拌した。有機層と水層を分離し、得られた有機層を5%クエン酸水溶液で3回洗浄した後、飽和塩化ナトリウム水溶液で洗浄した。得られた有機層を無水硫酸ナトリウムで乾燥した後、減圧下で濃縮し、[(2S)-4,4-ジブロモ-1-(2-シクロプロピルエトキシ)-3-オキソブタン-2-イル]カルバミン酸 tert-ブチルを含む残渣(10.7g)を油状物として得た。
[(2S)-4,4-ジブロモ-1-(2-シクロプロピルエトキシ)-3-オキソブタン-2-イル]カルバミン酸 tert-ブチル(9.6g)とトルエン(76ml)の混合物に、氷冷下で2M水酸化ナトリウム水溶液(57ml)を15分間かけて滴下し、その後、室温で2時間攪拌した。得られた反応混合物にトルエンと水を加えた後、有機層と水層を分離した。有機層を水で2回抽出し、先に得られた水層と合わせた後、AcOEtを加えた。得られた混合物を氷冷した後、2M塩酸を加え水層のpHを約1.5に調節した。得られた反応混合物の有機層と水層を分離し、水層をAcOEtで3回抽出した。得られた有機層を合わせ、無水硫酸ナトリウムで乾燥した。得られた有機層を減圧下で濃縮し、(3S)-3-[(tert-ブトキシカルボニル)アミノ]-4-(2-シクロプロピルエトキシ)-2-ヒドロキシブタン酸(4.53g)を油状物として得た。
3-{[2-シアノ-3-(ヘキサ-1-イン-1-イル)ピリジン-4-イル]スルファニル}プロパン酸 2-エチルヘキシル(271mg)とMeOH(5.5ml)の混合物に、室温でナトリウムメトキシド(28% MeOH溶液、0.2ml)を加え、40℃で2時間攪拌した後、ナトリウムメトキシド(28% MeOH溶液、0.14ml)を追加し、同温でさらに1.5時間攪拌した。得られた反応混合物に氷冷下3M塩酸(0.8ml)を加え、室温で2.5時間攪拌した。得られた反応混合物を減圧下で濃縮し、残渣に飽和炭酸水素ナトリウム水溶液を加えAcOEtで3回抽出した。得られた有機層を無水硫酸マグネシウムで乾燥し、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、2-ブチルチエノ[3,2-c]ピリジン-4-カルボン酸メチル(107mg)を油状物として得た。
[(1R)-1-[(4R)-4-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル]-2-(メチルスルファニル)エチル]カルバミン酸 tert-ブチル(105mg)とCH2Cl2(2ml)の混合物を氷冷した後、m-クロロ過安息香酸(約25%含水、48mg)を加え、同温で1時間攪拌した。得られた反応混合物に氷冷下で10%チオ硫酸ナトリウム水溶液を加え10分間攪拌した。水層と有機層を分離した後、有機層を飽和炭酸水素ナトリウム水溶液で2回洗浄した。得られた有機層を無水硫酸マグネシウムで乾燥後、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(CHCl3/MeOH)にて精製し、(1R)-1-[(4R)-4-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル]-2-(メチルスルフィニル)エチル]カルバミン酸 tert-ブチル(85mg)を泡状固体として得た。
[(1R)-1-[(4R)-4-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル]-2-(メチルスルファニル)エチル]カルバミン酸 tert-ブチル(121mg)とCH2Cl2(7ml)の混合物を氷冷した後、m-クロロ過安息香酸(約25%含水、111mg)を加え、室温で1時間攪拌した。得られた反応混合物を再度氷冷し、m-クロロ過安息香酸(約25%含水、11mg)を加え、室温で30分間攪拌した。得られた反応混合物に10%チオ硫酸ナトリウム水溶液を氷冷下で加え10分間攪拌した。水層と有機層を分離した後、有機層を飽和炭酸水素ナトリウム水溶液で2回洗浄した。得られた有機層を無水硫酸マグネシウムで乾燥後、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(CHCl3/AcOEt)にて精製し、[(1R)-1-[(4R)-4-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル]-2-(メチルスルホニル)エチル]カルバミン酸 tert-ブチル(83mg)を泡状固体として得た。
