Abstract

L'invention porte sur un procédé pour induire une différenciation de cellules iPS en cellules parenchymateuses hépatiques. Ledit procédé fournit des cellules parenchymateuses hépatiques sans danger, suffisamment fonctionnelles, et peut produire de grandes quantités. Le procédé fourni comprend : (a) une étape dans laquelle des cellules iPS sont cultivées dans une culture en suspension et des corps embryoïdes sont formés; (b) une étape dans laquelle 10 à 15 corps embryoïdes par puits sont amenés à adhérer à une plaque recouverte de gélatine et cultivés, induisant des cellules d'endoderme embryonnaires; et (c) une étape dans laquelle HFG et DMSO sont ajoutés et les cellules d'endoderme embryonnaires sont cultivées sur un substrat de membrane basale, induisant une différenciation en cellules parenchymateuses hépatiques.
