Abstract
본 발명은 ASM 억제제를 유효성분으로 포함하는 퇴행성 신경질환의 예방 또는 치료용 조성물에 관한 것이다. 본 발명에 따르면, 알츠하이머병 모델 마우스에 ASM을 부분적으로 결손시킨 경우, ASM 유전자가 부분적으로 결손된 마우스와 알츠하이머병 모델 마우스를병체 결합시킨 모델의 경우 및 ASM 유전자가 결손된 마우스의 혈청을 알츠하이머병 모델 마우스에 주입한 경우 등, 알츠하이머병 모델 마우스에서 ASM을 억제하였을 때, 뇌 조직 내 β-아밀로이드의 침착이 저해되고, 학습 및 기억력이 개선되는 우수한 효과가 있음을 확인하였다. 따라서, ASM 억제제는 알츠하이머병을 비롯한 퇴행성 신경질환의 예방 또는 치료에 유용하게 이용될 수 있다.
Description
본 발명은 ASM (acid sphingomyelinase) 억제제를 유효성분으로 포함하는 퇴행성 신경질환의 예방 또는 치료용 조성물에 관한 것이다.
치매(dementia)는 일상생활에 장애를 주는 기억과 인지 능력의 점차적인 악화로 정의되며, 크게 혈관성 치매와 알츠하이머형 치매로 나눌 수 있다. 혈관성 치매는 주로 혈관 내에 형성된 혈전에 의해 뇌경색 또는 뇌졸중 등이 발생하는 경우에 해당되며 발병 주변의 뇌 세포가 손상을 입어 기억력 상실 등의 증상이 유발되는 것으로 알려져 있다. 반면, 혈관성 치매보다 훨씬 더 많은 비중을 차지하고 있는 알츠하이머형 치매(Alzheimer's Diseases : AD)는 서서히 진행되는 뇌 질환으로서 처음에는 기억력 감퇴, 성격의 변화 그리고 사고력이 감소하는 질병으로서 대부분의 환자는 8-10년 내에 폐렴 등으로 사망한다. 전 세계적으로 65세 이상의 노인 중 3.5-10% 가량이 이 질환을 앓고 있으며 미국에서만 400만 명의 환자가 있는 것으로 추정된다. 이 질환을 치료하기 위하여 소요되는 사회적 비용만도 미국에서만 연 1000억불이 지출되고 있을 정도로 노인의 대표적인 질병이다.
현재까지 밝혀진 알츠하이머형 치매의 발생 기전으로는 아밀로이드 전구 단백질(APP)에서 β-아밀로이드가 유리되고, 유리된 β-아밀로이드는 서로 응집되어 불용성 아밀로이드 플라크를 생성하는데, 이 β-아밀로이드의 응집 및 아밀로이드 플라크(amyloid plaques)의 생성이 신경 세포의 퇴행을 일으키게 되고, 그 결과 이차적으로 신경섬유 농축체(neurofibrillary tangle)의 생성이 유발된다는 것이다. 이와 같이 뇌 조직 내 β-아밀로이드의 축적과 그로 인한 신경독성이 알츠하이머형 치매의 매우 중요한 발병원인으로 작용하고 있음이 밝혀짐에 따라 BACE-1 억제제와 같이 β-아밀로이드의 생성 저지, 응집 저지 또는 독성 저지 효과를 지니면서 상대적으로 부작용이 적은 물질의 탐색 연구가 전 세계적으로 집중 연구되고 있다. β-아밀로이드는 아밀로이드 단백질의 전구체인 APP가 감마-시크리타제 및 베타-시크리타제라는 단백분해효소들의 작용을 받아 생성된 아밀로이드 전구 단백질의 절편이다. β-아밀로이드의 생성에 가장 주요한 역할을 하고 있는 효소 베타-시크리타제는 일반적으로 BACE로 불리우고 있으며, BACE-1과 BACE-2 등 두 개의 타입이 알려져 있다. 이 중 BACE-1은 베타-시크리타제의 대부분의 활성(약 90%)을 지니고 있어 β-아밀로이드 생성 과정에 있어 BACE-2에 비하여 훨씬 더 중요한 역할을 담당하고 있다고 알려져 있다.
또한, 최근의 역학 연구에 의하면 고혈압, 당뇨병, 고지혈증 및 심장질환 등 뇌혈관 질환의 위험인자들이 혈관성 치매뿐만 아니라 알츠하이머형 치매의 발병률을 증가시킨다고 보고되었다. 알츠하이머형 치매(AD)로 인한 인지기능 저하현상에 대한 현대 의학적 관점은 대부분 뇌 콜린성 신경세포의 광범위한 변성 및 소실을 인지기능감퇴의 가장 주요한 원인으로 간주하고 있으며, 이를 극복하기 위한 방편으로 손상되지 않고 남아있는 콜린성 신경계의 활성을 증가시켜 손상된 인지기능을 부분적으로 회복시킬 수 있는 약물들을 개발하고자 하는 연구가 주종을 이루고 있다.
현재 미국 식품의약국(FDA)으로부터 알츠하이머형 치매의 치료제로 공인받은 네 가지 약물(타크린, 리바스티그민, 도네페질, 갈란타민)은 모두 아세틸콜린 분해 효소의 활성을 저해함으로써 인지기능을 항진시키고자하는 일명 아세틸콜린에스테라제 억제제(acetylcholinesterase inhibitors)들이다. 이와 같이 지금까지 알려진 알츠하이머형 치매의 치료제로 승인된 의약품은 아세틸콜린에스테라제 억제제가 유일한 실정이나, 이들 약제들은 일부 알츠하이머형 치매 환자(40-50%)에서 일시적인 병증 완화 효과만을 보이고 그 약효가 오래 지속되지 못하는 단점이 있다. 또한 질환의 특성상 장기 복용을 요하는데, 지금까지 개발된 아세틸콜린에스테라제 억제제는 간독성을 비롯한 여러 가지 부작용을 수반하는 것 또한 문제점으로 드러나고 있다. 즉, 현재까지 개발된 치료제들은 단지 일시적으로 증상만 완화시킬 뿐이므로, 병을 근본적으로 치료하거나 진행을 억제하는 약물의 개발이 절실히 요구되고 있다.
한편, 스핑고지질 대사는 정상적인 세포 신호전달을 조절하며, 스핑고지질 대사를 조절하는 효소인 ASM(acid sphingomyelinase)은 거의 모든 종류의 세포에서 발현되는 단백질로서, 스핑고지질 대사 및 세포막 턴오버(turnover)에 중요한 역할을 한다. 상기 ASM은 대부분 엔도좀/리소좀 부위 안에 위치하고, 세포 스트레스 반응 시, 세포막의 바깥쪽으로 이동한다. ASM은 윌슨병, 당뇨병, 낭포성 섬유증, 폐기종 등 다양한 질병에서 증가되어 있으며 질병의 발병과 상당한 연관성을 지니고 있다. 그러나 ASM의 상기와 같은 역할에도 불구하고, ASM과 알츠하이머병과의 관련성에 관한 연구는 현재 미미한 실정이다.
