また、本発明の方法によれば、ラタノプロストを含有する点眼剤等の水溶液に、界面活性剤、及び炭素数3~10の脂肪族モノ-もしくはジ-カルボン酸又はその塩を含有させることで、界面活性剤のみを添加する従来法と比較して、ラタノプロストの樹脂への吸着をより顕著に抑制することができる。
本明細書において「水性点眼剤」とは、水性の点眼用液剤をいい、エマルション形態(油中水型、水中油型等)の点眼剤を含まない。水性点眼剤としては、たとえば、水溶液等の一相系溶液、油溶性物質がミセルにより可溶化された水溶液、水を基剤とした不溶性の物質の分散液又は懸濁液等が挙げられる。
本明細書において「ラタノプロスト」とは、化学名:(+)-イソプロピル-(Z)-7-[(1R,2R,3R,5S)-3,5-ジヒドロキシ-2-[(3R)-3-ヒドロキシ-5-フェニルペンチル]シクロペンチル]-5-ヘプテノエートで表される物質をいう。ラタノプロストは、緑内障治療薬として用いられるプロスタグランジンF2α誘導体である。
本明細書において「界面活性剤」とは、分子内に親水性部分と疎水性部分とを分かれて有し、液体に溶けて表面張力を著しく低下させる作用を有する物質をいう。
本明細書において「非イオン性界面活性剤」とは、水に溶解したときに電離してイオンにならない界面活性剤をいう。

本明細書において「ポリテレフタル酸エステル製容器」とは、ポリテレフタル酸エステルを主材料として成形された容器をいう。
本明細書において「ポリテレフタル酸エステル」とは、テレフタル酸と二価アルコールからなるポリエステルをいう。
一つの局面において、本発明は、ラタノプロスト、界面活性剤、及び炭素数3~10の脂肪族モノ-もしくはジ-カルボン酸又はその塩を含有する、水性点眼剤を提供する。ここで、水性点眼剤の形態としては、たとえば、水溶液等の一相系溶液、油溶性物質がミセルにより可溶化された水溶液、水を基剤とした不溶性の物質の分散液又は懸濁液等が挙げられるが、これらに限定されない。かかる構成をとることにより、本発明の水性点眼剤は、優れた眼圧降下作用と、ラタノプロストの樹脂への吸着抑制効果を得ることができる。
他の実施形態において、本発明の水性点眼剤におけるラタノプロストの含有量は、約0.00005(w/v)%~約0.005(w/v)%、約0.0001(w/v)%~約0.001(w/v)%、約0.0002(w/v)%~約0.0008(w/v)%、約0.0003(w/v)%~約0.0007(w/v)%、約0.0004(w/v)%~約0.0006(w/v)%であり得る。
実際に投与されるラタノプロストの量は、処置が適用されるべき個体に依存し、好ましくは、眼圧降下作用が顕著な副作用を伴うことなく達成されるように最適化された量である。有効用量の決定は十分に当業者の能力内にある。
ラタノプロストは水に溶けにくい疎水性の化合物である。そのためラタノプロストは疎水結合によって樹脂表面に吸着しやすい。一方、界面活性剤は水中でミセルを形成し、疎水性の化合物を取り込むことができる。水溶液中にラタノプロストが存在する場合、疎水性のラタノプロストはこのミセルに取り込まれると考えられる。ミセルに取り込まれたラタノプロストは、樹脂表面には吸着せず、界面活性剤はラタノプロストの樹脂表面への吸着に抑制的に働くと考えられる。従って、本発明において、界面活性剤は種類には関係なく使用することが可能であり、どのような界面活性剤を採用してもラタノプロストの樹脂表面への吸着を抑制するという効果を発揮する。
脂肪族モノ-もしくはジ-カルボン酸は、分子内に疎水性の炭素鎖部分と親水性のカルボン酸を有する。これは、界面活性剤が分子内に親水性部分と疎水性部分を有することとよく類似している。脂肪族モノ-もしくはジ-カルボン酸はその程度に差はあるが、界面活性剤と同様に液体に溶けて表面張力を低下させる作用を有すると考えられる。従って、このような構造を有する脂肪族モノ-もしくはジ-カルボン酸であれば、ラタノプロストの樹脂表面への吸着を抑制する効果を発揮することができるので、本発明において使用することが可能である。
これらの中では、炭素数3~7の脂肪族モノ-もしくはジ-カルボン酸が好ましく用いられる。さらに、炭素数4~6の直鎖状又は分岐鎖状の飽和又は不飽和のモノカルボン酸又はジカルボン酸がより好ましく、特にソルビン酸が好ましい。
