変形性関節症(osteoarthritis:OA)は、関節軟骨の変性と軟骨下骨の増殖および再形成に特徴付けられる進行性の変性関節疾患である。症状は、硬直、動作制限、疼痛などであり、加齢とともに発生頻度が増加する。しかしながら、軟骨は修復能に乏しいため、変形性関節症の根治方法はなく、運動制限と鎮痛剤で疼痛をコントロールすることが治療の現状である。症状が進行し末期に至った場合には、変性した関節軟骨を切除して金属製の覆いをかぶせる人工関節置換術が行われている。
軟骨修復促進剤の開発をめざして、軟骨細胞の増殖を促進する化合物やタンパク質が探索されているが、実際に軟骨を組織として増大させることができるものは骨形成因子(BMP)のみである。しかしBMPは、軟骨を増大させた後に、軟骨細胞を肥大化させて骨に置換する作用を有しており、関節軟骨の治療に用いるには問題がある。近年、関節軟骨の変性に軟骨細胞の肥大化促進がかかわることが指摘され、軟骨細胞の肥大化を防ぐことが変形性関節症の治療における一つの主要なターゲットになっている。これまで明らかにされたターゲットとしては、Hedgehog(非特許文献1)、Hif-2alpha(非特許文献2、3)がある。また、一過性にbeta-cateninシグナルを活性化させると関節軟骨が肥厚することが報告されている(非特許文献4)。

軟骨形成異常症(chondrodysplasia)は骨系統疾患(skeletal dysplasia)に含まれ、軟骨原基および成長軟骨の先天性の障害により小人症や骨格の成長障害や変形がおきる種々疾患の総称である。軟骨形成異常症は、軟骨細胞分化にかかわる種々の遺伝子の変異によって引き起こされることが多い(非特許文献6)。その病態には軟骨細胞の分化の異常がかかわっている。すなわち、軟骨細胞分化が亢進している場合と、抑制されている場合がある。軟骨細胞の肥大化は、軟骨細胞の分化のひとつである。軟骨形成異常症に対する治療薬として、軟骨細胞分化を制御できる薬剤の開発が望まれている。また、軟骨形成異常症のいくつかの成長軟骨に対して、軟骨細胞を移植して成長を正常化させる治療が考えられる。

本発明は、軟骨細胞の分化または肥大化に関与する分子を見出し、軟骨疾患治療薬のスクリーニング方法を提供することを課題とする。また、軟骨疾患の治療および病態の解析に利用可能な改変軟骨細胞を提供することを課題とする。
本発明は、上記課題を解決するために、以下の各発明を包含する。
[1]軟骨量を増大させる作用および/または軟骨細胞分化を正常化させる作用を有する物質のスクリーニング方法であって、塩誘導性キナーゼ3を使用することを特徴とする方法。
[2]軟骨量が減少する病気および/または軟骨の成長が障害される病気の改善のために利用する、塩誘導性キナーゼ3の発現もしくは機能が抑制または増強されている改変軟骨細胞。
[1]軟骨量を増大させる作用および/または軟骨細胞分化を正常化させる作用を有する物質のスクリーニング方法であって、塩誘導性キナーゼ3を使用することを特徴とする方法。
[2]軟骨量が減少する病気および/または軟骨の成長が障害される病気の改善のために利用する、塩誘導性キナーゼ3の発現もしくは機能が抑制または増強されている改変軟骨細胞。
本発明のスクリーニング方法によれば、軟骨量が減少する病気や軟骨の成長が障害される病気の治療薬として有用な物質をスクリーニングすることができる。また、本発明の改変軟骨細胞は、軟骨量が減少する病気や軟骨の成長が障害される病気の改善に有用である。さらに、本発明の改変軟骨細胞は、軟骨細胞分化異常を再現した疾患モデル細胞として、軟骨疾患の病態解明や薬剤のスクリーニングに用いることができる。
本発明者らは、塩誘導性キナーゼ3(salt-inducible kinase 3、以下「SIK3」という)のノックアウトマウスを作製したところ、成長軟骨細胞の肥大化が著しく抑制され、関節軟骨量が増大し、関節軟骨組織が肥厚化していることを見出した。このような表現型はSIK2のノックアウトマウスには認められていない(Pigment Cell Melanoma Res. 2010:23 809-819)。そこで、この表現型がSIK3の欠失に基づくものであることを確認するために、SIK3遺伝子の変異アレルを持つヘテロ接合体マウスとSIK3遺伝子のトランスジェニックマウスを交配してレスキュー試験を行ったところ、SIK3Tg、SIK3-/-の遺伝子型を持つF1マウスは野生型マウスと同様の表現型を示した。これらの結果から、SIK3の発現または機能を抑制すれば軟骨細胞の肥大化を抑制し、軟骨量を増大できることが実証された。
また、本発明者らは、SIK3遺伝子のトランスジェニックマウスの膝関節の成長軟骨板が野生型マウスより早い時期に消失することを見出した。この知見から、SIK3の発現または機能を亢進させれば、軟骨細胞の肥大化を促進し、軟骨細胞の骨化を加速できるものと考えられた。
これらの知見から、SIK3を使用すれば、軟骨疾患の治療薬として有用な物質をスクリーニングすることができることが明らかとなった。また、SIK3の発現もしくは機能が抑制されている軟骨細胞、SIK3の発現もしくは機能が増強されている軟骨細胞は、軟骨疾患の改善または治療に有用であることが明らかとなった。
これらの知見から、SIK3を使用すれば、軟骨疾患の治療薬として有用な物質をスクリーニングすることができることが明らかとなった。また、SIK3の発現もしくは機能が抑制されている軟骨細胞、SIK3の発現もしくは機能が増強されている軟骨細胞は、軟骨疾患の改善または治療に有用であることが明らかとなった。
〔スクリーニング方法〕
本発明は、SIK3を使用して軟骨疾患の治療薬として有用な物質をスクリーニングする方法を提供する。より詳細には、SIK3を使用して軟骨量を増大させる作用および/または軟骨細胞分化を正常化させる作用を有する物質をスクリーニングする方法を提供する。軟骨細胞分化を正常化させる作用には、軟骨細胞分化のスピードが正常より抑制されている場合に、これを亢進して軟骨細胞分化を正常化させる作用、および、軟骨細胞分化のスピードが正常より亢進している場合に、これを抑制して軟骨細胞分化を正常化させる作用が含まれる。
