Abstract
본 발명의 조성물, 건강기능식품 또는 치료방법은 우수한 뇌신경세포 보호 효과를 가지는 동시에 감각운동기능 회복 촉진 효과 및 체중 증가 효과를 가진다. 따라서 본 발명은 뇌졸중 또는 퇴행성 뇌질환 예방 효과가 있고, 발병 직후부터 중장기적으로 뇌신경세포를 보호하고 뇌졸중 및 퇴행성 뇌질환으로 인해 감소된 감각운동기능의 회복을 촉진하고 감소된 체중을 증가시키는 효과를 갖는다. 따라서 효과적으로 뇌졸중 및 퇴행성 뇌질환을 예방, 치료, 관리하기 위해 이용될 수 있다.
Description
본 발명은 뇌졸중 또는 퇴행성 뇌질환의 예방 또는 치료용 약학적 조성물 및 건강기능식품에 관한 것이다.
뇌졸중은 뇌기능의 부분적 또는 전체적으로 급속히 발생한 장애가 상당 기간 이상 지속되는 것을 말하며, 뇌혈관이 막혀서 발생하는 뇌경색(허혈성 뇌졸중)과 뇌혈관의 파열로 인해 뇌 조직 내부로 혈액이 유출되어 발생하는 뇌출혈(출혈성 뇌졸중)이 있다. 미국심장뇌졸중학회 (American Heart Association/American Stroke Association, AHA/ASA) 통계에 따르면, 전 세계적으로 허혈성 뇌졸중은 전체 뇌졸중의 약 87%를 차지한다. 국내의 경우 2000년에는 64.7%, 2009년에는 76.1%로 증가하였다.(보건복지부지정 뇌졸중임상연구센터. 뇌졸중 진료지침(개정판). 2013. 허혈성 뇌졸중이 발병하면 뇌조직으로의 산소와 영양공급이 차단되어 신경세포가 사멸하고, 사멸된 신경세포에서 담당하던 기능의 장애 또는 손실이 발생하여 마비, 언어장애, 감각장애, 기억력 저하 등 증상이 나타난다. 뇌졸중 후 가장 흔하게 보고되는 증상은 근력 약화 및 손상된 측면의 팔다리 마비 등의 운동장애이다.
현재까지 허혈성 뇌졸중 치료제로 FDA 승인을 받은 약물은 혈전용해제인 조직 플라스미노겐 활성제 (tissue plasminogen activator, t-PA)가 유일하다. 하지만 뇌졸중 발생 3시간 이내에 투여해야 하는 제한이 있으며, 대부분의 환자들은 도착 시간 지연, 뇌출혈 증가 위험성 등으로 인해 소수의 환자만 투여 받는 실정이다.
퇴행성 뇌질환은 뇌의 신경세포가 감소하며 인지기능이나 감각운동기능이 저하되는 질환을 통틀어 일컫는 말이다. 퇴행성 뇌질환에는 치매, 알츠하이머병, 파킨슨병, 헌팅턴 무도병, 피크(Pick)병 및 크로이펠츠-야콥(Creutzfeld-Jacob)병 등이 있다. 상기 질환들은 각기 다양한 원인과 기전에 의해 발생하나 공통적으로 뇌의 신경세포가 감소하고 인지기능, 감각운동기능 저하를 보인다.
본 발명의 목적은 뇌신경세포 보호 효과를 갖는 동시에 감각운동기능 회복 촉진 및 체중을 증가시키는 효과를 갖는 뇌졸중 또는 퇴행성 뇌질환의 예방 또는 치료용 약학적 조성물 및 건강기능식품을 제공하는 것이다.
본 발명은 갈근 및 황금 복합추출물을 유효성분으로 포함하는 뇌졸중 또는 퇴행성 뇌질환의 예방 또는 치료용 약학적 조성물을 제공한다.
갈근(葛根)은 콩과(Leguminosae)에 속하는 다년생 등목인 칡(Pueraria lobata)의 수피를 제거한 뿌리이다.
황금은 꿀풀과(Labiatae)에 속하는 다년생 초본인 황금(Scutellaria baicalensis)의 수피를 제거한 뿌리이다.
