より好ましくは、配列番号1に示すアミノ酸配列において113番目のアミノ酸がバリン又はイソロイシン、114番目のアミノ酸がアルギニン、117番目のアミノ酸がグルタミン酸、118番目のアミノ酸がバリン又はイソロイシン、134番目のアミノ酸がグルタミン又はアルギニン、135番目のアミノ酸がグルタミン酸又はアスパラギン酸、136番目のアミノ酸がトレオニン又はアラニン、145番目のアミノ酸がグリシン、154番目のアミノ酸がトレオニン又はアラニン、162番目のアミノ酸がロイシン、163番目のアミノ酸がイソロイシン、フェニルアラニン又はメチオニン、165番目のアミノ酸がバリン、176番目のアミノ酸がチロシン又はヒスチジン、177番目のアミノ酸がプロリン、179番目のアミノ酸がトリプトファン、181番目のアミノ酸がグルタミン酸、アスパラギン酸又はアスパラギン、185番目のアミノ酸がイソロイシン、バリン又はメチオニン、201番目のアミノ酸がトリプトファン又はフェニルアラニン、215番目のアミノ酸がアラニン又はシステイン、216番目のアミノ酸がセリン又はトレオニン、217番目のアミノ酸がセリン、222番目のアミノ酸がメチオニン、226番目のアミノ酸がトレオニン、227番目のアミノ酸がアルギニン又はリシン、229番目のアミノ酸がロイシン、フェニルアラニン又はイソロイシン、239番目のアミノ酸がグルタミン酸又はリシン、257番目のアミノ酸がリシン又はアルギニン、260番目のアミノ酸がリシン、アルギニン又はヒスチジン、300番目のアミノ酸がプロリン、309番目のアミノ酸がロイシン又はイソロイシン、314番目のアミノ酸がロイシン、メチオニン又はバリン、315番目のアミノ酸がグルタミン酸又はアスパラギン酸、316番目のアミノ酸がチロシン、317番目のアミノ酸がアルギニン又はリシン、318番目のアミノ酸がアルギニン又はリシン、319番目のアミノ酸がグルタミン酸に保存されたアミノ酸配列であって、且つ231番目のアミノ酸がトレオニンからリシンに置換されたアミノ酸配列を有し、チオエステラーゼ活性を有する改変体である。また、配列番号1に示すアミノ酸配列において、上記で挙げた位置以外の領域において1から数個のアミノ酸が欠失、置換、挿入、及び/又は付加されたアミノ酸配列を有するチオエステラーゼ活性が保持された改変体も好ましい。この場合、欠失、置換、挿入、及び/又は付加されるアミノ酸は、1~10個であることが好ましく、1~5個であることがより好ましく、1~2個であること特に好ましい。
第3の態様として、上記(c)の改変体が挙げられる。これは、本発明において特に好ましい態様である。
配列番号1に示す野生型チオエステラーゼのアミノ酸配列においては、84番目から382番目までの領域はチオエステラーゼとして機能するために特に重要で、当該タンパク質がチオエステラーゼ活性を示すために必要十分な領域であることがわかっている(Voelker,T.A.,A.C.Worrell,L.Anderson,J.Bleibaum,C.Fan,D.H.Hawkins,S.E.Radke,and H.M.Davies,“Fatty acid biosynthesis redirected to medium chains in transgenic oilseed plants”,Science,1992,257,p.72-74参照)。配列番号1に示されたアミノ酸配列において、少なくとも84番目から382番目までの領域に相当するアミノ酸配列を有するタンパク質はチオエステラーゼ活性を示しうる。そのため、前記(a)又は(b)のアミノ酸配列において、少なくとも配列番号1に示すアミノ酸配列の84番目から382番目までの領域に相当するアミノ酸配列を有するタンパク質も、本発明のチオエステラーゼ改変体として用いることができる。
以下、本発明で用いる前記(a)~(c)のチオエステラーゼ改変体を纏めてチオエステラーゼ改変体ともいい、BTE(T231K)とも略記する。
しかしながら、野生型チオエステラーゼのアミノ酸配列を改変したチオエステラーゼ改変体が、野生型チオエステラーゼと比べて生体内における脂肪酸及びこれを含む脂質の生産性を向上させることについては何らの報告もなく、これは、今回本発明者らによって新たに得られた知見である。
