Description

分取クロマトグラフ装置
 本発明は、液体又は気体の試料から、カラムで分離された特定の成分の液体又は気体を分画して採取する分取クロマトグラフ装置に関する。
 液体クロマトグラフやガスクロマトグラフを利用して、試料に含まれる1ないし複数の成分を分画して採取する、いわゆる分取クロマトグラフ装置が知られている。これらの分取クロマトグラフ装置では、試料中の特定の目的成分の保持時間に基づいて、カラムから溶出する溶出物を所定の時間だけ分画して採取するフラクションコレクタにより、当該目的成分の溶出物を試料から分離する。こうした分取クロマトグラフ装置では、目的成分の溶出物を分画するタイミングは、手動又は自動で決めている。
 比較的多量の試料を用意できる場合には、その試料について予備的なクロマトグラフィー分析を実行してクロマトグラムを作成し、オペレータがそのクロマトグラムを見て分画のタイミングを決めればよい。しかしながら、例えば試料が少量しかない場合や高価で貴重な場合には、予備的なクロマトグラフィー分析を行うことができず、1回のクロマトグラフィー分析で分画を実行しなければならない。
 特許文献1には、所定の時間間隔で分画を終了して次の分画に移行するという時間分画を行いつつ、クロマトグラムのピークの開始点が検出されたときに分画を開始し、ピーク終了点が検出されたときにその分画を終了して次の分画に移行するというピーク検出に基づく分画を行う、分取クロマトグラフ装置が記載されている。これにより、ピークが検出された成分について確実に分画すると共に、ピークが検出されない微量な成分も、複数回行われた時間分画のいずれかにおいて分画することができる。

 上述したような分画の場合、試料中の異なった成分の保持時間が十分に離れていれば、目的成分を高い純度で分画することができる。しかしながら、多様な成分を含む試料の場合、目的成分の保持時間にかなり近い保持時間を持つ別の成分(不純物)が含まれていることも多く、そうした場合、複数の成分のピークが重なってしまう。このような場合には、分画した溶出物に、目的成分以外の不純物が混入してしまう。
 本発明が解決しようとする課題は、目的成分の保持時間に近い保持時間を持つ別の成分(不純物)が含まれていても、不純物が混入することなく目的成分を分画することができる分取クロマトグラフ装置を提供することである。
 上記課題を解決するために成された本発明に係る分取クロマトグラフ装置は、試料中の成分を時間的に分離して溶出物を溶出させるカラムと、該溶出物の吸光度スペクトルA(λ)を取得する検出器と、該吸光度スペクトルA(λ)に基づいてクロマトグラムS(t)をリアルタイムで作成してゆき、該クロマトグラムS(t)に基づいて前記溶出物から目的成分を分画するフラクションコレクタとを有する分取クロマトグラフ装置であって、
 a) 前記クロマトグラムS(t)に基づいて前記目的成分のクロマトグラムピークが現れたか否かを判定するピーク区間判定部と、
 b) 前記吸光度スペクトルA(λ)より、その波長微分d(A(λ))/dλの、前記目的成分の吸光度が極大値又は極小値を持つ既知の波長λ0における値である微分スペクトル値A'(λ0)=d(A(λ))/dλ|λ=λ0を求め、該微分スペクトル値A'(λ0)の絶対値が所定値以下であるか否かを判定する微分値判定部と、
 c) 前記目的成分のクロマトグラムピークが現れたと判定され、且つ、前記微分スペクトル値A'(λ0)の絶対値が前記所定値以下であると判定されている時間帯に、前記溶出物を分画させるよう前記フラクションコレクタを制御するフラクションコレクタ制御部と
を備えることを特徴とする。
 本発明に係る分取クロマトグラフ装置では、ピーク区間判定部は、リアルタイムで作成されるクロマトグラムS(t)に基づいて、目的成分に対応するクロマトグラムピークが現れたか否かを順次判定する。なお、「リアルタイム」とは、吸光度スペクトルA(λ)の取得とクロマトグラムS(t)が同時であることには限られず、分取に支障の無い範囲内の時間も含む。クロマトグラムS(t)は、目的成分に対応する吸光度スペクトルのピーク内にある特定の波長λiの吸光度A(λi)に基づいて作成してもよいし、吸光度スペクトルA(λ)の波長λに関する積分値に基づいて作成してもよい。データ処理の負荷の観点から、特定波長λiの吸光度A(λi)に基づいてクロマトグラムS(t)を作成することが望ましい。目的成分に対応するクロマトグラムS(t)のピークが現れたか否かの判定は、従来より用いられている方法、例えば特許文献1に記載のように、その時点t=taにおけるS(ta)が所定の閾値を超えたか否か、あるいはt=taにおけるS(t)の曲線の傾きが所定の値を超えたか否かに基づいて行うことができる。
