Abstract
本発明は、シクロ-トランス-4-L-ヒドロキシプロリル-L-セリンを有効成分とする涙液分泌促進剤、及び涙液分泌減少に起因するドライアイの予防・治療薬に関する。
Description
本発明は、涙液分泌促進剤、及び涙液分泌減少に起因するドライアイの予防・治療薬に関する。
ドライアイは、様々な要因による涙液及び角結膜上皮の慢性疾患であり、眼不快感や視機能異常を伴う眼疾患である。日本におけるドライアイ患者は推定2000万人である。
ドライアイの治療薬は、これまで、ヒアルロン酸ナトリウム点眼や人工涙液の投与に限られていた。しかし、近年、ジクアホソルナトリウムを有効成分とする、ドライアイ治療薬が市販されるようになった。ジクアホソルナトリウムは、涙液分泌作用を有することが知られている。
一方、シクロ-トランス-4-L-ヒドロキシプロリル-L-セリンは、ヒドロキシプロリン誘導体であり、細胞増殖作用、細胞保護作用を有すること(特許文献1)、アレルギー性疾患及び炎症性疾患の予防・治療に有効であること(特許文献2及び3)が知られている。
本発明の目的は、新たな涙液分泌促進剤を提供することにある。本発明の目的は、また、涙液分泌減少に起因するドライアイの予防・治療薬を提供することにある。
本発明者らは、シクロ-トランス-4-L-ヒドロキシプロリル-L-セリン(以下、化合物1ともいう。)が、濃度依存的に涙液の分泌を促進することを見出した。化合物1の涙液分泌促進作用は、化合物1の既知の作用とは全く異なるものであり、かかる作用を有することは予想外の結果であった。
以上の知見から、本発明者らは本発明を完成するに至った。すなわち、本発明は、化合物1を有効成分とする涙液分泌促進剤を提供する。また、本発明は、化合物1を有効成分とする涙液分泌減少に起因するドライアイの予防・治療薬を提供する。
本発明は更に以下の(1)~(6)を提供する。
(1)有効量のシクロ-トランス-4-L-ヒドロキシプロリル-L-セリンを患者に投与する、涙液分泌の促進方法。
(2)有効量のシクロ-トランス-4-L-ヒドロキシプロリル-L-セリンを患者に投与する、涙液分泌減少に起因するドライアイの予防・治療方法。
(3)涙液分泌の促進に使用するための、シクロ-トランス-4-L-ヒドロキシプロリル-L-セリン。
(4)涙液分泌減少に起因するドライアイの予防・治療に使用するための、シクロ-トランス-4-L-ヒドロキシプロリル-L-セリン。
(5)涙液分泌促進剤を製造するための、シクロ-トランス-4-L-ヒドロキシプロリル-L-セリンの使用。
(6)涙液分泌減少に起因するドライアイの予防・治療薬を製造するための、シクロ-トランス-4-L-ヒドロキシプロリル-L-セリンの使用。
(1)有効量のシクロ-トランス-4-L-ヒドロキシプロリル-L-セリンを患者に投与する、涙液分泌の促進方法。
(2)有効量のシクロ-トランス-4-L-ヒドロキシプロリル-L-セリンを患者に投与する、涙液分泌減少に起因するドライアイの予防・治療方法。
(3)涙液分泌の促進に使用するための、シクロ-トランス-4-L-ヒドロキシプロリル-L-セリン。
(4)涙液分泌減少に起因するドライアイの予防・治療に使用するための、シクロ-トランス-4-L-ヒドロキシプロリル-L-セリン。
(5)涙液分泌促進剤を製造するための、シクロ-トランス-4-L-ヒドロキシプロリル-L-セリンの使用。
(6)涙液分泌減少に起因するドライアイの予防・治療薬を製造するための、シクロ-トランス-4-L-ヒドロキシプロリル-L-セリンの使用。
本発明によれば、新たな涙液分泌促進剤が提供される。また、涙液分泌減少に起因するドライアイの予防・治療薬が提供される。
以下、本発明の好適な実施形態について詳細に説明する。ただし、本発明はこれらの実施形態に限定されるものではない。
本発明は、化合物1を有効成分とする涙液分泌促進剤を提供する。かかる涙液分泌促進剤を投与することにより、涙液の分泌が促進される。そのため、本実施形態に係る涙腺分泌促進剤は、正常眼がドライアイに罹患する確率を下げることができ、更に、ドライアイに罹患した患者の症状を低減することができる。
化合物1の製造方法は特に限定されないが、例えば、特許文献1及び3に記載の方法で得ることができる。
本実施形態に係る化合物1を有効成分とする涙液分泌促進剤は、適宜の薬学的に許容される添加剤等の製薬上に必要な成分と混合して使うことができる。かかる添加剤は例えば、担体、賦形剤、pH調整剤及び希釈剤である。
上記の涙液分泌促進剤の形態は特に限定されず、点眼薬、内服薬、注射薬の形態の医薬製剤に調製されることが好ましい。当該製剤の化合物1の含有量は当業者が適宜に調整することができ、また、当該製剤の調製方法は特に限定されず、公知の方法を用いることができる。
かかる涙液分泌促進剤の効果的な投与量及び投与スケジュールは、投与方法、病状、患者の体重及び年齢等によって当業者が適宜に決定することができる。
化合物1は、涙液分泌促進作用を有することから、涙液分泌の減少に起因するドライアイの予防・治療薬に対して、特に効果的である。
以下、実施例を挙げて本発明をより具体的に説明するが、本発明はこれらの実施例に限定されるものではない。
(実施例1:涙液分泌に対する化合物1の濃度依存性)
化合物1による涙液分泌促進剤を確認するため、白色家兎(Slc:JW/CSK系統)を用いたin vivo実験を行った。涙液量はシルマー試験により、測定した。