軟骨量を増大させる作用を有する物質、および、軟骨細胞分化のスピードを抑制する作用を有する物質は、SIK3の発現または機能を抑制する被験物質を選択することによりスクリーニングすることができる。一方、軟骨細胞分化のスピードを亢進する作用を有する物質は、SIK3の発現または機能を亢進する被験物質を選択することによりスクリーニングすることができる。
本発明は、SIK3を使用して軟骨疾患の治療薬として有用な物質をスクリーニングする方法を提供する。より詳細には、SIK3を使用して軟骨量を増大させる作用および/または軟骨細胞分化を正常化させる作用を有する物質をスクリーニングする方法を提供する。軟骨細胞分化を正常化させる作用には、軟骨細胞分化のスピードが正常より抑制されている場合に、これを亢進して軟骨細胞分化を正常化させる作用、および、軟骨細胞分化のスピードが正常より亢進している場合に、これを抑制して軟骨細胞分化を正常化させる作用が含まれる。
軟骨量を増大させる作用を有する物質、および、軟骨細胞分化のスピードを抑制する作用を有する物質は、SIK3の発現または機能を抑制する被験物質を選択することによりスクリーニングすることができる。一方、軟骨細胞分化のスピードを亢進する作用を有する物質は、SIK3の発現または機能を亢進する被験物質を選択することによりスクリーニングすることができる。
ここで、SIKについて説明する。SIK1(塩誘導性キナーゼ1)は血圧調節因子の候補として高塩食負荷したラットの副腎皮質から単離したタンパク質リン酸化酵素である。SIK1に似た別のタンパク質リン酸化酵素が2種存在し、SIK2、SIK3とそれぞれ命名されている。(以後、SIK1/2/3に共通する場合はSIKとのみ記載する。)SIKシグナルカスケードの顕著な役割はcAMPで活性化される転写因子CREBのフィードバック的な抑制である(JBC 2002:277 15629-37, Cell 2004:11461-76)。CREBはTORC(transducer of regulated CREB activity)と名付けられた転写共役因子が存在しないと転写能を示さない。SIKはTORCをリン酸化依存的に失活させることでCREBを抑制する。生理的な役割としては、SIK1およびSIK2は肝臓におけるCREB依存的糖新生の抑制(Nature 2005:4371109-11)、SIK2は膵臓でのCREB依存的インスリン分泌の抑制(Cell 2004:11461-76)、脳におけるCREB依存的な神経栄養因子(BDNF)の発現抑制(Neuron 2011:69 106-119)などが知られている。一方、SIK1はクラス2-HDAC(ヒストンデアセチラーゼ)を抑制することで、MEF2C等に依存する転写を亢進させること(Nat Med 2007:13 597-603)が報告されている。しかし、SIK3の生理的意義は明らかにされておらず、ノックアウトマウスおよびトランスジェニックマウスの解析が待たれていた。
本発明のスクリーニング方法のターゲットとなるSIK3は、どのような生物由来のSIK3でもよく、特に限定されないが、哺乳動物のSIK3が好ましい。哺乳動物としては、ヒト、チンパンジー、サル、イヌ、ウシ、マウス、ラット、モルモットなどが好ましく、より好ましくはヒトである。各種動物のSIK3をコードする遺伝子の塩基配列およびアミノ酸配列の情報は、例えば表1に示すアクセッション番号で公知のデータベース(DDBJ/GenBank/EMBL等)から取得することができる。
本発明のスクリーニング方法を適用する被験物質としては、例えば、例えば、核酸、ペプチド、タンパク、非ペプチド性化合物、合成化合物、発酵生産物、細胞抽出液、細胞培養上清、植物抽出液、哺乳動物の組織抽出液、血漿等が挙げられる。被験物質は、新規な物質であってもよいし、公知の物質であってもよい。これら被験物質は塩を形成していてもよく、被験物質の塩としては、生理学的に許容される酸や塩基との塩が用いられる。
本発明のスクリーニング方法によって得られる物質は、種々の軟骨疾患の治療薬の有効成分として有用である。軟骨疾患としては、例えば、変形性関節症、軟骨損傷、軟骨形成異常症が挙げられる。軟骨形成異常症には、軟骨無形成症、軟骨形成不全症、軟骨異栄養症、軟骨発育不全症、内軟骨腫症などが含まれる。これらのうち、変形性関節症および軟骨損傷は、軟骨量が減少する病気である。また、軟骨形成異常症は軟骨の成長が障害される病気である。軟骨形成異常症の中で、例えば軟骨無形成症は、軟骨細胞の分化が亢進していると考えられる(Deng C, et al. Cell. 1996 Mar 22; 84(6):911-21、Minina E, et al. Dev Cell. 2002 Sep; 3(3):439-49、Dailey L, et al. J Cell Biol. 2003 Jun 23; 161(6):1053-66)。また、軟骨形成異常症の中で、例えばJansen-type metaphyseal chondrodysplasiaは、軟骨細胞の分化が抑制されていると考えられる(Schipani E, et al. Science. 1995 Apr 7; 268(5207):98-100)。
本発明のスクリーニング方法の第1の実施形態は、被験物質と内因性のSIK3を発現している細胞とを接触させる工程と、前記細胞のSIK3のタンパク質量またはmRNA量を測定する工程と、被験物質依存的なSIK3のタンパク質またはmRNA量の変化を分析する工程とを含むスクリーニング方法である。本実施形態のスクリーニング方法により、SIK3の発現を抑制する物質またはSIK3の発現を亢進する物質を選択することができる。内因性のSIK3を発現している細胞は、内因性のSIK3を発現している限り特に限定されず、生体内の細胞でもよく、培養細胞でもよい。実施の簡便さの点で培養細胞が好ましい。培養細胞は、初代培養細胞でもよく、細胞株でもよい。内因性のSIK3を発現している細胞株として、具体的には、例えば、HepG2、COS-7、ATDC5などが挙げられ、これらはいずれも本発明のスクリーニング方法の第1の実施形態に好適に用いることができる。
