Abstract
Description
マイコプラズマ・ニューモニエ感染症は、市中非定型肺炎に分類され、市中肺炎に占める割合は成人では30~40%、15~25歳の若年成人に限ると60~70%に及ぶとされている。マイコプラズマ・ニューモニエの感染経路は経気道感染であり、学校などの施設や家族内での感染が拡大することも少なくない。またマイコプラズマ・ニューモニエ感染症において、肺炎は感染症の約3~5%に起こり、残りは気管支炎、上気道炎、不顕性感染となる。特徴的な症状として感染初期から喀痰を伴わない頑固な咳嗽で、発熱、頭痛、咽頭痛、悪寒、全身倦怠などの症状を伴う場合もある。
マイコプラズマ・ニューモニエ感染症の検査として、患者咽頭拭い液からの培養検査、患者血清を用いた抗体検査が一般的である。培養検査はマイコプラズマ・ニューモニエ自体が特殊な培地でしか発育しないこと、手技が困難なことや、最終的なマイコプラズマ・ニューモニエの同定にはPCR検査を実施する必要性があることから、限られた施設でしか検査できないのが現状であり、また培養に数週間が必要となるため、迅速に結果が得られる検査が求められている。
例えば、特許文献1は、約43キロダルトン(KDa)のマイコプラズマ・ニューモニエの膜抗原蛋白質に対するモノクローナル抗体を用いた免疫検出法が記載されている。また、特許文献2は、リボソーム蛋白質L7/L12に対する抗体を用い、マイコプラズマ・ニューモニエの検出を精度良く行うことができることが記載されている。また、特許文献3は、マイコプラズマ・ニューモニエのP1蛋白質に対するモノクローナル抗体で、マイコプラズマ属の他の種又は共存する菌叢の他の病原種とは1%もしくはそれ未満の交差反応性しか示さないモノクローナル抗体によって、マイコプラズマ・ニューモニエ感染の迅速かつ特異的な診断が可能となることが記載されている。
非淋菌性尿道炎の主要な起炎菌としてはクラミジア・トラコマチス(Chlamydia trachomatis)が知られている。しかし、非淋菌性尿道炎患者においてクラミジア・トラコマチスが検出されるのは30~40%程度であり、多くはその症状が何に由来するか明らかでない。クラミジア・トラコマチス以外については、マイコプラズマ属及びウレアプラズマ属の微生物が注目されており、特にマイコプラズマ・ジェニタリウムは、非淋菌性尿道炎及び性行為感染症の起炎菌の1つとして示されている。
マイコプラズマ・ジェニタリウム感染症は、論文では培養法やPCR法による報告例があるが、これらの方法は迅速な診断を行うことができないため、迅速かつ特異的に臨床検体中のマイコプラズマ・ジェニタリウムに属する微生物を検出する方法が求められている。
一方、マイコプラズマ・ニューモニエとマイコプラズマ・ジェニタリウムは、血清学的には非常に近いことが知られているが、上述したように、マイコプラズマ・ニューモニエとマイコプラズマ・ジェニタリウムの感染する場所(組織や器官など)が異なることから、両微生物を特異的に検出することが可能な分子を同定できれば、該分子を指標として、両感染症を診断することが可能となると考えた。
本発明は上記問題に鑑みてなされたものであり、マイコプラズマ・ニューモニエ及び/又はマイコプラズマ・ジェニタリウム感染症を迅速かつ特異的に診断するための分子を特定し、該分子を指標とした検出方法、検出キットを提供することを目的とした。
本発明はこの知見に基づいて成し遂げられたものである。

また、実施例8に示したようにマイコプラズマ・ニューモニエのDnaK遺伝子は、マイコプラズマ・ニューモニエのP1遺伝子の型が異なる株間においても100%一致し、また、異なる分離地および、過去50年間においても変異が認められなかった。そこのことから、マイコプラズマ・ニューモニエのDnaK遺伝子及びDnaK蛋白質の配列は安定していると考えられる。よって、前記NCBIで公開されている遺伝子配列あるいは蛋白質配列を参照することが好ましい。