이에 본 발명자들은 ASM의 알츠하이머병에 대한 발병기전을 발견하고, 알츠하이머병 모델 마우스에 ASM을 부분적으로 결손시킨 경우, ASM 유전자가 부분적으로 결손된 마우스와 알츠하이머병 모델 마우스를 병체 결합시킨 모델의 경우 및 ASM 유전자가 결손된 마우스의 혈청을 알츠하이머병 모델 마우스에 주입한 경우 등 알츠하이머병 모델 마우스에서 ASM을 억제하였을 때, 뇌 조직 내 β-아밀로이드의 침착이 저해되고, 학습 및 기억력이 개선됨을 확인함으로써, 본 발명을 완성하게 되었다.
본 발명의 목적은 ASM(acid sphingomyelinase) 활성 억제제 또는 발현 억제제를 유효성분으로 포함하는 퇴행성 신경질환의 예방 또는 치료용 조성물을 제공함에 있다.
본 발명의 다른 목적은 상기 조성물의 치료적 유효량을 개체에 투여하는 단계;를 포함하는, 퇴행성 신경질환의 예방 또는 치료방법을 제공함에 있다.
본 발명의 또 다른 목적은 ASM의 발현양 변화를 이용한 퇴행성 신경질환의 예방 또는 치료물질의 스크리닝 방법을 제공함에 있다.
상기와 같은 목적을 달성하기 위하여, 본 발명은 ASM(acid sphingomyelinase) 활성 억제제 또는 발현 억제제를 유효성분으로 포함하는 퇴행성 신경질환의 예방 또는 치료용 조성물을 제공한다.
또한, 본 발명은 상기 조성물의 치료적 유효량을 개체에 투여하는 단계;를 포함하는, 퇴행성 신경질환의 예방 또는 치료방법을 제공한다.
또한, 본 발명은 생물학적 시료에 후보물질을 처리하여 ASM의 발현양 변화를 측정하는 단계;를 포함하는 퇴행성 신경질환의 예방 또는 치료물질의 스크리닝 방법을 제공한다.
본 발명에 따르면, 알츠하이머병 모델 마우스에 ASM을 부분적으로 결손시킨 경우, ASM 유전자가 부분적으로 결손된 마우스와 알츠하이머병 모델 마우스를병체 결합시킨 모델의 경우 및 ASM 유전자가 결손된 마우스의 혈청을 알츠하이머병 모델 마우스에 주입한 경우 등, 알츠하이머병 모델 마우스에서 ASM을 억제하였을 때, 뇌 조직 내 β-아밀로이드의 침착이 저해되고, 학습 및 기억력이 개선되는 우수한 효과가 있음을 확인하였다. 따라서, ASM 억제제는 알츠하이머병을 비롯한 퇴행성 신경질환의 예방 또는 치료에 유용하게 이용될 수 있다.

이하, 본 발명을 상세히 설명한다.
본 발명은 ASM(acid sphingomyelinase)의 활성 억제제 또는 발현 억제제를 유효성분으로 포함하는 퇴행성 신경질환의 예방 또는 치료용 조성물을 제공한다.
상기 조성물은 약학적 조성물 또는 식품 조성물을 포함한다.
본 발명에 따르면, 알츠하이머병 모델 마우스에 ASM을 부분적으로 결손시킨 경우, ASM 유전자가 부분적으로 결손된 마우스와 알츠하이머병 모델 마우스를병체 결합시킨 모델의 경우 및 ASM 유전자가 결손된 마우스의 혈청을 알츠하이머병 모델 마우스에 주입한 경우 등, 알츠하이머병 모델 마우스에서 ASM을 억제하였을 때, 뇌 조직 내 β-아밀로이드의 침착이 저해되고, 학습 및 기억력이 개선되는 우수한 효과가 있음을 확인하였다. 따라서, ASM 억제제는 알츠하이머병을 비롯한 퇴행성 신경질환의 예방 또는 치료에 유용하게 이용될 수 있다.
본 발명에 따른 ASM 활성 억제제는 ASM 단백질에 특이적으로 결합하는 화합물, 펩티드, 펩티드 모방체, 기질유사체, 앱타머 및 항체로 구성된 군으로부터 선택된 1종 이상 일 수 있으며, 이에 제한되지 않는다.
상기 펩티드 모방체(Peptide mimetics)는 ASM 단백질의 결합 도메인을 억제하여 ASM 단백질의 활성을 억제하는 것이다. 펩티드 모방체는 펩티드 또는 비펩티드일 수 있고, psi 결합(Benkirane, N., et al. J.Biol. Chem., 271:33218-33224, 1996)과 같은, 비펩티드 결합에 의해 결합된 아미노산으로 구성될 수 있다. 또한, "구조적으로 강제된(conformationally constrained)" 펩티드, 사이클릭 모방체(cyclic mimetics), 적어도 하나의 엑소사이클릭 도메인(exocyclic domain), 결합 부분(결합 아미노산) 및 활성 부위를 포함하는 사이클릭 모방체일 수 있다. 펩티드 모방체는 UBAP2(Ubiquitin-Associated Protein 2) 단백질의 이차구조 특성과 유사하게 구조화되고 항체(Park, B. W. et al. Nat Biotechnol 18, 194-198, 2000) 또는 수용성 수용체(Takasaki, W. et al. Nat Biotechnol 15, 1266-1270, 1997)와 같은 거대한 분자의 억제 특성을 모방할 수 있으며, 천연의 길항제와 동등한 효과로 작용할 수 있는 신규한 소분자일 수 있다(Wrighton, N. C. et al. Nat Biotechnol 15, 1261-1265, 1997).
상기 앱타머(aptamer)는 단일 사슬 DNA 또는 RNA 분자로서, SELEX(systematic evolution of ligands by exponential enrichment)라 불리는 올리고 뉴클레오티드(oligonucleotide) 라이브러리를 이용한 진화적인 방법에 의해 특정 화학 분자나 생물학적 분자에 높은 친화력과 선별력을 갖고 결합하는 올리고머를 분리하여 수득할 수 있다(C. Tuerand L. Gold, Science 249, 505 - 510, 2005; A. D. Ellington and J. W. Szostak, Nature 346, 818 - 822, 1990; M. Famulok, et. al., Acc. Chem. Res. 33, 591 - 599, 2000; D. S. Wilson and Szostak, Annu. Rev. Biochem. 68, 611 - 647, 1999). 앱타머는 표적에 특이적으로 결합하고 표적의 활성을 조정할 수 있는데, 예컨대, 결합을 통하여 표적의 기능을 차단할 수 있다.
