Abstract
본 발명은 캠페리아 파비플로라 (Kaempferia parviflora Wall, ex Baker ) 추출물을 함유하는 근육 질환 치료용 약학적 조성물에 관한 것으로, 보다 상세하게는 캠페리아 파비플로라 추출물 또는 이로부터 분리된 플라본계 화합물을 유효성분으로 포함하는 근육 질환 치료용 약학적 조성물 또는 근 기능 개선용 식품 조성물에 관한 것이다. 본 발명은 캠페리아 파비플로라 추출물과 이로부터 분리된 플라본계 화합물은 근육 내 단백질의 이화 작용을 감소시키고 근육 세포 분화 및 동화 작용을 증가시키므로, 근육 질환 치료용 약학적 조성물 또는 근 기능 개선용 식품 조성물을 제조하는데 효과적이다.

본 발명은 캠페리아 파비플로라 (Kaempferia parviflora Wall, ex Baker) 추 출물 또는 플라본계 화합물의 근육 질환 예방 또는 치료 용도에 관한 것으로, 보다 상세하게는 캠페리아 파비플로라 추출물 또는 이로부터 분리된 플라본계 화합물을 유효성분으로 포함하는 근육 질환 치료용 약학적 조성물, 근 기능 개선용 식품 조 성물에 관한 것이다. -
【배경기술]
일반적으로 65-70세 인구 중 약 24%는 노화에 의한 근위축을 경험하고 있으 며, 20%정도가 근 기능이 상실되고 있는 것으로 알려지고 있다 (J. Gerontol. A Biol. Sci. Med. Sci. 60(2): 21316, 2005). 신체가 노화됨에 따라, 단백질을 합성 하는 작용인 동화 작용 (anabolism)에 관여하는 성장 호르몬, 성 호르몬, 유사 인슐 린 성장인자 등의 호르몬 분비가 감소되어 근육의 단백질 합성이 저하되고 순환 염 증성 사이토카인 (cytokine), 특히 종양괴사인자 -알파 (tumor necrosis factor-; T F-)의 수치가 증가하게 된다. 근섬유의 재생능력을 저하시키는 요인으로 염증성 사이토카인의 하나인 TNF- α는 손상된 근섬유와 융합하거나 분화를 억제하며, 근육 내 단백질을 분해하는 작용인 이화 작용 (catabolism)을 촉진하여 결국 골격근 조직 의 손실을 야기할 수 있다 (Clin. Nutr . 26(5): 52434, 2007) .
노화에 따른 만성 염증으로 근위축을 유발하는 주요 신호전달체계인 forkhead box(foxo)경로가 활성화되면 단백질 분해에 관여하는 E3 ubiquitin ligase인 atrogin-1과 MuRFl의 발현이 증가된다 (Cell Metab. 6(6): 47283, 2007). 반면, PI3K/AKT 신호전달 경로는 foxo의 활성을 억제시키고 단백질 합성 관련인자 mammalian target of rapamycin(mTOR)를 활성화시켜 근육성장을 유도한다 (Int. J. Biochem. Cell Biol. 37(10): 1985-96, 2005) . 특히, PI3K/AKT 및 mTOR는 운동에
의해 활성화되는 신호기작으로서 근육량을 증가시켜 운동능력을 향상시키는데 관여 한다 (Acta. Physiol. (Oxf). 191(1): 67-75, 2007) .
근육세포 분화는 myoD, myf5, myogenin, mrf4와 같은 다양한 근육 조절 민 자 (muscle regulatory factors; MRFs)에 의해 조절된다. MyoD는 근육 특이 유전자 의 발현을 개시하게 하고 중간엽줄기세포가 근육세포 계열로 분화하는 것을 유도한 다. Myogenin 발현의 유도는 myoD 활성에 의해 조절된다. 손상된 근육세포의 재생 은 노화 또는 만성 염증으로 발생한 근위축증을 예방 또는 치료할 수 있다 (J. Histochem. Cytochem. 54(11): 1177-91, 2006)
노화에 의한 근위축은 근 기능 자체의 문제만이 아니라 골다공증, 다양한 질병으로의 전이, 낙상과 관련된 부상 등과 밀접하게 관계되어있다. 따라서 가능한 노화로 인한 근육량의 손실을 제한하기 위하여 가능한 빠른 시기에 적절한 운동과 영양학적 처치가 중요하다 (Exp. Gerontol . 37(4): 477-89, 2002).
【발명의 상세한 설명】
[기술적 과제】
천연물 유래의 근 기능 조절 물질을 탐색한 결과, 캠페리아 파비플로라 (Kaempferia parvi flora Wall, ex Baker) 추출물 및 이에 포함된 플라본계 화합물 이 근육 내 단백질 이화 작용을 감소시키고 근육 세포 분화 및 동화 작용을 증가시 키는 활성이 있음을 발견하여 본 발명을 완성하게 되었다. 따라서 본 발명의 목적은 캠페리아 파비플로라 Tae/np/er/a parvi flora) 추출 물을 유효성분으로 포함하는 근육 질환 예방 또는 치료용 약학적 조성물을 제공한 다. 본 발명은 하기 화학식 4의 화합물 또는 이의 염을 유효성분으로 포함하는 근육 질환 예방또는 치료용 약학적 조성물을 제공한다.