本発明の水性点眼剤において、炭素数3~10の脂肪族モノ-もしくはジ-カルボン酸又はその塩の含有量は、界面活性剤の種類によって異なるが、通常、約0.01(w/v)%~約10(w/v)%程度、好ましくは約0.02(w/v)%~約5(w/v)%、より好ましくは約0.04(w/v)%~約2(w/v)%程度である。また、本発明の水性点眼剤にβ遮断薬(例えば、チモロール)を組み合せる場合には、β遮断薬の眼内移行性を考慮して、脂肪族モノ-もしくはジ-カルボン酸の濃度を適宜選択することは当業者の能力の範囲内である。
特に、炭素数3~7の脂肪族モノ-もしくはジ-カルボン酸及びその塩については、チモロール又はその塩の眼内移行性を亢進することが報告されている(国際公開第99/22715号パンフレット)。
本発明の水性点眼剤において、チモロール又はその塩を含有する場合は、上述の炭素数3~10の脂肪族モノ-もしくはジ-カルボン酸又はその塩のうち、炭素数3~7の脂肪族モノ-もしくはジ-カルボン酸又はその塩を用いることが好ましい。炭素数3~7の脂肪族モノ-もしくはジ-カルボン酸又はその塩を用いることにより、チモロール又はその塩の房水移行性が向上するという効果を有する。
本発明の水性点眼剤に、さらにチモロール又はその塩を含有させる場合、該薬物の房水移行性又は眼内移行性亢進効果を考慮すれば、炭素数3~10(特に炭素数3~7)の脂肪族モノ-もしくはジ-カルボン酸又はその塩の含有量は、好ましくは約0.01(w/v)%~約10(w/v)%、より好ましくは約0.04(w/v)%~約2(w/v)%の範囲である。
等張化剤は、通常、浸透圧比が約0.8~約1.2になるように添加することができる。また、緩衝剤は、約0.01(w/v)%~約2(w/v)%程度添加され得る。安定化剤は、約0.001(w/v)%~約1(w/v)%程度添加され得る。粘稠剤は、約0.001(w/v)%~約3(w/v)%程度添加され得る。
滅菌ろ過工程において、ろ過膜及びフィルターカートリッジの外枠に用いられる樹脂としては、たとえば、酢酸セルロース、芳香族ポリアミド、ポリビニルアルコール、ポリアクリロニトリル、ポリスルホン、ポリエチレン、ポリプロピレン、ポリフッ化ビニリデン、ポリエチレンテレフタレート等が挙げられ得るが、これらに限定されない。水性点眼剤の充填工程において、チューブ、パッキング、バルブ等に用いられる樹脂としては、たとえば、シリコーン樹脂、ポリテトラフルオロエチレン等が挙げられ得るが、これらに限定されない。水性点眼剤を充填する容器に用いられる樹脂としては、たとえば、ポリエチレン、ポリプロピレン、ポリ塩化ビニル、ポリ塩化ビニリデン、ポリテトラフルオロエチレン、ポリエチレンテレフタレート、ポリプロピレンテレフタレート、ポリブチレンテレフタレート、ポリスチレン、ポリ酢酸ビニル、ポリウレタン、ポリメタクリル酸メチル、ポリカーボネート等のプラスチック樹脂が挙げられ得るが、これらに限定されない。
本発明には、例えば、ポリエチレン製容器(低密度ポリエチレン(例えば、東ソー、175K)で成形した容器、低密度ポリエチレンでブロー成形したポリエチレン製容器(阪神化成工業株式会社、点眼容器))、ポリプロピレン製容器(例えば、ポリプロピレン(例えば、日本ポリプロ、NLB340G)でブロー成形した容器、ポリプロピレンでブロー成形したポリプロピレン製容器(西口アンプル製作所、点眼容器))等を使用することができる。本発明において使用される樹脂製部材及び樹脂製容器は、容器会社などから市販されているものをそのまま用いることが可能であり、あるいは、公知の方法により製造することが可能である。
一つの実施形態において、本発明の水性点眼剤の製造において用いられる、樹脂製部材及び樹脂製容器としては、ポリエチレン、ポリプロピレン等のポリオレフィン樹脂;ポリエチレンテレフタレート、ポリプロピレンテレフタレート、ポリブチレンテレフタレート等のポリアルキレンテレフタレート等のポリテレフタル酸エステル樹脂;ポリフッ化ビニリデン、ポリテトラフルオロエチレン、ポリスチレン等の、ハロゲン基又はフェニル基等により置換されたポリオレフィン樹脂などを主材料として形成されたものが好ましく用いられ得る。