상기 항체는 ASM에 특이적이고 직접적으로 결합하여 ASM의 활성을 효과적으로 억제할 수 있다. 상기 ASM에 특이적으로 결합하는 항체로는 폴리클로날(polyclonal) 항체 또는 모노클로날(monoclonal) 항체를 사용하는 것이 바람직하다. 상기 ASM에 특이적으로 결합하는 항체는 당업자에게 알려진 공지의 방법으로 제작하여도 무방하며, 상업적으로 알려진 ASM 항체를 구입하여 사용할 수 있다. 상기 항체는 당업자에게 알려진 종래 방법에 따라 면역원인 ASM 단백질을 외부 숙주에 주사함으로써 제조될 수 있다. 외부 숙주는 마우스, 랫트, 양, 토끼와 같은 포유동물을 포함한다. 면역원은 근육 내, 복강 내 또는 피하 주사방법으로 주사되며, 일반적으로 항원성을 증가시키기 위한 보조제(adjuvant)와 함께 투여할 수 있다. 외부 숙주로부터 정기적으로 혈액을 채취하여 형성된 역가 및 항원에 대한 특이성을 보이는 혈청을 수거하여 항체를 분리할 수 있다.
본 발명에 따른 ASM 발현 억제제는 ASM 유전자 또는 ASM의 발현을 촉진하는 유전자의 mRNA에 상보적으로 결합하는 안티센스 뉴클레오티드, 짧은 헤어핀 RNA(small hairpin RNA; shRNA), 작은 간섭 RNA(small interfering RNA; siRNA) 및 리보자임(ribozyme)으로 구성된 군으로부터 선택된 1종 이상 일 수 있으며, 이에 제한되지 않는다.
상기 siRNA는 ASM 단백질을 암호화하는 유전자의 mRNA의 염기서열 내에서 선택되는 15 내지 30머(mer)의 센스 서열 및 상기 센스 서열에 상보적으로 결합하는 안티센스 서열로 구성되며, 이때, 상기 센스 서열은 특별히 이에 제한되는 것은 아니나, 약 25개의 염기로 구성되는 것이 바람직하다.
상기 안티센스 뉴클레오티드는 왓슨-크릭(Watson-Crick) 염기쌍에 정의된 바에 따라, DNA, 미성숙-mRNA 또는 성숙된 mRNA의 상보적 염기서열에 결합(혼성화)하여 DNA에서 단백질로서 유전정보의 흐름을 방해하는 것이다. 표적 서열에 특이성이 있는 안티센스 뉴클레오티드의 성질은 그것들을 예외적으로 다기능이 되도록 한다. 안티센스 뉴클레오티드는 모노머 단위의 긴 사슬이기 때문에 이들은 표적 RNA 서열에 대해 쉽게 합성될 수 있다. 최근 많은 연구들은 표적 단백질을 연구하기 위한 생화학적 수단으로 안티센스 뉴클레오티드의 유용성을 증명하였다(Rothenberg et al., J. Natl. Cancer Inst., 81:1539-1544, 1999). 올리고뉴클레오티드 화학 및 향상된 세포흡착, 표적결합의 친화도 및 뉴클레아제(nuclease) 내성을 나타내는 뉴클레오티드 합성 분야에서 최근 많은 진보가 있었으므로 안티센스 뉴클레오티드의 사용은 새로운 형태의 억제제로 고려될 수 있다.
본 발명에 따른 퇴행성 신경질환은 알츠하이머병, 파킨슨병, 진행성 핵상마비, 다계통 위축증, 감람핵-뇌교-소뇌 위축증(OPCA), 샤이-드래거 증후군, 선조체-흑질 퇴행증, 헌팅톤병, 근위축성 측색 경화증(ALS), 본태성 진전증, 피질-기저핵 퇴행증, 미만성 루이 소체 질환, 파킨스-ALS-치매 복합증, 픽병, 뇌허혈 및 뇌경색을 포함하나, 이에 제한되지 않는다.
본 발명의 조성물은 상기 ASM의 활성 억제제 또는 발현 억제제와 함께 ASM의 발현 또는 활성 억제 효과를 갖는 공지의 유효성분 또는 퇴행성 신경질환의 치료 효과를 갖는 공지의 유효성분을 1 종 이상 함유할 수 있다.
또한, 본 발명의 약학적 조성물은 목적하는 방법에 따라 경구 투여하거나 비경구 투여(예를 들어 정맥 내, 피하, 복강 내 또는 국소에 적용) 할 수 있으며, 투여량은 개체의 체중, 연령, 성별, 건강상태, 식이, 투여시간, 투여방법, 투여기간 또는 간격, 배설율, 체질 특이성, 제제의 성질 등에 따라 그 범위가 다양하다. 본 발명의 ASM의 발현 또는 활성 억제제의 일일 투여량은 약 0.001~1000 mg/kg, 바람직하게는 약 0.1~500 mg/kg이나, 임상실험결과에 따라 가감될 수 있으며, 하루 일회 내지 수회에 나누어 투여하는 것이 바람직하다.
본 발명의 약학적 조성물은 투여를 위하여 여러 가지 제제로 제형화될 수 있다. 본 발명에 포함될 수 있는 부형제는 비독성이고 불활성인 약제학적으로 적합한 고상, 준고상 또는 액상의 모든 유형의 제형 보조물로서, 예를 들면, 충진제, 중량제, 결합제, 습윤제, 붕해제, 분산제, 계면활성제 또는 희석제 등을 들 수 있다.
본 발명의 약학적 조성물은 정제, 피복 정제, 캡슐제, 환제, 과립제, 좌약, 액제, 현탁액제 및 에멀전제, 페이스트제 (pastes), 연고제, 겔제, 크림제, 로션제, 산제 또는 분무제로 제형화될 수 있다.
본 발명의 조성물은 퇴행성 신경질환의 개선을 위한 건강기능식품에 첨가될 수 있다. 본 발명의 ASM의 발현 억제제 또는 활성 억제제를 식품 첨가물로 사용할 경우, 상기 유효성분을 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용할 수 있고, 통상적인 방법에 따라 적절하게 사용할 수 있다. 유효성분의 혼합양은 사용 목적(예방, 건강 또는 치료적 처치)에 따라 적합하게 결정될 수 있다. 일반적으로, 식품 또는 음료의 제조 시 본 발명의 유효성분은 원료에 대하여 15 중량% 이하, 바람직하게는 10 중량% 이하의 양으로 첨가된다. 그러나 건강 및 위생을 목적으로 하거나 또는 건강 조절을 목적으로 하는 장기간의 섭취의 경우 상기 양은 상기 범위 이하일 수 있으며, 안전성 면에서 아무런 문제가 없기 때문에 유효성분은 상기 범위 이상의 양으로도 사용될 수 있다.