本発明のマイコプラズマ・ニューモニエ又はマイコプラズマ・ジェニタリウムを検出する方法の第一の態様としては、該微生物に特異的な抗DnaK抗体を用いることを特徴としている。なお、特異的な抗体を選択する際には、該微生物への感度が少なくとも105CFU/mL以上、好ましくは104CFU/mL以上、更に好ましくは103CFU/mL以上あり、対象外の微生物への感度は少なくとも107CFU/mL以下、好ましくは108CFU/mL以下である。
本発明で用いることのできる抗体は、ポリクローナル抗体又はモノクローナル抗体のいずれも使用することができ、以下の方法あるいはその他の類似の方法によって取得することができるが、これらの方法に限定されるものではない。
また、既知の該遺伝子の両端部位におけるDNA配列をプライマーとしたPCR法による遺伝子増幅、相同部分配列を鋳型プローブとしたハイブリダイゼーション法など通常の遺伝子操作手法を用いることにより該遺伝子の全長配列を取得することができる。
その後、他の蛋白質遺伝子とのフュージョン遺伝子などを構築し、大腸菌等を宿主として公知の遺伝子導入手法により宿主内に該当フュージョン遺伝子を挿入し、大量に発現させた後にフュージョン蛋白質として用いた蛋白質に対する抗体アフィニティーカラム法などにより発現蛋白質を精製することにより目的とする蛋白質抗原を取得することができる。この場合、該DnaK蛋白質の全長蛋白質が抗原となるため対象外の微生物間で保存されているアミノ酸部分に対する抗体を取得しても本発明の目的に合致しない。従って、本法によって取得した抗原に対しては公知の手法によりモノクローナル抗体を産生するハイブリドーマを取得し、該当する微生物とのみ反応する抗体を産生するクローンを選択することにより目的の抗体を取得することができる。
目的とする蛋白質抗原の取得は、該蛋白質遺伝子と他の蛋白質遺伝子とのフュージョン遺伝子などを構築し、大腸菌等を宿主として公知の遺伝子導入手法により宿主内に該当フュージョン遺伝子を挿入し大量に発現させる。その後に、フュージョン蛋白質として用いた蛋白質に対する抗体アフィニティーカラム法などにより、発現蛋白質を取得することができる。この場合、該DnaK蛋白質の全長蛋白質が抗原となるため対象外の微生物間で保存されているアミノ酸部分に対する抗体を取得しても本発明の目的に合致しない。従って、本法によって取得した抗原に対しては公知の手法によりモノクローナル抗体を産生するハイブリドーマを取得し、該当する微生物とのみ反応する抗体を産生するクローンを選択することにより目的の抗体を取得することができる。
また、同様にして、マイコプラズマ・ジェニタリウムを免疫原として作製することもできる。上記微生物も免疫原とする場合、免疫原の調製は公知の方法によって行うことができる。公知の方法として、超音波処理、熱処理、界面活性剤処理、ホルマリン処理、凍結融解処理、塩酸処理があげられる。
本発明の抗体を少なくとも1種含んでいれば良く、使用する免疫測定法に合わせて、使用する抗体の数や種類や組み合わせは適宜変更することができる。更に、試料の前処理として、上記の抽出法による前処理液を含んでも良い。
DNAの抽出方法としては、公知の方法を適用することができる。例えば、試料を界面活性剤による可溶化や、除タンパク剤による除タンパク等の操作を行って、DNAを取得する方法等が挙げられる。好ましくは、後述する該DnaK遺伝子の解析が可能なものであれば、例えば、引き続いてPCR法による遺伝子増幅を行う場合には、PCR反応の阻害物質を含まない状態に調製しておくことが好ましい。
試料の前処理方法としては、上述した抗体を用いた該微生物の検出方法と同様な方法を使用することもできる。
一方、マイコプラズマ・ジェニタリウムDnaK遺伝子を増幅する際には、NCBIで公開されているマイコプラズマ・ジェニタリウムのDnaK遺伝子配列(NCBI番号:L43967 REGION: 374919..376706)を元にPCR増幅プライマーを設計することができる。