野生型チオエステラーゼのアミノ酸配列(配列番号1)及びそれをコードする塩基配列(例えば、配列番号2)は、GenBank等のデータベースから得ることができる(例えばGenBankでは、タンパク質配列;AAA34215.1,mRNA配列:M94159.1)。得られた配列をもとに、野生型チオエステラーゼをコードする遺伝子を人工合成により取得することができる。遺伝子の人工合成はインビトロジェン社等のサービスを利用することができる。また、カリフォルニアゲッケイジュから野生型チオエステラーゼをコードする遺伝子をクローニングによって取得することもでき、例えば、Molecular Cloning-A LABORATORY MANUAL THIRD EDITION[Joseph Sambrook,David W. Russell,Cold Spring Harbor Laboratory Press(2001)]記載の方法等により行うことができる。
部位特異的変異の導入方法としては、後述する実施例で用いたSplicing overlap extension(SOE)PCR反応(Horton et al.,Gene 77,61-68,1989)を利用した方法、ODA法(Hashimoto-Gotoh et al.,Gene,152,271-276,1995))、Kunkel法(Kunkel,T. A.,Proc. Natl. Acad. Sci. USA,1985,82,488)等が挙げられる。また、Site-Directed Mutagenesis System Mutan-SuperExpress Kmキット(タカラバイオ社)、Transformer TM Site-Directed Mutagenesisキット(Clonetech社)、KOD-Plus-Mutagenesis Kit(東洋紡社)等の市販のキットを利用することもできる。なかでも、本発明においては、SOE-PCR法によって行うことが好ましい。
94℃で30秒間行い、プライマー対を1本鎖DNAにハイブリダイズさせるアニーリング反応を55℃で約30秒間行い、DNAポリメラーゼを作用させる伸長反応を72℃で約70秒間行い、これらを1サイクルとしたものを30サイクル行うことが挙げられる。
本発明は、宿主に前記(a)~(c)のいずれかのチオエステラーゼ改変体をコードする遺伝子を導入して得られる形質転換体であって、脂肪酸又はこれを含有する脂質の生産能が向上した形質転換体を提供することができる。チオエステラーゼ改変体をコードする遺伝子を有する本発明の形質転換体は、野生型チオエステラーゼ遺伝子を有する形質転換体に比べ、脂肪酸及びこれを含有する脂質の生産能が有意に向上している。なお、本発明において、野生型チオエステラーゼ及びチオエステラーゼ改変体の脂肪酸及び脂肪酸含有脂質の生産能については、実施例で用いた方法により測定することができる。
(d)配列番号2に示す塩基配列において691~693番目のトレオニンをコードする塩基がリシンをコードする塩基に置換されている塩基配列(例えば、配列番号4に示す塩基配列)。
(e)(d)の塩基配列において、691~693番目以外の1から数個の塩基が欠失、置換、挿入、及び/又は付加された塩基配列であって、当該塩基配列からなるDNAによりコードされるタンパク質がチオエステラーゼ活性を有するもの。なお、欠失、置換、挿入及び/又は付加される塩基の位置及び数については、上述した(b)のチオエステラーゼ改変体のアミノ酸配列の保存領域を参照して、チオエステラーゼ活性を保持するよう適宜設計できる。
(f)(d)又は(e)の塩基配列において、配列番号2に示す塩基配列の250番目から1149番目までに相当する塩基配列を少なくとも含んでいる塩基配列
具体的なベクターとしては、微生物を宿主とする場合には、例えば、pBluescript II SK(-)(Stratagene社製)、pUC119(宝酒造社製)、pET系ベクター(タカラバイオ社製)、pGEX系ベクター(GEヘルスケア社製)、pCold系ベクター(タカラバイオ社製)、pHY300PLK(タカラバイオ社製)、pUB110(Mckenzie,T. et al.,(1986),Plasmid 15(2);p.93-103)、pBR322(タカラバイオ社製)、pRS403(ストラタジーン社製)、pMW218/219(ニッポンジーン社製)等を挙げることができる。植物細胞を宿主とする場合には、例えば、pRI系ベクター(タカラバイオ社製)、pBI系ベクター(クロンテック社製)、IN3系ベクター(インプランタイノベーションズ社製)等をあげることができる。特に、大腸菌を宿主とする場合には、pBluescript II SK(-)(Stratagene社製)、pMW218/219(ニッポンジーン社製)が好ましく用いられ、シロイヌナズナを宿主とする場合には、pRI系ベクター(タカラバイオ社製)、pBI系ベクター(クロンテック社製)が好ましく用いられる。