상기 식품의 종류에는 특별한 제한은 없다. 상기 물질을 첨가할 수 있는 식품의 예로는 육류, 소세지, 빵, 쵸코렛, 캔디류, 스넥류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차, 드링크제, 알콜 음료 및 비타민 복합제 등이 있으며, 통상적인 의미에서의 건강기능식품을 모두 포함한다.
본 발명의 건강음료 조성물은 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상술한 천연 탄수화물은 포도당 및 과당과 같은 모노사카라이드, 말토오스 및 수크로오스와 같은 디사카라이드, 덱스트린 및 시클로덱스트린과 같은 폴리사카라이드, 및 자일리톨, 소르비톨 및 에리트리톨 등의 당알콜이다. 감미제로서는 타우마틴, 스테비아 추출물과 같은 천연 감미제나, 사카린, 아스파르탐과 같은 합성 감미제 등을 사용할 수 있다. 상기 천연 탄수화물의 비율은 본 발명의 조성물 100 g당 일반적으로 약 0.01~0.20 g, 바람직하게는 약 0.04~0.10 g이다.
상기 외에 본 발명의 조성물은 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산 음료에 사용되는 탄산화제 등을 함유할 수 있다. 그 밖에 본 발명의 조성물은 천연 과일쥬스, 과일쥬스 음료 및 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 이러한 첨가제의 비율은 크게 중요하진 않지만 본 발명의 조성물 100 중량부 당 0.01~0.20 중량부의 범위에서 선택되는 것이 일반적이다.
또한, 본 발명은 상기 조성물의 치료적 유효량을 개체에 투여하는 단계;를 포함하는, 퇴행성 신경질환의 예방 또는 치료방법을 제공한다.
상기 치료적 유효량은 올바른 의학적 판단범위 내에서 결정될 수 있다는 것은 당업자에게 자명한 일이다. 특정 환자에 대한 구체적인 치료적 유효량은 달성하고자 하는 반응의 종류와 정도, 경우에 따라 다른 제제가 사용되는지의 여부를 비롯한 구체적 조성물, 환자의 연령, 체중, 일반 건강 상태, 성별 및 식이, 투여 시간, 투여 경로 및 조성물의 분비율, 치료기간, 구체적 조성물과 함께 사용되거나 동시 사용되는 약물을 비롯한 다양한 인자와 의약 분야에 잘 알려진 유사 인자에 따라 다르게 적용하는 것이 바람직하다. 따라서 본 발명의 목적에 적합한 조성물의 유효량은 전술한 사항을 고려하여 결정하는 것이 바람직하다.
또한, 경우에 따라, 본 발명의 조성물과 함께 공지의 퇴행성 신경질환 치료제를 병용 투여하여 퇴행성 신경질환 치료의 효과를 증대시킬 수 있다.
본 발명은 퇴행성 신경질환을 갖는 임의의 포유동물에 적용가능하고, 상기 포유동물은 인간 및 영장류뿐만 아니라, 소, 돼지, 양, 말, 개 및 고양이 등의 가축을 포함한다.
또한, 본 발명은
1) 생물학적 시료에 후보물질을 처리하는 단계; 및
2) 상기 1) 단계의 생물학적 시료에서 ASM(acid sphingomyelinase)의 mRNA 또는 단백질 발현양의 변화를 측정하는 단계;를 포함하는 퇴행성 신경질환의 예방 또는 치료물질의 스크리닝 방법을 제공한다.
상기 1) 단계의 생물학적 시료는 퇴행성 신경질환에 걸린 동물의 혈액, 소변, 타액 또는 조직 등을 포함하나, 이에 제한되지 않는다.
상기 2) 단계에서 mRNA 발현양의 변화를 측정하는 방법은 역전사 중합효소반응(RT-PCR), 경쟁적 역전사 중합효소반응(Competitive RT-PCR), 실시간 역전사 중합효소반응(Realtime RT-PCR), RNase 보호 분석법(RPA; RNase protection assay), 노던 블랏팅(Northern blotting) 및 DNA 칩 등을 포함하나, 이에 제한되지 않는다.
상기 2) 단계에서 단백질 발현양의 변화를 측정하는 방법은 웨스턴 블롯팅, ELISA(enzyme linked immunosorbent assay), 방사선면역분석(RIA: Radioimmunoassay), 방사 면역 확산법(radioimmunodiffusion), 오우크테로니(Ouchterlony) 면역확산법, 로케트(rocket) 면역전기영동, 조직면역 염색, 면역침전 분석법(Immunoprecipitation Assay), 보체 고정 분석법(Complement Fixation Assay), 유세포분석(Fluorescence Activated Cell Sorter, FACS) 및 단백질 칩(protein chip) 등을 포함하나, 이에 제한되지 않는다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
[실시예 1] ASM (acid sphingomyelinase) 돌연변이 마우스에서 ASM 억제의 알츠하이머병에 대한 치료 효과 확인
1. ASM 돌연변이 마우스의 제조
알츠하이머병의 실험 동물 모델인 APP/PS1(APP/presenilin) 이중 돌연변이(double mutant) 마우스 및 APP/PS1/ASM+/- 삼중 돌연변이 마우스(ASM의 부분적 유전적 결손)를 이용하여 실험을 수행하였다.
동물 실험은 IACUC(Kyungpook National University Institutional Animal Care and Use Committee)에서 승인을 받아 이루어졌다. C57BL/6 마우스(Charles River, UK)를 바탕으로 APPswe(hAPP695swe) 또는 PS1(presenilin-1M146V)을 과발현시킨 형질전환 마우스 라인을 이용하였다[이하, APP 마우스: APPswe를 과발현하는 마우스, PS1 마우스: presenilin-1M146V를 과발현하는 마우스; GlaxoSmithKline]. ASM+/- 마우스(한 쌍의 ASM 유전자 중 하나만 결손)와 APP/PS1 마우스를 교배시켜 APP/PS1/ASM+/- 마우스를 제조하였다. 그 구체적인 과정을 도 1에 나타내었다.