본 발명의 일 구현예로서, 상기 갈근 및 황금 복합추출물은 갈근 및 황금이 100:1 내지 1:100의 중량비로 포함된 혼합물로부터 추출된 것일 수 있다. 바람직하게는 상기 갈근 및 황금 복합추출물은 갈근 65 내지 95 중량% 및 황금 5 내지 35 중량%를 포함하는 혼합물로부터 추출된 것일 수 있고, 더욱 바람직하게는 상기 갈근 및 황금 복합추출물은 갈근 87.91 중량 % 및 황금 12.09 중량%를 포함하는 혼합물로부터 추출된 것일 수 있다. 상기 조성 비율은 본 발명자들이 우수한 뇌신경세포 보호 효과를 발휘하면서 체중 증가 및 감각운동기능 회복 촉진 효과를 나타내는 조성 비율에 대해 연구한 결과 발견한 조성 비율에 해당한다.
상기 뇌졸중은 허혈성 뇌졸중, 출혈성 뇌졸중을 모두 포함하며, 바람직하게는 허혈성 뇌졸중을 의미한다.
상기 퇴행성 뇌질환은 치매, 알츠하이머병, 파킨슨병, 헌팅턴 무도병, 피크(Pick)병 및 크로이펠츠-야콥(Creutzfeld-Jacob)병으로 이루어진 군으로부터 선택된 어느 하나일 수 있다. 그러나 퇴행성 뇌질환의 종류는 이에 해당되지 않으며, 본 발명의 조성물로 혜택을 볼 수 있는 질병은 모두 포함된다.
본 발명은 갈근 및 황금 복합추출물을 유효성분으로 포함하는 뇌신경세포보호 및 감각운동기능(sensorimotor function) 회복 촉진용 조성물을 제공한다.
본 발명은 갈근 및 황금 복합추출물을 유효성분으로 포함하는 뇌신경세포보호, 감각운동기능 회복 촉진, 및 체중 증가용 조성물을 제공한다. 뇌졸중 및 퇴행성 뇌질환 환자는 체중이 감소하는 경우가 많다. 예를 들어 뇌졸중 환자는 삼킴장애로 인하여 충분한 경구 영양공급이 이루어지지 못하는 경우가 많아 체중이 급격히 감소하는 경향이 있는데, 뇌졸중 후 체중 감소는 환자의 재활에 악영향을 미쳐 재활을 지연시키는 것으로 보고된 바 있다. 본 발명의 갈근 및 황금 복합추출물은 뇌졸중 후 감소한 체중을 증가시키는 효과가 있으므로, 뇌졸중 및 퇴행성 뇌질환에서 환자의 재활을 도울 수 있다.
본 발명의 조성물은 뇌신경세포 보호 효과를 가져 뇌졸중 및 퇴행성 뇌질환을 예방하거나 발병 직후부터 이에 대한 치료 효과를 가질 수 있고, 중장기적으로 뇌졸중 및 퇴행성 뇌질환으로 인해 감소된 감각운동기능의 회복을 촉진하고 감소된 체중을 증가시키는 효과를 갖는다. 따라서 효과적으로 뇌졸중 및 퇴행성 뇌질환을 예방, 치료, 관리하기 위해 이용될 수 있다.
본 발명의 약학적 조성물은 갈근 및 황금 복합추출물을 유효성분으로 포함하며, 유효성분의 함량은 사용 목적(예방, 또는 치료적 처치)에 따라 적합하게 결정될 수 있다. 일반적으로, 갈근 및 황금 복합추출물은 조성물의 총중량에 대하여 0.0001 내지 100 중량%로 첨가할 수 있다.
본 발명의 조성물은 경구 또는 비경구로 투여될 수 있으며, 바람직하게는 비경구, 더욱 바람직하게는 정맥내로 투여될 수 있다. 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제 및 계면활성제 등의 희석제 또는 부형제를 사용하여 조제될 수 있다.
비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조제제 및 좌제가 포함된다. 비수성용제와 현탁용제로는 프로필렌글리콜, 폴리에틸렌글리콜, 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈 61, 카카오지, 라우린지, 글리세롤 및 젤라틴 등이 사용될 수 있다. 본 발명의 약학적 조성물은 비경구 투여시 피하주사, 정맥주사 또는 근육내 주사를 통하여 투여될 수 있다.