 微分値判定部は、前記吸光度スペクトルA(λ)を取得する度に、前記波長λ0(以下、「吸光極値波長」と呼ぶ)における微分スペクトル値A'(λ0)を求め、該微分スペクトル値A'(λ0)の絶対値が所定値以下であるか否かを順次判定する。ここで、吸光度スペクトルA(λ)を取得する頻度は、クロマトグラムS(t)を作成するために吸光度スペクトルA(λ)を取得する頻度と同じであってもよいし、それより少なくてもよい。吸光極値波長λ0は、目的成分の吸光度が極大値又は極小値を持つ波長であり、目的成分によって相違する。ここで、一般的な試料の吸光度が短波長側で強くなる傾向があることから、目的成分の吸光度は極大値だけではなく極小値を持つことがある。以下では、簡単のため、主に吸光度が極大値を取る場合の吸光極値波長λ0を用いて説明するが、吸光度が極小値を取る場合も同様である。
 ある時間において溶出物に不純物が含まれていなければ、吸光度スペクトルA(λ)には目的成分以外の成分によるスペクトルが重畳されていないため、微分スペクトル値A'(λ0)は理想的には0になる。ただし、測定誤差等の影響があるため、A'(λ0)の絶対値がある程度の範囲内(前記所定値以下)にあるときには、溶出物に不純物が含まれていないとみなすことができる。一方、溶出物に不純物が含まれていれば、目的成分によるスペクトルに、不純物によるスペクトルが重畳されるため、微分スペクトル値A'(λ0)の絶対値は前記所定値よりも大きい値を持つ。従って、微分スペクトル値A'(λ0)の絶対値が所定値以下であるか否かを順次判定することは、その時点における溶出物に不純物が含まれるか否かを判別することを意味する。これにより、本発明では、目的成分と不純物の保持時間が近くクロマトグラムピークの分離が困難な場合であっても、不純物の有無を判別することができる。
 フラクションコレクタ制御部は、ピーク区間判定部において目的成分に対応するクロマトグラムピークが現れたと判定され、且つ、微分値判定部において微分スペクトル値A'(λ0) の絶対値が所定値以下である(すなわち、溶出物が不純物を含有しない)と判定された時間帯に、溶出物を分画させるよう、フラクションコレクタを制御する。これにより、不純物の含まれない目的成分を分画することができる。
 前記特定波長λiの吸光度A(λi)に基づいてクロマトグラムS(t)を作成する場合には、前述のように一般的な試料の吸光度が短波長側で強くなる傾向があるため、該特定波長λiは、目的成分の吸光極値波長λ0よりも短波長であることが望ましいが、それに限定されるものではない。
 本発明において、上記クロマトグラフは、液体クロマトグラフ、ガスクロマトグラフのいずれでもよい。上記検出器には、典型的には、フォトダイオードアレイのように同時に多数の波長に対して検出が可能なマルチチャンネル型検出器が用いられるが、波長走査を伴う紫外可視分光光度計、赤外分光光度計、近赤外分光光度計、蛍光分光光度計などを用いてもよい。
 前記微分値判定部は、前記波長λ0における微分スペクトル値A'(λ0)が0であるか否かを判定する代わりに、吸光度スペクトルA(λ)の波長微分d(A(λ))/dλの値が0になる波長λaを求めたうえで、該波長が前記波長λ0と一致するか否かを判定するようにしてもよい。λa=λ0であれば溶出物に不純物が含まれていないことを意味する。ただし、前述の場合と同様に、測定誤差等の影響があるため、λaとλ0の差の絶対値が所定値以下であれば、溶出物に不純物が含まれていないとみなすことができる。一方、λaとλ0の差の絶対値が所定値よりも大きければ、溶出物に不純物が含まれていることを意味する。