(3R,4S)-3-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-3-(メトキシメトキシ)-1-(メトキシメチル)-4-{[(トリイソプロピルシリル)オキシ]メチル}アゼチジン-2-オン(1.6g)とTHF(30ml)の混合物を氷冷した後、フッ化テトラ-n-ブチルアンモニウム(1M THF溶液、3ml)を加え、同温で1時間攪拌した。反応混合物に飽和塩化アンモニウム水溶液を加え、AcOEtで2回抽出した。有機層を飽和塩化ナトリウム水溶液で洗浄し、無水硫酸マグネシウムで乾燥後、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(CHCl3/MeOH)にて精製し、(3R,4S)-3-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-4-(ヒドロキシメチル)-3-(メトキシメトキシ)-1-(メトキシメチル)アゼチジン-2-オン(937mg)を油状物として得た。
(3R,4S)-3-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-4-(ヒドロキシメチル)-3-(メトキシメトキシ)-1-(メトキシメチル)アゼチジン-2-オン(936mg)、ピリジン(0.75ml)及びCH2Cl2(10ml)の混合物に、塩化メタンスルホニル(0.37ml)を室温で加え、同温で15時間攪拌した。得られた反応混合物にCHCl3を加え、0.5M塩酸、飽和炭酸水素ナトリウム水溶液、飽和塩化ナトリウム水溶液の順に洗浄した。得られた有機層を無水硫酸マグネシウムで乾燥し、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、メタンスルホン酸 [(2S,3R)-3-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-3-(メトキシメトキシ)-1-(メトキシメチル)-4-オキソアゼチジン-2-イル]メチル(1.11g)を油状物として得た。
メタンスルホン酸 [(2S,3R)-3-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-3-(メトキシメトキシ)-1-(メトキシメチル)-4-オキソアゼチジン-2-イル]メチル(1.11g)、DMF(20ml)及びチオ酢酸カリウム(400mg)の混合物を60℃で7時間攪拌した。得られた反応混合物に室温でチオ酢酸カリウム(53mg)を追加し、60℃でさらに12時間攪拌した。得られた反応混合物にAcOEtを加え、水、飽和炭酸水素ナトリウム水溶液、飽和塩化ナトリウム水溶液の順に洗浄し、無水硫酸マグネシウムで乾燥した。得られた有機層を減圧下で濃縮し、チオ酢酸S-{[(2R,3R)-3-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-3-(メトキシメトキシ)-1-(メトキシメチル)-4-オキソアゼチジン-2-イル]メチル}(1.02g)を油状物として得た。
窒素雰囲気下、チオ酢酸 S-{[(2R,3R)-3-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-3-(メトキシメトキシ)-1-(メトキシメチル)-4-オキソアゼチジン-2-イル]メチル}(80mg)、ヨードエタン(0.03ml)、DMF(0.8ml)及びMeOH(0.8ml)の混合物に炭酸カリウム(50mg)を加え、室温で16時間攪拌した。得られた反応混合物にAcOEtを加え、水、飽和炭酸水素ナトリウム水溶液、飽和塩化ナトリウム水溶液の順に洗浄し、無水硫酸マグネシウムで乾燥した。得られた有機層を減圧下で濃縮し、(3R,4R)-3-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-4-[(エチルスルファニル)メチル]-3-(メトキシメトキシ)-1-(メトキシメチル)アゼチジン-2-オン(67mg)を油状物として得た。
窒素雰囲気下、トリメチルシリルアセチレン(25ml)とTHF(170ml)の混合物を-78℃に冷却し、n-ブチルリチウム(1.6Mヘキサン溶液、115ml)を滴下した。得られた反応混合物を氷冷下15分間攪拌後、再度-78℃に冷却した。得られた反応混合物にN,N,N',N',N'',N''-ヘキサメチルリン酸 トリアミド(32ml)を加え、同温で30分間攪拌した後、(2-ブロモエチル)シクロプロパン(27g)を5分間かけて滴下し同温で30分間攪拌した。得られた反応混合物を室温まで昇温し、16時間攪拌した。得られた反応混合物に氷冷下で水を加え、有機層を分離した。得られた有機層を飽和塩化ナトリウム水溶液で洗浄し、無水硫酸マグネシウムで乾燥後、減圧下で濃縮して(4-シクロプロピルブタ-1-イン-1-イル)(トリメチル)シラン(31.8g)を油状物として得た。