このようなベクターにチオエステラーゼ改変体をコードする遺伝子を、制限酵素処理やライゲーション等の通常の手法によって組み込むことにより形質転換用のベクターを構築することができる。
また、目的遺伝子断片が導入された形質転換体の選択は、選択マーカー等を利用することで行うことができる。例えば、ベクター由来の薬剤耐性遺伝子が、形質転換時に目的DNA断片とともに宿主細胞中に導入された結果、形質転換体が獲得する薬剤耐性を指標に行うことができる。また、ゲノムを鋳型としたPCR法等によって、目的DNA断片の導入を確認することもできる。
本発明の脂肪酸又はこれを含有する脂質の製造方法は、上述したチオエステラーゼ改変体を用いる。具体的には、チオエステラーゼ改変体をコードする遺伝子を含む形質転換体を用いる方法が挙げられる。また、精製したAcyl-ACPとチオエステラーゼを用いてin vitroでAcyl-ACPからの脂肪酸の切り出しを行う方法(Yuan et al.,PNAS,1995,(92),p.10639-10643)等の方法により脂肪酸や脂質を製造することもできる。
本発明の製造方法においては、チオエステラーゼ改変体をコードする遺伝子を有する形質転換体を用いて、該形質転換体内において脂肪酸又はこれを含有する脂質を産生させ、これを採取することが好ましい。具体的には、上述のようにチオエステラーゼ改変体をコードする遺伝子が導入された形質転換体(組換え体)を得た後、当該形質転換体を適切な条件にて培養・生育し、培養物や形質転換体から脂肪酸又はこれを含む脂質を採取することが好ましい。
形質転換体の培養・生育条件は、遺伝子が導入された宿主の種類に応じて選択することができ、適宜好ましい条件を採用することができる。一例として、大腸菌を宿主として用いた形質転換体の場合、LB培地で30~37℃、0.5~1日間培養を行うことが挙げられる。また、シロイヌナズナを宿主として用いた形質転換体の場合、温度条件20~25℃、白色光を連続照射又は明期16時間・暗期8時間等の光条件下で1~2か月間育成を行うことが挙げられる。
また、脂肪酸及び脂質の生産効率の点から、培地中に、例えばチオエステラーゼの基質或いは脂肪酸生合成系に関与する前駆物質としてグリセロール、酢酸、マロン酸等を添加してもよい。
形質転換体において産生された脂肪酸又はこれを含む脂質を単離、回収する方法としては特に限定されず、通常生体内の脂質成分等を単離する際に用いられる方法により行うことができる。例えば、培養物や形質転換体から、ろ過、遠心分離、細胞の破砕、ゲルろ過クロマトグラフィー、イオン交換クロマトグラフィー、クロロホルム/メタノール抽出法、ヘキサン抽出法、エタノール抽出法等により脂肪酸又はこれを含む脂質成分を単離、回収することができる。またより大規模な場合は、培養物や形質転換体より油分を圧搾又は抽出により回収後、脱ガム、脱酸、脱色、脱蝋、脱臭等の一般的な精製を行い、脂質を得ることができる。このように脂肪酸を含む脂質成分を単離した後、単離した脂質を加水分解することで脂肪酸を得ることができる。脂質成分から脂肪酸を単離する方法として具体的には、アルカリ溶液中で70℃程度の高温で処理をする方法、リパーゼ処理をする方法、高圧熱水を用いて分解する方法等が挙げられる。
1.野生型チオエステラーゼ(BTE)遺伝子発現プラスミドの構築
配列番号2に示す塩基配列からなる野生型チオエステラーゼをコードする遺伝子を鋳型として、下記表1に示す配列番号5及び配列番号6のプライマー対を用いたPCR反応により、野生型チオエステラーゼ遺伝子のDNA断片を取得した。なお、配列番号2の塩基配列を有する遺伝子はインビトロジェン(株)の提供する受託合成サービスを利用して取得した。PCR反応により取得した野生型チオエステラーゼ遺伝子断片を制限酵素XbaIで消化した。また、プラスミドベクターpBluescriptII SK(-)(Stratagene社、サンディエゴ,カリフォルニア州)をXbaIで消化し、その後に脱リン酸化処理を行った。これら2つの制限酵素消化物をライゲーション反応により連結することでpBluescriptII SK(-)のXbaIサイトに野生型チオエステラーゼ遺伝子断片(配列番号2に示す塩基配列において、249番目~1149番目の塩基配列)を挿入し、ベクター由来のLacZタンパク質のN末端側の27番目のアミノ酸と融合する形で野生型チオエステラーゼが発現するプラスミドを構築した。得られたプラスミドは、DNAシークエンス法によって、野生型チオエステラーゼをコードする遺伝子が挿入されていることを確認した。
