該凹部の周縁に設けられ該凹部に収容されたバイオチップを該バイオチップの裏面が該凹部の底面から上方に離間した状態で支持する支持部と、を備えている、バイオチップ保持具が提供される。

前記支持部が、前記凹部の底部に形成されている。
また、凹部に該凹部の外部と連通する流路が設けられていてもよい。
前記凹部がプレート表面に複数形成されている。
前記バイオチップ保持具の少なくとも底面の一部が、シクロオレフィンコポリマーを含むフィルムによって構成されている。
上記のいずれかのバイオチップ保持具の凹部の底面に、シクロオレフィンコポリマーを含むフィルムを溶接するステップを備えている、

まず、バイオチップ保持具によって保持されるバイオチップの一例であるDNAチップ10の構成を説明するが、本発明ではDNAチップには限定されない。図1は、DNAチップ10の構成を示す模式的な斜視図である。本明細書において、バイオチップ保持具の凹部をウエルと称することがある。
その平面基板上に、所定の間隔をもって、所定の検出用試料を種類毎に固定したもの(スポッティング法等;Science 270, 467-470 (1995)等参照)、また、平面基板上の特定の位置で、所定の検出用試料を種類毎に逐次合成したもの(フォトリソグラフィー法等;Science 251, 767-773 (1991)等参照)も使用できる。
また、1つのウエルが、ウエルの外部に繋がるように、1または複数の流路を有していてもよい。複数のウエルを有するバイオチップ保持具の場合は、各ウエルどうしが流路により連結されている構成でもよい。
また、反応のため下面から加熱することもあるため、検出を行うまで、底面を耐熱性の保護フィルムで保護しておくことが好ましい。
より詳細には、ノルボルネンとエチレンをメタロセン触媒にて共重合したシクロオレフィンコポリマーとして知られているポリプラスチックス(株)製の「TOPAS」(商品名)や同様の性質を持つ日本ゼオン(株)の「ZEONEX」(商品名)を用いるのが好ましい。このような材料を使用することにより、ウエルプレートの上方から励起光を照射して蛍光を観察することができる。
一対の突起34、34のそれぞれは、押さえ具28上の反対位置に設けられ、押さえ具28がウエル24内に嵌合されたとき、ウエル24の内壁を押圧することにより、押さえ具28をウエル24から抜けにくくしている。
しかしながら、バイオチップの浮き上がりを防止できる形態であれば特に限定されず、押え具の材料を金属等の質量の重い材料にすることによって浮き上がりを防止してもよい。その際、当該押さえ具の形状は、枠状又はリング状であってもよいし、枠状又はリング状の一部が欠損したU字型、C字型又はL字型であってもよい。
まず、DNAチップ10を、検査対象のウエルプレート22の各ウエル24に導入し、ウエル24内の支持部26上にDNAチップ10を配置する(図4)。次いで、各ウエル24に押さえ具28を挿入し、DNAチップ10を支持部26と押さえ具28との間に挟持する。
ウエル間の配置がANSI/SBS規格に対応した(ウエル中心間隔9mm)スクエアタイプの96ウエルプレートを用意した。このウエルプレートには、各ウエルの底面の四隅に厚さ400μmの上記実施形態の支持部26と同様の支持部が形成されている。このウエルプレートは、プレート全体がシクロオレフィンコポリマー(ポリプラスチックス(株)製、商品名TOPAS)であり、底面から蛍光観察が可能とされている。
上記DNAチップを、ウエルプレートの各ウエル内に収納し、上記押え具で固定した。
各ウエルの底面の支持部が形成されていないスクエアタイプの96ウエルプレートを使用したことを除き、実施例1と同様にチップを収納して実験を行った。
