Description
固相合成法は、自動合成化が可能でありスピード面で有利であるが、設備制約上スケールアップに制限があり、また、反応性が低いため、ヌクレオチド伸長反応において使用される試薬であるモノマーを過剰に使用する必要があり、工業的な大量合成に向かない。また、途中段階での反応の進行状況の確認、中間体構造解析等も困難という欠点もある。
一方、液相合成法は、カラム精製など煩雑な処理が必要であり、アンチセンス医薬品として利用されうる20mer程度の鎖長のモルフォリノオリゴヌクレオチドを大量かつ迅速に合成することは困難であった。
すなわち、5’位水酸基側が当該保護基で保護されたモルフォリノヌクレオチドをモルフォリノオリゴヌクレオチド液相合成の原料として使用することにより、縮合反応自体は液相中で行うことができるので反応性が固相法に比べて格段に向上し、使用するモノマー当量を格段に低減でき、さらに反応後は晶析または抽出操作によりモルフォリノオリゴヌクレオチドを簡便に単離・精製することができるので、医薬品に利用されうる鎖長が20mer程度のモルフォリノオリゴヌクレオチドを液相法で効率的かつ高収率に合成可能となった。

また、本発明の別の態様は、5’位水酸基が活性化(チオ)ホスフェート化又は活性化(チオ)ホスホルアミダート化され、かつモルフォリン環窒素原子が酸性条件下で除去可能な一時保護基で保護されたp個重合モルフォリノオリゴヌクレオチド(pは、1以上の任意の整数を示す。)を、5’位水酸基又は5’位水酸基の置換基上に存在する水酸基が炭素数10以上300以下のアルキル基及び/又は炭素数10以上300以下のアルケニル基を有する保護基で保護され、かつモルフォリン環窒素原子が保護されていないn個重合モルフォリノオリゴヌクレオチド(nは、1以上の任意の整数を示す。)と、そのモルフォリン環窒素原子を介して(チオ)ホスホルアミダート結合又は(チオ)ホスホロジアミダート結合により縮合させる工程を含む、n+p個重合モルフォリノオリゴヌクレオチドの製造方法である。

文中で特に断らない限り、本明細書で用いるすべての技術用語および科学用語は、本発明が属する技術分野の当業者に一般に理解されるのと同じ意味をもつ。本明細書に記載されたものと同様または同等の任意の方法および材料は、本発明の実施または試験において使用することができるが、好ましい方法および材料を以下に記載する。本明細書で言及したすべての刊行物および特許は、例えば、記載された発明に関連して使用されうる刊行物に記載されている、構築物および方法論を記載および開示する目的で、参照として本明細書に組み入れられる。
モルフォリノヌクレオシド(1)は、自体公知の方法(例えば、WO91/09033A1に記載の方法)、又はそれに準じた方法に従い調製することができる。具体的には、下記スキームに示すように、対応するリボヌクレオシド(2)を過ヨウ素酸ナトリウム等で酸化開環して対応する2’,3’-ジアルデヒド(3)とし、ジアルデヒド(3)をアンモニアで閉環して2’,3’-ジヒドロキシモルフォリノヌクレオシド(4)とし、ジヒドロキシモルフォリノヌクレオシド(4)を還元剤(例、水素化シアノホウ素ナトリウム、水素化トリアセトキシホウ素ナトリウムなど)で還元することにより、モルフォリノヌクレオシド(1)を得ることができる。
m’は、1以上の任意の整数を示し、

本発明で使用される特定の保護基で5’位水酸基又は5’位水酸基の置換基上に存在する水酸基が保護されたモルフォリノヌクレオチドを使用することにより、液相合成に適したモルフォリノオリゴヌクレオチドの製造方法を提供することができる。
中でも目的とするモルフォリノオリゴヌクレオチドの製造方法において高効率かつ高収率が達成できるという観点から、5’位水酸基又は5’位水酸基の置換基上に存在する水酸基が炭素数10以上300以下のアルキル基及び/又は炭素数10以上300以下のアルケニル基を有する保護基で保護されたモルフォリノヌクレオチドが好ましい。

mが0であり、S1が単結合である一般式(I)(以下、「一般式(Ia)」と称する)で表される本発明化合物の製造方法としては、特に限定されないが、自体公知の方法またはこれらに準ずる方法(Richard T.Pon et al.,Nucleic Acids Research 2004,32,623-631.)に従って、上記保護基の前駆体から製造することができる。
上記一般式(Ia)のLがスクシニル基である化合物の一般的な製造方法を以下に示す。
3’末端モルフォリン環窒素原子が保護基P1により保護されたモルフォリノヌクレオシド(a)を、塩基存在下、コハク酸無水物と反応させることにより、5’位水酸基にコハク酸が導入された化合物(b)を得る。化合物(b)を縮合剤存在下、保護基の前駆体(Z-Y-H)(アルコールまたはアミン)と脱水縮合させることにより、一般式(Ia)で表される化合物を得ることができる。
S1が、*O-S2**(式中、各記号は前記と同義を示す。)である場合の化合物については、公知の方法(例えば、WO2008/008113に記載の方法)に準じて、モルフォリノヌクレオシド(a)の5’位水酸基に前記「水酸基を有する置換基」を導入した後、上記方法に従って製造することができる。
また、Yが単結合である場合の化合物については、Z-Y-Hの活性化誘導体(ハロゲン化物、酸ハロゲン化物、活性化カルボキシル基等)を、自体公知の方法に従って、モルフォリノヌクレオシド(a)と反応させるか、あるいは、Z-Y-Hとモルフォリノヌクレオシド(a)を縮合剤の存在下に反応させることにより、製造することができる。Z-Y-Hとモルフォリノヌクレオシド(a)の縮合反応は、Z-Y-Hと化合物(b)の縮合反応と同様に行うことができる。
mが1以上である一般式(I)で示される化合物は、出発原料として、一般式(Ia)で示される化合物を用いて、下記の本発明の製造方法に従い、5’末端伸長プロセスを繰り返すことにより製造することができる。
なお、原料化合物として使用する化合物、例えば、一般式(I)中のZを構成するR5、R7に対応するハロゲン化物等は、市販品として入手可能であるか、あるいは、自体公知の方法またはこれらに準ずる方法に従って製造することができる。
また、該保護基の前駆体(Z-Y-H)は、上記したとおり、自体公知の方法またはこれらに準ずる方法に従って製造することができるが、原料化合物が反応に影響を及ぼす置換基(例えば、水酸基、アミノ基、カルボキシ基)を有する場合には、原料化合物を予め公知の方法に従い、適当な保護基で保護した後に反応を行うことが一般的である。かかる保護基は、反応後に、酸処理、アルカリ処理、接触還元等の公知の方法に従い除去することができる。
例えば、P1がトリチル基である場合は、モルフォリノヌクレオシド(1)を、トリエチルアミン等の塩基存在下に塩化トリチルと反応させることにより、化合物(a)を得ることができる。
また、P1が水素原子である化合物(a)は、P1が一時保護基である化合物(a)を後述の脱保護工程(1)に付することにより得ることができる。
次に、本発明にかかるモルフォリノオリゴヌクレオチドの製造方法(以下、「本発明の製造方法」ともいう。)について説明する。具体的には、適宜保護されたn個重合モルフォリノオリゴヌクレオチドから、適宜保護されたn+p個重合モルフォリノオリゴヌクレオチドへの製造方法について説明していくが、例えば、n=1の場合には、n個重合モルフォリノオリゴヌクレオチドは、「モルフォリノヌクレオシド」と解し、p=1の場合には、p個重合モルフォリノオリゴヌクレオチドは、「モルフォリノヌクレオシド」と解し、n+p個重合モルフォリノオリゴヌクレオチドは、「モルフォリノジヌクレオチド」と解すべきものである。
本発明の製造方法は、好ましくは、以下の工程(2)を含有する。
nの上限は、特に限定されるものではないが、50以下が好ましく、30以下がより好ましく、20以下が更に好ましい。
pの上限は、特に限定されるものではないが、50以下が好ましく、30以下がより好ましく、20以下が更に好ましく、5以下が更に一層好ましく、3以下が特に好ましい。
(1)縮合工程(2)の前に、非極性溶媒中において、5’位水酸基又は5’位水酸基の置換基上に存在する水酸基が炭素数10以上300以下のアルキル基及び/又は炭素数10以上300以下のアルケニル基を有する保護基で保護され、かつモルフォリン環窒素原子が酸性条件下で除去可能な一時保護基で保護されたn個重合モルフォリノオリゴヌクレオチドを酸と反応させて、モルフォリン環窒素原子の一時保護基を除去する工程。
工程(1)は、好ましくは、モルフォリン環窒素原子の一時保護基を除去した後、有機塩基により中和する工程をさらに含む。これにより、工程(1)及び(2)を液中で連続的に行うことが可能になり、モルフォリノヌクレオチドが伸長したモルフォリノオリゴヌクレオチドを連続的に行うことができる。
(3)工程(1)及び/又は(2)で得られた反応液に極性溶媒を添加して、n+p個重合モルフォリノオリゴヌクレオチドを沈殿させて、固液分離により取得する工程。
工程(3)は、下記(A)~(C)に示すように、工程(1)及び(2)の反応液の両方で行ってもよいし、工程(1)及び(2)の反応液のどちらか一方のみで行ってもよい。