본 발명의 조성물에서, 상기 5, 7-디메록시플라본, 5,7,4、-트리메록시플라 본, 및 3,5,7,3、4、-펜타메록시플라본은 화학적 합성방법에 의하여 합성되거나, 캠 페리아 파비플로라혹은 다른 식물로 부터 추출되어 분리된 것일 수 있다.
본 발명의 상기 플라본계 화합물은 그 자체 또는 염 또는 약학적으로 허용 가능한 염의 형태로 사용될 수 있다. 상기에서 '약학적으로 허용 가능한 '이란 생리 학적으로 허용되고 인간에게 투여될 때, 통상적으로 알레르기 반응 또는 이와 유사 한 반웅을 일으키지 않는 것을 말하며, 상기 염으로는 약제학적으로 허용 가능한 유리산 (free acid)에 의하여 형성된 산 부가염이 바람직하다. 상기 유리산으로는 유기산과 무기산을 사용할 수 있다. 상기 유기산은 이에 제한되는 것은 아니나, 구 연산, 초산, 젖산, 주석산, 말레인산, 푸마르산, 포름산, 프로피은산, 옥살산, 트 리플로오로아세트산, 벤조산, 글루콘산, 메타술폰산, 글리콜산, 숙신산, 4-틀루엔 술폰산, 글루탐산 및 아스파르트산을 포함한다. 또한 상기 무기산은 이에 제한되는 것은 아니나, 염산, 브름산, 황산 및 인산을 포함한다.
캠페리아파비플로라는 생강과 (Zingiberaceae family)의 식물의 일종으로서 블랙 진저 (black ginger)라고도 불린다. 본 발명의 캠페리아 파비플로라 추출물은 플라본계 화합물 특히, 다량의 5, 7-디메톡시플라본, 5,7,4、-트리메특시플라본, 3,5,7,3、4、-펜타메록시플라본을 포함한다.
캠페리아 파비플로라의 추출물로부터 본 발명의 플라본계 화합물의 분리 및 정제는 실리카겔 (silica gel)이나 활성 알루미나 (alumina)등의 각종 합성수지를 층 진한 컬럼 크로마토그래피 및 고성능 액체 크로마토그래피 (HPLC) 등을 단독으로 혹 은 병행하여 사용할 수 있으나, 추출 및 분리정제 방법이 반드시 상기 방법에 한정 되는 것은 아니다. 본 발명인이 캠페리아 파비플로라 추출물 및 이로부터 정제한 화학식 4의 화 합물로 근육 단백질 이화 작용 억제, 동화 작용 촉진, 근육 세포 분화 촉진 효과 및 근육량 증가 효과를 확인하였다 (실험예 1내지 3 참조).
따라서 상기 캠페리아 파비플로라 추출물 및 이로부터 정제한 화학식 4의 화 합물은 근육 질환을 치료할 수 있는떼, 상기 근육 질환은 근육 소모 또는 퇴화로 인한 질환을 의미한다. 상기 질환의 예로는 긴장감퇴증 (atony), 근위축증 (muscular atrophy), 근이영양증 (muscular dystrophy), 근육 퇴화, 근무력증 및 근육감소증 (Sacopenia)을 들 수 있다. 근육 소모는 근육량의 점진적 손실, 근육, 특히 골격근 또는 수의근 및 심장근육의 약화 및 퇴행을 특징으로 한다. 근육 소모 및 퇴화는

