本申请要求于2014年09月03日提交中国专利局、申请号为201410445720.9、发明名称为“一种可以提高人体免疫力的电雾化水香烟烟液及包含该烟液的电雾化水香烟”的中国专利申请的优先权,其全部内容通过引用结合在本申请中。
技术领域
本发明属于电子烟技术领域,特别涉及一种电子烟烟液及包含该烟液的电子烟。
背景技术
烟草自16世纪中叶传入中国,距今已有400多年的历史。目前,我国约有3亿成年人是烟民,超过7亿人受二手烟的影响。众所周知,吸烟有害健康,会诱发多种疾病,如:肺癌、支气管炎、心肌梗塞、消化系统损伤、神经损伤等;而点燃香烟产生的烟雾中含有的焦油、亚硝胺、一氧化碳、放射性物质等,更对吸烟者本身及其周围的人们产生极大损害。
随着全球控烟运动的不断发展、《中国烟草控制规划(2012-2015》的出台、国务院公共场所禁烟的颁布等,吸烟越来越不得人心。但是,由于长期形成的吸烟文化、吸烟社会性功能、吸烟成瘾等原因,禁烟之路必定漫长。基于此类原因,各种模拟卷烟的产品相继出现,其中以电子烟(电雾化烟)最为盛行。
电子烟包括烟杆、雾化器、电池组件,其原理是通过加热将烟液雾化,直接进入吸烟者呼吸道;不燃烧、不产生烟灰、不形成二手烟。电子烟的口味,关键在于烟液的成分;为了尽可能地模拟普通香烟的香气、口味,目前市场上所售的电子烟烟液中不得不加入一定量的尼古丁;而尼古丁危害中枢神经系统、周边神经系统、副肾髓质等。现有技术中,通过向电子烟烟液中添加多种中药材,如:人参、黄芪、五味子、枸杞、淫羊藿、当归、白芷、鹿茸、桔梗、三七、曼陀罗、连翘、天冬、柴胡等,来试图改 善这一缺点,但是,这些电子烟的实用效果平平,并未引起关注。所以,还需要提供新的电子烟烟液,以实现在满足电子烟口感的情况下,提高电子烟的使用安全性。
发明内容
本发明的目的在于提供一种电子烟烟液及包含该烟液的电子烟。本发明提供的电子烟烟液制备而成的电子烟既能满足电子烟口感,又能够提高人体综合免疫能力,从而降低吸烟的危害,逐步达到戒烟的目的,还几亿烟民健康的体魄。
为了实现本发明的发明目的,本发明采用如下的技术方案:

在本发明中,当利用本发明提供的电子烟烟液与烟杆、雾化器、电池集成后,即获得电子烟,我们称之为“水香烟”。相应地,该电子烟烟液在本申请中也被称为“水香烟”烟液。
本发明提供了一种电子烟烟液及包含该烟液的电子烟。本发明提供的电子烟烟液包括皂苷类化合物、烟草香料和溶剂。实验结果证实,本发明提供的电子烟烟液制备而成的电子烟的雾化效果好、口感纯正、对咽喉刺激小;且使用该电子烟烟液制备而成的电子烟可以有益肺阴、清虚火、降血压、降血脂、提高抗氧化能力、消除疲劳、静心凝神、增强记忆力,能够提高人体免疫综合能力,从而降低吸烟的危害,逐步达到戒烟的目的。

本发明公开了一种电子烟烟液及包含该烟液的电子烟,本领域技术人员可以借鉴本文内容,适当改进工艺参数实现。特别需要指出的是,所有类似的替换和改动对本领域技术人员来说是显而易见的,它们都被视为包括在本发明。本发明的方法及应用已经通过较佳实施例进行了描述,相关人员明显能在不脱离本发明内容、精神和范围内对本文所述的方法和应用进行改动或适当变更与组合,来实现和应用本发明技术。
本发明提供的一种电子烟烟液及包含该烟液的电子烟中所用到的试剂和原料均可由市场购得。
下面结合实施例,进一步阐述本发明:
实施例1电子烟烟液的制备