被験物質と内因性のSIK3を発現している細胞とを接触させる方法は、被験物質と細胞が接触できる方法であればどのような方法でもよく、特に限定されない。例えば、培養細胞を用いる場合には、培地に被験物質を添加する方法などが挙げられる。例えば、生体において被験物質と細胞とを接触させる場合には、経口投与、静脈内投与、腹腔内投与等の全身投与、標的臓器や標的組織への局所投与などが挙げられる。また、被験物質を接触させない対照群を設けることが好ましい。
細胞のSIK3タンパク質量は、公知の方法で細胞からタンパク質を抽出し、公知のタンパク質量測定方法を用いて定量することができる。公知のタンパク質量測定方法としては、例えば、ウエスタンブロット法、EIA法、ELISA法、RIA法、タンパク質測定試薬を用いる方法などが挙げられる。細胞のSIK3mRNA量は、公知の方法で細胞からRNAを抽出し、公知のmRNA量測定方法を用いて定量することができる。公知のmRNA量測定方法としては、ノーザンブロット法、RT-PCR法、定量RT-PCR法、RNaseプロテクションアッセイなどが挙げられる。
被験物質依存的なSIK3のタンパク質量またはmRNA量の変化を分析する方法は特に限定されない。例えば、被験物質を接触させない対照群におけるSIK3のタンパク質量またはmRNA量と比較して、被験物質を接触させた場合にSIK3のタンパク質量またはmRNA量が減少または増加していれば、当該被験物質を目的物質として選択すれることができる。被験物質がSIK3のタンパク質量またはmRNA量を減少または増加させる程度は特に限定されないが、例えば、被験物質を接触させていない細胞のタンパク質量またはmRNA量と比較して50%以下または150%以上にさせる被験物質が好ましく、25%以下または175%以上にさせる被験物質がより好ましい。
本発明のスクリーニング方法の第2の実施形態は、被験物質とSIK3のプロモーター活性を確認可能な細胞とを接触させる工程と、前記細胞のSIK3のプロモーター活性を測定する工程と、被験物質依存的なSIK3のプロモーター活性の変化を分析する工程を含むスクリーニング方法である。本実施形態のスクリーニング方法により、SIK3の発現を抑制する物質またはSIK3の発現を亢進する物質を選択することができる。SIK3のプロモーター活性を確認可能な細胞としては、例えば、SIK3のプロモーター活性を有するDNA断片の下流にレポーター遺伝子を融合したベクターが導入された細胞が挙げられる。宿主細胞は特に限定されないが、例えば、HEK293、HepG2、COS-7、ATDC5などが挙げられる。
ヒトSIK3遺伝子のプロモーター領域の塩基配列を配列番号1に示した。また、マウスSIK3遺伝子のプロモーター領域の塩基配列を配列番号2に示した。SIK3のプロモーター活性を有するDNA断片として、配列番号1で表される塩基配列と同一または実質的に同一な塩基配列を有するDNA断片、および配列番号2で表される塩基配列と同一または実質的に同一な塩基配列を有するDNA断片を好適に用いることができる。SIK3のプロモーター活性を有するDNA断片はこれらに限定されるものではなく、他の生物由来のSIK3遺伝子のプロモーター領域を有するDNA断片を用いてもよい。
配列番号1または2で表される塩基配列と実質的に同一な塩基配列を有するDNA断片としては、例えば、配列番号1または2で表される塩基配列と約50%以上、好ましくは約60%以上、さらに好ましくは約70%以上、より好ましくは約80%以上、特に好ましくは約90%以上、最も好ましくは約95%以上の相同性がある塩基配列を有し、配列番号1または2で表される塩基配列を有するDNA断片と同質のプロモーター活性を有するDNA断片であればどのようなものでもよい。
配列番号1または2で表される塩基配列と実質的に同一な塩基配列を有するDNA断片としては、例えば、配列番号1または2で表される塩基配列と約50%以上、好ましくは約60%以上、さらに好ましくは約70%以上、より好ましくは約80%以上、特に好ましくは約90%以上、最も好ましくは約95%以上の相同性がある塩基配列を有し、配列番号1または2で表される塩基配列を有するDNA断片と同質のプロモーター活性を有するDNA断片であればどのようなものでもよい。
レポーター遺伝子は、一般に用いられているものであれば特に限定されないが、安定かつ活性の定量が容易なものが好ましい。例えば、ルシフェラーゼ、β-ガラクトシダーゼ、β-グルクロニダーゼ、クロラムフェニコールアセチルトランスフェラーゼ、アルカリホスファターゼ、ペルオキシダーゼ、緑色蛍光タンパク質(GFP)等をコードする遺伝子が挙げられる。
被験物質とSIK3のプロモーター活性を確認可能な細胞とを接触させる方法は、被験物質と細胞が接触できる方法であればどのような方法でもよく、特に限定されない。例えば、培地に被験物質を添加する方法などが挙げられる。また、被験物質を接触させない対照群を設けることが好ましい。
SIK3のプロモーター活性は、レポーター遺伝子の発現量により測定することができる。レポーター遺伝子の発現量は、レポーター遺伝子がコードするタンパク質(レポーター遺伝子産物)に応じて適宜選択される。例えば、レポーター遺伝子がルシフェラーゼをコードする場合、導入細胞を適当な方法により溶解し、この細胞溶解液の上清中に基質となるルシフェリンを添加した後、市販の検出器を用いて発光量を測定し、レポーター遺伝子の発現量とする。他のレポーター遺伝子を用いた場合も、公知の方法でレポーター遺伝子産物量を測定することができる。
被験物質依存的なSIK3のプロモーター活性の変化を分析する方法は特に限定されない。例えば、例えば、被験物質を接触させない対照群におけるSIK3のプロモーター活性と比較して、被験物質を接触させた場合にSIK3のプロモーター活性が低下または上昇していれば、当該被験物質を目的物質として選択すれることができる。被験物質がプロモーター活性を低下または上昇させる程度は特に限定されないが、例えば、被験物質を接触させていない細胞のプロモーター活性と比較して50%以下または150%以上にさせる被験物質が好ましく、25%以下または175%以上にさせる被験物質がより好ましい。
