雄性へアレスマウス(4~7週令)摘出皮膚をFranz型撹拌セルのレセプター相とドナー相の間にセットし、レセプター相にはMcIlvain緩衝液(pH 4.2)を加えた。撹拌子の回転速度は約650 rpm、実験温度は32℃とした。あらかじめMcIlvain緩衝液にて1時間水和させた皮膚の上(ドナー相)に直径12 mmの円形に切断した各種貼付剤を適用した。適用開始時点を0時間とし、8時間までは1時間ごとに、その後適用開始時点より24時間、30時間及び48時間にマニュアルにてサンプリングを行った。サンプリングは、Franz型撹拌セルのレセプター相に32℃に保温した緩衝液を0.5 mL加え、同量の試料を抜き取ることにより実施した。採取した試料を高速液体クロマトグラフィー(HPLC)にて定量し、EPZの皮膚透過量(n=4)を求めた。

1.各試料の調製
(1)エプタゾシン含有GP-1オルガノゲル
EPZ 63.56 mg(2%)、GP-1 124.73 mg(4%)及びGEFA-C8 153.47 mg(5%)を試験管に加え、さらにIPM 2677.55 mgを加えて全量 3019.31 mgとした。試験管を130℃に加熱したヒーティングブロックに入れ、混合溶液を加熱した。時々、試験管をヒーティングブロックより取り出して、ボルテックスミキサーにて撹拌し、GP-1を均一に溶解させた。GP-1が溶解した後、ヒーティングブロックの温度設定を80℃に設定し、時々撹拌して均一に混和しながら冷却した。90℃以下まで冷却したとき、ヒーティングブロックより取り出して、撹拌しながら放冷しEPZ含有オルガノゲルとした。
エプタゾシン臭化水素酸塩(EPZ・HBr) 84.42 mg(EPZとして2%)、GP-1 120.85 mg(4%)、及びGEFA-C8 147.96 mg(5%)を試験管に加え、さらにIPM 2632.08 mgを加えて全量 2985.31 mgとした。以下、上記1.(1)と同様に調製し、EPZ・HBr含有オルガノゲルとした。
EPZ 18.43 mg(0.6%)及びGEFA-C8 156.84 mg(5%)を試験管に加え、さらにIPMを加えて全量 3000 mgとした。この混液をよく撹拌した後、20分間の超音波処理で均一に分散させ、EPZ懸濁液とした。
EPZ・HBr 24.82 mg(EPZとして0.6%)及びGEFA-C8 149.60 mg(5%)を試験管に加え、さらにIPMを加えて全量 3000 mgとした。以下、上記1.(3)と同様に調製し、EPZ・HBr懸濁液とした。
(1)オルガノゲルのマウス皮膚透過性試験
雄性へアレスマウス(4~7週令)摘出皮膚をFranz型撹拌セルのレセプター相とドナー相の間にセットし、レセプター相にはMcIlvain緩衝液を加えた。撹拌子の回転速度は約650 rpm、実験温度は32℃とした。予めMcIlvain緩衝液にて1時間水和させた皮膚の上(ドナー相)に上記1.(1)及び(2)で調製した各オルガノゲルを約280 mg適用した。適用開始時点を0時間とし、8時間までは1時間ごとに、8~48時間は4時間ごとに自動経皮吸収試験器(商品名:Microette Plus、ハンソンリサーチ社製)によりサンプリングを行った。サンプリングは、Franz型撹拌セルのレセプター相から試料2.0 mLを抜き取り、32℃に保温した同量の緩衝液を補充することにより実施した。採取した試料を高速液体クロマトグラフィー(HPLC)にて定量し、EPZの皮膚透過量(n=5)を求めた。なお、HPLC条件は上記参考例1と同様である。
上記2.(1)と同様に、上記1.(3)及び(4)で調製したEPZ含有懸濁液及びEPZ・HBr含有懸濁液を1mL(比重:約0.85)適用してマウス皮膚透過性試験を行った。
1.エプタゾシン含有オルガノゲルの調製
(1)エプタゾシン含有GP-1オルガノゲル(第1法)
EPZ 60.52 mg(2%)、GP-1 121.1 mg(4%)及びGEFA-C8 151.5 mg(5%)を試験管に加え、さらにIPMを加えて全量 3000.8 mgとした。試験管を予め130℃に加熱したヒーティングブロックに入れ、混合溶液を加熱した。時々、試験管をヒーティングブロックより取り出して撹拌し、GP-1を均一に溶解させた。GP-1が溶解した後、ヒーティングブロックの温度設定を80℃に設定し、時々撹拌して薬物を均一に分散させながら冷却した。90℃位まで冷却したとき、ヒーティングブロックより取り出して、撹拌しながら放冷しオルガノゲルとした。
EPZ 100.53 mg、GEFA-C8 251.6 mg及びIPM 2011.9 mgを取り、乳鉢上にてEPZを粉砕しながら混和し、EPZ懸濁液とした。この懸濁液 1912.4 mg〔EPZ 81.32 mg(2%)、GEFA-C8 203.5 mg(5%)含有〕及びGP-1 160.1 mg(4%)を試験管に加え、さらにIPMを加えて全量 4021.2 mgとした。試験管を予め130℃に加熱したヒーティングブロックに入れ、混合溶液を加熱した。時々、試験管をヒーティングブロックより取り出して撹拌し、GP-1を均一に溶解させた。GP-1が溶解した後、ヒーティングブロックの温度設定を90℃に設定し、時々撹拌して薬物を均一に分散させながら冷却した。100℃位まで冷却したとき、ヒーティングブロックより取り出して、ゲル化が開始するまでゆっくり撹拌した。ゲル化が始まった時点で撹拌をやめて静置しオルガノゲルとした。