(B)工程(1)→工程(2)→工程(3)

また、副生物発生を厳格に管理・制御でき、高純度のモルフォリノオリゴヌクレオチドに導けるという観点で、工程(1)~工程(3)を含む上記(A)~(C)のいずれかを基本単位として繰り返すことが好ましい。
(4)得られたn+p個重合モルフォリノオリゴヌクレオチドの保護基を全て除去する工程。
以下に、各工程について詳細に説明する。
まず、工程(1)及び(2)の原料に用いられるn個重合モルフォリノオリゴヌクレオチドについて説明する。
工程(1)で使用されるn個重合モルフォリノオリゴヌクレオチドは、例えば、下記一般式(i)に示されるような、5’位水酸基又は5’位水酸基の置換基上に存在する水酸基が炭素数10以上300以下のアルキル基及び/又は炭素数10以上300以下のアルケニル基を有する保護基で保護され、かつモルフォリン環窒素原子が酸性条件下で除去可能な一時保護基で保護されたn個重合モルフォリノオリゴヌクレオチドを示し、工程(2)で使用されるn個重合モルフォリノオリゴヌクレオチドは、例えば、下記一般式(ii)に示されるような、5’位水酸基又は5’位水酸基の置換基上に存在する水酸基が炭素数10以上300以下のアルキル基及び/又は炭素数10以上300以下のアルケニル基を有する保護基で保護され、かつモルフォリン環窒素原子が保護されていないn個重合モルフォリノオリゴヌクレオチドを示す。
mは、n-1に相当する0以上の任意の整数を示し、
P1’は、酸性条件下で除去可能な一時保護基を示し、
P2は、炭素数10以上300以下のアルキル基及び/又は炭素数10以上300以下のアルケニル基を有する保護基を示し、

mの上限は、特に限定されるものではないが、通常99以下、好ましくは74以下、さらに好ましくは49以下、さらに好ましくは29以下である。
5’位水酸基又は5’位水酸基の置換基上に存在する水酸基の保護基として、炭素数10以上300以下のアルキル基及び/又は炭素数10以上300以下のアルケニル基を有する保護基、より好ましくは、式(II)で表される基を使用すれば、n個重合モルフォリノオリゴヌクレオチドおよびn+p個重合モルフォリノオリゴヌクレオチドの脂溶性、溶媒(特に、非極性溶媒)に対する溶解性を向上させることができ、工程(1)及び(2)を円滑に行うことができ、また、後述する工程(3)のように、n個重合モルフォリノオリゴヌクレオチド、又はn+p個重合モルフォリノオリゴヌクレオチドを簡便に単離・精製することができる。
式(II)におけるL、Y及びZの好ましい態様は、式(I)におけるL、Y及びZと同様である。
まず、工程(2)の原料に用いられるp個重合モルフォリノオリゴヌクレオチドについて説明する。
工程(2)で使用される「5’位水酸基が活性化(チオ)ホスフェート化又は活性化(チオ)ホスホルアミダート化され、かつモルフォリン環窒素原子が酸性条件下で除去可能な一時保護基で保護されたp個重合モルフォリノオリゴヌクレオチド(pは、1以上の任意の整数を示す。)」としては、当該構造要件を満たす限り特に限定されない。
*は、モルフォリノオリゴヌクレオチドの5’末端水酸基に結合する位置を示し、
L1は、脱離基を示し、
Xは、C1-6アルコキシ基、ジC1-6アルキルアミノ基、または4位窒素原子が保護基で保護され、さらに置換されていてもよいピペラジノ基を示し、

で表される基で修飾されていることを意味する。
「活性化チオホスフェート化」とは、上記式において、XがC1-6アルコキシ基であり、かつWが硫黄原子である場合を意味する。
「活性化ホスホルアミダート化」とは、上記式において、XがジC1-6アルキルアミノ基または4位窒素原子が保護基で保護され、さらに置換されていてもよいピペラジノ基であり、かつWが酸素原子である場合を意味する。
「活性化チオホスホルアミダート化」とは、上記式において、XがジC1-6アルキルアミノ基または4位窒素原子が保護基で保護され、さらに置換されていてもよいピペラジノ基であり、かつWが硫黄原子である場合を意味する。
X及びWの定義、例示、好ましい態様は、上記式(I)で説明した通りである。
「酸性条件下で除去可能な一時保護基」の定義、例示、好ましい態様は、上記式(I)で説明した通りである。
qは、p-1に相当する0以上の任意の整数を示し、