경피 투여제의 경우 연고제, 크림제, 로션제, 겔제, 외용액제, 파스타제, 리 니멘트제, 에어를제 등의 형태가 포함된다. 상기에서 "경피 투여"는약학적 조성물 을 국소적으로 피부에 투여하여 약학적 조성물에 함유된 유효한 양의 활성성분이 피부 내로 전달되는 것을 의미한다. 이들 제형은 제약 화학에 일반적으로 공자된 처방서인 ¾- Ί (Remington' s Pharmaceutical Science, 15th Edit ion, 1975, Mack Publishing Company, East on, Pennsylvania)에 기술되어 있다.

또한, 본 발명에 따른 약학적 조성물은 하나 이상의 완층제 (예를 들어, 식염 수 또는 PBS), 카보하이트레이트 (예를 들어, 글루코스, 만노즈, 슈크로즈 또는 덱 스트란), 항산화제, 정균제, 킬레아트화제 (예를 들어, EDTA 또는 글루타치은), 아 쥬반트 (예를 들어, 알루미늄 하이드록사이드), 현탁제, 농후제 및 /또는 보존제를 추가로 포함할수 있다.
본 발명의 추출물 또는 화합물의 바람직한 투여량은 환자의 상태 및 체중, 질병의 정도, 약물형태, 투여경로 및 기간에 따라 다르지만, 당업자에 의해 적절하 게 선택될 수 있다. 그러나 바람직한 효과를 위해서, 본 발명의 화합물은 0.001 내 지 300mg/day/체중 kg, 바람직하게는 0.01 내지 200mg/day/체중 kg으로 투여하는 것 이 좋다. 그러나, 상기 약학적으로 유효한 양은 질환 및 이의 중증정도, 환자의 연 령, 체중, 건강상태, 성별, 투여 경로 및 치료기간 등과 같은 여러 인자에 따라 적 절히 변화될 수 있다.
상기에서 "약학적으로 허용되는'' 이란 생리학적으로 허용되고 인간에게 투여 될 때, 활성성분의 작용을 저해하지 않으며 통상적으로 위장 장애 , 현기증과 같은 알레르기 반응 또는 이와 유사한 반응을 일으키지 않는 비독성의 조성물을 말한다.
상기 담체로는 모든 종류의 용매, 분산매질, 수증유 또는 유중수 에멀견, 수성 조 성물, 리포 좀, 마이크로비드 및 마이크로좀이 포함된다.

또한, 본 발명의 캠페리아 파비플로라 추출물 또는 화학식 4의 화합물 또는 이의 염을 식품 첨가제의 형태로 사용하기 위해서는 분말 또는 농축액 형태로 제조 하여 사용할 수 있다.

상기와 같이, 본 발명의 캠페리아 파비플로라 추출물과 이로부터 분리된 플 라본계 화합물은 근육 내 단백질의 이화 작용을 감소시키고 근육 세포 분화 및 동 화 작용을 증가시키므로, 근육 질환 치료용 약학적 조성물 또는 근 기능 개선용 식 품 조성물을 제조하는데 효과적이다.