EPZ 60.20 mg(2%)、EB-21 121.5 mg(4%)及びGEFA-C8 153.9 mg(5%)を試験管に加え、さらにIPMを加えて全量 2999.3 mgとした。試験管を予め140℃に加熱したヒーティングブロックに入れ、混合溶液を加熱した。時々、試験管をヒーティングブロックより取り出して撹拌し、EB-21を均一に溶解させた。EB-21が溶解した後、ヒーティングブロックの温度設定を100℃に設定し、時々撹拌して均一に混和しながら冷却した。120℃以下まで冷却したとき、ヒーティングブロックより取り出して、撹拌しながら放冷しオルガノゲルとした。
EPZ 100.11 mg、GEFA-C8 250.9 mg及びIPM 2151.3 mgを取り、乳鉢上にてEPZを粉砕しながら混和し、EPZ懸濁液とした。この懸濁液 2009.7 mg〔EPZ 80.40 mg(2%)、GEFA-C8 201.5 mg(5%)含有〕及びEB-21 160.0 mg(4%)を試験管に加え、さらにIPMを加えて全量 4000.1 mgとした。試験管を予め140℃に加熱したヒーティングブロックに入れ、混合溶液を加熱した。時々、試験管をヒーティングブロックより取り出して撹拌し、EB-21を均一に溶解させた。EB-21が溶解した後、ヒーティングブロックの温度設定を100℃に設定し、時々撹拌して薬物を均一に分散させながら冷却した。120℃位まで冷却したとき、ヒーティングブロックより取り出して、ゲル化が開始するまでゆっくり撹拌した。ゲル化が始まった時点で撹拌をやめて静置しオルガノゲルとした。
EPZ 60.03 mg(2%)、GP-1 89.7 mg(3%)、EB-21 30.7 mg(1%)及びGEFA-C8 150.8 mg(5%)を試験管に加え、さらにIPMを加えて全量 3004.5 mgとした。試験管を130℃に加熱したヒーティングブロックに入れ、混合溶液を加熱した。時々、試験管をヒーティングブロックより取り出して、ボルテックスミキサーにて撹拌し、GP-1及びEB-21を均一に溶解させた。GP-1及びEB-21が溶解した後、ヒーティングブロックの温度設定を80℃に設定し、時々撹拌して均一に混和しながら冷却した。90℃以下まで冷却したとき、ヒーティングブロックより取り出して、撹拌しながら放冷しオルガノゲルとした。
EPZ 59.76 mg(2%)、レオパールKL2 298.8 mg(10%)及びGEFA-C8 153.4 mg(5%)を試験管に加え、さらにIPMを加えて全量 3002.9 mgとした。試験管を95℃に加熱したヒーティングブロックに入れ、混合溶液を加熱した。時々、試験管をヒーティングブロックより取り出して、ボルテックスミキサーにて撹拌し、レオパールKL2を均一に溶解させた。レオパールKL2が溶解した後、ヒーティングブロックより取り出して、撹拌しながら放冷しオルガノゲルとした。
EPZ 60.20 mg(2%)、レオパールKS2 297.7 mg(10%)及びGEFA-C8 153.2 mg(5%)を試験管に加え、さらにIPMを加えて全量 3005.8 mgとした。以下、上記1.(6)と同様に調製し、オルガノゲルとした。
(1)トラマドール含有GP-1オルガノゲル
トラマドール(TRD) 60.76 mg(2%)、GEFA-C8 150.4 mg(5%)及びGP-1 120.8 mg(4%)を試験管に加え、さらにIPMを加えて全量 3007.2 mgとした。試験管を予め135℃に加熱したヒーティングブロックに入れ、混合溶液を加熱した。時々、試験管をヒーティングブロックより取り出して撹拌し、混合溶液を均一に溶解させた。均一に混和した後、ヒーティングブロックの温度設定を90℃に設定し、時々撹拌し100℃以下まで冷却したとき、ヒーティングブロックより取り出して、ゲル化が開始するまで室温にてゆっくり撹拌した。ゲル化が始まった時点で撹拌をやめて静置しオルガノゲルとした。
トラマドール塩酸塩(TRD・HCl) 91.23 mg(TRDとして2%)、GEFA-C8 199.0 mg及びIPM 1602.4 mgを取り、乳鉢上にてTRD・HClを粉砕しながら混和し、TRD・HCl 懸濁液とした。この懸濁液 1438.1 mg(TRD・HCl 69.32 mg[TRDとして60.89 mg](2%)、GEFA-C8 151.2 mg(5%)含有)及びGP-1 121.3 mg(4%)を試験管に加え、さらにIPMを加えて全量 3020.0 mgとした。試験管を予め135℃に加熱したヒーティングブロックに入れ、混合溶液を加熱した。時々、試験管をヒーティングブロックより取り出して撹拌し、GP-1を均一に溶解させた。GP-1が溶解した後、ヒーティングブロックの温度設定を90℃に設定し、時々撹拌して薬物を均一に分散させながら冷却した。105℃位まで冷却したとき、ヒーティングブロックより取り出して、ゲル化が開始するまで室温にてゆっくり撹拌した。ゲル化が始まった時点で撹拌をやめて静置しオルガノゲルとした。
(1)ペンタゾシン含有GP-1オルガノゲル