본 발명의 약학적 조성물의 투여량은 환자의 상태 및 체중, 질병의 정도, 약물형태, 투여경로 및 기간에 따라 다르며, 당업자에 의해 적절하게 선택될 수 있다. 그러나 바람직한 효과를 위해서 본 발명의 조성물은 1일 0.0001 내지 2000 mg/kg, 바람직하게는 0.1 내지 1000 mg/kg의 용량으로 투여할 수 있다. 투여는 하루에 한 번 투여할 수도 있고, 수회 나누어 투여할 수도 있다. 다만, 상기 투여량의 본 발명의 범위를 한정하는 것은 아니다.
본 발명의 조성물은 뇌졸중 또는 퇴행성 뇌질환 예방 및 치료를 위하여 단독으로, 또는 수술, 호르몬 치료, 화학 치료 및 생물학적 반응 조절제를 사용하는 방법들과 병용하여 사용할 수 있다.
본 발명은 갈근 및 황금 복합추출물을 유효성분으로 포함하는 뇌졸중 또는 퇴행성 뇌질환의 예방 또는 개선용 건강기능식품을 제공한다.

본 발명은 갈근 및 황금 복합추출물을 유효성분으로 포함하는 뇌신경세포보호, 감각운동기능 회복 촉진, 및 체중 증가용 건강기능식품을 제공한다.
본 발명의 상기 갈근 및 황금 혼합 추출물은 하기의 단계들을 포함하는 제조방법에 의해 제조되는 것이 바람직하나 이에 한정되지 않는다:
(1) 갈근 및 황금에 혼입된 이물질을 제거하고 세척, 분쇄하는 단계;
(2) 세척한 갈근 및 황금 분쇄물의 혼합물에 추출용매를 가하여 추출물을 추출하는 단계;
(3) 추출물을 감압농축하는 단계;
(4) 농축액을 여과하는 단계; 및
(5) 여과한 추출물을 건조하는 단계.
단계 (1)의 세척은 1 ~ 5회 반복하는 것이 바람직하다.
단계 (2)의 추출은 진탕 추출, 속실렛(Soxhlet) 추출 또는 환류 추출 방법을 이용할 수 있으나 이에 한정되지 않는다.
추출용매로는 물, C1~C4 알코올 또는 이들의 혼합물을 사용할 수 있으며, 에탄올을 이용하여 열수 추출하는 것이 가장 바람직하다. 상기 추출용매로는 준비된 갈근 및 황금 전체 중량의 3 내지 10배의 추출용매를 가하여 추출하는 것이 바람직하다.
상기 열수 추출은 압력은 가압하여 추출할 수 있고, 1.1 ~ 2 기압이 바람직하고, 1.2 ~ 1.5 기압이 더욱 바람직하다.
또한, 열수 추출시 온도는 60 ~ 100 ℃인 것이 바람직하며, 80 ~ 90 ℃인 것이 더욱 바람직하다. 또한, 추출시간은 2 내지 24시간인 것이 바람직하며, 추출회수는 1 내지 5회인 것이 바람직하다.
단계 (3)의 감압농축은 진공감압농축기 또는 진공회전증발기를 이용하는 것이 바람직하다. 단계 (3)에서 추출액을 고형분 40 % 이상이 되도록 농축하는 것이 바람직하다.
단계 (5)의 건조는 감압건조, 진공건조, 비등건조, 분무건조 또는 동결건조 방법을 이용할 수 있으나, 이에 한정되지 않는다.
본 발명의 건강기능식품은 갈근 및 황금 복합추출물을 유효성분으로 포함하며, 유효성분의 함량은 사용 목적(예방, 개선 또는 치료적 처치)에 따라 적합하게 결정될 수 있다. 일반적으로, 갈근 및 황금 복합추출물은 건강기능식품의 총중량에 대하여 0.0001 내지 100 중량%로 첨가할 수 있다.
또한, 상기 식품의 종류에는 특별한 제한이 없다. 본 발명이 적용될 수 있는 식품의 예로는 음료, 껌, 비타민 복합제, 드링크제 등이 있으며, 통상적인 의미에서의 건강기능식품을 모두 포함한다.
