まず図10A、10Bを参照しながらセンサチップを適用したイムノクロマト計測の基本的な放熱構造について説明する。図10Aは放熱構造の断面図、図10Bは平面図を示している。生体試料保持担体203には、計測対象となる生体試料に対する抗体が固定化されている。生体試料保持担体203上の抗体固定化部204とセンサチップセンサ部202とが一致するようにセンサチップ201を配置し、センサチップ201には熱拡散媒体205を密着させる。(図では見易いように、少し間を空けている。)熱拡散媒体205は、センサチップ201と生体試料保持担体203だけでなく、生体試料保持基板211にも密着している。これにより生体試料保持担体203、センサチップ201、熱拡散媒体205を熱的に接触させ、センサチップ201より生じる熱を熱拡散媒体により拡散させる。添加部208より添加された生体試料および試薬は、生体試料保持担体203中を拡散し、吸収パッド206により吸収される。これにより生体試料および試薬は、抗体固定化部204上の抗体と生体試料保持担体203の特性に依存する流速で反応する。さらに未反応な試薬および生体試料は速やかに吸収パッド206により吸収される。
センサと信号処理回路の集積型
集積型センサを有するPOCTデバイスに放熱構造を適用した計測装置の実施例を図2Aに示す。計測装置120の主要な構成要素は集積化センサチップ105と外部の制御器209、生体試料保持担体102、生体試料保持担体102中に形成された抗体固定部104および熱拡散媒体103である。計測にあたっては検出対象物に特異的に結合する2種の抗体を使う(第1抗体と第2抗体)。検出対象物を含む試料溶液が生体試料保持担体102に滴下されると検出対象物が固定化抗体(第1抗体)に捕捉される。ここに化学発光を触媒する酵素で修飾された第2抗体が滴下されると、第1抗体―検出対象物-第2抗体からなるサンドイッチ構造が形成され、抗体固定部104には検出対象物の濃度に応じて酵素が集積される。ここで生体試料保持担体102に該酵素が触媒作用を及ぼす化学発光基質を流すと、抗体固定部104において化学発光が生じ、集積化センサチップ105上のセンサ(本実施例ではフォトダイオード)202によって検出対象物に応じた化学発光が検出される。この実施例では集積化センサチップ105への電力供給および信号の授受は有線106によって行われる。図2Bは計測装置の機能ブロック図を示す。集積化センサ105は制御器209からの制御コマンドによって制御される。制御コマンドはインターフェースブロック213によって復調され、制御論理回路ブロック212によって復号化されてセンサインタフェース(センサIF)を通してセンサアナログ回路ブロック107を制御する。生体試料保持担体102の一部である抗体固定部104には検出対象物に特異的に結合する抗体があらかじめ固定されている。生体試料保持担体102に試料、続いて発光基質がセンサ202によって検出された信号はAD変換器(ADC)とアンプからなるセンサアナログ回路ブロック107により増幅された後、ディジタル電気信号に変換される。ディジタル化された検出信号は制御論理回路ブロック212で符号化され、インターフェースブロック213において変調されて、外部制御装置209に送信される。

集積型センサを有するPOCTデバイスに放熱構造を適用した計測装置の実施例として電磁的エネルギーの供給および制御コマンド、計測データの送受信の手段として無線を用いたPOCTデバイスに放熱構造を適用した計測装置121を図3Aに示す。集積化センサチップ108、生体試料保持担体102、リーダーコイル109、リーダ210、PC217から構成される。ここで図2A、2Bにおける外部制御器209のセンサチップ制御機能はリーダ210とPC217が担う。図3Bに機能ブロック図を示す。集積化センサチップ108はリーダ210を介したPC217からの制御コマンドによって制御される。制御コマンドはセンサチップに接続されたチップコイル216から共振回路ブロック215を経てRF回路ブロック214で復調、制御論理回路ブロック212によって復号化されてセンサインタフェース(センサIF)を通してセンサアナログ回路ブロック107を制御する。