Description
本発明は、医療分野において有用な、成熟した樹状細胞を含む細胞集団を製造する方法に関する。
樹状細胞(以下、「DC」ともいう)は、樹枝状の突起を伸展させていることを特徴とし免疫応答に関わる細胞の総称である。DCは、主要組織適合遺伝子複合体(MHC)クラスII陽性の抗原提示細胞であり、生体内の免疫系で重要な役割を持っている。DCは造血幹細胞よりミエロイド系やリンパ球系分化経路を経て未成熟DCへ分化し、そして成熟DCに至る。生体において、DCは分化系列(ミエロイド系及びリンパ球系)や成熟段階(未成熟及び成熟)の異なる多様なサブセットとして、末梢組織やリンパ組織に広く存在している。微生物、ウイルス、異物等の外来抗原に侵襲された末梢炎症組織では、未成熟DCがマンノースレセプターやtoll様レセプター(toll-like receptor(TLR))等のパターン認識レセプター(pattern-recognition receptor(PRR))により外来抗原を捕食して成熟DCへ分化し、所属二次リンパ組織に移行する。さらに、成熟DCはナイーブT細胞に抗原刺激と共刺激を与え、抗原特異的エフェクターT細胞を分化誘導し、一次免疫応答を惹起する。このようにDCは自然免疫と獲得免疫を繋ぐ最も強力な抗原提示細胞である。その臨床応用例として成熟DCを用いた癌ワクチン療法の臨床試験が進められている。
成熟DCは白血球の1%にも満たないため、成熟DCを末梢血から単離することは困難である。また、成熟DCを組織から抽出することも困難である。したがって、末梢血から未成熟DC及び成熟DCを作製する方法の研究が進められている。これらの方法は、出発材料としてCD34陽性造血前駆細胞を使用する方法と、CD14陽性単球を使用する方法に大きく分けられる。これらの細胞を、顆粒球単球コロニー刺激因子(GM-CSF)及びインターロイキン(IL)-4などのサイトカインにより未成熟DCに分化させ、次いで未成熟DCをDC成熟化因子(エンドトキシンや炎症性サイトカイン)により成熟DCに成熟させる2段階の方法が多数報告されている(非特許文献1)。
細胞の周りや細胞と細胞の間には、細胞外マトリックス(ECM)と総称される巨大なタンパク質の超分子複合体が存在し、細胞→組織→器官→個体という階層的な細胞社会の形成を制御している。また、細胞外マトリックスは、様々な増殖因子や分化誘導因子と同様、細胞の増殖・分化の制御に直接かかわっていることが近年明らかにされている。DCに対する細胞外マトリックスの作用についても、種々検討が行われている。例えば、I型コラーゲン及びフィブリノーゲンは未成熟DCを成熟化させることが報告されている。一方、フィブロネクチン(FN)はDCの成熟化に効果は無く、むしろ成熟DCのマーカー遺伝子の発現を抑制することが報告されている(非特許文献2、3)。また、FNフラグメントで刺激した単球は、抗CD3抗体により活性化したT細胞の増殖を抑制することが報告されている(非特許文献4)。
以上の通り、DCの成熟化をより促進し、かつエフェクターT細胞による免疫応答をより惹起する成熟DCの製造方法が求められているものの、効率的に多くの成熟DCを調製するのに十分な方法はいまだ確立されていない。

本発明の目的は、成熟DCを含有する細胞集団の製造方法、及びその方法により製造された細胞集団、さらにその細胞集団を含む医薬を提供することにある。
本発明者らは、未成熟DCを含有する細胞集団をDC成熟化因子及びFNフラグメントの存在下で培養することにより、DCの成熟化が促進され、エフェクターT細胞による免疫応答を惹起するサイトカインの産生が促進されることを見出し、本発明を完成するに至った。
すなわち本発明を概説すれば、

本発明の方法を用いれば、T細胞の活性化をはじめ、免疫反応を強く惹起する成熟DCを効率よく製造することができる。当該方法により製造された成熟DCを含む細胞集団は、免疫細胞療法、癌ワクチン療法等に有用である。
本明細書において「樹状細胞(Dendritic Cell)」とは、樹枝状の突起を伸展させていることを特徴とし強力な抗原提示機能を有する細胞の一群を意味する。「未成熟(Immature)DC」は成熟DCの前駆細胞を意味し、CD14陰性、CD83陽性、CD86陽性、MHCクラスII陽性であるDCを意味する。また、「成熟(Mature)DC」は、CD14陰性、CD83強陽性、CD86強陽性、MHCクラスII強陽性であるDCを意味する。表1にヒト単球及びヒトDCが発現している表面マーカーを例示する。なお、「陽性」は、「強陽性(bright)」及び「弱陽性(dim)」を含む。
本明細書において「フィブロネクチン(FN)フラグメント」とは、分子内にフィブロネクチン由来のアミノ酸配列の一部を含有するポリペプチドを意味する。FNフラグメントは、天然由来の単離されたポリペプチド、人為的に合成されたポリペプチド、又は遺伝子工学的に調製された組換えポリペプチドのいずれでもよい。
本発明に使用できるFNフラグメント、並びに該フラグメントの調製に関する有用な情報は、ジャーナル・オブ・バイオケミストリー(J.Biochem.)、第110巻、第284~291頁(1991)、EMBO ジャーナル(EMBO J.)、第4巻、第7号、第1755~1759頁(1985)、及びバイオケミストリー(Biochemistry)、第25巻、第17号、第4936~4941頁(1986)、国際公開第2007/142300号パンフレット等より得ることができる。また、フィブロネクチンをコードする核酸配列及びフィブロネクチンのアミノ酸配列は、Genbank Accession No.NM_002026、NP_002017にて開示されている。
