脂溶性化合物をリポソームへ封入する場合、比較的容易に高い封入率を達成できるが、アムホテリシンB(リポソーム医薬品アンビゾームの主薬)のような膜親和性の非常に高い化合物の場合を除き、一般的に血漿中での保持安定性は低く、体内動態が十分に改善されることは難しい。水溶性化合物のリポソームへの封入方法にはリピドフィルム法(ボルテックス法)、逆相蒸発法、界面活性剤除去法、凍結融解法やリモートローディング法(pH勾配法、イオン勾配法)等様々な方法がある。しかしながら、100%近い高い封入率が得られるのはリモートローディング法のみであり、その他の方法では5~30%程度の封入率しか得られない。
pH勾配法によりリポソームに封入される化合物の一例として、例えば、ドキソルビシン(DOX、pKa:8.2)が挙げられる。pH4の緩衝液によりリポソーム溶液を調製した後に、リポソーム外相をpH7の緩衝液に置換する。このリポソーム溶液にDOXを加えた場合、加えられたpH7の溶液中では分子型のDOXは脂溶性となるため、水相ではなくリポソーム膜に移行する。さらにリポソーム膜に移行したDOXがpH4のリポソーム内相と接触した場合、イオン型となりリポソーム内相に溶け込んでいく。このように、解離平衡の移動によってDOXはリポソーム外相から内相に輸送される(非特許文献1、非特許文献2、および特許文献1を参照。)。
非特許文献3には、コレステロールフリーリポソームという特別な組成のリポソームにおいてpH勾配法を行う際に、リポソーム外相に活性化合物と共にエタノールを添加することで、活性化合物の封入率を向上させる技術が開示されている。
特許文献2には、pH勾配に加えて、リポソーム内相に銅イオンを存在させることで、活性化合物の封入率を向上させる技術が開示されている。
特許文献4には、硫酸アンモニウムによるイオン勾配に加えて、リポソーム外相に活性化合物と共にボロン酸を添加することで、リポソーム内に活性化合物を取り込む技術が開示されている。
また、特許文献5には、硫酸アンモニウムによるイオン勾配に変えて、グルクロン酸アニオンによるイオン勾配を利用して活性化合物をリポソームに取り込むことで、硫酸アンモニウムを利用した場合に比べて、活性化合物の放出速度を向上させる技術が開示されている。

「リポソーム」とは、脂質二重層により包囲された内相を有する微細閉鎖小胞を意味する。本発明において、リポソームは、小さな1枚膜リポソーム(SUV:small unilamellar vesicle)、大きな1枚膜リポソーム(LUV:large unilamellar vesicle)、さらに大きな1枚膜リポソーム(GUV:giant unilamellar vesicle)、同心円状の複数の膜を有する多重層リポソーム(MLV:multilamellar vesicle)、同心円状でなく不規則に複数の膜を有するリポソーム(MVV:Multivesicular vesicle)等を含む。
「リポソーム内相」は、リポソームの脂質二重層に包囲された水性領域を意味し、「内水相」および「リポソーム内水相」と同義で用いる。「リポソーム外相」は、リポソームが液体中に分散している場合に、リポソームの脂質二重層に包囲されていない領域(すなわち、内相および脂質二重層以外の領域)を意味する。
「リポソーム分散液」とは、リポソームを含む組成物であって、リポソーム内相に活性化合物が導入される前の組成物を意味する。
「リポソーム準備液」とは、リポソームを含む組成物であって、リポソーム内相にメシル酸エリブリンを封入するために、リポソーム外相を調整する前の組成物を意味する。
「リポソーム試薬」とは、液体状の場合、リポソーム分散液を意味し、固体状の場合は、所定の溶媒に溶解または懸濁することでリポソーム分散液を得ることのできる試薬を意味する。溶媒については後述する。後述するように、固体状のリポソーム試薬は、例えばリポソーム分散液を乾燥させることで得られる。
なお、本明細書において、固体と液体を混合するとは、固体を液体に溶解することと懸濁することとを包含するものとし、混合、溶解、懸濁は互いに交換可能に用いる。同様に、溶媒と分散媒も互いに交換可能に用いる。
また、本発明のリポソーム組成物、リポソーム分散液、リポソーム準備液、リポソーム試薬は、シクロデキストリンを実質的に含まない。シクロデキストリンを実質的に含まないとは、シクロデキストリンを添加していないことを意味する。なお、シクロデキストリンによる活性化合物の溶解度(みかけの溶解度)の向上が有意に認められる量のシクロデキストリンを含まなければよく、活性化合物の溶解度の向上が有意に認められない量を加えた場合も、本発明の実施として排除されるものではない。
さらに、本発明の好ましい態様として、リポソーム分散液におけるリポソーム分散液が、リポソーム外相に実質的にアンモニウム塩を含まないとは、リポソーム分散液のリポソーム外相にアンモニウム塩を添加していないことを意味する。なお、本発明の目的を達成できる範囲の量のアンモニウム塩を加えることが、本発明の実施として排除されるものではない。また、リポソーム準備液のリポソーム外相にアンモニウム塩を含む場合、アンモニウム塩を実質的に含まない溶液を用いてリポソーム準備液のリポソーム外相を置換または希釈することにより、アンモニウム塩を実質的に含まないリポソーム分散液を調製することができる。