本発明のスクリーニング方法の第3の実施形態は、被験物質とSIK3とSIK3の基質とを接触させる工程と、SIK3とSIK3の基質との結合状態を確認する工程と、被験物質依存的な結合状態の変化を分析する工程を含むスクリーニング方法である。本実施形態のスクリーニング方法により、SIK3の機能を抑制する物質またはSIK3の機能を亢進する物質を選択することができる。SIK3の基質は、以下の式(1)で表されるアミノ酸配列を有するタンパク質またはペプチドであればどのようなものでもよく、例えば、Cell. 2004 119:61-74 や FEBS J. 2006 273:2730-2748 等に記載の方法が利用できる。具体的には、例えば、前述のTORC、HDAC、これらのフラグメント(式(1)で表されるアミノ酸配列を含む)等が挙げられる。

SIK3は、式(1)で表されるアミノ酸配列を有するタンパク質を基質として認識し、これと結合して当該アミノ酸配列の第6位のSerをリン酸化する。なお、式(1)で表されるアミノ酸配列の第1位のLeuおよび第10位のLeuは、他の疏水性アミノ酸に置換されていてもよい。

SIK3は、式(1)で表されるアミノ酸配列を有するタンパク質を基質として認識し、これと結合して当該アミノ酸配列の第6位のSerをリン酸化する。なお、式(1)で表されるアミノ酸配列の第1位のLeuおよび第10位のLeuは、他の疏水性アミノ酸に置換されていてもよい。
本実施形態のスクリーニング方法に用いるSIK3は、天然タンパク質および組換えタンパク質のいずれでもよい。SIK3は、例えば、内因性のSIK3を発現している細胞やSIK3発現ベクターが導入された細胞の培養上清または細胞抽出物から公知の方法(例えば、アフィニティーカラム)を用いて取得することができる。SIK3の基質も天然タンパク質および組換えタンパク質のいずれでもよく、SIK3と同様に、公知の方法で取得することができる。例えば、TORCのアミノ酸配列や、TORCをコードする遺伝子の塩基配列は、公知のデータベース(DDBJ/GenBank/EMBL等)から取得することができる。なお、ヒトTORC2のアミノ酸配列のアクセッション番号はNP_859066であり、これをコードする遺伝子の塩基配列のアクセッション番号はNM_181715である。ヒトTORC2のアミノ酸配列(693アミノ酸、配列番号3)において、上記式(1)の配列は第166~175位に存在する。
被験物質とSIK3とSIK3の基質とを接触させる方法は特に限定されない。例えば、SIK3とSIK3の基質とを含む反応系を準備し、ここに被験物質を添加する方法が挙げられる。接触時間、接触温度は特に限定されず、適宜選択すればよい。また、被験物質を接触させない対照群を設けることが好ましい。
SIK3とSIK3の基質との結合状態を確認する方法は、特に限定されず、公知の方法を適宜選択して使用することができる。例えば、ELISA法を好適に用いることができる。ELISA法を用いて本実施形態のスクリーニング方法を実施する場合、SIK3およびSIK3の基質のいずれか一方を固相化し、そこに他方および被験物質を添加して反応させ、SIK3とSIK3の基質の結合状態を適当な一次抗体および二次抗体を用いて検出すればよい。
SIK3とSIK3の基質との結合状態を確認する方法は、特に限定されず、公知の方法を適宜選択して使用することができる。例えば、ELISA法を好適に用いることができる。ELISA法を用いて本実施形態のスクリーニング方法を実施する場合、SIK3およびSIK3の基質のいずれか一方を固相化し、そこに他方および被験物質を添加して反応させ、SIK3とSIK3の基質の結合状態を適当な一次抗体および二次抗体を用いて検出すればよい。
被験物質依存的な結合状態の変化を分析する方法は特に限定されない。例えば、被験物質を接触させない対照群におけるSIK3とSIK3の基質との結合状態と比較して、被験物質を接触させた場合にSIK3とSIK3の基質との結合状態が減弱または増強していれば、当該被験物質を目的物質として選択すれることができる。被験物質がSIK3とSIK3の基質との結合状態を減弱または増強させる程度は特に限定されないが、例えば、被験物質を接触させていない場合の両者の結合状態と比較して、結合状態を50%以下または150%以上にさせる被験物質が好ましく、25%以下または175%以上にさせる被験物質がより好ましい。
本発明のスクリーニング方法の第4の実施形態は、被験物質とSIK3とSIK3の基質とを接触させる工程と、SIK3の基質のリン酸化状態を確認する工程と、被験物質依存的なリン酸化状態の変化を分析する工程を含むスクリーニング方法である。本実施形態のスクリーニング方法により、SIK3の機能を抑制する物質またはSIK3の機能を亢進する物質を選択することができる。
本実施形態では、上記第3の実施形態と同じSIK3およびSIK3の基質を使用することができ、同様の方法でこれらと被験物質を接触させることができる。SIK3の基質のリン酸化状態を確認する方法は特に限定されず、公知の方法を適宜選択して使用することができる。例えば、リン酸化状態を識別し得る抗体を用いる方法が挙げられる。本実施形態において、SIK3の基質として、上記のヒトTORC2を用いる場合には、ヒトTORC2のアミノ酸配列(配列番号3)の第171位のセリンがリン酸化されたタンパク質を特異的に認識する抗体を用いればよい。このような抗体は、ヒトTORC2の第171位のセリンがリン酸化されたタンパク質またはリン酸化部位を含むペプチドを免疫原として公知のポリクローナル抗体またはモノクローナル抗体の作製方法に従って取得することができる。例えば、特許第4568022号に記載の方法である。
