Abstract
Description
パーキンソン病の運動症状は中脳黒質のドパミンニューロンの変性脱落で生じるが、他にもコリン系、セロトニン系、ノルアドレナリン系など、いくつかの中枢神経系が系統的に変性脱落する。末梢自律神経系などにも変性を生じており、多彩な臨床症状の背景となっている。先ずは末梢に障害が始まり、延髄に及び、橋(RBD)、青斑核(うつの一部)などを巻き込みつつ上行し、中脳の黒質が障害された時点でパーキンソン病特有の運動障害を呈する。更に大脳に障害が及ぶと幻覚、妄想や認知機能障害を呈するようになる。約半数はこのような上行性進展様式をたどる。
一方、レストレスレッグス症候群(rest-less legs syndrome:RLS)は、「むずむず脚症候群」、「下肢静止不能症候群」とも呼ばれる。RLSはあまり知られておらず、一般診療では見逃されやすい疾患である。日本での有病率は2~5%、日常生活に支障をきたす患者は約200~300万人といわれている。欧米でも推定患者数は、人口の5~15%であるとされている。この病気は、40代以降に好発し、女性に多い(男性の1.2-1.4倍)と言われている。現在のところ、疾患の認知が進んでいないため、多くの患者が見逃されたり、不眠症や他の病気だと診断されたり、十分な治療を受けられないままになっている。特に、症状は夜間安静時、特に睡眠時間帯に増悪する。年単位で徐々に進行する慢性疾患であり、60代の睡眠-覚醒障害の35%がこの疾患であるとされている。このRLSの原因は明らかになっていないが、ドパミンの機能低下、中枢神経における鉄分不足による代謝異常、脊髄や末梢神経の異常、遺伝学的要素などが考えられている。
レストレスレッグス症候群治療に用いられる基本的薬剤は、軽症の症例の場合には、ベンゾジアゼピン系鎮静剤(クロナゼパム等)が主に使われている。中等症や重症の場合には、パーキンソン病治療薬として使われるドパミン受容体刺激薬(プラミペキソールとロピニロ一ル)、ドパミン製剤(レボドパ)などが有効とされている。
しかし、ドパミン受容体刺激薬は有効であるものの、投与初期に、はきけ、嘔吐、めまい、低血圧症、便秘、不眠症を起こすことが知られている。
これらの副作用を軽減させるためには、ドパミン受容体刺激薬の徐放性製剤化が一つの方法と考えられており、外用剤化の試みがなされてきた(特許文献1、2、3)。
現在、ドパミン受容体刺激薬のロチゴチンを経皮吸収剤化した製品が欧米で上市されている。しかしながら、副作用として、貼付部位にかぶれ等の炎症が、使用患者の約40~50%に起こるとされている。このような副作用を回避し、長期投与が可能な貼付剤は未だ見出されていない。そのため、経口投与で効果のあるドパミン受容体刺激薬(プラミペキソール)に関して、経口製剤ではなく、長期投与が可能な貼付剤が求められている状況であった。
テープ剤の基剤(膏体)は脂溶性のものであるため、薬効成分を溶解させるために水を使用すれば膏体に水がなじまないので、時間の経過と共にテープ剤中の水分が膏体から遊離することになってくる。従って、有機溶媒に難溶性のプラミペキソールをどのようにして溶解させるのかが大きな問題となっていた。
本発明者らは、炭素数14~18の脂肪酸のイオン液体を有機溶媒へのプラミペキソールの溶解剤として使用でき、2価アルコールと脂肪酸エステルを組合わせて使用することで、非水系のテープ剤が作製できることを見出した。更に、薬剤の膏体からの放出性を改善するために、充填剤を添加することで、経皮吸収性が顕著に向上することを見出した。
本発明者は、以上の知見に基づいて、本件発明を完成した。
(1)プラミペキソール及び/又はその薬学的に許容される塩と、炭素数14~18の脂肪酸系イオン液体、2価アルコールと脂肪酸エステルを含有することを特徴とする、テープ製剤。
(2)上記炭素数14~18の脂肪酸系イオン液体の脂肪酸が、イソステアリン酸、オレイン酸、パルミチン酸、ミリスチン酸、ステアリン酸の中から、一つ以上が選択されることを特徴とする、上記(1)記載のテープ製剤。
