本発明のカルシウム結合型発光蛋白質は、一般式(1)で表わされる化合物(本発明のセレンテラジン類縁体)と、カルシウム結合型発光蛋白質のアポ蛋白質とを接触させて、カルシウム結合型発光蛋白質を得ることにより、製造又は再生することができる。
(a)天然型アポ蛋白質のアミノ酸配列において1~複数個のアミノ酸が欠失、置換、挿入及び/又は付加したアミノ酸配列からなり、かつ、カルシウム結合型発光蛋白質のアポ蛋白質活性若しくは機能を有する蛋白質、
(b)天然型アポ蛋白質のアミノ酸配列に対して90%以上の同一性を有するアミノ酸配列からなり、かつ、カルシウム結合型発光蛋白質のアポ蛋白質活性若しくは機能を有する蛋白質、及び
(c)天然型アポ蛋白質の塩基配列に相補的な塩基配列からなるポリヌクレオチドとストリンジェントな条件下でハイブリダイズするポリヌクレオチドによってコードされるアミノ酸配列からなり、かつ、カルシウム結合型発光蛋白質のアポ蛋白質活性若しくは機能を有する蛋白質。
本発明で使用することができる組換えアポ蛋白質として、例えば、Shimomura, O. and Inouye, S.Protein Express. Purif. (1999) 16: 91-95に記載の組換えイクオリン、Inouye, S. and Sahara, Y. Protein Express. Purif. (2007) 53: 384-389に記載の組換えクライティン-I、又はInouye, S.J.Biochem. (2008)143: 711-717に記載の組換えクライティン-IIなどを挙げることができる。
(1)発光基質としての利用
本発明のいくつかの態様のセレンテラジン類縁体は、発光触媒の作用により発光するので、発光基質として利用できる。そこで、本発明は、本発明のセレンテラジン類縁体に、発光触媒を接触させることを含む、発光方法を提供する。ここで、「接触」とは、本発明のセレンテラジン類縁体と発光触媒とを同一の反応系に存在させることを意味し、例えば、セレンテラジン類縁体を収容した容器に発光触媒を添加すること、発光触媒を収容した容器にセレンテラジン類縁体を添加すること、又はセレンテラジン類縁体と発光触媒とを混合すること、などが含まれる。
本発明の一つの態様では、レニラルシフェラーゼが、Renilla reniformis由来のルシフェラーゼであり、例えば、配列番号14のアミノ酸配列からなるポリペプチドを含む。本発明の別の態様では、エビルシフェラーゼが、Oplophorus gracilorostris由来のルシフェラーゼであり、例えば、配列番号16のアミノ酸配列からなるポリペプチドを含む。本発明のさらに別の態様では、ガウシアルシフェラーゼが、Gaussia princeps由来のルシフェラーゼであり、例えば、配列番号18のアミノ酸配列からなるポリペプチドを含む。
上記のようにして得た本発明の発光蛋白質は、アポ蛋白質と、セレンテラジン類縁体及び分子状酸素より生成するセレンテラジン類縁体のペルオキシドとが非共有的な結合を形成したものであり、且つカルシウムイオンの作用によって発光する発光蛋白質(ホロ蛋白質)である。よって、本発明の発光蛋白質は、カルシウムイオンの検出又は定量に使用することができる。
本発明の発光蛋白質は、レポーター蛋白質としてプロモーターなどの転写活性の測定に利用することもできる。アポ蛋白質をコードするポリヌクレオチドを、目的のプロモーター又は他の発現制御配列(例えば、エンハンサーなど)に融合したベクターを構築する。前記ベクターを宿主細胞に導入し、さらに、これに、セレンテラジン又はその類縁体を接触させ、本発明の発光蛋白質に由来する発光を検出することにより、目的のプロモーター又は他の発現制御配列の活性を測定することができる。ここで、「接触」とは、宿主細胞とセレンテラジン又はその類縁体とを同一の培養系・反応系に存在させることを意味し、例えば、宿主細胞の培養容器にセレンテラジン又はその類縁体を添加すること、宿主細胞とセレンテラジン又はその類縁体とを混合すること、宿主細胞をセレンテラジン又はその類縁体の存在下で培養することなどが含まれる。また、セレンテラジン又はその類縁体は、本発明のセレンテラジン類縁体の他、前記したものが挙げられる。
