Abstract
本発明の目的は、グルタミン酸若しくはその塩及びアルギニン若しくはその塩、又はグルタミン酸の塩基性アミノ酸塩を有効成分とし、該有効成分を高含有率で含有する錠剤であって、優れた溶出性を有し、且つ簡便に製造できる錠剤を提供することである。 本発明は、有効成分として、グルタミン酸若しくはその塩及びアルギニン若しくはその塩、又はグルタミン酸の塩基性アミノ酸塩を含有し、該有効成分の含有率が錠剤総重量に対して50~90重量%であり、結晶セルロースを実質的に含有しない錠剤を提供する。
Description
本発明は、グルタミン酸若しくはその塩及びアルギニン若しくはその塩、又はグルタミン酸の塩基性アミノ酸塩を有効成分とし、該有効成分を高含有率で含有する錠剤に関する。
錠剤は、1錠当たりに1回投与量の有効成分が含有され、さらに小型化されて、利用者(患者)の服用コンプライアンスを向上させ得る製剤が望まれている。
有効成分の薬理効果が発揮されるためには、錠剤は所望の部位で適切に崩壊し、錠剤中の有効成分が溶解して体内に吸収されることが必要である。錠剤のような固形製剤は、液状製剤に比べ、一般に化学的及び物理的に安定性に優れており、種々の製剤技術によって薬効の持続化や有効成分の吸収部位の調節が可能である。例えば、有効成分の作用の発現部位は、添加剤等を加えて成形することにより調節され得る。しかしながら、有効成分を高含有率で含有する錠剤では、一般に添加剤等の配合量が少ないことから、有効成分の溶出速度は低下すると言われている。
グルタミン酸及びアルギニンの併用、並びにグルタミン酸の塩基性アミノ酸塩は、機能性消化管障害の予防及び改善に有用であることが報告されている(特許文献1、2)。有効成分がグルタミン酸若しくはその塩及びアルギニン若しくはその塩、又はグルタミン酸の塩基性アミノ酸塩であり、有効成分の含有量が錠剤総重量に対して50重量%~90重量%であり、かつ崩壊剤を含有する錠剤が開示されている(特許文献3)。特許文献3には、錠剤の崩壊性および有効成分の溶出性に優れた錠剤の調製のために、錠剤の製造工程において、崩壊剤を水と接触しない方法で加えることが好ましいことが記載されている。
湿式造粒法による錠剤の製造において、崩壊剤を水と接触しない方法で加えるためには、例えば、有効成分と崩壊剤以外の添加剤を湿潤剤(水)を用いて造粒し、乾燥した後に、別途崩壊剤を添加する工程が必要になる。崩壊剤を湿式造粒工程で他の添加剤と同時に加えることができれば、工程が省略でき、錠剤の製造が簡便になる。そこで、本発明者らは、高含有率で有効成分を含有する錠剤であって、錠剤の製造工程において、たとえ崩壊剤を水と接触する条件下で加えた場合であっても、優れた溶出性を示す錠剤の開発に着目した。
本発明は、有効成分を高含有率で含有することで錠剤の小型化を達成すると共に、製造工程において、たとえ崩壊剤を水と接触する条件下で加えて製造した場合であっても、優れた溶出性を示す錠剤を提供することを目的とする。
本発明者らは、上記課題を解決すべく鋭意検討した結果、結晶セルロースを実質的に含有しないことで、たとえ崩壊剤を水と接触する条件下で加える工程を含んで製造した場合であっても、本発明の有効成分を高含有率で含有し、且つ優れた溶出性を示す錠剤を製造できることを見出した。かかる知見に基づき、本発明者らは、さらに研究を重ねることによって本発明を完成するに至った。
即ち、本発明は以下に関する。
〔1〕有効成分として、グルタミン酸若しくはその塩及びアルギニン若しくはその塩、又はグルタミン酸の塩基性アミノ酸塩を含有し、該有効成分の含有率が錠剤総重量に対して50~90重量%であり、結晶セルロースを実質的に含有しない、錠剤。
〔2〕グルタミン酸の塩基性アミノ酸塩がグルタミン酸アルギニンである、〔1〕に記載の錠剤。
〔3〕有効成分の含有率が錠剤総重量に対して80~90重量%である、〔1〕又は〔2〕に記載の錠剤。
〔4〕結合剤を更に含有し、該結合剤の含有率が錠剤総重量に対して5重量%以下である、〔1〕~〔3〕のいずれか1つに記載の錠剤。
〔5〕結合剤がヒドロキシプロピルセルロースである、〔4〕に記載の錠剤。
〔6〕クロスポビドン、カルボキシメチルスターチナトリウム及びクロスカルメロースナトリウムからなる群より選択される少なくとも1種の崩壊剤を更に含有する、〔1〕~〔5〕のいずれか1つに記載の錠剤。
〔7〕更に崩壊剤を含有し、崩壊剤が水と接触する条件下で製造されてなる、〔1〕~〔5〕のいずれか1つに記載の錠剤。
〔8〕崩壊剤の含有率が錠剤総重量に対して10重量%以下である、〔6〕又は〔7〕に記載の錠剤。
〔9〕有効成分と崩壊剤とが造粒された造粒物を含有する、〔6〕~〔8〕のいずれか1つに記載の錠剤。
〔10〕錠剤総重量が400mg以下である、〔1〕~〔9〕のいずれか1つに記載の錠剤。
〔11〕流動層造粒法により得られた造粒物を打錠することによって製造される、〔1〕~〔10〕のいずれか1つに記載の錠剤。
〔12〕コーティング処理が施された〔1〕~〔11〕のいずれか1つに記載の錠剤。
〔13〕錠剤が素錠であり、第15改正日本薬局方溶出試験法でパドル法を用い、パドル回転数を毎分50回転とし、37℃の水900mLを溶出試験液とする溶出試験において、溶出時間が5分における有効成分の溶出率が60%以上である、〔1〕~〔11〕のいずれか1つに記載の錠剤。
〔14〕機能性消化管障害の予防又は改善用である、〔1〕~〔13〕のいずれか1つに記載の錠剤。