本発明における活性化合物は、エリブリンまたはその薬理学的に許容される塩である(以下、「エリブリン等」と記載する場合がある。)。薬理学的に許容される塩とは、無機酸塩、有機酸塩のいずれであってもエリブリンと塩を形成するものであれば特に限定されないが、例えば、塩酸塩、硫酸塩、クエン酸塩、臭化水素酸塩、ヨウ化水素酸塩、硝酸塩、重硫酸塩、リン酸塩、過リン酸塩、イソニコチン酸塩、酢酸塩、乳酸塩、サリチル酸塩、酒石酸塩、パントテン酸塩、アスコルビン酸塩、コハク酸塩、マレイン酸塩、フマル酸塩、グルコン酸塩、サッカリン酸塩、ギ酸塩、安息香酸塩、グルタミン酸塩、メタンスルホン酸塩、エタンスルホン酸塩、ベンゼンスルホン酸塩、p-トルエンスルホン酸塩、パモ酸塩(pamoate)等が挙げられ、好ましくは、塩酸塩、硫酸塩、酢酸塩、リン酸塩、クエン酸塩、メシル酸塩等が挙げられ、より好ましくはメシル酸塩である。すなわち、本発明における活性化合物は、好ましくは、メシル酸エリブリンである。また、エリブリンの薬理学的に許容される塩としては、エリブリンと、アルミニウム、カルシウム、リチウム、マグネシウム、カルシウム、ナトリウム、亜鉛、ジエタノールアミンとの塩であってもよい。エリブリンまたはその薬理学的に許容される塩は、国際公開第99/65894号パンフレットまたは米国特許第6214865号公報(これら特許文献に記載される内容は参照により取り込まれる)に記載される化合物またはその塩であり、抗腫瘍及び抗有糸分裂活性を含めて薬理学的活性を有する。そして、エリブリンまたはその薬理学的に許容される塩は、抗腫瘍剤として、メラノーマ、線維肉腫、単球性白血病、結腸ガン、卵巣ガン、乳ガン、骨肉腫、前立腺ガン、肺ガン、及びras-転換繊維芽細胞などに抗腫瘍活性を有する。
活性化合物としては、低分子化合物等が挙げられ、中でも、抗腫瘍剤、抗菌剤、抗炎症剤、抗心筋梗塞剤、造影剤として用いられる化合物が好適である。
活性化合物は、分子量が100~2000であるものが好ましく、200~1500であるものがより好ましく、300~1000であるものがさらに好ましい。一般にこれらの範囲において、活性化合物のリポソーム膜透過性が良好であり、好適に本発明を適用できる。
活性化合物は、水溶性化合物および脂溶性化合物を含み、水または水性溶媒へ少なくとも多少とも溶解するものであれば、本発明を適用することができる。
本発明のリポソームは、膜構成成分として、リン脂質および/またはリン脂質誘導体を含むことが好ましい。リン脂質およびリン脂質誘導体としては、例えば、ホスファチジルエタノールアミン、ホスファチジルコリン、ホスファチジルセリン、ホスファチジルイノシトール、ホスファチジルグリセロール、カルジオリピン、スフィンゴミエリン、セラミドホスホリルエタノールアミン、セラミドホスホリルグリセロール、セラミドホスホリルグリセロールホスファート、1,2-ジミリストイル-1,2-デオキシホスファチジルコリン、プラスマロゲン、ホスファチジン酸等を挙げることができる。リン脂質およびリン脂質誘導体は、これらのうち1種または2種以上の組み合わせであってもよい。
リン脂質およびリン脂質誘導体における脂肪酸残基は特に限定されないが、例えば、炭素数12~20の飽和または不飽和の脂肪酸残基を挙げることができ、具体的には、ラウリン酸、ミリスチン酸、パルミチン酸、ステアリン酸、オレイン酸、リノール酸等の脂肪酸由来のアシル基を挙げることができる。また、卵黄レシチン、大豆レシチン等の天然物由来のリン脂質、および不飽和脂肪酸残基に部分的にまたは完全に水素添加した部分水素添加卵黄レシチン、(完全)水素添加卵黄レシチン、部分水素添加大豆レシチン、(完全)水素添加大豆レシチン等を用いることもできる。
リポソームを調製する際に用いるリン脂質および/またはリン脂質誘導体の配合量(モル分率)は、特に限定されないが、リポソーム膜成分全体に対して10~80%が好ましく、30~60%がより好ましい。
リポソームを調製する際に用いるこれらステロール類の配合量(モル分率)は、特に限定されないが、リポソーム膜成分全体に対して、1~60%が好ましく、10~50%がより好ましく、30~50%がさらに好ましい。
また、脂肪酸類の配合量(モル分率)は、特に限定されないが、リポソーム膜成分全体に対して、0~30%が好ましく、0~20%がより好ましく、0~10%がさらに好ましい。酸化防止剤の配合量(モル分率)は酸化防止効果が得られる量が添加されれば十分であるが、リポソーム膜成分全体の0~15%が好ましく、0~10%がより好ましく、0~5%がさらに好ましい。
機能性脂質として、血中滞留性脂質誘導体、温度変化感受性脂質誘導体、pH感受性脂質誘導体等を挙げることができる。修飾脂質として、PEG化脂質、糖脂質、抗体修飾脂質、ペプチド修飾脂質等が挙げられる。
リポソームを調製する際に用いる血中滞留性脂質誘導体の配合量(モル分率)は、特に限定されないが、リポソーム膜構成成分脂質全体の0~50%が好ましく、0~30%がより好ましく、0~20%がさらに好ましい。
上記した通り、リポソームは脂質二重層により包囲された内相を有する微細閉鎖小胞である。
理想的には、リポソームは、a)エリブリン等を内相にいったん封入した後では、エリブリン等がリポソーム外相には漏れ出ないバリア能を有していることが好ましい。また、医薬として用いる場合、リポソームは生体内で安定であることが好ましく、また、リポソームは、生体に投与した際に血中ではエリブリン等がリポソーム外相には漏れ出ないバリア能を有することが好ましい。
実用可能なレベルでこのような膜透過性を有するリポソームのための膜構成成分の組成は、当業者であれば、必要に応じて後述する実施例を参照することで、活性化合物や標的組織などに応じて適宜設定することができる(菊池寛他、「リポソームI-調製法と検定法-」、細胞工学, (1983), 2(9):pp.1136-1149および当該文献に引用される文献等参照)。
また、リポソーム組成物は固形癌などの標的組織へのターゲティングばかりでなく、血液癌などへの活性化合物の送達にも用いることができ、血中での徐放性製剤(slow release formulation)や放出制御製剤(controlled release formulation)等として用いることも可能である。