被験物質依存的なリン酸化状態の変化を分析する方法は特に限定されない。例えば、被験物質を接触させない対照群におけるSIK3の基質のリン酸化状態と比較して、被験物質を接触させた場合にSIK3の基質のリン酸化状態が減弱または増強していれば当該被験物質を目的物質として選択すれることができる。被験物質がSIK3の基質のリン酸化状態を減弱または増強させる程度は特に限定されないが、例えば、被験物質を接触させていない場合のリン酸化状態と比較して、リン酸化状態を50%以下または150%以上にさせる被験物質が好ましく、25%以下または175%以上にさせる被験物質がより好ましい。
本発明のスクリーニング方法の第5の実施形態は、SIK3のシグナル伝達を評価し得る系に被験物質を添加する工程と、SIK3のシグナル伝達状態を確認する工程と、被験物質依存的なシグナル伝達状態の変化を分析する工程を含むスクリーニング方法である。本実施形態のスクリーニング方法により、SIK3の機能を抑制する物質またはSIK3の機能を亢進する物質を選択することができる。SIK3のシグナル伝達を評価し得る系は、SIK3活性が正常な場合とSIK3活性が抑制または亢進された場合において、シグナル伝達の下流の差異を評価できる系であれば特に限定されない。例えば、SIK3活性が正常な場合とSIK3活性が抑制または亢進された場合とで、レポーター遺伝子の発現量が変化するような系を構築して用いることができる。具体的には、例えば、後述する実施例1で使用した系(SIK3の基質の発現ベクターと、当該基質のリン酸化により活性化されるプロモーターにレポーター遺伝子が結合したベクターが導入された細胞を用いる系)などが挙げられる。
SIK3のシグナル伝達状態を確認する方法は、用いた系に応じて公知の方法を適宜選択すればよい。例えば、レポーター遺伝子を用いる系の場合は、用いたレポーター遺伝子に応じて、公知の方法でレポーター遺伝子産物量を測定すればよい。
被験物質依存的なシグナル伝達状態の変化を分析する方法は特に限定されない。例えば、被験物質を接触させない対照群におけるSIK3のシグナル伝達状態と比較して、被験物質を接触させた場合にSIK3の活性が低下または上昇しているとの評価が得られれば、当該被験物質を目的物質として選択すれることができる。被験物質がSIK3の活性を低下または上昇させる程度は特に限定されないが、例えば、被験物質を接触させていない場合と比較して、50%以下または150%以上にさせる被験物質が好ましく、25%以下または175%以上にさせる被験物質がより好ましい。
被験物質依存的なシグナル伝達状態の変化を分析する方法は特に限定されない。例えば、被験物質を接触させない対照群におけるSIK3のシグナル伝達状態と比較して、被験物質を接触させた場合にSIK3の活性が低下または上昇しているとの評価が得られれば、当該被験物質を目的物質として選択すれることができる。被験物質がSIK3の活性を低下または上昇させる程度は特に限定されないが、例えば、被験物質を接触させていない場合と比較して、50%以下または150%以上にさせる被験物質が好ましく、25%以下または175%以上にさせる被験物質がより好ましい。
〔スクリーニングにより得られた物質を含有する医薬〕
上記、本発明のスクリーニング方法を用いて得られる物質は、軟骨量を増大させる作用および/または軟骨細胞分化を正常化させる作用を有する物質であるので、種々の軟骨疾患の治療薬の有効成分として有用である。軟骨疾患としては、例えば、変形性関節症、軟骨損傷、軟骨形成異常症などが挙げられる。したがって、本発明には、SIK3の発現もしくは機能を抑制する物質、または、SIK3の発現もしくは機能を増強する物質を有効成分とする軟骨疾患の治療用医薬が含まれる。ここで、「軟骨量を増大させる作用を有する物質」とは、当該物質を正常個体に投与した場合に、当該物質を投与していない正常個体と比較して、同一部位(例えば四肢の関節)の軟骨量を増大させる作用を有する物質を意味する。軟骨量が増大しているか否かを確認する方法は特に限定されないが、例えば、比較する軟骨組織の組織標本を作製して顕微鏡観察することにより行うことができる。
上記、本発明のスクリーニング方法を用いて得られる物質は、軟骨量を増大させる作用および/または軟骨細胞分化を正常化させる作用を有する物質であるので、種々の軟骨疾患の治療薬の有効成分として有用である。軟骨疾患としては、例えば、変形性関節症、軟骨損傷、軟骨形成異常症などが挙げられる。したがって、本発明には、SIK3の発現もしくは機能を抑制する物質、または、SIK3の発現もしくは機能を増強する物質を有効成分とする軟骨疾患の治療用医薬が含まれる。ここで、「軟骨量を増大させる作用を有する物質」とは、当該物質を正常個体に投与した場合に、当該物質を投与していない正常個体と比較して、同一部位(例えば四肢の関節)の軟骨量を増大させる作用を有する物質を意味する。軟骨量が増大しているか否かを確認する方法は特に限定されないが、例えば、比較する軟骨組織の組織標本を作製して顕微鏡観察することにより行うことができる。
本発明のスクリーニング方法を用いて得られる物質を、軟骨疾患の治療薬として使用する場合、常套手段に従って製剤化することができる。例えば、経口投与のための製剤としては、固体または液体の剤形、具体的には錠剤(糖衣錠、フィルムコーティング錠を含む)、丸剤、顆粒剤、散剤、カプセル剤(ソフトカプセル剤を含む)、シロップ剤、乳剤、懸濁剤などが挙げられる。これらの製剤は公知の方法によって製造され、製剤分野において通常用いられる担体、希釈剤もしくは賦形剤を含有するものである。例えば、錠剤用の担体、賦形剤としては、乳糖、でんぷん、蔗糖、ステアリン酸マグネシウムなどが用いられる。非経口投与のための製剤としては、例えば、注射剤、坐剤などが用いられ、注射剤は静脈注射剤、皮下注射剤、皮内注射剤、筋肉注射剤、点滴注射剤、関節内注射剤などの剤形を包含する。このような注射剤は、公知の方法に従って、例えば、上記SIK3の発現または機能を抑制する物質またはその塩を通常注射剤に用いられる無菌の水性もしくは油性液に溶解、懸濁または乳化することによって調製する。注射用の水性液としては、例えば、生理食塩水、ブドウ糖やその他の補助薬を含む等張液などが用いられ、適当な溶解補助剤、例えば、アルコール(例えば、エタノール等)、ポリアルコール(例えば、プロピレングリコール、ポリエチレングリコール等)、非イオン界面活性剤(例えば、ポリソルベート80、HCO-50等)などと併用してもよい。油性液としては、例えば、ゴマ油、大豆油などが用いられ、溶解補助剤として安息香酸ベンジル、ベンジルアルコールなどを併用してもよい。直腸投与に用いられる坐剤は、上記SIK3の発現または機能を抑制する物質またはその塩を通常の坐薬用基剤に混合することによって調製される。