(3)上記炭素数14~18の脂肪酸系イオン液体の脂肪酸が、オレイン酸またはミリスチン酸であることを特徴とする、上記(1)記載のテープ製剤。
(4)上記炭素数14~18の脂肪酸系イオン液体の脂肪酸が、イソステアリン酸、オレイン酸の2種の混合物であることを特徴とする、上記(1)記載のテープ製剤。
(5)上記脂肪酸系イオン液体のアルカノールアミン化合物が、ジイソプロパノールアミン、トリエタノールアミン、トリイソプロパノールアミン、トロメタモールの中から一つ以上が選択されることを特徴とする、上記(1)~(3)のいずれかに記載のテープ製剤。

更に、本願発明では、有機系充填剤を添加することにより、膏体内部に薬剤の放出路を形成させることを行なった。その結果、膏体内部から薬剤の放出が良好になり、薬剤の皮膚透過量は大きく改善され、膏体中に残存する薬剤量は減少した。
これらの結果、結晶性のよいプラミペキソール塩酸塩を使用し、テープ剤中での結晶析出を回避した、安定で経皮吸収性の改善されたテープ剤を提供できるようになった。
本願発明の脂肪酸系イオン液体の含量は、薬効成分のプラミペキソールの使用量を1とすると、プラミペキソールの溶解性と経皮吸収性の点で、1.0倍量以上を使用することが好ましく、医薬品添加物事典における使用上限や製剤処方等を勘案すると、20倍量以下が好ましい。より好ましい脂肪酸系イオン液体の使用量は、薬効成分のプラミペキソールの使用量を1とすると、4倍量以上であり、16倍量以下である。
本発明の「膏体」とは、貼付剤の粘着剤層のことを言い、特に、膏体量として脂肪酸系イオン液体の含量等を対比する場合には、エラストマーと粘着付与剤の合計を表すものである。エラストマーとしては、公知のエラストマーが使用可能であるが、スチレン系熱可塑性エラストマー、特に、スチレン系ブロック共重合体が好ましく用いられる。具体的には、スチレン-ブタジエンブロック共重合体、スチレン-ブタジエン-スチレンブロック共重合体、スチレン-イソプレンブロック共重合体、スチレン-イソプレン-スチレンブロック共重合体(SIS)、スチレン-エチレン/ブチレンブロック共重合体、スチレン-エチレン/ブチレン-スチレンブロック共重合体、スチレン-エチレン/プロピレンブロック共重合体、スチレン-エチレン/プロピレン-スチレンブロック共重合体、スチレン-イソブチレンブロック共重合体、スチレン-イソブチレン-スチレンブロック共重合体などが挙げられる。これら、スチレン系ブロック共重合体は、1種のみ用いても、2種以上の組合せとして用いてもよい。より好ましいものとしては,SISを挙げることができる。また、粘着付与剤としては、通常貼付剤の分野で汎用される粘着付与剤が使用できる。例えば、ロジン系樹脂、テルペン樹脂、クマロン-インデン樹脂、石油系樹脂、テルペン-フェノール樹脂、脂環族飽和炭化水素樹脂等が挙げられる。好ましいものとしては、テルペン樹脂を挙げることができる。
なお、膏体(エラストマーと粘着付与剤)の添加量としては、含量が多くなると薬剤の放出性が悪くなり、含量が少ないと皮膚に対する粘着性が悪くなる。それ故、膏体(エラストマーと粘着付与剤)の含量としては、約10~50重量%が好ましい。より好ましくは、約20~45重量%を挙げることができる。
また、薬剤の放出性を良好にするためには、膏体の組成として、エラストマーを1重量部とすると、粘着付与剤が1/4~7/2重量部が好ましい。より好ましくは、粘着付与剤が1/3~5/2重量部であることが挙げられる。更にエラストマーの含量としては、8~25重量%であることが望ましい。
本発明の「脂肪酸エステル」とは、経皮吸収促進剤として使用される公知の脂肪酸エステルであればよく、例えばセバシン酸ジエチル、ミリスチン酸イソプロピル、アジピン酸ジイソプロピル、炭酸プロピレンを挙げることができる。好ましいものとしては、セバシン酸ジエチル、ミリスチン酸イソプロピル、炭酸プロピレンを挙げることができる。これらの脂肪酸エステルは、一種でも、また複数組み合わせても、使用することができる。好ましくは、セバシン酸ジエチル、ミリスチン酸イソプロピルの2種を組み合わせて使用することができる。
