Abstract

 検出装置(100B)は、発光素子(6)と、蛍光を受光するための受光素子(9)と、発光素子(6)で照射された捕集治具(12)からの蛍光の受光量に基づいて、捕集治具(12)で捕集された生物由来の粒子量を算出するための測定部(40)とを備え、捕集治具(12)のリフレッシュ動作としてヒータ(91)で捕集治具(12)を加熱することで捕集治具(12)上の捕集物を消失させる。

Description

検出装置および検出方法
 この発明は検出装置および検出方法に関し、特に、微生物などの、空気中の生物由来の粒子を検出する検出装置および検出方法に関する。
 従来、空気中の微生物の検出においては、落下菌法、衝突法、スリット法、多孔板法、遠心衝突法、インピンジャ法、およびフィルタ法などの方法で空気中の微生物を採取した後、培養し、出現するコロニーの計数を行なう。しかしながら、この方法では、培養に2日から3日が必要であり、リアルタイムでの検出は難しい。そこで、近年、特開2003-38163号公報(特許文献1)、特表2008-508527号公報(特許文献2)のように、空気中の微生物に紫外光を照射して、微生物からの蛍光発光を検出して個数を計測する装置が提案されている。
 特許文献1、2で提案されているような従来装置では、浮遊粒子が生物由来のものかどうかを判定する手段として、紫外線の照射により蛍光を発光するかどうかを判断する手法が採用されている。

 しかしながら、実際に空気中に浮遊する埃には、紫外光の照射により蛍光を発する化学繊維のくずなどが多く含まれている。それ故、特許文献1、2で提案されているような従来装置を用いると、空気中に存在する生物由来の粒子に加え、蛍光を発する埃も検出されてしまう。すなわち、特許文献1、2で提案されているような従来装置では、空気中に存在する生物由来の粒子だけを正確に評価できないという問題がある。
 本発明はこのような問題に鑑みてなされたものであって、蛍光を利用し、リアルタイムに、生物由来の粒子を、蛍光を発する埃から分離して検出することのできる検出装置および検出方法を提供することを目的としている
 上記目的を達成するために、本発明のある局面に従うと、検出装置は生物由来の粒子を検出するための検出装置であって、発光素子と、蛍光を受光するための受光素子と、発光素子で照射された捕集用部材からの蛍光の受光量に基づいて、捕集用部材で捕集された生物由来の粒子量を算出するための算出装置と、捕集用部材を清掃するための清掃機構とを備える。清掃機構は、捕集用部材を加熱するための機構を含む。
 好ましくは、検出装置は捕集用部材を加熱するためのヒータをさらに備え、算出装置は、ヒータで捕集用部材を加熱する前後の捕集用部材からの蛍光の受光量の変化に基づいて生物由来の粒子量を算出し、清掃手段に含まれる加熱するための機構は、ヒータが捕集用部材を加熱するための加熱温度よりも高い温度で捕集用部材を加熱する。
 より好ましくは、清掃機構はヒータを利用して捕集用部材を加熱し、ヒータは、捕集用部材に対して発光素子および受光素子と反対側に配置される。
 好ましくは、清掃機構に含まれる加熱するための機構は、捕集用部材に対して発光素子および受光素子と同じ側に配置される。
 より好ましくは、その加熱するための機構は、発熱を伴う光源である。
 好ましくは、清掃機構に含まれる加熱するための機構は、捕集用部材に対して発光素子および受光素子と反対側に配置される。
 好ましくは、清掃機構に含まれる加熱するための機構は、200℃以上で捕集用部材を加熱する。
 好ましくは、清掃機構は、加熱するための機構に加えて、捕集用部材に接触可能な部材を含み、加熱するための機構と接触可能な部材とを異なるタイミングに用いて捕集用部材を清掃する。
 本発明の他の局面に従うと、検出方法は生物由来の粒子を検出する方法であって、加熱前の捕集用部材の、発光素子の照射下での蛍光量を測定するステップと、加熱後の捕集用部材の、発光素子の照射下での蛍光量を測定するステップと、加熱前の捕集用部材から測定された蛍光量と、加熱後の捕集用部材から測定された蛍光量との変化量に基づいて、捕集用部材で捕集された生物由来の粒子量を算出するステップと、加熱後の捕集用部材をその加熱の後にさらに加熱することで、捕集用部材表面の捕集物を消失させるステップとを備える。
 この発明によると、リアルタイムかつ高精度で、生物由来の粒子を、蛍光を発する埃から分離して検出することができる。

 以下に、図面を参照しつつ、本発明の実施の形態について説明する。以下の説明では、同一の部品および構成要素には同一の符号を付してある。それらの名称および機能も同じである。
 実施の形態においては、図1に示される空気清浄機が微生物検出装置として機能するものとして以降の説明を行なうが、微生物検出装置単体として用いられるものであってもよい。
 <装置構成>
 図1を参照して、微生物検出装置100としての空気清浄機は、操作指示を受け付けるためのスイッチ110と、検出結果などを表示するための表示パネル130とを含む。その他、図示されない、空気を導入するための吸引口、排気するための排気口、などを含む。さらに、微生物検出装置100は、記録媒体を装着するための通信部150を含む。通信部150は、ケーブル400で外部装置としてのパーソナルコンピュータ(PC)300など接続するためのものであってもよい。または、通信部150は、インターネットを介して他の装置と通信するための通信回線を接続するためのものであってもよい。または、通信部150は、赤外線通信やインターネット通信などで他の装置と通信するためのものであってもよい。
 [第1の実施の形態]
 <微生物検出装置の装置構成>
 図2は、第1の実施の形態にかかる微生物検出装置100Aの基本構成を示す図である。
 空気清浄機の検出装置部分である微生物検出装置100Aには、検出機構と捕集機構と加熱機構とが含まれる。図2を参照して、微生物検出装置100Aは、孔5C’を有する区切り壁である壁5Cで隔てられた、捕集機構の少なくとも一部を含んだ捕集室5Aと、検出機構を含んだ検出室5Bとを備える。
 捕集室5Aには、捕集機構として針状の放電電極1および捕集治具12が配備され、検出室5Bには、検出機構として発光素子6、受光素子9、および集光レンズ13が配備される。
 捕集室5Aの放電電極1側および捕集治具12には、それぞれ、捕集室5A内に空気を導入するための導入孔10および排出孔11が設けられる。図2に示されるように、導入孔10にはフィルタ(プレフィルタ)10Bが設けられてもよい。
 導入孔10および排出孔11には、捕集室5A内への空気の出入りは可能として外部光の入射を遮断するための構成として、それぞれ、図3A、図3Bに表わされるような遮光部10Aおよび遮光部11Aが備えられてもよい。
 図3Aおよび図3Bを参照して、導入孔10に備えられる遮光部10Aおよび排出孔11に備えられる遮光部11A共に、4.5mm程度の間隔で遮光板10aおよび遮光板10bが交互に重ねられた構造を有する。遮光板10aおよび遮光板10bは、それぞれ図3Cおよび図3Dに示されるように導入孔10および排出孔11の形状(ここでは円形)に対応した形状であって、互いに重ならない部分に削孔を有する。具体的には、遮光板10aは周辺部分に削孔を有し、遮光板10bは中央部分に削孔を有する。遮光板10aおよび遮光板10bが重ねられたときに、それぞれの板に設けられた孔は重ならない。図3Aに示されるように、導入孔10に備えられる遮光部10Aは外部から内部へ遮光板10a、遮光板10b、遮光板10a、遮光板10bの順に遮光板が配置され、図3Bに示されるように、排出孔11に備えられる遮光部11Aは外部から内部へ遮光板10b、遮光板10a、遮光板10bの順に遮光板が配置される。この構成によって、捕集室5A内への空気の出入りは可能となるものの、外部光の入射が遮断され、捕集室5A内の迷光が抑えられる。