また、マイコプラズマ・ニューモニエのDnaK蛋白質の配列も安定していると考えられることから、これを用いて作製した抗体は、遺伝子型、地域、年代によって反応性に差はないと考えられ、広範な地域、年代において使用することができる。
また、別の態様としては、目的の該DnaK遺伝子に特異的な塩基配列を特定するための1種類のプローブを少なくとも含むことを特徴とする。
更に、試料の前処理として、上記の抽出法による前処理液を含んでも良い。
(1)マイコプラズマ・ニューモニエ及びマイコプラズマ・ジェニタリウムに特異的なモノクローナル抗体の作製
(1-1)免疫菌株の培養及び免疫抗原の調製
マイコプラズマ・ニューモニエ6株(FH・Bru・Mac・M52・PI1428・M129-B7株:ATCCより購入)を、株毎にそれぞれ、PPLOグルコースブロス(ウマ血清、新鮮酵母エキス、酢酸タリウム含有)に接種し、37℃で7日間、好気条件下で培養した。遠心回収した各菌体を洗浄後、PBSに懸濁して凍結融解処理したものを免疫抗原とした。
免疫感作には、雌性Balb/cマウス 6週齢(日本クレア社)を使用した。各菌体に由来する抗原溶液を、それぞれ、フロイントのコンプリートアジュバント(SIGMA社)で乳化し、抗原100μgをマウスに皮下注射した。この後、免疫原に対する抗体価の上昇が認められるまで、2週間おきにフロイントのインコンプリートアジュバント(SIGMA社)で乳化した抗原50μgを皮下注射した。更に、細胞融合3日前にPBSで希釈した抗原25μgを腹腔内に注射した。
定法に従って、以下の操作を行った。免疫感作マウスより無菌的に取り出した脾臓細胞と骨髄腫細胞(P3U1)をポリエチレングリコール1500(Roche社)を用いて融合し、96穴プレートに播種した。HAT培地を用いてハイブリドーマ細胞を選択培養し、これらの培養上清に対して以下のELISA条件でスクリーニングを実施した。ELISAの固相化には、免疫原とした6種類の各株に由来する、各マイコプラズマ・ニューモニエ抗原1μg/mLを使用した。ブロッキング処理後、培養上清を添加し、4℃で一晩反応させた。洗浄液で3回洗浄後、2000倍希釈HRP標識ウサギ抗マウスIg抗体(Dako社)を添加し、室温で1時間反応させた。洗浄液で3回洗浄後、基質(TMBZ)溶液を添加し、室温で10分間反応させた。反応停止後、450nmの吸光度を測定した。これにより選出されたハイブリドーマについて限界希釈法によるクローニングを実施し、クローン株を樹立した。その内16株より産生されるモノクローナル抗体について、以下の検討を行った。得られたクローン株は、6種の各株に由来する免疫抗原のいずれとも反応するものである。
ウェスタンブロットにより、上記16種のモノクローナル抗体の認識蛋白質の分子量を確認した。マイコプラズマ・ニューモニエ抗原(FH株)10μgを、SDS-PAGEし、続けてニトロセルロースメンブレンにブロットした。各クローンの培養上清をメンブレンに添加し、室温で1時間反応させた。洗浄液で3回洗浄後、1000倍希釈HRP標識ウサギ抗マウスIg抗体を添加し、室温で1時間反応させた。洗浄液で3回洗浄後、基質(4-クロロ-1-ナフトール)溶液を添加し、室温で10分間反応させた。発色後、蒸留水で洗浄し、反応を停止した。
その結果、10種が62~69KDa、6種が40~45KDaの分子を認識していることが判明した。そのことから、取得数が多く抗原性が高いと考えられる62~69KDaの分子について、続いて抗原の同定を試みた。
Iso Strip(Roche社)を用いて62~69KDaの分子を認識するモノクローナル抗体10種のサブクラスを確認したところ、6種がH鎖G1/L鎖κ、1種がH鎖G1/L鎖λ、1種がH鎖2b/L鎖κ、1種がH鎖2b/L鎖λ、1種がH鎖2a/L鎖λであることが判った。
(2-1)モノクローナル抗体認識抗原の精製
(2-1-1)菌体培養