なお、本発明において、リポソームの粒子径は、動的光散乱法(準弾性光散乱法)による重量平均粒子径を意味する。ここでは動的光散乱(Dynamic Light Scattering)測定器(例えば、Malvern Instruments Ltd社製Zetasizer Nano ZSモデルや大塚電子社製ELS-8000)により測定された粒子径を示す。測定器は粒子のブラウン運動を測定し、確立された動的光散乱法理論に基づいて粒子径を決定している。
本発明によるとリポソーム組成物が提供される。リポソーム組成物は、リポソームを含み、リポソーム内相にエリブリン等をさらに含む。上記した通り、リポソーム組成物は、固体状および液体状のものを含む。リポソーム組成物が固体状のものの場合、後述するように所定の溶媒に溶解または懸濁することで、液体状のものとすることができる。また、リポソーム組成物が凍結した固体の場合には室温放置等により融解させることで、液体状のものとすることができる。
リポソーム組成物におけるリポソームの濃度および活性化合物の濃度は、リポソーム組成物の目的、剤形等に応じて適宜設定することができる。リポソーム組成物が液剤の場合において、リポソームの濃度はリポソームの構成成分である全脂質の濃度として、0.2~100mM、好ましくは1~30mMとすることができる。リポソーム組成物を医薬として用いる場合における活性化合物の濃度(用量)については後述する。リポソーム組成物におけるシクロデキストリンの量は、エリブリン等に対して0.1モル当量未満が好ましく、検出限界以下であることがより好ましい。
本発明におけるリポソームは、脂質二重層にエリブリン等が分配されていてもよい。
この溶媒(分散媒)に分散したリポソームを安定に長期間保存するには、凝集などの物理的安定性の面から、溶媒(分散媒)中の電解質を極力なくすことが好ましい。また、脂質の化学的安定性の面から、溶媒(分散媒)のpHを酸性から中性付近(pH3.0~8.0)に設定することや窒素バブリングにより溶存酸素を除去することが好ましい。
リポソーム組成物に含まれる糖または多価アルコールの濃度は特に限定されないが、リポソームが溶媒に分散した状態において、例えば、糖の濃度は、2~20%(W/V)が好ましく、5~10%(W/V)がより好ましい。多価アルコールの濃度は、1~5%(W/V)が好ましく、2~2.5%(W/V)がより好ましい。これら溶媒をリポソーム分散液におけるリポソーム外相として用いることもでき、リポソーム準備液のリポソーム外相をこれら溶媒で置換または希釈することで、リポソーム外相の溶液をこれら溶液とすることができる。
本発明によると、エリブリンまたはその薬理学的に許容される塩を含有するリポソーム組成物を製造する方法が提供される。リポソーム組成物を製造する方法は、リポソームを含むリポソーム分散液を供するステップと、前記リポソーム分散液と前記活性化合物(エリブリンまたはその薬理学的に許容される塩)とを混合するステップと、前記リポソーム分散液のリポソーム内相に前記活性化合物を導入するステップとを含む方法である。
リポソーム準備液を調製する際のアンモニウム塩を含む溶液は、いずれかのアンモニウム塩を含む溶液であれば特に限定されるものではない。
アンモニウム塩としては、例えば、塩化アンモニウム、ホウ酸アンモニウム、硫酸アンモニウム、ギ酸アンモニウム、酢酸アンモニウム、クエン酸アンモニウム、酒石酸アンモニウム、コハク酸アンモニウム、リン酸アンモニウムが挙げられ、好ましくは、硫酸アンモニウム、酢酸アンモニウム、クエン酸アンモニウム、酒石酸アンモニウム、リン酸アンモニウムであり、より好ましくは、硫酸アンモニウム、クエン酸アンモニウム、酒石酸アンモニウムであり、特に好ましくは、硫酸アンモニウムである。
これらのアンモニウム塩のうち、いずれか2種以上のアンモニウム塩を組み合わせて用いてもよい。
アンモニウム塩を含む溶液のpH調整のために、pH調整剤を使用することができる。アンモニウム塩を含む溶液中の個々のpH調整剤の濃度は特に限定されないが、好ましくは1~300mMであり、より好ましくは5~100mMである。
pH調整剤は、例えば、アルギニン、ヒスチジン、グリシンなどのアミノ酸、アスコルビン酸、安息香酸、クエン酸、グルタミン酸、リン酸、酢酸、プロピオン酸、酒石酸、炭酸、乳酸、ホウ酸、マレイン酸、フマル酸、りんご酸、アジピン酸、塩酸、硫酸などの酸、上記酸のナトリウム塩、カリウム塩、アンモニウム塩などの塩、トリスヒドロキシメチルアミノメタン、アンモニア水(アンモニア)、水酸化ナトリウム、水酸化カリウムなどのアルカリ性化合物(塩基)等が挙げられる。pH調整剤としては、好ましくは、水酸化ナトリウム、塩酸、アンモニア水、酢酸、乳酸、酒石酸、コハク酸、クエン酸、およびリン酸であり、より好ましくは水酸化ナトリウム、アンモニア水、塩酸、酢酸、クエン酸、およびリン酸であり、さらに好ましくは、水酸化ナトリウム、アンモニア水、塩酸、クエン酸、およびリン酸である。pH調整剤は、いずれか2種以上のアンモニウム塩を組み合わせて用いてもよい。また、pH調整剤として、リン酸塩緩衝液、クエン酸塩緩衝液、リン酸塩緩衝化生理食塩液等の緩衝液を用いてもよい。
リポソーム準備液として、実質的にシクロデキストリンを含まずに、リポソームを調製することで得られる溶液を用いることが好適である。また、リポソーム準備液として、リポソーム内相に塩、酸、塩基および/またはアミノ酸をさらに含有していてもよく、この場合、リポソーム内相に、活性化合物、アンモニウム塩、および酸を含有することが好ましい。アンモニウム塩としては、硫酸アンモニウムが好ましい例示として挙げられ、酸としては、クエン酸が好ましい例示として挙げられる。
リポソームの調製における各種条件(膜構成成分の量、温度など)は、リポソームの調製方法や目的とするリポソームの組成、粒子径等に応じて適宜設定することができる(前掲、菊池(1983)等参照)。
遠心分離は、例えば、遠心加速度が、好ましくは100,000g以上、より好ましくは300,000g以上で実施することができる。遠心によりリポソーム外相を置換することで、リポソーム外相の置換とあわせて、リポソームの濃縮を行うことができる。