 排出孔11近傍には空気導入機構としてのファン50が設けられる。ファン50によって、吸引口からの空気が捕集室5Aに導入される。空気導入機構としては、たとえば、捕集室5A外に設置されたポンプおよびその駆動機構などであってよい。またたとえば、捕集室5A内に組み込まれた熱ヒータやマイクロポンプ、マイクロファン、およびその駆動機構などであってもよい。また、ファン50は、空気清浄機の空気清浄装置部分の空気導入機構と共通とする構成であってもよい。好ましくは、ファン50の駆動機構は、測定部40によって制御され、導入する空気の流速が制御される。好ましくは、ファン50で導入する空気の流速は1L(リットル)/minから50m3/minである。ファン50は測定部40によって制御される図示しない駆動機構により駆動することで、図中の点線矢印で表わされたように、導入孔10から捕集室5A外の空気を捕集室5A内に導入し、捕集室5A内の空気を排出孔11から捕集室5A外に排気する。
 捕集機構としては、公知の捕集機構を採用することができる。図2では、一例として特開2003-214997号公報に開示されている捕集機構を採用した場合を示している。すなわち、図2を参照して、捕集機構は、放電電極1、捕集治具12、および高圧電源2を含む。放電電極1は高圧電源2の正極に電気的に接続される。捕集治具12と高圧電源2の負極に電気的に接続される。
 捕集治具12は、導電性の透明の皮膜を有する、ガラス板などからなる支持基板である。支持基板は、ガラス板には限定されず、その他、セラミック、金属等であってもよい。また、支持基板表面に形成される皮膜は、透明に限定されない。他の例として、支持基板は、金属皮膜をセラミック等の絶縁材料の上に形成して構成されてもよい。また、支持基板が金属材料の場合は、その表面に皮膜を形成する必要もない。
 捕集治具12の皮膜側は高圧電源2の負極に電気的に接続される。これにより、放電電極1と捕集治具12と間に電位差が発生し、これらの間に図2の矢印Eに示される向きの電界が構成される。
 ファン50の駆動によって導入孔10から導入された空気中の浮遊粒子は、放電電極1付近にて負に帯電される。負に帯電した粒子は静電気力で捕集治具12の方向に移動して導電性の皮膜に吸着されることで、捕集治具12上に捕集される。ここで、放電電極1として針状電極を用いることによって、帯電した粒子を捕集治具12の放電電極1に対面する、(後述する)発光素子の照射領域15に対応したきわめて狭い範囲に吸着させることができる。これにより、後述する検出工程において、吸着された微生物を効率的に検出することができる。
 検出室5Bに含まれる検出機構は、光源である発光素子6と、受光素子9と、受光素子9の受光方向に備えられ、捕集機構により捕集治具12上に捕集された浮遊微粒子に発光素子6から照射することにより生じる蛍光を受光素子9に集光するための集光レンズ(またはレンズ群)13とを含む。その他、発光素子6の照射方向に備えられ、発光素子6からの光を平行光にする、または所定幅とするためのレンズ(またはレンズ群)、アパーチャ、照射光が受光素子9に入り込むのを防ぐためのフィルタ(またはフィルタ群)などが含まれてもよい。これらの構成は、従来技術を応用できる。集光レンズ13は、プラスチック樹脂製またはガラス製でよい。
 検出室5Bは、好ましくは、少なくとも内部に、黒色塗料の塗布または、黒色アルマイト処理等が施される。これにより、迷光の原因となる内部壁面での光の反射が抑えられる。捕集室5Aおよび検出室5B筐体の材質は特定の材質に限定されないが、好ましくは、プラスチック樹脂、アルミもしくはステンレスなどの金属、またはそれらの組み合わせが用いられる。導入孔10および排出孔11は、直径が1mmから50mmの円形である。導入孔10および排出孔11の形状は円形に限定されず、楕円形、四角形など他の形状であってもよい。
 発光素子6は、半導体レーザまたはLED(Light Emitting Diode)素子を含む。波長は、浮遊微粒子の生物由来の微粒子を励起して蛍光を発させるものであれば、紫外または可視いずれの領域の波長でもよい。好ましくは、特表2008-508527号公報に開示されているように、微生物中に含まれ、蛍光を発するトリプトファン、NaDH、リボフラビン等が効率よく励起される300nmから450nmである。受光素子9は、従来用いられている、フォトダイオード、イメージセンサなどが用いられる。
 発光素子6の発光は捕集治具12の表面に照射され、捕集治具12上に照射領域15を形成する。照射領域15の形状に限定はなく、円形、楕円形、四角形などであってよい。照射領域15は特定のサイズに限定されないが、好ましくは、円の直径または楕円の長軸方向の長さまたは四角形の1辺の長さが約0.05mmから50mmである。
 受光素子9は信号処理部30に接続されて、受光量に比例した電流信号を信号処理部30に対して出力する。従って、導入された空気中に浮遊し、捕集治具12表面に捕集された粒子に発光素子6から光が照射されることによって該粒子から発光された蛍光は、受光素子9において受光され、信号処理部30においてその受光量が検出される。
 検出室5B内の、捕集治具12表面に触れる位置には、捕集治具12表面をリフレッシュするためのブラシ60が設けられる。ブラシ60は、測定部40によって制御される図示しない移動機構に接続され、図中の両側矢印Bに示されるように、すなわち、捕集治具12上を往復するように移動する。これにより、捕集治具12表面に付着した埃や微生物が取り除かれる。
 加熱機構は、測定部40に電気的に接続され、測定部40によって加熱量(加熱時間、加熱温度等)が制御されるヒータ91を含む。ヒータ91としては、好適にはセラミックヒータが用いられる。ヒータ91は、捕集治具12上に捕集された空気中の浮遊粒子を加熱し得る位置であって、少なくとも加熱時には発光素子6、受光素子9等のセンサ機器から何かによって隔てられる位置に配備される。好ましくは、図2に表わされたように、捕集治具12の放電電極1から遠い側の面に配備される。より好ましくは、図4Aに示されるように、ヒータ91は周囲が断熱材で囲まれる。断熱材としては、好適にはガラスエポキシ樹脂が用いられる。このように構成することによって、セラミックヒータであるヒータ91が約2分で200℃に到達したときに断熱材を介してヒータ91に接続される部分(図示せず)の温度が30℃以下であったことを発明者らが確認している。
 捕集治具12とヒータ91とを含んだユニットをここでは捕集ユニット12Aと称する。捕集ユニット12Aは測定部40によって制御される図示しない移動機構に接続され、図中の両側矢印Aに示されるように、すなわち、捕集室5Aから検出室5Bへ、検出室5Bから捕集室5Aへ、壁5Cに設けられた孔5C’を通って移動する。なお、上述のように、ヒータ91は、捕集治具12上に捕集された空気中の浮遊粒子を加熱し得る位置であって、少なくとも加熱時には発光素子6、受光素子9等のセンサ機器から何かによって隔てられる位置に配備されればよいため、捕集ユニット12Aに含まれず、他の位置に備えられてもよい。後述するように加熱動作が捕集室5Aで行なわれる場合、ヒータ91は捕集ユニット12Aに含まれず、捕集室5Aの、捕集ユニット12Aがセットされる位置であって、捕集治具12の、発光素子6、受光素子9等のセンサ機器と反対側に固定されていてもよい。このようにすることよっても加熱時にはヒータ91は捕集治具12によって発光素子6、受光素子9等のセンサ機器から隔てられ、それにより発光素子6、受光素子9等への熱の影響を抑えることができる。この場合、捕集ユニット12Aには少なくとも捕集治具12が含まれていればよい。
 図4Bに示されるように、捕集ユニット12Aの壁5Cから最も遠い側の端部には、上下に突起を有したカバー65Aが備えられる。壁5Cの捕集室5A側の面であって、孔5C’の周囲には、カバー65Aに対応したアダプタ65Bが備えられる。アダプタ65Bには、カバー65Aの上記突起に嵌合する凹部が設けられ、これによりカバー65Aとアダプタ65Bとが完全に接合され、孔5C’を覆うことになる。すなわち、捕集ユニット12Aが図4B中の矢印A’の方向に、孔5C’を通って捕集室5Aから検出室5Bへ移動し、捕集ユニット12Aが完全に検出室5Bに入った時点で、カバー65Aがアダプタ65Bに接合されて孔5C’が完全に覆われ、検出室5B内が遮光される。これにより、検出室5Bで検出動作が行なわれている間には検出室5B内への入射が遮断される。
 信号処理部30は測定部40に接続されて、電流信号を処理した結果を測定部40に対して出力する。測定部40は、信号処理部30からの処理結果に基づいて、測定結果を表示パネル130に表示させるための処理を行なう。
 <検出原理>
 ここで、微生物検出装置における検出原理の一例について説明する。