一般式(iii’)の化合物は、例えば国際公開第91/09033号パンフレット等の公知の方法により調製することができる。
以下、工程(1)~(4)を、便宜上、式(i)、(ii)、(iii)等を参照して説明するが、これに限定されるものではない。
本工程は、縮合工程(2)の前に、非極性溶媒中において、5’位水酸基又は5’位水酸基の置換基上に存在する水酸基が炭素数10以上300以下のアルキル基及び/又は炭素数10以上300以下のアルケニル基を有する保護基で保護され、かつモルフォリン環窒素原子が酸性条件下で除去可能な一時保護基で保護されたn個重合モルフォリノオリゴヌクレオチド(i)を酸と反応させて、モルフォリン環窒素原子の一時保護基を除去し、5’位水酸基又は5’位水酸基の置換基上に存在する水酸基が炭素数10以上300以下のアルキル基及び/又は炭素数10以上300以下のアルケニル基を有する保護基で保護され、かつモルフォリン環窒素原子が保護されていないn個重合モルフォリノオリゴヌクレオチド(ii)を得る工程(脱保護工程)である。
良好な反応を達成できるという観点で、トリフルオロ酢酸、シアノピリジントリフルオロ酢酸塩、トリエチルアミントリフルオロ酢酸塩、シアノ酢酸がより好ましく、シアノピリジントリフルオロ酢酸塩、トリエチルアミントリフルオロ酢酸塩が更に好ましく、トリエチルアミントリフルオロ酢酸塩が特に好ましい。これら酸は、上記非極性溶媒で希釈しても構わない。また、前記酸を使用する際には、特定の塩基(例、トリエチルアミン等)を組み合わせて、酸性度を適宜調整して使用しても構わない。
中和に使用する有機塩基としては、前出の酸を中和することができ、得られた塩が縮合剤として機能しうるものであれば特に限定されないが、反応が良好に進行するという観点で、N,N-ジイソプロピルエチルアミン、ピリジン、4-シアノピリジン、トリエチルアミンが好ましく、N,N-ジイソプロピルエチルアミン、トリエチルアミンがより好ましく、N,N-ジイソプロピルエチルアミンが特に好ましい。
本工程は、5’位水酸基が活性化(チオ)ホスフェート化又は活性化(チオ)ホスホルアミダート化され、かつモルフォリン環窒素原子が酸性条件下で除去可能な一時保護基で保護されたp個重合モルフォリノオリゴヌクレオチド(iii)を、5’位水酸基又は5’位水酸基の置換基上に存在する水酸基が炭素数10以上300以下のアルキル基及び/又は炭素数10以上300以下のアルケニル基を有する保護基で保護され、かつモルフォリン環窒素原子が保護されていないn個重合モルフォリノオリゴヌクレオチド(ii)と、そのモルフォリン環窒素原子を介して(チオ)ホスホルアミダート結合又は(チオ)ホスホロジアミダート結合により縮合させて、n+p個重合モルフォリノオリゴヌクレオチド(iv)を得る工程である。
5’位水酸基が活性化(チオ)ホスフェート化又は活性化(チオ)ホスホルアミダート化され、かつモルフォリン環窒素原子が酸性条件下で除去可能な一時保護基で保護されたp個重合モルフォリノオリゴヌクレオチド(iii)としては、pが1である場合(すなわち、5’位水酸基が活性化(チオ)ホスフェート化又は活性化(チオ)ホスホルアミダート化され、かつモルフォリン環窒素原子が一時保護基P1’’により保護されたモルフォリノヌクレオシド)が好ましい。
或いは、工程(1)の後に、反応液を後述の工程(3)に付して、n個重合モルフォリノオリゴヌクレオチド(ii)を一旦単離し、所定の溶媒に溶解させた後に、p個重合モルフォリノオリゴヌクレオチド(iii)を添加してもよい。
なお、二重付加とは、前サイクルの縮合反応で使用されて残留した活性化モルフォリノヌクレオチドが次サイクルの縮合反応で反応し、同じ残基が二重に付加されることを言う。
上記工程(1)および/または(2)の工程において得られるn個重合モルフォリノオリゴヌクレオチド(ii)またはn+p個重合モルフォリノオリゴヌクレオチド(iv)は、5’末端の水酸基が、炭素数10以上300以下のアルキル基及び/又は炭素数10以上300以下のアルケニル基を有する保護基、好ましくは、式(II):Z-Y-L-(式中、各記号は、前記と同義である。)で表される保護基で保護されているため、モルフォリノオリゴヌクレオチドに非常に高い脂溶性が付与されており、カラム精製等の煩雑な操作を要することなく、晶析や抽出操作のみで簡便にモルフォリノオリゴヌクレオチドを単離・精製することができる。
以下に、晶析によるモルフォリノオリゴヌクレオチドの単離・精製法について説明するが、本発明はこれに限定されるものではない。
工程(1)及び/又は(2)で得られた反応液に直接、極性溶媒を添加してもよいし、工程(1)及び/又は(2)で得られた反応液を濃縮した後に極性溶媒を添加してもよい。
本発明のモルフォリノオリゴヌクレオチドの製造方法においては、工程(3)の後に、保護基の種類・性質に応じて、脱保護を行い、モルフォリノオリゴヌクレオチドを単離することができる。脱保護の方法としては、例えば、グリーンズ・プロテクティブ・グループス・イン・オーガニック・シンセシス(Greene’s PROTECTIVE GROUPS in ORGANIC SYNTHESIS)、第4版、ウィリー・インターサイエンス(Wiley-Interscience)出版(2006年)等に記載されている脱保護方法に従い、モルフォリノオリゴヌクレオチドの全ての保護基を除去する工程を行うことができる。具体的には、本願発明における炭素数10以上300以下のアルキル基及び/又は炭素数10以上300以下のアルケニル基を有する保護基、ならびに核酸塩基の保護基であるフェノキシアセチル基、アセチル基等は、アンモニア水、アンモニア水/エタノール溶液、またはアンモニア水とメチルアミン水溶液の混合液で処理することにより、全て除去することができる。また、モルフォリノオリゴヌクレオチド3’末端モルフォリン環窒素原子の保護基は、工程(1)で使用される酸またはそれらを適宜希釈した溶液で処理することにより除去することができる。

アルゴン雰囲気下、化合物1[mo(Tr)Cbz](573mg,1.0mmol)およびN,N-ジメチルアミノピリジン(183mg,1.5mmol)を乾燥ジクロロメタン(7.0ml)に溶解し、無水コハク酸(150mg,1.5mmol)を加えて、室温にて3時間撹拌した。薄層クロマトグラフィーで反応終了を確認した後、メタノール(1.0ml)を加え、反応液を減圧濃縮した。得られた濃縮残渣に酢酸エチル(8.0ml)および0.5mol/lKH2PO4水溶液(8.0ml)を加えて抽出分層し、更に水層へ酢酸エチル(8.0ml)を加えて抽出した。得られた酢酸エチル層を合わせて、0.5mol/lKH2PO4水溶液(13.0ml)、水(13.0ml)、飽和食塩水(13.0ml)で洗浄し、硫酸マグネシウムで乾燥、ろ過した。ろ液を濃縮乾固して化合物2[suc-mo(Tr)Cbz](680mg,quant)を得た。