本発明の「テープ製剤」とは、水を必須な成分として含有しない、非水系のテープ製剤である。本発明の貼付剤の膏体には、汎用の基剤を使用することができ、例えばアクリル酸樹脂の基剤、あるいはSIS樹脂と粘着付与剤、軟化剤等を添加した基剤を使用することができる。好ましいものとしては、SIS樹脂をエラストマーとする基剤を挙げることができる。
上記剥離紙は粘着剤層の保護を目的として使用され、例えば、ポリエチレンコート上質紙、ポリオレフィンコートグラシン紙、ポリエチレンテレフタレート(以下PETという)フィルム、ポリプロピレンフィルムなどの片面をシリコン処理したものが使用可能である。
抗酸化剤としては、例えばBHT、没食子酸プロピル、アスコルビン酸ナトリウム等の有機系抗酸化剤、例えばチオ硫酸ナトリウム、亜硫酸水素ナトリウム、亜硫酸ナトリウム、ピロ亜硫酸ナトリウム等の無機系抗酸化剤を挙げることができる。
pH調節剤としては、例えばクエン酸、酢酸、酒石酸等の有機酸、例えばリン酸、塩酸等の無機酸を挙げることができる。
更に、カーボポール等の増粘剤、紫外線吸収剤等を添加することができる。
プラミペキソール2塩酸塩1水和物を有効成分として使用し、以下の組成比(重量%)で各試薬を秤取した。まず、脂肪酸イオン液体と有機溶媒の混合液に、プラミペキソール2塩酸塩1水和物を溶解させる。次に、SISや流動パラフィン、粘着付与剤等の膏体基剤をヘキサンに溶解した溶液に、プラミペキソール塩酸塩の溶解液を添加して均一になるよう撹拌する。この均一撹拌された粘稠溶液を塗工機に掛けてテープ剤を作製した。
作製されたテープ剤の経皮吸収性は、フランツセルを用いて8時間後のプラミペキソールの透過量で評価した。その結果を以下の表1に併せて示す。
また、使用するイオン液体の量が増えれば、プラミペキソールの皮膚透過量も増大することが分かった。
脂肪酸系イオン液体における脂肪酸の組成を固定し、アルカノールアミンの種類を変化させ、その効果を確認した。以下の表2(重量%)に示されるように試薬を秤取し、実施例1と同様にしてテープ剤を作製した。
作製されたテープ剤の経皮吸収性は、フランツセルを用いて8時間後のプラミペキソールの透過量で評価した。その結果を以下の表3に併せて示す。
アルカノールアミンとして、トリエタノールアミンとジイソプロパノールアミン(重量比1:1)の混合したものを使用して、上記組成のイオン液体を作製し、経皮吸収促進効果を評価した。その結果、塩基性の強いジイソプロパノールアミン(pH8.9)が、トリエタノールアミン(pH7.8)よりも、経皮吸収促進効果に支配的な影響を与えることが分かった。
脂肪酸系イオン液体は、有機溶媒と一緒になってプラミペキソールを溶解し、溶解したプラミペキソール溶液を膏体の中に分散させる界面活性剤的な役割を果たしている。即ち、プラミペキソールを含有するイオン液体のミセルになって、膏体中に分散していると考えられる。しかし、膏体中の流動性が悪く、物質移動が抑制されていることから、このミセルも膏体から放出され難い状態であると考えられる。その結果、貼付剤の中に薬剤が残留し易く、経皮吸収性が膏体からの放出律速になる傾向があった。
これまでの研究の結果、膏体内部の物質移動を促進させるため、充填剤(有機溶媒、イオン液体と膏体に不溶な粉末状の固体)を膏体に添加すると、充填剤同士の会合によって、空隙が形成され、この形成された空隙を薬剤の放出チャンネル(放出路)として使えることが分かっている。
そこで、本発明においても、膏体内部に分散しているイオン液体のミセルを充填剤の空隙を伝わって膏体表面に放出され易くすることを試みた。そのために、どのような充填剤が適切であるのかを検討した。即ち、有機系充填剤として結晶セルロース(セオラス)を使用し、無機系充填剤として無水ケイ酸を使用した。以下の表4の組成(重量%)になるように各試薬を秤量し、実施例1と同様にしてテープ剤を作製した。