 特表2008-508527号公報にも開示されているように、空気中に浮遊する生物由来の粒子に紫外光または青色光を照射すると蛍光を発することは、従来から知られている。しかし、空気中には化学繊維の埃など同様に蛍光を発するものが浮遊しており、蛍光を検出するのみでは、生物由来の粒子からのものであるか化学繊維の埃などからのものであるかが区別されない。
 図5~図13は、生物由来の粒子と、化学繊維の埃などとのそれぞれに対して加熱処理を施し、加熱の前後における蛍光の変化を測定する実験を行なって得られた、具体的な測定結果を示す図である。この実験より、埃は加熱処理によって蛍光強度が変化しないのに対して、生物由来の粒子は加熱処理によって蛍光強度が増加することが見出された。
 具体的に、図5は、生物由来の粒子として、大腸菌を200℃にて5分間加熱処理したときの、加熱処理前(曲線71)および加熱処理後(曲線72)の蛍光スペクトルの測定結果である。図5に表わされた測定結果より、加熱処理を施すことによって大腸菌からの蛍光強度が大幅に増加していることが分かった。また、図6Aに示された加熱処理前の蛍光顕微鏡写真と、図6Bに示された加熱処理後の蛍光顕微鏡写真との比較によっても、加熱処理を施すことによって大腸菌からの蛍光強度が大幅に増加していることが明らかとなっている。
 同様に、図7は、生物由来の粒子として、バチルス菌を200℃にて5分間加熱処理したときの加熱処理前(曲線73)および加熱処理後(曲線74)の蛍光スペクトルの測定結果であり、図8Aが加熱処理前、図8Bが加熱処理後の蛍光顕微鏡写真である。また、図9は、生物由来の粒子として、アオカビ菌を200℃にて5分間加熱処理したときの加熱処理前(曲線75)および加熱処理後(曲線76)の蛍光スペクトルの測定結果であり、図10Aが加熱処理前、図10Bが加熱処理後の蛍光顕微鏡写真である。これらに示されるように、他の微生物でも大腸菌と同様に加熱処理によって蛍光強度が大幅に増加することが分かった。
 これに対して、図11Aおよび図11Bは、それぞれ、蛍光を発する埃を200℃にて5分間加熱処理したときの加熱処理前(曲線77)および加熱処理後(曲線78)の蛍光スペクトルの測定結果であり、図12Aが加熱処理前、図12Bが加熱処理後の顕微鏡写真である。図11Aに示された蛍光スペクトルと図11Bに示された蛍光スペクトルとを重ねると図13に示されるように、これらはほぼ重なることが検証された。すなわち、図13の結果や図12A、図12Bの比較に示されるように、埃からの蛍光強度は加熱処理の前後において変化がないことが分かった。
 微生物検出装置100Aにおける検出原理として、発明者らの検証した上述の現象が応用される。すなわち、空気中では、埃と、生物由来の粒子が付着した埃と、生物由来の粒子とが混合されている。上述の現象を基にすると、捕集した粒子に蛍光を発する埃が混ざっている場合、加熱処理前に測定される蛍光スペクトルには、生物由来の粒子からの蛍光と蛍光を発する埃からの蛍光とが含まれ、生物由来の粒子を化学繊維の埃などから区別して検出することができない。しかしながら、加熱処理を施すことで生物由来の粒子だけが蛍光強度が増加し、蛍光を発する埃の蛍光強度は変化しない。そのため、加熱処理前の蛍光強度と所定の加熱処理後の蛍光強度との差を測定することで、生物由来の粒子の量を求めることができる。
 <機能構成>
 以降の説明においては、微生物検出装置100Aが上述した原理を利用して空気中の微生物を検出するものとする。もちろん、空気中の生物由来の粒子を検出するために利用される原理は上述の原理に限定されるものではなく、上の原理は、単に、好適に用いられる一例として挙げているに過ぎない。
 図14は、上記原理を利用して空気中の微生物を検出するための微生物検出装置100Aの機能構成を示すブロック図である。
 図14では、信号処理部30の機能が主に電気回路であるハードウェア構成で実現される例が示されている。しかしながら、これら機能のうちの少なくとも一部は、信号処理部30が図示しないCPU(Central Processing Unit)を備え、該CPUが所定のプログラムを実行することによって実現される、ソフトウェア構成であってもよい。また、測定部40の構成がソフトウェア構成である例が示されている。しかしながら、これら機能のうちの少なくとも一部は、電気回路などのハードウェア構成で実現されてもよい。
 図14を参照して、信号処理部30は、受光素子9に接続される電流-電圧変換回路34と、電流-電圧変換回路34に接続される増幅回路35とを含む。
 測定部40は、制御部41、記憶部42、およびクロック発生部43を含む。さらに、測定部40は、スイッチ110の操作に伴ったスイッチ110からの入力信号を受け付けることで情報の入力を受け付けるための入力部44と、表示パネル130に測定結果等を表示させる処理を実行するための表示部45と、通信部150に接続された外部装置とのデータ等のやり取りに必要な処理を行なうための外部接続部46と、ファン50、ヒータ91、捕集ユニット12Aを往復移動させるための図示しない機構、およびブラシ60を往復移動させるための図示しない機構を駆動させるための駆動部48とを含む。
 捕集室5Aに導入され捕集治具12上に捕集された粒子に対して発光素子6から照射されることで、照射領域15にある当該粒子からの蛍光が、受光素子9に集光される。受光素子9から、受光量に応じた電流信号が信号処理部30に対して出力される。電流信号は、電流-電圧変換回路34に入力される。
 電流-電圧変換回路34は、受光素子9から入力された電流信号より蛍光強度を表わすピーク電流値Hを検出し、電圧値Ehに変換する。電圧値Ehは増幅回路35で予め設定した増幅率に増幅され、測定部40に対して出力される。測定部40の制御部41は信号処理部30から電圧値Ehの入力を受け付けて、順次、記憶部42に記憶させる。
 クロック発生部43はクロック信号を発生させ、制御部41に対して出力する。制御部41は、クロック信号に基づいたタイミングで、ファン50を回転させるための制御信号を駆動部48に対して出力して、ファン50による空気の導入を制御する。また、制御部41は発光素子6および受光素子9と電気的に接続され、それらのON/OFFを制御する。
 制御部41は計算部411を含み、計算部411において、記憶部42に記憶された電圧値Ehを用いて、導入された空気中の生物由来の粒子量を算出するための動作が行なわれる。具体的な動作について、図15の制御部41での制御の流れを示すフローチャートを用いて説明する。ここでは、生物由来の粒子量として、捕集室5A内に所定時間の間に導入された空気中の微生物濃度を算出するものとする。
 <測定動作>
 図15は、微生物検出装置100Aでの測定動作の具体的な流れを表わしたフローチャートである。図15のフローチャートに表わされた動作は、信号処理部30に含まれる図示しないCPUが所定のプログラムを実行して図14に示される各機能を発揮することによって実現される。
 図15を参照して、微生物検出装置100AがONされると、ステップS1で、予め規定されている捕集時間である時間ΔT1の間、捕集室5Aでの捕集動作が行なわれる。ステップS1での具体的な動作としては、制御部41は駆動部48に対して制御信号を出力してファン50を駆動させて捕集室5A内に空気を取り込む。捕集室5A内に導入された空気中の粒子は、放電電極1により負電荷に帯電され、ファン50による空気の流れと放電電極1および捕集治具12表面の皮膜3の間で形成される電界とにより、捕集治具12表面の照射領域15に対応した狭い範囲に捕集される。捕集時間ΔT1が経過すると制御部41は捕集動作を終了、すなわち、ファン50の駆動を終了させる。
 これにより、時間ΔT1の間、外部空気が捕集室5A内に導入孔10を通じて導入され、その空気中の粒子は、捕集治具12表面に時間ΔT1の間、捕集される。
 次に、ステップS3で制御部41は、駆動部48に対して制御信号を出力して捕集ユニット12Aを移動させるための機構を稼動させて、捕集ユニット12Aを捕集室5Aから検出室5Bに移動させる。移動が完了すると、ステップS5で検出動作が行なわれる。ステップS5では微生物検出装置100Aと同様に、制御部41は発光素子6に発光させ、規定の測定時間ΔT2の間、受光素子9により蛍光を受光させる。発光素子6からの光は、捕集治具12の表面の照射領域15に照射され、捕集された粒子から蛍光が発光される。その蛍光強度F1に応じた電圧値が測定部40に入力されて記憶部42に記憶される。これにより、加熱前の蛍光量S1が測定される。