ゲルろ過は、例えば、SephadexやSepharose等のカラムを用いて、分子量に基づいて分画することにより実施することができる。
pH調整剤としては、例えば、アルギニン、ヒスチジン、グリシンなどのアミノ酸、アスコルビン酸、安息香酸、クエン酸、グルタミン酸、リン酸、酢酸、プロピオン酸、酒石酸、炭酸、乳酸、ホウ酸、マレイン酸、フマル酸、リンゴ酸、アジピン酸、塩酸、硫酸などの酸、上記酸のナトリウム塩、カリウム塩、アンモニウム塩などの塩、トリスヒドロキシメチルアミノメタン、アンモニア水、水酸化ナトリウム、水酸化カリウムなどのアルカリ性化合物が挙げられ、好ましくは、水酸化ナトリウム、塩酸、ヒスチジン、酒石酸、コハク酸、クエン酸、およびリン酸であり、より好ましくは水酸化ナトリウム、塩酸、ヒスチジン、酒石酸、クエン酸、およびリン酸であり、さらに好ましくは水酸化ナトリウム、塩酸、ヒスチジン、リン酸である。
また、リポソーム外相のpHをリポソーム内相のpHよりも高く設定してpH勾配を付与することができる。好ましいpHの勾配は1~5であり、より好ましくは2~3である。
さらに、リポソーム外相のpHをエリブリン等のpKa付近により近づけることにより、リポソームへの封入率を高めることができる。好ましくは7.5~12.5、より好ましくは8.5~11.5、さらに好ましくは9~10.5である(メシル酸エリブリンのpKa=9.6)。
混合液を加熱する場合、一般的には、より高い温度に昇温することで、より効率的に活性化合物をリポソーム内相に導入できる。具体的には、活性化合物や用いるリポソーム膜構成成分の熱的安定性を考慮して、昇温する温度を設定することが好ましい。特に、昇温する温度は、リポソームの脂質二重膜の相転移温度以上とすることが好ましい。
昇温する温度は通常20~100℃であり、好ましくは40~80℃、さらに好ましくは45~65℃である。
外相の調整するpHは特に限定されないが、リポソームを構成するリン脂質の化学的安定性の観点から、好ましくは4~10、より好ましくは5~9、さらに好ましくは6~8の中性pHである。
すなわち、リポソーム組成物を液剤とする場合、上記導入するステップにより得られた液体状のリポソーム組成物をそのまま最終的なリポソーム組成物とすることができ、または得られた液体状のリポソーム組成物におけるリポソーム外相を調整(置換等)することで最終的なリポソーム組成物とすることができる。この際のリポソーム外相の調整は、リポソーム準備液におけるリポソーム外相の調整と同様に行うことができる。リポソーム組成物が液剤である場合、そのまま使用に供することができる。
本発明のリポソーム組成物は、医薬分野において、治療薬として用いることができる。具体的には、本発明のリポソーム組成物は抗腫瘍剤の医薬組成物とし用いることができる。
本発明によるとリポソーム組成物を調製するためのキットが提供される。キットは、医薬としてのリポソーム組成物を調製するために用いることができ、臨床現場で医者や患者自身が使用することができる。
<リポソーム内相用水溶液の調製>
硫酸アンモニウム396.4mgおよびクエン酸一水和物189.1mgを純水で溶解し、15mLにメスアップすることで200mM 硫酸アンモニウム/60mM クエン酸水溶液を作成した。リポソーム内相用水溶液は200mM 硫酸アンモニウム/60mM クエン酸水溶液2.5mLをアンモニア水でpH5.5に調整後、純水を用いて5mLにメスアップすることにより調製した。
水素添加大豆ホスファチジルコリン(Lipoid社製)317.9mg、コレステロール(Sigma社製)116.0mgおよびポリエチレングリコール2000-ホスファチジルエタノールアミン(Genzyme社製、MPEG2000-ジステアロイルホスファチジルエタノールアミン)130.4mgをクロロホルム10mLに溶解した後に正確に3本に分注し、そのうちの1本をロータリーエバポレーターでクロロホルムを減圧留去し、リピッドフィルムを作成した。得られたリピッドフィルムに、リポソーム内相用水溶液5mLを約60℃に加温して添加し、撹拌してリポソーム準備液を調製した。このリポソーム準備液を20分間超音波処理した後に、約65℃に加温したエクストルーダー(Lipex Biomembranes社製)を用いて整粒し、リポソーム準備液を得た。得られたリポソーム準備液中のリポソームの粒子径を動的光散乱法で測定したところ、いずれも90~100nmであった。
Sephadex G-50カラムを用い、得られたリポソーム準備液を0.9%塩化ナトリウム/10mM ヒスチジン水溶液(pH=7.6)で溶出することで、リポソーム外相を0.9%塩化ナトリウム/10mM ヒスチジン水溶液へと置換した。リポソーム外相を置換した後、400,000×gで30分間遠心した。遠心後、再分散し、0.9%塩化ナトリウム/10mM ヒスチジン水溶液を用いて液量を5mLに調整し、リポソーム分散液を得た。
メシル酸エリブリンを0.9%塩化ナトリウム/10mM ヒスチジン水溶液で溶解し、1mg/mLのメシル酸エリブリン溶液を得た。
10mLのガラス容器中、0.5mLのリポソーム分散液と0.5mLのメシル酸エリブリン溶液を混合し、55℃の水浴中で3分間インキュベートすることにより、リポソーム内にメシル酸エリブリンが導入されたリポソーム組成物を得た。
封入率は、以下のようにして求めた。
活性化合物が封入されたリポソーム組成物を400,000×gで30分間超遠心した。上清中の活性化合物濃度をHPLCで測定することで、リポソームに未封入の活性化合物量を定量した。封入率を下記式により計算した。
<リポソーム内相用水溶液の調製>
実施例1と同様にして、硫酸アンモニウム264.3mgおよびクエン酸一水和物126.1mgを純水で溶解し、メスフラスコを用いて10mLにメスアップすることで200mM 硫酸アンモニウム/60mM クエン酸水溶液を作成した。そのうち1mLを量り取り、アンモニア水でpHを5.5に調整後、純水で2mLにメスアップすることによりリポソーム内相用水溶液を調製した。