アルゴン雰囲気下、3-ブロモピリジン-4(1H)-オン(500mg)、(4-シクロプロピルブタ-1-イン-1-イル)(トリメチル)シラン(1.44g)、Et3N(2.8ml)及びDMF(5ml)の混合物にフッ化テトラ-n-ブチルアンモニウム(1M THF溶液、8.8ml)を加えた。得られた混合物を30秒間超音波処理した後、ビス(トリフェニルホスフィン)パラジウム(II)ジクロリド(420mg)を加え、マイクロ波照射下、110℃で1時間攪拌した。得られた反応混合物にAcOEtとシリカゲルを加え、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン(302mg)を油状物として得た。
窒素雰囲気下、2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン(300mg)とCHCl3(6ml)の混合物に氷冷下m-クロロ過安息香酸(約25%含水、555mg)を加えた。得られた反応混合物を室温で8時間攪拌した後、氷冷し、m-クロロ過安息香酸(約25%含水、300mg)を再度加えた。得られた反応混合物を室温でさらに16時間攪拌した。得られた反応混合物を氷冷した後、飽和炭酸水素ナトリウム水溶液と5%亜硫酸ナトリウム水溶液を加え、CHCl3で3回抽出した。得られた有機層を無水硫酸ナトリウムで乾燥した後、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(CHCl3/MeOH)にて精製し、2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン 5-オキシド(190mg)を油状物として得た。
窒素雰囲気下、2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン 5-オキシド(190mg)、Et3N(0.33ml)及びMeCN(4ml)の混合物にトリメチルシリルシアニド(0.183ml)を加え、85℃で16時間攪拌した。得られた反応混合物を室温まで放冷後、Et3N(0.65ml)とトリメチルシリルシアニド(0.35ml)を追加した。反応混合物を再度85℃で3.5時間攪拌した後、得られた反応混合物にAcOEtを加え、飽和炭酸水素ナトリウム水溶液、飽和塩化ナトリウム水溶液の順に洗浄した。得られた有機層を無水硫酸マグネシウムで乾燥し、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-カルボニトリル(148mg)を油状物として得た。
2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-カルボニトリル(148mg)とMeOH(1.5ml)の混合物に、氷冷下、ナトリウムメトキシド(28% MeOH溶液、0.14ml)を加え、室温で1.5時間攪拌した。得られた反応混合物に氷冷下3M塩酸(0.7ml)を加え、室温で18時間攪拌した。得られた反応混合物に水とMeOHを加え、減圧下で濃縮した。得られた残渣に飽和炭酸水素ナトリウム水溶液を加えAcOEtで2回抽出した。得られた有機層を飽和塩化ナトリウム水溶液で洗浄した後に、無水硫酸マグネシウムで乾燥し、有機層を減圧下で濃縮した。窒素雰囲気下、得られた残渣とMeOH(4ml)の混合物に、氷冷下でNaBH4(80mg)を加え、攪拌しながら1時間かけて徐々に室温へと昇温し、その後5時間攪拌した。得られた反応混合物を再度氷冷した後、NaBH4(80mg)を加え、攪拌しながら1時間かけて徐々に室温へと昇温し、その後15時間攪拌した。得られた反応混合物に室温でアセトンを加えた後、減圧下で濃縮した。得られた残渣に水を加え、AcOEtで3回抽出した。得られた有機層を無水硫酸マグネシウムで乾燥し、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEtで溶出したのち、CHCl3/MeOHで溶出)にて精製し、[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メタノール(123mg)を油状物として得た。
窒素雰囲気下、4-クロロ-3-ヨードピリジン 1-オキシド(21g)と1,2-ジクロロエタン(230ml)の混合物にN,N-ジメチルカルバモイルクロリド(12.5ml)を加え、室温で30分間攪拌した。得られた反応混合物にトリメチルシリルシアニド(20.5ml)を加え、60℃で6時間攪拌した。得られた反応混合物を室温まで放冷後、飽和炭酸水素ナトリウム水溶液(260ml)を加え、30分間攪拌した。得られた混合物をAcOEtで2回抽出し、有機層を飽和塩化ナトリウム水溶液で洗浄した後、無水硫酸マグネシウムで乾燥した。得られた有機層を減圧下で濃縮し、残渣をヘキサンとイソプロパノールの混合液で洗浄して、4-クロロ-3-ヨードピリジン-2-カルボニトリル(7.1g)を固体として得た。