本発明の発光蛋白質は、発光による検出マーカとして利用することができる。本発明の検出マーカは、例えば、イムノアッセイ又はハイブリダイゼーションアッセイなどにおける目的物質の検出に利用することができる。本発明の発光蛋白質を化学修飾法など通常用いられる方法により目的物質(蛋白質或いは核酸など)と結合させて使用することができる。このような検出マーカを用いた検出方法は、通常の方法によって行うことができる。また、本発明の検出マーカは、例えば、アポ蛋白質と目的物質との融合蛋白質として発現させ、マイクロインジェクション法などの手法により細胞内に導入し、さらに、これに、本発明のセレンテラジン類縁体を接触させることによって、本発明の発光蛋白質を生成させ、さらに、これに、セレンテラジン又はその類縁体を接触させて、前記目的物質の分布を測定するために利用することもできる。ここで、「接触」とは、細胞と本発明のセレンテラジン類縁体等とを同一の培養系・反応系に存在させることを意味し、例えば、細胞の培養容器に本発明のセレンテラジン類縁体等を添加すること、細胞と本発明のセレンテラジン類縁体等とを混合すること、宿主細胞を本発明のセレンテラジン類縁体等の存在下で培養することなどが含まれる。また、「セレンテラジン又はその類縁体」は、本発明のセレンテラジン類縁体の他、前記のものを例示することができる。
アポ蛋白質と本発明のセレンテラジン類縁体のペルオキシドとからなる複合体(本発明の発光蛋白質)は、微量のカルシウムイオンと結合するだけで発光する。よって、本発明の発光蛋白質は、アミューズメント用品の材料の発光基材として好適に使用することができる。アミューズメント用品としては、たとえば、発光シャボン玉、発光アイス、発光飴、発光絵の具等があげられる。本発明のアミューズメント用品は、通常の方法によって製造することができる。
本発明のいくつかの態様のセレンテラジン類縁体は、前記のように、発光触媒の作用により発光するので、生物発光共鳴エネルギー移動(BRET)法による分子間相互作用の原理を利用した生理機能の解析や酵素活性の解析(又は測定)等の分析方法に利用することができる。また、本発明の発光蛋白質も、生物発光共鳴エネルギー移動(BRET)法による分子間相互作用の原理を利用した生理機能の解析や酵素活性の測定等の分析方法に利用することができる。
なお、以下の実施例においてクロマトグラフィー用の混合溶媒の比率は特に説明が無い限りv/v である。
セレンテラジン類縁体(CTZ類縁体)の合成法の概要
ケトアセタールを用いた、i-セレンテラジン(i-CTZ (R1 = I, R2 = H, R3= H))、n-セレンテラジン(n-CTZ (R1, R2 = benzo, R3= H))、me-セレンテラジン(me-CTZ (R1 = CH3, R2 = H, R3 = H))、et-セレンテラジン(et-CTZ (R1 = C2H5, R2= H, R3 = H))、cf3-セレンテラジン(cf3-CTZ (R1 = CF3, R2 = H, R3 = H)、meo-セレンテラジン(meo-CTZ (R1 = OCH3, R2 = H, R3 = H))、3me-セレンテラジン(3me-CTZ (R1 =H, R2 = CH3, R3 = H))、3meo-セレンテラジン(3meo-CTZ (R1 =H, R2 = OCH3, R3 = H))、αmeh-セレンテラジン(αmeh-CTZ (R1=H, R2 = H, R3 = CH3))、及び3-イソセレンテラジン(3iso-CTZ (R1 =H, R2 = OH, R3= H))の合成法の概要は、下記に示す通りである。
CTZ類縁体の合成の際に、フラッシュカラムクロマトグラフィーは、シリカゲル(関東化学社製37563-85、silica gel 60 N(球状、中性)、粒径40-50μm)を用いて行った。ただし、CTZ類縁体の精製の際には、酸性シリカゲル(関東化学社製37562-79、 silica gel 60(球状)、粒径40-50μm)を用いた。
薄層クロマトグラフィー(TLC)は、あらかじめシリカゲルが塗布されたガラスプレート(MERCK社製1.05715, silica gel 60 F254)を用いて行った。
CTZ類縁体の純度は、高速液体クロマトグラフィー(HPLC)を用いて検定した:Agilent社製1100 シリーズHPLCシステム:測定条件、カラム:Lichrosorb(登録商標) RP-18(5μm, 4.0 mm i.d. ´ 125 mm, Merck Chemicals社製);移動相:gradient 60-100% methanol/0.1% aqueous TFA for 40 min;流速:0.45 mL/min;検出:UV 225 nm;インジェクトサンプル量:0.5 mg/5 mL in methanol/0.1% aqueous TFA = 6/4。
融点(Mp)は、ヤナコ(YANACO)機器開発研究所社製、MP-J3微量融点測定装置を用いて測定した(未補正値)。
CTZ類縁体(20 μMメタノール溶液)の紫外吸収スペクトル(UV)は、島津製作所(SHIMADZU)社製、UV-3100紫外可視近赤外分光光度計を用い、石英セル(光路長10 mm)中、25 ℃においてスキャン速度、高速の条件で測定した。