본 발명의 건강기능식품을 음료 형태로 제조하는 경우, 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상기 천연 탄수화물의 예로는 모노사카라이드, 예를 들어, 포도당, 과당 등; 디사카라이드, 예를 들어 말토스, 수크로스 등; 및 폴리사카라이드, 예를 들어 덱스트린, 시클로덱스트린 등과 같은 통상적인 당, 및 자일리톨, 소르비톨, 에리트릴톨 등의 당알콜이다. 또한, 상술한 것 이외의 향미제(사카린, 아스파르탐 등)을 사용할 수 있다.
본 발명의 건강기능식품은 여러 가지 영양제, 비타민, 광물(전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제 및 증진제(치즈, 초콜릿 등), 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산음료에 사용되는 탄산화제 등을 함유할 수 있다.
본 발명의 건강기능식품은 뇌졸중 또는 퇴행성 뇌질환 예방 및 치료를 위하여 단독으로, 또는 수술, 호르몬 치료, 화학 치료 및 생물학적 반응 조절제를 사용하는 방법들과 병용하여 사용할 수 있다.
본 발명은 치료가 필요한 대상체에 치료학적으로 유효한 양의 갈근 및 황금 복합추출물을 투여하여 뇌졸중 또는 퇴행성 뇌질환을 예방, 개선 또는 치료하는 방법을 제공한다.

본 발명의 약학적 조성물, 건강기능식품, 치료방법 및 약제학적 제제를 제조하는 데 사용하는 용도는 뇌허혈에 대한 뇌신경세포 보호 효과를 갖는 동시에 감각운동기능 회복 촉진 효과 및 체중 증가 효과도 뛰어나다. 즉 뇌허혈에 의한 뇌졸중 치료에 있어서, 시간적 시기(time window)상 발병 후 수일간 일어나는 지연성 신경세포사를 막는 뇌신경세포 보호 효과를 갖는 동시에, 급성기 이후에 감각운동기능을 회복시키고 감소된 체중을 증가시키는 효과도 갖는 것이다. 감각운동기능 회복 촉진 및 체중 증가 효과는 뇌신경세포 보호 효과와는 다른 기전으로 일어나는 것으로서, 이러한 효과들을 동시에 갖는 약물은 지금까지 개발된 바 없다.
본 발명의 약학적 조성물, 건강기능식품, 치료방법 및 약제학적 제제를 제조하는 데 사용하는 용도의 효과는 중대뇌동맥 폐쇄 랫드 모델에서 확인되었다. 지금까지 가장 널리 알려진 뇌허혈에 의한 뇌신경세포사 기전으로는 2가지가 있다. 하나는 뇌허혈에 의해서 세포 바깥에 과도한 글루타메이트(glutamate)가 축적되게 되며, 이러한 글루타메이트가 세포 내로 유입되어 결국 과도한 세포 내 칼슘의 축적으로 뇌신경세포사가 유발된다는 흥분성 뇌신경세포사 기전(Kang TC, et al., J. Neurocytol., 30: 945-955, 2001)이고, 다른 하나는 허혈-재관류시에 갑작스러운 산소 공급으로 인한 생체 내 라디칼의 증가로 인해 DNA 및 세포질에 손상을 입어 유발된다는 산화성 뇌신경세포사이다(Won MH 외, Brain Res., 836: 70-78, 1999; Sub AY., Chen YM., J. Biomed. Sci., 5: 401-414, 1998; Flowers F, Zimmerman JJ. New Horiz. 6: 169-180, 1998). 이 동물 모델은 산화성 뇌신경세포사 및 흥분성 뇌신경세포사가 모두 일어나 뇌허혈에 의한 뇌신경세포사의 예방 또는 치료 효과를 확인하기에 적합한 모델에 해당한다.
본 발명의 조성물, 건강기능식품 또는 치료방법은 우수한 뇌신경세포 보호 효과를 가지는 동시에 감각운동기능 회복 촉진 효과 및 체중 증가 효과를 가진다. 따라서 본 발명은 뇌졸중 또는 퇴행성 뇌질환 예방 효과가 있고, 발병 직후부터 중장기적으로 뇌신경세포를 보호하고 뇌졸중 및 퇴행성 뇌질환으로 인해 감소된 감각운동기능의 회복을 촉진하고 감소된 체중을 증가시키는 효과를 갖는다. 따라서 효과적으로 뇌졸중 및 퇴행성 뇌질환을 예방, 치료, 관리하기 위해 이용될 수 있다.