ペンタゾシン(PTZ) 79.86 mg(2%)、GEFA-C8 204.0 mg(5%)及びGP-1 162.7 mg(4%)を試験管に加え、さらにIPMを加えて全量 3998.3 mgとした。試験管を予め135℃に加熱したヒーティングブロックに入れ、混合溶液を加熱した。時々、試験管をヒーティングブロックより取り出して撹拌し、混合溶液を均一に溶解させた。均一に混和した後、ヒーティングブロックの温度設定を90℃に設定し、時々撹拌し100℃以下まで冷却したとき、ヒーティングブロックより取り出して、ゲル化が開始するまで室温にてゆっくり撹拌した。ゲル化が始まった時点で撹拌をやめて静置しオルガノゲルとした。
(1)エプタゾシン含有オルガノゲルのマウス皮膚透過性試験
雄性へアレスマウス(4~9週令)摘出皮膚をFranz型撹拌セルのレセプター相とドナー相の間にセットし、レセプター相にはMcIlvain緩衝液(pH 4.2)を加えた。撹拌子の回転速度は約650 rpm、実験温度は32℃とした。予めMcIlvain緩衝液にて1時間水和させた皮膚の上(ドナー相)に上記実施例2の1.で調製した各EPZ含有オルガノゲルを約300 mg適用した。適用開始時点を0時間とし、8時間までは1時間ごとに、20~30時間、44~48時間は2時間ごとにマニュアルにてサンプリングを行った。サンプリングは、Franz型撹拌セルのレセプター相に32℃に保温したMcIlvain緩衝液を0.5 mL加え、同量の試料を抜き取ることにより実施した。採取した試料を高速液体クロマトグラフィー(HPLC)にて定量し、EPZの皮膚透過量(n=4)を求めた。なお、HPLC条件は上記参考例1と同様である。
上記(1)と同様に、上記実施例2の2.で調製したTRD(フリー体又は塩酸塩)含有オルガノゲルノゲルについて、TRDの皮膚透過量(n=4)を求めた。

1.エプタゾシン含有オルガノゲルの調製
(1)ミリスチン酸イソプロピル(IPM)オルガノゲル
IPMオルガノゲルは上記実施例2の1.(2)で調製したものを用いた。
EPZ 80.19 mg、GEFA-C8 199.9 mg及びIPP 1729.1 mgを取り、乳鉢上にてEPZを粉砕しながら混和し、EPZ懸濁液とした。この懸濁液 1501.2 mg〔EPZ 59.92 mg(2%)、GEFA-C8 149.4 mg(5%)含有〕及びGP-1 120.9 mg(4%)を試験管に加え、さらにIPPを加えて全量 3002.4 mgとした。以下、上記実施例2の1.(2)と同様に調製し、IPPオルガノゲルとした。
IPPの代わりにEHPを用いて、上記(2)と同様に調製し、EHPオルガノゲルとした。
IPPの代わりにETSを用いて、上記(2)と同様に調製し、ETSオルガノゲルとした。
IPPの代わりにETOを用いて、上記(2)と同様に調製し、ETOオルガノゲルとした。
IPPの代わりにIPLiを用いて、上記(2)と同様に調製し、IPLiオルガノゲルとした。
IPPの代わりにDIAdを用いて、上記(2)と同様に調製し、DIAdオルガノゲルとした。
IPPの代わりにDESeを用いて、上記(2)と同様に調製し、DESeオルガノゲルとした。
上記1.で調製した各種脂肪酸エステルを用いたEPZ含有オルガノゲル(1)乃至(8)について、上記実施例3(1)と同様にマウス皮膚透過性試験を行い、EPZの皮膚透過量(n=4)を測定した。各々、EPZの皮膚透過量より、透過速度及び遅延時間を算出し、累積透過量(48時間後)と共に表6に示す。また、経時的累積透過量のグラフを図9に示す。
1.エプタゾシン含有オルガノゲルの調製
(1)5%モノカプリル酸グリセリン(GEFA-C8)添加オルガノゲル
5%GEFA-C8添加オルガノゲルは上記実施例2の1.(2)で調製したものを用いた。
GEFA-C8の代わりにGEFA-C10を用いて、上記(1)と同様に調製し、5%GEFA-C10添加オルガノゲルオルガノゲルとした。
GEFA-C8の代わりにGEFA-C12を用いて、上記(1)と同様に調製し、5%GEFA-C12添加オルガノゲルとした。
GEFA-C8の代わりにGEFA-C18:1を用いて、上記(1)と同様に調製し、5%GEFA- C18:1添加オルガノゲルとした。
GEFA-C8の代わりにGEFA-diC8を用いて、上記(1)と同様に調製し、5%GEFA-diC8添加オルガノゲルとした。
GEFA-C8の代わりにGEFA-triC8を用いて、上記(1)と同様に調製し、5%GEFA-triC8添加オルガノゲルとした。
EPZ 80.25 mg及びIPM 1919.0 mgを取り、乳鉢上にてEPZを粉砕しながら混和し、EPZ懸濁液とした。この懸濁液 1614.4 mg〔EPZ 64.80 mg(2%)含有〕及びGP-1 120.0 mg(4%)を試験管に加え、さらにIPMを加えて全量 3017.5 mgとした。以下、上記実施例2の1.(2)と同様に調製し、グリセリン脂肪酸エステル無添加オルガノゲルとした。
EPZ 80.59 mg、GEFA-C8 101.5 mg及びIPM 1825.1 mgを取り、乳鉢上にてEPZを粉砕しながら混和し、EPZ懸濁液とした。この懸濁液 1502.4 mg〔EPZ 60.32 mg(2%)、GEFA-C8 75.97 mg(2.5%)含有〕及びGP-1 121.4 mg(4%)を試験管に加え、さらにIPMを加えて全量 3003.2 mgとした。以下、上記実施例2の1.(2)と同様に調製し、2.5%GEFA-C8添加オルガノゲルとした。