CTZ類縁体(8 μg/mLメタノール溶液)の蛍光スペクトル(FL)は、日本分光(JASCO)社製、FP-6500分光蛍光光度計を用い、石英セル(光路長10 mm)中、25 ℃において励起波長330 nm、励起側バンド幅3 nm、蛍光側バンド幅3 nm、レスポンス0.5秒、感度Medium、走査速度100 nm/minの条件で測定した。
1H NMRスペクトルは、バリアン(Varian)社製、Unity Plus 400核磁気共鳴装置を用いて測定した。13C NMRスペクトルは、日本電子(JEOL)社製、JNM-EX270核磁気共鳴装置を用いて測定した。19F NMRスペクトルは、バリアン(Varian)社製、Mercury 300核磁気共鳴装置を用いて測定した。NMRスペクトル測定用溶媒には、CDCl3又はCD3OD(いずれもCIL社製)を使用した。
化学シフト(δ)は、テトラメチルシラン((CH3)4Si)(CDCl3中での1H NMR測定;δ 0 ppm)、測定溶媒の非重水素化体由来のピーク(CD3OD中での1H NMR測定;δ 3.31 ppm、CDCl3中での13C NMR測定;δ 77.0 ppm)、又はヘキサフルオロベンゼン(19F NMR測定;δ 0 ppm)を内部標準として相対的な値として表し、結合定数(J)はHzで示した。略号s、d、t、q、m、及びbrはそれぞれ単重線、二重線、三重線、四重線、多重線、及び幅広線を表す。
赤外分光スペクトル(IRスペクトル)は、DRS-8000A拡散反射測定装置を装着した島津製作所(SHIMADZU)社製、IRPrestige-21フーリエ変換赤外分光光度計を用いて拡散反射法により測定した。
高分解能質量分析スペクトル(HRMS)は、日本電子(JEOL)社製、JMS-700を用い、電子衝撃イオン化(EI+)法、又は高速原子衝突(FAB+)法により測定した。FAB+法の際のマトリックスには、m-ニトロベンジルアルコール(NBA)又はグリセロールを用いた。
アルゴン雰囲気下、4-ヨードフェニル酢酸(11)(Chen, Q.-H. et al., Bioorg. Med. Chem. 14, 7898-7909 (2006) に記載の方法によって製造)(1.06 g, 4.05 mmol)に塩化チオニル(5.00 mL, 68.6 mmol)を加え、1.5時間半加熱還流(100 ℃)した。室温まで放冷後、減圧濃縮し、4-ヨードフェニルアセチルクロリド(12)を茶色油状の粗生成物として得た。これ以上の精製は行わず、そのまま次の反応に用いた。
アルゴン雰囲気下、上記で調整した4-ヨードフェニルアセチルクロリド(12)をテトラヒドロフラン(THF)(2 mL)及びアセトニトリル(2 mL)に溶解し、0 ℃に冷却した。これにトリメチルシリルジアゾメタンのジエチルエーテル溶液(2.0 M, 4.00 mL, 8.00 mmol)をゆっくりと加え、室温まで昇温後、一晩(14時間)攪拌した。減圧濃縮し、残渣をシリカゲルフラッシュカラムクロマトグラフィー(n-ヘキサン/ジエチルエーテル=1/1)にて精製し、1-diazo-3-(4-iodophenyl)propan-2-one(13)を淡黄色固体として得た(635 mg, 2.22 mmol, 54.9%, 2 steps)。

アルゴン雰囲気下、1-diazo-3-(4-iodophenyl)propan-2-one(13)(1.11 g, 3.88 mmol)を脱水エタノール(10 mL)に溶解し、0 ℃に冷却した。これに次亜塩素酸tert-ブチル(440μL, 3.89 mmol)を加え、0 ℃のまま1時間攪拌した。減圧濃縮し、残渣をシリカゲルフラッシュカラムクロマトグラフィー(n-ヘキサン/酢酸エチル=9/1)にて精製し、1,1-diethoxy-3-(4-iodophenyl)propan-2-one(14)を黄色油状物質として得た(758 mg, 2.18 mmol, 56.1%)。

アルゴン雰囲気下、1,1-diethoxy-3-(4-iodophenyl)propan-2-one(14)(421 mg, 1.21 mmol)を1,4-ジオキサン(2 mL)及び水(0.4 mL)に溶解した。これにセレンテラミン(Adamczyk, M. et al., Org. Prep. Proced. Int., 33, 477-485 (2001) に記載の方法によって製造)(222 mg, 801μmol)を加え、0 ℃に冷却した後、さらに濃塩酸(0.20 mL)を加え、100 ℃で一晩(15時間)攪拌した。室温まで放冷後、減圧濃縮し、残渣をアルゴン気流下でシリカゲルフラッシュクロマトグラフィー(n-ヘキサン/酢酸エチル=1/2→酢酸エチル→酢酸エチル/メタノール=20/1→10/1)で精製した。得られた固体をさらに再沈殿(n-ヘキサン/アセトン)することで、i-セレンテラジン(15,i-CTZ)を黄土色粉末として得た(45.2 mg, 84.7μmol, 10.6%)。