アルゴン雰囲気下、3,4,5-トリス(オクタデシルオキシ)ベンジルアルコール(133mg,0.15mmol)、N,N-ジメチルアミノピリジン(21mg,0.18mmol)、化合物2[suc-mo(Tr)Cbz](123mg,0.18mmol)を乾燥ジクロロメタン(2.4ml)に溶解し、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド塩酸塩(101mg,0.53mmol)を加え、室温にて終夜撹拌した。薄層クロマトグラフィーで反応終了を確認後、メタノール(6.0ml)を加えて得られた混合液を減圧濃縮した。濃縮残渣にメタノール(6.0ml)を加えて得られたスラリーを15分間撹拌し、ろ過した。得られた固体を減圧加熱乾燥し、化合物3[TOB-suc-mo(Tr)Cbz](218mg,95%)を得た。

アルゴン雰囲気下、化合物5[TOB-suc-PMO[Cbz-T]-Tr](75.9mg,0.04mmol)に対して、2%CYTFA,1%エタノール,10%CF3CH2OHを含むジクロロメタン溶液(1.5ml)を加え、室温で30分間撹拌した。薄層クロマトグラフィーで反応終了を確認した後、5%N,N-ジイソプロピルエチルアミン,25%2-プロパノールを含むジクロロメタン溶液(2.3ml)を加え、室温で30分間撹拌した。その後、この反応液に、ClPONMe2-mo(Tr)Cbz(83.8mg,0.12mmol)のテトラヒドロフラン溶液(0.4ml)および25%N,N-ジイソプロピルエチルアミンを含む1,3-ジメチル-2-イミダゾリジノン溶液(0.5ml)を加えて室温で30分、更に38℃で45分撹拌した。反応液にメタノール(1.5ml)を加え、この溶液を減圧濃縮した。濃縮残渣にメタノール(1.5ml)を加えて室温で15分間撹拌した。得られたスラリーをろ過し、固体をメタノール(1.5ml)で洗浄した。この固体を真空加熱乾燥し、化合物7[TOB-suc-PMO[Cbz-T-Cbz]-Tr](90.8mg,97.9%)を得た。
化合物7[TOB-suc-PMO[Cbz-T-Cbz]-Tr](162mg,0.07mmol)に28%アンモニア水(3.5ml)とエタノール(14.0ml)の混合液を加え、オートクレーブに封入し55℃にて終夜加熱撹拌した。室温まで冷却し、薄層クロマトグラフィーで反応終了を確認し、内容物をメタノールおよびジクロロメタンで洗浄しながらメタノール中へ移液した。この混合液を減圧濃縮し、得られた濃縮残渣にメタノール(14.0ml)を加えて30分間スラリー洗浄した。固体をろ別し、ろ過液を濃縮して得られた固体を0.2mol/lKH2PO4水溶液(pH2.0)に溶解し15分間撹拌した。薄層クロマトグラフィーで原料消失を確認した後、2.0mol/l水酸化ナトリウム水溶液でpH13~pH14に調整し、メンブレンフィルターでろ過し、ろ過液をHPLC分析に供したところ、目的物分析標品と一致した。

化合物3[TOB-suc-mo(Tr)Cbz](94.1mg,0.06mmol)から実施例4および9に記載の方法に従ってトリチル基の脱保護を行い、実施例5および10に記載の方法に従って、対応するモノマーのカップリングを順次行った。各段階で単離した生成物重量と重量収率を下表に示す。
実施例15にて合成した化合物9[TOB-suc-PMO[Cbz-Cbz-T-Cbz-Cbz-Gpac-Gpac-T-T-Cbz-T-Gpac]-Tr](6.0mg,1.0μmol)に28%アンモニア水/エタノール=1/3(v/v)(0.2ml)を加え、オートクレーブに封入し、55℃で終夜加熱撹拌した。室温へ戻し、メタノールで洗浄しながら濾過して固形物を除去した。ろ過液を濃縮し、化合物10[PMO[CCTCCGGTTCTG]-Tr]の粗生成物(3.3mg,80%)を取得した。

実施例19で合成した化合物13[TOB-suc-PMO[Cbz-Cbz-T-Cbz-Cbz-Gce/pac-Gce/pac-T]-Tr](19.3mg,4.2μmol)を28%アンモニア水/エタノール=1/3(v/v)(2.0ml)に懸濁させ、オートクレーブに封入して一晩55℃に加熱撹拌した。室温に冷却した後、ジクロロメタン(1.5ml)およびメタノール(1.5ml)を用いてナスフラスコへ移液し、減圧濃縮した。濃縮残渣にメタノール(2.0ml)を加えて析出した固体をメンブレンフィルターでろ別し、ろ過液を濃縮乾固して化合物14[PMO[CCTCCGGT]-Tr]を定量的に得た。

実施例21で合成した化合物15[TOB-suc-PMO[Cbz-Cbz-T-Cbz-Cbz-Gce/pac-Gce/pac-T-T-Cbz-T-Gce/pac-Abz]-Tr](8.6mg,1.3μmol)を28%アンモニア水/エタノール=1/3(v/v)(1.0ml)に懸濁させ、オートクレーブに封入して一晩55℃に加熱撹拌した。室温に冷却した後、ジクロロメタン(1.0ml)およびメタノール(1.0ml)を用いてナスフラスコへ移液し、減圧濃縮した。濃縮残渣にメタノール(1.0ml)を加えて析出した固体をメンブレンフィルターでろ別し、ろ過液を濃縮乾固して化合物16[PMO[CCTCCGG]-Tr]を定量的に得た。

アルゴン雰囲気下、化合物3[TOB-suc-mo(Tr)Cbz](300mg,0.19mmol)に対して、2%CYTFA,1%エタノール,10%CF3CH2OHを含むジクロロメタン溶液(8.0ml)を加え、室温で90分間撹拌した。薄層クロマトグラフィーで反応終了を確認した後、5%N,N-ジイソプロピルエチルアミン,25%2-プロパノールを含むジクロロメタン溶液(12.0ml)を加え、室温で30分間撹拌した。この溶液を減圧濃縮して得られた濃縮残渣に、アセトニトリル(8.0ml)を加えて室温で15分間撹拌した。得られたスラリーをろ過し、固体をアセトニトリル(8.0ml)で洗浄した。この固体を真空加熱乾燥し、脱Tr化された化合物4[TOB-suc-moCbz](254mg,100%)を得た。
アルゴン雰囲気下、化合物4[TOB-suc-moCbz](242mg,0.18mmol)に対して、ClPONMe2-mo(Tr)Cbz(382mg,0.55mmol)のテトラヒドロフラン(1.8ml)溶液および25%N,N-ジイソプロピルエチルアミンを含む1,3-ジメチル-2-イミダゾリジノン溶液(2.0ml)を加えて室温で30分撹拌した。薄層クロマトグラフィーで反応終了を確認した後、モルフォリン(48μl,0.55mmol)を加え10分間撹拌した。その後、この溶液にアセトニトリル(7.3ml)を加えて減圧濃縮し、得られた濃縮残渣に再度アセトニトリル(7.3ml)を加えて得られたスラリーを15分撹拌した。このスラリーをろ過して得られた固体をアセトニトリル(7.3ml)で洗浄し、減圧加熱乾燥してTOB-suc-PMO[Cbz-Cbz]-Tr(355mg,98%)を得た。
以下同様に、脱トリチル化と対応するモノマーとのカップリングを順次行い、表題化合物17[TOB-suc-PMO[Cbz-Cbz-T-Cbz-Cbz-Gce/pac-Gce/pac]-Tr]を得た。各段階で単離した生成物重量と重量収率を下表に示す。