抗原精製には、これまでに全遺伝子配列が決定されているマイコプラズマ・ニューモニエM129-B7株菌体を用いた。マイコプラズマ・ニューモニエ(M129-B7株)を、PPLOグルコースブロス(ウマ血清、新鮮酵母エキス、酢酸タリウム含有)に接種し、37℃で7日間、好気条件下で培養した。遠心回収した菌体を洗浄後、PBSに懸濁して凍結処理した。
CNBr-activated Sepharose 4B(GEヘルスケア社)に、(1)で得られたモノクローナル抗体MCM12をカラム担体に結合させ、抗原精製用アフィニティーカラムを作製した。カラム担体への結合は、0.1 mol/L NaHCO3-NaOH、0.5 mol/L NaCl(pH8.3)下でIgG 5mg/mL gelを4℃、一晩反応させた。未反応基のブロックには、0.2 mol/L グリシン緩衝液(pH8)を用いた。
マイコプラズマ・ニューモニエ菌体より抽出した蛋白質をカラムにアプライし、非吸着画分を溶出した後、3 mol/Lチオシアン酸ナトリウムによりカラム吸着画分を溶出・回収した。これを50mmol/L PBS(pH7)で透析したものを精製品とした。
(2-2-1)認識蛋白質のSDS-PAGE上での分子量確認
精製抗原を、SDS-PAGE及びウェスタンブロットにより解析した。精製抗原0.1μgをSDS-PAGEし、続けてニトロセルロースメンブレンにブロットした。モノクローナル抗体MCM12又はモノクローナル抗体MCM19の 10μg/mL IgG溶液を、それぞれ、メンブレンに添加し、室温で1時間反応させた。洗浄液で3回洗浄後、1000倍希釈HRP標識ウサギ抗マウスIg抗体を添加し、室温で1時間反応させた。洗浄液で3回洗浄後、基質(4-クロロ-1-ナフトール)溶液を添加し、室温で反応させた。発色後、蒸留水で洗浄し、反応を停止した。
いずれの抗体も精製抗原を認識することが確認できた。
定法に従って、精製抗原蛋白のN末端10アミノ酸残基を解析した。精製抗原をSDS-PAGEし、PVDFメンブレンにブロットした試料を50%メタノール/0.1%トリフルオロ酢酸、メタノールで洗浄後、乾燥し、N末端より10サイクルのアミノ酸配列分析を実施した。解析装置には、プロテインシーケンサーPPSQ-23A(島津製作所)とPTHアナライザーSPD-10A(島津製作所)を使用した。
その結果、以下の配列が得られた。

定法に従って、データーベースSwiss-Protを用いた検索により、得られた配列はマイコプラズマ・ニューモニエのChaperone protein DnaKの2~11残基目の配列と完全に一致した。また、このアミノ酸配列より推定される該DnaKの分子量は65KDaであり、ウェスタンブロット上での抗体認識抗原の分子量とほぼ一致した。
以上より、上記で取得された抗体は、マイコプラズマ・ニューモニエ及びマイコプラズマ・ジェニタリウム特異的な抗DnaK抗体であることを確認することができた。
実施例1で得られたモノクローナル抗体の内、モノクローナル抗体MCM12及びモノクローナル抗体MCM19を使用して該抗体の感度及び交差性の検討を行った。
(1)試験菌株の培養及び調製
(1-1)感度試験用菌株
表1に示すマイコプラズマ・ニューモニエ8株をPPLOグルコースブロス(ウマ血清、新鮮酵母エキス、酢酸タリウム含有)に接種し、37℃、4日間好気培養し、ブロスのpHが6.8となったものを試験菌として用いた。ブロス中の菌数は滅菌PBSにて10段階希釈液を作製、その各希釈液をPPLO(ウマ血清、新鮮酵母エキス、酢酸タリウム含有)寒天培地上に10μL接種後、37℃、10日間培養した。寒天培地上の発育集落を40倍の光学顕微鏡にて集落数を測定し、菌数を算出した。