他の局面において、本発明は、界面活性剤ならびに炭素数3~10の脂肪族モノ-もしくはジ-カルボン酸又はその塩を含有させることにより、水溶液中のラタノプロストの樹脂への吸着を抑制する方法を提供する。
他の局面において、本発明は、界面活性剤、及び炭素数3~10の脂肪族モノ-もしくはジ-カルボン酸又はその塩を含有させることにより、水溶液中のラタノプロストの熱安定性を向上させる方法を提供する。
本発明の水性点眼剤Aには、(c)チモロール又はその塩、(d)炭素数3~10の脂肪酸又はその塩を含有させることができる。ラタノプロスト、チロキサポール、チモロール、樹脂等として、上述の(水性点眼剤)の項に記載されるものと同様のものが使用され得る。炭素数3~10の脂肪酸としては、上述の(水性点眼剤)の項に記載される炭素数3~10の脂肪族モノ-もしくはジ-カルボン酸と同様のものが使用され得る。
実施例1の水性点眼剤の処方を、比較例1及び2の水性点眼剤の処方とともに表1に示す。これらの調製は、まず、表1中の(1)を(8)(10%ベンザルコニウム塩化物溶液)に加えて約50℃で攪拌溶解しラタノプロスト原液を調製した。別容器に(2)~(7)及び適量の(11)を入れ、攪拌溶解して基剤原液を調製した。基剤原液をラタノプロスト原液へ全量加え、十分攪拌したのち、(9)及び(10)を加えてpH6.7に調整した後、(11)を加えて全量1Lとした。
実施例1、比較例1及び2で調製した水性点眼剤を、ポリフッ化ビニリデン製の樹脂膜(デュラポアメンブレンフィルターGVWP04700、日本ミリポア株式会社製)を用いてそれぞれ50mLずつろ過した。ろ液に含まれるラタノプロスト量を定量し、ろ過前の各水性点眼剤中のラタノプロスト含有量を100%とした回収率(%)を算出した。結果を図1に示す。なお、ラタノプロストの定量は、下記HPLC測定条件Iの条件下にて、高速液体クロマトグラフィー(HPLC)により行った。
<HPLC測定条件I>
測定サンプル調製:各ラタノプロスト点眼液2mLを正確に量り、希釈液1(pH2.0トリフルオロ酢酸溶液/アセトニトリル(1:2))を加えて正確に5mLとし、試料溶液とした。別にラタノプロスト標準品約0.1gを精密に量り、アセトニトリルを加えて溶かし、正確に100mLとし、標準原液とした。標準原液4mLを正確に量り、アセトニトリルを加えて正確に20mLとした。この液2mLを正確に量り、希釈液2(pH3.5トリフルオロ酢酸溶液/アセトニトリル(2:1))を加えて正確に20mLとし、標準溶液とした。試料溶液及び標準溶液100μLにつき、次の条件で液体クロマトグラフィーにより試験を行い、ラタノプロストのピーク面積Atl及びAslを自動積分法により測定した。

実施例1、比較例1及び2の水性点眼剤を、それぞれ5mLずつガラスアンプル(大和特殊硝子、5mL無色粉末ガラスアンプル)及び低密度ポリエチレンでブロー成形したポリエチレン製容器(阪神化成工業株式会社製)各2個に充填し、試料とした。各試料を40℃及び60℃でそれぞれ2週間保存した後、ラタノプロストを上記試験例1と同様にHPLC(HPLC測定条件I)により定量し、保存前の各水性点眼剤中の含有量に対する比(%)を算出し、表3に示した。
実施例1、比較例1及び2の水性点眼剤を、それぞれ5mLずつ、低密度ポリエチレンでブロー成形したポリエチレン製容器(阪神化成工業株式会社製)、及びポリプロピレンでブロー成形したポリプロピレン製容器(西口アンプル製作所製)の各6個に充填し、試料とした。各試料を25℃、40℃及び60℃でそれぞれ8日間又は16日間保存した。次いで、上記試験例1と同様の方法(HPLC測定条件I)を用いて、ラタノプロストをHPLCにより定量し、保存前の各水性点眼剤中の含有量に対する比(%)を算出した。その結果を、表4に示す。
実施例2~6の水性点眼剤の処方を、比較例3の水性点眼剤の処方とともに、表5に示した。表5中、各成分についての数値は含有量((w/v)%)を示す。また「適量」とは、塩酸及び水酸化ナトリウムについては、水性点眼剤のpHを6.7とするのに必要な量を示し、精製水については、全量を100(w/v)%とする量を示す。これらの水性点眼剤は、実施例1と同様の方法を用いて調製した。