(1)花旗参提取物的制备:将花旗参根部切成0.5~1.0cm的小段,与5倍体积的体积百分含量为75%的乙醇混合,之后于70℃条件下保温6小时,保温期间不断搅拌;静置、取上清;所得沉淀与5倍体积的体积百分含量为75%的乙醇混合,之后进行提取,如此重复3次。合并上清液,减压浓缩,热风吹干,得到花旗参提取物。
(2)花旗参提取物中皂苷类化合物的定性检测(定性检测方法):
采用薄层层析法。取人参皂苷Rb1、Rc、Rd、Re、Rg1、Rg3、Rh1、Rh2各10mg,分别溶于2mL甲醇,制成标准品液。用微量点样器分别吸取标准品液以及花旗参提取物液,点于薄层层析板上,用风筒吹干,然后置于盛有展开剂[V(氯仿):V(甲醇):V(水)=65:35:10(下层)]的层析缸中展 开,展开约距上边缘2~3mm时取出,挥干溶剂,喷10%H2SO4-乙醇溶液后,在105℃加热3min显色后观察。检测结果见图1,其中Rb1代表人参皂苷Rb1标准品,Rc代表人参皂苷Rc标准品,Re代表人参皂苷Re标准品、Rd代表人参皂苷Rd标准品、Rg1代表人参皂苷Rg1标准品、Rh1代表人参皂苷Rh1标准品、Rg3代表人参皂苷Rg3标准品、Rh2代表人参皂苷Rh2标准品;其中1代表实施例1制备获得的花旗参提取物、2代表实施例2制备获得的花旗参提取物、3代表实施例3制备获得的花旗参提取物、4代表实施例4制备获得的花旗参提取物、5代表实施例5制备获得的花旗参提取物,7代表实施例7制备获得的花旗参提取物,8代表实施例8制备获得的花旗参提取物。
根据样品点与标准品位置的比较可知(见图1),该花旗参提取物中皂苷类化合物主要为人参皂苷Rb1、Rc、Re、Rd、Rg1、Rh1
(3)花旗参提取物中皂苷类化合物的质量百分比的测定(定量检测方法):
a.标准曲线的绘制:精密称量人参皂苷Re对照品16.3mg,置于10mL容量瓶中,加入甲醇至刻度,摇匀,即得。分别精密量取人参皂苷Re标准品溶液10,20,40,60,80,100μL于具塞试管中,用甲醇定容到1mL,配成系列对照溶液。分别准确加入0.5mL 8%的香兰素乙醇溶液(用无水乙醇现配)、5mL 77%的硫酸溶液,混合均匀,置于60℃水浴上恒温10min,然后立即取出,放入冰水浴15min,取出放置至室温,显色溶液于544nm处测定吸光度,以吸光度(A)对皂苷类化合物浓度(C)作回归处理,得回归方程:C=0.2305A-0.0045,r=0.999。结果表明,人参皂苷Re在16.3~163μg范围内吸光度与浓度呈良好的线性关系。
b.取花旗参提取物0.5g,溶于5mL容量瓶中,取20μL溶液共3份,分别置于3个10mL磨口带塞试管中,按上述方法显色测定吸光度,由回归方程计算得到,该花旗参提取物中皂苷类化合物的平均质量百分比为7.34%。
(4)烟叶提取物的制备:将烟叶于40℃下烘干,冷却至室温,粉碎,与5倍体积的去离子水混合,15℃浸泡,不定期搅拌,48小时后抽滤,减压浓缩至1/2体积左右,得到烟叶提取物。
(5)电子烟烟液的制备:取所得花旗参提取物1份(相当于皂苷类化合物0.0734份,每份1g计)、所得烟叶提取物20份、丙三醇11.5份、丙二醇65份,混合均匀、充分溶解,即得电子烟烟液100mL,所得电子烟烟液中皂苷类化合物的含量为0.734g/L。本发明称之为“水香烟”烟液。
实施例2电子烟烟液的制备