〔15〕グルタミン酸若しくはその塩及びアルギニン若しくはその塩が、L-グルタミン酸若しくはその塩及びL-アルギニン若しくはその塩であり、グルタミン酸の塩基性アミノ酸塩がL-グルタミン酸L-アルギニンである、〔1〕~〔14〕のいずれか1つに記載の錠剤。
〔1〕有効成分として、グルタミン酸若しくはその塩及びアルギニン若しくはその塩、又はグルタミン酸の塩基性アミノ酸塩を含有し、該有効成分の含有率が錠剤総重量に対して50~90重量%であり、結晶セルロースを実質的に含有しない、錠剤。

本発明の錠剤は、有効成分を高含有率で含有すると共に、たとえ崩壊剤を水と接触する方法で加える工程を含んで製造した場合であっても、優れた溶出性を示す。このように、本発明によれば、有効成分を高含有率で含有し、優れた溶出性を示し、且つ簡便に製造できる錠剤を提供できる。本発明によれば、有効成分を高含有率で含有できるので、錠剤を小型化でき、その結果服用コンプライアンスを向上させ得る。
本発明の錠剤は、消化管、特に胃の中において優れた溶出性を示すため、優れたバイオアベイラビリティーを有する機能性消化管障害の予防又は改善剤として有用である。
本発明の錠剤は、有効成分として、グルタミン酸若しくはその塩及びアルギニン若しくはその塩、又はグルタミン酸の塩基性アミノ酸塩を含有する。有効成分は一種又は二種以上を組み合せて使用してもよい。
グルタミン酸、アルギニン及びそれらの塩は、動物や植物に由来する天然のもの、或いは化学合成法、発酵法、遺伝子組換え法によって得られるもののいずれを使用してもよい。
グルタミン酸及びその塩は、L体、D体、これらの混合物(例えば、ラセミ体)のいずれでもよいが、L体が好適に使用される。
またアルギニン及びその塩は、L体、D体、これらの混合物(例えば、ラセミ体)のいずれでもよいが、L体が好適に使用される。
本発明におけるグルタミン酸の塩及びアルギニンの塩としては、薬理的に許容される塩が挙げられる。このような塩としては、無機塩基との塩、無機酸との塩、有機酸との塩、塩基性アミノ酸との塩、及び酸性アミノ酸との塩等が挙げられる。
無機塩基との塩としては、ナトリウム、カリウム、リチウム等のアルカリ金属との塩、カルシウム、マグネシウム等のアルカリ土類金属との塩、アンモニウムとの塩等が挙げられる。
無機酸との塩としては、ハロゲン化水素酸(塩酸、臭化水素酸、ヨウ化水素酸等)、硫酸、硝酸、リン酸等との塩が挙げられる。
有機酸との塩としては、ギ酸、酢酸、プロピオン酸、シュウ酸、コハク酸、マレイン酸、フマル酸、クエン酸等との塩が挙げられる。
塩基性アミノ酸との塩としては、アルギニン、リジン、ヒスチジン及びオルニチン等との塩が挙げられる。
酸性アミノ酸との塩としては、グルタミン酸、アスパラギン酸等との塩が挙げられる。
グルタミン酸の塩としては、ナトリウム塩等のアルカリ金属との塩、及びアルギニン、ヒスチジン等の塩基性アミノ酸との塩が好適である。アルギニンの塩としては、塩酸等の無機塩との塩が好適である。
無機酸との塩としては、ハロゲン化水素酸(塩酸、臭化水素酸、ヨウ化水素酸等)、硫酸、硝酸、リン酸等との塩が挙げられる。
有機酸との塩としては、ギ酸、酢酸、プロピオン酸、シュウ酸、コハク酸、マレイン酸、フマル酸、クエン酸等との塩が挙げられる。
塩基性アミノ酸との塩としては、アルギニン、リジン、ヒスチジン及びオルニチン等との塩が挙げられる。
酸性アミノ酸との塩としては、グルタミン酸、アスパラギン酸等との塩が挙げられる。
グルタミン酸の塩としては、ナトリウム塩等のアルカリ金属との塩、及びアルギニン、ヒスチジン等の塩基性アミノ酸との塩が好適である。アルギニンの塩としては、塩酸等の無機塩との塩が好適である。
本発明におけるグルタミン酸又はその塩としては、具体的には、グルタミン酸、グルタミン酸アルギニン、L-グルタミン酸ナトリウム、D-グルタミン酸ナトリウム等が挙げられる。なかでも、グルタミン酸、グルタミン酸アルギニン、L-グルタミン酸ナトリウム等が好ましい。
本発明におけるアルギニン又はその塩としては、具体的には、アルギニン、アルギニン塩酸塩等が挙げられる。
本発明におけるグルタミン酸の塩基性アミノ酸塩としては、アルギニン、リジン又はオルニチン等の塩基性アミノ酸との塩が挙げられる。なかでもアルギニンとの塩(グルタミン酸アルギニン)が好ましい。
グルタミン酸アルギニンとは、グルタミン酸とアルギニンとが塩形成してなる公知物質であり、具体的には以下の化学式で表される。
本発明においてグルタミン酸アルギニンは、L-グルタミン酸とL-アルギニンとの結合体(即ち、L-グルタミン酸L-アルギニン)であることが好ましい。グルタミン酸アルギニンは、公知の方法に準じて製造することができ、市販品を用いることもできる。グルタミン酸アルギニンは、必要に応じて水和物及び溶媒和物とすることができる。
本発明の有効成分の含有率(有効成分が2種以上の場合は、合計した上での割合)は、錠剤の総重量に対して、通常50~90重量%であり、好ましくは55~90重量%、より好ましくは60~90重量%、更に好ましくは65~90重量%、特に好ましくは70~90重量%、最も好ましくは80~90重量%である。尚、本明細書中の錠剤の総重量は、特に言及する場合を除き、コーティングの重量は含まず、素錠の総重量を意味する。上記有効成分が、塩、水和物又は溶媒和物を形成している場合は、これらをフリー体に換算して、該有効成分の含有率を算出する。
