Abstract
Description

正常個体のアミロスフェロイド量よりも低いほど重症度が重い、及び/又は、前記被検体の以前のアミロスフェロイド量よりも高い/低い場合にそれぞれ重症度が重く/低くなったとする基準と、前記測定結果を比較することを含む、方法;

本明細書において、アミロスフェロイド(以下、「ASPD」ともいう。)とは、機能的に成熟した神経細胞に選択的に細胞死を誘発できるAβ集合体をいう。ASPDは、「合成ASPD」及び「ネイティブASPD」を含む。合成ASPDとは、合成Aβを用いてin vitroで調製及び単離される直径約10~15nmの球状体であるASPDをいう(非特許文献3)。また、ネイティブASPDとは、ヒト生体内で形成されたASPD、とりわけ、アルツハイマー病及び/又はレビー小体型認知症の患者の脳から単離されうるASPDをいう(非特許文献1)。合成ASPD及びネイティブASPDは、ともに、成熟ヒト神経細胞に細胞死を誘発する。ASPDに特異的な立体構造を認識できる抗ASPD抗体も作製されている(例えば、特許文献1及び2に開示されるhaASD1、haASD2、mASD3など)。これらの抗ASPDモノクローナル抗体の特性解析やASPDのNMR解析の結果から、ASPDは、これまで報告された他のAβ集合体とは異なるユニークな立体構造をもつことがわかっている(例えば、非特許文献1のSupplemental Table S1、厚生労働科学研究費補助金(医療機器開発研究促進事業)平成22年度報告書)。具体的には、まず抗ASPD抗体のエピトープマップの結果から、ASPDを形成するにあたって、その構成要素であるAβは折り畳まれてN末端と中間部位が共にASPDの表面に出ていることがわかっている。このことからASPDに特異的な表面3次元構造を作っていることがわかる。ASPDの二次元NMRスペクトルもこの結果を支持しており、本来Aβモノマーで検出されるべきシグナルのうち、ASPD表面に出ている部位ではシグナルが観測出来る一方で、ASPDの内側に折り畳まれた部位に由来するシグナルは内部に折り畳まれることで磁気的に非等価かつ運動性が抑制されるためシグナルが減弱し検出されなくなる。NMR解析から得られた結果も、AβのN末端と中間部位がASPD表面に露出していることが示されており、抗ASPD抗体が認識する部位と一致していた。ASPD表面に出ている中間部位は、通常の凝集体では内部に折り畳まれている部位であり、ASPDの特異な立体構造を持つ凝集体であることを示している。したがって、本明細書においてASPDは、特許文献1及び2に開示されるASPDに特異的な抗ASPD抗体に反応し、かつ、機能的に成熟した神経細胞に選択的に細胞死を誘発できるAβ集合体ともいうことができる。ASPDは、限定されない一又は複数の実施形態において、Aβモノマーの約20~150量体である。ASPDは、限定されない一又は複数の実施形態において、細胞毒性が高い形態として、分子量が約84kDa~約172kDaであり、平均分子量が約128kDaである。ASPDは、限定されない一又は複数の実施形態において、溶液中でのAFM(原子間力顕微鏡)で測定した直径が、約4.6nm~約9.8nmであり、平均約7.2nmである。