 すなわち、このような微分値判定部を有する分取クロマトグラフ装置は、試料中の成分を時間的に分離して溶出物を溶出させるカラムと、該溶出物の吸光度スペクトルA(λ)を取得する検出器と、該吸光度スペクトルA(λ)に基づいてクロマトグラムS(t)をリアルタイムで作成してゆき、該クロマトグラムS(t)に基づいて前記溶出物から目的成分を分画するフラクションコレクタとを有する分取クロマトグラフ装置であって、
 a) 前記クロマトグラムS(t)に基づいて前記目的成分のクロマトグラムピークが現れたか否かを判定するピーク区間判定部と、
 b) 前記吸光度スペクトルA(λ)より、その波長微分d(A(λ))/dλの値が0になる波長λaを求め、該波長λaと、前記目的成分の吸光度が極大値又は極小値を持つ既知の波長λ0の差の絶対値が所定値以下であるか否かを判定する微分値判定部と、
 c) 前記目的成分のクロマトグラムピークが現れたと判定され、且つ、波長λaと波長λ0の差の絶対値が前記所定値以下であると判定されている時間帯に、前記溶出物を分画させるよう前記フラクションコレクタを制御するフラクションコレクタ制御部と
を備えることを特徴とする。
 本発明に係る分取クロマトグラフ装置により、目的成分の保持時間に近い保持時間を持つ別の成分(不純物)が含まれていても、不純物が混入することなく目的成分を分画することができる。

 本発明に係る分取クロマトグラフ装置の実施例を、図1~図7を用いて説明する。
(1) 本実施例の分取クロマトグラフ装置の構成
 本実施例の分取クロマトグラフ装置は、液体クロマトグラフ(LC)を用いた分取LC装置である。この分取LC装置10は、図1に示すように、移動相容器11、送液ポンプ12、インジェクタ13、カラム14、検出器15、フラクションコレクタ16がこの順に送液流路19で接続されている。また、分取LC装置10は、検出器15から得られるデータに基づいてフラクションコレクタ16を制御するための制御部17を有している。
 分取LC装置10では、移動相容器11に貯留されている移動相が送液ポンプ12により吸引され、一定流量でインジェクタ13を介してカラム14に流される。インジェクタ13において、移動相中に試料が注入される。試料は移動相に乗ってカラム14に導入され、カラム14を通過する間に時間方向に分離されて溶出する。検出器15は、本実施例では紫外分光光度計であり、測定光を発する紫外光源151、溶出液が流れるフローセル152、回折格子などを含む分光器153、フォトダイオードアレイ154及び吸光度スペクトル生成部155を有している。紫外光源151から発せられた測定光は、フローセル152を流れる溶出液中を通過するが、その際に溶出液に含まれる成分に特有の波長の吸収を受ける。この透過光は分光器153で波長分散され、分散光は測定範囲内の波長において、フォトダイオードアレイ154により同時に検出される。これにより、透過光の波長毎の信号強度スペクトルI(λ)が得られる。吸光度スペクトル生成部155にて、分析開始時に得られる信号強度スペクトルI0(λ)を記憶し、時々刻々得られる信号強度スペクトルI(λ)から吸光度スペクトルA(λ)=log(I0(λ)/I(λ))が生成され、この信号が検出器15から出力される。
 検出器15を通過した溶出液はその全量(又は一部でもよい)がフラクションコレクタ16に導入される。フラクションコレクタ16は、電磁バルブ161と、多数の容器を収容する容器ラック162と、電磁バルブ161の下流側に設けられた、溶出液を滴下する分注ノズル163と、分注ノズル163を2軸方向に移動させることによって溶出液を採取する容器を切り替える駆動部164を有する。電磁バルブ161の開閉及び駆動部164による分注ノズル163の移動は、後述の制御部17により制御される。
 制御部17は、ピーク区間判定部171と、微分値判定部172と、フラクションコレクタ制御部173を有しており、CPU及びソフトウエアにより具現化されている。