2. ASM 돌연변이 마우스에서 ASM 농도 수준 확인
9개월령의 야생형(WT) 마우스, APP/PS1 마우스 및 상기 실시예 1-1에서 제조한 APP/PS1/ASM+/- 마우스의 혈청, 뇌 조직 및 섬유아세포에서 ASM의 농도 수준을 측정하였다. 보다 구체적으로, 각 마우스의 혈청, 뇌 조직 및 섬유아세포 시료 3 ㎕를 ASM 활성 완충액과 혼합하여 37 ℃에서 보관하였다. 상기 혼합액에 114 ㎕의 에탄올을 가하여 가수분해 반응을 종료시킨 후, 원심분리하였다. 30 ㎕의 상층액을 유리 바이알에 옮긴 후, 5 ㎕를 UPLC 시스템에 적용시켰다. ASM 농도 수준은 스핑고미엘린 및 세라마이드와 결합된 Bodipy(aminoacetaldehyde)와 비교함으로써 정량화하였다. 상기 스핑고미엘린 및 세라미이드의 추출 및 정량화는 공지된 방법에 따라 시료에서 지질을 추출하고, 상기 건조된 지질 추출물을 25 ㎕의 0.2% Igepal CA-630(Sigma-Aldrich)에 재부유시키고, 각각의 지질의 농도 수준을 UPLC 시스템을 이용하여 정량화함으로써 수행되었다. 그 결과를 도 2(왼쪽: 혈장; 중앙: 뇌 조직; 오른쪽: 섬유아세포)에 나타내었다.
도 2에 나타낸 바와 같이, APP/PS1 마우스의 혈장, 뇌 조직 및 섬유아세포에서의 ASM 농도의 수준과 비교하여, APP/PS1/ASM+/- 마우스의 혈장, 뇌 조직 및 섬유아세포에서의 ASM 농도의 수준이 현저히 낮음을 확인하였다.
3. ASM 돌연변이 마우스에서 뇌 조직 내 β-아밀로이드 침착 억제 확인
상기 실시예 1-2에서 확인한 바와 같이, APP/PS1/ASM+/- 마우스에서 ASM 농도 수준의 감소가 알츠하이머병의 병리학적인 측면에서 어떠한 영향을 미치는지 확인하기 위하여, 뇌 조직 내 β-아밀로이드 침착 정도를 분석하였다.
먼저, 실시예 1-1에서 제조된 각 마우스의 대뇌피질 및 해마 조직을 분리한 후, 조직 절편을 수득하고 종래 공지된 방법에 따라 티오플라빈 S로 염색하였다. 그 결과를 도 3(대뇌피질) 및 도 4(해마)에 나타내었다.
도 3 및 도 4에 나타낸 바와 같이, APP/PS1 마우스의 뇌 조직과 비교하여, APP/PS1/ASM+/- 마우스의 뇌 조직에 침착된 Aβ40 및 Aβ42가 현저히 감소함을 확인하였다.
또한, Aβ42에 대한 항-20G10(마우스, 1:1000) 및 Aβ40에 대한 항-G30(토끼, 1:1000), 항-Iba-1(토끼, 1:500, Wako), 항-GFAP(토끼, 1:500, DAKO) 및 항-활성 카스파제3(토끼, 1:50, Chemicon) 항체를 이용하여 종래 공지된 방법에 따라 면역형광법을 수행하였다. Fluoview SV1000 이미징 소프트웨어(Olympus FV1000, Japan)를 장착한 레이저 스캐닝 공초점 현미경 또는 Olympus BX51 현미경을 이용하여 조직 절편을 관찰하였으며, Metamorph software(Molecular Devices)를 이용하여 총 조직의 넓이에 대한 염색된 부위의 넓이의 퍼센트를 정량화하였다.
또한, 상업적으로 이용가능한 ELISA 키트(Biosource)를 이용하여 β-아밀로이드 침착을 확인하였다. 보다 구체적으로, 각 마우스의 뇌의 반구를 0.02 M의 구아니딘(guanidine)을 포함하는 완충액에서 균질화하였다. 이 후, 제조사의 메뉴얼에 따라, Aβ40 및 Aβ42에 대한 ELISA를 수행하였다.
그 결과를 도 5(면역형광법) 및 도 6(ELISA)에 나타내었다.
도 5 및 도 6에 나타낸 바와 같이, APP/PS1 마우스의 뇌 조직과 비교하여, APP/PS1/ASM+/- 마우스의 뇌 조직에 침착된 Aβ40 및 Aβ42가 현저히 감소함을 확인하였다.
4. ASM 돌연변이 마우스에서 학습 및 기억능력 향상 확인
상기 실시예 1-1에서 제조한 APP/PS1/ASM+/- 마우스에서 학습 및 기억능력 향상 효과가 있는지를 확인하기 위하여, 종래 공지된 방법에 따라 MWM(Morris water maze) 테스트를 수행하였다.
보다 구체적으로, 야생형 마우스, APP/PS1/ASM+/- 마우스, ASM+/- 마우스 및 APP/PS1 마우스를 이용하였으며, 10일 동안 하루에 4 번씩 과제를 학습시키고, 11일째 되는 날에 플랫폼을 제거하고, 탐침시험(probe trial)을 수행하였다. 실험 기간 동안의 탈출 시간 및 11일 째 되는 날 표적 플랫폼에서 보내는 시간을 측정하였다. 그 결과를 도 7 및 도 8에 나타내었다.
도 7 및 도 8에 나타낸 바와 같이, APP/PS1 마우스는 실험 기간 동안 탈출 시간에 변화가 없고, 표적 플랫폼과 비 표적 플랫폼에서 보내는 시간에도 차이가 없는 등 공간 기억 형성에 장애가 나타남을 확인하였으나, APP/PS1/ASM+/- 마우스는 야생형 마우스와 유사한 수준으로 학습 및 기억능력이 향상됨을 확인하였다.
상기 MWM 테스트 결과를 검증하기 위하여, 종래 공지된 방법에 따라 환경적 맥락 또는 조건자극을 전기쇼크와 연합시켜 학습과 기억을 평가하는 공포 조건화 (fear conditioning) 테스트를 수행하였다.
보다 구체적으로, 야생형 마우스, APP/PS1/ASM+/- 마우스, ASM+/- 마우스 및 APP/PS1 마우스를 이용하였으며, 실험 첫째 날, 각 마우스를 개별적으로 조건화 챔버에 위치시킨 후 60 초간의 탐험 시간이 지나고, 10 초간 소리자극(tone, 10 kHz, 70 dB)을 주었다. 이를 조건 자극(conditioned stimulus, CS)이라고 하였다. CS는 무조건 자극(unconditioned stimulus, US)인 전기적인 발바닥 쇼크(0.3 mA, 1 s)와 함께 종결되었다. CS-US는 짝지어 20초 간격으로 2회 시행되었다. 실험 둘째 날, 각 마우스를 CS 또는 발바닥 쇼크의 집행이 없는 공포-조건화 챔버에 다시 위치시킨 후, 5분간 동결반응(freezing response)을 관찰하였다. 실험 셋째 날, 각 마우스를 조건화 챔버와는 다른 실험 챔버에 위치시킨 후 60 초간의 탐험 시간이 지나고, 발바닥 쇼크 없이 60초간 소리 자극을 주었다. 공포에 대한 기억을 측정하기 위하여 동결 반응을 측정하였다. 그 결과를 도 9 및 도 10에 나타내었다.