測定に用いるサンプル点眼液は、終濃度100μM、1mM、10mMとなるように、化合物1を生理食塩水(0.9w/v%塩化ナトリウム水溶液)に溶解させたものを、それぞれ用いた。対照実験に用いる点眼液は、生理食塩水のみのものを用いた。
白色家兎の下眼瞼に、シルマー試験紙(昭和薬品化工株式会社製)の折り目5mm部分を挿入し、試験紙が濡れた長さを指標にして涙液量を測定した。
化合物1が100μM~10mMとなるように調製したサンプル点眼液又は生理食塩水を50μL白色家兎に点眼した。点眼してから12分経過後、ベノキシール点眼液(0.4w/v%、登録商標、参天製薬社製)を10μL点眼し、眼表面の局所麻酔を行った。局所麻酔してから3分経過後、白色家兎の下眼瞼にシルマー試験紙を挿入した。挿入した試験紙が1分間に濡れた長さ(シルマー試験値)を、涙液量として測定した。なお、サンプル点眼液又は生理食塩水を処置しない白色家兎に対しては、上述のベノキシール点眼液の点眼以降の工程のみを行った。測定は、各群についてn=8~10で行った。測定の結果を図1に示す。
図1に示されるように、白色家兎の涙液量(シルマー試験値)は、サンプル点眼液に含まれる化合物1の濃度依存的に上昇した。
(実施例2:化合物1による涙液分泌の経時的変化)
化合物1による涙液分泌の経時的変化は、化合物1が100μMとなるように調製したサンプル点眼液を用いて行った。対照実験には、生理食塩水のみの点眼薬を用いた。

化合物1が100μMとなるように調製したサンプル用点眼液又は生理食塩水を50μL白色家兎に点眼した。その後、ベノキシール点眼液を点眼するまでの時間を5分、15分、30分に変更したこと以外は、実施例1と同様にして涙液量を測定した。測定は、各群についてn=8~9で行った。測定の結果を図2に示す。
図2に示されるように、涙液量(シルマー試験値)は、化合物1を含有するサンプル点眼液を点眼してから顕著に増加し、少なくとも30分間は増加することが明らかになった。
以上の実施例の結果から、化合物1には、涙液の分泌を促進する作用があることが明らかになった。

- シクロ-トランス-4-L-ヒドロキシプロリル-L-セリンを有効成分とする、涙液分泌促進剤。
- シクロ-トランス-4-L-ヒドロキシプロリル-L-セリンを有効成分とする、涙液分泌減少に起因するドライアイの予防・治療薬。