本明細書において「アミロイドβモノマー」とは、アミロイド前駆体タンパク質(APP)がβ-及びγ-セレクターゼに切断されることよって切り出されるペプチドをいい、「βアミロイド」、「Aβ」又は「Aβモノマー」とも表記される。また、本明細書において、Aβはそのアミノ酸配列の長さからAβ1-39、Aβ1-40、Aβ1-41、Aβ1-42、及びAβ1-43と呼ばれるものを含みうる。本明細書において、Aβは、ヒト型(ヒトに存在する配列)であってもよく、非ヒト型(ヒト以外の動物に存在する配列)であってもよい。また、本明細書において、Aβは、生体内の(ネイティブな)Aβ、及び、合成されたAβを含みうる。合成Aβは、特に限定されないが、公知のペプチド合成法(例えば、Fmoc法やBoc法)で合成でき、例えば、公知のペプチド合成機を利用して製造できる。ヒト型のAβ1-42(「Aβ42」ともいう。)は、アミノ酸配列(N末端から):DAEFRHDSGYEVHHQKLVFFAEDVGSNKGAIIGLMVGGVVIA(配列番号2)で表わされるアミノ酸配列からなるペプチドである。また、ヒト型のAβ1-41(Aβ41)、Aβ1-40(Aβ40)、及びAβ1-39(Aβ39)は、配列番号2のアミノ酸配列のC末端からそれぞれA、IA、及びVIAを欠いたアミノ酸配列からなるペプチドである。さらに、ヒト型のAβ1-43(Aβ43)は、配列番号2のアミノ酸配列のC末端にトレオニン残基(T/Thr)が1つ付加されたアミノ酸配列からなるペプチドである。
本明細書において、Na+/K+-ATPaseα3(以下、「NAKα3」ともいう。)とは、ATP1A3遺伝子の遺伝子産物の膜タンパク質である「ナトリウム/カリウム輸送体ATPアーゼサブユニットα3」のことをいう。Na+/K+ATPアーゼは、細胞質膜を介したナトリウムとカリウムの電気化学勾配を維持する役割を果たすATPアーゼ輸送体である。α3は特に神経細胞においては、膜電位の維持に関与すると考えられている。本明細書において、ATP1A3遺伝子及びNAKα3は、ヒト型(ヒトに存在する配列)であってもよく、非ヒト型(ヒト以外の動物に存在する配列)であってもよい。なお、本明細書において「ヒト以外の動物」とは、特に限定されないが、ヒトAβ及び/又はヒトNAKα3のオーソログを発現する細胞を有する動物であって、一又は複数の実施形態において、例えば、哺乳類、例えば、霊長類やげっ歯類、例えば、サルやラットである。
NAKα3は、ASPDが結合して成熟神経細胞に細胞死を誘導する標的分子である。本明細書において「ASPDの標的分子」とは、「ASPDが結合して成熟神経細胞に細胞死を誘導する標的分子」を含む。ASPDの標的分子であるNAKα3は、以下のようにして同定された。すなわち、成熟神経細胞、幼若神経細胞、及びヒト胎児腎臓由来細胞であるHEK293細胞由来の抽出液をそれぞれ電気泳動で展開し、生体内で形成された(すなわち、ネイティブな)ASPDをリガンドとし抗ASPD抗体を用いて検出することでファーウエスタンを行った。その結果、成熟神経細胞の場合のみに現れた約105kDaのバンドからNAKα3が同定された。なお、このバンドは合成ASPDをリガンドとした場合でも検出されるが、Aβモノマーをリガンドとし抗Aβ抗体(6E10)を検出に使用したファーウエスタンでは現れない。また、成熟ラットの海馬ニューロン(21DIV)の細胞抽出物をASPD共存下において、抗ASPD抗体で免疫沈降すると、NAKα3が共沈する。
ASPDが標的であるNAKα3とタンパク質間相互作用して成熟神経細胞の細胞死を誘発するメカニズムは、以下のように考えられる。ASPDがNAKα3と相互作用すると、成熟神経細胞のNa+/K+ATPアーゼの機能が阻害されるため、細胞膜電位が上昇して神経細胞が過興奮状態となり電位依存性カルシウムチャネルを介して細胞外Ca2+が細胞内に多量に流入する。Ca2+の異常流入により細胞内Ca2+ホメオスタシスが失われ、その結果、細胞内Ca2+濃度が過剰に上昇して細胞死に至ると考えられる(図1参照)。より詳細には、細胞内Ca2+濃度の上昇は、カルパインの活性化、p25/サイクリン依存性キナーゼ5(CDK5)の活性化、グリコーゲン合成酵素キナーゼ3β(GKS3β)の活性化、及び、タウタンパク質の過剰リン酸化をこの順で引き起こし、成熟神経細胞に細胞死をもたらすと考えられる。但し、本開示は、これらのメカニズムに限定して解釈されなくてもよい。