脂質混合物(水素添加大豆ホスファチジルコリン:コレステロール:ポリエチレングリコール2000-ホスファチジルエタノールアミン=58.6:19.2:22.2(重量比))を80mgずつ秤量し、リポソーム内相用水溶液2mLを約80℃に加温して添加し、撹拌してリポソーム準備液を調製した。このリポソーム準備液を約80℃に加温したエクストルーダー(Lipex Biomembranes社製)を用いて整粒し、リポソーム準備液を得た。
得られたリポソーム準備液を0.9%塩化ナトリウム/10mM ヒスチジン水溶液(pH=7.6)で10mLにメスアップし、400,000×gで30分間遠心した。遠心後、上清を全て廃棄した。沈殿を0.9%塩化ナトリウム/10mM ヒスチジン水溶液で再分散し、メスフラスコを用いて液量を1mLに調整し、リポソーム分散液を得た。
メシル酸エリブリン(メシル酸エリブリン)を0.9% 塩化ナトリウム/10mM ヒスチジン水溶液で溶解し、5mg/mLのメシル酸エリブリン溶液を得た。
10mLのガラス容器中、0.96mLのリポソーム分散液と0.24mLのメシル酸エリブリン溶液を混合し、60℃の水浴中で3分間インキュベートすることにより、リポソーム内にメシル酸エリブリンが導入されたリポソーム組成物を得た。
調製したメシル酸エリブリン封入リポソーム0.2mLとラット血漿1.8mLを混合し、液相インキュベーターを用いて37℃で振盪した。調製直後、振盪開始6時間後、12時間後、24時間後、48時間後、72時間後にサンプリングを行い、メシル酸エリブリンのリポソーム内残存量をHPLCで測定した。
測定結果を図1に示す。図1からわかるように血漿中でも120時間という長期間に亘ってメシル酸エリブリンを安定に保持し、徐放することが可能であることが示された。
<リポソーム内相用水溶液の調製>
硫酸アンモニウム264.3mgおよびクエン酸一水和物126.1mgを純水で溶解し、約15mLにした。水酸化ナトリウム水溶液でpHを7.0に調整後、純水で20mLにメスアップすることによりリポソーム内相用水溶液(100mM硫酸アンモニウム/30mMクエン酸)を調製した。
脂質混合物(水素添加大豆ホスファチジルコリン:コレステロール:ポリエチレングリコール2000-ホスファチジルエタノールアミン=58.6:19.2:22.2(重量比))を378mg秤量し、上記リポソーム内相用水溶液10mLを約80℃に加温して添加し、撹拌してリポソーム準備液を調製した。50nmのポリカーボネートメンブレンフィルターを装着し約80℃に加温したエクストルーダー(Lipex Biomembranes社製)でリポソーム準備液を整粒し、粒子径約80nmのリポソーム準備液を得た。
Sephadex G-50カラムを用い、得られたリポソーム準備液を0.9%塩化ナトリウム/10mMヒスチジン水溶液(pH=7.6)で溶出することで、リポソーム外相を0.9%塩化ナトリウム/10mMヒスチジン水溶液へと置換した。リポソーム外相を置換した後、約400,000×gで30分間遠心した。遠心後、96mg/mLスクロース/10mMヒスチジン水溶液(pH=7.6)で再分散し、液量を10mLにメスアップすることでリポソーム分散液を得た。
メシル酸エリブリンを96mg/mLスクロース/10mM ヒスチジン水溶液(pH=7.6)で溶解し、5mg/mLのメシル酸エリブリン溶液を得た。
10mLのガラス容器中、9.6mLのリポソーム分散液と1.2mLのメシル酸エリブリン溶液を混合し、水酸化ナトリウムを用いてpHを9.5に調整した。60℃の水浴中で3分間インキュベートすることにより、リポソーム内にメシル酸エリブリンが導入されたリポソーム組成物を得た。冷却後、塩酸を用いてpHを7.5に調整した。実施例1と同様にして封入率を測定したところ、封入率は99%であった。
<リポソーム内相用水溶液の調製>
実施例1と同様にして、100mM 硫酸アンモニウム/30mM クエン酸水溶液(pH=5.5)を調製した。
水素添加大豆ホスファチジルコリンおよびコレステロールおよびポリエチレングリコール2000-ホスファチジルエタノールアミンを以下の表1に記載の量だけそれぞれ秤量した。それぞれクロロホルム3mLに溶解した後にロータリーエバポレーターでクロロホルムを減圧留去し、リピッドフィルムを作成した。得られたリピッドフィルムに、作成したリポソーム内相用水溶液10mLを約80℃に加温して添加し、撹拌してリポソーム準備液を調製した。約80℃に加温したエクストルーダー(Lipex Biomembranes社製)を用いて整粒し、整粒したリポソーム準備液を得た。得られたリポソーム準備液中のリポソームの粒子径を動的光散乱法で測定したところ、Rp.1は77nm、Rp.2は95nm、Rp.3は79nm、Rp.4は128nmであった。
実施例1と同様にして、リポソーム分散液を得た。また、メシル酸エリブリンを0.9% 塩化ナトリウム/10mM ヒスチジン水溶液で溶解し、5mg/mLのメシル酸エリブリン溶液を得た。
10mLのガラス容器中、4.8mLの各リポソーム分散液と0.6mLのメシル酸エリブリン溶液をそれぞれ混合し、60℃の水浴中で3分間インキュベートすることにより、リポソーム内にメシル酸エリブリンが導入されたリポソーム組成物を得た。それぞれのリポソーム組成物に24.6mLの0.9% 塩化ナトリウム/10mM ヒスチジン水溶液を添加し、0.22μmのポリフッ化ビニリデン(PVDF)フィルターを用いて濾過滅菌することで、投与検体(メシル酸エリブリン濃度:0.1mg/mL)を得た。
実施例1と同様にして、封入率を測定し、いずれの処方においても90%以上であることを確認した。