実施例14で記載の合成途上で得られる2merから5merの中間体について、実施例14記載の方法によって脱保護および担体からの切出しを行い、質量分析(LC-MS)にて不純物解析を実施した。同様に、実施例23に記載の合成途上で得られた2merから7merの中間体についても同様に脱保護と担体からの切出しを行い、質量分析(LC-MS)にて不純物解析を実施した。それぞれの分析結果を比較し、モルフォリンを用いて過剰モノマーをクエンチした実施例23に記載の方法では、n+1merなどの過剰に伸長した不純物は副生していないことが分かった。

DPM-suc-mo(Tr)Cbz(1.69g,1.13mmol)をクロロホルム(16.9mL)に溶解して氷冷し、2,2,2-トリフルオロエタノール(4.3mL) およびエタノール(0.52g,11.3mmol)を投入した後、トリフルオロ酢酸(1.03g,9.07mmol)とトリエチルアミン(0.57g,5.67mmol)のクロロホルム(7.0mL)溶液を滴下し、15℃で1時間撹拌した。反応終了をUHPLCで確認した後、反応液を氷冷し、N,N-ジイソプロピルエチルアミン(0.59g,4.54mmol)のクロロホルム(6mL)溶液を滴下した。反応液にアセトニトリル(16.9mL)を加え、溶媒を減圧留去した。得られた残渣にアセトニトリル(16.9mL)を加え、0℃で30分間撹拌した後、沈殿物をろ取した。得られた結晶をアセトニトリル(16.9mL)で再度洗浄して、DPM-suc-moCbzを得た。この結晶は乾燥することなくそのまま次の反応の原料として使用した。

DPM-suc-moCbz湿結晶(3.33g,1.13mmol相当)をクロロホルム(9.8mL)に溶解し、シクロヘキサン(4.2mL)を加えた。N,N-ジイソプロピルエチルアミン (0.26g,2.03mmol)およびClPONMe2-mo(Tr)Cbz(1.18g,1.69mmol)を投入し、40℃で21時間加熱撹拌した。反応終了をUHPLCで確認した後、反応液を室温に冷却し、モルフォリン(0.20g,2.25mmol)を投入して室温で1時間撹拌した。反応液にアセトニトリル(14mL)を加え、溶媒を減圧留去した。得られたスラリーを0℃で30分間撹拌した後、沈殿物をろ取した。得られた結晶をアセトニトリル(14mL)で再度洗浄し、減圧乾燥してDPM-suc-PMO[Cbz-Cbz]-Trを得た(1.96g、収率91%)。

TOB-suc-moCbz湿結晶(1.39g,0.64mmol相当)をクロロホルム(10mL)に溶解し、N,N-ジイソプロピルエチルアミン (0.15g,1.15mmol)およびClPONMe2-mo(MMTr)Cbz(695mg,0.96mmol)を投入し、室温で17時間撹拌した。反応終了をUHPLCで確認した後、得られた溶液にアセトニトリル(20mL)を加え、沈殿物をろ取した。得られた結晶をアセトニトリル(20mL)で再度洗浄し、減圧乾燥してTOB-suc-PMO[Cbz-Cbz]-MMTrを得た(1.19g、収率92%)。