 ピーク区間判定部171は、検出器15から吸光度スペクトルA(λ)を取得し、該吸光度スペクトルA(λ)に基づいてクロマトグラムS(t)をリアルタイムで作成したうえで、目的成分に対応するクロマトグラムピークが現れたか否かの判定を順次行う。本実施例では、判定時t=taが目的成分の保持時間(既知)の前後に亘る所定の時間範囲内にあって、S(ta)が閾値を超えているときには、目的成分に対応するクロマトグラムピークが現れていると判定する。微分値判定部172は、検出器15から吸光度スペクトルA(λ)の信号を取得する度に、既知の吸光極値波長λ0における微分スペクトル値A'(λ0)を順次求め、微分スペクトル値A'(λ0)の値が0であるか否かを順次判定するものである。
 フラクションコレクタ制御部173は、ピーク区間判定部171によってクロマトグラムピークが現れたと判定され、且つ、微分値判定部172によって微分スペクトル値A'(λ0)が0であると判定されている時間帯に、該溶出物を分画させるよう、フラクションコレクタ16を制御するものである。具体的には、上記判定がなされる前に目的成分を分取するための容器上に分注ノズル163を移動させておき、上記判定がなされたときに、電磁バルブ161を開放するように制御する。また、前記判定の条件を満たさなくなったときに、フラクションコレクタ制御部173は、電磁バルブ161を閉鎖するように制御する。
(2) 溶出物中の不純物の有無を判断する原理
 本実施例の分取LC装置10の微分値判定部172におけるデータ処理によって、溶出物中の不純物の有無を判断する原理を、図2~図5を用いて説明する。
 図2(a)は、時刻t、波長λ、及び吸光度Aという3つのディメンジョンを持つ3次元クロマトグラムデータの模式図である。3次元クロマトグラムデータのうち、特定の時刻t=taにおける吸光度Sと波長λの関係は、その時刻tにおける吸光度スペクトルA(λ)に該当する(図2(a)中の曲線)。また、本実施例では、特定の波長λ=λiにおける吸光度Sと時刻tの関係は、その波長λiにおけるクロマトグラムS(t)に該当する(図2(b))。本実施例では、吸光度スペクトルA(λ)を用いて、以下のように不純物の有無を判断する。
 いま、目的成分xと不純物yの2つの成分が試料に含まれる場合を考える。図3は、目的成分xと不純物成分yそれぞれの吸光度スペクトルを分離して示した一例の図である。図示するように、一般に、吸光度ピークの頂点(極大点)に対応した波長(吸光極大値波長)は物質毎に相違する。また、吸光度は短波長側で強くなる傾向があるため、吸光度スペクトルには極大点よりも短波長側に谷の底(極小点)が現れることがある。
 図4は、クロマトグラム上での目的成分xと不純物成分yそれぞれのピークプロファイルの一例と、それらピークプロファイルが重なった状態、つまり混合ピークプロファイルとを示す図である。測定される吸光度スペクトルから実際に得られるクロマトグラムは、混合ピークプロファイルの方である。目的成分xと不純物成分yの保持時間はかなり近接しており、混合ピークプロファイルからは、どの時間帯で不純物成分yが溶出物に混合しているのか否かを判断するのは困難である。
 そこで、本実施例では、不純物成分yの混入の有無を判定するために、以下のように、吸光度スペクトルの波長微分である微分スペクトルを用いる。図5は、図3に示した成分毎の吸光度スペクトルを波長方向に微分した微分スペクトルである。波長方向に波長スペクトルのカーブが上昇している局面では微分スペクトルの値は正、波長スペクトルのカーブが下降している局面では微分スペクトル値は負になり、吸光度ピークの極大点及び極小点では微分スペクトル値は0となる。本実施例では、目的成分xの吸光極大値波長、すなわち微分スペクトル値が正値から負値に変化する状況で0になる波長をλ0とする。
 前述のように、一般に吸光極大値波長は物質毎に相違するため、図示の通り、不純物成分yの吸光極大値波長は目的成分xの吸光極大値波長λ0と相違し、また、目的成分xの吸光極大値波長λ0における不純物成分yの微分スペクトル値は0ではない値を有する。そのため、溶出物が目的成分xのみを含有する(不純物成分yを含有しない)場合には、吸光極大値波長λ0において測定される微分スペクトル値は、理想的には0になる。一方、溶出物が目的成分xだけではなく不純物成分yも含有している場合には、吸光極大値波長λ0において測定される微分スペクトル値は0ではない値を示す。