4-クロロ-3-ヨードピリジン-2-カルボニトリル(8.4g)とDMF(170ml)の混合物に酢酸セシウム(30g)を加え、100℃で終夜攪拌した。得られた反応混合物に氷冷下、1M塩酸(191ml)を加え、AcOEtで2回抽出した。得られた有機層を5%塩化ナトリウム水溶液で2回洗浄した後、無水硫酸マグネシウムで乾燥した。得られた有機層を減圧下で濃縮し、残渣にトルエンを加え再度減圧下で濃縮する操作を3回繰り返した。得られた残渣にジイソプロピルエーテルを加え、生じた不溶物をろ取した。得られた固体をジイソプロピルエーテルとイソプロパノールの混合液で洗浄して4-ヒドロキシ-3-ヨードピリジン-2-カルボニトリル(4.4g)を固体として得た。
4-クロロ-3-ヨードピリジン-2-カルボニトリル(1.82g)、Et3N(1.92ml)及びDMF(18ml)の混合物に氷冷下、3-スルファニルプロパン酸 2-エチルヘキシル(1.73ml)を加え、その後、室温で18時間攪拌した。得られた反応混合物にAcOEtと水を加え、有機層を分離した。得られた有機層を飽和塩化ナトリウム水溶液で洗浄し、無水硫酸マグネシウムで乾燥した。得られた有機層を減圧下で濃縮し、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、3-[(2-シアノ-3-ヨードピリジン-4-イル)スルファニル]プロパン酸 2-エチルヘキシル(254mg)を油状物として得た。
窒素雰囲気下、3-[(2-シアノ-3-ヨードピリジン-4-イル)スルファニル]プロパン酸 2-エチルヘキシル(252mg)、1-ヘキシン(0.128ml)、Et3N(0.394ml)及びMeCN(5ml)の混合物にビス(トリフェニルホスフィン)パラジウム(II)ジクロリド(79mg)を加え、70℃で3.5時間攪拌した。得られた反応混合物を室温まで放冷後、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、3-{[2-シアノ-3-(ヘキサ-1-イン-1-イル)ピリジン-4-イル]スルファニル}プロパン酸 2-エチルヘキシル(178mg)を油状物として得た。
窒素雰囲気下、2-ブチルチエノ[3,2-c]ピリジン-4-カルボン酸メチル(112mg)とMeOH(3.4ml)の混合物に、氷冷下、NaBH4(102mg)を加え、攪拌しながら1時間かけて徐々に室温へと昇温し、その後12時間攪拌した。得られた反応混合物に室温でアセトンを加えた後、減圧下で濃縮した。得られた残渣に水を加え、AcOEtで3回抽出した。得られた有機層を無水硫酸マグネシウムで乾燥し、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(CHCl3/MeOH)にて精製し、(2-ブチルチエノ[3,2-c]ピリジン-4-イル)メタノール(95mg)を油状物として得た。
窒素雰囲気下、(2-ブチルチエノ[3,2-c]ピリジン-4-イル)メタノール(113mg)とCH2Cl2(1.2ml)の混合物に氷冷下で塩化チオニル(0.075ml)のCH2Cl2(0.6ml)溶液を加えた。得られた反応混合物を室温で1時間攪拌した。得られた反応混合物を減圧下で濃縮し、2-ブチル-4-(クロロメチル)チエノ[3,2-c]ピリジン 塩酸塩(140mg)を固体として得た。
窒素雰囲気下、2-ブチル-4-(クロロメチル)チエノ[3,2-c]ピリジン 塩酸塩(139mg)とCH2Cl2(5.6ml)の混合物にヨウ化ナトリウム(225mg)を加え、室温で30分間攪拌した。得られた反応混合物に飽和炭酸水素ナトリウム水溶液を加え、5分間攪拌した後、CH2Cl2で2回抽出した。得られた有機層を無水硫酸マグネシウムで乾燥した後、トルエンを加えて希釈した。得られた混合物を減圧下で約5mlになるまで濃縮した。得られた混合物に再度トルエンを加え、減圧下で約3mlになるまで濃縮する操作を2回繰り返した(混合物A)。
窒素雰囲気下、[(1R)-1-(2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル)-2-(メチルスルファニル)エチル]カルバミン酸 tert-ブチル(143mg)とTHF(2.8ml)の混合液に攪拌しながらLDA(1.13Mヘキサン-THF溶液、1ml)を-78℃で滴下した。得られた反応混合物を同温にて30分間攪拌した後、混合物Aを滴下した。得られた反応混合物を同温にて1時間攪拌した後、飽和塩化アンモニウム水溶液を加え、室温まで昇温した。得られた混合物をAcOEtで2回抽出し、有機層を無水硫酸マグネシウムで乾燥した。得られた有機層を減圧下で濃縮し、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製して、[(1R)-1-{(4R)-4-[(2-ブチルチエノ[3,2-c]ピリジン-4-イル)メチル]-2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル}-2-(メチルスルファニル)エチル]カルバミン酸 tert-ブチル(170mg)を油状物として得た。