ASPDが誘発する成熟神経細胞死がASPDとNAKα3とのタンパク質間相互作用に起因することは、これまで報告されていない、従来技術の予測を超える新たな知見である。ASPDとNAKα3とのタンパク質間相互作用を阻害できれば、ASPDが誘発する成熟神経細胞死を抑制でき、ひいては、アルツハイマー病及び/又はレビー小体型認知症の治療、診断、改善、及び/又は予防が可能となる。したがって、一又は複数の実施形態において、本開示は、アミロスフェロイドが結合して成熟神経細胞死を誘発する標的分子としてのNAKα3の使用に関する。ASPDの標的分子としてのNAKα3の使用は、一又は複数の実施形態において、ASPDとNAKα3とのタンパク質間相互作用を阻害することでASPDの神経細胞死を抑制すること、ASPDとNAKα3とのタンパク質間相互作用を阻害することでアルツハイマー病及び/又はレビー小体型認知症の治療、診断、改善、及び/又は予防をすること、ASPDとNAKα3との相互作用に基づく医薬組成物の開発をすることなどを含みうる。ASPDの標的分子としてのNAKα3の使用は、一又は複数の実施形態において、本明細書で開示される発明及びその実施形態を含みうる。なお、本明細書において「タンパク質間相互作用」とは、結合を含み、「ASPDとNAKα3とのタンパク質間相互作用」とは、結果として細胞死をもたらし得る作用をNAKα3に及ぼすことを含む。また、本明細書において「アルツハイマー病」は、アルツハイマー型認知症を含む。また、本明細書において「アルツハイマー病及び/又はレビー小体型認知症の治療、診断、改善、及び/又は予防」の対象は、特に限定されないが、ヒト及び/又はヒト以外の動物とすることができる。
ASPDは、前述のとおり、ユニークな立体構造を有するAβ集合体である。したがって、ASPDとNAKα3とのタンパク質間相互作用に関与するASPD又はNAKα3の部位の立体構造を模した物質(例えば、ポリペプチド、ペプチドミミック、低分子化合物、これらの塩、これらの溶媒和物など)であれば、ASPDとNAKα3とのタンパク質間相互作用を阻害することができる。相互作用する一方の立体構造を模した分子が存在することで競合阻害が生じると考えられるが、本開示はこの考え方に限定されなくてもよい。なお、本明細書において「物質」とは、化合物、その塩、その溶媒和物、及び/又はこれらを含む組成物を意味することができ、より具体的には、ポリペプチド、ペプチドミミック、低分子化合物、これらの塩、これらの溶媒和物、及び/又はこれらを含む組成物を意味することができる。
さらにまた、本開示は、一又は複数の実施形態において、NAKα3の細胞外ドメイン4、「RLNW」若しくは「LNW」を含む部分、及び/又は、「YGQQWT」若しくは「Y-NLWR」を含む部分の立体構造を特異的に認識する抗NAKα3抗体に関する。前記抗NAKα抗体は、一又は複数の実施形態において、ポリクローナル抗体、モノクローナル抗体、ヒト化抗体、及び/又は、完全ヒト抗体である。これらの抗体のなかでも、一又は複数の実施形態において、NAKα3とASPDとの相互作用を阻害しつつ、NAKα3のイオンチャネルの機能を阻害しないものが好ましい。したがって、本開示は、一又は複数の実施形態において、アミロスフェロイドが存在しうる条件下の成熟神経細胞のNAKα3に前記抗NAKα3抗体を結合させることを含む、ASPDが誘発する成熟神経細胞死を抑制する方法、及び、アルツハイマー病及び/又はレビー小体型認知症の予防、改善、及び/又は治療の方法に関する。また、本開示は、その他の態様において、ASPDが誘発する成熟神経細胞死の抑制、或いは、アルツハイマー病及び/又はレビー小体型認知症の予防、改善、及び/又は治療の方法に使用する、前記抗NAKα3抗体を含む組成物に関する。