(1-1)で示したマイコプラズマ・ニューモニエ以外のMycoplasma属、Ureaplasma属、Acholeplasma属のそれぞれの菌株を表2に示したブロス及び培養条件にて培養した。なお、培養は37℃で実施した。また、表2における「好気」及び「嫌気」は、それぞれ、好気培養及び嫌気培養であることを示す。ブロス中の菌数は滅菌PBSにて10段階希釈液を作製、その各希釈液をPPLO(ウマ血清、新鮮酵母エキス、酢酸タリウム含有)寒天培地上に10μL接種後、37℃、10日間培養した。寒天培地上の発育集落を40倍の光学顕微鏡にて集落数を測定し、菌数を算出した。試験菌数は106~107cfu/mLで実施した。
(1-1)及び(1-2)で使用したMycoplasma属、Ureaplasma属、Acholeplasma属以外の細菌、真菌の交差反応性試験に供した微生物及びその培養条件を、表3~6にそれぞれ示した。なお、使用培地としては、ハートインフュージョンアガー(ディフコ社)、トリプチケースソイIIヒツジ寒天培地(ベクトン・デッキンソン社)、チョコレート寒天培地(日水社)、変法GAM寒天培地(日水社)、スキロー寒天培地(ベクトン・デッキンソン社)、サブロー・デキストロースアガー(ディフコ社)を使用した。
これらの菌は寒天培地で培養後、滅菌PBSに107~108cfu/mLになるように浮遊させ、試験菌とした。菌数測定は滅菌PBSに浮遊させた試験菌液を同緩衝液にて10段階希釈し、そのそれぞれを寒天培地に50μL接種し、培地上に発育した集落を肉眼にて測定した。
なお、表中の菌株No.欄の空白部分は、臨床材料から分離同定されたものである。
(2-1)ELISA法の構築
(2-1-1)固相化抗体調製方法及び固相化方法
モノクローナル抗体MCM19を含む腹水を硫安分画後、rProteinA Sepharose FF(GEヘルスケア社)を用いてIgG精製し、BCA法にて蛋白質定量を実施した。96ウェルのマイクロプレートに精製IgG抗体(10μg/mL)を固相化した。
モノクローナル抗体MCM12を含む腹水を硫安分画後、MEP Hypercel(日本ポール社)を用いてIgG精製を実施した。更に、IgGをペプシンにてF(ab’)2化後、F(ab’)2をアルカリ性ホスファターゼと架橋反応を行い、アルカリ性ホスファターゼ標識抗体を作製した。
固相化96ウェルマイクロプレートを洗浄後、1%BSAを含む0.1mmol/L TBS(pH7.5)で室温、1時間ブロッキングした。試験菌液100μLを添加し、室温で1時間反応させた。洗浄後、アルカリ性ホスファターゼ標識抗体(10μg/mL)を添加し、室温で1時間反応させた。洗浄後、基質(pNPP)溶液で30分間発色させ、反応停止後に405nmでの吸光度を測定した。
試験菌(1-1)を前述のELISAにて測定し、吸光度が0.05以上認められた最高希釈倍率の試験液から菌数を算定して表7に示した。
表7の結果から、本モノクローナル抗体を使用したELISAによって、マイコプラズマ・ニューモニエに対する感度は、103~104cfu/mLであることが判った。
試験菌(1-2)(表2に記載のマイコプラズマ・ニューモニエ以外のMycoplasma属、Ureaplasma属、Acholeplasma属)及び試験菌(1-3)(表3~6に記載の他細菌、真菌)を前述のELISAにて測定した。
マイコプラズマ・ジェニタリウム以外の微生物では、全て、吸光度が0.010未満であった。一方、マイコプラズマ・ジェニタリウムでは、吸光度が0.05以上認められた最高希釈倍率の試験液から菌数を算定したところ、6.9×104cfu/mLであった。
以上より、マイコプラズマ・ニューモニエ又はマイコプラズマ・ジェニタリウム感染症を診断する際に障害になる可能性のある、多くの細菌及び真菌とは交差しないことが確認された。
(1)イムノクロマト法の構築
(1-1)検出用標識物である金コロイド標識抗マイコプラズマ・ニューモニエ抗体の作製