FNのドメイン構造は7つに分けられており、またそのアミノ酸配列中には3種類の類似の配列が含まれている。これらの各配列の繰り返しで全体が構成されている。3種類の類似の配列は、I型、II型及びIII型とそれぞれ呼ばれ、このうち、III型は71~96個のアミノ酸残基で構成されており、これらのアミノ酸残基の一致率は17~40%である。FN中には14個のIII型の配列が存在するが、そのうち、1番目、2番目及び3番目(それぞれIII1、III2及びIII3と称する)はフィブロネクチン結合ドメインに、8番目、9番目及び10番目(それぞれIII8、III9及びIII10と称する)は細胞接着ドメインに、また12番目、13番目及び14番目(それぞれIII12、III13及びIII14と称する)はヘパリン結合ドメインに含有されている。また、ヘパリン結合ドメインのC末端側に、IIICSと呼ばれる領域が存在する。IIICSには、25アミノ酸からなるVLA-4に対して結合活性を有するCS-1と呼ばれる領域が存在する。
以下、本発明を具体的に説明する。
(1)成熟したDCを含有する細胞集団の製造方法
本発明の成熟したDCを含有する細胞集団の製造方法は、未成熟のDCを含有する細胞集団を、DC成熟化因子及びFNフラグメントの存在下で培養する工程を包含することを特徴とする。
(1)成熟したDCを含有する細胞集団の製造方法
本発明の成熟したDCを含有する細胞集団の製造方法は、未成熟のDCを含有する細胞集団を、DC成熟化因子及びFNフラグメントの存在下で培養する工程を包含することを特徴とする。
本発明に使用できるFNフラグメントは、FN結合ドメイン、細胞接着ドメイン又はヘパリン結合ドメインに含まれるフラグメントから選択される。例えば、III1、III2、III3、III7、III8、III9、III11、III12、III13及びCS-1から選択される少なくとも1つのフラグメントを含めばよく、さらに複数のドメインが繰り返し連結されたフラグメントであってもよい。例えば、VLA-5へのリガンドを含む細胞接着ドメイン、ヘパリン結合ドメイン、VLA-4へのリガンドであるCS-1ドメイン、III1等を含有するフラグメントが本発明に使用される。前記フラグメントとしては、例えば、前記のJ.Biochem.、第110巻、第284~291頁(1991)に記載されたCH-271、CH-296、H-271、H-296、並びにこれらの誘導体や改変物が例示される。前記のCH-296はレトロネクチン(登録商標)の名称で市販されている。また、III1のC末端側の2/3のポリペプチドがFibronectin Fragment III1-Cの名称で市販されている。
FNフラグメントは、培地中に溶解して使用してもよく、適切な固相、例えば、細胞培養器材、ビーズ、メンブレン、スライドガラス等の細胞培養用担体に固定化して使用してもよい。固相へのFNフラグメントの固定化は、例えば、国際公開第00/09168号パンフレットに記載の方法に従って実施することができる。本発明において使用するFNフラグメントの濃度は、特に限定はなく、例えば最終濃度が0.001~500μg/mL、好適には0.01~500μg/mLとなるよう培地に添加する。また、FNフラグメントを固定化して使用する場合は、前記濃度のFNフラグメント溶液を使用して固相への固定化を実施すればよい。FNフラグメント存在下での細胞集団の培養は、国際公開第03/080817号パンフレットに詳細に記載されており、これを参照して実施することができる。
DC成熟化因子は、DCの成熟を促進する物質であればいずれも使用できるが、例えば、1)菌体製剤、2)CD40アゴニスト、3)腫瘍壊死因子(TNF)-α及びIL-1βを含むDC成熟化カクテル(MC:Maturation Cocktail)から選択される少なくとも1つの生物応答修飾剤を使用することができる。
本発明に使用する菌体製剤は、例えば、細菌由来製剤[OK-432、BCG(Bacillus Calmette Guerin)、Streptcoccus pyogenes、Corynebacterium parvum及びこれらの細胞壁骨格]、担子菌由来製剤(レンチナン、シゾフィラン、PSK等)、リポポリ多糖(LPS)を使用することができる。本発明にはOK-432を好適に使用できる。OK-432はA群3型溶血性連鎖球菌(Streptococcus pyogenes)の弱毒性自然変異株(Su株)をペニシリンで処理した細菌由来製剤の一般名である。本製剤はピシバニール(登録商標)の商品名で市販されている。本発明に使用する菌体製剤の添加量に特に限定はなく、使用される培地及び菌体製剤に応じて、適切な量を選択すればよい。特に本発明を限定するものではないが、菌体製剤は最終濃度が1~1000ng/mL、好ましくは50~200ng/mL程度になるように、特にOK-432を使用する場合、0.001~1KE/mL、好ましくは0.005~0.5KE/mL、さらに好ましくは0.01~0.2KE/mLの終濃度で培地に添加される。
本発明に使用する菌体製剤は、例えば、細菌由来製剤[OK-432、BCG(Bacillus Calmette Guerin)、Streptcoccus pyogenes、Corynebacterium parvum及びこれらの細胞壁骨格]、担子菌由来製剤(レンチナン、シゾフィラン、PSK等)、リポポリ多糖(LPS)を使用することができる。本発明にはOK-432を好適に使用できる。OK-432はA群3型溶血性連鎖球菌(Streptococcus pyogenes)の弱毒性自然変異株(Su株)をペニシリンで処理した細菌由来製剤の一般名である。本製剤はピシバニール(登録商標)の商品名で市販されている。本発明に使用する菌体製剤の添加量に特に限定はなく、使用される培地及び菌体製剤に応じて、適切な量を選択すればよい。特に本発明を限定するものではないが、菌体製剤は最終濃度が1~1000ng/mL、好ましくは50~200ng/mL程度になるように、特にOK-432を使用する場合、0.001~1KE/mL、好ましくは0.005~0.5KE/mL、さらに好ましくは0.01~0.2KE/mLの終濃度で培地に添加される。