 なお、上記測定時間ΔT2は制御部41に予め設定されているものであってもよいし、スイッチ110などの操作や、ケーブル400を介して通信部150に接続されたPC300からの信号や、通信部150に装着された記録媒体からの信号などによって入力、変更されるものであってもよい。
 このとき、別途設けたLED等の発光素子(図示せず)からの発光の、捕集治具12表面の粒子が捕集されない反射領域(図示せず)からの反射光を、別途設けた受光素子(図示せず)で受光し、その受光量を参照値I0として用いてF1/I0を記憶部42に記憶してもよい。参照値I0に対する比率を算出することで、発光素子や受光素子の温度、湿度等の環境条件や劣化等による特性変動に起因する蛍光強度の変動を補償することができるという利点が生じる。
 ステップS5の測定動作が終了すると、ステップS7で制御部41は、駆動部48に対して制御信号を出力して捕集ユニット12Aを移動させるための機構を稼動させて、捕集ユニット12Aを検出室5Bから捕集室5Aに移動させる。移動が完了すると、ステップS9で加熱動作が行なわれる。ステップS9では微生物検出装置100Aと同様に、制御部41は予め規定した加熱処理時間である時間ΔT3の間、ヒータ91に加熱を行なわせる。このときの加熱温度は予め規定されている。
 加熱動作後、ステップS11で冷却動作が行なわれる。ステップS11では、制御部41は、駆動部48に制御信号を出力して、所定の冷却時間、ファン50を逆回転させる。捕集ユニット12Aに外部の空気を触れさせることで冷却する。加熱処理時間ΔT3、加熱温度、および冷却時間も、制御部41に予め設定されているものであってもよいし、スイッチ110などの操作や、ケーブル400を介して通信部150に接続されたPC300からの信号や、通信部150に装着された記録媒体からの信号などによって入力、変更されるものであってもよい。
 ステップS7で捕集ユニット12Aを捕集室5Aに移動させた後に捕集室5A内で加熱動作および冷却動作が行なわれ、冷却後に捕集ユニット12Aが検出室5Bに移動することで、加熱時にヒータ91は発光素子6、受光素子9等のセンサ機器から隔てられた距離に位置し、また、壁5C等によっても隔てられ、それにより発光素子6、受光素子9等への熱の影響を抑えることができる。なお、このように加熱時にヒータ91は発光素子6、受光素子9等のセンサ機器とは壁5C等によっても隔てられた捕集室5A内にあることから、ヒータ91は捕集ユニット12A内の必ずしも放電電極1から遠い側の面、すなわち検出室5Bに捕集ユニット12Aが移動したときに発光素子6、受光素子9等から遠い側の面になくてもよく、たとえば放電電極1から近い側の面にあってもよい。
 ステップS9の加熱動作およびステップS11の冷却動作が終了すると、ステップS13で制御部41は、駆動部48に対して制御信号を出力して捕集ユニット12Aを移動させるための機構を稼動させて、捕集ユニット12Aを捕集室5Aから検出室5Bに移動させる。移動が完了すると、ステップS15で再度検出動作が行なわれる。ステップS15の検出動作はステップS5での検出動作と同じである。ここでの蛍光強度F2に応じた電圧値が測定部40に入力されて記憶部42に記憶される。これにより、加熱後の蛍光量S2が測定される。
 ステップS15で加熱後の蛍光量S2が測定されると、ステップS17で捕集ユニット12Aのリフレッシュ動作が行なわれる。ステップS17で制御部41は、駆動部48に対して制御信号を出力してブラシ60を移動させるための機構を稼動させて、捕集ユニット12A表面でブラシ60を所定回数往復移動させる。このリフレッシュ動作が完了すると、ステップS19で制御部41は、駆動部48に対して制御信号を出力して捕集ユニット12Aを移動させるための機構を稼動させて、捕集ユニット12Aを検出室5Bから捕集室5Aに移動させる。これにより、開始の指示を受けると直ちに次の捕集動作(S1)を開始することができる。
 計算部411は、記憶された蛍光強度F1と蛍光強度F2との差分を増大量ΔFとして算出する。上述のように、増大量ΔFは生物由来の粒子量(粒子数または粒子濃度等)に関連している。計算部411は、予め、図16に表わされたような、増大量ΔFと生物由来の粒子量(粒子濃度)との対応関係を記憶しておく。そして、計算部411は、算出された増大量ΔFと該対応関係とを用いて得られる生物由来の粒子濃度を、捕集室5A内に時間ΔT1の間に導入された空気中の生物由来の粒子濃度として算出する。
 増大量ΔFと生物由来の粒子濃度との対応関係は、予め実験的に決められる。たとえば、1m3の大きさの容器内に、大腸菌やバチルス菌やカビ菌などの微生物の一種を、ネブライザを利用して噴霧し、微生物濃度をN個/m3に維持して、微生物検出装置100Aを用いて、上述の検出方法により時間ΔT1の間微生物を捕集する。そして、所定加熱量(加熱時間ΔT3、所定の加熱温度)で捕集した微生物に対してヒータ91によって加熱処理を施し、その前後の蛍光強度の増大量ΔFを測定する。種々の微生物濃度について同様の測定がなされることで、図16に示された増大量ΔFと微生物濃度(個/m3)との関係が得られる。
 増大量ΔFと生物由来の粒子濃度との対応関係は、スイッチ110などの操作によって入力されることで計算部411に記憶されてもよい。または、該対応関係を記録した記録媒体が通信部150に装着され、外部接続部46が読み込むことで計算部411に記憶されてもよい。または、PC300によって入力および送信され、通信部150に接続されたケーブル400を介して外部接続部46が受け付けることで、計算部411に記憶されてもよい。または、通信部150が赤外線通信やインターネット通信を行なう場合には、外部接続部46が通信部150でのそれらの通信によって他の装置から受け付けることで、計算部411に記憶されてもよい。また、いったん計算部411に記憶された該対応関係が、測定部40により更新されてもよい。
 計算部411は、増大量ΔFが差分ΔF1と算出された場合、図16の対応関係から増大量ΔF1に対応する値を特定することで、生物由来の粒子濃度N1(個/m3)を算出する。
 ただし、増大量ΔFと微生物濃度との対応関係は、微生物の種類(たとえば菌種)によって異なる可能性がある。そこで、計算部411は、いずれかの微生物を標準の微生物と規定して、増大量ΔFと該微生物の濃度との対応関係を記憶する。これにより、様々な環境における微生物濃度が、標準の微生物を基準として換算された微生物濃度として算出される。その結果、様々な環境を比較することが可能となり、環境管理が容易となる。
 なお、上述の例では増大量ΔFには、所定の加熱量(所定の加熱温度、加熱時間ΔT3)の加熱処理の前後の蛍光強度の差分が用いられているが、これらの比率が用いられてもよい。
 計算部411で算出された捕集された粒子中の生物由来の粒子すなわち微生物の濃度は、制御部41から表示部45に対して出力される。表示部45は、入力された微生物の濃度を、表示パネル130に表示させるための処理を行なう。表示パネル130での表示の一例として、たとえば、図17Aに表わされるセンサ表示が挙げられる。詳しくは、表示パネル130には、濃度ごとのランプが備えられ、図17Bに示されるように、表示部45は、算出された濃度に対応したランプを点灯するランプとして特定し、該ランプを点灯する。他の例として、算出された濃度ごとに、ランプを異なる色に点灯させてもよい。また、表示パネル130はランプ表示に限定されず、数字を表示したり、濃度や対応して予め用意されているメッセージを表示したりしてもよい。また、測定結果は、外部接続部46によって、通信部150に装着された記録媒体に書き込まれてもよいし、通信部150に接続されたケーブル400を介してPC300に送信されてもよい。
 入力部44はスイッチ110からの操作信号に従って、表示パネル130での表示方法の選択を受け付けてもよい。または、測定結果を、表示パネル130に表示するか、外部装置に出力するか、の選択を受け付けてもよい。その内容を示す信号は、制御部41に対して出力され、制御部41から表示部45および/または外部接続部46に対して必要な制御信号が出力される。