本発明の錠剤において、グルタミン酸又はその塩とアルギニン又はその塩との割合は、フリー体に換算して、通常、グルタミン酸とアルギニンとが10:1~1:10のモル比で配合されている。なかでも5:1~1:5の範囲が好ましく、2:1~1:2の範囲がより好ましく、1:1が特に好ましい。
本発明の錠剤は、結晶セルロースを実質的に含まないことを特徴としている。
結晶セルロースは錠剤の成形性に大きく寄与しており、一般に、結晶セルロースを用いることにより錠剤中の粒子(顆粒)が絡み合い、錠剤が容易に成形できることが知られている。また、結晶セルロース自体には味はなく、化学的にも不活性であることから、結晶セルロースは、錠剤の賦形剤として一般的に利用されている。
本発明においては、結晶セルロースを実質的に含有しないことで、製造工程において、たとえ崩壊剤を水と接触する方法で加えた場合であっても、本発明の有効成分を高含有率で含有し、且つ優れた溶出性を示す錠剤を製造できる。
本発明においては、結晶セルロースを実質的に含有しなくても、後述のように添加剤を選択することにより十分な成形性を確保でき、結晶セルロースを用いる場合と同等程度の錠剤硬度を有する錠剤の製造が可能である。
結晶セルロースは錠剤の成形性に大きく寄与しており、一般に、結晶セルロースを用いることにより錠剤中の粒子(顆粒)が絡み合い、錠剤が容易に成形できることが知られている。また、結晶セルロース自体には味はなく、化学的にも不活性であることから、結晶セルロースは、錠剤の賦形剤として一般的に利用されている。
本発明においては、結晶セルロースを実質的に含有しないことで、製造工程において、たとえ崩壊剤を水と接触する方法で加えた場合であっても、本発明の有効成分を高含有率で含有し、且つ優れた溶出性を示す錠剤を製造できる。
本発明においては、結晶セルロースを実質的に含有しなくても、後述のように添加剤を選択することにより十分な成形性を確保でき、結晶セルロースを用いる場合と同等程度の錠剤硬度を有する錠剤の製造が可能である。
本発明において「実質的に含有しない」とは、本発明の効果(高い有効成分含有率、錠剤の小型化及び有効成分の溶出性の向上等)に影響を与える程度の量を含まないことを意味する。具体的には、結晶セルロースの含有率は、錠剤総重量に対し、通常10重量%以下、好ましくは8重量%以下、より好ましくは6重量%以下、さらに好ましくは4重量%以下、最も好ましくは2重量%以下である。
本発明の錠剤の総重量は、通常400mg以下、好ましくは390mg以下、より好ましくは380mg以下、さらに好ましくは370mg以下である。また、通常330mg以上である。
本発明の錠剤は、上記有効成分以外に、製剤上の必要に応じて任意の添加剤、例えば、薬学的に許容され得る担体を含むことができる。薬学的に許容され得る担体としては、例えば、賦形剤、結合剤、崩壊剤、滑沢剤、甘味剤、色素、香料、溶剤等が挙げられる。後述の各種添加剤は、それぞれ、一種のみで用いてもよく、二種以上を組み合わせてもよい。
本発明の錠剤においては、結合剤を配合することが好ましい。結合剤としては、ヒドロキシプロピルセルロース、トラガントガム、アラビアゴム、コーンスターチ、ゼラチン、ヒプロメロース、ポビドン、エチルセルロース、ポリビニルアルコール、グァーガム、低置換度ヒドロキシプロピルセルロース、メチルセルロース等が例示される。本発明では、結晶セルロースを実質的に含有せずとも錠剤の十分な成形性を獲得するという観点から、これらの中でもヒドロキシプロピルセルロースが特に好ましい。
上記結合剤の含有率は、錠剤総重量に対して通常10重量%以下、好ましくは8重量%以下、より好ましくは6重量%以下、さらに好ましくは5重量%以下、特に好ましくは4重量%以下である。また、通常1重量%以上、好ましくは2重量%以上である。結合剤の含有率が上記範囲内であることにより、有効成分を高含有率で含有すると共に優れた溶出性を示す錠剤を容易に製造することができる。
本発明の錠剤は、崩壊剤を含有することが好ましい。崩壊剤としては、クロスポビドン、カルボキシメチルスターチナトリウム、クロスカルメロースナトリウム、カルメロース又はその塩、低置換度ヒドロキシプロピルセルロース、コーンスターチ、馬鈴薯澱粉、アルギン酸等が例示される。本発明では、錠剤の崩壊性の観点から、クロスポビドン、カルボキシメチルスターチナトリウム及びクロスカルメロースナトリウムが好ましく、これらの中でも、有効成分を高含有率で含有させると共に、有効成分の溶出性をより向上させるという観点から、クロスポビドンが特に好ましい。
崩壊剤の含有率は、特に限定されないが、錠剤総重量に対して通常40重量%以下である。該含有率は、好ましくは30重量%以下、より好ましくは20重量%以下、さらに好ましくは10重量%以下、さらにより好ましくは10重量%未満、特に好ましくは8重量%以下、最も好ましくは6重量%以下であり、また、通常1重量%以上である。崩壊剤の含有率が上記範囲内であることにより、有効成分を高含有量で含有すると共に優れた溶出性を示す錠剤を容易に製造することができる。また、後述するように、錠剤の製造効率をさらに高めることができる。
その他の担体としては、部分アルファー化デンプン、二酸化珪素、リン酸二カルシウム等の賦形剤(好ましくは、部分アルファー化デンプン);ステアリン酸マグネシウム等の滑沢剤;スクロース等の甘味剤;色素;オレンジフレーバー等の香料;水、エタノール、グリセロール等の溶剤等を適宜使用することができる。