TOB-suc-moCbz(4.3g,3.23mmol)をクロロホルム(33mL)に溶解し、N,N-ジイソプロピルエチルアミン(0.73g,5.81mmol)およびClPONMe2-mo(Tr)T(2.95g,4.85mmol)を投入し、室温で19時間撹拌した。反応終了をUHPLCで確認した後、反応液を室温に冷却し、モルフォリン(0.28g,3.23mmol)を投入して室温で1時間撹拌した。反応液を氷冷し、2,2,2-トリフルオロエタノール(13.1mL)およびエタノール(1.49g,32.3mmol)を投入した後、トリフルオロ酢酸(2.95g,25.8mmol)とトリエチルアミン(1.21g,12.0mmol)のクロロホルム(9.9mL)溶液を滴下し、15℃で1.5時間撹拌した。反応終了をUHPLCで確認した後、反応液を氷冷し、N,N-ジイソプロピルエチルアミン(3.26g,25.8mmol)のクロロホルム(4.5mL)溶液を滴下した。反応液にアセトニトリル(86mL)を加え、溶媒を減圧留去した。得られたスラリーを0℃で30分間撹拌した後、沈殿物をろ取した。得られた結晶をアセトニトリル(43mL)で再度洗浄してTOB-suc-PMO[Cbz-T]を得た。この結晶は乾燥することなくそのまま次の反応の原料として使用した。
TOB-suc-PMO[Cbz-T]湿結晶(5.7g,3.20mmol相当)をクロロホルム(43mL)に溶解し、N,N-ジイソプロピルエチルアミン(0.73g,25.6mmol)およびClPONMe2-mo(Tr)Cbz(3.35g,4.8mmol)を投入し、室温で16時間撹拌した。反応終了をUHPLCで確認した後、反応液を室温に冷却し、モルフォリン(0.28g,3.20mmol)を投入して室温で1時間撹拌した。反応液を氷冷し、2,2,2-トリフルオロエタノール(13.0mL)およびエタノール(1.48g,32.0mmol)を投入した後、トリフルオロ酢酸(2.92g,25.6mmol)とトリエチルアミン(1.20g,11.8mmol)のクロロホルム(9.8mL)溶液を滴下し、15℃で1.5時間撹拌した。反応終了をUHPLCで確認した後、反応液を氷冷し、N,N-ジイソプロピルエチルアミン(3.23g,25.6mmol)のクロロホルム(4.5mL)溶液を滴下した。反応液にアセトニトリル(106mL)を加え、溶媒を減圧留去した。得られたスラリーを0℃で15分間撹拌した後、沈殿物をろ取した。得られた結晶をアセトニトリル(53mL)で再度洗浄してTOB-suc-PMO[Cbz-T-Cbz]を得た。この結晶は乾燥することなくそのまま次の反応の原料として使用した。
TOB-suc-moCbz湿結晶(4.9g,3.08mmol相当)をクロロホルム(34.9mL)に溶解し、塩化リチウム(143mg,3.38mmol)、N,N-ジイソプロピルエチルアミン(0.72g,5.54mmol)およびClPONMe2-mo(Tr)T(2.81g,4.61mmol)を投入し、40℃で16時間撹拌した。反応終了をUHPLCで確認した後、反応液を室温に冷却し、モルフォリン(0.32g,3.69mmol)を投入して室温で1時間撹拌した。反応液を氷冷し、2,2,2-トリフルオロエタノール(12.5mL)およびエタノール(1.41g,30.8mmol)を投入した後、トリフルオロ酢酸(1.75g,15.4mmol)とトリエチルアミン(0.49g,4.86mmol)のクロロホルム(5.9mL)溶液を滴下し、室温で1時間撹拌した。反応終了をUHPLCで確認した後、反応液を氷冷し、N,N-ジイソプロピルエチルアミン(2.0g,15.4mmol)のクロロホルム(2.7mL)溶液を滴下した。反応液にアセトニトリル(82mL)を加え、溶媒を減圧留去した。得られたスラリーを0℃で30分間撹拌した後、沈殿物をろ取した。得られた結晶をアセトニトリル(41mL)で再度洗浄してTOB-suc-PMO[Cbz-T]を得た。この結晶は乾燥することなくそのまま次の反応の原料として使用した。
TOB-suc-PMO[Cbz-T]湿結晶(5.5g,3.05mmol相当)をクロロホルム(44.6mL)に溶解し、塩化リチウム(142mg,3.36mmol)、N,N-ジイソプロピルエチルアミン(0.71g,5.49mmol)およびClPONMe2-mo(Tr)Cbz(3.19g,4.58mmol)を投入し、40℃で16時間撹拌した。反応終了をUHPLCで確認した後、反応液を室温に冷却し、モルフォリン(0.26g,3.05mmol)を投入して室温で1時間撹拌した。反応液を氷冷し、2,2,2-トリフルオロエタノール(12.4mL)およびエタノール(1.41g,30.5mmol)を投入した後、トリフルオロ酢酸(1.74g,15.3mmol)とトリエチルアミン(0.52g,5.19mmol)のクロロホルム(5.8mL)溶液を滴下し、室温で1時間撹拌した。反応終了をUHPLCで確認した後、反応液を氷冷し、N,N-ジイソプロピルエチルアミン(1.97g,15.3mmol)のクロロホルム(2.7mL)溶液を滴下した。反応液にアセトニトリル(100mL)を加え、溶媒を減圧留去した。得られたスラリーを0℃で15分間撹拌した後、沈殿物をろ取した。得られた結晶をアセトニトリル(50mL)で再度洗浄し、減圧乾燥してTOB-suc-PMO[Cbz-T-Cbz]を得た(6.74g、収率106%)。
TOB-suc-moCbz(4.3g,3.23mmol)をクロロホルム(42.8mL)に溶解し、塩化リチウム(150mg,3.55mmol)、N,N-ジイソプロピルエチルアミン(0.73g,5.81mmol)およびClPONMe2-mo(Tr)T(2.95g,4.85mmol)を投入し、室温で15時間撹拌した。反応終了をUHPLCで確認した後、反応液を室温に冷却し、無水酢酸(0.03mL,0.32mmol)および2,6-ルチジン(0.19mL,1.62mmol)を加えて室温で1時間撹拌した。反応液にモルフォリン(0.28g,3.23mmol)を投入して室温で1時間撹拌した。反応液を氷冷し、インドール(3.78g,32.3mmol)を投入した後、トリフルオロ酢酸(2.86g,25.8mmol)のクロロホルム(19.2mL)溶液を滴下し、室温で1.5時間撹拌した。反応終了をUHPLCで確認した後、反応液を氷冷し、N,N-ジイソプロピルエチルアミン(3.25g,25.8mmol)のクロロホルム(4.5mL)溶液を滴下した。反応液にアセトニトリル(86mL)を加え、溶媒を減圧留去した。得られたスラリーを0℃で15分間撹拌した後、沈殿物をろ取した。得られた結晶をアセトニトリル(43mL)で再度洗浄してTOB-suc-PMO[Cbz-T]を得た。この結晶は乾燥することなくそのまま次の反応の原料として使用した。
TOB-suc-PMO[Cbz-T]湿結晶(6.5g,3.23mmol相当)をクロロホルム(53.4mL)に溶解し、塩化リチウム(150mg,3.36mmol)、N,N-ジイソプロピルエチルアミン(0.73g,5.81mmol)およびClPONMe2-mo(Tr)Cbz(3.38g,4.85mmol)を投入し、室温で40時間撹拌した。反応終了をUHPLCで確認した後、反応液を室温に冷却し、無水酢酸(0.03mL,0.32mmol)および2,6-ルチジン(0.15mL,1.29mmol)を加えて室温で1時間撹拌した。反応液にモルフォリン(0.28g,3.23mmol)を投入して室温で1時間撹拌した。反応液を氷冷し、インドール(3.78g,32.3mmol)を投入した後、トリフルオロ酢酸(2.86g,25.8mmol)のクロロホルム(19.2mL)溶液を滴下し、室温で1.5時間撹拌した。反応終了をUHPLCで確認した後、反応液を氷冷し、N,N-ジイソプロピルエチルアミン(3.25g,25.8mmol)のクロロホルム(4.5mL)溶液を滴下した。反応液にアセトニトリル(107mL)を加え、溶媒を減圧留去した。得られたスラリーを0℃で15分間撹拌した後、沈殿物をろ取した。得られた結晶をアセトニトリル(53mL)で再度洗浄し、減圧乾燥してTOB-suc-PMO[Cbz-T-Cbz]を得た。この結晶は乾燥することなくそのまま次の反応の原料として使用した。