窒素雰囲気下、4-ヒドロキシ-3-ヨードピリジン-2-カルボニトリル(400mg)、エチニルシクロペンタン(0.205ml)、Et3N(0.91ml)及びMeCN(8ml)の混合物にCuI(93mg)とビス(トリフェニルホスフィン)パラジウム(II)ジクロリド(58mg)を加え、70℃で終夜攪拌した。得られた反応混合物を室温まで放冷後、減圧下で濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、2-シクロペンチルフロ[3,2-c]ピリジン-4-カルボニトリル(270mg)を油状物として得た。
[(1R)-1-[(4R)-4-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル]-2-(メチルスルファニル)エチル]カルバミン酸 tert-ブチル(200mg)、AcOH(2mL)及びEtOH(2mL)の混合物に10%Pd/C (50%含水、200mg)を加え、3.5気圧水素雰囲気下、室温で5日間攪拌した。得られた反応混合物にセライトを加え、不溶物をろ別した。ろ液を減圧下で濃縮し、得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、[(1R)-1-[(4R)-4-{[2-(2-シクロプロピルエチル)-2,3-ジヒドロフロ[3,2-c]ピリジン-4-イル]メチル}-2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル]-2-(メチルスルファニル)エチル]カルバミン酸 tert-ブチル(58mg)を油状物として得た。
窒素雰囲気下、4-ヒドロキシ-3-ヨードピリジン-2-カルボニトリル(800mg)、3-ヘキシン(1.12ml)、Et3N(1.82ml)及びDMF(16ml)の混合物にビス(トリフェニルホスフィン)パラジウム(II)ジクロリド(230mg)を加え、マイクロ波照射下、150℃で2時間攪拌した。得られた反応混合物に、水を加えAcOEtで抽出した。有機層を減圧下で濃縮し、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製し、2,3-ジエチルフロ[3,2-c]ピリジン-4-カルボニトリル(215mg)を油状物として得た。
2,3-ジエチルフロ[3,2-c]ピリジン-4-カルボニトリル(240mg)とMeOH(3ml)の混合物に、ナトリウムメトキシド(28% MeOH溶液、0.29ml)を加え、40℃で終夜攪拌した。得られた反応混合物に再度ナトリウムメトキシド(28% MeOH溶液、0.29ml)を加え、60℃で3時間攪拌した。得られた反応混合物に6M水酸化ナトリウム水溶液(3ml)とEtOH(3ml)を加え、80℃で終夜攪拌した。得られた反応混合物に氷冷下、6M塩酸(3.5ml)を加えて中和し、水を加えCHCl3で2回抽出した。得られた有機層を無水硫酸マグネシウムで乾燥し、2,3-ジエチルフロ[3,2-c]ピリジン-4-カルボン酸(150mg)を油状物として得た。
窒素雰囲気下、2,3-ジエチルフロ[3,2-c]ピリジン-4-カルボン酸(140mg)とTHF(1.5ml)の混合物に、氷冷下でクロロギ酸イソブチル(0.1ml)とN-メチルモリホリン(0.09ml)を加え、同温で30分間攪拌した。得られた反応混合物から不溶物をろ別し、得られたろ液をNaBH4(105mg)と水(1.5ml)の混合物に氷冷下で加え、室温で2時間攪拌した。得られた反応混合物を氷冷し、アセトン(0.2ml)を加え、AcOEtで抽出した。得られた有機層を飽和塩化ナトリウム水溶液で洗浄し、無水硫酸マグネシウムで乾燥した。得られた有機層を減圧下で濃縮し、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製して、(2,3-ジエチルフロ[3,2-c]ピリジン-4-イル)メタノール(70mg)を油状物として得た。
4-(クロロメチル)-2,3-ジエチルフロ[3,2-c]ピリジン 塩酸塩(73mg)とCH2Cl2(1.4ml)の混合物にヨウ化ナトリウム(155mg)を加え、室温で1時間攪拌した。得られた反応混合物に飽和炭酸水素ナトリウム水溶液を加え、5分間攪拌した後、トルエンで抽出した。得られた有機層を飽和塩化ナトリウム水溶液で洗浄し、無水硫酸マグネシウムで乾燥し、減圧下で濃縮した。得られた残渣をトルエンで2回共沸した後、トルエン(1ml)を加えて溶液を得た(混合物A)。