生体試料保持担体102の一部である抗体固定部104には検出対象物に特異的に結合する抗体があらかじめ固定されている。図3Aでは抗体は生体試料保持担体102の抗体固定部104にだけに固定化されている様に描いたが、実際にはセンサチップ108のセンサ部202領域の他、表裏面および側面の一部または全面に固定されていてもよい。図3Aに示す計測装置の表面はセンサ部202を除いて保護膜が形成されており、保護膜上に固定された抗体は計測装置の動作に有意な影響を与えないからである。生体試料保持担体102に試料、続いて発光基質がセンサ202によって検出された信号はAD変換器(ADC)とアンプからなるセンサアナログ回路ブロック107により増幅された後、ディジタル電気信号に変換される。ディジタル化された検出信号は制御論理回路ブロック2121で符号化され、インターフェースブロック213において変調されて、外部制御装置209に送信される。制御論理回路ブロック212には複数のセンサチップの中から特定のセンサチップを特定するための認識番号(UID:Unique Identifier)を格納または生成するUID回路が設けられている。各センサチップ108は異なるUIDを有している。これにより、一組のPC217、リーダ210により複数のセンサチップ108を制御することが出来る。図3Cに複数のセンサチップを一組のPC217、リーダ210を用いて同時に計測する計測装置122を示す。UIDは電磁波、磁場変化あるいは電場変化のいずれかにのせてPC217からリーダ210を経て各センサチップ108に送信される。これはコイル109の通信範囲内に置かれた複数のセンサチップ108a、108bに到達し、各センサチップ108に形成されたアンテナ216に受信され、整流検波、復調回路を経て各センサチップ108内にあらかじめ書き込まれた固有のUIDと照合される。照合は各センサチップ108の制御論理回路ブロック212内の照合回路において行われる。センサチップ108内の各回路ブロックやセンサで消費される電力は、リーダ210から送られる電磁波、磁場変化あるいは電場変化のいずれかを装置上のチップコイル216によって受け取られ、RF回路ブロック214内の整流回路と平滑回路、電圧レギュレータによって構成される直流電源から供給される。
図11A、11Bを参照して実施例4を説明する。光センサを搭載したテスト用センサチップ308は、光センサ部307が固定化された抗原302に密着するように配置する。生体試料保持担体である膜部材301は抗原抗体反応および酵素による化学発光反応の場となっており、センサチップから生じる熱の影響を受けにくい環境であることが望ましい。そこで図11Aに示すようにセンサチップ裏面(光センサ部の反対側)312に熱伝導シート309を密着させ、熱伝導シート309の一部を生体試料保持基板であるスライドガラス310に接触させる。これによりセンサチップより生じる熱は熱伝導シート309を介してスライドガラス310より効率的に放熱され、生体試料保持担体である生体試料保持担体301側への熱拡散が抑制される。
図11A、11Bを参照しながら実施例5について説明する。図11A、11Bにおいて生体試料保持担体301を支持基板となるスライドガラス310上に配置し、生体試料保持担体301上にラップフィルム306を配置し、その上にセンサチップ308、313を配置している。熱伝導シート309はセンサチップ308、313とスライドガラス310に密着しており、生体試料保持担体301とは熱的に接している。それぞれの熱伝導率は、生体試料保持担体301(PES):0.18W/m・K、ラップフィルム306(主成分ポリプロピレン/ナイロン):0.17~0.24W/m・K、センサチップ308、313(主成分シリコン):168W/m・K、スライドガラス310(一般ガラス):1.0W/m・K熱伝導シート309(主成分シリコンゲル):0.8W/m・Kであるので、センサチップ308、3131より生じた熱は熱伝導シート309を伝って熱伝導率の高いスライドガラス310より効率的に拡散される。ここでは支持基板310としてガラス基板を適用しているが、熱伝導シート309より熱伝導率が高い材質であれば支持基板を熱拡散媒体として熱伝導シート309と同様の放熱効果が得られる。