도 9 및 도 10에 나타낸 바와 같이, APP/PS1/ASM+/- 마우스는 APP/PS1 마우스에 비해 기억능력이 향상됨을 확인하였다.
[실시예 2] ASM (acid sphingomyelinase) 돌연변이 마우스에서 ASM 억제가 자가포식작용(autophagy)에 미치는 영향 확인
1. ASM 억제에 의한 자가포식작용 관련 유전자 발현 확인
유전적인 ASM 억제가 알츠하이머병의 자가포식작용 관련 경로에서 어떻게 작용하는지 확인하기 위하여, 9개월령의 야생형 마우스, APP/PS1 마우스 및 상기 실시예 1-1에서 제조한 APP/PS1/ASM+/- 마우스의 꼬리 섬유아세포 및 뇌 조직 시료를 분석하였다.
보다 구체적으로, LC3 (rabbit, 1:1000, Cell Signaling Technologies, 4108S) Beclin-1 (rabbit, 1:1000, Cell Signaling Technologies, 3738S), p62 (rabbit, 1:1000, Cell Signaling Technologies, 5114S), cathepsin D (goat, 1:500, R&D Systems, BAF1029) 및 β-actin (1:1000, Santa Cruz, SC-1615) 항체를 이용하여 종래 공지된 방법에 따라 웨스턴 블롯팅을 수행하였으며, ImageJ 소프트웨어(US National Institutes of Health)를 이용하여 밀도 정량화(densitometric quantification)를 수행하였다. 그 결과를 도 11 내지 도 14에 나타내었다.
도 11 내지 도 14에 나타낸 바와 같이, WT 마우스와 비교하여, APP/PS1 마우스에서 LC3-Ⅰ에서 LC3-Ⅱ로의 전환이 증가함을 확인하였고, APP/PS1/ASM+/- 마우스에서 증가된 LC3-Ⅱ의 발현이 감소함을 확인하였다. 베클린-1의 발현에는 각 군간 유의한 차이가 나타나지 않았다. 또한, 자가포식작용 턴오버(turnover)의 지표인 카텝신 D(lysosomal hydrolase) 및 p62의 발현은 알츠하이머 환자에서 증가하여 알츠하이머와 병리학적으로 관련이 있는데, WT 마우스와 비교하여, APP/PS1 마우스에서 카텝신 D 및 p62의 발현이 증가함을 확인하였고, APP/PS1/ASM+/- 마우스에서 상기 증가된 카텝신 D 및 p62의 발현이 감소함을 확인하였다.
2. ASM 억제에 의한 단백질 분해 활성 평가
9개월령의 야생형 마우스, APP/PS1 마우스 및 상기 실시예 1-1에서 제조한 APP/PS1/ASM+/- 마우스의 꼬리 섬유아세포에서 단백질 분해 활성을 확인하였다.
수명이 긴 단백질(long-lived protein)을 표지시키기 위하여 48시간 동안 [3H]-류신(leucine; 2μCi/ml)을 이용하여 펄스를 주어 펄스-채이스 실험(Pulse-chase experiment)을 수행하였다. 표지화된 세포를 세척하고, 이를 자가포식작용이 억제되는 환경의 완전 배지(complete medium) 또는 자가포식작용이 유도되는 혈청(serum)이 결핍된 배지에서 배양하였다. 배지의 분취량 (Aliquots)을 서로 다른 시점에서 수거하여 10% TCA로 침전시킨 후, 0.22 μm의 구멍을 갖는 막에서 여과하였고, 방사성 활성을 측정하여 단백질 분해 활성을 분석하였다. 그 결과를 도 15에 나타내었다.
도 15에 나타낸 바와 같이, 자가포식작용(혈청이 결핍된 배지에서 배양)이 유도되면, WT 마우스에서 유래한 세포에 비하여, APP/PS1/ASM+/- 마우스로부터 유래한 세포에서 단백질 분해가 높아짐을 확인하였다.
3. 투과전자현미경(TEM)을 이용한 마우스 뇌 조직 분석
9개월령의 야생형 마우스, APP/PS1 마우스 및 상기 실시예 1-1에서 제조한 APP/PS1/ASM+/- 마우스의 뇌 조직을 3%의 글루타르알데히드(glutaraldehyde)가 포함된 0.1M, pH 7.4의 인산염 완충액으로 고정시킨 후, 오스뮴 테트록사이드(osmium tetroxide)를 포함하는 Sorensen`s 인산염 완충액에서 추가적으로 고정하였다. 상기 조직을 에틸 알코올로 탈수시킨 후, Epon(Electron Microscopy Sciences)에 끼어넣었다(embbeded). 상기 조직을 순차적으로 자른 후, 투과전자현미경(Transmission Electron Microscope; Tecnai)을 이용하여 분석하였다. 디지털 카메라 및 Xplore3D tomography 소프트웨어를 이용하여 이미지를 수득하였다. 그 결과를 도 16에 나타내었다.
도 16에 나타낸 바와 같이, APP/PS1 마우스의 뇌 조직에서 자가포식액포(autophagic vacuole; AV)의 크기 및 수가 증가됨을 확인하였고, APP/PS1/ASM+/- 마우스의 뇌 조직에서는 WT 마우스보다는 다소 증가된 크기 및 수의 액포가 관찰되었으나, APP/PS1 마우스보다는 작은 크기 및 적은 수의 액포가 관찰됨을 확인하였다.
[실시예 3] 인간 세포에서
ASM 억제의 자가포식작용(autophagy)에 대한 영향 확인
1. 인간의 섬유아세포에서 재조합 ASM 단백질에 의한 자가포식작용 관련 유전자 발현 변화 확인
인간 섬유아세포주는 Coriell Institute에서 수득하였으며, 15% FBS를 포함하는 DMEM 배지에서 37℃ 및 5%의 CO2에서 배양하였다. 상기 세포주에 재조합 ASM(1 μM 내지 10 μM)을 처리한 후, 실시예 2-1과 같은 방법으로 웨스턴 블롯팅 및 밀도 정량화를 수행하였다. 그 결과를 도 17 및 도 18에 나타내었다.
도 17 및 도 18에 나타낸 바와 같이, 재조합 ASM의 처리 농도에 의존적으로 LC3-Ⅰ에서 LC3-Ⅱ로의 전환이 진행됨을 확인하였으며, 베클린-1의 발현에는 유의한 변화가 없음을 확인하였다.
2. M6P(mannose-6-phosphate)를 이용한 ASM의 작용 기전 확인
ASM이 어떠한 경로를 통하여 자가포식작용에 영향을 주는지 확인하기 위하여, 하기와 같은 실험을 수행하였다. 인간 섬유아세포에 ASM을 단독으로 처리하거나, 단백질을 리소좀에 위치시키는 작용과 관련 있는 M6P(mannose-6-phosphate; 10 mM)의 존재 하에서 처리한 후, 자가포식소체(autophagosome)의 축적을 웨스턴 블롯팅 및 밀도 정량을 이용하여 확인하였다. 그 결과를 도 19에 나타내었다.