本開示は、一又は複数の実施形態において、アルツハイマー病及び/又はレビー小体型認知症の予防、改善、及び/又は治療のための医薬組成物の有効成分の候補化合物のスクリーニング方法であって、テスト化合物を用いてASPDとNAKα3とのタンパク質間相互作用の阻害能を測定すること、及び、前記測定結果に基づき候補化合物を選択することを含むスクリーニング方法に関する。
ASPDとNAKα3との相互作用に起因して細胞外から細胞内にCa2+を流入させ神経を死に至らしめるのは、N型の電位依存性カルシウムチャネル(VGCC)である。さらに、細胞内小器官から細胞質へのCa2+流入も細胞死に関与すると考えられる。したがって、本開示は、その他の態様において、アミロスフェロイドと相互作用する成熟神経細胞において、細胞膜のN型電位依存性カルシウムチャネル(VGCC)、ミトコンドリアのNa+/Ca2+交換輸送体(mNCX)、ミトコンドリアの膜透過性遷移孔(mPTP)、及び、小胞体のリアノジン受容体(RyR)からなる群から選択される少なくとも1つのカルシウムチャネルを阻害することを含む、ASPDが誘発する成熟神経細胞死を抑制する方法、及び、アルツハイマー病及び/又はレビー小体型認知症の予防、改善、及び/又は治療の方法に関する。各カルシウムチャネルの阻害剤は、公知の阻害剤を用いてもよい。また、本開示は、その他の態様において、ASPDが誘発する成熟神経細胞死の抑制、或いは、アルツハイマー病及び/又はレビー小体型認知症の予防、改善、及び/又は治療の方法に使用する、細胞膜のN型電位依存性カルシウムチャネル(VGCC)、ミトコンドリアのNa+/Ca2+交換輸送体(mNCX)、ミトコンドリアの膜透過性遷移孔(mPTP)、及び、小胞体のリアノジン受容体(RyR)からなる群から選択される少なくとも1つのカルシウムチャネルを阻害できる化合物を含む組成物又は医薬組成物に関する。

本開示は、一又は複数の実施形態において、合成、単離、又は精製されたポリペプチドであって、前記ASPD相互作用モチーフを含み、アミロスフェロイドに対する結合能を有するポリペプチド(以下、「第1のポリペプチド」ともいう)に関する。第1のポリペプチドにおいて、前記ASPD相互作用モチーフの実施形態は上述のとおりである。第1のポリペプチドは、一又は複数の実施形態において、好ましくは、ASPDが誘発する成熟神経細胞死を抑制でき、より好ましくは、さらに/或いは、ASPDとNAKα3との相互作用を阻害できる。
本開示は、さらにその他の態様として、本開示のポリペプチドの構造をミミックしたペプチドミミックに関する。本開示のペプチドミミックは、ASPDとNAKα3との相互作用に対する阻害能を有する。本開示のペプチドミミックの好ましい実施形態として、前記式(I)で表わされるモチーフのペプチドミミックと脳移行性ペプチドとが直接又はリンカーを介して結合した形態が挙げられる。脳移行性ペプチドを結合させることで、本開示のペプチドミミックが血液脳関門を越えて脳に移行される効率を高めることができる。脳移行性ペプチドをとしては、従来公知のものを利用できる。
本開示は、さらにその他の態様として、本開示のポリペプチドをコードするポリヌクレオチドに関し、また、本開示のポリペプチドをコードするポリヌクレオチドを含み、前記ポリペプチドを発現するためのベクターに関する。前記ベクターとしては、前記ポリヌクレオチドを遺伝子導入できるものであれば特に限定されないが、安全性の観点からAVV(アデノ随伴ウイルス)ベクターが好ましい。本開示のベクターの好ましい実施形態として脳移行性ペプチドと結合した形態が挙げられる。脳移行性ペプチドを結合させることで、本開示のベクターが血液脳関門を越えて脳に移行される効率を高めることができる。脳移行性ペプチドをとしては、従来公知のものを利用できる。