本発明において、賦形剤の含有率は、錠剤総重量に対して通常0~50重量%、好ましくは0~40重量%、より好ましくは0~30重量%である。滑沢剤の含有率は、錠剤総重量に対して通常0~5重量%、好ましくは0~4重量%、より好ましくは0~3重量%である。
本発明において、賦形剤の含有率は、錠剤総重量に対して通常0~50重量%、好ましくは0~40重量%、より好ましくは0~30重量%である。滑沢剤の含有率は、錠剤総重量に対して通常0~5重量%、好ましくは0~4重量%、より好ましくは0~3重量%である。
また、蛋白質、アミノ酸、ビタミン、脂質、グルコース等の栄養素も担体として使用可能である。
さらに、医薬担体として、システイン、グルタチオン、アスコルビン酸、メタ亜硫酸ナトリウム、重亜硫酸ナトリウム等の薬学的に許容される抗酸化剤、炭酸カルシウム、水酸化アルミニウムゲル、ケイ酸アルミニウム等の酸中和剤が挙げられる。
さらに、医薬担体として、システイン、グルタチオン、アスコルビン酸、メタ亜硫酸ナトリウム、重亜硫酸ナトリウム等の薬学的に許容される抗酸化剤、炭酸カルシウム、水酸化アルミニウムゲル、ケイ酸アルミニウム等の酸中和剤が挙げられる。
本発明の錠剤は、経口的又は経腸的に投与することができる。投与方法としては経口投与が好ましく、さらに薬物徐放形態であってもよい。本発明の錠剤としては、素錠、糖衣錠、フィルムコーティング錠、口腔内崩壊錠、腸溶コーティング錠等が例示されるが、これらに限定されない。尚、上述した担体や錠剤の剤形は、当業者によく知られたものであり、例えば、Reimington's Pharmaceutical Science, ed. 16(1980), Mack Publishing Companyに記載されるような担体及び剤形を採用することができる。
本発明の錠剤は、素錠そのままであってもよく、或いは素錠にコーティング処理が施されたコーティング錠であってもよい。本発明の錠剤がコーティング錠であれば、服用性及び製剤強度等をさらに高めることができる。本発明の錠剤がコーティング錠である場合、そのコーティング量については、特に限定されるものではないが、素錠総重量(100重量部)に対して、好ましくは0.01重量部~50重量部、より好ましくは0.1重量部~20重量部、さらに好ましくは1重量部~10重量部である。
後述のコーティング基材、コーティング添加剤は、それぞれ、一種のみで用いてもよく、二種以上を組み合わせてもよい。

コーティング基剤の好適な例としては、糖衣基剤、フィルムコーティング基剤等が挙げられる。
糖衣基剤としては、例えば、白糖が挙げられ、さらに、タルク、沈降炭酸カルシウム、ゼラチン、アラビアゴム、プルラン、カルナバロウ等から選ばれる1種又は2種以上を併用してもよい。
フィルムコーティング基剤としては、例えば、ヒドロキシプロピルセルロース、ヒドロキシプロピルメチルセルロース(例、ヒプロメロース2910)、ヒドロキシエチルセルロース、メチルヒドロキシエチルセルロース等のセルロース系高分子;ポリビニルアセタールジエチルアミノアセテート、アミノアルキルメタアクリレートコポリマーE〔オイドラギットE(商品名)〕、ポリビニルピロリドン等の合成高分子;プルラン等の多糖類等が挙げられる。
コーティング添加剤の好適な例としては、酸化チタン、タルク、三二酸化鉄、黄色三二酸化鉄等の遮光剤及び/又は着色剤;ポリエチレングリコール(例、マクロゴール6000)、クエン酸トリエチル、ヒマシ油、ポリソルベート類等の可塑剤;クエン酸、酒石酸、リンゴ酸、アスコルビン酸等の有機酸等が挙げられる。
糖衣基剤としては、例えば、白糖が挙げられ、さらに、タルク、沈降炭酸カルシウム、ゼラチン、アラビアゴム、プルラン、カルナバロウ等から選ばれる1種又は2種以上を併用してもよい。
フィルムコーティング基剤としては、例えば、ヒドロキシプロピルセルロース、ヒドロキシプロピルメチルセルロース(例、ヒプロメロース2910)、ヒドロキシエチルセルロース、メチルヒドロキシエチルセルロース等のセルロース系高分子;ポリビニルアセタールジエチルアミノアセテート、アミノアルキルメタアクリレートコポリマーE〔オイドラギットE(商品名)〕、ポリビニルピロリドン等の合成高分子;プルラン等の多糖類等が挙げられる。
コーティング添加剤の好適な例としては、酸化チタン、タルク、三二酸化鉄、黄色三二酸化鉄等の遮光剤及び/又は着色剤;ポリエチレングリコール(例、マクロゴール6000)、クエン酸トリエチル、ヒマシ油、ポリソルベート類等の可塑剤;クエン酸、酒石酸、リンゴ酸、アスコルビン酸等の有機酸等が挙げられる。
本発明の錠剤は、例えば、機能性消化管障害の予防又は改善に使用することができる。患者のQOL(quality of life)を低下させる再現性のある機能性消化管障害、特に機能性胃腸症、胃食道逆流症等の上部消化管機能障害における下記記載の症状の発生の予防又は症状の改善に有効である。
「機能性消化管障害」とは、明らかな器質的変化がないにもかかわらず消化管機能が低下しているような状態をいい、消化管(咽頭、食道、胃、小腸(十二指腸、空腸、回腸)、大腸)のあらゆる機能の低下を言う。Rome IIIにおいては、「機能性消化管障害」は、AからHまでの区分がされており、機能性食道障害、機能性胃十二指腸障害、機能性腸障害、機能性腹痛症候群、機能性胆嚢・オッディ括約筋障害、機能性直腸肛門障害、新生児及び乳幼児の機能障害、小児、青年期の機能性障害と定義されている。