作製されたテープ剤の経皮吸収性(皮膚透過量)は、フランツセルを用いて8時間後のプラミペキソールの透過量で評価した。その結果を以下の表4に併せて示す。
このように、有機系充填剤を使用して適切な空隙(流出路)が形成された結果、皮膚透過量は、充填剤無添加の場合と比較して、約1.5倍に増加することが示されている。即ち、有機系充填剤を使用すると、膏体からのプラミペキソールの放出性が、約1.5倍増加することになる。
更に、無機系充填剤である無水ケイ酸を使用した場合には、充填剤無添加の試験No.K300Rと同様の皮膚透過量しか示していない。このことは、無水ケイ酸の場合には、酸性が高いため、シリカゲルカラムでイオン複合体や塩がトラップされるように、イオン複合体を形成しているプラミペキソールが無水ケイ酸にトラップされやすくなっており、放出が阻害されているためと考えられる。
そこで、結晶セルロースと無水ケイ酸の重量比1:1の組成のテープ剤を作成して、プラミペキソールの皮膚透過量を評価した。その結果、試験No.K312Jに示されるように、No.K301SとK301Tの平均値に近い皮膚透過量を示した。このことから、充填剤の添加効果は、相加的な効果であることが分かった。
膏体(SISとテルペン樹脂)は流動性が悪いので、膏体中に分散したイオン液体に含有されたプラミペキソールの物質移動は困難である。それ故、膏体の量が増加し、流動パラフィンの含量が減少すると、膏体が硬くなって、更に膏体中の物質移動が低下する。即ち、膏体の量が増えれば、膏体からのプラミペキソールの放出が低下するので、プラミペキソールの皮膚透過量は、減少することになる。そこで、皮膚透過量に対する膏体量の影響を評価するため、以下の表5の組成(重量%)になるように、各試剤を秤取し、実施例1に基づいてテープ剤を作製した。
作製されたテープ剤の経皮吸収性(皮膚透過量)は、フランツセルを用いて8時間後のプラミペキソールの透過量で評価した。その結果を以下の表5に併せて示す。
薬剤の皮膚透過量(経皮吸収性)は、2つの要因(皮膚の吸収律速と膏体からの放出律速)で決まる。イオン液体や他の経皮吸収促進剤等の影響により、薬剤の皮膚吸収性が高ければ、薬剤の膏体からの放出性が律速になって、薬剤の皮膚透過量が支配される。一方、薬剤の皮膚吸収性が悪ければ、それが律速になって、薬剤の皮膚透過量が支配される。
実施例4で示したように、膏体(SISとテルペン樹脂)の含量が多い場合、膏体中の物質移動は困難であるので、膏体中に分散されているプラミペキソールは、膏体から放出され難くなっている。即ち、膏体量が多くなると、膏体からの放出律速が支配要因となるので、本発明のイオン液体の経皮吸収促進効果を評価できないことになる。
そこで、プラミペキソールに関して、膏体からのプラミペキソールの放出性が膏体の組成(エラストマーと粘着付与剤の組成)にどのような影響を受けるかを評価することを行った。そのため、以下の表6の組成(重量%)になるように、各試剤を秤取し、実施例1に基づいてテープ剤を作製した。
作製されたテープ剤の経皮吸収性(皮膚透過量)は、フランツセルを用いて8時間後のプラミペキソールの透過量で評価した。その結果を以下の表6に併せて示す。
更に、エラストマーであるSISの妥当な範囲を評価するため、実施例4の結果を参照すると以下の表7として整理することができた。
そこで、これまでの結果を検証するため、膏体中のエラストマーと粘着付与剤の合計量と皮膚透過量の相関を、以下の表8にまとめた。
以上のことから、膏体中のエラストマーと粘着付与剤の合計量は、30重量%以下が望ましく、エラストマーの含有量としては、8~15重量%であり、粘着付与剤の量はエラストマーの1/3~2/3の範囲であることが、薬剤の放出律速を回避するために望ましいことが示された。