本発明に使用するCD40アゴニストは、哺乳動物CD40リガンド(CD40L)、好ましくはヒトCD40L(GenBank Accession:NP_000065)とその可溶性フラグメント及びバリアント、並びにCD40レセプターに対するアゴニスト抗体である。CD40Lは膜貫通型タンパク質であり、その細胞内ドメイン及び膜貫通ドメインを除去した細胞外ドメインからなるCD40L可溶性フラグメントが好適に使用される。また、CD40アゴニストと共に、TNF-αを添加しても良い。添加するCD40アゴニストの濃度は限定されることはなく適宜設定すればよいが、最終濃度が1~1000μg/mL、好ましくは1~50μg/mL程度である。
本発明に使用するDC成熟化カクテルは、TNF-α、IL-1βを含む生物応答修飾剤の混合物であり、さらに、IL-6、インターフェロン(IFN)-α、IFN-γ、プロスタグランジン(PG)E2、ポリ(I:C)などの核酸類からなる群より選択される物質を含有してもよい。例えば、TNF-α、IL-1β、IL-6、PGE2を含むDC成熟化カクテル、TNF-α、IL-1β、ポリ(I:C)、IFN-α、IFN-γを含むDC成熟化カクテルが例示される。使用する量は各成熟化カクテルの組成に応じて適宜設定すれば良い。例えば、TNF-α、IL-1β、IL-6、IFN-α、IFN-γなどのサイトカイン類は、最終濃度が1ng/mL~1000ng/mL、好ましくは5ng/mL~500ng/mL程度である。PGE2は、最終濃度が0.1μg/mL~100μg/mL、好ましくは1μg/mL~10μg/mL程度である。ポリ(I:C)は、最終濃度が1μg/mL~1000μg/mL、好ましくは10μg/mL~100μg/mL程度である。用いるサイトカインは生体成分から分離されたものでも遺伝子工学的手法により生産したものでもよく、市販品を入手することもできる。また、本発明には、サイトカインの作用を示し得る各サイトカインの変異体及びフラグメントも使用できる。
本発明に使用するDC成熟化因子として、合成物質(ピランコポリマー、レバミゾール等)を使用しても良い。
未成熟DCを含有する細胞集団を培養する工程において、培養開始時の細胞濃度は特に限定はないが、例えば、1~1×108cells/mL、好適には10~5×107cells/mL、さらに好適には1×102~2×107cells/mLが例示される。
未成熟DCを含有する細胞集団の培養は、公知の培養条件で行ってもよく、通常の細胞培養に使用される条件を適用することができる。例えば、34℃~38℃、好ましくは37℃、2%~10%、好ましくは5%CO2条件下で培養することができる。また、適当な時間間隔で細胞培養液に新鮮な培地を加えて希釈したり、培地を新鮮なものに交換したり、又は細胞培養器材を交換することができる。培養期間は、数時間から数週間程度が好ましく、より好ましくは12時間~10日間であり、1~5日間程度がさらに好ましい。
本発明の方法において、対象とする特定の抗原を成熟DCに提示させるために、抗原を負荷(パルス)させることができる。例えば、前記抗原をDC成熟化因子の代わりに使用することができる。また、前記抗原をDC成熟化因子と共存させて、抗原と未成熟DCを接触させることができる。さらに、抗原を未成熟DC内又は成熟DC内に強制的に導入することができ、抗原がポリペプチドの場合は抗原をコードする核酸を未成熟DC内又は成熟DC内に導入して細胞内で発現させることができる。核酸はウイルスベクター又はエレクトロポレーションを使用してDCに導入することができ、これらの方法に使用する試薬やキットが広く市販されている。
対象とする特定の抗原としては、例えば、病原体、病原体溶解産物、病原体抽出物、病原体ポリペプチド、ウイルス粒子、細菌、タンパク質、ポリペプチド、がん細胞、がん細胞溶解産物、がん細胞特異的ポリペプチド(がん抗原)があるが、それらに限定されない。また、これらの抗原は、天然由来又は組換体の抗原のいずれもが使用できる。
以上の、本発明の方法により未成熟のDCを含有する細胞集団の培養を実施することにより、成熟したDCを含有する細胞集団を得ることができる。
本発明の方法に使用される未成熟DCを含有する細胞集団は、単球又は造血前駆細胞を含有する細胞集団、すなわち単核細胞フラクションより調製することができる。これらの細胞集団は適切な出発材料から調製することができ、例えば末梢血から調製される末梢血単核細胞(PBMC)は本発明に好適である。末梢血からPBMCを分離する方法として、密度勾配遠心分離法が一般に用いられている。密度勾配遠心分離法には、例えば、市販のフィコール(登録商標)、パーコール(登録商標)などの密度勾配遠心用媒体を使用することができる。さらに血球分離用に調製されたLymphoprep(登録商標)、Mono-poly Resolving Solution(登録商標)などを利用してもよい。密度勾配遠心分離法における媒体の量、温度設定等は用いる媒体により、適宜、設定される。
単核細胞フラクションからの単球の分離は、プラスチック付着法によって行うことができる。プラスチック付着法は、例えば、まず通常の培養用のプラスチックフラスコにウシ胎児血清(FBS)を注入し底面に十分いきわたらせた後、リン酸緩衝生理食塩水(PBS)で3回程度洗浄する。10%FBSを添加したRPMI-1640培地などに浮遊させた単核細胞フラクションを上記フラスコ内に注入し、37℃で1時間程度培養する。上清を吸引後、培地によりフラスコを3回程度洗浄し、非付着細胞を除去する。さらに冷却した0.5%EDTA、5%FBS含有PBSを添加し4℃で30分程度放置する。その後、ピペット等でフラスコから遊離した細胞を回収することにより、単球を含有する細胞集団を調製することができる。