도 19에 나타낸 바와 같이, M6P를 이용하여 리소좀 ASM의 흡수를 억제하는 경우, LC3-Ⅱ로의 전환이 감소하여 ASM-유도 자가포식소체의 축적이 감소됨을 확인하였다.
ASM이 자가포식소체의 형성 속도를 증가시키는 것인지, 자가포식소체의 분해 속도를 감소시키는 것인지 확인하기 위하여, autophagic flux assay를 수행하였다. 보다 구체적으로, 자가포식소체의 분해를 억제하지만, 자가포식소체의 형성에는 영향을 주지 않는 NH4Cl의 존재 또는 비 존재하의 세포에서 LC3-Ⅰ에서 LC3-Ⅱ로의 전환 비율을 웨스턴 블롯팅을 이용하여 측정하였다. 그 결과를 도 20에 나타내었다.
도 20에 나타낸 바와 같이, 세포에 ASM 및 NH4Cl를 처리하는 경우에 LC3-Ⅱ의 양에 유의적인 변화가 없음을 확인하였다.
또한, 혈청이 없는 배지 또는 완전 배지에서 키운 알츠하이머병에 걸린 인간 섬유아세포를 배양한 후, NH4Cl또는 ASM을 처리한 경우에 LC3-Ⅰ에서 LC3-Ⅱ로의 전환 비율을 웨스턴 블롯팅을 이용하여 측정하였다. 그 결과를 도 21 및 도 22에 나타내었다.
도 21에 나타낸 바와 같이, 혈청이 없는 배지에서 세포를 배양하여 자가포식작용을 유도한 후, NH4Cl를 가하면 LC3-Ⅱ의 수준이 유의적으로 증가함을 확인하였으며, 도 22에 나타낸 바와 같이, 혈청 없는 배지에서 세포를 배양하여 자가포식작용을 유도한 후, ASM을 처리하는 경우, LC3-Ⅱ의 축적이 유의적으로 증가함을 확인하였다.
이러한 실험 결과를 통하여, 알츠하이머병에서 ASM이 자가포식소체의 형성을 유발하는 것이 아니라, 자가포식소체의 단백질 분해를 억제함을 확인하였다.
[실시예 4] 알츠하이머병 모델 마우스에서 ASM 억제의 병리학적인 개선 효과 검증
1. APP/PS1 마우스에 AMI 투여 후 알츠하이머병 치료 효과 검증
알츠하이머병 모델 마우스인 APP/PS1 마우스에 ASM의 알려진 억제제이며, BBB를 통과할 수 있다고 알려진 AMI(amitriptyline-hydrochloride)를 4개월간 투여한 후, WM(water maze) 테스트를 수행하였다(도 23). 마우스가 9개월령이 되었을 때 혈청 및 뇌 조직을 수득하여 ASM의 발현 정도를 측정하였으며(실시예 1-2와 동일한 방법), 뇌 조직 내 β-아밀로이드의 침착 정도를 측정하였다(실시예 1-3과 동일한 방법). 각 실험 결과를 도 24 내지 26에 나타내었다.
도 24에 나타낸 바와 같이, AMI의 투여를 통해 마우스의 혈청 및 뇌 조직에서 ASM이 감소되었음을 확인하였으며, 도 25에 나타낸 바와 같이, AMI를 투여한 마우스의 대뇌 피질 및 해마에서 β-아밀로이드 침착 정도가 억제됨을 확인하였다. 또한, 도 26에 나타낸 바와 같이, AMI를 투여한 마우스가 대조군에 비해 탈출 시간(escape latency)이 감소하여 기억력이 회복됨을 확인하였다.
2. 병체 결합 시스템(parabiotic system)을 이용한 알츠하이머병 치료 효과 검증
APP/PS1 마우스의 병체 결합 시스템(parabiotic system)을 이용하여 ASM 억제의 효과를 확인하였다. 보다 구체적으로, 마우스들을 수술적 기법으로 피부 및 연조직을 연결하고 새로운 신생혈관이 두 마우스 사이에 생기게 유도한 후 혈류를 공유하게 하여, 아이소크로닉(isochronic; APP/PS1-APP/PS1: 알츠하이머병 모델 마우스인 APP/PS1 마우스끼리 병체결합), 헤테로크로닉 Ⅰ(heterochronic Ⅰ;APP/PS1-ASM+/-: APP/PS1 마우스와 ASM+/-마우스간의 병체결합), 헤테로크로닉 Ⅱ(heterochronic Ⅱ; APP/PS1-WT: APP/PS1 마우스와 야생형 마우스간의 병체 결합) 마우스를 제조하였다 (도 27). 상기 실시예 4-1과 동일한 방법으로 혈청 및 뇌 조직을 수득하여 ASM의 발현 정도를 측정하였으며, 뇌 조직 내 β-아밀로이드 침착 정도 및 단백질 발현 정도를 측정하였다. 각 실험 결과를 도 28 내지 30에 나타내었다.
도 28에 나타낸 바와 같이, 아이소크로닉(isochronic; APP/PS1-APP/PS1) 및 헤테로크로닉 Ⅱ(APP/PS1-WT) 마우스에 비하여, 헤테로크로닉 Ⅰ(APP/PS1-ASM+/-)마우스의 혈청 및 뇌 조직에서 ASM의 농도가 낮음을 확인하였으며, 이를 통해 ASM+/- 마우스가 ASM의 농도를 낮추는 역할을 함을 확인하였다.
또한, 도 29에 나타낸 바와 같이, 아이소크로닉(APP/PS1-APP/PS1) 마우스에 비하여 헤테로크로닉 Ⅰ(APP/PS1-ASM+/-)마우스의 대뇌 피질 및 해마에서 β-아밀로이드 침착이 현저히 감소함을 확인하였다.
또한, 도 30에 나타낸 바와 같이, 아이소크로닉(APP/PS1-APP/PS1) 마우스에 비하여 헤테로크로닉 Ⅰ(APP/PS1-ASM+/-)마우스의 뇌 조직에서 LC3-Ⅱ로의 전환이 감소하였고, p62 및 카텝신 D의 발현이 감소함을 확인하였으며, ALP 기능과 관련된 단백질인 TFEB 및 Lamp1의 발현이 증가함을 확인하였다.