도 3은 동물 모델에서 샴군, 대조군, 갈근 및 황금 복합추출물 30 mg/kg 투여군, 갈근 및 황금 복합추출물 100 mg/kg 투여군, 갈근 및 황금 복합추출물 300 mg/kg 투여군의 신경학적 점수(neurological score)를 나타낸 도이다.
도 4는 동물 모델에서 샴군, 대조군, 갈근 및 황금 복합추출물 30 mg/kg 투여군, 갈근 및 황금 복합추출물 100 mg/kg 투여군의 체중 변화를 나타낸 도이다.
본 발명자들은 다양한 한약재 추출물들의 뇌졸중 또는 퇴행성 뇌질환 치료 효과를 관찰한 결과, 갈근 및 황금 복합추출물을 유효성분으로 포함하는 조성물이 동물 모델(MCAo)에서 뇌허혈에 의한 뇌신경세포 손상에 대하여 뇌신경세포 보호 효과가 우수하고, 감각운동기능 회복 촉진 효과 및 체중 증가 효과가 뛰어남을 확인하고 본 발명을 완성하였다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
<실시예 1> 갈근 및 황금 복합추출물의 제조
갈근과 황금은 약수당(서울 동대문구)에서 구입하였다. 각각 분쇄한 갈근 87.91 g 및 황금 12.09 g을 혼합하여 혼합물을 준비하였다. 상기 혼합물에 10배량의 30% 에탄올 1L을 넣고 90 ℃에서 3 시간 동안 1차 열수 추출하여 실온에서 약 2-3시간 방치한 다음 여과한 액을 1차 추출액으로 하였다. 다시 추출기에 30% 에탄올 1L을 넣고 80~90 ℃에서 3 시간 동안 2차 열수 추출하여 실온에서 약 2-3시간 방치한 다음 여과한 액을 2차 추출액으로 하였다. 1차 추출액과 2차 추출액을 혼합하여 50-60 ℃에서 약 20 브릭스(brix)가 될 때까지 감압농축 하였다. 얻어진 농축액을 예비동결기로 -45℃에서 20분간 예비 동결한 후, 동결건조기로 동결건조하여 분말상태로 제조하였다(수득율 약 25%).
<비교예 1> 갈근 추출물의 제조
갈근 100 g 을 실시예 1과 동일한 방법으로 추출하여 갈근 추출물을 제조하였다.
<비교예 2> 황금 추출물의 제조
황금 100 g 을 실시예 1과 동일한 방법으로 추출하여 황금 추출물을 제조하였다.
상기 실시예, 비교예 1 및 2의 추출물에 포함된 갈근 추출물 및 황금 추출물의 함량을 하기 표 1에 정리하였다.

<실험예 1> 동물 모델 제작
<1-1> 실험동물의 준비
실험동물로 300 g 내외의 8주령의 스프라그-다울리계(sprague-dawley, 샘타코, 대한민국) 수컷 랫드를 구입하여 사료와 물을 충분히 공급하면서 실험환경에 적응하도록 하였다. 1주일 정도의 적응기간을 준 후, 동물실험을 수행하였다.
<1-2> 동물 모델 제작
Zea Longa 등(Longa EZ, Weinstein PR, Carlson S, Cummins R. Reversible middle cerebral artery occlusion without craniectomy in rats. Stroke; a journal of cerebral circulation. 1989; 20(1): 84-91.)의 방법을 응용하여 국소뇌허혈을 유발하였다.