本実施例2~6及び比較例3の水性点眼剤について、試験例1と同様に樹脂膜への吸着性試験を行った。結果を表5に併せて示した。
本実施例では、界面活性剤とソルビン酸とが、ラタノプロストの樹脂への吸着を抑制するという効果をより正確に評価するため、ソルビン酸と相互作用するベンザルコニウム塩化物を含まない処方を用いた。
実施例7~11、比較例10~13の水性点眼剤の処方を、表6に示した。表7中、各成分についての数値は含有量(w/v%)を示す。
実施例7~11、比較例10~13の水性点眼剤は、表6中、(1)~(10)を(14)に加えて約80℃で攪拌し、溶解した後、(12)及び(13)を加えてpHを6.7に調整し、(14)で全量を200mLとして調製した。
実施例7~11、比較例10~13で調製した水性点眼剤を、ポリフッ化ビニリデン製の樹脂膜(デュラポアメンブレンフィルター GVWP04700、日本ミリポア株式会社製)を用いてそれぞれ50mLずつろ過した。ろ液に含まれるラタノプロスト量を定量し、ろ過前の各水性点眼剤のラタノプロスト含有量を100%とした回収率(%)を算出した。結果を表6に示す。なお、ラタノプロストの定量は、HPLC測定条件Iにて、高速液体クロマトグラフィーにより行った。
実施例12~15及び比較例14の水性点眼剤の処方を、表8に示した。表8中、各成分についての数値は含有量(w/v%)を示す。これらの調製は、まず、表8中の(1)を(11)(10%ベンザルコニウム塩化物溶液)に加えて約50℃で攪拌溶解しラタノプロスト原液を調製した。別容器に(2)~(10)及び適量の(14)を入れ、攪拌溶解して基剤原液を調製した。基剤原液をラタノプロスト原液へ全量加え、十分攪拌したのち、(12)及び(13)を加えてpH6.7に調整した後、(14)を加えて全量1Lとした。
実施例12~15、比較例14の水性点眼剤を、それぞれ5mLずつ、低密度ポリエチレンでブロー成型したポリエチレン製容器(阪神化成工業株式会社製)、及びポリプロピレンでブロー成型したポリプロピレン製容器(株式会社西口アンプル製作所製)に充填し、試料とした。各試料を60℃で4週間保存した後、試料中のラタノプロストを高速液体クロマトグラフィー(HPLC)により定量し、保存前の各水性点眼剤中の含有量に対する比(%)を算出し、表8に示した。なお、高速液体クロマトグラフィーの条件(HPLC測定条件II)を下記に示した。
測定サンプル調製:各ラタノプロスト点眼液2mLを正確に量り、希釈液3(0.2%酢酸溶液/アセトニトリル(48:52))を加えて正確に5mLとし、試料溶液とした。別にラタノプロスト標準品約0.03gを精密に量り、アセトニトリルを加えて溶かし、正確に30mLとし、標準原液とした。標準原液4mLを正確に量り、アセトニトリルを加えて正確に20mLとした。この液2mLを正確に量り、希釈液3を加えて正確に20mLとし、標準溶液とした。試料溶液及び標準溶液50μLにつき、次の条件で液体クロマトグラフィーにより試験を行い、ラタノプロストのピーク面積Atl及びAslを自動積分法により測定した。
・試料溶液中のラタノプロストの量(対表示率%)
=ラタノプロスト標準品の秤取量(mg)×100/30×ラタノプロスト標準品の純度×Atl/Asl
・Atl:試料溶液のラタノプロストのピーク面積
・Asl:標準溶液のラタノプロストのピーク面積
検出器:紫外吸光光度計(測定波長:210nm)
カラム:内径4.6mm、長さ250mmのステンレス管に5μmの液体クロマトグラフィー用
オクタデシルシリル化シリカゲルを充填した。
(TSK-GEL,ODS-80TS QA,4.6mm×250mm、5μm、東ソー株式会社製)
カラム温度:40℃付近の一定温度
移動相A:0.2%酢酸溶液/アセトニトリル(48:52)
移動相B:100%アセトニトリル
移動相の送液:移動相A及び移動相Bの混合比率を表8のように変えて直線濃度勾配制御した。
WO99/22715において、チモロールマレイン酸塩及びソルビン酸が配合された点眼液をウサギの眼に点眼した場合、チモロールマレイン酸塩の点眼液よりも房水へのチモロールの移行性が亢進することが記載されている。
