例えば、黄色ブドウ球菌(Staphylococcus aureus)は、化膿性疾患、肺炎、食中毒等の起因菌として知られるが、抗生物質メチシリン等の多くの薬剤に対する耐性を獲得したメチシリン耐性黄色ブドウ球菌(MRSA)の出現が、臨床上大きな問題となっている。現在、MRSAに対する代表的な治療薬としては、バンコマイシン、テイコプラニン、アルベカシン、リネゾリドなどが使用されているが、完全にMRSAを排除することは一般に困難であるとされており、また、これらのうち、バンコマイシンについては、既にバンコマイシン耐性黄色ブドウ球菌(VRSA)の出現が報告されており、その使用には十分な注意が必要であるとされている。
このような薬剤耐性菌の問題を克服するために、従来の抗菌剤とは異なった新しい作用機構による微生物に対する新規抗微生物剤の開発が望まれている(例えば、非特許文献1参照)。
細菌の二成分制御系としては、グラム陽性菌のYycFおよびYycGが関与する情報伝達系が存在し、これを阻害すると細菌が死滅することが知られており(例えば、非特許文献3~6参照)、前記情報伝達系を阻害することによるグラム陽性細菌に抗菌力を示す抗菌剤が期待される。
また、ハクサイ、ジャガイモといった農作物に感染し、農業生産に甚大な被害をもたらす軟腐病菌の病原性は3種の二成分制御系PehS/PehR(例えば、非特許文献7参照)、PmrB/PmrA(例えば、非特許文献8参照)、ExpS/ExpA(例えば、非特許文献9参照)によって病原性が調節されていることが知られており、これら病原性を抑制することによる軟腐病菌の防除が期待される。

本発明の化合物、即ち「シグナマイシン(signamycin)A」、「シグナマイシン(signamycin)B」の製造方法は、培養工程と、採取工程とを少なくとも含み、必要に応じてさらにその他の工程を含む。
前記培養工程は、ストレプトマイセス(Streptomyces)属に属し、「シグナマイシン(signamycin)A」、及び「シグナマイシン(signamycin)B」の少なくともいずれかを生産する能力を有する微生物を培養する工程である。
前記栄養培地としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、従来放線菌の培養に利用されている公知のものを使用することができる。
前記栄養培地に添加する栄養源としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、窒素源として、市販されている大豆粉、ペプトン、酵母エキス、肉エキス、コーン・スティープ・リカー、硫酸アンモニウムなどが使用でき、炭素源として、トマトペースト、グリセリン、でん粉、グルコース、ガラクトース、デキストリンなどの炭水化物、脂肪などが使用できる。さらに、食塩、炭酸カルシウムなどの無機塩を培地に添加して使用することもでき、その他、必要に応じて微量の金属塩を培地に添加して使用することもできる。
これらの材料は、シグナマイシン類生産菌が利用し、「シグナマイシン(signamycin)A」、及び「シグナマイシン(signamycin)B」の少なくともいずれかの生産に役立つものであればよく、公知の培養材料はすべて用いることができる。
前記培養の温度としては、シグナマイシン類生産菌の発育が実質的に阻害されずに、「シグナマイシン(signamycin)A」、及び「シグナマイシン(signamycin)B」の少なくともいずれかを生産しうる範囲であれば、特に制限はなく、使用する生産菌に応じて適宜選択することができるが、25℃~35℃が好ましい。
前記培養の期間としては、特に制限はなく、「シグナマイシン(signamycin)A」、及び「シグナマイシン(signamycin)B」の少なくともいずれかの蓄積に合わせて適宜選択することができる。通常、培養3日間~10日間で「シグナマイシン(signamycin)A」、及び「シグナマイシン(signamycin)B」の少なくともいずれかの蓄積が最高となる。