「消化管」とは、口腔から肛門までの一連の消化に携わる管腔臓器をいい、例えば、咽頭、食道、胃、小腸(十二指腸、空腸、回腸)、大腸が挙げられる。さらに「上部消化管」とは、咽頭、食道、胃、十二指腸をいう。
「機能性胃腸症」とは、これまで胃排出遅延が認められる慢性胃炎や胃炎として診断されてきた疾患をいい、腹部痛、胃もたれ、胸やけ等の症状を呈することを特徴とする。Rome IIIによると、煩わしい食後膨満感、早期飽満感、心窩部痛、心窩部灼熱感のうち一症状以上が慢性的に(厳密には3ヶ月以上)みられ、且つ器質的疾患が除外された場合に機能性胃腸症と診断される。
「胃食道逆流症」は逆流性食道炎も含み、胃酸が逆流することで発症し、胸やけ、胃酸が口まで上がってくる等の特有の症状がある症状をいう。
本発明の錠剤により予防又は改善可能な機能性消化管障害の具体的な症状(不定愁訴)としては、悪心、嘔吐、吐き気、胸焼け、膨満感、胃もたれ、ゲップ、胸中苦悶感、胸痛、胃部不快感、食欲不振等の代表的な上部消化管不定愁訴、腹痛、便秘、下痢等の下部消化管不定愁訴及び関連した愁訴、例えば、息切れ、息苦しさ、意欲低下、喉頭閉塞・異物感(漢方でいう「梅核気」)、易疲労感、肩こり、緊張、口のかわき(口渇・口乾)、呼吸促迫、四肢熱感・冷感、集中困難、焦燥感、睡眠障害、頭痛、全身倦怠感、動悸、寝汗、不安感、ふらつき感、めまい感、熱感、のぼせ、発汗、腹痛、便秘、抑鬱感等が挙げられる。
本発明の錠剤は、ヒト、動物(例えば、ウシ、ウマ、ブタ、ヒツジ、イヌ、トリ等の家畜や家禽、及びマウス、ラット等の実験動物)に安全に投与することができる。
本発明の錠剤は、上記有効成分の有効量を投与対象に投与することで機能性消化管障害等を改善することができる。
本発明において、有効成分の投与量は、投与対象の性別、年齢、体重、食餌、投与の形態、消化管の器質性疾患の症状等によって異なるが、例えば、成人(体重60kg、以下同様)1日当たりの投与量は、グルタミン酸としては、通常0.01~10gであり、0.1~3gが好ましい。アルギニンとしては、成人1日当たりの投与量は、通常0.01~10gであり、0.1~3gが好ましい。グルタミン酸の塩及びアルギニンの塩の場合は、それぞれフリー体に換算した上で投与量が前記範囲内であればよい。
グルタミン酸とアルギニンとをあわせた有効成分の成人1日当たりの投与量は、通常0.01~20gであり、0.02~20gが好ましく、0.02~10gがより好ましく、0.1~6gが更に好ましい。グルタミン酸の塩及びアルギニンの塩の場合は、それぞれフリー体に換算した上で投与量が前記範囲内であればよい。
上述した有効成分の1日当たりの投与量は1回又は数回に分けることができる。本発明の錠剤は、上記有効成分の1回の投与量を1錠に含有するように設計することが可能である。
本発明の錠剤は、他の薬剤を併用してもよく、かかる薬剤として、H2受容体拮抗薬、プロトンポンプ阻害薬等の酸分泌抑制剤、5-HT受容体作用剤、D2拮抗剤等の運動機能改善剤、ムスカリン受容体拮抗薬、抗ガストリン薬、抗コリン薬等の制酸剤、テプレノン、プラウノトール、オルノプロスチル、エンプロスチル、ミソプロストール、レバミピド、スクラルファート、ポラプレジンク、アズレン、エグアレンナトリウム、グルタミン、アルジオキサ、ゲファルナート、エカベトナトリウム等の粘膜保護剤、スルファサラジン、5-ASA製剤、ステロイド、レミケード等の炎症性大腸炎治療剤を含有することができる。これらは一種又は二種以上を含有することができる。また本発明の錠剤と他の薬剤は、両者を単一製剤又は別個の製剤としてもよく、同時投与又は時間差をつけて投与してもよい。
本発明の錠剤はまた、本発明の有効成分を含有し、機能性消化管障害の改善、機能性胃腸症又は胃食道逆流症の予防又は改善といった特定の目的のために摂取する食品であってもよい。また、いわゆる健康食品を含む一般食品としてもよい。さらに、本発明の錠剤は、消費者庁の特別用途食品制度に規定される、特定用途食品(病者用食品、特定保健用食品)、さらにダイエタリーサプリメント(栄養補助食品)とすることができる。この場合、グルタミン酸及びアルギニン、又はそれらの塩を2種以上混合して使用することができる。
本発明の錠剤を上述した食品として使用する場合、グルタミン又はその塩の摂取量、アルギニン又はその塩の摂取量、グルタミン又はその塩とアルギニン又はその塩とを合わせた摂取量、グルタミン酸の塩基性アミノ酸塩の摂取量はそれぞれ、成人1日当たりについて、上記に示した成人1日当たりの投与量と同様である。
本明細書において「1回投与量」及び「1回当たりの投与量」は、それぞれ「1食摂取量」及び「1食当たりの摂取量」に置き換えて解釈することができる。
次に本発明の錠剤の製造方法を説明する。尚、以下の製造方法は、本発明において一例を示すものであって、特に限定されるわけではなく、当業者であれば所望により適宜変更及び修飾することができる。
本発明の錠剤は、上述の有効成分、添加剤を原料として、以下の(1)~(3)の工程により製造することができる。
(1)有効成分、及び任意に添加してもよい添加剤(例えば、崩壊剤(例えば、クロスポビドン)、賦形剤(例えば、部分アルファー化デンプン)、結合剤(例えば、ヒドロキシプロピルセルロース))を混合し、造粒する工程、
(2)工程(1)により得られた造粒物を必要により整粒する工程、並びに
(3)工程(2)により得られた顆粒を、任意に添加してもよい添加剤(例えば、滑沢剤(例えば、ステアリン酸マグネシウム))と混合し、打錠する工程。