실험예 <1-1>에서 준비한 랫드를 70% N2O와 30% O2가 섞인 혼합가스에 5 %의 이소플루란(isoflurane)으로 전신마취를 한 후, 목 전방 부위 중앙의 피부를 절개하고 왼쪽 경동맥과 외경동맥(ECA)을 주변 조직과 신경들로부터 분리하였다. 외경동맥 분지인 상부갑상선동맥과 후두동맥, 내경동맥의 분지인 익돌근구개동맥을 전기소작하였다. 외경동맥을 자르고 프로브(Doccol, Redlands, CA, USA)를 외경동맥에서 내경동맥으로 삽입하되 총경동맥분지에서 약 18~19 ㎜ 정도 삽입한 후 실로 고정하였다. 피부절개부위를 다시 봉합한 후 마취에서 자연회복시켰다. 유발 60분 후 상기와 같은 방법으로 재마취한 후 프로브를 후퇴시켜 재관류(reperfusion)시켰다. 모든 수술과정은 이소플루란을 2 %로 유지하면서 수술현미경하에서 시행하였으며, 체온이 37± 0.5 ℃ 이하로 떨어지지 않도록 체온 유지장치를 이용하였다.
<실시예 2> 동물 모델에서 비교예 1, 2와 실시예 1의 신경보호 효과 비교
<실험예 1>에서 언급한 방법으로 랫드에 중대뇌동맥 폐쇄를 90분 동안 유발하였다. 랫드를 4개 군으로 나누어 제1군에는 비히클(5% DMSO), 제2군에는 비교예 1의 갈근 추출물, 제3군에는 비교예 2의 황금 추출물, 제4군에는 실시예 1의 갈근 및 황금 복합추출물을 각각 100 mg/kg 용량으로 유발 직후 1회 경구투여 하였다. 재관류 24시간 후에 랫드를 희생시키고 2분 이내에 뇌를 적출한 후 랫드 뇌 매트릭스(rat brain matrix)를 이용하여 2 ㎜ 두께로 6개의 절편으로 나누었다. 2 % 2,3,5-트리페닐테트라졸리움 클로라이드(2,3,5-triphenyltetrazolium chloride, TTC) 용액이 들어 있는 12 웰 플레이트에 조직을 충분히 담그고 37℃에서 30분 동안 염색하였다. 염색된 뇌 조직은 디지털 카메라로 촬영한 후 이미지 분석프로그램인 이미지 프로 플러스 5.0(image Pro plus 5.0)을 이용하여 뇌경색 부피를 측정하였다. 뇌경색 부피(Infarct volume) (%)은 아래의 수학식으로 계산하였다.
뇌경색 부피 (%) = (A-B/A)×100
A: 정상 좌반구의 부피(㎣)
B: 손상된 뇌경색의 부피 (㎣)
갈근 및 황금 복합추출물 투여군은 뇌경색 부피가 대조군 대비 47.9 % 유의하게 감소하였다(p<0.05). 반면 갈근 추출물 투여군 및 황금 추출물 투여군은 대조군 대비 통계적으로 유의한 효과가 나타나지 않았다(도 1). 상기 실험 결과는 하기 표 2에 정리하였다.

<실시예 3> 동물 모델에서 감각운동기능 저하에 대한 복합추출물의 기능회복 촉진효과 확인
랫드에 <실험예 1>에서 언급한 방법으로 중대뇌동맥 폐쇄를 60분 동안 유발하였다. 랫드를 샴(Sham)군(가장수술군), 대조군, 실시예 1의 갈근 및 황금 복합추출물 30 mg/kg 투여군, 실시예 1의 갈근 및 황금 복합추출물 100 mg/kg 투여군 4개 군으로 나누고, 수술 3일 후부터 1일 1회 14일간 복합추출물을 kg 당 5 ml의 부피로, 또는 샴군과 대조군의 경우 비히클 (5% DMSO)을 kg 당 5 ml 부피로 경구투여하였다.
랫드의 운동협응(motor coordination) 및 균형조절(balance alteration)에 대한 능력을 평가하기 위하여 Jones BJ 등의 방법(Jones BJ, Roberts DJ. The quantiative measurement of motor inco-ordination in naive mice using an acelerating rotarod. The Journal of pharmacy and pharmacology. 1968; 20(4): 302-4.)을 응용하여 수술 하루 전날, 수술 1, 2, 3, 4주 후에 로타로드 시험(rotarod test)을 수행하였다. 로타로드(B1001, B.S Technolab INC., 한국)의 회전봉이 5에서 30 rpm까지 5분간 회전하는 가속회전법을 사용하였고, 뇌허혈이 유발된 랫드를 봉의 가운데에 균형을 잡아 놓은 후, 회전을 시작한 시간부터 랫드가 떨어질 때까지의 시간(유지시간, latency time)을 측정하였다. 각 랫드마다 5회 실시하여 최고치와 최저치를 제외한 중간 3개 값의 평균을 수치로 사용하였다.