단, 하기 실시 예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실 시 예에 한정되는 것은 아니다.
<실시예 1>
캠페리아파비플로라 (혹생강) 에탄올추출물의 체조 건조된 캠페리아 파비플로라 근경 (rhizome)을 믹서로 분쇄한 다음, 분쇄한 캠페리아 파비플로라 시료 100 g을 95% 에탄을 1L에 넣고 48시간 상은에서 넁침하 여 추출하였다. 추출된 시료는 와트만 (Whatman) 2번 여과지로 여과하고, 여과된 추 출액을 진공 회전 농축기로 농축하여 용매성분을 제거한 후 캠페리아 파비플로라 에탄올 추출물 (9.56 g)을 얻었다.
<실시예 2>
혹생강에탄올추출물로부터 폴라본계 화합물분리 <2-1> 5, 7-디메특시플라본의 분리 및 구조결정
상기 실시예 1에서 얻은 농축된 캠페리아 파비플로라 에탄올 추출물을 실리 카겔을 6x15 cm로 층진한 컬럼에 적재하고 에틸아세테이트 및 메탄을을 10:0.5 (v/v)의 비율로 흔합한 용매시스템을 이용하여 분취하였다. 상기 분취 순서에 따라 서 총 5개의 분획으로 나누어 각각의 분획을 농축 건조하였다. 5개의 분획 중 3번 분획 (분획 3)을 Rp-18 컬럼 크로마토그래피 (Lichroprep RP-18 25~40um, Merck & Co., 미국)를 이용하여 전개 용매 70% 메탄을로 분취하였다. 상기 분취 순서에 따 라서 총 2개의 분획으로 나누어 농축 건조하였다. 상기 2개의 분획 중 2번 분획 (분 획 3-2)을 실리카겔을 6x15 cm로 층진한 컬럼에 적재하고 에틸아세테이트 및 메탄 을을 10:0.4 (v/v)의 비율로 흔합한 용매시스템을 이용하여 분취하여 최종적으로 상기 2개의 분획 중 1번 분획 (분획 3-2-1)을 농축 건조시켜 자외선 차단물질을 분 리하였다.
상기에서 분리된 단일 활성물질의 구조결정을 위하여 -NMR스펙트럼과 13C-NMR 스펙트럼을 각각 500 MHz 와 125 MHz (용매: CDC13)에서 측정하였다. 본
화합물은 EI/MS에서 [M]가 m/z 282에서 관측되어 분자량이 282로 판명되었다. 이 상의 — NMR, 13C-NMR, 및 EI/MS에 대한 결과와 기존에 발표된 연구보고 (Sutthanut K. et al., J. Chromatogr A., 1143:22그 233, 2007)를 비교 분석하여 동정한 결과, 상기에서 분리된 단일물질은 하기 화학식 1로 표시되는 5, 7-디메특시플라본으로 확 인되었다.

USA)를 DMEM(10% FBS, 100 U/mL penicillin, 100 g/mL streptomycin)에서 배양하였 다. 세포밀도가 약 80~85%가 되었을 때 세포 배지를 2%의 FBS를 첨가한 DMEM 성장 배지로 교체하여 6일간 분화한 후 실험을 진행하였다. 분화된 L6 세포에 근 감소를 유도시키기 위하여 TNF- α를 24시간 처리한 후, 상기 실시예 2-1에서 제조한 5, 7- 디메특시플라본을 놈도별 (1, 5 μΜ)로 처리하였다. 근육 내 단백질 이화 작용에 관여하는 주요 ligase인 atrogin-1과 MuRFl의 mRNA 발현량을 알아보기 위해 RT-PCR 을 수행하였다. 분화한 세포로부터 1 12 시약(111 0 11, Carlsbad, CA, USA)을 사용하여 총 RNA를 수확하여 역전사시킨 후, RT-PCR 분석을 다음과 같이 수행하였 다. 먼저 cDNA 합성을 위해 상기 RNA를 reverse transcriptase로 역전사시켰다. RT-PCR은 다음의 특정 프라이머로 수행하였다:

<2-2>근육세포 단백질 동화작용증가효과
상기 실험예 1과 같은 방법으로 처리된 L6세포를 proteinase inhibitor cocktail이 포함된 RIPA 완층용액으로 용해시켰다. 상기시료를 5분간 끓인 후, 동 량의 단백질 (20 yg)을 10% SDS-PAGE로 전기영동하여 분리하였다. 전기영동 후 분 리된 단백질들을 니트로셀롤로스 막으로 전달하고 웨스턴 블롯을 수행하였다. 1차 항체에 반응시킨 다음 TBST를 이용하여 10분간 3회 세척하였다. 이때, 본 발명에서 사용된 1차 항체의 종류와 회석률은 1:1000이다. 2차 항체반웅은 상기 1차 항체반 웅올 수행한 막에 2차 항체 (ant i -rabbit horseradish)를 넣고 상온에서 2시간 동안 반응하였다. 이때 2차 항체의 회석률은 1:5000으로 하였다. Protein band는 ECL western blotting detection r eagent s ( Amer sham , Tokyo , Japan)를 사용하여 발색하 였다. 근육 내 단백질 동화 작용에 관여하는 PI3K, p-AKT, p-mTOR의 단백질 발현을 확인하였으며, a-tubulin으로 단백질 로딩량이 일정함을 나타내었다. 상기 방법과 동일하게 진행하되, 처리물질을 5, 7-디메록시플라본 외에도 캠페리아 파비플로라 추출물 (1, 10 g/ml)로 하여 진행하였고, 5, 7-디메록시플라본 처리 결과는 도 3,
캠페리아 파비플로라 추출물 처리 결과는 도 5에 표시하였다.
도 3 및 도 5에 나타난 바와 같이, 5,7-디메톡시플라본과 캠페리아 파비플로 라 추출물이 PI3K, p-AKT, p-mTOR의 단백질 발현량 증가를 알 수 있었다. 이러한 결과는 처리물질들이 근육량을 증가시키고 운동능력을 향상시키는 것을 의미한다.
<실험예 3>
동물모델에서 근육량증가효과확인
5주령된 Wi star rat을 1주간 적응시키고 TNF- α 100 ng/g을 2주간 공급하여 근위축을 유도시킨 다음, 체중을 기초로 무작위적으로 군을 배치하여 각각 8마리씩 총 4군으로 나누어 실험에 이용하였다. 실험군으로는 캠페리아 파비플로라 추출물 을 500 mg/kg체중, 5,7-디메특시플라본, 5,7,4、-트리메특시플라본, 3,5,7,3、4、-펜 타메특시플라본을 300 mg/kg체중의 농도로 0.25% 카르복시메틸셀를로오스 (carboxymethylcelhilose)에 현탁하여 1일 1회씩 8주간 일정한 시간에 투여하였다. 이때, 대조군으로는 실험군이 섭취하는 .동일한 양의 0.25% 카르복시메틸셀를로오스 에 TNF-α를 투여한 군을사용하였다.
8주간 시료를 투여한후 오른쪽 종아리 아래 근육을 절제하여 microbalance (Mettler PE160, USA)로 무게를 측정하였다, 그 결과, 표 1에 나타난 바와 같이, 대조군과 비교하여 캠페리아 파비플로라 추출물, 5,7-디메톡시플라본, 5,7,4、-트리 메특시플라본, 3,5,7,3、4、-펜타메록시플라본을 투여한 군에서 근육의 무게가 각각 유의적 (p<0.01)으로 125%, 126%, 118%, 120% 증가하였다. 이와 같은 결과로 본 발 명의 캠페리아 파비플로라 추출물과 이로부터 분리된 플라본계 화합물이 근육량 증 대에 효율적으로 작용하고 있음을 의미한다.

제 1항, 제 3항, 제 5항 및 제 7항 중 어느 한 항에 있어서, 상기 근육 질환은 긴장감퇴증 (atony), 근위축증 (muscular atrophy), 근이영양증 (muscular dystrophy), 근육 퇴화, 근무력증 및 근육감소증 (Sacopenia)으로 이루어진 군에서 선택되는 하나 이상의 질환인 것을 특징으로 하는약학적 조성물.