アルゴン雰囲気下、ジエトキシ酢酸エチル(900 μL, 5.06 mmol)のTHF溶液(15 mL)に-78 ℃にて、(2-naphtalenylmethyl)magnesium bromide(21)のジエチルエーテル溶液(0.25 M, 25.0 mL, 6.25 mmol)をゆっくりと滴下した。-78 ℃にて2.5時間撹拌した後、徐々に室温まで昇温し、15時間撹拌した。これに20%塩化アンモニウム水溶液(10 mL)を加え、酢酸エチルで抽出した(×3)。有機層を水(×1)と飽和食塩水(×1)で順次洗浄し、無水硫酸ナトリウムで乾燥した。ろ過後、減圧濃縮し、残渣をシリカゲルフラッシュカラムクロマトグラフィー(n-ヘキサン/酢酸エチル=19/1)にて精製し、1,1-diethoxy-3-(2-naphthalenyl)propan-2-one(22)を無色油状物質として得た(675 mg, 2.48 mmol, 49.0%)。

真空下で切削片状マグネシウム(270 mg, 11.1 mmol)をヒートガン用いて加熱乾燥した。室温まで放冷し、アルゴン雰囲気下にした後、THF(8 mL)を加え、これに4-メチルベンジルクロリド(31)(1.35 mL, 10.2 mmol)を室温にてゆっくりと滴下した。反応液は、反応熱によりあたたかくなり、ほとんどのマグネシウム片が消費された。これを室温まで放冷し、(4-methylbenzyl)magnesium chloride(32)のTHF溶液として、そのまま次の反応に用いた。
アルゴン雰囲気下、ジエトキシ酢酸エチル(1.80 mL, 10.1 mmol)のTHF溶液(20 mL)に-78 ℃にて、上記で調製した32のTHF溶液をゆっくりと滴下した。-78 ℃にて1時間撹拌した後、これに20%塩化アンモニウム水溶液(10 mL)を加え、酢酸エチルで抽出した(×3)。有機層を水(×1)と飽和食塩水(×1)で順次洗浄し、無水硫酸ナトリウムで乾燥した。ろ過後、減圧濃縮し、残渣をシリカゲルフラッシュカラムクロマトグラフィー(n-ヘキサン/酢酸エチル=9/1)にて精製し、1,1-diethoxy-3-(4-methylphenyl)propan-2-one(33)を無色油状物質として得た(1.96 g, 8.27 mmol, 82.4%)。

アルゴン雰囲気下、1,1-diethoxy-3-(4-methylphenyl)propan-2-one(33)(216 mg, 914μmol)を1,4-ジオキサン(2 mL)及び水(0.4 mL)に溶解した。これにセレンテラミン(196 mg, 707μmol)を加え、0 ℃に冷却した後、さらに濃塩酸(0.20 mL)を加え、100 ℃で一晩(14時間)攪拌した。室温まで放冷後、減圧濃縮し、残渣をアルゴン気流下でシリカゲルフラッシュクロマトグラフィー(n-ヘキサン/酢酸エチル=1/2→酢酸エチル→酢酸エチル/メタノール=50/1→20/1)で精製し、得られた固体をさらに再沈殿(n-ヘキサン/アセトン)することで、me-coelenterazine(34, me-CTZ)を黄色粉末として得た(180 mg, 427 μmol, 60.4%)。

真空下で切削片状マグネシウム(273 mg, 11.2 mmol)をヒートガン用いて加熱乾燥した。室温まで放冷し、アルゴン雰囲気下にした後、THF(8 mL)を加え、これに4-エチルベンジルクロリド(41)(1.48 mL, 9.95 mmol)を室温にてゆっくりと滴下した。反応液は、反応熱によりあたたかくなり、ほとんどのマグネシウム片が消費された。これを室温まで放冷し、(4-ethylbenzyl)magnesium chloride(42)のTHF溶液として、そのまま次の反応に用いた。

Soc., 130, 7148-7152 (2008) に記載の方法により製造)(7.94 g, 31.4 mmol)をジクロロメタン(150 mL)に溶解し、0 °Cに冷却した。これに、トリエチルアミン(9.10 mL, 66.7 mmol)およびメタンスルホニルクロリド(3.80 mL, 49.1 mmol)を順次加え、室温まで昇温後、25時間撹拌した。これに水を加え、ジクロロメタンで抽出した(×3)。有機層を水(×1)と飽和食塩水(×1)で順次洗浄し、無水硫酸ナトリウムで乾燥した。ろ過後、減圧濃縮し、残渣をシリカゲルフラッシュカラムクロマトグラフィー(n-ヘキサン/酢酸エチル=95/1)にて精製し、3-(tert-butyldimethylsilyloxy)benzyl chloride(102)を無色油状物質として得た(5.90 g, 23.0 mmol, 73.1%)。