このようにして得られる製剤は安全で低毒性であるので、例えば、ヒトまたは温血動物(例えば、マウス、ラット、ウサギ、ヒツジ、ブタ、ウシ、ウマ、トリ、ネコ、イヌ、サル、チンパンジーなど)に対して経口的にまたは非経口的に投与することができる。
このようにして得られる製剤は安全で低毒性であるので、例えば、ヒトまたは温血動物(例えば、マウス、ラット、ウサギ、ヒツジ、ブタ、ウシ、ウマ、トリ、ネコ、イヌ、サル、チンパンジーなど)に対して経口的にまたは非経口的に投与することができる。
〔改変軟骨細胞〕
本発明は、SIK3の発現または機能が抑制されている改変軟骨細胞、および、SIK3の発現または機能が増強されている改変軟骨細胞を提供する。これらの改変軟骨細胞は、軟骨量が減少する病気および/または軟骨の成長が障害される病気の改善のために利用することができる。軟骨量が減少する病気としては、変形性関節症、軟骨損傷などが挙げられ、軟骨の成長が障害される病気としては、軟骨形成異常症などが挙げられる。具体的には、例えば、SIK3の発現または機能が抑制されている改変軟骨細胞は、軟骨細胞の成熟や変性が抑制されて軟骨細胞の形態を維持することができるので、軟骨が減少する疾患の再生医療に用いる細胞として非常に有用である。また、例えば、SIK3の発現または機能が抑制または増強されている改変軟骨細胞は、それぞれ、軟骨細胞の分化が亢進または抑制されている軟骨形成異常症の成長軟骨への移植に用いる細胞として非常に有用である。さらに、例えば、SIK3の発現または機能が抑制または増強されている改変軟骨細胞は、疾患モデル細胞として、軟骨形成異常症の病態解析や薬剤スクリーニングに用いる細胞として非常に有用である。
本発明は、SIK3の発現または機能が抑制されている改変軟骨細胞、および、SIK3の発現または機能が増強されている改変軟骨細胞を提供する。これらの改変軟骨細胞は、軟骨量が減少する病気および/または軟骨の成長が障害される病気の改善のために利用することができる。軟骨量が減少する病気としては、変形性関節症、軟骨損傷などが挙げられ、軟骨の成長が障害される病気としては、軟骨形成異常症などが挙げられる。具体的には、例えば、SIK3の発現または機能が抑制されている改変軟骨細胞は、軟骨細胞の成熟や変性が抑制されて軟骨細胞の形態を維持することができるので、軟骨が減少する疾患の再生医療に用いる細胞として非常に有用である。また、例えば、SIK3の発現または機能が抑制または増強されている改変軟骨細胞は、それぞれ、軟骨細胞の分化が亢進または抑制されている軟骨形成異常症の成長軟骨への移植に用いる細胞として非常に有用である。さらに、例えば、SIK3の発現または機能が抑制または増強されている改変軟骨細胞は、疾患モデル細胞として、軟骨形成異常症の病態解析や薬剤スクリーニングに用いる細胞として非常に有用である。
本発明のSIK3の発現または機能が抑制されている改変軟骨細胞は、例えば、軟骨細胞に分化誘導可能な細胞のSIK3遺伝子を改変してSIK3遺伝子を不活化し、当該細胞を軟骨細胞に分化誘導することにより取得することができる。また、例えば、軟骨細胞に、siRNA、shRNAまたはアンチセンスオリゴヌクレオチドを導入することにより取得することができる。また、例えば、SIK3のノックアウトマウス(参考例1参照)から、軟骨細胞を採取することにより取得することができる。
本発明のSIK3の発現または機能が増強されている改変軟骨細胞は、例えば、軟骨細胞に分化誘導可能な細胞にSIK3発現ベクターを導入してSIK3の発現が増強された細胞を選択し、当該細胞を軟骨細胞に分化誘導することにより取得することができる。また、軟骨細胞にSIK3発現ベクターを導入してSIK3の発現が増強された軟骨細胞を選択することにより取得することができる。また、例えば、SIK3遺伝子のトランスジェニックマウス(参考例2参照)から、軟骨細胞を採取することにより取得することができる。
本発明のSIK3の発現または機能が抑制されている改変軟骨細胞の製造方法として、例えば、軟骨細胞に分化誘導可能な細胞のSIK3遺伝子を改変して、SIK3遺伝子を不活化する工程と、得られた細胞を軟骨細胞に分化誘導する工程を含む方法を好適に用いることができる。軟骨細胞に分化誘導可能な細胞としては、例えば幹細胞、未熟な軟骨細胞などが挙げられる。幹細胞は、自己複製能と多分化能を持った細胞を意味し、体性幹細胞、多能性幹細胞などが含まれる。軟骨細胞に分化誘導可能な体性幹細胞としては、間葉系幹細胞が挙げられる。軟骨細胞に分化誘導可能な体性幹細胞としては、ES細胞(胚性幹細胞)、iPS細胞(人工多能性幹細胞)、mGS細胞(多能性生殖幹細胞)、ES細胞と体細胞との融合細胞などが挙げられる。好ましくは、間葉系幹細胞、ES細胞およびiPS細胞である。
SIK3遺伝子を改変する方法は特に限定されず、公知の遺伝子改変方法を適宜選択して用いることができる。例えば、紫外線や変異原物質を用いてランダムに変異を導入する方法、部位特異的に変異を導入する方法、ターゲティングベクターを用いて変異を導入する方法などが挙げられる。SIK3遺伝子を破壊するためのターゲティングベクターとしては、例えば、本発明者らがSIK3のノックアウトマウスを作製する際に使用したターゲティングベクター(図1(a)参照)などが挙げられる。SIK3遺伝子が不活化していることは、例えば、SIK3遺伝子を改変した細胞のSIK3のmRNA量やタンパク質量を測定すること、SIK3遺伝子を改変した細胞から単離したSIK3タンパク質のキナーゼ活性を測定することにより確認することができる。