(1)有効成分、及び任意に添加してもよい添加剤(例えば、崩壊剤(例えば、クロスポビドン)、賦形剤(例えば、部分アルファー化デンプン)、結合剤(例えば、ヒドロキシプロピルセルロース))を混合し、造粒する工程、
(2)工程(1)により得られた造粒物を必要により整粒する工程、並びに
(3)工程(2)により得られた顆粒を、任意に添加してもよい添加剤(例えば、滑沢剤(例えば、ステアリン酸マグネシウム))と混合し、打錠する工程。
工程(1)の原料として、有効成分は、適度な大きさ(例えば、平均粒子径が500μm以下)に粉砕されたものが好適に用いられる。このような有効成分の粉砕物は、公知の方法により粉砕して得ることができるが、既に粉砕済みの市販品を用いることもできる。
工程(1)における混合、造粒は、自体公知の方法により行うことができる。混合は、例えば、V型混合機、タンブラー混合機、コンテナ混合機等の混合機を用いて行うことができる。造粒は、例えば、流動層造粒乾燥機、高速攪拌造粒機、押し出し造粒機等を用いて行うことができる。本発明の錠剤は、流動層造粒法により造粒されるのが好ましい。
工程(1)における混合、造粒は、自体公知の方法により行うことができる。混合は、例えば、V型混合機、タンブラー混合機、コンテナ混合機等の混合機を用いて行うことができる。造粒は、例えば、流動層造粒乾燥機、高速攪拌造粒機、押し出し造粒機等を用いて行うことができる。本発明の錠剤は、流動層造粒法により造粒されるのが好ましい。
工程(1)における造粒は、湿式造粒法及び乾式造粒法のいずれを用いてもよいが、造粒のしやすさや得られる造粒物の性質等の観点から湿式造粒法が好適に用いられる。ここで、本明細書において「湿式造粒法」とは、有効成分それ自体に、或いは該有効成分に賦形剤、崩壊剤、結合剤等の適当な添加剤を加えた混合物に、適当な湿潤剤を加えて又は噴霧しながら造粒し、造粒物を製造する方法をいう。また、本明細書において「乾式造粒法」とは、有効成分をそのまま、或いは該有効成分に賦形剤、崩壊剤、結合剤等の適当な添加剤を加えた混合物を、湿潤剤を加えることなく造粒し、造粒物を製造する方法をいう。
本発明において湿式造粒法が用いられる場合、湿潤剤としては、好ましくは水(特に精製水)が用いられる。また、さらに好ましくは、結合剤(例えばヒドロキシプロピルセルロース)が予め溶解された水溶液が用いられる。湿潤剤は、好ましくは有効成分等に対して噴霧される。
ここで、崩壊剤は吸水により膨潤し、錠剤を崩壊する作用を有するが、結晶セルロースも同様の性質を有している。従って、崩壊剤等と水とが接触する工程を含む製造方法で製造された錠剤では有効成分の溶出性が低下すると考えられた。それゆえ、従前の製造方法では崩壊剤及び結晶セルロースは、特に湿式造粒法による造粒工程では添加されず、得られた造粒物を乾燥、整粒後に別途工程を設けて添加・混合されていた。しかしながら、結晶セルロースを実質的に含有しない本発明の錠剤であれば、驚くべきことに、崩壊剤を湿式造粒法による造粒工程において添加・混合しても、有効成分の溶出性が低下しない錠剤の製造が可能となることが本発明者らにより初めて見出された。本発明の錠剤は、湿式造粒法のような、崩壊剤が水と接触する条件下で製造しても、優れた溶出性を示す。従って、本発明の錠剤は、特に湿式造粒法を利用した製造方法において好適である。
工程(2)では、周知の整粒機等を用いて、造粒物の平均粒子径が100μm~500μm程度に整粒すればよい。
工程(3)では、上記工程(2)で得られた顆粒、或いはこれと滑沢剤等の添加剤との混合物が打錠される。混合、打錠は、自体公知の方法により行うことができる。混合は、例えば、上述の混合機を用いて行うことができる。打錠は、例えば、単発錠剤機、ロータリー式打錠機等を用いて行うことができる。
本発明の錠剤は、上述した通りコーティング処理が施されていてもよい。コーティング操作は、特に限定されず、当業者に周知のコーティング装置等を用いて、自体公知の方法により行うことができる。
本発明の錠剤の溶出率は、素錠の場合、下記測定条件下で、好ましくは、5分で60%以上であり、より好ましくは、5分で65%以上であり、さらに好ましくは、5分で70%以上である。なお、コーティングされた錠剤の場合は、コーティング基剤の性状等によって、これらの溶出率は当然変動しうるものである。
測定方法(測定条件)は、日本薬局法第15改正一般試験法・溶出試験法・装置2パドル法を用い、パドル回転数は毎分50回転、37℃の水900mLを溶出試験液として用いる。試験開始後5分の溶出試験液中の有効成分量をアミノ酸アナライザー法又は高速液体クロマトグラフィー法にて定量し、溶出率を算出する。
本発明の錠剤の硬度は、素錠の場合、好ましくは50~100N、より好ましくは80~100Nである。
錠剤の硬度は、例えば錠剤硬度測定装置(TABLET TESTER 8M:フロイント産業(株))により、測定できる。
測定方法(測定条件)は、日本薬局法第15改正一般試験法・溶出試験法・装置2パドル法を用い、パドル回転数は毎分50回転、37℃の水900mLを溶出試験液として用いる。試験開始後5分の溶出試験液中の有効成分量をアミノ酸アナライザー法又は高速液体クロマトグラフィー法にて定量し、溶出率を算出する。
本発明の錠剤の硬度は、素錠の場合、好ましくは50~100N、より好ましくは80~100Nである。