前記1.で構築した野生型チオエステラーゼ発現プラスミドを鋳型として、下記表1に示す配列番号5及び配列番号8のプライマー対、及び配列番号6及び配列番号7のプライマー対を用いたPCR反応により2種類の遺伝子断片を増幅した。これら2種類の断片を鋳型として、再度、配列番号5及び配列番号6のプライマー対を用いてSplicing overlap extension(SOE)PCR反応(Horton et al.,Gene,77,61-68,1989)を行い、配列番号2に示した野生型チオエステラーゼの691~693番目の塩基配列であるACA(Thr)がAAG(Lys)へと置換されたチオエステラーゼ改変体遺伝子のDNA断片を取得した。取得したチオエステラーゼ改変体遺伝子のDNA断片を制限酵素XbaIで消化した。また、プラスミドベクターpBluescriptII SK(-)(Stratagene社)をXbaIで消化し、その後に脱リン酸化処理を行った。これら2つの消化物をライゲーション反応により連結することでpBluescriptII SK(-)のXbaIサイトにチオエステラーゼ改変体遺伝子断片(配列番号4に示す塩基配列において、249番目~1149番目の塩基配列)を挿入し、ベクター由来のLacZタンパク質のN末端側27番目のアミノ酸と融合する形でチオエステラーゼ改変体が発現するプラスミドを構築した。得られたプラスミドは、DNAシークエンス法によって、チオエステラーゼ改変体をコードする遺伝子が挿入されていることを確認した。
前記1.及び2.で構築した野生型チオエステラーゼ又はチオエステラーゼ改変体を発現するプラスミドを用いて、大腸菌突然変異株であるK27株(fadD88)(Overath et al, Eur.J.Biochem.7,559-574,1969)をコンピテントセル形質転換法により形質転換した。形質転換処理をしたK27株を30℃で一晩静置して得られたコロニーをLBAmp液体培地(Bacto Trypton 1%,Yeast Extract 0.5%,NaCl 1%,アンピシリンナトリウム50μg/mL)1mLに接種し、30℃で12時間振とう培養した。12時間後の培養液を別のLBAmp液体培地2.5mLに25μL添加し、30℃で振とう培養し、培養開始から15時間経過した培養液中に含まれている脂質成分を後述の方法にて解析した。また15時間後の培養液の600nmにおける吸光度(OD600)を計測し、培養液に含まれる大腸菌の量を測定した。なお、陰性対照として、プラスミドベクターpBluescriptII SK(-)にて形質転換した大腸菌K27株についても同様に実験を行った。
培養開始後15時間経過した培養液900μLに、酢酸40μL、および内部標準としてメタノールに溶解した7-ペンタデカノン(0.5mg/mL)を40μL添加した。この液に対してクロロホルム0.5mLとメタノール1mLを添加し、十分に攪拌した後に15分間静置した。さらに、1.5%塩化カリウム水溶液0.5mLとクロロホルム0.5mLを添加し、十分に攪拌した後に15分間静置した。室温、1500rpmで5分間遠心分離を行った後、下層部分を採取し、窒素ガスで乾燥した。乾燥したサンプルに3-フッ化ホウ素メタノール錯体溶液を1mL添加し、80℃で10分間恒温することにより脂肪酸のメチルエステル化処理を行った。その後、飽和食塩水1mLとヘキサン1mLを添加し、十分に攪拌した後に30分間静置した。上層部分を採取し、ガスクロマトグラフィー(Hewlett Packard 6890)解析に供した。使用したガスクロマトグラフィーは[キャピラリーカラム:DB-1 MS 30m×200μm×0.25μm(J&W Scientific)、移動層:高純度ヘリウム、カラム内流量:1.0mL/min、昇温プログラム:100℃(1分間)→10℃/min→300℃(5分間)、平衡化時間:1分間、注入口:スプリット注入(スプリット比:100:1),圧力14.49psi,104mL/min,注入量1μL、洗浄バイアル:メタノール・クロロホルム、検出器温度:300℃]の条件で行った。
ガスクロマトグラフィー解析により得られた波形データのピーク面積より、各脂肪酸のメチルエステル量を定量した。なお、各ピーク面積を内部標準である7-ペンタデカノンのピーク面積と比較することで試料間の補正を行い、培養液1リットルあたりの各脂肪酸量を算出した。さらに、算出した各脂肪酸量を、事前に計測した培養液中に含まれる大腸菌量(OD600)で標準化した。結果を図1に示す。
また、上記で算出された各脂肪酸生産量を合計して得られた総脂肪酸生産量(脂質量)を図2に示す。