감각운동기능 저하에 대한 복합추출물의 개선 효과를 확인하기 위하여 로타로드 시험을 수술 하루 전날(수술 전), 수술 1, 2, 3, 4주 후 수행한 결과는 도 2와 같다. 수술 전날 모든 군의 유지시간을 측정한 결과 샴군, 대조군, 갈근 및 황금 복합추출물 30 mg/kg 투여군, 갈근 및 황금 복합추출물 100 mg/kg 투여군은 각각 296.0±2.6, 283.9±9.7, 278.9±11.7, 284.4±6.6 초의 유지시간을 보여 수술 전 군간 차이가 없음을 확인하였다. 중대뇌동맥폐쇄 수술 1주 후 유지시간을 측정하였을 때 대조군의 경우 샴군과 비교하여 급격하게 유지시간이 감소하였다(137.2±30.3 vs. 253.8±13.8 sec, p<0.05). 그러나 갈근 및 황금 복합추출물을 투여한 군에서는 유지시간이 이보다 길었다. 2주 후 갈근 및 황금 복합추출물 100 mg/kg 투여군과 대조군을 비교하였을 때, 갈근 및 황금 복합추출물 100 mg/kg 투여군은 대조군에 비하여 원통 위에서 머무는 시간이 1.6배 증가한 것을 확인하였다 (210.2±23.6 vs. 129.3±20.8 sec, p<0.05).
<실시예 4> 신경학적 손상에 대한 복합추출물의 회복 촉진 효과 확인
랫드를 샴군, 대조군, 갈근 및 황금 복합추출물 30 mg/kg 투여군, 갈근 및 황금 복합추출물 100 mg/kg 투여군, 갈근 및 황금 복합추출물 300 mg/kg 투여군 5개 군으로 나눈 후 <실험예 1>에서 언급한 방법으로 중대뇌동맥 폐쇄를 60분 동안 유발하였다. 수술 4일 후부터 1일 1회 14일간 복합추출물을 kg 당 5 ml의 부피로 경구투여 하였다. 샴군과 대조군의 경우 비히클(5% DMSO)을 복합추출물 투여군과 동일한 방법으로 투여하였다.
신경학적 기능을 평가하기 위하여 HS Kim 등(Kim HS. Histological and Functional Changes after Transplantation of Human Mesenchymal Stem Cell in the Ischemic Rat Model. Kor J Cerebrovascular Surgery. 2005; 7(1): 61-8.)의 방법을 응용하여 수술 후 7, 14, 21일째에 변형된 신경학적 중증도 점수(modified Neurological Severity Score, mNSS) 시험을 수행하였다. 점수가 높을수록 기능이상의 정도가 심한 것으로 정상은 0점이고 가장 심한 신경학적 점수는 7점이다.
mNSS 시험을 수술 1, 2, 3주 후 수행한 결과는 도 3과 같다. 뇌허혈 유발 1주 후 mNSS 점수를 측정하였을 때, 대조군의 점수가 샴군에 비하여 유의하게 증가하였다 (4.3±0.6 vs. 1.6±0.2점,p<0.05). 유발 2주 후 갈근 및 황금 복합추출물 300 mg/kg 투여군의 점수는 3.1±0.5로 대조군 점수인 4.5±0.4점과 비교하였을 때 유의하게 감소하는 것을 확인하였다 (p<0.05). 유발 3주 후 갈근 및 황금 복합추출물 100 mg/kg과 300 mg/kg 투여군의 점수는 각각 3.1±0.4, 2.9±0.4점으로 대조군 점수인 4.5±0.5점과 비교하였을 때 유의하게 감소하는 것을 확인하였다 (모두 p<0.05).