PKパラメーターはnon-compartment model解析ソフトウェア(WinNonlin v.5.0.1)を用いて算出した。メシル酸エリブリンの血漿PKパラメーターおよび腫瘍組織PKパラメーターの結果を、それぞれ表2および表3に示す。
<リポソーム内相用水溶液の調製>
実施例1と同様に、100mM 硫酸アンモニウム/30mM クエン酸水溶液(pH=5.5)を調製した。
水素添加大豆ホスファチジルコリン221.8mgおよびコレステロール72.5mgおよびポリエチレングリコール2000-ホスファチジルエタノールアミン86.9mgを秤量した。クロロホルム3mLに溶解した後にロータリーエバポレーターでクロロホルムを減圧留去し、リピッドフィルムを作成した。得られたリピッドフィルムに、作成したリポソーム内相用水溶液10mLを約80℃に加温して添加し、撹拌してリポソーム準備液を調製した。約80℃に加温したエクストルーダー(Lipex Biomembranes社製)を用いて整粒し、整粒したリポソーム準備液を得た。得られたリポソーム準備液中のリポソームの粒子径を動的光散乱法で測定したところ、約90nmであった。
Sephadex G-50カラムを用い、得られたリポソーム準備液を0.9%塩化ナトリウム/10mMヒスチジン水溶液(pH=7.6)で溶出することで、リポソーム外相を0.9%塩化ナトリウム/10mMヒスチジン水溶液へと置換した。リポソーム外相を置換した後、400,000×gで30分間遠心した。遠心後、再分散し、0.9%塩化ナトリウム/10mMヒスチジン水溶液を用いて液量を10mLに調整し、リポソーム分散液を得た。
メシル酸エリブリンを0.9% 塩化ナトリウム/10mM ヒスチジン水溶液で溶解し、1mg/mLのメシル酸エリブリン溶液を得た。また、フリー体の投与検体として、0.9% 塩化ナトリウム/10mM ヒスチジン水溶液でメシル酸エリブリン溶液を希釈し、0.22μmのPVDFフィルターを用いて濾過滅菌することで、投与検体(メシル酸エリブリン濃度:0.3mg/mLおよび0.4mg/mL)を得た。
10mLのガラス容器中、1.8mLのリポソーム分散液と1.2mLのメシル酸エリブリン溶液をそれぞれ混合し、60℃の水浴中で3分間インキュベートすることにより、リポソーム内にメシル酸エリブリンが導入されたリポソーム組成物を得た。得られたリポソーム組成物を0.9% 塩化ナトリウム/10mM ヒスチジン水溶液で希釈し、0.22μmのPVDFフィルターを用いて濾過滅菌することで、投与検体(メシル酸エリブリン濃度:0.2mg/mL)を得た。実施例1と同様にして、封入率を測定し、90%以上であることを確認した。
検体投与後の平均腫瘍体積の推移結果を図2に示す。
図2に示すように、FaDuは、メシル酸エリブリンに感受性の低い細胞株であるため、フリー体では最大耐用量である4mg/kg投与でも腫瘍縮小効果は得られなかった。一方、リポソーム組成物の場合には、最大耐用量以下である2mg/kg投与でも腫瘍縮小効果が明確に認められ、これまでメシル酸エリブリンが奏功しなかった癌種に対しても非常に高い薬理効果が得られることが示された。
<リポソーム内相用水溶液の調製>
実施例1と同様に、100mM 硫酸アンモニウム/30mM クエン酸水溶液(pH=5.5)を調製した。
実施例5と同様に実施し、フリー体の投与検体(メシル酸エリブリン濃度:0.2mg/mLおよび0.3mg/mLおよび0.4mg/mL)を得た。
上述のように作製したリポソーム内相溶水溶液を用いた以外は実施例5と同様にして、リポソーム組成物(メシル酸エリブリン濃度:0.3mg/mL)を得た。実施例1と同様にして、封入率を測定し、90%以上であることを確認した。
検体投与後の平均腫瘍体積の推移結果を図3に示す。
図3に示すように、ACHNは、メシル酸エリブリンに耐性のある細胞株であるため、フリー体投与では2mg/kg投与群、3mg/kg投与群、および4mg/kg(最大耐用量)投与群のいずれにおいても検体投与開始45日後において無処置群との有意な差は認められなかった。一方、リポソーム組成物3mg/kg投与群においては腫瘍増殖抑制効果が認められ、検体投与開始45日後において無処置群およびフリー体投与群に対し有意に小さい腫瘍体積値を示した。このようにこれまでのメシル酸エリブリンではまったく治療効果が得られないような癌種に対してもリポソーム製剤化することで、腫瘍の増殖を遅延させることが可能となることが示された。
<リポソーム内相用水溶液の調製>
以下の表4に示す12種類の内相用水溶液を作成した。
脂質混合物(水素添加大豆ホスファチジルコリン:コレステロール:ポリエチレングリコール2000-ホスファチジルエタノールアミン=58.6:19.2:22.2(重量比))を120mgずつ試験管に秤量し、80℃に加温した各内相用水溶液3mLで水和した。
このリポソーム準備液を約80℃に加温したエクストルーダーを用いて整粒し、リポソーム準備液を得た。
Sephadex G-50カラムを用い、得られたリポソーム準備液を0.9%塩化ナトリウム/10mM ヒスチジン水溶液で溶出することで、リポソーム外相を0.9%塩化ナトリウム/10mM ヒスチジン水溶液へ置換した。
リポソーム外相を置換した後、400,000×gで1時間遠心し、上清を完全に除去した。沈殿物を96mg/mLスクロース/10mM ヒスチジン水溶液(pH=7.5)で約2mLとなるように再懸濁した。
得られたリポソーム分散液の粒子径を動的光散乱法で測定したところ、いずれも約80nmであった。
メシル酸エリブリンを96mg/mLスクロース/10mM ヒスチジン水溶液で溶解し、5mg/mLのメシル酸エリブリン溶液を得た。
10mLのガラス容器中、リポソーム分散液とメシル酸エリブリン溶液をメシル酸エリブリン:0.2mg/mL,総脂質濃度:16μmol/mLとなるように混合した。60℃で5分間昇温し、リポソーム内にメシル酸エリブリンが導入されたリポソーム組成物を得た。