得られた細胞を軟骨細胞に分化誘導する方法は、例えば、文献 J Clin Invest. 2011 121:640-57. に記載の方法を好適に用いることができる。

以下、参考例および実施例により本発明を詳細に説明するが、本発明はこれらに限定されるものではない。
〔参考例1:SIK3ノックアウトマウスにおける軟骨細胞の分化解析〕
(1)SIK3ノックアウトマウス(以下「SIK3KOマウス」という)
マウスSIK3遺伝子の5’側のゲノム配列(5’arm)、pgkプロモーター、ネオマイシン耐性遺伝子、マウスSIK3遺伝子の3’側のゲノム配列(3’arm)を順に持つターゲティングベクター(図1(a)参照)を、定法に従い作製した。得られたターゲティングベクターを、マウスES細胞に導入し、G418を含む培地で培養してネオマイシン耐性ES細胞を得た。得られたネオマイシン耐性ES細胞からDNAを抽出し、サザンブロット解析により相同組換えクローンを同定した(図1(b)参照)。
得られた相同組換えクローンをBDF2マウスより得た胚盤胞へマイクロインジェクションし、操作胚を子宮へ移植した。操作胚を移植し妊娠したマウスより、キメラマウスを出産させた。キメラマウスをC57BL/6マウスと交配することによって、変異アレルを持つヘテロ接合体マウス(以下「SIK3ヘテロKOマウス」という)を得た。SIK3ヘテロKOマウス同士の交配によってホモ接合体のSIK3KOマウスを得た。胎児または出生児の遺伝子型は、尾、耳または皮膚の一部からゲノムDNAを抽出し、PCRにて確認した。PCRには、以下の3種類のプライマーを用いた。プライマーaおよびプライマーbから、野生型アレル由来の270bpのバンドが増幅され、プライマーaおよびプライマーcから、変異アレル由来の429bpのバンドが増幅される(図1(c)参照)。なお、SIK3ヘテロKOマウスは、独立行政法人医薬基盤研究所から入手可能である。

SIK3ヘテロKOマウス同士の交尾を確認し、所定の日数経過後に妊娠マウスを二酸化炭素投与により安楽死させ、14.5日胚(14.5dpc)および交尾後18.5日胚(18.5dpc)を得た。胎児の肢芽を採取し、ホルマリン固定した。
(3)生後マウスからの四肢採取
生後3月齢のSIK3KOマウスおよび同腹の野生型(WT)マウスを二酸化炭素投与により安楽死させ、解剖して四肢を採取し、ホルマリン固定およびEDTAによる脱灰を行った。
(4)組織標本作製
採取、固定した四肢の1つをパラフィン包埋し、5マイクロメートルの厚みの切片を作製した。切片をスライドガラスに貼付し、脱パラフィン後サフラニンO-ファーストグリーン-アイアンヘマトキシリン染色を行った。軟骨マトリックスはサフラニンOにより赤~オレンジ色に染色される。
SIK3ヘテロKOマウス同士の交尾を確認し、所定の日数経過後に妊娠マウスを二酸化炭素投与により安楽死させ、14.5日胚(14.5dpc)および交尾後18.5日胚(18.5dpc)を得た。胎児の肢芽を採取し、ホルマリン固定した。
(3)生後マウスからの四肢採取

(5)標本の観察
得られた標本を光学顕微鏡で観察した。図2(a)、(b)に14.5dpc胚の大腿骨を含む切片の組織染色像を示した。(a)は野生型マウス、(b)はSIK3KOマウスである。また、図において大腿骨内で濃く染まっているのが肥大化していない軟骨細胞である。図2(a)、(b)から明らかなように、14.5dpc胚において、(a)野生型マウスでは大腿骨の中央部において軟骨細胞が肥大化し、染まりが薄くなっているが、(b)KOマウスでは大腿骨の中央部の中央部がほとんど薄くなっておらず、軟骨細胞の肥大化が抑制されていることが分かった。
図3(a)、(b)に18.5dpc胚の大腿骨を含む切片の組織染色像を示した。(a)は野生型マウス、(b)はSIK3KOマウスである。図3(a)、(b)から明らかなように、野生型マウス(a)では大腿骨の中央部において骨化が進行しているが、KOマウス(b)では未だ軟骨細胞の肥大化が抑制されており、骨化は生じていないことが分かった。
図4(a)、(b)に3月齢マウスの膝関節を含む切片の組織染色像を示した。(a)は野生型マウス、(b)はSIK3KOマウスである。図4(a)、(b)から明らかなように、(b)KOマウスの関節軟骨は、(a)野生型マウスの関節軟骨と比較して、顕著に肥厚していることが分かった。
以上の結果から、SIK3KOマウスにおいて軟骨細胞の肥大化の抑制および関節軟骨の肥厚が観察されたことから、SIK3は、軟骨細胞肥大化抑制および軟骨肥厚化のターゲットとなる可能性が示唆された。
得られた標本を光学顕微鏡で観察した。図2(a)、(b)に14.5dpc胚の大腿骨を含む切片の組織染色像を示した。(a)は野生型マウス、(b)はSIK3KOマウスである。また、図において大腿骨内で濃く染まっているのが肥大化していない軟骨細胞である。図2(a)、(b)から明らかなように、14.5dpc胚において、(a)野生型マウスでは大腿骨の中央部において軟骨細胞が肥大化し、染まりが薄くなっているが、(b)KOマウスでは大腿骨の中央部の中央部がほとんど薄くなっておらず、軟骨細胞の肥大化が抑制されていることが分かった。

以上の結果から、SIK3KOマウスにおいて軟骨細胞の肥大化の抑制および関節軟骨の肥厚が観察されたことから、SIK3は、軟骨細胞肥大化抑制および軟骨肥厚化のターゲットとなる可能性が示唆された。
〔参考例2:SIK3トランスジェニックマウスを用いたレスキュー試験〕
(1)SIK3トランスジェニックマウス(以下「SIK3Tgマウス」という)
軟骨特異的発現をもたらすXI型コラーゲンα2鎖遺伝子(Col11a2)プロモーター/エンハンサー(Tsumaki N, et al. J Cell Biol. 1996; 134:1573-1582.)にヒトSIK3cDNA(ACCESSION NM_025164)を結合してトランスジーンコンストラクトを作製した。このトランスジーンインサートをベクターバックボーンから切り出して精製した後、マウス受精卵にマイクロインジェクションし、それらを偽妊娠マウスの卵管に移植した。偽妊娠マウスから生まれたマウスからSIK3Tgマウスを選別し、SIK3Tgマウスラインを樹立した。胎児または出生児の遺伝子型は、尾または耳組織よりゲノムDNAを抽出し、PCRにて確認した。PCRには、以下のプライマーを用いた。