本開示が医薬組成物である場合、その剤形は、投与方法に応じて適宜選択でき、例えば、注射剤、液体製剤、カプセル剤、咀嚼剤、錠剤、懸濁剤、クリーム剤、軟膏等が挙げられる。また、投与方法も特に制限されず、経口投与、非経口投与が挙げられる。本開示の医薬組成物は、投与形態や剤形に応じた従来公知の添加物(例えば、賦形剤又は希釈剤)を含有してもよい。
本明細書において、アルツハイマー病及び/又はレビー小体型認知症の予防とは、アルツハイマー病及び/又はレビー小体型認知症の発症を抑制すること、或いは可逆的な軽度な認知障害よりも病態の進行を進めないことを含む。また、本明細書において、アルツハイマー病及び/又はレビー小体型認知症の改善とは、アルツハイマー病及び/又はレビー小体型認知症の可逆的な軽度な認知障害の病態の進行が止まること、或いは、病態が軽度になることを含む。さらに、本明細書において、アルツハイマー病及び/又はレビー小体型認知症の治療とは、病態の進行を遅らせる、或いはほとんど止まらせることを含む。
本開示は、その他の態様において、ASPDイメージングプローブ又はその前駆体であって、本開示のポリペプチド又はペプチドミミックをASPDとの結合部分として備える、イメージング用プローブ又はその前駆体に関する。本開示のポリペプチド又はペプチドミミックに適切な標識放射性化合物で標識することで、ASPDのイメージング用プローブとすることができる。
本開示のASPDイメージング方法で得られるシグナルデータ又はイメージデータに基づき、ASPD量を算出してもよい。アルツハイマー病及び/又はレビー小体型認知症の患者の大脳皮質におけるネイティブASPD量は、アルツハイマー病及び/又はレビー小体型認知症の重症度に相関して増加することから、本開示のイメージング方法を行うことにより、アルツハイマー病及び/又はレビー小体型認知症の病態の重症度を判定又は診断できる。したがって、本開示は、その他の態様において、ASPDイメージング方法で得られるシグナルデータ又はイメージデータに基づき、アルツハイマー病及び/又はレビー小体型認知症の病態の重症度を判定することを含む診断方法に関する。
(1-1)ASPD結合実験
合成ASPDをフィルターで精製した後、ヒト胎児由来HEK293細胞、成熟ラット海馬由来初代培養神経細胞に投与し、1時間後に固定、免疫組織染色を行った。ASPDはHEK293細胞には結合せず、成熟神経細胞に結合した。その結果を図3の左に示す。同図に示すとおり、ASPDと成熟神経細胞との結合は、抗ASPD抗体により阻害された。
(1-2)ASPD毒性実験
一定濃度のASPD(5μM)をそれぞれの細胞に投与し一晩置いた後、アポトーシス活性をロッシュ社製Cell Death ELISAを用いて定量した。ASPDは非神経細胞であるHEK293細胞、ラット海馬由来未成熟神経細胞には毒性を発揮せず、成熟神経細胞にのみ毒性を発揮した。その結果を図3の右に示す。同図に示す通り、ASPDの成熟神経細胞に対する毒性は抗ASPD抗体により阻止された。
(1-3)上記及び図3から、ASPDがその特異な立体構造により、成熟神経細胞表面にのみ存在するターゲット分子と結合し、神経毒性を発揮していることが示唆された。
(2-1)ファーウエスタン法による分析
ラット海馬由来初代培養神経細胞(図4に表示の培養日数)とHEK293細胞のそれぞれからRIPA(RadioImmunoPrecipitation Assay)により抽出液を調製、タンパク質量を定量後、一定量をSDS-PAGEにて電気泳動分離を行い、ニトロセルロース膜に転写後、ネイティブASPD又はAβモノマーと一定時間反応させた。ネイティブASPDの結合を抗ASPD抗体haASD1にて、Aβモノマーの結合は市販の抗Aβモノマー抗体である6E10にて、検出を行った(ファーウエスタンブロッティング)。その結果を図4に示す。