複数センサ使用
図11A、11Bを参照しながら実施例6について説明する。図11A、11Bではテスト部(抗体固定部)302とブランク部303の間から計測対象サンプル(生体試料)および試薬類を添加している。図11Aではテスト部302およびブランク部303における化学発光反応を同一条件にすることを目的に、添加部304から各部までの距離(拡散条件)を同一に設計している。ここで設計する距離は、適用している化学発光基質(CDP-Star)のメンブレンにおける拡散距離に依存する。図11A、11Bではメンブレンの多孔質構造の表面を疎水化して発光基質(CDP-Atar)のメンブレン内における拡散を促進させるエンハンサー(Nitro Block II,Tropix)を使用する条件において、添加部304からテスト部302および添加部304からブランク部303までの距離範囲(拡散範囲)は0~5mm以内である。計測対象サンプル(生体試料)および試薬類の添加部位置は、テスト部302およびブランク部303までの距離条件が同一であればテスト部302とブランク部303の間以外の場所であってもよい。
熱拡散媒体と一体化
図7を参照しながら実施例7について説明する。センサチップ101は自身の発熱を拡散するために熱拡散媒体114と熱的に接している必要がある。熱拡散媒体114による効果は、発熱源と間接的に接するより直接的に接する場合に効果的であり、その接触面積が大きいほどより効果が得られる。従って図7に示すように熱拡散媒体114にセンサチップ101を組み込んだ構造は、より効果的な構造である。

図17A、17B、図18A、18B、図19A、19Bを参照しながら実施例8について説明する。図17A、17Bに樹脂基板を用いたセンサチップによるイムノクロマト計測構造を示す。図17Aには溶液添加部409から見た断面図を示し、図17Bには平面図を示す。図17A、17Bで適用している樹脂基板404の材質は環状ポリオレフィン(COC、熱伝導率:0.21W/m・K)である。樹脂素材は加工が容易なことから抗体固定部403直下にテスト用センサチップ401と抗体が固定化されていない領域にブランク用センサチップ402を樹脂基板404に埋め込んだ構造を適用している。樹脂基板404上には、溶液添加部409から添加した生体試料および試薬を保持し且つ、効率的な反応場を形成する流路406を導入している。(図では見易いように、間を空けている。)流路406は、高さ0.02mm、幅5mm、長さ10mmの流路を形成し、樹脂基板404に埋め込んだテスト用センサチップ401およびブランク用センサチップ402上に流路を形成している。流路406の溶液添加部409側にはサンプルパッド407、溶液排出側には吸収パッド408を配置している。サンプルパッド407には図22Aに示すように酵素標識した二次抗体を含むコンジュゲートパッド410を用いても良い。また図22Bのようにコンジュゲートパッド410を積層しても良い。溶液排出側に配置している吸収パッド408は、添加する生体試料および試薬の過剰分の吸収に加え、溶液の流速を制御する働きを持っている。吸収パッド408の材質、規格、配置位置を変えることで流路内を流れる溶液の流速を制御する効果が得られる。吸収パッド408の代わりにポンプを付けて溶液の排出および流速の制御を行っても良い。樹脂基板404下には、センサチップ401、402を樹脂基板404に密着させるためにセンサチップ支持基板410を配置している。図17A、17Bではこのセンサチップ支持基板410に樹脂基板404と同じCOC樹脂を適用しているが、これより熱伝導率が高い材質であればより効率的な放熱効果が得られる。図18A、18Bに温度センサを搭載したセンサチップを用いて、図17A、17Bに示す樹脂基板を適用した計測構造におけるセンサチップ自身の温度変化を示す。センサチップ401、402にリーダーコイル405から電力を供給し始めると、センサチップ401、402の発熱により室温26℃から徐々にセンサチップ温度が上昇し始める。センサチップ温度が一定になりフォトダイオード暗電流が安定化する温度(T1)は33℃であった。続いて発光基質溶液を添加すると溶液温度に依存して温度が低下し、発光反応中(1000~1800秒の平均)温度(T3)は30.4℃に低下した。このように樹脂基板404とセンサチップ401、402が熱的に接触している状態ではセンサチップ401、402の室温からの温度上昇は7℃に抑えられ、溶液添加による温度変動(T4=T1-T3)も2.6℃と小さくなっている。以上の結果から、樹脂基板404は熱拡散媒体と同様に放熱効果を有する材質であることが示される。