さらに、図2に示すように、本発明のチオエステラーゼ改変体遺伝子が導入された形質転換体では、野生型チオエステラーゼ遺伝子を導入した形質転換体に比べて、総脂肪酸生産量(脂質量)が約1.6倍と大幅に増加していることがわかった。
1.Napin遺伝子のプロモーター領域及びターミネーター領域のクローニング
茨城県潮来市にて採取した野生のアブラナ様植物からBrassica rapa由来のNapin遺伝子のプロモーター領域を、また、栃木県益子町にて採取した野生のアブラナ様植物からNapin遺伝子のターミネーター領域をそれぞれ下記の手法によって取得した。
上記植物からPowerPlant DNA Isolation Kit(MO BIO Laboratories,USA)を用いてゲノムDNAを抽出し、得られたゲノムDNAをテンプレートとし、DNAポリメラーゼPrimeSTARを用いたPCR反応により各プロモーターとターミネーター領域の増幅を試みた。具体的には表2に示す配列番号12及び配列番号13のプライマー対を用いてBrassica rapa由来のNapin遺伝子プロモーター領域を、配列番号14及び配列番号15のプライマー対を用いてBrassica rapa由来のNapin遺伝子ターミネーター領域をそれぞれ増幅した。増幅が見られたBrassica rapaのNapin遺伝子由来のプロモーター及びターミネーターについて、PCR産物をテンプレートとし、再度PCR反応を行った。この時、Napin遺伝子プロモーターは表2に示す配列番号16及び配列番号17のプライマー対、Napin遺伝子ターミネーターは配列番号14及び配列番号18のプライマー対を用いた。PCRにより増幅したDNA断片はMighty TA-cloning Kit(タカラバイオ社製)を用いて両末端にデオキシアデニン(dA)を付加した後、pMD20-T vector(タカラバイオ社製)にライゲーション反応により挿入して、Napin遺伝子プロモーターを含むプラスミドpPNapin1、及びNapin遺伝子ターミネーターを含むプラスミドpTNapin1をそれぞれ構築した。これらのプラスミドをシーケンス解析に供し、プロモーター領域の塩基配列(配列番号9)及びターミネーター領域の塩基配列(配列番号10)を決定した。
カリフォルニア・ベイ由来Acyl-ACPチオエステラーゼ(BTE)遺伝子の葉緑体移行シグナルペプチドをコードする遺伝子(配列番号11)を、Invitrogen社(Carlsbad,California)の提供する受託合成サービスを利用して取得した。
上記配列を含むプラスミドをテンプレートとし、表2に示す配列番号19及び配列番号20のプライマー対を用いて葉緑体移行シグナルペプチドをコードする遺伝子断片をPCR反応(PrimeSTARを使用)により増幅した。また、野生型チオエステラーゼ(BTE)遺伝子を含むプラスミドをテンプレートとし、表2に示す配列番号21及び配列番号22のプライマー対を用いてPCR反応(PrimeSTARを使用)によりBTE遺伝子断片を増幅した。2つの増幅断片をテンプレートとし、PrimeSTAR、表2に示す配列番号19及び配列番号22のプライマー対を用いたSplicing overlap extension(SOE)PCR反応(Horton et al.,Gene,1989)により2つの遺伝子断片を連結し、葉緑体移行シグナルペプチドを連結した野生型チオエステラーゼ(BTE)遺伝子の全長に相当する遺伝子断片を構築した。
同様の手法により、チオエステラーゼ改変体(BTE(T231K))遺伝子についても、表2に示す配列番号19及び配列番号22のプライマー対を用いて、葉緑体移行シグナルペプチドを連結したチオエステラーゼ改変体(BTE(T231K))遺伝子の全長に相当する遺伝子断片を構築した。
増幅した遺伝子断片はMighty TA-cloning Kit(タカラバイオ社製)を用いて両末端にデオキシアデニン(dA)を付加した後、pMD20-T vector(タカラバイオ社製)にライゲーション反応により挿入し、BTE遺伝子が挿入されたプラスミドpBTEsig1及びBTE(T231K)遺伝子が挿入されたプラスミドpBTE(T231K)sig1をそれぞれ構築した。
植物導入用ベクターとしてpRI909(タカラバイオ社製)を用いた。