アルゴン雰囲気下、(3R,4R)-4-[(エチルスルファニル)メチル]-3-(メトキシメトキシ)-1-(メトキシメチル)アゼチジン-2-オン(70mg)とTHF(2.8ml)の混合液に攪拌しながらLDA(1.13Mヘキサン-THF溶液、0.3ml)を-78℃で滴下した。得られた反応混合物を同温にて20分間攪拌した後、混合物Aを滴下した。得られた反応混合物を同温にて2時間攪拌した後、AcOH(0.025ml)を加え室温まで昇温した。得られた混合物に水を加えCHCl3で抽出した。得られた有機層を飽和塩化ナトリウム水溶液で洗浄し、無水硫酸マグネシウムで乾燥した。得られた有機層を減圧下で濃縮し、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製して、(3R,4R)-3-[(2,3-ジエチルフロ[3,2-c]ピリジン-4-イル)メチル]-4-[(エチルスルファニル)メチル]-3-(メトキシメトキシ)-1-(メトキシメチル)アゼチジン-2-オン(95mg)を油状物として得た。
メタンスルホン酸 [(2S,3R)-3-(メトキシメトキシ)-1-(メトキシメチル)-4-オキソアゼチジン-2-イル]メチル(900mg)のDMF(9ml)溶液に、氷冷下、ナトリウムエタンチオラート(505mg)を加え、同温で1時間攪拌した。得られた反応混合物にAcOEtを加え、水、飽和炭酸水素ナトリウム水溶液、飽和塩化ナトリウム水溶液の順に洗浄し、無水硫酸マグネシウムで乾燥した。得られた有機層を減圧下で濃縮し、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製して、(3R,4R)-4-[(エチルスルファニル)メチル]-3-(メトキシメトキシ)-1-(メトキシメチル)アゼチジン-2-オン(557mg)を油状物として得た。
窒素雰囲気下、2-ブチル-7-メチルフロ[3,2-c]ピリジン(3.3g)とCHCl3(40ml)の混合物に氷冷下で過酸化水素尿素(2.46g)とメチルトリオキソレニウム(VII)(85.7mg)を加えた。得られた反応混合物を室温で30分間攪拌後、再度氷冷し、過酸化水素尿素(2.47g)とメチルトリオキソレニウム(VII)(131mg)を加えた。得られた反応混合物を室温で1時間攪拌後、二酸化マンガン(150mg)を加え、さらに1.5時間攪拌した。不溶物をろ別し、CHCl3-MeOH(98:2)で洗浄した。得られたろ液に5%亜硫酸ナトリウム水溶液を加え、得られた混合物をCHCl3-MeOH(98:2)で3回抽出した。得られた有機層を無水硫酸マグネシウムで乾燥後、減圧下濃縮し、2-ブチル-7-メチルフロ[3,2-c]ピリジン 5-オキシド(3.97g)を固体として得た。
窒素雰囲気下、チオ酢酸 S-{[(2R,3R)-3-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-3-(メトキシメトキシ)-1-(メトキシメチル)-4-オキソアゼチジン-2-イル]メチル}(80mg)、(ブロモメチル)シクロプロパン(0.035ml)、ヨウ化ナトリウム(52mg)、DMF(0.8ml)及びMeOH(0.8ml)の混合物に炭酸カリウム(50mg)を加え、室温で4時間攪拌した。得られた反応混合物にAcOEtを加え、水、飽和炭酸水素ナトリウム水溶液、飽和塩化ナトリウム水溶液の順に洗浄した。得られた有機層を無水硫酸マグネシウムで乾燥したのち減圧下で濃縮し、(3R,4R)-3-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-4-{[(シクロプロピルメチル)スルファニル]メチル}-3-(メトキシメトキシ)-1-(メトキシメチル)アゼチジン-2-オン(73mg)を油状物として得た。
(3S)-3-アミノ-2-ヒドロキシ-4-(メチルスルファニル)ブタン酸 塩酸塩(27.7g)、THF(139ml)、水(139ml)、Et3N(57.4ml)の混合物に二炭酸ジ-tert-ブチル(60ml)を加え、室温で終夜攪拌した。得られた反応混合物に、N,N-ジメチルエチレンジアミン(7.5ml)を加え、室温で2時間攪拌した。得られた反応混合物に1M硫酸水素ナトリウム水溶液(371ml)を加え、水層と有機層を分離した。得られた水層をAcOEtで3回抽出した。得られた有機層を混合し、飽和塩化ナトリウム水溶液で洗浄した後、無水硫酸ナトリウムで乾燥した。得られた有機層を減圧下で濃縮し、残渣にAcOEt(416ml)とジシクロヘキシルアミン(27.3ml)を加え、室温で終夜攪拌した。得られた混合物を減圧下で濃縮し、残渣にジイソプロピルエーテル(416ml)を加え、室温で1時間、氷冷下で2時間攪拌したのち、室温で終夜攪拌した。得られた混合物から不溶物をろ別した後、ろ液を減圧下で濃縮した。得られた残渣にジイソプロピルエーテル(416ml)を加え、室温で3日間攪拌した。生じた不溶物をろ取し、減圧下で乾燥した。得られた固体に再度ジイソプロピルエーテル(416ml)を加え、室温で1時間攪拌した後に不溶物をろ取して、(3S)-3-[(tert-ブトキシカルボニル)アミノ]-2-ヒドロキシ-4-(メチルスルファニル)ブタン酸 - ジシクロヘキシルアミン(1:1)(30.2g)を固体として得た。