さらに別の方法として、後述する実施例で示すように、PBMCから抗CD14固定化ビーズを用いてCD14陽性の単球を含有する細胞集団を回収する方法も挙げられる。
単球を含有する細胞集団を、IL-4、顆粒球・マクロファージ-コロニー刺激因子(GM-CSF)、ステムセルファクター(SCF)、IL-13、TNF-αから選択される少なくとも1つのサイトカインを含有する未成熟DC分化誘導因子の存在下で培養することにより、単球から未成熟DCへの分化を誘導し、未成熟DCを含有する細胞集団を得ることができる。この操作にはIL-4及びGM-CSFの組合せが好適である。また、必要に応じてIL-1、IL-2、IL-3等のサイトカインをさらに添加することが好ましい。用いるサイトカインは生体成分から分離されたものでも遺伝子工学的手法により生産したものでもよく、市販品を入手することもできる。また、本発明には、サイトカインの作用を示し得る各サイトカインの変異体及びフラグメントも使用できる。添加する各サイトカインの濃度は適宜設定すればよいが、最終濃度が1ng/mL~2000ng/mL、好ましくは10ng/mL~1000ng/mL程度である。培養期間は、数時間から数週間程度が好ましく、より好ましくは1~10日間であり、4~8日間程度がさらに好ましい。また、培養条件は、34℃~38℃、好ましくは37℃、2%~10%、好ましくは5%CO2条件下が好ましい。
また、未成熟DCを含有する細胞集団を造血前駆細胞から調製する場合は、PBMCから調製したCD34陽性細胞をGM-CSF、TNF-α、flt-3リガンド(FL)、c-kitリガンド(SCF)又はトロンボポエチン(TPO)を単独、もしくはその組合せの存在下で培養する。この操作により、未成熟DCを含有する細胞集団を得ることができる。
未成熟DCを含有する細胞集団を調製する工程は、未成熟DCを含有する細胞集団から未成熟DCを採取する工程や、未成熟DC以外の細胞を除去する工程を包含することができる。この工程により、未成熟DCを濃縮することができる。例えば、未成熟DCの細胞表面マーカー(例えばCD1a)を指標に、フローサイトメーターや前記マーカーに結合する分子を固定化したビーズ等により分離すればよい。
なお、単球、造血前駆細胞及び未成熟DCを含有する細胞集団を培養する各工程で使用する培地は、培養する細胞及び細胞集団に応じて適宜適切な培地を使用すれば良いが、例えば、RPMI-1640培地、ダルベッコ改変イーグル(DMEM)培地、TIL培地(免疫生物研究所)、表皮角化細胞(KBMと)培地、イスコフ(IMEM)培地等、細胞培養に使用されている市販の培地を使用することができる。また、必要に応じて5~20%のウシ血清、FBS、ヒト血清、血漿などを添加することができる。0.1~5%の血清を添加した低血清培地を使用しても良い。なお、血清又は血漿の由来としては、自己(培養する細胞と由来が同じであることを意味する)又は非自己(培養する細胞と由来が異なることを意味する)のいずれでもよいが、安全性の観点から、自己由来のものが好適に使用される。また、AIM-V培地、X-Vivo15培地、Cell Gro GMP Serum-Free DC Mediumなどの無血清培地を使用することもできる。全ての培養工程を同一の培地を使用しても良く、各培養工程で異なる培地を使用しても良い。
各工程の培養に使用される細胞培養器材としては、特に限定はないが、例えば、シャーレ、フラスコ、バッグ、バイオリアクター等を使用することができる。なお、バッグとしては、例えば、細胞培養用CO2ガス透過性バッグを使用することができる。また、工業的に大量の細胞集団を製造する場合には、バイオリアクターの使用が有利である。また、培養は開放系又は閉鎖系のいずれでも実施することができるが、得られる成熟DCの安全性の観点から、閉鎖系で培養を行うことが好ましい。
(2)本発明の製造方法により得られる細胞集団
本発明の細胞集団は、前記(1)の方法により得られる細胞集団である。本発明の細胞集団は、成熟したDCを含有する。DCの成熟は、当該分野で公知の方法によって、細胞表面マーカー、産生するサイトカイン量又はDC特異的遺伝子の発現量でモニタリングすることができる。これらのモニタリングは、当該分野で通常のアッセイ(例えば、免疫組織化学、mRNA定量法など)を用いて実施することができる。さらに、酵素免疫定量(ELISA)法、Fluorescence Activated Cell Sorting(FACS、フローサイトメトリー)法などのアッセイにより、細胞表面マーカー及びサイトカイン産生量をモニタリングすることができる。本発明の細胞集団は、CD80、CD86及びHLA-DRが強陽性で、CD83を発現する細胞を含有する細胞集団である。さらに、本発明の細胞集団はIL-1β、IL-6、TNF-α、IL-12、IFN-γなどのサイトカインを産生する。特にIL-12の産生増強は、1型(Th-1)の免疫応答を強く促進する。成熟DCはまた、貪食による抗原取り込み能力を失う。貪食活性は、通常の取り込みアッセイによって測定することができる。