pRI909にBrassica rapa由来のNapin遺伝子のプロモーターとNapin遺伝子のターミネーターを導入した。まず、前記1.で作製したプラスミドpPNapin1をテンプレートとし、PrimeSTAR、表2に示す配列番号16及び配列番号23のプライマー対を用いて、PCR反応によりプロモーター領域のDNA断片を増幅した。また、プラスミドpTNapin1をテンプレートとし、PrimeSTAR、表2に示す配列番号18及び配列番号24のプライマー対を用いて、PCR反応によりターミネーター領域のDNA断片を増幅した。増幅断片は、Mighty TA-cloning Kit(タカラバイオ社製)を用いて両末端にデオキシアデニン(dA)を付加した後、pMD20-T vector(タカラバイオ社製)にライゲーション反応により挿入し、プラスミドpPNapin2およびpTNapin2をそれぞれ構築した。プラスミドpPNapin2をSalIとNotIで、プラスミドpTNapin2をSmaIとNotIでそれぞれ処理し、SalIとSmaIで処理したpRI909にライゲーション反応により挿入し、プラスミドp909PTnapinを構築した。
続いて、BTE遺伝子又はBTE(T231K)遺伝子を、Napin遺伝子プロモーターの下流に導入した。まず、前記2.で作製したプラスミドpBTEsig1又はpBTE(T231K)sig1をテンプレートとし、PrimeSTAR、表2に示す配列番号25及び配列番号26のプライマー対を用いて遺伝子断片をそれぞれ増幅した。増幅した遺伝子断片は、Mighty TA-cloning Kit(タカラバイオ社製)を用いて両末端にデオキシアデニン(dA)を付加した後、pMD20-T vector(タカラバイオ社製)にライゲーション反応により挿入し、プラスミドpBTEsig2およびpBTE(T231K)sig2を構築した。プラスミドpBTEsig2とp909PTnapinをそれぞれNotIで処理し、得られた断片をライゲーション反応で連結することで、Napin遺伝子プロモーターとNapin遺伝子ターミネーターの間にBTE遺伝子が挿入された植物導入用ベクターp909BTEを構築した。同様の手法で、BTE(T231K)遺伝子が挿入された植物導入用ベクターp909BTE(T231K)を構築した。
構築した植物導入用ベクターp909BTE及びp909BTE(T231K)を、インプランタイノベーションズ(株)によるシロイヌナズナ形質転換受託サービスに供し、BTE遺伝子又はBTE(T231K)遺伝子が導入されたシロイヌナズナ(Arabidopsis thaliana(Colombia株))の形質転換体Pnapin-BTE及びPnapin-BTE(T231K)をそれぞれ得た。
シロイヌナズナの野生株及び形質転換体Pnapin-BTE及びPnapin-BTE(T231K)を、室温22℃で、蛍光灯照明を用いて明期16時間(約4000ルクス)、暗期8時間の条件で育成した。約2か月の栽培の後、種子を収穫した。
(1)種子からの脂質抽出と脂肪酸のメチルエステル化
収穫したシロイヌナズナ種子をシードスプーン(200粒用、バイオメディカルサイエンス製)で2杯程度すくい、Lysing Matrix D(MP Biomedicals,USA)に入れた。Lysing Matrix DをFastPrep(MP Biomedicals)にセットし、速さ6.0で20秒間振動させて種子を粉砕し、そこにメタノールに溶解した20μLの7-ペンタデカノン(0.5mg/mL)(内部標準)と20μLの酢酸を添加した。CHCl3 0.25mL,メタノール0.5mLを加え、十分に攪拌した後に15分間静置した。さらに、1.5%塩化カリウム水溶液0.25mLとCHCl3 0.25mLとを添加し、十分に攪拌した後に15分間静置した。室温にて1500rpmで5分間遠心分離を行った後、下層部分を採取し、窒素ガスで乾燥した。乾燥したサンプルにメタノールに溶解した0.5N KOH 100μL加え、70℃で30分間恒温することによりトリアシルグリセロールを加水分解した。3-フッ化ホウ素メタノール錯体溶液を0.3mL添加して乾燥物を溶解し、80℃で10分間恒温することにより脂肪酸のメチルエステル化処理を行った。その後、飽和食塩水0.2mLとヘキサン0.3mLを添加し、十分に攪拌した後に30分間静置した。脂肪酸のメチルエステルが含まれるヘキサン層(上層部分)を採取し、ガスクロマトグラフィ(GC)分析に供した。なお種指数計測のためにLysing Matrix Dに入れる直前の種子を薬包紙上に広げデジタルカメラで撮影した。その画像をもとにして脂質解析に供した種子数を計測し、後述の種子100粒あたりの脂質量の補正に用いた。