封入率は、実施例1と同様にして測定した。結果を表4に示す。表4から分かるとおり、いずれのアンモニウム塩を内相に用いた場合でもメシル酸エリブリンの封入率が向上することが明らかとなった。特に硫酸アンモニウム、クエン酸アンモニウム、リン酸アンモニウム、酒石酸アンモニウムを用いた際に顕著に封入率が向上した。
<リポソーム内相用水溶液の調製>
実施例7と同様にして、100mM 硫酸アンモニウム/30mM クエン酸水溶液(pH=7.5)のリポソーム内相用水溶液を調製した。
上記リポソーム内相用水溶液を用いて実施例7と同様にリポソーム準備液を調製した。
Sephadex G-50カラムを用い、得られたリポソーム準備液を0.9%塩化ナトリウム/10mM ヒスチジン水溶液で溶出することで、リポソーム外相を0.9%塩化ナトリウム/10mM ヒスチジン水溶液へ置換した。
リポソーム外相を置換した後、400,000×gで1時間遠心し、上清を完全に除去した。沈殿物を96mg/mL/10mM ヒスチジン水溶液(pH=7.5)で再懸濁し、リポソーム外相を96mg/mLスクロース/10mM ヒスチジン水溶液(pH=7.5)に置換し、リポソーム分散液を得た。得られたリポソーム分散液の粒子径を動的光散乱法で測定したところ、約80nmであった。
リポソーム分散液を7本に分注し、既知量の硫酸アンモニウム(水酸化ナトリウム水溶液によりpHを7.5に調整済)を表5の各濃度になるようにリポソーム外相にそれぞれ添加し、リポソーム外相中に硫酸アンモニウムが既知濃度存在するリポソーム分散液を得た。
メシル酸エリブリンを96mg/mLスクロース/10mM ヒスチジン水溶液で溶解し、5mg/mLのメシル酸エリブリン溶液を得た。
10mLのガラス容器中、リポソーム分散液とメシル酸エリブリン溶液をメシル酸エリブリン:0.2mg/mL,総脂質濃度:16mMとなるように混合した。60℃で5分間昇温し、リポソーム内にメシル酸エリブリンが導入されたリポソーム組成物を得た。
封入率は、実施例1と同様にして測定した。結果を表5に示す。リポソーム外相中に硫酸アンモニウムが0.4mM存在するだけで封入率は顕著に低下し、10mM存在するとほとんど封入されないことが示された。
<リポソーム内相用水溶液の調製>
実施例7と同様にして、100mM 硫酸アンモニウム/30mM クエン酸水溶液(pH=7.5)のリポソーム内相用水溶液を調製した。
上記リポソーム内相用水溶液を用いて実施例7と同様にリポソーム準備液を調製した。
Sephadex G-50カラムを用い、得られたリポソーム準備液を0.9%塩化ナトリウム/10mM ヒスチジン水溶液で溶出することで、リポソーム外相を0.9%塩化ナトリウム/10mM ヒスチジン水溶液へ置換した。

その後、リポソーム分散液を7本に分注した。
メシル酸エリブリンを96mg/mLスクロース/10mM ヒスチジン水溶液で溶解し、5mg/mLのメシル酸エリブリン溶液を得た。
10mLのガラス容器中、リポソーム分散液とメシル酸エリブリン溶液をメシル酸エリブリン:0.2mg/mL,総脂質濃度:16mMとなるようにそれぞれ混合した。1M 水酸化ナトリウム水溶液を用いて表6に示すようにそれぞれリポソーム外相のpHを調整した。60℃で5分間昇温し、リポソーム内にメシル酸エリブリンが導入されたリポソーム組成物を得た。次いで、塩酸を用いて外相のpHを7.5に調整した。
封入率は、実施例1と同様にして測定した。結果を表6に示す。リポソーム外相のpHの上昇に伴いエリブリンの封入率は大幅に向上し、ほぼ100%の封入率を達成した。
<リポソーム内相用水溶液の調製>
実施例7と同様にして、100mM 硫酸アンモニウム/30mM クエン酸水溶液(pH=7.5)のリポソーム内相用水溶液を調製した。
上記リポソーム内相用水溶液を用いて実施例7と同様にリポソーム準備液を調製した。
Sephadex G-50カラムを用い、得られたリポソーム準備液を0.9%塩化ナトリウム/10mM ヒスチジン水溶液で溶出することで、リポソーム外相を0.9%塩化ナトリウム/10mM ヒスチジン水溶液へ置換した。
リポソーム分散液を4本に分注し、400,000×gで1時間遠心し、上清を完全に除去した。そのうち2本の沈殿物は96mg/mLスクロース/10mM ヒスチジン水溶液(pH=7.5)で再懸濁し、リポソーム外相を96mg/mLスクロース/10mM ヒスチジン水溶液(pH=7.5)に置換した。残りの2本の沈殿物は0.9%塩化ナトリウム/10mM ヒスチジン水溶液(pH=7.5)で再懸濁し、リポソーム外相を0.9%塩化ナトリウム/10mM ヒスチジン水溶液(pH=7.5)に置換した。得られたリポソーム分散液の粒子径を動的光散乱法で測定したところ、いずれも約80nmであった。
メシル酸エリブリンを96mg/mLスクロース/10mM ヒスチジン水溶液で溶解し、5mg/mLのメシル酸エリブリン溶液を得た。また、同様にメシル酸エリブリンを0.9%塩化ナトリウム/10mM ヒスチジン水溶液で溶解し、5mg/mLのメシル酸エリブリン溶液を得た。
10mLのガラス容器中、リポソーム分散液とメシル酸エリブリン溶液をメシル酸エリブリン:0.2mg/mL,総脂質濃度:16mMとなるようにそれぞれ混合した。リポソーム外相が96mg/mLスクロース/10mM ヒスチジン水溶液(pH=7.5)の2本のうち1本のリポソーム外相のpHを水酸化ナトリウムの添加により9.5に調整した。同様に、リポソーム外相が0.9%塩化ナトリウム/10mM ヒスチジン水溶液(pH=7.5)の2本のうち1本のリポソーム外相のpHを水酸化ナトリウムの添加により9.5に調整した。60℃で5分間昇温し、リポソーム内にメシル酸エリブリンが導入されたリポソーム組成物を得た。