TOB-suc-moCbz湿結晶(16.1g,3.2mmol相当)をクロロホルム(42mL)に溶解し、N,N-ジイソプロピルエチルアミン (0.74g,5.76mmol)およびClPONMe2mo(Tr)Cbz(3.35g,4.80mmol)を投入し、室温で15時間撹拌した。反応終了をUHPLCで確認した後、反応液を室温に冷却し、モルホリン(0.28g,3.2mmol)を投入して室温で1時間撹拌した。反応液を氷冷し、2,2,2-トリフルオロエタノール(14.1mL)およびエタノール(1.5g,32.0mmol)を投入した後、トリフルオロ酢酸(2.92g,25.6mmol)とトリエチルアミン(1.13g,11.2mmol)のクロロホルム(40mL)溶液を滴下し、15℃で30分間撹拌した。反応液に2,2,2-トリフルオロエタノール(3.5mL)を追加し、さらに15℃で1時間撹拌した。反応終了をUHPLCで確認した後、反応液を氷冷し、N,N-ジイソプロピルエチルアミン (3.31g,25.6mmol)のクロロホルム(30mL)溶液を滴下した。反応液にアセトニトリル(75mL)を加え、溶媒を減圧留去した。得られた残渣にアセトニトリル(25mL)を加え、溶媒を減圧留去した後、アセトニトリル(100mL)を加えた。得られたスラリーを0℃で30分間撹拌した後、沈殿物をろ取した。得られた結晶をアセトニトリル(100mL)で再度洗浄し、減圧乾燥してTOB-suc-PMO[Cbz-Cbz]を得た(5.4g、収率96.6%対TOB-suc-moCbz)。
TOB-suc-PMO[Cbz-Cbz](5.4g,3.09mmol)をクロロホルム(54mL)に溶解し、N,N-ジイソプロピルエチルアミン (0.72g,5.56mmol)およびClPONMe2-mo(Tr)T(2.82g,4.64mmol)を投入し、室温で23時間撹拌した。反応終了をUHPLCで確認した後、反応液を室温に冷却し、モルフォリン(0.27g,3.09mmol)を投入して室温で1時間撹拌した。反応液を氷冷し、2,2,2-トリフルオロエタノール(17mL)およびエタノール(1.42g,30.9mmol)を投入した後、トリフルオロ酢酸(2.82g,24.7mmol)とトリエチルアミン(1.16g,11.4mmol)のクロロホルム(39mL)溶液を滴下し、15℃で1.5時間撹拌した。反応終了をUHPLCで確認した後、反応液を氷冷し、N,N-ジイソプロピルエチルアミン(3.19g,24.7mmol)のクロロホルム(30mL)溶液を滴下した。反応液にアセトニトリル(160mL)を加え、溶媒を減圧留去した。得られた残渣にアセトニトリル(80mL)を加え、溶媒を減圧留去した後、アセトニトリル(100mL)を加えた。得られたスラリーを0℃で30分間撹拌した後、沈殿物をろ取した。得られた結晶をアセトニトリル(100mL)で再度洗浄し、減圧乾燥してTOB-suc-PMO[Cbz-Cbz-T]を得た。この結晶は乾燥することなくそのまま次の反応の原料として使用した。
TOB-suc-mo(Tr)Cbz(5.0g,3.2mmol)から実施例37および38に記載の方法に従って、対応するモノマーのカップリングおよびトリチル基の脱保護をワンポットで順次行った。各段階での工程収率を下表に示す。なお、数値のない鎖長の生成物は、乾燥せずに次工程で使用した。
TOB-suc-PMO[Cbz-Cbz-T-Cbz-Cbz-Gce/pac-Gce/pac-T-T]湿結晶(12.9g,2.37mmol相当)をクロロホルム(100mL)に溶解し、塩化リチウム(150mg,3.56mmol)、N,N-ジイソプロピルエチルアミン(0.53g,4.27mmol)およびClPONMe2mo(Tr)Cbz(2.48g,3.56mmol)を投入し、室温で33時間撹拌した。反応終了をUHPLCで確認した後、反応液を室温に冷却し、モルフォリン(0.78g,9.0mmol)を投入して室温で1時間撹拌した。反応液を氷冷し、2,2,2-トリフルオロエタノール(10.2mL)およびエタノール(1.1g,23.7mmol)を投入した後、トリフルオロ酢酸(1.89g,16.6mmol)のクロロホルム(10mL)溶液を滴下し、室温で2時間撹拌した。反応終了をUHPLCで確認した後、反応液を氷冷し、N,N-ジイソプロピルエチルアミン(2.07g,16.0mmol)のクロロホルム(10mL)溶液を滴下した。反応液にアセトニトリル(200mL)を加え、溶媒を減圧留去した。得られた残渣にアセトニトリル(100mL)を加えた。得られたスラリーを0℃で30分間撹拌した後、沈殿物をろ取した。得られた結晶をアセトニトリル(100mL)で再度洗浄してTOB-suc-PMO[Cbz-Cbz-T-Cbz-Cbz-Gce/pac-Gce/pac-T-T-Cbz]を得た。この結晶は乾燥することなくそのまま次の反応の原料として使用した。
TOB-suc-PMO[Cbz-Cbz-T-Cbz-Cbz-Gce/pac-Gce/pac-T-T-Cbz]湿結晶(12.6g,2.37mmol)をクロロホルム(110mL)に溶解し、塩化リチウム(150mg,3.56mmol)、N,N-ジイソプロピルエチルアミン(0.53g,4.27mmol)およびClPONMe2-mo(Tr)T(2.17g,3.56mmol)を投入し、室温で19時間撹拌した。反応終了をUHPLCで確認した後、反応液を室温に冷却し、モルフォリン(0.78g,9.01mmol)を投入して室温で1時間撹拌した。反応液を氷冷し、2,2,2-トリフルオロエタノール(10.2mL)およびエタノール(1.09g,23.7mmol)を投入した後、トリフルオロ酢酸(1.89g,16.6mmol)のクロロホルム(10mL)溶液を滴下し、室温で2時間撹拌した。反応終了をUHPLCで確認した後、反応液を氷冷し、N,N-ジイソプロピルエチルアミン(2.07g,16.0mmol)のクロロホルム(10mL)溶液を滴下した。反応液にアセトニトリル(150mL)を加え、溶媒を減圧留去した。得られた残渣にアセトニトリル(200mL)を加え、溶媒を減圧留去した後、アセトニトリル(50mL)を加えた。得られたスラリーを0℃で30分間撹拌した後、沈殿物をろ取した。得られた結晶をアセトニトリル(100mL)で再度洗浄し、減圧乾燥してTOB-suc-PMO[Cbz-Cbz-T-Cbz-Cbz-Gce/pac-Gce/pac-T-T-Cbz-T]を得た。この結晶は乾燥することなくそのまま次の反応の原料として使用した。
TOB-suc-mo(Tr)Cbz(5.0g,3.2mmol)から実施例37、38、40および41に記載の方法に従って、対応するモノマーのカップリングおよびトリチル基の脱保護をワンポットで順次行った。各段階での工程収率を下表に示す。なお、数値のない鎖長の生成物は、乾燥せずに次工程で使用した。
3,4,5-トリ(オクタデシル)ベンジルアルコール(91.4mg,0.1mmol)をクロロホルム(1.0mL)に溶解し、suc-teg-Tr(72.6mg,0.12mmol)、EDC・HCl(23.0mg,0.12mmol)および4-ジメチルアミノピリジン(1.2mg,0.01mmol)を加えて、室温で65時間撹拌した。反応終了をUHPLCで確認した後、アセトニトリル(2mL)を加え、溶媒を減圧留去した。得られた残渣にアセトニトリル(4mL)を加え、沈殿物をろ取し、減圧乾燥してTOB-suc-teg-Trを得た(140mg、収率93%)。