SIK3ヘテロKOマウスとSIK3Tgマウスを交配し、交尾確認後15.5日で妊娠マウスを二酸化炭素投与により安楽死させ、15.5日胚(15.5dpc)を得た。各胎児の皮膚よりゲノムDNAを抽出し、PCRにてSIK3ノックアウトとSIK3トランスジェニックの遺伝子型を解析した。胎児の肢芽を採取し、ホルマリン固定した。
(3)組織標本作製
参考例1と同様の手順で切片を作製し、染色を施した。
SIK3ヘテロKOマウスとSIK3Tgマウスを交配し、交尾確認後15.5日で妊娠マウスを二酸化炭素投与により安楽死させ、15.5日胚(15.5dpc)を得た。各胎児の皮膚よりゲノムDNAを抽出し、PCRにてSIK3ノックアウトとSIK3トランスジェニックの遺伝子型を解析した。胎児の肢芽を採取し、ホルマリン固定した。

(4)標本の観察
得られた標本を光学顕微鏡で観察した。図5(a)~(d)に15.5dpc胚の大腿骨を含む切片の組織染色像を示した。(a)はSIK3Tg、SIK3+/+の遺伝子型を持つマウス、(b)はSIK3Tg、SIK3+/-の遺伝子型を持つマウス、(c)はSIK3-/-の遺伝子型を持つマウス、(d)はSIK3Tg、SIK3-/-の遺伝子型を持つマウスである。図5から明らかなように、(c)SIK3-/-の遺伝子型を持つマウス(SIK3KOマウス)は、参考例1と同様に大腿骨中央部の軟骨細胞の肥大化が抑制されていたが、(d)SIK3Tg、SIK3-/-の遺伝子型を持つマウスは、軟骨細胞の肥大化が抑制されていなかった。したがって、SIK3の機能がレスキューされていることが明らかとなった。
この結果から、軟骨細胞の肥大化抑制は、SIK3の機能欠失に基づくものであることが明らかとなった。
得られた標本を光学顕微鏡で観察した。図5(a)~(d)に15.5dpc胚の大腿骨を含む切片の組織染色像を示した。(a)はSIK3Tg、SIK3+/+の遺伝子型を持つマウス、(b)はSIK3Tg、SIK3+/-の遺伝子型を持つマウス、(c)はSIK3-/-の遺伝子型を持つマウス、(d)はSIK3Tg、SIK3-/-の遺伝子型を持つマウスである。図5から明らかなように、(c)SIK3-/-の遺伝子型を持つマウス(SIK3KOマウス)は、参考例1と同様に大腿骨中央部の軟骨細胞の肥大化が抑制されていたが、(d)SIK3Tg、SIK3-/-の遺伝子型を持つマウスは、軟骨細胞の肥大化が抑制されていなかった。したがって、SIK3の機能がレスキューされていることが明らかとなった。
この結果から、軟骨細胞の肥大化抑制は、SIK3の機能欠失に基づくものであることが明らかとなった。
〔参考例3:SIK3トランスジェニックマウスにおける膝関節成長軟骨板の観察〕
生後3月齢、6月齢および8月齢のSIK3Tgマウスおよび野生型マウスを二酸化炭素投与により安楽死させ、解剖して四肢を採取し、ホルマリン固定およびEDTAによる脱灰を行った。その後、参考例1と同様の手順で切片を作製し、染色を施した。
得られた標本を光学顕微鏡で観察した。図6に各月齢マウスの膝関節を含む切片の組織染色像を示した。左が野生型マウス(図中Wt)、右がSIK3Tgマウス(図中LI-hSIK3Tg)である。図6から明らかなように、3月齢では野生型マウスおよびSIK3Tgマウスのどちらにも成長軟骨板が観察されたが、6月齢および8月齢のSIK3Tgマウスでは、成長軟骨板が消失していた。一方、6月齢および8月齢の野生型マウスでは、成長軟骨板が観察された。
この結果から、SIK3の機能亢進は、軟骨細胞の肥大化を促進させ、軟骨細胞の骨化を加速するものと考えられた。

〔実施例1:スクリーニング方法〕
HEK293細胞に、レポーターベクターGAL4-Luc(ホタルルシフェラーゼ:pTAL-5XGAL4(150ng))、内因性レポターターベクター(リニーラルシフェラーゼ:pRL-Int(-)(30ng))、TORC発現ベクターの有り無し(pM-TORC2またはpMのみ(50ng))およびSIK3発現ベクターの有り無し(pTarget-SIK3またはpTargetのみ(50ng)をLipofectamine2000を利用して導入し、24時間後に被験物質を加えさらに6時間処理した。レポーター活性の測定には、プロメガ社のDual Luciferaseアッセイキットを利用した。被験物質には、スタウロスポリン(STS)、SIK2阻害剤であるCompoundCを用いた。コントロールにはDMSOのみを加えた。当該スクリーニング系の模式図を図6に示した。

結果を図7に示した。図7において縦軸はTORC2の活性を示し、ホタルルシフェラーゼ活性をリニーラルシフェラーゼで割り算した後、pM-TORC2の活性をpMの倍数で表示してある。白棒と黒棒はSIK3の有り無しの時のTORC2の活性を示しており、白棒(SIK3無し)と黒棒(SIK3有り)の差が大きければSIK3の活性が大きく、白棒(SIK3無し)と黒棒(SIK3有り)の差が小さければSIK3の活性が阻害されたと判定できる。図7から明らかなように、スタウロスポリン(STS)にSIK3阻害活性が検出された。
なお本発明は上述した各実施形態および実施例に限定されるものではなく、請求項に示した範囲で種々の変更が可能であり、異なる実施形態にそれぞれ開示された技術的手段を適宜組み合わせて得られる実施形態についても本発明の技術的範囲に含まれる。また、本明細書中に記載された学術文献および特許文献の全てが、本明細書中において参考として援用される。
Claims (2)
- 軟骨量を増大させる作用および/または軟骨細胞分化を正常化させる作用を有する物質のスクリーニング方法であって、塩誘導性キナーゼ3を使用することを特徴とする方法。
- 軟骨量が減少する病気および/または軟骨の成長が障害される病気の改善のために利用する、塩誘導性キナーゼ3の発現もしくは機能が抑制または増強されている改変軟骨細胞。