図20に示すラップフィルムによる発光基質溶液の拡散促進および均一性の向上効果を参照しながら実施例9について説明する。図20には、図11に示す構造において生体試料保持担体とした膜部材307上にラップフィルム312を配置し、ラップフィルム312の上にセンサチップ301、304を配置して物理的に分離した場合と膜部材307上に直接センサチップ301、304を配置(ラップフィルム312を挿入しない)した場合を比較したデータを示す。図20A、20BではhCG抗原濃度0.8ng/mlにおけるセンサチップイムノクロマト計測を実施して膜部材上の発光基質の分布をCCDカメラにて観察した。ラップフィルムを配置しない場合の図20Aでは、センサチップの辺縁に発光基質が滞留してしまい非常に高いバックグラウンドが生じている。一方、ラップフィルムを膜部材上に配置した場合の図20Bでは、センサチップ辺縁で観察された滞留現象が解消されている。また、図20Cに示すhCG抗原濃度0.1ng/mlにおけるセンサチップイムノクロマト計測データでは、ラップフィルムを配置することにより信号強度が増加している事が示されており、これらの結果からラップフィルムによる発光基質の拡散促進効果を確認した。
図12A、B、C、D、Eを参照しながら実施例10について説明する。図12B、Dでは熱拡散媒体の一部を更に熱伝導率の高いアルミニウムに接触させた場合の放熱効果について調査した。具体的には、図12Bは熱伝導シートをセンサチップとスライドガラスに密着させ、さらに熱伝導シートの末端をアルミ箔に密着させた場合のセンサチップの温度変化を調べた結果を示している。熱伝導シートをαゲルに変え同様にしてセンサチップの温度変化を調べた結果を図12Dに示す。熱伝導シートだけ図12Aに比べ、熱伝導シートの一端をアルミ箔に密着させた場合の図12Bではセンサチップの温度が一定になる温度(T1)が6.9℃低下し、センサチップの温度変動幅(T3)も0.3℃低下した。この結果から熱拡散媒体の一部を熱伝導率の高いアルミニウムに密着させることは、センサチップから生じる熱をより効率的に拡散させる効果があることが確認された。ここでは、アルミニウムを適用したが使用する熱拡散媒体より高い熱伝導率を有する材質(金、銀、銅、鉄、白金、水晶など)であれば効率的な放熱効果が見込まれる。
図11に示すように熱拡散媒体309は、センサチップ308、313からの発熱をなるべく生体試料保持担体301に伝えないように熱を拡散することを目的としているため、生体試料保持担体301より大きい熱伝導率を有する材質であることが望ましい。図11では生体試料保持担体301として膜部材(PESメンブレン)を適用しており、熱伝導率は0.18W/m・Kである。従ってセンサチップ308、313を用いた計測における熱拡散媒体309としては熱伝導率が0.2W/m・K以上であることが望ましい。
センサチップを適用したイムノクロマト計測において算出している信号強度とは、テスト部における発光強度からブランク部における発光強度(フォトダイオード暗電流)を差し引いた値である。従ってテスト部とブランク部における計測条件は同一であることが前提となる。図11に示すようにテスト部302とブランク部303に配置したセンサチップ308、313の位置は添加部304から同じ距離に配置している。センサチップ308、313上に配置している熱伝導シート309についても、同じ大きさ(2.5mm×25mm)に調製したものをセンサチップ308、313上に配置している。以上のように熱伝導シート309を適用した放熱構造は、テスト部302とブランク部303における熱拡散効率が同一条件であることを特徴とする。