前記採取工程は、前記培養工程で得られた培養物から「シグナマイシン(signamycin)A」、及び「シグナマイシン(signamycin)B」の少なくともいずれかを採取する工程である。
前記「シグナマイシン(signamycin)A」、及び「シグナマイシン(signamycin)B」は、上述した物理化学的性状を有するので、その性状に従って培養物から採取することができる。
また、分離した菌体からは、適当な有機溶媒を用いた抽出方法や、菌体破砕による溶出方法などにより、前記「シグナマイシン(signamycin)A」や、「シグナマイシン(signamycin)B」を菌体から抽出し、上記と同様に単離精製して採取することができる。
本発明の微生物は、ストレプトマイセス(Streptomyces)属に属し、上述した本発明の化合物、即ちシグナマイシン(signamycin)A、及びシグナマイシン(signamycin)Bの少なくともいずれかを生産する能力を有することを特徴とする。前記微生物は、シグナマイシン(signamycin)A、及びシグナマイシン(signamycin)Bの少なくともいずれかを生産する能力を有し、そのために、上述した本発明の化合物の製造方法において、シグナマイシン(signamycin)A、及びシグナマイシン(signamycin)Bの少なくともいずれかの生産菌として使用され得る微生物であれば、特に制限はなく、目的に応じて適宜選択することができる。
MK851-mF8株は、分枝した基生菌糸より、比較的長い気菌糸を伸長し、その先端は8回転~10回転のらせんを形成する。成熟した胞子鎖は10個~50個の卵円形~円筒形の胞子を連鎖する。胞子の大きさは約0.5μm~0.6μm×0.9μm~1.1μmで、胞子の表面はとげ状である。輪生枝、菌糸束、胞子のう、及び運動性胞子は認められない。
色の記載について[ ]内に示す標準は、コンティナー・コーポレーション・オブ・アメリカのカラー・ハーモニー・マニュアル(Container Corporation of Americaのcolor harmony manual)を用いた。
(1)イースト・麦芽寒天培地(ISP-培地2、27℃培養)
赤茶[7 pi, Dk Wine]の発育上に、明るい茶灰[4 ge, Lt Fawn]の気菌糸を着生し、赤茶の可溶性色素を産生する。発育の色、及び可溶性色素は、0.1モル塩酸の添加でにぶ赤紫に変化するが、0.1モル水酸化ナトリウムの添加による変化は認められない。
(2)オートミール寒天培地(ISP-培地3、27℃培養)
灰味黄茶[3 ni, Clove Brown]の発育上に、灰白[b, Oyster White]の気菌糸をうっすらと着生する。可溶性色素は、かすかに灰味赤を呈する。
(3)スターチ・無機塩寒天培地(ISP-培地4、27℃培養)
にぶ黄[2 ne, Mustard Gold]の発育上に、黄味灰[2 ca, Lt Ivory]~明るい灰[d]の気菌糸を着生する。可溶性色素は、かすかにうす黄だいだいを呈する。0.1モル塩酸、及び0.1モル水酸化ナトリウムの添加による発育の色、及び可溶性色素の変化は認められない。
(4)グリセリン・アスパラギン寒天培地(ISP-培地5、27℃培養)
にぶ黄[2 ne, Mustard Gold~3 ne, Topaz]の発育上に、黄味白の気菌糸をうっすらと着生し、可溶性色素はうす赤を帯びる。
(5)チロシン寒天培地(ISP-培地7、27℃培養)
黄茶[3 ng, Yellow Maple~3 pi, Golden Brown]の発育上に、白の気菌糸をわずかに着生し、可溶性色素は茶色味を帯びる。
(6)シュクロース・硝酸塩寒天培地(27℃培養)
うす黄[2 gc, Bamboo]の発育上に、白の気菌糸をうっすらと着生し、可溶性色素は認められない。
(1)生育温度範囲