GEFA-C8 最終濃度が7.5%となるように、上記(8)と同様に調製し、7.5%GEFA-C8添加オルガノゲルとした。
GEFA-C8 最終濃度が10%となるように、上記(8)と同様に調製し、10%GEFA-C8オルガノゲル添加とした。
上記1.で調製した各種グリセリン脂肪酸エステルを用いたEPZ含有オルガノゲル(1)乃至(10)について、上記実施例3(1)と同様にマウス皮膚透過性試験を行い、EPZの皮膚透過量(n=4)を測定した。各々、EPZの皮膚透過量より、透過速度及び遅延時間を算出し、累積透過量(48時間後)と共に表7に示す。
1.エプタゾシン含有オルガノゲルの調製
(1)0.5%エプタゾシン含有GP-1オルガノゲル
EPZ 40.17 mg、GEFA-C8 400.3 mg及びIPM 3559.6 mgを取り、乳鉢上にてEPZを粉砕しながら混和し、EPZ懸濁液とした。この懸濁液 1518.8 mg〔EPZ 15.25 mg(0.5%)、GEFA-C8 152.0 mg(5%)含有〕及びGP-1 120.5 mg(4%)を試験管に加え、さらにIPMを加えて全量 3008.6 mgとした。以下、上記実施例2の1.(2)と同様に調製し、0.5%EPZ含有GP-1オルガノゲルとした。
EPZ が最終濃度1%となるように、上記(1)と同様に調製し、1%EPZ含有GP-1オルガノゲルとした。
2%EPZ含有GP-1オルガノゲルは上記実施例2の1.(2)で調製したものを用いた。
EPZ が最終濃度5%となるように、上記(1)と同様に調製し、5%EPZ含有GP-1オルガノゲルとした。
EPZ が最終濃度10%となるように、上記(1)と同様に調製し、10%EPZ含有GP-1オルガノゲルとした。
EPZ が最終濃度15%となるように、上記(1)と同様に調製し、15%EPZ含有GP-1オルガノゲルとした。
(1)2%トラマドール含有EB-21オルガノゲル
TRD 60.25 mg (2%) 、GEFA-C8 153.5 mg (5%) 及びEB-21 119.1 mg (4%) を試験管に加え、更にIPMを加えて全量 3002.0 mgとした。試験管をあらかじめ145℃に加熱したヒーティングブロックに入れ、混合溶液を加熱撹拌し、均一に溶解させた。均一に溶解した後、ヒーティングブロックの温度設定を100℃に設定し、時々撹拌し120℃以下まで冷却したとき、ヒーティングブロックより取り出して、ゲル化が開始するまで室温にてゆっくり撹拌した。ゲル化が始まった時点で撹拌をやめて静置し、2%TRD含有EB-21オルガノゲルとした。
TRD 300.21 mg (10%) 、GEFA-C8 150.2 mg (5%) 及びEB-21 120.8 mg (4%) を試験管に加え、更にIPMを加えて全量 3001.7 mgとした。以下、上記(1)と同様に調製し、10%TRD含有EB-21オルガノゲルとした。
3.マウス皮膚透過性試験
上記1.で調製したEPZ含有オルガノゲルについては上記実施例3(1)と同様に、また、上記2.で調製したTRD含有オルガノゲルについては上記実施例3(2)と同様に、各々、マウス皮膚透過性試験を行い、薬剤の皮膚透過量(n=4)を測定した。各々、薬剤の皮膚透過量より、透過速度及び遅延時間を算出し、累積透過量(48時間後)と共に表8に示す。
1.エプタゾシン含有オルガノゲルの調製
(1)2%GP-1オルガノゲル
EPZ 200.78 mg、GEFA-C8 500.4 mg及びIPM 4302.9 mgを取り、乳鉢上にてEPZを粉砕しながら混和し、EPZ懸濁液とした。この懸濁液 1504.7 mg〔EPZ 60.37 mg(2%)、GEFA-C8 150.5 mg(5%)含有〕及びGP-1 60.8 mg(2%)を試験管に加え、さらにIPMを加えて全量 3001.0 mgとした。以下、上記実施例2の1.(2)と同様に調製し、2%GP-1オルガノゲルとした。
4%GP-1オルガノゲルは上記実施例2の1.(2)で調製したものを用いた。
GP-1が最終濃度6%となるように、上記(1)と同様に調製し、6%GP-1オルガノゲルとした。
GP-1が最終濃度10%となるように、上記(1)と同様に調製し、10%GP-1オルガノゲルとした。
上記1.で調製した各種濃度のGP-1を用いたオルガノゲル(1)乃至(4)について、上記実施例3(1)と同様にマウス皮膚透過性試験を行い、EPZの皮膚透過量(n=4)を測定した。各々、EPZの皮膚透過量より、透過速度及び遅延時間を算出し、累積透過量(48時間後)と共に表9に示す。
1.エプタゾシン含有オルガノゲルの調製
(1)20%エタノールを添加して調製した GP-1オルガノゲル