アルゴン雰囲気下、(3S)-3-[(tert-ブトキシカルボニル)アミノ]-2-ヒドロキシ-4-(メチルスルファニル)ブタン酸 - ジシクロヘキシルアミン (1:1)(30.1g)とAcOEt(301ml)の混合物に2,2-ジメトキシプロパン(83ml)及びp-トルエンスルホン酸 一水和物(14.1g)を加え、70℃で16時間攪拌した。得られた反応混合物を室温まで放冷後、不溶物をろ別した。得られたろ液を2.5%炭酸水素ナトリウム水溶液で1回、0.5M塩酸で3回、2.5%炭酸水素ナトリウム水溶液で1回の順に洗浄した。得られた有機層を飽和塩化ナトリウム水溶液で洗浄し、無水硫酸ナトリウムで乾燥した。得られた有機層を減圧下で濃縮し、残渣にAcOEt(90ml)、ヘキサン(180ml)、シリカゲル(28g)及び活性炭(2.8g)を加えて室温で1時間攪拌した。得られた混合物から不溶物をろ別し、得られたろ液を減圧下で濃縮して[(1S)-1-(2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル)-2-(メチルスルファニル)エチル]カルバミン酸 tert-ブチル(12.7g)を油状物として得た。
製造例37に記載の方法と同様にして、[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メタノール(1.41g)から4-(クロロメチル)-2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン 塩酸塩(1.76g)を固体として得た。窒素雰囲気下、得られた4-(クロロメチル)-2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン 塩酸塩(668mg)とCH2Cl2(7.5ml)の混合物にヨウ化ナトリウム(810mg)を加え、室温で1時間攪拌した。得られた反応混合物に飽和炭酸水素ナトリウム水溶液を加え、5分間攪拌した後、トルエンで2回抽出した。得られた有機層を飽和塩化ナトリウム水溶液で洗浄し、無水硫酸ナトリウムで乾燥したのち、減圧下で濃縮した。得られた混合物に再度トルエンを加え、減圧下で濃縮する操作を3回繰り返し、最終的に約5mlのトルエン溶液とした(混合物A)。
アルゴン雰囲気下、[(1S)-1-(2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル)-2-(メチルスルファニル)エチル]カルバミン酸 tert-ブチル(500mg)とTHF(5ml)の混合液に攪拌しながらLDA(1.13Mヘキサン-THF溶液、1.45ml)を-78℃で滴下した。得られた反応混合物を同温にて10分間攪拌した後、クロロトリメチルシラン(0.207ml)を加え、同温にて30分間攪拌した。得られた反応混合物を氷冷下30分間攪拌し、その後、-78℃で攪拌しながらLDA(1.13Mヘキサン-THF溶液、1.74ml)を滴下した。得られた反応混合物を同温で30分間攪拌した後、混合物Aを滴下した。得られた反応混合物を同温にて2時間攪拌した後、AcOH(0.094ml)を加え、同温にて10分間攪拌した。得られた反応混合物に9.5Mジメチルアミン水溶液(0.34ml)を加え、氷冷下で30分間攪拌した。得られた混合物に1M塩酸を加えて中和したのち、AcOEtで2回抽出した。
得られた有機層を飽和塩化ナトリウム水溶液で洗浄し、無水硫酸ナトリウムで乾燥した。得られた有機層を減圧下で濃縮し、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/AcOEt)にて精製して、[(1S)-1-[(4R)-4-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2,2-ジメチル-5-オキソ-1,3-ジオキソラン-4-イル]-2-(メチルスルファニル)エチル]カルバミン酸 tert-ブチル(230mg)を油状物として得た。
Claims (16)
- 式(I)の化合物又はその塩。
Xは、O又はSであり、
点線は、単結合又は二重結合であり、
R1は、G1群から選択される1~4個の置換基を有していてもよい低級アルキル、G2群から選択される1~5個の置換基を有していてもよいシクロアルキル、又は、-低級アルキレン-(G2群から選択される1~5個の置換基を有していてもよいシクロアルキル)であり、
R2及びR5は、同一又は異なって、H、低級アルキル又はシクロアルキルであり、