イースト・スターチ寒天培地(溶性デンプン 1.0%、イーストエキス 0.2%、ひも寒天 2.6%、pH7.0)を用い、10℃、20℃、24℃、27℃、30℃、37℃、及び50℃の各温度で試験した結果、10℃、50℃での生育は認められず、20℃~37℃の範囲で生育した。生育至適温度は、30℃付近である。
(2)スターチの加水分解(スターチ・無機塩寒天培地、ISP-培地4、27℃培養)
培養後5日目にはスターチの加水分解が認められ、その作用は中等度である。
(3)メラニン様色素の生成(トリプトン・イースト・プロス、ISP-培地1;ペプトン・イースト・鉄寒天培地、ISP-培地6;チロシン寒天培地、ISP-培地7;いずれも27℃培養)
ペプトン・イースト・鉄寒天培地、及びチロシン寒天培地では陽性である。トリプトン・イースト・プロスでは判然としない。
(4)炭素源の利用性(プリドハム・ゴドリーブ寒天培地、ISP-培地9;27℃培養)
D-グルコース、L-アラビノース、D-フルクトース、スクロース、イノシトール、ラムノース、ラフィノース、及びD-マンニトールを利用して発育し、D-キシロースもおそらく利用する。
細胞壁中の2,6-ジアミノピメリン酸はLL-型である。
16S rRNA遺伝子の部分塩基配列(1481bp)を決定し、DNAデータベースに登録された公知菌株のデータと比較した。その結果、MK851-mF8株の塩基配列は以下に示すように、ストレプトマイセス(Streptomyces)属放線菌の16S rRNA遺伝子と高い相同性を示した。即ち、Streptomyces canus(99%)、S. ciscaucasicus(99%)、S. viridochromogenes(99%)、S. pseudovenezuelae(99%)、S. purpureofuscus subsp. acoagulans(99%)、S. resistomycificus(99%)、S. roseogriseus(99%)、S. panayensis(99%)等である。なお、括弧内は塩基配列の相同値を表記した。
MK851-mF8株の細胞壁中の2,6-ジアミノピメリン酸はLL-型である。
MK851-mF8株の16S rRNA遺伝子の部分塩基配列を解析し、公知菌株のデータと比較したところ、ストレプトマイセス属放線菌と高い相同性を示した。
なお、前記MK851-mF8株は、独立行政法人製品評価技術基盤機構 特許微生物寄託センターに寄託申請し、平成20年7月23日、NITE P-612として受託された。
-化合物含有組成物-
本発明の化合物含有組成物は、上述した本発明の化合物、即ちシグナマイシン(signamycin)A、及びシグナマイシン(signamycin)Bの少なくともいずれかを含み、必要に応じて適宜その他の成分を含む。
なお、前記化合物含有組成物は、単独で使用されてもよいし、他の成分を有効成分とする医薬と併せて使用されてもよい。また、前記化合物含有組成物は、他の成分を有効成分とする医薬中に、配合された状態で使用されてもよい。
本発明の抗菌剤は、上述した本発明の化合物、即ちシグナマイシン(signamycin)A、及びシグナマイシン(signamycin)Bの少なくともいずれかを含み、必要に応じて適宜その他の成分を含む。
なお、前記抗菌剤は、単独で使用されてもよいし、他の成分を有効成分とする医薬と併せて使用されてもよい。また、前記抗菌剤は、他の成分を有効成分とする医薬中に、配合された状態で使用されてもよい。
したがって、前記抗菌剤は、薬剤耐性菌などに起因する感染症の予防、又は治療に好適に利用可能である。また、前記抗菌剤は、農園芸用殺菌剤としても好適に利用可能である。
本発明の酵素活性阻害剤は、上述した本発明の化合物、即ちシグナマイシン(signamycin)A、及びシグナマイシン(signamycin)Bの少なくともいずれかを含み、必要に応じて適宜その他の成分を含む。
前記酵素活性阻害剤は、ヒスチジンキナーゼ活性を好適に阻害することができる。
なお、前記酵素活性阻害剤は、単独で使用されてもよいし、他の成分を有効成分とする医薬と併せて使用されてもよい。また、前記酵素活性阻害剤は、他の成分を有効成分とする医薬中に、配合された状態で使用されてもよい。