 従って、本実施例では、目的成分xの吸光極大値波長λ0において、微分スペクトル値A'(λ0)を順次(時々刻々と)求め、該微分スペクトル値A'(λ0)が0であるか否かを判定することにより、その時点において溶出物が不純物成分yを含有する(A'(λ0)≠0)かしない(A'(λ0)=0)かを判定する。ただし、測定誤差などの影響を考慮して、微分スペクトル値A'(λ0)の絶対値がある程度の範囲内にある場合には、その時点において溶出物が不純物成分yを含有しないと判断してもよい。そして、本実施例では、この判定方法に従って、溶出物が不純物成分yを含有しない時間帯にのみ、目的成分xの分取を実行する(図4の「分取区間」)。
 なお、ここまでは吸光度ピークの極大点に対応する波長を吸光極大値波長として説明したが、吸光度ピークの極小点に対応する波長である吸光極小値波長として用いても同様である。
(3) 本実施例の分取クロマトグラフ装置の動作フロー
 本実施例の分取LC装置10の動作フローを、図6のチャートを用いて説明する。分取LC装置10は、ステップS7の条件を満たすまで、ステップS1~S7を繰り返し実行する。この繰り返しの動作の1ループについて、以下に説明する。
 まず、ピーク区間判定部171は、検出器15から吸光度スペクトルA(λ)を取得し(ステップS1)、現時点(t=taとする)における波長λ=λiの吸光度を、taにおけるクロマトグラムS(t)の値S(ta)として、それまでに得られているクロマトグラムS(t)に追加することにより、現時点taまでのクロマトグラムS(t)を作成する(ステップS2)。続いて、ピーク区間判定部171は、得られたS(t)に基づいて、上述の方法により、現時点が目的成分のピーク期間にあるか否かを判定する(ステップS3)。ここで、現時点が目的成分のピーク期間であればステップS4に進み、現時点が目的成分のピーク期間ではなければ、ステップS1に戻り、これまでに説明したステップS3までの動作を再度実行する。
 次に、微分値判定部172は、現時点で得られている吸光度スペクトルA(λ)に基づいて、吸光極大値波長(または吸光極小値波長)λ0における微分スペクトル値A'(λ0)を求め(ステップS4)、この微分スペクトル値A'(λ0)が0であるか否かを判定する(ステップS5)。
 ステップS5において微分スペクトル値A'(λ0)が0であると判定された場合にはステップS6-1に進み、フラクションコレクタ制御部173は、上述のように、溶出液を目的成分の溶液として分取する操作を実行する。ここで、前回までのループにおいて既に当該分取の操作を実行済である場合には、ステップS6-1ではその操作を維持するのみである。ステップS6-1の動作の終了後、ステップS7に進む。
 一方、ステップS5において微分スペクトル値A'(λ0)が0ではないと判定された場合には、ステップS6-2に進む。ステップS6-2では、フラクションコレクタ制御部173は、溶出液を目的成分の溶液として分取しないように、必要な操作を実行する。すなわち、その時点において分取中であれば電磁バルブ161を閉鎖し、分取中でなければそのままの状態を維持する。ステップS6-2の動作の終了後、ステップS7に進む。
 ステップS7では、操作の開始から所定時間が経過しているか否かを判定する。既知の目的成分では、カラムからの流出が終了する時間も既知であるため、この終了時間を前記所定時間とすればよい。所定時間が経過していなければ、ステップS1に戻ってこれまでの操作を再度実行し、所定時間が経過していれば、操作を終了する。
(4) 第1の変形例
 本発明は上記実施例には限定されない。
 上記実施例では、微分値判定部172は、目的成分xの吸光極大値波長λ0における微分スペクトル値A'(λ0)が0であるか否かを判定したが、その代わりに、以下のように判定を行ってもよい。
 前述の通り、通常、不純物成分yの吸光極大値波長(又は吸光極小値波長。以下同じ。)は目的成分xの吸光極大値波長λ0と相違している(図5)。そのため、溶出物が目的成分xのみを含有する(不純物成分yを含有しない)場合には、当該溶出物の吸光度スペクトルA(λ)の波長微分A'(λ)=d(A(λ))/dλの値は、当該吸光極大値波長λ0において0になる。一方溶出物が目的成分xだけではなく不純物成分yも含有している場合には、溶出物が目的成分xだけではなく不純物成分yも含有している場合には、当該溶出物のA'(λ)は、波長λ0以外の波長において0になる。