図4に示す通り、ASPDは、成熟神経細胞由来抽出液においてのみ105kDaのバンドを認識することがわかった。一方、Aβモノマーではこのバンドに結合せず、50kDa程度のバンドを認識していた。ネイティブASPDと105kDaのバンドの結合は、ASPDに特異的であり、抗ASPD抗体だけではバンドは検出されず、ネイティブASPDを予め過剰量の抗ASPD抗体で処理した場合にもバンドは検出されなかった。これはASPDの成熟神経細胞への結合と毒性が、抗ASPD抗体の事前処理で失われることと相関している。
図5の左に、図4のネイティブASPDに換えて合成ASPDを使用した場合のファーウエスタンブロッティングの結果を示す。ネイティブASPD(図4左)と同様に、成熟神経細胞でのみ105kDaのバンドに合成ASPDが結合していることが示されている。従って、合成ASPDはネイティブASPDと等価と考えられ、今後これを用いて解析を実施した。
図5の右に、図5左で用いた細胞由来抽出液の銀染色パターンを示す。四角で囲ったところに、未成熟神経細胞由来抽出液やHEK293細胞由来抽出液では検出出来ないバンドが認められた。これを切り出して質量分析計(MALDI-TOF-MS、ブルカーダルトニクス社製、商品名:Ultraflex)によりMS/MS解析を行ったところ、Na+/K+-ATPaseαサブユニットが検出された。
図4左及び図5左のファーウエスタンブロッティングに用いた抽出液を使って、Na+/K+-ATPaseα1サブユニットとα3サブユニットに選択的な抗体によるウエスタンブロッティングを行った。その結果を図6に示す。図6に示すとおり、成熟神経細胞に選択的に発現しているのはα3サブユニットであり、ASPDが結合しているターゲットはNa+/K+-ATPaseα3サブユニット(NAKα3)であると考えられた。
成熟したラット海馬由来初代培養神経細胞の抽出液、その抽出液に合成ASPDを添加し、抗ASPD抗体(haASD1)あるいは対照として正常マウスIgGを用いた免疫沈降物をそれぞれ、電気泳動を行い、NAKα3特異的抗体でウエスタンブロッティングを行った。その結果を図7に示す。同図に示す通り、抗ASPD抗体によりNAKα3がASPDと共沈してくることが明らかとなった。
上記(2-4)の実験から、ASPDとNAKα3が直接相互作用することが示された。その相互作用が生きている細胞上で起こることを示すために、ビオチン化したAβを一定比率配合したビオチン化ASPDを調製し、それを生きた成熟神経細胞(ラット海馬由来初代培養神経細胞)に投与し1時間以内に、ASPDの構造を破壊しない条件下で細胞膜を回収し、ビオチンに強い親和性を持つアビジンで分画を行った。銀線色の結果を図8に示す。同図が示す通り、ビオチン化ASPD投与条件でのみ105kDaあたりに認められるバンドがあり、タンデム質量分析(MS/MS)解析からNAKαであることが明らかとなった。
以上の異なる2つの実験(2-4)及び(2-5)から、ASPDとNAKα3が十分な結合の強さを持って直接相互作用することが示された。
合成ASPDを投与後、成熟神経細胞(ラット海馬由来初代培養神経細胞)上のASPDの結合部位を抗ASPD抗体によって蛍光染色し、NAKa3の存在部位を特異的抗体によって蛍光染色し、そして蛍光顕微鏡観察した結果を図9に示す。同図が示す通り、ASPDが結合する部位はNAKα3の存在部位と合致することが示された。