実施例5で記載したように、薬剤の皮膚透過量(経皮吸収性)は、2つの要因(皮膚の吸収律速と膏体からの放出律速)で決まる。イオン液体と溶媒の含量が多い場合、膏体が軟化し、プラミペキソールが膏体から放出され易くなる。即ち、溶媒量が一定で、イオン液体量が多くなると、皮膚吸収律速が支配要因となる。逆に、イオン液体の量が少なくなると、プラミペキソールの放出律速が支配要因となる。そこで、イオン液体の添加量とプラミペキソールの皮膚透過量の相関性を評価し、皮膚吸収律速が支配要因になるイオン液体の好適な添加量の範囲を評価することを行った。そこで、以下の表9の組成(重量%)になるように、各試剤を秤取し、実施例1に基づいてテープ剤を作製した。
作製されたテープ剤の経皮吸収性(皮膚透過量)は、フランツセルを用いて8時間後のプラミペキソールの透過量で評価した。その結果を以下の表9に併せて示す。
上記実施例3~6では、プラミペキソールの膏体からの放出性を評価し、製剤処方の検討を行った。即ち、貼付剤における放出律速に留意した製剤処方の検討であった。そのため、貼付剤における皮膚吸収律速に留意した製剤処方の検討が充分ではなかった。その背景としては、これまでの評価指標である皮膚透過量を使用していては、皮膚吸収律速であるのか、放出律速であるのかが充分区別できないことが挙げられる。
そこで、本発明者らは、膏体組成を一定にして薬剤の放出性が関与する割合を一定にすると共に、更に、評価指標を変更し、皮膚透過量ではなく、単位時間当たりの皮膚透過速度(貼付後4時間目)を指標とすることを行った。これにより、皮膚での吸収性を的確に評価できるようになった。即ち、皮膚での吸収性を最適化するための製剤処方の検討が適切にできるようになったと考えられる。
そこで、実施例1で得られたデータから、プラミペキソールの皮膚透過速度を算出し、その結果を以下の表10と表11に併せて示す。
また、皮膚透過速度は、皮膚内の拡散速度の影響も受けることから考えると、上記表10と表11の結果は、イオン液体の皮膚吸収促進効果と共に、皮膚内の拡散促進効果を反映したものになっていると考えられる。
なお、実施例1の表1の結果と対比すると、概略として以下の表12になる。
しかし、イオン液体の単体使用では、経皮吸収促進効果が高くない、イソステアリン酸とパルミチン酸において、この混合イオン液体の場合には、皮膚透過量と皮膚透過速度が相加的ではなく、相乗的であることが示されている。
また、図1と図3によれば、貼付後4時間目の皮膚透過速度は、最大皮膚透過速度に到達する前のポイントであり、この測定時間は、膏体からの薬剤の放出性の影響が出難い時間帯であると考えられる。即ち、貼付後4時間目の皮膚透過速度が、イオン液体の皮膚吸収効果を主に反映していると考えられる。一方、貼付後8時間後の皮膚透過量は、イオン液体の皮膚吸収効果だけではなく、皮膚内の薬剤の拡散速度と膏体からの薬剤の放出速度も加味されたテープ剤の全体的な経皮吸収性を表わす指標になっていると考えられる。
このように、皮膚透過速度と皮膚透過量を評価指標として併用することにより、脂肪酸系イオン液体の経皮吸収促進効果が皮膚吸収律速を反映しているのか、膏体からの放出律速を反映しているのかを区別して評価できるようになった。その結果、使用目的の薬剤に関して、皮膚を透過させるために、より適切なイオン液体あるいはより適切な混合イオン液体を選択できるようになった。即ち、プラミペキソールについては、オレイン酸系イオン液体とミリスチン酸系イオン液体を基本的な経皮吸収促進剤として使用するか、あるいはこれらのイオン液体とイソステアリン酸系イオン液体等の混合イオン液体を使用することが望ましいことが分かった。
脂肪酸系イオン液体の経皮吸収促進効果を評価する指標として、実施例7では皮膚透過速度が指標になることを検証した。そこで、皮膚透過速度に対するアルカノールアミンの影響を評価するため、実施例2のデータから皮膚透過速度を算出し、表3の結果と共に、以下の表13に概要を示す。