TOB-suc-teg-Tr(139mg,0.09mmol)をクロロホルム(1.4mL)に溶解して氷冷し、2,2,2-トリフルオロエタノール(0.4mL)およびエタノール(42.8g,0.93mmol)を投入した後、トリフルオロ酢酸(82.7mg,0.75mmol)とトリエチルアミン(47.4mg,0.47mmol)のクロロホルム(0.5mL)溶液を滴下し、室温で2時間撹拌した。反応終了をUHPLCで確認した後、反応液を氷冷し、N,N-ジイソプロピルエチルアミン(97mg,0.75mmol)のクロロホルム(0.5mL)溶液を滴下した。反応液にアセトニトリル(5mL)を加え、溶媒を減圧留去した。得られた残渣にアセトニトリル(4mL)を加え、0℃で30分間撹拌した後、沈殿物をろ取してTOB-suc-teg-Hを得た。この結晶は乾燥することなくそのまま次の反応の原料として使用した。

TOB-suc-teg-H湿結晶(168mg,0.09mmol相当)をクロロホルム(1.2mL)に溶解し、N,N-ジイソプロピルエチルアミン(20.7mg,0.16mmol)およびClPONMe2-mo(Tr)Cbz(92.8mg,0.13mmol)を投入し、室温で13時間加熱撹拌した。反応終了をUHPLCで確認した後、反応液を室温に冷却し、モルフォリン(11.3mg,0.09mmol)を投入して室温で1時間撹拌した。反応液にアセトニトリル(2.4mL)を加え、溶媒を減圧留去した。得られた残渣にアセトニトリル(3mL)を加え、0℃で30分間撹拌した後、沈殿物をろ取し、減圧乾燥してTOB-suc-teg-mo(Tr)Cbzを得た(148mg、収率86%、対TOB-suc-teg-Tr)。

2-{3,4,5-トリス[3,4,5-トリ(2’,3’-ジヒドロフィチルオキシ)ベンジル]ベンジルアミノカルボニル}エチルカルボニル基からなる群から選択される、
請求項1または2に記載のモルフォリノヌクレオチド。 - P1が、トリチル基、モノメトキシトリチル基、またはジメトキシトリチル基である、請求項1~7のいずれか一項に記載のモルフォリノヌクレオチド。
- (2)5’位水酸基が活性化(チオ)ホスフェート化又は活性化(チオ)ホスホルアミダート化され、かつモルフォリン環窒素原子が酸性条件下で除去可能な一時保護基で保護されたp個重合モルフォリノオリゴヌクレオチド(pは、1以上の任意の整数を示す。)を、5’位水酸基又は5’位水酸基が水酸基を有する置換基を有する場合は該置換基上に存在する水酸基が炭素数10以上300以下のアルキル基及び/又は炭素数10以上300以下のアルケニル基を有する保護基で保護され、かつモルフォリン環窒素原子が保護されていないn個重合モルフォリノオリゴヌクレオチド(nは、1以上の任意の整数を示す。)と、そのモルフォリン環窒素原子を介して(チオ)ホスホルアミダート結合又は(チオ)ホスホロジアミダート結合により縮合させる工程を含む、n+p個重合モルフォリノオリゴヌクレオチドの製造方法。
- pが1である、請求項9に記載の製造方法。
- 反応終了後、反応混合物をクエンチ剤で処理する、請求項9または10記載の製造方法。
- 更に、下記工程(1)を含有する、請求項9~11のいずれか一項に記載の製造方法。
(1)縮合工程(2)の前に、非極性溶媒中において、5’位水酸基又は5’位水酸基が置換基を有する場合は該置換基上に存在する水酸基が炭素数10以上300以下のアルキル基及び/又は炭素数10以上300以下のアルケニル基を有する保護基で保護され、かつモルフォリン環窒素原子が酸性条件下で除去可能な一時保護基で保護されたn個重合モルフォリノオリゴヌクレオチドを酸と反応させて、モルフォリン環窒素原子の一時保護基を除去する工程。 - 一時保護基の除去がカチオン捕捉剤の存在下に行われる、請求項12記載の製造方法。
- 工程(1)において、モルフォリン環窒素原子の一時保護基を除去した後、有機塩基により中和する工程をさらに含む、請求項12又は13に記載の製造方法。
- 炭素数10以上300以下のアルキル基及び/又は炭素数10以上300以下のアルケニル基を有する保護基が、下記式(II):
**は、S1との結合位置を示し;
L1は、置換されていてもよい2価のC1-22炭化水素基を示し;かつ
L2は、C(=O)を示すか、または***N(R3)-R1-N(R2)C(=O)**(式中、**は、L1との結合位置を示し、***は、Yとの結合位置を示し、R1は、置換されていてもよいC1-22アルキレン基を示し、R2およびR3は、それぞれ独立して、水素原子もしくは置換されていてもよいC1-22アルキル基を示すか、またはR2およびR3が一緒になって、置換されていてもよいC1-22アルキレン結合を形成していてもよい。)で表される基を示す。]で示される基を示し、
Yは、単結合、酸素原子、またはNR(Rは、水素原子、アルキル基またはアラルキル基を示す。)を示し、ならびに
Zは、式(a2):
R4は、水素原子を示すか、あるいはRbが、下記式(a3)で表される基である場合には、R6と一緒になって単結合または-O-を示して、環Bと共にフルオレニル基またはキサンテニル基を形成していてもよく;
k個のQは、独立してそれぞれ単結合を示すか、あるいは-O-、-S-、-OC(=O)-、-NHC(=O)-または-NH-を示し;
k個のR5は、独立してそれぞれ炭素数10以上300以下のアルキル基及び/又は炭素数10以上300以下のアルケニル基を有する有機基を示し;
kは、1~4の整数を示し;
環Aは、k個のQR5に加えて、更にハロゲン原子、ハロゲン原子で置換されていてもよいC1-6アルキル基、およびハロゲン原子で置換されていてもよいC1-6アルコキシ基からなる群から選択される置換基を有していてもよく;
Raは、水素原子を示し;
Rbは、水素原子、または式(a3):
jは、0~4の整数を示し;
j個のQは、前記と同意義を示し;
j個のR7は、独立してそれぞれ炭素数10以上300以下のアルキル基及び/又は炭素数10以上300以下のアルケニル基を有する有機基を示し;
R6は、水素原子を示すか、またはR4と一緒になって単結合または-O-を示して、環Aと共にフルオレニル基またはキサンテニル基を形成していてもよく;かつ
環Bは、j個のQR7に加えて、更にハロゲン原子、ハロゲン原子で置換されていてもよいC1-6アルキル基、およびハロゲン原子で置換されていてもよいC1-6アルコキシ基からなる群から選択される置換基を有していてもよい。)で表される基を示し、あるいは
Ra及びRbは一緒になって、酸素原子を形成する。]で表される基である、請求項9~14のいずれか一項に記載の製造方法。 - 更に、下記工程(3)を含有する、請求項15記載の製造方法。
(3)工程(1)及び/又は(2)で得られた反応液に極性溶媒を添加して、モルフォリノオリゴヌクレオチドを沈殿させて、固液分離により取得する工程。 - 非極性溶媒が、ハロゲン系溶媒、芳香族系溶媒、エステル系溶媒、脂肪族系溶媒、非極性エーテル系溶媒、およびこれらの組合せからなる群より選択される溶媒である、請求項12~16のいずれか一項に記載の製造方法。
- 極性溶媒が、アルコール系溶媒、またはニトリル系溶媒である、請求項16又は17に記載の製造方法。
- 更に、下記工程(4)を含有する、請求項9~18のいずれか一項に記載の製造方法。
(4)得られたn+p個重合モルフォリノオリゴヌクレオチドの保護基を全て除去する工程。 - 酸性条件下で除去可能な一時保護基が、トリチル基、ジメトキシトリチル基、またはモノメトキシトリチル基である、請求項9~19のいずれか一項に記載の製造方法。