EPZ 159.15 mg、GEFA-C8 400.5 mg及びIPM 3446.2 mgを取り、乳鉢上にてEPZを粉砕しながら混和し、EPZ懸濁液とした。この懸濁液 1519.1 mg〔EPZ 60.35 mg(2%)、GEFA-C8 151.9 mg(5%)含有〕及びGP-1 120.2 mg(4%)を試験管に加え、さらにIPMを加えて全量 3092.2 mgとした。ここに99.5% エタノール 760μL(20%)を加え、試験管を予め100℃に加熱したヒーティングブロックに入れ、混合溶液を加熱した。時々、試験管をヒーティングブロックより取り出して撹拌し、均一に溶解させた。そのまま加熱を続け、エタノールを一部留去した。ヒーティングブロックより取り出して、ゲル化が開始するまでゆっくり撹拌した。ゲル化が始まった時点で撹拌をやめて静置して、40℃にて終夜乾燥し、20%エタノールを添加して調製した GP-1オルガノゲルとした。
混合溶液の50%のエタノールを加えて、上記(1)と同様に調製し、50%エタノールを添加して調製した GP-1オルガノゲルとした。
EPZ 79.95 mg、GEFA-C8 600.6 mg及びIPM 1719.1 mgを取り、乳鉢上にてEPZを粉砕しながら混和し、EPZ懸濁液とした。この懸濁液 1505.9 mg〔EPZ 60.21 mg(2%)、GEFA-C8 151.1 mg(5%)含有〕及びゲルオールD 9.08 mg(0.3%)を試験管に加え、さらにIPMを加えて全量 3007.6 mgとした。ここに99.5% エタノール 1200μL(30%)を加え、試験管を予め100℃に加熱したヒーティングブロックに入れ、混合溶液を加熱した。以下、上記(1)と同様に調製し、30%エタノールを添加して調製したゲルオールDオルガノゲルとした。
上記1.でエタノールを添加して調製したEPZ含有オルガノゲル(1)乃至(3)について、上記実施例3(1)と同様にマウス皮膚透過性試験を行い、EPZの皮膚透過量(n=4)を測定した。各々、EPZの皮膚透過量より、透過速度及び遅延時間を算出し、累積透過量(48時間後)と共に表10に示す。
1.エプタゾシン含有オルガノゲルの調製
上記実施例2の1.(2)と同様に調製したEPZ含有GP-1オルガノゲルを用いた。
ヘアレスマウス摘出皮膚の上(ドナー相)に、薬物放出膜(多孔性ポリプロピレン膜 4×4cm、商品名:Celgard2400、ポリポア社製)、又は、3種のメンブレンフィルターA〔ニトロセルロース、0.20μm、133μm〕、メンブレンフィルターB〔四フッ化エチレン樹脂、0.20μm、80μm 〕、メンブレンフィルターC〔親水性特殊処理四フッ化エチレン樹脂、0.20μm、35μm〕(各々括弧内は、材質、孔径、厚さを示す。いずれもADVANTEC社製。)の各々を載せてからオルガノゲルを適用する以外は上記実施例3(1)と同様に、上記1.で調製したEPZ含有GP-1オルガノゲルのEPZの皮膚透過量(n=4)を測定した。各々、EPZの皮膚透過量より、透過速度及び遅延時間を算出し、累積透過量(48時間後)と共に表11に示す。
1.粘着剤の調製
本実施例で使用する粘着剤のうち、Eudragit E 粘着剤(アクリル系粘着剤、EVONIC DEGUSSA JAPAN社製)については、以下の通りに調製したもの粘着剤として使用した。アセトン、エタノール及び2-プロパノールをそれぞれ19.9g、11.1g及び2.2gをビーカーに量り取り、撹拌して均一に混和した。ここに、アミノアルキルメタクリレートコポリマーE(Eudragit E PO、EVONIC DEGUSSA JAPAN社製)37.5gを撹拌しながら少量ずつ加え、溶解させた。その後、可塑剤であるセバシン酸ジブチル19.9gをすばやく加え、10分間撹拌した。最後に、架橋剤であるコハク酸3.4gを撹拌しながら少量ずつ加えて固形成分を完全に溶解させ、Eudragit E粘着剤 (固形成分含量64.7%) とした。
A.GP-1オルガノゲルマトリックス型貼付剤(粘着剤、塗工厚)
(1)2%エプタゾシン含有貼付剤(Eudragit E 50%、250μm)
EPZ 20.27 mg (2%) 、GEFA-C8 52.1 mg (5%) 、GP-1 39.6 mg (4%) 及びIPM 388.4 mg (39%)をそれぞれ秤取し、試験管に加えた。ここに、Eudragit E粘着剤785.7 mg (固形成分500.5 mg) を加えて全量 (固形成分1000.87 mg) とした後、エタノール500μLを加えた。試験管を予め90℃に加熱したヒーティングブロックに入れ、混合溶液を加熱撹拌して均一に溶解させた。この溶液を平面ガラス板上に固定した支持体 (商品名:Scotchpak 9732 Backing、3M社製) 上に、フィルムアプリケーター(商品名:MULTICATOR 411、ERICHSEN Gmbh & Co. KG製)を用いて厚さ250μmで塗工した。次に、送風定温乾燥器内で、60℃で30分間乾燥した。乾燥後、剥離フィルム (商品名:Scotchpak 1022 Release Liner、3M社製) のフッ素樹脂加工面を製剤の接着面と張り合わせ、2%EPZ含有貼付剤 (Eudragit E 50%) とした。
EPZ 20.72 mg (2%) 、GEFA-C8 50.3 mg (5%) 、GP-1 41.3 mg (4%) 及びIPM 396.8 mg (39%)をそれぞれ秤取し、試験管に加えた。ここに、Duro-Tak 87-9301粘着剤1239.9 mg (固形成分496.0 mg) を加えて全量 (固形成分1005.1 mg) とした後、エタノール700μLを加えた。以下、上記(1)と同様に調製し、2%EPZ含有貼付剤 (Duro-Tak 87-9301 50%) とした。