アルゴン雰囲気下、4-iodophenylacetic acid(11)(Chen, Q.-H. et al., Bioorg. Med. Chem., 14, 7898-7909 (2006) に記載の方法によって製造)(1.06 g, 4.05 mmol)に塩化チオニル(5.00 mL, 68.6 mmol)を加え、1時間半加熱還流(100 ℃)した。室温まで放冷後、減圧濃縮し、4-iodophenylacetyl chloride(12)を茶色油状の粗生成物として得た。
1-Diazo-3-(4-iodophenyl)propan-2-one(13)(1.27 g, 4.43 mmol)を酢酸(20 mL)に溶解し、0 ℃に冷却した。これに47%臭化水素酸(1.55 mL, 13.3 mmol)を加え、室温まで昇温後、さらに1時間攪拌した。これに飽和炭酸水素ナトリウム溶液を加えて中和した後、酢酸エチルで抽出した(×3)。有機層を飽和炭酸水素ナトリウム溶液(×1)と飽和食塩水(×1)で順次洗浄し、無水硫酸ナトリウムで乾燥した。ろ過後、減圧濃縮し、残渣をシリカゲルフラッシュカラムクロマトグラフィー(n-ヘキサン/酢酸エチル = 7/1)にて精製し、1-bromo-3-(4-iodophenyl)propan-2-one(16)を無色固体として得た(1.42 g, 4.18 mmol, 94.4%)。

1-Bromo-3-(4-iodophenyl)propan-2-one(16)(460 mg, 1.36 mmol)をアセトニトリル(4 mL)に溶解し、これにアセトニトリル(4 mL)に溶かした硝酸銀(596 mg, 3.51 mmol)を加え、室温で一晩(13時間)攪拌した。反応液をセライトろ過し、ろ液を水(×1)と飽和食塩水(×1)で順次洗浄した。有機層を無水硫酸ナトリウムで乾燥し、ろ過後、減圧濃縮し3-(4-iodophenyl)-2-oxopropyl nitrate(17)を無色固体として得た(490 mg, <1.36 mmol, <100%)。これ以上の精製は行わず、そのまま次の反応に用いた。

3-(4-Iodophenyl)-2-oxopropyl nitrate(17)(crude, 450 mg)をジメチルスルホキシド(20 mL)に溶解し、これに酢酸ナトリウム三水和物(211 mg, 1.55 mmol)を加え、室温で1時間攪拌した。これに水を加え、ジエチルエーテルで抽出し(×3)、有機層を水(×1)、飽和炭酸水素ナトリウム水溶液(×1)及び飽和食塩水(×1)で順次洗浄した。無水硫酸ナトリウムで乾燥した後、ろ過し、減圧濃縮することで3-(4-iodophenyl)-2-oxopropanal(18)をオレンジ色固体として得た(crude, 346 mg, <1.26 mmol, <90.1%)。これ以上の精製は行わず、そのまま次の反応に用いた。
アルゴン雰囲気下で3-(4-iodophenyl)-2-oxopropanal(18)(343 mg, 1.25 mmol)を1,4-ジオキサン(2.0 mL)及び水(0.4 mL)に溶解した。これにセレンテラミン(233 mg, 834μmol)を加え、0 ℃に冷却した後、濃塩酸(0.20 mL)を加え、100 ℃で6時間攪拌した。室温まで放冷後、ジクロロメタンと少量のメタノールで抽出した(×4)。有機層を水(×1)と飽和食塩水(×1)で順次洗浄後、無水硫酸ナトリウムで乾燥した。ろ過後、減圧濃縮し、残渣をアルゴン気流下でシリカゲルフラッシュクロマトグラフィー(脱気ジクロロメタン/脱気メタノール = 50/1 → 10/1)で精製した。得られた固体をさらに再沈殿(n-ヘキサン/アセトン)し、i-coelenterazine(15,i-CTZ)を黄土色粉末として得た(52.6 mg, 98.6μmol, 11.8%)。
基質特異性解析用の半合成イクオリンの作製及び発光活性の測定
以下の半合成イクオリンの作製には、チッソ社製の組換えアポイクオリンを使用した。この組換えアポイクオリンは、Inouye, S. and Sahara, Y. Protein Express. Purif. (2007) 53: 384-389に記載の発現ベクターpiP-HEΔEにヒスチジン配列を挿入したpiP-H-HEを用い、同文献に記載の方法に従って発現し、精製したものである。
10 mM EDTAを含む30 mM Tris-HCl(pH7.6)1 ml に 2-メルカプトエタノール 1μl、組換えアポイクオリン溶液(チッソ社製)1.31μgを加えて混合した。次いでエタノールに溶解したセレンテラジン又はその類縁体 1μl を加えて4 ℃で放置し、半合成イクオリンへと変換した。また、アポイクオリンから半合成イクオリンへの再生時間及び再生効率を確認するために、各再生過程(再生開始から0.5時間、1時間、1.5時間、2時間、3時間及び18時間の各時点)で発光活性を測定した。その結果を図1に示した。