したがって、前記酵素活性阻害剤は、薬剤耐性菌などを含む幅広いグラム陽性細菌、及びグラム陰性細菌の病原性を抑制することができ、前記細菌に起因する感染症の予防、又は治療に好適に利用可能である。また、農園芸用殺菌剤の有効成分としても好適に利用可能である。
前記化合物含有組成物、抗菌剤、及び酵素活性阻害剤の剤型としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、粉末状、カプセル状、錠剤状、液状等の剤型とすることができる。これらの剤型の前記化合物含有組成物、抗菌剤、及び酵素活性阻害剤は、常法に従い製造することができる。
また、乳剤、水和剤、液剤、フロアブル(ゾル)剤、粉剤、粒剤、微粒剤、錠剤などの目的で各種の界面活性剤(または、乳化剤)が使用される。このような界面活性剤としては、非イオン型(ポリアルキルエーテル、ポリオキシエチレンアルキルエステル、ポリオキシエチレンソルビタンアルキルエステルなど)、陰イオン型(アルキルオコシエチレンアルキルサルフェート、アリールスルホネートなど)、陽イオン型(アルキルアミン類、ポリオキシアルキルアミン類など)、両性型(硫酸エステル塩など)が挙げられるが、もちろんこれらの例示のみに限定されるものではない。また、これらの他にポリビニルアルコール、カルボキシメチルセルロース、アラビアゴム、ポリビニルアセテート、アルギン酸ソーダ、ゼラチン、トラガカントガムなどの各種補助剤を使用することができる。
前記化合物含有組成物、抗菌剤、及び酵素活性阻害剤の投与方法としては、特に制限はなく、例えば、前記化合物含有組成物、抗菌剤、及び酵素活性阻害剤の剤型などに応じて適宜選択することができ、経口又は非経口で投与することができる。
前記化合物含有組成物、抗菌剤、及び酵素活性阻害剤の投与量としては、特に制限はなく、投与対象個体の年齢、体重、体質、症状、他の成分を有効成分とする医薬の投与の有無など、様々な要因を考慮して適宜選択することができる。
前記化合物含有組成物、抗菌剤、及び酵素活性阻害剤の投与時期としては、特に制限はなく、目的に応じて適宜選択することができる。
前記化合物含有組成物、抗菌剤、及び酵素活性阻害剤の投与対象となる動物種としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、ヒト、サル、ブタ、ウシ、ヒツジ、ヤギ、イヌ、ネコ、マウス、ラット、トリなどが挙げられる。
また、前記農園芸用殺菌剤として用いる場合の投与方法、投与量、投与時期、及び投与対象としては、特に制限はなく、目的に応じて適宜選択することができる。
-培養工程-
寒天斜面培地に培養したストレプトマイセス エスピー(Streptomyces sp.)MK851-mF8株(NITE P-612として寄託)を、ガラクトース 2%、デキストリン 2%、グリセリン 1%、バクトソイトン(ディフコ社製) 1%、コーン・スティープ・リカー 0.5%、硫酸アンモニウム 0.2%、炭酸カルシウム 0.2%を含む液体培地(pH7.0に調整)を三角フラスコ(500mL容)に110mLずつ分注して、常法により120℃で20分滅菌した培地に接種した。その後に30℃で4日間回転振とう培養し、種母培養液を得た。
このようにして得られた培養液3リットルを遠心分離して、培養ろ液と菌体に分離した。続いて、菌体に1リットルのメタノールを加えてよく撹拌し、菌体からシグナマイシンAと、シグナマイシンBとをメタノールで抽出し、シグナマイシンA、及びシグナマイシンBを含む菌体抽出液1.37リットルを得た。菌体抽出液1.37リットルに等量の水を加え、十分撹拌した後、ダイヤイオンCHP20P(内径60mm×220mm、三菱化学社製)カラムに吸着させ、1.8リットルの50%メタノール水で洗浄した後、1.8リットルの80%メタノール水でシグナマイシンA、及びシグナマイシンBを含む活性画分を溶出した。得られた80%メタノール水1.8リットルは、減圧下で濃縮乾固して、シグナマイシンA、及びシグナマイシンBを含む粗精製物0.842gを得た。