EPZ 20.17 mg (2%) 、GEFA-C8 49.9 mg (5%) 、GP-1 41.0 mg (4%) 及びIPM 390.4 mg (39%)をそれぞれ秤取し、試験管に加えた。ここに、BIO-PSA 7-4202粘着剤837.6 mg (固形成分502.6 mg) を加えて全量 (固形成分1004.1 mg) とした後、エタノール700μLを加えた。以下、上記(1)と同様に調製し、2%EPZ含有貼付剤 (BIO-PSA 7-4202 50%) とした。
EPZ 20.32 mg (2%) 、GEFA-C8 50.4 mg (5%) 、GP-1 40.8 mg (4%) 及びIPM 393.1 mg (39%)をそれぞれ秤取し、試験管に加えた。ここに、Eudragit E粘着剤787.8 mg (固形成分501.8 mg) を加えて全量 (固形成分1006.4 mg) とした後、エタノール500μLを加えた。以下、塗工の厚さを400μmとする以外は、上記(1)と同様に調製し、2%EPZ含有貼付剤 (Eudragit E 50%) とした。
EPZ 50.20 mg (5%) 、GEFA-C8 51.5 mg (5%) 、GP-1 40.1 mg (4%) 及びIPM 376.8 mg (36%)をそれぞれ秤取し、試験管に加えた。ここに、Eudragit E粘着剤789.2 mg (固形成分502.7 mg) を加えて全量 (固形成分1021.3 mg) とした後、エタノール1500μLを加えた。以下、上記(4)と同様に調製し、5%EPZ含有貼付剤 (Eudragit E 50%) とした。
(1)2%エプタゾシン含有貼付剤(Eudragit E 50%、400μm)
EPZ 20.27 mg (2%) 、GEFA-C8 52.1 mg (5%) 、EB-21 39.6 mg (4%) 及びIPM 388.4 mg (39%)をそれぞれ秤取し、試験管に加えた。ここに、Eudragit E粘着剤785.7 mg (固形成分500.5 mg) を加えて全量 (固形成分1000.87 mg) とした後、エタノール500μLを加えた。以下、上記A.(4)と同様に調製し、2%EPZ含有貼付剤 (Eudragit E 50%) とした。
EPZ 50.68 mg (5%) 、GEFA-C8 49.6 mg (5%) 、EB-21 40.5 mg (4%) 及びIPM 360.0 mg (36%)をそれぞれ秤取し、試験管に加えた。ここに、Eudragit E粘着剤786.4 mg (固形成分500.9 mg) を加えて全量 (固形成分1001.7 mg) とした後、エタノール1000μLを加えた。以下、上記A.(4)と同様に調製し、5% EPZ含有貼付剤 (Eudragit E 50%) とした。
EPZ 99.90 mg (10%) 、GEFA-C8 50.6 mg (5%) 、EB-21 40.7 mg (4%) 及びIPM 310.6 mg (31%)をそれぞれ秤取し、試験管に加えた。ここに、Eudragit E粘着剤790.1 mg (固形成分503.3 mg) を加えて全量 (固形成分1005.1 mg) とした後、エタノール2500μLを加えた。以下、上記A.(4)と同様に調製し、10%EPZ含有貼付剤 (Eudragit E粘着剤50%) とした。
EPZ 151.66 mg (15%) 、GEFA-C8 49.6 mg (5%) 、EB-21 40.8 mg (4%) 及びIPM 363.8 mg (36%)をそれぞれ秤取し、試験管に加えた。ここに、Eudragit E粘着剤636.3 mg (固形成分405.3 mg) を加えて全量 (固形成分1011.2 mg) とした後、エタノール3300μLを加えた。試験管を予め90℃に加熱したヒーティングブロックに入れ、混合溶液を加熱撹拌して均一に溶解させた。この溶液100 μLをアルミ箔で覆った鋳型 (φ15×1 mm) に分注した。次に、送風定温乾燥器内で、60℃で3時間、40℃ で15時間乾燥した。乾燥後、剥離フィルム (Scotchpak 1022 Release Liner) のフッ素樹脂加工面を製剤の接着面と張り合わせ、15%EPZ含有貼付剤 (Eudragit E 40%) とした。
EPZ 40.72 mg (2%) 、GEFA-C8 101.4 mg (5%) 、EB-21 80.5 mg (4%) 及びIPM 782.5 mg (39%)をそれぞれ秤取し、試験管に加えた。ここに、Eudragit E粘着剤1566.1 mg (固形成分1003.9 mg) を加えて全量 (固形成分2008.9 mg) とした後、エタノール500μLを加えた。試験管を予め90℃に加熱したヒーティングブロックに入れ、混合溶液を加熱撹拌して均一に溶解させた。この溶液を平面ガラス板上に固定した支持体 (Scotchpak 9732 Backing) 上に、フィルムアプリケーター(MULTICATOR 411)を用いて厚さ250μmで塗工した。次に、送風定温乾燥器内で、60℃で1.5時間、40℃で15時間乾燥した。乾燥後、剥離フィルム (Scotchpak 1022 Release Liner) のフッ素樹脂加工面を製剤の接着面と張り合わせ、2%EPZ含有貼付剤 (Eudragit E 50%) とした。