(1)花旗参提取物的制备:将花旗参根部切成0.5~1.0cm的小段,与4倍体积的体积百分含量为80%的乙醇混合,之后于65℃条件下保温4小时,保温期间不断搅拌;静置、取上清;所得沉淀与4倍体积的体积百分含量为80%的乙醇混合,之后进行提取,如此重复3次。合并上清液,减压浓缩,热风吹干,得到花旗参提取物。按照实施例1提供的定性和定量检测方法进行定性和定量测定,得到:该花旗参提取物中皂苷类化合物的质量百分比为7.65%,主要成分为人参皂苷Rb1、Rc、Re、Rd、Rg1(见图1)。
(2)合成香料的制备:按照质量比为6:2:1:1:1:3:5:1:1;分别取香豆素、芸香苷、槲皮素、绿原酸、异戊酸、β-甲基戊酸、香叶醛、儿茶酚、茄尼醇,混合均匀,即得。
(3)电子烟烟液的制备:取所得花旗参提取物10份(相当于皂苷类化合物0.765份,每份1g计)、所得合成香料20份、去离子水5份、丙三醇11.5份、丙二醇80份,混合均匀、充分溶解,即得电子烟烟液100mL,所得电子烟烟液中皂苷类化合物的含量为7.65g/L。本发明称之为“水香烟”烟液。
实施例3电子烟烟液的制备
(1)花旗参提取物的制备:将花旗参根部切成0.5~1.0cm的小段,与6倍体积的体积百分含量为50%的乙醇混合,之后于75℃条件下保温 10小时,保温期间不断搅拌;静置、取上清;所得沉淀与6倍体积的体积百分含量为50%的乙醇混合,之后进行提取,如此重复3次。合并上清液,减压浓缩,热风吹干,得到花旗参提取物。按照实施例1提供的定性和定量检测方法进行定性和定量测定,得到:该花旗参提取物中皂苷类化合物的质量百分比为7.73%,主要成分为人参皂苷Rb1、Rc、Re、Rd、Rg1、Rh1(见图1)。
(2)烟叶提取物的来源:深圳市佩里沃生物科技有限公司。
(3)电子烟烟液的制备:取所得花旗参提取物5份(相当于皂苷类化合物0.387份,每份1g)、所得烟草提取物30份、去离子水5份、山梨醇6.7份、丙二醇55份,混合均匀、充分溶解,即得电子烟烟液100mL,所得电子烟烟液中皂苷类化合物的含量为3.87g/L。本发明称之为“水香烟”烟液。
实施例4电子烟烟液的制备
(1)花旗参提取物的制备:取花旗参茎叶,加5倍体积的蒸馏水,煎煮3次,每次3、3、2小时,分次过滤,合并滤液,静置24小时,取上清液,加氢氧化钠调节pH值至10,通过大孔树脂柱吸附,用水洗柱至中性后,用75%乙醇洗脱,洗脱液回收乙醇,浓缩至相对密度1.15(50℃热测)的浸膏,喷雾干燥,得到花旗参提取物。按照实施例1提供的定性和定量检测方法进行定性和定量测定,得到:该花旗参提取物中皂苷类化合物的质量百分比为81.02%,主要成分为人参皂苷Rb1、Rc、Re、Rd、Rg1、Rh1(见图1)。
(2)烟叶提取物的制备:将烟叶于30℃下烘干,冷却至室温,粉碎,与6倍体积的去离子水混合,30℃浸泡,不定期搅拌,40小时后抽滤,减压浓缩至1/2体积,得到烟叶提取物。
(3)电子烟烟液的制备:取所得花旗参提取物1份(相当于皂苷类化合物0.81份,每份1g)、所得烟叶提取物25份、去离子水5份、丙二醇60份、山梨醇6.7份,混合均匀、充分溶解,即得电子烟烟液100mL,所得电子烟烟液中皂苷类化合物的含量为8.1g/L。本发明称之为“水香烟”烟 液。
实施例5电子烟烟液的制备
(1)花旗参提取物的制备:取花旗参茎叶,加3倍体积的3%(W/V)硫酸铵溶液,煎煮3次,每次3、3、2小时,分次过滤,合并滤液,静置24小时,取上清液,加氢氧化钠调节pH值至12,通过大孔树脂柱吸附,用水洗柱至中性后,用80%乙醇洗脱,洗脱液回收乙醇,浓缩至相对密度1.15(50℃热测)的浸膏,喷雾干燥,得到花旗参提取物。按照实施例1提供的定性和定量检测方法进行定性和定量测定得到:该花旗参提取物中皂苷类化合物的质量百分比为91.33%,主要成分为人参皂苷Rb1、Rc、Re、Rd、Rg1、Rh1(见图1)。