合成ASPDを成熟ラット海馬由来初代培養神経細胞に投与し、1時間後に細胞を固定し、抗ASPD抗体を用いて細胞に結合したASPD量を定量した結果のスキャチャードプロットの一例を図10に示す。また、同図のスキャチャードプロットから合成ASPDの解離定数がKd=1.5±7.8×10-7Mであることが示された。
合成ASPDを成熟ラット海馬由来初代培養神経細胞に投与し約24時間後に細胞膜を回収し、ATPase活性を測定し、NAKの選択的阻害剤であるouabainを用いてNAK活性を求めた。ラットにおいては、ouabainのKiはα3=3.1±0.3x10-8M、α1=4.3±1.9x10-5Mである。そこで、低濃度のouabain(10nM)存在下ではNAKα3のみが阻害されるため、NAKα3活性を求めることが可能である(図11の左欄のグラフ)。そして、10nM ouabain存在下では阻害されず100μM ouabainで阻害される活性としてNAKα1活性(図11の中欄のグラフ)を求めることが可能である。図11に示したとおり、ASPD処理によりNAKα3活性が20%以下になっていることがわかった。一方、NAKα1活性はNAKα3活性ほど低下していないことが明らかとなった。従って、ASPDは特にNAKα3活性を強く阻害していることが明らかとなった。
Na+/K+-ATPase(NAK)の機能と構造と分布を図12に示す。図12上図は、NAKの機能である、ATP加水分解と共役したナトリウム-カリウムポンプ機能を説明図の一例である。図12中図は、NAKのαサブユニット及びβサブユニットの構造の一例を示す図である。同図に示すように、βサブユニットは、αサブユニットの細胞外ドメイン4に結合することが報告されている(Lemas et al. vol 269, 8255-8259, 1994)。図12下図の表に示す通りα1サブユニットは全ての細胞に存在する普遍的タイプである。一方、α3サブユニットは成熟神経細胞に選択的に発現している。幼若神経ではα2サブユニットが発現しているが、成熟するとα3サブユニットに切り替わることが知られている。これらのNAKα3の知見は、図3~11の結果とよく合致している。
(3-1)細胞内カルシウム濃度を測定
ASPDが結合することによりNAKα3の活性が阻害されるとしたならば、(1)流入したNaがくみ出されないために膜電位が上昇し、その結果、膜電位依存的なCaチャンネルが開口し、あるいは、(2)Naの上昇に応答して細胞膜上のNa/Ca exchanger (NCX)が反応してCaを取り込み、最終的に細胞内に異常なカルシウムの流入が起こることが予想された。そこで、成熟したラット海馬由来初代培養神経細胞にASPDを投与したときの細胞内カルシウム濃度をFura PE3蛍光カルシウム指示薬にて観察した。その結果を図13に示す。同図に示す通り、ASPD投与直後から細胞内カルシウム濃度の上昇が検出され、それが最終的には細胞内カルシウム濃度が振り切れて細胞が死に至ることがわかった。これはASPD濃度に応じてより早く起きることも示された。
成熟ラット海馬由来初代培養神経細胞に、下記表1に示される各種阻害剤を同表に示される濃度で予め投与し、30分後に合成ASPD 4μMを投与し、およそ24時間後にアポトーシス活性を測定した。その結果を表1及び図14に示す。同表及び同図が示す通り、ASPD投与後、NAKα3活性が抑制されることにより起きる神経細胞死シグナルカスケードには、膜電位依存的カルシウムチャンネルのうち、プレシナプスに多く存在するN型チャンネルが関わっていることがわかった。さらに、ミトコンドリアに存在するNCX、mPTPもそれぞれ貢献していることがわかった。従って、図15に示したように、細胞内へのカルシウム流入はまずシナプス上の膜電位依存性N型カルシウムチャンネルが活性化することで起こり、それに続いてミトコンドリアのカルシウム代謝異常が起きて、細胞死に至ることが示された。