EPZ 40.66 mg (2%) 、GEFA-C8 100.9 mg (5%) 、EB-21 80.7 mg (4%) 及びIPM 382.5 mg (19%)をそれぞれ秤取し、試験管に加えた。ここに、Eudragit E粘着剤2186.0 mg (固形成分1401.3 mg) を加えて全量 (固形成分2006.0 mg) とした後、エタノール500μLを加えた。以下、上記(5)と同様に調製し、2%EPZ含有貼付剤 (Eudragit E 70%) とした。
EPZ 20.77 mg (2%) 、GEFA-C8 49.8 mg (5%) 、EB-21 41.4 mg (4%) 及びIPM 288.7 mg (29%)をそれぞれ秤取し、試験管に加えた。ここに、Eudragit E粘着剤775.7 mg (固形成分497.2 mg) 及びDuro-Tak 87-9301 (固形成分含量 60%) 277.7 mg (固形成分 111.1 mg) を加えて全量 (固形成分1009.0 mg) とした後、エタノール500μLを加えた。以下、上記(5)と同様に調製し、2%EPZ含有貼付剤 (Eudragit E 50%+Duro-Tak 87-9301 10%) とした。
EPZ 20.48 mg (2%) 、GEFA-C8 53.0 mg (5%) 、EB-21 40.3 mg (4%) 及びIPM 289.6 mg (29%)をそれぞれ秤取し、試験管に加えた。ここに、Eudragit E粘着剤797.8 mg (固形成分511.4 mg) 及びエタノール500μLを加えた。試験管を予め90℃に加熱したヒーティングブロックに入れ、混合溶液を加熱撹拌して均一に溶解させた。ここにQuintone M100を99.4 mg (10%) を加えて全量 (固形成分1014.1 mg) とした後、さらに酢酸エチル 1000μLを加えた。以下、上記(5)と同様に調製し、2%EPZ含有貼付剤 (Eudragit E 50%+Quintone M100 10%) とした。
EPZ 21.51 mg (2%) 、GEFA-C8 50.7 mg (5%) 、EB-21 41.7 mg (4%) 及びIPM 401.1 mg (40%)をそれぞれ秤取し、試験管に加えた。ここにエタノール500 μLを加え、試験管を予め120℃に加熱したヒーティングブロックに入れ、混合溶液を加熱撹拌して均一に溶解させた。ここにQuintone M100 269.3 mg (27%) 及びQuintac 3421 220.6 mg (22%) を加えて全量 (固形成分1004.8 mg) とした後、さらにトルエン 1500μLを加えた。120℃に加熱したヒーティングブロックに入れ、混合溶液を加熱撹拌して均一に溶解させた。以下、上記(5)と同様に調製し、2% EPZ含有貼付剤 (Quintone M100 27%+Quintac 3421 22%) とした。
EPZ 23.0 mg (2%) 、GEFA-C8 51.5 mg (5%) 、EB-21 40.3 mg (4%) 及びIPM 430.9 mg (43%)をそれぞれ秤取し、試験管に加えた。ここに、Eudragit E粘着剤552.1 mg (固形成分353.9 mg) 及びエタノール500μLを加えた。試験管を予め120℃に加熱したヒーティングブロックに入れ、混合溶液を加熱撹拌して均一に溶解させた。ここにQuintone M100 62.4 mg (6%) 及びQuintac 3421 53.9 mg (5%) を加えて全量 (固形成分1015.8 mg) とした後、さらにトルエン 1500μLを加えた。以下、上記(9)と同様に調製し、2%EPZ含有貼付剤 Eudragit E 35%+Quintone M100 6%+Quintac 3421 5%) とした。
EPZ 20.76 mg (2%) 、GEFA-C8 51.7 mg (5%) 、EB-21 39.8 mg (4%) 及びIPM 389.3 mg (39%)をそれぞれ秤取し、試験管に加えた。ここに、Eudragit E粘着剤488.1 mg (固形成分309.5 mg) 及びエタノール200μLを加えた。試験管を予め100℃に加熱したヒーティングブロックに入れ、混合溶液を加熱撹拌して均一に溶解させた。ここにQuintac 3421 104.6 mg (10%) 及びクリアロン P125 100.8 mg (10%) を加えて全量 (固形成分1016.4 mg) とした後、さらにトルエン 1000 μLを加えた。以下、上記(9)と同様に調製し、2% EPZ含有貼付剤(Eudragit E 30%+Quintac 3421 10%+クリアロン P125 10%) とした。
EPZ 20.05 mg (2%) 、GEFA-C8 51.0 mg (5%) 、EB-21 40.2 mg (4%) 及びIPM 349.8 mg (35%)をそれぞれ秤取し、試験管に加えた。ここにエタノール400μLを加え、試験管を予め120℃に加熱したヒーティングブロックに入れ、混合溶液を加熱撹拌して均一に溶解させた。ここにQuintac 3421 268.0 mg (27%) 及びエステルガムH 270.5 mg (27%) を加えて全量 (固形成分999.4 mg) とした後、さらにトルエン 1500μLを加えた。以下、上記実施例(9)と同様に調製し、2%EPZ含有貼付剤 (Quintac 3421 27%+エステルガムH 27%) とした。