なお、試験No.K304Xに対する試験No.K303Uの相対比は、皮膚透過速度では2.5倍になるが、皮膚透過量では2.1倍になる。即ち、脂肪酸系イオン液体の経皮吸収促進効果(皮膚の透過律速の場合)を評価するためには、皮膚透過速度で評価する方が、より感度良く、経皮吸収促進効果(皮膚の透過律速の場合)を評価できることが示された。
上記のように、皮膚透過速度と皮膚透過量が同じ倍率にならないことは、貼付後8時間目になれば、膏体の皮膚接触面の近傍部分の薬剤はほぼ放出されてしまっており、更に膏体内部の薬剤が放出されるためのタイムラグ(薬剤の膏体からの放出性)が徐々に影響してきているからであると考えられる。そこで、皮膚透過量についても貼付後4時間目で測定した数値で評価し直すと、以下の表14のようになる。
実施例3で示したように、充填剤として紛体を膏体に加えることにより、膏体中に分散する薬剤含有のイオン液体溶液の放出路が形成されるため、膏体から薬剤が放出され易くなると考えられる。即ち、薬剤の放出が良好になれば、薬剤の吸収は皮膚吸収律速になるので、イオン液体と溶媒の組成が同じであれば、皮膚透過速度は同一になる筈である。即ち、充填剤の影響を評価するためには、皮膚透過速度で評価する方が適切であろうと考えられた。そこで、実施例3の実験結果から皮膚透過速度を算出し、表3の結果と共に、その概略を以下の表15に示した。
実施例4~5で示したように、膏体の含量と組成によって、膏体から薬剤の放出され易さは異なっている。そこで、薬剤の膏体からの放出性を評価するため、膏体の含量と組成の影響を、皮膚透過速度で評価し直すことを行った。即ち、実施例4~5の結果から皮膚透過速度を算出し、実施例5の表8の結果と共に、その概略を以下の表16に示した。
以上のことから、実施例5の結論と同様に、膏体中のエラストマー(SIS)と粘着付与剤(テルペン樹脂)の合計量は、30重量%以下が望ましく、エラストマーの含有量としては、8~15重量%であり、粘着付与剤の量はエラストマーの1/3~1の範囲であることが、薬剤の放出性を改善するために望ましいことが示された。また、流動パラフィンの含量は約30~50重量%が望ましく、より好ましくは約40~50重量%であることが示されている。
実施例6では、イオン液体の添加量とプラミペキソールの皮膚透過量の相関性を評価し、薬剤の膏体からの放出性ではなく、薬剤の皮膚吸収性が支配要因になるイオン液体の好適な添加量の範囲を評価した。本実施例では、皮膚透過速度を評価基準とするため、実施例6の結果から皮膚透過速度を算出し、実施例6の表9の結果と併せて、その概要を以下の表17に示す。
薬剤の皮膚透過量は、試験例1で示すように、フランツ拡散セルのレセプター液に滲出してくる薬剤の量を、透過量として測定している。従って、フランツ拡散セルに貼付したテープ剤の薬剤含有量のどれだけの量がレセプター液の側に移行しているかを測定し、百分率で薬剤の皮膚透過率を表した。
実施例3の試験No.K300RとK301Tの貼付後の各時間における皮膚透過率(貼付剤の薬剤のどれだけがレセプター液に透過したかを%で表す)を、以下の表18と図6に示す。
充填剤を添加すれば、実施例3に示されているように、膏体からの放出性が約1.5倍増加する。このことは、プラミペキソールの皮膚透過率からもサポートされている。即ち、貼付後4~12時間の間では、充填剤が添加されていれば、皮膚透過率が約1.5倍の高くなっている。このことは、充填剤を添加すると膏体から薬剤の放出性が高くなり、皮膚上への薬剤供給が多いことを示している。
上記表18に示されるように、充填剤を添加することにより、テープ剤に含有された薬剤の40%近くが、皮膚透過することは、画期的なことであると考えられる。
図5に示されるように、0~2時間目の皮膚透過量と2~4時間目の皮膚透過量が大きく異なる理由として、薬剤の皮膚浸透のタムラグと、皮膚に一定量の薬剤が貯留され、オーバーフローするまで、レセプター液側に薬剤が放出されないことを考えられる。そこで、0~2時間目の皮膚透過量と2~4時間目の皮膚透過量の差を、皮膚に貯留した薬剤量であると仮定すると、皮膚の残存薬物量は、約5~9μg/cm2と推定される。そうであれば、薬剤の約3%近くが皮膚内に残留していることになる。