直径40nmの金コロイド溶液(田中貴金属社)18mLに50mmol/L リン酸緩衝液(pH11)2mLを加えることでpHを調製した金コロイド溶液に、100μg/mLのモノクローナル抗体MCM12溶液2.5mLを加えて攪拌した。1時間攪拌した後、1質量%ポリエチレングリコール(Mw.20000、和光純薬社)水溶液を1mL加え攪拌し、続いて10質量%BSA水溶液(SIGMA社)を2mL加え攪拌した。この溶液を4℃、8000G、15分間遠心した後、1mL程度を残して上清を取り除き、超音波発生機により金コロイドを再分散した。この後、20mLのBSAを含むリン酸緩衝液に分散し、再び4℃、8000G、15分間遠心した後、1mL程度を残して上清を取り除き、超音波発生機により金コロイドを再分散し、金コロイド標識抗体溶液を作製した。
(1-1)で作製した金コロイド標識抗体溶液を上記BSAを含むリン酸緩衝液で希釈し、20mm×300mmに切ったガラスファイバーパッド(ミリポア社)に染み込ませ、一晩室温で乾燥し、金コロイド抗体保持パッドを作製した。
30mm×300mmに切断したニトロセルロースメンブレン(ミリポア社)に関して以下のような方法により、抗体を固定化し抗体固定化メンブレンを作製した。メンブレンの長辺を下にし、下から16mmの位置に、5mg/mLとなるように調製した固定化用モノクローナル抗体MCM19溶液を塗布機(BioDot社)を用いて幅1mm程度のライン状に塗布し、乾燥させて、抗体固定化メンブレンを作製した。
バック粘着シート(日栄化工社)に抗体固定化メンブレン、金コロイド保持パッド、吸収パッド(日本ポール社)を重ねて貼り付け、これら重ね貼り部材を、部材の長辺側を6mm幅になるようにカッターで切断していくことでイムノクロマトグラフ用テストストリップを作製した。これらをハウジングケースに入れ、試験用イムノクロマトグラフキットとした。
TritonX-100を含むリン酸緩衝液で培養菌、PBS洗浄菌、培養上清、培養菌の沈殿を溶解し、各濃度の試験用マイコプラズマ・ニューモニエ抗原(又は菌株)溶液を作製した。各試験用イムノクロマトキットに、各濃度の試験用マイコプラズマ・ニューモニエ抗原(又は菌株)溶液を100μL滴下し、15分後に抗体固定メンブレンの抗マイコプラズマ・ニューモニエモノクローナル抗体を塗布した部位に目視で染色が見られた場合は「陽性」、染色が見られなかった場合は「陰性」と判別した。
実施例2の試験菌(1-1)を前述のイムノクロマト法にて測定し、テストラインに出現した染色が認められた最高希釈倍率の試験液から菌数を算定して、表8に示した。
実施例2の試験菌(1-2)(表2に記載のマイコプラズマ・ニューモニエ以外のMycoplasma属、Ureaplasma属、Acholeplasma属)及び試験菌(1-3)(表3~6に記載の他細菌、真菌)を前述のイムノクロマト法にて測定した。
マイコプラズマ・ジェニタリウム以外の微生物は、全て、陰性、すなわち、染色が認められなかった。一方、マイコプラズマ・ジェニタリウムでは染色が認められ、最高希釈倍率の試験液から菌数を算定したところ、6.9×104cfu/mLであった。
以上より、マイコプラズマ・ニューモニエ又はマイコプラズマ・ジェニタリウム感染症を診断する際に障害になる可能性のある、多くの細菌及び真菌とは交差しないことが確認された。
マイコプラズマ感染症の疑いがある患者3名及び健常人33名から、咽頭拭い液を採取し、実施例3のイムノクロマト法に従って、マイコプラズマ・ニューモニエの検出を実施した。その結果、表9に示す通り、マイコプラズマ感染症の疑いがある患者3名では陽性反応が認められ、健常人33名では陰性であった。
また、同検体より定法に従ってDNAを抽出し、マイコプラズマ・ニューモニエP1遺伝子の一部(M.pneumoniae M129-B7 NCBI番号:NC_000912)を増幅する、Jensenら(APMIS. 1989;97(11):1046-8.)の定性PCR法の改法を用いて、マイコプラズマ・ニューモニエの遺伝子検出を実施し、両者を比較した。その結果、表9に示す通り、陽性、陰性共に両者は一致した。