本発明の細胞集団は、前記(1)の方法により得られる細胞集団である。本発明の細胞集団は、成熟したDCを含有する。DCの成熟は、当該分野で公知の方法によって、細胞表面マーカー、産生するサイトカイン量又はDC特異的遺伝子の発現量でモニタリングすることができる。これらのモニタリングは、当該分野で通常のアッセイ(例えば、免疫組織化学、mRNA定量法など)を用いて実施することができる。さらに、酵素免疫定量(ELISA)法、Fluorescence Activated Cell Sorting(FACS、フローサイトメトリー)法などのアッセイにより、細胞表面マーカー及びサイトカイン産生量をモニタリングすることができる。本発明の細胞集団は、CD80、CD86及びHLA-DRが強陽性で、CD83を発現する細胞を含有する細胞集団である。さらに、本発明の細胞集団はIL-1β、IL-6、TNF-α、IL-12、IFN-γなどのサイトカインを産生する。特にIL-12の産生増強は、1型(Th-1)の免疫応答を強く促進する。成熟DCはまた、貪食による抗原取り込み能力を失う。貪食活性は、通常の取り込みアッセイによって測定することができる。
本発明の細胞集団は、インビトロでT細胞と接触させることによりT細胞を活性化させることができる。その際、GM-CSF、IL-12、又はIL-2などのサイトカインを培地に添加することができる。特に、対象とする特定の抗原を負荷させたDCは、前記抗原を認識するT細胞を活性化することから、特定の抗原に対する免疫応答を生じさせるために有用である。このように活性化されたT細胞は、ヒト又はヒト以外の哺乳動物に投与することができる。
(3)本発明の細胞集団を含有する医薬
本発明の医薬は、前記(2)の細胞集団を含有することを特徴とする。本発明の医薬は、従来技術に従って、有効量の前記細胞集団を許容される担体と混合するなどして、経口/非経口製剤として製造することが出来る。本発明の医薬は、通常は、注射剤、懸濁剤、点滴剤等の非経口製剤として製造される。当該非経口製剤に含まれ得る担体としては、例えば、生理食塩水、ブドウ糖やその他の補助薬を含む等張液(例えば、D-ソルビトール、D-マンニトール、塩化ナトリウムなど)などの注射用の水性液を挙げることが出来る。また、例えば、緩衝剤(例えば、リン酸塩緩衝液、酢酸ナトリウム緩衝液)、無痛化剤(例えば、塩化ベンザルコニウム、塩酸プロカインなど)、安定剤(例えば、ヒト血清アルブミン、ポリエチレングリコールなど)、保存剤、酸化防止剤、免疫抑制剤(ラパマイシンなど)、抗体製剤、細胞製剤(制御性樹状細胞)などと配合してもよい。さらに、必要に応じてサイトカインを添加してもよい。
本発明の医薬は、前記(2)の細胞集団を含有することを特徴とする。本発明の医薬は、従来技術に従って、有効量の前記細胞集団を許容される担体と混合するなどして、経口/非経口製剤として製造することが出来る。本発明の医薬は、通常は、注射剤、懸濁剤、点滴剤等の非経口製剤として製造される。当該非経口製剤に含まれ得る担体としては、例えば、生理食塩水、ブドウ糖やその他の補助薬を含む等張液(例えば、D-ソルビトール、D-マンニトール、塩化ナトリウムなど)などの注射用の水性液を挙げることが出来る。また、例えば、緩衝剤(例えば、リン酸塩緩衝液、酢酸ナトリウム緩衝液)、無痛化剤(例えば、塩化ベンザルコニウム、塩酸プロカインなど)、安定剤(例えば、ヒト血清アルブミン、ポリエチレングリコールなど)、保存剤、酸化防止剤、免疫抑制剤(ラパマイシンなど)、抗体製剤、細胞製剤(制御性樹状細胞)などと配合してもよい。さらに、必要に応じてサイトカインを添加してもよい。
本発明の医薬が含有する細胞集団の細胞量は、投与対象、対象臓器、症状、投与方法などにより差異はあるが、通常、成人の患者(体重60kgとして)においては、例えば非経口投与の場合、1日につき、105~1012cellsであることが好ましく、より好ましくは106~107cellsである。他の動物の場合も、60kg当たりに換算して同等の量を投与することができる。本発明の医薬は、1回又は複数回、例えば毎日又は毎月患者に投与することができる。
本発明の医薬は、静脈内、皮内、皮下等へ注射することも、所属リンパ節へ注射等により注入することもできる。また、直接病変部に注入してもよく、全身に投与することも可能である。あるいは、病変部近傍の動脈から注入することも可能である。
本発明の医薬を投与することにより患者体内のT細胞を活性化することができる。特に対象とする特定の抗原を負荷させたDCを含有する医薬は、特定の抗原に特異的な細胞傷害活性を有するT細胞を活性化させるため、例えば、癌や感染症の治療に用いることが可能である。医薬に含有される細胞は、投与する患者に対して同種異系でも又は自家でもよい。
したがって、本発明は、本発明の医薬を投与することを特徴とする治療方法を提供する。さらに、本発明は、医薬の製造における本発明の細胞集団の使用を提供する。また、本発明は、治療において使用するための、成熟したDCを含有する細胞集団を提供する。さらに、本発明によれば、本発明の細胞集団を投与することによる、T細胞の活性化方法が提供される。
以下に実施例を挙げて本発明をさらに具体的に説明するが、本発明は以下の実施例のみに限定されるものではない。
実施例1 FNフラグメントを用いた樹状細胞(DC)の製造-1
(1)単球の調製
インフォームドコンセントが得られた健常人から調製したヒト末梢血単核細胞(PBMC)よりDynabeads Untouched Human Monocytes(ライフテクノロジーズ社製)を用いて、CD14陽性細胞(単球)を多く含む細胞集団を回収した。
(1)単球の調製
インフォームドコンセントが得られた健常人から調製したヒト末梢血単核細胞(PBMC)よりDynabeads Untouched Human Monocytes(ライフテクノロジーズ社製)を用いて、CD14陽性細胞(単球)を多く含む細胞集団を回収した。
(2)樹状細胞への分化