実施例1及び比較例15(表9)をウサギの左右眼に投与し、最高濃度到達時点(Tmax)である0.5時間において、房水中チモロール濃度を測定した。実施例1及び比較例15の投与後0.5時間における房水中チモロール濃度(平均値±標準偏差、n=10)はそれぞれ3491±1448ng/mL及び3239±1675ng/mLを示し、統計学的に有意な差は認められなかった。
以上のことから、Tmaxにおいて実施例1及び比較例15を投与後の房水中チモロール濃度は同程度であり、ラタノプロストが添加されても、ソルビン酸によるチモロールの移行性が亢進することについては影響がないことが確認された。
Claims (20)
- ラタノプロスト、界面活性剤、及び炭素数3~10の脂肪族モノ-もしくはジ-カルボン酸又はその塩を含有する、水性点眼剤。
- さらに、チモロール又はその塩を含有する、請求項1に記載の水性点眼剤。
- 前記炭素数3~10の脂肪族モノ-もしくはジ-カルボン酸がソルビン酸である、請求項1または2に記載の水性点眼剤。
- 前記界面活性剤が、非イオン性界面活性剤である、請求項1~3のいずれかに記載の水性点眼剤。
- 前記非イオン性界面活性剤が、ポリオキシエチレン硬化ヒマシ油類、ポリオキシエチレン脂肪酸エステル類、ポリオキシエチレンソルビタン脂肪酸エステル類、ポリオキシエチレンアルキルエーテル類、ポリオキシエチレンアルケニルエーテル類、ポリオキシエチレンアルキルフェニルエーテル類、ポリオキシエチレンアルケニルフェニルエーテル類及びポリオキシエチレンアルキニルフェニルエーテル類からなる群より選択される、請求項4に記載の水性点眼剤。
- 前記非イオン性界面活性剤が、チロキサポール、ポリオキシエチレン(20)ソルビタンオレイン酸エステル、ポリオキシエチレン硬化ヒマシ油40及びモノステアリン酸ポリエチレングリコール(40)からなる群より選択される、請求項4に記載の水性点眼剤。
- 前記界面活性剤の濃度が、約0.01(w/v)%~約0.1(w/v)%である、請求項1~6のいずれかに記載の水性点眼剤。
- 前記界面活性剤の濃度が、約0.03(w/v)%~約0.1(w/v)%である、請求項1~6のいずれかに記載の水性点眼剤。
- 樹脂製容器に充填されている、請求項1~8のいずれかに記載の水性点眼剤。
- 前記樹脂製容器が、置換されていてもよいポリオレフィン製容器又はポリテレフタル酸エステル製容器である、請求項9に記載の水性点眼剤。
- 前記樹脂製容器が、ポリエチレン製容器、ポリプロピレン製容器、ポリフッ化ビニリデン製容器又はポリエチレンテレフタレート製容器である、請求項9に記載の水性点眼剤。
- 界面活性剤、及び炭素数3~10の脂肪族モノ-もしくはジ-カルボン酸又はその塩を含有させることにより、水溶液中のラタノプロストの樹脂への吸着を抑制する方法。
- 前記炭素数3~10の脂肪族モノ-もしくはジ-カルボン酸がソルビン酸である、請求項12に記載の方法。
- 前記界面活性剤が、非イオン性界面活性剤である、請求項12又は13に記載の方法。
- 前記非イオン性界面活性剤が、ポリオキシエチレン硬化ヒマシ油類、ポリオキシエチレン脂肪酸エステル類、ポリオキシエチレンソルビタン脂肪酸エステル類、ポリオキシエチレンアルキルエーテル類、ポリオキシエチレンアルケニルエーテル類、ポリオキシエチレンアルキルフェニルエーテル類、ポリオキシエチレンアルケニルフェニルエーテル類及びポリオキシエチレンアルキニルフェニルエーテル類からなる群より選択される、請求項14に記載の方法。
- 前記非イオン性界面活性剤が、チロキサポール、ポリオキシエチレン(20)ソルビタンオレイン酸エステル、ポリオキシエチレン硬化ヒマシ油40及びモノステアリン酸ポリエチレングリコール(40)からなる群より選択される、請求項14に記載の方法。
- 前記界面活性剤を、約0.01(w/v)%~約0.1(w/v)%となるように含有させる、請求項12~16のいずれかに記載の方法。
- 前記界面活性剤を、約0.03(w/v)%~約0.1(w/v)%となるように含有させる、請求項12~16のいずれかに記載の方法。
- 前記樹脂が、置換されていてもよいポリオレフィン又はポリテレフタル酸エステルである、請求項12~18のいずれかに記載の方法。
- 前記樹脂が、ポリエチレン、ポリプロピレン、ポリフッ化ビニリデン又はポリエチレンテレフタレートである、請求項12~18のいずれかに記載の方法。