 従って、図7に示すように、微分値判定部172は、吸光度スペクトルA(λ)の波長微分A'(λ)の値が0になる波長λaを求めたうえで(ステップS4A)、波長λaが波長λ0と一致しているか否かを判定する(ステップS5A)ようにしてもよい。そして、波長λaが波長λ0と一致していれば上記ステップ6-1を実行し、波長λaが波長λ0と一致していなければ上記ステップ6-2を実行すればよい。なお、測定誤差等を考慮して、波長λaが波長λ0の差の絶対値がある程度の範囲内にあるときには上記ステップ6-1を実行するようにしてもよい。
(5) その他の変形例
 上記実施例では、検出器15として、フォトダイオードアレイを有する紫外分光光度計を用いたが、波長走査を行う紫外分光光度計を用いてもよいが、この場合には、波長スペクトルは同時に取得できず、走査する時間が必要である。また、検出器15には、赤外分光光度計検出器、分光蛍光検出器などの他の方式による検出器を用いてもよい。
 また、上記実施例では分取液体クロマトグラフ装置について説明したが、気体の試料を対象とするガスクロマトグラフにおいても同様の構成を取ることができる。なお、ガスクロマトグラフの場合には、検出器15には赤外分光光度計や近赤外分光光度計を用いることが望ましい。

Claims (4)

  1.  試料中の成分を時間的に分離して溶出物を溶出させるカラムと、該溶出物の吸光度スペクトルA(λ)を取得する検出器と、該吸光度スペクトルA(λ)に基づいてクロマトグラムS(t)をリアルタイムで作成してゆき、該クロマトグラムS(t)に基づいて前記溶出物から目的成分を分画するフラクションコレクタとを有する分取クロマトグラフ装置であって、
     a) 前記クロマトグラムS(t)に基づいて前記目的成分のクロマトグラムピークが現れたか否かを判定するピーク区間判定部と、
     b) 前記吸光度スペクトルA(λ)より、その波長微分d(A(λ))/dλの、前記目的成分の吸光度が極大値又は極小値を持つ既知の波長λ0における値である微分スペクトル値A'(λ0)=d(A(λ))/dλ|λ=λ0を求め、該微分スペクトル値A'(λ0) の絶対値が所定値以下であるか否かを判定する微分値判定部と、
     c) 前記目的成分のクロマトグラムピークが現れたと判定され、且つ、前記微分スペクトル値A'(λ0)の絶対値が前記所定値以下であると判定されている時間帯に、該溶出物を分画させるよう前記フラクションコレクタを制御するフラクションコレクタ制御部と
    を備えることを特徴とする分取クロマトグラフ装置。
  2.  試料中の成分を時間的に分離して溶出物を溶出させるカラムと、該溶出物の吸光度スペクトルA(λ)を取得する検出器と、該吸光度スペクトルA(λ)に基づいてクロマトグラムS(t)をリアルタイムで作成してゆき、該クロマトグラムS(t)に基づいて前記溶出物から目的成分を分画するフラクションコレクタとを有する分取クロマトグラフ装置であって、
     a) 前記クロマトグラムS(t)に基づいて前記目的成分のクロマトグラムピークが現れたか否かを判定するピーク区間判定部と、
     b) 前記吸光度スペクトルA(λ)より、その波長微分d(A(λ))/dλの値が0になる波長λaを求め、該波長λaと、前記目的成分の吸光度が極大値又は極小値を持つ既知の波長λ0の差の絶対値が所定値以下であるか否かを判定する微分値判定部と、
     c) 前記目的成分のクロマトグラムピークが現れたと判定され、且つ、波長λaと波長λ0の差の絶対値が前記所定値以下であると判定されている時間帯に、前記溶出物を分画させるよう前記フラクションコレクタを制御するフラクションコレクタ制御部と
    を備えることを特徴とする分取クロマトグラフ装置。
  3.  前記クロマトグラムS(t)が、前記目的成分に対応する吸光度スペクトルのピーク内にある特定の波長λiの吸光度A(λi)に基づいて作成されることを特徴とする請求項1又は2に記載の分取クロマトグラフ装置。
  4.  前記波長λiが前記波長λ0よりも短いことを特徴とする請求項3に記載の分取クロマトグラフ装置。