実施例1-(1)で得られた細胞を、FBS(ライフテクノロジーズ社製)を最終濃度5%、ペニシリン-ストレプトマイシン(ライフテクノロジーズ社製)を最終濃度100U/mLとなるように添加したRPMI-1640培地(シグマアルドリッチ社製)(以下、FS培地と記載)に、1×106cells/mLとなるように懸濁した。さらに、この細胞懸濁液にGM-CSF(R&Dシステムズ社製)及びIL-4(R&Dシステムズ社製)をそれぞれ最終濃度100ng/mLとなるように添加し、細胞培養用12ウェルプレートにて37℃、5%CO2インキュベーターで樹状細胞誘導培養を開始した。培養2日後、培地を半量除き、GM-CSF及びIL-4をそれぞれ最終濃度100ng/mL含むFS培地を、除いた量と等量加えさらに3日間培養した。
実施例1-(1)で得られた細胞を、FBS(ライフテクノロジーズ社製)を最終濃度5%、ペニシリン-ストレプトマイシン(ライフテクノロジーズ社製)を最終濃度100U/mLとなるように添加したRPMI-1640培地(シグマアルドリッチ社製)(以下、FS培地と記載)に、1×106cells/mLとなるように懸濁した。さらに、この細胞懸濁液にGM-CSF(R&Dシステムズ社製)及びIL-4(R&Dシステムズ社製)をそれぞれ最終濃度100ng/mLとなるように添加し、細胞培養用12ウェルプレートにて37℃、5%CO2インキュベーターで樹状細胞誘導培養を開始した。培養2日後、培地を半量除き、GM-CSF及びIL-4をそれぞれ最終濃度100ng/mL含むFS培地を、除いた量と等量加えさらに3日間培養した。
(3)固定化プレートの調製
フィブロネクチン(FN)フラグメントであるCH-296[レトロネクチン(登録商標)、タカラバイオ社製]、ヒト血漿由来フィブロネクチン(シグマアルドリッチ社製)をそれぞれ25μg/mLとなるようにPBS(ライフテクノロジーズ社製)に溶解し、表面未処理96ウェル平面プレート(ベクトン・ディッキンソン社製)の各ウェルに0.07mLずつ加え、4℃で3日間放置した後、37℃で4時間保持した。また、対照として溶媒(PBS)のみを表面未処理96ウェル平面プレートの各ウェルに0.07mL加え、4℃で3日間放置した後、37℃で4時間保持した。以下、FNフラグメントCH-296(レトロネクチン)を固定化したプレートをRNプレート、ヒト血漿由来FNを固定化したプレートをFNプレート、溶媒のみを使用したプレートをNTプレートと記載する。こうして調製したプレートは、溶液を除去しPBSで2回洗浄した後に使用した。
フィブロネクチン(FN)フラグメントであるCH-296[レトロネクチン(登録商標)、タカラバイオ社製]、ヒト血漿由来フィブロネクチン(シグマアルドリッチ社製)をそれぞれ25μg/mLとなるようにPBS(ライフテクノロジーズ社製)に溶解し、表面未処理96ウェル平面プレート(ベクトン・ディッキンソン社製)の各ウェルに0.07mLずつ加え、4℃で3日間放置した後、37℃で4時間保持した。また、対照として溶媒(PBS)のみを表面未処理96ウェル平面プレートの各ウェルに0.07mL加え、4℃で3日間放置した後、37℃で4時間保持した。以下、FNフラグメントCH-296(レトロネクチン)を固定化したプレートをRNプレート、ヒト血漿由来FNを固定化したプレートをFNプレート、溶媒のみを使用したプレートをNTプレートと記載する。こうして調製したプレートは、溶液を除去しPBSで2回洗浄した後に使用した。
(4)固定化プレートでの樹状細胞の成熟化
実施例1-(2)で調製した樹状細胞誘導培養後の細胞を500×g、8分間の遠心で回収し、DC成熟化因子としてOK-432(日本標準商品分類番号874299、中外製薬社製)を最終濃度0.05KE/mL含むFS培地に6.7×105cells/mLとなるように懸濁した。この細胞懸濁液0.15mLを実施例1-(3)で調製した各プレートにて37℃、5%CO2インキュベーターで2日間培養した。
実施例1-(2)で調製した樹状細胞誘導培養後の細胞を500×g、8分間の遠心で回収し、DC成熟化因子としてOK-432(日本標準商品分類番号874299、中外製薬社製)を最終濃度0.05KE/mL含むFS培地に6.7×105cells/mLとなるように懸濁した。この細胞懸濁液0.15mLを実施例1-(3)で調製した各プレートにて37℃、5%CO2インキュベーターで2日間培養した。
また、細胞をDC成熟化因子を含有しないFS培地に懸濁し、37℃、5%CO2インキュベーターで2日間培養した群を調製した。
実施例2 樹状細胞の表面抗原発現解析-1
実施例1-(4)で得られた培養細胞を、ウシ胎児血清を最終濃度1%、ペニシリン-ストレプトマイシンを最終濃度100U/mLとなるように添加したPBS(以下、1%FBS/PBS培地と記載)に懸濁し、下記の蛍光色素標識抗体をそれぞれ添加し4℃で20分間抗体反応を行った。その後、1%FBS/PBS培地で洗浄した細胞をフローサイトメーターFACS CantoII(ベクトン・ディッキンソン社製)での解析に供した。解析では前方散乱光(FSC)と側方散乱光(SSC)でミエロイド系列細胞にゲートを掛け、平均蛍光強度(MFI:Mean Fluorescence Intensity)としてGeo Meanの値により、細胞表面抗原の発現量を測定した。
実施例1-(4)で得られた培養細胞を、ウシ胎児血清を最終濃度1%、ペニシリン-ストレプトマイシンを最終濃度100U/mLとなるように添加したPBS(以下、1%FBS/PBS培地と記載)に懸濁し、下記の蛍光色素標識抗体をそれぞれ添加し4℃で20分間抗体反応を行った。その後、1%FBS/PBS培地で洗浄した細胞をフローサイトメーターFACS CantoII(ベクトン・ディッキンソン社製)での解析に供した。解析では前方散乱光(FSC)と側方散乱光(SSC)でミエロイド系列細胞にゲートを掛け、平均蛍光強度(MFI:Mean Fluorescence Intensity)としてGeo Meanの値により、細胞表面抗原の発現量を測定した。