特異的抗体を用いてNAKの分布をヒト脳で検証した。正常脳での解析からは、まずNAKα1は、全体的に分布しており、これはNAKα1が普遍的に存在することと合致していた(図16左上)。一方、NAKα3はNAKα1とは全く異なった分布を示し、大脳皮質においては神経突起や軸索に沿ってドット状に、あるいは錐体細胞(神経細胞)を取り巻くように分布していることがわかった(図16右上)。小脳においては、NAKα3は神経突起や軸索に沿ってドット状に存在するほかにpurkinje cellを取り巻くように分布していた(図17左上)。これはBasket cellの分布と非常に良く重なる。Basket cellは大脳皮質にも存在しており、NAKα3がmRNAの発現解析において抑制性神経にその発現がよく認められることと合致している。
患者脳では、アルツハイマー病で障害が大きく認められる大脳皮質ではNAKα3の染色は失われていた(図16右下)。一方、NAKα1の染色は本来の存在部位では失われているが、それ以外の部分ではむしろ量が増大しているという結果が得られた(図16左下)。これは過去のmRNAの解析と合致している結果である(Siegel 1998)。同時にASPD量を調べてみたところ、患者小脳では正常と同じレベルの極小量のASPDしか検出されない一方で、障害が大きい大脳皮質では多量のASPDが存在しており、これはNAKα3の減少と相関していた(表2)。
抗ASPD抗体がASPDに結合することで、ターゲット分子であるNAKα3との相互作用を阻止し、神経細胞死を抑制することから、同様の効果を持つペプチドの探索を行った。そのために、マイクロタイタープレートに固定したASPDに対して、ランダムな12アミノ酸のライブラリをウイルス表面に発現しているphage display library(市販)を利用して、スクリーニングを行い、収束した配列を解析した。3回のphage displayから、同じ配列を持つphageが複数回重複して得られていることから、何らかの生物学的濃縮が起こり、ASPDに結合活性があるペプチドが得られている可能性を示唆している。また、得られた配列すべてをバイオインフォマティクス的に解析すると、H(His)およびW(Trp)が有意に高頻度で出現していた。H/Wを多く含むペプチドモチーフの中に、ASPDに特異的に結合するものが含まれていることを示唆している。その中から実際にASPD毒性を中和するものと中和しないものを調べると、毒性を中和するものは共通のモチーフを持つことがわかった。この中和活性を持つペプチドをAnti-AD toxicity (AAT)ペプチドと名付けた。得られたAATペプチド01~05を表3に示す。また、中和活性を持たないペプチド(PD)の2例を表3に示す。
前記AATペプチド01~03のC末端側にビオチンを結合させチップ上に固相化し、ASPDをリガンドとして表面プラズモン共鳴(SPR)により解離定数を求めた。その結果を下記表5に示す。

- 成熟神経細胞死を誘発するアミロスフェロイドの結合標的分子としてのNa+/K+-ATPaseα3の使用。
- アミロスフェロイドとNa+/K+-ATPaseα3との相互作用に基づき医薬品開発を行うことを含む、請求項1記載の使用。
- アミロスフェロイドが誘発する成熟神経細胞死を抑制する方法であって、
アミロスフェロイドとNa+/K+-ATPaseα3とのタンパク質間相互作用を阻害することを含む、方法。 - アミロスフェロイドとNa+/K+-ATPaseα3とのタンパク質間相互作用の阻害が、両者又は一方の表面立体構造に基づく競合阻害を行い得る物質とアミロスフェロイドとの接触により行われる、請求項3記載の方法。