上記発光活性の測定は、具体的には、次のように行った。各再生過程の半合成イクオリン溶液2μlに、50 mM 塩化カルシウム溶液を含む50 mM Tris-HCl(pH7.6)100μlを加えることにより発光反応を開始した。発光活性は、発光測定装置Luminescencer-PSN AB2200(アトー社製)にて10秒間測定し、最大発光強度値(Imax)で示した。また、60秒間の発光積算を行ない発光能力とした。
半合成イクオリンの発光パターンと半減時間の測定法
再生した半合成イクオリン溶液を、0.1% BSA(シグマ社製)及び0.01 mM EDTA、150 mM NaCl を含む20 mM Tris-HCl (pH7.6)にて10倍希釈し、96 穴マイクロプレート(Nunc #236108)に5μl/ウェルで分注した。発光プレートリーダーCentro LB960 (ベルトールド社製)を用いて、50 mM 塩化カルシウム溶液を含む50 mM Tris-HCl(pH7.6)を100μl/ウェルで注入することにより発光反応を開始した。60秒間の発光パターンを測定し、発光の半減衰時間(最大発光強度値の半分になるまでの時間)を求めた。その結果を図2及び下記表1にまとめた。
半合成イクオリンの発光スペクトル測定法
光路長10 mmの石英セルに、1 mM EDTAを含む50 mM Tris-HCl(pH7.6)1 ml、再生した半合成イクオリン溶液100μl(100μg蛋白質)を加えて混合し、次いで0.1 mlの10 mM 塩化カルシウム溶液を含む50 mM Tris-HCl(pH7.6)100μlを加えて発光反応を開始させ、蛍光測定装置(日本分光社製、FP-6500)の励起光源をオフにして測定した。測定条件は、バンド幅:20 nm、レスポンス:0.5秒、走査速度:2000 nm/分、22~25℃である。その結果を図3に示した。尚、図3中、AQは、セレンテラジンを発光基質とする半合成イクオリンを示す。
図3に示されているように、me-CTZ、et-CTZ、cf3-CTZ、meo-CTZ、3me-CTZ、3meo-CTZ及び3iso-CTZ を発光基質とする半合成イクオリンは、公知のセレンテラジン類縁体(h-CTZ、及びi-CTZ)と異なる発光スペクトルを示す。
カルシウム標準液の調製
1 g/L の炭酸カルシウム標準液(和光純薬社製)1 mlを50 mM Tris-HCl(pH7.6)9 mlに溶解し、10-3 M 炭酸カルシウム溶液を調製した。
得られた10-3 M 炭酸カルシウム溶液を1 ml 取り、50 mM Tris-HCl(pH7.6)9 mlに加えて、10-4M 炭酸カルシウム溶液を調製した。更に、得られた10-4M 炭酸カルシウム溶液を3 ml取り、50 mM Tris-HCl(pH7.6)6 mlに加えて、3×10-4M 炭酸カルシウム溶液を調製した。得られた10-4M 炭酸カルシウム溶液を1 ml取り、50 mM Tris-HCl(pH7.6)9 mlに加えて、10-5 M 炭酸カルシウム溶液を調製した。更に得られた10-5 M 炭酸カルシウム溶液を3 ml取り、50 mM Tris-HCl(pH7.6)6 mlに加えて、3×10-5 M 炭酸カルシウム溶液を調製した。上記の方法により順次希釈系列を作製し、10-3 M~10-8 Mのカルシウム標準液を調製した。
カルシウム濃度検出用の半合成イクオリンの作製
組換えアポイクオリン(チッソ社製)5 mgを、10 mM DTT及び30 mM EDTAを含む50 mM Tris-HCl(pH7.6)5 mlに溶解し、エタノールに溶解した1.2 倍等量のセレンテラジン類縁体 100μgを加えて4 ℃で一昼夜放置し、半合成イクオリンへと変換した。得られた半合成イクオリンは、アミコンウルトラ-4(ミリポア社製、分子量10.000カット)で濃縮後、0.05 mM EDTAを含む30 mM Tris-HCl (pH7.6)3 mlで3 回洗浄し、余剰のセレンテラジン類縁体を除き、EDTA濃度を0.05 mMとした。
この半合成イクオリン溶液(2.5 mg/ml)を0.1 % BSA(シグマ社製)及び0.01 mM EDTA、150 mM NaCl を含む20 mM Tris-HCl(pH7.6)にて希釈した。
カルシウム標準曲線の作成
上記の様に作製したカルシウム標準液を96 穴マイクロプレート(Nunc #236108)に50μl/ウェル分注し、発光プレートリーダーCentro LB960 (ベルトールド社製)にて、希釈した半合成イクオリン溶液を10μl/ウェル注入して発光強度を60秒間測定し、最大発光強度値(Imax)で示した。それぞれの半合成イクオリンにて同様に測定を行い、得られた各最大発光強度値(Imax)から、各半合成イクオリンのカルシウム標準曲線を作成した。
その結果を図4に示した。