蛍光色素標識抗体は、FITC標識抗ヒトCD83抗体(eBioscience社製)、PE標識抗ヒトCD80抗体(eBioscience社製)、PE標識抗ヒトCD86抗体(eBioscience社製)をそれぞれ使用した。
その結果を図1に示す。図1の縦軸は平均蛍光強度(MFI)を示す。図1より、OK-432存在下(以下、図中で「OK432」と記載する)にRNプレートで成熟させたDCは、NTプレートやFNプレートで成熟させたDCより成熟化マーカーであるCD83、CD80及びCD86の細胞表面発現量が上昇した。一方、OK-432非存在下(以下、図中「-」と記載する)では、いずれのプレート(NTプレートやRNプレートやFNプレート)を使用しても成熟化マーカーの発現量の上昇は見られなかった。以下、図中で、RNプレートは「RN」、FNプレートは「FN」、NTプレートは「NT」と記載する。
以上の実施例から、FNフラグメントはDC成熟化因子であるOK-432存在下にDC成熟化マーカーの発現量を増加させること、すなわち、FNフラグメントはDCの成熟化を促進することが明らかとなった。
実施例3 FNフラグメントを用いた樹状細胞の製造-2
(1)単球の調製
実施例1-(1)と同様の方法で行った。

実施例3-(1)で得られた細胞をFS培地に1.2×106cells/mLとなるように懸濁し、さらにGM-CSF及びIL-4をそれぞれ最終濃度100ng/mLとなるように添加し、細胞培養用12ウェルプレートにて37℃、5%CO2インキュベーターで樹状細胞誘導培養を開始した。培養3日後、培地を半量除き、GM-CSF及びIL-4をそれぞれ最終濃度100ng/mL含むFS培地を、除いた量と等量加えさらに3日間培養した。
実施例3-(1)で得られた細胞をFS培地に1.2×106cells/mLとなるように懸濁し、さらにGM-CSF及びIL-4をそれぞれ最終濃度100ng/mLとなるように添加し、細胞培養用12ウェルプレートにて37℃、5%CO2インキュベーターで樹状細胞誘導培養を開始した。培養3日後、培地を半量除き、GM-CSF及びIL-4をそれぞれ最終濃度100ng/mL含むFS培地を、除いた量と等量加えさらに3日間培養した。
(3)固定化プレートの調製
FNフラグメントCH-296(レトロネクチン)を25μg/mLとなるようにPBSに溶解し、表面未処理48ウェル平面プレート(ベクトン・ディッキンソン社製)の各ウェルに0.3mL加え、4℃で3日間放置した後、37℃で4時間保持してRNプレートを調製した。また、対照として溶媒(PBS)のみを表面未処理48ウェル平面プレートの各ウェルに0.3mL加え、4℃で3日間放置した後、37℃で4時間保持してNTプレートを調製した。こうして調製したプレートは、溶液を除去しPBSで2回洗浄した後に使用した。
FNフラグメントCH-296(レトロネクチン)を25μg/mLとなるようにPBSに溶解し、表面未処理48ウェル平面プレート(ベクトン・ディッキンソン社製)の各ウェルに0.3mL加え、4℃で3日間放置した後、37℃で4時間保持してRNプレートを調製した。また、対照として溶媒(PBS)のみを表面未処理48ウェル平面プレートの各ウェルに0.3mL加え、4℃で3日間放置した後、37℃で4時間保持してNTプレートを調製した。こうして調製したプレートは、溶液を除去しPBSで2回洗浄した後に使用した。
(4)固定化プレートでの樹状細胞の成熟化
実施例3-(2)で調製した樹状細胞誘導培養後の細胞を500×g、8分間の遠心で回収し、DC成熟化因子としてOK-432を最終濃度0.05KE/mL含むFS培地に4.2×105cells/mLとなるように懸濁した。この細胞懸濁液0.5mLを実施例3-(3)で調製した各プレートにて37℃、5%CO2インキュベーターで2日間培養した。
実施例3-(2)で調製した樹状細胞誘導培養後の細胞を500×g、8分間の遠心で回収し、DC成熟化因子としてOK-432を最終濃度0.05KE/mL含むFS培地に4.2×105cells/mLとなるように懸濁した。この細胞懸濁液0.5mLを実施例3-(3)で調製した各プレートにて37℃、5%CO2インキュベーターで2日間培養した。
実施例4 樹状細胞の培養上清中サイトカインの定量解析-1
実施例3-(4)で得られた培養上清を試料とし、Human Th1/Th2 11plex FlowCytomix Multiplex(eBioscience社製)を用い、フローサイトメーターFACS CantoIIにて、DCから産生される培養上清中のサイトカイン(IL-1β、TNF-α、IL-12、IFN-γ)量を測定した。解析はFlowCytomix Pro(eBioscience社製)を用いた。
実施例3-(4)で得られた培養上清を試料とし、Human Th1/Th2 11plex FlowCytomix Multiplex(eBioscience社製)を用い、フローサイトメーターFACS CantoIIにて、DCから産生される培養上清中のサイトカイン(IL-1β、TNF-α、IL-12、IFN-γ)量を測定した。解析はFlowCytomix Pro(eBioscience社製)を用いた。
その結果を図2に示す。図2の縦軸はサイトカイン量を示す。図2より、OK-432存在下にRNプレートで成熟させたDCはNTプレートで成熟させたDCより、その培養上清中にIL-1β、TNF-α、IL-12、IFN-γが多く存在した。
以上の実施例から、FNフラグメントは、DC成熟化因子であるOK-432存在下にDCからのIL-1β、TNF-α、IL-12、IFN-γ産生を促進することが明らかとなった。
実施例5 FNフラグメントを用いた樹状細胞の製造-3
(1)単球の調製
実施例1-(1)と同様の方法で行った。