<실시예 5> 체중감소에 대한 복합추출물의 개선 효과 확인
랫드를 샴군, 대조군, 갈근 및 황금 복합추출물 30 mg/kg 투여군, 갈근 및 황금 복합추출물 100 mg/kg 투여군의 4개 군으로 나눈 후 <실험예 1>에서 언급한 방법으로 중대뇌동맥 폐쇄를 60분 동안 유발하였다. 수술 3일 후부터 1일 1회 14일간 복합추출물을 kg 당 5 ml의 부피로 경구투여 하였다. 샴군과 대조군의 경우 비히클(5% DMSO)을 복합추출물 투여군과 동일한 방법으로 투여하였다.
수술 당일부터 실험 종료일인 수술 후 28일째까지 매일 동일한 시간에 체중을 측정하였다.
수술 당일부터 실험종료일인 28일째까지 측정한 군간 체중변화는 도 4와 같다. 수술 직전(0일차) 측정한 체중은 군간 유의한 차이가 없었다. 수술 후 대조군 체중은 급격히 감소하여 수술 4일차에 258.3±9.0 g으로 수술 직전 체중인 302.7±1.2g에 비하여 14.7% 감소한 체중을 보였다. 수술 후 샴군과 대조군 간의 체중은 10일째까지 유의한 차이를 보였다. 복합추출물 100 mg/kg 투여군은 투여 후 체중이 증가하기 시작하여 7일째부터 실험종료일인 28일째까지 대조군 대비 유의한 체중 차이를 보였다. 특히 유발 14일째에는 복합추출물 100 mg/kg 투여한 군의 체중은 359.8±6.7 g으로 샴군의 체중인 359.0±13.0 g과 거의 비슷한 수치를 보였다.
본 발명은 효과적으로 뇌졸중 또는 퇴행성 뇌질환을 예방, 치료 또는 개선시킬 수 있다.
Claims (12)
- 갈근 및 황금 복합추출물을 유효성분으로 포함하는 뇌졸중 또는 퇴행성 뇌질환의 예방 또는 치료용 약학적 조성물.
- 제 1항에 있어서, 상기 갈근 및 황금 복합추출물은 갈근 65 내지 95 중량% 및 황금 5 내지 35 중량%를 포함하는 혼합물로부터 추출된 약학적 조성물.
- 제 2항에 있어서, 상기 갈근 및 황금 복합추출물은 갈근 87.91 중량% 및 황금 12.09 중량%를 포함하는 혼합물로부터 추출된 약학적 조성물.
- 제1항에 있어서, 상기 퇴행성 뇌질환은 치매, 알츠하이머병, 파킨슨병, 헌팅턴 무도병, 피크(Pick)병 및 크로이펠츠-야콥(Creutzfeld-Jacob)병으로 이루어진 군으로부터 선택된 어느 하나인 약학적 조성물.
- 갈근 및 황금 복합추출물을 유효성분으로 포함하는 뇌졸중 또는 퇴행성 뇌질환의 예방 또는 개선용 건강기능식품.
- 제 5항에 있어서, 상기 갈근 및 황금 복합추출물은 갈근 65 내지 95 중량% 및 황금 5 내지 35 중량%를 포함하는 혼합물로부터 추출된 건강기능식품.
- 제 6항에 있어서, 상기 갈근 및 황금 복합추출물은 갈근 87.91 중량% 및 황금 12.09 중량%를 포함하는 혼합물로부터 추출된 건강기능식품.
- 제 5항에 있어서, 상기 퇴행성 뇌질환은 치매, 알츠하이머병, 파킨슨병, 헌팅턴 무도병, 피크(Pick)병 및 크로이펠츠-야콥(Creutzfeld-Jacob)병으로 이루어진 군으로부터 선택된 어느 하나인 건강기능식품.
- 갈근 및 황금 복합추출물을 유효성분으로 포함하는 뇌신경세포 보호 및 감각운동기능(sensorimotor function) 회복 촉진용 약학적 조성물.
- 제 9항에 있어서, 상기 조성물은 체중 증가용인 조성물.
- 갈근 및 황금 복합추출물을 유효성분으로 포함하는 뇌신경세포 보호 및 감각운동기능(sensorimotor function) 회복 촉진용 건강기능식품.
- 제 11항에 있어서, 상기 건강기능식품은 체중 증가용인 건강기능식품.