R3は、-低級アルキレン-X3-低級アルキル、-低級アルキレン-X3-低級アルキレン-(G2群から選択される1~5個の置換基を有していてもよいシクロアルキル)、-低級アルキレン-(G2群から選択される1~5個の置換基を有していてもよいシクロアルキル)、又は、-低級アルキレン-X3-(G2群から選択される1~5個の置換基を有していてもよいシクロアルキル)であり、
X3は、O又はS(O)nであり、nは0、1又は2であり、
R4は、OH、NH2又は-O-低級アルキルであり、且つR6はHであるか、又はR4とR6は、それらが結合する-C(=O)-C-O-と一体となり、2,2-ジ(低級アルキル)-4-オキソ-1,3-ジオキソラン-5,5-ジイルを形成し、
G1群は、ハロゲン、OH、-O-低級アルキル、-S-低級アルキル及び-O-ハロゲノ低級アルキルであり、及び
G2群は、低級アルキル、ハロゲン、ハロゲノ低級アルキル、OH、-O-低級アルキル、-S-低級アルキル及び-O-ハロゲノ低級アルキルである。) - R4が、OH、NH2又は-O-低級アルキルであり、且つR6がHである請求項1記載の化合物又はその塩。
- XがOであり、且つ点線が単結合又は二重結合であるか、又は、XがSであり、且つ点線が二重結合である、請求項2記載の化合物又はその塩。
- R1が、ハロゲン、OH、及び-O-低級アルキルからなる群から選択される1~4個の置換基を有していてもよい低級アルキル;1~2個の低級アルキルで置換されていてもよいシクロアルキル;又は、-低級アルキレン-(1~2個の低級アルキルで置換されていてもよいシクロアルキル)であり、
R3が、-低級アルキレン-X3-低級アルキル、-低級アルキレン-X3-低級アルキレン-(1~2個の低級アルキルで置換されていてもよいシクロアルキル)、-低級アルキレン-(1~2個の低級アルキルで置換されていてもよいシクロアルキル)、又は、-低級アルキレン-X3-(1~2個の低級アルキルで置換されていてもよいシクロアルキル)であり、及び、
R2及びR5が、同一又は異なって、H又は低級アルキルである、請求項3記載の化合物又はその塩。 - R1が、ハロゲン及びOHからなる群から選択される1~4個の置換基を有していてもよい低級アルキル;シクロアルキル;又は、-低級アルキレン-シクロアルキルであり、及び
R3が、-低級アルキレン-S(O)n -低級アルキル、-低級アルキレン-O-低級アルキレン-シクロアルキル、-低級アルキレン-S-低級アルキレン-シクロアルキル、-低級アルキレン-シクロアルキル、又は、-低級アルキレン-S-シクロアルキルである、請求項4記載の化合物又はその塩。 - R1が、低級アルキル、シクロアルキル、又は、-低級アルキレン-シクロアルキルであり、R3が、-低級アルキレン-S -低級アルキル、-低級アルキレン-O-低級アルキレン-シクロアルキル、-低級アルキレン-S-低級アルキレン-シクロアルキル、-低級アルキレン-シクロアルキル、又は、-低級アルキレン-S-シクロアルキルであり、R2がH又は低級アルキルであり、R5がHであり、及び、R4がOHである、請求項5記載の化合物又はその塩。
- XがOであり、且つ点線が二重結合であり、R1が、低級アルキル、又は、-低級アルキレン-シクロアルキルであり、及び、R3が、-低級アルキレン-S-低級アルキル、又は、-低級アルキレン-シクロアルキルである、請求項6記載の化合物又はその塩。
- XがOであり、且つ点線が二重結合である、請求項2記載の化合物又はその塩。
- (2R,3R)-3-アミノ-2-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-4-(エチルスルファニル)-2-ヒドロキシブタン酸、
(2R,3S)-3-アミノ-5-シクロプロピル-2-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2-ヒドロキシペンタン酸、
(2R,3R)-3-アミノ-2-{[2-(2-シクロプロピルエチル)フロ[3,2-c]ピリジン-4-イル]メチル}-2-ヒドロキシ-4-(メチルスルファニル)ブタン酸、及び
(2R,3R)-3-アミノ-2-[(2-ブチル-7-メチルフロ[3,2-c]ピリジン-4-イル)メチル]-2-ヒドロキシ-4-(メチルスルファニル)ブタン酸、
からなる群から選択される化合物又はその塩である、請求項1記載の化合物又はその塩。 - 請求項1記載の化合物又はその塩を含有する医薬組成物。
- 医薬組成物が夜間頻尿治療剤である、請求項10記載の医薬組成物。
- 請求項1記載の化合物又はその塩と賦形剤を含有する医薬組成物。
- 夜間頻尿治療用医薬組成物の製造のための、請求項1記載の化合物又はその塩の使用。
- 夜間頻尿治療のための、請求項1記載の化合物又はその塩の使用。
- 夜間頻尿治療のための請求項1記載の化合物又はその塩。
- 請求項1記載の化合物又はその塩の有効量を対象に投与することからなる、夜間頻尿治療方法。