 <第1の実施の形態の効果>
 このように、微生物検出装置100Aは、生物由来の粒子からの蛍光と蛍光を発する埃からの蛍光との加熱処理による性質の差を利用し、所定の加熱処理後の増大量に基づいて生物由来の粒子を検出するものである。すなわち、微生物検出装置100Aは、捕集された生物由来の粒子と埃とに加熱処理を施すと微生物は蛍光強度が増加し埃は変化しない、という現象を利用して生物由来の粒子を検出するものである。そのため、導入された空気中に蛍光を発する埃が含まれている場合であっても、リアルタイムに、かつ精度よく、生物由来の粒子を、蛍光を発する埃から分離して検出することができる。
 またこのように、微生物検出装置100Aでは、捕集室5Aと検出室5Bとが区切られ、その間を捕集ユニット12Aが行き来して捕集と検出とが行なわれるため、捕集と検出とを連続して行なうことができる。また、上述のように捕集室5Aで捕集治具12が加熱され、冷却された後に検出室5Bに移動させるため、検出室5B内にあるセンサ等への熱の影響を抑えることができる。
 さらに、微生物検出装置100Aでは捕集室5Aでの捕集工程から検出室5Bでの検出工程に捕集ユニット12Aが移動する際に捕集ユニット12Aに備えられたカバーが壁5Cの孔5C’を遮蔽する。そのため、検出室5B内への外部光の入射が遮断される。これにより、蛍光測定中に浮遊粒子による散乱等での迷光が抑えられ、測定精度を向上させることができる。
 なお微生物検出装置100Aでは壁5Cで区切られた捕集室5A、検出室5Bが備えられているが、それぞれを完全に分離された別の個体である捕集装置、検出装置で構成し、それらの間で捕集ユニット12Aを移動させる構成、またはそれぞれの装置に捕集ユニット12Aをセットする構成であってもよい。この場合、捕集治具12の加熱は発光素子6、受光素子9等のセンサ機器から隔てられた位置として、検出装置以外の箇所で行なわれればよい。たとえば、上述のように捕集室5Aに対応した捕集装置内で行なわれてもよいし、捕集装置および検出装置のいずれでもないその他の位置(たとえば捕集装置から検出装置への移動の途中等)で行なわれてもよい。ヒータ91は捕集ユニット12Aに含まれてもよいし、検出装置以外の箇所である加熱を行なう箇所に設けられてもよい。また、捕集装置と検出装置とをセットとして用いるのみならず、上記捕集室5Aに対応した捕集装置、または上記検出室5Bに対応した検出装置単体で用いてもよい。その場合、用いる方の装置に信号処理部30および測定部40等に対応した機能が含まれる。
 さらに微生物検出装置100Aでは捕集ユニット12Aが1つ設けられ、両側矢印Aで表わされた往復運動を行なうことで捕集室5Aと検出室5Bとの間を往復移動するものとしている。しかしながら、捕集ユニット12Aの他の例として回転可能な円盤の上に2以上設けられ、回転に伴って捕集室5Aと検出室5Bとの間を移動するものとしてもよい。この場合、複数の捕集ユニット12Aのうちの1つを捕集室5Aに位置させ、他の1つを検出室5Bに位置させることで、捕集動作と検出動作とを並行して行なわせることができる。このような構成にすることで、連続的に捕集動作を行なうことが可能となり、それと並行して検出動作を連続して行なうことが可能となる。
 [第2の実施の形態]
 上のような構成とすることで、第1の実施の形態にかかる微生物検出装置100Aでは、連続して検出動作を行なわせることが可能となる。その際、微生物検出装置100Aに設けられたブラシ60で捕集治具12表面をリフレッシュすることで、次の検出動作での検出精度が維持される。
 第2の実施の形態では、微生物検出装置100Aとは異なる方法で捕集治具12表面をリフレッシュすることで、連続して検出動作を行なうことを可能とする構成について説明する。
 <微生物検出装置の装置構成>
 図18は、第2の実施の形態にかかる微生物検出装置100Bの基本構成を示す図である。図18において、微生物検出装置100Aと同じ構成に関しては同じ参照符号で示し、その説明を繰り返さない。
 図18を参照して、第2の実施の形態にかかる微生物検出装置100Bは、一方端に導入孔10、他方端にファン50が配された排出孔11を備えた、内空状の筒状筐体5Dを有し、その内部に、捕集機構として針状の放電電極1および捕集治具12が配備されている。導入孔10にはフィルタ(プレフィルタ)10Bが設けられてもよい。
 放電電極1は高圧電源2の正極に電気的に接続される。捕集治具12と高圧電源2の負極に電気的に接続される。
 ファン50の図示しない駆動機構は測定部40によって制御され、その回転が制御される。ファン50が回転することによって、図中の点線矢印で表わされたように、導入孔10から外部空気が筒状筐体5D内に導入され、排出孔11から筒状筐体5D外に排気される。
 図19の例では、筒状筐体5D内のその高さ方向を遮る位置に略中央に孔を有する絞り板が設けられ、放電電極は、その孔をくぐるように設置されている。これにより、導入孔10から導入された空気の流路は絞り板の孔の径に狭められ、絞り板を通過する際に放電電極1にて電荷される。そして、その狭められた流路のまま捕集治具12に到達するため、捕集治具12上のある程度狭い範囲に浮遊粒子が吸着することになる。
 さらに、検出機構として、光源である発光素子6と受光素子9とが配される。発光素子6は好適には半導体レーザが用いられ、レーザ光を照射する。受光素子9は好適にはフォトダイオードが用いられ、蛍光を受光する。
 なお、図18において発光素子6と受光素子9とは筒状筐体5Dの外部に配されている例が示されている。これは、図の概略化のためにそのように表わされているだけであって、発光素子6と受光素子9とは筒状筐体5Dの内部に配置されていてよい。
 または、図18に表わされた構成のまま、つまり、発光素子6と受光素子9とが筒状筐体5Dの外部に配された構成としてもよい。
 この構成の場合、図18に表わされたように、筒状筐体5Dの外部にある発光素子6から筒状筐体5D内の捕集治具12の表面まで照射光を導入するためのガイド6Aが設けられる。または、筒状筐体5Dの壁面の、少なくとも発光素子6から捕集治具12の表面までの間に存在する部分が照射光の透過率の高い素材で形成されていてもよい。
 同様に、筒状筐体5D内の捕集治具12の表面から筒状筐体5Dの外部にある受光素子9まで蛍光を導出させるためのガイド9Aが設けられる。または、筒状筐体5Dの壁面の、少なくとも捕集治具12の表面から受光素子9までの間に存在する部分が蛍光の透過率の高い素材で形成されていてもよい。
 受光素子9は信号処理部30に接続され、受光量に比例した電流信号を信号処理部30に対して出力する。信号処理部30は測定部40に接続されて、電流信号を処理した結果を測定部40に対して出力する。従って、導入された空気中に浮遊し、捕集治具12表面に捕集された粒子に発光素子6から光が照射されることによって該粒子から発光された蛍光は、受光素子9において受光され、信号処理部30においてその受光量が検出される。測定部40は、信号処理部30からの処理結果に基づいて、測定結果を表示パネル130に表示させるための処理を行なう。
 捕集治具12の放電電極1より遠い側には加熱機構としてのヒータ91が配され、捕集治具12とヒータ91とを含んだユニットが捕集ユニット12Aを構成している。ヒータ91は測定部40に電気的に接続され、測定部40によって加熱量(加熱時間、加熱温度等)が制御される。
 図19Aおよび図19Bは捕集ユニット12Aの構成を表わす概略図であって、図19Aは捕集ユニット12Aの放電電極1側から見た平面図、図19Bは断面図を表わしている。
 図19Aを参照して、捕集治具12は断熱材上に配置されて、両端が捕集板押さえ板電極で放電電極1側から断熱材に向かう方向に押さえられている。
 図19Bを参照して、捕集治具12の放電電極1の反対側には、間に断熱材を挟まずにヒータ91が配され、ヒータ91の周囲が断熱材で覆われている。
 <動作概要>
 微生物検出装置100Bでは、ファン50が回転することで導入された外気が、針状の放電電極1から放出されるイオンによりチャージされ、捕集治具12表面と放電電極1との間の電位差により捕集治具12表面に捕集される。捕集治具12表面に対して発光素子6からレーザ光が照射されることで、捕集治具12表面の微生物を含む捕集物から蛍光が発せられる。