3. 혈청주입을 이용한 알츠하이머병 치료 효과 검증
APP/PS1 또는 ASM-/- 마우스의 혈청을 APP/PS1 마우스에 주입하였다. 보다 구체적으로, APP/PS1 또는 ASM-/- 마우스의 심장으로부터 혈액을 수득한 후 EDTA로 코팅된 튜브에 수집하였다. 수집된 혈액을 원심분리하여 혈청을 수득하였으며, 혈청 100 μl를 3주 동안 총 8번, 8개월령의 APP/PS1 마우스의 정맥 내에 주입하였다(도 31). 실험 종료 후, 상기 실시예 4-1과 동일한 방법으로 혈청 및 뇌 조직을 수득하여 ASM의 발현 정도를 측정하였으며, 뇌 조직 내 β-아밀로이드 침착 정도 및 단백질 발현 정도를 측정하였다. 또한, 상기 실시예 1-4와 동일한 방법으로 행동 실험을 수행하였다. 각 실험 결과를 도 32 내지 36에 나타내었다.
도 32에 나타낸 바와 같이, APP/PS1 마우스의 혈청을 제공받은 APP/PS1 마우스에 비하여, ASM-/- 마우스의 혈청을 제공받은 APP/PS1 마우스의 혈청 및 뇌 조직에서 ASM이 감소되었음을 확인하였다.
도 33 및 도 34에 나타낸 바와 같이, APP/PS1 마우스의 혈청을 제공받은 APP/PS1 마우스에 비하여, ASM-/- 마우스의 혈청을 제공받은 APP/PS1 마우스의 뇌 조직 내 β-아밀로이드 침착이 감소하였으며(도 33), LC3-Ⅱ로의 전환이 감소하였고, p62 및 카텝신 D의 발현이 감소함을 확인하였다(도 34).
도 35 및 도 36에 나타낸 바와 같이, MWM 및 공포 조건화 테스트 결과, APP/PS1 마우스의 혈청을 제공받은 APP/PS1 마우스에 비하여, ASM-/- 마우스의 혈청을 제공받은 APP/PS1 마우스에서 기억력이 향상됨을 확인하였다.
이하 본 발명의 약학적 조성물 및 식품 조성물의 제제예를 설명하나, 본 발명을 한정하고자 함이 아닌 단지 구체적으로 설명하고자 함이다.
제제예 1. 약학적 제제의 제조

상기의 비타민 및 미네랄 혼합물의 조성비는 비교적 건강식품에 적합한 성분을 바람직한 실시예로 혼합 조성하였지만, 그 배합비를 임의로 변형 실시하여도 무방하며, 통상의 건강식품 제조방법에 따라 상기의 성분을 혼합한 다음, 과립을 제조하고, 통상의 방법에 따라 건강식품 조성물 제조에 사용할 수 있다.
Claims (10)
- ASM(acid sphingomyelinase)의 활성 억제제 또는 발현 억제제를 유효성분으로 포함하는 퇴행성 신경질환의 예방 또는 치료용 약학적 조성물.
- 제 1항에 있어서, 상기 ASM 활성 억제제는 ASM 단백질에 특이적으로 결합하는 화합물, 펩티드, 펩티드모방체, 기질유사체, 앱타머, 및 항체로 이루어진 군으로부터 선택된 1종 이상인 것을 특징으로 하는, 퇴행성 신경질환의 예방 또는 치료용 약학적 조성물.
- 제 1항에 있어서, 상기 ASM 발현 억제제는, ASM 유전자의 mRNA에 상보적으로 결합하는 안티센스 뉴클레오티드, siRNA, shRNA 및 리보자임으로 이루어진 군으로부터 선택된 1종 이상인 것을 특징으로 하는, 퇴행성 신경질환의 예방 또는 치료용 약학적 조성물.
- 제 1항 내지 제 3항 중 어느 한 항에 있어서, 상기 퇴행성 신경질환은 알츠하이머병, 파킨슨병, 진행성 핵상마비, 다계통 위축증, 감람핵-뇌교-소뇌 위축증(OPCA), 샤이-드래거 증후군, 선조체-흑질 퇴행증, 헌팅톤병, 근위축성 측색 경화증(ALS), 본태성 진전증, 피질-기저핵 퇴행증, 미만성 루이 소체 질환, 파킨스-ALS-치매 복합증, 픽병, 뇌허혈 및 뇌경색으로 구성된 군으로부터 선택된 1종 이상인 것을 특징으로 하는 퇴행성 신경질환의 예방 또는 치료용 약학적 조성물.
- ASM(acid sphingomyelinase)의 활성 억제제 또는 발현 억제제를 유효성분으로 포함하는 퇴행성 신경질환의 예방 또는 개선용 식품 조성물.
- 제 1항 내지 제 3항 중 어느 한 항에 따른 조성물의 치료적 유효량을 개체에 투여하는 단계;를 포함하는, 퇴행성 신경질환의 예방 또는 치료방법.
- 1) 생물학적 시료에 후보물질을 처리하는 단계; 및2) 상기 1) 단계의 생물학적 시료에서 ASM(acid sphingomyelinase)의 mRNA 또는 단백질 발현양 변화를 측정하는 단계;를 포함하는 퇴행성 신경질환의 예방 또는 치료물질의 스크리닝 방법.
- 제 7항에 있어서, 상기 1) 단계의 생물학적 시료는 퇴행성 신경질환에 걸린 동물의 혈액, 소변, 타액 및 조직으로 이루어진 군으로부터 선택된 1종 이상인 것을 특징으로 하는, 퇴행성 신경질환의 예방 또는 치료물질의 스크리닝 방법.
- 제 7항에 있어서, 상기 2) 단계에서 mRNA 발현양 변화는 역전사 중합효소반응(RT-PCR), 경쟁적 역전사 중합효소반응(Competitive RT-PCR), 실시간 역전사 중합효소반응(Realtime RT-PCR), RNase 보호 분석법(RPA; RNase protection assay), 노던 블랏팅(Northern blotting) 및 DNA 칩으로 이루어진 군으로부터 선택된 1 종 이상의 방법을 이용하여 측정하는 것을 특징으로 하는, 퇴행성 신경질환의 예방 또는 치료물질의 스크리닝 방법.
- 제 7항에 있어서, 상기 2) 단계에서 단백질 발현양 변화는 웨스턴 블롯팅, ELISA (enzyme linked immunosorbent assay), 방사선면역분석(RIA: Radioimmunoassay), 방사 면역 확산법(radioimmunodiffusion), 오우크테로니(Ouchterlony) 면역확산법, 로케트(rocket) 면역전기영동, 조직면역 염색, 면역침전 분석법(Immunoprecipitation Assay), 보체 고정 분석법(Complement Fixation Assay), 유세포분석(Fluorescence Activated Cell Sorter, FACS) 및 단백질 칩(protein chip)으로 이루어진 군으로부터 선택된 1종 이상의 방법을 이용하여 측정하는 것을 특징으로 하는, 퇴행성 신경질환의 예방 또는 치료물질의 스크리닝 방법.