前記粗精製物660mgを少量のメタノールに溶解し、C18逆層カラムクロマトグラフィー(Capcell pak UG120、内径30mm×長さ250mm、資生堂製)でシグナマイシンAと、シグナマイシンBとを分離した。即ち、展開溶媒としてアセトニトリル:水:トリフルオロ酢酸=60:40:0.001を用い、流速15mL/分でクロマトグラフィーを行うと、シグナマイシンAは33分~34分に、シグナマイシンBは42分~48分に溶出し、これらを集めて減圧下で濃縮乾固し、純粋なシグナマイシンAを22.5mgと、シグナマイシンBを206.4mgとを得た。

図6に、シグナマイシンBの重クロロホルム中で30℃にて測定した、150MHzにおける炭素13核磁気共鳴スペクトルのチャートを示す。
薬剤耐性菌(メチシリン耐性菌、バンコマイシン耐性菌等)を含む各種の微生物に対するシグナマイシンA、及びシグナマイシンBの抗菌スペクトルを、日本化学療法学会標準法に基づき、ミュラ・ヒントン寒天培地上で倍数希釈法により測定した。最小発育阻止濃度(MIC)の測定結果を表1に示す。
中でも、シグナマイシンAは、黄色ブドウ球菌(S. aureus)に対して抗菌活性が高く、シグナマイシンBは、黄色ブドウ球菌(S. aureus)と、腸球菌(E. faecalis、E. faecium)に対して抗菌活性が高かった。
-(1)VicKヒスチジンキナーゼ活性阻害試験-
う蝕菌(Streptococcus mutans)のVicKに対する、シグナマイシンA、及びシグナマイシンBの酵素阻害活性を調べた。
ヒスチジンキナーゼ活性の測定は、Biosci. Biotechnol. Biochem., 64, 919-923, 2000に報告された方法を改変して行った。
VicKのキナーゼ活性ドメインのみを含む領域(N-末端から31番目のアミノ酸から450番目のアミノ酸を含む)をPCR法により、う蝕菌の染色体DNAから調製し、発現ベクターpET21a(+)にクローニングした。このようにして得られたプラスミドpET-SMvicK31-450を大腸菌に形質転換した株の培養液から、VicKのヒスチジンキナーゼ活性ドメインのみを発現させたタンパク質(VicK-31-450)を精製した。
ヒスチジンキナーゼ活性測定のための反応溶液の組成は、0.5M VicK-31-450、50mM Tris-HCl(pH7.5)、50mM KCl、10mM MgCl2であり、この反応液 7μLにシグナマイシンA、又はシグナマイシンBを 1μL加え、25℃で5分間インキュベートした。その後、[32P]ATPを含む12.5M ATPを2μL加え(終濃度2.5M)反応を開始し、25℃で20分間インキュベート後、反応を終了させ、SDS-ポリアクリルアミドゲル電気泳動を行うことによって、う蝕菌のVicKに対する50%阻害濃度(IC50)を求めた。結果を表2に示す。
枯草菌168株(B. subtilis 168)のYycGに対する、シグナマイシンA、及びシグナマイシンBの酵素阻害活性を調べた。
ヒスチジンキナーゼ活性の測定は、Biosci. Biotechnol. Biochem., 64, 919-923, 2000に報告された方法にしたがって行った。
YycGのキナーゼ活性ドメインのみを含む領域(N-末端から204番目のアミノ酸から611番目のアミノ酸を含む)をPCR法により枯草菌168株の染色体DNAから調製し、発現ベクターpET-21a(+)にクローニングした。このようにして得られたプラスミドpET-yycGtruを大腸菌に形質転換した株の培養液から、YycGのヒスチジンキナーゼ活性ドメインのみを発現させたタンパク質(YycG-204-611)を精製した。