 蛍光は受光素子9で受光されることで、信号処理部30において検出される。検出された蛍光には、生物由来の粒子から発せられた蛍光の他、他の物質から発せられた蛍光が含まれることが考えられる。そこで、一例として、微生物検出装置100Bでも上述した微生物検出装置100Aでの検出原理と同じ検出原理を利用して生物由来の粒子を検出する。すなわち、上述のように、生物由来の粒子から発せられる蛍光は加熱処理することにより強度が増加するのに対し、生物由来でない埃からの蛍光は加熱処理の前後で強度が変化しないという差異を利用して、加熱前後の蛍光強度の差分に基づいて生物由来の粒子を検出する。
 このような検出方法は、捕集治具12表面に静電誘導力を利用して浮遊粒子を吸着させて捕集することを前提としている。そのため、微生物検出装置100Bを用いて連続して検出動作を行なう場合、捕集治具12表面のリフレッシュが不可欠となる。
 第1の実施の形態にかかる微生物検出装置100Aでは、ブラシ60を捕集治具12表面に物理的に接触させることでその表面に付着した粒子を取り除くものであった。第2の実施の形態にかかる微生物検出装置100Bでは、熱処理を利用して捕集治具12表面に付着した粒子を取り除く。
 微生物検出装置100Bは、この動作を行なうための機能として、図14に示された第1の実施の形態にかかる微生物検出装置100Aの機能構成と同様の機能を備える。
 <動作フロー>
 図20は、微生物検出装置100Bでの測定動作の具体的な流れを表わしたフローチャートである。図20のフローチャートに表わされた動作は、概ね、図15に表わされた微生物検出装置100Aでの測定動作と同じである。ただし、微生物検出装置100Bは微生物検出装置100Aと異なって捕集ユニット12Aを移動させる動作が含まれないため、図15の動作のうちのステップS3,S7,S13,S19の捕集ユニット12Aを移動させる動作が不要となる。
 すなわち、図20を参照して、第2の実施の形態にかかる微生物検出装置100Bでは、ステップS1で第1の実施の形態にかかる微生物検出装置100Aと同様の捕集動作が行なわれた後、ステップS5で捕集治具12の表面の蛍光量S1が測定される。
 その後、ステップS9でヒータ91に所定の加熱量で捕集治具12を加熱させる。ここでの加熱量の一例として、たとえば200℃で2分の加熱が挙げられる。
 そして、ステップS11でファン50を駆動させて捕集治具12を冷却した後、ステップS15で加熱後の捕集治具12の表面の蛍光量S2が測定される。
 以上で加熱前後の蛍光量の測定が終了し、第2の実施の形態にかかる微生物検出装置100Bでも、リフレッシュ動作(ステップS17’)が行なわれる。
 ただし、微生物検出装置100Bでのリフレッシュ動作はブラシなどの物理的な接触によるものではなく、捕集治具12を加熱することにより行なわれる。すなわち、ステップS17’では、ヒータ91に所定の加熱量で捕集治具12を加熱させる。ここでの加熱量は上記ステップS9での加熱量よりも大きく、具体的には、生物由来の粒子から発光された蛍光が消失する温度(蛍光発光消失温度)以上の温度である。一例として、たとえば300℃で2分の加熱が挙げられる。場合によっては、400℃や500℃であってもよい。
 <第2の実施の形態の効果>
 微生物検出装置100Bにおいて高温での加熱によるリフレッシュ動作が行なわれることで、先の検出動作で捕集治具12表面に付着した生物由来の粒子からの蛍光発光を消失させることができる。さらに、塵埃等の生物由来の粒子以外の粒子も熱消失させることができる。これにより、次の検出動作を開始するにあたって捕集治具12表面をリフレッシュさせることができる。
 捕集治具12表面からの蛍光は生物由来の粒子からの蛍光および蛍光埃からの蛍光が大部分を占める。捕集治具12表面のリフレッシュがなされないと、先に説明した検出原理を利用する場合、加熱後の蛍光強度の増加分が受光素子9からのセンサ出力として測定部40に入力される。さらに次の検出動作が実行されると、その検出動作での蛍光強度は、先の検出動作における加熱後の蛍光強度に新たに捕集された増加分が加算されることになる。従って、検出動作を繰り返すに連れてセンサ出力の信号のベースが上昇することになる。センサの出力信号の最大値は、センサの駆動電圧で決定される。そのため、信号ベースの上昇は、その後のセンサの測定回数の減少につながる。
 微生物検出装置100Bで上述のようなリフレッシュ動作が行なわれることで捕集治具12表面が高効率にリフレッシュされるため、センサの測定回数の減少を抑えることができる。つまり、連続して検出動作を行なっても高精度で生物由来の粒子が検出されることになる。
 なお、第1の実施の形態にかかる微生物検出装置100Aにも上述のようにブラシ60が設けられ、ブラシ60を捕集治具12上を往復移動させることによる物理的な接触によって捕集治具12の付着物を取り除くことがなされている。そのため、もちろん、微生物検出装置100Aでも、同様に、検出動作を連続して行なうことが可能であり、その回数を増加させることができる。
 しかしながら、物理的な接触を利用して付着物を取り除く場合、接触が完全でない箇所が生じるとその部分の付着物が取り除かれないことになる。そのため、微生物検出装置100Bのように加熱処理を利用したリフレッシュ動作を行なうことで、より効率的に捕集治具12上の付着物を取り除くことができる。
 さらに、図示していないものの、図18に表わされた微生物検出装置100Bの構成に微生物検出装置100Aと同様のブラシをさらに加えて、ブラシによる物理的接触でのリフレッシュ動作と加熱処理でのリフレッシュ動作とを組み合わせることで、より効率的に捕集治具12上の付着物を取り除くことができる。ブラシによる物理的接触でのリフレッシュ動作と加熱処理でのリフレッシュ動作とを組み合わせる場合、これら動作は同時ではなく、異なるタイミングで実行される。好ましくは、先にブラシで捕集治具12上の付着物を取り除き、その後に加熱処理を行なう。このように両方の動作を組み合わせることによって、より完全に捕集治具12上の付着物を取り除くことができる。
 <実験の説明>
 発明者らは、第2の実施の形態にかかる微生物検出装置100Bの効果を確認するため、カビ菌をサンプルとして用いて、加熱量と蛍光強度との変化を測定した。
 図21は、カビ菌を付着させた捕集治具を200℃に加熱した後、さらに、250℃まで加熱した場合、および300℃まで加熱した場合の、捕集治具からの蛍光強度の時間変化の測定結果を示す図である。
 また、図22は、検出動作前の捕集治具表面、検出動作後加熱前の捕集治具表面、200℃に加熱した後の捕集治具表面、および300℃まで加熱した後の捕集治具表面からの蛍光強度の測定結果を表わす図である。
 図21を参照して、200℃で加熱した場合、加熱時間の経過に伴って蛍光強度の減少が顕著には見られず、20分経過しても捕集治具からの蛍光強度が蛍光消失レベルには達さないことが測定された。そのため、200℃での加熱では蛍光消失レベルに達するまでかなりの時間を要することがわかる。
 250℃で加熱した場合、加熱時間の経過に伴って蛍光強度は減少するものの、同様に、20分経過しても捕集治具からの蛍光強度が蛍光消失レベルには達さないことが測定された。そのため、250℃での加熱でも蛍光消失レベルに達するまでかなりの時間を要することがわかる。
 300℃まで加熱した場合、加熱後、ほぼ1分間で蛍光強度は消失レベルに到達したことが測定された。また、図22を参照して、また、300℃の加熱によって、捕集治具からの蛍光強度は、捕集後のそれより低下することが測定された。これは、300℃の加熱によって生物由来の粒子のみならず、蛍光埃等の生物由来の粒子以外の粒子も消失していることを表わしている。
 この実験より、捕集治具表面のリフレッシュ動作としての加熱処理には300℃が適しているとの考察が得られた。
 次に、発明者らは、リフレッシュ動作としての300℃での加熱処理を伴って微生物検出装置100Bで検出動作を繰り返し行なった場合の蛍光強度と、リフレッシュ動作を伴わない場合の蛍光強度とを測定した。図23は、リフレッシュ動作を伴って5回検出動作を繰り返した場合(ハッチングあり)とリフレッシュ動作を伴わずに5回(ハッチングなし)との測定結果を、検出動作ごとに比較して示した図である。