図4に示されているように、本発明のセレンテラジン類縁体(me-CTZ、et-CTZ、cf3-CTZ、meo-CTZ、3me-CTZ, 3meo-CTZ, αmeh-CTZ, 3iso-CTZ)から製造した半合成イクオリンを用いて、カルシウム標準曲線を作成することができることから、本発明の発光蛋白質は、カルシウムイオンの検出又は定量等に使用することができることが分かる。
エビルシフェラーゼの19 kDa蛋白の基質特異性解析及び発光活性の測定法
エビ(Oplophorus)ルシフェラーゼの19 kDa蛋白を、Inouye, S. and Sasaki, S. Protein Express. and Purif. (2007) 56: 261-268に記載の方法で精製し、使用した。
10 mM EDTA を含む30 mM Tris-HCl (pH7.6) 100μlに、1 mM DTTを含むエビルシフェラーゼの19 kDa蛋白(2.3 mg/ml)1μlを溶解し、エタノールに溶解したセレンテラジン又はその類縁体(1μg/μl)1μlを混合して発光反応を開始させ、発光測定装置Luminescencer-PSN AB2200(アトー社製)で60秒間発光活性を測定した。発光活性は、前記した発光活性の測定を3回行い、発光活性の最大発光強度値(Imax)で示した。また、10秒間の発光積算を行ない発光能力とした。
発光スペクトルは、セレンテラジン又はその類縁体5μg (5μl のエタノール溶解)を、10 mM EDTA を含む30 mM Tris-HCl (pH7.6) 990μl に加え、エビルシフェラーゼ20μl (46μg) を添加することにより、発光を開始した。測定は、蛍光測定装置(日本分光社製、FP-6500)の励起光源をオフにして、バンド幅:20 nm、レスポンス:0.5秒、走査速度:2000 nm/分、22~25℃で行なった。発光活性、発光能力、及び最大発光波長についての結果を下記表2に示した。また、発光スペクトルチャートは、図5に示した。
ガウシアルシフェラーゼの基質特異性解析及び発光活性の測定法
ガウシア(Gaussia)ルシフェラーゼを、特開2008-099669号公報記載の方法で精製し、使用した。
0.01% Tween20(シグマ社製)、10 mM EDTA を含むリン酸緩衝生理食塩水(シグマ社製)100μlに、ガウシアルシフェラーゼ(0.16 mg/ml)1μl を溶解し、エタノールに溶解したセレンテラジン又はその類縁体(1μg/μl)1μlを混合して発光反応を開始させ、発光測定装置Luminescencer-PSN AB2200(アトー社製)で10秒間発光活性を測定した。発光活性は、前記した発光活性の測定を3回行い、発光活性の最大発光強度値(Imax)で示した。発光スペクトルは、セレンテラジン又はその類縁体5μg (5μl のエタノール溶解)を、10 mM EDTA及び0.01% Tween20を含むリン酸緩衝生理食塩水 1000μl に加え、ガウシアルシフェラーゼ1μl (0.23μg) を添加することにより、発光を開始した。測定は、蛍光測定装置(日本分光社製、FP-6500)の励起光源をオフにし、バンド幅:20 nm、レスポンス:0.5秒、走査速度:2000 nm/分、22~25℃で行なった。発光活性、発光能力、及び最大発光波長についての結果を下記表3に示した。また、発光スペクトルチャートは、図6に示した。
レニラルシフェラーゼの基質特異性解析及び発光活性の測定法
レニラ(Renilla)ルシフェラーゼを、Inouye, S. & Shimomura, O. Biochem. Biophys. Res. Commun. (1997) 233: 349-353に記載の方法で精製し、使用した。
10 mM EDTA を含む30 mM Tris-HCl (pH7.6) 100μlに、レニラルシフェラーゼ(0.45 mg/ml)1μlを溶解し、エタノールに溶解したセレンテラジン又はその類縁体(1μg/μl)1μlを混合して発光反応を開始させ、発光測定装置Luminescencer-PSN AB2200(アトー社製)で10秒間発光活性を測定した。発光活性は、前記した発光活性の測定を3回行い、発光活性の最大発光強度値(Imax)で示した。発光スペクトルは、セレンテラジン又はその類縁体5μg (5μl のエタノール溶解)を、10 mM EDTA を含む30 mM Tris-HCl (pH7.6) 1000μl、レニラルシフェラーゼ5μl (2.3μg) に添加することにより、発光を開始した。測定は、蛍光測定装置(日本分光社製、FP-6500)の励起光源をオフにし、バンド幅:20 nm、レスポンス:0.5秒、走査速度:2000 nm/分、22~25℃で行なった。発光活性、発光能力、最大発光波長についての結果を下記表4に示した。また、発光スペクトルチャートは、図7に示した。