上記でメチルエステル化した試料を、GCにて分析した。使用したGCはカラム:DB1-MS(J&W Scientific,Folsom,California)、分析装置:6890(Agilent technology,Santa Clara,California)を用いて、[カラムオーブン温度:100℃保持1分→100~300℃(10℃/分昇温)→300℃保持5分(ポストラン2分)、注入口検出器温度:300℃、注入法:スプリットモード(スプリット比=193:1)、サンプル注入量1-2μL、カラム流速:0.5mL/minコンスタント、検出器:FID、キャリアガス:水素、メイクアップガス:ヘリウム]の条件で行った。GC解析により得られた波形データのピーク面積より、各脂肪酸のメチルエステル量を定量した。なお、各ピーク面積を内部標準である7-ペンタデカノンのピーク面積と比較することで試料間の補正を行い、解析に供した全種子中に含まれる脂肪酸量を算出した。また、算出した脂肪酸量を事前に計測した種子数で除することにより、種子100粒あたりの脂肪酸量を算出した。なお、種子中の各脂質に対応するGCのピークは、各脂肪酸の標準品のメチルエステルの保持時間(Retention Time,RT)、及び下記のGC/MSによる解析により同定した。
GC分析に供した試料について、必要に応じてGC/マススペクトル(MS)分析を、キャピラリーカラム:DB1-MS、GC分析装置:7890A(Agilent technology)、MS分析装置:5975C(Agilent technology)を用いて以下の条件にて行った。[カラムオーブン温度:100℃保持2分→100~300℃(10℃/分昇温)→300℃保持5分(平衡時間2分、ポストラン320℃で5分)もしくは100℃保持2分→100~200℃(10℃/分昇温)→200~320℃(50℃/分昇温)→320℃保持5分(平衡時間2分、ポストラン320℃で5分)、注入口検出器温度:250℃、注入法:スプリットレスモード、サンプル注入量1μL、カラム流速:1mL/minコンスタント、検出器:FID、キャリアガス:水素、メイクアップガス:ヘリウム、溶媒待ち時間:7分もしくは3.5分、イオン化法:EI法、イオン源温度:250℃、インターフェース温度:300℃、測定モード:スキャンモード(m/z:20~550若しくは10~550)]
1.チオエステラーゼ改変体(BTE(MRR197RRH))遺伝子発現プラスミドの構築
野生型チオエステラーゼ遺伝子の197~199位のアミノ酸であるメチオニン-アルギニン-アルギニン(MRR)が、アルギニン-アルギニン-ヒスチジン(RRH)へと置換された2重改変体であるチオエステラーゼ改変体BTE(MRR197RRH)を作製した(配列番号27及び28)。なお、改変体BTE(MRR197RRH)は231番目のアミノ酸がトレオニンであり、本発明で用いるチオエステラーゼ改変体には含まれないものである。
実施例1にて構築した野生型チオエステラーゼ発現プラスミドを鋳型として、実施例1の表1に示す配列番号5及び下記表3に示す配列番号29のプライマー対、並びに実施例1の表1に示す配列番号6及び下記表3に示す配列番号30のプライマー対を用いたPCR反応により2つの遺伝子断片に分けて増幅した。これら2つの断片を鋳型として、再度オリゴヌクレオチドプライマー対(表1に示す配列番号5及び6のプライマー対)を用いてSplicing overlap extension(SOE)PCR反応(Horton et al.,Gene,77,61-68,1989)を行い、配列番号28に示した野生型チオエステラーゼの589~591番目の塩基配列であるATG(Met)がCGG(Arg)へ、また595~597番目の塩基配列であるCGT(Arg)がCAT(His)へと置換されたチオエステラーゼ改変体BTE(MRR197RRH)の遺伝子断片を取得した。取得したDNA断片を制限酵素XbaIで消化した。また、プラスミドベクターpBluescriptII SK(-)(Stratagene社)をXbaIで消化し、その後に脱リン酸化処理を行った。これら2つの消化物をライゲーション反応により連結することでpBluescriptII SK(-)のXbaIサイトにチオエステラーゼ改変体遺伝子断片を挿入し、ベクター由来のLacZタンパク質のN末端側27番目のアミノ酸と融合する形でチオエステラーゼ改変体BTE(MRR197RRH)が発現するプラスミドを構築した。