錠剤の硬度は、例えば錠剤硬度測定装置(TABLET TESTER 8M:フロイント産業(株))により、測定できる。
以下、実施例により更に詳細に説明するが、本発明はこれらの実施例に限定されるものではない。
(実施例1)流動層造粒法
(1)L-グルタミン酸L-アルギニン(水分値3.64%)2490.7g、部分アルファー化デンプン101.3g、クロスポビドン168.0gを流動層造粒乾燥機(フローコーター5型:フロイント産業(株))中で混合した後、ヒドロキシプロピルセルロース96.0gを含有する水溶液(1200g)を加えて造粒し、これを乾燥して造粒顆粒を得た。
(2)得られた造粒顆粒を整粒し、整粒顆粒を得た。
(3)得られた整粒顆粒495.8gに対して、ステアリン酸マグネシウム4.2gを加え、混合し、打錠用顆粒を得た。
(4)得られた打錠用顆粒を打錠し、素錠を得た。
(5)水90重量部、ヒプメロース6.3重量部、マクロゴール6000 1.5重量部、酸化チタン1.2重量部を混合してコーティング液を得た。
(6)(4)で得られた素錠に(5)で得られたコーティング液をフィルムコーティング装置(ハイコーター・ミニ型:フロイント産業(株))を用いて噴霧し、これを乾燥してコーティング錠を得た。
上記方法により得られたコーティング錠の組成を下表1に示す。
なお、有効成分の水分値補正を賦形剤で実施した。
(1)L-グルタミン酸L-アルギニン(水分値3.64%)2490.7g、部分アルファー化デンプン101.3g、クロスポビドン168.0gを流動層造粒乾燥機(フローコーター5型:フロイント産業(株))中で混合した後、ヒドロキシプロピルセルロース96.0gを含有する水溶液(1200g)を加えて造粒し、これを乾燥して造粒顆粒を得た。
(2)得られた造粒顆粒を整粒し、整粒顆粒を得た。

(4)得られた打錠用顆粒を打錠し、素錠を得た。
(5)水90重量部、ヒプメロース6.3重量部、マクロゴール6000 1.5重量部、酸化チタン1.2重量部を混合してコーティング液を得た。
(6)(4)で得られた素錠に(5)で得られたコーティング液をフィルムコーティング装置(ハイコーター・ミニ型:フロイント産業(株))を用いて噴霧し、これを乾燥してコーティング錠を得た。
上記方法により得られたコーティング錠の組成を下表1に示す。
なお、有効成分の水分値補正を賦形剤で実施した。
(実施例2)流動層造粒法
L-グルタミン酸L-アルギニン(水分値3.64%)2490.7g、部分アルファー化デンプン125.3g、クロスポビドン168.0gを流動層造粒乾燥機(フローコーター5型:フロイント産業(株))中で混合した後、ヒドロキシプロピルセルロース72.0gを含有する水溶液(900g)を加えて造粒し、これを乾燥して造粒顆粒を得た。その他は、実施例1と同様の方法で素錠を調製した。
L-グルタミン酸L-アルギニン(水分値3.64%)2490.7g、部分アルファー化デンプン125.3g、クロスポビドン168.0gを流動層造粒乾燥機(フローコーター5型:フロイント産業(株))中で混合した後、ヒドロキシプロピルセルロース72.0gを含有する水溶液(900g)を加えて造粒し、これを乾燥して造粒顆粒を得た。その他は、実施例1と同様の方法で素錠を調製した。
(実施例3)流動層造粒法
L-グルタミン酸L-アルギニン(水分値3.64%)2490.7g、部分アルファー化デンプン77.3g、クロスポビドン168.0gを流動層造粒乾燥機(フローコーター5型:フロイント産業(株))中で混合した後、ヒドロキシプロピルセルロース120.0gを含有する水溶液(1500g)を加えて造粒し、これを乾燥して造粒顆粒を得た。その他は、実施例1と同様の方法で素錠を調製した。
L-グルタミン酸L-アルギニン(水分値3.64%)2490.7g、部分アルファー化デンプン77.3g、クロスポビドン168.0gを流動層造粒乾燥機(フローコーター5型:フロイント産業(株))中で混合した後、ヒドロキシプロピルセルロース120.0gを含有する水溶液(1500g)を加えて造粒し、これを乾燥して造粒顆粒を得た。その他は、実施例1と同様の方法で素錠を調製した。
(比較例1)攪拌造粒法
(1)L-グルタミン酸L-アルギニン(水分値4%)2348g、部分アルファー化デンプン201g、ヒドロキシプロピルセルロース135g、クロスポビドン159gを撹拌造粒乾燥機(ハイスピードミキサー10型:深江パウテック(株))中で混合した後、精製水を加えて造粒をし、湿顆粒を得た。
(2)得られた湿顆粒を流動層造粒乾燥機(フローコーター5型:フロイント産業(株))中で乾燥して、造粒顆粒を得た。
(3)得られた造粒顆粒を整粒し、整粒顆粒を得た。
(4)得られた整粒顆粒200gに対して、結晶セルロース21g、ステアリン酸マグネシウム2.6gを加え、混合し、打錠用顆粒を得た。
(5)得られた打錠用顆粒を打錠し、素錠を得た。
(1)L-グルタミン酸L-アルギニン(水分値4%)2348g、部分アルファー化デンプン201g、ヒドロキシプロピルセルロース135g、クロスポビドン159gを撹拌造粒乾燥機(ハイスピードミキサー10型:深江パウテック(株))中で混合した後、精製水を加えて造粒をし、湿顆粒を得た。
(2)得られた湿顆粒を流動層造粒乾燥機(フローコーター5型:フロイント産業(株))中で乾燥して、造粒顆粒を得た。

(4)得られた整粒顆粒200gに対して、結晶セルロース21g、ステアリン酸マグネシウム2.6gを加え、混合し、打錠用顆粒を得た。