図11A、11Bを参照しながら実施例13について説明する。センサチップ308、313への電力供給と通信は、生体試料保持担体の下側に設置したリーダーコイル311とチップコイルとの誘導結合によってリーダから供給される形態が望ましい。図11A、11Bではシリコンゲルを主成分とする熱伝導シート309を適用している。図12A、B、C、D、E~図16に示す通りシリコンゲルを主成分とする熱拡散媒体については、図11A、11Bに示す構造においてセンサチップ308、313への電力供給および通信に対する影響は認められない。
実施例1、実施例10、実施例13において熱拡散媒体の材料としてシリコンゲルやαゲルを使ったセンサチップ放熱構造を説明した。一般に金属など電気伝導度の高い材料は熱伝導率が高いが、無線通信における搬送波の伝播を阻害するので好ましくない。前述のシリコンゲルの電気伝導度は5.0×1012Ω・cm程度であり、搬送波の伝播は阻害されない。一般に熱拡散媒体としてシリコンゲルのほか、電気伝導度が1.0×106Ω・cm以下の材料を用いることにより無線通信を維持しながら熱拡散の効果を得ることができる。
実施例14以外の実施例として図21に示すように高透磁率材料705を用いてよい。高透磁率材料としては、Fe-Ni合金、フェライト、などが挙げられる。この構造により、電気伝導度の高い熱拡散媒体704を使った場合でも、センサチップ701のコイルに磁力線を十分に貫通させることができる。
図13を参照しながら実施例16について説明する。図13に実施例1に従って作成したhCG抗原の検量線を示す。図13で生体試料保持担体として適用したPESメンブレンは、共有結合によるタンパク質の固定化に適した表面修飾が施されている。同様に図17A、17Bに示す樹脂基板404に関しても、タンパク質無吸着処理をすることで生体試料保持担体として適用している。図18A、18Bに樹脂基板を用いた実施例として温度センサを搭載した温度センサチップによる温度の計測例を示す。さらに図19A、19Bに光センサを搭載した光センサチップを用いたイムノアッセイ結果を示す。計測対象としてhCG抗原(濃度500mIU/ml)を用いている。図19A、19Bではタンパク質の固定化に適した表面修飾を施した環状ポリオレフィン(COC)樹脂505を生体試料保持担体として適用している。抗hCG-αサブユニット抗体を固定し、実施例1と同様にアルカリフォスファターゼ修飾した抗hCG-βサブユニット抗体を用いたサンドイッチ法によるイムノアッセイを実施している。樹脂基板修飾面の反対側には凹部を設けてあり、センサチップ501、502は光センサ部503、504を修飾面に向けて埋め込んである。図19Aにテスト用センサチップ501およびブランク用センサチップ502の樹脂基板配置図を示す。樹脂基板上のテスト部506にはセンサチップ光センサ部に一致する位置に抗体が固定化されている。テスト部506とブランク部507の間には、テスト部で生じる発光のクロストークを防止するために、光を吸収または遮断する領域508を設けている。図19Bは、図19Aに示す構造を用いて実施したhCG抗原濃度500mIU/mlにおけるイムノアッセイ結果である。横軸は時間、縦軸は光センサチップより送信されたフォトダイオード出力値を示している。テスト509はテスト部からの信号をセンサチップ501で計測した結果であり、テスト部506における発光が検出できることを示している。
図12A、B、C、D、Eおよび図19A、19Bでは、生体試料保持担体に抗体タンパク質を固定化し、生体試料として抗原タンパク質を用いたイムノクロマトアッセイを実施したが、同様に血液や血清などを生体試料に用いたイムノクロマトも可能である。さらに、細胞や組織の粗抽出液、タンパク分画液を生体試料に用いることにより、新規タンパク質および機能未知なタンパク質のスクリーニングも可能である。また、固定化するタンパク質として、ある疾患に特異的な標的タンパク質を用いた場合には、生体試料に低分子化合物を用いて新薬候補リード化合物のスクリーニングも可能である。