前記1.で構築したチオエステラーゼ改変体BTE(MRR197RRH)を発現するプラスミドを用いて、大腸菌突然変異株であるK27株(fadD88)(Overath et al, Eur.J.Biochem.7,559-574,1969)を形質転換した。形質転換処理をしたK27株を30℃で一晩静置して得られたコロニーをLBAmp液体培地(Bacto Trypton 1%,Yeast Extract 0.5%,NaCl 1%,アンピシリンナトリウム50μg/mL)1mLに接種し、30℃で12時間振とう培養した。12時間後の培養液を新鮮なLBAmp液体培地2.5mLに25μL添加し、30℃で振とう培養し、培養開始から15時間経過した培養液中に含まれている脂肪酸成分及び脂肪酸量を前記実施例1の4.及び5.と同様の方法にて解析した。また15時間後の培養液の600nmにおける吸光度(OD600)を計測し、培養液に含まれる大腸菌の量を測定した。なお、コントロール及び比較のために、プラスミドベクターpBluescriptII SK(-)にて形質転換した大腸菌K27株、野生型チオエステラーゼ(BTE)遺伝子又は本発明のチオエステラーゼ改変体(BTE(T231K))遺伝子を有する形質転換体についても同様に実験を行った。
培養液中のC12:0からC18:1までの各脂肪酸生産量を合計して得られた総脂肪酸量を図4に示す。
Claims (10)
- 下記(a)~(c)のいずれかのチオエステラーゼ改変体を用いる脂肪酸又はこれを含む脂質の製造方法。
(a)配列番号1に示すアミノ酸配列において231番目のアミノ酸がトレオニンからリシンに置換されたアミノ酸配列を有するチオエステラーゼ改変体
(b)(a)のアミノ酸配列において、231番目以外の1から数個のアミノ酸が欠失、置換、挿入、及び/又は付加されたアミノ酸配列を有し、かつチオエステラーゼ活性を有するチオエステラーゼ改変体
(c)(a)又は(b)のアミノ酸配列において、配列番号1に示すアミノ酸配列の84番目から382番目までに相当するアミノ酸配列を少なくとも有し、かつチオエステラーゼ活性を有するチオエステラーゼ改変体 - 宿主に前記(a)~(c)のいずれかのチオエステラーゼ改変体をコードする遺伝子を導入して、脂肪酸又はこれを含有する脂質の生産能が向上した形質転換体を得る工程と、得られた形質転換体から脂肪酸含有脂質を採取する工程とを含む、請求項1記載の脂肪酸又はこれを含む脂質の製造方法。
- 前記脂肪酸が炭素数12以上の長鎖脂肪酸であることを特徴とする請求項1又は2記載の脂肪酸又はこれを含む脂質の製造方法。
- 前記脂肪酸がラウリン酸であることを特徴とする請求項1~3のいずれか1項に記載の脂肪酸又はこれを含む脂質の製造方法。
- 前記宿主が植物又は微生物であることを特徴とする請求項2~4のいずれか1項に記載の脂肪酸又はこれを含む脂質の製造方法。
- 前記宿主がシロイヌナズナ又は大腸菌であることを特徴とする請求項2~5のいずれか1項に記載の脂肪酸又はこれを含む脂質の製造方法。
- 宿主に下記(a)~(c)のいずれかのチオエステラーゼ改変体をコードする遺伝子を導入して、脂肪酸又はこれを含む脂質の生産能が向上した形質転換体を得る工程を含む、脂肪酸含有脂質の生産性を向上させる方法。
(a)配列番号1に示すアミノ酸配列において231番目のアミノ酸がトレオニンからリシンに置換されたアミノ酸配列を有するチオエステラーゼ改変体
(b)(a)のアミノ酸配列において、231番目以外の1から数個のアミノ酸が欠失、置換、挿入、及び/又は付加されたアミノ酸配列を有し、かつチオエステラーゼ活性を有するチオエステラーゼ改変体
(c)(a)又は(b)のアミノ酸配列において、配列番号1に示すアミノ酸配列の84番目から382番目までに相当するアミノ酸配列を少なくとも有し、かつチオエステラーゼ活性を有するチオエステラーゼ改変体 - 宿主に下記(a)~(c)のいずれかのチオエステラーゼ改変体をコードする遺伝子を導入して得られる脂肪酸又はこれを含む脂質の生産能が向上した形質転換体。
(a)配列番号1に示すアミノ酸配列において231番目のアミノ酸がトレオニンからリシンに置換されたアミノ酸配列を有するチオエステラーゼ改変体
(b)(a)のアミノ酸配列において、231番目以外の1から数個のアミノ酸が欠失、置換、挿入、及び/又は付加されたアミノ酸配列を有し、かつチオエステラーゼ活性を有するチオエステラーゼ改変体
(c)(a)又は(b)のアミノ酸配列において、配列番号1に示すアミノ酸配列の84番目から382番目までに相当するアミノ酸配列を少なくとも有し、かつチオエステラーゼ活性を有するチオエステラーゼ改変体 - 宿主が植物又は微生物であることを特徴とする請求項8記載の形質転換体。
- 宿主がシロイヌナズナ又は大腸菌であることを特徴とする請求項8又は9記載の形質転換体。