なお、この手法で、マイコプラズマ・ジェニタリウムの遺伝子の増幅が可能なことは確認されている。
測定対象は、ATCCより購入したマイコプラズマ・ニューモニエ8株(M.pneumoniae FH:ATCC No.15531、M.pneumoniae Bru:ATCC No.15377、M.pneumoniae Mac:ATCC No.15492、M.pneumoniae Mutant 22:ATCC No.39505、M.pneumoniae M52:ATCC No.15293、M.pneumoniae PI1428:ATCC No.29085、M.pneumoniae M129-B7:ATCC No.29342および、M.pneumoniae UTMB-10P:ATCC No.49894)を用いた。これらのマイコプラズマ・ニューモニエ8株をPPLO培地にて培養し、DNAを抽出した。
DNA抽出は、スマイテストEX-R&Dキット(医学生物学研究所)を用い、10mmol/L Tris-HCl、1mmol/L EDTA Buffer pH8.0(ニッポンジーン)(以下、TE Buffer)に懸濁し、-40℃に凍結保存した。
16s rRNA領域のマイコプラズマ共通定量PCRは、以下の通り実施した。
16s rRNA遺伝子領域を増幅するマイコプラズマ属に共通なプライマーを設計し、リアルタイムPCR法を用い、標準品を元に、抽出したM.pneumoniae DNAの遺伝子コピー数を算出した。リアルタイムPCRは、LightCycler FastStart DNA Master SYBR Green I (Roche Applied Science)を用いた。
プライマー配列は以下を使用した。
M.pneumoniae M129-B7 complete genome : GenBank Accession No.NC_000912

PCR条件は、95℃で10分反応させ、次いで、94℃、10秒で変性、60℃、2秒でアニーリング、72℃で12秒を50サイクル行った。
標準品には、M.pneumoniae(M129株)の16s rRNAの一部(771bp:16s rRNAの302~1072番目)を組み換えしたpT7Blue T-Vector(タカラバイオ)の希釈系列(107、105、103、102、101copies/test)を用いた。
標準品のコピー数は、以下の計算式に基づき算出した。
DNA 濃度(μg/mL)= ABS(260nm)×50および、1 Kbp DNAの1 pmol = 0.66μgより
MpDnaK_S:5’-CTCAAACGCTAAAAGTGCTAACG-3’ 23mer(配列番号4)
MpDnaK_A:5’-AAACCATTATTACAGGTCAAATAAGAC-3’ 27mer(配列番号5)
次にPCR反応は、Mastercycler(エッペンドルフ)を使用し、94℃、30秒で変性、50℃、30秒でアニーリング、72℃、2分を50サイクル行い、最後に72℃、5分インキュベートの条件で実施した。反応後、PCR産物5μLを用い、2%アガロース電気泳動し、エチジウムブロマイドで染色後、紫外線照射により、約1,900bpの増幅バンドを確認した。
上記で調製したマイコプラズマ・ニューモニエ8株に対して検討した結果、8株すべてにおいて、102copies/testまで増幅が確認された。