R2は、水素、水酸基、フッ素、メチル、エチル、プロピル、アダマンチルメチル、シクロペンチルメチル、シクロヘキシルメチル、シクロヘキシルエチル、トリフルオロメチル、メトキシ、エトキシ、n-プロポキシ、iso-プロポキシ、sec-プロポキシ、n-ブトキシ、iso-ブトキシ、sec-ブトキシ、又はtert-ブトキシであることを特徴とする請求項15又は16に記載の方法。 - 一般式(1)において、
R3は、水素、メチル、エチル、プロピル、アダマンチルメチル、シクロペンチルメチル、シクロヘキシルメチル、シクロヘキシルエチル、メトキシ、エトキシ、n-プロポキシ、iso-プロポキシ、sec-プロポキシ、n-ブトキシ、iso-ブトキシ、sec-ブトキシ、又はtert-ブトキシであることを特徴とする請求項15~17のいずれか1項に記載の方法。 - 一般式(1)において、
R4は、フェニル、p-ヒドロキシフェニル、ベンジル、α-ヒドロキシベンジル、フェニルエチル、フェニルビニル、シクロヘキシル、シクロヘキシルメチル、シクロヘキシルエチル、メチル、エチル、プロピル、2-メチルプロピル、2-メチルプロペニル、アダマンチルメチル、シクロペンチルメチル、又はチオフェン-2-イルであること、
を特徴とする、請求項15~18のいずれか1項に記載の方法。 - 一般式(1)において、
R1は、水素、メチル、エチル、トリフルオロメチル、又はメトキシであり、
R2は、水素、水酸基、メチル、又はメトキシであり、
R3は、水素、又はメチルであり、
R4は、ベンジルであり、
R5は、水素であり、
X1は、水酸基であり、
X2は、水素であり、
但し、R2及びR3が水素の時は、R1は、メチル、エチル、トリフルオロメチル、又はメトキシであり、
R1及びR3が水素の時は、R2は、水酸基、メチル、又はメトキシであり、
R1及びR2が水素の時は、R3は、メチルであること、
を特徴とする、請求項15~19のいずれか1項に記載の方法。 - 請求項1~14のいずれか1項に記載の化合物と、カルシウム結合型発光蛋白質のアポ蛋白質とを接触させて、カルシウム結合型発光蛋白質を得ることを含む、カルシウム結合型発光蛋白質の製造方法。
- 請求項21に記載の方法によって製造したカルシウム結合型発光蛋白質を用いることを含む、カルシウムイオンを検出又は定量する方法。
- 請求項21に記載の方法によって製造したカルシウム結合型発光蛋白質をドナー蛋白質として用いて、生物発光共鳴エネルギー移動(BRET)法を行うことを特徴とする、生理機能又は酵素活性の解析方法。
- 請求項1~14のいずれか1項に記載の化合物と、レニラ(Renilla sp.)、エビ(Oplophorus sp.)、又はガウシア(Gaussia sp.)由来のルシフェラーゼとを用いることを含む、転写活性の測定又は目的物質の検出方法。
- 請求項1~14のいずれか1項に記載の化合物と、レニラ(Renilla sp.)、エビ(Oplophorus sp.)、又はガウシア(Gaussia sp.)由来のルシフェラーゼとをドナー蛋白質として用いて、生物発光共鳴エネルギー移動(BRET)法を行うことを特徴とする、生理機能又は酵素活性の解析方法。
- レニラ(Renilla sp.)がRenilla reniformisである、請求項24又は25に記載の方法。
- Renilla reniformis由来のルシフェラーゼが、配列番号14のアミノ酸配列からなるポリペプチドを含む、請求項26に記載の方法。
- エビ(Oplophorus sp.)がOplophorus gracilorostrisである、請求項24又は25に記載の方法。
- Oplophorus gracilorostris由来のルシフェラーゼが、配列番号16のアミノ酸配列からなるポリペプチドを含む、請求項28に記載の方法。
- ガウシア(Gaussia sp.)がGaussia princeps.である、請求項24又は25に記載の方法。
- Gaussia princeps.由来のルシフェラーゼが、配列番号18のアミノ酸配列からなるポリペプチドを含む、請求項30に記載の方法。
- 請求項1~14のいずれか1項に記載の化合物と、レニラ(Renilla sp.)、エビ(Oplophorus sp.)、及びガウシア(Gaussia sp.)からなる群から選択される少なくとも1つの生物由来のルシフェラーゼを含む、転写活性の測定又は目的物質の検出用キット。
- 請求項1~14のいずれか1項に記載の化合物と、レニラ(Renilla sp.)、エビ(Oplophorus sp.)、及びガウシア(Gaussia sp.)からなる群から選択される少なくとも1つの生物由来のルシフェラーゼと、有機化合物及び蛍光蛋白質からなる群から選択される少なくとも1つとを含む、生物発光共鳴エネルギー移動(BRET)法による分子間相互作用の原理を利用した生理機能の解析又は酵素活性の解析用キット。