(2)合成香料的制备:按照质量比为1:10:0.5:3:3:1:1:2:5;分别取香豆素、芸香苷、槲皮素、绿原酸、异戊酸、β-甲基戊酸、香叶醛、儿茶酚、茄尼醇,混合均匀,即得。
(3)电子烟烟液的制备:取所得花旗参提取物5份(相当于4.57份的皂苷类化合物,每份1g)、所得合成香料5份、去离子水15份、丙二醇70份、丙三醇11.5份,混合均匀、充分溶解,即得电子烟烟液100mL,所得电子烟烟液中皂苷类化合物的含量为45.7g/L。本发明称之为“水香烟”烟液。
实施例6电子烟烟液的制备
(1)花旗参提取物的制备:实施例5提供的制备方法制备获得,所得花旗参提取物中皂苷类化合物的质量百分比为91.33%,主要成分为人参皂苷Rb1、Rc、Re、Rd、Rg1、Rh1(见图1)。
(2)烟叶提取物的制备:将烟叶于50℃下烘干,冷却至室温,粉碎,与4倍体积的去离子水混合,25℃浸泡,不定期搅拌,55小时后抽滤,减压浓缩至1/2体积,得到烟叶提取物。
(3)电子烟烟液的制备:取所得花旗参提取物20份(相当于18.27份的皂苷类化合物,每份1g)、所得烟叶提取物20份、丙二醇65份、丙三醇11.5份,混合均匀、充分溶解,即得电子烟烟液100mL,所得电子烟烟液 中皂苷类化合物的含量为182.7g/L。本发明称之为“水香烟”烟液。
实施例7电子烟烟液的制备
(1)花旗参提取物的制备:取花旗参茎叶,加6倍体积的蒸馏水,煎煮3次,每次3、3、2小时,分次过滤,合并滤液,静置24小时,取上清液,加氢氧化钠调节pH值至11,通过大孔树脂柱吸附,用水洗柱至中性后,用78%乙醇洗脱,洗脱液回收乙醇,浓缩至相对密度1.15(50℃热测)的浸膏,喷雾干燥,得到花旗参提取物。按照实施例1提供的定性和定量检测方法测定得到:该花旗参提取物中皂苷类化合物的质量百分比为80.56%,主要成分为人参皂苷Rb1、Rc、Re、Rd、Rg1、Rh1(见图1)。
(2)烟草提取物的来源:广东华邦香料科技有限公司。
(3)电子烟烟液的制备:取所得花旗参提取物5份(相当于4.03份的皂苷类化合物,每份1g)、所得烟草提取物70份、去离子水5份、丙二醇15份、山梨醇6.7份,混合均匀、充分溶解,即得电子烟烟液100mL,所得电子烟烟液中皂苷类化合物的含量为40.03g/L。本发明称之为“水香烟”烟液。
实施例8电子烟烟液的制备
(1)花旗参提取物的制备:取花旗参根10g装于250mL锥形瓶中,加入20mL去离子水,封口,121℃高压蒸汽灭菌30min。冷却后于超净工作台中紫外灭菌30min后,接种黑曲霉(Aspergillus nige,来源于沂源康源生物科技有限公司)5mL,37℃恒温培养7d。将培养好的发酵液于121℃高压蒸汽灭菌30min,加入同等体积的水饱和正丁醇,超声萃取30min,过滤、离心(5000r/min,10min),取上清。将上清液减压浓缩,热风吹干,得到花旗参提取物。按照实施例1提供的定性检测和定量检测方法进行定性和定量测定得到:该花旗参提取物中皂苷类化合物的质量百分比为10.09%,主要成分为人参皂苷Rb1、Rc、Re、Rd、Rg1、Rg3、Rh1、Rh2(见图1)。