(5)得られた打錠用顆粒を打錠し、素錠を得た。
(比較例2)流動層造粒法
(1)L-グルタミン酸L-アルギニン(水分値3.79%)2494.5g、クロスポビドン168.0gを流動層造粒乾燥機(フローコーター5型:フロイント産業(株))中で混合した後、ヒドロキシプロピルセルロース120.0gを含有する水溶液(1500g)を加えて造粒し、これを乾燥して造粒顆粒を得た。
(2)得られた造粒顆粒を整粒し、整粒顆粒を得た。
(3)得られた整粒顆粒347.8gに対して、結晶セルロース9.2g、ステアリン酸マグネシウム3.0gを加え、混合し、打錠用顆粒を得た。
(4)得られた打錠用顆粒を打錠し、素錠を得た。
上記方法により得られた素錠の組成を下表2に示す。
(1)L-グルタミン酸L-アルギニン(水分値3.79%)2494.5g、クロスポビドン168.0gを流動層造粒乾燥機(フローコーター5型:フロイント産業(株))中で混合した後、ヒドロキシプロピルセルロース120.0gを含有する水溶液(1500g)を加えて造粒し、これを乾燥して造粒顆粒を得た。
(2)得られた造粒顆粒を整粒し、整粒顆粒を得た。
(3)得られた整粒顆粒347.8gに対して、結晶セルロース9.2g、ステアリン酸マグネシウム3.0gを加え、混合し、打錠用顆粒を得た。
(4)得られた打錠用顆粒を打錠し、素錠を得た。
上記方法により得られた素錠の組成を下表2に示す。
(試験例)
錠剤の溶出性評価を日本薬局方第15改正一般試験法・溶出試験法・装置2パドル法に準じて行った。
実施例1~3及び比較例1~2の素錠について、パドル回転数毎分50回転、37℃の水900mLを溶出試験液として用い、試験開始後5分の溶出試験液中の有効成分量をアミノ酸アナライザー法又は高速液体クロマトグラフィー法にて定量し、溶出率を算出した。その結果を表2に示す。
また、実施例1~3及び比較例1~2の素錠について、錠剤硬度測定装置(TABLET TESTER 8M:フロイント産業(株))により、錠剤硬度を測定した。その結果を表2に示す。
錠剤の溶出性評価を日本薬局方第15改正一般試験法・溶出試験法・装置2パドル法に準じて行った。
実施例1~3及び比較例1~2の素錠について、パドル回転数毎分50回転、37℃の水900mLを溶出試験液として用い、試験開始後5分の溶出試験液中の有効成分量をアミノ酸アナライザー法又は高速液体クロマトグラフィー法にて定量し、溶出率を算出した。その結果を表2に示す。
また、実施例1~3及び比較例1~2の素錠について、錠剤硬度測定装置(TABLET TESTER 8M:フロイント産業(株))により、錠剤硬度を測定した。その結果を表2に示す。
本発明の錠剤は、錠剤総重量に対する有効成分の含有率が高く、有効成分の溶出性にも優れ、且つ簡便に製造することができる。また、本発明の錠剤は従来品よりも小型化することが可能であり、対象への服用性にも優れている。本発明における有効成分は、機能性消化管障害の予防又は改善作用を有している。従って、本発明の錠剤は医薬分野及び食品分野等において有用である。
本出願は、日本で出願された特願2012-098111を基礎としており、その内容は本出願にすべて包含されるものである。
Claims (15)
- 有効成分として、グルタミン酸若しくはその塩及びアルギニン若しくはその塩、又はグルタミン酸の塩基性アミノ酸塩を含有し、該有効成分の含有率が錠剤総重量に対して50~90重量%であり、結晶セルロースを実質的に含有しない、錠剤。
- グルタミン酸の塩基性アミノ酸塩がグルタミン酸アルギニンである、請求項1に記載の錠剤。
- 有効成分の含有率が錠剤総重量に対して80~90重量%である、請求項1又は2に記載の錠剤。
- 結合剤を更に含有し、該結合剤の含有率が錠剤総重量に対して5重量%以下である、請求項1~3のいずれか1項に記載の錠剤。
- 結合剤がヒドロキシプロピルセルロースである、請求項4に記載の錠剤。
- クロスポビドン、カルボキシメチルスターチナトリウム及びクロスカルメロースナトリウムからなる群より選択される少なくとも1種の崩壊剤を更に含有する、請求項1~5のいずれか1項に記載の錠剤。
- 更に崩壊剤を含有し、崩壊剤が水と接触する条件下で製造されてなる、請求項1~5のいずれか1項に記載の錠剤。
- 崩壊剤の含有率が錠剤総重量に対して10重量%以下である、請求項6又は7に記載の錠剤。
- 有効成分と崩壊剤とが造粒された造粒物を含有する、請求項6~8のいずれか1項に記載の錠剤。
- 錠剤総重量が400mg以下である、請求項1~9のいずれか1項に記載の錠剤。
- 流動層造粒法により得られた造粒物を打錠することによって製造される、請求項1~10のいずれか1項に記載の錠剤。
- コーティング処理が施された請求項1~11のいずれか1項に記載の錠剤。
- 錠剤が素錠であり、第15改正日本薬局方溶出試験法でパドル法を用い、パドル回転数を毎分50回転とし、37℃の水900mLを溶出試験液とする溶出試験において、溶出時間が5分における有効成分の溶出率が60%以上である、請求項1~11のいずれか1項に記載の錠剤。
- 機能性消化管障害の予防又は改善用である、請求項1~13のいずれか1項に記載の錠剤。
- グルタミン酸若しくはその塩及びアルギニン若しくはその塩が、L-グルタミン酸若しくはその塩及びL-アルギニン若しくはその塩であり、グルタミン酸の塩基性アミノ酸塩がL-グルタミン酸L-アルギニンである、請求項1~14のいずれか1項に記載の錠剤。