実施例1では熱伝導シートによるセンサチップ熱の拡散作用が、センサチップのフォトダイオード暗電流の安定化に効果的であることを説明した。図12A、C、Eでは熱拡散媒体(熱伝導シート、αゲル)によるセンサチップ熱の拡散効果を示している。図12Eに示すようにセンサチップより生じる熱に対して何も対策をしない場合(放熱対策無)ではセンサチップの温度変動幅(T4)は16.8℃である。この放熱対策をしないセンサチップイムノクロマト計測においては、図14に示すようにセンサチップのフォトダイオード暗電流にバラツキが生じていた。一方、図12Aに示すようにセンサチップに熱伝導シートを密着させてセンサチップ熱を拡散させた場合では、センサチップの温度変動幅(T4)は8.1~8.4℃まで小さくなり、この放熱構造を適用したセンサチップイムノクロマト計測では、図15、図16に示すようにセンサチップのフォトダイオード暗電流の安定化が認められた。以上の結果からセンサチップを適用する計測においては、反応前後の温度変動幅(T4)が10℃以下であることが望ましい。
実施例8で説明した通り、樹脂は表面にタンパク質無吸着処理をすることで生体試料保持担体として適用できる。また、図17A、17B~図19A、19Bで適用しているCOC樹脂の熱伝導率は0.21W/m・Kで図18A、18Bに示すように樹脂基板は熱拡散媒体と同様の効果を有している。従って、樹脂基板を適用した場合には生体試料保持担体と放熱媒体が一体化した構造となる。

Claims (20)
- 固定化試料を保持する保持担体と、
測定対象の試料と前記固定化試料との反応を検出するセンサと、
前記センサから発生した熱を拡散する熱拡散媒体を有し、
前記熱拡散媒体は、前記センサに熱的に接触していることを特徴とする試料分析チップ。 - 前記センサは、信号検出部と信号処理部を有することを特徴とする請求項1記載の試料分析チップ。
- 前記センサはさらに無線通信部を有することを特徴とする請求項2記載の試料分析チップ。
- 前記熱拡散媒体は、前記保持担体又は/及び前記センサと物理的な接触をすることにより熱的に接触していることを特徴とする請求項1記載の試料分析チップ。
- 前記保持担体を保持する基板を有し、前記熱拡散媒体は、前記基板に接触していることを特徴とする請求項1記載の試料分析チップ。
- 前記センサとして、試料測定用のセンサとブランク用のセンサを備え、前記試料測定用のセンサと前記ブランク用のセンサの間には、前記測定対象の試料を添加させる溝を有することを特徴とする請求項1記載の試料分析チップ。
- 前記センサは、前記熱拡散媒体に組み込まれていることを特徴とする請求項1記載の試料分析チップ。
- 前記保持担体は、前記センサから発生した熱を拡散する前記熱拡散媒体であることを特徴とする請求項1記載の試料分析チップ。
- 前記センサと前記保持担体との間に、添加される前記測定対象の試料を拡散する層を有することを特徴とする請求項1記載の試料分析キット。
- 前記熱拡散媒体は、アルミ層を含むことを特徴とする請求項1記載の試料分析チップ。
- 前記熱拡散媒体は、0.2W/m・K以上の熱伝導率を有することを特徴とする請求項1記載の試料分析チップ。
- 前記熱拡散媒体は、前記センサと前記ブランク用のセンサから発生した熱を同等に拡散することを特徴とする請求項1記載の試料分析チップ。
- 前記熱拡散媒体は、シリコンゲルを含むことを特徴とする請求項1の試料分析チップ。
- 前記熱拡散媒体は、電気抵抗率100Ωcm以上の材質であることを特徴とする請求項1記載の試料分析チップ。
- 前記センサと前記熱拡散媒体との間に高透磁率材料の層を有することを特徴とする請求項1記載の試料分析チップ。
- 前記保持担体は、半導体、多孔質膜、ガラス、樹脂のいずれかであることを特徴とする請求項1に記載の試料分析チップ。
- 前記測定対象の試料は、生体高分子、低分子化合物のいずれかであることを特徴とする請求項1記載の試料分析チップ。
- 前記反応の前後において、前記保持担体の温度変化が、10℃以下であることを特徴とする請求項1記載の試料分析チップ。
- 前記保持担体は、0.2W/m・K以上の熱伝導率を有することを特徴とする請求項1記載の試料分析チップ。
- 固定化試料を保持する保持担体と、測定対象の試料と前記固定化試料との反応を検出するセンサと、前記センサから発生した熱を拡散し、前記センサに熱的に接触した熱拡散媒体とを有する試料分析チップと、前記試料分析チップと信号を授受する外部制御装置と、を備えたことを特徴とする計測システム。