以上のことから、プラミペキソールの本発明のテープ剤においては、充填剤を含まない試験No.K300Rにおいては、約35%の薬剤が皮膚に移行したことになる。また、充填剤を添加したK301Tにおいては、約45%の薬剤が皮膚に移行したことになると想定される。
上記実施例1の各サンプルから約3cm2(プラミペキソールとして約210μg)を裁断し、試験温度32℃でフランツ拡散セル(透過面積:1cm2、レセプター液容量:8mL)を用いて以下のように経皮吸収性の評価試験を行なった。
(1)ラット皮膚: 5週齢のウィスターラット(雄)の市販の腹部摘出皮膚を使用。
(2)レセプター溶液:生理食塩水:エタノール(9:1)
(3)透過薬物の濃度測定:HPLC法(260~266nm)
上記ラット腹部皮膚(5週齢のウイスター系ラット)を縦型拡散セル(有効拡散面積:1cm2)に挟み、角質層側に表1~8に記載の約3cm2に裁断したサンプルを貼付し、また、真皮層側に生理的食塩水/エタノール(9:1)溶液を適用した。実験開始後2、4、6、8、12、20、22および24時間目にレセプター液を100μLサンプリングし、皮膚を透過して溶出した薬物濃度をHPLCにより測定し、各時間における薬物の累積透過量を測定した。また、各時点における薬物の皮膚透過速度については、各時点の薬物透過量と直前の時点の薬物透過量の差を経過時間で割ることで算出した。その結果を表1~18と図1~4に示す。これにより、各製剤サンプル間の経皮吸収性の優劣を評価することが出来た。
Claims (15)
- プラミペキソール及び/又はその薬学的に許容される塩と、炭素数14~18の脂肪酸系イオン液体、2価アルコールと脂肪酸エステルを含有することを特徴とする、テープ製剤。
- 上記炭素数14~18の脂肪酸系イオン液体の脂肪酸が、イソステアリン酸、オレイン酸、パルミチン酸、ミリスチン酸、ステアリン酸の中から、一つ以上が選択されることを特徴とする、請求項1に記載のテープ製剤。
- 上記炭素数14~18の脂肪酸系イオン液体の脂肪酸が、オレイン酸またはミリスチン酸であることを特徴とする、請求項1又は2に記載のテープ製剤。
- 上記炭素数14~18の脂肪酸系イオン液体の脂肪酸が、イソステアリン酸、オレイン酸の2種の混合物であることを特徴とする、請求項1~3のいずれかに記載のテープ製剤。
- 上記脂肪酸系イオン液体のアルカノールアミン化合物が、ジイソプロパノールアミン、トリエタノールアミン、トリイソプロパノールアミン、トロメタモールの中から一つ以上が選択されることを特徴とする、請求項1~4のいずれかに記載のテープ製剤。
- 上記脂肪酸系イオン液体のアルカノールアミン化合物が、ジイソプロパノールアミンであることを特徴とする、請求項1~5のいずれかに記載のテープ製剤。
- 上記2価アルコールが、プロピレングリコール、ポリエチレングリコール又は1,3-ブタンジオールの中から一つ以上が選択されることを特徴とする、請求項1~6のいずれかに記載のテープ製剤。
- 上記2価アルコールが、プロピレングリコールであることを特徴とする、請求項1~7のいずれかに記載のテープ製剤。
- 上記脂肪酸エステルが、セバシン酸ジエチル、ミリスチン酸イソプロピル、炭酸プロピレン、アジピン酸ジイソプロピルの中から一つ以上が選択されることを特徴とする、請求項1~8のいずれかに記載のテープ製剤。
- 上記脂肪酸エステルが、セバシン酸ジエチル及び/又はミリスチン酸イソプロピルであることを特徴とする、請求項1~9のいずれかに記載のテープ製剤。
- 充填剤が添加されていることを特徴とする、請求項1~10のいずれかに記載のテープ製剤。
- 充填剤が、有機系充填剤であることを特徴とする、請求項11に記載のテープ剤。