 この実験では、微生物検出装置100Bの捕集治具12として捕集板シリコンウェハー15mmを用い、発光素子6としてレーザダイオードを用いてレーザ光を捕集治具12に対して照射し、受光素子9としてフォトダイオードを用いて捕集治具12から発光する蛍光を検出した。検出動作前の捕集治具12からの蛍光強度を初期値として測定する。
 その後、捕集治具12の電位をグランド電位とし、放電電極1に5KV印加して、電位差を+5kVに設定した。ファン50Aにより、20L/minの流量で外気を微生物検出装置100B内に導入し、15分間、空気中の浮遊粒子を静電捕集した。
 捕集終了後、捕集治具12からの蛍光強度を測定し、ヒータ91により捕集治具12を200℃2分間加熱して冷却した後、再度、蛍光強度を測定した。
 リフレッシュ動作を伴う検出動作の場合、さらにその後、300℃で2分間の加熱を行なった。
 図23を参照して、リフレッシュ動作を伴わない場合、捕集治具12からの蛍光強度は、捕集後に捕集前よりも増加している。そして、200℃加熱で蛍光強度が更に増加している。先述のように、これは、生物由来の粒子からの蛍光強度の変化によるものであるため、この200℃加熱前後の蛍光強度の差分により、導入された空気中の生物由来の粒子の量が検出される。
 リフレッシュ動作を伴わずに次の検出動作を実行すると、次の検出動作では前回の検出動作で得られた蛍光強度がベースとなり、今回の検出動作で捕集された粒子による蛍光強度の増加分が、先の蛍光強度に追加されることになる。すなわち、リフレッシュ動作を伴わずに検出動作を繰り返すと、蛍光強度が累積される、つまりベースが増加するため、受光素子9での測定可能な回数が限定されることになる。
 これに対して、リフレッシュ動作を伴う検出動作の場合、200℃の加熱による蛍光強度の増加を確認した後に、リフレッシュ動作としてさらに300℃で加熱処理を実行する。これにより、捕集された粒子のうちの生物由来の粒子が消失するため、測定された蛍光強度のうちの200℃で2分間加熱することで増加した生物由来の粒子からの蛍光強度の分が減少する。
 図22の実験結果より、先述のように、300℃の加熱によって生物由来の粒子のみならず、蛍光埃等の生物由来の粒子以外の粒子も消失することが分かっている。つまり、この実験での300℃の加熱によって、捕集された塵埃の一部も消失すると考えられる。図23の実験結果でも、300℃の加熱を行なうことで、蛍光強度が200℃での加熱の前、つまり捕集後の蛍光強度よりも小さいことがわかる。
 そのため、リフレッシュ動作を伴って次の検出動作を実行する場合、前回の検出動作で得られた蛍光強度よりも次の検出動作の開始時の蛍光強度よりも小さくなり、蛍光のベースが低下することになる。
 つまり、図23の実験結果より、リフレッシュ動作を伴わずに検出動作を繰り返した場合、ベースの増加である、先の検出動作で測定された蛍光強度から次の検出動作で測定された蛍光強度までの増加分は、1400[mV]となるのに対して、リフレッシュ動作を伴った場合にはベースの増加は300[mV]程度であることがわかった。つまり、リフレッシュ動作としての300℃での加熱処理を伴うことで、ベースの増加は約1/5に抑えられることがわかった。したがって、この実験より、リフレッシュ動作である300℃での加熱処理を行なうことにより、微生物検出装置100Bで可能な検出動作の連続回数を大幅に増加できることがわかった。
 なお、図21での実験結果より、リフレッシュ動作として加熱する温度を、200℃よりもさらに高温とすることでより効果があると考えられる。すなわち、300℃よりも高い450℃や500℃とすることにより、より大きな捕集物も消失することになり、さらにベースの増加が抑えられると考えられる。
 そこで、発明者らは、微生物検出装置100Bを用いてアオカビおよび花粉を含む空気を導入して検出動作を行ない、その捕集治具を450℃~500℃で加熱した場合の、捕集治具上の粒子の消失状況を観察した。アオカビの胞子の大きさは、2~3μmであり、花粉は30μm程度である。
 図24~図26は、加熱処理後の捕集治具表面の顕微鏡写真である。図24はリフレッシュ動作としての加熱前、つまり200℃加熱後の450倍の顕微鏡写真である。図25は、図24の写真中のアオカビ部分を拡大して示した顕微鏡写真であって、2000倍の顕微鏡写真である。図26は、450℃~500℃で加熱した後の、図24の撮影箇所と概ね同一箇所の450倍の顕微鏡写真である。
 図24の写真より、200℃で加熱した後の捕集治具上にはアオカビ胞子および花粉の粒子が観察できる。図26の写真より、450℃~500℃で加熱されることによって、捕集治具上のアオカビおよび花粉のいずれもが消失しているのがわかる。
 すなわち、450℃~500℃での加熱によって、蛍光が消失すると同時に、300℃では消失し切れなかった数μm~数10μm程度の大きな粒子が縮小または消失する。これにより、300℃での加熱を行なうよりも450℃~500℃での加熱の方が蛍光強度がさらに低下することがわかる。従って、この実験より、リフレッシュ動作として450℃~500℃で加熱することで微生物検出装置100Bで可能な検出動作の連続回数をさらに増加できることがわかった。
 [第2の実施の形態の変形例]
 なお、上の例では、図18に示されたように、加熱機構としてのヒータ91を利用してリフレッシュ動作としての加熱処理を行なうものとしている。このようにすることで、リフレッシュ動作用の構成を加熱機構と別個に備える必要がなくなる。
 しかしながら、リフレッシュ動作としての加熱処理は、加熱機構としてのヒータ91で用いられた板状ヒータを用いる方法に限らず、ハロゲンランプ(赤外線ランプ)、レーザ等の、発熱を伴う光源からの照射光による加熱でもよい。
 ただし、発熱を伴う光源からの照射光を用いて捕集治具を加熱する場合、捕集治具12の上方に配置し、上方から捕集治具12表面に向けて照射させる必要がある。つまり、ヒータ91のように捕集治具12の表面から遠い位置に配置されると、その表面を効率的に加熱することができない。
 そこで、第2の実施の形態の変形例として、たとえば光を照射して加熱する場合などの、捕集治具12の上方にリフレッシュ動作としての加熱処理を行なうための熱源を備える構成について説明する。
 図27は、変形例にかかる微生物検出装置100B’の基本構成を示す図である。
 図27を参照して、微生物検出装置100B’には、検出動作として捕集治具12を加熱するためのヒータ91とは別に、リフレッシュ動作として捕集治具12を加熱するためのランプ92が設けられる。ランプ92として、好適にはハロゲンランプが用いられる。ハロゲンランプの他、赤外ランプなどの照射光源が考えられる。また、上方からの熱源はランプに限定されず、ヒータ91と同じような板状ヒータなどが用いられてもよい。
 図27の例では、一例として、ランプ92が筒状筐体5Dとは異なる筒状筐体5E内に設けられる例が示されている。この例において、筒状筐体5Dは捕集治具12より上方の一部が着脱可能に構成され、筒状筐体5Eはその部分と同形状を有している。図27では、筒状筐体5Dの絞り板よりも上方が着脱可能な構成となっており、その部分と筒状筐体5Eとは、回転部材5E’を挟んで接合されている。
 回転部材5E’は測定部40からの制御信号に従って回転可能であって、回転することで、捕集治具12の上方に絞り板を含んだ筒状筐体5Dの上方部分がある状態と、筒状筐体5Eがある状態とに切り替わる。筒状筐体5E内にはランプ92が含まれるため、捕集の後に回転部材5E’が回転することで、捕集治具12上にランプ92が位置するようになる。
 ランプ92の図示しない駆動機構は測定部40に接続されており、測定部40からの制御信号に従った照射量の光を照射する。
 微生物検出装置100B’では、捕集治具12で空気中の浮遊粒子が捕集されてヒータ91で加熱の前後の蛍光強度が測定された後、リフレッシュ動作として筒状筐体5Dの上方を付け替えるための回転部材5E’を回転させる制御がなされた後に、ランプ92から光が照射される。
 なお、図27の例ではランプ92が筒状筐体5D外に設置される例が示されている。このように構成することで、捕集の際にランプ92が気流の妨げとならず、効率的な捕集が可能となる。