(2)合成香料的制备:按照质量比为10:1:0.1:1:1:1:5:5:5;分别 取香豆素、芸香苷、槲皮素、绿原酸、异戊酸、β-甲基戊酸、香叶醛、儿茶酚、茄尼醇,混合均匀,即得。
(3)电子烟烟液的制备:取所得花旗参提取物5份(相当于皂苷类化合物0.505份,每份1g)、所得合成香料5份、去离子水15份、丙二醇70份、丙三醇11.5份,混合均匀、充分溶解,即得电子烟烟液100mL,所得电子烟烟液中皂苷类化合物的含量为5.05g/L。本发明称之为“水香烟”烟液。
实施例9电子烟烟液的制备
(1)皂苷类化合物的制备:按照质量比为1:1:1,分别取人参皂苷Rb1、人参皂苷Rc、人参皂苷Rd(来源于成都汉禾生化制品有限公司),混合均匀,即得。
(2)合成香料的制备:按照质量比为1:5:0.5:2:2:2:4:4:4;分别取香豆素、芸香苷、槲皮素、绿原酸、异戊酸、β-甲基戊酸、香叶醛、儿茶酚、茄尼醇,混合均匀,即得。
(3)电子烟烟液的制备:取所得皂苷类化合物66份、所得合成香料1份、去离子水15份、丙二醇70份、山梨醇11.5份,混合均匀、充分溶解,即得电子烟烟液100mL,所得电子烟烟液中皂苷类化合物的含量为660g/L。本发明称之为“水香烟”烟液。
实施例10电子烟烟液的制备
(1)烟草提取物的制备:将烟叶于40℃下烘干,冷却至室温,粉碎,与4倍体积的去离子水混合,30℃浸泡,不定期搅拌,40小时后抽滤,减压浓缩至1/2体积,得到烟叶提取物。
(2)电子烟烟液的制备:取人参皂苷Re(来源于成都汉禾生化制品有限公司)1份(每份1g)、所得烟叶提取物25份、丙二醇65份、山梨醇6.7份,混合均匀、充分溶解,即得电子烟烟液100mL,所得电子烟烟液中皂苷类化合物的含量为10g/L。本发明称之为“水香烟”烟液。
实施例11电子烟烟液的质量评价
对照组电子烟烟液的获得:
(1)烟叶提取物的制备:将烟叶于40℃下烘干,冷却至室温,粉碎,与5倍体积的去离子水混合,15℃浸泡,不定期搅拌,48小时后抽滤,减压浓缩至1/2体积,得到烟叶提取物。
(2)电子烟烟液的制备:取所得烟叶提取物20份、丙三醇11.5份、丙二醇65份,混合均匀、充分溶解,即得。
分别取实施例1至实施例10、制备获得的电子烟烟液、对照组提供的电子烟烟液。
感官质量评价实验:
由本发明实施例1至实施例10制备获得的电子烟烟液制成的电子烟作为实验组,以不添加花旗参提取物的电子烟烟液制成的电子烟作为对照组。所得电子烟样品由国内具有国家或省级卷烟感官质量评吸资格证书的20位专业技术人员进行评吸,进而进行感官质量评价考核,感官质量评价标准见表1。评吸试验结果见表2。

根据实验结果可知,相比对照组,实验组的电子烟产品(即由本发明实施例1至实施例10制备获得的电子烟烟液制成的电子烟)的香气、余味得到了进一步的提升,而刺激性显著减小,差异显著(P<0.05),明显改善了电子烟对咽喉的刺激性。相比烟雾量,对照组与实验组实验结果相当,说明在电子烟烟液中添加花旗参提取物不影响雾化效果。
综上所述,本发明提供的电子烟烟液雾化效果好、口感纯正、显著减小了对咽喉的刺激性。
电子烟烟液对人体正常机能的影响:
取由本发明实施例1制备获得的电子烟烟液制成的电子烟,考察其对人体正常机能的影响。