測定対象は、ATCCより購入したマイコプラズマ17株(M.genitalium:ATCC No.33530、M.hominis:ATCC No.23114、Ureaplasma parvum:ATCC No.700970、U. urealyticum:ATCC No.27618、M.fermentans:ATCC No.19989、Acholeplasma laidlawii:ATCC No.23206、A.oculi:ATCC No.51735、M.penetrans:ATCC No.55252、M.pirum:ATCC No.25960、M.orale:ATCC No.23714、M.salivarium:ATCC No.23064、M.arthritidis:ATCC No.19611、M.buccale:ATCC No.23636、M.faucium:ATCC No.25293、M.lipophilum:ATCC No.27104、M.primatum:ATCC No.25948および、M.spermatophilum:ATCC No.49695)を用いた。これらのマイコプラズマ17株を各PPLO培地にて培養し、実施例5と同様にDNAを抽出し、16s rRNA領域の定量PCRにて、遺伝子コピー数を測定し、それぞれ2×105 copies/μLに希釈した。
測定対象の検体は、国立感染症研究所「病原体検出マニュアル」p.1309-1344 「マイコプラズマ肺炎」の項に記載されているマイコプラズマ・ニューモニエP1遺伝子領域のnested PCRにて、陽性であった臨床検体46例(咽頭拭い液40例、鼻汁2例、鼻咽頭吸引液1例、鼻咽頭拭い液3例)および、陰性であった30例(健常人咽頭拭い液10例、臨床検体咽頭拭い液10例、鼻汁4例、鼻咽頭吸引液3例、鼻咽頭拭い液3例)の抽出DNAを用いた。
DnaK遺伝子PCRにて増幅の認められた、実施例5のATCC株8株、実施例7の臨床検体8例(咽頭拭い液7例、鼻咽頭拭い液1例)のPCR産物の塩基配列を、BigDye Terminator v3.1(Applied Biosystems)を使用し、3130xl Genetic Analyzer(Applied Biosystems)にて解読した。
その結果、上記、ATCC株8株および、臨床検体8例のPCR産物は、DnaK遺伝子1,788bp(配列番号6)が、すべて100%一致し、GenBankに登録のあるM129株(Acc No. NC_000912)およびFH株(Acc No. CP002077)とも100%一致した。図1~図3にM129株とFH株の相同性を示す。
一方、P1遺伝子について、参考文献(JOURNAL OF CLINICAL MICROBIOLOGY, 1996, p. 447-449 Vol. 34, No. 2)に従って、PCR-RLFP法によって型鑑別を行ったところ、実施例5のATCC株8株は2群に分かれた。具体的には、M129-B7株、M52株、PI1428株、Mutant 22株の4株がI型に分類でき、FH株、Bru株、Mac株、UTMB-10P株の4株がII型に分類できた。図4~図10に代表的なM129株(配列番号7)とFH株(配列番号8)の相同性を示す。
この結果から、DnaK遺伝子は、P1遺伝子型の異なる株間でも100%一致し、また、異なる分離地および、過去50年間に分離された株おいても、塩基配列の変異が認められないため、獲得した抗体は、遺伝子型、地域、年代によって反応性に差はないと考えられた。
以上、本発明を特定の態様に沿って説明したが、当業者に自明の変形や改良は本発明の範囲に含まれる。

- マイコプラズマ・ニューモニエ又はマイコプラズマ・ジェニタリウムのDnaKを指標とすることを特徴とする、マイコプラズマ・ニューモニエ又はマイコプラズマ・ジェニタリウムの検出方法。
- DnaK蛋白質を免疫学的に分析する、請求項1に記載の方法。
- マイコプラズマ・ニューモニエ又はマイコプラズマ・ジェニタリウムに特異的な抗DnaK抗体。
- 請求項3に記載の抗DnaK抗体を含む、マイコプラズマ・ニューモニエ又はマイコプラズマ・ジェニタリウムの検出用キット。
- DnaK遺伝子を指標とする、請求項1に記載の方法。
- マイコプラズマ・ニューモニエ又はマイコプラズマ・ジェニタリウムに特異的なプライマー又はプローブ。
- 請求項6に記載のプライマー又はプローブを含む、マイコプラズマ・ニューモニエ又はマイコプラズマ・ジェニタリウムの検出用キット。