 しかしながら、ランプ92の配置は図27のような配置に限定されず、予め筒状筐体5D内に設置されていてもよい。この場合、好ましくは、絞り板に設けられている孔の直下以外の位置に設けられる。このようにすることで、気流の妨げとなることを防止すると共に、図27の例のようなランプ92の付け替え動作を不要とすることができる。
 <第2の実施の形態の変形例にかかる効果>
 このように、変形例にかかる微生物検出装置100B’で捕集治具12の上方に熱源を配して上方から加熱することで、捕集治具12に直接熱が加えられることになるため、下方のヒータ91を用いる場合よりも効率的に加熱することができる。
 <実験の説明>
 発明者らは、第2の実施の形態の変形例にかかる微生物検出装置100B’の効果を確認するための実験を行なった。すなわち、捕集治具12を上方から直接加熱する場合と下方から間接的に加熱する場合のとの効果の相違を確認するため、図28Aおよび図28Bのような簡易な装置を用いて実験を行なった。
 図28Aは捕集治具表面を上方から加熱するための実験装置の概略図であり、図28Bは捕集治具表面を下方から間接的に加熱するための実験装置の概略図である。図28Aの装置を用いた実験を実験A、図28Bの装置を用いた実験を実験Bと称する。
 実験Aにおいては、支柱に対してシリコン基板で捕集治具を水平または略水平に支え、熱源として24V、75Wのハロゲンスポットランプを用いて捕集治具から約10mm離れた位置に設置した。実験Bにおいては、支柱に対してシリコン基板で捕集治具を水平または略水平に支え、そのシリコン基盤の下面に、熱源としてセラミック面状ヒータを設置した。
 図29A~図34Bは、実験Aおよび実験Bでの捕集治具表面の顕微鏡写真であって、図29A,図29Bおよび図30A,図30Bは捕集サンプルとしてカビ菌を用いた場合、図31A~図34Bは捕集サンプルとして花粉を用いた場合の、実験Aおよび実験Bでの捕集治具表面の顕微鏡写真である。
 図29Aは、ハロゲンスポットランプによって、カビ菌を捕集した捕集治具表面を直接200℃で7分加熱した後の捕集治具表面の2000倍の顕微鏡写真、図29Bは、その後、さらに、450℃で3分加熱した後の捕集治具表面の2000倍の顕微鏡写真であって、概ね同じ位置を撮影したものである。
 図30Aは、セラミック面状ヒータによって、カビ菌を捕集した捕集治具を間接的に200℃で7分加熱した後の捕集治具表面の2000倍の顕微鏡写真、図30Bは、その後、さらに、450℃で3分加熱した後の捕集治具表面の2000倍の顕微鏡写真であって、概ね同じ位置を撮影したものである。
 図30Aと図30Bとを比較すると、セラミック面状ヒータによって間接的に加熱した場合には、450℃で3分、追加的に加熱する前後で、カビ菌に変化がないことが分かる。
 これに対して、図29Aと図29Bとを比較すると、ハロゲンスポットランプによって直接、捕集治具表面を加熱した場合には、450℃で3分、追加的に加熱する前後で、数珠状のカビ菌が離脱したり、縮小化したりしていることが分かる。
 図31Aは、ハロゲンスポットランプによって、花粉を捕集した捕集治具表面を直接200℃で7分加熱した後の捕集治具表面の50倍の顕微鏡写真、図31Bは、その後、さらに、450℃で3分加熱した後の捕集治具表面の50倍の顕微鏡写真であって、概ね同じ位置を撮影したものである。図32Aおよび図32Bは、それぞれ、図31Aおよび図31Bの条件での、2000倍の顕微鏡写真である。
 また、図33Aは、セラミック面状ヒータによって、花粉を捕集した捕集治具を間接的に200℃で7分加熱した後の捕集治具表面の50倍の顕微鏡写真、図33Bは、その後、さらに、450℃で3分加熱した後の捕集治具表面の50倍の顕微鏡写真であって、概ね同じ位置を撮影したものである。図34Aおよび図34Bは、それぞれ、図33Aおよび図33Bの条件での、2000倍の顕微鏡写真である。
 図33Aの顕微鏡写真では、その撮影範囲に花粉が32個計測され、図33Bの顕微鏡写真では、その撮影範囲に花粉が30個計測され、セラミック面状ヒータによって間接的に加熱した場合には、450℃で3分、追加的に加熱する前後で、花粉の個数に大きな変化がないことがわかる。
 また、図32Aと図32Bとを比較すると、撮影された花粉のサイズが概ね同じであり、色のみ黒色に変化している。つまり、セラミック面状ヒータによって間接的に加熱した場合には、450℃で3分、追加的に加熱する前後で、花粉が黒化しているのみで、大きさに大きな変化がないことがわかる。
 これに対して、図31Aの顕微鏡写真では、その撮影範囲に花粉が77個計測され、図31Bの顕微鏡写真では、その撮影範囲に花粉が36個計測され、ハロゲンスポットランプによって直接、捕集治具表面を加熱した場合には、450℃で3分、追加的に加熱すると花粉の個数が約半分まで減少したことがわかる。
 また、図32Aと図32Bとを比較すると、ハロゲンスポットランプによって直接、捕集治具表面を加熱した場合には、450℃で3分、追加的に加熱することで撮影された花粉のサイズが約1/3に縮小したことがわかる。
 つまり、同じ加熱条件であっても、セラミック面状ヒータなどを用いて間接的に加熱するよりもハロゲンスポットランプなどを用いてその上方から直接的に加熱する方が効率的に捕集治具表面を加熱できることがわかった。
 今回開示された実施の形態はすべての点で例示であって制限的なものではないと考えられるべきである。本発明の範囲は上記した説明ではなくて請求の範囲によって示され、請求の範囲と均等の意味および範囲内でのすべての変更が含まれることが意図される。

Claims (9)

  1.  生物由来の粒子を検出するための検出装置であって、
     発光素子と、
     蛍光を受光するための受光素子と、
     前記発光素子で照射された捕集用部材からの蛍光の受光量に基づいて、前記捕集用部材で捕集された生物由来の粒子量を算出するための算出装置と、
     前記捕集用部材を清掃するための清掃機構とを備え、
     前記清掃機構は、前記捕集用部材を加熱するための機構を含む、検出装置。
  2.  前記捕集用部材を加熱するためのヒータをさらに備え、
     前記算出装置は、前記ヒータで前記捕集用部材を加熱する前後の前記捕集用部材からの蛍光の受光量の変化に基づいて前記生物由来の粒子量を算出し、
     前記清掃機構に含まれる前記加熱するための機構は、前記ヒータが前記捕集用部材を加熱するための加熱温度よりも高い温度で前記捕集用部材を加熱する、請求項1に記載の検出装置。
  3.  前記清掃機構は前記ヒータを利用して前記捕集用部材を加熱し、
     前記ヒータは、前記捕集用部材に対して前記発光素子および前記受光素子と反対側に配置される、請求項2に記載の検出装置。
  4.  前記清掃機構に含まれる前記加熱するための機構は、前記捕集用部材に対して前記発光素子および前記受光素子と同じ側に配置される、請求項1に記載の検出装置。
  5.  前記加熱するための機構は、発熱を伴う光源である、請求項3に記載の検出装置。
  6.  前記清掃機構に含まれる前記加熱するための機構は、前記捕集用部材に対して前記発光素子および前記受光素子と反対側に配置される、請求項1に記載の検出装置。
  7.  前記清掃機構に含まれる前記加熱するための機構は、200℃以上で前記捕集用部材を加熱する、請求項1に記載の検出装置。
  8.  前記清掃機構は、前記加熱するための機構に加えて、前記捕集用部材に接触可能な部材を含み、前記加熱するための機構と前記接触可能な部材とを異なるタイミングに用いて前記捕集用部材を清掃する、請求項1に記載の検出装置。
  9.  生物由来の粒子を検出する方法であって、
     加熱前の捕集用部材の、発光素子の照射下での蛍光量を測定するステップと、
     加熱後の前記捕集用部材の、前記発光素子の照射下での蛍光量を測定するステップと、
     前記加熱前の捕集用部材から測定された蛍光量と、前記加熱後の捕集用部材から測定された蛍光量との変化量に基づいて、前記捕集用部材で捕集された生物由来の粒子量を算出するステップと、
     前記加熱後の前記捕集用部材を前記加熱の後にさらに加熱することで、前記捕集用部材表面の捕集物を消失させるステップとを備える、検出方法。