TRD 100.38 mg (10%) 、IPM 312.8 mg (31%) 、GEFA-C8 51.30 mg (5%) 及びEB-21 43.40 mg (4%) をそれぞれ秤取し、試験管に加えた。ここに、Eudragit E粘着剤790.5 mg (固形成分507.3 mg) を加えて全量 (固形成分1015.2 mg) とした後、エタノール300μLを加えた。試験管をあらかじめ100℃に加熱したヒーティングブロックに入れ、混合溶液を加熱撹拌して均一に溶解させた。この溶液を平面ガラス板上に固定した支持体 (Scotchpak 9732 Backing) 上に、フィルムアプリケーター(MULTICATOR 411)を用いて厚さ250μmで塗工した。次に、送風定温乾燥器内で、60℃で1時間、40℃で終夜乾燥した。乾燥後、剥離フィルム (Scotchpak 1022 Release Liner) のフッ素樹脂加工面を製剤の接着面と張り合わせ、10%TRD含有貼付剤 (Eudragit E 50%) とした。
PTZ 101.00 mg (10%) 、IPM 320.1 mg (31%) 、GEFA-C8 61.1 mg (5%) 及びEB-21 40.6 mg (4%) をそれぞれ秤取し、試験管に加えた。ここに、Eudragit E粘着剤783.5 mg (固形成分502.2 mg) を加えて全量 (固形成分1025.0 mg) とした後、エタノール300μLを加えた。試験管を予め90℃に加熱したヒーティングブロックに入れ、混合溶液を加熱撹拌して均一に溶解させた。この溶液を平面ガラス板上に固定した支持体 (Scotchpak 9732 Backing) 上に、フィルムアプリケーター(MULTICATOR 411)を用いて厚さ250μmで塗工した。次に、送風定温乾燥器内で、60℃ で1時間、40℃で15時間乾燥した。乾燥後、剥離フィルム (Scotchpak 1022 Release Liner) のフッ素樹脂加工面を製剤の接着面と張り合わせ、10%PTZ含有貼付剤 (Eudragit E 50%) とした。
ヘアレスマウス摘出皮膚の上(ドナー相)に、オルガノゲルの代わりに上記2.乃至4.で調製した各薬剤含有貼付剤(φ12mm)を適用する以外は、上記1.で調製したEPZ含有貼付剤については上記実施例3(1)と同様に、また、上記3.で調製したTRD含有貼付剤ついては上記実施例3(2)と同様に、また、上記4.で調製したPTZ含有オルガノゲルについては上記実施例3(3)と同様に、各々、マウス皮膚透過性試験を行い、各薬剤の皮膚透過量(n=4)を測定した。各々、薬剤の皮膚透過量より、透過速度及び遅延時間を算出し、累積透過量(上記2.B.(5)乃至(12)は28時間後、その他は48時間後)と共に表12及び表13に示す。また、上記2.B.(1)乃至(4)のEPZ含有貼付剤、上記3.のTRD含有貼付剤及び上記4.のPTZ含有貼付剤について、各々、経時的累積透過量のグラフを図10乃至12に示す。
参考例1において示したとおり、表1に処方を示した種々のアクリル系粘着剤を使用したマトリックス型貼付剤では、図1及び表2に示した結果のように、好ましいEPZの皮膚透過性は得られなかった。すなわち、10%又は20%という高濃度の薬剤含有マトリックスであっても、2%EPZ含有のオルガノゲルと比べて、透過速度及び累積透過量は極めて低かった(表2、表5)。
また、へアレスマウスよりも皮膚の形態や薬物透過性がヒトに近いとされるブタ(ユカタンミニブタ)の表皮(肩部)を用いてEPZ含有オルガノゲル(実施例2の1.(2))の皮膚透過性試験を行った結果においても、ヘアレスマウスの皮膚と比較して48時間後の累積透過量で62%の皮膚透過量が認められ、EPZが確実にブタ皮膚を透過することが示された。
また同様に、表7に示したように、本発明は各種のグリセリン脂肪酸エステルを用いることができる。特にモノカプリル酸グリセリン、モノカプリン酸グリセリン及びモノラウリン酸グリセリンを添加して製造したEPZ含有オルガノゲル組成物は、優れた皮膚透過性を示した。グリセリン脂肪酸エステルの添加濃度については、表7の(1)及び(8)乃至(10)のモノカプリル酸グリセリンの結果に示されるように、7.5%まで濃度依存的に皮膚透過性が向上し、10%では7.5%と同程度の皮膚透過性を示した(実施例5参照)。

- マトリックス型貼付剤が、アクリル樹脂、シリコン樹脂、スチレンイソプレンブロック共重合体、脂肪族系炭化水素樹脂、脂環族飽和炭化水素樹脂、テルペン樹脂、ロジンエステル樹脂及びポリイソブチレン樹脂から選ばれる1種又は複数の粘着剤を組み合わせて作製する請求項43記載の貼付剤。
- 請求項24乃至40のいずれか一項に記載の製造方法で製造された外用医薬組成物を用いて製剤化したゲル剤。
- 請求項24乃至40のいずれか一項に記載の製造方法で製造された外用医薬組成物を用いて製剤化した軟膏剤又はクリーム剤。