ヒスチジンキナーゼ活性測定のための反応溶液の組成は、0.5μM YycG-204-611、50mM Tris-HCl(pH8.5)、100mM KCl、100mM NH4Cl、5mM MgCl2であり、この反応溶液に2.5μM ATP-10μCi[γ-32P]ATP混合溶液を加えて10μLとし、反応を開始し、30℃で10分間インキュベート後、反応を終了させ、SDS-ポリアクリルアミド電気泳動を行った。阻害活性を調べる際は、ATP混合溶液を加える前に所定の濃度のシグナマイシンA、又はシグナマイシンBを反応溶液中に加え、30℃で5分間インキュベートし、枯草菌のYycGに対する50%阻害濃度(IC50)を求めた。結果を表2に示す。
軟腐病菌MAFF301393株(Erwinia carotovora subsp. carotovora MAFF301393)のPehSに対する、シグナマイシンA、及びシグナマイシンBの酵素阻害活性を調べた。
ヒスチジンキナーゼ活性の測定は、Biosci. Biotechnol. Biochem., 64, 919-923, 2000に報告された方法にしたがって行った。
PehSのキナーゼ活性ドメインのみを含む領域(N-末端から209番目のアミノ酸から484番目のアミノ酸を含む)をPCR法により軟腐病菌MAFF301393株の染色体DNAから調製し、発現ベクターpET-21a(+)にクローニングした。このようにして得られたプラスミドpET-pehScM2-2を大腸菌に形質転換した株の培養液から、PehSヒスチジンキナーゼ活性ドメインのみを発現させたタンパク質(PehS-209-484)を精製した。
ヒスチジンキナーゼ活性測定のための反応溶液の組成は、4μM PehS-209-484、50mM Tris-HCl(pH8.5)、100mM KCl、100mM NH4Cl、5mM MgCl2であり、この反応溶液に2.5μM ATP-10μCi[γ-32P]ATP混合溶液を加えて10μLとし、反応を開始し、30℃で20分間インキュベート後、反応を終了させ、SDS-ポリアクリルアミド電気泳動を行った。阻害活性を調べる際は、ATP混合溶液を加える前に所定の濃度のシグナマイシンA、又はシグナマイシンBを反応溶液中に加え、30℃で5分間インキュベートし、軟腐病菌のPehSに対する50%阻害濃度(IC50)を求めた。結果を表2に示す。
Claims (9)
- 請求の範囲第1項に記載の化合物、及び請求の範囲第2項に記載の化合物の少なくともいずれかの製造方法であって、
ストレプトマイセス(Streptomyces)属に属し、請求の範囲第1項に記載の化合物、及び請求の範囲第2項に記載の化合物の少なくともいずれかを生産する能力を有する微生物を培養する培養工程と、
前記培養工程で得られた培養物から請求の範囲第1項に記載の化合物、及び請求の範囲第2項に記載の化合物の少なくともいずれかを採取する採取工程と
を含むことを特徴とする化合物の製造方法。 - ストレプトマイセス(Streptomyces)属に属し、請求の範囲第1項に記載の化合物、及び請求の範囲第2項に記載の化合物の少なくともいずれかを生産する能力を有する微生物が、受託番号NITE P-612のストレプトマイセス エスピー(Streptomyces sp.)MK851-mF8株である請求の範囲第3項に記載の化合物の製造方法。
- ストレプトマイセス(Streptomyces)属に属し、請求の範囲第1項に記載の化合物、及び請求の範囲第2項に記載の化合物の少なくともいずれかを生産する能力を有することを特徴とする微生物。
- 受託番号NITE P-612のストレプトマイセス エスピー(Streptomyces sp.)MK851-mF8株である請求の範囲第5項に記載の微生物。
- 請求の範囲第1項に記載の化合物、及び請求の範囲第2項に記載の化合物の少なくともいずれかを含むことを特徴とする化合物含有組成物。
- 請求の範囲第1項に記載の化合物、及び請求の範囲第2項に記載の化合物の少なくともいずれかを含むことを特徴とする抗菌剤。
- 請求の範囲第1項に記載の化合物、及び請求の範囲第2項に記載の化合物の少なくともいずれかを含むことを特徴とする酵素活性阻害剤。