実施例5-(2)で調製した樹状細胞誘導培養後の細胞を500×g、8分間の遠心で回収し、DC成熟化因子としてOK-432を最終濃度0.05KE/mL含むHS培地に4.0×105cells/mLとなるように懸濁した。この細胞懸濁液0.15mLを実施例5-(3)で調製した各プレートにて37℃、5%CO2インキュベーターで2日間培養した。
また、成熟化因子を加えずにHS培地で懸濁後、37℃、5%CO2インキュベーターで2日間培養した群を調製した。
実施例6 樹状細胞の表面抗原発現解析-2
実施例5-(4)で得られた培養細胞を、実施例2と同様の方法で測定した。

蛍光色素標識抗体は、FITC標識抗ヒトHLA-DR抗体(ベクトン・ディッキンソン社製)、FITC標識抗ヒトCD83抗体、PE標識抗ヒトCD80抗体、PE標識抗ヒトCD86抗体をそれぞれ使用した。
その結果を図3に示す。図3の縦軸は平均蛍光強度を示す。図3より、OK-432存在下にRNプレートで成熟させたDCはNTプレートで成熟させたDCより、DC成熟化マーカーであるHLA-DR、CD83、CD80、CD86の細胞表面発現量が上昇した。一方、OK-432非存在下(図中「-」)では、いずれのプレート(NTプレート及びRNプレート)を使用しても発現量の上昇は見られなかった。
以上の実施例から、ヒトAB血清含有培地を用いてもFNフラグメントは、DC成熟化因子であるOK-432存在下にDC成熟化マーカーの発現を促進することが明らかとなった。
実施例7 FNフラグメントを用いた樹状細胞の製造-4
(1)単球の調製
実施例1-(1)と同様の方法で行った。

FNフラグメントCH-296(レトロネクチン)、ヒト血漿由来フィブロネクチン、フィブロネクチン断片III1―Cヒト(シグマアルドリッチ社製)を50μg/mLとなるようにPBSに溶解し、細胞培養用96ウェル平面プレート(コーニング社製)の各ウェルに0.07mL加え、4℃で3日間放置した後、37℃で4時間保持した。また、対照として溶媒(PBS)のみを細胞培養用96ウェル平面プレートの各ウェルに0.07mL加え、4℃で3日間放置した後、37℃で4時間保持した。以下、FNフラグメントIII1―Cを固定化したプレートをIIICプレートと記載する。こうして調製したプレートは、溶液を除去しPBSで2回洗浄した後に使用した。
FNフラグメントCH-296(レトロネクチン)、ヒト血漿由来フィブロネクチン、フィブロネクチン断片III1―Cヒト(シグマアルドリッチ社製)を50μg/mLとなるようにPBSに溶解し、細胞培養用96ウェル平面プレート(コーニング社製)の各ウェルに0.07mL加え、4℃で3日間放置した後、37℃で4時間保持した。また、対照として溶媒(PBS)のみを細胞培養用96ウェル平面プレートの各ウェルに0.07mL加え、4℃で3日間放置した後、37℃で4時間保持した。以下、FNフラグメントIII1―Cを固定化したプレートをIIICプレートと記載する。こうして調製したプレートは、溶液を除去しPBSで2回洗浄した後に使用した。
(4)固定化プレートでの樹状細胞の成熟化
実施例7-(2)で調製した樹状細胞誘導培養後の細胞を500×g、8分間の遠心で回収し、DC成熟化因子としてOK-432を最終濃度0.05KE/mL含むHS培地に3.0×105cells/mLとなるように懸濁した。この細胞懸濁液0.125mLを実施例1-(3)で調製した各プレートにて37℃、5%CO2インキュベーターで2日間培養した。

実施例9-(4)で得られた培養上清を試料とし、Human Th1/Th2 11plex FlowCytomix Multiplexを用い、フローサイトメーターFACS CantoIIにて、DCから産生される培養上清中のIL-6、TNF-α量を測定した。解析はFlowCytomix Proを用いた。

以上の実施例から、ヒトAB血清含有培地を用いて分化させたDCの場合も、FNフラグメントは、DC成熟化因子であるOK-432存在下にDCからのIL-6、TNF-α産生を促進することが明らかとなった。

以上の実施例から、無血清培地(XV培地、AM培地又はCG培地)をDC成熟化因子であるMCと組み合わせた場合でも、FNフラグメントはDC成熟化マーカーの発現を促進することが明らかとなった。
本発明により、成熟したDCを含む細胞集団の製造方法が提供される。当該方法により、活性の高いDCを含有する細胞集団が得られる。本発明の細胞集団は、免疫応答の誘導において優れており、DCワクチン療法、養子免疫療法をはじめとする医療分野において極めて有用である。
Claims (10)
- 未成熟の樹状細胞を含有する細胞集団を、樹状細胞成熟化因子及びフィブロネクチンフラグメントの存在下で培養する工程を包含することを特徴とする、成熟した樹状細胞を含有する細胞集団の製造方法。
- 樹状細胞成熟化因子が、下記1)~3)からなる群より選択される、請求項1記載の方法。
1)菌体製剤、
2)CD40アゴニスト、及び
3)腫瘍壊死因子-α及びインターロイキン-1βを含む樹状細胞成熟化カクテル。 - 菌体製剤が、OK-432である、請求項2記載の方法。
- フィブロネクチンフラグメントが、細胞接着ドメイン、ヘパリン結合ドメイン及びフィブロネクチン結合ドメイン部位からなる群より選択されるドメインを有する組換えポリペプチドである請求項1~3のいずれか1項記載の方法。
- フィブロネクチンフラグメントが、CH-296及びIII1-Cから選択される組換えポリペプチドである請求項4記載の方法。
- 単球を含有する細胞集団より未成熟の樹状細胞を含有する細胞集団を調製する工程をさらに含む、請求項1~5のいずれか1項記載の方法。

- 無血清培地を使用して培養を実施する、請求項1~7のいずれか1項記載の方法。
- 請求項1~8のいずれか1項記載の製造方法により得られる細胞集団。
- 請求項9記載の細胞集団を含有する医薬。