在全国东北、华北、西南地区选取吸卷烟三年以上,且最近3个月平均每天吸0.5~2包卷烟的烟民150人,提供实施例1提供的制备方法制备获得的电子烟烟液制成的电子烟。使用方法相同:受试者每天1.5mL,连续使用3个月。分别在使用本发明提供的电子烟之前和使用3个月之后检测受试者的与心血管系统、血液系统、抗氧化能力相关的指标:血压、总胆固 醇、甘油三脂、低密度脂蛋白、高密度脂蛋白、丙二醛、超氧化物歧化酶、谷胱甘肽过氧化物酶、过氧化脂质。同时检测巨噬细胞数量与功能、进行体外淋巴细胞增殖试验以检验机体免疫力。
结果显示,经使用本发明提供的电子烟烟液后,94.67%受试者的血压(血压指标)有显著改善(P<0.05);90.67%受试者的血脂(总胆固醇、甘油三脂、低密度脂蛋白、高密度脂蛋白指标)有显著改善(P<0.05);92%受试者体内的抗氧化能力(丙二醛、超氧化物歧化酶、谷胱甘肽过氧化物酶、过氧化脂质指标)有显著提高(P<0.05)。受试者体内巨噬细胞数量与功能检测试验、体外淋巴细胞增殖试验结果显示,受试者的免疫力获得显著提高(P<0.05)。
取实施例2至实施例10制备获得的电子烟烟液制成的电子烟,按照相同的实验方法,考察本发明提供的电子烟烟液对人体正常机能的影响,获得了类似的实验结果,即经使用本发明实施例2至实施例10制备获得的电子烟烟液后,受试者的血压有显著改善、血脂得到显著改善、抗氧化能力显著提高,综合免疫力得到显著提高。
因此,利用本发明提供的电子烟烟液制备而得的电子烟长期使用,可降血压、降血脂、抗疲劳、延缓衰老,达到提高综合免疫力的功效。
以上对本发明所提供的电子烟烟液及包含该烟液的电子烟进行了详细介绍。本文应用了具体个例对本发明的原理及实施方式进行了阐述,以上实施例的说明只是用于帮助理解本发明的方法及其核心思想。应当指出,对于本技术领域技术人员来说,在不脱离本发明原理的前提下,还可以对本发明进行若干改进和修饰,这些改进和修饰也落入本发明权利要求的保护范围内。

Claims (9)

  1. 一种电子烟烟液,其特征在于,其包括皂苷类化合物、烟草香料和溶剂。
  2. 根据权利要求1所述的电子烟烟液,其特征在于,所述皂苷类化合物为人参皂苷Ra1、人参皂苷Ra2、人参皂苷Ra3、人参皂苷Rb1、人参皂苷Rb2、人参皂苷Rc、人参皂苷Rd、人参皂苷Re、人参皂苷Rf、人参皂苷Rg1、人参皂苷Rg2、人参皂苷Rg3、人参皂苷Rh1、人参皂苷Rh2、人参皂苷Rs1、人参皂苷Rs2、人参皂苷Rs3、人参皂苷F1、人参皂苷F2、人参皂苷F3、人参皂苷F5、人参皂苷R0、人参皂苷IH901、人参皂苷RAO或拟人参皂苷F11中的一种或两者以上混合物。
  3. 根据权利要求1或2所述的电子烟烟液,其特征在于,所述皂苷类化合物由花旗参提取物提供。
  4. 根据权利要求1至3中任一项所述的电子烟烟液,其特征在于,其中所述皂苷类化合物的含量为0.734g/L~660g/L。
  5. 根据权利要求1所述的电子烟烟液,其特征在于,所述烟草香料为烟草提取物和/或合成香料。
  6. 根据权利要求5所述的电子烟烟液,其特征在于,所述合成香料由香豆素、芸香苷、槲皮素、绿原酸、异戊酸、β-甲基戊酸、香叶醛、儿茶酚、茄尼醇组成。
  7. 根据权利要求1所述的电子烟烟液,其特征在于,所述溶剂为山梨醇、丙三醇、丙二醇、水中的一种或两者以上混合物。
  8. 根据权利要求1至7中任意一项所述的电子烟烟液,其特征在于,其包括0.0734~66重量份的所述皂苷类化合物、1~70重量份的所述烟草香料、26.7~96.5重量份的所述溶剂。
  9. 一种电子烟,其特征在于,其包括以下构件:烟杆、雾化器、电池组件;如权利要求1至8中任意一项所述的电子烟烟液放置于所述雾化器之中。