封入率は、実施例1と同様にして測定した。結果を表7に示す。リポソーム外相が非電解質であるスクロースの場合と比べ、電解質である塩化ナトリウムの場合では非常に高い封入率が得られることが明らかとなった。また、電解質の効果に加え、リポソーム外相をアルカリにするpH勾配を適用することで100%の封入率を達成した。
本発明のリポソーム組成物は、エリブリンまたはその薬理学的に許容される塩の薬理作用により治療用途に用いることが好適である。
Claims (39)
- リポソームを含み、リポソーム内相に活性化合物を含むリポソーム組成物であって、活性化合物がエリブリンまたはその薬理学的に許容される塩である、リポソーム組成物。
- リポソーム組成物が固体状または液体状である、請求項1に記載のリポソーム組成物。
- リポソーム内相にアンモニウム塩をさらに含む、請求項1または2に記載のリポソーム組成物。
- 前記アンモニウム塩の濃度が10mM以上である、請求項3に記載のリポソーム組成物。
- リポソーム内相に塩、酸、塩基および/またはアミノ酸をさらに含む、請求項1~4のいずれかに記載のリポソーム組成物。
- 前記塩の濃度が1~300mMである、請求項5に記載のリポソーム組成物。
- 前記酸の濃度が1~300mMである、請求項5または6に記載のリポソーム組成物。
- 前記アミノ酸の濃度が1~300mMである、請求項5~7のいずれかに記載のリポソーム組成物。
- 前記塩基の濃度が1~300mMである、請求項5~8のいずれかに記載のリポソーム組成物。
- 前記活性化合物の濃度が0.01~300mg/mLである、請求項1~9のいずれかに記載のリポソーム組成物。
- 前記活性化合物がメシル酸エリブリンである、請求項1~10のいずれかに記載のリポソーム組成物。
- リポソーム内相に硫酸アンモニウム、クエン酸、および活性化合物をさらに含む、請求項1~11のいずれかに記載のリポソーム組成物。
- リポソーム外相に糖、電解質、および/またはアミノ酸を含む、請求項1~12のいずれかに記載のリポソーム組成物。
- リポソーム外相に糖または電解質、およびアミノ酸を含む、請求項1~13のいずれかに記載のリポソーム組成物。
- 前記糖の濃度が2~20%である、請求項13または14に記載のリポソーム組成物。
- 前記アミノ酸の濃度が1~300mMである、請求項13~15のいずれかに記載のリポソーム組成物。
- リポソーム外相にショ糖または塩化ナトリウム、およびヒスチジンを含む、請求項1~16のいずれかに記載のリポソーム組成物。
- 前記リポソーム内相にシクロデキストリンを実質的に含まない、請求項1~17のいずれかに記載のリポソーム組成物。
- リポソームが水素添加ホスファチジルコリンを含む、請求項1~18のいずれかに記載のリポソーム組成物。
- リポソームがコレステロールを含む、請求項1~19のいずれかに記載のリポソーム組成物。
- リポソームがメトキシポリエチレングリコール縮合体を含む、請求項1~20のいずれかに記載のリポソーム組成物。
- 前記メトキシポリエチレングリコール縮合体がジステアロイルホスファチジルエタノールアミン-ポリエチレングリコール縮合体である、請求項21に記載のリポソーム組成物。
- リポソームが水素添加ホスファチジルコリン、コレステロール、およびジステアロイルホスファチジルエタノールアミン-ポリエチレングリコール縮合体を含む、請求項1~22のいずれかに記載のリポソーム組成物。
- 前記水素添加ホスファチジルコリンを10~80%、前記コレステロールを1~60%、前記ジステアロイルホスファチジルエタノールアミン-ポリエチレングリコール縮合体を0~50%含む、請求項23に記載のリポソーム組成物。
- リポソームが水素添加大豆ホスファチジルコリン、コレステロール、およびポリエチレングリコール2000-ホスファチジルエタノールアミンを含む、請求項1~24のいずれかに記載のリポソーム組成物。
- 請求項1~25のいずれかに記載のリポソーム組成物を製造する方法であって、
リポソームを含む、リポソーム分散液を供するステップと、
前記リポソーム分散液と前記活性化合物とを混合するステップと、
前記リポソーム分散液のリポソーム内相に前記活性化合物を導入するステップと
を含む方法。 - 前記リポソーム分散液がリポソーム外相にアンモニウム塩を実質的に含まない、請求項26に記載の方法。
- 前記リポソーム分散液のリポソーム外相のpHが3~10である、請求項26または27に記載の方法。
- 前記リポソーム分散液のリポソーム外相のpHが7~10である、請求項26~28のいずれかに記載の方法。
- 前記pHが、前記リポソーム分散液と前記活性化合物とを混合するステップにおける前記リポソーム分酸液のリポソーム外相のpHである、請求項28または29に記載の方法。
- 前記リポソーム分散液を供するステップが、リポソームを含み、リポソーム内相およびリポソーム外相にアンモニウム塩を含むリポソーム準備液を供するステップと、
前記リポソーム準備液のリポソーム外相を置換または希釈するステップと
を含む、請求項26~30のいずれかに記載の方法。 - 前記リポソーム外相を置換または希釈するステップが、リポソーム外相のpHをリポソーム内相のpHより高くするステップである、請求項31に記載の方法。
- 前記リポソーム外相を置換または希釈するステップが、リポソーム内相のpHとリポソーム外相のpHの差を1~5とするステップである、請求項31または32に記載の方法。
- 前記リポソーム内相のpHが3~9である、請求項26~33のいずれかに記載の方法。
- 前記リポソーム内相のpHが4~9である、請求項26~34のいずれかに記載の方法。
- 前記リポソーム内相のpHが5~8である、請求項26~35のいずれかに記載の方法。
- 前記活性化合物を導入するステップにおいて、リポソーム外相が電解質を含む溶液である、請求項26~36のいずれかに記載の方法。
- 前記リポソーム分散液が、シクロデキストリンをリポソーム内相に実質的に含まない、請求項26~37のいずれかに記載の方法。
